Abstract This study aims to explore the perspectives of community leaders (CL) on trusted sources of health information, examine their roles, challenges, and needs in disseminating COVID-19 health information, and identify opportunities to improve public health reach and access for underserved communities. This qualitative study included seven focus groups conducted via online video conferencing from April-June 2021. The participants (n = 9) were CL recruited from the Texas Community Engagement Alliance (CEAL) for their experience working with underserved communities in Dallas, Texas. Data were coded deductively and analyzed thematically. The analysis resulted in three themes: (1) Information fatigue, mistrust, and desensitization were experienced by community members and leaders; (2) CL were expected to process, verify, and share accurate and timely COVID-19 health information; (3) CL identified potential strategies to improve COVID-19 health info access and reach. CLs were critical in addressing mis/disinformation and fostering trust in public health communication, particularly in underserved communities. They acted as vital intermediaries for health information, striving to debunk mis/disinformation and disseminate accurate health information. Public health strategies could benefit from prioritizing the integration of CL in communicating health information during crises and rebuilding trust within historically and racially marginalized and underserved communities. Supporting CLs, for example, through resources and training programs to counter misinformation and provide access to culturally and linguistically tailored health resources, may significantly enhance the access and reach of evidence-based health communication during health crises.

Abstract In recent funding calls, the National Center for Advancing Translational Sciences has shifted its focus from “translational research,” which applies to studies in specific therapeutic areas, toward “translational science” interventions which aim to modify the system of translational discovery in the health sciences. To date, the social responsibilities of translational science have not been adequately articulated. In this paper, we argue that the ethical practice of translational science should include explicit social responsibilities that contribute to improved health outcomes and decreased disparities. Articulating social responsibilities specific to translational science is justified based on three of the field’s foundational elements: (1) the social contract regarding public funding of research, (2) the goals of translational science, and (3) the increased risk of direct, indirect, and systemic harms from translational science, which involve system-level changes. We integrate social responsibilities into a framework which prioritizes developing relevant, usable, and sustainable innovations in translational science and provide three illustrative examples to demonstrate the practical application of this framework.

Abstract Vaccination uptake is essential to controlling the ongoing COVID-19 pandemic. Healthcare workers (HCWs) play a critical role in receiving, recommending and delivering COVID-19 vaccination. Understanding the specific influences on each behaviour enables the development of targeted and tailored interventions to improve vaccination uptake. This paper presents a qualitative synthesis of HCWs’ individual and context barriers and drivers to these three vaccination behaviours across 10 countries in Europe and Central Asia. Qualitative data from interviews and focus group discussions with 378 HCWs between December 2020 and March 2022 were synthesised and organised by four COM (capability, physical and social opportunity, motivation) factors. Differences by stage of COVID-19 vaccine roll-out (in preparation, early and late delivery) were explored. Receiving vaccination related to all four factors. Recommending vaccination mostly related to capability and motivation. HCWs were generally well-informed by official sources and viewed vaccination as the way to end the pandemic, acknowledging their important role in this. Colleagues, family and friends were positive influences on personal vaccination decisions. However, knowledge gaps were evident, particularly amongst nurses who relied on (social) media. Concerns about safety and effectiveness, often connected to knowledge gaps, were heightened by the accelerated timeline for COVID-19 vaccine development and approval. This impeded some HCWs’ motivation to receive and recommend vaccination even in the later roll-out countries. Delivering vaccination was facilitated by support from public health organisations, teamwork and service re-organisation, more evident amongst later roll-out countries. Ongoing high workloads, stress and burnout hindered delivery. Complex and inter-related factors affecting HCWs’ vaccination behaviours were identified. These insights should inform the design of multifaceted interventions (e.g., communication skills training, management support for HCWs’ mental health, and engaging them in decision-making for service redesign); not only for COVID-19 vaccination as it is integrated into routine services but for routine immunization as a whole.

Abstract Workshop manuals on lithography tend to be written with art students in mind and the information they contain largely focuses on technical aspects of the process. It is, however, difficult to put into words the nuances of this printmaking practice, and consequently, handbooks rarely refer to sensory information and phenomenological experience. In light of this issue, my intention in Listen is to test the potential and limitations of written language as a means through which to describe the tacit and embodied knowledge of a lithographer. To aid this task, I created a two-minute video recording of myself preparing a lithography stone and this video features as a central element in the text. Prompted by a process of transcribing its sound, this video became the protagonist of a transdisciplinary encounter between lithographic sound and words. Structured as an intertextual narrative, Listen couples the transcription of the video with a historic, geological and cultural survey of sonorous stones. Punctuating the dialogue, are quotations from lithography handbooks that tether this serendipitous exchange to its intention: that being to speak about the perceptual realms of lithographic practice. At the core of Listen, is the subject of graining limestone—a process that requires both careful attention, to ensure that the surface is even and free from unwanted marks, and a tolerant sensitivity to the abrasive noise of graining stone. These two aspects, attention and noise, are entwined in the content, critical interests, and metaphorical dimensions of Listen. As a piece of written material from ongoing practice-led research that explores the intersection between lithography and language, Listen knowingly tests the protocols of academic language. My intention through this unconventional approach, is not to present the results of an enquiry, but to offer the reader a scriptural space for contemplative reflection. Somewhat akin to the practices of stone lithography, I suggest that the act of engagement that Listen proposes is rewarded by intimate attention and sensitivity to the presence of noise.

