Abstract Age at first farrowing (AFF) is a reproductive trait with low heritability and high importance in the pig industry. To enhance the statistical power of genome‐wide association study (GWAS) and further explore the genetic nature of AFF, we first conducted GWAS meta‐analysis using three Yorkshire populations, and then integrated the Pig Genotype‐Tissue Expression (PigGTEx) resources to interpret their potential regulatory mechanism. Additionally, we compared the AFF in pig with the age at first birth (AFB) of human using GWAS summary statistics. We detected 18 independent variants in GWAS meta‐analysis and 8 genes in gene‐based association analysis significantly associated with AFF. By integrating the PigGTEx resource, we conducted transcriptome‐wide association study (TWAS) and colocalization analysis on 34 pig tissues. In TWAS, we detected 18 significant gene‐tissue pairs, such as DCAF6 in uterus and CREG1 in blood. In colocalization, we found 111 potential candidate tissue‐gene pairs, such as GJD4 and LYPLAL1. We found that the homologous gene, CHST10, might be the potential candidate gene between humans in AFB and pigs in AFF. In conclusion, integrating GWAS meta‐analysis and PigGTEx resources is a meaningful way to decipher the genetic architecture of complex traits. We found that DCAF6, CREG1, GJD4, and LYPLAL1 are candidate genes with high reliability for AFF in swine. The comparative analysis showed that CHST10 might play a potentially critical role in AFB/AFF across human and pigs.

Abstract Persister cells (PS) selected for anticancer therapy have been recognized as a significant contributor to the development of treatment‐resistant malignancies. It is found that imposing glutamine restriction induces the generation of PS, which paradoxically bestows heightened resistance to glutamine restriction treatment by activating the integrated stress response and initiating the general control nonderepressible 2‐activating transcription factor 4‐alanine, serine, cysteine‐preferring transporter 2 (GCN2‐ATF4‐ASCT2) axis. Central to this phenomenon is the stress‐induced ATF4 translational reprogramming. Unfortunately, directly targeting ATF4 protein has proven to be a formidable challenge because of its flat surface. Nonetheless, a G‐quadruplex structure located within the promoter region of ATF4 (ATF4‐G4) is uncovered and resolved, which functions as a transcriptional regulator and can be targeted by small molecules. The investigation identifies the natural compound coptisine (COP) as a potent binder that interacts with and stabilizes ATF4‐G4. For the first time, the high‐resolution structure of the COP‐ATF4‐G4 complex is determined. The formation of this stable complex disrupts the interaction between transcription factor AP‐2 alpha (TFAP2A) and ATF4‐G4, resulting in a substantial reduction in intracellular ATF4 levels and the eventual death of cancer cells. These seminal findings underscore the potential of targeting the ATF4‐G4 structure to yield significant therapeutic advantages within the realm of persister cancer cells induced by glutamine‐restricted therapy.

Objective Dynamization of the external fixator, defined as gradually decreasing construct‐stability of the fixator, is widely accepted as a method for treatment during the late phase of the bone healing process. However, the dynamization is mostly based on the subjective experience of orthopaedists at present, without unified standards and a clear theoretical basis. The objective of the study is to investigate the influence of the dynamization operations on the tibial mechanical properties with a hexapod circular external fixator and standardize the dynamization process. Methods A 3D‐printed tibial defects model with Young's modulus of 10.5 GPa and Poisson's ratio of 0.32 simulated the clinically fractured bone. A 10 × ∅ 45 mm silicone sample with Young's modulus of 2.7 MPa and Poisson's ratio of 0.32 simulated the callus in the fracture site. Furthermore, a hexapod circular external fixator whose struts were coded from #1 to #6 was fixed on the model with six half‐pins (5 mm diameter). Corresponding to the action of removing and loosening the struts, 17 dynamization operations are designed. For each construct after different dynamization operations, the mechanical environment changes in the fracture site were recorded by a triaxle forces sensor under gradually increasing external load from 0 to 500 N. Results The results show that the bone axial load‐sharing ratio of each construct in the removal group was generally higher than that in the loosening group. The ratio increased from 92.51 ± 0.74% to 102.68 ± 0.27% with the number of operated struts rising from 2 to 6. Besides, the constructions with the same number of operated struts but with different strut codes such as constructions 3–5, had similar bone axial load‐sharing ratios. In addition, the proposed dynamization method of the hexapod circular external fixator can gradually increase the bone axial load‐sharing ratio from 90.73 ± 0.19% to 102.68 ± 0.27% and maintain the bone radial load‐sharing ratio below 8%. Conclusion The laboratory study verified the effects of the type of operations and the number of operated struts on the bone axial load‐sharing ratio, as well as the slight influence of the choice of the strut code. Besides, a dynamization method of the hexapod circular external fixator was proposed to increase the bone axial load‐sharing ratio gradually.

