This study investigated the interactive effects of drought stress with biochar on plant growth, the physiological and biochemical attributes of soybean seedlings. A pot trial tested the effect of amending a loamy sandy soil with biochar at three doses: 0%, 3% and 6% in the alleviation of drought stress, imposed by adjusting soil moisture content at three levels (in terms of soil field capacity, FC): 100% FC, 75% FC and 50% FC. Water deficit negatively influenced the growth of soybean seedlings and chlorophyll content of leaves, particularly in non-amended plants whereas biochar amendment boosted the growth and chlorophyll content, particularly under drought stress. Drought stress elevated malondialdehyde (MDA), hydrogen peroxide (H2O2), proline, sucrose and abscisic acid (ABA) contents as well as the antioxidant enzyme activity [peroxidase (POD), catalase (CAT) and superoxide dismutase (SOD)] of soybean leaves but reduced the contents of indole acetic acid (IAA) and gibberellic acid (GA); biochar amendment counteracted the impact of drought. Previous research has explored the demands of amateur boxing-specific activity; however, no holistic review of the acute responses to such activity currently exists. This paper aimed to provide a systematic review of the available literature on the acute physiological, endocrine, biochemical, and performance responses to amateur boxing-specific activity. Following a search of EBSCOhost, SPORTDiscus, PubMed and Google Scholar databases, 25 studies were identified as meeting the inclusion criteria for the review. The methodological quality of the included studies was assessed via a modified Downs and Black checklist. Random-effects meta-analysis of standardised mean differences (SMD) revealed large (SMD = 4.62) increases in pre-post blood lactate (BLa), cortisol (SMD = 1.33), myoglobin (Mb) (SMD = 1.43), aspartate transaminase (AST) (SMD = 1.37), and alanine aminotransferase (ALT) (SMD = 0.97), in addition to moderate increases in creatine kinase (CK) (SMD = 0.65). Small pre–post increases in counter-movement jump (CMJ) height (SMD = 0.33) were observed. Consistently greater pre–post alterations were observed in competitive bouts, followed by sparring, and less so in boxing-specific simulations. Considerable physiological, endocrine, and biochemical responses are elicited following amateur boxing. Interestingly, neuromuscular and task-specific performance may not deteriorate following boxing-specific activity. The findings of the review may assist in the designing and periodising of boxing-specific training, dependent on the desired physical adaptations, training phase, and recovery status of the amateur boxer. Highlights Amateur boxing elicits a considerable acute physiological, hormonal, and biochemical response. Such responses are typically greater in competitive bouts, followed by sparring, and less so in simulated activity. The considerable demands of amateur boxing-specific activity do not appear to negatively affect neuromuscular or task-specific performance. Amateur boxers may be conditioned to preserve performance despite the acute demands of the sport, or the lack of performance decrement may reflect the short duration of amateur boxing. The aim of this study was to investigate the effects of bottled water left in the car and those left in the refrigerator on the physiological and biochemical parameters of male rats. Eighteen male albino rats were randomly divided into control, warmed bottled water (WBW), and cooled bottled water (CBW) groups. The concentrations of bisphenol A (BPA) and antimony (Sb) in the water samples were measured. Results showed that the levels of BPA and Sb were higher in bottled water left in the car and those kept at room temperature (control), relative to the bottled water samples kept in the fridge. Bodyweight, some metabolic indicators, cTnT and CRP levels, hematological parameters, testosterone level, and sperm quality were significantly affected by storage conditions. These results indicate that storage conditions can potentially affect the quality of bottled water, which in turn can affect the biological parameters of living organisms. Moreover, since the concentrations of BPA and Sb were least in bottled water kept in the refrigerator, we recommend that manufacturers and households should endeavor to keep bottled water under cold storage. Military personnel are required to complete physically demanding tasks when performing work and training, which may be quantified through the physical stress imposed (external load) or the resultant physiological strain (internal load). The aim of this narrative review is to provide an overview of the techniques used to monitor work and training load in military settings, summarise key findings, and discuss important practical, analytical, and conceptual considerations. Most investigations have focused upon measuring external and internal load in military training environments; however, limited data exist in operational settings. Accelerometry has been the primary tool used to estimate external load, with heart rate commonly used to quantify internal load. Supplemental to heart rate, psychophysiological and biochemical measures have also been investigated to elucidate aspects of internal load. Broadly, investigations have revealed that military training requires personnel to perform relatively large volumes of physical activity (e.g. averaging ∼15,000 steps·day−1) of typically low-moderate intensity activity (<6 MET), although considerable temporal and inter-individual variability is observed from these gross mean estimates. There are limitations associated with these measures and, at best, estimates of external and internal load can only be inferred. These limitations are particularly pertinent for military tasks such as load carriage and manual material handling, which often involve complex activities performed individually or in teams, in a range of operational environments, with multiple layers of protection, over a protracted duration. This study determined the extent of diversity and relationships among 74 Aloe germplasm collected mainly from the Indian arid zone by characterizing 18 morphological traits (8 quantitative and 10 qualitative). High coefficient of variation (CV) was observed in quantitative traits (aloin content [AC] [43.1%], leaf width [LW] [32.0], polysaccharide [30.7], and inflorescence length [18.5]) through CV. Range and mean values for important biochemical traits of Aloe vera were 0.009-10.94 and 5.77, respectively, for aloin and 0.16-0.61 and 0.40, respectively, for polysaccharide (mg 100 mL-1) content. Leaf rind thickness (LRT) showed the highest variability (0.8-1.8 mm), followed by leaf length (LL) (30-57.5 mm) and LW (99.2-17.3 mm). Pearson's correlation study revealed both positive and negative relationships among the quantitative traits of aloe genotypes ranging from −0.19 (AC and LL/LRT) to 0.48 (leaf thickness and LRT). Dendrogram was generated for the six quantitative traits by using Euclidean distance-based unweighted pair group method with arithmetic mean method of cluster analysis which formed four major clusters at coefficient value of 3.7 with two subclusters each. Cluster analysis clearly brought out two groups as non-bitter and bitter ones. The pattern obtained through cluster analysis confirmed by two-dimensional scatter plot of principal components analysis (PCA) accounted for 67.73% of cumulative variance. Despite limitation in estimating the total genetic variation, the present study indicated that morphological and biochemical traits were useful in preliminary evaluation and can be used as a general approach for assessing genetic