The development of neural circuits has long-lasting effects on brain function, yet our understanding of early circuit development in humans remains limited. Here, periodic EEG power features and aperiodic components were examined from longitudinal EEGs collected from 592 healthy 2–44 month-old infants, revealing age-dependent nonlinear changes suggestive of distinct milestones in early brain maturation. Developmental changes in periodic peaks include (1) the presence and then absence of a 9-10 Hz alpha peak between 2-6 months, (2) nonlinear changes in high beta peaks (20-30 Hz) between 4-18 months, and (3) the emergence of a low beta peak (12-20 Hz) in some infants after six months of age. We hypothesized that the emergence of the low beta peak may reflect maturation of thalamocortical network development. Infant anesthesia studies observe that GABA-modulating anesthetics do not induce thalamocortical mediated frontal alpha coherence until 10-12 months of age. Using a small cohort of infants (n = 23) with EEG before and during GABA-modulating anesthesia, we provide preliminary evidence that infants with a low beta peak have higher anesthesia-induced alpha coherence compared to those without a low beta peak. Research into Family Language Policy (FLP) focuses on language planning by and among family members within home spaces. In this study we gather data from multilingual young adults with different backgrounds across Mexico through an online questionnaire. We use a trajectory approach to look at how FLP changes over time and space from the perspective of young adults. Our data provides a diversity of young adult perspectives on the construction of linguistic trajectories, their relationship with the interventions, practices, and ideologies in the negotiation and emergence of FLPs, as well as how young adults are, or are not, using multilingual resources in digitally mediated family communication. Our findings show the dynamism of FLP over time and space, the diversity inherent in multilingual FLPs in Mexico, and how questions of mobility, generational differences, orality vs. literacy, digital access, globalization, and neoliberalism impact linguistic trajectories and FLP in Mexican multilingual young adults. Preoperative knowledge of expected postoperative pain can help guide perioperative pain management and focus interventions on patients with the greatest risk of acute pain. However, current methods for predicting postoperative pain require patient and clinician input or laborious manual chart review and often do not achieve sufficient performance. We use routinely collected electronic health record data from a multicenter dataset of 234,274 adult non-cardiac surgical patients to develop a machine learning method which predicts maximum pain scores on the day of surgery and four subsequent days and validate this method in a prospective cohort. Our method, POPS, is fully automated and relies only on data available prior to surgery, allowing application in all patients scheduled for or considering surgery. Here we report that POPS achieves state-of-the-art performance and outperforms clinician predictions on all postoperative days when predicting maximum pain on the 0–10 NRS in prospective validation, though with degraded calibration. POPS is interpretable, identifying comorbidities that significantly contribute to postoperative pain based on patient-specific context, which can assist clinicians in mitigating cases of acute pain. Purpose: The use of a tourniquet in total knee replacement has advantages and drawbacks. Some studies suggest that using ischaemia at low pressures could reduce its negative effects. Our objective is to verify whether the use of ischaemia at low pressures (100 mmHg above the systolic blood pressure) produces greater pain and loss of strength than surgery without a tourniquet. Methods: By the means of a prospective randomized clinical trial, patients were assigned to the control group (no tourniquet, NT) or the experimental group (tourniquet, T). The main variables measured were pain (VAS) and isometric muscle strength (preoperatively, 10 days and 3 months after surgery). Secondary variables were haemoglobin at 24 h, transfusion index, need for rescue drugs and days of admission. Results: A total of 71 patients (73 prosthesis) were studied. Both groups were homogeneous in terms of age, body mass index, sex ratio, preoperative strength and level of anesthetic risk. We did not find significative differences in any of the main variables (pain and strength) nor in the secondary ones. We could only find differences in the days of admission (2.77 vs. 3.05; p = 0.031). Conclusions: Use of a tourniquet at low pressures (100 mmHg above systolic blood pressure) did not result in an increase in postoperative pain or a decrease in quadriceps extension force within the first 3 months after surgery. Nowadays, Artificial Intelligence (AI) is growing by leaps and bounds in almost all fields of technology, and Autonomous Vehicles (AV) research is one more of them. This paper proposes the using of algorithms based on Deep Learning (DL) in the control layer of an autonomous vehicle. More specifically, Deep Reinforcement Learning (DRL) algorithms such as Deep Q-Network (DQN) and Deep Deterministic Policy Gradient (DDPG) are implemented in order to compare results between them. The aim of this work is to obtain a trained model, applying a DRL algorithm, able of sending control commands to the vehicle to navigate properly and efficiently following a determined route. In addition, for each of the algorithms, several agents are presented as a solution, so that each of these agents uses different data sources to achieve the vehicle control commands. For this purpose, an open-source simulator such as CARLA is used, providing to the system with the ability to perform a multitude of tests without any risk into an hyper-realistic urban simulation environment, something that is unthinkable in the real world. The results obtained show that both DQN and DDPG reach the goal, but DDPG obtains a better performance. DDPG perfoms trajectories very similar to classic controller as LQR. The long-term ecological dynamics of a population inhabiting a seasonal environment is analyzed using a semi-discrete or impulsive system to represent the consumer–resource interaction. The resource corresponds to an incoming energy flow for consumers that is allocated to reproduction as well as to maintenance in each non-reproductive season. The energy invested in these life-history functions is used in reproductive events, determining the size of the offspring in each reproductive season. Two long-term dynamic patterns are found, resulting in either the persistence or the extinction of the population of consumers. In addition, our model indicates that only one energy allocation strategy provides an optimal combination between individual consumption and long-term population size. The current study contributes to the understanding of how the individual-level and the population-level are interrelated, exhibiting the importance of incorporating phenotypic traits in population dynamics. Cholesterol Gallstone Disease (GSD) is a common multifactorial disorder characterized by crystallization and aggregation of biliary cholesterol in the gallbladder. The global prevalence of GSD is ~10–20% in the adult population but rises to 28% in Chile (17% among men and 30% among women). The small intestine may play a role in GSD pathogenesis, but the molecular mechanisms have not been clarified. Our aim was to identify the role of the small intestine in GSD pathogenesis. Duodenal biopsy samples were obtained from patients with GSD and healthy volunteers. GSD status was defined by abdominal ultrasonography. We performed a transcriptome study in a discovery cohort using Illumina HiSeq. 2500, and qPCR, immunohistochemistry and immunofluorescence were used to validate differentially expressed genes among additional case-control cohorts. 548 differentially expressed genes between GSD and control subjects were identified. Enriched biological processes related to cellular response to zinc, and immune and antimicrobial responses were observed in GSD patients. We validated lower transcript levels of metallothionein, NPC1L1 and tight junction genes and higher transcript levels of genes involved in immune and antimicrobial pathways in GSD patients. Interestingly, serum zinc and phytosterol to cholesterol precursor ratios were lower in GSD patients. A significant association was observed between serum zinc and phytosterol levels. Our results support a model where proximal small intestine plays a key role in GSD pathogenesis. The molecular mechanisms underlying inclination responses in trees are unclear. In this study, we identified a MADS-box transcription factor differentially expressed early after inclination in the stems of Pinus radiata D. Don. PrMADS10 has a CDS of 582 bp and encodes a group II MADS-box transcription factor. We measured highest accumulation of this transcript on the lower side of inclined pine stems. In an effort to identify putative targets, we stably transformed Arabidopsis thaliana with a 35S::PrMADS10 construct. Transcriptome analysis revealed 1,219 genes differentially-expressed, with 690 and 529 genes up- and down-regulated respectively, when comparing the transgenic and wild-type. Differentially-expressed genes belong to different biological processes, but were enriched in cell wall remodeling and phenylpropanoid metabolic functions. Interestingly, lignin content was 30% higher in transgenic as compared to wild-type plants consistent with observed changes in gene expression. Differentially expressed transcription factors and phenylpropanoid genes were analyzed using STRING. Several MYB and NAC transcription factors showed interactions with genes of the phenylpropanoid pathway. Together, these results implicate PrMADS10 as a regulatory factor, triggering the expression of other transcription factors and genes involved in the synthesis of lignin. [ABSTRACT FROM AUTHOR]

