The current study explored the influence of dietary supplementation of Chlorella vulgaris dried powder (CV) with zinc-oxide-nanoparticles (ZnO-NPs), and/or selenium-nanoparticles (Se-NPs) on broilers' growth, antioxidant capacity, immune status, histological responses, and gene expression of some related genes. Several 200 one-day-old Cobb-500 male chicks were distributed into 5 groups with four replicates each. In the 1st group, birds were fed the basal diet (BD). In the 2nd, 3rd, 4th, and 5th groups, birds received the BD supplemented with CV only, CV + ZnO-NPs, CV + Se-NPs, and CV + ZnO-NPs + Se-NPs, respectively. The CV dried powder, ZnO-NPs, and Se-NPs were added to the BD at a rate of 1 g, 40 mg, and 0.3 mg/kg diet, respectively. After 6 weeks of feeding, increases in final body weights (P < 0.05), body weight gain (P < 0.05), and feed intake (P < 0.05) were linked with improvements in FCR (P < 0.05) and intestinal morphometric indices (P < 0.05), and marked up-regulations of MYOS (P < 0.05), GHR (P < 0.05), and IGF (P < 0.05) genes were established. Additionally, distinct increases in antioxidant enzyme activities of SOD (P < 0.05), and GPX (P < 0.05) with increases in the mRNA copies of their genes were measured. Moreover, slight improvement in immunity indices, WBCs count (P > 0.05), and phagocytic and lysozyme activities (P > 0.05) were found. However, distinct increases in phagocytic index (P < 0.05) and up-regulations of IL-1β and TNF, and down-regulation of IL-10 mRNA levels were reported (P < 0.05). These findings were prominent in the case of the separate supplementation of CV with ZnO-NPs or Se-NPs confirming the synergistic mechanisms of CV with ZnO-NPs or Se-NPs. Naringenin (NRG) inhibits the fungal 17β-hydroxysteroid dehydrogenase accountable for ergosterol synthesis in Candida albicans (C. albicans), a causative agent for cutaneous candidiasis. In present research, NRG was complexed with ZnO nanomaterial (NRG-Zn 2+ ) to synthesize NRG-Zn 2+ nanocomposites. The particle size and ζ-potential of NRG-Zn 2+ nanocomposites were respectively estimated to be 180.33 ± 1.22-nm and - 3.92 ± 0.35-mV. In silico data predicted the greater affinity of NRG-Zn 2+ nanocomposite for 14α-demethylase and ceramide in comparison to NRG alone. Later, NRG-Zn 2+ nanocomposites solution was transformed in to naringenin-zinc oxide nanocomposites loaded chitosan gel (NRG-Zn-CS-Gel) with viscosity and firmness of 854806.7 ± 52386.43 cP and 698.27 ± 10.35 g, respectively. The ex-vivo skin permeation demonstrated 70.49 ± 5.22% skin retention, significantly greater (P < 0.05) than 44.48 ± 3.06% of naringenin loaded chitosan gel (NRG-CS-Gel) and 31.24 ± 3.28% of naringenin solution (NRG Solution). NRG-Zn-CS-Gel demonstrated 6.71 ± 0.84% permeation of NRG with a flux value of 0.046 ± 0.01-µg/cm 2 /h. The MIC 50 of NRG-Zn-CS-Gel against C. albicans was estimated to be 0.156-µg/mL with FICI (fractional inhibitory concentration index) of 0.018 that consequently exhibited synergistic efficacy. Further, NRG-Zn-CS-Gel demonstrated superior antifungal efficacy in C. albicans induced cutaneous candidiasis infection in Balb/c mice. The fungal burden in NRG-Zn-CS-Gel treated group was 109 ± 25 CFU/mL, significantly lower (P < 0.05) than positive control (2260 ± 446 CFU/mL), naringenin loaded chitosan gel (NRG-CS-Gel; 928 ± 127 CFU/mL) and chitosan gel (CS-Gel; 2116 ± 186 CFU/mL) treated mice. Further, histopathology examination and cytokine profiling of TNF-α, IL-1β and IL-10 revealed the healing of skin and inflammation associated with cutaneous candidiasis infection. In conclusion, NRG-Zn-CS-Gel may be a potential candidate for translating in to a clinical viable topical nanotherapeutic., (© 2024. The use of engineered nanoparticles against pathogenic bacteria has gained attention. In this study, zinc oxide nanoparticles conjugated with rutin were synthesized and their antivirulence properties against Pseudomonas aeruginosa and Staphylococcus aureus. The physicochemical characteristics of ZnO-Rutin NPs were investigated using SEM, FT-IR, XRD, DLS, EDS, and zeta potential analyses. Antimicrobial properties were evaluated by well diffusion, microdilution, growth curve, and hemolytic activity assays. The expression of quorum sensing (QS) genes including the lasI and rhlI in P. aeruginosa and agrA in S. aureus was assessed using real-time PCR. Swimming, swarming, twitching, and pyocyanin production by P. aeruginosa were evaluated. The NPs were amorphous, 14-100 nm in diameter, surface charge of -34.3 mV, and an average hydrodynamic size of 161.7 nm. Regarding the antibacterial activity, ZnO-Rutin NPs were more potent than ZnO NPs and rutin, and stronger inhibitory effects were observed on S. aureus than on P. aeruginosa. ZnO-Rutin NPs inhibited the hemolytic activity of P. aeruginosa and S. aureus by 93.4 and 92.2%, respectively, which was more efficient than bare ZnO NPs and rutin. ZnO-Rutin NPs reduced the expression of the lasI and rhlI in P. aeruginosa by 0.17-0.43 and 0.37-0.70 folds, respectively while the expression of the agrA gene in S. aureus was decreased by 0.46-0.56 folds. Furthermore, ZnO-Rutin NPs significantly reduced the swimming and twitching motility and pyocyanin production of P. aeruginosa. Key Message: Nanoparticle pretreatment improved the health of aged Cajanus cajan seeds viz., regulation of redox status, gene expression, and restoration of hormonal homeostasis. Ageing deteriorates the quality of seeds by lowering their vigor and viability, and terminating with loss of germination. These days, nanotechnology has been seen to revolutionize the agricultural sectors, and particularly nano zinc oxide (nZnO) has gained considerable interests due to its distinctive properties. The aim of the present work was to decipher the possibilities of using nZnO to rejuvenate accelerated aged (AA) seeds of Cajanus cajan. Both chemically (CnZnO) and green (GnZnO; synthesized using Moringa oleifera) fabricated nZnOs were characterized via standard techniques to interpret their purity, size, and shape. Experimental results revealed erratic germination with a decline in viability and membrane stability as outcomes of reactive oxygen intermediate (ROI) buildup in AA seeds. Application of nZnO substantially rebated the accrual of ROI, along with enhanced production of antioxidants, α-amylase activity, total sugar, protein and DNA content. Higher level of zinc was assessed qualitatively/ histologically and quantitatively in nZnO pulsed AA seeds, supporting germination without inducing toxicity. Meantime, augmentation in the gibberellic acid with a simultaneous reduction in the abscisic acid level were noted in nZnO invigorated seeds than that determined in the AA seeds, suggesting possible involvement of ROI in hormonal signalling. Furthermore, nZnO-subjected AA seeds unveiled differential expression of aquaporins and cell cycle regulatory genes. Summarizing, among CnZnO and GnZnO, later one holds better potential for a revival of AA seeds of Cajanus cajan by providing considerable tolerance against ageing-associated deterioration via recouping the cellular redox homeostasis, hormonal signaling, and alteration in expression patterns of aquaporin and cell cycle regulatory genes., (© 2024. An 8-week feeding trial was carried out to examine the effect of different sources of dietary Zn on some physiological responses (performance, digestive enzymes activity, hemato-biochemical parameters, antioxidant status and liver histology) of Siberian sturgeon, Acipenser baerii. For this purpose, fish with an average weight of 100 g ± 5 were randomly allocated into four groups including control, inorganic zinc (Zn-sulfate), organic zinc (Zn-gluconate), and zinc-oxide nanoparticles (ZnO-NPs) at 50 mg Zn kg - 1 feed. Improved growth indices, namely weight gain (WG) and specific growth rate (SGR) and feed conversion ratio (FCR) were observed in fish fed Zn-gluconate supplemented diet (P < 0.0.5). The highest digestive enzymes activity was recorded in fish fed Zn-gluconate supplementation (P < 0.0.5). Hematological indices significantly increased in fish fed diet containing ZnO-NPs (P < 0.0.5). Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) of fish fed ZnO-NPs contained diet were the highest (P < 0.0.5). The highest serum superoxide dismutase (SOD) and catalase (CAT) enzymes activity were observed in fish fed ZnO-NPs and inorganic/organic Zn contained diets, respectively. While liver tissue SOD and glutathione peroxidase (GPx) enzymes activity Zn were significantly increased in fish fed inorganic/organic Zn supplemented diet (P < 0.0.5). Based on liver histological results, a severe tissue changes such as necrosis and pyknosis were observed in fish fed with Zn-sulfate in comparison to other forms. Black heart rot is a serious disease of apricot and it has been reported to be caused by Alternaria solani, around the world. The present research was designed to control this disastrous disease using zinc oxide nanoparticles (b-ZnO NPs). These NPs were synthesized in the filtrate of a useful bacterium (Bacillus safensis) and applied to control black heart rot of apricot. After synthesis, the reduction of b-ZnO NPs was confirmed by UV-visible spectroscopy, at 330 nm. Fourier transform infrared (FTIR) spectra ensured the presence of multiple functional groups (alcohols, phenols, carboxylic acids, nitro compounds and amines) on the surface of b-ZnO NPs. X-Ray diffraction (XRD) analysis elucidated their average size (18 nm) while scanning electron microscopy (SEM) micrograph described the spherical shape of b-ZnO NPs. The synthesized b-ZnO NPs were applied in four different concentrations (0.25 mg/ml, 0.50 mg/ml, 0.75 mg/ml, 1.0 mg/ml) under both in vitro and in vivo conditions. These NPs were very efficient in inhibiting mycelial growth (85.1%) of A. solani at 0.75 mg/ml concentration of NPs, in vitro. Same concentration also performed best, in vivo, and significantly reduced disease incidence (by 67%) on self-inoculated apricot fruit. Apart from this, application of b-ZnO NPs helped apricot fruit to maintain its quality under fungal-stress conditions. The decay of apricot fruit was reduced and they maintained greater firmness and higher weight. Moreover, b-ZnO NPs treated fruits controlled black heart rot disease by maintaining higher contents of ascorbic acid, soluble sugars and carotenoids. A total of 150 21-day-old weaned piglets [(Yorkshire × Landrace) × Duroc] were randomly assigned to 3 groups (CON, TRT1, TRT2) to evaluate the effects of dietary supplementation of probiotic, paraprobiotic, and hydrolyzed yeast mixture (PPY) on growth performance, nutrient digestibility, fecal bacterial counts, fecal calprotectin contents, and diarrhea rate in a 42-day experiment (phase 1: days 1-14; phase 2: days 15-42). There were 10 replicate pens per treatment with 5 pigs per pen (three gilts and two barrows). Pigs in CON were only provided with a basal diet. Pigs in TRT1 were provided with a basal diet + 3000 mg/kg zinc oxide during phase 1 and a basal diet during phase 2. Pigs in TRT2 were provided with a basal diet + 200 mg/kg probiotic (Saccharomyces cerevisiae boulardii) + 800 mg/kg paraprobiotic (inactivated yeast strains of Saccharomyces cerevisiae and Cyberlindnera jadinii) + 10 g/kg hydrolyzed yeast mixture during phase 1, and a basal diet + 100 mg/kg probiotic + 400 mg/kg paraprobiotic mixture during phase 2. Pigs in TRT1 and TRT2 were significantly heavier at day 14 and 42 than CON pigs. Growth rate during days 1-14, 15-42, and 1-42 and feed efficiency during days 1-14 were similarly affected by treatment while feed efficiency was significantly higher for TRT2 pigs between 15-42 and 1-42 days. Moreover, nitrogen and energy digestibility in both TRT1 and TRT2 were higher than that in CON. During experimental periods, diarrhea rate in TRT1 and TRT2 was lower than that in CON. Therefore, we demonstrated that PPY supplementation had comparable effects as ZnO in improving growth performance and nutrient digestibility as well as ameliorating post-weaning diarrhea in weaned piglets., (© 2022. The increasing number of cancer patients has cast attention on developing new anti-cancer modalities. Photodynamic therapy is a safe anti-cancer approach, which encompasses (1) local administration of a photosensitizer and (2) light irradiation. Zinc oxide (ZnO) quantum dots (QDs) are photosensitizers that can be utilized for this purpose. In the present study, to better appreciate the likely more efficient cytotoxic effect of the combination of ZnO QDs and the visible 470-nm blue light in comparison to the QDs alone, several assays were to be conducted upon breast cancer MDA-MB 231 cells. MTT assay showed that in certain groups the combination displayed higher cytotoxic effects compared to those following QD treatment alone. LDH leakage and lipid peroxidation rates by the combination were significantly higher than treatment with either the blue laser or QDs. Although the combination managed to meaningfully reduce the number of colonies and CAT activity compared to QD treatment, there were no palpable differences between them. Lastly, the combination was able to increase the apoptotic genes, including BAX, TP53, caspase 3, and caspase 9 compared to QD, while, in the case of Bcl-2, an anti-apoptotic gene, none of the groups managed to make any tangible differences on its expression levels. Our findings propose that there may be synergistic effects between the blue laser and QD that can possibly be adopted in anti-cancer therapy in the future. Bordeaux mixture is commonly used in agricultural production due to its certain antibacterial activity. However, it has been observed to promote plant growth at a slow pace. Therefore, it is crucial to explore an effective antibacterial agent that can enhance the antibacterial activity and promote plant growth in commercially available Bordeaux mixture, which can significantly contribute to the development of the agricultural economy. The investigation into inorganic agents with both bacteriostatic and plant-promoting properties has a broad application potential in agriculture. Fe 3 O 4 /ZnO (FZ) composites were synthesized from FeCl 3 , ZnCl 2 , and NaAc in a "one-pot approach" and analyzed using transmission electron microscopy (TEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and a vibrating sample magnetometer (VSM). To investigate the antibacterial activity and mechanism of FZ nanocomposites, Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus) were used as model bacteria, and human mammary epithelial cells and model plant mung bean were used as targets to study the effects of FZ on human and plant growth. The results revealed that at 300 µg/mL for 80 min, the antibacterial efficacy of FZ composites was 99.8% against E. coli, which was 20% greater than that of Bordeaux liquid (FC), and 99.9% against S. aureus, which was 28.6% higher than that of FC. The inhibitory mechanism demonstrated that the substance could efficiently damage the bacterial cell wall of a concentration of 300 µg/mL. The IC 50 of the material to human mammary epithelial cells was 49.518 µg/mL, and it also increased mung bean germination, root growth, and chlorophyll content, indicating that the application performance was 1.5 times better than that of FC. Biofilms are made up of bacterial colonies and their extracellular polymeric substances (EPS) matrix, which protects the bacteria from adverse environmental conditions. The increasing drug resistivity of pathogenic bacteria is becoming an emergency for developing new antibacterial agents. In this study, we have synthesized the zinc oxide nanoparticles (ZnO NPs) using the leaf extract of Saraca asoca plant, and the antibacterial and antibiofilm activity of green synthesized ZnO NPs was measured against the biofilm-producing bacteria Bacillus subtilis. The disk diffusion data reveals that the zone of inhibition (ZOI) starts at a concentration of 0.5 mg/mL and minimum inhibition concentration (100 µg/mL) and minimum bactericidal concentration (150 µg/mL) values were also evaluated for green synthesized ZnO nanomaterials. Crystal violet test and microscopic examination were used to assess the impact of produced nanoparticles on biofilm development. The findings indicated a nearly 45%, 64%, and 83% suppression of biofilm development at 0.5 × MIC, 0.75 × MIC, and 1 × MIC value, respectively. The biofilm biomass of the preformed or matured biofilms by the ZnO NPs was evaluated to be 68%, 50%, and 33% at concentrations of 0.5 × MIC, 0.75 × MIC, and 1 × MIC which was concentration-dependent. Moreover, flow cytometry results suggest damage to the bacterial cell membrane. The data indicated that the proportion of dead cells increased with NP concentration in comparison to the control. Therefore, it can be concluded that the green synthetic ZnO nanoparticles showed excellent antibacterial and antibiofilm activity against the Bacillus subtilis bacteria that produce biofilms and that they could be a promising substitute agent for the treatment of biofilms and drug-resistant bacteria., (© 2023. Currently, ascorbic acid (AA) is widely used as a skin whitening material, but, AA, an unstable hydrophilic molecule, cannot penetrate the skin easily, due to the hydrophobic character of the stratum corneum. Therefore, we conjugated AA with hydrated zinc oxide-an inorganic matrix with positive surface charge, to improve the stability of AA. The metal-conjugated-ascorbic acid (ZnAA) was then combined with yeast vacuole through the vacuolar membrane proteins that relate to metal transportation to create an enhanced vacuole that contained ZnAA. The characteristics of vacuole with ZnAA (ZnAA&#95;Vac) were next examined by various tests that included X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), Field emission scanning electron microscopy (FE-SEM), and energy-dispersive X-ray (EDX) analysis. Furthermore, the ability of ZnAA&#95;Vac to degrade melanin was confirmed in both melanoma cell line B16F10, and the artificial human skin MelanoDerm. The results showed that ZnAA&#95;Vac possessed a higher depigmenting effect than the wild-type vacuole or ascorbic acid by reducing 75% of melanin color. Interestingly, ZnAA&#95;Vac was found to be harmless, and did not cause any cytotoxicity to the cells. Nano materials have found developing interest in biogenic approaches in the present times. In this study, metal oxide nanoparticles (NPs) such as cobalt oxide (Co 3 O 4 ), copper oxide (CuO), nickel oxide (NiO) and zinc oxide (ZnO), were synthesized using a convenient and rapid method. The structural features of synthesized metal oxide NPs were studied using various microscopic and spectroscopic techniques like SEM, TEM, XRD, FTIR and EDX. The characterization results confirmed that the prepared NPs possess highly pure, unique and crystalline geometry with size ranging between 10 and 20 nm. The synthesized nanoparticles were successfully employed for pharmacological applications. Enzyme inhibition potential of NPs was evaluated against the urease and tyrosinase enzymes. The percent inhibition for the urease enzyme was observed as 80 to 90% by using Co 3 O 4 , CuO, NiO and ZnO NPs while ZnO NPs were found to have best anti-urease and anti-tyrosinase activities. Moreover, effective inhibition was observed in the case of ZnO NPs at IC 50 values of 0.0833 and 0.1732 for urease and tyrosinase enzymes which were comparable to reference drugs thiourea and kojic acid. The lower the IC 50 value, higher the free radical scavenging power. Antioxidant activity by DPPH free radical scavenging method was found moderately high for the synthesized metal oxide NPs while best results were obtained for Co 3 O 4 and ZnO NPs as compared to the standard ascorbic acid. Antimicrobial potential was also evaluated via the disc diffusion and well diffusion methods. CuO NPs show a better zone of inhibition at 20 and 27 mm by using both methods. This work studied the effects of the inclusion of Purple Garlic Powder (PGP) and Oregano Essential Oil (OEO) in the feed, at different doses and combinations, on intestinal health and the growth performance of 140 and 3000 piglets, respectively, weaned at 21 days of age. Seven dietary treatments were used: a negative control group (basal diet), a positive control group with ZnO (3000 mg/Kg of feed), two groups with OEO at 0.4% and 1.2% respectively, two groups with PGP at 0.4% and 2% respectively and one group with OEO at 1.2% combined with PGP at 2%. Only the positive control group received ZnO in the diet. Each group of piglets received the treatment for seven weeks, from weaning, and were later sacrificed to obtain jejunum and ileum samples for counting of goblet cells, intraepithelial lymphocytes, and IgA-producing cells. The growth performance were measured at the beginning and at the end of the seven weeks. In jejunum and ileum, the number of goblet cells increased in the groups with ZnO, PGP 2%, OEO 1.2% and PGP 2% + OEO 1.2%, presenting significant differences with the rest of the groups. The results obtained for the intraepithelial lymphocyte count were in line with those obtained for the count of goblet cells. Regarding IgA-producing cells, the groups that showed significantly favourable results in the jejunum and ileum were OEO 1.2%, PGP 2% and their combination, but the groups that showed the most similar means to ZnO were the OEO 0.4% and the PGP 0.4%. Regarding the growth performance, PGP 2%, OEO 1.2% and their combination had similar results to ZnO. The intestinal health of piglets could be improved, without harming the growth performance, by means of the supplementation of PGP 2%, OEO 1.2% and PGP 2% + OEO 1.2% offering a natural alternative to the use of ZnO., (© 2022. Nowadays, there are concerns about the inadequacy of new antimicrobials and the rise of antimicrobial resistance. Hence, novel antibacterial agents need to be discovered. In this respect, the use of nanoparticles (NPs) seems promising. Zinc oxide nanoparticles (ZnONPs) are functional and inexpensive NPs that possess antimicrobial characteristics, stability, microbial selectivity, and an easy manufacturing procedure. Imidazolium is one of the quaternary ammonium compounds (QACs) frequently employed as antimicrobial materials in industrial and clinical fields. The present study successfully employed imidazolium to couple with ZnONPs to improve their antimicrobial properties. The antimicrobial activities of ZnONPs doped with imidazolium (IM@ZnONPs) compared to ZnONPs and zinc (Zn) ions against some pathogen microorganism species including Streptococcus aureus (S. aureus), Enterococcus faecalis (E. faecalis), methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant enterococci (VRE), and Candida albicans (C. albicans) were evaluated by the microdilution method. The minimum inhibitory concentration (MIC) results revealed that the antimicrobial activities of Zn ions, ZnONPs, and IM@ZnONPs were concentration-dependent. Moreover, we found that the nanoparticulate forms of Zn had considerably stronger antibacterial activities, particularly against VRE and MRSA, compared to Zn ions which failed to restrain the microbial strains at the tested microdilutions of this experiment (MIC: ≥512 