The investigation dealt with the effect of the replacement of a part of wheat flour by pea flour on the properties of batters and cakes. As the protein composition of pea flour differs from that of wheat, the effect of its incorporation on batter formation and cake properties was monitored throughout the different steps of cake processing. The incorporation of air, which influences the cell structure and density of the cake, was the subject of particular attention. Four orders of incorporation were first investigated to identify their effects on a standard recipe made with 100% wheat flour. Mixing first egg and sugar together allows introducing air, but most of it is lost after oil and flour introduction. Whatever the order of incorporation, the density of the batter ends around 1.1 ± 0.2 g.cm(−1). However, batter consistencies are significantly different and resulting cakes show different crumb structures. These results are discussed in terms of physicochemical mechanisms, and a schematic representation of the phenomena occurring at the different steps of mixing depending on the order of ingredient incorporation is proposed. When 20 and 40% of the wheat flour was replaced by pea flour using the two most energy-efficient orders of incorporation, more air was incorporated into the batter. However, the resulting cakes were denser, but surprisingly softer. Differences in cell structure explain this apparent contradiction.

To trace the geographical origin of wheat flour used in breads, we examined the stable carbon, nitrogen, and oxygen isotope ratios (δ13C, δ15N, and δ18O) of wheat flour proteins. The presence or absence of auxiliary materials, such as yeast, butter, and non-fat dry milk powder, had little influence on the protein composition of wheat glutenin fractions extracted from breads. We determined the δ13C, δ15N, and δ18O values of the wheat glutenin fractions from bread samples made from wheat flour obtained from Canada, USA, and Japan. Objective: To develop clinical tools assessing the refluxogenic potential of foods and beverages (F&B) consumed by patients with laryngopharyngeal reflux (LPR). Methods: European experts of the LPR Study group of the Young-Otolaryngologists of the International Federation of Oto-rhino-laryngological societies were invited to identify the components of Western European F&B that would be associated with the development of LPR. Based on the list generated by experts, four authors conducted a systematic review to identify the F&B involved in the development of esophageal sphincter and motility dysfunctions, both mechanisms involved in the development of gastroesophageal reflux disease and LPR. Regarding the F&B components and the characteristics identified as important in the development of reflux, experts developed three rational scores for the assessment of the refluxogenic potential of F&B, a dish, or the overall diet of the patient. Results: Twenty-six European experts participated to the study and identified the following components of F&B as important in the development of LPR: pH; lipid, carbohydrate, protein composition; fiber composition of vegetables; alcohol degree; caffeine/theine composition; and high osmolality of beverage. A total of 72 relevant studies have contributed to identifying the Western European F&B that are highly susceptible to be involved in the development of reflux. The F&B characteristics were considered for developing a Refluxogenic Diet Score (REDS), allowing a categorization of F&B into five categories ranging from 1 (low refluxogenic F&B) to 5 (high refluxogenic F&B). From REDS, experts developed the Refluxogenic Score of a Dish (RESDI) and the Global Refluxogenic Diet Score (GRES), which allow the assessment of the refluxogenic potential of dish and the overall diet of the LPR patient, respectively. Conclusion: REDS, RESDI and GRES are proposed as objective scores for assessing the refluxogenic potential of F&B composing a dish or the overall diet of LPR patients. Future studies are needed to study the correlation between these scores and the development of LPR according to impedance–pH study.

Bread wheat is the primary bread crop in the majority of countries in the world. The most important product that is manufactured from its grain and flour is yeast bread. In order to obtain an excellent bread, grain with high physical properties is needed for flour and dough. The Russian spring wheat cultivar Saratovskaya 29 is characterized by its exclusively high physical properties of flour and dough. The purpose of this work was to identify the chromosomes carrying the main loci for these traits in Saratovskaya 29 and to map them using recombinant substitution lines genotyped with molecular markers. A set of inter-varietal substitution lines Saratovskaya 29 (Yanetzkis Probat) was used to identify the “critical” chromosomes. The donor of individual chromosomes is a spring cultivar with average dough strength and tenacity. Substitution of 1D and 4D*7A chromosomes in the genetic background of Saratovskaya 29 resulted in a significant decrease in the physical properties of the dough. Such a deterioration in the case of 1D chromosome might be related to the variability of gluten protein composition. With the help of recombinant substitution double haploid lines obtained from a Saratovskaya 29 (Yanetzkis Probat 4D*7A) substitution line the region on the 4D chromosome was revealed in the strong-flour cultivar Saratovskaya 29, with the microsatellite locus Xgwm0165 to be associated with the unique physical properties of flour and dough. The detected locus is not related to the composition gluten proteins. These locus may be recommended to breeders for the selection of strong-flour cultivars. Additionally, a QTL associated with vitreousness of grain was mapped in the short arm of chromosome 7A.

