Abstract Objective This study aims to comprehensively analyze the clinical characteristics and identify the differentially expressed genes associated with drug‐resistant epilepsy (DRE) in patients with focal cortical dysplasia (FCD). Methods A retrospective investigation was conducted from July 2019 to June 2022, involving 40 pediatric cases of DRE linked to FCD. Subsequent follow‐ups were done to assess post‐surgical outcomes. Transcriptomic sequencing and quantitative reverse transcription polymerase chain reaction (qRT‐PCR) were used to examine differential gene expression between the FCD and control groups. Results Among the 40 patients included in the study, focal to bilateral tonic–clonic seizures (13/40, 32.50%) and epileptic spasms (9/40, 22.50%) were the predominant seizure types. Magnetic resonance imaging (MRI) showed frequent involvement of the frontal (22/40, 55%) and temporal lobes (12/40, 30%). In cases with negative MRI results (13/13, 100%), positron emission tomography/computed tomography (PET‐CT) scans revealed hypometabolic lesions. Fused MRI/PET‐CT images demonstrated lesion reduction in 40.74% (11/27) of cases compared with PET‐CT alone, while 59.26% (16/27) yielded results consistent with PET‐CT findings. FCD type II was identified in 26 cases, and FCD type I in 13 cases. At the last follow‐up, 38 patients were prescribed an average of 1.27 ± 1.05 anti‐seizure medications (ASMs), with two patients discontinuing treatment. After a postoperative follow‐up period of 23.50 months, 75% (30/40) of patients achieved Engel class I outcome. Transcriptomic sequencing and qRT‐PCR analysis identified several genes primarily associated with cilia, including CFAP47, CFAP126, JHY, RSPH4A, and SPAG1. Significance This study highlights focal to bilateral tonic–clonic seizures as the most common seizure type in patients with DRE due to FCD. Surgical intervention primarily targeted lesions in the frontal and temporal lobes. Patients with FCD‐related DRE showed a promising prognosis for seizure control post‐surgery. The identified genes, including CFAP47, CFAP126, JHY, RSPH4A, and SPAG1, could serve as potential biomarkers for FCD. Plain Language Summary This study aimed to comprehensively evaluate the clinical data of individuals affected by focal cortical dysplasia and analyze transcriptomic data from brain tissues. We found that focal to bilateral tonic–clonic seizures were the most prevalent seizure type in patients with drug‐resistant epilepsy. In cases treated surgically, the frontal and temporal lobes were the primary sites of the lesions. Moreover, patients with focal cortical dysplasia‐induced drug‐resistant epilepsy exhibited a favorable prognosis for seizure control after surgery. CFAP47, CFAP126, JHY, RSPH4A, and SPAG1 have emerged as potential pathogenic genes for the development of focal cortical dysplasia.

Abstract Objective Defects in RARS2 cause cerebellopontine hypoplasia type 6 (pontocerebellar hypoplasia type 6, PCH6, OMIM: #611523), a rare autosomal recessive inherited mitochondrial disease. Here, we report two male patients and their respective family histories. Methods We describe the clinical presentation and magnetic resonance imaging (MRI) findings of these patients. Whole‐exome sequencing was used to identify the genetic mutations. Results One patient showed hypoglycemia, high lactic acid levels (fluctuating from 6.7 to 14.1 mmol/L), and frequent seizures after birth, with progressive atrophy of the cerebrum, cerebellum, and pons. The other patient presented with early infantile developmental and epileptic encephalopathies (EIDEEs) with an initial developmental delay followed by infantile epileptic spasm syndrome (IESS) at 5 months old, with no imaging changes. Whole‐exome sequencing identified compound heterozygous RARS2 variants c.25A>G (p.I9V) with c.1261C>T (p.Q421*) and c.1A>G (p.M1V) with c.122A>G (p.D41G) in these two patients. Of these loci, c.1261C>T and c.122A>G have not been previously reported. Significance Our findings have expanded the RARS2 gene variant spectrum and present EIDEEs and IESS as phenotypes which deepened the association between PCH6 and RARS2. Abstract Continuous cultivation of faba beans often results in a high occurrence of Fusarium wilt. Nevertheless, this issue can be successfully managed through wheat‐faba bean intercropping. Our objective is to elucidate the function of non‐host wheat in combating faba bean Fusarium wilt. We assessed the impact of wheat on the development of faba bean Fusarium wilt through field and pot experiments, and examined and identified the compounds in the root exudates of wheat. The influence of wheat root exudates on Fusarium commun mycelial growth and spore multiplication was investigated through indoor culture experiments. Wheat‐faba bean intercropping significantly reduced the occurrence of Fusarium wilt in field trials. Root separation experiments in pots indicated that wheat root exudates might play a vital role in this outcome. The exogenous addition of wheat root exudates to faba bean grown alone notably decreased the occurrence and severity of Fusarium wilt. Furthermore, the exogenous addition of wheat root exudates effectively hindered the growth and reproduction of Fusarium commun's mycelium. And, wheat root exudates contained six compounds with relatively high concentrations, including salicylic acid, p‐hydroxybenzoic acid, tartaric acid, malic acid, glutamic acid, and threonine, all of which inhibited the growth and spore formation of Fusarium commun. In a wheat‐faba bean intercropping system, these six compounds found in the root exudate of non‐host wheat can help control Fusarium wilt in faba bean by inhibiting the mycelial growth and sporulation of Fusarium commun.

