In this study, it was aimed to develop a simple, accurate, and sensitive analytical strategy for the determination of nitrite in processed meat products using a solid optical chemosensor and digital imaging system. The method is simply based on formation of azo dyes obtained by mixing the powdered 1-naphthy lamine, N, N-dimethyl aniline, and nitrite solution under acidic conditions and the colorimetric analysis of the color formed on glass microfiber filter paper by a digital-based imaging system. All the system parameters in color formation and detection steps were optimized to lower the detection limit. In the optimal experimental conditions, detection limit, percent relative standard deviation, and R2 values were determined as 0.64 mg/L, 0.64%, and 0.994, respectively. Recovery results (99.2–121.6%) were calculated for different sausage and salami samples spiked at two different concentrations. Unlike the standard azo dyestuff–based methods, the method is environmentally friendly, fast, simple, and cheap since organic solvents are not used during the analysis.

In this research, simultaneous quantification of cysteine (Cys) and histidine (His) using a paper-based sensor was investigated for the first time. A microfluidic paper-based sensor is a promising amino acid determination tool due to its low cost, low sample consumption, and fast analysis and is easy to make. We used SBA-15 as porous materials that are crystalline compounds formed with surfactants and TEOS as a silica source. The SBA-15 sample was characterized by X-ray diffraction, Fourier transform infrared, and scanning electron microscope techniques. The microfluidic paper-based sensors were fabricated using a wax pen, and the mesoporous SBA-15 modified filter paper. The fabricated sensors for Cys and His determination are operated based on an indicator-displacement assay. A comparative determination study of Cys and His on the sensors was carried out. The results illustrated that the addition of silica nanoporous material led to an immediate and uniform color change. The sensors were successfully exploited in the simultaneous determination of urinary Cys and His levels, thus providing the potential opportunity for clinical diagnosis. The linear range of 1.0 to 90.0 μM and 1.0 to 100 μM for His and Cys was obtained from the calibration data. The detection limits were also calculated (S/N = 3) for His and Cys of 0.5 μM and 1.5 μM, respectively. The proposed method showed a good agreement with the results achieved by a standard method.

Phosphatidylethanol (PEth), which is formed by enzymatic reaction between ethanol and phosphatidylcholine, is a direct marker for alcohol usage. PEth has a long elimination half-life (~5-10 d) and specimens can be sampled using minimally invasive microsampling strategies. In combination with rapid analysis procedures PEth has proved to be advantageous for the detection of abstinence over other direct (e.g., ethyl glucuronide in blood, urine or hair) and indirect (e.g., carbohydrate-deficient transferrin in serum) alcohol markers. Although PEth determination is widely applied around the world, laboratory protocols are not standardized. Here we provide general guidelines for the analysis of PEth in dried blood spots (DBSs), including reference material evaluation, synthesis of a deuterated internal standard, preparation of calibration samples (reference material in teetotaller blood), and analyte separation and detection. The protocol contains information to extract the DBSs either manually or with a fully automated autosampler. Extraction of the analytes from DBS filter paper cards is performed using an organic extraction, followed by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). For accurate and reliable measurement of PEth, the two most abundant analogs, PEth 16:0/18:1 and PEth 16:0/18:2, are quantified. We show data that provide guidelines on how to interpret the results for both demographic studies and forensic applications. Body surface mucus was scraped off from white croaker Pennahia argentata after washing with two different procedures, suspended in cold water and filtered through a filter paper. The filtrate thus obtained was lyophilized and defined as the fish body surface substances. The relative level of the substances obtained after high-pressure water washing was 28.8% and significantly lower than those obtained after conventional washing (68.8%) and without washing (100%). The breaking strength of thermally induced gels prepared by pre-heating at 65 °C mimicking modori for 60 min after the high-pressure water washing was 3.8 N and was significantly higher than that after the conventional washing (2.6 N) and without washing (2.1 N). These results suggest that rheological properties of thermally induced gels can be improved with an extensive washing of fish material. The degradation of myosin heavy chain was recognized, when the water soluble fraction of the lyophilized powder of the fish body surface mucus was added into the ordinary muscle homogenate at around 60 °C and pH 8.15, suggesting that proteases were present in the fish body surface mucus. Such novel findings are useful for avoiding the modori phenomenon and thus improving the quality of surimi-based products.

The present work describes the development and validation of a novel approach to determine methadone (MTD) and its main metabolite (EDDP) in oral fluid samples, using the dried saliva spots (DSS) sampling approach and gas chromatography-tandem mass spectrometry (GC-MS/MS). Oral fluid samples (50 μL) were applied into Whatman™ 903 protein saver filter paper cards and were allowed to dry overnight. The extraction was carried out by immersion of the spot in 1 mL of isopropyl alcohol with agitation for 1 min. Afterwards, the extract was centrifuged for 15 min at 3500 rpm and the supernatant evaporated to dryness and reconstituted with 50 μL of methanol. The procedure was considered linear in the range of 10 to 250 ng/mL for both compounds, with determination coefficients greater than 0.99. Intra- and inter-day precision and accuracy revealed coefficients of variation (CVs) lower than 15% at the studied concentrations, with mean relative errors within ± 15% of the nominal concentrations. Recoveries ranged from 45 to 74%. The limits of detection and quantification were 5 and 10 ng/mL respectively for both analytes. All studied parameters complied with the defined criteria and the method enabled the successful determination of MTD and EDDP in oral fluid samples from patients undergoing opiate substitution/maintenance therapy.

Cellulose nanocrystals (CNCs) have gained increased interest worldwide for their unique properties. CNC production by acid hydrolysis of cellulose-rich biomass is well established, but its' quantification is still complicated. In this study, a rapid method for the determination of CNC concentration using Calcofluor White (CW) fluorescence dye is demonstrated for both purified and homemade CNCs recovered from Whatman filter paper. The method is robust, selective for crystalline cellulose, suitable for routine measurement in CNC production and that the pH must be basic for staining using CW.

The objective of this work was to compare the quality, purity and quantity of DNA isolated from dried blood spots (DBS) by three methods (Chelex-100, QIAamp DNA mini kit, and TE (Tris EDTA)-Buffer). Sample collection was performed in six districts in Odisha, India and screened for cases of clinical malaria and dengue and vector density. Mosquito abdomens were spotted on Whatman 3MM (MERCK) Filter paper and dried for 10 min at room temperature. DNA was isolated from DBS using three methods (Chelex-100, QIAamp DNA mini kit, and TE-Buffer), and PCR was used to determine the feeding behaviours of vector mosquitoes. DNA was quantified using a UV-spectrophotometer, and q-PCR was used to determine the target gene copy number to compare the methods. The QIAamp DNA mini kit method was used as the reference method. The yield and purity of DNA extracted with Chelex-100 and TE were 14–72 ng/µl and 1.51–1.85 and 9–50 ng/µl and 1.68–2.1, respectively. DNA extracted using the Chelex-100 method was stored for over 1 month at − 20 °C and was suitable for later use. The Chelex-100 method had a sensitivity of 99.5% and specificity of 78%. A Bland–Altman plot suggested that the Chelex-100 method was similar to the QIAamp DNA mini kit method for determining the feeding behaviours of vector mosquitoes. The Chelex-100 method is simple, cost-effective, and safe and requires minimal time for DNA extraction from dried blood spots. In malaria and dengue research, detecting the feeding behaviours from mosquito DNA from dried blood spots on filter paper by PCR is an easy, minimally invasive and inexpensive molecular technique that can be performed in remote areas.