diversity among morphologically distinguishable aloe genotypes. The constrained economic context leads laboratories to centralize their routine analyses on high-throughput platforms, to which blood collection tubes are sent from peripheral sampling sites that are sometimes distantly located. Providing biochemistry results as quickly as possible implies to consolidate the maximum number of tests on a minimum number of blood collection tubes, mainly serum tubes and/or tubes with anticoagulants. However, depending on the parameters and their pre-analytical conditions, the type of matrix - serum or plasma - may have a significant impact on results, which is often unknown or underestimated in clinical practice. Importantly, the matrix-related effects may be a limit to the consolidation of analyses on a single tube, and thus must be known by laboratory professionals. The purpose of the present critical review is to put forward the main differences between using serum and plasma samples on clinical biochemistry analyses, in order to sensitize laboratory managers to the need for standardization. To enrich the debate, we also provide an additional comparison of serum and plasma concentrations for approximately 30 biochemistry parameters. Properties, advantages, and disadvantages of serum and plasma are discussed from a pre-analytical standpoint - before, during, and after centrifugation - with an emphasis on the importance of temperature, delay, and transport conditions. Then, differences in results between these matrices are addressed for many classes of biochemistry markers, particularly proteins, enzymes, electrolytes, lipids, circulating nucleic acids, metabolomics markers, and therapeutic drugs. Finally, important key-points are proposed to help others choose the best sample matrix and guarantee quality of clinical biochemistry assays. Uranium is a radioactive heavy metal ubiquitous in the natural environment. In its chemical form, it is known to induce nephrotoxicity both in human and in animals. Its toxicity is dose and time dependent, also varies with form of uranium. In the present study, we assessed the nephrotoxicity induced by a single dose of uranyl nitrate (UN) in mice at different time intervals and recovery from its toxicity. Two doses of 2 and 4 mg/kg body weight of uranyl nitrate was injected intraperitoneally and animals were sacrificed after 1, 3, 5, 14, and 28 d of administration. Histopathological and biochemical alterations of post-UN dosing in comparison to control were evaluated. Tubular damage to about 75% was observed after 3 d (4 mg/kg) and the biochemical parameters such as serum creatinine, urea, and blood urea nitrogen levels were also significantly increased. Progression of tubular damage was not found after 5 d. Dose-dependent recovery of uranyl nitrate-treated animals was observed after 14 and 28 d of dosing. Objectives: The aim of this study was to assess monocyte Fc receptor (FcR) status and function in patients with active psoriatic arthritis (PsA) in relation to healthy controls (HC) and to disease activity.Method: The study population comprised 23 patients with active polyarticular PsA and 33 age- and gender-matched HC. Immunoglobulin (Ig) levels, inflammatory laboratory parameters, patient-reported outcomes of joint disease activity, skin scoring (Psoriasis Area and Severity Index, PASI), and joint status were determined in the patients. Monocytes were analysed for the expression of FcRs for IgG (FcγR) class I (CD64), IIa (CD32a), IIb (CD32b), and III (CD16), the FcR for IgA (FcαR) (CD89), and surface-bound IgG. The monocytic FcγR function was assessed by evaluating IgG immune complex (IC) binding and tumour necrosis factor (TNF)-α production following IgG-IC stimulation. The monocytes were further subdivided and analysed according to their CD14 and CD16 expression.Results: The PsA patients presented elevated serum levels of IgG1, 2, and 3 and increased numbers of CD64(+) monocytes. Furthermore, the PsA monocytes exhibited increased cell-bound IgG, and the FcγR function was affected in terms of reduced IgG-IC-mediated TNF-α release. These findings correlated significantly with different markers of joint disease activity. PsA was also accompanied by an increase in the CD16 low-expressing monocyte subset.Conclusions: An intensified humoral immune response affects monocytes and their FcR status in active polyarticular PsA. The up-regulated CD64(+) monocytes seem to be have an important role in psoriatic joint inflammation. These cells may prove to be a useful target in future PsA therapeutic interventions. A greenhouse study was conducted to evaluate effects of phosphorus (P) levels (0, 50 and 100 mg kg−1soil) under saline (0, 1000 and 2000 mg sodium chloride (NaCl) kg−1soil) conditions on growth and chemical composition of pistachio seedlings (Pistacia veraL.) cv. ‘Badami-zarand’ in a completely randomized design (CRD) with four replications. Results showed that salinity application decreased leaf, stem, and root dry weights, number of leaf, length of stem and leaf area, while this effect diminished with P fertilization. By increasing salinity levels, all of the nutrients concentration in leaf, stem and root except sodium (Na) content were reduced. P application increased P and potassium (K) concentrations in the leaves, stem and root, while decreased Na and Zinc (Zn) leaf, stem and root concentrations. However, the results indicated that proline accumulation and reducing sugar content were increased by salinity, P and their interaction application. The results suggest that fertilization of phosphorus can diminish some adverse effects of high salinity on growth and chemical composition of pistachio seedlings. [ABSTRACT FROM AUTHOR]

Comparative efficiency of three extraction solutions, including the universal sodium phosphate buffer (USPB), the Tris-HCl buffer (UTHB), and the specific buffers, were compared for assays of soluble protein, free proline, superoxide radical (), hydrogen peroxide (H2O2), and the antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), guaiacol peroxidase (POD), ascorbate peroxidase (APX), glutathione peroxidase (GPX), and glutathione reductase (GR) in Populus deltoide. Significant differences for protein extraction were detected via sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional electrophoresis (2-DE). Between the two universal extraction buffers, the USPB showed higher efficiency for extraction of soluble protein, CAT, GR, , GPX, SOD, and free proline, while the UTHB had higher efficiency for extraction of APX, POD, and H2O2. When compared with the specific buffers, the USPB showed higher extraction efficiency for measurement of soluble protein, CAT, GR, and , parallel extraction efficiency for GPX, SOD, free proline, and H2O2, and lower extraction efficiency for APX and POD, whereas the UTHB had higher extraction efficiency for measurement of POD and H2O2. Further comparisons proved that 100 mM USPB buffer showed the highest extraction efficiencies. The biochemical status in the saliva of 12 males before/after using mobile phone has been evaluated. Radio frequency signals of 1800 MHz (continuous wave transmission, 217 Hz modulate and Global System for Mobile Communications [GSM - non-DTX]) with 1.09 w/kg specific absorption rate (SAR) value were used for 15 and 30 min. Cell phone radiation induced a significant increase of superoxide dismutase (SOD); there was a statistically significant effect of talking time on the levels of SOD, F(2, 33) = 8.084, p < 0.05, ω = 0.53. The trend analysis suggests a significant quadratic trend, F(1, 33) = 4.891, p < 0.05; indicating that after 15 min of talking the levels of