Background Radix isatidis (Isatis indigotica Fort.) is an ancient medicinal herb, which has been applied to the prevention and treatment of influenza virus since ancient times. In recent years, the antioxidant activity of Radix isatidis has been widely concerned by researchers. Our previous studies have shown that Radix isatidis protein (RIP) has good antioxidant activity in vitro. In this study, the composition of the protein was characterized and its antioxidant activity in vivo was evaluated. Methods The model of oxidative damage in mice was established by subcutaneous injection of D-galactose for 7 weeks. Commercially available kits were used to determine the content of protein and several oxidation indexes in different tissues of mice. The tissue samples were stained with hematoxylin and eosin (H&E) and the pathological changes were observed by optical microscope. The molecular weight of RIP was analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The amino acid composition of RIP was determined by a non-derivative method developed by our research group. Results RIP significantly increased the activities of antioxidant enzymes such as SOD, CAT, GSH-Px and total antioxidant capability (TAOC) but decreased the MDA level in the serum, kidney and liver. H&E stained sections of liver and kidney revealed D-galactose could cause serious injury and RIP could substantially attenuate the injury. The analysis of SDS-PAGE showed that four bands with molecular weights of 19.2 kDa, 21.5 kDa, 24.8 kDa and 40.0 kDa were the main protein components of RIP. Conclusions The results suggested that RIP had excellent antioxidant activity, which could be explored as a health-care product to retard aging and a good source of protein nutrition for human consumption.

Quality of durum wheat is of importance for pasta production. Our aim was to evaluate the quality of released durum wheat at Ethiopia by the use of pheno-quality traits as well as composition and amount and size distribution of proteins; these attributes were also compared with the same ones for Spanish released wheat grown in the same environment. Large variation was obtained among the released durum wheat for all parameters investigated. The evaluation of protein parameters indicated the varieties Klinto, LD-357, Tob-66 and Ude to have suitable protein composition for pasta production, while Tob-66 showed high protein concentration and Tob-66, LD-357 and Yerer showed high gluten strength. The most promising Ethiopian variety for pasta production as to the results from the present study was Tob-66. Also, LD-357 showed promising protein characteristics, although the kernels were white, which is not desired for pasta production. Some of the evaluated varieties might be of interest for production of local leavened bread although the quality is not good enough for pasta production.

The physicochemical, functional, antioxidant and anticancer properties of protein isolates from the seeds of Soybean (SP), Black soybean (BSP), Adzuki bean (ABP), and Mung bean (MBP) were comparatively characterized. The difference was found in the protein composition and physicochemical properties of the four types of legume proteins, which affected the functional properties and bioactivities. BSP and SP had six predominant proteins with a molecular weight (MW) range of 20–95 kDa, whereas ABP and MBP showed the most intense bands of 48 kDa. ABP with higher essential amino acids content and the highest solubility exhibited the highest antioxidant activities among the four types of proteins. While BSP with higher content of acidic amino acids, low content of the hydrophobic amino acids and higher WHC, may have potential nutraceutical uses.

As a part of development of medicine for extreme situations, we determined, for the first time, the protein composition of the exhaled breath condensate (EBC) of cosmonauts before and after long-lasting orbital flights on the International Space Station (ISS). The analysis was performed by electrospray ionization ion cyclotron resonance mass spectrometry. The pattern of variation of the protein composition of the exhaled air during preflight preparation, at the time of landing, and seven days after landing was elucidated. Forty-four proteins, which appeared in the exhaled breath condensate of the cosmonauts after a long orbital flight and landing and were absent before the flight, were detected. These proteins included the immune response proteins, indicating cell and blood vessel damage. In some cases, a temporary appearance of cancer markers was noted.

ProCos (Protein Composition Server, script version), one of the machine learning techniques, was used to classify nitrilases as aliphatic and aromatic nitrilases. Some important feature vectors were used to train the algorithm, which included pseudo-amino acid composition (PAAC) and five-factor solution score (5FSS). This clearly differentiated into two groups of nitrilases, i.e., aliphatic and aromatic, achieving maximum sensitivity of 100.00%, specificity of 90.00%, accuracy of 95.00% and Mathew Correlation Coefficient (MCC) of about 0.90 for the pseudo-amino acid composition. On the other hand, five-factor solution score achieved a sensitivity of 96.00%, specificity of 84.00%, accuracy of 90.00% and Mathew Correlation Coefficient (MCC) of about 0.81. The total count of aliphatic amino acids, Ala (A), Gly (G), Leu (L), Ile (I), Val (V), Met (M) and Pro (P), was found to be higher, i.e., 42.7 in case of aliphatic nitrilases, whereas it was 40.1 in aromatic nitrilases. On the other hand, aromatic amino acids, Tyr (Y), Trp (W), His (H) and Phe (F) number, were found to be higher, i.e., 12.7 in aromatic nitrilases as compared to aliphatic nitrilases which was 10.7. This approach will help in predicting a nitrilase as aromatic or aliphatic nitrilase based on its amino acid sequence. The protein composition of animal venoms is usually determined by peptide-centric proteomics approaches (bottom-up proteomics). However, this technique cannot, in most cases, distinguish among toxin proteoforms, herein called toxiforms, because of the protein inference problem. Top-down proteomics (TDP) analyzes intact proteins without digestion and provides high quality data to identify and characterize toxiforms. Denaturing top-down proteomics is the most disseminated subarea of TDP, which performs qualitative and quantitative analyzes of proteoforms up to ~30 kDa in high-throughput and automated fashion. On the other hand, native top-down proteomics provides access to information on large proteins (> 50 kDA) and protein interactions preserving non-covalent bonds and physiological complex stoichiometry. The use of native and denaturing top-down venomics introduced novel and useful techniques to toxinology, allowing an unprecedented characterization of venom proteins and protein complexes at the toxiform level. The collected data contribute to a deep understanding of venom natural history, open new possibilities to study the toxin evolution, and help in the development of better biotherapeutics.