Abstract In this paper, an optimal planning strategy is proposed for EV charging stations by considering travel demand based on non‐cooperative game theory. In the proposed methodology, a realistic and stochastic charging demand distribution was obtained with the travel chain of an EV and the associated probability density function. Based on the charging demand of EV owners and a multi‐operator scenario, a two‐stage effective and practical charging station planning model was established. The first stage of the planning model optimized the location of charging stations to decrease the total deadhead mileage of the EVs with the cooperation of the charging station operators. In the second stage, the operators were committed to increasing their economic benefits by developing a charging station capacity strategy in the framework of the non‐cooperative game. Furthermore, the combination of an adaptive mutation particle swarm optimization algorithm (AMPSO) and approximate Nash equilibrium had a better effect in reducing the difficulty of solving the game model and obtaining an optimal planning strategy. Finally, the results of the case study demonstrated the effectiveness of the proposed methodology for improving the charging experience of the EV owners and achieving a balance of benefits among multiple charging station operators.

Abstract Objective This study aimed to explore the correlation between T1 and T2 relaxation times of synthetic MRI (SyMRI) and gestational age (GA) in each hemisphere of preterm and term newborns at the initial 28 days of birth. Methods Seventy preterm and full‐term infants were prospectively included in this study. All subjects completed 3.0 T routine MRI and SyMRI (MAGiC) one‐stop scanning within 28 days of birth (aged 34–42 W at examination). The SyMRI postprocessing software (v8.0.4) was used to measure the T1 and T2 relaxation values of each brain region. The linear regression equations of quantitative relaxation values with GA were established to compare the variation speed in each brain region. Abstract Aim Attitudes toward older persons could affect services that are provided to them and change the value of the care they receive. The attitude of nursing students toward older persons is very important as it will affect their care of older patients in future. However, in China, the attitude of higher vocational nursing students toward older persons has not been widely studied. The purpose of this study is to explore the attitudes and factors toward older people among nursing students in higher vocational colleges in Heilongjiang Province. Design A cross‐sectional study. Methods A cross‐sectional study was conducted using cluster sampling. A total of 1106 nursing students from two higher vocational colleges in Heilongjiang Province participated. Multiple linear regression analysis was conducted to identify the factors influencing nursing students’ attitudes toward older people. Conclusion This study showed that the vocational college nursing students have a positive attitude towardolder persons. However, the proportion of respondents taking care of older persons as their first choice in their future career needs to be increased. Advanced vocational colleges should pay attention to the attitudes of nursing students toward older persons, and strengthen the education of career development planning, which has practical significance in meeting the diversified health needs of older persons and ensuring healthy aging.

