2. Identification of a sugarcane bacilliform virus promoter that is activated by drought stress in plants.

Sugarcane (Saccharum spp.) is an important sugar and biofuel crop in the world. It is frequently subjected to drought stress, thus causing considerable economic losses. Transgenic technology is an effective breeding approach to improve sugarcane tolerance to drought using drought-inducible promoter(s) to activate drought-resistance gene(s). In this study, six different promoters were cloned from sugarcane bacilliform virus (SCBV) genotypes exhibiting high genetic diversity. In β-glucuronidase (GUS) assays, expression of one of these promoters (P
SCBV-YZ2060 ) is similar to the one driven by the CaMV 35S promoter and >90% higher compared to the other cloned promoters and Ubi1. Three SCBV promoters (PSCBV-YZ2060 , PSCBV-TX , and PSCBV-CHN2 ) function as drought-induced promoters in transgenic Arabidopsis plants. In Arabidopsis, GUS activity driven by promoter PSCBV-YZ2060 is also upregulated by abscisic acid (ABA) and is 2.2–5.5-fold higher when compared to the same activity of two plant native promoters (PScRD29A from sugarcane and PAtRD29A from Arabidopsis). Mutation analysis revealed that a putative promoter region 1 (PPR1) and two ABA response elements (ABREs) are required in promoter PSCBV-YZ2060 to confer drought stress response and ABA induction. Yeast one-hybrid and electrophoretic mobility shift assays uncovered that transcription factors ScbZIP72 from sugarcane and AREB1 from Arabidopsis bind with two ABREs of promoter PSCBV-YZ2060 . After ABA treatment or drought stress, the expression levels of endogenous ScbZIP72 and heterologous GUS are significantly increased in PSCBV-YZ2060 :GUS transgenic sugarcane plants. Consequently, promoter PSCBV-YZ2060 is a possible alternative promoter for genetic engineering of drought-resistant transgenic crops such as sugarcane. The promoter PSCBV-YZ2060 cloned from the sugarcane bacilliform virus is shown to be induced by drought in sugarcane and Arabidopsis plants and is a potential alternative promoter for genetic engineering of drought-resistant transgenic crops. Triple-negative breast cancer (TNBC) is a highly aggressive cancer with distant metastasis. Accumulated evidence has demonstrated that exosomes are involved in TNBC metastasis. Elucidating the mechanism underlying TNBC metastasis has important clinical significance. In the present study, exosomes were isolated from clinical specimens and TNBC cell lines. Colony formation, EdU incorporation, wound healing, and transwell assays were performed to examine TNBC cell proliferation, migration, and metastasis. Macrophage polarization was evaluated by flow cytometry and RT-qPCR analysis of polarization markers. A mouse model of subcutaneous tumor was established for assessment of tumor growth and metastasis. RNA pull-down, RIP and Co-IP assays were used for analyzing molecular interactions. Here, we proved that high abundance of circRHCG was observed in exosomes derived from TNBC patients, and increased exosomal circRHCG indicated poor prognosis. Silencing of circRHCG suppressed TNBC cell proliferation, migration, and metastasis. TNBC cell-derived exosomes promoted M2 polarization via delivering circRHCG. Exosomal circRHCG stabilized BTRC mRNA via binding FUS and naturally enhanced BTRC expression, thus promoting the ubiquitination and degradation of TFEB in THP-1 cells. In addition, knockdown of BTRC or overexpression of TFEB counteracted exosomal circRHCG-mediated facilitation of M2 polarization. Furthermore, exosomal circRHCG promoted TNBC cell proliferation and metastasis by facilitating M2 polarization. Knockdown of circRHCG reduced tumor growth, metastasis, and M2 polarization through the BTRC/TFEB axis in vivo. In summary, exosomal circRHCG promotes M2 polarization by stabilizing BTRC and promoting TFEB degradation, thereby accelerating TNBC metastasis and growth. Pediatric high-grade gliomas of the subclass MYCN (HGG-MYCN) are highly aggressive tumors frequently carrying MYCN amplifications, TP53 mutations, or both alterations. Due to their rarity, such tumors have only recently been identified as a distinct entity, and biological as well as clinical characteristics have not been addressed specifically. To gain insights into tumorigenesis and molecular profiles of these tumors, and to ultimately suggest alternative treatment options, we generated a genetically engineered mouse model by breeding hGFAP-cre::Trp53
Fl/Fl ::lsl-MYCN mice. All mice developed aggressive forebrain tumors early in their lifetime that mimic human HGG-MYCN regarding histology, DNA methylation, and gene expression. Single-cell RNA sequencing revealed a high intratumoral heterogeneity with neuronal and oligodendroglial lineage signatures. High-throughput drug screening using both mouse and human tumor cells finally indicated high efficacy of Doxorubicin, Irinotecan, and Etoposide as possible therapy options that children with HGG-MYCN might benefit from.Paediatric high-grade gliomas with MYCN amplification (HGG-MYCN) are rare and highly aggressive. 6. Simulation study of hydrogen sulfide removal in underground gas storage converted from the multilayered sour gas field.

