Fatty acid (FA) content and composition of zooplankton in Puget Sound, Washington (USA) was studied to investigate the nutritional quality of diverse zooplankton prey for juvenile salmon (Oncorhynchus spp.) in terms of their essential fatty acid (EFA) content. The study focus was on eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and arachidonic acid (ARA) as these are key FA needed to maintain growth and development of juvenile fish. The different zooplankton taxa varied in their FA composition. Much of the variation in FA composition was driven by 18:1ω9 (a biomarker of carnivory), ARA, DHA, and FA characteristic of diatoms, which are linked to zooplankton diet sources. Gammarid and hyperiid amphipods contained the highest amount of EFA, particularly the gammarid amphipod Cyphocaris challengeri, while shrimp and copepods had much lower EFA content. Crab larvae, which are important prey for juvenile salmon in Puget Sound, had intermediate EPA + DHA content and the lowest DHA/EPA ratio, and were rich in diatom biomarkers. Temporal and spatial trends in zooplankton lipids were less apparent than the taxonomic differences, although the EFA content increased from spring to summer in Cancridae zoeae and the amphipod C. challengeri. These results on taxon-specific EFA content provide baseline information on the nutritional quality of zooplankton that can be applied in food web models. Combining zooplankton fatty acid data (quality) with taxon-specific zooplankton biomass data (quantity) enables development of new, sensitive indicators of juvenile fish production to help assess recent declines in salmon production in the Pacific Northwest and predict future adult returns. The River Continuum Concept implies that consumers in headwater streams have greater dietary access to terrestrial basal resources, but recent studies have highlighted the dietary importance of high-quality algae. Algae provide consumers with physiologically important omega-3 (n-3) polyunsaturated fatty acids (PUFA), particularly eicosapentaenoic acid (EPA). However, terrestrial plants and most benthic stream algae lack the long-chain (LC) n-3 PUFA docosahexaenoic acid (DHA, 22:6n-3), which is essential for neural development in fish and other vertebrates. We sampled subalpine streams to investigate how the PUFA composition of neural (brain and eyes), muscle, and liver tissues of freshwater fish is related to their potential diets (macroinvertebrates, epilithon, fresh and conditioned terrestrial leaves). The PUFA composition of consumers was more similar to epilithon than to terrestrial leaves. Storage lipids of eyes most closely resembled dietary PUFA (aquatic invertebrates and algae). However, DHA and arachidonic acid (ARA, 20:4n-6) were not directly available in the diet but abundant in organs. This implies that algal PUFA were selectively retained or were produced internally via enzymatic PUFA conversion by aquatic consumers. Mixing models have become requisite tools for analyzing biotracer data, most commonly stable isotope ratios, to infer dietary contributions of multiple sources to a consumer. However, Bayesian mixing models will always return a result that defaults to their priors if the data poorly resolve the source contributions, and thus, their interpretation requires caution. We describe an application of information theory to quantify how much has been learned about a consumer’s diet from new biotracer data. We apply the approach to two example data sets. We find that variation in the isotope ratios of sources limits the precision of estimates for the consumer’s diet, even with a large number of consumer samples. Thus, the approach which we describe is a type of power analysis that uses a priori simulations to find an optimal sample size. Biotracer data are fundamentally limited in their ability to discriminate consumer diets. We suggest that other types of data, such as gut content analysis, must be used as prior information in model fitting, to improve model learning about the consumer’s diet. Ultrathin-layer chromatography (UTLC) potentially offers faster analysis, reduced solvent and sample volumes, and lower costs. One novel technique for producing UTLC plates has been glancing angle deposition (GLAD), a physical vapor deposition technique capable of aligning macropores to produce interesting separation properties. To date, however, GLAD-UTLC plates have been restricted to model dye systems, rather than realistic analytes. This study demonstrates the transfer of high-performance thin-layer chromatography (HPTLC) sugar analysis methods to GLAD-UTLC plates using the office chromatography framework. A consumer inkjet printer was used to apply very sharp low volume (3-30 nL) bands of water-soluble analytes (lactose, sucrose, and fructose). Analytic performance measurements extrapolated the limits of detection to be 3-5 ng/zone, which was experimentally proven down to 60-70 ng/band, depending on the sugar. This qualitative analysis of sugars in a commercially available chocolate sample is the first reported application