Earthen construction is one of the world’s oldest and most popular construction methods, and it is still the target of prejudice due to the loss of ancestral knowledge. Due to the need for more effective and healthy building solutions, this study conducted a survey to determine the interest and knowledge of construction professionals regarding sustainable and natural materials and building techniques to understand how open these professionals are to changes in their working methods and if they identify urgency in that change. With the intent of proving the durability of earthen construction materials, laboratory research was developed which involved the preparation and performance evaluation of samples of earthen elements from the most-used techniques: rammed earth and compressed earth blocks. This evaluation was performed using the accelerated erosion test, simulating periods of rainfall and drying, and the post-test loss of resistance was also evaluated. According to the results obtained from the research survey, there is a predominant lack of knowledge about earthen construction and other traditional and sustainable materials. On the other hand, the experiments demonstrated that earthen construction can be durable when using either a small percentage of stabilizing material or a covering plaster.

This research pioneers the application of microwave irradiation as an innovative strategy for one-pot synthesis and surfactant elimination (cetyltrimethylammonium bromide—CTAB) from MCM-41, introducing a rapid and efficient methodology. MCM-41 silica is widely utilized in various applications due to its unique textural and structural properties. Nonetheless, the presence of residual surfactants after synthesis poses a challenge to its effective application. MCM-41 synthesis, conducted in a microwave reactor at 60 °C, provided a result within 0.5 to 1 h. Comprehensive analyses of structural, chemical, morphological, and surface characteristics were undertaken, with a focus on the impact of synthesis time on these properties. Surfactant extraction involved the use of ethanol as a solvent at 120 °C for 6 min within the microwave reactor. The acquired particles, coupled with the properties of textural and structural features, affirmed the efficacy of the synthesis process, resulting in the synthesis of MCM-41 within 36 min. This study presents the first instance of one-pot synthesis and surfactant removal from MCM-41 using a microwave reactor. The proposed method not only addresses the surfactant removal challenge, but also substantially accelerates the synthesis process, thereby enhancing the potential for MCM-41’s application in diverse fields.

Protein kinases function as pivotal regulators in biological events, governing essential cellular processes through the transfer of phosphate groups from ATP molecules to substrates. Dysregulation of kinase activity is frequently associated with cancer, ocasionally arising from chromosomal translocation events that relocate genes encoding kinases. Fusion proteins resulting from such events, particularly those involving the proto-oncogene tyrosine-protein kinase ROS (ROS1), manifest as constitutively active kinases, emphasizing their role in oncogenesis. Notably, the chromosomal reallocation of the ros1 gene leads to fusion of proteins with the ROS1 kinase domain, implicated in various cancer types. Despite their prevalence, targeted inhibition of these fusion proteins relies on repurposed kinase inhibitors. This review comprehensively surveys experimentally determined ROS1 structures, emphasizing the pivotal role of X-ray crystallography in providing high-quality insights. We delve into the intricate interactions between ROS1 and kinase inhibitors, shedding light on the structural basis for inhibition. Additionally, we explore point mutations identified in patients, employing molecular modeling to elucidate their structural impact on the ROS1 kinase domain. By integrating structural insights with in vitro and in silico data, this review advances our understanding of ROS1 kinase in cancer, offering potential avenues for targeted therapeutic strategies.

Background. Human and wild rodent infection rates with Leishmania (Viannia) braziliensis are needed to differentiate transmission pathways in anthropogenically altered habitats. Methods. Human participants in northeast Brazil were tested by the leishmanin skin test (LST) and inspected for lesions/scars characteristic of American clinical leishmaniasis (ACL). Molecular (PCR/qPCR) test records of free-ranging rodents were available from a concurrent capture–mark–recapture study. Force of Infection (λ) and recovery (ρ) rates were estimated from cross-sectional and longitudinal datasets. Results. Cumulative prevalences of human LST+ves and ACL scar+ves were 0.343–0.563 (n = 503 participants) and 0.122–0.475 (n = 503), respectively. Active ACL lesions were not detected. Annual rates of LST conversions were λ = 0.03–0.15 and ρ = 0.02–0.07. The probability of infection was independent of sex and associated with increasing age in addition to the period of exposure. Rodents (n = 596 individuals of 6 species) showed high rates of exclusively asymptomatic infection (λ = 0.222/month) and potential infectiousness to the sand fly vector. Spatially concurrent rodent and household human infection prevalences were correlated. Conclusions. Human exposure to L. (V.) braziliensis continues to be high despite the substantial drop in reported ACL cases in recent years. Spill-over transmission risk to humans from rodents in peridomestic habitats is likely supported by a rodent infection/transmission corridor linking houses, plantations, and the Atlantic Forest.

