This paper addresses the self-supervised learning of several critical factors in Visual Simultaneous Localization and Mapping (VSLAM) in low-level vision: interest point learning, descriptor learning, ego-motion estimation, and depth estimation. The key insight we have is that appearance and geometry constraints can be used to couple these fundamental vision issues. We propose a self-supervised framework for joint training of neural networks for multiple objectives to address complicated issues, simplify systems, and provide important information for deep monocular VSLAM systems. First, we input two adjacent images into pose and depth networks to obtain their corresponding depth maps and camera poses. Then, we employ a differentiable geometry module and utilize the depth maps and camera poses to generate pseudo-input images needed for the interest point network and construct the geometry loss. Further, we input the pseudo-input image and source image into the interest point network to obtain the corresponding interest points, descriptors, and scores. Subsequently, we construct the appearance loss. Finally, we combine the geometry and appearance losses to constrain the whole network in an unsupervised manner. The novelty of this paper is that it integrates the key information necessary in monocular VSLAM into a unified framework that takes into account interest point learning, descriptor learning, ego-motion estimation, and depth estimation at the same time. Host immune responses are tightly controlled by various immune factors during infection, and protozoan parasites also manipulate the immune system to evade surveillance, leading to an evolutionary arms race in host‒pathogen interactions; however, the underlying mechanisms are not fully understood. We observed that the level of superoxide dismutase 3 (SOD3) was significantly elevated in both Plasmodium falciparum malaria patients and mice infected with four parasite species. SOD3-deficient mice had a substantially longer survival time and lower parasitemia than control mice after infection, whereas SOD3-overexpressing mice were much more vulnerable to parasite infection. We revealed that SOD3, secreted from activated neutrophils, bound to T cells, suppressed the interleukin-2 expression and concomitant interferon-gamma responses crucial for parasite clearance. Overall, our findings expose active fronts in the arms race between the parasites and host immune system and provide insights into the roles of SOD3 in shaping host innate immune responses to parasite infection. Scale ambiguity is a difficulty in monocular visual odometry. This challenge has not yet been well solved, regardless of whether it is a traditional method or a learning-based method. Simultaneously, the performance of unsupervised monocular visual odometry in tricky situations is limited. To tackle these issues, we propose an accurate and efficient end-to-end system that can favorably deal with the problem of scale ambiguity in unsupervised monocular visual odometry, particularly in long-sequence videos in challenging environments. To acquire the depth map and optical flow, we first employ the depth network and the optical flow network. Then, we combine the optical flow information and the camera height using a ground plane geometric model to recover the absolute scale of the camera translation and depth map using our scale recovery algorithm. Finally, we use the recovered scale and our flow-depth loss to unify the training and testing processes across the entire network. Extensive experiments and analyses reveal that, when compared to existing unsupervised approaches, our method achieves state-of-the-art performance on the KITTI dataset not only for depth estimation and optical flow estimation but also for monocular visual odometry. As a new promising detection technology in the terahertz research field, the terahertz time-domain spectroscopy (THz-TDS) has very broad application potential in many fields because its advantage on the characteristic spectrum, wide spectrum and non-destructive analysis of interested substances. In this paper, the terahertz absorption spectra of gases mixed with 12CO and 13CO in the spectrum range of 0.5–2.5 THz are measured by terahertz time-domain spectroscopy for the first time. Several isotopologues can be clearly distinguished based on the difference in their rotational energies and the consequent terahertz spectrum. The experimental results show that 12CO and 13CO have obvious characteristic absorption peaks in the spectrum range of 0.5–2.5 THz due to the difference in rotational energy, and the rotational constant B can be calculated according to the experimental values to distinguish the two gaseous isotopologues. The frequency positions of the characteristic absorption peak measured by this experiment and the rotation constant B calculated according to the experimental values are compared with those previous theoretical calculations and experimental results, and they are in good agreement. Ticks are