Purpose In this study, an aqueous two-phase micellar system (ATPMS), formed by the non-ionic surfactant Triton X-114, was used to investigate the partitioning of cellulolytic enzymes produced by the filamentous fungus Aspergillus fumigatus CCT 7873. Methods Performance of the ATPMS on the partitioning of CMCase (activity on carboxymethyl cellulose) and FPase (activity on filter paper) was investigated by varying the temperature (35, 40, 45, 50, 55, 60, and 65 °C), enzyme crude extract concentration (20, 40, 60, and 80% w/w), and Triton X-114 concentration (2, 4, 6, and 8% w/w) and by adding different inorganic salts (NaCl, CaCl2, MgSO4, and MnSO4) in the system. Results An ATPMS formed with 8% (w/w) Triton X-114 and 40% (w/w) enzymatic crude extract at a system temperature of 55 °C was most favorable for partitioning the tested enzymes. Under these conditions, a purification factor for CMCase and FPase of 10.89 and 0.65 was reached, respectively. The addition of inorganic salts changed the distribution of enzymes. Of these, CaCl2 contributed to a higher distribution coefficient (50.0), whereas for FPase, the presence of MnSO4 in the system improved the purification factor to 3.94. Conclusion The highest values obtained for the yield and purification factors demonstrate that ATPMS is an interesting option for recovering and purifying cellulolytic enzymes.

Glucose is a typical diagnostic marker of diabetes mellitus but it may be also helpful during diagnosis of some types of poisoning, immunological disorder or long-term stress at hospitalized patients. Diagnosis of diabetes mellitus in time is indispensable for prevention of life-threatening complication and so, accurate and easy device for non-invasive and accurate self-diagnosis is necessary. Simple assay based on filter paper with chitosan layer and with immobilized enzymes glucose oxidase and peroxidase and phone camera detection has been proposed for home-care glucose detection in urine. Data were processed in RGB (red–green–blue) color model when blue color channel showed the best sensitivity to changing glucose concentrations. Assay exerted linear regression from 0 to 4 mmol/l and limit of detection was set to be 166 µmol/l. Assay was not influenced by any interfering substances including maltose, sucrose, sorbitol or fructose or by urine matrix substance such as bovine serum albumin, ascorbic acid, uric acid or urea. Measurement of glucose from spiked urine samples reveals also no influence of the matrix on the assay. Long-term stability was measured and filter papers coated by enzymes showed excellent stability for at least 9 weeks without any special treatment or package. In comparison with differently modified filter paper, filter paper with both chitosan and enzymes showed the best sensitivity to changing glucose concentration. In a conclusion, simple (phone camera detection) and fast (3-min reaction) assay for quantitative detection of glucose from urine was optimized.

A rapid and accurate detection of pathogens is essential for bedside or on-site diagnosis. Filter paper is an ideal diagnostic tool as it requires no equipment, possesses a high surface-area-to-volume ratio and a high capacity of capillary force. The functionalization of the surface of cellulose filter paper was explored by using glutaric anhydride, N-hydroxysuccinimide, and N,N'-dicyclohexylcarbodiimide. The activated surface systems enable aminated DNA to be immobilized on the surface of filter paper. Both synthetic oligonucleotides and bacterial genomic DNA of Staphylococcus aureus, Escherichia coli, and Campylobacter jejuni were detected successfully. The system produces a clear, consistent and highly visible brown signal within 1-5 min. The digital image can also be analyzed quantitatively due to the brown color resulting from the presence of magnetic beads. Bacterial DNA detection was accomplished by using 16S rDNA probe on the activated paper surface for universal bacterial diagnosis. The method is stable and repeatable. It can detect at least 0.5 pmol of a 120-base synthetic oligonucleotide per assay and 5-10 ng of bacterial DNA per assay. A novel and simple method utilizing a chromogenic reaction on filter paper is introduced for the rapid screening of banned aromatic amines released from azo dyes. The proposed method involves the sample preparation protocols outlined by the current standard method and the chromogenic reaction of extracted aromatic amines on filter paper. Based on the principle of the reaction between primary amines and aldehydes, p-dimethylaminobenzaldehyde (DMAB) was used as the chromogenic reagent for the rapid screening of 24 carcinogenic aromatic amines and aniline without any chromatographic instruments under optimized experimental conditions. The detection limit for all the aromatic amines in this study was less than 15 mg/kg. A total of 727 dyed textile samples were analyzed using both the present standard method and the proposed method simultaneously. Using the proposed method, a total of 471 samples did not require further instrumental analysis, which can dramatically save instrumental detection time (61.2%), can decrease instrumental detection costs, and can avoid the use of large amounts of toxic reagents. The proposed method has been applied to detect banned aromatic amines in some inspection institutions and dye factories and has large social and economic benefits. The authors describe a filter paper modified with mesoporous silica that carries phenylboronic acid on its surface. The material possesses the advantages of the flexibility of filter paper and the large surface of mesoporous silica. Bisphenol A (BPA) is a typical endocrine disruptor. It exists everywhere in the environment for its migration from the products of polycarbonate and epoxy resin. It is feasible to develop a fast and sensitive detection method for the effective monitoring of BPA. In this paper, we utilized the advantages of Ag nanoparticle-modified filter paper and cysteamine hydrochloride (Cys) to establish a surface-enhanced Raman spectroscopy (SERS) detection method of BPA. Good uniformity and more “hot spots” can be afforded with the SERS substrate fabricated by vacuum filtration of Ag nanoparticles on filter papers. Cys was used to catch BPA by the electrostatic interaction between positively charged groups of −NH3 + and hydroxy of BPA. Then, BPA-tailed Cys self-assembled to the surface of SERS substrate. Due to the preconcentration of BPA and high coverage of BPA-tailed Cys on the Ag nanoparticle-decorated filter paper, a highly sensitive detection of BPA in water samples was achieved. The method exhibited a good linear correlation ranging from 0.05 to 20 ng/mL with a limit detection of 0.005 ng/mL. Excellent recoveries in water samples from 90.2 to 121.1% were obtained.

A nematicidal fungus strain was isolated and identified as a novel member of the genus Arthrobotrys based on the morphologic and phylogenetic analysis and designed as CX1. Strain CX1 could make the filter paper translucent, resulting in an increasing hydrature index by 73.7 % and a reducing crystalline index by 32 % in comparison with the untreated filter paper. Scanning electron microscopy showed swollen microfibrils in the presence of water, and Fourier transformed infrared spectroscopy indicated the change in absorbance and/or wavenumber for hydrogen bonds. The cellulose degradability was increased by 3.5 times when the filter paper was treated with Arthrobotrys sp. CX1 but no cellulase activities could be detected in the culture supernatant of strain CX1. The isolate CX1 could gelatinize the filter paper cellulose causing decrystallization with high water imbibition. It is significant to apply it to the initial cellulose deconstruction to facilitate the cellulose saccharification.