Wine producers have to label the use of certain animal proteins for fining of must and wine since 2012 [regulation (EC) No. 579/2012] under specific conditions, which has encouraged the search for alternative fining agents produced out of plants or yeasts. Yeast proteins are naturally present in wine and first studies of yeast protein extracts (YPE) for the fining of wine have been published, which were mainly focused on the technical properties of the YPE as fining agents. The present study used YPE of different production techniques for fining of red wine and evaluated their technical properties. Furthermore, the sensory effect of the new fining agents, their protein composition and the presence of glycans and glycoproteins were examined. The YPE showed differences in their protein profile and in protein glycosylation, but all YPE that were investigated were successfully applied for fining of red wine. Mannoproteins were detected in all YPE, and they did not prevent the mutual precipitation of proteins and tannins that is necessary for successful fining as reported earlier in case of glycans and glycoproteins. The chemical parameters of the red wine were modified by fining with YPE in a way comparable to fining with gelatine, a traditional fining agent used as reference, and the initial quality of wine flavour was maintained during the treatments.

Whereas the protein composition and overall shape of several giant virus capsids have been described, the mechanism by which these large capsids assemble remains enigmatic. Here, we present a reconstruction of the capsid of Cafeteria roenbergensis virus (CroV), one of the largest viruses analyzed by cryo-electron microscopy (cryo-EM) to date. The CroV capsid has a diameter of 3,000 Å and a Triangulation number of 499. Unlike related mimiviruses, the CroV capsid is not decorated with glycosylated surface fibers, but features 30 Å-long surface protrusions that are formed by loops of the major capsid protein. Based on the orientation of capsomers in the cryo-EM reconstruction, we propose that the capsids of CroV and related giant viruses are assembled by a newly conceived assembly pathway that initiates at a five-fold vertex and continuously proceeds outwards in a spiraling fashion.

WOS: 000391814900008, PubMed: 27867819, Background: A total of 150 bread wheat genotypes representing 121 Indian and 29 Turkish origin were screened for nutrient concentrations and grain protein content. Elemental and grain protein composition were studied by Inductively Coupled Plasma-Atomic Emission Spectrophotometer and LECO analyser, respectively. The study was performed to determine the variability in nutrient concentrations present in the collected wheat genetic material from two countries. Results: Several fold variations among genotypes existed for almost all the elements. Three major components of principal component analysis (PCA) revealed 60.8% variation among the genotypes. Nutrient variables segregated into two groups, one group containing all the macroelements except sulphur; and another cluster containing proteins and all the microelements except Zn and Mn. Pearson correlation analysis and heat-map were in accordance with each other determining strong positive association between P-K, Mn-Zn, Mg-S and Cu-protein content. Also, PCA and hierarchical grouping divided all the Indian and Turkish genotypes in two main clusters. Conclusions: Nutritional profile differentiated the genotypes from two countries into separate groups. However, some of the varieties were closely associated and indicated the success of global wheat exchange programs. While most of the correlations were in agreement with the previous studies, non-association of zinc with grain protein content directed towards its control by some other genetic factors. Some of the experimental wheat varieties with promising nutrient content have been suggested for future wheat advancement programs. In this paper volatile organic compounds (VOCs) from durum wheat cultivars and landraces were analyzed using PTR-TOF-MS. The aim was to characterize the VOC’s profile of the wholemeal flour and of the kernel to find out if any VOCs were specific to varieties and sample matrices. The VOC data is accompanied by SDS-PAGE analyses of the storage proteins (gliadins and glutenins). Statistical analyses was carried out both on the signals obtained by MS and on the protein profiles. The difference between the VOC profile of two cultivars or two preparations of the same sample - matrices, in this case kernel vs wholemeal flour - can be very subtle; the high resolution of PTR-TOF-MS - down to levels as low as pptv - made it possible to recognize these differences. The effects of grinding on the VOC profiles were analyzed using SIMPER and Tanglegram statistical methods. Our results show that it is possible describe samples using VOC profiles and protein data.