Edible films have been studied mainly as primary packaging materials, but they may be used as barrier layers between food components, e.g., by reducing the moisture migration between components with contrasting water activities. Since edible films are part of the food itself, components adding sensory appeal (e.g., fruit purees) are usually desirable. The objective of this study was to develop a film to be applied as a moisture barrier between nachos and guacamole. Ten film formulations were prepared according to a simplex centroid design with three components—a polysaccharide matrix (consisting of a 5:1 mixture of bacterial cellulose—BC—and carboxymethyl cellulose), tomato puree (for sensory appeal), and palm olein (to reduce hydrophilicity)—and produced by bench casting. The film with the highest palm olein content (20%) presented the lowest water vapor permeability, and its formulation was used to produce a film by continuous casting. The film was applied as a layer between nachos and guacamole, and presented to 80 panelists. The film-containing snack was preferred and considered as crispier when compared to the snack without the film, suggesting that the film was effective in reducing the moisture migration from the moist guacamole to the crispy nachos.

The incidence and prevalence of diabetes mellitus (DM) are increasing worldwide, and the resulting cardiac complications are the leading cause of death. Among these complications is diabetes-induced cardiomyopathy (DCM), which is the consequence of a pro-inflammatory condition, oxidative stress and fibrosis caused by hyperglycemia. Cardiac remodeling will lead to an imbalance in cell survival and death, which can promote cardiac dysfunction. Since the conventional treatment of DM generally does not address the prevention of cardiac remodeling, it is important to develop new alternatives for the treatment of cardiovascular complications induced by DM. Thus, therapy with mesenchymal stem cells has been shown to be a promising approach for the prevention of DCM because of their anti-apoptotic, anti-fibrotic and anti-inflammatory effects, which could improve cardiac function in patients with DM.

Pulmonary arterial hypertension (PAH) is characterized by the remodeling of pulmonary arteries, with an increased pulmonary arterial pressure and right ventricle (RV) overload. This work investigated the benefit of the association of human umbilical cord mesenchymal stem cells (hMSCs) with lodenafil, a phosphodiesterase-5 inhibitor, in an animal model of PAH. Male Wistar rats were exposed to hypoxia (10% O2) for three weeks plus a weekly i.p. injection of a vascular endothelial growth factor receptor inhibitor (SU5416, 20 mg/kg, SuHx). After confirmation of PAH, animals received intravenous injection of 5.105 hMSCs or vehicle, followed by oral treatment with lodenafil carbonate (10 mg/kg/day) for 14 days. The ratio between pulmonary artery acceleration time and RV ejection time reduced from 0.42 ± 0.01 (control) to 0.24 ± 0.01 in the SuHx group, which was not altered by lodenafil alone but was recovered to 0.31 ± 0.01 when administered in association with hMSCs. RV afterload was confirmed in the SuHx group with an increased RV systolic pressure (mmHg) of 52.1 ± 8.8 normalized to 29.6 ± 2.2 after treatment with the association. Treatment with hMSCs + lodenafil reversed RV hypertrophy, fibrosis and interstitial cell infiltration in the SuHx group. Combined therapy of lodenafil and hMSCs may be a strategy for PAH treatment.