SOD increased, but as talking time increased the SOD activity started to drop. In contrast to this, there was no statistically significant effect of talking time on the level of salivary albumin, cytochrome c, catalase or uric acid. Results suggest that exposure to electromagnetic radiation may exert an oxidative stress on human cells as evidenced by the increase in the concentration of the superoxide radical anion released in the saliva of cell phone users. Ubiquitination, a fundamental post-translational modification (PTM) resulting in the covalent attachment of ubiquitin (Ub) to a target protein, is currently implicated in several key cellular processes. Although ubiquitination was initially associated with protein degradation, it is becoming increasingly evident that proteins labeled with polyUb chains of specific topology and length are activated in an ever-expanding repertoire of specific cellular processes. In addition to their involvement in the classical protein degradation pathways they are involved in DNA repair, kinase regulation and nuclear factor-κB (NF-κB) signaling. The sorting and processing of distinct Ub signals is mediated by small protein motifs, known as Ub-binding domains (UBDs), which are found in proteins that execute disparate biological functions. The involvement of UBDs in several biological pathways has been revealed by several studies which have highlighted the vital role of UBDs in cellular homeostasis. Importantly, functional impairment of UBDs in key regulatory pathways has been related to the development of pathophysiological conditions, including immune disorders and cancer. In this review, we present an up-to-date account of the crucial role of UBDs and their functions, with a special emphasis on their functional impairment in key biological pathways and the pathogenesis of several human diseases. Crude ethanol extracts (CEE) of purple rice was fractionated to obtain hexane soluble (HSF) and ethyl acetate soluble fractions (EASF). Total antioxidant capacity was higher in CEE than the HSF and EASF. However, HSF exhibited strong antiproliferation and apoptosis induction against colon cancer cell lines, both p53 wild-type (RKO) and mutant (SW620) strains. Then, the CEE was used to determine the effects on the progression of aberrant crypt foci (ACF), a preneoplastic lesion seen in colon carcinogenesis in rats. Male Wistar rats were subcutaneously injected of 40 mg/kg body weight dimethylhydrazin (DMH) once weekly for 2 wk. After 2 wk, rats were orally administered ethanol extract at 100 and 1000 mg/kg body weight, for 4 wk. Rats fed with only the high dose of CEE had significantly decreased numbers of ACF per rat (45.56%) and crypt multiplicity (AC/focus) (16.67%) compared to rats that received DMH alone. The result also demonstrated that CEE induced apoptosis in colonic epithelium cells of rat received colon carcinogen as detected the increasing of caspase-3 activity. This finding could be concluded that purple rice extracts inhibited aberrant colonic epithelial cell progression via apoptosis induction. [ABSTRACT FROM PUBLISHER]

Antioxidant ellagic acid is a herbal polyphenolic compound shown to possess growth-inhibiting and apoptotic activities in cancer. Protein kinase C (PKC) plays an important role in cell proliferation, apoptosis, and differentiation. Apoptosis of tumor cells is induced by inactivation of glycolytic enzyme of anaerobic metabolism, lactate dehydrogenase (LDH)-A, and by activating apoptotic protein caspase-3 via PKCδ. The present study aims to analyze the role of ellagic acid on regulation of novel and atypical isozymes of PKC to modulate apoptosis and anaerobic metabolism to prevent lymphoma growth as its role on classical PKCs is reported earlier. Expression of novel and atypical isozymes of PKC, activity of PKCδ, expression and activity of caspase-3, and LDH-A have been analyzed. Expression is measured by RT-PCR, activities of PKCδ as level of its catalytic fragment, caspase-3 as level of its p17 fragment, and LDH-A by specific staining. Lymphoma bearing mice were treated with 3 different doses of ellagic acid. The treatment enhanced expression of all novel and atypical PKCs, activity and expression of caspase-3, and activity of PKCδ but decreased activity and expression of LDH-A. Our results suggest that ellagic acid induces apoptosis via novel and atypical PKCs in association with caspase-3 and induces cancer cell death by blocking the energy metabolism. Zinc deficiency and excess influence cellular homeostasis and are believed to modulate apoptosis. Zinc also regulates cell growth and proliferation. Understanding of the role of zinc in the mechanisms associated with these changes is limited because of its diverse, complex, and cell-specific effects. Therefore, we investigated the oxidative stress responses and the underlying molecular mechanisms associated with the disruption of intracellular zinc homeostasis in H4IIE rat hepatoma cells. We found that zinc excess (100 μM) and DTPA (diethylenetriaminepentaacetic acid; 50–100 μM) induced zinc deficiency both generate reactive oxygen species (ROS) and decrease viability in H4IIE cells. However, cotreatment with the antioxidant, N-acetyl-L-cysteine (NAC) both reduced ROS production and protected cells from death. We additionally observed an increase in Bax mRNA and cytochrome c release from the mitochondria in DTPA-treated cells and an elevated expression of Fas/Fas ligand mRNA with zinc treatment. Both treatments increased p53 and MdM2 protein concentrations along with caspase 3/7 activity. These results suggest that zinc deficiency stimulates mitochondrial-dependent apoptosis whereas zinc activates the extrinsic-apoptotic pathway. Both decreasing and increasing cellular zinc concentrations modulate ROS mediated apoptosis and warrant further research on zinc mediated cancer chemoprevention in this and other cancer cell lines. Multiple studies have indicated that selective cyclooxygenase-2 (COX-2) inhibitors possess clinically chemopreventive and preclinically anticancer activities. Their long-term use, however, may be limited by the cardiovascular toxicity. This study tried to investigate whether an apple oligogalactan (AOG) could enhance the growth inhibitory effect of celecoxib on colorectal cancer. Caco-2 and HT-29 cell lines were exposed to different concentrations of AOG (0–1 g/L), celecoxib (0–25 μmol/L), and their combination. COX-2 levels were assessed by reverse transcription PCR and Western blot. COX-2 activity was evaluated by measuring prostaglandin E2 concentration. A colitis-associated colorectal cancer (CACC) mouse model was used to determine the effect of the combination in vivo. AOG (0.1–0.5 g/L) could potentiate the inhibitory effect of physiologic doses of celecoxib (5 μmol/L) on cell growth and decrease COX-2 expressions both at RNA and protein levels. In vivo, the combination (2.5% AOG plus 0.04% celecoxib, w/w) prevented against CACC in mice effectively. Our data indicate that AOG could potentiate the growth inhibitory effect of celecoxib on colorectal cancer both in vitro and in vivo through influencing the expression and function of COX-2 and phosphorylation of MAPKs, which suggests a new possible combinatorial strategy in colorectal cancer therapy. The article presents a case study of a 34-year-old man suffering from chylothorax. The patient was presented at the emergency department of the hospital with a stab wound on the neck. He underwent a chest X-ray and a computed tomography angiogram, which revealed a pleural effusion on the left side of the chest. The patient was diagnosed with a chylothorax that was caused due to a penetrating trauma to the neck.