of GLAD-UTLC to food samples. The potential utility of GLAD-UTLC is further exemplified by successful coupling with electrospray ionization mass spectrometry for the first time to characterize underivatized sugars. Hood Canal, Washington, USA, is a poorly ventilated fjord-like sub-basin of Puget Sound that commonly experiences hypoxia. This study examined the influence of watershed soils, vegetation, physical features, and population density on nitrogen (N) export to Hood Canal from 43 tributaries. We also linked our watershed study to the estuary using a salinity mass balance model that calculated the relative magnitude of N loading to Hood Canal from watershed, direct precipitation, and marine sources. The overall flow-weighted total dissolved N (TDN) and particulate N input concentrations to Hood Canal were 152 and 49 μg l, respectively. Nitrate and dissolved organic N comprised 64 and 29% of TDN, respectively. The optimal regression models for TDN concentration and areal yield included a land cover term suggesting an effect of N-fixing red alder ( Alnus rubra) and a human population density term (suggesting onsite septic system (OSS) discharges). There was pronounced seasonality in stream water TDN concentrations, particularly for catchments with a high prevalence of red alder, with the lowest concentrations occurring in the summer and the highest occurring in November-December. Due to strong seasonality in TDN concentrations and in particular stream flow, over 60% of the TDN export from this watershed occurred during the 3 month period of November-January. Entrainment of marine water into the surface layer of Hood Canal accounted for ≈98% of N loading to the euphotic zone of this estuary, and in a worst case scenario OSS N inputs contribute ≈0.5% of total N loading. Domestic wastewater discharges and red alders appear to be a very important N source for many streams, but a minor nutrient source for the estuary as a whole. [ABSTRACT FROM AUTHOR]

We developed a mechanistic model of nutrient, phytoplankton, zooplankton and fish interactions to test the effects of phytoplankton food quality for herbivorous zooplankton on planktonic food web processes. When phytoplankton food quality is high strong trophic cascades suppress phytoplankton biomass, the zooplankton can withstand intense zooplanktivory, and energy is efficiently transferred through the food web sustaining higher trophic level production. Low food quality results in trophic decoupling at the plant-animal interface, with phytoplankton biomass determined primarily by nutrient availability, zooplankton easily eliminated by fish predation, and poor energy transfer through the food web. At a given nutrient availability, food quality and zooplanktivory interact to determine zooplankton biomass which in turn determines algal biomass. High food quality resulted in intense zooplankton grazing which favored fast-growing phytoplankton taxa, whereas fish predation favored slow-growing phytoplankton. These results suggest algal food quality for herbivorous zooplankton can strongly influence the nature of aquatic food web dynamics, and can have profound effects on water quality and fisheries production. Glancing angle deposition (GLAD) is a physical vapour deposition method used to fabricate highly functional thin films with an engineerable columnar morphology. Recent developments in GLAD technology have produced columnar nanostructures of increased complexity, including periodic, nanofibrous, perforated, and graded porosity thin films for use in applications ranging from sensors and actuators to optical filters, microfluidics, and catalysis. We conducted statistical analyses of a 10-year record of stream nutrient and sediment concentrations for 17 streams in the greater Seattle region to determine the impact of urban non-point-source pollutants on stream water quality. These catchments are dominated by either urban (22-87%) or forest (6-73%) land cover, with no major nutrient point sources. Stream water phosphorus concentrations were moderately strongly (r2?=?0.58) correlated with catchment land-cover type, whereas nitrogen concentrations were weakly (r2?=?0.19) and nonsignificantly (at a<0.05) correlated with land cover. The most urban streams had, on average, 95% higher total phosphorus (TP) and 122% higher soluble reactive phosphorus (SRP) and 71% higher turbidity than the most forested streams. Nitrate (NO3), ammonium (NH4), and total suspended solids (TSS) concentrations did not vary significantly with land cover. These results suggest that urbanization markedly increased stream phosphorus concentrations and modestly increased nitrogen concentrations. During a 1-year period, we sampled stream water total phosphorus (TP) concentrations daily and soluble reactive phosphorus (SRP) concentrations weekly in four Seattle area streams spanning a gradient of forested to urban-dominated land cover. The objective of this study was to develop time series models describing stream water phosphorus concentration dependence on seasonal variation in stream