Traumatic brain injury (TBI) is the most important cause of disability in individuals under the age of 45 years and thus represents a significant social and economic burden. Evidence strongly suggests that oxidative stress is a cornerstone event leading to and propagating secondary injury mechanisms such as excitotoxicity, mitochondrial dysfunction, apoptosis, autophagy, brain edema, and inflammation. TBI has defied conventional approaches to diagnosis and therapy development because of its heterogeneity and complexity. Therefore, it is necessary to explore alternative approaches to therapy development for TBI. The aim of this review is to present a therapeutic approach for TBI, taking into account the evidence supporting the role for oxidative stress in the pathophysiological processes of secondary brain injury. The role of agents such as mitochondria-targeted antioxidants (melatonin and new mitochondria-targeted antioxidants), nicotinamide adenine dinucleotide phosphate (NADPH) inhibitors (antioxidant vitamins and apocynin), and other compounds having mainly antioxidant properties (hydrogen-rich saline, sulforaphane, U-83836E, omega-3, and polyphenols) is covered. The rationale for innovative antioxidant therapies based on current knowledge and particularly the most recent studies regarding this field is discussed. Preeclampsia is a high-prevalence systemic pregnancy disorder associated with maternal and foetal mortality. Its pathogenesis is unknown, but it is thought that oxidative stress and endothelial dysfunction may play a fundamental role. Von Willebrand factor (vWF), a marker of endothelial cell injury, can be found in different cells and zones of the placenta. To determine the differential immunoexpression of vWF at different tissue types of preeclamptic placenta and endothelial dysfunction markers at maternal serum of preeclamptic pregnancies. A case-control study was performed on a population of pregnant women with preeclampsia (n = 14), and normal pregnancies (n = 8). Placental and blood plasma samples were withdrawn at delivery. Immunohistochemical vWF expression in the placental tissue was determined. Endothelial dysfunction was assessed through plasminogen activator inhibitor (PAI) 1 and 2 ratio and vWF concentration in maternal plasma. P values less than 0.05 were considered statistically significant. Preeclamptic women showed increased plasma PAI-1/PAI-2 ratio ( P < 0.05). There was diminished placental vWF expression in syncytiotrophoblast and increased in the intervillous space of preeclamptic placentas ( P < 0.05). No significant differences in vWF expression were found in the villous endothelium and stroma, but it was significantly higher in maternal plasma ( P < 0.05). In preeclampsia occurs endothelial damage and placental cell injury. Cell damage in syncytiotrophoblast that occurs in preeclampsia could liberate vWF from syncytiotrophoblast to the placental intervillous space, and this may have pathogenic implications. This study examined the participation of a group of middle school students in an after-school mathematics club as they worked on cryptography problems. The analysis focused on interactions characterized by collective problem-solving activity, when intellectual work was distributed and various students took on active problem-solving roles, paying particular attention to intersections between task structures and positioning moves. We found that all open-ended tasks—those tasks that afforded multiple strategies and had multiple solutions—resulted in at least some collective problem solving, though it was not always sustained (Turner, Gutiérrez, & Sutton, 2009). We also found that the task structures, in combination with interactive positioning moves by facilitators and students, served to sustain or disrupt collective problem-solving activity. In this study, genome-wide expression profiling based on Affymetrix ATH1 arrays was used to identify discriminating responses of Arabidopsis thaliana to five herbicides, which contain active ingredients targeting two different branches of amino acid biosynthesis. One herbicide contained glyphosate, which targets 5-enolpyruvylshikimate- 3-phosphate synthase (EPSPS), while the other four herbicides contain different acetolactate synthase (ALS) inhibiting compounds. In contrast to the herbicide containing glyphosate, which affected only a few transcripts, many effects of the ALS inhibiting herbicides were revealed based on transcriptional changes related to ribosome biogenesis and translation, secondary metabolism, cell wall modification and growth. The expression pattern of a set of 101 genes provided a specific, composite signature that was distinct from other major stress responses and differentiated among herbicides targeting the same enzyme (ALS) or containing the same chemical class of active ingredient (sulfonylurea). A set of homologous genes could be identified in Brassica napus that exhibited a similar expression pattern and correctly distinguished exposure to the five herbicides. Our results show the ability of a limited number of genes to classify and differentiate responses to closely related herbicides in A. thaliana and B. napus and the transferability of a complex transcriptional signature across species. The technique RT-qPCR for viral RNA detection is the current worldwide strategy used for early detection of the novel coronavirus SARS-CoV-2. RNA extraction is a key pre-analytical step in RT-qPCR, often achieved using commercial kits. However, the magnitude of the COVID-19 pandemic is causing disruptions