Background: Pediatric lower extremity vascular injury (PLEVI) is uncommon and the availability of granular data is sparse. This study evaluated the surgical management of PLEVIs between a Level I adult (ATC) vs pediatric (PTC) trauma center. Methods: We performed a retrospective review of PLEVIs (< 18 years) managed surgically between 01/2009–12/2022. Demographics and outcome data were obtained. Primary outcomes included amputation and fasciotomy rates. Secondary outcomes included type of vessel repair, mortality, and hospital length of stay. Results: Seventy-nine patients were identified, 41 at the ATC and 38 at the PTC, totaling 112 vessels injured. ATC patients were older (median years 16.0 vs 12.5) and almost exclusively (97.6% vs 29.0%) gunshot wounds. Vascular surgeons managed 50% of injuries at the ATC vs 73.7% at the PTC (p = 0.10). Amputations were uncommon and not significantly different between centers. Seventeen patients (44.7%) required fasciotomies at the PTC vs 21 (51.2%) at the ATC (p = 0.56). Rates of vessel repair, ligation, grafting, mortality, and hospital length of stay were not significantly different. Conclusions: PLEVI can be managed safely at ATCs and PTCs with acceptable outcomes. However, important nuances in patient triage and management need to be considered. Objectives: Maternal mortality and morbidity disproportionately affect birthing people from racialized populations. Unfortunately, researchers can often compound these poor outcomes through a lack of authentic community engagement in research beyond the role of the research subject, leading to ineffective strategies for improving care and increasing equity. This article details the real-life strategies utilized to develop a community-engaged research project of a phased federally funded grant employing community engagement principles of co-leadership and co-creation. It also includes reflections from the researchers and advisory board on promising practices and lessons learned for equitably engaging patients and community partners in research. Methods: This article details the application of principles of community-engaged research in a federally funded phased research project focused on understanding disparities in maternal sepsis to develop better clinical and community interventions. Specifically, it discusses early steps in the research partnership to create a sustainable partnership with a Community Leadership Board guided by the principles of transparency, respect, compensation, and increasing research justice. Results: Based on the authors' experience, recommendations are provided for funders, researchers, and institutions to improve the quality and outcomes of communityengaged research. This work adds to community-based participatory and community-engaged research literature by providing concrete and practical steps for equitably engaging in research partnerships with a variety of collaborators. Conclusions: In conclusion, integrated patient and community co-leadership enhances research by providing insight, access to communities for education and dissemination of information, and identifying critical areas needing change. This report may help others address fundamental principles in this journey. Significance: Community-engaged research can be an equitable approach to research that involves communities most affected by empowering them to guide the research as equal partners. This article serves as a guide for researchers to form these critical partnerships, sustain relationships, and recommend solutions to common challenges based on the reflections of the researchers and advisory board. Abstract Infections with rabies virus (RABV) and related lyssaviruses are uniformly fatal once virus accesses the central nervous system (CNS) and causes disease signs. Current immunotherapies are thus focused on the early, pre‐symptomatic stage of disease, with the goal of peripheral neutralization of virus to prevent CNS infection. Here, we evaluated the therapeutic efficacy of F11, an anti‐lyssavirus human monoclonal antibody (mAb), on established lyssavirus infections. We show that a single dose of F11 limits viral load in the brain and reverses disease signs following infection with a lethal dose of lyssavirus, even when administered after initiation of robust virus replication in the CNS. Importantly, we found that F11‐dependent neutralization is not sufficient to protect animals from mortality, and a CD4 T cell‐dependent adaptive immune response is required for successful control of infection. F11 significantly changes the spectrum of leukocyte populations in the brain, and the FcRγ‐binding function of F11 contributes to therapeutic efficacy. Thus, mAb therapy can drive potent neutralization‐independent T cell‐mediated effects, even against an established CNS infection by a lethal neurotropic virus.

Abstract This paper examines the anti-mask and anti-lockdown online movement in connection to the COVID-19 pandemic. To combat the spread of the coronavirus, health officials around the world urged and/or mandated citizens to wear facemasks and adopt physical distancing measures. These health policies and guidelines have become highly politicized in some parts of the world, often discussed in association with freedom of choice and independence. We downloaded references to the anti-mask and anti-lockdown social media posts using 24 search terms. From a total of 4209 social media posts, the researchers manually filtered the explicit visual and textual content that is related to discussions of different genders. We used multimodal discourse analysis (MDM) which analyzes diverse modes of communicative texts and images and focuses on appeals to emotions and reasoning. Using the MDM approach, we analysed posts taken from Facebook and Instagram from active anti-mask and anti-lockdown users, and we identified three main discourses around the gendered discussion of the anti-mask movement including hypermasculine, sexist and pejorative portrayals of “Karen”, and appropriating freedom and feminism discourses. A better understanding of how social media users evoke gendered discourses to spread anti-mask and anti-lockdown messages can help researchers identify differing reactions toward pandemic measures.

Invasive species pose a significant threat to biodiversity, particularly when local communities view them positively, presenting a key challenge for effective control. Acacia trees, native to Australia, have been introduced in various regions worldwide, including Chile. Our study focuses on the geographical distribution, perceptions, memories, and willingness to control Acacia species, especially dealbata and A. melanoxylon, in a basin in Central Chile. Landsat 8 OLI/TIRS images captured during the tree's flowering season were employed to analyse the species' distribution. We conducted an open survey with 81 participants to gauge the community's perceptions and memories of these tree species. A binomial model was constructed, incorporating social and geographic variables, and analyses were conducted to assess the significance of memories. We found in the basin 613.200 hectares of Acacia, constituting 1.7% of the analysed basin. 59 sentences of expressing memories were analyzed; most of them were positive towards Acacia. 45.68% of interviewed believed that Acacia is a native tree, and 35.8% recognized it as a harmful species. Despite 72.84% considering Acacia a useful tree, 60.49% expressed a willingness to limit its expansion. Counterintuitively, the willingness to control the species was positively correlated with its use. The use of Acacia also proved positive interaction with the perception that it is expanding and is harmful. Younger and moderately educated participants exhibited more interest in controlling Acacia trees, and older participants when these variables interacted with their use. We interpret these results as an indication that individuals with greater knowledge of Acacia possess increased awareness of the importance of limiting its expansion. Furthermore, perceptions appear to transition from positive in childhood and youth to negative in adult life. Abstract Analysis of circulating tumor DNA (ctDNA) to monitor cancer dynamics and detect minimal residual disease has been an area of increasing interest. Multiple methods have been proposed but few studies have compared the performance of different approaches. Here, we compare detection of ctDNA in serial plasma samples from patients with breast cancer using different tumor‐informed and tumor‐naïve assays designed to detect structural variants (SVs), single nucleotide variants (SNVs), and/or somatic copy‐number aberrations, by multiplex PCR, hybrid capture, and different depths of whole‐genome sequencing. Our results demonstrate that the ctDNA dynamics and allele fractions (AFs) were highly concordant when analyzing the same patient samples using different assays. Tumor‐informed assays showed the highest sensitivity for detection of ctDNA at low concentrations. Hybrid capture sequencing targeting between 1,347 and 7,491 tumor‐identified mutations at high depth was the most sensitive assay, detecting ctDNA down to an AF of 0.00024% (2.4 parts per million, ppm). Multiplex PCR targeting 21–47 tumor‐identified SVs per patient detected ctDNA down to 0.00047% AF (4.7 ppm) and has potential as a clinical assay.

