Previous studies examining the covariation among Attention Deficit Hyperactivity Disorder (ADHD), Oppositional Defiant Disorder (ODD) and Conduct Disorder (CD) have yielded inconsistent results. Some studies have concluded that the covariation among these symptoms is due to common genetic influences, whereas others have found a common environmental overlap. The present study investigated the genetic and environmental correlations among these three childhood disorders, based on a sample of 1,219 twins, age 9-10 years. A latent externalizing behavior factor was found to explain the covariance among ADHD, ODD and CD symptoms. Genetic influences explained more than half of the variance in this externalizing factor in both boys and girls. There were also unique genetic and environmental influences in each set of symptoms, suggesting some etiological independence of the three disorders. Inputs of fertilizer nutrients in agriculture are estimated to have contributed to > 40% increase in crop production over the past century, resulting in widespread benefits to food security and prosperity. However, fertilizer nutrient redistribution has fundamentally altered global and local nutrient cycles alike, yielding trade-offs in socioeconomic and environmental outcomes. David Pimentel's body of work on the management of energy, water, and soil resources in agriculture, along with his perspectives on agronomy and sustainable resource management, resonates with a critical understanding of the consequences of nutrient redistribution in agriculture. With Pimentel's legacy in mind, we consider trade-offs of global nutrient redistribution, improved recycling of nutrients in agricultural systems, as well as the challenges of, and opportunities for, transformations that seek to adjust nutrient cycles in modern agriculture. Pimentel's legacy and contributions provide valuable insight into agriculture's wicked nutrient challenge, as he framed the costs and opportunities of production systems across different scales of food production, developed foundational understanding of global resource challenges, promoted often marginalized or underemployed management strategies to enhance agriculture's ecosystem services, confronted conventional wisdom and popular trends, and appropriately, attacked the use of "silver bullets" as singular solutions to ecological challenges and instead promoted systems-level analyses. To reduce nutrient losses from the food system, it is necessary to improve biomass management and foster change. Such a change is often hindered by a lack of stakeholder interaction. Therefore, a qualitative case study and a practical application of the innovation platform approach in the Dutch-German border region Rhine-Waal were carried out to determine challenges and opportunities in the agro-food-waste system towards circular nutrient management in a nutrient-saturated and intensive animal production-dominated localized area. Twenty-one actors participated in a half-day workshop. A bottom-up approach was chosen as it increases trust between stakeholders and supports the acceptance of research processes. This study identified opportunities and challenges perceived by stakeholders participating in the innovation platform approach to facilitate a transition towards local circular nutrient management. We observed that challenges and opportunities exist at three levels: the individual actor's level, the system level and the interconnection of the system with its wider environment. With a variety of stakeholders from animal and crop production to the food processing industry being present in the study area, the current demand and supply of biomass is very diverse. This diversity has been identified as a distinct opportunity for the establishment of a biomass exchange network in the area. However, information on demand and supply of nutrients between actors is currently scattered and information sharing hindered by the lack of direct monetary benefits. Excessive exposure to sunlight can contribute for skin photo-damage, such as sunburn, dryness, wrinkles, hyperpigmentation, immunosuppressive events and skin sensitization reactions. The use of aftersun products is an effective strategy to reduce the visible signs and symptoms of acute photodamage in the skin. Aiming to unveil the active ingredients able to offset acute sun damage, this work focuses on the characterization of the aftersun products market. A total of 84 after-sun formulations from 41 international brands currently marketed in Portugal were analyzed concerning the composition described on the product label, identifying natural and synthetic/semi-synthetic ingredients with the ability to mitigate solar-induced effects. The majority of aftersun formulations contained ingredients derived from terrestrial and marine sources (> 80%). An in-depth examination of these compounds is also offered, revealing the top of the most used natural and synthetic/semi-synthetic ingredients present in aftersun products, as well as their mechanism of action. A critical appraisal of the scientific data was made aiming to highlight the scientific evidence of ingredients able to mitigate skin photodamage. Amino acids and peptides, and A. barbadensis extract were tested for their in vivo efficacy. Nevertheless, all the ingredients were analyzed with in vitro studies as preliminary screening before in vivo, ex vivo and/or clinical studies. In summary, this study provides an overview of the use of active ingredients in commercial aftersun products to understand better the benefits associated with their use in cosmetic formulations and identify opportunities for innovation. Over the past two decades, family structures have diversified. International migration has led to a rise in the number of families in which at least one parent is foreign-born. Increases have also been observed in both the rate of partnership separation, leading to a greater number of single-parent households and an increase in the number of families where grandparents have assumed caring responsibilities for their grandchildren. Evidence indicates a strong relationship between family structure and children's educational outcomes. Parental involvement is well documented as a key ingredient for the educational success of children. Drawing on Taiwanese multi-wave survey data (Taiwan Assessment of Student Achievement) and a machine-learning-based propensity score algorithm for multiple treatments, this paper aims to determine the various relationships between children from different household structures (two-parent households, skipped generation households, single-parent households, and immigrant households) and their cognitive knowledge (measured by test scores). Key findings reveal that children from skipped generation households achieve the lowest performance scores and that those from immigrant households tend to perform even better than children from traditional two-parent households in certain disciplines. Our results suggest that policy interventions targeted at providing