base flows, short-term flow fluctuations, antecedent flow conditions, and rainfall. Stream water SRP concentrations varied on average by ±18% or ±5.7 μg/L from one week to another, whereas TP varied ±48% or ±32.5 μg/L from one week to another. On average, SRP constituted about 47% of TP. Stream water SRP concentrations followed a simple sine-wave annual cycle with high concentrations during the low-flow summer period and low concentrations during the high-flow winter period in three of the four study sites. These trends are probably due to seasonal variation in the relative contributions of groundwater and subsurface flows to stream flow. In forested Issaquah Creek, SRP concentrations were relatively constant throughout the year except during the fall, when a major salmon spawning run occurred in the stream and SRP concentrations increased markedly. Stream water SRP concentrations were statistically unrelated to short-term flow fluctuations, antecedent flow conditions, or rainfall in each of the study streams. Stream water TP concentrations are highly variable and strongly influenced by short-term flow fluctuations. Each of the processes assessed had statistically significant correlations with TP concentrations, with seasonal base flow being the strongest, followed by antecedent flow conditions, short-term flow fluctuations, and rainfall. Times series models for each individual stream were able to predict ∼70% of the variability in the SRP annual cycle in three of the four streams ( r2 = 0.57–0.81), whereas individual TP models explained ∼50% of the annual cycle in all streams ( r2 = 0.39–0.59). Overall, time series models for SRP and TP dynamics explained 82% and 76% of the variability for these variables, respectively. Our results indicate that SRP, the most biologically available and therefore most important phosphorus fraction, has simpler and easier-to-predict seasonal and weekly dynamics. Determining the factors that control food web interactions is a key issue in ecology. The empirical relationship between nutrient loading (total phosphorus) and phytoplankton standing stock (chlorophyll a) in lakes was described about 30 years ago and is central for managing surface water quality. The efficiency with which biomass and energy are transferred through the food web and sustain the production of higher trophic levels (such as fish) declines with nutrient loading and system productivity, but the underlying mechanisms are poorly understood. Here we show that in seston (fine particles in water) during summer, specific ?3-polyunsaturated fatty acids (?3-PUFAs), which are important for zooplankton, are significantly correlated to the trophic status of the lake. The ?3-PUFAs octadecatetraenoic acid, eicosapentaenoic acid (EPA) and docosahexaenoic acid, but not a-linolenic acid, decrease on a double-logarithmic scale with increasing total phosphorus. By combining the empirical relationship between EPA-to-carbon content and total phosphorus with functional models relating EPA-to-carbon content to the growth and egg production of daphnids, we predict secondary production for this key consumer. Thus, the decreasing efficiency in energy transfer with increasing lake productivity can be explained by differences in ?3-PUFA-associated food quality at the plant-animal interface. Three-dimensional (3D) simulation of combined lattice and grain-boundary diffusion of impurities in thin-film diffusion barriers for eemiconductor device metallizations is performed. Calculated results of impurity concentration profiles demonstrate quantitatively an obvious underestimation of the frequently used two-dimensional (2D) analysis with respect to the influence of film geometry and grain-boundary diffusion coefficient. As for the average concentration at the backside of diffusion barriers, approximately a factor of two difference between the 2D and 3D simulation results is found over an interesting range of times and grain size structures. Graphs for predicting the effectiveness of diffusion barriers are presented with several normalized parameters associated with position and time. Particular application examples of aluminum diffusion in titanium nitride films justify the use of this material as an effective diffusion barrier in silicon microelectronic devices. This study examined the long term effects of predation by larvae of the midge Chaoborus and simulated fish predation on experimental Daphnia longispina populations. Chaoborus predation, relative to fish predation, led to populations composed of larger individuals as a whole, larger egg-bearing individuals, and a larger primiparous instar. Daphnia retained helmets beyond the first instar in response to the presence of Chaoborus. Both types of predation, relative to predator-free controls, reduced prey population size and rates of increase, but increased population death rates. The reduction in population size due to predation led to increased resource availability for individuals remaining in the populations and increased individual fecundity in the predation treatments.
