Facility agriculture, which involves agricultural production in controlled environments such as greenhouses, indoor farms, and vertical farms, aims to maximize efficiency, yield, and quality while minimizing resource consumption and environmental impact. Energy-saving technologies are essential to the green and low-carbon development of facility agriculture. Recently, phase change heat storage (PCHS) systems using phase change materials (PCMs) have gained significant attention due to their high thermal storage density and excellent thermal regulation performance. These systems are particularly promising for applications in facility agriculture and related buildings, such as solar thermal utilization, greenhouse walls, and soil insulation. However, the low thermal conductivity of PCMs presents a challenge for applications requiring rapid heat transfer. This study aims to provide a comprehensive review of the types, thermophysical properties, and various forms of PCMs, including macro-encapsulated PCMs, shape-stabilized PCMs, and phase change capsules (PCCs), as well as their preparation methods. The research methodology involves an in-depth analysis of these PCMs and their applications in active and passive PCHS systems within facility agriculture and related buildings. The major conclusion of this study highlights the critical role of PCMs in advancing energy-saving technologies in facility agriculture. By enhancing PCM performance, optimizing latent heat storage systems, and integrating intelligent environmental control, this work provides essential guidelines for designing more efficient and sustainable agricultural structures. The article will serve as the fundamental guideline to design more robust structures for facility agriculture and related buildings.

The use of low-fluence picosecond (LFPS) 1064 nm neodymium-doped yttrium aluminum garnet (Nd:YAG) lasers, referred to as laser toning, is increasingly acknowledged as an effective treatment for pigmentation disorders in the Asian skin phenotype. This study aimed to conduct a comparative analysis on the effectiveness and safety of utilizing LFPS 1064 nm Nd:YAG lasers against picosecond 532 nm Nd:YAG lasers in treating pigmented lesions among Chinese patients. A retrospective photographic analysis and chart reviews were performed on 31 subjects exhibiting Fitzpatrick skin types III–VI who underwent LFPS 1064 nm Nd:YAG or picosecond 532 nm Nd:YAG treatments at a single tertiary center. Utilizing VISIA Complexion Analysis, comparative photographs were taken. Two independent physicians evaluated treatment efficacy using a visual analog scale (VAS) to assess the percentage of pigmentary clearance in standard photographs. Solar lentigines were the most prevalent pigmentary disorder, followed by post-inflammatory hyperpigmentation (PIH), nevus zygomaticus, melasma, freckles, and nevus of Ota. The clinical effectiveness of picosecond 532 nm and LFPS 1064 nm laser treatments proved comparable for lesions on the face, with mean VAS scores of 2.2 ± 1.1 and 1.8 ± 0.8, respectively. There were two cases of PIH in the picosecond 532 nm group, which resolved within one month. Overall, the LFPS 1064 nm laser demonstrates promise as a safe and efficient therapeutic modality for managing pigmented lesions in Chinese patients.

Variational Mode Decomposition (VMD) has typically been used in weak fault feature extraction in recent years. The problem analyzed in this study is weak fault feature extraction and the enhancement of AUV thrusters based on Artificial Rabbits Optimization (ARO) and VMD. First, we introduce ARO to solve the problem of long-running times when using VMD for weak fault feature extraction. Then, we propose a VMD denoising method based on an improved ARO algorithm to address the issue of deteriorations in the fault feature extraction effect after introducing ARO. In this method, chaotic mapping and Gaussian mutation are used to improve ARO to optimize the parameters of VMD. This leads to a reduced running time and improved fault feature extraction performance. We then perform fault feature enhancement. Due to the unsatisfactory enhancement effect of traditional modified Bayes (MB) methods for weak fault features, we introduce energy operators to transform the fault signals into the energy domain for fault feature enhancement. Finally, we add differential processing to the signal to address the issue of certain fault feature values decreasing after introducing energy operators. In the end, the effectiveness of the proposed methods is verified via pool experiments on a “Beaver II” AUV prototype.

Complex hierarchical structures and diverse personnel mobility pose challenges for many multi-level organizations. The difficulty of reasonable human resource planning in multi-level organizations is mainly caused by ignoring the hierarchical structure. To address the above problems, firstly, a multi-level organization human resource network optimization model is constructed by representing the turnover situation of multi-level organizations in a dimensional manner as a multi-level network. Secondly, we propose an improved late acceptance hill climbing based on tabu and retrieval strategy (TR-LAHC) and designed two intelligent optimization operators. Finally, the TR-LAHC algorithm is compared with other classical algorithms to prove that the algorithm provides the best solution and can effectively solve the personnel mobility planning problem in multi-level organizations.

