Abstract The exceptional optoelectronic properties of lead halide perovskite nanocrystals (PeNCs) in the ultraviolet and visible spectral regions have positioned them as a promising class of semiconductor materials for diverse optoelectronic and photovoltaic applications. However, their limited response to near‐infrared (NIR) light due to the intrinsic bandgap (>1.5 eV) has hindered their applications in many advanced technologies. To circumvent this limitation, it is of fundamental significance to integrate PeNCs with lanthanide‐doped upconversion nanoparticles (UCNPs) that are capable of efficiently converting low‐energy NIR photons into high‐energy ultraviolet and visible photons. By leveraging the energy transfer from UCNPs to PeNCs, this synergistic combination can not only expand the NIR responsivity range of PeNCs but also introduce novel emission profiles to upconversion luminescence with multi‐dimensional tunability (e.g., wavelength, lifetime, and polarization) under low‐to‐medium power NIR irradiation, which breaks through the inherent restrictions of individual PeNCs and UCNPs and thereby opens up new opportunities for materials and device engineering. In this review, we focus on the latest advancements in the development of PeNCs‐UCNPs nanocomposites, with an emphasis on the controlled synthesis and optical properties design for advanced optoelectronic applications such as full‐spectrum solar cells, NIR photodetectors, and multilevel anticounterfeiting. Some future efforts and prospects toward this active research field are also envisioned.

Abstract Textures in molybdenum‐rhenium (Mo‐Re) alloys are inevitable during thermal fabrication. [110] and [100] are common orientations in Mo‐Re alloys and effect mechanical responses. However, orientation dependence of mechanical responses in Mo‐Re alloys is not quite clear yet. To clarity this problem, micro‐pillar compression tests are conducted in grains with orientation [100] and [110] separately. Orientation‐dependent compressive properties are found in Mo‐14Re and Mo‐42Re (wt.%), but are not found in Mo and Mo‐5Re, which may be attributed to activated multi‐slip planes as increased Re. Solid solution effect of Re not only relies on orientations, but also on Re contents. Softening effect occurs in both [100] and [110] Mo‐5Re. while, strong strengthening effect happens in [110] Mo‐14Re and Mo‐42Re. Our research clarifies that Mo‐Re alloys with [110] orientation/texture could be preferred to obtain good strengthening effect.

Abstract Dryland ecosystems cover 40% of our planet's land surface, support billions of people, and are responding rapidly to climate and land use change. These expansive systems also dominate core aspects of Earth's climate, storing and exchanging vast amounts of water, carbon, and energy with the atmosphere. Despite their indispensable ecosystem services and high vulnerability to change, drylands are one of the least understood ecosystem types, partly due to challenges studying their heterogeneous landscapes and misconceptions that drylands are unproductive “wastelands.” Consequently, inadequate understanding of dryland processes has resulted in poor model representation and forecasting capacity, hindering decision making for these at‐risk ecosystems. NASA satellite resources are increasingly available at the higher resolutions needed to enhance understanding of drylands' heterogeneous spatiotemporal dynamics. NASA's Terrestrial Ecology Program solicited proposals for scoping a multi‐year field campaign, of which Adaptation and Response in Drylands (ARID) was one of two scoping studies selected. A primary goal of the scoping study is to gather input from the scientific and data end‐user communities on dryland research gaps and data user needs. Here, we provide an overview of the ARID team's community engagement and how it has guided development of our framework. This includes an ARID kickoff meeting with over 300 participants held in October 2023 at the University of Arizona to gather input from data end‐users and scientists. We also summarize insights gained from hundreds of follow‐up activities, including from a tribal‐engagement focused workshop in New Mexico, conference town halls, intensive roundtables, and international engagements.

Abstract l‐2‐Hydroxyglutarate (l‐2‐HG) is a functionally compartmentalized metabolite involved in various physiological processes. However, its subcellular distribution and mitochondrial transport remain unclear owing to technical limitations. In the present study, an ultrasensitive l‐2‐HG biosensor, sfLHGFRH, composed of circularly permuted yellow fluorescent protein and l‐2‐HG‐specific transcriptional regulator, is developed. The ability of sfLHGFRH to be used for analyzing l‐2‐HG metabolism is first determined in human embryonic kidney cells (HEK293FT) and macrophages. Then, the subcellular distribution of l‐2‐HG in HEK293FT cells and the lower abundance of mitochondrial l‐2‐HG are identified by the sfLHGFRH‐supported spatiotemporal l‐2‐HG monitoring. Finally, the role of the l‐glutamate transporter SLC1A1 in mitochondrial l‐2‐HG uptake is elucidated using sfLHGFRH. Based on the design of sfLHGFRH, another highly sensitive biosensor with a low limit of detection, sfLHGFRL, is developed for the point‐of‐care diagnosis of l‐2‐HG‐related diseases. The accumulation of l‐2‐HG in the urine of patients with kidney cancer is determined using the sfLHGFRL biosensor.

Abstract Simultaneously achieving high‐energy‐density and high‐power‐density is a crucial yet challenging objective in the pursuit of commercialized power batteries. In this study, atomic layer deposition (ALD) is employed combined with a coordinated thermal treatment strategy to construct a densely packed, electron‐ion dual conductor (EIC) protective coating on the surface of commercial LiNi0.5Co0.2Mn0.3O2 (NCM523) cathode material, further enhanced by gradient Al doping (Al@EIC‐NCM523). The ultra‐thin EIC effectively suppresses side reactions, thereby enhancing the stability of the cathode‐electrolyte interphase (CEI) at high‐voltages. The EIC's dual conduction capability provides a potent driving force for Li+ transport at the interface, promoting the formation of rapid ion deintercalation pathways within the Al@EIC‐NCM523 bulk phase. Moreover, the strategic gradient doping of Al serves to anchor the atomic spacing of Ni and O within the structure of Al@EIC‐NCM523, curbing irreversible phase transitions at high‐voltages and preserving the integrity of its layered structure. Remarkably, Al@EIC‐NCM523 displays an unprecedented rate capability (114.7 mAh g−1 at 20 C), and a sustained cycling performance (capacity retention of 74.72% after 800 cycles at 10 C) at 4.6 V. These findings demonstrate that the proposed EIC and doping strategy holds a significant promise for developing high‐energy‐density and high‐power‐density lithium‐ion batteries (LIBs).

Abstract The rodent running‐wheel recording apparatus is a reliable approach for studying circadian rhythm. This study demonstrated how to construct a simple and intelligent running‐wheel recording system. The running wheel was attached to the cage's base, whereas the Hall sensor was attached to the cage's cover. Then, the RJ25 adaptor relayed the running signal to the main control board. Finally, the main control board was connected to the USB port of the computer with the USB connection. Data were collected using the online‐accessible, self‐created software Magturning. Through Magturning, generated data were saved and exported in real time. Afterward, the device was validated by collecting data on the locomotor activities of mice under different light conditions. In conclusion, this new device can record circadian activity of rodents. Our device is appropriate for interdisciplinary investigations related to biological clock research.

Abstract With 75 known species, the freshwater fish genus Sinocyclocheilus is the largest cavefish radiation in the world and shows multiple adaptations for cave‐dwelling (stygomorphic adaptations), which include a range of traits such as eye degeneration (normal‐eyed, micro‐eyed and eyeless), depigmentation of skin, and in some species, the presence of “horns”. Their behavioural adaptations to subterranean environments, however, are poorly understood. Wall‐following (WF) behaviour, where an organism remains in close contact with the boundary demarcating its habitat when in the dark, is a peculiar behaviour observed in a wide range of animals and is enhanced in cave dwellers. Hence, we hypothesise that wall‐following is also present in Sinocyclocheilus, possibly enhanced in eyeless species compared to eye bearing (normal‐/micro‐eyed species). Using 13 species representative of Sinocyclocheilus radiation and eye morphs, we designed a series of assays, based on pre‐existing methods for Astyanax mexicanus behavioural experiments, to examine wall‐following behaviour under three conditions. Our results indicate that eyeless species exhibit significantly enhanced intensities of WF compared to normal‐eyed species, with micro‐eyed forms demonstrating intermediate intensities in the WF distance. Using a mtDNA based dated phylogeny (chronogram with four clades A–D), we traced the degree of WF of these forms to outline common patterns. We show that the intensity of WF behaviour is higher in the subterranean clades compared to clades dominated by normal‐eyed free‐living species. We also found that eyeless species are highly sensitive to vibrations, whereas normal‐eyed species are the least sensitive. Since WF behaviour is presented to some degree in all Sinocyclocheilus species, and given that these fishes evolved in the late Miocene, we identify this behaviour as being ancestral with WF enhancement related to cave occupation. Results from this diversification‐scale study of cavefish behaviour suggest that enhanced wall‐following behaviour may be a convergent trait across all stygomorphic lineages.

