Background: Concurrent Exercise Training (CET) is defined as an exercise training mode in which aerobic and resistance exercises are performed in the same session. Objectives: The effects of CET in type 2 diabetes mellitus (T2DM) are inconclusive. Therefore, the objective of the present systematic review with a meta-analysis is to evaluate the effects of CET on metabolic markers, body composition, lipid profile, inflammation, and cardiorespiratory fitness (CRF) in people with T2DM. Methods: Five electronic databases were searched from inception up to January 2023. The eligibility criteria included prospective studies that had T2DM participants aged between 35 and 65 years who performed CET at least 2 times/week for a minimum duration of 8 weeks with a control group. Results: Seven studies were identified for qualitative review. Sufficient data were not available for one study, therefore, meta-analysis was performed in six studies. A significant effect was observed in glucose level (p <.01); glycated hemoglobin (p =.02); Homeostatic Model Assessment for Insulin Resistance (p =.02); body mass index (p =.02); %body fat (p <.01); %fat-free mass (p <.01); waist circumference (p =.03); triglyceride (p <.01); peak oxygen uptake (p <.01); and tumor necrotic factor -α (p <.01). Conclusions: The CET is effective in improving metabolic markers, body composition, lipid profile, inflammation, and CRF in people with T2DM and can be included in the management of T2DM. Lipids participate in many important biological functions through energy storage, membrane structure stabilization, signal transduction, and molecular recognition. Previous studies have shown that patients with esophageal squamous cell carcinoma (ESCC) have abnormal lipid metabolism. However, studies characterizing lipid metabolism in ESCC patients through lipidomics are limited. Plasma lipid profiles of 65 ESCC patients and 42 healthy controls (HC) were characterized by lipidomics-based ultraperformance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Single-factor and multi-factor statistical analysis were used to screen the differences in blood lipids between groups, and combined with component ratio analysis and receiver operating characteristic (ROC) curve diagnostic efficiency assessment, to reveal the potential mechanisms and biomarkers of ESCC. There were significant differences in lipid profiles between the ESCC and HC groups. Thirty-six differential lipids (11 up-regulated and 25 down-regulated) were selected based on the criteria of p <.05 and fold change > 1.3 or < 0.77. Glycerophospholipids were the major differential lipids, suggesting that these lipid metabolic pathways exhibit a significant imbalance that may contribute to the development of esophageal squamous cell carcinoma. Among them, the seven candidate biomarkers for esophageal squamous cell carcinoma with the highest diagnostic value are three phosphatidylserine (PS), three fatty acids (FA) and one phosphatidylcholine (PC). Few studies have reported the cardiovascular health effects of different high-intensity interval training (HIIT) protocols among sedentary young women. We investigated the impact of a traditional HIIT programme and a high-intensity circuit training (HICT) programme on lipid profiles and inflammatory cytokine levels in sedentary young women. Forty-two women were randomly assigned to HICT (body weight-based training), HIIT (cycling-based training), or control groups (n = 14 each). HICT and HIIT participants completed an 8-week training programme of three sessions per week. Total cholesterol (TC), triglyceride, high- and low-density lipoprotein, leptin, resistin, tumour necrosis factor-alpha (TNF-α), interleukin-8, and interferon-gamma levels were measured before and after the intervention. Post-intervention, TC and leptin were decreased in the HICT group. The HICT group also demonstrated increased lean mass, upper and lower limb strength, and balance, while the HIIT group displayed improved lower limb strength. Additionally, the control group showed significant increases in triglyceride levels, weight, body mass index, and fat mass. In conclusion, although both HICT and HIIT interventions showed improvements in cardiovascular health and physical fitness, participants in the HICT group experienced more health benefits. The detection of specific markers of dementia and mild cognitive decline (MCI) could be the key to disease prevention and forehanded treatment. Female gender is one of the major risk factor for dementia. The aim of our study was to compare serum concentration of some factors related to lipid metabolism and the immune system in patients with MCI and dementia. The study was performed on women >65 years old: controls (n = 75), diagnosed with dementia (n = 73) and MCI (n = 142). Patients were evaluated using Mini-Mental State Examination, Clock Drawing Test and Montreal Cognitive Assessment scales in the period 2020–2021. The level of Apo A1 and HDL was significantly decreased in patients with dementia; the level of Apo A1 was also decreased in MCI. EGF, eotaxin-1, GRO-α, and IP-10 were elevated in patients with dementia compared to the controls. IL-8, MIP-1β, sCD40L, and TNF-α levels were decreased in MCI patients and increased in patients with dementia compared to the control. Serum VEGF levels were decreased in MCI and dementia patients in comparison with the control. We hypothesize that no single marker can indicate a neurodegenerative process. High concentrations of matrix metalloproteinase-2 (MMP-2) and −9 (MMP-9) are associated with inflammation early after stroke. In chronic stages, the elevation of some cytokines is related to the presence of co-morbid conditions in these individuals. In addition to this, some characteristics such as sensorimotor impairment, atrophy, and sedentary lifestyle predispose the system to an inflammatory response. To quantify MMP-2 and −9 serum activity in chronic post-stroke individuals and correlate it with variables of physical activity level, body composition, functional and walking capacity, and with inflammatory biomarkers. Additionally, gelatinase activity was characterized according to motor impairment. Fourteen patients with stroke onset >6 months and seven healthy individuals were enrolled in this study. The clinical assessment included: body composition, measure by bioelectrical impedance analyzer; Fugl-Meyer Motor Assessment Scale; six-minute and ten-meter walk tests, and physical activity level assessed by the StepWatch® Activity Monitor. Blood samples were collected from antecubital vein and serum MMP-2 and −9 activity was analyzed using gelatin Zymography, and the TNFα, IL-6, IL-1β, IL-10 biomarkers using ELISA kits. Chronic post-stroke individuals presented an increased activity of MMP-2 and −9 compared to healthy individuals. Positive correlations with time and steps in low cadence and negative ones with medium cadence and peak activity index were observed. According to the motor impairment, the MMP-2 activity was increased in the mild-moderate group compared to the control group. The associations between components of metabolic syndrome (MetS), insulin resistance (IR), and several malignancies have been reported. However, the possible link between IR and dermal malignancies, including non-melanoma skin cancers (NMSCs), has not been investigated to date. In this