Background The discovery of bioactive compounds in traditional Chinese medicine (TCM) has become an important field in TCM modernization. Ligand fishing is a suitable method for discovery of bioactive compounds in complex mixtures such as TCM with high selectivity. Because of unique advantage of low cost and convenience, paper-based microdevices can be good carriers for enzyme immobilized ligand fishing. Methods As an important enzyme for glucose metabolism, α-glucosidase was immobilized on polycaprolactone–chitosan-modified paper to prepare the microdevice with unique microfluid structure generated by 3D printing technology, which can be easily applied to screen active compounds in herbal extracts. The preparation conditions of the paper microarray were optimized. The activity of immobilized α-glucosidase was verified by colorimetric reactions which can be easily monitored by cellphone. The paper microarray with α-glucosidase immobilized was used to screen active compounds in the water extracts of mulberry leaves and lotus leaves. Results Several key parameters including Na2CO3 solution concentration, Na2CO3 solution volume, glutaraldehyde concentration, crosslinking time of glutaraldehyde and time of α-glucosidase immobilization were optimized. The proposed paper-based microarray was successfully applied in screening active compounds in two herbal extracts. Four compounds including chlorogenic acid, quercetin-3-O-glucuronide, isoquercetin, and quercetin were identified as α-glucosidase inhibitors. The compounds with significant non-specific adsorption caused by chitosan, such as isoquercitrin, astragalin, quercetin, were also found to be active compounds. Conclusions An enzyme immobilized paper microarray was designed and fabricated in this work. Polycaprolactone and chitosan were used to modify filter paper to prepare paper microarrays. Parameters of paper device preparation were optimized. Our findings suggested that 3D-printing paper-based microarrays can be a simple and low-cost approach for discovery of active compounds of TCM. Point-of-care testing (POCT) for uropathogen detection and chemical screening has great benefits for the diagnosis of urinary tract infections (UTIs). The goal of this study was to develop a portable and inexpensive paper-based analytical device (PAD) for cultivating bacteria in situ and rapidly testing for nitrite on the same device. The PAD was fabricated using a wax printing technique to create a pattern on Whatman No. 1 filter paper, which was then combined with a cotton sheet to support bacterial growth. Nitrite detection was based on the principle of the Griess reaction, and a linear detection range of 0–1.6 mg/dL (R2 = 0.989) was obtained. Scanning electron microscopy (SEM) analysis demonstrated that the bacteria were able to grow and formed a cluster on the cellulose fibres within 2 hours. The enzyme β-glucuronidase, which is specifically produced by Escherichia coli, was able to convert the pre-immobilized 5-bromo-4-chloro-3-indolyl-β-D-glucuronide sodium salt (X-GlcA), a colourless substrate, generating a blue colour. Under optimum conditions, the proposed device allowed bacterial concentrations in the range of 104–107 colony forming units (CFU)/mL to be quantified within 6 hours. Moreover, the use of this device enables the identification of E. coli pathogens with selectivity in real urine samples. In conclusion, the PAD developed in this study for UTI screening provides a rapid, cost-effective diagnostic method for use in remote areas.

A new paper platform was developed for the colorimetric detection of creatinine. The filter paper was coated with 3-propylsulfonic acid trimethoxysilane and used as the platform. Creatinine in a cationic form was extracted onto the paper via an ion-exchange mechanism and detected through the Jaffé reaction, resulting in a yellow-orange color complex. The color change on the paper could be observed visually, and the quantitative detection of creatinine was achieved through monitoring the color intensity change. The color intensity of creatinine complexes on the paper platform as a function of the creatinine concentration provided a linear range for creatinine detection in the range of 10 - 60 mg L(-1) and a detection limit of 4.2 mg L(-1). The accuracy of the proposed paper-based method was comparable to the conventional standard Jaffé method. This paper platform could be applied for simple and rapid detection of creatinine in human urine samples with a low consumption of reagent.

An effective wipe sampling and LC–MS/MS method was developed to simultaneously analyze six commonly administered antineoplastic drugs in stainless steel surface. The analyzed drugs were methotrexate, paclitaxel, cyclophosphamide, 5-fluorouracil, vincristine, and oxaliplatin, a frequently prepared antineoplastic drug that has not been included among any of the published simultaneous detection methods. The established method was used to evaluate the recoveries of antineoplastic drugs on brand new and worn stainless steel surfaces by wiping the plates with a Whatman filter paper wetted with 0.5 mL of water/methanol (20:80) with 0.1 % formic acid followed by LC–MS/MS before desorbing the filter with a water/methanol (50:50) solution. A significant decrease in the recovery of all evaluated drugs was found when worn plates were used. Additionally, the inter-personnel variability on drug recoveries during wiping procedures was evaluated. Significantly higher recoveries were achieved by the personnel with more training and experience versus personnel without prior experience. Finally, a laboratory stability test was developed to assess the degradation of the antineoplastic drugs during replicated shipping conditions. Artificial materials with controllable wettability are of global interests in oil–water related applications. In the present report, a surface with superhydrophilicity/superoleophilicity and underwater superoleophobicity was constructed on filter paper via dip-coating ZnO colloids. However, the wettability of the filter paper was converted to be superhydrophobic and superoleophilic after being modified by octyltrimethoxysilane, and water contact angle on the surface is larger than 150° while oil contact angle is smaller than 5°. The filter paper can be employed to selectively adsorb oil floating on water or separate water–oil mixtures via filtration process because of its superhydrophobicity and superoleophilicity. Additionally, the filter paper has also the capacity to hold water, which is expected to be used as a material for manufacturing labwares. A filter paper with superhydrophobicity and superoleophilicity was fabricated by dip-coating and subsequent modification. Water contact angle and sliding angle on the filter paper are 157°and 6°, respectively. The filter paper with superhydrophobicity and superoleophilicity could be employed to separate oil–water mixture with high efficiency.