Abstract In vivo imaging in the deep near‐infrared (NIR) spectral region, that is, beyond 800 nm, has become popular due to its penetration depth. While imaging of the neutral medium/tissue has been repeatedly showcased, imaging of the high‐acidic medium remains challenging partly because of the high‐lying HOMO orbital and hence a high pKa of the electron‐donating group of the NIR fluorophores. We devised a novel electron‐donating group (D6) with which we further synthesized ECJ. ECJ exhibits an absorption wavelength beyond 900 nm and is fluorescent. Its pKa was found to be lower than zero, rendering it suitable for bioimaging of a highly‐acidic medium. Its potential for practical applications was showcased in proof‐of‐concept in vivo imaging with a mouse model.

Abstract Foodborne protein hydrolysates exhibit biological activity that may be therapeutic in a number of human disease settings. Hemp peptides (HP) generated by controlled hydrolysis of hemp proteins have a number of health benefits and are of pharmaceutical value. In the present study, we produce small molecular weight HP from hemp seed and investigate its anticancer properties in Hep3B human liver cancer cells. We demonstrate that HP treatment increased apoptosis, reduced cell viability, and reduced cell migration in Hep3B human liver cancer cells without affecting the normal liver cell line L02. We correlate these phenotypes with increased cellular ROS levels, upregulation of cleaved caspase 3 and Bad, and downregulation of antiapoptotic Bcl‐2. HP treatment led to increased Akt and GSK‐3β phosphorylation, with subsequent downregulation of β‐catenin, suggesting β‐catenin signaling modulation as a critical mechanism by which HP exhibits anticancer properties. Our findings suggest HP are of potential therapeutic interest for liver cancer treatment.

Logo design is related to the image of a company, unit, product, etc. A good logo can leave a deep impression on people and intuitively understand the content of the product. With the increasing emphasis on intellectual property protection, the design of logos should also pay more attention to the protection of property rights. It can be said that the logo of some big brands is enough to represent the enterprise itself; for example, seeing the red cross as a hospital. This paper aims to propose a logo design process and method under the background of intellectual property big data, so as to make the logo design more procedural and efficient, and to better focus on the protection of property rights. By introducing the relevant knowledge of logo design and trademark registration, it is indicated that trademark search can be used as an auxiliary tool for logo design. Combined with digital media and big data technology, a logo image recognition and classification method is designed so that the trademark registration and screening stage can avoid the infringement design to the greatest extent, so as to put forward the design process and method to improve the innovation ability of logo design. According to the research, the classification accuracy rate of the algorithm proposed in this paper is 91.5%, and the recognition rate of various defects is also above 95%. In addition, in design practice, the stability of design proposals should be monitored through multiple search methods using trademark search tools. If a high similarity is found in the search results, the design direction should be adjusted in time until the final design scheme is determined.

Rock brittleness is a critical factor affecting the stimulation of shale oil reservoirs. For efficient development of the shale oil in Lucaogou Formation, Jimusar Sag, the brittleness of the sweet spots needs to be evaluated. In this paper, the triaxial compression, acoustic wave measurements, and three-point bending tests were carried out on the reservoir cores. Based on the prepeak energy characteristics of the stress-strain curve, the brittleness of different horizons was calculated, with the largest difference of 24.89%. An improved model based on dynamic elastic properties was proposed to evaluate the brittleness along the vertical pay zones, by which the continuous brittleness in the upper sweet spot was found more changeful than that in the lower sweet spot. The linear correlation coefficient between the brittleness from the improved model and that from the laboratory tests is 0.85, improving the accuracy by 21% and 27% respectively, compared with the conventional elastic property methods. From the characteristics of compression fractures and the length of the fracture process zone, it was found that the compression fractures were more complex and the fracture process zone length was shorter in a more brittle rock, verifying the reliability of the improvement model. The improved method based on dynamic elastic properties proposed in this paper is expected to guide the brittleness evaluation in other regions.