Oat prolamin (avenin) is a family of proteins that contain several polymorphic components. The high interspecific variability of avenin among cultivars, electrophoretic patterns have been used for grain protein comparison and species relationships. Therefore, the present study was carried out to evaluate the effect of environment on avenin constitution following an effort to increase protein content of oat using different approaches. The avenin electrophoretic patterns of 25 samples resulting from 20 oat cultivars and 5 mutated oat lines grown or developed in different fields and greenhouses were compared. Quantification of total proteins and SDS-PAGE analysis of avenin protein extracts were carried out. Different pattern of avenin allele in electrophoretic gel allowed the comparison within and between groups. This analysis shows that the cultivars obtained from Swedish fields had differences in the number of alleles compared with the parental cultivar, so were not stable across sites. The study revealed that oats treated with nitrogen and the selected mutated lines showed increase in protein concentration, with consistent avenin loci to the parental line, so these lines are now being used as potential candidates for developing high-protein oat lines in the future. Our results provide useful information, on the stability of modifications, for breeders aiming to increasing the content and nutritional value of oat protein. The study will increase our knowledge in the stability of different modifications and thus to accordingly select oat lines with stable and improved nutritional values.

Physicochemical properties of Korean wheat flours were evaluated to determine the effect of flour characteristics on yellow alkaline noodles by comparing commercial and imported wheat flours. Optimum water absorption, thickness, and color of noodle dough significantly correlated with protein content-related parameters of flour. Korean waxy wheats showed shorter cooking time (8 min) and softer texture of cooked noodles than other Korean wheats. Fish meat was brought to a supercooled state through slow cooling, and changes in the texture, histology, and protein composition of the meat were investigated. The groups whose storage temperature were lowered by 1.0°C per day (the 1.0°C group) and 0.5°C per day (the 0.5°C group) began to freeze in the vicinity of −3.5 and −5.0°C, respectively. The freezing point depended on the fish species; the lowest freezing point was −8.5°C, for the red sea bream in the 1.0°C group. The breaking strength tended to decrease more slowly in the 1.0°C group, but the collagen fibers collapsed more rapidly in the 1.0°C group. In SDS electrophoresis, a slight change in the banding patterns was observed, but the relationship between this observation and changes in histology and physical properties was unclear. This study shows that it is possible to produce a supercooled state in fish meat, and demonstrates that supercooling is a potential new storage method that lowers the temperature without generating ice crystals.

– The protein composition of 24 individual Maiwa yak milk samples that were collected in the Sichuan province of China was determined by reversed-phase high-performance liquid chromatography. Yak milk differed from cow milk by its high concentration of total proteins (46.2– 58.4 g·L−1), total casein (40.2 g·L−1 on average) and the proportion of individual caseins. The high content of β-casein (more than 45%) and consequently the lower proportion of αs-casein (about 40%) together with a small increase in κ-casein (15%) appear to make this milk more favorable for infant nutrition as used by the Tibetan nomads. Also, the whey proteins, β-lactoglobulin and serum albumin, showed similarities in centesimal composition with the homologous cow whey proteins. However, this initial study on yak milk proteins has highlighted the lack of knowledge in the literature and underlines that much research is required to optimize the utilization of all components of this milk for people living in extreme climatic environments.

White flours from 23 einkorn breeding lines (assortment 1) and wholemeal flours from 24 einkorn lines (assortment 2) were investigated for their qualitative and quantitative protein compositions by means of a combined extraction/HPLC procedure. The HPLC patterns of the gliadin fractions enabled the differentiation of most einkorn samples. The absence of a group of γ-gliadins at the beginning of the γ-gliadin elution region was unique for einkorn compared to all other wheat species. Differences in the patterns of γ-gliadins allowed the classification of einkorns into four groups; a further subdivision of these groups was possible by the number of ω5-gliadins and the different patterns of α-gliadins and low-molecular-weight glutenin subunits. The total gluten protein (gliadins + glutenins) contents of einkorn flours were similar to or even higher than those of common wheat and spelt. Typical for einkorn flours was the extreme excess of gliadins over glutenins with ω5-gliadins being most abundant and high-molecular-weight glutenin subunits being extremely rare. Micro-tests on the mixing properties and baking performance of assortment 2 flours revealed remarkable differences. Dough development time was negatively correlated with the ratio of gliadins to glutenins and positively with the content of glutenins; bread volume was mainly dependent on the content of glutenins. In conclusion, the determination of the quantitative gluten protein compositions offers a reliable indication of the expected baking quality during the early stages of breeding.