µg/mL). Interestingly, the incorporation of imidazolium into ZnONPs resulted in significant inhibition of microbial growth in antimicrobial-resistant pathogens at low concentrations (MIC: 32 µg/mL) and effectively improved the monodispersity of the final coated NPs in terms of size and morphology. To sum up, IM@ZnONPs can be a favorable substitute for conventional antimicrobial agents to combat antimicrobial resistance in many fields, including pharmaceuticals, dental materials, and cosmetic products., (© 2023. Due to their distinctive characteristics and widespread application across all scientific disciplines, nanoparticles have attracted a lot of attention in the current millennium. Green synthesis of ZnO-NPs is gaining a lot of interest at the moment due to a number of its advantages over traditional methods, including being quicker, less expensive, and more environmentally friendly. In the current study, two distinct plant extracts are used to quickly, cheaply, and environmentally friendly synthesize zinc oxide nanoparticles (ZnO-NPs). Mint (Mentha spicata) and basil (Ocimum basilicum) were the plants employed in this study as stabilizing agents to synthesize ZnO-NPs with a green chemistry approach. The innovative aspect of the study is the use of mint and basil extracts in the conversion of zinc chloride to zinc oxide and then determining the effect of these two types of nanoparticles produced by green synthesis on the growth parameters of the plant when they reach the plants by foliar spraying and their uptake by plants and evaluating the antibacterial properties of these nanoparticles. The physical properties of the produced nanoparticles were investigated using XRD, SEM, and FTIR. Moreover, Escherichia coli and Staphylococcus aureus were used to demonstrate the antibacterial properties of ZnO-NPs against both gram-positive and gram-negative bacteria, respectively. Synthesized ZnO-NPs were also given as foliar treatment in order to determine Zn +2 uptake by plants and potential toxic effects on the growth of wheat. The shape of ZnO-NPs was triangular, as revealed by SEM analysis. In the X-ray diffraction study, strong and clearly discernible sharp peaks were seen, with an average size of 24.5 nm for M-ZnO-NPs and 26.7 nm for B-ZnO-NPs determined using Scherrer's formula. The phytoconstituents of the plant extract served as capping/stabilizing agents during the synthesis of ZnO-NPs, as demonstrated by Fourier transform-infrared spectroscopy. The produced nanoparticles were applied to the green parts of wheat plants by spraying, and the development of the plants and the change of zinc uptake were investigated. At the same time, the effect of these three types of nanoparticles on the germination of wheat seeds in the soil medium containing these nanoparticles was investigated. According to experimental results, M-ZnO-NPs (produced from mint) and B-ZnO-NPs (produced from basil) improved the germination percentage of wheat at 400 mg/L concentration (100%), while raw ZnO-NPs showed 90% germination at the same concentration. When the Zn +2 uptake of the plant by the leaves depending on the Zn +2 concentration in the environment after spraying was examined, it was determined that the Zn +2 uptake of the plants increased due to the increase in the applied Zn +2 concentration. The highest Zn +2 uptake of the plant was determined as 50, 25, and 50 mg/L for M-ZnO-NP, B-ZnO-NPs, and raw ZnO-NPs, respectively. Therefore, it has been determined that plant growth varies depending on the type and concentration of ZnO-NPs, and therefore, if foliar nanoparticle applications are made to wheat, the threshold concentrations, sizes, and types of ZnO-NPs should be carefully evaluated. Due to an inevitable disadvantage of chemical or physical synthesis routes, biosynthesis approach to nanoparticles, especially metallic oxide is attractive nowadays. Metallic oxides nanoparticles present a new approach to the control of plant pathogens. ZnO nanoparticles (ZNPs) have very important role in phytopathology. In current study, biosynthesized ZNPs were tested against two devastating bacterial pathogens including Xanthomonas campestris pv. vesicatoria and Ralstonia solanacearum causing bacterial leaf spot and bacterial wilt in tomato. ZNPs were produced using a new extract from the plant Picea smithiana using an environmentally friendly, cost-effective and simple procedure. Zinc acetate was added to P. smithiana extract, stirred and heated to 200 °C. The white precipitation at the bottom were clear indication of synthesis of nanoparticles, which were further dried by subjecting them at 450 °C. X-ray diffraction pattern determined that the ZNPs had a crystallite size of about 26 nm, Fourier transform infrared spectroscopy indicated a peak between 450 and 550 cm -1 and the particle size estimated by dynamic light scattering was about 25 nm on average. Scanning electron microscopic analysis indicated that the particles were hexagonal in shape 31 nm in diameter. Antibacterial tests showed ZNPs synthesized by P. smithiana resulted in clear inhibition zones of 20.1 ± 1.5 and 18.9 ± 1.5 mm and 44.74 and 45.63% reduction in disease severity and 78.40 and 80.91% reduction in disease incidence in X. compestris pv. vesicatoria and R. solanacearum respectively at concentration of 100 µg/ml. Our findings reveal that the concentration of ZNPs was important for their efficient antibacterial activity. BiSnSbO 6 -ZnO composite photocatalytic material with type II heterojunction structure was synthesized by a simple solid-phase sintering method, it was characterized by XRD, UV-vis, and PT methods. The photocatalytic antibacterial experiments were carried out under LED light irradiation. The experimental results showed that the photocatalytic antibacterial properties of BiSnSbO 6 -ZnO composites against bacteria and fungi were significantly stronger than those of single BiSnSbO 6 and ZnO. Under light conditions, the antibacterial efficiencies of 500 mg/L BiSnSbO 6 -ZnO composites against E. coli, S. aureus, and P. aeruginosa reached 99.63%, 100%, and 100% for 6 h, 4 h, and 4 h, respectively. The best antibacterial concentration of BiSnSbO 6 -ZnO composite against the eukaryotic microorganism Candida albicans was 250 mg/L, and the antibacterial efficiency reached the highest 63.8% at 6 h. Antibacterial experiments were carried out on domestic livestock and poultry wastewater, which showed that the BiSnSbO 6 -ZnO composite photocatalytic material has broad-spectrum antibacterial activity against bacteria, and the antibacterial effect has species differences. Through the MTT experiment, it is proved that the prepared BiSnSbO 6 -ZnO composite photocatalytic material has no toxicity at the experimental concentration. According to the free radical scavenging experiment and SEM observation of the morphological changes of the bacteria after light treatment, the prepared BiSnSbO6-ZnO composite photocatalytic material can generate active species OH, h + , and e - through light irradiation to achieve the purpose of sterilization, where e - play a major role, indicating that the BiSnSbO 6 -ZnO composite photocatalytic material has broad application prospects in the actual antibacterial field., (© 2023. In this study, the manufacture of zinc oxide nanoparticles (ZnO-NPs) was completed via the sol-gel method with Trigonella foenum-graecum L extract for the first time to function as the stabilizing and reducing agent. The obtained product was investigated by various analyzing procedures such as TGA/DTG, FT-IR, UV-Vis, XRD, and EDX/FESEM. The calcination of our product was conducted at temperatures of 400, 500, and 600 °C. In conformity to the XRD pattern, heightening the temperature of calcination caused an enlargement in the size of nanoparticles. The photocatalytic performance of ZnO-NPs was evaluated to degrade methylene blue and Eriochrome black T (EBT) dyes under UV light, which resulted in a degradation percentage of about 96% and 94%, after 90 min, respectively. There has been some evidence suggesting that the green synthesis of ZnO-NPs has increased their use in medicine. The outcomes of examining the cytotoxicity effect of this product against the Huh-7 cell line by the performance of the MTT assay were indicative of an IC 50 of around 62.5 µg/mL. Finally, according to the results of the broth microdilution method, which was performed to assess the antibacterial activity of ZnO-NPs towards gram-positive and gram-negative bacteria, the value of MIC was in the range of 31 to 125 µg/mL. Here, silica sand-supported heterojunction composite of nano zinc oxide (nZnO) and graphene oxide nanosheet (nZnO-GO@SS) was prepared, and its potential as an efficient photocatalyst for the degradation of methylene blue (MB) and Rhodamine-B (Rh-B) under sunlight was demonstrated. Transmission electron microscopy confirmed the uniform distribution of spherically shaped nZnO of average size of approximately 8 nm over graphene oxide nanosheet (GO) in the composites. Photodegradation yields of 95.3% and 97.5% for 100 ppm of MB and Rh-B dye within 150 and 220 min, respectively, were achieved under sunlight by the prepared nanocatalyst (nZnO-GO), while sand microparticle-supported nanocatalyst (nZnO-GO@SS) demonstrated faster degradation of MB and Rh-B, i.e., within 120 and 160 min, respectively. Furthermore, when the recyclability of the photocatalyst was studied, the nZnO-GO exhibited more than 80% degradation efficiency after five cycles for both the dyes and nZnO-GO@SS demonstrated 10% higher (~90%) removal capability after five cycles of reuse. Furthermore, the antibacterial assay showed complete inactivation of Escherichia coli and Staphylococcus aureus bacterial strain by nZnO-GO@SS. Hence, our proposed strategy for the removal of toxic dyes from the aquatic environment under sunlight proved that sand microparticle-supported nanocatalyst (nZnO-GO@SS) might be a superior, cost-effective, and suitable photocatalytic system for industrial applications toward toxic dye removal and decontamination from industrial wastewater., (© 2022. The present study represents new evidence of the successful use of metal oxide nanoparticles in the remediation of heavy metals. Zinc oxide nanoparticles (ZnO NP) and copper oxide nanoparticles (CuO NP) were assessed to alleviate cadmium (Cd) toxicity in Phaseolus vulgaris L. seedlings and plants. Monitoring physiological and metabolic parameters allowed to elucidate Cd mechanism and process whereby it exerts phytotoxic effects on bean. The response of P. vulgaris seedlings is NP dose-dependent (10 mg/L, 50 mg/L, 100 mg/L, and 200 mg/L). Similarly, applied concentrations triggered a differential response of growing plants in terms of length and biomass. Our physiological data allowed to select 100 mg/L as the most appropriate concentration to apply, in order to avoid any risk of phytotoxicity. The regulatory mechanisms by which ZnO NP and CuO NP act are for the first time compared in the embryonic axes of bean seedlings under Cd stress. Both NP were able to reduce the hypergeneration of hydrogen peroxide (H 2 O 2 ). They also acted via enhancing ROS scavenging enzymatic capacity, and activity of antioxidant enzymes CAT, APX, GPOX, GPX, and GR, and inhibited the activity of ROS producing enzymes such as GOX and NOX. Another mechanistic effect of NP consisted of the modulation of redox enzymes Trx, NTR, Fd, and FNR evolved in cellular homeostasis and maintaining reduced status in cells. Chitosan (CS) is one of the most abundant biopolymers in nature with superior properties such as biocompatibility, biodegradability, lack of toxicity, antimicrobial activity, acceleration of wound healing, and stimulation of the immune system. In this study, chitosan was extracted from the exoskeletons of beetles (Pimelia payraudi latreille) and then used for the biosynthesis of highly pure MgO NPs and ZnO NPs by a facile greener route. The extracted chitosan exhibited excellent physicochemical properties, including high extraction yield (39%), high degree of deacetylation (90%), low ash content (1%), high fat-binding capacity (366%), and unusual crystallinity index (51%). The MgO NPs and ZnO NPs exhibited a spherical morphology with crystallite sizes of 17 nm and 29 nm, particle sizes of about 20-70 nm and 30-60 nm, and band gap energies of 4.43 and 3.34 eV, respectively. Antibacterial assays showed that the extracted chitosan exhibited high antibacterial activity against Gram-positive and -negative bacteria, while ZnO NPs showed much stronger antibacterial activity against Gram-positive bacteria than against Gram-negative bacteria. For MgO NPs, the antibacterial activity against Gram-positive bacteria was lower than against Gram-negative bacteria. Investigation of ZnO nanoparticles (nano-ZnO) and graphene quantum dots (GQDs) toxicology on dinoflagellate Gymnodinium helps to understand the effects of different surface characteristic nanoparticles on marine algae. The growth and biological responses of the algae exposed to 1, 10, 20 mg L -1 nano-ZnO and GQDs in f/2 media were explored. Nano-ZnO showed slight effects on algal cells growth, while the growth inhibition rates of Gymnodinium increased as GQDs concentration increasing. Both nanoparticle treatments induced accumulation of reactive oxygen species and activated intracellular antioxidant defensive system, including SOD and ATPase, which were related to the two nanoparticles concentration. Under combined exposure of nano-ZnO and GQDs, the inhibitory effects decreased compared to the single GQDs and showed antagonistic effect. The addition of nano-ZnO could decrease the toxicity of GQDs due to aggregation and sedimentation interaction between nanoparticles. Energy-levels well-matched direct Z-scheme ZnNiNdO/CdS heterojunction was successfully fabricated using facile co-precipitation and ultra-sonication techniques and characterized with XRD, FTIR, Raman, PL, UV-vis, and FE-SEM. The XRD diffractograms confirmed the co-doping of Ni-Nd in ZnO and the formation of heterostructured nanocomposite. FTIR and Raman data showed the presence of metal-oxygen vibration and optical phonon modes of ZnO and CdS. FE-SEM images exhibited the network type morphology. The energy bandgap was redshifted by co-doping (3.37-2.9 eV) and was further reduced (2.6 eV) by making a composite with CdS. The ZnNiNdO/CdS catalyst degraded 99.7, 49, 96.6, 98.6, and 98.6% methylene blue (MB), p-nitroaniline (P-Nitro), methyl orange (MO), methyl red (MR), and rhodamine B (RhB) dyes under 50 min sunlight irradiation. Moreover, ZnNiNdO/CdS showed intense inhibition activity towards Staphylococcus aureus, Escherichia coli, Proteus vulgaris, and Pseudomonas aeruginosa bacterial strains with maximum inhibition zone diameters 30, 33, 27, and 31 mm, respectively. The synergistic effects arising from band alignment can lead to efficient vectorial charge separation, transportation, and lower recombination of photoinduced charge carriers, ultimately boosting photocatalytic and antibacterial performance. The ZnNiNdO/CdS photocatalyst has higher stability up to the 7th cycle towards MB dye with ~ 5% deficit in degradation efficiency. The higher generation of superoxide and hydroxyl radical was confirmed by species trapping experiments responsible for photodegradation of dyes molecules. The comparative effects of potentiated zinc oxide (pZnO) and antibiotic growth promoters (AGP) supplementation on intestinal morphometry and nutrient digestibility in broiler chickens were studied. Four hundred straight-run Cobb 500-day-old broiler chicks were randomly allotted to four dietary treatments replicated 10 times with 10 birds per replicate. Dietary treatments were as follows: T1: basal diets without AGP (negative control; NC), T2: basal diets with 500 g/t maduramicin 10 g and 500 g/t zinc bacitracin 150 (positive control; PC), T3: NC added with 150 g/t pZnO, and T4: PC added with 150 g/t pZnO in a 2 × 2 factorial design in RCBD. At days 18 and 35, 10 birds were randomly selected per treatment for morphometry of the duodenum, jejunum, and ileum. At day 38, eight birds per treatment were used for the nutrient digestibility study. Results showed significant interaction effects (P < 0.05) of AGP and pZnO supplementation on day 35 intestinal morphometry of duodenum's villi height and villi height: crypt depth, and ileum's crypt depth; apparent COD GE , AME, CP, DM, and EE. Significant differences (P < 0.05) with pZnO supplementation were only observed on feed intake and FCR of birds fed with pZnO at days 8-14 and fecal quality at days 0-7. The antimicrobial efficacy of rhamnolipid is well established against a wide range of pathogens. However little is known about the enhancement of antimicrobial efficacy of rhamnolipid in the form of nanoparticles. With a curiosity of enhancing antimicrobial activity, a study has been carried out to evaluate the antimicrobial efficacy of rhamnolipid-coated zinc oxide nanoparticles. The zinc oxide nanoparticles were synthesized with rhamnolipid, produced by Pseudomonas aeruginosa JS29. The rhamnolipid-coated zinc oxide nanoparticles were characterized by FTIR, XRD, TGA, TEM, and SAED. The antimicrobial and antibiofilm efficacy of the nanoparticles was evaluated against Staphylococcus aureus MTCC 96. FTIR, XRD, TEM, and SAED analyses confirmed that the nanoparticles contain both rhamnolipid and zinc as constituents and are polycrystalline with sizes ranging from 40 to 50 nm. At a concentration of 250 µg/ml, rhamnolipid-coated zinc oxide nanoparticles exhibited 80% growth inhibition of the pathogen. Again, at the same concentration, the nanoparticle was observed to inhibit 78% of biofilm formation while disrupting 100% of preformed biofilm. The nanoparticles demonstrated an enhanced inhibitory and antibiofilm efficacy against the pathogen compared to the individual effect of both rhamnolipid and zinc oxide nanoparticles. With the established non-toxicity of rhamnolipid-coated zinc oxide nanoparticles in fibroblast cell lines, the nanoparticles could be a promising pharmaceutical alternative., (© 2021. Sunscreen safety and efficacy is generally evaluated based upon the properties of the individual chemicals in a formulation. However, the photostability of sunscreens has been shown to be highly dependent on the mixture of chemicals present. To better understand how sunscreen formulation influences stability, and to establish a foundation for probing the influence of zinc oxide additives, we formulated five different small-molecule based ultraviolet-filter (UV-filter) mixtures with a Sun Protection Factor (SPF) of 15. These mixtures contained active ingredients approved in either the United States or European Union and were designed to represent formulations of actual products on the market. We evaluated the photostability and toxicity of these mixtures in the absence and presence of zinc oxide after UV exposure for two hours. Changes in UV absorbance were minimal for all five small-molecule-based mixtures without zinc oxide. The presence of either micro- or nano-sized zinc oxide caused significant small-molecule photodegradation and the degraded mixtures exhibited higher levels of toxicity in embryonic zebrafish assays. Zinc oxide (ZnO) nanoparticles (NPs) are generally utilized in cosmetic goods, sheds, biosensors, and delivery of drug. As in vitro ideal systems, mesenchymal stem cells (MSCs) are used to test acute toxicity. In the present study, size-dependent cytotoxicity effects of ZnO NPs on MSCs were assessed. Bone marrow and adipose MSCs were treated with ZnO NPs with average sizes of 10-30 and 35-45 nm. The 5 and 10 µg/ml concentrations of ZnO NP were found to be the safe concentrations for the NP sizes of 10-30 and 35-45 nm, respectively. Cell-cycle analysis indicated that the small size of ZnO NPs has more negative effects on the process of cell entry to DNA synthesis when compared to the larger size. The results of the β-galactosidase test showed the promotion of the aging process in the cells treated with the smaller size of ZnO NPs. Both sizes of the NP were found to upregulate the aging-related genes NF-kB and p53 and downregulate the anti-aging gene Nanog. Cell density-based intercellular signaling mechanism is known as Quorum sensing (QS); it serves a significant role in regulating the pathogenic factors. The objective of the present study was to assess the influence of chitosan-zinc oxide nanocomposite (CH-ZnO nanocomposite), alone and in combination with gentamicin, on the sensitivity to hydrogen peroxide (H 2 O 2 ), the production of pathogenic factors and QS-regulated genes of Pseudomonas aeruginosa. The efficacy of the minimum inhibitory concentration (MIC) and 1/4 MIC of the CH-ZnO nanocomposite, alone and in combination with gentamicin, on the sensitivity to H 2 O 2 , pyocyanin secretion, swarming and twitching motilities was evaluated. In addition, the expression of some QS-regulated genes including rhlI, rhlR, lasI and lasR genes was measured by Real-time quantitative PCR (RT-qPCR) following exposure to the nanocomposite. The results demonstrated that at MIC concentrations, the gentamicin-loaded CH-ZnO nanocomposite significantly inhibited QS-regulated phenotypes such as pyocyanin secretion (82.4%), swarming (76%) and twitching (73.6%) motilities; further it increased the inhibition growth zone (134.5%), as well as, at 1/4 MIC concentration decreased the expression of lasI (72%), lasR (78%), rhlI (76%) and rhlR (82%) genes; as compared to untreated P. aeruginosa PAO1 (P < 0.05). Diabetes mellitus is a common chronic metabolic disorder. This study aimed to investigate the effects of co-treatment with L-carnitine (LC) and zinc oxide nanoparticles (ZnONPs) on serum levels of sex hormones, oxidative stress, and ovarian histopathology in streptozotocin (STZ)-induced diabetic rats. Female Wistar rats (n = 56, 180-220 g) received a single intraperitoneal (IP) injection of STZ (65 mg/kg). They were randomly assigned into the following groups: diabetic group (Dia), Dia+Met group (100 mg metformin/kg/day), Dia+LC group (200 mg/kg/day), Dia+ZnONPs group (10 mg/kg/day), and Dia+LC+ZnONPs group (200 mg LC/kg/day and 10 mg ZnONPs/kg/day). Control group (Ctl) received the same volume of STZ solvent. After 21 days of treatment, blood serum was centrifuged for sex hormone assays. The right ovary was used for biochemical analysis, and the left ovary was fixed in 10% neutral buffered formalin for histological assessment. The levels of estradiol, progesterone, FSH, and LH significantly increased in the Dia+ZnONPs+LC group (P < 0.001) compared with the Dia group. Co-treatment with LC and ZnONPs reduced malondialdehyde and carbonyl protein and increased glutathione, catalase, and superoxide dismutase activities in ovarian tissue compared with the Dia group (P < 0.05). Moreover, the number of all ovarian follicles significantly increased in this group compared with the Dia group (P < 0.05). The results of this study indicated that co-treatment with LC and ZnONPs could preserve ovarian function by increasing sex hormones levels and antioxidant activity and decreasing lipid peroxidation in diabetic rats. Therefore, this compound supplementation may improve ovulation and fertility in people with diabetes mellitus.