Building energy consumption is increasingly becoming a matter of global concern. A key aspect of this is the nature of building cold source systems and their effectiveness. However, choosing the cold source for a building is a complex decision-making process. The traditional evaluation method is relatively simple, and it is difficult to comprehensively consider multiple factors and their mutual influences, and it is even more difficult to increase the consideration of the whole life-cycle cost on this basis. Especially in China, the cost of extra-large public projects invested and constructed by the government plays a significant role in the decision-making process, and it should not be affected by other factors to change the decision-making. Therefore, this paper proposes a new decision-making system, which is based on value engineering (VE) and combined with analytic network process (ANP) to conduct a comprehensive study of project decision-making. Based on existing approaches to construction project evaluation and the selection of cold source systems, we establish an evaluation index for building cold source system functional models and network structures. This approach can take into account the impact of cost on decisions. We then present the results of a simulation-based case study of potential central air-conditioning cold source system solutions for the new Xiang’an New Airport in Xiamen, China. The corresponding value coefficients for conventional electric refrigeration, ice storage, and water storage were found to be 0.8262, 1.0049, and 1.2442, respectively. This suggests that water storage-based cooling offers the best technical and economic benefits for this project. It also confirms that the proposed decision support system can identify the modes with the highest value coefficients, which is one of the most effective methods for selecting a building cold source system scheme. Therefore, the system has the potential to support the effective selection of other building cold source schemes.

Background Aspirin is a classic anti‐inflammatory drug and its anticancer effect has been previously explored in many types of cancer including colorectal cancer therapy. Programmed cell death‐ligand 1 (PD‐L1) is widely expressed in tumor cells and displays an inhibitory role in antitumor immunity. This study aimed to clarify the role of PD‐L1 in aspirin‐suppressed lung cancer. Methods The inhibitory effect of aspirin on lung cancer cell proliferation was assessed using an MTT cell viability assay. The role of aspirin in the modulation of PD‐L1 expression was analyzed by western blot or RT‐PCR assays. In lung cancer cells, the influence of aspirin on PD‐L1 promoter activity was detected using a luciferase reporter assay. The interaction of TAZ with PD‐L1 promoter in the cells, with or without aspirin administration, was tested via chromatin immunoprecipitation (ChIP) analysis. The function of PD‐L1 in aspirin‐mediated growth inhibition of lung cancer was examined using a cell viability assay. Results Following treatment with aspirin, lung cancer cell growth was markedly suppressed. Aspirin was able to markedly decrease the expression of PD‐L1 at the mRNA and protein levels in lung cancer cells. For the mechanism study, we found that the promoter of PD‐L1 was inactivated by aspirin via TAZ transcriptional coactivator in the cells. With regard to the functional investigation, aspirin was capable of resisting cell proliferation and PD‐L1 overexpression abolished aspirin‐depressed cell proliferation in lung cancer. Conclusions Aspirin suppressed the growth of lung cancer cells via targeting the TAZ/PD‐L1 axis.

The balancing holes in centrifugal pumps with seals mounted in both suction and discharge sides are one of the approaches used by pump manufacturers to reduce the axial thrust. The balance hole diameter directly affects the axial force of the centrifugal pump. The flow characteristics in the balance chamber are closely related to the balance hole diameter. However, research is not very clear on the internal flow of the balanced chamber, due to the small axial and radial sizes and the complicated flow conditions in the chamber. In this paper, we analyzed the influence of the balance hole diameter on the liquid leakage rate, flow velocity, and vortex motion in the balance chamber. The results indicated that when the balance hole diameter was lower than the design value, the volume flow rate of leakage flow was proportional to the diameter. The liquid flow rate and vortex distribution rules in the balance chamber were mainly associated with the coeffect of radial leakage flow in the rear sealing ring interval and the axial balance hole leakage flow. The research has revealed the mechanisms of leakage flow of the balance chamber in the centrifugal pump and that this is of great significance for accurate calculation and balancing of the axial force.