The parallel advances of different scientific fields provide a contemporary scenario where collaboration is not a differential, but actually a requirement. In this context, crystallography has had a major contribution on the medical sciences, providing a “face” for targets of diseases that previously were known solely by name or sequence. Worldwide, cancer still leads the number of annual deaths, with 9.6 million associated deaths, with a major contribution from lung cancer and its 1.7 million deaths. Since the relationship between cancer and kinases was unraveled, these proteins have been extensively explored and became associated with drugs that later attained blockbuster status. Crystallographic structures of kinases related to lung cancer and their developed and marketed drugs provided insight on their conformation in the absence or presence of small molecules. Notwithstanding, these structures were also of service once the initially highly successful drugs started to lose their effectiveness in the emergence of mutations. This review focuses on a subclassification of lung cancer, non-small cell lung cancer (NSCLC), and major oncogenic driver mutations in kinases, and how crystallographic structures can be used, not only to provide awareness of the function and inhibition of these mutations, but also how these structures can be used in further computational studies aiming at addressing these novel mutations in the field of personalized medicine.

Coxsackievirus B (CVB) is a common human enterovirus that causes systemic infection but specifically replicates to high titers in the pancreas. It was reported that certain viruses induce mitochondrial fission to support infection. We documented that CVB triggers mitochondrial fission and blocking mitochondrial fission limits infection. The transient receptor potential channels have been implicated in regulating mitochondrial dynamics; namely, the heat and capsaicin receptor transient receptor potential cation channel subfamily V member 1 (TRPV1) contributes to mitochondrial depolarization and fission. When we transiently warmed HeLa cells to 39 °C prior to CVB exposure, infection was heightened, whereas cooling cells to 25 °C reduced infection. Inducing “cold” by stimulating transient receptor potential cation channel subfamily M member 8 (TRPM8) with menthol led to reduced infection and also resulted in lower levels of mitochondrial fission during infection. Additionally, menthol stabilized levels of mitochondrial antiviral signaling (MAVS) which is known to be tied to mitochondrial dynamics. Taken together, this highlights a novel pathway wherein CVB relies on TRPV1 to initiate proviral mitochondrial fission, which may contribute to the disruption of antiviral immunity. TRPM8 has been shown to antagonize TRPV1, and thus we hypothesize that stimulating TRPM8 blocks TRPV1-mediated mitochondrial fragmentation following CVB exposure and attenuates infection.

Herpes simplex virus infections have been described in the medical literature for centuries, yet the the drugs available nowadays for therapy are largely ineffective and low oral bioavailability plays an important role on the inefficacy of the treatments. Additionally, the details of the inhibition of Herpes Virus type 1 are still not fully understood. Studies have shown that several viruses encode one or more proteases required for the production new infectious virions. This study presents an analysis of the interactions between HSV-1 protease and benzoxazinone derivatives through a combination of structure-activity relationships, comparative modeling and molecular docking studies. The structure activity relationship results showed an important contribution of hydrophobic and polarizable groups and limitations for bulky groups in specific positions. Two Herpes Virus type 1 protease models were constructed and compared to achieve the best model which was obtained by MODELLER. Molecular docking results pointed to an important interaction between the most potent benzoxazinone derivative and Ser129, consistent with previous mechanistic data. Moreover, we also observed hydrophobic interactions that may play an important role in the stabilization of inhibitors in the active site. Finally, we performed druglikeness and drugscore studies of the most potent derivatives and the drugs currently used against Herpes virus.