Sunscreens have now been around for decades to mitigate the Sun's damaging ultraviolet (UV) radiation which, although essential for the existence of life, is a recognized prime carcinogen. Accordingly, have suncreams achieved their intended purposes towards protection against sunburns, skin photo-ageing and the like? Most importantly, however, have they provided the expected protection against skin cancers that current sunscreen products claim to do? In the last two decades, there have been tens, if not hundreds of studies on sunscreens with respect to skin protection against UVB (280‒320 nm)-traditionally sunscreens with rather low sun protection factors (SPF) were intended to protect against this type of radiation-and UVA (320‒400 nm) radiation; a distinction between SPF and UVA protection factor (UVA-PF) is made. Many of the studies of the last two decades have focused on protection against the more skin-penetrating UVA radiation. This non-exhaustive article reviews some of the important facets of what is currently known about sunscreens with regard (i) to the physical UV filters titanium dioxide (TiO 2 ) and zinc oxide (ZnO) and the mostly photo-unstable chemical UVB/UVA filters (e.g., octinoxate (OMC) and avobenzone (AVO), among others), (ii) to novel chemical sunscreen agents, (iii) to means that minimize the breakdown of chemical filters and improve their stability when exposed to UV sunlight, (iv) to SPF factors, and (v) to a short discussion on non-melanoma skin cancers and melanoma. Importantly, throughout the article we allude to the safety aspects of sunscreens and at the end ask the question: do active ingredients in sunscreen products pose a risk to human health, and what else can be done to enhance protection? Significant loss of skin protection from two well-known commercial suncreams when exposed to simulated UV sunlight. Cream I: titanium dioxide, ethylhexyl triazone, avobenzone, and octinoxate; Cream II: octyl salicylate, oxybenzone, avobenzone, and octinoxate.