remedial education and/or financial assistance are needed to support children from skipped generation families to redress existing educational disadvantages in Taiwan. Urogenital reconstructive malformation surgery is sometimes hampered by lack of tissue for the repair. We have previously shown that autologous micrografting allows for single-staged scaffold cellularization after surgical implantation. Here, a collagen-based scaffold reinforced with biodegradable mesh and a stent was implanted as a bladder conduit in ten full-grown female minipigs. We aimed to assess short-term regenerative outcomes, safety, and feasibility of implanting tubular urinary micrografted scaffolds versus acellular controls. Five scaffolds were embedded with autologous urothelial micrografts harvested perioperatively. After six weeks, all animals were assessed by cystoscopy, CT-urography, and microanatomical assessment of the urinary conduits. The procedure proved technically feasible within the confines of a regular surgical theater, with duration-times comparable to corresponding conventional procedures. No animals experienced postoperative complications, and all implanted conduits were patent at follow-up. Improved tissue regeneration was observed in the micrografted conduits compared with the acellular controls, including increased luminal epithelialization, increased cell proliferation, decreased cell apoptosis, and increased conduit vascularization. We concluded that single-staged on-site construction and implantation of tissue engineered urinary conduits proved feasible and safe, with improved regenerative potentials in micrografted conduits. Since the birth of cognitive science, researchers have used reaction time and accuracy to measure cognitive ability. Although recognition of these two measures is often based on empirical observations, the underlying consensus is that most cognitive behaviors may be along two fundamental dimensions: cognitive processing speed (CPS) and cognitive processing accuracy (CPA). In this study, we used genomic-wide association studies (GWAS) data from 14 cognitive traits to show the presence of those two factors and revealed the specific neurobiological basis underlying them. We identified that CPS and CPA had distinct brain phenotypes (e.g. white matter microstructure), neurobiological bases (e.g. postsynaptic membrane), and developmental periods (i.e. late infancy). Moreover, those two factors showed differential associations with other health-related traits such as screen exposure and sleep status, and a significant causal relationship with psychiatric disorders such as major depressive disorder and schizophrenia. Utilizing an independent cohort from the Adolescent Brain Cognitive Development (ABCD) study, we also uncovered the distinct contributions of those two factors on the cognitive development of young adolescents. These findings reveal two fundamental factors underlying various cognitive abilities, elucidate the distinct brain structural fingerprint and genetic architecture of CPS and CPA, and hint at the complex interrelationship between cognitive ability, lifestyle, and mental health. Hirschsprung disease (HSCR) and its associated disorders (AD-HSCR) often result in severe hypoperistalsis caused by enteric neuropathy, mesenchymopathy, and myopathy. Notably, HSCR involving the small intestine, isolated hypoganglionosis, chronic idiopathic intestinal pseudo-obstruction, and megacystis-microcolon-intestinal hypoperistalsis syndrome carry a poor prognosis. Ultimately, small-bowel transplantation (SBTx) is necessary for refractory cases, but it is highly invasive and outcomes are less than optimal, despite advances in surgical techniques and management. Thus, regenerative therapy has come to light as a potential form of treatment involving regeneration of the enteric nervous system, mesenchyme, and smooth muscle in affected areas. We review the cutting-edge regenerative therapeutic approaches for managing HSCR and AD-HSCR, including the use of enteric nervous system progenitor cells, embryonic stem cells, induced pluripotent stem cells, and mesenchymal stem cells as cell sources, the recipient intestine's microenvironment, and transplantation methods. This study examines whether and to what extent the availability of grandparents in the home plays a buffering role in the labor supply of parents of children aged 0–5 during the COVID-19 pandemic. The role of grandparents as a childcare resource and its association with parents' labor supply have received increasing attention in the literature. Limited childcare options during the pandemic underscore the need to investigate how working parents manage the double burden of family and work and what role grandparents could play. I use monthly data from the Current Population Survey (CPS) from January to May 2019 and 2020. Using a propensity score matching method, parents in three-generational households were matched to those in two-generational households. Then, I employ a difference-in-differences approach with propensity score-matched samples. Results show that parents living with grandparents are more likely to have worked last week and worked longer during the pandemic relative to parents in a two-generational household. The effects of the availability of coresident grandparents are more pronounced among single and low-educated parents than their counterparts. Furthermore, mothers and fathers show heterogeneous responses to the presence of grandparents during the pandemic. In conclusion, results highlight that grandparents played a buffering role in mitigating the adverse impact of the pandemic on parental labor supply. This study sheds light on the importance of grandparental care specifically and informal care and home-based care in general. Supply chain efficiency is a major challenge in today's business environment, where efficient resource allocation and coordination of activities are essential for competitive advantage. Traditional efficiency strategies often struggle for resources for the complex and dynamic network. In response, bio-inspired metaheuristic algorithms have emerged as powerful tools to solve these optimization problems. Referring to the random search nature of metaheuristic algorithms and emphasizing that no metaheuristic algorithm is the best optimizer for all optimization applications, the No Free Lunch (NFL) theorem encourages researchers to design newer algorithms to be able to provide more effective solutions to optimization problems. Motivated by the NFL theorem, the innovation and novelty of this paper is in designing a new meta-heuristic algorithm called Bobcat Optimization Algorithm (BOA) that imitates the natural behavior of bobcats in the wild. The basic inspiration of BOA is derived from the hunting strategy of bobcats during the attack towards the prey and the chase process between them. The theory of BOA is stated and then mathematically modeled in two phases (i) exploration based on the simulation of the bobcat's position change while moving