Abstract Synergistic changes between tumor‐associated macrophages (TAMs) and cancer‐associated fibroblasts (CAFs) aggravated immune evasion of hepatocellular carcinoma (HCC), however, the underlying molecular mechanisms remain elusive. Their continuous and dynamic interactions are subject to bioactive molecule changes. A real‐time and in situ monitoring method suitable for in vivo research of these processes would be indispensable but is scarce. In this study, a dual imaging strategy that tracing the TAMs and CAFs simultaneously was developed using a new arginase‐specific probe and established CAFs‐specific probe. The emerging roles of arginase in mediating CAFs activation in mice were explored. Results showed arginase up‐regulation in TAMs, followed by proline increase. Subsequently, proline produced by TAMs initiated the activation of CAFs. Through the JAK‐STAT signaling, CAFs up‐regulated the PD‐L1 and CTLA‐4, ultimately promoting immune evasion of HCC. This study revealed a new mechanism by which TAMs and CAFs collaborate in immune evasion, providing new targets for HCC immunotherapy.

Abstract Background Electroacupuncture (EA) has been shown to facilitate brain plasticity‐related functional recovery following ischemic stroke. The functional magnetic resonance imaging technique can be used to determine the range and mode of brain activation. After stroke, EA has been shown to alter brain connectivity, whereas EA's effect on brain network topology properties remains unclear. An evaluation of EA's effects on global and nodal topological properties in rats with ischemia reperfusion was conducted in this study. Methods and results There were three groups of adult male Sprague‐Dawley rats: sham‐operated group (sham group), middle cerebral artery occlusion/reperfusion (MCAO/R) group, and MCAO/R plus EA (MCAO/R + EA) group. The differences in global and nodal topological properties, including shortest path length, global efficiency, local efficiency, small‐worldness index, betweenness centrality (BC), and degree centrality (DC) were estimated. Graphical network analyses revealed that, as compared with the sham group, the MCAO/R group demonstrated a decrease in BC value in the right ventral hippocampus and increased BC in the right substantia nigra, accompanied by increased DC in the left nucleus accumbens shell (AcbSh). The BC was increased in the right hippocampus ventral and decreased in the right substantia nigra after EA intervention, and MCAO/R + EA resulted in a decreased DC in left AcbSh compared to MCAO/R. Conclusion The results of this study provide a potential basis for EA to promote cognitive and motor function recovery after ischemic stroke.

Abstract Background Compliance with colonoscopy among elderly individuals participating in colorectal cancer (CRC) screening programs is unsatisfactory, despite a high detection rate of bowel‐related diseases. In this study, our aim was to analyze the impact of risk factors on the trends of compliance and detection rates in colonoscopy among high‐risk individuals aged 60–74. Methods A retrospective study was conducted on the high‐risk individuals aged 60–74 participating in the 2021 CRC screening program in Tianjin, China. Logistic regression analyses, including both univariate and multivariate analyses, were performed to explore the impact of different risk factors on colonoscopy compliance among the high‐risk individuals. Besides, the study investigated the influence of various risk factors on the detection rates of bowel‐related diseases among the high‐risk individuals who underwent colonoscopy. Results A total of 24,064 high‐risk individuals were included, and 5478 individuals received a free colonoscopy, with an overall compliance of 22.76%. Among them, the adenoma detection rate was 55.46%. Males and individuals with a positive FIT had high compliance and detection rates for CRC, advanced adenomas (AA), advanced colorectal neoplasia (ACN), and colorectal neoplasm (CN). Individuals aged 70–74 were associated with low compliance but high CRC, ACN, and CN detection rates. Individuals who reported a history of chronic constipation, bloody mucous, and CRC in first‐degree relative showed high compliance but no significantwere associated with the detection rates of CRC, AA, and CN. Conclusion This study reported several risk factors associated with the screening behaviors for CRC. Patterns and trends in CRC, AA, ACN, and CN compliance and detection rates correlate with risk factors.

Abstract The availability of an ever‐expanding portfolio of 2D materials with rich internal degrees of freedom (spin, excitonic, valley, sublattice, and layer pseudospin) together with the unique ability to tailor heterostructures made layer by layer in a precisely chosen stacking sequence and relative crystallographic alignments, offers an unprecedented platform for realizing materials by design. However, the breadth of multi‐dimensional parameter space and massive data sets involved is emblematic of complex, resource‐intensive experimentation, which not only challenges the current state of the art but also renders exhaustive sampling untenable. To this end, machine learning, a very powerful data‐driven approach and subset of artificial intelligence, is a potential game‐changer, enabling a cheaper – yet more efficient – alternative to traditional computational strategies. It is also a new paradigm for autonomous experimentation for accelerated discovery and machine‐assisted design of functional 2D materials and heterostructures. Here, the study reviews the recent progress and challenges of such endeavors, and highlight various emerging opportunities in this frontier research area.

Abstract Background Corylifol A (CYA) is one of the main active components of Psoralea corylifolia L. CYA had been reported to have ameliorating effects on dexamethasone‐induced atrophy of C2C12 mouse skeletal myotubes, but its effects on cancer cachexia were unclear. Here, we checked the influence of CYA on muscle atrophy in cancer cachexia mice and tried to clarify its mechanisms. Methods C26 tumour‐bearing mice were applied as the animal model to examine the effects of CYA in attenuating cachexia symptoms. The in vitro cell models of TNF‐α‐induced C2C12 myotubes or ad‐mRFP‐GFP‐LC3B‐transfected C2C12 myotubes were used to check the influence of CYA on myotube atrophy based on both ubiquitin proteasome system (UPS) and autophagy‐lysosome system. The possible direct targets of CYA were searched using the biotin‐streptavidin pull‐down assay and then confirmed using the Microscale thermophoresis binding assay. The levels of related signal proteins in both in vitro and in vivo experiments were examined using western blotting and immunocytochemical assay. Results The administration of CYA prevented body weight loss and muscle wasting in C26 tumour‐bearing mice without affecting tumour growth. At the end of the experiment, the body weight of mice treated with 30 mg/kg of CYA (23.59 ± 0.94 g) was significantly higher than that of the C26 model group (21.66 ± 0.56 g) with P

Abstract CO2 adducts of hydrophobically modified polyethylenimines (PEIs) are promising alternatives to global warming halogen‐containing blowing agents of polyurethanes (PUs), despite the high cost of the raw material PEIs. Herein, an economical synthesis of PEI‐like polymers was explored via condensation between pentaethylenehexamine and terephthalaldehyde, followed by chemical reduction of the as‐formed Schiff base linkages. The resultant p‐phenylene‐containing polyamine polymers (PEIPs) could be grafted with alkyl (Cn) side chains and adducted with CO2 to form a new type of CO2‐releasing blowing agents for PUs designated as yCn‐xPEIP‐CO2s, where x and y represent the backbone molecular weight and the side chain grafting rate, respectively. Among them, the specimen 10%C8−3.6 kPEIP‐CO2 was the most effective in terms of good dispersibility in PU raw materials, low foam density (about 51 kg/m3), and uniform pore morphology. Moreover, the phenylene linkages enhanced the hydrophobicity of the consequent CO2 adducts and weakened the intermolecular hydrogen bonding and ionic attraction, both facilitating the dispersion of the corresponding blowing agents into a castor oil‐derived polyol, Polycin M‐365. The specimen 10%C8−3.6 kPEIP‐CO2 could disperse as suspended fine floccules that finally aggregated into a flocculent, liquid‐like bottom layer, being easily redispersed into the bulk. The unique compatibility with plant oil‐derived polyols and the economic availability would make the PEIP‐based blowing agents suitable for the next generation of sustainable and biomass‐based PU foams.