Abstract Sphingosine‐1‐phosphate (S1P), the circulating HDL‐bound lipid mediator that acts via S1P receptors (S1PR), is required for normal vascular development. The role of this signaling axis in vascular retinopathies is unclear. Here, we show in a mouse model of oxygen‐induced retinopathy (OIR) that endothelial overexpression of S1pr1 suppresses while endothelial knockout of S1pr1 worsens neovascular tuft formation. Furthermore, neovascular tufts are increased in Apom−/− mice which lack HDL‐bound S1P while they are suppressed in ApomTG mice which have more circulating HDL‐S1P. These results suggest that circulating HDL‐S1P activation of endothelial S1PR1 suppresses neovascular pathology in OIR. Additionally, systemic administration of ApoM‐Fc‐bound S1P or a small‐molecule Gi‐biased S1PR1 agonist suppressed neovascular tuft formation. Circulating HDL‐S1P activation of endothelial S1PR1 may be a key protective mechanism to guard against neovascular retinopathies that occur not only in premature infants but also in diabetic patients and aging people.

Abstract Neurodegenerative diseases are increasingly prevalent in the aging population, yet no disease‐modifying treatments are currently available. Increasing the expression of the cold‐shock protein RBM3 through therapeutic hypothermia is remarkably neuroprotective. However, systemic cooling poses a health risk, strongly limiting its clinical application. Selective upregulation of RBM3 at normothermia thus holds immense therapeutic potential. Here we identify a poison exon within the RBM3 gene that is solely responsible for its cold‐induced expression. Genetic removal or antisense oligonucleotide (ASO)‐mediated manipulation of this exon yields high RBM3 levels independent of cooling. Notably, a single administration of ASO to exclude the poison exon, using FDA‐approved chemistry, results in long‐lasting increased RBM3 expression in mouse brains. In prion‐diseased mice, this treatment leads to remarkable neuroprotection, with prevention of neuronal loss and spongiosis despite high levels of disease‐associated prion protein. Our promising results in mice support the possibility that RBM3‐inducing ASOs might also deliver neuroprotection in humans in conditions ranging from acute brain injury to Alzheimer's disease.

The Covid-19 pandemic moved focus from face-to-face learning to hybrid in Higher Education; many educators did not have previous experience of this mode prior to this shift in learning locations. One form of hybrid learning is "synchronous hybrid learning" where both face-to-face and online students simultaneously attend learning activities. This study set out to explore students' experiences of, and attitudes towards, synchronous hybrid learning, using a multi-method approach: a student survey (n = 66) and interviews with both students (n = 22) and lecturers (n = 8). Findings reveal technology challenges for both face-to-face and online learning but the majority of students preferred hybrid learning. Students acknowledged that hybrid formats provided fewer opportunities to collaborate and that they felt more engaged when face-to-face, but they could manage their time better with hybrid. Staff observed equity-related benefits, while acknowledging increased complexity of delivery. The study makes recommendations for a future that encompasses good quality, post-pandemic hybrid learning. [ABSTRACT FROM AUTHOR]

Levels of a predictor variable that are absent when a classification tree is grown can not be subject to an explicit splitting rule. This is an issue if these absent levels are present in a new observation for prediction. To date, there remains no satisfactory solution for absent levels in random forest models. Unlike missing data, absent levels are fully observed and known. Ordinal encoding of predictors allows absent levels to be integrated and used for prediction. Using a case study on source attribution of Campylobacter species using whole genome sequencing (WGS) data as predictors, we examine how target-agnostic versus target-based encoding of predictor variables with absent levels affects the accuracy of random forest models. We show that a target-based encoding approach using class probabilities, with absent levels designated the highest rank, is systematically biased, and that this bias is resolved by encoding absent levels according to the a priori hypothesis of equal class probability. We present a novel method of ordinal encoding predictors via principal coordinates analysis (PCO) which capitalizes on the similarity between pairs of predictor levels. Absent levels are encoded according to their similarity to each of the other levels in the training data. Many studies have shown that the prevalence of degenerative spinal cord compression increases with age. However, most cases at early stages are asymptomatic, and their diagnosis remains challenging. Asymptomatic cervical spinal cord compression (ASCC) patients are more likely to experience annular tears, herniated disks, and later develop symptomatic compression. Asymptomatic individuals do not typically undergo spinal cord imaging; therefore, an assessment test that is both sensitive and specific in diagnosing ASCC may be helpful. It has been demonstrated that the Patient Reported Outcome Measure Information System (PROMIS) mobility test is sensitive in detecting degenerative cervical myelopathy (DCM) symptoms. We investigated the use of the PROMIS mobility test in assessing clinical dysfunction in ASCC. In this study, 51 DCM patients and 42 age-matched healthy control (HC) were enrolled. The degree of cervical spinal cord compression was assessed using the high-resolution cervical spinal cord T2 Weighted (T2w) MRIs, which were available for 14 DCM patients. Measurements of the spinal cords anterior–posterior (AP) diameter at the region(s) that were visibly compressed as well as at different cervical spine levels were used to determine the degree of compression. The age-matched HC cohort had a similar MRI to establish the normal range for AP diameter. Twelve (12) participants in the HC cohort had MRI evidence of cervical spinal cord compression; these individuals were designated as the ASCC cohort. All participants completed the PROMIS mobility, PROMIS pain interference (PI), PROMIS upper extremity (UE), modified Japanese orthopedic association (mJOA), and neck disability index (NDI) scoring scales. We examined the correlation between the AP diameter measurements and the clinical assessment scores to determine their usefulness in the diagnosis of ASCC. Furthermore, we examine the sensitivity and specificity of PROMIS mobility test and mJOA. Compared to the HC group, the participants in the ASCC and DCM cohorts were significantly older (p = 0.006 and p < 0.0001, respectively). Age differences were not observed between ASCC and DCM (p > 0.999). Clinical scores between the ASCC and the HC group were not significantly different using the mJOA (p > 0.99), NDI (p > 0.99), PROMIS UE (p = 0.23), and PROMIS PI (p = 0.82). However, there were significant differences between the ASCC and HC in the PROMIS mobility score (p = 0.01). The spinal cord AP diameter and the PROMIS mobility score showed a significant correlation (r = 0.44, p = 0.002). Decreasing PROMIS mobility was significantly associated with a decrease in cervical spinal cord AP diameter independent of other assessment measures. PROMIS mobility score had a sensitivity of 77.3% and specificity of 79.4% compared to 59.1% and 88.2%, respectively, for