In order to deepen the understanding of the role and regulation mechanisms of prokaryotic global transcription regulators in complex processes, including virulence, the associations between the affinity and binding sequences of Mycobacterium tuberculosis MtrA have been explored extensively. Analysis of MtrA 294 diversified 26 bp binding sequences revealed that the sequence similarity of fragments was not simply associated with affinity. The unique variation patterns of GC content and periodical and sequential fluctuation of affinity contribution curves were observed along the sequence in this study. Furthermore, docking analysis demonstrated that the structure of the dimer MtrA-DNA (high affinity) was generally consistent with other OmpR family members, while Arg 219 and Gly 220 of the wing domain interacted with the minor groove. The results of the binding box replacement experiment proved that box 2 was essential for binding, which implied the differential roles of the two boxes in the binding process. Furthermore, the results of the substitution of the nucleotide at the 20th and/or 21st positions indicated that the affinity was negatively associated with the value of minor groove width precisely at the 21st position. The dimerization of the unphosphorylated MtrA facilitated by a low-affinity DNA fragment was observed for the first time. However, the proportion of the dimer was associated with the affinity of substrate DNA, which further suggested that the affinity was actually one characteristic of the stability of dimers. Based on the finding of 17 inter-molecule hydrogen bonds identified in the interface of the MtrA dimer, including 8 symmetric complementary ones in the conserved α4-β5-α5 face, we propose that hydrogen bonds should be considered just as important as salt bridges and the hydrophobic patch in the dimerization. Our comprehensive study on a large number of binding fragments with quantitative affinity values provided new insight into the molecular mechanism of dimerization, binding specificity and affinity determination of MtrA and clues for solving the puzzle of how global transcription factors regulate a large quantity of target genes.

The effects of deoxynivalenol (DON, 50 µg/mL) on the zebrafish liver and intestine were studied. Differentially expressed genes (DEGs) from mRNA and lncRNA were analyzed by RNA seq. Gene Ontology (GO) and signaling pathways were studied where the top 30 DEGs of each type of RNA were involved. The results showed there were 2325 up-regulated and 934 down-regulated DEGs of lncRNA in the intestinal tract, and 95 up-regulated genes and 211 down-regulated genes in the liver, respectively. GO functional annotation analysis showed that lncRNA was enriched in the biological processes, involving the RNA splicing, CSF1-CSF1R complexes, and MAP kinase activity. DEGs of lncRNA located in the KEGG signal pathways include the C-type lectin receptor signaling and the NOD-like receptor signaling pathways. Metabolism involves the biosynthesis of indole alkaloids, cancer pathways for human disease, MAPK and Rap1signaling pathways for environmental information processing, necroptosis and focal adhesion for cell processes. The mRNA gene expression analysis showed there were 1939 up-regulated, 1172 down-regulated genes and 866 up-regulated, 1211 down-regulated genes in the intestine and liver of zebrafish, respectively. This study provides transcriptome analysis and toxicological investigation of DON in the zebrafish liver and intestine, offering insights into gene expression patterns and potential detoxification pathways.

Breast cancer (BC) ranks in the top five malignant tumors in terms of morbidity and mortality rates. Among BC subtypes, TNBC has a high recurrence rate and metastasis rate and the worst prognosis. However, the exact mechanism by which TNBC develops is unclear. Here, we show that the deubiquitinase USP53 contributes to tumor growth and metastasis in TNBC. USP53 is overexpressed in TNBC, and this phenotype is linked to a poor prognosis. Functionally, USP53 promotes TNBC cell proliferation, migration, invasion, and epithelial–mesenchymal transition (EMT). More importantly, USP53 decreases the chemosensitivity of BC cells by enhancing v-crk sarcoma virus CT10 oncogene homologue (avian)-like (CRKL) expression. Mechanistically, USP53 directly binds CRKL to stabilize and deubiquitinate it, thereby preventing CRKL degradation. Overall, we discovered that USP53 deubiquitinates CRKL, encourages tumor development and metastasis, and reduces chemosensitivity in TNBC. These findings imply that USP53 might represent a new therapeutic target for the treatment of TNBC.

A hydraulic engine mount (HEM) is an advanced tuned mass damper (TMD) system used to isolate the noise and vibration from the engine to the chassis. This paper aims to design a semiactive HEM based on the TMD model, which only tunes damping without affecting the other lumped parameters of the TMD system. Firstly, the dynamic equations of an HEM modeled as a TMD system are derived based on the linear lumped parameter model (LPM). Each lumped parameter of the HEM is analyzed to identify the relevant parameters that affect damping. Secondly, a newly designed semiactive HEM design is proposed that utilizes a helical moving plate to simultaneously control both the inertia track area and length, resulting in precise damping tuning. To illustrate the dynamic performance of the newly designed HEM, calculations are presented based on various parameters for its tunable damping range and dynamic stiffness spectrum. Additionally, to demonstrate the performance of vibration isolation, this paper determines the optimal length and dynamic stiffness of the inertia track to minimize the transmissibility. Thirdly, to reveal the superiority of the newly designed HEM over the MR fluid mount, an example is presented where the MR fluid medium is used in place of conventional hydraulic fluid in the HEM while keeping all other parameters constant. Specifically, the novel semiactive HEM employs conventional hydraulic fluid and is spiral-driven by a moving plate while the MR fluid-based HEM is controlled by an additional tunable magnetic intensity controller. The nonlinear LPM of the newly designed HEM is verified by comparing the dynamic stiffness spectrum with the experimental results in the published literature. Then, the nonlinear LPM of the MR fluid mount is established, and its dynamic stiffness spectrum is calculated and compared with that of the newly designed HEM. The results indicate that the newly designed HEM and MR fluid mount have similar ranges of dynamic stiffness control, but the newly designed HEM does not require expensive MR/ER fluid or additional continuous external energy input to regulate the dynamic stiffness. Moreover, when using inexpensive low-viscosity hydraulic fluid, the newly designed HEM can provide a wider range of dynamic stiffness control.