to the global supply chains used by many diagnostic laboratories to procure the commercial kits required for RNA extraction. Shortage in these essential reagents is even more acute in developing countries with no means to produce kits locally. We sought to find an alternative procedure to replace commercial kits using common reagents found in molecular biology laboratories. Here we report a method for RNA extraction that takes about 40 min to complete ten samples, and is not more laborious than current commercial RNA extraction kits. We demonstrate that this method can be used to process nasopharyngeal swab samples and yields RT-qPCR results comparable to those obtained with commercial kits. Most importantly, this procedure can be easily implemented in any molecular diagnostic laboratory. Frequent testing is crucial for individual patient management as well as for public health decision making in this pandemic. Charting a temporal path in gene networks requires linking early transcription factor (TF)-triggered events to downstream effects. We scale-up a cell-based TF-perturbation assay to identify direct regulated targets of 33 nitrogen (N)-early response TFs encompassing 88% of N-responsive Arabidopsis genes. We uncover a duality where each TF is an inducer and repressor, and in vitro cis-motifs are typically specific to regulation directionality. Validated TF-targets (71,836) are used to refine precision of a time-inferred root network, connecting 145 N-responsive TFs and 311 targets. These data are used to chart network paths from direct TF1-regulated targets identified in cells to indirect targets responding only in planta via Network Walking. We uncover network paths from TGA1 and CRF4 to direct TF2 targets, which in turn regulate 76% and 87% of TF1 indirect targets in planta, respectively. These results have implications for N-use and the approach can reveal temporal networks for any biological system. The rhizosphere is considered the primary place for soil microbiome differentiation and plays a key role in plant survival, especially for those subjected to environmental stress. Using high-throughput sequencing of the 16S rRNA gene, we analyzed and compared soil bacterial communities associated to four of the most abundant high altitude native plant species of the Chilean Andean grasslands. We examined three soil compartments: the rhizosphere (bacteria firmly attached to the roots), the rhizosphere-surrounding soil (bacteria loosely attached to the roots) and the bulk soil (plant-free soil). The rhizosphere microbiome was in all cases the least diverse, exposing that the bulk soil was a more complex environment. Taxonomic analysis revealed an abrupt change between the rhizosphere and the rest of the non-rhizospheric soils. Thus, while rhizobacterial communities were enriched in Proteobacteria (mainly Alphaproteobacteria), Actinobacteria (mostly Blastocatellia) dominated in bulk soils. Finally, we detected certain taxonomic rhizosphere signatures, which could be attributed to a particular genotype. Overall, our results indicate that the thin layer of soil surrounding the roots constitute a distinctive soil environment. This study contributes to expand the knowledge about soil bacterial communities in the Chilean highlands and takes the first step to understand the processes that might lead to the rhizosphere differentiation in that area. Modal properties of dynamically tested wind turbine blades (WTBs) of a utility-scale wind turbine are identified. A comprehensive experimental program including free vibration and short- and long-term forced vibrations representing resonance and simplified fatigue conditions was carried out to investigate vibration-based features for damage diagnosis and prognosis. A set of 12 undamaged WTBs were tested to study the variability of the identified modal parameters. Results indicate that the variability of the natural frequencies was rather low, while the obtained damping ratios exhibited significant differences. Forced vibration tests were then conducted. To reach the failure of the blades, approximately 1.9 × 104 and 4.2 × 107 cycles were induced in the short- and long-term tests, respectively. Modal properties identified during testing protocols suggest that natural frequencies correlate well with damage. A linear finite element model was also developed, and its modal properties are compared to the identified modal parameters of the undamaged blades. [ABSTRACT FROM AUTHOR]