By using a silver nanoparticle wiper as a surface-enhanced Raman scattering substrate, a highly sensitive, convenient, and rapid platform for detecting dye adulteration of medicinal herbs was obtained. Commercially available filter paper was functionalized with silver nanoparticles to transform it into the flexible wiper. This device was found to collect dye molecules with unprecedented ease. Experiments were performed to optimize various factors such as the type of wiper used, the wetting reagent, and the wetting/wiping mode and time. Excellent wiper performance was observed in the detection of the simulated adulteration of samples with dyes at various concentrations. The limits of detection for nine dyes, including 10(-6) g/mL for malachite green, 10(-7) g/mL for Rhodamine 6G, and 5 × 10(-8) g/mL for methylene blue, were discerned. The results of this investigation show that this proposed method is potentially highly advantageous for field-based applications. Dried blood spot (DBS) sampling, a technique used for taking whole blood samples dried on a filter paper, was initially reported in 1963 by Robert Guthrie. While the diagnostic analysis of metabolic disorders in newborns was the focus of investigations at that time, the number of established applications for preclinical drug development, toxicological studies, and therapeutic drug monitoring increased enormously in the last decades. As a consequence of speed, simplicity, and minimal invasiveness, DBS recommends itself as the preferential technique in sports drug testing. The present approach highlights for the first time the development of a screening assay for the analysis of the synthetic human adrenocorticotropic hormone tetracosactide hexaacetate (Synacthen(®)) in DBS using liquid chromatography tandem mass spectrometry. Highly purified sample extracts were obtained by an advanced sample preparation procedure including the addition of an internal standard (d8-tetracosactide) and immunoaffinity purification. The method's overall recovery was 27.6 %, and the assay's imprecision was calculated between 8.1 and 17.9 % for intraday and 12.9 to 20.5 % for interday measurements. Stability of the synthetic peptide in DBS was shown for at least 10 days at room temperature and presents a major benefit, since a rapid degradation in conventionally applied matrices such as urine or plasma is well known. With a limit of detection of 50 pg/mL, a detection window of several hours is expected considering reported steady-state plasma levels of 300 pg/mL after intramuscular application of Synacthen(®) Depot (1 mg). The analysis of authentic DBS samples within the scope of an administration study with 250 μg Synacthen(®) (short stimulation test) demonstrated the great potential of the developed assay to simplify the analysis of Synacthen(®) for doping control purposes.

The development of a convenient method for oil-removal is of great significance for environmental protection. Here, we present a simple method for the removal of oils from water surface based on sponges that we fabricated by solution-immersion processes. The sponges exhibited high selectivity and absorption capacities for various kinds of oils when they were employed as absorptive materials. More importantly, the superhydrophobic sponge could be sustained 400 cycles of compressing test without losing their superhydrophobicity, exhibiting the high elasticity, robustness, and durability. To extend application field, superhydrophobic filter paper was used for oil–water separation. Interestingly, tunable wettability was received when oleophobic silica was employed instead of hydrophobic silica. We expected that this low-cost process can be used for oil-spill cleanup.

We evaluated whether a fingerprint, which consists of secretions from the fingertips, is a suitable biological sample for drug testing. A commercially available cold medicine containing ibuprofen, dihydrocodeine, chlorpheniramine, and methylephedrine was administered to healthy subjects. The subjects washed their hands with tap water and hand soap to remove the external contaminants, and then pressed a fingertip onto wet filter paper at fixed sampling times (from 2 h to 7 days). The analytes on the filter paper were dissolved in 25 % methanol–water, and a large volume (50 μl) of the extract was analyzed by liquid chromatography–tandem mass spectrometry. The relationship between the sampling times and the concentration of analytes in fingerprints was examined. The results were compared with drug concentrations in blood samples. Most of the drugs and their metabolites were detected from fingerprints at 7 days after drug administration. The fingerprint sample preparation is rapid (ca. 3 min) and simple, and the limits of detection were 0.1 pg/fingerprint for dihydrocodeine, chlorpheniramine, and methylephedrine. We demonstrate that drugs can be detected in fingerprints at later sampling times with more rapid and simpler sample preparation than in blood. The method should be applicable to drug testing in criminal investigations.

Primaquine (PQ) is the only 8-aminoquinoline antimalarial drug in clinical use because of its unique action on hypnozoites and gametocytes of Plasmodium species. We report here simple, sensitive and specific assay methods for the determination of PQ in human whole blood and dried blood spot (DBS) samples using high-performance liquid chromatography and liquid chromatography-mass spectrometry, respectively. Sample preparation was performed by a single or two-step liquid-liquid extraction with organic solvents. For whole blood analysis, separation was obtained on a reversed-phase C18 column with the mobile phase consisting of 0.25 % diethylamine and acetonitrile (7:3, v:v) running at a flow rate of 1.0 ml min−1. UV detection was at the wavelength 263 nm. For DBS analysis, separation was obtained on a reversed-phase column with the mobile phase consisting of methanol and 0.1 formic acid (1:3, v:v) running at a flow rate of 0.5 ml min−1. The selected ions generated by electrospray ionization were detected using mass spectrometer. Good precision and accuracy (both within-day and day-to-day assays) were obtained at the concentration ranges under investigation. Limits of quantification for PQ were accepted as 25 ng ml−1 using 500 μl whole blood and 5 ng ml−1 using 80 μl DBS samples. The mean recoveries for PQ and internal standard pyrimethamine (PYR) for both whole blood and DBS were over 70 %. The methods were successfully applied for a clinical pharmacology study of PQ in patients with Plasmodium vivax. Excellent correlation (r 2 = 0.997) was observed between the analysis of PQ in paired whole blood and DBS samples.

Herein we present a novel approach for the quantification of active pharmaceutical ingredients (APIs) using mass spectrometry imaging. This strategy uses a filter paper previously “eluted” with a MALDI matrix solution as a support for analyte application. Samples are submitted to mass spectrometry imaging (MSI) and quantification through characteristic fingerprints is ultimately performed. Results for the content of rosuvastatin from a known formulation are comparable to those obtained with a validated HPLC method.

A method has been developed that integrates filters directly into centrifugal microfluidic devices. This technique is suitable for both rapid prototyping and commercial applications. Commercially available filter paper was sealed into the centrifugal microfluidic device with a simple manual fabrication procedure. The method was validated using soil slurry in water and a variety of filter papers with pore sizes ranging from 0.7 to 11 μm. Filtration times of 4 s to several minutes were obtained for 100 μL samples depending on the type of filter paper and rotation rate utilized. The validity of the method was demonstrated by assessing the amount of light lost due to the scatter or absorption caused by particles in the filtered sample while the device was in motion. Filtration and sedimentation were compared and after 30 min of centrifugation, sedimentation had not removed particles as well as filtration. This technique opens up centrifugal microfluidic devices to a wide range of samples.

Phosphite application mitigates diseases caused by oomycete plant pathogens. Tissue concentrations of phosphite above 1 mM are generally required for disease protection. Determining the concentration of phosphite in plant material requires extensive extraction and derivatisation procedures prior to separation by gas-liquid chromatography (GLC). This paper describes a direct chemical method to estimate the concentration of phosphite using a silver nitrate reagent. Glass fiber filter papers were saturated with a 1 M aqueous solution of silver nitrate (adjusted to pH 2.5 with nitric acid) and dried for 2 h at 60 ° C. 20 μL of polyvinylpolypyrrolidone treated aqueous plant extract was adsorbed onto the filter paper and incubated in the dark at room temperature (25 ° C) for 1 h. The presence of phosphite in the extract reduces the silver ions to elemental silver resulting in a grey-black precipitate that is clearly visible. The method is rapid, sensitive and inexpensive, and can detect phosphite at concentrations of 1 mM in 20 μl of aqueous extract from 100 mg of fresh plant material. Samples analysed by this method gave similar results to analysis by GLC, indicating the method can be used in the field or the laboratory to determine uptake and distribution phosphite in the plant, the retention of phosphite over time and the timing of phosphite reapplication.