study, we aimed to examine the possible association between components of MetS, IR, adhesion molecules, and NMSC for the first time. This was a case-control study comprising 73 confirmed cases of NMSC and 72 unrelated healthy controls. Anthropometric and biochemical assessments including fasting blood lipid profile, glucose and insulin assays were performed. To evaluate IR, HOMA-IR formula was used. Though fasting serum glucose showed no significant between-group difference, serum concentrations of insulin (p = 0.048) as well as HOMA-IR (p = 0.037) were both significantly higher in NMSC group than in controls. Logistic regression analysis revealed significant associations between waist circumference (OR: 1.04, 95% CI: 1.007-1.080, p = 0.018), percent of visceral fat (OR: 1.10, 95% CI: 1.024-1.190, p = 0.01), HOMA-IR (OR: 1.169, 95% CI: 1.004-1.360, p = 0.044), circulating VCAM-1 concentrations (OR: 1.005, 95% CI: 1.003-1.007, p < 0.001) and NMSC risk. Interestingly, the occurrence of MetS was significantly higher in subjects with NMSC than in healthy controls (p = 0.038). MetS and its components were associated with increased NMSC risk. Household air pollution is a leading risk factor for morbidity and premature mortality. Numerous cookstoves have been developed to reduce household air pollution, but it is unclear whether such cookstoves meaningfully improve health. In a controlled exposure study with a crossover design, we assessed the effect of pollution emitted from multiple cookstoves on acute differences in blood lipids and inflammatory biomarkers. Participants (n = 48) were assigned to treatment sequences of exposure to air pollution emitted from five cookstoves and a filtered-air control. Blood lipids and inflammatory biomarkers were measured before and 0, 3, and 24 hours after treatments. Many of the measured outcomes had inconsistent results. However, compared to control, intercellular adhesion molecule-1 was higher 3 hours after all treatments, and C-reactive protein and serum amyloid-A were higher 24 hours after the highest treatment. Cancer-related fatigue (CRF) is a common side effect impacting breast cancer survivors. Research points to a relationship between obesity and CRF in breast cancer survivors related to elevated systemic inflammation and metabolic alterations. This cross-sectional study examined the relationship of obesity to CRF, inflammatory markers and serum lipids through a secondary analysis of a nationwide randomized controlled trial. Breast cancer survivors with CRF were categorized based on BMI category. Symptoms of CRF, inflammatory markers and serum fatty acids were assessed among groups. There were 105 breast cancer survivors in the analysis. BMI was positively associated with CRF based on MFSI General (p = 0.020; 95% C.I. 0.024, 0.273) and MFSI Physical (p = 0.013; 95% C.I. 0.035, 0.298) subscales. TNF-α (p = 0.007; 95% C.I. 0.007, 0.044), and IL-6 (p = 0.020; 95% C.I. 0.006, 0.073) were elevated in the obese. Monounsaturated fatty acid levels (p = 0.047; 95% C.I. 0.000, 0.053) and the omega-6 to omega-3 fatty acid ratio were associated with obesity (p = 0.047; 95% C.I. 0.002, 0.322). Obese breast cancer survivors had greater levels of CRF, inflammatory markers and certain fatty acids. We investigated the effects of a recreational training (RET) protocol in elderly women with type-2 diabetes mellitus (T2DM). We hypothesized that non-conventional physical activities of RET protocol might improve clinical outcomes regarding cardiovascular function, metabolic profile and mental health as participants keep the adherence to the protocol during the 3-month follow-up. Cardiovascular parameters (heart rate, systolic and diastolic blood pressure), circulating biomarkers (glucose and lipids) and salivary cortisol were attenuated in response to exercise. Introduction:Laurus nobilis is known in the field of herbal medicine and in vitro studies that it has beneficial effects such as antibacterial, antifungal, antidiabetes, and anti-inflammatory properties. Objective: We investigated whether L. nobilis tea consumption affects the plasma levels of lipid biomarkers in healthy volunteers. Methods: Thirty healthy Tunisian volunteers aged between 20 and 57 years old consumed L. nobilis infusion, prepared from 5 g of dried L. nobilis leaves in 100 ml boiled water, once a day during 10 days. Plasma concentrations of serum low-density lipoprotein (LDL) cholesterol, triglycerides and HDL (high-density lipoprotein) cholesterol were measured by Beckman Coulter D × 600 analyzer before L. nobilis consumption and at the end of the experiment. Results:L. nobilis tea consumption significantly increased the concentration of HDL cholesterol ([HDL cholesterol] D0 = 1.34 ± 0.25 pg/mL, D11=1.42 ± 0.29, p = 0.01). However, a slight decrease that was statistically non-significant in LDL cholesterol and triglycerides levels was observed (p < 0.05). Conclusions: These findings highlight the improving blood lipidic profiles, which means a possible positive effect on reducing the risk of cardiovascular disease of L. nobilis tea consumption in healthy volunteers. The antioxidant and hepatoprotective properties of the n-hexane fraction obtained by successive liquid fractionation of the crude cold ethanol extract (95%) of the dried powdered Ficus lutea leaves were evaluated by saponification and GC/MS analysis with column chromatography using silica gel. In vitro antioxidant effect was assessed by DPPH free radical technique and in vivo evaluation carried out on rats after carbon tetrachloride (CCl4) induction. The evaluation was done by determination of oxidative stress markers and liver function enzymes and examining histopathological picture of liver. Triterpenoidal and steroidal compounds lupenone (lupane skeleton): methyl isomasticadienonate (8-tirucallene skeleton), α-amyrin acetate, 20, 22- Epoxyeupha-24-ene-3-one, and stigmasterol were isolated and their structure elucidations were confirmed by spectral analysis MS, IR, and H1-NMR spectra. Purpose: To evaluate markers of cardiovascular disease in women with normal insulin sensitivity who had recently initiated use of depot medroxyprogesterone acetate (DMPA) as a contraceptive.Materials and Methods: A prospective, non-randomised, comparative study. Data of 30 women of 18-40 years of age, with normal hyperinsulinaemic-euglycaemic clamp at baseline and body mass index (BMI) < 30, who opted to use DMPA or a copper intrauterine device (IUD) as a contraceptive, were analysed. Serum samples were collected for evaluation of the lipid and hepatic profile, C-reactive protein, TNF-α, interleukin-6, leptin/adiponectin ratio and free fatty acids; body composition was evaluated using DXA, blood pressure and carotid intima-media thickness were measured. Evaluations were performed at baseline and 12 months later. The groups were compared using repeated measures analysis of variance (ANOVA). Significance level was 5%.Results: The mean age of the women in the DMPA and IUD group was 28.7 ± 6.5 and 28.3 ± 5.8 years, respectively; the BMI was 23.0 ± 3.4 and 24.3 ± 2.7 kg/m2, respectively, in the same groups. At 12 months, triglyceride levels were higher in the DMPA group compared to the IUD group and there was an increase in apolipoprotein B-100 in relation to baseline in DMPA group; there were no other difference between the groups.Conclusions: No clinical cardiovascular effect was found but deterioration in CV markers was observed in the first year of use of DMPA. Purpose of the study: There was inconsistent evidence about the benefit of vitamin D plus evening primrose oil (EPO) supplement intake on lipid profiles and reduced oxidative stress among women with polycystic ovary syndrome (PCOS). The current study was performed to evaluate the effects of vitamin D plus EPO supplementation on lipid profiles and biomarkers of oxidative stress in vitamin D-deficient women with PCOS.Materials and methods: This randomized, double-blind, placebo-controlled trial was performed among 60 vitamin D-deficient women with PCOS. Participants were randomly assigned into two groups to receive either 1000 IU vitamin D3 plus 1000 mg EPO (n= 30) or placebo (n= 30) for 12 weeks. Metabolic profiles were quantified at baseline and after the 12-week intervention.Results: Compared with the placebo group, women in vitamin D and EPO co-supplementation group had significant increases in serum 25-hydroxyvitamin D (25(OH)D) (+10.7 ± 8.4 vs. −0.5 ± 1.6 ng/mL,p< 0.001) and plasma total glutathione (GSH) (+62.7 ± 58.0 vs. −0.7 ± 122.7 µmol/L,p= 0.01), while there were significant decreases in triglycerides (−7.3 ± 23.8 vs. +6.9 ± 26.3 mg/dL,p= 0.03), very low-density lipoprotein (VLDL) cholesterol levels (−1.5 ± 4.7 vs. +1.4 ± 5.3 mg/dL,p= 0.03), total/high-density lipoprotein cholesterol ratio (−0.3 ± 0.4 vs. −0.02 ± 0.4,p= 0.02), and malondialdehyde (MDA) concentration (−0.4 ± 0.4 vs. +0.5 ± 1.8 µmol/L,p= 0.008).Conclusion: Overall, vitamin D and EPO co-supplementation for 12 weeks among vitamin D-deficient women with PCOS significantly improved triglycerides, VLDL cholesterol, GSH, and MDA levels. Objectives: Fatty acid profiles and desaturase (SCD-16, SCD018, D5D, D6D) and elongase (ELOVL6) enzyme activity have been associated with adiposity and metabolic disease. While this has been studied in adults, few studies have included children. The objective of this study was to evaluate these markers in children and identify relationships with markers of metabolic health. It was hypothesized that these lipid markers would be correlated to adiposity and metabolic disease.Methods: This study was a cross-sectional analysis of fourth- and fifth-grade children (n = 86, aged 9-12) participating in a comprehensive nutrition program. Any student enrolled in the program was eligible for inclusion in this study. Fasting plasma was collected and analyzed for total fatty acids, glucose, insulin, and full lipid panels. Insulin resistance was estimated using calculated homeostatic model assessment for insulin resistance (HOMA-IR) values.Results: There were no differences in lipid markers, glucose, insulin, or HOMA-IR among children classified as normal weight, overweight, or obese. SCD-16, D5D, and ELOVL6 activity was significantly correlated to HOMA-IR values (r = 0.39, p = 0.001; r = -0.33, p = 0.006; r = -0.37, p = 0.005, respectively). In regression analysis, body mass index for age percentile, D6D activity, ELOVL6 activity, and systolic blood pressure were the most significant predictors of HOMA-IR values (adjusted r2 = 0.39, p ≤ 0.001).Conclusions: There was no relationship between these lipid markers and adiposity in this population; however, there were correlations with HOMA-IR. Objectives: Cardiometabolic index (CMI) has been recognized as a novel and practical marker for the assessment of cardiometabolic risk as it is independently related to diabetes and atherosclerotic progression. This study tested the hypothesis that CMI represents a risk of ischemic stroke in a general population of rural China.Methods: From July 2012 to August 2013, we examined data from a large cross-sectional study of 11,345 participants (mean age 53.8 years; 60.8% females) who underwent biochemical determinations and anthropometric measurements in rural areas of northeast China. Ischemic stroke was documented as a history of cerebrovascular events and verified by medical record review.Results: The prevalence of ischemic stroke was given to 3.1% of females and 3.2% of males. The cardio-metabolic profile was notably more adverse in ischemic stroke groups, irrespective of gender. A dose-response manner was detected for the prevalence of ischemic stroke, exhibiting a significant increase from the lowest to the highest quartiles of CMI (1.2% to 6.4% in females, P for trend<0.001; 2.3% to 4.3% in males, P for trend = 0.017). In multivariable analysis, for every 1 SD increment in CMI, the probability of ischemic stroke increased by 18% in females and 14% in males, respectively. The odds ratios for ischemic stroke comparing the top versus bottom quartiles of CMI were 2.047 (95%CI: 1.168-3.587) for females and 1.722 (95%CI: 1.019-2.910) for males. According to the area under receiver operating characteristic (AUC), the discrimination power of CMI in predicting ischemic stroke was relatively higher for females (AUC: 0.685) than males (AUC: 0.573).Conclusion: The strong and independent association of CMI with ischemic stroke in females, in comparison with the much lesser degree in males, provides further insight to better stratify by sex in investigations of ischemic stroke and solidly corroborates the potential role of ischemic stroke prevention targeted at CMI. Objectives:Despite progress in identifying molecular pathophysiological processes in schizophrenia, valid biomarkers are lacking for both the disease and treatment response. Methods:This comprehensive review summarises recent efforts to identify molecular mechanisms on the level of protein and gene expression and epigenetics, including DNA methylation, histone modifications and micro RNA expression. Furthermore, it summarises recent findings of alterations in lipid mediators and highlights inflammatory processes. The potential that this research will identify biomarkers of schizophrenia is discussed. Results:Recent studies have not identified clear biomarkers for schizophrenia. Although several molecular pathways have emerged as potential candidates for future research, a complete understanding of these metabolic pathways is required to reveal better treatment modalities for this disabling condition. Conclusions:Large longitudinal cohort studies are essential that pair a thorough phenotypic and clinical evaluation for example with gene expression and proteome analysis in blood at multiple time points. This approach might identify biomarkers that allow patients to be stratified according to treatment response and ideally also allow treatment response to be predicted. Improved knowledge of molecular pathways and epigenetic mechanisms, including their potential association with environmental influences, will facilitate the discovery of biomarkers that could ultimately be effective tools in clinical practice. Sources of lipid biomarkers in two Lake Bled sediment cores deposited under oxic (Zaka Bay) and anoxic (WB) conditions were determined using stable carbon isotopes. Lipid biomarkers were of autochthonous origin mainly from planktonic and microbial sources. δ13C values for long-chain (LC) n-alkanes, short-chain (SC) and LC n-alcohols, SC n-fatty acids (FAs) were similar for both sediments, showing that identification of their sources was not related to different depositional regimes. A larger contribution of more