In recent years, the industrial use of ZnO quantum dots (QDs) and nanoparticles (NPs) has risen and there is a high chance of these nanoparticles affecting human health. In this study, different sizes of ZnO-NPs (6-100 nm) were prepared and characterized. The generation of reactive oxygen species (ROS) and its involvement in apoptosis when HepG2 cells were exposed to QDs (6 nm) and NPs of different sizes (15-20, 50, and 100 nm) was also investigated. At a concentration of 25-200 μg/mL, NPs induced dose-dependent cytotoxicity in HepG2 cells. The engineered NPs increased oxidative stress in a dose- and size-dependent manner, as seen by an increase in ROS production, lipid peroxidation, and glutathione reduction. Furthermore, cell-cycle analysis of HepG2 cells treated with different sizes of NPs showed an increase in the apoptotic peak after a 24-h exposure period. Quantitative real-time PCR data showed that the mRNA levels of apoptotic marker genes such as p53, bax, and caspase-3 were upregulated, whereas bcl-2, an anti-apoptotic gene, was downregulated; therefore, apoptosis was mediated through the p53, bax, caspase-3, and bcl-2 pathways, suggesting a possible mechanism by which QDs and NPs of ZnO mediate their toxicity.Graphic abstract.

Photoprotection, including the use of sunscreen, has been shown to decrease the development of keratinocyte cancers and melanoma. Due to concerns about the environmental effects of some organic UVR filters, several locations across the world have begun to pass legislation banning the use of these ingredients in sunscreens. Furthermore, the health effects of several organic UVR filters have also been called into question and a recent proposal by the US FDA has resulted in public confusion about the safety of sunscreens. The aim of this article is to discuss FDA regulation of sunscreens and to review the environmental and health effects of oxybenzone and octinoxate. Ultimately, as dermatologists, our recommendations are to continue to encourage people to practice proper photoprotection including photoprotective clothing, staying in the shade while outdoors, and applying sunscreen to exposed areas. For those concerned about the potential environmental and health effects of organic UVR filters, inorganic/mineral UVR filter (namely, zinc oxide and titanium dioxide)-based sunscreens can be used.

