1. Comparative Analysis of Nonlinear Methods for Multivariable Water Level Prediction: The Case Study of the Atrato River

Abstract

This paper presents an innovative multivariate forecasting application for hydrological variables, using an online model based on backpropagation (BP) neural networks. The effectiveness of this approach is verified by using the Nonlinear Autoregressive with Exogenous Inputs (NARX) and the linear Autoregressive with Exogenous Inputs (ARX) methods. The proposed methodology is implemented in two hydrological stations in the department of Chocó, Colombia, located along the Atrato River. The BP neural network structure is applied to establish correlations between water level, flow, and precipitation. This structure is designed to handle the dynamic complexity of the hydrological variables and their interconnections. In addition, a short-term water level prediction is developed for incorporation into a flood early warning system. Validation of the approach is performed by comparing various estimation errors using regression metrics such as root mean square error (RMSE), relative mean square error (MSRE), mean absolute error (MAE), relative mean absolute error (MARE), and Nash–Sutcliffe efficiency coefficient (NSE). The computation time and the accuracy of the estimated weights versus real data are also considered. The results show that the proposed structure is the most suitable for accurately predicting water levels. It is shown that the backpropagation neural network structure of the proposed model is more effective for water level prediction. This is observed when evaluating the real data compared to the estimated data, obtaining values corresponding to the regression metrics. In particular, for output 2, the RMSE is 0.0742. Compared to the NARX model (RMSE of 0.0985) and the ARX model (RMSE of 0.1265), it is observed that the backpropagation neural network model improves the estimation by 32.6 over the NARX model and by 70.4% over the ARX model. As for the NSE coefficient for output 2 of the model, the backpropagation neural network is the closest to 1 compared to the other two models. For output 2, the backpropagation model obtains a value of 0.9966, the NARX model 0.9958, and the ARX model 0.9914. Another evaluation metric applied to this model is the Kling–Gupta efficiency coefficient (KGE), which is used to evaluate the performance of hydrological models. For the output of the two hydrological stations located in the Atrato River, we observed that for the backpropagation model a value of 0.9992 was obtained, for the NARX model a value of 0.9988 was obtained, and for the linear ARX model with nonlinear structure a value of 0.9984 was obtained. This indicates that the backpropagation neural network model provides a better estimate, as its value is closer to 1. Furthermore, the computational time executed in the algorithm of the three proposed nonlinear models is calculated, where the backpropagation obtains a calculated value of 350.9 microseconds, for the NARX model the computational time is 5908.7 microseconds and for the ARX nonlinear model, the computational time is 5172.6 microseconds. The model put forward demonstrates its effectiveness in accurately predicting water levels at two key hydrological stations along Colombia’s Atrato River. By incorporating intricate interactions among hydraulic structures and dynamic hydrological conditions, it enhances prediction accuracy. This not only aids in disaster risk management but also streamlines planning processes, particularly within flood early warning systems.

Abstract

Abstract Species delimitation among closely related species is challenging because traditional phenotype‐based approaches, for example, using morphology, ecological, or chemical characteristics, may not coincide with natural groupings. With the advent of high‐throughput sequencing, it has become increasingly cost‐effective to acquire genome‐scale data which can resolve previously ambiguous species boundaries. As the availability of genome‐scale data has increased, numerous species delimitation analyses, such as BPP and SNAPP+Bayes factor delimitation (BFD*), have been developed to delimit species boundaries. However, even empirical molecular species delimitation approaches can be biased by confounding evolutionary factors, for example, hybridization/introgression and incomplete lineage sorting, and computational limitations. Here, we investigate species boundaries and the potential for micro‐endemism in a lineage of lichen‐forming fungi, Niebla Rundel & Bowler, in the family Ramalinaceae by analyzing single‐locus and genome‐scale data consisting of (a) single‐locus species delimitation analysis using ASAP, (b) maximum likelihood‐based phylogenetic tree inference, (c) genome‐scale species delimitation models, e.g., BPP and SNAPP+BFD, and (d) species validation using the genealogical divergence index (gdi). We specifically use these methods to cross‐validate results between genome‐scale and single‐locus datasets, differently sampled subsets of genomic data and to control for population‐level genetic divergence. Our species delimitation models tend to support more speciose groupings that were inconsistent with traditional taxonomy, supporting a hypothesis of micro‐endemism, which may include morphologically cryptic species. However, the models did not converge on robust, consistent species delimitations. While the results of our analysis are somewhat ambiguous in terms of species boundaries, they provide a valuable perspective on how to use these empirical species delimitation methods in a nonmodel system. This study thus highlights the challenges inherent in delimiting species, particularly in groups such as Niebla, with complex, relatively recent phylogeographic histories.