In this paper, a type of vapor chamber with a gradient pore size wick (VC-G) was developed, and its thermal performance was experimentally tested and compared with two types of VCs with uniform pore size wick (which can be defined as VC-U (200) and VC-U (50) as the powder size of the wick is 50-mesh and 200-mesh, respectively) and a VC without a wick (VC-N). In addition, a VC heat transfer ability experiment platform was built, and the thermal resistance, temperature distribution and thermal response time of the VC with different wick structures were experimentally investigated. The experiment results show that the capillary driving force provided by gradient pore size wick increases gradually from outside, which can not only promote the condensation fluid to gather in the central heat source but also facilitate the vapor to spread around. Therefore, compared with VC-U (200), VC-U (50) and VC-N, VC-G shows the best heat transfer performance, temperature uniform performance and start-up performance.

The Xiejiagou gold deposit located in the Zhaoyuan-Laizhou gold belt is composed of altered-rock-type gold mineralization and superposed auriferous quartz veins, showing unique two-stage gold mineralization. Oxygen and hydrogen isotopic analyses yielded the following results: δ18OH2O = 0.8‰ to 4.4‰ and δD = −106‰ to −85‰ for altered-rock-type mineralization, and δ18OH2O = 3.6‰ to 5.6‰ and δD = −98‰ to −89‰ for auriferous quartz-veins. Combined studies on Co/Ni, Sb/Bi and As/Ag ratios of pyrites, it can be inferred that the ore-forming fluids were dominated by magmatic water mixed with very little meteoric water. The fractured altered rocks in the ore-hosting fault zones are characterized by mylonitization, cataclastic lithification, and structural lenses, reflecting a compressional (closed) ore-forming system. In contrast, the occurrence of auriferous quartz veins in fissures of altered-rock-type orebodies and the fact that altered-rock-type ores commonly occur as breccias cemented by auriferous quartz veins that reflect an extensional (open) ore-forming system for the vein mineralization. The increase in δ34S values from stage I (5.8‰–7.2‰) to stage II (6.6‰–9.0‰) indicate that the altered-rock-type mineralization was the result of intense water–rock interaction, while the occurrence of immiscible inclusions in auriferous quartz veins demonstrates that fluid immiscibility contributes significantly to gold deposition in the vein-type mineralization. Fluid-inclusion microthermometric data indicate that the fluids for the altered-rock-type mineralization are characterized by moderate-to-high temperature (262–368 °C), and low-to-moderate salinity (4.3–10.8 wt.% NaCl equivalent). In contrast, halite-bearing inclusions are found in auriferous quartz veins, and its fluids are characterized by moderate-to-high temperature (290–376 °C) and moderate-to-high salinity (5.1–41.9 wt.% NaCl equivalent). From early stages (I and II) to the late stage (III), homogenization temperature and high temperature element (W, Sn, and Mo) concentrations in pyrite first decrease and then increase, δ34S values and metallization-related element (Au, Ag, and Bi) concentrations in pyrite first increase and then decrease. Therefore, it can be inferred that the two distinct types of gold mineralization in the Xiejiagou gold deposit may be two separate mineralization events. The presence of magnetite in the auriferous veins suggests an increase in oxidation state during the vein mineralization. Importantly, the Xiejiagou gold deposit preserves two types of mineralization in a single deposit and uniquely records a metallogenic transition from a compressional, reduced environment to an extensional, oxidized environment, as a result of a regional stress field transition that occurred in the Zhaoyuan-Laizhou gold belt at ca. 120 Ma.