vectors and reservoirs of a variety of pathogens including protozoa, bacteria and viruses which cause tick-borne diseases (TBDs) in humans and livestock. TBDs pose serious constraints to the improvement of livestock production in tropical and subtropical regions of the world. Despite their wide distribution, information on the tick and pathogen relationship is scarce in Tanzania. We used nested PCR and sequencing to screen pathogens of public and veterinary health importance in ticks collected by flagging from four districts of Tanzania. In total, 2021 ticks comprising nine species were identified. DNA from ticks was pooled according to tick species, developmental stage, and location, then screened for Babesia bigemina, Babesia bovis, Theileria parva and Coxiella burnetii. Out of 377 pools, 34.7% were positive for at least one pathogen. Theileria parva was the most abundant with a minimum infection rate (MIR) of 2.8%, followed by B. bigemina (MIR = 1.8%) and B. bovis (MIR = 0.8%). Multiple pathogens detection was observed in 7.2% of the tested pools. However, PCR screening of individual tick DNA revealed that only 0.3% of the examined pools had co-infection. DNA of C. burnetii was never detected in any tick DNA pool. The MIR of tick-borne pathogens (TBPs) differed significantly among districts, seasons, tick species, and tick developmental stages. Sequence analysis showed that B. bigemina RAP-1a, B. bovis SBP-4, and T. parva p104 genes were conserved among pathogens in the four districts. Despite the absence of C. burnetii in ticks, considering its pathogenic potential, it is essential to continue monitoring for its possible recurrence in ticks. Dihydroartemisinin (DHA), a potent antimalarial drug, also exhibits distinct property in modulation on Treg and B cells, which has been recognized for decades, but the underlying mechanisms remain understood. Herein we revealed that DHA could promote Treg proliferation, meanwhile, suppress B cell expansion in germinal centers, and consequently decrease the number of circulating plasma cells and the content of serum immunoglobulins. Further, DHA-activated Treg significantly mitigated lipopolysaccharide-induced and malaria-associated inflammation. All these scenarios were attributed to the upregulation of c-Fos expression by DHA and enhancement of its interaction with target genes in both Treg and circulating plasma cells with bilateral cell fates. In Treg, the c-Fos-DHA complex upregulated cell proliferation-associated genes and promoted cell expansion; whereas in plasma cells, it upregulated the apoptosis-related genes resulting in decreased circulating plasma cells. Thus, the bilateral immunoregulatory mechanism of DHA was elucidated and its application in the treatment of autoimmune diseases is further justified. The scramjet isolator length is a very important influence factor on the stability of scramjet engines. The effects of the isolator length on the pseudo-shock wave (PSW) were studied through a numerical method. The steady and unsteady PSW characteristics were analyzed to explore the influence mechanism of the isolator length. The numerical model was validated with the existing experimental results, and the validation results show that the numerical model has high accuracy. The effects of the structural variation of the PSW on self-excited oscillation were explored at different isolator lengths. Result shows that increasing the isolator length increases the length of the PSW. The leading shock becomes stable with the increasing of PSW length, and the following shocks become unstable. This phenomenon indicates that the unsteady characteristics of self-excited oscillation are closely related to the PSW structure. The immunomodulatory potential of dihydroartemisinin (DHA) has recently been highlighted; however, the potential mechanism remains to be clarified. Single-cell RNA sequencing was explored in combination with cellular and biochemical approaches to elucidate the immunomodulatory mechanisms of DHA. In this study, we found that DHA induced both spleen enlargement and rearrangement of splenic immune cell subsets in mice. It was revealed that DHA promoted the reversible expansion of effective regulatory T cells and interferon-γ+ cytotoxic CD8+ T cells in the spleen via induction of superoxide dismutase 3 (SOD3) expression and increased phosphorylation of c-Jun N-terminal kinases (JNK) and its downstream activator protein 1 (AP-1) transcription factors. Further, SOD3 knockout mice were resistant to the regulatory effect of DHA. African trypanosomatid parasites escape host acquired immune responses through periodic antigenic variation of their surface coat. In this study, we describe a mechanism by which the parasites counteract innate immune responses. Two TatD DNases were identified in each of Trypanosoma evansi and Trypanosoma brucei. These DNases are bivalent metal-dependent endonucleases localized in the cytoplasm and flagella of the parasites that can also be secreted by the parasites. These enzymes possess