The purpose of this study was to investigate the possibility of using zinc oxide nanoparticles (ZnONPs) instead of silver nanoparticles (AgNPs) for removing Aeromonas hydrophila from water used to culture Cyprinus carpio juvenile. Antibacterial materials as filter media were prepared by coating ZnONPs (two coating methods, referred as ZnA and ZnB) or AgNPs (referred as Ag) on the porous surfaces of zeolite beads. The characterization of coated samples was determined using FESEM, EDS, and GFAAS. The antibacterial activities of prepared samples were evaluated by the zone of inhibition test, tube test, and flow test. The diameter of inhibitory zones formed by ZnONP- and AgNP-coated zeolite beads was significantly higher than uncoated zeolite (control) (P < 0.05). Also, the tube test results revealed 100% killing of the bacterial cells after 24 h of contact to all coated materials. In the flow test (without fish), the antibacterial efficiency of filter columns that contained ZnA, ZnB, and Ag found to be 34.84, 23.77, and 100% after 96 h, respectively. The mortality rate of carp juveniles cultured in infected water treated with AgNP filters was significantly lower than those cultured in infected water or treated with ZnONPs filters (P < 0.05). The results indicated that although ZnONP filter media have somewhat antimicrobial properties (especially in vitro), their ability to complete removal of microorganisms from the water is not as high as AgNP filters. So, it still seems that zeolite coated with AgNPs has a higher potential for water disinfection in aquaculture.