mJOA in detecting cervical spinal cord compression. Certain elements of ASCC are not adequately captured with the traditional mJOA and NDI scales used in DCM evaluation. In contrast to other evaluation scales utilized in this investigation, PROMIS mobility score shows a significant association with the AP diameter of the cervical spinal cord, suggesting that it is a sensitive tool for identifying early disability associated with degenerative change in the aging spine. In a comparative analysis of PROMIS mobility test against the standard mJOA, the PROMIS mobility demonstrated higher sensitivity for detecting cervical spinal cord compression. Acquired resistance remains a major challenge for therapies targeting oncogene activated pathways. KRAS is the most frequently mutated oncogene in human cancers, yet strategies targeting its downstream signaling kinases have failed to produce durable treatment responses. Here, we developed multiple models of acquired resistance to dual-mechanism ERK/MAPK inhibitors across KRAS-mutant pancreatic, colorectal, and lung cancers, and then probed the long-term events enabling survival against this class of drugs. These studies revealed that resistance emerges secondary to large-scale transcriptional adaptations that are diverse and cell line-specific. Transcriptional reprogramming extends beyond the well-established early response, and instead represents a dynamic, evolved process that is refined to attain a stably resistant phenotype. Mechanistic and translational studies reveal that resistance to dual-mechanism ERK/MAPK inhibition is broadly susceptible to manipulation of the epigenetic machinery, and that Mediator kinase, in particular, can be co-targeted at a bottleneck point to prevent diverse, cell line-specific resistance programs. Marsh environments, characterized by their flora and fauna, change laterally in response to shoreline erosion, water levels and inundation, and anthropogenic activities. The Grand Bay coastal system (USA) has undergone multiple large-scale geomorphic and hydrologic changes resulting in altered sediment supply, depositional patterns, and degraded barrier islands, leaving wetland salt marshes vulnerable to increased wave activity. Two shore-perpendicular transect sites, one along a low-activity shoreline and the other in a high activity area of the same bay-marsh complex, were sampled to investigate how the marshes within 50 m of the modern shoreline have responded to different levels of increased wave activity over the past century. Surface sediments graded finer and more organic with increased distance from the shoreline while cores generally exhibited a coarsening upwards grain-size trend; all cores contained multiple large sedimentological shifts. 210Pb-based mass accumulation rates over the last two decades were greater than the long-term (centurial) average at each site with the fastest accumulation rates of 7.81 ± 1.58 and 7.79 ± 1.63 kg/m2/year at the sites nearest the shoreline. A shoreline change analysis of three time-slices (1848–2017, 1957–2017, 2016–2017) shows increased erosion at both sites since 1848 with modern rates of −0.95 and −0.88 m/year. Downcore sedimentology, mass accumulation rates, and shoreline change rates paired with foraminiferal biofacies and identification of local estuarine indicator species, Paratrochammina simplissima, aided in identifying paleo marsh types, their relative proximity to the shoreline, and sediment provenance. The high-energy marsh site transitioned from middle marsh to low marsh in the 1960s, and the low-energy marsh site transitioned later, at the end of the twentieth and early twenty-first century, due to its more protected location. Marsh type transition corresponds chronologically with the coarsening upwards grain-size trend observed and the degradation of Grand Batture Island; since its submergence, signatures of multiple storm event have been preserved downcore. Purpose: People with advanced or metastatic cancer and their caregivers may have different care goals and face unique challenges compared to those with early-stage disease or those nearing the end-of-life. These MASCC-ASCO standards and practice recommendations seek to establish consistent provision of quality survivorship care for people affected by advanced or metastatic cancer. Methods: An expert panel comprising MASCC and ASCO members was formed. Standards and recommendations relevant to the provision of quality survivorship care for people affected by advanced or metastatic cancer were developed through conducting: (1) a systematic review of unmet supportive care needs; (2) a scoping review of cancer survivorship, supportive care, and palliative care frameworks and guidelines; and (3) an international modified Delphi consensus process. Results: A systematic review involving 81 studies and a scoping review of 17 guidelines and frameworks informed the initial standards and recommendations. Subsequently, 77 experts (including 8 people with lived experience) across 33 countries (33% were low-to-middle resource countries) participated in the Delphi study and achieved ≥ 94.8% agreement for seven standards (1. Person-Centred Care; 2. Coordinated and Integrated Care; 3. Evidence-Based and Comprehensive Care; 4. Evaluated and Communicated Care; 5. Accessible and Equitable Care; 6. Sustainable and Resourced Care; 7. Research and Data-Driven Care) and ≥ 84.2% agreement across 45 practice recommendations. Conclusion: Standards of survivorship care for people affected by advanced or metastatic cancer are provided. These MASCC-ASCO standards will support optimization of health outcomes and care experiences by providing guidance to stakeholders in cancer care (healthcare professionals, leaders, and administrators; governments and health ministries; policymakers; advocacy agencies; cancer survivors and caregivers. Abstract Biological and biomedical research is increasingly conducted in large, interdisciplinary collaborations to address problems with significant societal impact, such as reducing antibiotic resistance, identifying disease sub-types, and identifying genes that control for drought tolerance in plants. Many of these projects are data driven and involve the collection and analysis of biological data at a large-scale. As a result, life-science projects, which are frequently diverse, large and geographically dispersed, have created unique challenges for collaboration and training. We examine the communication and collaboration challenges in multidisciplinary research through an interview study with 20 life-science researchers. Our results show that both the inclusion of multiple disciplines and differences in work culture influence collaboration in life science. Using these results, we discuss opportunities and implications for designing solutions to better support collaborative tasks and workflows of life scientists. In particular, we show that life science research is increasingly conducted in large, multi-institutional collaborations, and these large groups rely on “mutual respect” and collaboration. However, we found that the interdisciplinary nature of these projects cause technical language barriers and differences in methodology affect trust. We use these findings to guide our recommendations for technology to support life science. We also present recommendations for life science research training programs and note the necessity for incorporating training in project management, multiple language, and discipline culture.