conserved functional domains and have efficient DNA hydrolysis activity. Host neutrophil extracellular traps (NETs) induced by the parasites could be hydrolyzed by native and recombinant TatD DNases. NET disruption was prevented, and the survival rate of parasites was decreased, in the presence of the DNase inhibitor aurintricarboxylic acid. Lithium-nickel-cobalt-aluminum (NCA) material coated by a 10-nm thick LiTiO2 (LTO) layer was prepared using a method based on hydrolysis combined with a solvothermal process. The effects of LTO modifications on the structure, morphology, gas generation, and electrochemical performance of this NCA material were investigated by X-ray diffraction, scanning electron microscopy, transmission electron microscopy, Gas Chromatography-Mass Spectrometer, and electrochemical tests. The results vividly showed that the LTO layer was uniformly coated on NCA surfaces and titanium addition (1 wt%) for these modified results was crucial. The residual lithium on the surface of NCA material was efficiently resumed by LTO synthesis reaction without extra effects on the morphology and structure of NCA material. LTO treatment can significantly improve the processability, gas production performance, and the electrochemical properties of NCA material. After cycling 100 times at 25 °C with the current rate of 1C, optimized NCAT1 electrodes maintained a high discharge capacity at 164.6 mA·h·g−1 compared to NCA electrodes at 152.3 mA·h·g−1. On the other hand, the high-rate discharge capacity of NCAT1 electrodes at 10 C was also maintained at 135.0 mA·h·g−1, contrasted with NCA electrodes at only 125.0 mA·h·g−1. The enhancement of electrochemical performance was ascribed to the LTO-coating layer, which improved ionic conductivity, reduced interface resistance, and protected the core NCA material. Pseudo-shock waves in a scramjet isolator are worth studying. These waves are generally subjected to back pressure fluctuation induced by combustion. The characteristics and unsteadiness mechanism of pseudo-shock waves are investigated using a numerical method. A second nozzle with a movable pintle is utilized to simulate the back pressure fluctuation of a combustion chamber in the numerical model. A 2D numerical method with the kω-shear stress transport turbulence model is adopted to investigate the behavior of pseudo-shock waves. Validation of the numerical model is performed by comparing its results with existing experimental results. The accuracy of the numerical model is proven. The present study aims to investigate the unsteady characteristics of pseudo-shock waves in a scramjet isolator. As indicated by the results, the amplitude of pseudo-shock wave motion can be reduced by increasing the exciting frequency of back pressure fluctuation. Artemisia annua is an anti-fever herbal medicine first described in traditional Chinese medicine 1,000 years ago. Artemisinin, the extract of A. annua, and its derivatives (dihydroartemisinin (DHA), artemether, and artesunate) have been used for the treatment of malaria with substantial efficacy. Recently, DHA has also been tested for the treatment of lupus erythematosus, indicating that it may function to balance the immune response in immunocompromised individuals. In the present study, the regulatory effect of artemisinin on the murine immune system was systematically investigated in mice infected with two different protozoan parasites (Toxoplasma gondii and Plasmodium berghei). Our results revealed that the mouse spleen index significantly increased (spleen enlargement) in the healthy mice after DHA administration primarily due to the generation of an extra number of lymphocytes and CD8+ T lymphocytes in both the spleen and circulation. DHA could increase the proportion of T helper cells and CD8+ T cells, as well as decrease the number of splenic and circulatory B cells. Further, DHA could reduce the production of proinflammatory cytokines. Equine piroplasmosis (EP) is a severe disease of horses caused by the tick-borne protozoa Theileria equi (T. equi) and Babesia caballi (B. caballi). Infectious carriers are not always symptomatic, meaning there is a risk to non-enzootic areas. Regulatory tests for EP include sero-epidemiological methods for equine babesiosis, but these lack specificity due to cross-reactivity with other Babesia species. In this study, we present a real-time quantitative recombinase polymerase amplification (qRPA) method for fast simultaneous detection of both T. equi and B. caballi. In this method, primers and probes targeting the 18S rRNA gene of both T. equi and B. caballi, the ema-1 gene of T. equi and the bc48 gene of B. caballi were designed and evaluated. The sensitivity of qRPA was evaluated using the pUC57 plasmid DNA containing the target gene. For the pUC57-bc48 gene DNA, the R2 value was 0.983 for the concentration range 0.2 ng (4.1 × 107 DNA copies) to 2.0 fg (4.1 × 101 DNA copies). For the pUC57-ema gene DNA, the R2 value was 0.993 for the concentration range 0.2 ng (5.26 × 