Difficulties associated with the integration of liquids into a UHV environment make surface-science style studies of mineral dissolution particularly challenging. Recently, we developed a novel experimental setup for the UHV-compatible dosing of ultrapure liquid water, and studied its interaction with TiO2 and Fe3O4 surfaces. Here, we describe a simple approach to vary the pH through the partial pressure of CO2 (pCO2) in the surrounding vacuum chamber, and use this to study how these surfaces react to an acidic solution. The TiO2(110) surface is unaffected by the acidic solution, except for a small amount of carbonaceous contamination. The Fe3O4(001)-(rt2 x rt2)R45 surface begins to dissolve at a pH 4.0-3.9 (pCO2 = 0.8-1 bar) and, although it is significantly roughened, the atomic-scale structure of the Fe3O4(001) surface layer remains visible in scanning tunneling microscopy (STM) images. X-ray photoelectron spectroscopy (XPS) reveals that the surface is chemically reduced, and contains a significant accumulation of bicarbonate (HCO3-) species. These observations are consistent with Fe(II) being extracted by bicarbonate ions, leading to dissolved iron bicarbonate complexes (Fe(HCO3)2), which precipitate onto the surface when the water evaporates.

Mithilfe einer Kombination aus Photoemissionsspektroskopie, Rasterkraft- und Rastertunnelmikroskopie/-spektroskopie wird gezeigt, dass sich uberschussige Elektronen auf einer Anatas-TiO2(101)-Oberflache an Stufenkanten ansammeln. Stufenkanten fungieren daher als bevorzugte Adsorptionsplatze fur O2. Auch wenn in Dichtefunktionalrechnungen bereits bei sauberen Stufenkanten eine Lokalisierung der Elektronen festgestellt wird, wird dieser Effekt durch Sauerstofffehlstellen und Hydroxylierung noch weiter verstarkt. Die vorliegenden Befunde illustrieren die Bedeutung von Defekten bei den vielfaltigen Anwendungen von Titandioxid.

HAMP domains mediate input-output transactions in many bacterial signalling proteins. To clarify the mechanistic logic of HAMP signalling, we constructed Tsr-HAMP deletion derivatives and characterized their steady-state signal outputs and sensory adaptation properties with flagellar rotation and receptor methylation assays. Tsr molecules lacking the entire HAMP domain or just the HAMP-AS2 helix generated clockwise output signals, confirming that kinase activation is the default output state of the chemoreceptor signalling domain and that attractant stimuli shift HAMP to an overriding kinase-off signalling state to elicit counter-clockwise flagellar responses. Receptors with deletions of the AS1 helices, which free the AS2 helices from bundle-packing constraints, exhibited kinase-off signalling behaviour that depended on three C-terminal hydrophobic residues of AS2. We conclude that AS2/AS2' packing interactions alone can play an important role in controlling output kinase activity. Neither kinase-on nor kinase-off HAMP deletion outputs responded to sensory adaptation control, implying that out-of-range conformations or bundle-packing stabilities of their methylation helices prevent substrate recognition by the adaptation enzymes. These observations support the previously proposed biphasic, dynamic-bundle mechanism of HAMP signalling and additionally show that the structural interplay of helix-packing interactions between HAMP and the adjoining methylation helices is critical for sensory adaptation control of receptor output.

HAMP domains mediate input-output communication in many bacterial signalling proteins. To explore the dynamic bundle model of HAMP signalling (Zhou et al., Mol. Microbiol. 73: 801, 2009), we characterized the signal outputs of 118 HAMP missense mutants of the serine chemoreceptor, Tsr, by flagellar rotation patterns. Receptors with proline or charged amino acid replacements at critical hydrophobic packing residues in the AS1 and AS2 HAMP helices had locked kinase-off outputs, indicating that drastic destabilization of the Tsr-HAMP bundle prevents kinase activation, both in the absence and presence of the sensory adaptation enzymes, CheB and CheR. Attractant-mimic lesions that enhance the structural stability of the HAMP bundle also suppressed kinase activity, demonstrating that Tsr-HAMP has two kinase-off output states at opposite extremes of its stability range. HAMP mutants with locked-on kinase outputs appeared to have intermediate bundle stabilities, implying a biphasic relationship between HAMP stability and kinase activity. Some Tsr-HAMP mutant receptors exhibited reversed output responses to CheB and CheR action that are readily explained by a biphasic control logic. The findings of this study provide strong support for a three-state dynamic bundle model of HAMP signalling in Tsr, and possibly in other bacterial transducers as well.

During chemotactic signalling by Escherichia coli, the small cytoplasmic CheW protein couples the histidine kinase CheA to chemoreceptor control. Although essential for assembly and operation of receptor signalling complexes, CheW in stoichiometric excess disrupts chemotactic behaviour. To explore the mechanism of the CheW excess effect, we measured the physiological consequences of high cellular levels of wild-type CheW and of several CheW variants with reduced or enhanced binding affinities for receptor molecules. We found that high levels of CheW interfered with trimer assembly, prevented CheA activation, blocked cluster formation, disrupted chemotactic ability and elevated receptor methylation levels. The severity of these effects paralleled the receptor-binding affinities of the CheW variants. Because trimer formation may be an obligate step in the assembly of ternary signalling complexes and higher-order receptor arrays, we suggest that all CheW excess effects stem from disruption of trimer assembly. We propose that the CheW-binding sites in receptor dimers overlap their trimer contact sites and that high levels of CheW saturate the receptor-binding sites, preventing trimer assembly. The CheW-trapped receptor dimers seem to be improved substrates for methyltransferase reactions, but cannot activate CheA or assemble into clusters, processes that are essential for chemotactic signalling.