3. Likeability and perceived effectiveness of messages designed to encourage physical activity participation among older adults

Abstract

ISSUE ADDRESSED: Older adults are significantly less likely than their younger counterparts to engage in physical activity. Comprehensive policies to support healthy ageing thus need to include components encouraging greater participation in physical activity in later life. This study tested potential messages for use in health communication campaigns aimed at increasing physical activity among older adults. METHODS: Twelve written messages designed to encourage older adults to increase their levels of physical activity were rated by Australians aged 60-92 years (n = 369; 54% female) on the variables of likeability and perceived effectiveness. RESULTS: Ratings for all the tested messages were high across both outcome variables. The message Move more, live longer scored most favourably, with large majorities of participants liking this message (87%) and considering it effective (81%). Messages featuring rhyming or alliteration were rated significantly higher on both outcomes than messages without these attributes (all Ps < .001). CONCLUSIONS: Results provide insights into the types of messages that are likely to be accepted by older adults. Statements that use the phonological patterns of rhyming or alliteration are likely to be especially well received. SO WHAT?: Increasing participation in physical activity among older adults is critical to promoting the sustainability of health care systems and enhancing quality of life. The specific messaging attributes identified in the present study as being effective could be used by public health practitioners to inform their approach to physical activity messaging to older adults and incorporated into future health communication campaigns to increase their potential effectiveness with this target group.

6. Communicating with older people about physical activity

Abstract

OBJECTIVE: Little is known about how to effectively encourage higher levels of activity among older people. This study tested the effectiveness of a public service advertisement designed according to recommendations for communicating with older audiences and featuring five types of moderate-to-vigorous physical activity: tennis, line dancing, cycling, swimming and jogging. METHODS: A survey administered to 1,200 Australians aged 50+ years assessed effects of the public service advertisement on: motivation (intrinsic and extrinsic); perceived believability, relevance, and effectiveness; and feelings elicited (e.g. interest, hope, guilt). Open-ended questions enabled respondents to describe aspects of the ad they considered to be most and least effective. RESULTS: Moderate to high scores were obtained on the motivation measures and the ad evaluation criteria of perceived effectiveness, likeability, believability and personal relevance. Mean scores for the feelings measures were generally low, with the exception of the positive feelings of being interested, inspired, hopeful and determined. CONCLUSIONS: The results suggest physical activity promotion ads can be motivating across age and socioeconomic subgroups of older people. Implications for public health: Ads aiming to encourage older people to be more physically active may be accepted and effective if they depict everyday older people enjoying a range of relevant activities.

7. Development of a scale assessing retired older adults' attitudes to volunteering

OBJECTIVE: Given positive attitudes to volunteering are likely to be associated with support for volunteering and individual participation, this study aimed to develop and test a 10-item 'attitudes to formal volunteering' scale for retired older adults that could be used to inform intervention efforts. METHOD: A sample of 801 Australian retirees (62% female; mean age: 71.9 years) completed a survey that included the attitude scale, demographic items and questions assessing engagement in volunteering. RESULTS: A principal component analysis identified two factors (general attitude to volunteering and attitude to participating in volunteering) that together accounted for 64% of the variance in attitude scores. A confirmatory factor analysis indicated the two-factor model was an excellent fit to the data. Cronbach's alphas for both factors were >0.80, and both were positively associated with volunteering engagement. CONCLUSION: The proposed scale is a potentially useful instrument for measuring attitudes to volunteering among retired older adults.

Abstract

Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives.