A simulation study was carried out to investigate the temporal evolution of H
2 S in the Huangcaoxia underground gas storage (UGS), which is converted from a depleted sulfur-containing gas field. Based on the rock and fluid properties of the Huangcaoxia gas field, a multilayered model was built. The upper layer Jia-2 contains a high concentration of H2 S (27.2 g/m3 ), and the lower layer Jia-1 contains a low concentration of H2 S (14.0 mg/m3 ). There is also a low-permeability interlayer between Jia-1 and Jia-2. The multi-component fluid characterizations for Jia-1 and Jia-2 were implemented separately using the Peng-Robinson equation of state in order to perform the compositional simulation. The H2 S concentration gradually increased in a single cycle and peaked at the end of the production season. The peak H2 S concentration in each cycle showed a decreasing trend when the recovery factor (RF) of the gas field was lower than 70%. When the RF was above 70%, the peak H2 S concentration increased first and then decreased. A higher reservoir RF, a higher maximum working pressure, and a higher working gas ratio will lead to a higher H2 S removal efficiency. Similar to developing multi-layered petroleum fields, the operation of multilayered gas storage can also be divided into multi-layer commingled operation and independent operation for different layers. When the two layers are combined to build the storage, the sweet gas produced from Jia-1 can spontaneously mix with the sour gas produced from Jia-2 within the wellbore, which can significantly reduce the overall H2 S concentration in the wellstream. When the working gas volume is set constant, the allocation ratio between the two layers has little effect on the H2 S removal. After nine cycles, the produced gas's H2 S concentration can be lowered to 20 mg/m3 . Our study recommends combining the Jia-2 and Jia-1 layers to build the Huangcaoxia underground gas storage. This plan can quickly reduce the H2 S concentration of the produced gas to 20 mg/m3 , thus meeting the gas export standards as well as the HSE (Health, Safety, and Environment) requirements in the field. This study helps the engineers understand the H2 S removal for sulfur-containing UGS as well as provides technical guidelines for converting other multilayered sour gas fields into underground storage sites. Purpose: Primary brain tumors are a leading cause of cancer-related death in children, and medulloblastoma is the most common malignant pediatric brain tumor. The current molecular characterization of medulloblastoma is mainly based on protein-coding genes, while little is known about the involvement of long non-coding RNAs (lncRNAs). This study aimed to elucidate the role of the lncRNA OTX2-AS1 in medulloblastoma. Methods: Analyses of DNA copy number alterations, methylation profiles, and gene expression data were used to characterize molecular alterations of OTX2-AS1 in medulloblastoma tissue samples. In vitro analyses of medulloblastoma cell models and orthotopic in vivo experiments were carried out for functional characterization of OTX2-AS1. High-throughput drug screening was employed to identify pharmacological inhibitors, while proteomics and metabolomics analyses were performed to address potential mechanisms of drug action. Results: We detected amplification and consecutive overexpression of OTX2 and OTX2-AS1 in a subset of medulloblastomas. In addition, OTX2-AS1 promoter methylation was linked to OTX2-AS1 expression. OTX2-AS1 knockout reduced medulloblastoma cell viability and cell migration in vitro and prolonged survival in the D283 orthotopic medulloblastoma mouse xenograft model. Pharmacological inhibition of BCL-2 suppressed the growth of OTX2-AS1 overexpressing medulloblastoma cells in vitro. Conclusions: Our study revealed a pro-tumorigenic role of OTX2-AS1 in medulloblastoma and identified BCL-2 inhibition as a potential therapeutic approach to target OTX2-AS1 overexpressing medulloblastoma cells. Pilocytic astrocytoma (PA), the most common pediatric brain tumor, is driven by aberrant mitogen-activated protein kinase signaling most commonly caused by BRAF gene fusions or activating mutations. While 5-year overall survival rates exceed 95%, tumor recurrence or progression constitutes a major clinical challenge in incompletely resected tumors. Here, we used similarity network fusion (SNF) analysis in an integrative multi-omics approach employing RNA transcriptomic and mass spectrometry-based proteomic profiling to molecularly characterize PA tissue samples from 62 patients. Thereby, we uncovered that PAs segregated into two molecularly distinct groups, namely, Group 1 and Group 2, which were validated in three non-overlapping cohorts. Patients with Group 1 tumors were significantly younger and showed worse progression-free survival compared to patients with group 2 tumors. Ingenuity pathways analysis (IPA) and gene set enrichment analysis (GSEA) revealed that Group 1 tumors were enriched for immune response pathways, such as interferon signaling, while Group 2 tumors showed enrichment for action potential and neurotransmitter signaling pathways. Analysis of immune cell-related gene signatures showed an enrichment of infiltrating T Cells in Group 1 versus Group 2 tumors. Taken together, integrative multi-omics of PA identified biologically distinct and prognostically relevant tumor groups that may improve risk stratification of this single pathway driven tumor type. 9. Effectiveness of couple-based interventions for prostate cancer patients and their spouses on their quality of life: a systematic review and meta-analysis.

