Glycosylation is a ubiquitous cellular or microenvironment-specific post-translational modification that occurs on the surface of normal cells and tumor cells. Tumor cell-associated glycosylation is involved in hematogenous metastasis. A wide variety of tumors undergo aberrant glycosylation to interact with platelets. As platelets have many opportunities to engage circulating tumor cells, they represent an important avenue into understanding the role glycosylation plays in tumor metastasis. Platelet involvement in tumor metastasis is evidenced by observations that platelets protect tumor cells from damaging shear forces and immune system attack, aid metastasis through the endothelium at specific sites, and facilitate tumor survival and colonization. During platelet-tumor-cell interactions, many opportunities for glycan- ligand binding emerge. This review integrates the latest information about glycans, their ligands, and how they mediate platelet-tumor interactions. We also discuss adaptive changes that tumors undergo upon glycan-lectin binding and the impact glycans have on targeted therapeutic strategies for treating tumors in clinical settings. Despite proven effectiveness in reducing motorist crashes involving wildlife, systematic inclusion of wildlife-mitigation measures during highway planning and projects continues to vary by jurisdiction in the United States and Canada. Some agencies invest significant resources to reduce wildlife–vehicle collisions (WVCs), while others invest few or no resources. Various factors have been identified as relevant in explaining why some jurisdictions are more proactive than others in addressing WVCs. This analysis builds on existing literature by comparing select legal decisions in the United States and Canada regarding public authority liability for failures to remediate known WVC hot spots, with the goal of assessing the potential role of liability in creating incentives for government decision makers to systematically address motorist crashes involving animals. Recess is an integral part of a comprehensive school physical activity program. Immediately prior to the pandemic, state legislation doubled the amount of required recess, creating an opportunity to better understand how policies and practices influence recess. To describe changes to recess due to COVID-19 and extended recess legislation and to quantify student physical activity during recess following these changes. This mixed-method study was conducted in Spring 2021 in four elementary schools. Recess physical activity was measured using waist-worn accelerometers, recess was observed, and researchers conducted interviews with teachers and principals. Females spent 41.7% (95%CI: 38.2, 44.5) and males spent 47.9% (95%CI: 44.6, 51.2) of recess in accelerometer-measured moderate-to-vigorous physical activity (n = 164). Participants (n = 13) discussed increased supervision, classroom separation on the playground, and benefits of extended recess. States should consider extending recess requirements but need to consider options for scheduling and indoor recess. Recess continues to be a key physical activity opportunity during school. Health Educators considering changes to recess policies and practices may want to consider scheduling of recess including class integration, options for indoor recess, and professional development for recess supervision to help maximize the benefits of recess. Friction stir spot welding (FSSW) is a variation of linear Friction Stir Welding (FSW), which was invented to compete with resistance spot welding (RSW) and riveting of lightweight alloys in the automobile, shipbuilding and aerospace industries. Recently, the application of FSSW has rapidly extended to a variety of metals and nonmetals. This article provides a comprehensive review of the recent progress on the process fundamentals, parameters optimization, microstructural evolution and mechanical properties, and relevant simulation and modeling of FSSW. The article also evaluates the energy generation, temperature distribution, plastic flow and joining mechanisms. The optimizations of tool design and welding parameters are obtained through experiments and modeling. Furthermore, a particular emphasis is given to microstructural characterization of the recovery, recrystallization and grain growth, and related annealing phenomena after in the welded alloys. The mechanisms of defect formation and liquidation cracking are discussed in detail. The mechanical properties, including hardness, static strength, fatigue performance and failure mechanisms and the relationship between mechanical properties and microstructures are also addressed along with residual stress and corrosion behavior. Protein glycosylation influences characteristics such as folding, stability, protein interactions, and solubility. Therefore, glycan moieties of therapeutic proteins and proteins that are likely associated with disease pathogenesis should be analyzed in-depth, including glycan heterogeneity and modification sites. Recent advances in analytical methods and instrumentation have enabled comprehensive characterization of highly complex glycosylated proteins. The following aspects should be considered when analyzing glycosylated proteins: sample preparation, chromatographic separation, mass spectrometry (MS) and fragmentation methods, and bioinformatics, such as software solutions for data analyses. Notably, analysis of glycoproteins with heavily sialylated glycans or multiple glycosylation sites requires special considerations. Here, we discuss recent methodological advances in MS that provide detailed characterization of heterogeneous glycoproteins. As characterization