107 DNA copies) to 2.0 fg (5.26 × 102 DNA copies). For the pUC57-Bc18S gene DNA the R2 value was 0.976 for the concentration range 2.0 ng (4.21 × 108 DNA copies) to 2.0 fg (4.21 × 102 DNA copies). For the pUC57-Te18S gene DNA, the R2 value was 0.952 (Fig. S3b) for the concentration range 2.0 ng (4.16 × 108 DNA copies) to 2.0 fg (4.16 × 102 DNA copies). Furthermore, a duplex qRPA analysis was developed and optimized and the results showed that primers and probes targeting for the bc48 gene of B. caballi and the 18S rRNA gene of T. equi is the best combination for a duplex qRPA analysis in one reaction. The developed duplex qRPA assay has good specificity, and had negative amplification for several similar parasite. For DNA extracted from real horse blood specimens, this qRPA method has comparable sensitivity to traditional qPCR, but a simpler and more rapid operating process to obtain positive amplification. The qRPA, including the duplex strategy described here, could allow fast identification of the EP-causing T. equi and B. caballi, showing great potential for on-site EP screening of horses. Tachaea chinensis Thielemann, 1910 (Isopoda: Corallanidae) is a branchial ectoparasite that attaches onto shrimps and prawns. However, the distribution of T. chinensis in China, especially its epidemiology, remains unclear. We determined the prevalence of T. chinensis on the ridgetail white prawn (Exopalaemon carinicauda Holthuis, 1950) in Jiangsu Province. Fifty ponds in 10 shrimp farms were assessed. Isopod species were identified by morphological features and mitochondrial 16S rRNA gene analysis. A literature review was performed to determine the geographical distribution of T. chinensis in China. Published data revealed that T. chinensis was geographically distributed throughout five provinces in China, including Liaoning, Tianjin, Henan, Hubei, and Guangxi. A total of 998 T. chinensis were collected from 50 ridgetail white prawn ponds in Yancheng City and Rudong County. Tachaea chinensis prevalence ranged from 0.98% to 4.42% in Yancheng City and 0.62% to 0.92% in Rudong County. This is the first study to investigate the geographical distribution of T. chinensis in China and determine the prevalence of T. chinensis on the ridgetail white prawn in Jiangsu Province. In this work, we have developed a novel deep learning inverse solution or identification method to determine and identify the impact load conditions of shell structures based on their final state of damage or inelastic deformation. This artificial intelligence approach offers a practical solution to solve the inverse problem of engineering failure analysis based on final material and structure damage state and permanent plastic deformation. More precisely, the machine learning inverse problem solver may provide a practical solution to characterize failure load parameters and conditions based on the final permanent plastic deformation distribution of the shell structure that is under examination. In this work, we have demonstrated that the proposed deep learning method can accurately identify a "practically unique" static loading condition as well as the impact dynamic loading condition for a hemispherical shell structure based the permanent plastic deformation after the impact event as the forensic signatures. The data-driven based method developed in this work may provide a powerful tool for forensically diagnosing, determining, and identifying damage loading conditions for engineering structures in various accidental failure events, such as car crashes, pressure vessel failure, or thin-walled infrastructure structure collapses. For the central pattern generation inspired biped walking control algorithm, it is hard to coordinate all the degrees of freedom of a robot by regulating the parameters of a neutral network to achieve stable and adaptive walking. In this work, a hybrid rhythmic–reflex control method is presented, which can realize stable and adaptive biped walking. By integrating zero moment position information, the walking stability can be improved on flat terrain. The robot's body attitude information is used to modulate the control system in real-time to realize sloped terrain adaptive walking. A staged parameter evolution process is used to derive the parameters. Through the entrainment of the oscillatory network and the feedback information, the real-time joint control signals can be regulated to realize adaptive walking. The presented control strategy has been verified by using a biped robot restricted in sagittal plane and the experiments reveal that the robot can successfully achieve changing sloped terrain adaptive walking. Trypanosoma evansi is the causative agent of the animal trypanosomiasis surra, a disease with serious economic burden worldwide. The availability of the genome of its closely related parasite Trypanosoma brucei allows us to compare their genetic and evolutionarily shared and distinct biological features. The complete genomic sequence of the T. evansi YNB strain was