- Abstract
Purpose: This systematic review and meta-analysis aimed to synthesize the evidence on the effect of couple-based interventions on quality of life (QOL) among prostate cancer patients and their spouses. Method: Six English databases and two Chinese databases were systematically searched to identify relevant RCTs that examined the effect of couple-based interventions on QOL. The data from the included studies were extracted by two independent reviewers using a standardized data extraction form. Methodological quality was assessed by using the Cochrane Risk of Bias Tool. Meta-analysis was conducted among the suitable studies that the available data were sufficient. Results: One thousand ninety-five studies were identified, and 11 studies met the inclusion criteria for qualitative synthesis and 7 studied for meta-analysis. Couple-based interventions involve different formats of physical and psychosocial interventions. Physical exercise-based interventions were popular among couples, and these interventions had the highest level of adherence among all interventions examined herein. However, the meta-analysis of total QOL and physical and mental health revealed a non-significant effect on both prostate cancer patients and their spouses. More RCTs examining couple-based interventions may be needed in developing countries, especially in Asian countries. Conclusion: Couple-based interventions had non-significant effect on improving the total QOL and physical and mental health of prostate cancer patients and their spouses. However, the current evidence is limited because the sample size of the studies is small. Thus, more studies with large sample sizes need to be included to detect the efficacy of couple-based interventions on prostate cancer patients and their spouses. [ABSTRACT FROM AUTHOR]

10. Experimental study on uniaxial creep characteristics of sandstone with pre-peak unloading damage.

- Abstract
Deep resource extraction has been affected by the complex geological environment of "three highs and one disturbance" for a long time. The surrounding rocks experience strong unloading stress disturbance during the underground resource extraction. The creep characteristics of the perimeter rocks are of great theoretical and practical value. Firstly, the triaxial pre-peak loading and unloading tests on the prepared samples are conducted on the intact rock samples using the TAW-200 rock mechanics test system. Then the rock samples with different degrees of pre-peak unloading damage were prepared under the condition of pre-peak yield. Finally, the uniaxial creep tests were carried out to study the uniaxial creep mechanical properties. The results show that the wave velocities of the damaged rock samples are reduced to different degrees compared with those of the intact rock samples, the creep of the pre-peak unloading damaged rock has a time-dependent damage effect, and the study results are similar to those of the conventional uniaxial creep test of the white sandstone. The instantaneous strain at all stress levels of the damaged rock samples increased gradually with the stress level, which is consistent with the non-linear functional relationship; Based on the time-dependent damage effect, the two-parameter Weibull distribution function was introduced into the West Plains viscoelasticity model, and a creep damage model for white sandstone was proposed. The improved Nishihara model can simulate the uniaxial creep characteristics of each damaged rock sample at various stress levels. [ABSTRACT FROM AUTHOR]

Most lncRNAs display species-specific expression patterns suggesting that animal models of cancer may only incompletely recapitulate the regulatory crosstalk between lncRNAs and oncogenic pathways in humans. Among these pathways, Sonic Hedgehog (SHH) signaling is aberrantly activated in several human cancer entities. We unravel that aberrant expression of the primate-specific lncRNA HedgeHog Interacting Protein-AntiSense 1 (HHIP-AS1) is a hallmark of SHH-driven tumors including medulloblastoma and atypical teratoid/rhabdoid tumors. HHIP-AS1 is actively transcribed from a bidirectional promoter shared with SHH regulator HHIP. Knockdown of HHIP-AS1 induces mitotic spindle deregulation impairing tumorigenicity in vitro and in vivo. Mechanistically, HHIP-AS1 binds directly to the mRNA of cytoplasmic dynein 1 intermediate chain 2 (DYNC1I2) and attenuates its degradation by hsa-miR-425-5p. We uncover that neither HHIP-AS1 nor the corresponding regulatory element in DYNC1I2 are evolutionary conserved in mice. Taken together, we discover an lncRNA-mediated mechanism that enables the pro-mitotic effects of SHH pathway activation in human tumors. Long non-coding RNAs (lncRNAs) can contribute to cancers that are driven by Sonic hedgehog (SHH) signaling. Tomato early blight and grey mould are common diseases that affect tomato yield and quality. Endophytic bacteria are a class of biocontrol microorganisms that exist widely in plants that can, along with their metabolites, be used to develop new bacterial agents to control plant diseases and regulate plant growth while reducing the need for chemical pesticides. We previously isolated an endophytic bacterium ZJ1, Bacillus velezensis, from the stem of Buddleja lindleyana. The volatile organic compounds (VOCs) from ZJ1 exhibited significant antifungal activity against Alternaria solani and Botrytis cinerea. The VOCs were identified via gas chromatography–mass spectrometry of solid-phase microextraction samples (SPME–GC–MS). The results revealed eight main VOCs released by the strain ZJ1, among which isooctanol and 2-nonanol showed inhibitory effects on the mycelia growth of A. solani and B. cinerea. Furthermore, 2-nonanol and isooctanol showed good in vitro antifungal activity and in vivo control effect against the two pathogens. The 500 million CFU mL
−1 ZJ1 strain oil suspension with 200 times dilution had the highest control effect, the protective and therapeutic effects were both greater than 80%. Additionally, treatment with ZJ1 and its VOCs caused the mycelium of A. solani to become thin, twisted, deformed, bifurcated, and fractured, with a wrinkled and cracked surface. The VOCs caused leakage of the mycelia intracellular contents, thereby causing withering. Collectively, these findings suggest that the preparation of the ZJ1 strain and its VOC products, 2-nonanol and isooctanol, has good application prospects for the control of tomato early blight and grey mould. Deep high-stress roadway excavation under unloading disturbance inevitably leads to damage deterioration of the surrounding rock, which poses a serious threat to its stability. To explore the energy characteristics of white sandstone damaged by peak front unloading, uniaxial compression tests were conducted on damaged rock samples. The results show that the peak strength and modulus of elasticity of the rock sample gradually decrease with increasing damage degrees. The external work input energy, releasable elastic strain energy and dissipation energy all decreased with increasing damage. Damage evolution curves and equations of the rock samples were obtained based on the damage instantiation model established by the principle of energy dissipation and release. The effects of unloading damage on the fracture characteristics of the rock samples were analysed from both macro and microscopic viewpoints, and the results showed that a micro fracture in the rock is transformed from brittle–ductile damage, while macroscopic damage occurs in the form of a "shear"-"splitting"-"mixed shear-splitting" damage process. Prior research shows that noisy prices can introduce biases in returns causing equal-weighted portfolios to outperform value-weighted portfolios. In this paper, we reevaluate the superiority of EW portfolios in the presence of market frictions. We find that trading costs have limited impact on the performance of EW portfolios, while taxes cause the performance of both EW and VW portfolios to deteriorate by a similar magnitude leaving the superiority of EW portfolios intact. Besides mispricing, the results for small cap portfolios may also be affected by the size effect. Overall, EW portfolios earn annualized risk-adjusted returns between 1.23% and 1.79% even after accounting for trading costs and taxes. [ABSTRACT FROM AUTHOR]