Deciphering the protein composition of complex extracts or discovering biologically relevant polypeptides is frequently hindered by large dynamic concentration ranges. The presence of high-abundance proteins suppresses the signal of low-abundance ones, and the most rare proteins are frequently below the sensitivity of available analytical methods. This protocol addresses this problem by using highly diversified hexapeptide ligand libraries for capturing proteins. A protein extract is mixed with the library and because the library has equal amounts of each ligand, theoretically the maximum amount of each protein bound is the same. Under overloading conditions this has the effect of diluting those proteins present in excess of the ligand concentration and concentrating those of relatively lower abundance. Unbound components are washed out and captured species are finally desorbed. The entire sample treatment process takes about half a day and yields a protein solution that could be used as such for mass spectrometry investigations.

Several studies show the relationships between technological parameters and flour protein composition issued from different cultivars. Our objective was to investigate the protein and technological complementarity of the Soissons cultivar flour, blended with the Tremie or Galibier cultivar flours according to different proportions. For each mixture level, the technological parameters (P and W of the Alveograph) and the rearrangement of the glutenin aggregates (SE-HPLC), after kneading, were determined. We noticed that some technological parameters of flour mixtures were the average of the parameters of the pure flour while others were significantly different from those expected. The SDS insoluble glutenin dough content increased according to the proportion of Soissons flour in the Soissons/Tremie blend. However, this content was maximum when the proportion of Soissons flour reached 50% in the Soissons/Galibier blend. This content is well correlated with the P and W for the Soissons/Tremie blend while such relations are not confirmed in the Soissons/Galibier blend. We also studied the Molecular Weight Distribution of SDS insoluble fraction in the flour and dough by HPSEC-MALLS ; the results showed that molecular mass is a good predictive parameter for P and W in dough whereas gyration radius is better on flour. Nevertheless, none of these parameters is appropriate to predict baking values.

The purpose of this study was to compare the relative amount of low molecular weight salivary proteins in patients with head and neck tumours treated with radiotherapy and healthy subjects. Reverse-phase high-pressure liquid chromatography was used for protein separation. Nine protein fractions (including acidic and basic proline-rich proteins (PRPs), cystatins, histatins and statherin) were identified in saliva from irradiated patients as well as healthy subjects. However, compared with non-irradiated healthy subjects, the fraction of acidic PRPs was significantly reduced in irradiated patients. These data indicate an alteration of the relative amount of low molecular weight salivary proteins in irradiated patients besides the reduction of salivary flow.

As soon as they enter a biological medium (cell culture medium for in vitro, blood or plasma for in vivo studies), nanoparticles, in most cases, see their surface covered by biomolecules, especially proteins. What the cells see is thus not the ideal nanoparticle concocted by chemists, meaning the biomolecular corona could have great biological and physiological repercussions, sometimes masking the expected effects of purposely grafted molecules. In this review, we will mainly focus on gold nanoparticles. In the first part, we will discuss the fate of these particles once in a biological medium, especially in terms of size, and the protein composition of the corona. We will highlight the parameters influencing the quantity and the identity of the adsorbed proteins. In a second part, we will resume the main findings about the influence of a biomolecular corona on cellular uptake, toxicity, biodistribution and targeting ability. To be noticed is the need for standardized experiments and very precise reports of the protocols and methods used in the experimental sections to extract informative data. Given the biological consequences of this corona, we suggest that it should be taken into account in theoretical studies dealing with nanomaterials to better represent the biological environment.

Rapid signal propagation along vertebrate axons is facilitated by their insulation with myelin, a plasma membrane specialization of glial cells. The recent application of proteomic approaches to biochemically purified myelin established that its protein composition is much more complex than previously thought. Bioinformatic integration of the myelin proteome with other systematic datasets such as mRNA abundance profiles of myelinating glial cells provides valuable resources for the molecular dissection of myelin biology and pathology.

Herpesvirus infection is associated with an increase of LPO and of the membrane lipid and lipid-protein layers microviscosity in erythrocyte membranes of pregnant patients. This results in modification of the erythrocyte stromal protein composition. Erythrocyte deformability increases sharply under these conditions, this leading to disorders in tissue metabolism in the organs of pregnant patients.

The sarcomere of striated muscle is a very efficient machine transforming chemical energy into movement. However, a wrong distribution of the generated forces may lead to self-destruction of the engine itself. A well-known example for this is eccentric contraction (elongation of the sarcomere in the activated state), which damages sarcomeric structure and leads to a reduced muscle performance. The goal of this review is to discuss the involvement of different cytoskeletal systems, in particular the M-band filaments, in the mechanisms that provide stability during sarcomeric contraction. The M-band is the transverse structure in the center of the sarcomeric A-band, which is responsible both for the regular packing of thick filaments and for the uniform distribution of the tension over the myosin filament lattice in the activated sarcomere. Although some proteins from the Ig-superfamily, like myomesin and M-protein, are the major candidates for the role of M-band bridges, the exact molecular organisation of the M-band is not clear. However, the protein composition of the M-band seems to modulate the mechanical characteristics of the thick filament lattice, in particular its stiffness, adjusting it to the specific demands in different muscle types. The special M-band design in slow fibers might be part of structural adaptations, favouring sarcomere stability for a continuous contractile activity over a broad working range. In conclusion, we discuss why the interference with M-band structure might have fatal consequences for the integrity of the working sarcomere.