towards the prey and (ii) exploitation based on simulating the bobcat's position change during the chase process to catch the prey. The performance of BOA is evaluated in optimization to handle the CEC 2017 test suite for problem dimensions equal to 10, 30, 50, and 100, as well as to address CEC 2020. The optimization results show that BOA has a high ability in exploration, exploitation, and balance them during the search process in order to achieve a suitable solution for optimization problems. The results obtained from BOA are compared with the performance of twelve well-known metaheuristic algorithms. The findings show that BOA has been successful in handling the CEC 2017 test suite in 89.65, 79.31, 93.10, and 89.65% of the functions for the problem dimension equal to 10, 30, 50, and 100, respectively. Also, the findings show that in order to handle the CEC 2020 test suite, BOA has been successful in 100% of the functions of this test suite. The statistical analysis confirms that BOA has a significant statistical superiority in the competition with the compared algorithms. Also, in order to analyze the efficiency of BOA in dealing with real world applications, twenty-two constrained optimization problems from CEC 2011 test suite and four engineering design problems have been selected. The findings show that BOA has been successful in 90.90% of CEC2011 test suite optimization problems and in 100% of engineering design problems. In addition, the efficiency of BOA to handle SCM applications has been challenged to solve ten case studies in the field of sustainable lot size optimization. While the simultaneous degradation of muscle composition and postural stability in aging are independently highly investigated due to their association with fall risk, the interplay between the two has received little attention. Thus, the purpose of this study is to explore how age-related changes in muscle composition relate to postural stability. To that aim, we collected posturography measures and ultrasound images of the dominant Vastus Lateralis and Biceps Brachii from 32 young (18–35 year old) and 34 older (65–85 year old) participants. Muscle properties were quantified with echo-intensity and texture-based metrics derived from gray-level co-occurrence matrix analysis, and postural stability with the variability of the center of pressure during bipedal stance tasks. Ultrasound parameters revealed that young muscle possessed lower echo-intensity and higher homogeneity compared to the elderly. Echo-intensity and muscle thickness, and several texture-based parameters possessed outstanding young versus older classification performance. A canonical correlation analysis demonstrated a significant relationship between ultrasound and postural measures only within the young group (r = 0.53, p < 0.002), where those with 'better' muscle composition displayed larger postural sways. Our results indicate that, in older individuals, postural stability and muscle composition, two common fall risk factors, are unrelated. In view of this decoupling, both may contribute independently to fall risk. Furthermore, our data support the view that texture-based parameters provide a robust alternative to echo-intensity in providing markers of muscle composition. [ABSTRACT FROM AUTHOR]

Testicular torsion is a common disorder in males and results in blockage of testicular circulation with subsequent damage of testicular germ cells. The current work aimed to compare the therapeutic effect of platelet-rich plasma (PRP) and injectable platelet-rich fibrin (i-PRF) on torsion/detorsion (T/D) injury in rats. Forty mature male Wister rats were arranged into 4 groups; (1) Control, (2) T/D, (3) T/D + PRP, and (4) T/D+ i-PRF. The right testis was twisting 1080° clockwise for 3 h in groups 2, 3 and 4, then 10 μl of PRP or i-PRF was injected intra-testicular 3 h after detorsion in groups 3 and 4, respectively. After 30 days postoperatively, the semen quality and hormonal assay were improved in PRP and i-PRF-treated groups with superiority of i-PRF (P < 0.001). High significance of Catalase, Glutathione Peroxidase (GPx), Superoxide Dismutase, Interleukin-1β (IL-1β), Caspase-3 and Tumor necrosis factor-α (TNF-α) was reported in treated rats with PRP and i-PRF (P < 0.001) with superiority to i-PRF-treated rats (P < 0.001). Testicular histoarchitectures were improved in PRP and i-PRF-treated rats with superiority of i-PRF-treated rats. Background: More research is needed to improve measurement selection and to better understand informant differences in reports of reactive and proactive aggression. Objective: Toward this goal, the current study evaluated the psychometrics (i.e., reliability, factor structure, and validity) and correlates of two measures of reactive and proactive aggression (i.e., Dodge & Coie, in J Pers Soc Psychol 53:1146, 1987; Raine et al. in Aggress Behav 32:15–171, 2006) across three informants (i.e., parent, teacher, and youth). Method: Parent, teacher and youth reports of measures were collected in a community recruited sample of 9–12 year-old youth (M = 10.44; 56% male). Results: Both measures demonstrated adequate to good internal consistency by parent- and teacher-report, and borderline to adequate internal consistency by youth-report. Additionally, aggression subscales were correlated within and across measures and informants, and an appropriate 2-factor structure was identified for both measures across informants. Consistent with prior research, reactive aggression was more robustly associated with depression symptoms and effortful control than proactive aggression across measures, but there were some links with proactive aggression. Conclusions: Overall findings suggest that both measures are psychometrically appropriate to use with parents, teachers, and youth. However, there were distinctions between the two measures, and relying solely on youth reports in this age group is not recommended. Spinal muscular atrophy (SMA) is a neuromuscular disease caused by deletions or mutations in the survival of motor neuron 1 (SMN1) gene resulting in reduced levels of SMN protein. SMN protein is produced by cells throughout the body, and evidence suggests that low SMN protein can have systemic implications, including in male reproductive organs. However, a paucity of research exists on this important topic. This article will discuss findings from non-clinical studies on the role of SMN in the male reproductive system; additionally, real-world observational reports of individuals with SMA will be examined. Furthermore, we will review the non-clinical reproductive findings of risdiplam, a small-molecule SMN2 splicing modifier approved for the treatment of SMA, which has widespread distribution in both the central nervous system and peripheral organs. Specifically, the available non-clinical evidence of the effect of risdiplam on male reproductive organs and spermatogenesis is examined. Lastly, the article will highlight available capabilities to assess male fertility as well as the advanced