The effect of pH on the production of cellulases and xylanases by Penicillium echinulatum S1M29 was evaluated in a shake flask and in a bioreactor. To control the pH in a shake flask, a buffer made with citric acid and disodium phosphate was used. The buffer was capable of maintaining the culture pH values for the first 48 h. In the bioreactor, the pH was controlled automatically by the addition of NaOH and H2SO4. In the shake flask, the highest activities of xylanases (18.5 IU/mL) and endoglucanases (8.2 IU/mL), as well as the highest filter paper activity (FPA) (0.9 IU/mL), were obtained at initial pH values of between 6.0 and 7.0. In the bioreactor, the highest activities of these enzymes were obtained in a pH range of 5.5 to 6.5. Different isoforms of the endoglucanases were found in the various cultures depending on the pH. More acidic pH ranges favored the production of β-glucosidases in both the shake flask and the bioreactor.

Bovine serum albumin (BSA) was added to filter paper during the hydrolysis of cellulase. Adding BSA before the addition of the cellulase enhances enzyme activity in the solution, thereby increasing the conversion rate of cellulose. After 48 h of BSA treatment, the BSA adsorption quantities are 3.3, 4.6, 7.8, 17.2, and 28.3 mg/g substrate, each with different initial BSA concentration treatments at 50 °C; in addition, more cellulase was adsorbed onto the filter paper at 50 °C compared with 35 °C. After 48 h of hydrolysis, the free-enzyme activity could not be measured without the BSA treatment, whereas the remaining activity of the filter paper activity was approximately 41 % when treated with 1.0 mg/mL BSA. Even after 96 h of hydrolysis, 25 % still remained. Meanwhile, after 48 h of incubation without substrate, the remaining enzyme activities were increased 20.7 % (from 43.7 to 52.7 %) and 94.8 % (from 23.3 to 45.5 %) at 35 and 50 °C, respectively. Moreover, the effect of the BSA was more obvious at 35 °C compared with 50 °C. When using 15 filter paper cellulase units per gram substrate cellulase loading at 50 °C, the cellulose conversion was increased from 75 % (without BSA treatment) to ≥90 % when using BSA dosages between 0.1 and 1.5 mg/mL. Overall, these results suggest that there are promising strategies for BSA treatment in the reduction of enzyme requirements during the hydrolysis of cellulose.

The pH within cell wall pores of filter paper fibers and hemp fibers was assessed by Fluorescence Ratio Imaging (FRIM). It was found that the Donnan effect affected the pH measured within the fibers. When the conductivity of the added liquid was low (0.7 mS), pH values were lower within the cell wall than in the bulk solution. This was not the case at high conductivity (22 mS). The occurrence of the Donnan effect allowed the pH values within pores in normal regions of the cell wall to be compared to the pH in regions with misaligned microfibrils (dislocations) when FRIM was carried out in a low conductivity solution. Surprisingly, no pH difference was observed between normal regions and dislocations, suggesting that pore sizes within the two different regions are approximately the same. In another experiment the Donnan effect was shown to have an effect on hydrolysis of hydrothermally pretreated wheat straw only when conducted in a low conductivity solution and only for xylanase, not cellulases. The hydrolysis experiments indicate that under typical conditions where conductivity is high, the Donnan effect does not lower the pH close to the substrate to an extent that affects enzymatic activity during hydrolysis of lignocellulose.

Inorganic elements are responsible for essential bodily functions, such as osmotic regulation, cardiac frequency and contractibility, blood clotting and neuromuscular excitability. The determination of inorganic elements in corporeal fluids such as blood, serum, plasma and urine is used as a monitor for a part or the whole organism; their values, then, are compared with reference interval values. In this study, the energy dispersive X-ray fluorescence spectrometry (EDXRF), applying the Fundamental Parameters method, for the determination of inorganic elements in whole blood samples from humans and laboratory animals, was used. Peripheral blood samples were collected and, before coagulation, 100 μL of sample were deposited onto Whatman No. 41 filter paper and dried, using infrared spotlight. The reference interval values for healthy Brazilian population of Na were found to be 1,788–1,826 μg g−1, of Mg 63–75 μg g−1, of P 602–676 μg g−1, of S 1,519–1,718 μg g−1, of Cl 2,743–2,867 μg g−1, of K 1,508–1,630 μg g−1, of Ca 214–228 μg g−1, of Fe 170–184 μg g−1, of Cu 4–6 μg g−1 and of Zn 1–3 μg g−1. The reference interval values for golden hamster (Mesocricetus auratus) of Na were found to be 1,714–1,819 μg g−1, Mg 51–79 μg g−1, P 970–1,080 μg g−1, S 1,231–1,739 μg g−1, Cl 2,775–2,865 μg g−1, of K 1,968–2,248 μg g−1, of Ca 209–257 μg g−1, of Fe 145–267 μg g−1, of Cu 4–6 μg g−1 and of Zn 3–5 μg g−1. A comparative study between EDXRF and instrumental neutron activation analysis data was carried out and the results for both techniques are statistically equal (α = 0.05). The results contribute for the establishment of reference interval values for Na, Mg, P, S, Cl, K, Ca, Cu and Zn in the healthy Brazilian population and the referred laboratory animal species.

For the conservation of artworks on paper, powdery paint layers on the paper matrix are stabilized with dilute solutions of adhesives (0.25–1 %), commonly gelatin or methylcellulose, which are applied as aerosols. This technique allows non-contact application. The distribution of the adhesives must be carefully controlled: they have to be delivered to unstable paint layers in the right quantity to avoid visual alterations of the artwork during the stabilization treatment. To visualize the distribution of aerosol-misted adhesives in porous substrates, gelatin and methylcellulose were labeled with fluorescent dyes, purified from excess label, and applied on sample specimen featuring powdery pigment layers on handmade rag paper. As blank comparisons, sample papers without pigment layers were included to verify whether aerosol-misted adhesives are a suitable method to stabilize fragile papers. Penetration of the adhesive-label-conjugates was observed at thin sections of the samples by fluorescence microscopy. The fluorescence labeling of gelatin with Texas Red™ allowed an excellent visualization of aerosol-misted adhesive (0.5–1 %) in all sample types. Methylcellulose (Methocel™ A4C) labeled with Texas Red™ C2-dichlorotriazine enabled fluorescence tracing if applied in 0.5 % solutions by immersion. Aerosol application permitted local adhesive application, making it a suitable technique for stabilizing fragile papers. If applied to samples with low porosity, aerosol-misted gelatin was mainly deposited at the surface, whereas in porous filter paper, penetration dominated over surface deposition. Intermediate drying between repetitive applications apparently limited the penetration of aerosol-misted gelatin.