Object recognition is among the basic survival skills of human beings and other animals. To date, artificial intelligence (AI) assisted high-performance object recognition is primarily visual-based, empowered by the rapid development of sensing and computational capabilities. Here, we report a tactile-olfactory sensing array, which was inspired by the natural sense-fusion system of star-nose mole, and can permit real-time acquisition of the local topography, stiffness, and odor of a variety of objects without visual input. The tactile-olfactory information is processed by a bioinspired olfactory-tactile associated machine-learning algorithm, essentially mimicking the biological fusion procedures in the neural system of the star-nose mole. Aiming to achieve human identification during rescue missions in challenging environments such as dark or buried scenarios, our tactile-olfactory intelligent sensing system could classify 11 typical objects with an accuracy of 96.9% in a simulated rescue scenario at a fire department test site. The tactile-olfactory bionic sensing system required no visual input and showed superior tolerance to environmental interference, highlighting its great potential for robust object recognition in difficult environments where other methods fall short. For the object recognition in lightless environments, the authors propose the olfactory-tactile machine learning approach, inspired by the star-nose mole's neural system. 17. Relative slipping at the interfaces of the flexible fiber Bragg grating sensor.

- Abstract
To discuss the relative slipping at the interface between a flexibly embedded fiber Bragg grating sensor and a substrate, the strain transfer was derived for an ideal case between the materials of an embedded fiber sensor. ANSYS software was used to establish a simulation model and to analyze the effects of the axial tensile force and the semi-embedded length and encapsulation substrate for the axial strain relative errors with and without relative slipping. The results of the numerical simulations show that the relative strain errors are smaller at the ends of the fibers and larger in the middle for the same tensile force, indicating that the strain transfer effect is location dependent and that the choice of a semi-embedded length of the fibers greater than 40 mm helps to reduce the relative errors. Meanwhile, five flexible sensors with different half-embedding lengths were experimentally encapsulated and subjected to axial tension-strain experiments, which showed the best strain transfer at a half-embedding length of 60 mm, and the experimental results were consistent with the numerical simulation results. The experimental results provide some theoretical and experimental basis for parameter optimization of flexible fiber grating sensors. [ABSTRACT FROM AUTHOR]

Electron beam lithography (EBL) is renowned to provide fabrication resolution in the deep nanometer scale. One major limitation of current EBL techniques is their incapability of arbitrary 3d nanofabrication. Resolution, structure integrity and functionalization are among the most important factors. Here we report all-aqueous-based, high-fidelity manufacturing of functional, arbitrary 3d nanostructures at a resolution of sub-15 nm using our developed voltage-regulated 3d EBL. Creating arbitrary 3d structures of high resolution and high strength at nanoscale is enabled by genetically engineering recombinant spider silk proteins as the resist. The ability to quantitatively define structural transitions with energetic electrons at different depths within the 3d protein matrix enables polymorphic spider silk proteins to be shaped approaching the molecular level. Furthermore, genetic or mesoscopic modification of spider silk proteins provides the opportunity to embed and stabilize physiochemical and/or biological functions within as-fabricated 3d nanostructures. Our approach empowers the rapid and flexible fabrication of heterogeneously functionalized and hierarchically structured 3d nanocomponents and nanodevices, offering opportunities in biomimetics, therapeutic devices and nanoscale robotics. 19. Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity.