refractory terrestrial organic matter (OM) was observed in δ13C values obtained from SC n-FAs in both environments and from SC n-alkanes in Zaka Bay. In contrast, the δ13C values observed in LC n-FAs, and especially in sterols, were lower in the WB than in Zaka Bay, indicating the pronounced contribution of an anaerobic, bacterial origin. Overall, the data indicated that the main source of lipids was autochthonous in both sediment environments including, LC n-alkanes, n-alcohols, n-FAs and C29 sterols previously assigned to an allochthonous higher plant source. Objectives:The phospholipid vesicle-based permeation assay (PVPA) is a recently establishedin vitro stratum corneummodel to estimate the permeability of intact and healthy skin. The aim here was to further evolve this model to mimic thestratum corneumin a compromised skin barrier by reducing the barrier functions in a controlled manner.Methods:To mimic compromised skin barriers, PVPA barriers were prepared with explicitly defined reduced barrier function and compared with literature data from both human and animal skin with compromised barrier properties. Caffeine, diclofenac sodium, chloramphenicol and the hydrophilic marker calcein were tested to compare the PVPA models with established models.Results and discussions:The established PVPA models mimicking thestratum corneumin healthy skin showed good correlation with biological barriers by ranking drugs similar to those ranked by the pig ear skin model and were comparable to literature data on permeation through healthy human skin. The PVPA models provided reproducible and consistent results with a distinction between the barriers mimicking compromised and healthy skin. The trends in increasing drug permeation with an increasing degree of compromised barriers for the model drugs were similar to the literature data from otherin vivoandin vitromodels.Conclusions:The PVPA models have the potential to provide permeation predictions when investigating drugs or cosmeceuticals intended for various compromised skin conditions and can thus possibly reduce the time and cost of testing as well as the use of animal testing in the early development of drug candidates, drugs and cosmeceuticals. Purpose: To investigate the expression of lipid peroxidation markers in the tear film and ocular surface and their correlation with disease severity in patients with dry eye disease. Methods: The concentrations of hexanoyl-lysine (HEL), 4-hydroxy-2-nonenal (HNE), and malondialdehyde (MDA) were measured with enzyme-linked immunosorbent assays in tears obtained from 44 patients with non-Sjogren syndrome dry eye and 33 control subjects. The correlations between the marker levels and the tear film and ocular surface parameters, including tear film break-up time (BUT), Schirmer tear value, tear clearance rate, keratoepitheliopathy scores, corneal sensitivity, conjunctival goblet cell density, and symptom score, were analyzed. The expression of the lipid peroxidation markers HEL, 4-HNE, and MDA in the conjunctiva was evaluated using immunohistochemistry. Results: The concentrations of HEL, 4-HNE, and MDA were 279.84 ± 69.98 nmol/L, 0.02 ± 0.01 μg/mL, and 3.80 ± 1.05 pmol/mg in control subjects and 283.21 ± 89.67 nmol/L (p= 0.97), 0.20 ± 0.03 μg/mL (p< 0.01), and 13.32 ± 4.03 pmol/mg (p< 0.01) in dry eye patients. 4-HNE and MDA levels significantly correlated with BUT, Schirmer tear value, tear clearance rate, keratoepitheliopathy scores, conjunctival goblet cell density, and symptom score (p< 0.05), whereas HEL levels did not correlate with these parameters. Staining intensities for 4-HNE and MDA increased in dry eye patients. Conclusions: The expression of late lipid peroxidation markers, 4-HNE and MDA, increases in the tear film and ocular surface of patients with dry eye. Objective: Nutritional strategies can be effective for the prevention of menopause-related diseases, such as osteoporosis and cardiovascular disease. Our aim was to evaluate the effects of a dairy product enriched in polyunsaturated fatty acids, calcium, oleic acid, and vitamins on cardiovascular markers and bone metabolism in postmenopausal women with moderate cardiovascular risk.Methods: One hundred seventeen healthy postmenopausal women (aged 45 ± 7.7 years) were allocated to 2 groups: the intervention group (IG; n = 63), who consumed 0.5 L/day of a low-lactose skimmed milk enriched with 40 mg/100 mL of eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA), 0.54 g/100 mL oleic acid, and vitamins, and the control group (CG; n = 54), who consumed 0.5 L/day of semiskimmed milk 0.5 L/day enriched with vitamins A and D.Results: After 12 months, in the IG there was an improvement in lipid profile: a -5.78% decrease in total cholesterol (p = 0.010), -9.79% (p = 0.004) in low-density lipoprotein (LDL) cholesterol, -9.56% (p < 0.001) in total cholesterol (TC)/high-density lipoprotein (HDL) ratio, and -3.38% in LDL/HDL ratio (p < 0.001). No changes were observed in the CG. In the IG we observed a decrease of -28.20% in high-sensitivity C-reactive protein (hs-CRP; p = 0.012). There was no effect on bone turnover markers or serum osteoprotegerin (OPG) in either of the study groups. In the IG, receptor activator of nuclear factor κB ligand (RANKL) was reduced -17.64% (p = 0.003), with no effect in the CG.Conclusion: In postmenopausal women with moderate cardiovascular risk, dietary supplementation with a dairy drink enriched with fatty acids (EPA+DHA), oleic acid, minerals, and vitamins induces a positive effect on cardiovascular risk and parameters of bone metabolism. In order to investigate the potential application of blood biomarkers as surrogate indicators of carcinogen–adduct formation in target-specific tissues, temporal formation of benzo[a]pyrene (BaP)-associated DNA adducts, protein adducts, or lipid damage in target tissues such as lung, liver, and kidney was compared with globin adduct formation or plasma lipid damage in blood after continuous intraperitoneal (ip) injection of [3H]BaP into female ICR mice for 7 d. Following treatment with [3H]BaP, formation of [3H]BaP–DNA or –protein adducts in lung, liver, and kidney increased linearly, and persisted thereafter. This finding was similar to the observed effects on globin adduct formation and plasma lipid damage in blood. The lungs contained a higher level of DNA adducts than liver or kidneys during the treatment period. Further, the rate of cumulative adduct formation in lung was markedly greater than that in liver. Treatment with a single dose of [3H]BaP indicated that BaP–globin adduct formation and BaP–lipid damage in blood reached a peak 48 h after treatment. Overall, globin adduct formation and lipid damage in blood were significantly correlated with DNA adduct formation in the target tissues. These data suggest that peripheral blood biomarkers, such as BaP–globin adduct formation or BaP–lipid damage, may be useful for prediction of target tissue-specific DNA adduct formation, and for risk assessment after exposure. Work ability is based on the balance between personal resources and work demand. This study focused on the personal resources component of work ability. The aims of this study were to elucidate the association between work ability and cardiovascular (CV) risk factors, particularly oxidative stress, and to estimate the effect of a community-implemented