AIDS is a pandemic responsible for more than 35 million deaths. The emergence of resistant mutations due to drug use is the biggest cause of treatment failure. Marine organisms are sources of different molecules, some of which offer promising HIV-1 reverse transcriptase (RT) inhibitory activity, such as the diterpenes dolabelladienotriol (THD, IC50 = 16.5 µM), (6R)-6-hydroxydichotoma-3,14-diene-1,17-dial (HDD, IC50 = 10 µM) and (6R)-6-acetoxydichotoma-3,14-diene-1,17-dial (ADD, IC50 = 35 µM), isolated from a brown algae of the genus Dictyota, showing low toxicity. In this work, we evaluated the structure-activity relationship (SAR) of THD, HDD and ADD as anti HIV-1 RT, using a molecular modeling approach. The analyses of stereoelectronic parameters revealed a direct relationship between activity and HOMO (Highest Occupied Molecular Orbital)-LUMO (Lowest Unoccupied Molecular Orbital) gap (ELUMO–EHOMO), where antiviral profile increases with larger HOMO-LUMO gap values. We also performed molecular docking studies of THD into HIV-1 RT wild-type and 12 different mutants, which showed a seahorse conformation, hydrophobic interactions and hydrogen bonds with important residues of the binding pocket. Based on in vitro experiments and docking studies, we demonstrated that mutations have little influence in positioning and interactions of THD. Following a rational drug design, we suggest a modification of THD to improve its biological activity.

Zeolites are microporous crystalline aluminosilicates with a number of useful properties including acidity, hydrothermal stability, and structural selectivity. However, the exclusive presence of micropores restricts diffusive mass transport and reduces the access of large molecules to active sites. In order to resolve this problem, mesopores can be created in the zeolite, combining the advantages of microporous and mesoporous materials. In this work, mesospores were created in the Ultrastable USY zeolite (silicon/aluminum ratio of 15) using alkaline treatment (NaOH) in the presence of cetyltrimethylammonium bromide surfactant, followed by hydrothermal treatment. The effects of the different concentrations of NaOH and the surfactant on the textural, chemical, and morphological characteristics of the modified zeolites were evaluated. Generating mesoporosity in the USY zeolite was possible through the simultaneous presence of surfactant and alkaline solution. Among the parameters studied, the concentration of the alkaline medium had the greatest influence on the textural properties of the zeolites. The presence of Cetyltrimethylammonium Bromide (CTA+) prevented the amorphization of the structure during the modification and also avoided desilication of the zeolite.

The behavior of lyotropic biomimetic systems in drug delivery was reviewed. These behaviors are influenced by drug properties, the initial water content, type of lyotropic liquid crystals (LLC), swell ability, drug loading rate, the presence of ions with higher or less kosmotropic or chaotropic force, and the electrostatic interaction between the drug and the lipid bilayers. The in vivo interaction between LCC—drugs, and the impact on the bioavailability of drugs, was reviewed. The LLC with a different architecture can be formed by the self-assembly of lipids in aqueous medium, and can be tuned by the structures and physical properties of the emulsion. These LLC lamellar phase, cubic phase, and hexagonal phase, possess fascinating viscoelastic properties, which make them useful as a dispersion technology, and a highly ordered, thermodynamically stable internal nanostructure, thereby offering the potential as a sustained drug release matrix for drug delivery. In addition, the biodegradable and biocompatible nature of lipids demonstrates a minimum toxicity and thus, they are used for various routes of administration. This review is not intended to provide a comprehensive overview, but focuses on the advantages over non modified conventional materials and LLC biomimetic properties.

Background: Diabetic kidney disease (DKD) is a leading cause of end-stage kidney disease (ESKD) worldwide. This review examines the potential differences in clinical presentation, outcomes, and management between individuals with proteinuric DKD (P-DKD) and non-proteinuric DKD (NP-DKD). Methods: We analyzed articles published globally from 2000 and 2024. Results: Individuals with NP-DKD generally have lower blood pressure levels and a more favorable lipid profile. In contrast, histological studies show that P-DKD is associated with more severe glomerulosclerosis, mesangial expansion, arteriolar hyalinosis, interstitial-fibrosis/tubular atrophy, and immune complex deposits. Additionally, those with P-DKD are more likely to develop diabetic retinopathy and have a higher risk of all-cause mortality and progression to ESKD. Strategies to slow DKD progression, applicable to both NP-DKD and P-DKD, include non-pharmacologic and pharmacologic interventions such as renin–angiotensin system blockers, sodium-glucose co-transporter-2 inhibitors, finerenone, and glucagon-like protein receptor agonists. Conclusions: NP-DKD and P-DKD represent different presentations of the same underlying disease.