Future climate change has the potential to alter the distribution and prevalence of plant pathogens, which may have significant implications for both agricultural crops and natural plant communities. However, there are few long-term datasets against which modelled predictions of pathogen responses to climate change can be tested. Here, we use 18S metabarcoding of 28 rodent middens (solidified deposits of rodent coprolites and nesting material) from the Central Atacama, spanning the last ca. 49 ka, to provide the first long-term late Quaternary record of change in plant pathogen communities in response to changing climate. Plant pathogen richness was significantly greater in middens deposited during the Central Andean Pluvial Event (CAPE); a period of increased precipitation between 17.5-8.5 ka. Moreover, the occurrence frequency of Pucciniaceae (rust fungi) was significantly greater during the CAPE, and the highest relative abundances for five additional potentially pathogenic taxa also occurred during this period. The results demonstrate the promising potential for ancient DNA analysis of late Quaternary samples to reveal insights into how plant pathogens responded to past climatic and environmental change, which could help predict how pathogens may responded to future change. Gas breakthrough pressure is a significant parameter for the gas exploration and safety evaluation of engineering barrier systems in the carbon dioxide storage, remediation of contaminated sites, and deep geological repository for disposal of high-level nuclear waste, etc. Test for determining gas breakthrough pressure is very difficult and time-consuming, due to the low/ultra-low conductivity of the specimen. It is also difficult to get a comprehensive and high-precision model based on limited results obtained through individual experiments, as the measurements of gas breakthrough pressure were influenced by many factors. In this study, a collected database was built that covered a lot of former test data, and then, two models were developed by the random forest (RF) algorithm and multiexpression programming (MEP) method. The MEP model constructed with explicit expressions for the gas breakthrough pressure overcame the drawbacks of common "black box" models. Meanwhile, five significant indicators were selected from ten common features using the permutation importance algorithm. The RF model was interpreted by the Shapley value and the PDP/ICE plots, while the MEP model was analyzed through the proposed explicit expression, showing strong consistence with that in former studies. Hydrated bentonite is considered an alternative subsurface sealing/plugging material for deep geo-energy wells. However, the knowledge related to this application and the corresponding properties of bentonite is still lacking. This includes the mechanical properties at the interface of bentonite plugs with the adjacent materials (surrounding rock or casing steel) and the mechanical stability of plugs under downhole in-situ conditions. In this work, we performed experiments investigating the interface shear properties and shear strength of a bentonite plug under various settings for deep geo-energy applications, such as hydrocarbon and geothermal wells. The interface's shear properties against various adjacent materials and fluid conditions were characterized. The influence of chemical exposure, the salinity of the curing fluid, fluid pH, pressure, and temperature on bentonite's mechanical stability was evaluated in a small- and large-scale setting. The latter was performed using realistic casing sizes and placement methods, relevant for the field application. The experimental results show that the averaged shear strength of the bentonite plug interface is 13.3 kPa and 9.1 kPa when cured in freshwater and seawater, respectively. The increase in strength with increasing curing pressure, temperature, and fluid pH was characterized for the first time. The interfacial properties of cohesion and friction angle vary with different surrounding materials. They are also influenced by the saturating condition and salinity of the saturating fluid. Based on the experimental results, a bentonite plug with a minimum length of 15–43 m placed in casings of 7–5/8″ to 13–3/8″ would be sufficient to meet the necessary criteria of the Dutch regulators as an isolating material for well abandonment. To ensure the long-term safety and performance efficiency of a deep geological repository for disposal of high-level radioactive waste, understanding and assessment of gas migration behaviour in its engineered barrier system are of significant importance. In this study, a coupled gas transport model is utilised to simulate and to analyse a series of gas injection/breakthrough experiments on saturated bentonite under rigid boundary or constant volume conditions. To explain the laboratory gas migration and breakthrough results, a diffusion and solubility-controlled gas transport mechanism, instead of controversial visco-capillary flow or dilatancy-controlled flow mechanisms, is implemented in the model. The aim is to examine the extent to which this mechanism can describe helium migration and breakthrough behaviours in rigidly confined, saturated bentonite specimens. The predicted results are found to be in good agreement, both qualitatively and quantitatively, with the observed experimental results indicating the adequacy of this mechanism to describe the transport processes. The model represents the gas breakthrough phenomenon as a function of gas solubility. When the dissolved concentration of the injected gas reaches the maximum soluble concentration in the entire porewater domain, gas breakthrough occurs. Since, the system reaches a steady state and no further gas can be dissolved in the rigidly confined specimens, any injected/dissolved gas must be equated by the amount dissipated to comply with the principle of mass conservation. The maximum