Studies have found that miR-665 acted as a tumor suppressor or an oncogene in different malignancies. miR-665 expression was elevated in gastric adenocarcinoma tissues; however, its role and mechanism in this disease are not fully clarified. The expression of miR-665 and its target gene was detected in human gastric adenocarcinoma tissues and cells. Moreover, we analyzed the effects of miR-665 on the proliferation, migration, and epithelial-mesenchymal transition (EMT) of gastric adenocarcinoma cells as well as tumor growth in vivo. The mechanisms of miR-665 in gastric adenocarcinoma were investigated by using molecular biology techniques. We found miR-665 was upregulated and suppressor of cytokine signaling 3 (SOCS3) was downregulated in gastric adenocarcinoma tissues and cells. Elevated miR-665 was positively correlated with tumor size, lymph node metastasis, invasion depth, TNM stage, and poor differentiation in gastric adenocarcinoma patients. Overexpression of miR-665 promoted, whereas knockdown of miR-665 suppressed the proliferation, migration, and EMT of gastric adenocarcinoma cells. Furthermore, we demonstrated that miR-665 functioned through targeting SOCS3, followed by activation of the FAK/Src signaling pathway in gastric adenocarcinoma cells. miR-665 antagomir inhibited tumor growth as well as the activation of the FAK/Src pathway but increased SOCS3 expression in nude mice. In addition, miR-665 expression was negatively regulated by long noncoding RNA maternally expressed gene 3 (MEG3). In conclusion, miR-665 acted as an oncogene in gastric adenocarcinoma by inhibiting SOCS3 followed by activation of the FAK/Src pathway and it was negatively modulated by MEG3. miR-665 may be a promising therapeutic target for the treatment of gastric adenocarcinoma.

Background At present, the expression of MOR1 and its function in gastric cancer remains unclear with evidence suggesting that it is to be involved in tumor progression and metastasis. The study was to assess the clinicopathologic relevance and prognostic value of MOR1 expression in gastric cancer. Methods Real-time quantitative RT-PCR and immunohistochemical staining were used to detect MOR1 expression in primary gastric cancerous surgical specimens and adjacent nontumorous tissues. Results High MOR1 expression was detected in cancerous tumor compared with their adjacent nontumorous tissues. In addition, the chi-square test revealed that high MOR1 expression was significantly correlated with depth of invasion (p = 0.006), lymph node metastasis (p = 0.001), distant metastasis (p = 0.017), and TNM staging (p = 0.027). Moreover, Kaplan–Meier analysis revealed a significant association between MOR1 expression and overall survival. High expression of MOR1 was identified as an independent and significant predictor gene of reduced postoperative survival. Conclusion We conclude that MOR1 expression may be a useful biomarker for better prediction of the clinical outcome and management of gastric cancer patients.

The application of small specimen creep test techniques in the evaluation of creep properties of materials in-service has been increasing. To obtain the creep data accurately and conveniently, a new creep test method with small cantilever beam specimens is proposed. Analytical equations are derived that can convert the load to equivalent uniaxial stress and the displacement rate to equivalent uniaxial strain rate. Three types of the cantilever beam specimens are designed. The optimal configuration of the cantilever beam specimens is recommended with the aid of finite element method, which is further validated by the cantilever beam and uniaxial specimen tests. The results show that parameters obtained from the cantilever beam tests correspond reasonably well with those from uniaxial tests at low stress levels. With a relatively large equivalent gauge length, the cantilever beam specimen allows the small creep strain rate data obtained with a high accuracy.

With a new approach, the general current expressions of two typical second order catalytic reactions are obtained for disk, hemispherical and spherical microelectrodes under steady-state conditions. This approach is based on the concept of reaction layer. For the second order EC reactions, we also discussed how it is possible to observe pseudo-first order or second order behavior. This consideration allows the study of the fast chemical reactions and systems where the bulk concentrations of the reactants are nearly equal. Regeneration of Fe3+(EDTA) with H202 was selected as an example of a rapid catalytic reaction.