The replication of RNA viruses relies on the activity of RNA-dependent RNA polymerases (RdRps). Despite large variations in their genomic sequences, viral RdRps share a common architecture generally known as a closed right hand. The P2 polymerase of cystovirus φ6 is currently among the best characterized viral RdRps. This polymerase is responsible for carrying out both replication and transcription of the viral double-stranded RNA genome using de novo initiation. Despite the extensive biochemical and structural studies conducted on φ6 P2, further structural information on other cystoviral RdRps is crucial to elucidate the structural and functional diversity of viral RdRps. Here, we have determined the atomic X-ray structure of the RdRp P2 from the φ6-related cystovirus φ8 at 3Å resolution. This structure completes the existing set of structural information on the φ8 polymerase complex and sheds light on the difference and similarities with related cystoviral RdRps. Studies investigating the association between type 2 diabetes mellitus and central adiposity are lacking. Therefore, this study aimed to investigate trends in type 2 diabetes mellitus stratified by central adiposity using waist-to-height ratio (WHtR). Trends in type 2 diabetes mellitus were examined by central adiposity, using WHtR, with data from the Korea National Health and Nutrition Examination Survey (2005–2022). Individuals aged 30 years and over who participated in the survey were selected. Type 2 diabetes mellitus was identified based on serum glucose or HbA1c levels, the use of diabetes medications, or a prior diagnosis by a physician. Weighted β-coefficients or odd ratios (ORs) with 95% confidence intervals (CIs) were calculated to assess changes in disease prevalence. A total of 79,368 participants were included in the database (female: 45,163 [56.9%]). from 2005 to 2022, the prevalence of type 2 diabetes mellitus increased from 3.3 to 5.8% in the healthy central adiposity group, from 11.2 to 17.1% in the increased central adiposity group, and from 18.0 to 26.7% in the high central adiposity group. Males, older population, lower education level, lower household income, and smoking are associated with a higher risk of type 2 diabetes. In the high central adiposity group, overweight and obese individuals had higher susceptibility than underweight or normal-weight individuals, with ORs of 5.85 (95% CI, 2.54–13.47) and 8.24 (3.79–17.94), respectively. The prevalence of type 2 diabetes mellitus has increased in all central adiposity groups in the past decade. Green biorefineries can support the reduction of soybeans imports to Europe, by producing protein-rich animal feed from alternative feedstock such as perennial grass and legume species. Once the protein-rich green juice is extracted, a fiber-rich pulp is left as a residue. This work investigates the thermochemical processing of the pulp via pyrolysis as an option to improve the energy balance and climate footprint of a green biorefinery, by producing non-fossil energy and a high-value biochar product. Laboratory-scale pyrolysis and biochar activation were carried out on pulp samples obtained from different perennial species, different pressing method, and maturity at harvest. The results highlighted the importance of the activation stage to obtain a porous biochar, potentially suitable as animal feed additive. The effects on the overall energy balance and climate impact of the system following the integration of pulp drying and pyrolysis, plus a possible activation step for the biochar, were evaluated with a techno-environmental assessment. The pulp sample composition had only limited influence on the climate impact potentials identified. In all cases, it was found that the integration of a combined drying-pyrolysis-activation system in the green biorefinery may provide substantial additional climate benefits but also that the magnitude of these is strongly dependent on the substitution use-value of the energy products. Background: Targeted axillary dissection (TAD) facilitates nodal staging in cN1 breast cancer after neoadjuvant chemotherapy (NAC). Completion axillary node dissection (cALND) remains the standard of care for TAD-positive patients. This study investigated factors associated with additional positive nodes at cALND (cALND+) and the impact on the residual cancer burden (RCB). Methods: Retrospective review of cN1 breast cancer patients treated with NAC and TAD was conducted from July 2013 to June 2023. The review defined cN1 status by ultrasound (US) and biopsy. Patient, tumor, and treatment characteristics were evaluated. Multivariate analysis was performed to identify factors associated with cALND+, and RCB was calculated. Results: Of 902 patients who underwent TAD, 554 (61.4%) were TAD-positive. 457 underwent cALND, and 124 (27%) were cALND+ (average 4.1 additional +nodes). The cALND+ patients had larger primary tumors at diagnosis (4 vs 3.5 cm; p = 0.04), more than three suspicious nodes on initial US (30% vs 13%; p ≤ 0.0001), larger residual primary tumors on pathology (median, 3 vs 2.1 cm; p = 0.0004), and more positive TAD nodes (median, 2 vs 1; p ≤ 0.0001). In the multivariate analysis, the factors associated with cALND+ were more than three suspicious nodes on initial US (odds ratio [OR], 2.9; p ≤ 0.0001), more positive TAD nodes (OR, 1.1; p ≤ 0.0001), larger clipped node metastasis (OR, 1.1; p ≤ 0.0001), and larger residual tumor on pathology (OR, 1.1; p = 0.006). Of 65 cALND+ patients with RCB class I or II, 29 (45%) had an increase in RCB based on cALND. Conclusion: Of cN1 breast cancer patients treated with NAC who are TAD-positive, approximately 25% will have additional nodal disease on cALND. Background: Granulomatous mastitis (GM) is a benign, chronic, inflammatory disease lacking clear treatment guidelines. The purpose of this American Society of Breast Surgeons (ASBrS) prospective, multisite registry was to characterize the presentation of GM and identify treatment strategies associated with symptom resolution and optimal cosmesis. Methods: ASBrS members entered data into a registry on patient demographics, treatment, symptoms, and cosmesis over a 1-year period. Initial symptoms were graded as mild, moderate, or severe. The Chi-square test and logistic regression were used to identify factors related to symptom improvement and cosmesis. Results: Overall, 112 patients with a mean age of 36 years were included. More patients were Hispanic (49.1%) and from the Southwest (41.1%), and management included observation (4.5%), medical (70.5%), surgical (5.4%), or combination treatment (19.6%). Immunosuppression was used in 83 patients (74.1%), including 43 patients who received intralesional steroid injections. Patients with severe symptoms were more likely to undergo surgical intervention compared with those with mild or moderate symptoms (21.4% vs. 0% and 7.5%, respectively; p = 0.004). Within 1 year, 85 patients (75.9%) experienced symptom improvement and/or resolution at a median of 3 months. Receipt of immunosuppressive therapy was predictive of improvement or resolution at 1 month (odds ratio 4.22; p = 0.045). One-year physician-assessed cosmesis was excellent or good for 20/35 patients (57.1%) and was not associated with type of treatment or symptom severity. Conclusion: Although GM can have a protracted course, the majority of patients in this registry resolved within 1 year, with good cosmetic result. Algal physiological ecology on submarine groundwater discharge (SGD) influenced reefs is likely shaped by intermittent, tidally-driven estuarine conditions that occur with SGD fluxes of fresh-to-brackish groundwater from the subterranean estuary to reef ecosystems. SGD is a common inconspicuous feature worldwide on reefs of basaltic