There is a growing trend to implement biosecurity measures in small commercial broiler flocks and trying to replace ineffective antimicrobial with alternative materials to interevent a strategy for the control of Campylobacter bacteria in these farms. This study was designed to determine the prevalence rate of Campylobacter spp. in broiler flocks and their environment. Thereafter, assess the efficiency of chitosan, zinc oxide nanoparticles (ZnO NPs), and chitosan/ZnO NPs composite against Campylobacter strains to adopt a novel control strategy based on the ability to use those nanocomposites. A total of 220 samples were collected from broiler flocks, their environment, and farm attendants that direct contact with birds. All samples were subjected to microbiological investigation for isolation, then molecular identification of bacteria using PCR. ZnO NPs and chitosan/ZnO NPs composite were synthesized then characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), Fourier-transform infrared spectrum (FT-IR), and X-ray diffraction (X-RD). The efficiency of testing compounds was examined against 30 strains of Campylobacter coli (C. coli) to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The highest percentages of C. coli were isolated from the manure storage area, and broiler litter followed by flies, and feeders (66.7, 53.3, 40.0, and 33.3%, respectively). Both chitosan/ZnO NPs and ZnO NPs at a concentration of 0.5 μg/mL and 1.5 μg/mL, respectively showed complete efficiency (100%) against C. coli compared with chitosan compound. In conclusion, manure storage area and broiler litter represented the main reservoir of Campylobacter bacterial contaminant followed by flies in broiler poultry farms. Chitosan/ZnO NPs composite can be used in any biosecurity program of poultry farms as an alternative to ineffective antimicrobial agents.

Ingestion of nanoparticles (NPs) with antimicrobial properties may disrupt the balance of intestinal microbiota. To investigate the effects of zinc oxide (ZnO) NPs on intestinal flora, common carp Cyprinus carpio were fed a commercial feed containing 500 mg kg -1 ZnO NPs for 6 weeks and compared to a control group receiving a similar feed-only regime. Sequencing data were analyzed both in individual fish and in pooled samples. Sequencing of 16S rRNA encoding gene of individual specimens revealed high variation in intestinal microbial composition. Assessment of pooled results can obscure high individual variation in data. ZnO NPs consumption was not associated with a significant difference in the intestinal microbial community compared to untreated controls. Our results indicated a high individual variation in the intestinal microbiome, which may further point out the importance of functional study over microbial composition to address nanomaterials-microbiome relationship.

Diabetes mellitus is the most common metabolic disorder with a high mortality and morbidity rate. A new promising strategy to treat DM is pancreatic tissue engineering. We described a 3D culture system accompanied by signaling factors to differentiate hEnSCs into IPCs in the presence of nZnO. We isolated EnSCs and cultured it in DMEM/F12 medium. Nanofibrous PLA/Cs scaffold was prepared through the electrospinning method. The morphological properties of the scaffolds and cells were evaluated by SEM. MTT assay was used to investigate the metabolic activity of the hEnSCs cultured on the scaffolds and a four-stage protocol was applied to differentiate hEnSCs. The differentiated cells were tested for pancreatic markers by immunocytochemistry, qRT-PCR and DTZ staining. The results of this study revealed that hEnSCs cultured on PLA/Cs scaffold and treated with nZnO can efficiently differentiate into IPCs. The examination of differentiated cell morphology showed their near similarity with pancreatic islet cells, and DTZ staining emphasized the presence of insulin granules inside their cytoplasm. Moreover, qRT-PCR and immunofluorescent staining results showed the efficient expression of specific gene markers of IPCs in resultant differentiated cells. Moreover, PLA/CS and nZnO were able to provide a good nanoenvironment for the differentiation of hEnSCs into IPCS the in presence of other molecules.