of complex glycosylated proteins is still analytically challenging, the function or pathophysiological significance of these proteins is not fully understood. To reproducibly produce desired forms of therapeutic glycoproteins or to fully elucidate disease-specific patterns of protein glycosylation, a highly reproducible and robust analytical platform(s) should be established. In addition to advances in MS instrumentation, optimization of analytical and bioinformatics methods and utilization of glycoprotein/glycopeptide standards is desirable. Introduction: The review provides a comprehensive overview about applicability of serological detection of autoantibodies against aberrant glycans as cancer biomarkers. Areas covered: Clinical usefulness of autoantibodies as cancer biomarkers is discussed for seven types of cancers with sensitivity and specificity of such biomarkers provided. Moreover, an option of using serological antibodies against a non-natural form of sialic acid – N-glycolylneuraminic acid (Neu5Gc), which is taken into our bodies together with red meat, as a potential cancer biomarker is discussed shortly as well. Expert opinion: In the final part of the review, we discuss what measures need to be applied for selective implementation of autoantibody assays into a clinical practice. Moreover, we discuss key challenges ahead for reliable and robust detection of autoantibodies against aberrant glycans as biomarkers for disease diagnostics and for stratification of cancer patients. Objectives: The aim of our study was to investigate molecular mechanisms of lithium action by studying the gene expression profile of peripheral cell models generated from bipolar patients (BD) and healthy controls (HC). Methods: EBV-immortalised lymphoblastoid cells (LCLs) and fibroblast cells from BD and HC were incubated with either lithium chloride or plain medium for 3 weeks. We first conducted a microarray gene expression study. The most promising differentially regulated genes in terms of lithium-associated or disorder-associated pathways were then replicated by quantitative real-time PCR (qRT-PCR). Results: The pooled microarray analysis showed 459 genes to be differentially regulated in BD compared to HC and 58 due to lithium treatment in LCLs, and 295 genes to be differentially regulated in BD compared to HC and five due to lithium treatment in fibroblasts. After correction for multiple comparison, EPHB1 disorder × treatment interactions remained significant in LCLs validated by qRT-PCR. In the control group, lithium influenced the expression of ANP32E, PLEKHA2, KCNK1, PRKCH, ST3GAL6 and AIF1. In bipolar and control fibroblast cells lithium treatment decreased FGF9 expression. Conclusions: The differentially regulated genes in our study add evidence for the relevance of inflammation, neuronal/glial development, phosphatidylinositol second-messenger pathway and ion channels in the mode of action of lithium. Friction stir welding (FSW), a highly efficient solid-state joining technique, has been termed as “green” technology due to its energy efficiency and environment friendliness. It is an enabling technology for joining metallic materials, in particular lightweight high-strength aluminum and magnesium alloys which were classified as unweldable by traditional fusion welding. It is thus considered to be the most significant development in the area of material joining over the past two decades. Friction stir processing (FSP) was later developed based on the basic principles of FSW. FSP has been proven to be an effective and versatile metal-working technique for modifying and fabricating metallic materials. FSW/FSP of aluminum alloys has prompted considerable scientific and technological interest since it has a potential for revolutionizing the manufacturing process in the aerospace, defense, marine, automotive, and railway industries. To promote widespread applications of FSW/FSP technology and ensure the structural integrity, safety and durability of the FSW/FSP components, it is essential to optimize the process parameters, and to evaluate thoroughly the microstructural changes and mechanical properties of the welded/processed samples. This review article is thus aimed at summarizing recent advances in the microstructural evolution and mechanical properties of FSW/FSP aluminum alloys. Particular attention is paid to recrystallization mechanism, grain boundary characteristics, phase transformation, texture evolution, characteristic microstructures, and the effect of these factors on the hardness, tensile and fatigue properties as well as superplastic behavior of FSW/FSP aluminum alloys. The literature indicates that glucose deprivation, dehydration, decreased sleep quality and quantity, and mood changes, independently and adversely can influence cognitive functions and therefore learning. The Ramadan fast is an annual religious act undertaken by Muslims where individuals refrain from consuming food and fluid during daylight hours, daily over 30 days. Alteration in eating times and sleep–wake cycles lead to a disruption in the body's daily circadian rhythm resulting in sleep perturbations and mood swings. Consequently, Ramadan fasting is linked to daytime lower glucose concentration, dehydration, decreased sleep quality and quantity, mood swings and elevated subjective feelings of fatigue and malaise. This review highlights how Ramadan fasting potentially poses learning difficulties as a result of the independent and/or combined effects of these factors. [ABSTRACT FROM PUBLISHER]