Abstract

Establishing integration of an individual bird into a wild flock is particularly important in species that are highly gregarious and are reliant on the flock to increase their likelihood of survivorship. When individuals, rehabilitated and reintroduced to wild flocks, are tracked through satellite or global positioning system (GPS) telemetry, it is of further significance to establish integration to assess rehabilitation success and whether the individual can serve as a marker of the flock. To date, for black cockatoos (Calyptorhynchus spp.) this has been achieved through visual observations, which requires field‐based observations that may not be possible because of inaccessible terrain, or may be untenable because the tagged individual can move quickly or is cryptic within a given land cover type. To establish whether an individual had joined a flock, we proposed the use of behavioral change point analysis. Our analyses showed that for GPS data of 6 individuals of the 3 black cockatoo species endemic to Western Australia we could demonstrate behavioral differences in their movement paths that were either individual or flocked (integrated) behavior. We undertook field observations to validate integration into wild flocks. We characterized flocked behavior as a constant, repeated pattern at variable levels of velocity. Individual behavior manifested in 2 different forms: resident and exploratory behavior. The analysis showed that all birds had integrated into a wild flock within a month of release. Behavioral change point analysis is a useful method to characterize movement behavior in black cockatoos and to confirm their membership within a flock. Furthermore, confirmation of flock membership makes it possible to use the data from 1 tagged bird to indicate movement behavior at the flock level.

Ruzasvir (MK-8408, an NS5A inhibitor) and uprifosbuvir (MK-3682, a nonstructural protein 5B nucleotide inhibitor) are highly potent direct-acting antiviral agents for the treatment of hepatitis C virus (HCV) infection. A phase III clinical trial evaluating the two-drug combination of ruzasvir 60 mg plus uprifosbuvir 450 mg suggested suboptimal efficacy in certain HCV genotypes (C-BREEZE 1; NCT02759315). The aim of the present study was to evaluate the efficacy and safety of ruzasvir in combination with uprifosbuvir administered at a higher dose than that assessed in the earlier study (C-BREEZE 2: NCT02956629/Merck protocol PN041). Treatment-naïve or interferon (with or without ribavirin)-experienced participants with or without compensated cirrhosis were enrolled. All participants received ruzasvir 180 mg plus uprifosbuvir 450 mg once daily for 12 weeks. The primary objectives were the proportion of participants with HCV RNA <15 lU/mL at 12 weeks after the end of study therapy (SVR12), and safety and tolerability of the study drug. Overall, 282 participants were enrolled. SVR12 (n/N) was 91.3% (42/46) in participants infected with HCV genotype (GT) 1a; GT1b, 96.7% (29/30); GT2, 91.5% (43/47); GT3, 73.8% (45/61); GT4, 98.2% (55/56); GT5, 100.0% (18/18); and GT6, 90.9% (20/22). Adverse events (AEs) were reported by 61.3% of participants; drug-related AEs were reported by 33.3%. The most frequent (≥5% of participants) drug-related AEs in all participants were fatigue (7.8%) and headache (7.4%). In conclusion, the two-drug combination of ruzasvir 180 mg plus uprifosbuvir 450 mg for 12 weeks was highly effective and well tolerated in participants infected with HCV GT1, GT2, GT4, GT5 and GT6, with a lower efficacy in GT3-infected persons.

16. Clinical decision support improves quality of care in patients with ulcerative colitis

Abstract

BACKGROUND: Decision support tools may facilitate shared decision-making and improve quality of care. AIM: To assess the effectiveness of a decision support tool on improving quality of care in ulcerative colitis. METHODS: A prospective quality of care intervention was conducted at two Australian hospitals comparing out-patient-based ulcerative colitis care with, and without, a tablet-based decision support tool. This included questions on disease activity management; psychological well-being; and preventive care, with 13 process indicators relevant to each domain. Participants included adult out-patients with mild-to-moderate ulcerative colitis and their clinicians who were divided into two cohorts. The first cohort were followed up immediately after their clinical review to check whether their clinician had discussed the 13 process indicators during the consultation. The second cohort of patients used the decision support tool immediately prior to their consultation which then generated a suggested management plan for the patient and clinician to discuss during the consultation. Management between the 2 cohorts was compared to assess the effectiveness of the decision support tool in improving the primary outcome, defined as the proportion of quality process indicators used for ulcerative colitis care, with and without the decision support tool. RESULTS: Thirteen physicians and 100 patients participated. Fifty patients were managed without the decision support tool using standard care (median age 40; 44% male), and 50 patients used the decision support tool (median age 40; 46% male) over a 20-week period. Increase in the median use of process indicators overall was observed following use of the decision support tool (27% vs 100%; P < 0.001). Abstract