reproductive technologies utilized to treat male infertility. Ferulic acid and its derivative γ-oryzanol are common in Rice. They have the same molecular skeleton with some sunscreens that used commonly, but there were no reports on them as rice UV protection. In this work, UV absorption and fluorescence emission spectrum are performed to measure ferulic acid and γ-oryzanol effect of fighting with ultraviolet irradiation. We demonstrated that both ferulic acid and γ-oryzanol have good absorption intensity at UVA, UVB and UVC regions, with superior photostability. Furthermore, the optimized molecular conformation and potential energy surfaces (PES) in ground and excited states are obtained by using the time-dependent density functional theory (TD-DFT) method. From the calculation results, it can conclude that their S1 states decay to the S0 state via internal conversion (IC) process at the S1/S0 conical intersection (CI) point of PES followed by trans–cis photoisomerization. For the first time, we elucidated the mechanism of UV protection for ferulic acid and γ-oryzanol as UV protection in rice from our excited state non-adiabatic dynamics. [ABSTRACT FROM AUTHOR]

Purpose: To synthesize the effect of exercise training on functional capacity, muscle strength, exercise capacity, dialysis efficacy, and quality of life (QOL) in children and adolescents with CKD. Methods: PubMed/Medline, Scopus, PEDro, Web of Science, CINAHL, Cochrane, and Embase were searched from inception to September 30, 2023. Randomized control trials (RCTs) and clinical trials that assessed the effect of exercise training programs on functional capacity, muscle strength, exercise capacity, dialysis efficacy, and QOL in children and adolescents with CKD were included. Random effect model and meta-regression were used for the meta-analysis. Results: Four clinical trials and three RCTs were included. The results showed that exercise training improves strength, but meta-analysis did not show a significant effect of exercise on functional capacity (WMD: 1.02; 95% CI: − 0.14 to 2.18; p = 0.083) and QOL (WMD: 8.00; 95% CI: − 3.90 to 19.91; p = 0.187). Subgroup analysis revealed that more than 25 sessions and 45 min per session of intervention, a PEDro score of more than 5, and being younger than 12 years of age had a large effect on functional capacity and QOL results. Due to the limited number of studies that reported the effect of exercise on dialysis efficacy and exercise capacity, the findings were inconclusive. Conclusion: Exercise training could benefit children and adolescents with CKD by increasing their strength. Longer exercise interventions may be beneficial for improving functional capacity and QOL. Aim: To investigate the associations of the weekend warrior and other physical activity (PA) patterns with all-cause, cardiovascular disease (CVD) mortality risk in people with diabetes mellitus (DM) and chronic kidney disease (CKD). Methods: This study pooled the data from NHANES 2007–2020. Participants with DM and CKD were included. PA was assessed using a self-reported questionnaire. According to the characteristics of recreational activities, individuals were categorized as inactive (no activities), insufficiently active (total PA duration < 150 min/week), weekend warrior (total PA duration ≥ 150 min/week for 1–2 sessions), and regularly active (total PA duration ≥ 150 min/week for ≥ 3 sessions). Weighted Cox regression models with adjusting sociodemographic, behavioral, and metabolic factors were performed to investigate the relationship of PA patterns with all-cause and CVD mortality risk. Stratification and interaction analyses were further performed. Results: Among 1702 participants (46.53% female; 64 ± 0.46 years old), 536 died (163 cardiovascular) during the follow-up of 68 (39–104) months. The hazard ratio (HR) of all-cause death was 0.618 (95% CI 0.406–0.942) for insufficiently active PA pattern, 0.338 (95% CI 0.116–0.988) for weekend warrior PA pattern, and 0.536 (95% CI 0.395–0.726) for regularly active PA pattern compared with inactive PA pattern. HR of CVD death was 0.545 (95% CI 0.250–1.189) for the PA pattern of insufficiently active, 0.165 (95% CI 0.020–1.343) for weekend warrior, and 0.393 (95% CI 0.218–0.710) for regularly active compared with the inactive PA pattern. The associations present no difference in subgroups. Moreover, there was no discernible difference between weekend warrior and regularly active PA patterns for all-cause and CVD deaths. The risk of death declined relatively quickly When exercise was initiated and to a total of 450 min or 4 times per week. Conclusion: In a population of DM and CKD, the weekend warrior pattern was similar to regular activity to lower the risk of all-cause mortality, compared with inactivity. The weekend warrior pattern was recommended for people who only have time to exercise on the weekend. However, longer and larger sample cohort studies are needed to validate our findings. This study aimed to examine the relations between perceived teacher emotional support, Covid-19 anxiety, resilience, and mental well-being among Chinese vocational college students during Covid-19 pandemic. A sample of Chinese vocational college students (n = 1469) were surveyed with an online questionnaire composed of Teacher Emotional Support, Covid-19 Anxiety Scale, Brief Resilience Scale and Mental Well-being in Chinese version. Path analysis was employed in the study and the results showed that teacher emotional support was an important promoter for building up mental well-being but not a buffer for Covid-19 within Chinese cultural context, and the Covid-19 anxiety was significantly and negatively related to mental well-being. Resilience hindered the incidences of Covid-19 anxiety and was a significant protector for mental well-being. Covid-19 anxiety mediated the both relations between teacher emotional support and mental well-being, and resilience and mental well-being. These findings provided practical implications for coping with psychological problems and flourishing mental well-being among Chinese vocational college students. [ABSTRACT FROM AUTHOR]