obtained using a combination of genomic and transcriptomic sequencing, de novo assembly, and bioinformatic analysis. The genome size of the T. evansi YNB strain was 35.2 Mb, showing 96.59% similarity in sequence and 88.97% in scaffold alignment with T. brucei. A total of 8,617 protein-coding genes, accounting for 31% of the genome, were predicted. Approximately 1,641 alternative splicing events of 820 genes were identified, with a majority mediated by intron retention, which represented a major difference in post-transcriptional regulation between T. evansi and T. brucei. Disparities in gene copy number of the variant surface glycoprotein, expression site-associated genes, microRNAs, and RNA-binding protein were clearly observed between the two parasites. Since even a small disturbance may lead to the passive dynamic walker falling, former researches tried to enlarge the basin of attraction of the walking motion to resist disturbances. However, experience with real walking robots reveals that a new disturbance in one step will be added to the total disturbances accumulated from last steps, which results in falling due to a small accidental disturbance. Therefore, fast speed of convergence to the periodic walking motion is necessary to inhibit the accumulation of disturbances. In this paper, the internal features in the basin of attraction of the simplest walking model are investigated for the consideration that the performance for handling disturbances of a walking motion is determined by not only the size of basin of attraction, but also the internal features, such as the speed of convergence. The features of the basin of attraction are obtained by the cell mapping method with defining a convergent cell. The results show that it needs fewer steps for some stripe regions far away to converge than points close to the long period solution. Especially in some stripe regions, the convergence speed is very fast. There is no obvious relation between the size of the basin of attraction and the convergence speed, which motivates us to continue the research to find solutions for designing the walking model with high performance on both aspects. The digestive systems of mammals harbor a complex gut microbiome, comprising bacteria and other microorganisms that confer metabolic and immunological benefits to the host. Ruminants that digest plant-based foods have a four-compartment stomach consisting of the rumen, reticulum, omasum, and abomasum. The microorganisms in the stomach are essential for providing the host with critical nutrients. However, the majority of these microorganisms are unknown species. The microbiome of the stomach is diverse, and the majority of these organisms cannot be cultured. Next-generation sequencing (NGS) combined with bioinformatic analysis tools have allowed the dissection of the composition of the microbiome in samples collected from a specific environment. In this study, for the first time, the bacterial composition in two compartments, the reticulum and the omasum, of bovine were analyzed using a metagenomic approach and compared to the bacterial composition of the rumen. These data will assist in understanding the biology of ruminants and benefit the agricultural industry. The diversity and composition of the bacterial community in samples collected from the rumen, reticulum, and omasum of bovines in the Changchun Region of Northeast China were analyzed by sequencing the V3 region of the 16S rRNA gene using a barcoded Illumina paired-end sequencing technique, and the primary composition of the microbiome in the rumen, reticulum, and omasum of the bovines was determined. These microbiomes contained 17 phyla and 107 genera in all three samples. Five phyla, Bacteroidetes, Firmicutes, Proteobacteria, Spirochaetes, and Lentisphaerae, were the most abundant taxonomic groups. Bone tissue adapts to its functional environment by optimizing its morphology for mechanical demand. Among the mechanosensitive cells that recognize and respond to forces in the skeleton are osteocytes, osteoblasts, and mesenchymal progenitor cells (MPCs). Therefore, the ability to use mechanical signals to improve bone health through exercise and devices that deliver mechanical signals is an attractive approach to age-related bone loss; however, the extracellular or circulating mediators of such signals are largely unknown. Using SDS-PAGE separation of proteins secreted by MPCs in response to low-magnitude mechanical signals and in-gel trypsin digestion followed by HPLC and mass spectroscopy, we identified secreted proteins up-regulated by vibratory stimulation. We exploited a cell senescence-associated secretory phenotype screen and reasoned that a subset of vibration-induced proteins with diminished secretion by senescent MPCs will have the capacity to promote bone formation in vivo. We identified one such vibration-induced bone-enhancing ( vibe) gene as R-spondin 1, a Wnt pathway modulator, and demonstrated that it has the capacity to promote bone formation in three mouse