Abstract The rapid increase of photovoltaic (PV) penetration in active distribution networks (ADN) is posing great challenges to traditional voltage control schemes. A two‐stage voltage control strategy of ADN is proposed by the authors based on the alternating direction method of multipliers (ADMM) with refined power to hydrogen (P2H) model, considering multiple types of PV forms such as residential photovoltaic cluster (RPVC) and small‐scale PV power stations. In the day‐ahead stage, the optimal power flow is performed to determine the optimal scheduling results of on‐load tap changers and capacitor banks etc. In the intra‐day stage, the real‐time voltage control strategy is implemented at the distribution network layer to regulate the power of each type of PV, energy storage systems and P2H to further reduce the voltage deviation. At the customer layer, the residential photovoltaic (RPV) within the RPVC is precisely controlled based on the ADMM algorithm to achieve the minimum voltage deviation at each RPV access point. The proposed strategy is tested on the modified IEEE 33‐bus and IEEE 69‐bus distribution systems, and the simulation results verify its effectiveness in mitigating voltage violations.

Abstract Introduction This study aimed to investigate the presentations and survival outcomes of the distant metastatic non‐small cell lung cancer (NSCLC) without lymph node involvement to obtain a clearer picture of this special subgroup of metastatic NSCLC. Method A least absolute shrinkage and selection operator (LASSO) penalized Cox regression analysis was used to select the prognostic variables. A nomogram and corresponding risk‐classifying systems were constructed. The C‐index and calibration curves were used to evaluate the performance of the model. Overall survival (OS) curves were plotted using the Kaplan–Meier method, and the log‐rank test was used to compare OS differences between groups. Propensity score matching (PSM) was performed to reduce bias. Result A total of 12 610 NSCLC patients with M1 category (N0 group: 3045 cases; N1‐3 group: 9565 cases) were included. Regarding the N0 group, multivariate analysis demonstrated that age, sex, race, surgery, grade, tumor size, and M category were independent prognostic factors. A nomogram and corresponding risk‐classifying systems were formulated. Favorable validation results were obtained from the C‐index, calibration curves, and survival comparisons. Survival curves demonstrated that N0 NSCLC patients had better survival than N1‐3 NSCLC patients both before and after PSM. Furthermore, the survival of resected N0M1 patients was superior to that of those without surgery. Conclusion In this study, a prognostic nomogram and risk‐classifying systems designed for the T1‐4N0M1 NSCLC patients showed acceptable performance. Primary lung tumor resection might be a feasible treatment for this population subset. Additionally, we proposed that lymph node stage might have a place in the forthcoming tumor‐node‐metastasis (TNM) staging proposal for NSCLC patients with M1 category.

Abstract Background Increased preoperative red cell distribution width (RDW) is associated with poor prognosis in several cancers, but the relationships between preoperative RDW and changes in RDW (ΔRDW) and colorectal cancer (CRC) prognosis remain unclear. Our study aimed to demonstrate the prognostic significance of increased preoperative RDW and ΔRDW for CRC. Methods In this retrospective analysis, we enrolled 833 patients who underwent CRC surgery between 2015 and 2019 at the Affiliated Hospital of Xuzhou Medical University, China. ΔRDW in our study was defined as RDW at 1 month after discharge minus preoperative RDW. According to receiver operating characteristic (ROC) curve analysis, we used cut‐off values of 13.5% for RDW, 0.9% for ΔRDW. The cumulative survival rate was determined using the Kaplan–Meier method, and significant differences were evaluated by the log‐rank test. Multivariable Cox regression model was applied to clarify the independent risk factors for overall survival (OS), which were used to construct a nomogram prediction model. The competing risk method was also applied, and we analyzed only patients with early‐stage disease (stage 0‐II) for sensitivity analysis. Results Multivariable Cox regression analysis demonstrated that age, RDW, ΔRDW, postoperative adjuvant chemotherapy, CEA, CA19‐9, ASA, TNM stage, and pathological type were independent factors for OS in CRC patients (all p

Abstract Background Neoadjuvant chemotherapy followed by radical cystectomy (RC) is the standard of care for patients with muscle‐invasive bladder cancer (MIBC). However, treatment outcomes are suboptimal. Camrelizumab, a PD‐1 blockade, has shown benefits in several tumors. This study aimed to investigate the efficacy and safety of neoadjuvant camrelizumab in combination with gemcitabine plus cisplatin (GC) followed by RC for MIBC patients. Methods This was a multi‐center, single‐arm study that enrolled MIBC patients with a clinical stage of T2‐4aN0‐1M0, and scheduled for RC. Patients received three 21‐day cycles of camrelizumab 200 mg on day 1, gemcitabine 1000 mg/m2 on day 1 and 8, and cisplatin 70 mg/m2 on day 2, followed by RC. The primary endpoint was pathologic complete response (pCR, pT0N0). Results From May 2020 to July 2021, 43 patients were enrolled and received study medications at nine centers in China. Three of them were deemed ineligible and excluded from efficacy analysis but included in safety analysis. In total 10 patients were unevaluable as they declined RC (two due to adverse events [AEs] and eight due to patient's willingness). Among 30 evaluable patients, 13 patients (43.3%) achieved pCR, and 16 patients (53.3%) achieved pathologic downstaging. No AEs leading to death were observed. The most common AEs were anemia (69.8%), decreased white blood cell count (65.1%), and nausea (65.1%). Immune‐related AEs were all grade 1 or 2. Pathologic response was not correlated with PD‐L1 expression status or tumor mutation burden. Individual genes as a biomarker for pathologic response were not identified. Conclusions Neoadjuvant treatment with camrelizumab and GC regimen demonstrated preliminary anti‐tumor activity for MIBC patients with manageable safety profiles. The study met its primary endpoint, and the following randomized trial is ongoing.

Abstract Introduction Farmlands are key sources of greenhouse gas (GHG) emissions, which are susceptible to changes in precipitation regimes. The soils of seasonal fallow contribute approximately half of annual GHG emissions from farmlands, but the effect of precipitation frequency on soil GHG emissions from seasonal fallow croplands remains virtually unknown. Materials and Methods We conducted a microcosm study to evaluate the response of nitrous oxide (N2O), methane (CH4) and carbon dioxide (CO2) fluxes from typical paddy and upland soils to the changes in watering frequency simulating precipitation scenarios of subtropical regions during seasonal fallow. We also analyzed changes of soil properties and biotic characteristics associated with GHG emissions, including abundances of soil denitrifiers (nirK, nirS, nosZI and nosZII genes), methanotrophs (pmoA gene) and methanogens (mcrA gene) to altered watering frequency. Results Increased watering frequency led to overall increases in soil N2O and CO2 fluxes compared with low frequency. Compared with low frequency, high watering frequency decreased CH4 flux from the paddy soil by 3.5 times, while enhanced CH4 flux from the upland soil by 60%. Furthermore, the increased watering frequency had positive effects on cumulative N2O and CO2 fluxes from the upland soil, whereas no similar trend was observed for the paddy soil. Hierarchical partitioning analyses showed that N2O fluxes from the paddy soil were mostly related to nitrogen availability, and mcrA gene abundance had more than 90% of relative independent effects on CH4 and CO2 fluxes from the paddy soil. For the upland soil, nosZ (60.34%), pmoA (53.18%) and nir (47.07%) gene abundances were important predictors of N2O, CH4 and CO2 fluxes, respectively. Conclusion Our results demonstrate that increased watering frequency facilitates GHG emissions by changing soil properties and functional gene abundances. These findings provide new insights into GHG fluxes from seasonal fallow croplands in response to altered precipitation patterns.