Diabetes mellitus-related morbidity and mortality are primarily caused by long-term complications such as retinopathy, nephropathy, cardiomyopathy, and neuropathy. Diabetic neuropathy (DN) involves the progressive degeneration of axons and nerve fibers due to chronic exposure to hyperglycemia. This metabolic disturbance leads to excessive activation of the glycolytic pathway, inducing oxidative stress and mitochondrial dysfunction, ultimately resulting in nerve damage. There is no specific treatment for painful DN, and new approaches should aim not only to relieve pain but also to prevent oxidative stress and reduce inflammation. Given that existing therapies for painful DN are not effective for diabetic patients, mesenchymal stromal cells (MSCs)-based therapy shows promise for providing immunomodulatory and paracrine regulatory functions. MSCs from various sources can improve neuronal dysfunction associated with DN. Transplantation of MSCs has led to a reduction in hyperalgesia and allodynia, along with the recovery of nerve function in diabetic rats. While the pathogenesis of diabetic neuropathic pain is complex, clinical trials have demonstrated the importance of MSCs in modulating the immune response in diabetic patients. MSCs reduce the levels of inflammatory factors and increase anti-inflammatory cytokines, thereby interfering with the progression of DM. Further investigation is necessary to ensure the safety and efficacy of MSCs in preventing or treating neuropathic pain in diabetic patients.

This study presents the selection of 20 Extended Pseudo Invariant Calibration Sites (EPICS) for radiometric calibration and the derivation of their hyperspectral profiles using the DLR Earth Sensing Imaging Spectrometer (DESIS) and Hyperion data. The hyperspectral profile of one of these clusters, the GONA-EPICS cluster, was validated against ground truth measurements from the RadCalNet Gobabeb Namibia (GONA) site, demonstrating statistical agreement within their respective uncertainties through Welch’s test. The applicability of these hyperspectral profiles was further evaluated by generating Spectral Band Adjustment Factor (SBAF) between Landsat 8 and Sentinel-2A using the GONA-EPICS hyperspectral profile and comparing them to SBAF values derived from RadCalNet GONA site measurements. SBAF results were statistically the same, while SBAF derived from the combined DESIS and Hyperion data exhibited reduced uncertainty compared to those derived using Hyperion data alone, which is attributed to DESIS’s finer spectral resolution (2.5 nm vs. 10 nm). To assess EPICS applicability in cross-calibration, Cluster 13-GTS, which includes pixels from the Libya 4 CNES ROI, was used as a target. Cross-calibration gains obtained using EPICS and the T2T cross-calibration methodology were compared to those from the traditional cross-calibration approach using Libya 4 CNES ROI. Results demonstrated statistically similar gains, with EPICS achieving an uncertainty better than 6% across all bands compared to 4.4% for the traditional method, while enabling global coverage for daily cross-calibration opportunities. This study introduces globally distributed EPICS with validated hyperspectral profiles, offering enhanced spectral resolution and reliability for radiometric calibration and stability monitoring. The methodology supports efficient global scale sensor calibration and prepares for future hyperspectral missions.

Using fiber optics as a tool for different kinds of geotechnical monitoring can be highly attractive and cost-effective when compared to conventional instruments, such as piezometers and inclinometers, among others. A single fiber optic cable may cover a larger monitoring area compared to conventional instrumentation and allows for monitoring more than one physical quantity with the same fiber optic cable. The literature provides several different examples of distributed fiber optic systems usage. For using any sensor, a calibration curve and parameters are required. In the case of strain sensors, calibration is required to derive strain values from the frequency measurement quantity. However, fiber optic sensor cable manufacturers do not often provide cable calibration parameters, and researchers should consult the specialized literature. This article thus presents a bench adjusted for tests with single-mode fiber optic cables, as well as results of tensile tests for defining the function of strain variations in two different optical fiber cables manufactured by different companies using two different distributed interrogators. This paper also proposes a methodology for calibrating fiber optic cable deformation. A few manufacturers of fiber optic cables aim at civil engineering applications. Therefore, we propose a calibration methodology to show the possibility of obtaining calibration parameters of any fiber optic cable, even those manufactured for telecommunications purposes and not only for cables manufactured for civil engineering use. Thus, researchers will not be restricted to the acquisition of special cables for their applications. The results allowed us to conclude that the application of calibrated fiber optic sensors to experimental pile foundations permits the evaluation of the load–displacement behavior of these elements under different loading conditions.