models of age-related bone loss. By virtue of their secretory status, some vibe proteins may be candidates for pre-clinical development as anabolic agents for the treatment of osteoporosis. This paper presents a central pattern generator (CPG) and vestibular reflex combined control strategy for a quadruped robot. An oscillator network and a knee-to-hip mapping function are presented to realize the rhythmic motion for the quadruped robot. A two-phase parameter tuning method is designed to adjust the parameters of oscillator network. First, based on the numerical simulation, the influences of the parameters on the output signals are analyzed, then the genetic algorithm (GA) is used to evolve the phase relationships of the oscillators to realize the basic animal-like walking pattern. Moreover, the animal's vestibular reflex mechanism is mimicked to realize the adaptive walking of the quadruped robot on a slope terrain. Coupled with the sensory feedback information, the robot can walk up and down the slope smoothly. The presented bio-inspired control method is validated through simulations and experiments with AIBO. Under the control of the presented CPG and vestibular reflex combined control method, AIBO can cope with slipping, falling down and walk on a slope successfully, which demonstrates the effectiveness of the proposed walking control method. A least squares support vector fuzzy regression model (LS-SVFR) is proposed to estimate uncertain and imprecise data by applying the fuzzy set principle to weight vectors. This model only requires a set of linear equations to obtain the weight vector and the bias term, which is different from the solution of a complicated quadratic programming problem in existing support vector fuzzy regression models. Besides, the proposed LS-SVFR is a model-free method in which the underlying model function doesn't need to be predefined. Aquaporins are implicated in a wide variety of plant physiological processes, although the mechanisms involved in their regulation are not fully understood. To gain further insight into the regulatory factors involved in this process, we used a yeast two-hybrid system to screen for potential binding partners to the Arabidopsis tonoplast intrinsic protein (TIP) AtTIP1;1. This was the first protein identified to be associated with high water permeability in vacuolar membranes from Arabidopsis thaliana. Using AtTIP1;1 as bait, a novel binding protein was identified in both yeast and plant cells. This prey protein, named AtSM34, was a 309 aa polypeptide with a predicted molecular mass of 34 kD and contained a single MYB/SANT-like domain. AtSM34 promoter:: GUS histochemical staining analysis detected AtSM34 expression in flowers, stems and leaves, particularly in the vascular tissues, in response to osmotic stress. AtSM34 expression was localized in the endoplasmic reticulum membrane, and sequence deletion analysis revealed that the N-terminal coding region (amino acids 1-83) was critical for this localization. Overexpression of AtSM34 resulted in hypersensitivity to exogenous mannitol, sorbitol and abscisic acid, and caused a significant delay in germination. AtSM34 interacted with AtTIP1;2 and AtTIP2;1, which are essential proteins for modulation of tonoplast permeability and highly expressed in germinating seedlings. This paper is concerned with the problems of H-two filtering for discrete-time Markovian jump linear systems subject to logarithmic quantization. We assume that only the output of the system is available, and therefore the mode information is nonaccessible. In this paper, a mode-independent quantized H-two filter is designed such that filter error system is stochastically stable. To this end, sufficient conditions for the existence of an upper bound of H-two norm are presented in terms of linear matrix inequalities. Considering uncertainty of system matrices, a robust H-two filter is designed. The proposed method is also applicable to cover the case where the transition probability matrix is not exactly known but belongs to a given polytope. Compared with wheeled mobile robots, legged robots can easily step over obstacles and walk through rugged ground. They have more flexible bodies and therefore, can deal with complex environment. Nevertheless, some other issues make the locomotion control of legged robots a much complicated task, such as the redundant degree of freedoms and balance keeping. From literatures, locomotion control has been solved mainly based on programming mechanism. To use this method, walking trajectories for each leg and the gaits have to be designed, and the adaptability to an unknown environment cannot be guaranteed. From another aspect, studying and simulating animals’ walking mechanism for engineering application is an efficient way to break the bottleneck of locomotion control for legged robots. This has attracted more and more attentions. Inspired by central pattern generator (CPG), a control method has been proved to be a successful attempt within this