Abstract Purpose Distant metastasis (DM) and neoadjuvant treatment response prediction remain critical challenges in the management of locally advanced rectal cancer (LARC). The aim of this study was to investigate the clinical relevance of viable circulating tumor cells (CTCs) for DM or response in patients with LARC in a neoadjuvant setting. Methods The detection of viable CTCs at different stages of treatment was planned for consecutive patients from a prospective trial. The Kaplan–Meier method, Cox proportional hazards model, and logistic regression model were utilized to analyze factors associated with DM or pathological complete response (pCR) and clinical complete response (cCR). Results Between December 2016 and July 2018, peripheral blood samples from 83 patients were collected before any treatment (median follow‐up time, 49.3 months). CTCs were present in 76 of 83 patients (91.6%) at baseline, and more than three CTCs detected in the blood sample was considered high risk. Only the CTC risk group was significantly associated with 3‐year metastasis‐free survival (MFS) (high risk vs. low risk, 57.1% (95% CI, 41.6–72.6) vs. 78.3% (95% CI, 65.8–90.8), p = 0.018, log‐rank test). When all the important variables were entered into the Cox model, the CTC risk group remained the only significant independent factor for DM (hazard ratio (HR), 2.74; 95% CI, 1.17–6.45, p = 0.021). The pCR and continuous cCR rates were higher in patients with a decreased number of CTCs of more than one after radiotherapy (HR, 4.00; 95% CI, 1.09–14.71, P = 0.037). Conclusions The dynamic detection of viable CTCs may strengthen pretreatment risk assessment and postradiotherapy decision making for LARC. This observation requires further validation in a prospective study.

Abstract Background and Aims Colonoscopy is an important colorectal cancer (CRC) screening modality; however, not all high‐risk groups identified by fecal immunochemical test (FIT) and/or high‐risk factor questionnaire (HRFQ) undergo colonoscopy in time. The impact of delays in colonoscopy on CRC detection among high‐risk populations remains poorly understood, warranting further clarification. Methods A retrospective study was conducted among CRC high‐risk population identified by Tianjin CRC screening program. According to the colonoscopy results after HRFQ and FIT, patients were classified into CRC, advanced adenoma, non‐advanced adenoma, and normal groups. The time interval between CRC screening and colonoscopy was investigated and its relationship with colonoscopy results. Logistic regression was performed to explore the risk factors of CRC detection. Results Among the high‐risk population without a history of CRC or polyps, 49,810 underwent HRFQ, FIT, and colonoscopy, and a time interval of fewer than 6 months was found for 79.56% of patients (n = 39,630). People with positive FIT were more likely to undergo colonoscopy within 6 months, and detection rates of CRC and/or advanced adenoma were positively related to time intervals. Similar results were found in people with a negative FIT but positive HRFQ. A time interval longer than 6 months was a significant predictor of CRC detection in high‐risk populations. Conclusion For high‐risk people identified by CRC screening, especially those with a positive FIT, a time interval of 6 months was associated with an increased probability of CRC detection. Our findings emphasize that populations at high risk should undergo colonoscopy at least within 6 months.

Abstract Genomic signatures of local adaptation have been identified in many species but remain sparsely studied in amphibians. Here, we explored genome‐wide divergence within the Asiatic toad, Bufo gargarizans, to study local adaptation and genomic offset (i.e., the mismatch between current and future genotype‐environment relationships) under climate warming scenarios. We obtained high‐quality SNP data for 94 Asiatic toads from 21 populations in China to study spatial patterns of genomic variation, local adaptation, and genomic offset to warming in this wide‐ranging species. Population structure and genetic diversity analysis based on high‐quality SNPs revealed three clusters of B. gargarizans in the western, central‐eastern, and northeastern portions of the species' range in China. Populations generally dispersed along two migration routes, one from the west to the central‐east and one from the central‐east to the northeast. Both genetic diversity and pairwise FST were climatically correlated, and pairwise FST was also correlated with geographic distance. Spatial genomic patterns in B. gargarizans were determined by the local environment and geographic distance. Global warming will increase the extirpation risk of B. gargarizans.

Objective Gastrointestinal dysfunction seriously affects the prognosis and quality of life of patients with multiple fractures. However, experimental evidence of this relationship is lacking. Here we describe a newly developed mouse model of postoperative gastrointestinal dysfunction after multiple fractures. Methods Trauma severity was assessed using the injury severity score (ISS). Based on the ISS, a multiple fracture model was established in mice as follows: limb fractures with pelvic fractures and multiple rib fractures; limb fractures with multiple rib fractures; closed fracture of both forelegs with pelvic fracture and rib fractures; closed limb fractures; limb fracture with pelvic fracture; spinal fractures; hind leg fractures with pelvic fractures; pelvic fracture with multiple rib fractures; closed fracture of both fore legs with pelvic fracture; and closed fracture of both fore legs with multiple rib fractures. In each model group, gastrointestinal motility was assayed and the histopathology of the small intestine was examined. Western blot and immunohistochemical analyses of jejunal tissue were performed to detect c‐kit protein expression, the level of which was compared with that of a control group. The results of ANOVA are expressed as mean ± standard deviation. Results In mice with multiple fractures, food intake was greatly reduced, consistent with histopathological evidence of an injured intestinal epithelium. The jejunal tissue of mice in groups a, c, f, and h was characterized by extensively necrotic and exfoliated intestinal mucosal epithelium and inflammatory cell infiltration in the lamina propria. In traffic engineering design in saline soil environments, a salt barrier is usually installed in the embankment to prevent secondary salinization damage on the upper embankment and road surface. However, using this salt barrier method in saline soil subgrade projects in cold and arid highland areas has led to the problem of salt barrier failure, and the failure mechanism remains unclear. The study was performed in the saline soil distribution area in the Qaidam Basin on the northeastern part of the Qinghai–Tibet Plateau. We collected samples from the natural saline soil foundation and the structural layer of the subgrade soil using the method of excavating a road section that had undergone salt barrier failure. Laboratory tests and theoretical calculations were carried out on the particle gradation of the natural saline soils and the structural subgrade soil layers. In addition, their water and salt transport heights and the failure mechanism of the subgrade salt barrier were studied. The results showed that salt chemistry increases the capillary force between the soil grains in highly saline soil environments. This increases the water–salt transport of more than 850.00 mm in saline soils, which is much greater than the capillary water rise in ordinary soils (463.0–650.0 mm). When no water barrier geotextile layer is set in the subgrade structure, there is frequently a high headwater and salt transport in the soil layer of the subgrade structure for a long time. There is continuous vapor and salt transport in the large pores of the coarse-grained salt barrier. The vibration load of the road surface causes the fine soil particles to migrate downward into the coarse-grained salt barrier, reducing the thickness of the salt barrier. Under the combined effect of these three effects, the salt barrier of the saline soil subgrade in cold and arid areas fails to resist salt migration, and the subgrade is prone to secondary salinization.

Abstract This study aimed to predict the 5‐year overall survival (OS) benefit of pola‐R‐CHP versus R‐CHOP in the POLARIX trial based on the 2‐year event‐free survival (EFS) and progression‐free survival (PFS) rates in diffuse large B‐cell lymphoma (DLBCL). We identified randomized controlled trials (RCT) published before 31 May 2023. The correlation between the logarithmic (log) hazard ratio (HR) for EFS (HREFS) or PFS (HRPFS) and the HR for OS (HROS) was estimated at the trial‐level. Correlation analysis was performed between 2‐year PFS or EFS and 5‐year OS rates at the treatment arm‐level. Linear regression models were used to calculate the 5‐year OS of pola‐R‐CHP and R‐CHOP. In the included 20 RCTs, a linear correlation between HREFS (r = 0.765) or HRPFS (r = 0.534) and HROS was observed at the trial‐ level. Two‐year EFS (r = 0.918) or 2‐year PFS (r = 0.865) correlated linearly with 5‐year OS. Linear regression analysis between 2‐year EFS/PFS and 5‐year OS gave estimated 5‐year OS rates between pola‐R‐CHP and R‐CHOP of 6.4% and 6.3%, respectively. Two‐year EFS and PFS are feasible early endpoints in patients with DLBCL treated primarily with immunochemotherapy. The pola‐R‐CHP regimen is expected to improve 5‐year OS.