We developed a one-step method for the chromatographic purification of hyaluronidase extract on Sepharose Blue, which produced the purified enzyme with a high yield (95%). Purification of hyaluronidase allowed us to obtain biological preparations standardized in protein composition and enzyme activity, which are useful for basic and applied studies.

Three cowpea varieties were used in the present study,Insect susceptible popular variety, `Ife-brown' (IFB), an improved varietyfor insect resistance IT 81D-975 (1975) and a local black cowpea varietywith a white hilium adapted to diverse habitats, `Akidi' (AKD).Cowpea seeds were infested with Callosobruchus maculatus. Crudeprotein content of infested and uninfested whole cowpea flour and proteinconcentrate was determined. Prolamin contents of flour and proteinconcentrates were evaluated. Fat content, free fatty acid and peroxidevalues of cowpea seeds were also determined. All determinations werecarried out in infested and uninfested cowpea seeds in 3 replicates for eachdetermination. Emulsion properties of cowpea seeds stored in 3 containers,plastic cans (PC) and polyethylene bags (white and black WPB/BPB) weremonitored on a biweekly basis for 24 weeks.Varietal differences existed in the protein, fat and prolamin contents ofcowpeas. Infestation reduced the contents of these nutrients in all cases. Free fatty acid contents and peroxide values were increased by >100%. Infestation reduced the prolamin content by > 25% inall cases. Varietal differences also existed in emulsion capacities of cowpeaflours; IFB and 975 formed better and more stable emulsions than AKDflour. Storage in plastic cans led to better retention of the emulsionproperties of the cowpeas.

Proteomics provides an extremely powerful tool for the study of variations in protein expression between individuals, different disease states and different conditions. One of the major challenges facing the medical profession in the forthcoming decades is to understand the changes that occur in individuals as they become older and to attempt to develop means to improve the quality of life for the otherwise healthy ageing population. The present study describes the first phase of such an investigation in which the protein composition of human skeletal muscle samples from young and aged subjects are compared. These results provide the beginning of a Human Aged Skeletal Muscle Profile reference map which is an essential first step to further investigations.

Changes in protein synthesis in the hypothalamus, pituitary, adrenals, liver, myocardium, and femoral muscle during exposure to thermal factor were studied. Protein synthesis was activated in all studied tissues during the initial period of exposure to environmental temperatures of 35 and 40 degrees C. At higher temperature (45 degrees C) the synthesis was notably suppressed in all tissues except the pituitary, where it remained high (higher than in the control). Analysis of protein composition showed a series of polypeptides, identical to stress proteins, synthesized de novo.

The formation and maturation of focal adhesions involves significant changes in protein composition and requires acto-myosin contractility. A mass spectrometry approach reveals changes to the focal adhesion proteome on myosin inhibition, providing a valuable resource for the cell adhesion field.

Electronegative low-density lipoprotein (LDL) found in human plasma is highly atherogenic, and its level is elevated in individuals with increased cardiovascular risk. In this review, we summarize the available data regarding the elevation of the levels of electronegative LDL in the plasma of patients with various diseases. In addition, we discuss the harmful effects and underlying mechanisms of electronegative LDL in various cell types. We also highlight the known biochemical properties of electronegative LDL that may contribute to its atherogenic functions, including its lipid and protein composition, enzymatic activities, and structural features. Given the increasing recognition of electronegative LDL as a potential biomarker and therapeutic target for the prevention of cardiovascular disease, key future goals include the development of a standard method for the detection of electronegative LDL that can be used in a large-scale population survey and the identification and testing of strategies for eliminating electronegative LDL from the blood.

The structure and the composition of the wheat (Triticum aestivum L.) grain are heterogeneous. In order to study the composition of grain compartments, we developed a method to easily isolate a relative large amount of endosperms and embryos. The analysis of proteins were made as an example of compartimentation and heterogeneity of grain composition. Proteins were sequentially extracted and analysed by RP-HPLC. Gliadin and glutenin are specific to the endosperm, whereas some albumins-globulins are found in the endosperm, some in the embryo and others are common in both sites. The quantitative contribution of embryo proteins to the whole grain proteins however was not significant because of the low proportion of embryo weight to whole grain dry matter. Thus for proteins, the analyses of flour could be considered as equivalent as the analysis of the whole grain.

