The viral RNA of SARS-Coronavirus-2 is known to be contaminating municipal wastewater. We aimed to assess if COVID-19 disease is spreading through wastewater. We studied the amount of viral RNA in raw sewage and the efficiency of the sewage treatment to remove the virus. Sewage water was collected before and after the activated sludge process three times during summer 2020 from three different sewage treatment plants. The sewage treatment was efficient in removing SARS-CoV-2 viral RNA. Each sewage treatment plant gathered wastewater from one hospital, of which COVID-19 admissions were used to describe the level of disease occurrence in the area. The presence of SARS-CoV-2 viral RNA–specific target genes (N1, N2, and E) was confirmed using RT-qPCR analysis. However, hospital admission did not correlate significantly with viral RNA. Moreover, viral RNA loads were relatively low, suggesting that sewage might preserve viral RNA in a hot climate only for a short time. [ABSTRACT FROM AUTHOR]

Coxsackie B4 (CV-B4), is a major cause of viral myocarditis, dilated cardiomyopathy, and pancreatitis. Like other human enteroviruses, CV-B4 is ubiquitous, excreted in the stool, transmitted by fecal-oral route, and persists in the environment. In the context of studies on CV-B4 infection, it is important to investigate how this virus can be eliminated and to show the possibility of contamination risk with a CV-B4 E2 infected Swiss albino mice. Swiss albino female mice were inoculated with CV-B4 E2 strain and divided in two groups: the first was intraperitoneally (I.P.) infected; the second was orally infected. In order to study the CV-B4 E2 infection in mice, total RNA was extracted from thymus, spleen, pancreas, and intestine, and viral genome was detected using semi-nested (RT-PCR). To further demonstrate infection or immunization of mice, Sera obtained from infected mice were assayed in vitro for their neutralizing antibody. To detect virus in stool of infected mice, stool samples were collected at different post-infection (p.i.) times. Neutralizing antibodies were detectable all along the follow-up period (Day 0, 1, 3, 7, 9, 17, 22, 30, 45, 60 p.i.) in I.P and oral infected mice. Our results showed that when mice were inoculated successively at day 0 and day 8, neutralizing activity was increased in I.P route more than in the oral route. Viral isolation in HEp-2 cells showed negative results. Stool viral analyses reveal a low detection of the CV-B4 E2 genome for all infected mice. In conclusion, our experiments demonstrated that there are no risks linked with the stool of CV-B4 E2 of Swiss albino mice. A variety of biological and chemical contaminants can adversely impact cells in culture, ranging from outright destruction of the culture, mutation, phenotypic changes to relatively minor changes in morphology, or growth rate. There are various approaches to detecting and mitigating the risk of biological or microbial contaminants in cell cultures, and these are discussed in this article. Chemical contaminants typically arise from improper handling or sourcing of cell culture reagents, glassware, or other types of consumables. These and other sources of chemical contaminants of cell cultures are discussed. The occurrence of chemical contamination is mitigated through adherence to best practices in sourcing and handling of such materials and by avoiding the use of volatile solvents within incubators that are used for maintaining cell cultures. The aim of this study was to evaluate the efficiency of a passive point-of-use treatment system, namely, a polyvinyl (alcohol) (PVA) nanofiber membrane/activated carbon column, for the treatment of harvested rainwater. The efficiency of SMI-Q10 [quaternized poly (styrene- co-maleimide)] nanofiber membrane disks placed in a filtration assembly for the treatment of surface water (Plankenburg River, Western Cape, South Africa) and harvested rainwater was also assessed. Two rainwater harvesting tanks were installed at the Welgevallen Experimental farm, Stellenbosch, South Africa, with the filtration system intermittently attached to the tanks for collection of rainwater samples throughout the study period. Parameters used to monitor the filtration systems included heterotrophic bacteria, Escherichia coli, and total coliform enumeration and the presence/absence of adenovirus. When compared to drinking water guidelines, the results indicated that 3 L of potable water could be produced by the synthesized PVA nanofiber membrane/activated carbon column. However, PCR assays indicated that adenovirus and numerous bacteria such as Klebsiella spp., Legionella spp., Pseudomonas spp., and Yersinia spp. were not effectively removed by the filtration system utilized. Additionally, the SMI-Q10 nanofiber membrane disks did not remove viruses from the river or tank water samples as bovine adenovirus 3 strain, simian adenovirus, and human adenovirus A strain were detected in all water samples analyzed. Thus, while the microfiltration system was efficient in reducing the level of indicator organisms to within drinking water standards, further optimization of the electrospun filtration membranes is required as molecular analysis revealed that numerous opportunistic bacterial pathogens and viruses persisted after filtration. We use a technique from engineering (Xia and Moog, in IEEE Trans. Autom. Contr. 48(2):330-336, 2003; Jeffrey and Xia, in Tan, W.Y., Wu, H. (Eds.), Deterministic and Stochastic Models of AIDS Epidemics and HIV Infections with Intervention, 2005) to investigate the algebraic identifiability of a popular three-dimensional HIV/AIDS dynamic model containing six unknown parameters. We find that not all six parameters in the model can be identified if only the viral load is measured, instead only four parameters and the product of two parameters (N and λ) are identifiable. We introduce the concepts of an identification function and an identification equation and propose the multiple time point (MTP) method to form the identification function which is an alternative to the previously developed higher-order derivative (HOD) method (Xia and Moog, in IEEE Trans. Autom. Contr. 48(2):330-336, 2003; Jeffrey and Xia, in Tan, W.Y., Wu, H. (Eds.), Deterministic and Stochastic Models of AIDS Epidemics and HIV Infections with Intervention, 2005). We show that the newly proposed MTP method has advantages over the HOD method in the practical implementation. We also discuss the effect of the initial values of state variables on the identifiability of unknown parameters. We conclude that the initial values of output (observable) variables are part of the data that can be used to estimate the unknown parameters, but the identifiability of unknown parameters is not affected by these initial values if the exact initial values are measured with error. These noisy initial values only increase the estimation error of the unknown parameters. However, having the initial values of the latent (unobservable) state variables exactly known may help to identify more parameters. In order to validate the identifiability results, simulation studies are performed to estimate the unknown parameters and initial values from simulated noisy data.We also apply the proposed methods to a clinical data set to estimate HIV dynamic parameters. Although we have developed the identifiability methods based on an HIV dynamic model, the proposed methodologies are generally applicable to any ordinary differential equation systems. Objective: To investigate the viral contamination of invasive medical instruments in dentistry and to provide health administrative institutions with surveillance data. Methods: Sterilized samples were randomly collected from the department of dentistry to detect HBV-DNA, HCV-RNA, HIV-RNA and HBsAg. Results: Of the invasive medical instruments that were sterilized with 2% glutaraldehyde, one of the samples was positive for HBV-DNA, and another sample was positive for HBsAg. Conclusion: Though massive virus contamination of invasive medical instruments in dentistry has been reduced to a low level, the occurrence of contamination still remains. [ABSTRACT FROM AUTHOR]