Peroxisome proliferator-activated receptor γ (PPARG) might play a protective role in the development of myocardial infarction (MI) with limited mechanisms identified. Genes associated with both PPARG and MI were extracted from Elsevier Pathway Studio to construct the initial network. The gene expression activity within the network was estimated through a mega-analysis with eight independent expression datasets derived from Gene Expression Omnibus (GEO) to build PPARG and MI connecting pathways. Sox9 is an intrinsic transcription factor related to the determination and maintenance of chondrogenic lineage of bone marrow mesenchymal stem cells (BMSCs). In recent research, we have proved that fragmented chondrocyte aggregates (cell bricks) could promote chondrogenesis of BMSCs in vivo. However, it is still unknown whether the ratio of BMSCs/chondrocyte bricks has a significant influence on 3-D cartilage regeneration and related molecular mechanism. To address this issue, the current study subcutaneously injected three groups of cell complex with different rabbit BMSCs/chondrocyte bricks’ ratios (1 : 2, 1 : 1, and 2 : 1) into nude mice. Gross morphology observation, histological and immunohistochemical assays, biochemical analysis, gene expression analysis, and western blot were used to compare the influence of different BMSCs/chondrocyte bricks’ ratios on the properties of tissue-engineered cartilage and explore the related molecular mechanism. The constructs of 1 : 1 BMSCs/chondrocyte bricks, (B1CB1) group resulted in persistent chondrogenesis with appropriate morphology and adequate central nutritional perfusion without ossification. The related mechanism is that increased expression of Sox9 in the B1C1 group promoted chondrogenesis and inhibited the osteogenesis of BMSCs through upregulating Col-II as well as downregulating RUNX2 and downstream of Col-X and Col-I by upregulating Nkx3.2. This study demonstrated that BMSCs/chondrocyte bricks 1:1 should be a suitable ratio and the Sox9-Nkx3.2-RUNX2 pathway was a related mechanism which played an important role in the niche for stable chondrogenesis of BMSCs constructed by chondrocyte bricks and PRP.

Natural fractures usually develop in shale reservoirs. Thereby, in the process of hydraulic fracturing, it is inevitable that hydraulic fractures will intersect with natural fractures. In order to reveal the interaction mechanism between hydraulic-induced fractures and natural fractures, a two-dimensional fracture intersection model based on the extended finite element method (XFEM) is proposed, and the different types of intersecting criteria reported in the literature are compared. Then, the effects of natural fracture azimuth, fluid pressure in hydraulic fracture, and in situ principal stress difference on hydraulic fracturing are studied in detail. The results show that the fracture morphology is different under different criteria and working conditions. And the stress concentration phenomenon mainly concentrates on the tip in the obtuse angle side of natural fracture. Meanwhile, different fluid pressures in hydraulic fracture can also induce different intersection patterns. The obtained results in this work are of great benefit to understand the intersection mechanism between hydraulic fractures and natural fractures.

Isoorientin has anti-inflammatory effects; however, the mechanism remains unclear. We previously found isoorientin is an inhibitor of glycogen synthase kinase 3β (GSK3β) in vitro. Overactivation of GSK3β is associated with inflammatory responses. GSK3β is inactivated by phosphorylation at Ser9 (i.e., p-GSK3β). Lithium chloride (LiCl) inhibits GSK3β and also increases p-GSK3β (Ser9). The present study investigated the anti-inflammatory effect and mechanism of isoorientin via GSK3β regulation in lipopolysaccharide- (LPS-) induced RAW264.7 murine macrophage-like cells and endotoxemia mice. LiCl was used as a control. While AKT phosphorylates GSK3β, MK-2206, a selective AKT inhibitor, was used to activate GSK3β via AKT inhibition (i.e., not phosphorylate GSK3β at Ser9). The proinflammatory cytokines TNF-α, IL-6, and IL-1β were detected by ELISA or quantitative real-time PCR, while COX-2 by Western blotting. The p-GSK3β and GSK3β downstream signal molecules, including NF-κB, ERK, Nrf2, and HO-1, as well as the tight junction proteins ZO-1 and occludin were measured by Western blotting. The results showed that isoorientin decreased the production of TNF-α, IL-6, and IL-1β and increased the expression of p-GSK3β in vitro and in vivo, similar to LiCl. Coadministration of isoorientin and LiCl showed antagonistic effects. Isoorientin decreased the expression of COX-2, inhibited the activation of ERK and NF-κB, and increased the activation of Nrf2/HO-1 in LPS-induced RAW264.7 cells. Isoorientin increased the expressions of occludin and ZO-1 in the brain of endotoxemia mice. In summary, isoorientin can inhibit GSK3β by increasing p-GSK3β and regulate the downstream signal molecules to inhibit inflammation and protect the integrity of the blood-brain barrier and the homeostasis in the brain.