The perspectives of individuals with intellectual disabilities (ID) about family relationships are underrepresented in the literature. The topic of family relationships emerged in a grounded theory exploratory focus group study that involved thirty dually diagnosed participants with moderate or mild intellectual disabilities and histories of challenging behaviors. Because of the dearth of existing information and the salience of the topic, this analysis explored properties of the participant's disclosures associated with family relationships. The aims were to offer treatment providers empirically based information that may inform service provision and increase the availability of ID-specific, psychological supports for dually diagnosed individuals. Participants reported different types and statuses of family relationships. Transactional processes described in positive family relationships included properties such as reciprocity, flexibility, accommodation, trusting, and expressing affection. Conversely, participants described transactional relationship barriers (e.g., victimizing, behavioral dyscontrol, and substance abuse) that involved dysregulated behaviors of both the participants and family members in conflicted and severed family relationships. These factors appeared to lead to co-dysregulation versus co-regulation within the family relationships. These findings are relevant given the consensus in the literature that environmental factors are associated with challenging behaviors. Not only do treatment providers need to understand potential family relationship patterns to provide individual supports, but these multilayered factors may warrant seeking additional treatment modalities that address emotion regulation deficits of the participants and family members, trauma-informed treatment, and family therapy. Additionally, conceptualizing family relationships as transactional may help families and collateral supports co-construct positive, collaborative transactions with dually diagnosed individuals that improve the quality of life of all involved. Immunotherapy has been studied for many years in lung cancer without significant results, making the majority of oncologists quite skeptical about its possible application for non-small cell lung cancer treatment. However, the recent knowledge about immune escape and subsequent 'cancer immunoediting' has yielded the development of new strategies of cancer immunotherapy, heralding a new era of lung cancer treatment. Cancer vaccines, including both whole-cell and peptide vaccines have been tested both in early and advanced stages of non-small cell lung cancer. New immunomodulatory agents, including anti-CTLA4, anti-PD1/PDL1 monoclonal antibodies, have been investigated as monotherapy in metastatic lung cancer. To date, these treatments have shown impressive results of efficacy and tolerability in early clinical trials, leading to testing in several large, randomized Phase III trials. In 1981 the National Bureau of Standards, now the National Institute of Standards and Technology (NIST), issued the first natural matrix Standard Reference Material (SRM) for the determination of polycyclic aromatic compounds (PACs), SRM 1580 Organics in Shale Oil. In the next 10 years, additional natural matrix SRMs were developed including air and diesel particulate matter, petroleum crude oil, coal tar, sediment, and mussel tissue. The SRMs represented the "first generation" of natural matrix SRMs for the determination of PACs. The SRMs had "certified" values for only 5 to 12 PACs; however, their development established the foundation for the implementation of the "two or more independent analytical techniques" approach for certification of individual PACs in environmental matrices. The requirement for use of different analytical techniques spurred the development of both gas chromatography (GC) and liquid chromatography (LC) approaches for the determination of PACs. Since the mid-1990s, the "second generation" of natural matrix SRMs has been issued by NIST with certified values for over 20 PACs in each material. The greater number of certified values in these SRMs was the result of the combination of measurements using reversed-phase LC with fluorescence detection, multidimensional LC, and GC with mass spectrometric detection using two or more stationary phases with different selectivity for polycyclic aromatic hydrocarbon (PAH) separations. This article discusses the significant developments in analytical methods and chromatographic separation of PAHs during the past 20 years that have resulted in the certification of over 25 SRMs, which are now used worldwide for the validation of analytical methods for the measurement of PACs in environmental matrices. Using phenanthrene as a model analyte, we show that extraction from aqueous solution is possible via adsorption onto the surface of a poly(tetrafluoroethylene)-coated magnetic stir bar. Desorption from the stir-bar via ultrasonic agitation in acetonitrile solution allows quantitation of the analyte in solution. The technique is presented as a simple strategy, using commonly available laboratory apparatus, for the extraction of analytes from solution based on a surface adsorption processes: stir-bar adsorptive extraction (SBAE). In this work, a comparison between Soxhlet, ultrasonic, and microwave-assisted extraction (MAE) (using hexane:acetone 1:1) of polycyclic aromatic hydrocarbons (PAHs) from SRM 1650 diesel particulate matter was made. As good recoveries were not obtained for the higher molecular weight compounds, MAE with toluene and dichloromethane was also tested. Achieved recoveries were compared with the results obtained in the same assays made with SRM 1648 urban particulate matter and real atmospheric particulate samples. In order to explain the difficulty of the extraction of PAHs from SRM 1650 in comparison to SRM 1648 and real atmospheric particulate samples, elemental analysis, X-ray diffraction, and particle analysis using scanning electron microscopy coupled to an automatic computer imaging system were also made. It is important to notice that SRM 1650 shows a high percentage of carbon and differences in the morphology of the particles between SRM 1650 and SRM 1648 were observed. [ABSTRACT FROM AUTHOR]