lifestyle modification programme on work ability and CV risk factors. Urinary 8-iso-prostaglandin F2α(PGF2α), a biomarker of oxidative stress, was negatively correlated with psychological resources, as measured by the Work Ability Index (WAI). Overall WAI score was unchanged following the programme, while CV risk factors and antioxidative activity improved. A reduction in PGF2αlevels was correlated with an improvement in subjective work ability relative to job demands, as assessed by a WAI item. Taken together, the results suggest that lifestyle modification programmes enhance the personal resources component of work ability and are associated with a reduction in oxidative stress. Practitioner Summary:We demonstrated the capacity of a lifestyle modification programme to augment personal resources by reducing CV risk factors such as oxidative stress. In addition, since the community-implemented lifestyle modification programme minimally affected work ability, ‘worksite’ health promotion programmes might more effectively enhance personal resources. The marine planktonic archaea are dominated by Thaumarchaeotal Marine Group I, which is characterized by the lipid biomarker thaumarchaeol. The marine benthic archaea are characterized by greater diversity of currently unknown species whose lipid biomarker signatures are uncertain. In this study, a sediment core from the northwestern part of the South China Sea (SCS) (water depth 1474 m) was analyzed using molecular DNA and lipid biomarker approaches. While 16S rRNA gene analysis showed changing archaeal community structures with sediment depth, this change had little impact on the fossil record of archaeal lipids that are characteristic of the planktonic community. As a result, the fossil archaeal lipids recorded paleo sea surface temperature of the SCS since the last glacial maxima by the TEX86proxy, which agreed generally with the winter temperature recorded by planktonic foraminiferas collected from the same area of the SCS that hosted mass-transported deposits. This suggests that this deep water deposit may have partially preserved paleoclimate record reflecting seasonal temperature variation in a near shore setting, which is in contrast to annual sea surface temperature or sub sea surface temperature variation recorded by TEX86in the open ocean. The most recent guidelines released by the EAS/ESC and the Canadian Cardiovascular Society (CCS) retain low-density lipoprotein cholesterol (LDL-C) as the primary measure of the atherogenic risk of the apolipoprotein B (apoB) lipoproteins and the primary target of LDL-C lowering therapy. Both organizations endorse non-high-density lipoprotein cholesterol (non-HDL-C) and apoB as 'alternate/secondary' targets, but neither group offers evidence supporting the continued preference of LDL-C as the primary target over non-HDL-C and apoB. Further, both suggest that non-HDL-C and apoB more or less measure the same thing and, therefore, are essentially interchangeable. But what is the evidence that LDL-C should remain the primary target, and are apoB and non-HDL-C mirror images of one another? Furthermore, are estimation of risk and establishment of treatment targets the only relevant issues, or is diagnosis also an essential objective? These are the questions this article will address. Our principal objectives are: (1) to clarify the differences between LDL-C, non-HDL-C, and apoB and to distinguish what they measure; (2) to summarize the evidence relating to LDL-C, non-HDL-C, and apoB as predictors of cardiovascular risk and as targets for treatment; and (3) to demonstrate that diagnosis of atherogenic dyslipoproteinemias should be a fundamental clinical priority. The fatty acid, hydrocarbon and sterol composition were used to investigate the organic sources and compositional changes in particulate organic matter (POM) and trap material in the water column in the deepest part of Lake Bled (NW Slovenia) in October 2006. Fatty acids distribution of algal origin were abundant in particles and trap material in epilimnion, while in anoxic zone POM and trap material were enriched in bacterial fatty acids (e.g. 16:1n-7, 18:1n-7, 18:1n-9,iso-C14-C16andanteiso-C15). The lowest δ13C value of −51.7‰ was observed in 18:1n-7FA in trap material and was the only FA which could be linked to methanotrophic bacteria. In addition Zooplankton left a marked imprint on particulate lipids and trap material at 12 m by predominance ofn-C18:0overn-C16:0FA, short-chain, even-carbonn-alkenols, the high proportion of cholest-5-en-3β-ol (44.8% of total sterol concentration (TST)), cholesterol/phytosterol ratio of 0.49 and δ15NPNvalues of 6.8 and 11.7‰. It was shown that lacustrine phytoplankton biosynthesize 24-ethylcholest-5-en-3β-ol which is often used as a marker of terrigenous organic matter. Phytoplankton represented an important source of cholest-5-en-3β-ol and 24-methylcholest-5,22(E)-dien-3β-ol, while 24-ethylcholesta-5,22E-dien-3β-ol and 24-methylcholest-5-en-3β-ol were of terrestrial origin. Objective: To investigate whether lipid and protein oxidation products are elevated and correlated with routine clinical markers of hepatic and renal function in patients anesthetized with halothane, isoflurane, or sevoflurane. Methods: Urine and blood samples were collected from patient groups. Excretion of aldehydes, acetone, and o,o'-dityrosine was measured before and after anesthesia. Blood samples were analysed for clinical markers. Results: Urinary concentrations of aldehydes, acetone, o,o'-dityrosine and glucose were significantly increased after anesthesia in halothane and sevoflurane groups earlier than clinical markers. Significant correlations were found in sevoflurane group. Conclusion: Lipid and protein oxidation contributes to subclinical sevoflurane nephrotoxicity. Aortic stenosis is the most prevalent valve pathology and calcific aortic valve disease (CAVD) is its most frequent etiology in developed countries. There is extensive evidence that CAVD represents an active disease process similar to that of atherosclerosis with similar classical cardiovascular risk factors and pathological mechanisms. Given that in the vast majority of situations the only treatment available is valve replacement there is a need to develop pharmacological therapies that retard the disease progression. Lipid-lowering therapies have been the focus of research, however, so far with negative results. Future studies, including animal models, shall provide an opportunity to further evaluate the disease mechanisms of CAVD and to discover potential disease biomarkers and pharmacological interventions that can reduce the need for valve replacement. The sources of organic matter in three typical mangrove systems of Cochin estuary were assessed for the first time with help of sediment bulk parameters and fatty acid biomarkers. The C/N and δ13C analysis showed intermediate values to that of autochthonous and terrestrial inputs of organic matter, signaling to a mixed origin. The biochemical composition of sedimentary organic matter revealed a dominance of lipids over proteins and carbohydrates, indicating the nutritive or energetic value as well as the freshness of organic matter in all the systems. Fatty acids ranging from C8 to C24 were identified in the study. C20:5n-3 was one of the major fatty acids at station 1, indicating