The N7-methylguanosine cap is a hallmark of the 5' end of eukaryotic mRNAs and is required for gene expression. Loss of the cap was believed to lead irreversibly to decay. However, nearly a decade ago, it was discovered that mammalian cells contain enzymes in the cytoplasm that are capable of restoring caps onto uncapped RNAs. In this review, we summarize recent advances in our understanding of cytoplasmic RNA recapping and discuss the biochemistry of this process and its impact on regulating and diversifying the transcriptome. Although most studies focus on mammalian RNA recapping, we also highlight new observations for recapping in disparate eukaryotic organisms, with the trypanosome recapping system appearing to be a fascinating example of convergent evolution. We conclude with emerging insights into the biological significance of RNA recapping and prospects for the future of this evolving area of study. Abstract

The RASopathies are a group of genetic syndromes caused by germline mutations in genes that encode components of the Ras/mitogen-activated protein kinase (MAPK) pathway. Some of these syndromes are neurofibromatosis type 1, Noonan syndrome, Costello syndrome, cardio-facio-cutaneous syndrome, LEOPARD syndrome and Legius syndrome. Their common underlying pathogenetic mechanism brings about significant overlap in phenotypic features and includes craniofacial dysmorphology, cardiac, cutaneous, musculoskeletal, GI and ocular abnormalities, and a predisposition to cancer. The proceedings from the symposium “Genetic Syndromes of the Ras/MAPK Pathway: From Bedside to Bench and Back” chronicle the timely and typical research symposium which brought together clinicians, basic scientists, physician-scientists, advocate leaders, trainees, students and individuals with Ras syndromes and their families. The goals, to discuss basic science and clinical issues, to set forth a solid framework for future research, to direct translational applications towards therapy and to set forth best practices for individuals with RASopathies was successfully meet with a commitment to begin to move towards clinical trials.

Abstract

Ras, a signal-transducing protein involved in mediating growth factor-stimulated proliferation, is mutationally activated in over 30% of human tumors. To be functional Ras must bind to the inner surface of the plasma membrane, with post-translational lipid modifications being necessary for this localization. The essential, first modification of Ras is farnesylation catalyzed by the enzyme farnesyl: proteintransferase (FPTase). Inhibitors of FPTase (FTIs) are currently being tested to determine if they are capable of tumor growth inhibition. Here we describe our efforts, along with those of other groups, in testing the biological and biochemical effects of FTIs.

Abstract

AIMS: This report describes the baseline characteristics of patients in the Reduction of Events with Darbepoetin alfa in Heart Failure trial (RED-HF) which is testing the hypothesis that anaemia correction with darbepoetin alfa will reduce the composite endpoint of death from any cause or hospital admission for worsening heart failure, and improve other outcomes. METHODS AND RESULTS: Key demographic, clinical, and laboratory findings, along with baseline treatment, are reported and compared with those of patients in other recent clinical trials in heart failure. Compared with other recent trials, RED-HF enrolled more elderly [mean age 70 (SD 11.4) years], female (41%), and black (9%) patients. RED-HF patients more often had diabetes (46%) and renal impairment (72% had an estimated glomerular filtration rate < 60 mL/min/1.73 m2). Patients in RED-HF had heart failure of longer duration [5.3 (5.4) years], worse NYHA class (35% II, 63% III, and 2% IV), and more signs of congestion. Mean EF was 30% (6.8%). RED-HF patients were well treated at randomization, and pharmacological therapy at baseline was broadly similar to that of other recent trials, taking account of study-specific inclusion/exclusion criteria. Median (interquartile range) haemoglobin at baseline was 112 (106-117) g/L. CONCLUSION: The anaemic patients enrolled in RED-HF were older, moderately to markedly symptomatic, and had extensive co-morbidity. Fil: McMurray, John J. V.. Abstract

Ibutilide fumarate, racemic N-[4-[4-(ethyl-n-heptylamino)-1-hydroxybutyl]phenyl]methanesulfonamide hemifumarate, and artilide, the R-(+)-enantiomer of N-[4-[4-(di-n-butylalmino)-1-hydroxybutyl]-phenyl]methanesulfonamide hemifumerate, are under clinical investigation as Class III antiarrhythmic agents. For conducting drug disposition studies, we synthesized carbon-14 labeled ibutilide, as well as its two enantiomeric forms. In addition, high specific activity tritium labeled ibutilide was also prepared to facilitate development of a radioimmunoassay and for studying receptor site characteristics of this agent. Results of metabolism studies with [14C]ibutilide led us to prepare tritium labeled artilide, which is more readily accessible than the C-14 labeled drug. The optical antipode of artilide was also labeled with tritium for comparative drug disposition investigations on the two enantiomers.