In order to control bacterial adhesion and metal corrosion in the circulating cooling water system, it is necessary to prepare a nanocomposite-modified coating with antibacterial and anticorrosive functions. Copper and zinc composite oxide (CuZnO) was synthesized to prepare CuZnO@RGO nanocomposites. The antibacterial mechanism of CuZnO@RGO nanocomposites was investigated using gram-negative bacteria E. coli and gram-positive bacteria S. aureus as the two model microorganisms. The antibacterial properties of CuZnO@RGO nanocomposites on mixed bacteria were researched in the cooling water system. In addition, the CuZnO@RGO waterborne polyurethane (WPU) composite coating (CuZnO@RGO/WPU) was synthesized. The antibacterial performance, hardness, and corrosion inhibition performance of CuZnO@RGO/WPU composite coating in the cooling water system were also investigated. The results showed that after adding CuZnO@RGO nanocomposites to E. coli or S. aureus suspension, the protein leakage after 20 h was 9.3 times or 7.2 times higher than that in the blank experiment. The antibacterial rate of CuZnO@RGO nanocomposites in circulating cooling water reached 99.70% when the mass fraction of RGO was 15%. When the mass fraction of CuZnO@RGO accounting for CuZnO@RGO/WPU composite coating was 2%, the antibacterial rate, hardness, and corrosion inhibition efficiency were 94.35%, 5H, and 93.30%, respectively.

Aim: This was to assess the antimicrobial effectiveness of pastes used as intracanal medicaments against Enterococcus faecalis in necrotic immature teeth., Method: An agar-disc diffusion method over a period of 30 days was used. The following medications were analysed: triple antibiotic paste (TAP) I; double antibiotic paste (DAP) I; DAP I with calcium hydroxide; calcium hydroxide paste with saline solution 0.9 %; calcium hydroxide paste with chlorhexidine 2 % gel; DAP II; DAP II with zinc oxide; DAP II with calcium hydroxide; and saline solution 0.9 % as control. The diameters of the halos of inhibition (in mm) of tested medicaments were determined and analysed with one-way analysis of variance and Tukey's post hoc tests., Results: TAP I (G1) was significantly more effective against E. faecalis, and showed the largest halo of inhibition during all the experiments. DAP groups (G2 and G6) also showed inhibiting bacterial growth with said inhibition remaining stable throughout the 30-day period. In contrast, G4 and G5 groups, in which Ca(OH)2 was added to saline and chlorhexidine, had no antibacterial effect., Conclusions: TAP and DAP showed better antibacterial efficacy and remained active for 30 days. Combination of Ca(OH)2 with antibiotics should be avoided, due to the possibility of antibiotic hydrolysis.

In the current study, antidiabetic activity and toxic effects of zinc oxide nanoparticles (ZnO) were investigated in diabetic rats compared to zinc sulfate (ZnSO4) with particular emphasis on oxidative stress parameters. One hundred and twenty male Wistar rats were divided into two healthy and diabetic groups, randomly. Each major group was further subdivided into five subgroups and then orally supplemented with various doses of ZnO (1, 3, and 10 mg/kg) and ZnSO4 (30 mg/kg) for 56 consecutive days. ZnO showed greater antidiabetic activity compared to ZnSO4 evidenced by improved glucose disposal, insulin levels, and zinc status. The altered activities of erythrocyte antioxidant enzymes as well as raised levels of lipid peroxidation and a marked reduction of total antioxidant capacity were observed in rats receiving ZnO. ZnO nanoparticles acted as a potent antidiabetic agent, however, severely elicited oxidative stress particularly at higher doses.

The short-term combined effects of two most extensively used nanoparticles (NPs) TiO2 NPs (n-TiO2) and ZnO NPs (n-ZnO) versus their individual cytotoxicities on a model ammonia-oxidizing bacterium, Nitrosomonas europaea, were investigated at both physiological and transcriptional levels. n-ZnO exerted more serious impairment effects on cell morphology, cell density, membrane integrity, and ammonia monooxygenase activity than n-TiO2. However, the co-existing n-TiO2 displayed a dose-dependent mitigation effect on n-ZnO cytotoxicity. Consistently, the n-TiO2 and n-ZnO mixture-impacted global transcriptional expression profile, obtained with the whole-genome microarray technique, was more comparable to the n-TiO2-impacted one than that impacted by n-ZnO. The expressions of numerous genes associated with heavy metal scavenging, DNA repair, and oxidative stress response were less up-regulated under the binary impacts of NP mixture than n-ZnO. Moreover, only n-ZnO alone stimulated the up-regulations of heavy metal resistance genes, which further implied the capacity of co-existing n-TiO2 to alleviate n-ZnO cytotoxicity. In addition, the damage of cell membrane structures and the suppression of cell membrane biogenesis-related gene expressions under the influence of either individual NPs or their combinations strongly suggested that the interruption of cell membranes and the associated metabolic activities would probably be one of NPs' critical cytotoxicity mechanisms.

Streptococcus mitis from the oral cavity causes endocarditis and other systemic infections. Rising resistance against traditional antibiotics amongst oral bacteria further aggravates the problem. Therefore, antimicrobial and antibiofilm activities of zinc oxide and titanium dioxide nanoparticles (NPs) synthesized and characterized during this study against S. mitis ATCC 6249 and Ora-20 were evaluated in search of alternative antimicrobial agents. ZnO and TiO2-NPs exhibited an average size of 35 and 13 nm, respectively. The IC50 values of ZnO and TiO2-NPs against S. mitis ATCC 6249 were 37 and 77 µg ml(-1), respectively, while the IC50 values against S. mitis Ora-20 isolate were 31 and 53 µg ml(-1), respectively. Live and dead staining, biofilm formation on the surface of polystyrene plates, and extracellular polysaccharide production show the same pattern. Exposure to these nanoparticles also shows an increase (26-83 %) in super oxide dismutase (SOD) activity. Three genes, namely bapA1, sodA, and gtfB like genes from these bacteria were identified and sequenced for quantitative real-time PCR analysis. An increase in sodA gene (1.4- to 2.4-folds) levels and a decrease in gtfB gene (0.5- to 0.9-folds) levels in both bacteria following exposure to ZnO and TiO2-NPs were observed. Results presented in this study verify that ZnO-NPs and TiO2-NPs can control the growth and biofilm formation activities of these strains at very low concentration and hence can be used as alternative antimicrobial agents for oral hygiene.

The antibacterial activity of ZnO (0-15.53 mol%) based SiO2-CaO-P2O5-Na2O-CaF2 bioactive glass-ceramics synthesized by controlled crystallisation were studied against eight micro-organisms using modified Kirby Bauer method. The antibacterial activity of the specimens was statistically evaluated using one-way analysis of variance and P < 0.05 was used as the level of significance. In vitro dissolution tests were performed in stimulated body fluid for 48 h at 37 °C for different time intervals to correlate the dissolution behaviour of test samples with antibacterial effects. The results illustrate that specimen BZn15.53 having the highest concentration of ZnO (15.53 mol%) demonstrated the strongest effect against Staph.aureus, S. epidermidis, B. subtilis and K. pneumonia. The effectiveness of BZn15.53 in inhibiting bacteria was due to accumulation of Zn(+2) ions around the surface of the bacteria cell release that caused the death of the cell, besides the presence of hydroxyapatite phase was also responsible for damaging the cell membrane of bacteria.

Oxidative damage to cells and tissues from free radicals induced by ultraviolet (UV) irradiation can be attenuated by sunscreen components, such as ZnO and TiO2 nanoparticles (NPs). Although it is known that reactive oxygen species (ROS) are generated by cells upon exposure to ZnO and TiO2 NPs, it is unknown to what extent the amount generated is altered with UV co-exposure. As it is a critical component for determining the relative risk of these NPs when used in sunscreen formulations, we have investigated ROS generation by these NPs in human THP-1 monocyte immune cells following UVA co-exposure. Whilst the applied UVA dose (6.7 J cm(-2)) did not alter cell viability after 24 h, it induced significant ROS production - causing a 7-fold increase in intracellular peroxide and 3.3-fold increase in mitochondrial superoxide levels after 1 h. However, co-exposure to NPs and UVA generated the same or less ROS than with UVA exposure alone, with the exception of anatase TiO2, which showed significantly increased levels. These findings indicate that ROS generation from nanosunscreens is, in most cases, an insignificant contributor to the overall risk associated with oxidative stress from UVA exposure itself.