Abstract Lanthanide (Ln3+) oxysulfide nanocrystals (NCs) have great prospect in many advanced technologies; however, they suffer from a low photoluminescence efficiency due to the volatility of sulfur and deleterious surface quenching effect. Herein, we report a novel sandwiched luminescent heterostructure based on Ln3+‐doped Gd2O2S@NaYF4 core/shell NCs with tunable sulfur content in the sandwich layer. By means of Eu3+ as the sensitive structural probes, we unravel the ligand‐mediated structure control of the NCs from Gd2O3: Ln3+@NaYF4 to Gd2O2S: Ln3+@NaYF4 with tailored S2– deficiency. Such a sandwich‐type core/shell heterostructure enables us to achieve efficient and multicolor downshifting and upconversion luminescence (UCL), with up to 208.8 folds of enhancement in UCL intensity as compared to that of their core‐only counterparts. These findings provide a general approach for the controlled synthesis of lanthanide oxysulfide@fluoride heterostructure, which offers a new way for the materials design towards diverse emerging applications.

Abstract Microneedles (MNs) have been broadly used for transdermal delivery of a variety of drugs, ranging from small chemicals to biological macromolecules, due to the properties of increased drug permeability, minimal invasiveness and improved patient compliance. Despite these MNs can be made of different materials, such as metal, silicon, and glass, polymers have attracted the most attention as a microneedle (MN) matrix because of their excellent biocompatibility and biodegradability, which eliminates the requirement of MN removal after drug release. To satisfy different needs of transdermal drug delivery, polymeric MNs have been fabricated with several special designs. In this review, we summarize the advancement of the fabrication designs of polymeric MNs, including integrated MNs, two‐segment MNs, core‐shell or multi‐layered MNs, and arrowhead MNs. The related biomedical applications of MNs with these different specific designs are also discussed. Finally, we provide our perspectives on the future development of polymeric MNs.

Abstract A novel stripline diplexer design using frequency‐dependent couplings (FDC) to achieve multiple transmission zeros is developed in this paper. The transmission zeros generated by FDCs are flexible and controllable; on the basis of the existing cross‐coupled, more transmission zeros are introduced to improve the frequency selection characteristics. Based on this characteristic, the authors synthesized and designed a 2.6‐GHz diplexer, its transmitting channel filter is five orders with three transmission zeros, and the receiving channel filter is four orders with three transmission zeros. The authors fabricated and measured it, in narrowband, the tested results, synthesis responses and simulation results are well matched with each other, which will provide more flexibility in the design of diplexers for wireless communication system.

Abstract Background Magnetic resonance imaging (MRI) is the gold standard for evaluating body composition. However, the reference ranges have not been established. Methods Three lean tissue and seven adipose tissue parameters based on MRI data from the UK Biobank were used in this study. Participants with European ancestry and data on at least one parameter were screened. Age‐ and sex‐specific percentile curves were generated using the lambda–mu–sigma method. Three levels of reference ranges were provided, which were equivalent to the mean ± 1 standard deviation (SD), 2 SDs and 2.5 SDs. Results The final analysis set for each parameter ranged from 4842 to 14 148 participants (53.4%–56.6% women) with a median age of 61. For lean tissue parameters, compared with those at age 45, the median total lean tissue volume and total thigh fat‐free muscle volume at age 70 were 2.83 and 1.73 L, and 3.02 and 1.51 L lower in men and women, respectively. The median weight‐to‐muscle ratios at age 45 were 0.51 and 0.83 kg/L lower compared with those at age 70 in men and women, respectively. Adipose tissue parameters showed inconsistent differences. In men, the median muscle fat infiltration, visceral adipose tissue (VAT) volume, total abdominal adipose tissue index and abdominal fat ratio were 1.48%, 0.32 L, 0.08 L/m2 and 0.4 higher, and the median abdominal subcutaneous adipose tissue (ASAT) volume and total adipose tissue volume were 0.47 and 0.41 L lower, respectively, at age 70 than at age 45. The median total trunk fat volume was approximately 9.53 L at all ages. In women, the median muscle fat infiltration and VAT volume were 1.68% and 0.76 L higher, respectively, at age 70 than at age 45. The median ASAT volume, total adipose tissue volume, total trunk fat volume, total abdominal adipose tissue index and abdominal fat ratio were 0.35 L, 0.78 L, 1.12 L, 0.49 L/m2 and 0.06 higher, respectively, at age 60 than at age 45. The medians of the former three parameters were 0.33 L, 0.14 L and 0.20 L lower, at age 70 than at age 60. The medians of the latter two parameters were approximately 3.64 L/m2 and 0.55 at ages between 60 and 70. Conclusions We have established reference ranges for MRI‐measured body composition parameters in a large community‐dwelling population. These findings provide a more accurate assessment of abnormal adipose and muscle conditions.

Abstract Background Liver transplantation (LT), resection (LR), and ablation (LA) are three curative‐intent treatment options for patients with early hepatocellular carcinoma (HCC). We aimed to develop a prognostic calculator to compare the long‐term outcomes following each of these therapies. Methods A total of 976 patients with HCC within the Milan criteria who underwent LT, LR, and LA between 2009 and 2019 from four institutions were evaluated. Multistate competing risks prediction models for recurrence‐free survival (RFS), recurrence within the Milan criteria (RWM), and HCC‐specific survival (HSS) were derived to develop a prognostic calculator. Results During a median follow‐up of 51 months, 420 (43%) patients developed recurrence. In the multivariate analysis, larger tumor size, multinodularity, older age, male, higher alpha‐fetoprotein (AFP), higher albumin‐bilirubin (ALBI) grade, and the presence of portal hypertension were significantly associated with higher recurrence and decreased survival rates. Abstract Retinal ischemia is involved in the occurrence and development of various eye diseases, including glaucoma, diabetic retinopathy, and central retinal artery occlusion. To the best of our knowledge, few studies have reported self‐assembling peptide natural products for the suppression of ocular inflammation and oxidative stress. Herein, a self‐assembling peptide GFFYE is designed and synthesized, which can transform the non‐hydrophilicity of rhein into an amphiphilic sustained‐release therapeutic agent, and rhein‐based therapeutic nanofibers (abbreviated as Rh‐GFFYE) are constructed for the treatment of retinal ischemia‐reperfusion (RIR) injury. Rh‐GFFYE significantly ameliorates oxidative stress and inflammation in an in vitro oxygen‐glucose deprivation (OGD) model of retinal ischemia and a rat model of RIR injury. Rh‐GFFYE also significantly enhances retinal electrophysiological recovery and exhibits good biocompatibility. Importantly, Rh‐GFFYE also promotes the transition of M1‐type macrophages to the M2 type, ultimately altering the pro‐inflammatory microenvironment. Further investigation of the treatment mechanism indicates that Rh‐GFFYE activates the PI3K/AKT/mTOR signaling pathway to reduce oxidative stress and inhibits the NF‐κB and STAT3 signaling pathways to affect inflammation and macrophage polarization. In conclusion, the rhein‐loaded nanoplatform alleviates RIR injury by modulating the retinal microenvironment. The findings are expected to promote the clinical application of hydrophobic natural products in RIR injury‐associated eye diseases.

Abstract As an indigenous breed, the Tibetan chicken is found in highland regions and shows physiological adaptations to high altitude; however, the genetic changes that determine these adaptations remain elusive. We assumed that the microevolution of the Tibetan chicken occurred from lowland to highland regions with a continuous elevation range. In this study, we analyzed the genome of 188 chickens from lowland areas to the high‐altitude regions of the Tibetan plateau with four altitudinal levels. Phylogenetic analysis revealed that Tibetan chickens are significantly different from other altitude chicken populations. Reconstruction of the demographic history showed that the migration and admixture events of the Tibetan chicken occurred at different times. The genome of the Tibetan chicken was also used to analyze positive selection pressure that is associated with high‐altitude adaptation, revealing the well‐known candidate gene that participates in oxygen binding (HBAD), as well as other novel potential genes (e.g., HRG and ANK2) that are related to blood coagulation and cardiovascular efficiency. Our study provides novel insights regarding the evolutionary history and microevolution mechanisms of the high‐altitude adaptation in the Tibetan chicken.