solution concentrations of helium are predicted to be 2.01 × 10–5, 7.75 × 10–5, 1.07 × 10–4 mol/L for GMZ bentonite specimens with dry densities of 1.3, 1.5 and 1.7 g/cm3, respectively. The analysis of the injection pressure effects on gas migration behaviour revealed that, if sufficient time is permitted, gas breakthrough may occur at pressures lower than the laboratory observed injection pressures. Unlike regular elastic materials, when auxetic materials are compressed, they become thinner in the direction perpendicular to the applied force. Despite their outstanding mechanical properties, a systematic design of new and controlled auxetics remains underdeveloped. Here we establish a unified framework to describe bidimensional perfect auxetics with potential use in the design of new materials. Inspired by a natural connection between rotating rigid units and antiferromagnetic spin systems, we unveil the conditions for the emergence of a non-trivial floppy mode responsible for the auxetic behaviour. This model establishes three simple steps to design new auxetics. In particular, we constructed an exotic crystal, a Penrose quasi-crystal and the long-desired isotropic auxetic. The auxeticity of these designs is robust under small structural disturbances, as seen from experiments and numerical simulations. We expect that this work will allow the implementation of auxetic behaviour into advanced materials to enhance their functionalities, with a promising extension into 3D auxetics. Auxetics are an unusual family of materials that, for instance, when stretched in a particular direction will exhibit an expansion of the dimensions that are perpendicular to the applied stress; however, despite many known examples of auxectics there is no universal description of the material properties. Here, the authors report a model based on antiferromagnetic spins and demonstrate how this can be used to design a auxetic material with a Poisson ratio of -1 over a range of finite strain. Metal mining extraction and concentration need water. The recycling of liquid effluents reduces its consumption. This recycled water must contain low concentrations of metals. This project's first objective was to start up a laboratory-scale upflow anaerobic sludge blanket (UASB) reactor for biotransforming metal sulfates into metal sulfides of effluent from flotation units of a mining plant (FE). The second objective was to determine the effect of pH and chemical oxygen demand:sulfate ratio (COD:SO42−) in the precipitation of heavy metal sulfides. The third objective, which is the subject of this part of the research, was to evaluate the feasibility of this proposal through a mass balance of the metals separated by precipitation through the formation of sulfides by sulfate-reducing microorganisms (SRM), using as energy source carbonaceous compounds from flotation residual organic reagents enriched with lactic acid. To monitor bioconversion effectiveness, various parameters were used: pH-alkalinity factor (α), temperature (T), COD, SO42−, and sulfides (S2−). Four metals were considered for this part of the research: Cu, Pb, Zn, and Fe. The UASB system achieved a sulfate bioconversion of 69% and an organic matter removal as COD of 88% after transient state with the best COD:SO42− ratio found. Using a statistical analysis by clusters, metal sulfide production of 74 mg L−1 and removals of 39, 70, 79, and 65%, for Pb, Cu Zn, and Fe, respectively, were obtained. These values were calculated with an initial ratio of COD:SO42− of 0.66 ± 0.2 and pH values around 6 inside the reactor, through anaerobic microbial biomass, indicating that the solution proposed to recycle water in the metal extraction and concentration is feasible after using these systems. Main conclusion: The plasticity of plant growth response to differing nitrate availability renders the identification of biomarkers difficult, but allows access to genetic factors as tools to modulate root systems to a wide range of soil conditions. Nitrogen availability is a major determinant of crop yield. While the application of fertiliser substantially increases the yield on poor soils, it also causes nitrate pollution of water resources and high costs for farmers. Increasing nitrogen use efficiency in crop plants is a necessary step to implement low-input agricultural systems. We exploited the genetic diversity present in the worldwide Arabidopsis thaliana population to study adaptive growth patterns and changes in gene expression associated with chronic low nitrate stress, to identify biomarkers associated with good plant performance under low nitrate availability. Arabidopsis accessions were grown on agar plates with limited and sufficient supply of nitrate to measure root system architecture as well as shoot and root fresh weight. Differential gene expression was determined using Affymetrix ATH1 arrays. We show that the response to differing nitrate availability is highly variable in Arabidopsis accessions. Analyses of vegetative shoot growth and root system architecture identified accession-specific reaction modes to cope with limited nitrate availability. Transcription and epigenetic factors were identified as important players in the