We described the simple, selective, and rapid method for determination of fucoidans using methylene blue staining of sulfated polysaccharides, immobilized into filter paper and consequent optic density (at A (663) nm) measurement of the eluted dye from filter paper. This solid-phase method allows selective determination of 1-20 μg fucoidan in presence of potentially interfering compounds (alginic acid, DNA, salts, proteins, and detergents). Further, we demonstrated the alternative way of using image processing software for fucoidan quantification without extraction of methylene blue dye from stained spots of fucoidan-dye complex.

Carbon dots synthesized from citric acid and ethylene diamine by a one-step hydrothermal technique were used to develop a carbon dot-based paper immunoassay (CDPIA) for rapid detection of HIV-1 p24 antigen. In the present study, the 96-well template was hand patterned using a wax pencil, as a prototype method, on two types of paper, (1) Whatman filter paper and (2) nitrocellulose paper. The sandwich immunoassay was performed on both paper microplates for detection of HIV-1 p24 antigen which is an early marker of HIV infection. The detection range was from 10 μg/mL to 1 ng/mL for the Whatman filter paper while the nitrocellulose paper exhibited a higher range from 10 μg/mL to 250 pg/mL. CDPIA on the nitrocellulose paper (CDNIA) exhibited a fourfold increase in sensitivity and reduced the assay time by threefold compared with CDPIA on Whatman paper (CDWIA). HIV-negative and HIV-positive plasma samples were tested using CDNIA for the presence of HIV-1 p24 antigen. This immunoassay exhibited no false-positive and false-negative results with the clinical samples tested. This simple and sensitive paper-based HIV-1 p24 antigen assay may be useful in preventing HIV transmission by blood transfusion in resource-limited settings by reducing the antibody negative, infectious window period in blood donors and for early diagnosis of HIV infected individuals where nucleic acid-based testing is not practical or feasible.

Methylmalonic aciduria is a common inherited metabolic disorder. Methylmalonic acid (MMA), a key indicator of methylmalonic aciduria, increases in the amniotic fluid of affected fetuses. For prenatal diagnosis, the MMA in amniotic fluid can be measured by stable-isotope dilution gas chromatography-mass spectrometry. Here, we quantified the MMA in cell-free amniotic fluid samples that had been dried on filter paper and transported at ambient temperatures, and compared the results with data obtained from the original amniotic fluid. Our results indicated that the filter paper method was reproducible and accurate enough to be applied to clinical analysis. We also used the filter paper method to screen at-risk fetuses and obtained a clear diagnosis in each case. We conclude that our method enables the prenatal diagnosis of methylmalonic aciduria using practical procedures and a simplified method for transporting the samples.

A in-line desorption device was developed, which allows for direct analysis of dried blood spots eliminating the need for punching disks from the filter paper cards. Using this device, we have validated a method to quantify biomarkers related to maple syrup urine disease (MSUD), a metabolism disorder that often requires a second-tier test for confirmation. Direct analysis of newborn screening cards is conducted in-line with a high-resolution chromatographic separation with mass spectrometry using electrospray ionization and multiple-reaction monitoring. Quantification of leucine and isoleucine using an isotopically labeled internal standard encompasses a range suitable for MSUD assessment. Precision and accuracy of the technique was acceptable with relative standard deviations within 10% at three fortified concentrations and an unfortified level. A post-column infusion test shows minimum matrix suppression was observed using this direct sampling technique.

Chrysosporium keratinophilum is known to be a keratinophilic fungus and an effective mosquito control agent. This fungus was grown on Sabauraud dextrose broth in the laboratory at 25°C, while the relative humidity was maintained at 75 ± 5% for 15 days. Filtration process of metabolites was done using whatman-1 filter paper, column chromatography and flash, chromatography. Larvicidal efficacy was performed against all instars of Culex quinquefasciatus. Larvicidal efficacy was performed at six different concentrations with different effective ratios (ethanol/metabolites: 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7, 2:8, and 1:9). The mortality values were then subjected by the probit analysis. The larval mortalities were observed for a period of 24, 48, and 72 h, respectively. The first and second instars were highly susceptible to 2:8 ratio. In the first instar after column chromatography, LC(50) =26.66 ppm, LC(90) =121.96 ppm, LC(99) =231.86 ppm were observed after 72 h, while after flash chromatography the LC(50) =20 ppm, LC(90) =123.02 ppm, LC(99) =281.83 ppm were observed after 48 h. In the second instar after column chromatography, LC(50) =18.19 ppm, LC(90) =102.32 ppm, LC(99) =162.18 ppm were observed after 72 h, while doing flash chromatography 100% mortality could be recorded after 24 h. In the third instar after column chromatography, the LC(50) =38.01 ppm, LC(90) =131.82 ppm, LC(99) =245.47 ppm were observed after 72 h, while after flash chromatography the LC(50) =17.78 ppm, LC(90) =100 ppm, LC(99) =151.35 ppm. In the fourth instar, LC(50) =61.65 ppm, LC(90) =181.97 ppm, LC(99) =436.51 ppm, while after flash chromatography LC(50) =40 ppm, LC(90) =120 ppm, and LC(99) =223.87 ppm were observed after 72 h. The extracellular metabolites of C. keratinophilum could be a fungal based larvicides resource for the control of C. quinquefasciatus larvae. This could be another agent for biotechnological exploitation, if found suitable in field trials.

Fabry disease (FD) is an X-linked lysosomal storage disorder resulting from a deficiency of α-galactosidase A, which leads to the progressive accumulation of one biomarker, globotriaosylceramide (Gb3), prominently elevated in the urine of affected patients. Using filter paper discs saturated with urine, we evaluated the analytical performance and clinical usefulness of the urinary Gb3 assay by tandem mass spectrometry (LC-MS/MS), with respect to linearity, precision, and reproducibility. We used healthy control urine samples to validate the reference interval of urinary Gb3. This method showed a good linearity (R2= 0.9998) in the range of 0.05–10 μg/mL. Within-run CVs were less than 5% and total CVs were within 10%. The mean recovery of Gb3 from the urine filter paper was 96.7% and the limit of quantification (S/N≥5) was 0.05 μg/mL, which was sensitive enough for the diagnosis of FD. The mean concentration of Gb3 in urine samples from healthy Korean controls was 5.93 ±3.6 μg/mmol Cr (range 0.9–16.43 μg/mmol Cr). The urinary Gb3 assay by LC-MS/MS showed good analytical performance required for the diagnosis of FD in its linearity, precision, and accuracy. Therefore, this technique could be used for a rapid and reliable first line screening, monitoring and/or diagnosis of individuals at high risk for FD.

Here, we present a significantly improved version of our previously published method for the extraction of fungal genomic DNA from pure cultures using Whatman FTA® filter paper matrix technology. This modified protocol is extremely rapid, significantly more cost effective than our original method, and importantly, substantially reduces the problem of potential cross-contamination between sequential filters when employing FTA technology.