Abstract This article has conducted a comprehensive study on the stator and rotor vibration characteristics of the doubly‐fed induction generator (DFIG) in the case of radial air gap eccentricity (RAGE). Differently from other previous studies, this paper not only investigates the vibration characteristics of stator and rotor in radial static air gap eccentricity (RSAGE) and radial dynamic air‐gap eccentricity (RDAGE) fault but also takes the radial hybrid air gap eccentricity (RHAGE) into account. Through theoretical derivation, the detailed expressions of stator magnetic pull per unit area (MPPUA) and rotor unbalanced magnetic pull (UMP) are obtained. The finite element analysis (FEA) and experiment based on a two pairs of poles DFIG, and the results are consistent with theoretical calculations. It is shown that RAGE will enlarge the magnitude of stator MPPUA/vibration and rotor UMP/vibration. Specifically, RSAGE will not change the frequency components of stator MPPUA/vibration but will bring new frequency components to rotor UMP/vibration. However, both stator MPPUA/vibration and rotor UMP/vibration will generate new frequency components during RDAGE fault. In addition, the effect of RHAGE is the superposition of RSAGE and RDAGE. The research conclusions obtained in this paper can be used as a supplementary criterion for diagnosing the eccentricity of DFIG.

Abstract Osteoblasts play an important role in the regulation of bone homeostasis throughout life. Thus, the damage of osteoblasts can lead to serious skeletal diseases, highlighting the urgent need for novel pharmacological targets. This study introduces chemical genetics strategy by using small molecule forskolin (FSK) as a probe to explore the druggable targets for osteoporosis. Here, this work reveals that transglutaminase 2 (TGM2) served as a major cellular target of FSK to obviously induce osteoblast differentiation. Then, this work identifies a previously undisclosed allosteric site in the catalytic core of TGM2. In particular, FSK formed multiple hydrogen bonds in a saddle‐like domain to induce an “open” conformation of the β‐sandwich domain in TGM2, thereby promoting the substrate protein crosslinks by incorporating polyamine. Furthermore, this work finds that TGM2 interacted with several mitochondrial homeostasis‐associated proteins to improve mitochondrial dynamics and ATP production for osteoblast differentiation. Finally, this work observes that FSK effectively ameliorated osteoporosis in the ovariectomy mice model. Taken together, these findings show a previously undescribed pharmacological allosteric site on TGM2 for osteoporosis treatment, and also provide an available chemical tool for interrogating TGM2 biology and developing bone anabolic agent.

Abstract The behavior of nickel (Ni) isotopes during magmatic differentiation is not adequately explored. Here, we find that tholeiitic rocks in the Kīlauea Iki (KI) lava lake, Hawai'i, show progressively lighter Ni isotopic compositions with increasing magmatic differentiation, whereas calc‐alkaline rocks from the thick Kamchatka arc (30–45 km), located at the convergent boundary of the Eurasian and Pacific plates show increasing Ni isotope values as MgO and Ni decrease. Forty‐three global intermediate‐felsic continental igneous rocks analyzed in this study display large Ni isotopic variations, with the Eoarchean samples having light Ni isotopic compositions that fall in the trend defined by the KI lavas, and the post‐Eoarchean samples showing systematically heavier Ni isotopic compositions overlapping those of Kamchatka arc rocks. The isotopic dichotomy results from the crystallization of isotopically heavy magnetite during low‐pressure differentiation of KI lavas, whereas the participation of sulfide separation that removes isotopically light Ni during high‐pressure differentiation of magmas traversing thick continental crust. Combined with Rhyolite‐MELTS and sulfur concentration at sulfide saturation simulations, we demonstrate that the Ni isotope fractionation during magmatic differentiation is mainly controlled by the tempo of magnetite crystallization and sulfide formation, which is a function of pressure, oxygen fugacity, and water activity. High‐pressure calc‐alkaline differentiation usually suppresses magnetite crystallization while stabilizing sulfide, leading to heavy Ni isotopic compositions for the evolved magmas, significantly deviating from the low‐pressure fractionation trend seen in the KI lavas. Ni isotopes can be used in the future as a tracer of magmatic differentiation and processes of continent formation and differentiation.

Abstract Objective Roberts syndrome (RBS), also known as Roberts‐SC phocomelia syndrome, is a rare autosomal recessive developmental disorder caused by mutations in the ESCO2 gene. Cardinal clinical manifestations are pre‐ and postnatal growth retardation and craniofacial and limb malformations. Here, we report RBS in a Chinese adolescent with novel biallelic ESCO2 variations and complex cerebrovascular diseases. Methods Medical history, neurological examinations, neuroimaging, and pathology were collected in the proband and the family. Whole exome sequencing (WES) with copy number variation analysis was performed to screen for genetic variations. Results The clinical features of the proband were craniofacial and limb malformations together with complex cerebrovascular diseases. She suffered ischemic stroke at 6 years old and died of cerebellar hemorrhage secondary to an aneurysm at 13 years old. Besides, neuroimaging showed the triad of leukoencephalopathy, calcifications, and cysts. Brain histopathology revealed angiomatous changes and perivascular cysts suggesting chronic small cerebral vasculopathy. Whole exome sequencing (WES) identified novel biallelic variations in the ESCO2 gene (c.1220A>T, p.H407L and c.1562delC, p.A521fs). Conclusions We describe complex cerebrovascular diseases in Roberts syndrome caused by novel ESCO2 biallelic variations. This case expands not only the cerebral involvement in Roberts syndrome but also the disease spectrum of the neuroimaging triad with leukoencephalopathy, calcifications, and cysts.

Abstract Aim To identify subgroups of cancer patients with distinct self‐care self‐efficacy profiles and to explore factors that can be used to predict those at risk of low self‐efficacy. Design A secondary analysis of data pooled from two cross‐sectional surveys was performed. Methods In total, 1,367 Chinese cancer survivors were included in the analysis. Latent profile analysis (LPA) was performed to categorize participants into latent subgroups with distinct self‐efficacy profiles. Multinomial logistic regression was conducted to identify predictors of self‐care self‐efficacy subgroup classification. Results We identified three distinct subgroups: low, medium and high self‐care self‐efficacy. Patients with the “low” profile, which was characterized by a low education level, single marital status, complications, late cancer stage and a lower level of social support, had the poorest self‐care behaviour.

Abstract Background Cancer incidence and mortality have received critical attention during the long‐term management of morbidities in patients with autoimmune diseases (AIDs). This study aimed to investigate and compare the risk of cancer associated with five major AIDs in a large‐scale Chinese cohort. Methods A total of 8,120 AID patients consecutively admitted to a national tertiary referral center in China were included and followed‐up for 38,726.55 patient‐years, including those with systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), Sjögren's syndrome (SS), systemic scleroderma (SSc), and idiopathic inflammatory myositis (IIM). Demographic data, cancer incidence, predilecting sites and cancer onset time were recorded and compared among the five AIDs. Results Four hundred and thirty (5.3%) patients developed cancer. Their median age was 57.5 years and AID duration was 79.8 months. The estimated total standardized incidence ratio (SIR) of cancer in AIDs patients was 3.37, with the highest SIR observed in IIM (4.31), followed by RA (3.99), SSc (3.77), SS (2.88) and SLE (2.58). Abstract Sodium metal, with a high theoretical specific capacity of 1165 mAh g−1, is the ultimate anode for sodium batteries, yet how to deal with the inhomogeneous and dendritic sodium deposition and the infinite relative dimension change of sodium metal anodes during sodium depositing/stripping is still challenging. Here, a facile fabricated sodiuphilic 2D N‐doped carbon nanosheets (N‐CSs) are proposed as sodium host material for sodium metal batteries (SMBs) to prevent dendrite formation and eliminate volume change during cycling. Revealing from combined in situ characterization analyses and theoretical simulations, the high nitrogen content and porous nanoscale interlayer gaps of the 2D N‐CSs can not only concede dendrite‐free sodium stripping/depositing but also accommodate the infinite relative dimension change. Furthermore, N‐CSs can be easily process into N‐CSs/Cu electrode via traditional commercial battery electrode coating equipment that pave the way for large‐scale industrial applications. On account of the abundant nucleation sites and sufficient deposition space, N‐CSs/Cu electrodes demonstrate a superior cycle stability of more than 1500 h at a current density of 2 mA cm−2 with a high coulomb efficiency of more than 99.9% and ultralow nucleation overpotential, which enable reversible and dendrites‐free SMBs and shed light on further development of SMBs with even higher performance.