On November 28, 2012 Alexander (Alex) Bohm, a bacterial geneticist, died at age 41, only a few months after taking up a position as an assistant professor at the LOEWE Center for Synthetic Microbiology in Marburg, Germany. Earlier in 2012 Alex had been diagnosed with an aggressive form of thyroid cancer that left him little time to live his scientific and personal dreams.

Chemical signals sensed on the periplasmic side of bacterial cells by transmembrane chemoreceptors are transmitted to the flagellar motors via the histidine kinase CheA, which controls the phosphorylation level of the effector protein CheY. Chemoreceptor arrays comprise remarkably stable supramolecular structures in which thousands of chemoreceptors are networked through interactions between their cytoplasmic tips, CheA, and the small coupling protein CheW. To explore the conformational changes that occur within this protein assembly during signalling, we used in vivo crosslinking methods to detect close interactions between the coupling protein CheW and the serine receptor Tsr in intact E. coli cells. We identified two signal-sensitive contacts between CheW and the cytoplasmic tip of Tsr. Our results suggest that ligand binding triggers changes in the receptor that alter its signalling contacts with CheW (and/or CheA).

Summary The serine chemoreceptor Tsr and other methylaccepting chemotaxis proteins (MCPs) control the swimming behaviour of Escherichia coli by generating signals that influence the direction of flagellar rotation. MCPs produce clockwise (CW) signals by stimulating the autophosphorylation activity of CheA, a cytoplasmic histidine kinase, and counterclockwise signals by inhibiting CheA. CheW couples CheA to chemoreceptor control by promoting formation of MCP/CheW/CheA ternary complexes. To identify MCP structural determinants essential for CheA stimulation, we inserted fragments of the tsr coding region into an inducible expression vector and used a swimming contest called ‘pseudotaxis’ to select for transformant cells carrying CW-signalling plasmids. The shortest active fragment we found, Tsr (350‐470), stimulated CheA in a CheW-dependent manner, as full-length Tsr molecules do. It spans a highly conserved ‘core’ (370‐420) that probably specifies the CheA and CheW contact sites and other determinants needed for stimulatory control of CheA. Tsr (350‐470) also carries portions of the left and right arms flanking the core, which probably play roles in regulating MCP signalling state. However, this Tsr fragmentlacksall of the methylation sitescharacteristic of MCP molecules, indicating that methylation segments are not essential for generating receptor output signals.

The effect of reel speed, can size, and percent air during steam and steam/air processing in a Steritort on the heating rate of tomato concentrate (an induced convection-heating product) was investigated. A Taylor expansion equation, relating Ball process times with the above three variables, considering first order, second order, and interaction effects, was presented. A correlation equation for the Nusselt number as a function of the Reynolds and Prandtl numbers, the can length over the diameter ratio, and the steam content was also presented. Percent reduction in process lethality increased with increasing can size and air content, and decreasing reel speed and target lethality. Increase in percent air in the steam/air mixture could be compensated by an increase of the rotational speed.

Effects of residual air were evaluated on the heating rates of a convectionheating product processed in a continuous rotary sterilizer. Heat penetration tests were conducted in a Steritort at various reel speeds (1-10 rpm) and air overpressures (0-69.0 kPa) using 211 × 300, 300 × 407 and 603 × 700 cans containing Washington white beans in brine. Sterilization times calculated by Ball's formula method were longer for test cans processed with overpressure than those processed with no overpressure. Maximum reduction in calculated lethality for 69.0 kPa air overpressure was 15.2% for 211 × 300 and 300 × 407 experiments and 49.4% for 603 × 700 experiments. For each can size and reel speed, average process time was a linear function of overpressure.

Plasma oxalate and erythrocyte glutamic oxaloacetate transaminase activity (EGOT) (an indicator of nutritional status with respect to pyridoxine) were measured in 21 patients maintained on regular continuous ambulatory peritoneal dialysis or haemodialysis before and after a 4-month period of supplementation with pyridoxine, 100 mg day-1. Prior to supplementation 10/21 patients showed subnormal EGOT activity, although the increment in activity on addition of pyridoxal-5-phosphate in vitro was within the normal range in all cases. Mean plasma oxalate was 31.5 μmol 1-1 (SEM 2.9) prior to supplementation and did not change significantly with supplementation, despite normalization of EGOT activity in all but 2/21 patients. We conclude that pyridoxine deficiency does not contribute significantly to hyperoxalaemia in patients receiving dialysis and that 100 mg of pyridoxine daily is insufficient to reduce oxalate generation by a pharmacological action on glycine transamination.