Psoraleae Fructus (PF) is a well-known traditional herbal medicine in China, and it is widely used for osteoporosis, vitiligo, and other diseases in clinical settings. However, liver injury caused by PF and its preparations has been frequently reported in recent years. Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury (IDILI), but the mechanism underlying its hepatotoxicity remains unclear. This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity. Bavachin boosts the secretion of IL-1β and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C), monosodium urate crystal, or intracellular lipopolysaccharide. Bavachin does not affect AIM2 or NLRC4 inflammasome activation. Mechanistically, bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome. Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-1β via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI. These results suggest that bavachin specifically enhances the ATP- or nigericin-induced activation of the NLRP3 inflammasome. Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity. Moreover, bavachin and PF should be evaded among patients with diseases linked to the ATP- or nigericin-mediated activation of the NLRP3 inflammasome, which may be a dangerous factor for liver injury. The recent emergence of new fentanyl analogues and synthetic opioids on the drug market poses a global public health threat. However, these compounds cannot typically be identified using existing analytical methods. In this study, we aimed to develop and validate a rapid and sensitive method based on ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) for the simultaneous determination of 37 fentanyl analogues and novel synthetic opioids in hair samples. Hair samples (20 mg) were extracted by cryogenic grinding in an extraction medium of methanol, acetonitrile, and 2 mmol/L ammonium acetate (pH 5.3). Following centrifugation of the samples, the analytes were separated using a WATERS Acquity UPLC HSS T3 column. The limits of detection (LODs) and limits of quantification (LOQs) ranged from 0.5 to 2.5 pg/mg and from 2 to 5 pg/mg, respectively. The intraday and interday precisions were within 13.32% at LOQ, low, medium, and high levels. The accuracies were within the range of 85.63–116.1%. The extraction recoveries were in the range of 89.42–119.68%, and the matrix effects were within the range of 44.81–119.77%. Furthermore, the method was successfully applied to the detection and quantification of fentanyl and sufentanil in hair samples from two authentic cases. Epimedii Folium (EF) combined with Psoraleae Fructus (PF) is a common modern preparation, but liver injury caused by Chinese patent medicine preparations containing EF and PF has been frequently reported in recent years. Zhuangguguanjiewan pills (ZGW), which contain EF and PF, could induce immune idiosyncratic liver injury according to clinical case reports and a nonhepatotoxic dose of lipopolysaccharide (LPS) model. This present study evaluated the liver injury induced by EF or PF alone or in combination and investigated the related mechanism by using the LPS model. Liver function indexes and pathological results showed that either EF or PF alone or in combination led to liver injury in normal rats; however, EF or PF alone could lead to liver injury in LPS-treated rats. Moreover, EF combined with PF could induce a greater degree of injury than that caused by EF or PF alone in LPS-treated rats. Furthermore, EF or PF alone or in combination enhanced the LPS-stimulated inflammatory cytokine production, implying that IL-1β, which is processed and released by activating the NLRP3 inflammasome, is a specific indicator of EF-induced immune idiosyncratic hepatotoxicity. Thus, EF may induce liver injury through enhancing the LPS-mediated proinflammatory cytokine production and activating the NLRP3 inflammasome. In addition, the metabolomics analysis results showed that PF affected more metabolites in glycerophospholipid and sphingolipid metabolic pathways compared with EF in LPS model, suggesting that PF increased the responsiveness of the liver to LPS or other inflammatory mediators via modulation of multiple metabolic pathways. Therefore, EF and PF combination indicates traditional Chinese medicine incompatibility, considering that it induces idiosyncratic hepatotoxicity under immunological stress conditions. An aggressive proliferation of synoviocytes is the hallmark of rheumatoid arthritis (RA). Emerging evidence shows that inhibiting the NF-κB signaling pathway with 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] may be a therapeutic approach for controlling inflammatory diseases. In this study, we demonstrated the protective effects of three different 1,25(OH)2D3 concentration on adjuvant-induced arthritis (AA) rats through the NF-κB signaling pathway and their pro-apoptotic roles in cultured adjuvant-induced arthritis synoviocytes (AIASs). AA rats were prepared by injecting complete Freund's adjuvant and independently given daily intraperitoneal injection of 1,25(OH)2D3 at concentrations of 50, 100, and 300 ng/day/kg. Subsequently, AIASs were isolated from the inflamed joints of AA rats to test the effects of 1,25(OH)2D3 on AIASs in vitro. Intraperitoneal injection of 1,25-(OH)2D3 was found to induce a concentration- and time-dependent improvement in relieving the symptoms of AA. We found an increased paw withdrawal thermal latency (PWTL) in the affected paw of AA rats as the concentration of 1,25-(OH)2D3 increased. 1,25-(OH)2D3 treatment reduced levels of inflammatory factors in synovial tissues of AA rats. In the case of cultured AIASs, 1,25-(OH)2D3 was shown to inhibit cell proliferation and induce cell apoptosis in a concentration-dependent manner. Additionally, 1,25-(OH)2D3 inhibited the activation of the NF-κB signaling pathway. In conclusion, our study provides evidence emphasizing that 1,25(OH)2D3 has the potential to attenuate disease severity in RA potentially due to its contributory role in synoviocyte proliferation and apoptosis. 1,6- O, O-Diacetylbritannilactone (OODBL), a plant sesquiterpene lactone, was previously reported to show multiple pharmacological effects such as anti-cancer and anti-inflammatory activities. In this study, we investigated the anti-inflammatory effect of OODBL on interleukin (IL)-4-induced signal transducer and activator of transcription 6 (STAT6) signaling pathway in human lung A549 cells. We found that OODBL dramatically inhibited IL-4-induced messenger RNA (mRNA) expression of eotaxin-1 and arachidonate 15-lipoxygenase-1 (ALOX15) in a dose-dependent manner. To clarify the action mechanism of OODBL, we examined the effect of OODBL on activation of STAT6. OODBL decreased both STAT6 phosphorylation and reporter gene activity. Furthermore, OODBL suppressed phosphorylation of Janus Kinase 3 (JAK3) without affecting JAK1. Taken together, OODBL abolished IL-4-induced eotaxin-1 and ALOX15 mRNA expressions by repressing the activation of STAT6 and JAK3. These results suggest that OODBL has a potential therapeutic efficacy on inflammatory diseases especially allergic airway disease as a lead compound. [ABSTRACT FROM AUTHOR]