Herpes simplex virus type 1 (HSV-1) is human specific virus. The intercellular transmission of HSV-1 is essential in its pathogenesis. The tunneling nanotube (TNT), a new mode connecting distant cells, has been found to play an important role in the spread of various viruses like human immunodeficiency virus (HIV) and influenza virus. However, whether HSV-1 can be transmitted through TNTs has not been confirmed. The purpose of this study was to clarify this, and further to determine the effect of inhibiting the actin-related protein 2/3 (Arp2/3) complex on the intercellular transmission of HSV-1. A scanning electron microscope and fluorescence microscope detected the formation of TNTs between HSV-1 infected cells. Envelope glycoprotein D (gD) and envelope glycoprotein E (gE) of HSV-1 and viral particles were observed in TNTs. Treatment with CK666, an inhibitor of the Arp2/3 complex, reduced the number of TNTs by approximately 40–80%. At the same time, the DNA level of HSV-1 in cells and the number of plaque formation units (PFU) were also reduced by nearly 30%. These findings indicated that TNT contributes to HSV-1 transmission and that the inhibition of the Arp2/3 complex could impair HSV-1 transmission, which not only provides a novel insight into the transmission mode of HSV-1, but also a putative new antiviral target.

Collagens serve essential mechanical functions throughout the body, particularly in the connective tissues. In articular cartilage, collagens provide most of the biomechanical properties of the extracellular matrix essential for its function. Collagen plays a very important role in maintaining the mechanical properties of articular cartilage and the stability of the ECM. Noteworthily, many pathogenic factors in the course of osteoarthritis and rheumatoid arthritis, such as mechanical injury, inflammation, and senescence, are involved in the irreversible degradation of collagen, leading to the progressive destruction of cartilage. The degradation of collagen can generate new biochemical markers with the ability to monitor disease progression and facilitate drug development. In addition, collagen can also be used as a biomaterial with excellent properties such as low immunogenicity, biodegradability, biocompatibility, and hydrophilicity. This review not only provides a systematic description of collagen and analyzes the structural characteristics of articular cartilage and the mechanisms of cartilage damage in disease states but also provides a detailed characterization of the biomarkers of collagen production and the role of collagen in cartilage repair, providing ideas and techniques for clinical diagnosis and treatment.

Background: Human endothelial progenitor cells (hEPCs), originating from hemangioblasts in bone marrow (BM), migrate into the blood circulation, differentiate into endothelial cells, and could act as an alternative tool for tissue regeneration. In addition, trimethylamine-N-oxide (TMAO), one of the gut microbiota metabolites, has been identified as an atherosclerosis risk factor. However, the deleterious effects of TMAO on the neovascularization of hEPCs have not been studied yet. Results: Our results demonstrated that TMAO dose-dependently impaired human stem cell factor (SCF)-mediated neovascularization in hEPCs. The action of TMAO was through the inactivation of Akt/endothelial nitric oxide synthase (eNOS), MAPK/ERK signaling pathways, and an upregulation of microRNA (miR)-221. Docosahexaenoic acid (DHA) could effectively inhibit the cellular miR-221 level and induce the phosphorylation level of Akt/eNOS, MAPK/ERK signaling molecules, and neovascularization in hEPCs. DHA enhanced cellular amounts of reduced form glutathione (GSH) through an increased expression of the gamma-glutamylcysteine synthetase (γ-GCS) protein. Conclusions: TMAO could significantly inhibit SCF-mediated neovascularization, in part in association with an upregulation of miR-221 level, inactivation of Akt/eNOS and MAPK/ERK cascades, suppression of γ-GCS protein, and decreased levels of GSH and GSH/GSSG ratio. Furthermore, the DHA could alleviate the detrimental effects of TMAO and induce neovasculogenesis through suppression of miR-221 level, activation of Akt/eNOS and MAPK/ERK signaling cascades, increased expression of γ-GCS protein, and increment of cellular GSH level and GSH/GSSG ratio in hEPCs.

Vertical displacement acceleration and the pitch angle record produce the phenomenon of beat vibration when testing a 200 m electro-dynamic suspension (EDS) magnetic levitation (maglev) test vehicle with high-temperature superconducting (HTS) at the CRRC Changchun Railway Vehicles Co., Ltd., where the vehicle is clamped and in planar motion. First, to examine this phenomenon, this paper establishes dynamic equations of the vehicle with three degrees of freedom (DOF), and the levitation force on each superconducting magnet (SCM) is calculated by dynamic circuit theory. Second, the theory vertical equilibrium point is obtained from the average of the levitation force for a different velocity and the magneto-motive force (MMF) of the SCM. Third, this paper decouples SCM levitation forces from each other using MATLAB/SIMULINK, and a multi-body dynamic model with six DOF is developed in SIMPACK. All vertical displacements and acceleration responses, as well as the pitch angle and acceleration response from the simulation, appear to show the phenomenon of beat vibration since there are two closing natural frequencies of approximately 2 Hz and 2.4 Hz. Finally, based on the traversing method considering the influence of the velocity, initial vertical displacement, and the MMF of the SCM, the multi-body dynamic model is frequently utilized to study the response of the mean and amplitude of vertical displacement and that of the pitch angle. The results show that increasing the MMF or velocity could decrease the vertical displacement and pitch angle; the mean vertical displacement is a little larger than the theory equilibrium point; and the amplitude of vertical displacement is small when the initial vertical displacement is near the theory equilibrium point. Both the numerical and experimental results verify the validity of the dynamic circuit model and mechanical model in this paper.