Studies undertaken with alpha-lactalbumin-enriched formulae never addressed infants with colic. This study evaluated the nutritional adequacy, the gastrointestinal tolerance and the effect on colic of an alpha-lactalbumin-enriched and probiotic-supplemented formula. A double-blind, placebo-controlled study enrolled 66 healthy infants with colic, aged 3 weeks to 3 months, fed during 1 month with the either experimental formula (EF, Modilac Digest 1) or control formula (CF) and evaluated for efficacy and safety parameters at days 15 and 30. Weight and height gains were identical in the two groups and complied with standards (1023.4+/-360.4 g (EF) and 1047.4+/-372.1 g (CF), NS; 4.2+/-1.4 cm (EF) and 4.3+/-1.9 cm (CF), NS). No differences were found between groups for crying duration. 'Feeding-related' gastrointestinal side effects were significantly lower with EF than with CF (P=0.011). An alpha-lactalbumin-enriched and probiotic-supplemented formula guaranteed good weight and length gains to infants with colic and seemed to provide good gastrointestinal tolerance.

Proteomics is a modern, rapidly developing branch of biology aimed at the integral study of genome expression as the diversity of proteins. Proteomics is based on a variety of methods allowing one to isolate, detect, identify, and to analyze quantitatively thousands of proteins comprising the cellular proteome. Up to date, high-resolution two-dimensional electrophoresis is the key proteomic technology; it allows one to analyze simultaneously the protein composition of cells and tissues and to reveal quantitative and qualitative changes in the synthesis of proteins modulating multiple cell functions. The present status of proteomic technologies, i.e., of 2D electrophoresis, with accompanying methods of identification of proteins at the maps obtained, as well as a brief description of organization of human mouse 2D database suggested for global analysis of genome expression in normal and pathological conditions (http:/biobase.dk/cgi-bin/celis) are discussed.

The effect of retardants on cellulose biosynthesis and protein composition in seedlings and fiber of two cotton species is investigated. The activity of glucansynthetase and the synthesis of “de novo” polypeptides increase under the influence of morphonol and Pix.

The enzymes and proteins of the fibers of two lines of cotton plant differing in the strength of the fiber have been investigated. It has been shown that the activities of glucan synthetase and peroxidase rise as the fiber matures, while the activities of β-(1-3)-glucanase and cellulase fall. The specific enzymatic activities of peroxidase and glucan synthetase in the L-175 line, distinguished by a stronger fiber, are higher than for the L-466 line with a weaker fiber. The activity of glucanase changes according to the strength of the fiber. In a study of the protein composition of cotton fibers, polypeptides with molecular masses of 28 and 39 kDa were found among the proteins responsible for the strength of the fiber.

Synthetic surfactant peptides offer an opportunity to standardize the protein composition of surfactant. We tested the effect of phospholipids (PL) with synthetic full-length SP-B1-78 (B), mutant B (Bser), KL4 peptide (UCLA-KL4), and palmitoylated SP-C1-35 (C) on oxygenation and lung function in a surfactant-deficient rat model. Sixty-four adult rats were ventilated with 100% oxygen, a tidal volume of 7.5 mL/kg, and a rate of 60/min. Their lungs were lavaged with saline until the arterial PO2 dropped below 80 torr, when 100 mg/kg surfactant was instilled. Surfactant preparations included: PL (PL surfactant), PL + 3% B (B surfactant), PL + 3% B and 1% C (BC surfactant), PL + 3% UCLA-KL4 (KL4 surfactant), PL + 3% Bser (Bser surfactant), and PL + 3% B and 1% UCLA-KL4 (BKL4 surfactant). Sixty minutes after surfactant instillation, positive end-expiratory pressure was applied for 5 min, and pressure-volume curves were determined in situ. The six surfactant preparations had a minimum surface tensions10 mN/m on a Langmuir/Wilhelmy balance. Instillation of PL, Bser, and BKL4 surfactant increased mean arterial/alveolar PO2 (aADO2) ratios by 50-100% over postlavage values, whereas KL4 surfactant increased aADO2 ratios by 118%, B surfactant by 191%, and BC surfactant by 225%. Lung volumes at 30 cm H2O pressure were highest after treatment with BC surfactant, intermediate after B and KL4 surfactants, and lowest after BKL4, Bser, and PL surfactants. These data suggest that a surfactant preparation with a combination of synthetic B and C peptides surpasses synthetic B and KL4 surfactants in improving oxygenation and lung compliance in surfactant-deficient rats.

Arcelin seed proteins of common bean (Phaseolus vulgaris L.) confer resistance to bruchid pests, and in vitro results suggested that greater resistance could be achieved by increasing the concentration of arcelin. We created backcross lines having arcelin alleles (SMARC lines), or alleles for the related protein phytohemagglutinin (PHA) (SMPHA lines), and a null allele for phaseolin to determine if seeds lacking phaseolin would contain increased quantities of arcelin or PHA proteins. To test the affects of genetically removing phaseolin, SMARC and SMPHA lines were derived as pairs of phaseolin-containing and phaseolin-null lines. Parental, SMARC, and SMPHA lines were grown in a replicated greenhouse trial and measured for days-to-flower, days-to-maturity, seed weight, and for quantities of phaseolin, arcelin dimer, PHA, and total proteins. There were no differences between pairs of phaseolin and phaseolin-null lines for days-to-flower, seed weight or total protein, and inconsistent differences for days-to-maturity. Arcelin concentrations were significantly increased in two of four pairs of SMARC lines, and PHA concentration was significantly greater in four of five pairs of SMPHA lines. These or other changes in the seed protein composition in phaseolin null lines may improve resistance to bruchids.