27. Numerical investigation of characteristics of water-exit ventilated cavity collapse.

- Abstract
The collapse of the ventilated cavitation occurring on a cylindrical vehicle during the water-exit is numerically researched. The numerical model employs the finite volume method to solve the multiphase Reynoldsaveraged Navier-Stokes (RNNS) equations and uses the volume of fluid (VOF) method to capture the free surface. A practical water wave environment that the vehicle usually encounters is reproduced in a numerical wave flume, so that the water wave's effect on the cavity collapse flow regime is investigated. The main feature of the waterexit collapse of a ventilated cavity is studied under the wave-free condition. The result indicates that a collapsing ventilated cavity experiences two stages, in which the pattern of cavity evolution is different. In the early stage, the cavity undergoes a rapid shrinkage as a closed body. In the late stage, the cavity releases gas from the front due to the increase of the cavity pressure. The water wave effect is investigated at three typical wave phases: the wave crest, the wave trough and the wave node. Results show that when the vehicle is launched under the wave node, the cavity collapse regime remains fairly axisymmetric and is similar to the wave-free case. However, when the vehicle is launched under the wave crest and trough, the cavity evolution presents highly three-dimensional (3D) features. The results of predicted cavity size, pressure distributions and hydrodynamic forces at different wave phases show that the wave effect is relatively weak at the wave node but becomes apparent at the wave crest and trough. Genetic diversity analysis, which refers to the elaboration of total extent of genetic characteristics in the genetic makeup of a certain species, constitutes a classical strategy for the study of diversity, population genetic structure, and breeding practices. In this study, fluorescence-labeled seven gSSR and eight EST-SSR primer pairs and a capillary electrophoresis genotyping platform were used to assess the genetic diversity among 181 sugarcane clones. The clones were sorted into 14 series based on their origin. A total of 205 polymorphic SSR alleles were identified. The mean polymorphic information content ( PIC) value was 0.94 for gSSRs and 0.93 for EST-SSRs, respectively. Gene differentiation coefficient ( Gst) of inter-series variation (13.71 %) was much lower than intra-series variation (86.29 %). Gene flow value ( Nm = 3.15) suggested that there was no significant genetic differentiation or population structure variations among the 14 series. The 181 clones could be clustered into seven groups based on neighbor-joining cluster analysis. Three major groups, namely the USA Group, the Guangxi-Hainan-Fujian Group, and the Guangdong Group, consisted of 36, 64, and 39 clones, respectively. Alterations in this process may entail a high-risk for disease in later life. However, the potential factors affecting this process in the infant are not well known. Moreover, the potential impact of orally administered vaccines upon the establishing microbiome remains unknown. Here we assessed the intestinal microbiome establishment process and evaluated the impact of rotavirus vaccination upon this process. Metagenomic, PCR-DGGE and faecal short chain fatty acids analyses were performed on faecal samples obtained from three infants before and after the administration of each dose of vaccine. We found a high inter-individual variability in the early life gut microbiota at microbial composition level, but a large similarity between the infants'microbiomes at functional level. Rotavirus vaccination did not show any major effects upon the infant gut microbiota. Thus, the individual microbiome establishment and development process seems to occur in a defined manner during the first stages of life and rotavirus vaccination appears to be inconsequential for this process. A phytochemical investigation of Smilax riparia A. DC. resulted in the isolation of a new aurone ( 1) and 13 known compounds ( 2-14). The structure of the new aurone was determined to be 4,5,3′,4′,5′-pentamethoxy-6-hydroxyaurone on the basis of spectroscopic methods. Compounds 3-11 were isolated from this genus for the first time. Compounds 3 and 4 significantly inhibited the migration of MDA-MB-231 breast cancer cells induced by the chemokine epithelial growth factor (EGF). In addition, compounds 3, 4, and 8 showed significant effects on enhancing GLUT4 translocation in skeletal muscle cells L6myc. [ABSTRACT FROM AUTHOR]

The previous studies all focus on the effect of probiotics and antibiotics on infection after liver transplantation. Here, we focus on the effect of gut microbiota alteration caused by probiotics and antibiotics on hepatic damage after allograft liver transplantation. Brown-Norway rats received saline, probiotics, or antibiotics via daily gavage for 3 weeks. Orthotopic liver transplantation (OLT) was carried out after 1 week of gavage. Alteration of the intestinal microbiota, liver function and histopathology, serum and liver cytokines, and T cells in peripheral blood and Peyer's patch were evaluated. Distinct segregation of fecal bacterial diversity was observed in the probiotic group and antibiotic group when compared with the allograft group. As for diversity of intestinal mucosal microbiota and pathology of intestine at 2 weeks after OLT, antibiotics and probiotics had a significant effect on ileum and colon. The population of Lactobacillus and Bifidobacterium in the probiotic group was significantly greater than the antibiotic group and the allograft group. The liver injury was significantly reduced in the antibiotic group and the probiotic group compared with the allograft group. The CD4/CD8 and Treg cells in Peyer's patch were decreased in the antibiotic group. The intestinal Treg cell and serum and liver TGF-β were increased markedly while CD4/CD8 ratio was significantly decreased in the probiotic group. It suggested that probiotics mediate their beneficial effects through increase of Treg cells and TGF-β and deduction of CD4/CD8 in rats with acute rejection (AR) after OLT. [ABSTRACT FROM AUTHOR]