Breast cancer (BC) is the world’s second most frequent malignancy and the leading cause of mortality among women. All in situ or invasive breast cancer derives from terminal tubulobular units; when the tumor is present only in the ducts or lobules in situ, it is called ductal carcinoma in situ (DCIS)/lobular carcinoma in situ (LCIS). The biggest risk factors are age, mutations in breast cancer genes 1 or 2 (BRCA1 or BRCA2), and dense breast tissue. Current treatments are associated with various side effects, recurrence, and poor quality of life. The critical role of the immune system in breast cancer progression/regression should always be considered. Several immunotherapy techniques for BC have been studied, including tumor-targeted antibodies (bispecific antibodies), adoptive T cell therapy, vaccinations, and immune checkpoint inhibition with anti-PD-1 antibodies. In the last decade, significant breakthroughs have been made in breast cancer immunotherapy. This advancement was principally prompted by cancer cells’ escape of immune regulation and the tumor’s subsequent resistance to traditional therapy. Photodynamic therapy (PDT) has shown potential as a cancer treatment. It is less intrusive, more focused, and less damaging to normal cells and tissues. It entails the employment of a photosensitizer (PS) and a specific wavelength of light to create reactive oxygen species. Recently, an increasing number of studies have shown that PDT combined with immunotherapy improves the effect of tumor drugs and reduces tumor immune escape, improving the prognosis of breast cancer patients. Therefore, we objectively evaluate strategies for their limitations and benefits, which are critical to improving outcomes for breast cancer patients. In conclusion, we offer many avenues for further study on tailored immunotherapy, such as oxygen-enhanced PDT and nanoparticles.

Numerical and analytical investigations were performed to study the panel flutter generated by the coupling of elastic and aerodynamic loads with thermal loads. Based on large deflection theory and piston aerodynamic theory, the nonlinear dynamic differential equations of heated panels with pre-stretch displacement are derived. The Galerkin method is applied to transform the continuous partial differential equations into a nonlinear system of ordinary differential equations. The analytical expressions of the flutter critical dynamic pressure and flutter frequency, the static divergence stability boundary and the Hopf bifurcation fluttering stability boundary for the initial equilibrium of the panel can be obtained through the algebraic criterion of the Hopf bifurcation. The results show that, compared to the non-pre-stretch condition, when the pre-strain of the panel was merely 0.0328%, the flutter critical dynamic pressure and flutter frequency increased by 380.78% and 223.43%, respectively. Moreover, the pre-stretching method can significantly enhance the capacity of the supersonic panel to sustain temperature loads.

Deoxynivalenol, a mycotoxin that may present in almost all cereal products, can cause huge economic losses in the agriculture industry and seriously endanger food safety and human health. Microbial detoxifications using microbial consortia may provide a safe and effective strategy for DON mitigation. In order to study the interactions involving DON degradation and change in microbial flora, four samples from different natural niches, including a chicken stable (expJ), a sheep stable (expY), a wheat field (expT) and a horse stable (expM) were collected and reacted with purified DON. After being co-incubated at 30 °C with 130 rpm shaking for 96 h, DON was reduced by 74.5%, 43.0%, 46.7%, and 86.0% by expJ, expY, expT, and expM, respectively. After DON (0.8 mL of 100 μg/mL) was co-cultivated with 0.2 mL of the supernatant of each sample (i.e., suspensions of microbial communities) at 30 °C for 96 h, DON was reduced by 98.9%, 99.8%, 79.5%, and 78.9% in expJ, expY, expT, and expM, respectively, and was completely degraded after 8 days by all samples except of expM. DON was confirmed being transformed into de-epoxy DON (DOM-1) by the microbial community of expM. The bacterial flora of the samples was compared through 16S rDNA flux sequencing pre- and post the addition of DON. The results indicated that the diversities of bacterial flora were affected by DON. After DON treatment, the most abundant bacteria belong to Galbibacter (16.1%) and Pedobacter (8.2%) in expJ; Flavobacterium (5.9%) and Pedobacter (5.5%) in expY; f_Microscillaceae (13.5%), B1-7BS (13.4%), and RB41 (10.5%) in expT; and Acinetobacter (24.1%), Massilia (8.8%), and Arthrobacter (7.6%) in expM. This first study on the interactions between DON and natural microbial flora provides useful information and a methodology for further development of microbial consortia for mycotoxin detoxifications.

The transportation and surface deposition of aerosols from sneezing in a small indoor farmers’ market are studied numerically. The effects of numbers and locations of the entrances and exits of the market are discussed under the condition of natural convection. The results indicate that aerosols leave the indoor environment more quickly when two doors are designed face to face on the walls perpendicular to the natural wind direction compared to other natural ventilation strategies. The concentrations of aerosols accumulated on the surfaces of the stalls and human bodies inside the market are also lower. In this case, the risk of contacting the virus is relatively low among susceptible individuals in the indoor farmers’ markets. Moreover, opening more doors on the walls parallel to the natural wind direction is not beneficial for the fast exhaust of aerosols.