scope. In this paper, we will review the biological mechanism, the existence evidences, and the network properties of CPG. From the engineering perspective, we will introduce the engineering simulation of CPG, the property analysis, and the research progress of CPG inspired control method in locomotion control of legged robots. The expression of the gene OsITL1 coding for the inositol 1,3,4-trisphosphate 5/6-kinase protein was induced by 200 mM NaCl or dehydration. The expression of OsITL1 in response to NaCl and dehydration suggests the possible functions of OsITL1 in osmotic stress responses; however, physiological tests indicate that the expression of OsITL1 in tobacco decreases tolerance to NaCl during germination and seedling development. This result was consistent with that obtained on treatment of mature tobacco seedlings with NaCl (200 mM), suggesting that OsITL1 inversely regulates plant responses to osmotic stress. There is increasing evidence for considerable interlinking between the responses to heat stress and oxidative stress, and recent researches suggest heat shock transcription factors (Hsfs) play an important role in linking heat shock with oxidative stress signals. In this paper, we present evidence that AtHsfA2 modulated expression of stress responsive genes and enhanced tolerance to heat and oxidative stress in Arabidopsis. Using Northern blot and quantitative RT-PCR analysis, we demonstrated that the expression of AtHsfA2 was induced by not only HS but also oxidative stress. By functional analysis of AtHsfA2 knockout mutants and AtHsfA2 overexpressing transgenic plants, we also demonstrated that the mutants displayed reduced the basal and acquired thermotolerance as well as oxidative stress tolerance but the overexpression lines displayed increased tolerance to these stress. The phenotypes correlated with the expression of some Hsps and APX1, ion leakage, H2O2 level and degree of oxidative injuries. These results showed that, by modulated expression of stress responsive genes, AtHsfA2 enhanced tolerance to heat and oxidative stress in Arabidopsis. Critical telomere shortening (for example, secondary to partial telomerase deficiency in the rare disease dyskeratosis congenita) causes tissue pathology, but underlying mechanisms are not fully understood. Mice lacking telomerase (for example, mTR−/− telomerase RNA template mutants) provide a model for investigating pathogenesis. In such mice, after several generations of telomerase deficiency telomeres shorten to the point of uncapping, causing defects most pronounced in high-turnover tissues including intestinal epithelium. Here we show that late-generation mTR−/− mutants experience marked downregulation of Wnt pathway genes in intestinal crypt epithelia, including crypt base columnar stem cells and Paneth cells, and in underlying stroma. The importance of these changes was revealed by rescue of crypt apoptosis and Wnt pathway gene expression upon treatment with Wnt pathway agonists. Rescue was associated with reduced telomere-dysfunction-induced foci and anaphase bridges, indicating improved telomere capping. Thus a mutually reinforcing feedback loop exists between telomere capping and Wnt signalling, and telomere capping can be impacted by extracellular cues in a fashion independent of telomerase. Schistosomiasis is a major human parasitic disease with a global impact. Schistosoma japonicum, the most difficult to control, can survive within host veins for decades. Mechanisms of immune evasion by the parasite, including antigenic variation and surface masking, have been implicated but not well defined. In this study, we defined the immunoglobulin-binding proteomes of S. japonicum using human IgG, IgM, and IgE as the molecular bait for affinity purification, followed by protein identification by liquid chromatography with tandem mass spectrometry (LC-MS/MS). Several proteins situated at the tegument of S. japonicum were able to nonselectively bind to the Fc domain of host immunoglobulins, indicating a mechanism for the avoidance of host immune attachment and recognition. Linear B-cell epitopes are ideal biomarkers for the serodiagnosis of infectious diseases. However, the long-predicted diagnostic value of epitopes has not been realized. Here, we demonstrated a method, diagnostic epitopes in four steps (DEIFS), that delivers a combination of epitopes for the serodiagnosis of infectious diseases with a high success rate. Using DEIFS for malaria, we identified 6 epitopes from 8 peptides and combined them into 3 chimeric peptide constructs. Along with 4 other peptides, we developed a rapid diagnostic test (RDT), which is able to differentiate Plasmodium falciparum (P. falciparum) from Plasmodium vivax (P. vivax) infections with 95.6% overall sensitivity and 99.1% overall specificity. In addition to applications in diagnosis, DEIFS could also be used in the diagnosis of virus and bacterium infections, discovery of vaccine candidates, evaluation of vaccine potency, and study of disease progression.