Our research group showed that E-salignone inhibited the invasion of human breast cancer MDA-MB-231 cells induced by the chemokine epidermal growth factor (EGF). Due to the limited natural resources, we had to prepare sizable quantities of E-salignone for the antimetastasis investigation. E-Salignone was synthesized by methylation of the corresponding amine by reductive amination with paraformaldehyde in the presence of sodium triacetoxyhydroborate. The synthesized E-salignone exhibited significant inhibitory effects on the invasion of human MDA-MB-231 breast cancer cells and non-small cell lung cancer cells (A549) induced by the chemokine EGF with IC values of 0.36 μM and 5.77 μM, respectively. Since angiogenesis is required for tumor metastasis, anti-angiogenesis is consider a promising approach for tumor therapy. In our investigation of antimetastatic activity, E-salignone potently inhibited the migration of HUVEC and MDA-MB-231 in the wound healing assay. [ABSTRACT FROM AUTHOR]

Liver cirrhosis occurs as a consequence of many chronic liver diseases that are prevalent worldwide. Here we characterize the gut microbiome in liver cirrhosis by comparing 98 patients and 83 healthy control individuals. We build a reference gene set for the cohort containing 2.69 million genes, 36.1% of which are novel. Quantitative metagenomics reveals 75,245 genes that differ in abundance between the patients and healthy individuals (false discovery rate < 0.0001) and can be grouped into 66 clusters representing cognate bacterial species; 28 are enriched in patients and 38 in control individuals. Most (54%) of the patient-enriched, taxonomically assigned species are of buccal origin, suggesting an invasion of the gut from the mouth in liver cirrhosis. Biomarkers specific to liver cirrhosis at gene and function levels are revealed by a comparison with those for type 2 diabetes and inflammatory bowel disease. On the basis of only 15 biomarkers, a highly accurate patient discrimination index is created and validated on an independent cohort. 38. Novel Cell Membrane Capillary Chromatography for Screening Active Compounds from Natural Products.

- Abstract
Cell membrane chromatography (CMC) is a useful method for the simultaneous isolation and identification of active compounds from natural products. However, it suffers from high cell membrane consumption and is time-consuming to operate. In this study, CMC was performed for the first time with a silica capillary, termed cell membrane capillary chromatography (CMCC). Pancreatic islet cell membranes from a mouse were immobilized onto the capillary inner wall functionalized with aldehyde groups. Scanning electron microscopy observation of the prepared column showed that the cell membrane was evenly coated onto the capillary inner wall. Three model analytes with the pharmacological property of hypoglycemic activity including glibenclamide, glipizide and berberine were tested. They were all retained by the prepared column. Furthermore, the retention factors of the analytes in CMCC correlated well with their pharmacological action. The analytical procedure including washing (to obtain a flat baseline), injection and separation was accomplished within 10 min. The CMCC column was also used for screening active compounds from a natural plant ( Coptis chinensis). The hypoglycemia activity of active components such as berberine was verified using the method. Here, we describe a novel method utilizing double stable isotope ultra performance liquid chromatography-tandem mass spectrometry to measure tissue contents and activity of phenylethanolamine N-methyltransferase (PNMT), the enzyme responsible for synthesis of the stress hormone, epinephrine. The method is based on measurement of deuterium-labeled epinephrine produced from the reaction of norepinephrine with deuterium-labeled S-adenosyl- l-methionine as the methyl donor. In addition to enzyme activity, the method allows for determination of tissue contents of PNMT using human recombinant enzyme for calibration. The calibration curve for epinephrine was linear over the range of 0.1 to 5,000 pM, with 0.5 pM epinephrine representing the lower limit of quantification. The calibration curve relating PNMT to production of deuterium-labeled epinephrine was also linear from 0.01 to 100 ng PNMT. Intra- and inter-assay coefficients of variation were respectively 12.8 % ( n = 10) and 10.9 to 13.6 % ( n = 10). We established utility of the method by showing induction of the enzyme by dexamethasone in mouse pheochromocytoma cells and strong relationships to PNMT gene expression and tissue epinephrine levels in human pheochromocytomas. Development of this assay provides new possibilities for investigations focusing on regulation of PNMT, the crucial final enzyme responsible for synthesis of epinephrine, the primary fight-or-flight stress hormone. A novel quantum secure direct communication scheme based on four-qubit cluster states is proposed. In this scheme, the quantum channel between the sender Alice and the receiver Bob consists of an ordered sequence of cluster states which are prepared by Alice. After ensuring the security of quantum channel, Alice prepares the encoded Bell-state sequence, and performs Bell-basis measurements on the qubits at hand. Then Alice tells the measured results to Bob, and Bob also performs Bell-basis measurements on his own qubits. Finally Bob can get the secret information through the analysis of their measured results. In our scheme, the qubits carrying the secret message do not need to be transmitted in public channel. 41. Molecular characterization and phylogenetic analysis of Sugarcane yellow leaf virus isolates from China.

