Abstract

New 2-mercapto-quinazolin-4-one analogs were synthesized and tested for their in vitro anticancer activity, dihydrofolate reductase (DHFR) inhibition, and epidermal growth factor tyrosine kinase (EGFR-TK) inhibition activities. Abstract

Acylation of the amino group of (I) and subsequent cyclization on N-3 afford the annulated imidazooxa/thiadiazolones (III) and (V) while reaction of (I) with malononitrile (VI) in the presence of a calculated amount of triethylamine yields the oxa/thiadiazolopyrimidines (VII).