Given the popularity of ketogenic diets, their potential long-term consequences deserve to be more carefully monitored. Mitochondrially derived formate has a critical role in mammalian one-carbon (1C) metabolism and development. The glycine cleavage system (GCS) accounts for another substantial source for mitochondrially derived 1C units. Objective: We investigated how the ketogenic state modulates mitochondrial formate generation and partitioning of 1C metabolic fluxes. Design: HepG2 cells treated with physiological doses (1 mM and 10 mM) of β-hydroxybutyrate (βHB) were utilized as the in vitro ketogenic model. Eight-week male C57BL/6JNarl mice received either a medium-chain fatty-acid-enriched ketogenic diet (MCT-KD) or a control diet AIN 93M for 8 weeks. Stable isotopic labeling experiments were conducted. Results and Conclusions: MCT-KD is effective in weight and fat loss. Deoxythymidine (dTMP) synthesis from the mitochondrial GCS-derived formate was significantly suppressed by βHB and consumption of MCT-KD. Consistently, plasma formate concentrations, as well as the metabolic fluxes from serine and glycine, were suppressed by MCT-KD. MCT-KD also decreased the fractional contribution of mitochondrially derived formate in methionine synthesis from serine. With the worldwide application, people and medical professionals should be more aware of the potential metabolic perturbations when practicing a long-term ketogenic diet.

Piezoelectric waveguide transducers possess great potential for the online monitoring of high temperature critical components, in order to improve their operational safety. Due to the use of a waveguide bar, the sensory device is not susceptible to high temperature environments, which enables the long-term service of the piezoelectric transducers. However, the coupling between the waveguide bar and the high-temperature component has been proven to be the most important part of the monitoring system. In order to effectively transmit waves through the junction of the waveguide bar and the monitoring target, it is necessary to research a reliable coupling method to connect the waveguide transducers with the host structure. In the present research, the feasibility of brazing coupling for wave propagation through the junction was investigated through experiments. Piezoelectric waveguide transducers were welded using various kinds of brazing filler metals. The experimental results indicate that the coupling effects of the brazing welding depend on the filler metals. At the same time, some filler metals for the effective coupling of the transducer and the target monitoring component were identified. The brazing coupling method was verified that it can non-dispersively and effectively propagate waves into the host structure with much better reliability than the conventional dry coupling approach. Moreover, the high-temperature experimental results show that the brazing-coupled waveguide bar system can work reliably and stably in high temperatures at 300 °C for a long time. This work strives to pave a solid foundation for the application of piezoelectric waveguide transducers for the structural health monitoring of high temperature critical components.

The DNA-dependent protein kinase catalytic subunit (DNA-PKcs) is a key component of the DNA-PK complex that has a well-characterized function in the non-homologous end-joining repair of DNA double-strand breaks. Since its identification, a large body of evidence has demonstrated that DNA-PKcs is frequently overexpressed in cancer, plays a critical role in tumor development and progression, and is associated with poor prognosis of cancer patients. Intriguingly, recent studies have suggested novel functions beyond the canonical role of DNA-PKcs, which has transformed the paradigm of DNA-PKcs in tumorigenesis and has reinvigorated the interest to target DNA-PKcs for cancer treatment. In this review, we update recent advances in DNA-PKcs, in particular the emerging roles in tumor metastasis, metabolic dysregulation, and immune escape. We further discuss the possible molecular basis that underpins the pleiotropism of DNA-PKcs in cancer. Finally, we outline the biomarkers that may predict the therapeutic response to DNA-PKcs inhibitor therapy. Understanding the functional repertoire of DNA-PKcs will provide mechanistic insights of DNA-PKcs in malignancy and, more importantly, may revolutionize the design and utility of DNA-PKcs-based precision cancer therapy.

(1) Background: Ferroptosis is an apoptosis-independent cell death program implicated in many diseases including cancer. Emerging evidence suggests ferroptosis as a promising avenue for cancer therapy, but the paucity of mechanistic understanding of ferroptosis regulation and lack of biomarkers for sensitivity to ferroptosis inducers have significantly hampered the utility of ferroptosis-based therapy. (2) Methods: We performed integrated dataset analysis by correlating the sensitivity of small-molecule compounds (n = 481) against the transcriptomes of solid cancer cell lines (n = 659) to identify drug candidates with the potential to induce ferroptosis. Generalizable gene signatures of ferroptosis sensitivity and resistance are defined by interrogating drug effects of ferroptosis inducers (n = 7) with transcriptomic data of pan-solid cancer cells. (3) Results: We report, for the first time, the comprehensive identification of drug compounds that induce ferroptosis and the delineation of generalizable gene signatures of pro- and anti-ferroptosis in pan-cancer. We further reveal that small cell lung cancer (SCLC) and isocitrate dehydrogenase (IDH1/2)-mutant brain tumors show enrichment of pro-ferroptosis gene signature, suggesting a unique vulnerability of SCLC and IDH-mutant tumors to ferroptosis inducers. Finally, we demonstrate that targeting class I histone deacetylase (HDAC) significantly enhances ferroptotic cell death caused by Erastin, an ferroptosis inducer, in lung cancer cells, revealing a previously underappreciated role for HDAC in ferroptosis regulation. (4) Conclusions: Our work reveals novel drug compounds and gene networks that regulate ferroptosis in cancer, which sheds light on the mechanisms of ferroptosis and may facilitate biomarker-guided stratification for ferroptosis-based therapy.