Therapeutically intervening the function of RNA in vivo remains a big challenge. We here developed an exosome-based strategy to deliver engineered RNA-binding protein for the purpose of recruiting specific RNA to the lysosomes for degradation. As a proof-of-principle study, RNA-binding protein HuR was fused to the C-terminus of Lamp2b, a membrane protein localized in both exosome and lysosome. The fusion protein was able to be incorporated into the exosomes. Moreover, exosomes engineered with Lamp2b-HuR successfully decreased the abundance of RNA targets possibly via lysosome-mediated degradation, especially when the exosomes were acidified. The system was specifically effective in macrophages, which are lysosome enriched and resistant to routine transfection mediated RNAi strategy. In the CCl4-induced liver injury mouse model, we found that delivery of acidified exosomes engineered with Lamp2b-HuR significantly reduced liver fibrosis, together with decreased miR-155 and other inflammatory genes. In summary, the established exosome-based RNA-binding protein delivery strategy, namely “exosome-mediated lysosomal clearance”, takes the advantage of exosome in targeted delivery and holds great promise in regulating a set of genes in vivo.

Physalin B from Physalis angulata L. (Solanaceae) is a naturally occurring secosteroid with multiple biological activities. But its anti-inflammatory activity and mechanism remain unclear. Physalin B effects on RAW264.7 macrophages stimulated by lipopolysaccharide (LPS) were observed in this study. The expression and secretion of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) induced by LPS were significantly inhibited by physalin B. Meanwhile, the NF-κB nuclear translocation induced by LPS was inhibited by physalin B. The anti-inflammatory effects of physalin B could not be inhibited by mifepristone (RU486), the blocker of glucocorticoid receptor. In conclusion, physalin B can suppress inflammatory response to LPS in macrophages by inhibiting the production of inflammatory cytokines via NF-κB signaling.

Clustering and routing are two key techniques to improve the energy efficiency of wireless sensor networks. As clustering and routing for improving the energy efficiency of wireless sensor networks are NP-hard problems, increasing meta-heuristic algorithms are introduced for solving them. However, due to their discreteness and strong constraints, most meta-heuristics are unsuitable or inefficient to optimize them. Harmony search algorithm is one of the most suitable meta-heuristics for solving these problems. This article proposes a new energy-efficient clustering and routing algorithm based on harmony search algorithm to improve the energy efficiency of wireless sensor networks. The proposed approach contains two parts: clustering phase and routing phase. First, a new objective function model, which has considered balancing the energy consumption of both gateways and regular nodes as well as considered routing, is established for the clustering phase. Then, a new energy-efficient clustering algorithm is designed based on several improvements made to harmony search algorithm: (1) a discrete encoding scheme of a harmony for clustering is proposed; (2) a roulette wheel selection method is designed to choose a gateway for a regular sensor node to join, which is employed by two steps (i.e. initialization of harmony and improvisation of a new harmony); (3) the dynamically changed harmony memory considering rate is designed for improvisation of a new harmony; (4) a local search scheme is proposed to improve the best harmony within the harmony memory in iterations. In addition, the improved harmony search based energy-efficient routing algorithm that we proposed previously is employed to balance the energy consumption of gateways in the routing phase. The proposed approach is compared with several popular meta-heuristic-based clustering algorithms over extensive wireless sensor networks cases. The experimental results clearly demonstrate the superiority of the proposed approach.

A control strategy of permanent magnet synchronous motors (PMSMs), which is different from the traditional vector control (VC) and direct torque control (DTC), is proposed. Firstly, the circular rotating magnetic field is analyzed on the simplified model and discredited into stepping magnetic field. The stepping magnetomotive force will drive the rotor to run as the stepping motor. Secondly, the stator current orientation is used to build the control model instead of rotor flux orientation. Then, the discrete current control strategy is set and adopted in positioning control. Three methods of the strategy are simulated in computer and tested on the experiment platform of PMSM. The control precision is also verified through the experiment.
