Absorbed lipid residues from 24 seventh- to ninth-century coarseware potsherds from the major Anglo- Saxon trading centre of Hamwic (Southampton, UK) were analysed by gas chromatography - mass spectrometry ( GC- MS) in order to reconstruct the dietary habits of its population. The results show that the vessels were used for preparing ruminant fats and leafy vegetables. In addition, evidence was found for a minor contribution of aquatic foods. Beeswax was found once and most probably relates to a sealing function or to honey. Remarkable features were: (i) the isomeric mixture of octadecenoic acid ( C18:1 Δ7,8,9,10,11,12,13,14,15,16) and 8- to 16-hydroxyoctadecanoic acid, and (ii) the co-occurrence of C17:1, C19:1 and isoprenoid fatty acids. These features were proposed as biomarkers for ruminant and aquatic food sources, respectively. Furthermore, the carbonyl position distribution in mid-chain ketones was used to identify mixtures of animal- and plant-derived ketones. The paper highlights the difficulty in interpreting complex lipid signatures that show a mixture of various foods, as observed in the majority of the samples. This was linked to the preparation of stews or the recycling of vessels. Background and aims: The United Kingdom (UK) Psychoactive Substances Act (PSA), implemented on the 26th May 2016, made the production, supply and sale of all non‐exempted psychoactive substances illegal. The aim of this study was to measure trends in hospital presentations for severe toxicity following analytically confirmed synthetic cannabinoid receptor agonist (SCRA) exposure before and after implementation of the PSA. Design: Observational study. Setting: Thirty‐four hospitals across the UK participating in the Identification of Novel Psychoactive Substances (IONA) study. Participants: A total of 627 (79.9% male) consenting individuals who presented to participating hospitals between July 2015 and December 2019 with severe acute toxicity and suspected novel psychoactive substances exposure. Measurements Toxicological analyses of patient samples were conducted using liquid‐chromatography tandem mass‐spectrometry. Time‐series analysis was conducted on the monthly number of patients with and without analytically confirmed SCRA exposure using Poisson segmented regression. Findings SCRAs were detected in 35.7% (n = 224) of patients. After adjusting for seasonality and the number of active sites, models showed no clear evidence of an upward or downward trend in the number of SCRA exposure cases in the period before (incidence rate ratio [IRR], 1.12; 95% CI, 0.99–1.26; P = 0.068) or after (IRR, 0.97; 95% CI, 0.94–1.01; P = 0.202) the implementation of the PSA. There was also no clear evidence of an upward or downward trend in non‐SCRA exposure cases before (IRR, 1.12; 95% CI, 0.98–1.27; P = 0.105) or after (IRR, 1.01; 95% CI, 0.98–1.04; P = 0.478) implementation of the PSA. Conclusions: There is no clear evidence of an upward or downward trend in the number of patients presenting to UK hospitals with severe acute toxicity following analytically confirmed synthetic cannabinoid receptor agonist exposure since the implementation of the Psychoactive Substances Act. Wearable physical activity (PA) monitors have improved the ability to estimate free‐living total energy expenditure (TEE) but their application during arduous military training alongside more well‐established research methods has not been widely documented. This study aimed to assess the validity of two wrist‐worn activity monitors and a PA log against doubly labeled water (DLW) during British Army Officer Cadet (OC) training. For 10 days of training, twenty (10 male and 10 female) OCs (mean ± SD: age 23 ± 2 years, height 1.74 ± 0.09 m, body mass 77.0 ± 9.3 kg) wore one research‐grade accelerometer (GENEActiv, Cambridge, UK) on the dominant wrist, wore one commercially available monitor (Fitbit SURGE, USA) on the non‐dominant wrist, and completed a self‐report PA log. Immediately prior to this 10‐day period, participants consumed a bolus of DLW and provided daily urine samples, which were analyzed by mass spectrometry to determine TEE. Bivariate correlations and limits of agreement (LoA) were employed to compare TEE from each estimation method to DLW. Average daily TEE from DLW was 4112 ± 652 kcal·day−1 against which the GENEActiv showed near identical average TEE (mean bias ± LoA: −15 ± 851 kcal.day−1) while Fitbit tended to underestimate (−656 ± 683 kcal·day−1) and the PA log substantially overestimate (+1946 ± 1637 kcal·day−1). Wearable physical activity monitors provide a cheaper and more practical method for estimating free‐living TEE than DLW in military settings. Standard mode and collision/reaction cell mass spectroscopy methods have been utilized in order to overcome spectral interferences and provide ultra‐low quantification of selenium (Se) and tellurium (Te) in British Carboniferous coals for the first time. The accurate detection of Se and Te in coals is becoming increasingly important, as coals and pyrite have been identified as potentially significant trace element sources. The mean Se concentration of British coals bear comparison to that of world coals, with anomalous Se content (concentrations above 4 mg/kg) across westerly exposures, often coinciding with high sulphur (S) content and visible pyrite. New Te data for British coals gives a mean concentration of 0.02 mg/kg, with anomalous Te in Ayrshire. There is a positive correlation in the Te/Se ratio across the sample set. The close relationship between Se and Te, as well as Se–Te with both early syngenetic and later cleat‐filling pyrite, confirms an important role for sulphides in Se and Te sequestration in British coals. The high Se‐Bowland Shale and/or Ordovician volcanics may have provided the trace element source for British coals of similar or younger age. Regional intrusive activity (shallow tabular intrusions or more extensive plutons) and episodes of intense deformation can alter the thermal maturity of coals and may have driven the movement of trace element‐rich fluids through strata, locally enriching coals in Se and Te. Aim To determine the levels of LL-37 in and its susceptibility to degradation by components of gingival crevicular fluid ( GCF) in periodontal health and disease. Materials and Methods Levels of LL-37 in GCF from periodontitis patients and periodontally healthy subjects were determined by ELISA. In addition, degradation of synthetic/exogenous LL-37 by components of GCF in the presence and absence of inhibitors was determined by matrix-assisted laser desorption/ionization time of flight mass spectrometry. Results The concentration of native LL-37 in GCF from Porphyromonas gingivalis positive (Pg+) and P. gingivalis negative (Pg-) sites in periodontitis patients was significantly higher than in GCF from healthy subjects. When synthetic LL-37 was added to healthy GCF, the peptide was not degraded. Conversely, GCF from Pg+ sites rapidly degraded synthetic LL-37 which was prevented in the presence of Arg− and Lys− gingipain inhibitors. Synthetic LL-37 was degraded more slowly by GCF from Pg− sites. Conclusions LL-37 is detectable in GCF in periodontal health and disease. The rapid degradation of synthetic LL-37 in periodontitis GCF, particularly in Pg+ sites, limits its role as a potential therapeutic in the gingival crevice. The article reports on research which was conducted to investigate the pharmacokinetics of intranasal crushed oxycontin and intravenous oxycodone in nondependent prescription opioid abusers. Researchers evaluated eight healthy volunteers. They found that the intranasal and intravenous methods of delivery of the drugs resulted in the drugs getting into the subjects' bloodstreams faster than if they had been administered in tablet form and that all drug doses were well tolerated by subjects.

ABSTRACT Aims To assess the patterns of use, subjective effect profile and dependence liability of mephedrone, supported by corroborative urine toxicology. Design Cross-sectional structured telephone interview. Setting UK-based drug users associated with the dance music scene. Participants A total of 100 mephedrone users, recruited through their involvement with the dance music scene. Measurements Assessment of pattern of use, acute and after effects, DSM dependence criteria and gas chromatography-mass spectrometry urinalysis. Findings Mephedrone consumption results in typical stimulant-related subjective effects: euphoria, increased concentration, talkativeness, urge to move, empathy, jaw clenching, reduced appetite and insomnia. Thirty per cent of the sample potentially met criteria for DSM-IV dependence and there was evidence of a strong compulsion to use the drug (47% had used the drug for 2 or more consecutive days). Self-reported recent consumption of mephedrone was confirmed by toxicological analysis in all of the 14 participants who submitted a urine sample. Conclusion Mephedrone has a high abuse and health risk liability, with increased tolerance, impaired control and a compulsion to use, the predominant reported dependence symptoms. Tigecycline belongs to a new class of tetracyclines, the glycylcyclines, less than 20% of which is metabolized in the liver. Twenty-five patients with cirrhosis with varying degrees of functional hepatic reserve (Child-Pugh A, n = 10; B, n = 10; C, n = 5) and 23 healthy adults, matched by age, sex, weight, and smoking habits, received 100 mg of tigecycline infused intravenously over 60 minutes. Serum and urine samples were collected up to 120 hours after dosing. Pharmacokinetic data were derived using non-compartmental methods. The most common treatment-emergent adverse events in healthy volunteers were nausea (56.5%), vomiting (21.7%), and headache (21.7%) and in the patients with cirrhosis, albuminuria (12%). Mean (±1 SD) tigecycline clearance values were 29.8 ± 11.3 L/h in healthy subjects and 31.2 ± 13.9 L/h (Child-Pugh A), 22.1 ± 9.3 L/h (Child-Pugh B), and 13.5 ± 2.7 L/h (Child-Pugh C) in the patients. A single intravenous dose of tigecycline 100 mg was safe and well-tolerated in patients with cirrhosis with varying degrees of hepatic functional reserve. No adjustment of tigecycline maintenance dosage is warranted in patients with compensated or moderately decompensated cirrhosis; doses should be reduced by 50%, to 25 mg, every 12 hours in patients with severely decompensated disease.