Spatial compartmentalization is a key facet of protein quality control that serves to store disassembled or non-native proteins until triage to the refolding or degradation machinery can occur in a regulated manner. Yeast cells sequester nuclear proteins at intranuclear quality control bodies (INQ) in response to various stresses, although the regulation of this process remains poorly understood. Here we reveal the SUMO modification of the small heat shock protein Btn2 under DNA damage and place Btn2 SUMOylation in a pathway promoting protein clearance from INQ structures. Along with other chaperones, and degradation machinery, Btn2-SUMO promotes INQ clearance from cells recovering from genotoxic stress. These data link small heat shock protein post-translational modification to the regulation of protein sequestration in the yeast nucleus. Maintaining a healthy nuclear proteome during DNA damage is important but its regulation is poorly understood. Background: Children with prune belly syndrome (PBS) are at higher risk of developing kidney dysfunction and requiring kidney replacement therapy (KRT). While studies have described surgical and survival outcomes in these populations, there has yet to be a focused synthesis of evidence regarding kidney outcomes in this population. Here, the focus of this scoping review was to highlight knowledge gaps and report standards on kidney outcomes in PBS of all ages. Methods: Following scoping review methodology, EMBASE, MEDLINE, and Scopus were searched for peer-reviewed literature that describe kidney outcomes in PBS. All studies with a broad set of kidney outcomes (such as kidney function measures, chronic kidney disease (CKD), KRT and associated outcomes) were included. Findings were summarized and qualitatively synthesized. Results: Of the 436 unique records identified, 25 were included for synthesis. A total of 17 studies (441 patients) reported on kidney insufficiency outcomes, with an estimated prevalence of CKD ranging from 8 to 66%. A total of 15 studies (314 patients) described KRT, primary kidney transplant, and outcomes. Of these, the age for KRT ranged from 4 to 21 years, and graft survival ranged from 22 to 87% by last follow-up (range 1.3–27 years). Conclusions: There is significant variability in studies reporting kidney outcomes in PBS which limits meaningful synthesis. The central question of nephron-sparing surgery in unilateral non-syndromic Wilms tumour sits at a crossroads between surgery, oncology, and nephrology. There has been a significant paradigm shift in paediatric oncology towards reducing toxicity and addressing long-term treatment-related sequalae amongst childhood cancer survivors. After paediatric nephrectomy and 30–50 years of follow-up, 40% of patients will have chronic kidney disease, including 22% with hypertension and 23% with albuminuria. It is difficult to predict which patients will progress to develop hypertension, reduced glomerular filtration rate, albuminuria, and a higher cardiovascular risk. For these reasons, nephron-sparing surgery when it is technically feasible must be considered. To decrease the incidence of positive surgical margins (viable tumour present at a resection margin), incomplete lymph node sampling, and complications, these procedures should be performed at specialist and experienced reference centres. Based on the impacts of individual treatment pathways, survivors of childhood WT need to be followed through adulthood for early detection of chronic kidney disease, hypertension, and prevention of cardiovascular events. Genomic selection (GS) offers a promising opportunity for selecting more efficient animals to use consumed energy for maintenance and growth functions, impacting profitability and environmental sustainability. Here, we compared the prediction accuracy of multi-layer neural network (MLNN) and support vector regression (SVR) against single-trait (STGBLUP), multi-trait genomic best linear unbiased prediction (MTGBLUP), and Bayesian regression (BayesA, BayesB, BayesC, BRR, and BLasso) for feed efficiency (FE) traits. FE-related traits were measured in 1156 Nellore cattle from an experimental breeding program genotyped for ~ 300 K markers after quality control. Prediction accuracy (Acc) was evaluated using a forward validation splitting the dataset based on birth year, considering the phenotypes adjusted for the fixed effects and covariates as pseudo-phenotypes. The MLNN and SVR approaches were trained by randomly splitting the training population into fivefold to select the best hyperparameters. The results show that the machine learning methods (MLNN and SVR) and MTGBLUP outperformed STGBLUP and the Bayesian regression approaches, increasing the Acc by approximately 8.9%, 14.6%, and 13.7% using MLNN, SVR, and MTGBLUP, respectively. Acc for SVR and MTGBLUP were slightly different, ranging from 0.62 to 0.69 and 0.62 to 0.68, respectively, with empirically unbiased for both models (0.97 and 1.09). Our results indicated that SVR and MTGBLUBP approaches were more accurate in predicting FE-related traits than Bayesian regression and STGBLUP and seemed competitive for GS of complex phenotypes with various degrees of inheritance. As the number of young adults moving to urban areas increases, so does the number of older parents who are left behind in rural areas, where they face loneliness, isolation, hunger, and a lack of social support and care. The lack of remittances and alternative arrangements for caring for the migrant children's elderly parents, as well as the lack of social support networks, makes it difficult for them to cope on their own. The purpose of this study is to ascertain how the migration of younger adults affects the well-being of the left-behind older parents in Nsukka LGA, Enugu State, and to recommend ways to improve their living conditions. Twenty-four elderly parents were purposively chosen for the study. An in-depth interview method was employed for data collection. Using themes and quotes, the generated data were transcribed and analyzed. According to the study, young adults migrate to cities due to poverty, lack of employment and business opportunities, poor hospitals and schools, and lack of social amenities in rural areas. The study also discovered that elderly parents who are left behind in rural areas face hardship and neglect. As a result, to discourage unnecessary migration, we advocated for massive and aggressive rural infrastructure development. Relatively unmanaged interstitial areas at the residential–wildland interface can support the development of novel woody plant communities. Community assembly processes in urban areas involve interactions between spontaneous and cultivated species pools that include native, introduced (exotic/non-native) and invasive species. The potential of these communities to spread under changing climate conditions has implications for the future trajectories of forests within and beyond urban areas. We quantified woody vegetation (including trees and shrubs) in relatively unmanaged "interstitial" areas at the residential–wildland interface and in exurban reference natural areas in six metropolitan regions across the continental USA. In addition, we analyzed soil N and C cycling processes to ensure that there were no major anthropogenic differences between reference and interstitial sites such as compaction, profile disturbance or fertilization, and to explore effects of novel plant communities on soil processes. We observed marked differences in woody plant community composition between interstitial and reference sites in most metropolitan regions. These differences appeared to be driven by the expanded species pool in urban areas. There were no obvious anthropogenic effects on soils, enabling us to determine that compositional differences between interstitial and reference areas were associated with variation in soil N availability. Our observations of the formation