Folate-mediated one-carbon (1C) metabolism is a major target of many therapies in human diseases. Studies have focused on the metabolism of serine 3-carbon as it serves as a major source for 1C units. The serine 3-carbon enters the mitochondria transferred by folate cofactors and eventually converted to formate and serves as a major building block for cytosolic 1C metabolism. Abnormal glycine metabolism has been reported in many human pathological conditions. The mitochondrial glycine cleavage system (GCS) catalyzes glycine degradation to CO2 and ammonium, while tetrahydrofolate (THF) is converted into 5,10-methylene-THF. GCS accounts for a substantial proportion of whole-body glycine flux in humans, yet the particular metabolic route of glycine 2-carbon recycled from GCS during mitochondria glycine decarboxylation in hepatic or bone marrow 1C metabolism is not fully investigated, due to the limited accessibility of human tissues. Labeled glycine at 2-carbon was given to humans and primary cells in previous studies for investigating its incorporations into purines, its interconversion with serine, or the CO2 production in the mitochondria. Less is known on the metabolic fate of the glycine 2-carbon recycled from the GCS; hence, a model system tracing its metabolic fate would help in this regard. We took the direct approach of isotopic labeling to further explore the in vitro and in vivo metabolic fate of the 2-carbon from [2-13C]glycine and [2-13C]serine. As the 2-carbon of glycine and serine is decarboxylated and catabolized via the GCS, the original 13C-labeled 2-carbon is transferred to THF and yield methyleneTHF in the mitochondria. In human hepatoma cell-lines, 2-carbon from glycine was found to be incorporated into deoxythymidine (dTMP, dT + 1), M + 3 species of purines (deoxyadenine, dA and deoxyguanine, dG), and methionine (Met + 1). In healthy mice, incorporation of GCS-derived formate from glycine 2-carbon was found in serine (Ser + 2 via cytosolic serine hydroxy methyl transferase), methionine, dTMP, and methylcytosine (mC + 1) in bone marrow DNA. In these experiments, labeled glycine 2-carbon directly incorporates into Ser + 1, A + 2, and G + 2 (at C2 and C8 of purine) in the cytosol. It is noteworthy that since the serine 3-carbon is unlabeled in these experiments, the isotopic enrichments in dT + 1, Ser + 2, dA + 3, dG + 3, and Met + 1 solely come from the 2-carbon of glycine/serine recycled from GCS, re-enters the cytosolic 1C metabolism as formate, and then being used for cytosolic syntheses of serine, dTMP, purine (M + 3) and methionine. Taken together, we established model systems and successfully traced the metabolic fate of mitochondrial GCS-derived formate from glycine 2-carbon in vitro and in vivo. Nutritional supply significantly alters formate generation from GCS. More GCS-derived formate was used in hepatic serine and methionine syntheses, whereas more GCS-derived formate was used in dTMP synthesis in the bone marrow, indicating that the utilization and partitioning of GCS-derived 1C unit are tissue-specific. These approaches enable better understanding concerning the utilization of 1C moiety generated from mitochondrial GCS that can help to further elucidate the role of GCS in human disease development and progression in future applications. More studies on GCS using these approaches are underway.

Background: Malignant pleural mesothelioma (MPM) is driven by the inactivation of tumor suppressor genes (TSGs). An unmet need in the field is the translation of the genomic landscape into effective TSG-specific therapies. Methods: We correlated genomes against transcriptomes of patients’ MPM tumors, by weighted gene co-expression network analysis (WGCNA). The identified aberrant biochemical networks and potential drug targets induced by tumor suppressor loss were validated by integrative data analysis and functional interrogation. Results: CDKN2A/2B loss activates G2/M checkpoint and PI3K/AKT, prioritizing a co-targeting strategy for CDKN2A/2B-null MPM. CDKN2A deficiency significantly co-occurs with deletions of anti-viral type I interferon (IFN-I) genes and BAP1 mutations, that enriches the IFN-I signature, stratifying a unique subset, with deficient IFN-I, but proficient BAP1 for oncolytic viral immunotherapies. Aberrant p53 attenuates differentiation and SETD2 loss acquires the dependency on EGFRs, highlighting the potential of differentiation therapy and pan-EGFR inhibitors for these subpopulations, respectively. LATS2 deficiency is linked with dysregulated immunoregulation, suggesting a rationale for immune checkpoint blockade. Finally, multiple lines of evidence support Dasatinib as a promising therapeutic for LATS2-mutant MPM. Conclusions: Systematic identification of abnormal cellular processes and potential drug vulnerabilities specified by TSG alterations provide a framework for precision oncology in MPM.