Localization and protein composition of plastid nucleoids was analyzed in light-grown pea seedlings at various stages of leaf development. In young plastids of unopened leaf buds, nucleoids were abundant and localized in the periphery of plastids, whereas, in mature leaves, chloroplasts contained nucleoids within narrow spaces restricted by thylakoids or grana. The migration of nucleoids into the interior of plastids preceded the formation of grana, and hence, the maturation of the photosynthetic apparatus. The protein composition of nucleoids was considerably different in young plastids and mature chloroplasts. Polypeptides with a molecular mass of 70–100 kDa predominated in the nucleoids of young plastids, whereas polypeptides with molecular mass of 20–30 kDa were abundant in the nucleoids of mature chloroplasts. Immuno-blot analysis with antibodies against the nucleoids of young plastids identified various polypeptides that were significantly more abundant in the nucleoids of young plastids than in the nucleoids of mature chloroplasts. These results demonstrate that plastid nucleoids are subject to dynamic changes in both localization and composition during the normal development of chloroplasts in the light.

The total protein content amounts to 60% of the defatted flour or 14-15% of the weight of watermelon seeds. Water-, salt-, alcohol-, and alkali-soluble fractions of the proteins have been isolated and their percentage amounts in the seeds have been determined. More than half the total protein (53.3 %) consists of globulins - the reserve proteins of the seeds. The seeds contain about 30% of lipids, which form a valuable component of them and may serve as an additional source of nutrient plant oil. Using the method of amino acid scales a comparative analysis has been made of the biological value of the watermelon seed proteins with standard proteins.

Breast secretions can be classified into two types according to their major protein components. Type I fluids contain Zn-alpha 2-glycoprotein, apolipoprotein D, and gross cystic disease fluid protein-15, while Type II fluids are characterized by the presence of lactoferrin, lysozyme, and alpha-lactalbumin. In this study, the polypeptide composition of breast secretions from 719 nonlactating women was evaluated by using polyacrylamide gel electrophoresis. The required amount for the analysis (1 microliter) was obtained from 50% of control women and from 75% of women with mammary disease. There were more secretors in premenopausal than in postmenopausal women, as well as in parous than in nulliparous women. Evaluation of factors affecting protein composition of breast secretions revealed that Type II fluids were found in the majority of women who had given birth in the last four years and in a high proportion of oral contraceptive users. After excluding both of these groups, Type II fluids were detected in 47% of patients with breast cancer, but only in 8% of control women and in 16% of women with benign breast diseases. Taken together, these results suggest that protein analysis of breast secretions could be an useful tool for the study of breast pathologies.

Trypticase soy broth and Mueller Hinton broth lacking yeast extract resulted in morphological alterations (pleomorphism) ofCardiobacterium hominis, as demonstrated by light and transmission electron microscopy. Outer membrane protein profiles were influenced by the composition of growth media, especially in the region of porin proteins. In contrast, the protein composition of inner membranes was not influenced by media composition.C. hominis isolates tested were susceptible to 16 β-lactam antibiotics. All isolates exhibited minimal inhibitory concentrations of ≤1 µg/ml to 13 of the 16 β-lactams tested. The changes in outer membrane profiles induced by media composition did not influence susceptibility to β-lactam antibiotics. None of the isolates examined contained detectable plasmids, β-lactamase, or resistant mutants. Characteristic β-lactam-induced morphological defects were observed.

The protein composition and the kinetics of the quantitative changes in the various protein fractions during the ontogenesis of protoplast and hybrid cultures ofTrichoderma harzianum have been studied. In the exponential growth phases of both cultures the highest amount of protein was contained in the water-soluble (33.0–33.2%) and salt-soluble (31.0–32.0%) fractions. The protein contents of the water-soluble fractions of the cultures studied were similar to those in meat and clover and were 3.5 times greater than in yeast.

The authors have studied 122 medical histories of extrauterine pregnancies from archives and directly followed up 122 patients who underwent emergency surgery for extrauterine pregnancy. Particular disturbances of rheological properties of blood in patients with ectopic pregnancy are revealed. The authors prove the necessity of rheologically active infusion-transfusion therapy, which normalizes rheological properties of blood, its acidic and basic conditions, and protein composition both during the operation and in the postoperational period. The following solutions were used: acesol, Haemodesum, rheopolyglucine, rheoglumane, gelatinol, 10% albumin solution, and fresh frozen plasma.