high islands and continental margins. Yet, SGD-driven dynamics of algal physiology are not well understood. To understand how invasive species have physiologically outcompeted native species on many SGD-influenced reefs, physiology in tissue water potential (TWP) regulation, photosynthesis, nitrogen storage, and cellular anatomy were measured across a gradient of SGD-influence, for four Rhodophyte species. Compared with non-SGD conditions, SGD was associated with higher TWP, larger medulla cells with thinner walls, and thinner cortical cell walls for two invasives, Gracilaria salicornia and Acanthophora spicifera, higher photosynthetic rates in G. salicornia, greater nitrogen concentration for A. spicifera and G. salicornia, and increased δ 15N ratios for A. spicifera, G. salicornia, and native Laurencia dendroidea. Distinct physiological strategies were measured for the two invasive species across the gradient of SGD-influence, and for L. dendroidea and Gracilaria perplexa offshore. In naive individuals, sensory neurons directly detect and respond to allergens, leading to both the sensation of itch and the activation of local innate immune cells, which initiate the allergic immune response1,2. In the setting of chronic allergic inflammation, immune factors prime sensory neurons, causing pathologic itch3–7. Although these bidirectional neuroimmune circuits drive responses to allergens, whether immune cells regulate the set-point for neuronal activation by allergens in the naive state is unknown. Here we describe a γδ T cell–IL-3 signalling axis that controls the allergen responsiveness of cutaneous sensory neurons. We define a poorly characterized epidermal γδ T cell subset8, termed GD3 cells, that produces its hallmark cytokine IL-3 to promote allergic itch and the initiation of the allergic immune response. Mechanistically, IL-3 acts on Il3ra-expressing sensory neurons in a JAK2-dependent manner to lower their threshold for allergen activation without independently eliciting itch. This γδ T cell–IL-3 signalling axis further acts by means of STAT5 to promote neuropeptide production and the initiation of allergic immunity. These results reveal an endogenous immune rheostat that sits upstream of and governs sensory neuronal responses to allergens on first exposure. Workplace ostracism is a prevalent and painful experience. The majority of studies focus on negative outcomes of ostracism, with less work examining employees' potential adaptive responses to it. Further, scholars have suggested that such responses depend on employee attributions, yet little research has taken an attributional perspective on workplace ostracism. Drawing on sociometer theory and attribution theory we develop and test a model that investigates why and under what circumstances ostracized employees engage in adaptive responses to ostracism. Specifically, we argue that ostracized employees feel greater levels of shame and, in turn, are motivated to engage in greater ingratiation behavior toward their ostracizers. However, we predict that perceptions of ostracizers' competence and warmth shape different attributional processes, which influence the degree to which the ostracized employee experience shame and, in turn, is motivated to engage in ingratiation behavior. Results of a three-wave, time-lagged survey support our prediction that shame mediates the relationship between coworker ostracism and ingratiation behavior. Moreover, results support our three-way interaction, such that coworkers who report higher levels of ostracism and who perceive their coworkers as more (vs. less) competent and more (vs. less) warm report higher shame, and, in turn, ingratiation behavior. Seasonal epidemics of influenza viruses are responsible for a significant global public health burden. Vaccination remains the most effective way to prevent infection; however, due to the persistence of antigenic drift, vaccines must be updated annually. The selection of vaccine strains occurs months in advance of the influenza season to allow adequate time for production in eggs. RNA vaccines offer the potential to accelerate production and improve efficacy of influenza vaccines. We leveraged the nucleoside-modified RNA (modRNA) platform technology and lipid nanoparticle formulation process of the COVID-19 mRNA vaccine (BNT162b2; Comirnaty®) to create modRNA vaccines encoding hemagglutinin (HA) (modRNA-HA) for seasonal human influenza strains and evaluated their preclinical immunogenicity and toxicity. In mice, a monovalent modRNA vaccine encoding an H1 HA demonstrated robust antibody responses, HA-specific Th1-type CD4+ T cell responses, and HA-specific CD8+ T cell responses. In rhesus and cynomolgus macaques, the vaccine exhibited durable functional antibody responses and HA-specific IFN-γ+ CD4+ T cell responses. Immunization of mice with monovalent, trivalent, and quadrivalent modRNA-HA vaccines generated functional antibody responses targeting the seasonal influenza virus(es) encoded in the vaccines that were greater than, or similar to, those of a licensed quadrivalent influenza vaccine. Monovalent and quadrivalent modRNA-HA vaccines were well-tolerated by Wistar Han rats, with no evidence of systemic toxicity. Purpose: Practical considerations precluding health-related quality of life (HRQOL) monitoring in population and clinical research have spawned development of improved items for more brief surveys of frequently measured HRQOL outcomes. The aim of this study was to validate the use of the Quality of Life General (QGEN-8), a shorter 8-item alternative to the longer 36-item short form (SF)-36 Health Survey for measuring the same eight HRQOL domains across groups of adults with varying severity of acute respiratory symptoms, such as cough and sore throat. Methods: National Opinion Research Center (NORC) representative probability (N = 1,648) and supplemental opt-in (N = 5,915) U.S. adult samples were surveyed cross-sectionally online in 2020. Parallel analyses compared QGEN-8 and SF-36 estimates of group means for each of eight matching profile domains and summary physical and mental scores across groups differing in severity of acute symptoms and chronic respiratory conditions using analysis of covariance (ANCOVAs) controlling for socio-demographics and presence of chronic respiratory conditions. Results: In support of discriminant validity, ANCOVA estimates of QGEN-8 means with SF-36 estimates revealed the same patterns of declining HRQOL with the presence and increasing severity of symptoms and chronic condition severity. Conclusion: QGEN-8® shows satisfactory validity and warrants further testing in cross-sectional and longitudinal population and clinical survey research as a more practical method for estimating group differences in SF-36 profile and summary component HRQOL scores. Summary: Upper respiratory tract infections (URTI) with symptoms such as cough and sore throat are highly prevalent and negatively impact on health-related quality of life (HRQOL). Existing instruments that comprehensively measure HRQOL are lengthy, potentially increasing respondent burden and restricting their use in clinical studies and research. The aim of this study was to evaluate whether eight newly constructed survey items, the QGEN-8®, measure the same HRQOL outcomes as the 36-item SF-36 Health Survey well enough to serve as a more practical alternative for purposes of detecting the physical and mental HRQOL effects on differing severity of acute URTI symptoms, specifically cough and sore throat. The results showed that the QGEN-8® was psychometrically sound and able to differentiate between different levels of URTI symptoms, even in cases where respondents had chronic respiratory conditions. This indicates that the briefer QGEN-8® with 75% shorter