adaption to limited nitrogen in a global gene expression analysis. Five nitrate-responsive genes emerged as possible biomarkers for NUE in Arabidopsis. Seagrasses are key marine foundation species, currently declining due to the compounded action of global and regional anthropogenic stressors. Eutrophication has been associated with seagrass decline, while grazing has been traditionally considered to be a natural disturbance with a relatively low impact on seagrasses. In the recent years, this assumption has been revisited. Here, by means of a 16-month field-experiment, we investigated the molecular mechanisms driving the long-term response of Posidonia oceanica to the combination of nutrient enrichment, either as a chronic (press) or pulse disturbance, and herbivory. Changes in expression levels of 19 target genes involved in key steps of photosynthesis, nutrient assimilation, chlorophyll metabolism, oxidative-stress response and plant defense were evaluated through reverse transcription-quantitative polymerase chain reaction (RT-qPCR). High herbivore pressure affected the molecular response of P. oceanica more dramatically than did enhanced nutrient levels, altering the expression of genes involved in plant tolerance and resistance traits, such as photosynthesis and defense mechanisms. Genes involved in carbon fixation and N assimilation modulated the response of plants to high nutrient levels. Availability of resources seems to modify P. oceanica response to herbivory, where the upregulation of a nitrate transporter gene was accompanied by the decline in the expression of nitrate reductase in the leaves, suggesting a change in plant-nutrient allocation strategy. Finally, press and pulse fertilizations altered nitrate uptake and reduction-related genes in opposite ways, suggesting that taking into account the temporal regime of nutrient loading is important to assess the physiological response of seagrasses to eutrophication. Evaporation tests in concrete columns have been analysed by numerical models to characterize the thermo-hydraulic properties and the processes in concrete. Two evaporation tests were performed: a column heated by a lamp and a column kept in room conditions. The conceptual model considers unsaturated liquid flow and transport of vapour and energy. We also calculated models that take into account the dissolved salts to study its effect on vapour pressure and evaporation. A retention curve has been obtained from relative humidity and gravimetric water content measured after dismantling the tests. The models have been calibrated by adjusting the model's results to the measured data of water loss, relative humidity and temperature inside the concrete. The parameters obtained with the calibration are the permeability, thermal conductivity, boundary conditions and a tortuosity factor for vapour diffusion. Results show that the vapour diffusion is the dominant water transport process above an evaporation front, and liquid advection is dominant below it. [ABSTRACT FROM AUTHOR]

A Y-linked two-sex branching process with blind choice is a suitable model for analyzing the evolution of the number of carriers of two alleles of a Y-linked gene in a two-sex monogamous population where each female chooses her partner from among the male population without caring about his type (i.e., the allele he carries). This work focuses on the development of Bayesian inference for this model, considering a parametric framework with the reproduction laws belonging to the power series family of distributions. A sample is considered given by the observation of the total number of females and males (regardless of their types) up to some generation as well as the number of each type of male in the last generation. Using a simulation method based on the Gibbs sampler, we approximate the posterior distributions of the main parameters of this model. Assembling composite DNA modules from custom DNA parts has become routine due to recent technological breakthroughs such as Golden Gate modular cloning. Using Golden Gate, one can efficiently assemble custom transcription units and piece units together to generate higher-order assemblies. Although Golden Gate cloning systems have been developed to assemble DNA plasmids required for experimental work in model species, they are not typically applicable to organisms from other kingdoms. Consequently, a typical molecular biology laboratory working across kingdoms must use multiple cloning strategies to assemble DNA constructs for experimental assays. To simplify the DNA assembly process, we developed a multi-kingdom (MK) Golden Gate assembly platform for experimental work in species from the kingdoms Fungi, Eubacteria, Protista, Plantae, and Animalia. Plasmid backbone and part overhangs are consistent across the platform, saving both time and resources in the laboratory. We demonstrate the functionality of the system by performing a variety of experiments across kingdoms including genome editing, fluorescence microscopy, and protein interaction assays. The versatile MK system therefore streamlines the assembly of modular DNA constructs for biological assays across a range of model organisms. [ABSTRACT FROM AUTHOR]