Research on matrix metalloproteinases (MMPs) and in particular on gelatinase B, alias MMP-9, has grown exponentially in the decade 2003-2012. Structural details about flexibility of MMP-9 monomers, together with glycosylation, oligomerization, heterogeneity and instability of the wildtype enzyme explain why crystallography experiments have not yet been successful for the intact enzyme. MMP-9 may be viewed as a multidomain enzyme in which the hemopexin, the O-glycosylated and the catalytic domains yield support for attachment, articulation and catalysis, respectively. The stepwise proteolytic activation of the inactive zymogen into a catalytically active form becomes gradually better understood. Priming of activation by MMP-3 may be executed by meprins that destabilize the interaction of the aminoterminus with the third fibronectin repeat. Alternatively, autocatalytic activation may occur in the presence of molecules that tightly bind to the catalytic site and that push the cystein residue in the prodomain away from the catalytic zinc ion. Thanks to the development of degradomics technologies, substrate repertoires of MMP-9 have been defined, but it remains a challenge to determine and prove which substrates are biologically relevant. The substrate repertoire has been enlarged from extracellular to membrane-bound and efficient intracellular substrates, such as crystallins, tubulins and actins. Biological studies of MMP-9 have tuned the field from being primarily cancer-oriented towards vascular and inflammatory research. In tumor biology, it has been increasingly appreciated that MMP-9 from inflammatory cells, particularly neutrophils, co-determines prognosis and outcome. Aside from the catalytic functions executed by aminoterminal domains of MMP-9, the carboxyterminal hemopexin (PEX) domain of gelatinase B exerts non-catalytic anti-apoptotic signaling effects. The recognition that gelatinase B is induced by many pro-inflammatory cytokines, whereas its inhibitors are increased by anti-inflammatory cytokines, has generated interest to target MMP-9 in acute lethal conditions, such as bacterial meningitis, sepsis and endotoxin shock, and in acute exacerbations of chronic diseases. Previously described transcriptional regulation of MMP-9 is complemented by epigenetic checkpoints, including histone modifications and microRNAs. Because activation of proMMP-9 may be executed by other MMPs, the therapeutic dogma that MMP inhibitors need to be highly selective may be keyed down for the treatment of life-threatening conditions. When inflammation and MMP-9 fulfill beneficial functions to clear damaging protein complexes, such as in systemic autoimmune diseases, therapeutic MMP inhibition has to be avoided. In Mmp9 gene knockout mice, specific spontaneous phenotypes emerged with effects on the skeletal, reproductive and nervous systems. These findings not only have clinical correlates in bone growth and fertility, but also stimulate research on the roles of MMPs and MMP-9 in endocrinology, immunology and the neurosciences. Mmp9-deficient mice are valuable tools to define MMP-9 substrates in vivo and to study the role of this enzyme in animal models of inflammatory, vascular, neoplastic and degenerative diseases. Future challenges include solving the crystal structure, definition of the functions of covalent oligomers and heteromers in biology and pathology, life-imaging of MMP-9 activity, substrate determination in situ and the study of inhibitor effects on fertility, cancer and inflammation and in neurobiology and regenerative medicine. The existing literature on Problem-based learning (PBL) mostly paints a positive picture. If there were more evidence of the limitations of PBL, more could be done to apply the appropriate interventions to optimize the experience of PBL for students. The purpose of this article is to discuss second year medical students’ perceptions of the difficulties they face in knowledge acquisition in Biochemistry, Physiology and Anatomy during the first two years of transition from traditional education approaches to PBL. The main findings of the empirical study were that, through PBL, knowledge acquisition in Biochemistry and Physiology were perceived to be more difficult than in Anatomy. It is for this reason that, when transforming to PBL, measures need to be in place to address the challenges students might experience in acquiring knowledge in subjects like Biochemistry and Physiology. The aim of the study was to assess the age dependence on some physiological and serum chemistry parameters of male Wistar rats for the estimation of reference values in controlled environment. We are presenting values obtained from a large number of animals such as survival, average life span, body mass, food and water intake, serum chemistry parameters as total protein, albumin, transferrin and ferritin in serum. One part of this work compares the relationship between rat and human age. The maximal life span of our rats was determined to be about 4.4 years. The average life span was 3.75 years. The body weight quickly rose to the 85th week of life and then remained in the range of about 640–660 g up to the 163rd week when it began to decline. Food intake rose from the beginning to the maximum of about 39 g in the 33rd week and then decreased to about 20 g in the 163rd week. The water intake had a similar dynamics (about 43 mL in the 33rd week and 33 mL in the 163rd week). Levels of total protein in serum increased with age, in contrast, albumin levels decreased. Recent evidence suggests that fatty acid synthase mediating de novo fatty acid synthesis plays a crucial role in the carcinogenesis process of various cancers. Moreover, HER2 and related PI3K/Akt signaling pathway, which links intimately with cellular metabolism, influence cancer biological behavior. However, it remains unknown whether malignant phenotype of colorectal cancer cells is regulated by the HER2-PI3K/Akt-FASN signaling pathway. In this study, Caco-2 cells were selected for functional characterization, and treated with ZSTK474, followed by RT-qPCR and Western blot assays examining PI3K, Akt, HER2, and FASN expression. The MTT and colony formation assays were used to assess proliferation. The migration was investigated by transwell, apoptosis, and cell-cycle analysis. We found that the blockade of PI3K/Akt signaling pathway by ZSTK474 treatment led to downregulation of PI3K, Akt, HER2, and FASN expression. The proliferation was decreased upon treatment which was consistent with an increased percentage of G1 arrested cells instead of apoptosis. The migration of Caco-2 cells was also impaired by ZSTK474 treatment. Folate-binding