Sugarcane yellow leaf virus (SCYLV) (genus Polerovirus, family Luteoviridae), the causal agent of sugarcane yellow leaf disease (YLD), was first detected in China in 2006. To assess the distribution of SCYLV in the major sugarcane-growing Chinese provinces, leaf samples from 22 sugarcane clones ( Saccharum spp. hybrid) showing YLD symptoms were collected and analyzed for infection by the virus using reverse transcription PCR (RT-PCR), quantitative RT-PCR, and immunological assays. A complete genomic sequence (5,879 nt) of the Chinese SCYLV isolate CHN-FJ1 and partial genomic sequences (2,915 nt) of 13 other Chinese SCYLV isolates from this study were amplified, cloned, and sequenced. The genomic sequence of the CHN-FJ1 isolate was found to share a high identity (98.4-99.1 %) with those of the Brazilian (BRA) genotype isolates and a low identity (86.5-86.9 %) with those of the CHN1 and Cuban (CUB) genotype isolates. The genetic diversity of these 14 Chinese SCYLV isolates was assessed along with that of 29 SCYLV isolates of worldwide origin reported in the GenBank database, based on the full or partial genomic sequence. Phylogenetic analysis demonstrated that all the 14 Chinese SCYLV isolates clustered into one large group with the BRA genotype and 12 other reported SCYLV isolates. In addition, five reported Chinese SCYLV isolates were grouped with the Peruvian (PER), CHN1 and CUB genotypes. We therefore speculated that at least four SCYLV genotypes, BRA, PER, CHN1, and CUB, are associated with YLD in China. Interestingly, a 39-nt deletion was detected in the sequence of the CHN-GD3 isolate, in the middle of the ORF1 region adjacent to the overlap between ORF1 and ORF2. Understanding the growth and physiological responses of rice to upland conditions would be helpful for designing treatments to improve the tolerance of rice under a rainfed system. The objective of this study was to investigate the initiation, elongation and membrane stability of seminal, lateral and adventitious roots of upland rice after 9-d upland condition treatment. Compared with control roots under waterlogged conditions, upland water deficiency conditions favor seminal and lateral root growth over adventitious root growth by accelerating seminal root elongation, promoting lateral root initiation and elongation, and reducing the elongation and number of adventitious roots. Enhanced total root number and length resulted in increase of total root dry weight and thereby increasing the root-to-shoot ratio. Organic compound leakage from seminal root tips and adventitious roots increased progressively to some extent with upland culture duration, while significant increases in semial root tips were the consequence of loss of membrane integrity caused by the upland-condition enhanced growth. The efficacy of Radix Astragali membranaceus (AM) oral liquor combined with routine therapy and routine therapy alone on T-lymphocyte subsets of peripheral blood in viral myocarditis patients have been studied comparatively. The results showed that the T-lymphocyte subsets profile and OKT/OKT ratio of peripheral blood were significantly lower in viral myocarditis patients than that in healthy control ( P < 0.05, 0.01). Routine therapy combined with AM could singificantly enhance OKT, OKT and OKT/OKT ratio in the above-mentioned patients ( P <0.05, 0.01). Intestinal microbiota plays a crucial role in immune development and disease progression in mammals from birth onwards. The gastrointestinal tract of newborn mammals is rapidly colonized by microbes with tremendous biomass and diversity. Understanding how this complex of segmental communities evolves in different gastrointestinal sites over time has great biological significance and medical implications. However, most previous reports examining intestinal microbiota have focused on fecal samples, a strategy that overlooks the spatial microbial dynamics in different intestinal segments. Using intestinal digesta from six intestinal segments (duodenum, jejunum, ileum, cecum, colon and rectum) of newborn piglets, we herein conducted a large-scale 16S rRNA gene sequencing-based study to characterize the segmental dynamics of porcine gut microbiota at eight postnatal intervals (days 1, 7, 14, 21, 28, 35, 120 and 180). A total of 4,465 OTUs were obtained and showed that the six intestinal segments could be divided into three parts; in the duodenum-jejunum section, the most abundant genera included Lactobacillus and Bacteroides; in the ileum, Fusobacterium and Escherichia; and in the cecum-rectum section, Prevotella. Although the microbial communities of the piglets were similar among the six intestinal segments on postnatal day 1, they evolved and quickly differentiated at later intervals. An examination of time-dependent alterations in the dominant microbes revealed that the microbiome in the large intestine was very different from and much more stable than that in the small intestine. The gut microbiota in newborn piglets exhibited apparent temporal and spatial variations in different intestinal segments. In order to understand the genetic diversity and structure within and between the genera of Saccharum and Erianthus, 79 accessions from five species (S. officinarum, S. spontaneum, S. robustum, S. barberi, S. sinense), six accessions of E. arundinaceus, and 30 Saccharum spp. hybrids were analyzed using 21 pairs of fluorescence-labeled highly poloymorphic SSR primers and a capillary electrophoresis (CE) detection system. A total of 167 polymorphic SSR alleles were identified by CE with a mean value of polymorphic information content (PIC) of 0.92. Genetic diversity parameters among these 115 accessions revealed that Saccharum spp. hybrids were more diverse than those of Saccharum and Erianthus species. Based on the SSR data, the 115 accessions were classified into seven main phylogenetic groups, which corresponded to the Saccharum and Erianthus genera through phylogenetic analysis and principle component analysis (PCA). We propose that seven core SSR primer pairs, namely, SMC31CUQ, SMC336BS, SMC597CS, SMC703BS, SMC24DUQ, mSSCIR3, and mSSCIR43, may have a wide appicability in genotype identification of Saccharum species and Saccharum spp. hybrids.