of novel communities in interstitial spaces in six cities across a very broad range of climates, suggest that our results have relevance for how forests within and beyond urban areas are assessed and managed to provide ecosystem services and resilience that rely on native biodiversity. Transcription, a critical process in molecular biology, has found many applications in RNA synthesis, including mRNA vaccines and RNA therapeutics. However, current RNA characterization technologies suffer from amplification and enzymatic biases that lead to loss of native information. Here, we introduce a strategy to quantitatively study both transcription and RNA polymerase behaviour by sizing RNA with RNA nanotechnology and nanopores. To begin, we utilize T7 RNA polymerase to transcribe linear DNA lacking termination sequences. Surprisingly, we discover alternative transcription termination in the origin of replication sequence. Next, we employ circular DNA without transcription terminators to perform rolling circle transcription. This allows us to gain valuable insights into the processivity and transcription behaviour of RNA polymerase at the single-molecule level. Our work demonstrates how RNA nanotechnology and nanopores may be used in tandem for the direct and quantitative analysis of RNA transcripts. This methodology provides a promising pathway for accurate RNA structural mapping by enabling the study of full-length RNA transcripts at the single-molecule level. Although platelet-rich plasma-derived exosomes (PRP-Exos) hold significant repair potential, their efficacy in treating rotator cuff tear (RCT) remains unknown. In light of the potential for clinical translation of fibrin gel and PRP-Exos, we evaluated their combined impact on RCT healing and explored suitable gel implantation techniques. In vitro experiments demonstrated that PRP-Exos effectively enhanced key phenotypes changes in tendon stem/progenitor cells. Multi-modality imaging, including conventional ultrasound, shear wave elastography ultrasound, and micro-computed tomography, and histopathological assessments were performed to collectively evaluate the regenerative effects on RCT. The regenerated tendons exhibited a well-ordered structure, while bone and cartilage regeneration were significantly improved. PRP-Exos participated in the healing process of RCT. In-situ gelation of fibrin gel-encapsulated PRP-Exos at the bone-tendon interface during surgery proved to be a feasible gel implantation method that benefits the healing outcome. Comprehensive multi-modality postoperative evaluations were necessary, providing a reliable foundation for post-injury repair. In this study, the authors have investigated if the implantation of fibrin gel loaded with platelet-rich plasma-derived exosomes is a feasible strategy to treat rotator cuff injuries. In recent years groundwater contamination through nitrate contamination has increased rapidly in the managementof water research. In our study, fourteen nitrate conditioning factors were used, and multi-collinearity analysis is done. Among all variables, pH is crucial and ranked one, with a value of 0.77, which controls the nitrate concentration in the coastal aquifer in South 24 Parganas. The second important factor is Cl−, the value of which is 0.71. Other factors like—As, F−, EC and Mg2+ ranked third, fourth and fifth position, and their value are 0.69, 0.69, 0.67 and 0.55, respectively. Due to contaminated water, people of this district are suffering from several diseases like kidney damage (around 60%), liver (about 40%), low pressure due to salinity, fever, and headache. Youths with high levels of callous-unemotional (CU) traits and aggression are at an increased risk for developing antisocial behaviours into adulthood. In this population, neurostructural grey matter abnormalities have been observed in the prefrontal cortex. However, the directionality of these associations is inconsistent, prompting some to suggest they may vary across development. Although similar neurodevelopmental patterns have been observed for other disorders featuring emotional and behavioural dysregulation, few studies have tested this hypothesis for CU traits, and particularly not for aggression subtypes. The current study sought to examine grey matter correlates of CU traits and aggression (including its subtypes), and then determine whether these associations varied by age. Fifty-four youths (10–19 years old) who were characterized for CU traits and aggression underwent MRI. Grey matter volume and surface area within the anterior cingulate cortex was positively associated with CU traits. The correlation between CU traits and medial orbitofrontal cortex (mOFC) volume varied significantly as a function of age, as did the correlation between reactive aggression and mOFC surface area. These associations became more positive with age. There were no significant findings for proactive/total aggression. Cancer is associated with a high degree of heterogeneity, encompassing both inter- and intra-tumor heterogeneity, along with considerable variability in clinical response to common treatments across patients. Conventional models for tumor research, such as in vitro cell cultures and in vivo animal models, demonstrate significant limitations that fall short of satisfying the research requisites. Patient-derived tumor organoids, which recapitulate the structures, specific functions, molecular characteristics, genomics alterations and expression profiles of primary tumors. They have been efficaciously implemented in illness portrayal, mechanism exploration, high-throughput drug screening and assessment, discovery of innovative therapeutic targets and potential compounds, and customized treatment regimen for cancer patients. In contrast to conventional models, tumor organoids offer an intuitive, dependable, and efficient in vitro research model by conserving the phenotypic, genetic diversity, and mutational attributes of the originating tumor. Nevertheless, the organoid technology also confronts the bottlenecks and challenges, such as how to comprehensively reflect intra-tumor heterogeneity, tumor microenvironment, tumor angiogenesis, reduce research costs, and establish standardized construction processes while retaining reliability. This review extensively examines the use of tumor organoid techniques in fundamental research and precision medicine. It emphasizes the importance of patient-derived tumor organoid biobanks for drug development, screening, safety evaluation, and personalized medicine. Additionally, it evaluates the application of organoid technology as an experimental tumor model to better understand the molecular mechanisms of tumor. Twisted testicular appendages had difficult differential diagnosis with testicular torsion. The objective of this paper is to evaluate the number, shape, size and determine the laterality pattern of the testicular and epididymal hydatids and evaluate the correlations between the length and width of the testicular and epididymal hydatids with