Abstract

Farnesyl-Protein transferase catalyses a post-translational modification of Ras that is obligatory for the cell transforming activity of this oncogene protein. The screening of natural products to identify inhibitors of this enzyme as a potential anticancer agents, has led to the isolation of two novel dicarboxylic acids, named chaetomellic acids from Chaetomella acutiseta, as potent and selective inhibitors which appear to be the first examples of nonphosphorous containing FPP mimics.

35. Semivariograms from a forest transect gap model compared with remotely sensed data

Abstract

A spatially linked version of a forest gap model, ZELIG, parameterized for the H. J. Andrews Experimental Forest, Oregon, was used to generate structural properties (i.e. biomass, leaf area, and maximum tree height) of young (80 yr), mature (140 yr), and old-growth (450 yr) Pseudotsuga menziesii (Douglas fir) forests. Semivariograms were pro- duced at 10 and 30 m resolution to describe the spatio-tempo- ral patterns of variation of the simulated structural features along a 5 km transect of contiguous 10 m x 10 m grid cells. These semivariograms from the simulations were compared with semivariograms from matrices of pixel digital values obtained from aerial videography of similarly aged stands. Although autocorrelative spatial patterning was absent from both the remotely sensed imagery (except at < 20 m for the 450 yr stand) and the model output, the pixel-to-pixel and plot-to- plot variances exhibited similar patterns across the chronosequence at both resolutions. This suggests that gap models are able to capture temporal aspects of landscape dynamics associated with canopy texture of Pacific Northwest forests.

Abstract

Recent evidence has accumulated showing that activation of PLC-catalysed hydrolysis of phosphatidylcholine (PC-PLC) is a critical step in mitogenic signal transduction both in fibroblasts and in oocytes from Xenopus laevis. The products of ras genes activate PC-PLC, bind guanine nucleotides, have intrinsic GTPase activity, and are regulated by a GTPase-activating protein (GAP). It has been suggested that, in addition to its regulatory properties, GAP may also be necessary for ras function as a downstream effector molecule. In this study, evidence is presented that strongly suggests that the functional interaction between ras p21 and GAP is sufficient and necessary for activation of maturation promoting factor (MPF) H1-kinase activity in oocytes, and that PC hydrolysis is critically involved in this mechanism. Therefore, we identify GAP as a further step required for signalling through PC-PLC, and necessary for the control of oocyte maturation in response to ras p21/insulin but not to progesterone.

39. Fire decreases near-surface hydraulic conductivity and macropore flow in blanket peat

Abstract

Many peatlands have been subjected to wildfire or prescribed burning, but it is not known how these fires influence near-surface hydrological processes. Macropores are important flowpaths in the upper layers of blanket peat and were investigated through the use of tension disc infiltrometers, which also provide data on saturated hydraulic conductivity. Measurements were performed on unburnt peat (U), where prescribed burning had taken place 2years (B2), 4years (B4) and >15 (B15+) years prior to sampling, and where a wildfire (W) had taken place 4months prior to sampling. Where there had been recent burning (B2, B4 and W), saturated hydraulic conductivity was approximately three times lower than where there was no burning (U) or where burning was last conducted >15years ago (B15+). Similarly, the contribution of macropore flow to overall infiltration was significantly lower (between 12% and 25% less) in the recently burnt treatments compared to B15+ and U. There were no significant differences in saturated hydraulic conductivity or macropore flow between peat that had been subject to recent wildfire (W) and those that had undergone recent prescribed burning (B2 and B4). The results suggest that fire influences the near-surface hydrological functioning of peatlands but that recovery in terms of saturated hydraulic conductivity and macropore flow may be possible within two decades if there are no further fires.