Anthracocystis panici-leucophaei, causal agent of smut on Digitaria insularis (sourgrass), was evaluated as a biological control agent for this weed. Two types of inocula (teliospore and sporidia) were tested to assess its infectivity. The effects of teliospore and sporidia inoculations at different phenological stages of sourgrass were compared, as well as the potential of sporidia and teliospores in post-emergence sourgrass management. Virulence tests were conducted with the isolates obtained from D. insularis and evaluation of specificity of A. panici-leucophaei. Both teliospores and sporidia of A. panici-leucophaei are infective to D. insularis in three different phenological stages. Newly emerged plants with one pair of leaves are more sensitive to A. panici-leucophaei. Infection by A. panici-leucophaei inhibits the growth of sourgrass, decreasing several physiological parameters of D. insularis plants. The fungus produces systematic infection of sourgrass plants and may induce the formation of sori in a significant proportion of the plant panicles, partly castrating those plants. Among sixteen A. panici-peucophaei isolates tested, isolate 46 was the most virulent and inhibited the growth of sourgrass plants, and thus appears to have good potential as a biological control agent to be deployed against sourgrass. A. panici-leucophaei was demonstrated to be specific to D. insularis.

This study introduces a global land cover clustering using an unsupervised algorithm, incorporating the novel step of filtering data to retain only temporally stable pixels before applying K-means clustering. Unlike previous approaches that did not assess the pixel-level temporal stability, this method provides more reliable clustering results. The K-means identified 160 distinct clusters, with Cluster 13 Global Temporally Stable (Cluster 13-GTS) showing significant improvements in temporal stability. Compared to Cluster 13 Global (Cluster 13-G) from earlier research, Cluster 13-GTS reduced the coefficient of variation by up to 1% and increased the number of calibration locations from 23 to over 50. This study also validated these clusters using TOA reflectance from ground-truth measurements collected at the Radiometric Calibration Network (RadCalNet) Gobabeb (RCN-GONA) site, incorporating data from Landsat 8, Landsat 9, Sentinel-2A, and Sentinel-2B. The GONA Extended Pseudo Invariant Calibration Sites (EPICS) GONA-EPICS cluster used for the validation provided statistically comparable mean TOA reflectance to RCN-GONA, with a reduced chi-square test indicating minimal differences within the cluster’s uncertainty range. Notably, the difference in reflectance between RCN-GONA and GONA-EPICS was less than 0.023 units across all the bands. Tannins are compounds present in forage plants that, in small quantities in the diet of ruminants, produce protein complexes that promote passage through the rumen and use in the intestine. This study tested the hypothesis that beeswax (BW) and carnauba wax (CW) lipid matrices are effective encapsulants for creating bypass lysine (Lys) for ruminants, with tannin extracted from the Mimosa tenuiflora hay source enhancing material protection. Microencapsulated systems were made using the fusion–emulsification technique with a 2:1 shell-to-core ratio and four tannin levels (0%, 1%, 2%; 3%). The following eight treatments were tested: BWLys0%, BWLys1%, BWLys2%, BWLys3%, CWLys0%, CWLys1%, CWLys2%, and CWLys3%. Tannin inclusion improved microencapsulation yield and efficiency. CWLys3% had the highest microencapsulation efficiency and retained Lys. Lysine in BW and CW matrices showed higher thermal stability than in its free form. Material retention was greater in BW than CW. Rumen pH and temperature remained unaffected, indicating that BW and CW as the shell and tannin as the adjuvant are efficient encapsulants for Lys bypass production. The formulation CWLys3% is recommended as it is more efficient in protecting the lysin amino acid from rumen degradation.