testicular measurements. We analyzed 60 fixed cadavers and 16 patients with prostate cancer without previous hormonal treatment undergoing bilateral orchiectomy, totalizing 76 units and 152 testicles. In relation to the testicular appendices, we analyzed the following situations: absence of testicular and epididymis appendages, presence of a testicular appendix, presence of epididymis appendix, and presence of testicular and epididymis appendix. We measured the length, width and thickness of the testis and classified the appendages as sessile or pedicled. Chi-square test was used to verify associations between categorical variables. McNemar Test was used to verify differences between the percentages of right and left appendages. Correlations between quantitative measures were evaluated using the Pearson Correlation Coefficient (p < 0.05). In 50 cases (65.78%) we observed the presence of some type of appendices, in 34 cases (44.72%) we observed the presence of testicular appendices and in 19 cases (25%) the presence of epididymal appendices. We observed the presence of pedicled appendices in 39 cases (51.32%), with 25 of the cases (32.89%) of pedicled testicular appendices and 14 of the cases (18.42%) of pedicled epididymal appendages, with a significant association between the occurrence of appendices on the right and left sides (p < 0.001). Testicular hydatids were present in around two thirds of our sample being pedunculated in almost half of the cases with bilateral similarity. There is a significant chance in cases of twisted appendices that the same anatomical characteristics are present on the opposite side, which is a factor that tends to indicate the need for contralateral surgical exploration in cases of torsion, however studies with larger samples are needed to confirm these findings. Childhood head injuries and conduct problems increase the risk of aggression and criminality and are well-known correlates. However, the direction and timing of their association and the role of their demographic risk factors remain unclear. This study investigates the bidirectional links between both from 3 to 17 years while revealing common and unique demographic risks. A total of 8,603 participants (50.2% female; 83% White ethnicity) from the Millennium Cohort Study were analysed at 6 timepoints from age 3 to 17. Conduct problems were parent-reported for ages 3 to 17 using the Strengths and Difficulties Questionnaire (SDQ) and head injuries at ages 3 to 14. A cross-lagged path model estimated the longitudinal bidirectional effects between the two whilst salient demographic risks were modelled cumulatively at three ecological levels (child, mother, and household). Conduct problems at age 5 promoted head injuries between 5 and 7 (Z = 0.07; SE = 0.03; 95% CI, 0.02–0.13), and head injuries at ages 7 to 11 promoted conduct problems at age 14 (ß =.0.06; SE =.0.03; 95% CI, 0.01–0.12). Head injuries were associated with direct child-level risk at age 3, whereas conduct problems were associated with direct risks from all ecological levels until 17 years. The findings suggest a sensitive period at 5–11 years for the bidirectional relationship shared between head injuries and conduct problems. They suggest that demographic risks for increased head injuries play an earlier role than they do for conduct problems. Both findings have implications for intervention timing. Mitigating the negative environmental impacts caused by nitrogen (N) requires an understanding of its flows across and through various systems. This study fills a research gap by identifying the key N sink and sources with flows at three geographical scales (city, region, and country) using a comprehensive literature review. For a systematic literature review, the PRISMA 2020 method was used to select thirty-three studies spanning from 1995 to 2023. The findings of the study indicate that at the city scale, N enters primarily as food and miscellaneous (e.g., fossil fuel) flow and exits as emission and soil losses, while the primary stock of N occurs in landfills. At the region scale, the inflow of N is via feed and fertilizers and the main outflow is emissions and food; the N stock occurs in the soil available in the ecosystem. At the country scale, N imports come from Feed and Fertilizer, while the N outflows occur through emission and food with majority of N stocks held in the soil. The major outflows and stocks identified in this assessment can have potential to improve N management in future studies by acting as a guide for selection of key sectors, appropriate geographical scales, and flows. The main limitation in analysis originates from the varying scope of studies available in the literature. Parental phubbing, as a new risk factor for adolescents' healthy development, has been noticed by scholars. However, few studies have clarified the associations between parental phubbing and adolescents' proactive and reactive aggression and the potential mediating mechanisms. To address the gaps, the current study explored the longitudinal associations among parental phubbing, self-esteem, and adolescents' proactive and reactive aggression by using the parallel process latent growth curve model and the cross-lagged panel model. The current study involved 2407 Chinese adolescents (girls were 1202, Mage = 12.75, SD = 0.58 at baseline, range = 11–16). Results indicated that parental phubbing significantly positively predicted adolescents' reactive aggression but not proactive aggression. Self-esteem significantly predicted the adolescents' reactive and proactive aggression. Self-esteem significantly mediated the associations between parental phubbing and adolescents' proactive and reactive aggression, respectively. These findings suggest that intervening with parental phubbing is a promising way to reduce adolescents' reactive aggression, and promoting adolescents' self-esteem is an effective approach to prevent their proactive and reactive aggression. Atom-controlled sub-nanometer MoS 2 pores have been recently designed thanks to precise defect control techniques. This opens the path for several technological applications, such as electronics, sensing, and energy production and storage. Also, it is well known that oxidative environments are of particular interest for these applications. In this work, we carried out first-principles calculations of oxygen adsorption in pristine and sub-nanometer MoS 2 nanopores. The chemical stability of the 2 H-MoS 2 monolayers and nanopores towards oxygen was verified using density-functional theory and ab initio molecular dynamics (AIMD). Dissociation and diffusion barriers have been calculated in order to understand surface oxidation and electronic properties at the atomic scale. We show that oxygen diffusion depends on the oxygen molecule orientation. Furthermore, we show that oxygen substitutional is stable at room temperature, which helps solving the puzzle of oxygen interaction with 2 H-MoS 2 . [ABSTRACT FROM AUTHOR]