protein (FBP) was discovered in cow's milk around 40 years ago. Bovine FBP belongs to a family of several folate-binding proteins. In milk, it is a soluble whey protein with the ability to sequester folate from blood plasma. Bovine FBP is a well-characterized protein in terms of amino acid sequence and binding characteristics. Affinity and binding kinetics towards various folate forms have been intensively studied because they are crucial in using bovine FBP as an analytical tool. Shortly after the identification of bovine FBP, a competitive protein-binding assay for measuring serum and blood folate concentrations was introduced. Another analytical application of bovine FBP is in affinity chromatography, as a clean-up/concentration step for analysis of folates in foods and biological samples by liquid chromatographic methods. Concentrations of FBP in milk and dairy products have been determined by ELISA and Surface Plasmon Resonance-biosensor techniques. Since the initial reports of FBP in cow's milk, its physiological role has been discussed, especially regarding its effects on folate absorption from milk and dairy products. The balance of putative pro- and antiinflammatory lipoxygenase (LOX)-derived S-hydroxyeicosatetraenoic acids (S-HETEs) in colon mucosa is a potential target for modulating colon cancer risk and progression. The biological effects of S-HETEs and R-hydroxyeicosatetraenoic acids (produced by distinct pathways) may differ, but levels of these compounds in the colon are unknown. The objective of this study was to develop chiral methods to characterize hydroxyeicosatetraenoic (HETE) enantiomers in colonic mucosa and evaluate the effects of fish oil on HETE formation. C57BL/6 mice (COX-1 null, COX-2 null, wild-type) were fed a diet supplemented with either olive oil or menhaden oil for 11 wk, and R-/S-HETEs in colonic mucosa were quantified by chiral LC-MS/MS. The R-enantiomer comprised 60–72% of 5-HETE, 18–58% of 15-HETE, and 1–16% of 12-HETE in colonic mucosa, suggesting that non-LOX sources contribute to HETE profiles. Fish oil reduced levels of both R- and S-HETEs, and increased the preponderance of the R-enantiomers (particularly 12- and 15-HETEs). There was apparent shunting of arachidonic acid to 12-/15-LOX in the COX-1 null animals. There is conflicting evidence for the role diet and lifestyle play in the development of mismatch repair (MMR)-deficient colorectal cancers (CRC). In this study, associations between MMR deficiency, clinicopathological characteristics, and dietary and lifestyle factors in sporadic CRC were investigated. Tumor samples from 185 individuals in the EPIC-Norfolk study were analyzed for MLH1 gene promoter methylation and microsatellite instability (MSI). Dietary and lifestyle data were collected prospectively using 7-day food diaries (7dd) and questionnaires. MMR-deficient tumor cases (MLH1 promoter methylation positive, MSI-H) were more likely to be female, older at diagnosis, early Dukes’ stage (A/B), and proximal in location (MSI-H P = 0.03, 0.03, 0.02, and 0.001, respectively). Tumors with positive MLH1 promoter methylation (>20%) were associated with poor differentiation (P = 0.03). Low physical activity was associated with cases without MSI (P = 0.05). MMR deficiency was not significantly associated with cigarette smoking or alcohol, folate, fruit, vegetable, or meat consumption. We conclude that MMR-deficient tumors represent a distinct subset of sporadic CRC that are proximal in location, early Dukes’ stage, and poorly differentiated, in cases that are female and older at diagnosis. The incidence of squamous cancer of the esophagus varies up to a hundredfold in different regions of the world. In Transkei, South Africa, a particularly high incidence of the disease is observed. We have previously proposed an association between a maize-rich diet and elevated levels of intragastric prostaglandin E2 production (PGE2). Here we investigate the molecular mechanisms by which a high-maize diet could lead to increased incidence of squamous cancer of the esophagus. We confirm that levels of PGE2 are high (606.8 pg/ml) in the gastric fluid of individuals from Transkei. We also show that treatment of esophageal cells with linoleic acid, which is found at high levels in maize and is a precursor to PGE2, leads to increased cell proliferation. Similarly, treatment of cells with PGE2 or with gastric fluid from Transkeians also leads to increased proliferation. Our data suggest that the high levels of PGE2 associated with a maize-rich diet stimulate cell division and induce the enzyme COX 2, resulting in a positive feedback mechanism that predisposes the esophagus to carcinoma. In this study, the physiological and biochemical response of Nile tilapia (Oreochromis niloticus) after 96 and 24 h exposure to aqueous extracts of neem (Azadirachta indica) in extract concentrations ranging from 0 to 32,000 mg/l was evaluated. After 96 h and 24 h exposure, the LC50 of neem extract was estimated at 3,200 and 6,800 mg/l, respectively. Plasma cortisol increased beyond pre-treatment levels at neem extract concentrations above 2,000 mg/l over 96 h and above 4,000 mg/l over 24 h. Blood glucose increased at neem extract concentrations above 1,000 and 5,000 mg/l at 24 and 96 h, respectively. Neem extract concentration had little effect on serum sodium and plasma chloride. Hematocrit was higher than the control at neem extract concentrations above 1,000 mg/l in the 96 h exposure and above 2,000 mg/l in the 24 h exposure. Plasma ammonia increased significantly at neem extract concentrations above 2,000 mg/l for both the 96 h and 24 h tests. Immediately after beginning treatment, cortisol levels increased significantly at neem extract concentrations above 2,000 mg/l in the 96 h test and 4,000 mg/l in the 24 h toxicity test. Exposure to neem extract interfered with the antioxidant defense system of the fish by reducing liver catalase activity. Body composition is a key consideration in the physical make-up of professional soccer players. The aims of the present study were to determine whether the body composition of professional soccer players varied according to playing position, international status or ethnicity, and to establish which variables best distinguished the soccer players from a reference group. Body composition was assessed using dual-energy X-ray absorptiometry in 64 male professional soccer players. Measured variables included bone mineral density and the relative amounts of lean and fat mass. Data were analysed using analysis of variance and stepwise discriminant function. The soccer players recorded better values than a reference group (n = 