Concentrations of selected particle-borne parent, alkylated, oxygenated, and nitrated polycyclic aromatic compounds (PACs) have been monitored before, during, and after a large national-scale wood-burning pollution episode (5 November 1994 and 4-5 November 1995). Toluene-modified supercritical CO 2 was used for the extraction of PACs from the collected particulate matter. Analysis was performed using an on-line multidimensional chromatographic technique using coupled liquid and gas chromatography (LC-GC) with loop interface. Comparison of average urban and wood smoke particulate PAC profiles identified a marked change in the ambient PAC profile for the pollution episode period, caused by the change in emission source from predominantly vehicular to wood smoke. A chemical mass balance model has been used to calculate the relative contributions from general urban and wood smoke PAC sources and from them, an estimation of the atmospheric life-time of the PAC emissions has been made. When monitored with 24 hr resolution in 1994, the concentrations of quinone-PAC oxidation products were found to peak 1 day after the maximum concentrations in the parent PAH were recorded, consistent with photochemically driven reactions with singlet state molecular oxygen. When concentrations were monitored on a more frequent basis in 1995, the concentrations of oxygenated species were seen to begin increasing in concentration before sunrise but still peaking after the maximum concentrations of parent PAH were recorded. A study was conducted to investigate the effects of heat stress on time-sharing performance. Twelve participants performed three dual-task scenarios and a multiple-task scenario for 2 h in each of six climates. The climates were obtained by generating each of three wet bulb globe temperatures (WBGT; 22, 28 and 34°C) with two relative humidity levels (30 and 70%). The dual tasks selected from the Criterion Task Set (CTS) were: (1) display monitoring with mathematical processing; (2) memory search with mathematical processing; and (3) unstable tracking with memory search. The multiple task scenario was generated using the SYNTASK software. The results indicated a significant heat stress effect on CTS display monitoring and unstable tracking performance and on the SYNTASK visual monitoring and auditory discrimination tasks. Additionally, at 34°C WBGT, 70% relative humidity was more detrimental to performance than 30% relative humidity. Results were interpreted using the Maximal Adaptability Model and Shingledecker's information processing stage/resource framework. To describe the results in an orderly manner, the authors propose the concept of heat stress selectivity effects. In addition, the results were used to evaluate whether the most recent NIOSH recommended heat stress standard, which is based solely on physiological and medical criteria, protects time-sharing performance. Radioisotope tracer techniques were used to study the process of in-situ chelation / supercritical fluid extraction (SFE) of La3+ and Lu3+ from a solid matrix using mixed ligand hexafluoroacetylacetone (HFA) and tributylphosphate (TBP) as chelating agents. A lab-built SFE extractor was used in this study and the extractor design was optimized based on the experimental results. Quantitative recovery of La and Lu was achieved when the extractor design was optimized. Extraction of uranium from real world samples was also investigated to demonstrate the feasibility of this chelation / SFE technology for environmental remediation applications. The extraction of Cu2+, Pb2+, Cd2+ and Zn2+ utilising supercritical fluid carbon dioxide containing dissolved organophosphorus reagents is shown to be feasible. Using the solubility parameter concept and investigation of a variety of physical parameters favourable extraction conditions of 60 °C and 400 atm pressure were determined. Cyanex 302 was found to be the most favourable ligand in terms of stability and ability to complex a range of metal ions. A soil containing substantial amounts of Pb2+ and Cd2+ was studied using SFE and the technique was found to reduce the levels of leachable metal ions to near US EPA regulatory levels. The wet bulb globe temperature (WBGT) is the de facto standard to assess environmental contributions to heatstress. A practical problem emerges when the heat stress conditions vary over many locations or during the day. To address this problem, investigators have suggested empirical relationships and thermodynamic models. The purpose of this effort was to examine a thermodynamic model in the laboratory and to predict WBGTs in an aluminum smelter by both the empirical and thermodynamic models. In the laboratory, there was no real difference between the experimental data and the thermodynamic model. In the application to an aluminum smelter, there was a small overall tendency for the predicted values to be greater than the actual values, but there was no practical difference between the models. The empirical model provided a good match with a slight over-prediction by 0.5 C with a standard deviation of 3.0 C. For the same data, the thermodynamic model had an average over-prediction of 0.7 C with a standard deviation of 2.8 C. Either method of predicting WBGT was effective.