Polyketide or polyketide-like macrolides (pMLs) continue to serve as a source of inspiration for drug discovery. However, their inherent structural and stereochemical complexity challenges efforts to explore related regions of chemical space more broadly. Here, we report a strategy termed the Targeted Sampling of Natural Product space (TSNaP) that is designed to identify and assess regions of chemical space bounded by this important class of molecules. Using TSNaP, a family of tetrahydrofuran-containing pMLs are computationally assembled from pML inspired building blocks to provide a large collection of natural product-like virtual pMLs. By scoring functional group and volumetric overlap against their natural counterparts, a collection of compounds are prioritized for targeted synthesis. Using a modular and stereoselective synthetic approach, a library of polyketide-like macrolides are prepared to sample these unpopulated regions of pML chemical space. Validation of this TSNaP approach by screening this library against a panel of whole-cell biological assays, reveals hit rates exceeding those typically encountered in small molecule libraries. Li-ion batteries (LIBs) have dominated energy-storage techniques for portable electronic devices and electric cars, and are expanding their territory into the large-scale energy storage. The energy storage of LIBs is realized by the reversible shuttle of lithium ions between electrodes. It is essential to track the lithium diffusion and obtain a profound insight into the lithiation mechanism during the work cycle of LIBs. Transmission electron microscopy (TEM) is a powerful tool for the structural characterization, which can provide the information about the lithiation at the atomic scale. In this review, we summarize the research frontiers of TEM applications on LIBs. We introduce the techniques for the direct observation of Li species in LIB-related materials. Especially, the application of cryo-TEM is highlighted. Moreover, in-situ TEM technique is further discussed since it shows great advantages in studying the dynamical structure changes of LIBs. The emergence of migrant grandparent caregivers in urban China calls for research attention to the impact on their well-being. Given the stressful nature of grandparent caregiving and changes to social support under migration, in this study, we aimed to investigate the impact of grandparental stress and social support on migrant grandparent caregivers' affective well-being. Cross-sectional data were collected from nine kindergartens using multistage clustered sampling in Eastern China. The total sample consisted of 483 migrant Chinese grandparent caregivers (mean age = 56.54 years, 67.7% female). Structural equation modeling was used to validate the hypotheses. The results indicated that grandparental stress had a direct negative impact on positive affect (r = -0.130) and a direct positive effect on negative affect (r = 0.125). Both informal (r = 0.368) and formal social support (r = 0.126) were positively associated with positive affect, while only informal social support (r = -0.105) had a significant negative association with negative affect. Additionally, grandparent stress decreased informal social support (r = -0.414) and indirectly influenced both positive and negative affect. In turn, a lack of social support may intensify grandparental stress. The findings validate the integrated theoretical models for studying migrant grandparent caregivers in urban China. Cancer is one of the deadliest diseases affecting the health of human beings. With limited therapeutic options available, complementary and alternative medicine has been widely adopted in cancer management and is increasingly becoming accepted by both patients and healthcare workers alike. Chinese medicine characterized by its unique diagnostic and treatment system is the most widely applied complementary and alternative medicine. It emphasizes symptoms and ZHENG (syndrome)-based treatment combined with contemporary disease diagnosis and further stratifies patients into individualized medicine subgroups. As a representative cancer with the highest degree of malignancy, pancreatic cancer is traditionally classified into the "amassment and accumulation". Emerging perspectives define the core pathogenesis of pancreatic cancer as "dampness-heat" and the respective treatment "clearing heat and resolving dampness" has been demonstrated to prolong survival in pancreatic cancer patients, as has been observed in many other cancers. This clinical advantage encourages an exploration of the essence of dampness-heat ZHENG (DHZ) in cancer and investigation into underlying mechanisms of action of herbal formulations against dampness-heat. However, at present, there is a lack of understanding of the molecular characteristics of DHZ in cancer and no standardized and widely accepted animal model to study this core syndrome in vivo. The shortage of animal models limits the ability to uncover the antitumor mechanisms of herbal medicines and to assess the safety profile of the natural products derived from them. This review summarizes the current research on DHZ in cancer in terms of the clinical aspects, molecular landscape, and animal models. The association between the systemic immune-inflammation index (SII) and the risk of sarcopenia has not yet been revealed. The purpose of this study was to investigate the relationship between the SII and sarcopenia in individuals aged 18–59 years. All data for this study are from the National Health and Nutrition Examination Survey (NHANES) database, including 7258 participants (age range: 18–59 years). We divided SII values by quartiles (quartiles 1–4: 0.3–3.1, 3.2–4.4, 4.4–6.2, and 6.2–58.5). We constructed a multivariate logistic regression model to assess the association between the SII and the risk of sarcopenia, and an interaction test was run to test the stability of the model and identify high-risk individuals with sarcopenia. Compared to nonsarcopenia participants, sarcopenia patients had a significantly higher SII value (weighted average: 6.65 vs. 5.16) (P = 0.002). Multivariate logistic regression results showed a positive linear relationship between the SII and sarcopenia (OR [odds ratio] = 1.12, 95% CI [confidence interval] 1.03–1.21). Compared to the quartile 1 group, the quartile 4 group was associated with a higher risk of sarcopenia (OR = 3.94, 95% CI 1.42–10.94). Compared with the quartile 1 group, the OR value of the quartile 2 to quartile 4 groups showed an upwards trend (Ptrend < 0.001) as the level of SII increased. Subgroup analysis also indicate that the correlation between higher SII values and the risk of sarcopenia was stable. There was a significant positive linear relationship between SII and sarcopenia, indicating that higher SII values can increase the risk of sarcopenia in individuals aged 18–59 in the United States. The findings of this study will be beneficial in promoting the use of SII alone or in combination with other tools for the risk screening of sarcopenia in communities or large populations.