Abstract As primary producers, plants provide food, oxygen, and other resources for global ecosystems, and should therefore be given priority in biodiversity protection. Most biodiversity research focuses on biodiversity hotspots, while biodiversity coldspots, such as deserts, are largely ignored. We propose that the factors shaping plant species diversity differ between biodiversity hot spots and cold spots, especially for desert ecosystems. To test this hypothesis, we investigated plant species diversity along the Modern Silk Road in the Northwest China desert, an area characterized by low precipitation, scarce vegetation, a limited number of species, and variable human activities. Surface soil was sampled from 144 plots, environmental DNA (eDNA) was extracted from soil samples, and seed plant species were identified using DNA metabarcoding technology. A total of 671 seed plant species were detected, which was more diverse than indicated by plot survey data. Plant species diversity gradually decreased from east to west along the Silk Road. In this area, temperature determines plant species diversity more than precipitation. Additionally, human activity has altered plant species diversity by introducing crops and invasive plants and eliminating environmentally adapted indigenous plants. Our results demonstrate the potential of eDNA metabarcoding technology for plant species diversity surveying. Desert plants have adapted to dry environments by relying on underground water or utilizing occasional rainfall as ephemerals, which are often not visible during surface surveys because of their short aboveground life cycle but can be detected with eDNA metabarcoding technology. Groundwater maintenance and human activity control are recommended for plant species diversity conservation and desertification control.

Abstract Background There is no consensus on whether triplet regimen is better than doublet regimen in the first‐line treatment of advanced gastric cancer (AGC). We aimed to compare the efficacy and safety of oxaliplatin plus capecitabine (XELOX) and epirubicin, oxaliplatin, plus capecitabine (EOX) regimens in treating AGC. Methods This phase III trial enrolled previously untreated patients with AGC who were randomly assigned to receive the XELOX or EOX regimen. The primary endpoint was non‐inferiority in progression‐free survival (PFS) for XELOX as compared with EOX on an intention‐to‐treat basis. Results Between April 10, 2015 and August 20, 2020, 448 AGC patients were randomized to receive XELOX (n = 222) or EOX (n = 226). The median PFS (mPFS) was 5.0 months (95% confidence interval [CI] = 4.5‐6.0 months) in the XELOX arm and 5.5 months (95% CI = 5.0‐6.0 months) in the EOX arm (hazard ratio [HR] = 0.989, 95% CI = 0.812‐1.203; Pnon‐inferiority = 0.003). There was no significant difference in median overall survival (mOS) (12.0 vs. 12.0 months, P = 0.384) or objective response rate (37.4% vs. 45.1%, P = 0.291) between the two groups. In patients with poorly differentiated adenocarcinoma and liver metastasis, the EOX arm had a significantly longer mOS (P = 0.021) and a trend of longer mPFS (P = 0.073) than the XELOX arm. The rate of grade 3/4 adverse events (AEs) was 42.2% (90/213) in the XELOX arm and 72.5% (156/215) in the EOX arm (P = 0.001). The global health‐related quality of life (QoL) score was significantly higher in the XELOX arm than in the EOX arm during chemotherapy. Conclusions This non‐inferiority trial demonstrated that the doublet regimen was as effective as the triplet regimen and had a better safety profile and QoL as a first‐line treatment for AGC patients. However, the triplet regimen might have a survival advantage in patients with poorly differentiated adenocarcinoma and liver metastasis.

Abstract Presented is a new heterometallic compound, namely [Cd2Ba2(pdc)4(H2O)4(DMA)2]}n·n(H2O) (1, H2pdc = terephthalic acid, DMA is N, N′‐dimethylacetamide), which was created with the solvothermal reaction. The architectural analysis of X‐ray indicated that the 1 reveals a three‐dimensional skeleton according to one‐dimensional infinite chains. After removal of the free along with coordinated solvent molecules, it contains 49.8% solvent‐accessible volume and unsaturated coordination sites. The solvent‐free form of 1 exhibits excellent catalytic effect for the addition reaction between diethylzinc and benzaldehyde. Its therapeutic effect and mechanism exploration on diabetes was conducted in this present research. Firstly, the abundance of intestinal flora in the gut was detected with real time RT‐PCR assay. Besides, the measurement of the levels of sugar was conducted with blood glucose meter in the T2DM animal after compound treatment. The cytotoxicity of the 1 was evaluated with CCK‐8 assay.

Background/Aims. The underlying mechanisms of differential responsiveness to IFN remain poorly understood. The elongation factor Tu GTP binding domain-containing protein 2 (EFTUD2), involved in RNA splicing and pre-mRNA processing, was recognized as a novel innate immune regulator of the IFN antihepatitis C virus (HCV) effect. The aim of this study is to elucidate the mechanisms underlying EFTUD2’s involvement in IFN-mediated anti-HBV responses. Methods. Using a CRISPR/Cas9 gene-editing system, EFTUD2 single allele knockout HepG2.2.15 cells were constructed. Subsequently, the HBV biomarkers in EFTUD2+/– HepG2.2.15 cells and wild-type (WT) cells with or without IFN-α treatment were detected. And the EFTUD2-regulated genes were then identified using mRNA sequence. Selected gene mRNA variants and their proteins were examined by qRT-PCR and Western blotting. To confirm the effects of EFTUD2 on HBV replication and IFN-stimulated gene (ISG) expression, a rescue experiment in EFTUD2+/– HepG2.2.15 cells was performed by EFTUD2 overexpression. Results. IFN-induced anti-HBV activity was found to be restricted in EFTUD2+/– HepG2.2.15 cells. The mRNA sequence showed that EFTUD2 could regulate classical IFN and virus response genes. Mechanistically, EFTUD2 single allele knockout decreased the expression of ISG-encoded proteins, comprising Mx1, OAS1, and PKR (EIF2AK2), through mediated gene splicing. However, EFTUD2 did not affect the expression of Jak-STAT pathway genes. Furthermore, EFTUD2 overexpression could restore the attenuation of IFN anti-HBV activity and the reduction of ISG resulting from EFTUD2 single allele knockout. Conclusion. EFTUD2, the spliceosome factor, is not IFN-inducible but is an IFN effector gene. EFTUD2 mediates IFN anti-HBV effect through regulation of gene splicing for certain ISGs, including Mx1, OAS1, and PKR. EFTUD2 does not affect IFN receptors or canonical signal transduction components. Therefore, it can be concluded that EFTUD2 regulates ISGs using a novel, nonclassical mechanism.

Multipotent mesenchymal stromal cells (MSCs) expand in vitro and undergo replicative senescence, thereby restricting their clinical utilization. Thus, an effective strategy is required to impede MSC senescence. Since spermidine (SPD) supplementation can prolong the lifespan of yeast by inhibiting oxidative stress, spermidine is a potential option for delaying MSC senescence. In this study, to test our hypothesis, we first isolated primary human umbilical cord mesenchymal stem cells (hUCMSCs). Subsequently, the appropriate SPD dose was administered during continuous cell cultivation. Next, we evaluated the antisenescence effects by SA-β-gal staining, Ki67 expression, reactive oxygen species (ROS) levels, adipogenic or osteogenic ability, senescence-associated markers, and DNA damage markers. The results revealed that early SPD intervention significantly delays the replicative senescence of hUCMSCs and constrains premature H2O2-induced senescence. Additionally, by silencing SIRT3, the SPD-mediated antisenescence effects disappear, further demonstrating that SIRT3 is necessary for SPD to exert its antisenescence effects on hUCMSCs. Besides, the findings of this study also suggest that SPD in vivo protects MSCs against oxidative stress and delays cell senescence. Thus, MSCs maintain the ability to proliferate and differentiate efficiently in vitro and in vivo, which reflects the potential clinical utilization of MSCs in the future.