significant benthic phytoplankton contribution. The zooplankton biomarkers C20:1 and C24:1 were also present in appreciable levels, suggesting the tidal influx. Principal fatty acids at station 2 were the zooplankton biomarkers C20:1 and C24:1 implying significant estuarine contribution through tidal influx. The major fatty acids at station 3 were diatom marker C20:5n-3 and the higher plant marker C24:0. Dominant fatty acids in mangrove leaves; C18:2n-6 and C18:3n-3 were also present at station 3. The low contributions of C20:1 and C24:1 confirms the limited tidal exchange at this station. Presence of significant amount polyunsaturated fatty acids in the mangrove sediments confirms the freshness of organic matter. Thus, fatty acid biomarker study of the three mangrove systems of Cochin estuary could substantiate the bulk parameter approaches effectively; distinguishing the multiple sources of the organic matter and the observed spatial variations. In the present study, the main goal was to compare the effects of ionic copper versus copper nanoparticles in Enchytraeus albidus assessing the effect at the biomarker level, testing different concentrations and exposure times. Measured parameters were lipid peroxidation (LPO), total, reduced and oxidized glutathione content (TG, GSH and GSSG), the enzymatic activity of superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), glutathione peroxidase (GPx), glutathione S-transferases (GSTs) and cholinesterases (ChEs). Results showed that both salt- and nano-copper caused oxidative stress and damage to E. albidus, as confirmed by LPO levels, and effects could be discriminated between the copper forms. Nevertheless and despite the visible discrimination between nano and the salt form (time and exposure dependent), there was no single or a set of biomarkers that provided the best discrimination. Diet has been shown to modulate M1dG adduct, a biomarker of oxidative stress and lipid peroxidation. Thus, we analysed the association between diet and M1dG in 120 controls and 67 Map Ta Phut industrial estate workers in Rayong, Thailand, to evaluate the influence of fruit and vegetables, and fried and charcoal-grilled/barbecued food consumption on M1dG. M1dG was decreased in controls reporting to consume 14-17 servings/week of fruit and vegetables (mean ratio [MR]== 0.35, CI 0.18-0.69, p< 0.05). Conversely, a non-statistically significant M1dG increment was detected in controls consuming 9-18 servings/week of fried food (MR == 1.33, CI 0.88-2.00, p == 0.168). No effect of charcoal-grilled/barbecued food was found. No effect of diet was observed in workers. An association with smoking was observed in controls (MR == 1.88, CI 1.14-3.10, p < 0.05), but not in workers. The mineral, organic and elemental composition of medicine clays from three shrines in the Tong Hills in northern Ghana (Gbankil, Kusanaab, and Yaane) are assessed to ascertain what additives they might contain and the implications for their recognition, for example in archaeological contexts. These are clays that are widely used for healing purposes being perceived efficacious in curing multiple ailments and which are given a divine provenance, but their collection is ascribed human agency. The Yaane clay is also supplied as part of the process of obtaining the right to operate the shrine elsewhere making it widely dispersed. Organic geochemical analyses revealed a predominance of plant-derived material with a substantial contribution of microbial origin. Based on these (supported by elemental and mineral analyses), no unnatural organic material could be detected, making an exogenous contribution to these clays unlikely. The implications are that these are wholly natural medicinal substances with no anthropogenic input into their preparation, as the traditions suggest. The very similar mineralogy of all the clays, including a non-medicine clay sampled, suggests that, unless the geology radically differed, differentiating between them analytically in an archaeological contexts would be doubtful. Over the years, there has been concern about the changes taking place in heated oils and the effects on individuals consuming them. The present study investigated the effects of a diet containing thermally oxidized soybean oil (TO) or TO supplemented with probiotic Bifidobacteria (TO++Pro) on the serum lipid profile and oxidative stress biomarkers of male rats. The data showed several indicators of oil deterioration after thermal processing, including high levels of % free fatty acid (FFA; 15-fold), acid value (AV; 14-fold), peroxide value (8-fold), p-anisidine value (AnV; 39-fold), total oxidation value (TOTOX; 19-fold), thiobarbituric acid--reactive substances (TBARS) value (8.5-fold), and trans-FA (TFA) isomers (2.5-fold) compared to the control. The rats that were fed a diet containing TO showed a significant ( p < 0.05) decrease in body weight gain, food efficiency values, and liver weight. Furthermore, the total cholesterol and low-density lipoprotein (LDL) levels were increased, while the high-density lipoprotein (HDL) level was decreased in blood serum samples. High levels of TBARS, superoxide dismutase (SOD), and glutathione reductase (GR) activities were also detected in the livers, kidneys, testes, and brains of rats. Interestingly, a diet containing TO++Pro restored all biological parameters to their control values. The present data suggested that Bifidobacteria may ameliorate the serum lipid profile and oxidative stress biomarkers that are generated in animals that are fed a TO diet. [ABSTRACT FROM AUTHOR]

The fatty acid profiles in total, neutral and polar lipids in the heart tissues of five freshwater fish species (Nile perch Lates niloticus, Nile tilapia Oreochromis niloticus, marbled lungfish Protopterus aethiopicus, Bagrus docmak and African catfish Clarias gariepinus) from Lakes Victoria and Kyoga were determined chemometrically by methanolysis of the lipid extracts and gas chromatography of the resulting methyl esters. The analytical data were treated using multivariate principal component analysis. In neutral and polar lipids palmitic acid (16:0) and stearic acid (18:0) were the dominant saturated fatty acids. The polyunsaturated fatty acids in the polar lipid fraction ranged from 33% to 56% and in the neutral lipid fraction from 23% to 30%, dominated by arachidonic acid (20:4n6) and docosahexaenoic acid (22:6n3). The amount of cholesterol in the neutral fraction was 57-89 mg g-1 lipid. The amount of plasmalogen was 12-27 mg g-1 and 14-37 mg g-1 lipid in the polar fraction and total lipid, respectively. In the principal component plots, the fish species from the two lakes were distinguished by fatty acid profiles in neutral, polar and total lipid from the heart tissue. However, polar lipid fractions were better markers of fish species and populations than the neutral fraction and total lipids. Heart tissues have low lipid levels dominated by polar lipids (phospholipids), with polyunsaturated fatty acids as the principal components. A new bis heterocycle comprising both bioactive 2-aminopyrimidine and thiazolidin-4-one nuclei namely 3-(4′′-(4″″-fluorophenyl)-6′′-phenylpyrimidin-2′′-yl)-2-phenylthiazolidin-4-one 3 was synthesized, characterized with the help of melting point, elemental analysis, FT-IR, MS, one-dimensional NMR (1H, 13C) spectra and we evaluated the