Abstract Background The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial, and more high‐level clinical evidence is needed. This study aimed to evaluate the outcome of extended pancreatoduodenectomy (EPD) with retroperitoneal nerve resection in pancreatic head cancer. Methods This multicenter randomized trial was performed at 6 Chinese high‐volume hospitals that enrolled patients between October 3, 2012, and September 21, 2017. Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments (preoperative chemotherapy or chemoradiation) within three months were randomly assigned to undergo standard pancreatoduodenectomy (SPD) or EPD, with the latter followed by dissection of additional lymph nodes (LNs), nerves and soft tissues 270° on the right side surrounding the superior mesenteric artery and celiac axis. The primary endpoint was overall survival (OS) by intention‐to‐treat (ITT). The secondary endpoints were disease‐free survival (DFS), mortality, morbidity, and postoperative pain intensity. Results The R1 rate was slightly lower with EPD (8.46%) than with SPD (12.56%). The morbidity and mortality rates were similar between the two groups. The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort (23.0 vs. 20.2 months, P = 0.100), while the median DFS was superior in the EPD group (16.1 vs. 13.2 months, P = 0.031). Patients with preoperative CA19–9 < 200.0 U/mL had significantly improved OS and DFS with EPD (EPD vs. SPD, 30.8 vs. 20.9 months, P = 0.009; 23.4 vs. 13.5 months, P < 0.001). The EPD group exhibited significantly lower locoregional (16.48% vs. 35.20%, P < 0.001) and mesenteric LN recurrence rates (3.98% vs. 10.06%, P = 0.022). The EPD group exhibited less back pain 6 months postoperation than the SPD group. Conclusions EPD for pancreatic head cancer did not significantly improve OS, but patients with EPD treatment had significantly improved DFS. In the subgroup analysis, improvements in both OS and DFS in the EPD arm were observed in patients with preoperative CA19–9 < 200.0 U/mL. EPD could be used as an effective surgical procedure for patients with pancreatic head cancer, especially those with preoperative CA19–9 < 200.0 U/mL.

Background Cardiac failure is the primary cause of death in most patients with pulmonary arterial hypertension (PH). As pleiotropic cytokines, human resistin (Hresistin) and its rodent homolog, resistin‐like molecule α, are mechanistically critical to pulmonary vascular remodeling in PH. However, it is still unclear whether activation of these resistin‐like molecules can directly cause PH‐associated cardiac dysfunction and remodeling. Methods and Results In this study, we detected Hresistin protein in right ventricular (RV) tissue of patients with PH and elevated resistin‐like molecule expression in RV tissues of rodents with RV hypertrophy and failure. In a humanized mouse model, cardiac‐specific Hresistin overexpression was sufficient to cause cardiac dysfunction and remodeling. Dilated hearts exhibited reduced force development and decreased intracellular Ca2+ transients. In the RV tissues overexpressing Hresistin, the impaired contractility was associated with the suppression of protein kinase A and AMP‐activated protein kinase. Mechanistically, Hresistin activation triggered the inflammation mediated by signaling of the key damage‐associated molecular pattern molecule high‐mobility group box 1, and subsequently induced pro‐proliferative Ki67 in RV tissues of the transgenic mice. Intriguingly, an anti‐Hresistin human antibody that we generated protected the myocardium from hypertrophy and failure in the rodent PH models. Conclusions Our data indicate that Hresistin is expressed in heart tissues and plays a role in the development of RV dysfunction and maladaptive remodeling through its immunoregulatory activities. Targeting this signaling to modulate cardiac inflammation may offer a promising strategy to treat PH‐associated RV hypertrophy and failure in humans.