24) for all body composition compartments. Percent lean mass and bone mineral density were the variables best able to identify the soccer players (95.5% correctly classified). Differences in body composition were evident between goalkeepers and outfield players, but not between outfield playing positions. No differences were found on the basis of international status. The non-Caucasian players demonstrated significantly lower percent body fat (9.2 ± 2.0%) than the Caucasian players (10.7 ± 1.8%). It was concluded that body composition is important for elite soccer players, but that homogeneity between players at top professional clubs results in little variation between individuals. In this study, the first experiment analyzed the effects of 0, 85, 171, 256, and 342 mM sodium chloride (NaCl) on the germination of Acacia seyal seeds. In the second in vitro experiment, the effects of inoculation with two rhizobial strains on growth, physiological, and biochemical responses of A. seyal seedlings was evaluated at four levels (0, 85, 171, and 256 mM) of NaCl. Results showed that at 342 mM NaCl, germination rate of A. seyal seeds declined by 56% from the control value, while any germination was recorded for Acacia bivenosa and Acacia sclerosperma seeds at the same salt level. Salt stress gradually decreased the growth, the soluble protein and the leaf-chlorophyll contents of A. seyal seedlings. However, rhizobial inoculation limits these adverse effects of salt on physiological and metabolic processes responses. The north-western Indian Himalayan hill region contains around 40 phytophagous species of white grub (Coleoptera: Scarabaeidae). Anomala dimidiata and Holotrichia seticollis are the predominant species. Under natural conditions, up to one-fifth of the population of the grubs was found to exhibit symptoms of bacterial infection. Therefore, we attempted to isolate and characterize the most potent bacterium for the management of white grubs. Of 27 bacterial isolates tested against A. dimidiata, WGPSB-2 was found to be highly toxic. Based on morphology, physiology, biochemistry and sequencing of the 16S rRNA gene, the isolate WGPSB-2 was identified as Bacillus cereus. A dose determination bioassay indicated that first instar grubs of A. dimidiata and H. seticollis were more susceptible than second instars. Although the LT90 values for second instar grubs exceeded 8 weeks, the grubs stopped feeding after ingestion of spores, mostly after the third week. In outdoor microplots, a dose of 1.7 × 1010 spores/m2 applied to the root zone was found to be potentially satisfactory for control of the pests. Conclusion. Since aquaporin-6 (Aqp6) protein was located in the membrane of intracellular vesicles of the stria vascularis, endolymphatic sac, and vestibule, Aqp6 might be involved in some distinct physiological function of acid-base metabolism and water balance in endolymphatic fluid homeostasis. However, its lack of expression on the plasma membrane indicates that Aqp6 does not have a direct role in water flux via the plasma membrane. Objective. Aqp6 mRNA expression in the rat inner ear was investigated in the vestibulum as well as in the cochlea and endolymphatic sac using the reverse transcription-polymerase chain reaction (RT-PCR) method, and detailed immunolocalization of Aqp6 in the rat inner ear was investigated using immunohistochemical methods including immunofluorescence microscopy and immunoelectron microscopy. Results. We obtained novel data showing that not just Aqp6 mRNA but also Aqp6 protein is expressed in the cochlea, endolymphatic sac, and vestibule. Immunoelectron microscopic studies revealed that the immunolabelled gold was diffusely seen in the intracellular area of the stria vascularis, endolymphatic sac, and vestibule, but never in the plasma membranes. Azo dyes are widely used in the food, pharmaceutical, paper, and textile industries. Some azo dyes are known to produce carcinogenic compounds upon reductive cleavage of the azo bond (N =N) by intestinal flora. There is not much information available on the effect of these dyes on the physiology of the gut microflora as well as their kinetics of reduction in different environments. The azoreductase activity of Enterococcus faecalis, an important opportunistic intestinal pathogen, was tested using the azo dyes Methyl Red, Direct Blue 15 and tartrazine under the following conditions: a complex supplemented brain heart infusion (BHIS) medium under aerobic and anaerobic conditions, a simple phosphate-buffered saline glucose (PBSG) medium under anaerobic conditions, and cell extracts using NADH and NADPH as electron donors, under aerobic conditions. The kinetics of reduction in all these conditions was determined. The effect of high and low concentrations of each dye on the growth of E. faecalis was also tested. Our results show that the sulfonated azo dye tartrazine was completely reduced extracellularly in BHIS under aerobic and anaerobic conditions while the other sulfonated azo dye, Direct Blue 15 was completely reduced only under anaerobic conditions. The non-polar azo dye, Methyl Red was also completely reduced, with the highest rate of reduction in both conditions. The generation time of E. faecalis in the presence of high concentration of tartrazine was significantly lower than the control, while the generation time was unaffected in the presence of low and high concentrations of Methyl Red, Direct Blue 15 and 20 µM tartrazine. Our results suggest that the metabolism of different azo dyes by E. faecalis depends on the physiochemical properties of the azo dyes and the conditions of the surrounding environment in the presence of actively dividing cells. 1. The pharmacokinetics and metabolism of KW-4490, a selective phosphodiesterase 4 inhibitor, were investigated in rats and monkeys. After oral administration, KW-4490 was rapidly absorbed, and then its plasma concentrations apparently declined with half-lives of approximately 5 h in rats and 3.5 h in cynomolgus monkeys; however, a number of secondary peaks were apparent in the profiles for both species. The plasma pharmacokinetics of KW-4490 were comparable between rats and monkeys. 2. After oral administration, KW-4490 was mainly eliminated by metabolism to acylglucuronides and renal excretion in the unchanged form. KW-4490 acylglucuronides were found in monkey but not rat urine. In rats, KW-4490 acylglucuronides were excreted only in bile. Although the pathway of excretion of acylglucuronides differed between rats and monkeys, cumulative excretion in the two animals was very similar, as expected from comparable hepatic clearance for glucuronidation in rat and monkey liver microsomes. 