chemopreventive potential of 3-(4′′-(4″″-fluorophenyl)-6′′-phenylpyrimidin-2′′-yl)-2-phenylthiazolidin-4-one based on in vivo inhibitory effects on 7,12-dimethylbenz[ a]anthracene (DMBA)-induced hamster buccal pouch carcinogenesis. Administration of 3 effectively suppressed oral carcinogenesis initiated with DMBA as revealed by the reduced incidence of neoplasms. Lipid peroxidation, glutathione (GSH) content, and the activities of glutathione peroxidase (GPx), glutathione S-transferase (GST) were used to biomonitor the chemopreventive potential of 3. Lipid peroxidation was found to be significantly decreased, whereas GSH, GPx, GST, and GGT were elevated in the oral mucosa of tumor-bearing animals. Cancer recurrence after radical surgery might happen even in the case of patients with localized prostate carcinoma treated by radical prostatectomy. Therefore, identifying predictive markers of tumour recurrence is very important, so this study evaluated the presence of lipid peroxidation product acrolein in primary prostate carcinomas, assuming that acrolein could be involved in prostate carcinogenesis as was recently shown for colon cancer. Samples obtained by radical prostatectomy of 70 patients were analysed, out of which 27 patients suffered afterwards from tumour recurrence, while 43 patients were disease free. Immunohistochemistry using genuine monoclonal antibodies against acrolein-protein adducts revealed the association of acrolein with progression of carcinoma. The logistic regression combining clinical parameters together with the biochemical markers of disease and acrolein immunohistochemistry has shown that the relapse might be predicted with 90% accuracy if tumour-positive surgical margins, stage of disease and the intensity of acrolein presence in tumour stroma were taken together. Sediments of upwelling regions off Namibia, Peru, and Chile contain dense populations of large nitrate-storing sulfide-oxidizing bacteria, Thiomargarita, Beggiatoa, and Thioploca. Increased contents of monounsaturated C16 and C18 fatty acids have been found at all stations studied, especially when a high density of sulfide oxidizers in the sediments was observed. The distribution of lipid biomarkers attributed to sulfate reducers (10MeC16:0 fatty acid, ai-C15:0 fatty acid, and mono-O-alkyl glycerol ethers) compared to the distribution of sulfide oxidizers indicate a close association between these bacteria. As a consequence, the distributions of sulfate reducers in sediments of Namibia, Peru, and Chile are closely related to differences in the motility of the various sulfide oxidizers at the three study sites. Depth profiles of mono-O-alkyl glycerol ethers have been found to correlate best with the occurrence of large sulfide-oxidizing bacteria. This suggests a particularly close link between mono-O-alkyl glycerol ether-synthesizing sulfate reducers and sulfide oxidizers. Coprostanol (contribution characteristic from anthropogenic pollution) and other lipid biomarkers (sterols, fatty alcohols and fatty acids) were identified and quantified in recent sediment extracts from Rodrigo de Freitas Lagoon, a touristy spot of Rio de Janeiro- Brazil, using gas chromatography with mass selective detector (GC-MSD). The determination of lipid biomarkers profile indicates an autochthonous biogenic contribution due to the presence of phytoplankton, zooplankton, bacteria and dinoflagellates. The allochthonous biogenic contribution was confirmed by detection of biomarkers from higher plants in the sediments due to the influence of the Atlantic Forest inserted in the studied region. The concentration of the studied compounds varied from 5.53 to 216.47 μ g.g-1 for sterols, 0.47 to 5.35 μ g.g-1 for fatty alcohols, 20.15 to 66.22 μ g.g-1 for fatty acids and 0.08 to 3.98 μ g.g-1 for coprostanol. Estrogens exert beneficial effects on the vascular system, while progestogens generally have a negative impact (e.g. vasoconstrictor effects on the arterial system). In contrast, dydrogesterone appears to be largely neutral in terms of biochemical markers and indirect clinical endpoints, such as blood pressure, that act as surrogate markers for vascular function. Studies on lipid and carbohydrate metabolism, which can also influence vascular function, demonstrate that the addition of dydrogesterone intensifies rather than attenuates beneficial estrogenic effects. Dydrogesterone also has largely neutral effects on hemostasis. Since there are relatively few data available on clinical parameters such as blood flow measurements, especially in women with pre-existing cardiovascular diseases, increased risks cannot be excluded for a combination of estrogen replacement with dydrogesterone. Further studies should focus on this open question since dydrogesterone, with its largely neutral properties, might be a suitable option, including for older women already at increased cardiovascular risk. Microbial mats were collected from hot springs in California (Eagleville) and Nevada (Paradise Valley and Crescent Valley) to determine bacterial community structure and pathways of carbon cycling in different geothermal environments of the western United States. Phospholipid fatty acids (PLFA) at Eagleville contained even-numbered fatty acids, with 16:0 being the most abundant (48.8%), followed by 18:1ω 9c (17.2%), 16:1ω 7c/t (6.3%), and 18:0 (6.2%), which are consistent with lipid profiles of cyanobacteria or other phototrophic bacteria. The PLFA profiles at Paradise Valley and Crescent Valley were dominated by similar even-numbered fatty acids; however, branched fatty acids such as iso- and anteiso- 15:0 and 17:0 were also abundant (up to 7.1% compared to 2.0% at Eagleville), suggesting greater relative abundance of heterotrophic bacteria in these springs. Analysis of neutral lipids was only performed on Eagleville and Paradise Valley springs, which revealed abundant bacterial hopanoids including the 2-methylbacteriohopane-32,33,34,35-tetrol (2-methylBHT) that is specific to cyanobacteria; however, the diversity of hopanoid compounds was significantly lower at Eagleville than at Paradise Valley. The carbon-isotope composition of individual PLFA averaged -30.7 ± 1.3‰ (n = 7) at Eagleville, -28.0 ± 1.8‰ (n = 3) at Crescent Valley, and -29.7 ± 3.1‰ (n = 12) at Paradise Valley. Carbon isotope fractionation between PLFA and CO2 was only available for Eagleville (-11.7‰) and Paradise Valley (-21.7‰), which indicated the predominance of the Calvin cycle for CO2 fixation in these hot springs. Bacterial 16S rRNA genes were extracted from environmental samples at Eagleville and Paradise Valley but not Crescent Valley. Clone libraries indicated the predominance of cyanobacteria (50-75%) at these locations, which is consistent with the lipid profiles. Phylogenetic tree of the 16S rRNA genes indicated that most of the cyanobacterial sequences are unknown and may be specific to the Nevada and California hot springs. Phototrophic green non-sulfur bacteria were also present at Eagleville (13%) and Paradise Valley (7%). The remaining sequences were related to α-, β -, and γ -Proteobacteria, Acidobacteria, Deinococcus/Thermus, Bacteroidetes, and Spirochaetes. However, not all of these sequences were present at each of the springs. Results of this study demonstrate the consistency among lipid profiles (phenotypes), carbon isotopes (biogeochemistry), and 