response time is able to provide HRQOL measurements comparable to those derived from lengthier instruments thereby lending itself more readily to use in clinical studies and research of URTI symptoms, such as cough and sore throat. To support implementation of integrated behavioral health care (IBHC) models in local settings, providers may benefit from clinical decision-making support. The present analysis examines perspectives on patient characteristics appropriate or inappropriate for, and currently managed within, IBHC at a large medical center to inform recommendations for provider decision-making. Twenty-four participants (n = 13 primary care providers; n = 6 behavioral health providers; n = 5 administrators) in an IBHC setting were interviewed. Thematic analysis was conducted with acceptable interrater reliability (κ = 0.75). Responses indicated behavioral health symptom and patient characteristics that impact perceptions of appropriateness for management in IBHC, with high variability between providers. Many patients with characteristics identified as inappropriate for IBHC were nonetheless currently managed in IBHC. Interactions between patient ability to engage in care and provider ability to manage patient needs guided decisions to refer a patient to IBHC or specialty care. A heuristic representing this dimensional approach to clinical decision-making is presented to suggest provider decision-making guidance informed by both patient and provider ability. The human otolithic system (utricle and saccule), housed within the bony vestibule of the inner ear, establishes our sense of balance in conjunction with the semicircular canals. Yet, while the morphological evolution of the semicircular canals is actively explored, comparative morphological analyses of the otolithic system are lacking. This is regrettable because functional links with head orientation suggest the otolithic system could be used to track postural change throughout human evolution and across primates more broadly. In this context, we present the first analysis of the evolution of the human otolithic system within an anthropoid primate setting. Using the vestibule as a morphological proxy for the utricle and saccule, we compare humans to 13 other extant anthropoid species, and use phylogenetically-informed methods to find correlations with body size, endocranial flexion, and head-neck posture. Our results, obtained through micro-CT of 136 inner ears, reveal two major evolutionary transitions in hominoids, leading to distinctive vestibular morphology in humans, characterized by otolithic morphology resembling squirrel monkeys (possibly due to reversal), with a pronounced supraovalic fossa. Finally, we find a positional signal embedded in the anthropoid bony vestibule, providing the foundation to further explore the evolution of human head-neck posture using inner ear morphology. Pelvic organ prolapse (POP), a downward descent of the vagina and/or uterus through the vaginal canal, is a prevalent condition affecting up to 40% of women. Several risk factors of POP have been identified, including childbirth, connective tissue defects, and chronic intra-abdominal pressure; however, the underlying etiologies of POP development are not fully understood, leading to a high burden on patients and the healthcare systems. The uterosacral ligaments are key support structures of the uterus and upper vagina. Our previous work describes observed histopathological changes in uterosacral ligament (USL) tissue and demonstrates the presence of neutrophils in a subgroup of POP individuals. This presence of neutrophils prompted an examination for the presence of a broader spectrum of inflammatory cell types in the USL. Immunohistochemical staining was performed to identify neutrophils, lymphocytes, macrophages, and mast cells outside of the vasculature. All 4 inflammatory cell types were increased in the POP-HQ system-defined POP-Inflammatory (POP-I) phenotype USL tissue relative to the USL tissues of control or other POP-HQ phenotypes. Focal T-lymphocyte and macrophage co-accumulations were observed in the arterial walls from some patients of the POP-vascular (POP-V) phenotype suggesting previous arterial injury. In addition, 1 control and 2 POP-V subjects' USLs contained arterial wall foamy macrophages, evidence of atherosclerosis. These findings further support a complex etiology for POP and indicate that personalized approaches to preventing and treating the condition may be warranted. [ABSTRACT FROM AUTHOR]

Introduction: This study investigated exposures involving ∆8-tetrahydrocannabinol (∆8-THC) reported to US poison centers (PCs), including variation among states and regions. It evaluated whether the ∆8-THC exposure rate was lower among states with ∆8-THC regulations and states where cannabis (∆9-THC) use was legal. Methods: National Poison Data System data for ∆8-THC exposures in 2021–2022 were analyzed, including comparisons of state and regional population-based exposure rates. Results: There were 4,925 exposures involving ∆8-THC as the primary substance reported to US PCs from January 1, 2021, to December 31, 2022, with 69.8% of these reported in the US South. The rate of exposure per 100,000 US population increased by 79.2% from 0.53 in 2021 to 0.95 in 2022. In 2022, the mean rate of ∆8-THC exposures in states where cannabis use was illegal was 1.64 per 100,000 population (95% CI: 1.08–2.20) compared with 0.52 (95% CI: 0.29–0.76) in states where cannabis use was legal (P = 0.0010). In 2022, the mean rate of ∆8-THC exposures in states where ∆8-THC was unregulated was 1.36 per 100,000 population (95% CI: 0.95–1.77) compared with 0.17 (95% CI: 0.06–0.27) in states where ∆8-THC was banned (P < 0.0001). Conclusions: The rate of ∆8-THC exposures reported to US PCs increased by 79% from 2021 to 2022, with the US South accounting for more than two-thirds of exposures. The rate of ∆8-THC exposures reported to PCs was significantly lower among states where ∆8-THC was banned and among states where cannabis use was legal. Only recently has the use of 3D-scanning technology enhanced our ability to consider shape using landmark approaches to geometric morphometrics (GM). Studies examining several types of anthropological datasets have provided mixed reviews of the utility of the third dimension in landmark GM shape analyses. In this paper we present the results of a study examining the methodological utility of three dimensions in landmark GM shape analysis of prehistoric stone points. We used GM to generate principal components (PCs) of shape variation from independent data sets of Clovis and Dalton points in different shape spaces using 2D and 3D versions of the same sample and examined variation in the distribution of shape variables on PCs generated for each data set. Results of uniform multivariate statistical tests performed on each dataset’s PCs were compared to observe whether 2D or 3D data is more effective at determining group membership. We then generated PCs of shape variation in the same shape space using dependent 2D and 3D datasets to observe whether 2D versions of the data cluster with corresponding 3D versions of each point in a PC biplot and multivariate cluster analysis. Results suggest that 2D GM analysis is as capable of discriminating between Clovis and Dalton points as analyses conducted with 3D data. However, those interested in manufacturing technology will benefit from information provided by 3D data sets, which can capture information such as original blank form and thinning strategies. Systemic lupus erythematosus (SLE) can present with a diverse array of hematologic manifestations, among which atypical hemolytic uremic syndrome (aHUS) is a rare entity. SLE-triggered aHUS has significant morbidity