Inhaled zinc oxide nanoparticles (ZnO-NPs) can induce lung inflammation through released inflammatory mediators, such as interleukin 8 (IL-8), from airways. However, the mechanisms underlying ZnO-NP-induced IL-8 gene expression have not been fully characterized. The transcription inhibitor actinomycin D (Act D) and the BEAS-2B cells stably overexpressing wild-type or mutated IL-8 promoter at the NFκB or C/EBPβ binding site were used to determine the involvement of transcriptional mechanisms. The effect of ZnO-NPs on IL-8 mRNA stability was examined using mRNA decay assay. The phagocytosis inhibitor cytochalasin B (CB) was utilized to define the role of endocytosis in ZnO-NP-induced IL-8 expression. In addition, the solubility of ZnO-NPs in culture medium was assessed using atomic absorption spectroscopy. Exposure to ZnO-NPs significantly increased the expression of IL-8 mRNA and protein in a dose-dependent manner. Pretreatment with Act D blocked ZnO-NP-induced IL-8 expression. Both NFκB and C/EBPβ transcription factors were required for ZnO-NP-induced IL-8 transcription. mRNA decay assay showed that ZnO-NP stimulation delayed IL-8 mRNA degradation in BEAS-2B cells. Pretreatment of BEAS-2B cells with CB blocked ZnO-NP-induced IL-8 expression by 30 %. Exposure to ZnO-NPs induced IL-8 gene expression through transcriptional activation and mRNA stabilization. Internalization of nanoparticles was partially involved in ZnO-NP-induced IL-8 expression.

Objective: To evaluate the effects of zinc oxide nanoparticles on mouse spermatogenesis., Methods: Thirty two adult male NMRI mice were used. Experimental Groups (ZNP-1-ZNP-3) received one of the following treatments daily for 35 days: 5, 50 and 300 mg/kg zinc oxide nanoparticles respectively. Control group received only distilled water. Epididymal sperm parameters, testicular histopathology, morphometric analysis and spermatogenesis assessments were performed for evaluation of the zinc oxide nanoparticles effects on testis., Results: Epididymal sperm parameters including sperm number, motility and percentage of abnormality were significantly changed in 50 and 300 mg/kg zinc oxide nanoparticles treated mice (p < 0.01). Histopathological criteria such as epithelial vacuolization, sloughing of germ and detachment were significantly increased in 50 and 300 mg/kg zinc oxide nanoparticles treated mice (p < 0.001). 300 mg/kg zinc oxide nanoparticles induced formation of multinucleated giant cells in the germinal epithelium. 50 and 300 mg/kg zinc oxide nanoparticles also caused a significant decrease in seminiferous tubule diameter, seminiferous epithelium height and maturation arrest (p < 0.001)., Conclusion: Zinc oxide nanoparticles act as testicular toxicant and further studies are needed to establish its mechanism of action upon spermatogenesis.

The full-length cDNA of catalase (EfCAT) from Eisenia fetida was cloned (GenBank accession no. JN617999). Sequence characterization revealed that EfCAT protein sequence contained proximal heme-ligand signature sequence ((351)RLFSYSDTH(359)), two glycosylation sites (N(145) and N(436)), the proximal active site signature ((61)FDRERIPERVVHAKGAGA(78)), and 12 amino acids (N(145), H(191), F(195), S(198), R(200), N(210), Y(212), K(234), I(299), W(300), Q(302), and Y(355)), which were identified as putative residues involved in NADPH binding. These conserved motifs and catalase signature sequences were essential for the structure and function of EfCAT. The present study also investigated the effect of the veterinary food additive zinc oxide on antioxidant processes in E. fetida, at different concentrations and exposure durations. A significant increase (by 106.0 % compared to controls) in CAT activity at 500 mg/kg was registered at day 15. The superoxide dismutase (SOD) activity at 500 mg/kg increased to the maximum value (by 44.0 %) measured at day 15. There was a significant increase in glutathione peroxidase (GPx) activity for all concentrations after 5 days. The results showed that dietary Zn (500 mg/kg) causes oxidative damage to earthworms. At early stages of earthworms exposed to ZnO, GPx is the main enzyme to impair the oxidative status; while at later stages the enzymes CAT and SOD were the main indicators of oxidative stress. The antioxidant enzymatic variations may be an adaptive response of earthworms to survive in contaminated soils.

The wide scale use of Zinc oxide (ZnO) nanoparticles in the world consumer market makes human beings more prone to the exposure to ZnO nanoparticles and its adverse effects. The liver, which is the primary organ of metabolism, might act as a major target organ for ZnO nanoparticles after they gain entry into the body through any of the possible routes. Therefore, the aim of the present study was to assess the apoptotic and genotoxic potential of ZnO nanoparticles in human liver cells (HepG2) and the underlying molecular mechanism of its cellular toxicity. The role of dissolution in the toxicity of ZnO nanoparticles was also investigated. Our results demonstrate that HepG2 cells exposed to 14-20 μg/ml ZnO nanoparticles for 12 h showed a decrease in cell viability and the mode of cell death induced by ZnO nanoparticles was apoptosis. They also induced DNA damage which was mediated by oxidative stress as evidenced by an increase in Fpg sensitive sites. Reactive oxygen species triggered a decrease in mitochondria membrane potential and an increase in the ratio of Bax/Bcl2 leading to mitochondria mediated pathway involved in apoptosis. In addition, ZnO nanoparticles activated JNK, p38 and induced p53(Ser15) phosphorylation. However, apoptosis was found to be independent of JNK and p38 pathways. This study investigating the effects of ZnO nanoparticles in human liver cells has provided valuable insights into the mechanism of toxicity induced by ZnO nanoparticles.

Staphylococcus aureus, particularly methicillin-resistant S. aureus (MRSA), is an important cause of pyogenic skin and soft tissue infections (SSTIs). MRSA is an important pathogen in the healthcare sector that has neither been eliminated from the hospital nor community environment. In humans, S. aureus causes superficial lesions in the skin and localized abscesses, pyogenic meningitis/encephalitis, osteomyelitis, septic arthritis, invasive endocarditis, pneumonia, urinary tract infections and septicemia. Investigations focused in the search of other alternatives for the treatment of MRSA infections are in progress. Among the range of compounds whose bactericidal activity is being investigated, ZnO nanoparticles (ZnO-NPs) appears most promising new unconventional antibacterial agent that could be helpful to confront this and other drug-resistant bacteria. The aim of present study is to investigate the antibacterial potential of ZnO-NPs against Staphylococcus species isolated from the pus and wounds swab from the patients with skin and soft tissue infections in a tertiary care hospital of north India. ZnO-NPs (≈19.82 nm) synthesized by sol-gel process were characterized using scanning electron microscopy, X-ray diffraction , and Atomic force microscopy. The antibacterial potential was assessed using time-dependent growth inhibition assay, well diffusion test, MIC and MBC test and colony forming units methods. ZnO-NPs inhibited bacterial growth of methicillin-sensitive S. aureus (MSSA), MRSA and methicillin-resistant S. epidermidis (MRSE) strains and were effective bactericidal agents that were not affected by drug-resistant mechanisms of MRSA and MRSE.

ZnO nanostructures of diverse shape were grown via a solution process with different precursors and conditions. Morphological investigation of the nanostructures was carried out using field emission scanning electron microscopy and transmission microscopy observations and revealed that the nanostructures exhibit a wurtzite phase with an ideal lattice fringe distance of approximately 0.52 nm. The powder crystallinity was examined via X-ray diffraction spectroscopy. Screening results from anticancer studies of the effects on human brain tumor U87, cervical cancer HeLa, and normal HEK cells of ZnO nanostructures of diverse shape were obtained and indicate promising activity that varies with changes in the structure and the size of the particles. Treatment-induced cell death [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and survival assay], growth inhibition, cytogenetic damage (formation of micronuclei), and apoptosis were studied as parameters for the cellular response. Treatment with nanostructures enhanced growth inhibition and cell death in a concentration-dependent manner in both U87 and HeLa cell lines. At higher concentrations (above 15.6 μg/ml) the cytotoxic effects of the nanoparticles were highly synergistic and mainly mediated through apoptosis, implying the possible interactions of lesions caused by the agents. The enhanced cell death due to nanoparticles was accompanied by a significant increase (2-3 fold at 31.25 μg/ml) in the formation of micronuclei in U87 cells. The increase in the formation of micronuclei observed after treatment indicates that these structures may interfere with the rejoining of DNA strand breaks. Among all the nanostructures, nanoparticles and sheets exhibited potent activity against both HeLa and U87 cells. However, despite potent in vitro activity, all nanostructures exhibited diminished cytotoxicity against normal human HEK cells at all effective concentrations.

The specific role of size scale, surface capping, and aspect ratio of zinc oxide (ZnO) particles on toxicity toward prokaryotic and eukaryotic cells was investigated. ZnO nano and microparticles of controlled size and morphology were synthesized by wet chemical methods. Cytotoxicity toward mammalian cells was studied using a human osteoblast cancer cell line and antibacterial activity using Gram-negative bacteria (Escherichia coli) as well as using Gram-positive bacteria (Staphylococcus aureus). Scanning electron microscopy (SEM) was conducted to characterize any visual features of the biocidal action of ZnO. We observed that antibacterial activity increased with reduction in particle size. Toxicity toward the human cancer cell line was considerably higher than previously observed by other researchers on the corresponding primary cells, suggesting selective toxicity of the ZnO to cancer cells. Surface capping was also found to profoundly influence the toxicity of ZnO nanoparticles toward the cancer cell line, with the toxicity of starch-capped ZnO being the lowest. Our results are found to be consistent with a membrane-related mechanism for nanoparticle toxicity toward microbes.