Purpose. To investigate the changes in thromboelastography (TEG) in patients with dyslipidemia to study its effect on the blood coagulation status. Methods. 131 patients hospitalized in Fujian Provincial Jinshan Hospital from January 2018 to December 2020 were selected, and 64 cases in the hyperlipidemia (HL) group and 67 cases in the non-HL group were set according to whether their blood lipids were abnormal. By measuring the changes of each parameter of TEG in patients, the relevant parameters R value, K value, α angle, and MA value were calculated. And routine blood coagulation (PT, APTT, INR, FIB, and TT) and routine blood (platelet count) tests were performed on all study subjects to analyze the changes of each index of the coagulation function and each parameter of TED in both groups and explore the clinical value of TEG on HL diseases. Results. Compared with the non-HL group, R and K values decreased, and angle and MA values increased in the HL group (P0.05). State-of-the-art facial expression methods outperform human beings, especially, thanks to the success of convolutional neural networks (CNNs). However, most of the existing works focus mainly on analyzing an adult’s face and ignore the important problems: how can we recognize facial expression from a baby’s face image and how difficult is it? In this paper, we first introduce a new face image database, named BabyExp, which contains 12,000 images from babies younger than two years old, and each image is with one of three facial expressions (i.e., happy, sad, and normal). To the best of our knowledge, the proposed dataset is the first baby face dataset for analyzing a baby’s face image, which is complementary to the existing adult face datasets and can shed some light on exploring baby face analysis. We also propose a feature guided CNN method with a new loss function, called distance loss, to optimize interclass distance. In order to facilitate further research, we provide the benchmark of expression recognition on the BabyExp dataset. Experimental results show that the proposed network achieves the recognition accuracy of 87.90% on BabyExp.

Multimetallic Prussian blue compound Ni0.25Mn1.25[Fe(CN)6]·6.1H2O has been prepared by coprecipitation. The temperature-dependent magnetic susceptibilities show the magnet transition for the compound at 8.5 K. According to DC variable-temperature magnetic susceptibility paramagnetic Curie temperature θ is −9.32 K. The observed value of coercive field (Hc) and the remanent magnetization (Mr) for the compound are 0.32 KOe and 0.36 μB. According to study of zero-field-cooled (ZFC) and field-cooled (FC) magnetization curves and AC magnetization curves, there exists a spin-glass behaviour in the compound, which exhibits freezing temperature Tg=7.76 K, below magnetic transition TC=8.5 K; that glass behavior is termed “reentrant” spin glass.

Abstract Specific emitter identification (SEI) represents a prominent research direction within the electronic countermeasures domain aimed at discerning carrier identity attributes by analysing subtle radar characteristics. At present, most established SEI techniques assume that both the source and target domain (TD) data adhere to the same distribution. However, this assumption is invalidated by semantic drift which frequently occurs between TD and source domain (SD) samples owing to variations in the collection environment, equipment, and other factors. Considering the aforementioned challenges, this article introduces a transfer learning approach for SEI to leverage pseudo‐label integration within the framework of meta‐learning. This approach employs the bispectral perimeter integral for extracting emitter signal features to construct a feature extractor and basic learner based on CNN13. To label and filter the TD samples, the proposed approach utilises the multiple pseudo‐label serial filtering mechanism, which comprises positive and negative pseudo‐labelling strategies, label uncertainty prediction methods, and hard sample filtering strategies. Ultimately, to address algorithmic real‐time requirements, the labelled TD samples are integrated into the feature extractor and learner of the SD through meta‐learning to facilitate the transfer of TD features to the SD training model. Experimental validation conducted on a real radar dataset demonstrated that the proposed algorithm significantly enhances identification accuracy, exhibiting an improvement from approximately 50% to approximately 90%. Furthermore, the algorithm exhibits a short runtime and robust adaptability, effectively catering to the demands of practical scenarios.