16S rRNA genes (genotypes) of the bacterial community in these hot springs, which cumulatively suggest the importance of cyanobacteria in primary production of biomass under the environmental conditions examined. Oxidized low density lipoproteins (oxLDLs) may exert several pro-inflammatory effects that can contribute to the development of coronary artery disease (CAD). Evaluating a possible correlation between oxLDLs and clinical expression of CAD, we measured specific lipid peroxidation indices in healthy subjects and in patients at different clinical stages of CAD. We observed a slight, but not significant, increase in plasma content of cholesterol oxidation products, i.e. oxysterols, in all CAD patients, and a slight, but not significant, increase of 4-hydroxynonenal-protein adducts only in subjects with acute CAD. Moreover, CAD patients showed a plasma rise of specific inflammatory proteins, i.e. C-reactive protein, intercellular adhesion molecule-1, and interleukin-8, but not of monocyte chemotactic protein-1. These preliminary data, without excluding an involvement of oxidative stress and inflammation in CAD, do not show a strict correlation between relevant plasma markers, other than C-reactive protein, and acute phase of the disease. Ethylene and ethane as biomarkers for lipid peroxidation in humans can be detected in the exhaled breath by means of very sensitive gas sensors based on high-resolution molecular spectroscopy. To this purpose, a laser photoacoustic spectroscopy sensor (PAS) for online ethylene monitoring and a tunable diode laser absorption spectroscopy sensor (TDLAS) for ethane detection were developed at ENEA Frascati Molecular Spectroscopy Laboratory. The sensors have been calibrated by certified mixtures. The main advantages of the breath analysis by spectroscopic technique are the high sensitivity and the absolutely non-invasive character. During an experimental study in cooperation with Umberto I Hospital (Radiology Institute) of Rome, we were able to detect very low concentrations (under 1&ThickSpace;ppb) of trace ethylene content in the air exhaled by patients, following X-ray therapy; results are reported in this article. Lipid-modified proteins are classified based on the identity of the attached lipid, a post- or co-translational modification required for their biological function. At least five different lipid modifications of cysteines, glycines and other residues on the COOH- and NH 2 -terminal domains have been described. Cysteine residues may be modified by the addition of a 16-carbon saturated fatty acyl group by a labile thioester bond (palmitoylation) or by prenylation processes that catalyze the formation of thioether bond with mevalonate derived isoprenoids, farnesol and geranylgeraniol. The NH 2 -terminal glycine residues may undergo a quite distinct process involving the formation of an amide bond with a 14-carbon saturated acyl group (myristoylation), while glycine residues in the COOH-terminal may be covalently attached with a cholesterol moiety by an ester bond. Finally, cell surface proteins can be anchored to the membrane through the addition of glycosylphosphatidylinositol moiety. Several lines of evidence suggest that lipid-modified proteins are directly involved in different steps of the development of lesions of atherosclerosis, from leukocyte recruitment to plaque rupture, and their expression or lipid modification are likely altered during atherogenesis. This review will briefly summarize the different enzymatic pathways of lipid modification and propose a series of lipid-modified proteins that can be used as biomarkers for cardiovascular disease. The neuroprotective effect of maltol on oxidative damage in the brain of mice challenged with kainic acid was examined. Male ICR mice, 6-8 weeks of age, were administered orally with maltol (50 or 100 mg/kg) for 5 consecutive days. Thirty minutes after the final administration, the animals were challenged s.c. with kainic acid (50 mg/kg), and neurobehavioral activities were monitored. In addition, biomarkers of oxidative stress and neuronal loss in hippocampus for the biochemical and morphological evaluations were analyzed 2 days after the kainic acid challenge. During 5-day treatment with maltol, the body weight gain was not significantly different from that of vehicle-treated control animals. Administration of kainic acid alone induced severe epileptiform seizures, causing a lethality of approximately 50%, and injuries of pyramidals cells in hippocampus of mice survived the challenge. Kainic acid exposure also resulted in marked decreases in total glutathione level and glutathione peroxidase activity, and an increase in thiobarbituric acid-reactive substances (TBARS) value in brain tissues. In comparison, coadministration with maltol (100 mg/kg) remarkably attenuated the neurobehavioral signs and neuronal loss in hippocampus, leading to a decrease in mortality of animals to 12.5% ( p <0.05), although maltol at a dose of 50 mg/kg failed to show any remarkable protection. In addition, the changes in glutathione and TBARS values and glutathione peroxidase activity induced by kainic acid were restored to control levels by pretreatment with maltol (100 mg/kg). Background: The incidence of obesity and related metabolic diseases is high and increasing in sub-Saharan African women. Evidence on the determinants of these diseases is limited, particularly in black South African women. Objective: This PhD review attempts to understand the determinants of obesity and metabolic syndrome (MetS) in a population of ageing urban-dwelling black South African women. Methods: Drawing on the longitudinal Birth-to-Twenty-Plus cohort, data were collected in 2002/03 and 2012/13, including information on behavioural factors (smoking, sitting time, physical activity, smokeless tobacco, and alcohol consumption), body-size perception, body composition (measures of adiposity and lean mass), blood pressure, and cardiometabolic biomarkers (lipid profile, fasting insulin, fasting blood glucose, insulin resistance, leptin, and adiponectin). Results: The prevalence of obesity and related cardiovascular disease risk was high and increased significantly over the 10 year period. Despite most of the study population being physically active, sitting time was high and associated with elevated blood pressure and hypertriglyceridaemia. Two groups of people were observed, those who were happy and those who were unhappy with their body size. In logistic regression analysis, the risk of MetS was lowered by abdominal subcutaneous adipose tissue and adiponectin, and increased by age, smoking, truncal lean mass, and insulin resistance. Conclusions: Obesity was confirmed to be increasing in black South African women, despite most women being sufficiently active according to guidelines of ≥150 min activity/week. Nevertheless, the contribution of sitting time to poor health outcomes is evident in this study population and must be addressed, particularly in women who are content with being obese. The novel finding of the effects of abdominal subcutaneous tissue and truncal lean mass with MetS requires further investigation. The protective effect of adiponectin against MetS is an important finding which highlights the novel interaction between adiposity and cardiometabolic diseases in black South African women.