NMR spectroscopy of urine samples is able to diagnose many inborn errors of metabolism (IEM). However, urinary metabolites have a poor stability, requiring special care for routine analysis (storage of urine at -20 or -80 degrees C, fast transport). The aim of our study was to investigate the reliability of dried urine filter paper for urine storage and transport and to evaluate the ability of NMR to detect several IEM using this method. Urine samples from five healthy subjects were analyzed by (1)H NMR following different storage conditions (-20 vs 4 degrees C vs dried on filter paper) and at different time points (24 h, 48 h, 96 h, and 7 days). Urine pattern of fresh urine was considered as a reference. We analyzed the conservation of some amino acids and organic acids using Bland and Altman plot with intraclass correlation coefficient determination. Then, we evaluated the use of filter paper to detect four different IEM (methylmalonic and isovaleric acidurias, ornithine transcarbamylase deficiency, and cystinuria). Analysis of urine samples from healthy subjects revealed a high stability of studied molecules (ICC > 0.8) even after 7 days of storage on filter paper. Moreover, an excellent preservation of metabolites specifically accumulated in IEM was observed when analysis of dried urine filter paper was compared to fresh urine (coefficient of variation < 15%). This preliminary study demonstrates that storage of dried urine on filter paper is reliable for (1)H NMR spectroscopy analysis. Preservation of urine molecules over time using that method is convenient for routine clinical practice.

Cellobiohydrolases (CBH-s) attack primarily from the cellulose chain ends. It is known that CBH-s can differ in their chain end preference. Here the cellulose reducing end-specific fluorescence labeling was studied with the aim to find the most suitable labeled cellulose for easy determination of CBH-s chain end preference. Anthranilic acid (AA) was used as fluorescence label. Bacterial cellulose, amorphous Avicel and filter paper were subjected to the labeling. Suitability of modified bicincoic acid method for determination of reducing groups on cellulose was also confirmed. AA labeled celluloses were hydrolysed with family 7 and 6 CBH-s from Trichoderma reesei and Phanerochaete chrysosporium. Hydrolysis data were plotted as released label (%) versus total degradation (%) (progress curves). Most characteristic progress curves were obtained with AA labeled regenerated amorphous Avicel where the family 6 CBH-s produced clearly upward curved progress curves confirming their preference for non-reducing ends.

A sensitive, specific and efficient high-performance liquid chromatography-tandem mass spectrometry assay for the simultaneous determination of vincristine and actinomycin-D in human dried blood spots is presented. Dried blood spots were punched out of a collection paper with a 0.25-in.-diameter punch. The analytes were extracted from the punched-out disc using sonication during 15 min in a mixture of acetonitrile-methanol-water (1:1:1, v/v/v) containing the internal standard vinorelbine. Twenty-microlitre volumes were injected onto the HPLC system. Separation was achieved on a 50 x 2.1 mm ID Xbridge C(18) column using elution with 1 mM ammonium acetate-acetonitrile (70:30, v/v) adjusted to pH 10.5 with ammonia and run in a gradient with methanol at a flow rate of 0.4 mL/min. HPLC run time was 6 min. The assay quantifies vincristine from 1 to 100 ng/mL and actinomycin-D from 2 to 250 ng/mL using a blood sample obtained by a simple finger prick. Validation results demonstrate that vincristine and actinomycin-D can be accurately and precisely quantified in human dried blood spots with the presented method. The assay can now be used to support clinical pharmacologic studies with vincristine and actinomycin-D.

Newborn screening programs use whole blood dried on filter paper as the standard specimen. Metabolites are reasonably stable and can easily be sent to screening laboratories by regular mail. The recommended sample collection procedure is to spot native blood without anticoagulants onto the filter paper, because anticoagulants can interfere with the different laboratory methods. However, visual examination of the blood spots cannot always detect contamination. In this study, whole blood was drawn by venous puncture from a healthy volunteer, spiked with the corresponding metabolites and EDTA, and spotted onto filter paper. TSH and 17alpha-hydroxyprogesterone were determined by time resolved fluoroimmunoassays with the AutoDelfia system. Total galactose, biotinidase activity, and galactose-1-phosphate uridyltransferase activity were measured photometrically or fluorometrically. Succinyl acetone was estimated indirectly through the inhibition of porphobilinogen synthase activity (PBGS assay). EDTA, amino acids, and acylcarnitines were converted to the corresponding butyl esters, after extraction with methanol, and analysed by LC-MS/MS. EDTA contamination gives falsely elevated 17-OHP values and falsely reduced TSH and PBGS values. The inclusion of an EDTA determination in routine screening revealed that at least 0.06% of newborn screening samples were contaminated with EDTA. In conclusion, non-conformity during the pre-analytical phase is a source of false positive and false negative screening results. Determination of EDTA from NBS blood spots can reliably identify these samples and prevent screening errors.

The authors investigated the effect of Bacillus mucilaginosus on weathering of phosphorite. Analysis of different proteins was of significance in exploring the molecular biological mechanism in the bacterial weathering process. The concrete methods are described as follows: Mineral powder was put into liquid culture medium and B. mucilaginosus was incubated in the medium. The control (group) had no mineral powder in the medium. The treatments and controls were cultured simultaneously under the same condition. In a few days, the supernatant was filtrated, the main cations (Ca2+, Mg2+, Na+, Mn2+, Al3+, Fe3+, K+) were measured by ICP-OES, and the contents of water soluble phosphorus (Pws) and silicon (Siws) were determined by colorimetry. The residual solid was weighed on the filter paper, followed by digestion with concentrated HNO3. The concentrations of the main cations and Pws, Siws in the digest liquid were measured by using the method mentioned above. After the supernatant was centrifuged, the precipitation was used to analyze the protein differences between the treatment groups and the control groups by 2-dimentional gel electrophoresis (2-DE). The experimental results showed that apatite and quartz were partially weathered, but kaolinite was dissolved completely. The population of bacteria increased when mineral powder was added in the liquid medium. Software analysis and comparison of the 2-DE pictures of bacterial proteins revealed 1134 visible protein spots in the treatment group, and 729 visible protein spots in the control group. To compare the bacterial protein expression contents of the treatment group with those of the control group, there were 496 different protein spots, including 214 protein spots which indicated that the protein contents increased, 75 protein spots were indicative of a decrease, and 207 proteins were newly synthesized. It is proposed that the increased bacterial contents may be related to some protein expression and activation after the input of mineral powder. Newly synthesized proteins, the increase of expression contents and the reduction of proteins may be connected with bacterial metabolic regulations, cell activation and signal transmission.