3. The glomerular filtration rate of unbound KW-4490 indicated that renal tubular secretion was significant in monkeys, whereas reabsorption was significant in rats. These species differences in urinary excretion of KW-4490 and its acylglucuronide metabolites are most likely due to substrate specificity of active transporters in rat and monkey kidney. Biochemical markers of inflammation are emerging as new predictors of risk of cardiovascular disease (CVD) and may alter acutely with exercise. Few studies have been conducted on the effects of walking on these markers or whether different walking intensities elicit varied effects. As there is growing interest in modifiable lifestyle factors such as walking to reduce CVD risk, these inflammatory responses warrant investigation. The aim of this study was to compare the effects of walking at 50% versus 70% of predicted maximal heart rate on C-reactive protein (CRP), plasma fibrinogen, and trigylcerides in sedentary post-menopausal women. Twelve post-menopausal women (mean age 58 years, s ± 6; stature 1.62 m, s ± 0.06; body mass 66.8 kg, s ± 6.2) completed two 30-min treadmill walks in a randomized cross-over design. Fasted blood samples were taken (for the determination of plasma fibrinogen, CRP, and lipids) before, immediately after, and 1 and 24 h after exercise. Triglyceride concentrations decreased from pre-exercise to 24 h post exercise at both walking intensities (time×group interaction, P < 0.05). No significant effects were observed for plasma fibrinogen, CRP, total cholesterol, low-density or high-density lipoprotein cholesterol (time x group interaction, P > 0.05). The results of this study suggest that fasting plasma triglycerides are decreased on the morning after 30 min of brisk walking at either 50% or 70% of maximal heart rate (moderate and vigorous intensity). Biochemical changes in the body components during gonad maturation of the tropical abalone Haliotis varia were investigated using wild collected specimens from the Gulf of Mannar, on the southeast coast of India. The gonadosomatic index (GSI) and the hepatosomatic index (HSI) showed negative correlations throughout the study period as well as during the progression of gonad maturation stages. The highest GSI for both the sexes were in the ripe stages followed by late maturing stages. The HSI ranged from 2.97 to 6.71 in females, and 3.55 to 5.09 in males. Among the biochemical components analysed, lipid and carbohydrate contents showed significant variations in the different tissues of H. varia during the progress of gonad maturation. The highest protein content was in the foot muscle and the lowest was in the digestive gland. Total lipids in the ovary were always higher compared with that of the testis and the values ranged from 12.60 to 26.49%, registering the highest value in the ripe ovary. Gonad carbohydrate content was lower when the lipid content was higher, suggesting the conversion of carbohydrate to lipids. Toxicologic and physiologic roles of CYP1A enzyme induction, the major biochemical effect of aryl hydrocarbon receptor activation by TCDD and other receptor ligands, are unknown. Evidence is presented that CYP1A exerts biologic effects via metabolism of endogenous substrates (i.e., arachidonic acid, other eicosanoids, estrogens, bilirubin, and melatonin), production of reactive oxygen, and effects on K + and Ca 2+ channels. These interrelated pathways may connect CYP1A induction to TCDD toxicities, including cardiotoxicity, vascular dysfunction, and wasting. They may also underlie homeostatic roles for CYP1A, especially when transiently induced by common chemical exposures and environmental conditions (i.e., tryptophan photoproducts, dietary indoles, and changes in oxygen tension). The disposition of compound A, a potent inhibitor of the hepatitis C virus (HCV) NS5B polymerase, was characterized in animals in support of its selection for further development. Compound A exhibited marked species differences in pharmacokinetics. Plasma clearance was 44&ThickSpace;ml&ThickSpace;min -1 &ThickSpace;kg -1 in rats, 9&ThickSpace;ml&ThickSpace;min -1 &ThickSpace;kg -1 in dogs and 16&ThickSpace;ml&ThickSpace;min -1 &ThickSpace;kg -1 in rhesus monkeys. Oral bioavailability was low in rats (10%) but significantly higher in dogs (52%) and monkeys (26%). Compound A was eliminated primarily by metabolism in rats, with biliary excretion accounting for 30% of its clearance. Metabolism was mainly mediated by cyclohexyl hydroxylation, with N-deethylation and acyl glucuronide formation constituting minor metabolic pathways. Qualitatively, the same metabolites were identified using in vitro systems from all species studied, including humans. The low oral bioavailability of compound A in rats was mostly due to poor intestinal absorption. This conclusion was borne out by the findings that hepatic extraction in the rat was only 30%, intraperitoneal bioavailability was good, and compound A was poorly absorbed from the rat isolated intestinal loop, with no detectable intestinal metabolism. Compound A was not an inhibitor of major human cytochrome P450 enzymes, indicating minimal potential for clinical drug–drug interactions. The metabolic clearance of compound A in rat, dog and monkey hepatocytes correlated with the systemic clearance observed in these species. The metabolism and disposition of 14 C-labelled juglone in male F344 rats following oral, intravenous and dermal administration were studied. Approximately 40–50% of an oral dose (0.1 to 10&ThickSpace;mg&ThickSpace;kg -1 ) and less than 20% of the dermal dose (4&ThickSpace;mg&ThickSpace;kg -1 ) were absorbed within 24&ThickSpace;h. Most of the oral dose was excreted in faeces and urine within 24&ThickSpace;h and only 1–3% remained in the tissues. High concentrations of juglone-derived radioactivity were found in kidney for all three dosing routes. The accumulation in kidney can be attributed to covalent binding of juglone and/or metabolites to cytosolic protein. Five metabolites were identified in the urine of rats treated with an oral dose: 1,4,5-trihydroxynaphthalene di-glucuronide, 1,4,5-trihydroxynaphthalene mono-glucuronide mono-sulfate, 2-sulfo-2,3-dihydrojuglone, 4,8-dihydroxy-1-tetralone mono-glucuronide and 1,4,5-trihydroxynaphthalene mono-glucuronide. Apoptosis is a physiological process that controls tissue homeostasis, in combination with survival signals delivered by distinct receptors that bind hormones, growth factors or extracellular matrix components. The extrinsic pathway of apoptosis is due to the triggering of death receptors and the activation of the caspase cascade; the intrinsic