and mortality without timely intervention, yet its frequency remains uncertain and optimal strategies for complement-directed therapies are largely expert-driven. We performed a comprehensive literature review and present a case of a 23-year-old female newly diagnosed with SLE/class IV lupus nephritis who developed aHUS that rapidly responded to the C5 antagonist, eculizumab. Review of the current literature identified forty-nine published cases of SLE with concurrent aHUS and revealed a predilection for aHUS in younger SLE patients, concurrent presentation with lupus nephritis, anti-dsDNA positivity, and complement system abnormalities. Over seventy percent of cases used eculizumab as complement-directed therapy with a trend towards faster time to improvement in laboratory parameters, though reported outcomes were highly variable. Early recognition of aHUS in SLE is pivotal in guiding appropriate therapeutic interventions, and prompt initiation of eculizumab may reduce the potential morbidity associated with plasmapheresis and additional immunosuppression. While eculizumab showcases promising results, its optimal timing and duration remain elusive. An understanding of a patients' complement genetics could aid management strategies, and ongoing research into complement-targeted therapies offers promising avenues for both SLE and aHUS treatment. The aim of this research was to investigate how young students can be supported to use Socratic Seminar as a tool for critical thinking skills and deep, dialogic discussion of complex texts through use of picturebooks. In this teacher action research study, a first-time kindergarten classroom teacher supported by a university professor, enhanced the literacy instruction of their 19 students through the use of Socratic Seminar. Data were systematically collected through weekly Harkness models and a speaking and listening rubric. Results from the Harkness models indicate a meaningful change in the dynamics of the student conversation from teacher-led to interactive student engagement. Across the 5-week intervention, participation increased from approximately one-half of the students to full participation. The speaking and listening rubrics demonstrate a positive increase in (a) tracking the speaker, (b) repeating back what is heard, (c) contributing to conversation with multiple exchanges, and (d) following agreed-upon rules for conversation. Students also became better at listening to others' responses, and responding on topic. Through iterative design and intentional reflection, the teacher and university researcher found that while the Socratic Seminar was effective in improving students' participation, multiple scaffolds were necessary to support young participants such as visual supports (e.g., interactive word walls and printed pages from the texts), physical supports (e.g., question cubes and talking chips), and written supports (e.g., student writing samples). This work aims to serve as a comprehensive guide to properly characterize solid-contact ion-selective electrodes (SC-ISEs) for long-term use as they advance toward calibration-free sensors. The lack of well-defined SC-ISE performance criteria limits the ability to compare results and track progress in the field. Laser-induced graphene (LIG) is a rapid and scalable method that, by adjusting the CO2 laser parameters, can create LIG substrates with tunable surface properties, including wettability, surface chemistry, and morphology. Herein, we fabricate laser-induced graphene (LIG) solid-contact electrodes using different laser settings and subsequently convert them into ion-selective sensors using a potassium-selective membrane. We measure the aforementioned tunable surface properties and correlate them with resultant low-frequency capacitance and water layer formation in an effort to pinpoint their effects on the sensitivity (Nernstian response), reproducibility (E°' variation), and potential stability of the LIG-based SC-ISEs. More specifically, we demonstrate that the surface wettability of the LIG substrate, which can be tuned by controlling the lasing parameters, can be modified to exhibit hydrophobic (contact angle > 90°) and even highly hydrophobic surfaces (contact angle ≈ 130°) to help reduce sensor drift. Recommendations are also provided to ensure proper and robust characterization of SC-ISEs for long-term and continuous measurements. Ultimately, we believe that a comprehensive understanding of the correlation between LIG tunable surface properties and SC-ISE performance can be used to improve the electrochemical behavior and stability of SC-ISEs designed with a wide range of materials beyond LIG. Purpose: Previous work comparing ASD to a normative population demonstrated that a large proportion of lumbar lordosis is lost proximally (L1-L4). The current study expands on these findings by collectively investigating regional angles and spinal contours. Methods: 119 asymptomatic volunteers with full-body free-standing radiographs were used to identify age-and-PI models of each Vertebra Pelvic Angle (VPA) from L5 to T10. These formulas were then applied to a cohort of primary surgical ASD patients without coronal malalignment. Loss of lumbar lordosis (LL) was defined as the offset between age-and-PI normative value and pre-operative alignment. Spine shapes defined by VPAs were compared and analyzed using paired t-tests. Results: 362 ASD patients were identified (age = 64.4 ± 13, 57.1% females). Compared to their age-and-PI normative values, patients demonstrated a significant loss in LL of 17 ± 19° in the following distribution: 14.1% had "No loss" (mean = 0.1 ± 2.3), 22.9% with 10°-loss (mean = 9.9 ± 2.9), 22.1% with 20°-loss (mean = 20.0 ± 2.8), and 29.3% with 30°-loss (mean = 33.8 ± 6.0). "No loss" patients' spine was slightly posterior to the normative shape from L4 to T10 (VPA difference of 2°), while superimposed on the normative one from S1 to L2 and became anterior at L1 in the "10°-loss" group. As LL loss increased, ASD and normative shapes offset extended caudally to L3 for the "20°-loss" group and L4 for the "30°-loss" group. Conclusion: As LL loss increases, the difference between ASD and normative shapes first occurs proximally and then progresses incrementally caudally. Understanding spinal contour and LL loss location may be key to achieving sustainable correction by identifying optimal and personalized postoperative shapes. [ABSTRACT FROM AUTHOR]

Avida-ED is a model system that lets students explore evolution and the nature of science by observing and manipulating the evolutionary dynamics of digital organisms. Over 5 years, we ran eight 2.5-day in-person professional development workshops for 105 primarily college biology instructors to introduce them to Avida-ED and digital evolution and to help them to plan implementations. In this paper, based upon 60-min interviews with 46 of the attendees, we describe what they found to be of value in the workshop itself and the implementations of Avida-ED that they subsequently carried out. The Active LENS workshops were universally valued by the interviewees as a professional development experience; they valued the overall experience of the workshops, their organization and content, and the instructor support materials. Of the 46 teachers that we interviewed, 41 implemented Avida-ED in their classrooms, in 66 separate implementations. We characterized these with respect to the nature of the implementation and its duration, and examined the data in relation to course type, course level, and stated learning goals of the instructors. The most common use was to have students learn evolutionary concepts by observing them in action. A smaller fraction used it to provide a complete research experience. [ABSTRACT FROM AUTHOR]