The effects of surfactants on the pretreatment and enzymatic hydrolysis stages of recycled newspaper processing were examined. Newspaper substrate was pretreated with surfactants at 40°C and 400 rpm for 1 h, and the enzymatic digestibilities of the pretreated substrate were compared. NP-20 was 10–20% more effective as a surfactant than Tween-20 and Tween-80. To investigate the effects of the surfactants on the subsequent enzymatic hydrolysis stage, the newspaper was pretreated with NP-20 and then hydrolyzed in the presence of TW-20 or TW-80. TW-80 showed an approximate 7% higher digestibility than TW-20. The surfactant effect on the hydrolysis of the untreated newspaper was significant, whereas the surfactant effect on the hydrolysis of the surfactant-pretreated newspaper was marginal. When the digestibilities of the pure cellulose substrates (α-cellulose and filter paper) were examined, markedly different surfactant effects were observed. In contrast to the newspaper substrate, the surfactant-pretreated pure cellulose substrates had a significant effect on digestibility when they were hydrolyzed in the presence of a surfactant, indicating that the surfactant effect on digestibility is highly dependent on substrate type.

Screening programs for pediatric lead (Pb) poisoning routinely collect blood by finger stick onto specially designed filter paper (FP) for subsequent laboratory testing. Here, laser ablation inductively coupled plasma time-of-flight mass spectrometry is evaluated as a novel method for the determination of Pb in blood spotted onto FP cards. The method requires no sample pretreatment and may be particularly suitable for rapidly screening a large number of samples. Signal intensity from the ICP-MS strongly correlated (r (2) = 0.996) with Pb target values for five blood cards from a laboratory proficiency testing program. Better precision was obtained by replicate ablation line scans (15% rsd) compared to a grid of 25 individual ablation spots (30% rsd). Because2% of the sample is consumed in the laser ablation analysis, selected samples can also be analyzed by traditional techniques or archived. Lead isotope ratios determined during the same total-Pb analysis appear to be effective for discerning contamination extraneous to the blood sample. Determining that an elevated result was due to contamination may negate the need for retesting with invasive pediatric venipuncture. Some elements monitored along with Pb exhibited signal profiles conducive to potential quantitation (Ca, V, Fe, Cu Zn), whereas others did not (Cr, Mn, Co, Ni, Cd, Hg, Tl, U). Copper, Ca, and V were inversely correlated with Pb (r = -0.89, -0.90, and -0.92, respectively). Scanning electron microscope images revealed surface modality differences between FP spotted with fresh whole blood and reconstituted freeze-dried blood reference material.

We developed a simple method to achieve semiquantitative detection of an amphiphilic biosample through measuring the length of flow on a microfluidic analytical device (μPAD) based on paper. When an amphiphilic sample was dripped into a straight microchannel defined with a printed wax barrier (hydrophobic) on filter paper (hydrophilic), the length of flow was affected by the reciprocal effect between the sample, the filter-paper channel and the wax barrier. The flow length decreased with increasing concentration of an amphiphilic sample because of adsorption of the sample on the hydrophobic barrier. Measurement of the flow length enabled a determination of the concentration of the amphiphilic sample. The several tested samples included surfactants (Tween 20 and Triton X-100), oligonucleotides (DNA), bovine serum albumin (BSA), human albumin, nitrite, glucose and low-density lipoprotein (LDL). The results show that the measurement of the flow length determined directly the concentration of an amphiphilic sample, whereas a non-amphiphilic sample was not amenable to this method. A method was developed for the fabrication of microfluidic paper-based analytical devices (μPAD). This method was based on the silanisation of cellulose in filter paper using alkyltrimethoxysilane coupled with UV radiation. The filter paper sheet was hydrophobised by immersion in an octadecyltrimethoxysilane/heptane (OTMS/heptane) solution (0.25 vol. %) containing 5 vol. % of ethyl acetate (EtOAc). The hydrophobic-hydrophilic contrast was generated on the filter paper after the hydrophobised paper sheet was exposed to UV light with a metal mask creating the desired pattern on the sheet. The exposed area was oxidised to create a hydrophilic area, while the hydrophobic area was protected by the metal mask. The optimal conditions for the fabrication of μPAD were studied; these included ethyl acetate concentration (CEtOAc), immersion time, octadecyltrimethoxysilane concentration (COTMS) and exposure time. This method is cost-effective and simple. In addition, different functional groups could be further grafted for various assay purposes. To demonstrate the feasibility of the μPAD in analytical applications, a flower-shaped μPAD with eight channels and eight detection units was fabricated and used to determine the nitrite content in pickled vegetables. The nitrite content (124 µg g−1) in the sample determined by this method compared favourably with that measured using a standard method (137 µg g−1).

Metabolic studies utilizing stable isotope tracer in humans have typically used intravenous tracer infusions and venous blood sampling. These studies explore subcutaneous infusion of isotope and "finger stick" capillary blood sampling to measure glucose turnover. Five subjects received simultaneous 8-h infusions of glucose labeled with isotope: [1-(13)C]glucose subcutaneously and [6,6-(2)H(2)]glucose intravenously. At regular intervals, venous and finger stick blood specimens were obtained. Finger stick blood was applied to filter paper. Substrate and isotopic steady state was reached after 7.0 h with both routes of infusion. The isotopic enrichments of finger stick and venous specimens did not differ significantly for the subcutaneously infused [1-(13)C]glucose (p = 0.33 and p = 0.23, respectively) but the finger stick [6,6-(2)H(2)]glucose enrichment was slightly higher (p0.03) than that of the venous sample. Using [6,6-(2)H(2)]glucose infusion and venous plasma sampling as the reference method, the [1-(13)C]glucose gave estimates of glucose R(a) that were 13% (plasma) and 17% (finger stick) lower (p0.001 and p0.02, respectively). This difference could be attributed to recycling of (13)C label. In conclusion, subcutaneous infusion and finger stick specimen collection onto filter paper represent a potential method of conducting in vivo studies of substrate metabolism outside of a hospital-based research unit.

There are reports of fabrication of taste sensor by adsorbing lipids into Millipore filter paper. With this lipid based sensor, it has been found that the taste sensing efficiency of membrane can be remarkably improved. We have made an attempt to prepare taste sensor material by using functionalized polymer without any lipid. PVA-cellulose composite has been modified to use as the sensor material. The research work covers polymer membrane preparation, morphology study and structural characterization of the membrane and study of the taste sensing characteristics of this membrane for five different taste substances. PVA-cellulose composite membrane was modified by phosphorylation with POCl3. FTIR spectroscopic analysis, XRD analysis and SEM were done to get an idea about the structure and morphology of the prepared phosphorylated PVA-cellulose composite membrane. The sensor characteristics like temporal stability, response stability, response to different taste substances, and reproducibility of sensing performance were studied using phosphorylated PVA-cellulose composite membrane. Sensor device prepared with this membrane has shown distinct response patterns for different taste substances in terms of membrane potential. Threshold concentrations of phosphorylated PVA-cellulose composite membrane for HCl, NaCl, Q-HCl, sucrose and MSG are 0001 mM, 0.001 mM, 0.001 mM, 0.001 mM and 0.009 mM, respectively. The threshold concentrations are below human threshold concentrations. Membranes also showed characteristic response patterns for organic acids like acetic acid, citric acid, formic acid etc, mineral acids like HCl, H2SO4 and HNO3 salts, bitter substances, sweet substances and umami substances. Sensor device prepared with this membrane has excellent shelf life.