Objective. COVID-19 has evolved into a major global public health event. The number of people reporting insomnia is growing exponentially during the pandemic. This study aimed to explore the relationship between aggravated insomnia and COVID-19-induced psychological impact on the public, lifestyle changes, and anxiety about the future. Methods. In this cross-sectional study, we used the questionnaires from 400 subjects who were obtained from the Department of Encephalopathy of the Wuhan Hospital of Traditional Chinese Medicine between July 2020 and July 2021. The data collected for the study included demographic characteristics of the participants and psychological scales consisting of the Spiegel Sleep Questionnaire, the Fear of COVID-19 Scale (FCV-19S), the Zung Self-Rating Anxiety Scale (SAS), and the Zung Self-Rating Depression Scale (SDS). The independent sample t-test and one-way ANOVA were used to compare the results. Correlation analysis of variables affecting insomnia was performed using Pearson correlation analysis. The degree of influence of the variables on insomnia was determined using linear regression, and a regression equation was derived. Results. A total of 400 insomnia patients participated in the survey. The median age was 45.75 ± 15.04 years. The average score of the Spiegel Sleep Questionnaire was 17.29 ± 6.36, that of SAS was 52.47 ± 10.39, that of SDS was 65.89 ± 8.72, and that of FCV-19S was 16.09 ± 6.81. The scores of FCV-19S, SAS, and SDS were closely related to insomnia, and the influencing degree was in the following order: fear, depression, and anxiety (OR = 1.30, 0.709, and 0.63, respectively). Conclusion. Fear of COVID-19 can be one of the primary contributors to worsening insomnia.

Abstract Aims and objectives This study aimed to evaluate the effect of a virtual simulation education programme with debriefing in undergraduate nursing students. Perceived clinical competence and learning needs of students in a simulation environment were also measured. Background Evidence showed virtual simulation education programmes provided better knowledge acquisition. However, these studies to date did not incorporate virtual simulation in the combination of a debriefing model in nursing students. Design A one‐group pre‐test and post‐test design. Methods 188 final year undergraduate nursing students participated in the study. Linear mixed model analysis was conducted to evaluate the effect of the programme. Results Students have perceived a significant improvement in clinical competence and nursing process. Self‐efficacy has also boosted. Communication and critical thinking were applied better in the traditional clinical environment. Conclusion Perceived clinical competence of Chinese nursing students has significant improvements by using virtual simulation combining a debriefing model during the COVID‐19 period. Virtual simulation met students' learning needs. Future studies should include a control group for comparison and long‐term measurement. Relevance to clinical practice The study provided an innovative clinical learning pedagogy to serve as a potential alternative with traditional clinical practicum during the COVID‐19 period as this is substantially limited.

There were approximately 1.93 million new cases and 940 000 deaths from colorectal cancer in 2020. The first‐line chemotherapeutic drugs for colorectal cancer are mainly based on 5‐fluorouracil, although the use of these drugs is limited by the development of drug resistance. Consequently, there is a need for novel chemotherapeutic drugs for the efficient treatment of colorectal cancer patients. In the present study, we screened 160 drugs approved by the Food and Drug Administration and identified that cabazitaxel (CBT), a microtube inhibitor, can suppress colony formation and cell migration of colorectal cancer cells in vitro. CBT also induces G2/M phase arrest and apoptosis of colorectal cancer cells. Most importantly, it inhibits the growth of colorectal cancer cell xenograft tumors in vivo. Transcriptome analysis by RNA‐sequencing revealed that Tub family genes are abnormally expressed in CBT‐treated colorectal cancer cells. The expression of several p53 downstream genes that are associated with cell cycle arrest, apoptosis, and inhibition of angiogenesis and metastasis is induced by CBT in colorectal cancer cells. Overall, our results suggests that CBT suppresses colorectal cancer by upregulating the p53 pathway, and thus CBT may have potential as an alternative chemotherapeutic drug for colorectal cancer.

Abstract In this study, the rotor loss as well as the temperature variations due to the static air‐gap eccentricity (SAGE) and the rotor inter‐turn short circuit (RISC) in synchronous generators is presented. Different from the previous studies, this study investigates the loss/temperature changes under not only the single SAGE fault and the single RISC fault but also the combined faults (CF) that are composed of these two single faults. Detailed formula derivation on the hysteresis loss (HL) and the eddy current loss (ECL), together with the rotor temperature rise due to varied fault types and different operating conditions, are first carried out based on a theoretical analysis of the magnetic flux density (MFD) variation. Then, 3‐D finite element analysis (FEA) and experimental verifications are performed on a CS‐5 prototype generator that has two poles and a rated rotating speed of 3000 rpm. It shows that the occurrence of either SAGE or RISC will increase the rotor loss and temperature. Comparatively, the single RISC fault overweighs the single SAGE fault in the rotor temperature increment, while the combined fault overweighs both of these single faults. In the same fault cases, the rotor temperature will have the highest value under full‐load operation.

Abstract Textile materials have been enriched in function at the composite level with continuous developments in the textile industry. Zinc oxide (ZnO) nanoparticles (ZnO‐NPs) are strongly influenced by ultraviolet (UV) filter, antifungal, high catalysis, and semiconductor/piezoelectric coupling characteristics. Therefore, the antibacterial property and UV resistance of ZnO‐NP materials are zcomprehensively analysed to provide a basis for applying ZnO‐NP in the textile industry. In addition, the textile preparation and application of ZnO‐NP in piezoelectric power generation is discussed. Based on relevant documents for ZnO‐textile industry applications, scanning electron microscopy analysis, biological activity analysis, and UV transmittance analysis of textiles containing composite materials prove that textiles based on ZnO‐based composite materials (ZnO‐NP materials) have antibacterial properties and UV resistance. The antibacterial property and UV resistance of ZnO‐NP materials are analysed comprehensively to provide a basis for applying ZnO‐NP in the textile industry. After the photocatalytic reaction, its practical application as slurry type suspensions is limited because of the difficulty of separating the catalyst particles. In terms of its piezoelectric power generation characteristics, intensity of current voltage analysis and X‐ray diffraction analysis reveal that textiles based on ZnO‐NP materials have obvious semiconductor characteristic and obvious current enhancement signals locally, indicating that the textiles can achieve better piezoelectric properties.

Abstract The main proteases (Mpro), also termed 3‐chymotrypsin‐like proteases (3CLpro), are a class of highly conserved cysteine hydrolases in β‐coronaviruses. Increasing evidence has demonstrated that 3CLpros play an indispensable role in viral replication and have been recognized as key targets for preventing and treating coronavirus‐caused infectious diseases, including COVID‐19. This review is focused on the structural features and biological function of the severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) main protease Mpro (also known as 3CLpro), as well as recent advances in discovering and developing SARS‐CoV‐2 3CLpro inhibitors. To better understand the characteristics of SARS‐CoV‐2 3CLpro inhibitors, the inhibition activities, inhibitory mechanisms, and key structural features of various 3CLpro inhibitors (including marketed drugs, peptidomimetic, and non‐peptidomimetic synthetic compounds, as well as natural compounds and their derivatives) are summarized comprehensively. Meanwhile, the challenges in this field are highlighted, while future directions for designing and developing efficacious 3CLpro inhibitors as novel anti‐coronavirus therapies are also proposed. Collectively, all information and knowledge presented here are very helpful for understanding the structural features and inhibitory mechanisms of SARS‐CoV‐2 3CLpro inhibitors, which offers new insights or inspiration to medicinal chemists for designing and developing more efficacious 3CLpro inhibitors as novel anti‐coronavirus agents.