The article presents the statement issued by the Indian Academy of Pediatrics Committee of Immunization (IAP COI) and the Strategic Advisory Group of Experts (SAGE) on Immunization on the issue of contamination of the Rotarix, a rotavirus vaccine from GlaxoSmithKline (GSK), with porcine circovirus type-1 (PCV-1). A chronology of events about contamination of the rotavirus vaccine with PCV1 is provided. According to the IAP COI, the use of the vaccine in India poses no immediate health threats to vaccinated individuals.

Microbicides play critical roles in infection prevention and control of Ebola virus by decontaminating high-touch environmental surfaces (HITES), interrupting the virus-HITES-hands nexus. We evaluated the efficacy of formulations containing different microbicidal actives for inactivating Ebola virus–Makona strain (EBOV/Mak) on stainless-steel carriers per ASTM E2197-11. Formulations of sodium hypochlorite (NaOCl) (0.05–1%), ethanol (70%), chloroxylenol (PCMX) (0.12–0.48% by weight) in hard water, and a ready-to-use disinfectant spray with 58% ethanol (EDS), were tested at contact times of 0, or 0.5 to 10 min at ambient temperature. EBOV/Mak was inactivated (> 6 log10) by 70% ethanol after contact times ≥ 2.5 min, by 0.5% and 1% NaOCl or EDS (> 4 log10) at contact times ≥ 5 min, and by 0.12–0.48% PCMX (> 4.2 log10) at contact times ≥ 5 min. Residual infectious virus in neutralized samples was assessed by passage on cells and evaluation for viral cytopathic effect. No infectious virus was detected in cells inoculated with EBOV/Mak exposed to NaOCl (0.5% or 1%), PCMX (0.12% to 0.48%), or EDS for ≥ 5 min. These results demonstrate ≥ 6 log10 inactivation of EBOV/Mak dried on prototypic surfaces by EDS or formulations of NaOCl (≥ 0.5%), PCMX (≥ 0.12%), or 70% ethanol at contact times ≥ 5 min. Despite strong evidence to the contrary, speculation continues that the AIDS virus, human immunodeficiency virus type 1 (HIV-1), may have crossed into humans as a result of contamination of the oral polio vaccine (OPV). This ‘OPV/AIDS theory’ claims that chimpanzees from the vicinity of Stanleyville - now Kisangani in the Democratic Republic of Congo - were the source of a simian immunodeficiency virus (SIVcpz) that was transmitted to humans when chimpanzee tissues were allegedly used in the preparation of OPV. Here we show that SIVcpz is indeed endemic in wild chimpanzees of this region but that the circulating virus is phylogenetically distinct from all strains of HIV-1, providing direct evidence that these chimpanzees were not the source of the human AIDS pandemic.