pathway is due to withdrawal of growth factors and mainly related to mitochondrial metabolism. The choice between survival or apoptosis, which is the result of such different integrated environmental signals, is crucial for the maintainance of bone marrow reservoir of hematopoietic precursors (HPC). CD34  +  HPC can receive multiple survival signals during homing and maturation, due to different interactions with adhesion molecules expressed on endothelial and bone marrow stromal cells, proteins of the extracellular matrix and chemokines or growth factors. Indeed, this molecule, originally described as an adhesion receptor belonging to the immunoglobulin superfamily, capable of homophilic and heterophilic interactions, turned out to be a signalling molecule, containing an immunoreceptor tyrosine-based inhibitory motifs (ITIM) within its cytoplasmic domain. In particular, it has been shown that PECAM-1 binds to different kinases and phosphatases, including the phosphatidylinositide-3-kinase that phosphorylates Akt, which, in turn can upregulate transcription and function of antiapoptotic proteins, such as Bcl-2 and Bcl-x or A1, responsible for the rescue from mitochondrial apoptosis. Role of α-tocopherol (vitamin E), β-carotene and/or their combination as antioxidants against the toxicity of fenvalerate on blood hematology, free radicals, biochemical parameters, and semen quality were studied in male rats. Fenvalerate (20 mg/kg BW), vitamin E (100 mg/kg BW), β-carotene (10 mg/kg BW), and vitamin E plus β-carotene (100+10 mg/kg BW, respectively) were given alone or in combination with fenvalerate. The tested doses were given to rats every other day for 30 days. Results obtained showed that fenvalerate significantly (P<0.05) induced free radicals in plasma and brain and insignificantly in liver and testes. While, vitamin E, β-carotene alone and/or in combination decreased the levels of free radicals in plasma, liver, testes, and brain. The activities of glutathione S-transferase (liver), alkaline phosphatase (plasma and liver), aspartate amino-transferase (plasma, liver, and testes) and alanine aminotransferase (plasma and liver) were significantly (P<0.05) increased due to fenvalerate administration. The activity of acetylcholinesterase was significantly (P<0.05) decreased in brain and plasma, while plasma glucose, urea, creatinine, and bilirubin concentrations were significantly (P<0.05) increased in rats treated with fenvalerate. Also, results showed a significant (P<0.05) alterations in plasma proteins, hematological parameters, body weight, and relative weights of organs. Sperm concentration and motility (%) were significantly (P<0.05) decreased, while dead and abnormal sperm increased in rats exposed to fenvalerate. Vitamin E, b-carotene alone and/or in combination did not cause any changes in the investigated parameters, but improved semen quality and minimized the toxic effect of fenvalerate. The obtained results demonstrated the beneficial influences of vitamin E, b-carotene alone and/or in combination in reducing the harmful effects of fenvalerate. [ABSTRACT FROM AUTHOR]

Multiple interacting mechanisms control the formation and dissolution of clots to maintain blood in a state of delicate balance. In addition to a myriad of biochemical reactions, rheological factors also play a crucial role in modulating the response of blood to external stimuli. To date, a comprehensive model for clot formation and dissolution, that takes into account the biochemical, medical and rheological factors, has not been put into place, the existing models emphasizing either one or the other of the factors. In this paper, after discussing the various biochemical, physiologic and rheological factors at some length, we develop a model for clot formation and dissolution that incorporates many of the relevant crucial factors that have a bearing on the problem. The model, though just a first step towards understanding a complex phenomenon, goes further than previous models in integrating the biochemical, physiologic and rheological factors that come into play. Objective: In the development of autoimmune diseases, dendritic cells (DC) play critical roles. Here, we examined the effect of aspirin on lipopolysaccharide (LPS)-induced DC activation.Methods: The monocyte-derived DC were established. The cytokine production was measured by ELISA, reverse transcriptase/polymerase chain reaction, or intracellular staining analyzed by flow cytometry. The expression of cell surface molecules was determined by flow cytometry.Results: Aspirin inhibited LPS-induced DC maturation and costimulatory molecules expression. Aspirin, at therapeutic concentrations, also decreased LPS-induced IL-12 and IL-10 production. In contrast, the LPS-induced TNF-alpha production was enhanced by aspirin. The differential effects of aspirin on IL-12 and TNF-alpha production may not be due to down-regulation of cyclooxygenase activities.Conclusion: The various effects of aspirin on LPS-stimulated DC may influence the understanding of the diverse immunomodulatory mechanisms of this anti-inflammatory drug. The effect of oxidative stress catalysed by transition metals appears tohave a critical relevance for the structure and function not only of membranelipids but also of integral membrane proteins in a complex lipid-protein assembling,and membrane-dependent function. The integral membrane enzyme 5′-nucleotidaseis susceptible to Fe[sup [sup 2+] ] -ion catalysed oxidative modification,and the extent of enzyme inhibition is in inverse relationship (r = -0.820)with lipid peroxidation (MDA) level. This work is also a comparative studyabout possible effectiveness of different Fe-ion chelators (deferoxamine,Na-citrate, Na-salicylate, ammonium oxalate and EDTA), antioxidants (GSH,GSH/GSH-Px system, Cu, Zn-SOD and mannitol) and metal cations (Mg[sup 2+] and Mn[sup 2+] ) to protect or restore Fe[sup 2+] -ion induced 5′-nucleotidaseinhibition and to supress Fe[sup 2+] -ion enhanced lipid peroxidation.Among the examined chelators it was only deferoxamine and Na-citrate thatexerted a fully protective and reactivating ability; among the antioxidantsit was only GSH; among the metal cations it was only Mn[sup 2+] . Theability to protect or restore 5′-nucleotidase activity and to diminishchain-induced lipid peroxidation is explicable in terms of: metal-bindingability, capacity of taking iron away from a biological molecule, or abilityof transferring the damage to itself. After a short incubation period, theiron associated with enzyme or lipid hydroperoxides could be in a labile coordinativelinkage, still able to interact with possible ligands or metal cations.
