The annual Computational Neuroscience Meeting (CNS) began in 1990 as a small workshop called Analysis and Modeling of Neural Systems. The goal of the workshop was to explore the boundary between neuroscience and computation. Riding on the success of several seminal papers, physicists had made'Neural Networks'fashionable, and soon the quantitative methods used in these abstract model networks started permeating the methods and ideas of experimental neuroscientists. This volume contains the proceedings of an International Symposium on `Second Messenger Systems - Molecular, Cellular and Behavioural Aspects', which was held at Tobago on June 16-17, 1994. The interaction of an extracellular agonist (First Messenger) with its plasma membrane receptor leads to the transmission of a signal across the cell membrane and results in the production and/or activation of other signalling molecules (Second Messengers). These Second Messengers control the action of many protein kinases and protein phosphatases and so lead to cellular responses. Although the biochemical basis of the transduction of signals in the main signalling systems in eukaryotic cells is probably largely known, intensified research is ongoing in the following areas: the discovery of specific substrates for many protein kinases, elucidation of the biological significance of the differential tissue expression and heterogeneity of many signalling proteins, and the unravelling of diverse interactions (such as signal potentiation, synergism, antagonism and neuronal co-transmission) between signalling systems. As knowledge from such studies accumulates, it is becoming clear that the `cross talk'interactions between signalling systems are important features of dynamic cell regulation. Primary sensory neurons respond to peripheral stimulation and project to the spinal cord. Specifically, the population of neurons which respond to damaging stimuli terminate in the superficial layers of the dorsal horn. Therefore, the dorsal horns constitute the first relay site for nociceptive fibre terminals which make synaptic contacts with second order neurons. It has recently become clear that the strength of this first pain synapse is plastic and modifiable by several modulators, including neuronal and non-neuronal regulators, and studies on the fundamental processes regulating the plasticity of the first pain synapse have resulted in the identification of new targets for the treatment of chronic pain. The synapse is a fascinating structure for many reasons. Biologically, it is an exquisitely organized subcellular compartment that has a remarkable capacity for fidelity and endurance. Nervous systems, including our own brains, possess an extraordinary capacity for adaptation and memory because the synapse, not the neuron, constitutes the basic unit for information storage. Brain functions are realized by the activity of neuronal networks composed of a huge number of neurons. The efficiency of information transfer within the networks is changeable. Even the networks themselves can change through experience. Information transfer between neurons is performed at the synapse (the site of the neurons'contact) by release of neurotransmitters from the pre-synaptic cell and capture of neurotransmitters by the post-synaptic cell. The amount of released neurotransmitter or the efficacy of capture can change. Moreover, synapses are found to be newly formed upon activity or abandoned upon inactivity. These changes are called'synaptic plasticity'. This text focuses on one component of synaptic plasticity called transsynaptic signaling, or communication of synapses during their formation.