The development of novel, noble metal-free semiconductor catalysts with high efficiency is of great importance for the degradation of organic compounds. Among them, amorphous materials have been extensively studied for their unique and commercially useful properties. Here, a completely amorphous, noble metal-free photocatalyst C3N4ZnxOy (x, y = 0.32–1.10) was successfully synthesized from urea and ZnO by a simple high-temperature polymerization method. As the Zn content increased, the short-range ordered structures of the amorphous samples were still retained, as revealed by XPS, FTIR, and ssNMR. Meanwhile, the -CN3 structures were observed to be gradually destroyed, which may make the amorphous state more favorable for photocatalytic reactions. Compared with g-C3N4, the amorphous samples showed significantly reduced intensities in the photoluminescence spectra, indicating that the recombination rate of the photo-generated charge carriers was greatly reduced. It was confirmed that the optimized sample (C3N4Zn0.61O0.61) achieved a photocatalytic efficiency of 86.1% in the degradation of tetracycline hydrochloride under visible light irradiation within 1 h. This is about 2 times higher than that of both g-C3N4 and ZnO. This study emphasizes the importance of the amorphous structure in photocatalytic reactions, and this synthetic strategy may provide an effective model for designing other novel catalysts.

Uncertainty caused by a parameter measurement error or a model error causes difficulties for the implementation of the control method. Experts can divide the uncertain system into a definite part and an uncertain part and solve each part using various methods. Two uncertainty problems of the control system arise: problem A for the definite part—how does one find out the optimal number and position of actuators when the actuating force of an actuator is smaller than the control force? Problem B for the uncertain part—how does one evaluate the effect of uncertainty on the eigenvalues of a closed-loop control system? This paper utilizes an interval to express the uncertain parameters and converts the control system into a definite part and an uncertain part using interval theory. The interval state matrix is constructed by physical parameters of the system for the definite part of the control system. For Problem A, the paper finds out the singular value element sensitivity of the modal control matrix and reorders the optimal location of the actuators. Then, the paper calculates the state feedback gain matrix for a single actuator using the receptance method of pole assignment and optimizes the number and position of the actuators using the recursive design method. For Problem B, which concerns the robustness of closed-loop systems, the paper obtains the effects of uncertain parameters on the real and imaginary parts of the eigenvalues of a closed-loop system using the matrix perturbation theory and interval expansion theory. Finally, a numerical example illustrates the recursive design method to optimize the number and location of actuators and it also shows that the change rate of eigenvalues increases with the increase in uncertainty.

The joint control problem of the underwater manipulator is addressed in this paper, under the influence of uncertainty factors such as model uncertainty, external disturbance, and manipulator joint lag. In general, for the uncertainty factors, it is usually approximated online, but it is difficult to select a reasonable value for the approximation error boundary, too conservative estimated values would cause chattering problem easily. And the influence of joint lag on the manipulator control should be considered in actual work. Unlike most previous control method, in this paper, the function approximation technique (FAT), which uses the Legendre polynomial, is adopted to approximate the uncertainty factors online. Then, based on the proportion integral differential (PID) sliding manifold with the integral and differential of tracking error, a sliding model PID controller is designed to speed up the response and reduce the effects of joint lag. For the error boundary, the adaptive law is proposed, and it will reduce chattering of the control quantity under the steady state of the system. It was proved that the joint error of the control system is uniformly asymptotic convergence through the stability analysis. Finally, the effectiveness of the proposed approach is demonstrated with pool comparison experiments of the underwater manipulator installed in the autonomous underwater vehicles (AUVs).

Polyethylene oxide-based solid polymer electrolytes (SPEs) are of research interest because of their potential applications in all-solid-state Li+ batteries. However, despite their advantages in terms of compatibility with the electrodes and easy processing, polyethylene oxide (PEO)/Li+ complexes often suffer from low conductivity at room temperature. Understanding the conduction mechanism and, in turn, developing strategies to improve the conductivity have long been the main objectives underlying research into PEO/Li+ complex electrolytes. Here, we prepared several special PEO/Li+ complex samples where the PEO/Li+ complex structures were located on the surfaces of PEO crystals and consisted of high content chain ends. We found two different Li+ species in the PEO/Li+ complex structures via solid-state nuclear magnetic resonance (NMR). The 2D 7Li exchange NMR showed the exchange process between the different Li+ species. The exchange dynamics of the Li+ ions provide a molecular mechanism of the Li+ transportation in the surface of PEO crystal lamella, which is further correlated with the ionic conduction mechanism of the PEO/Li+ complex structure.

The dried roots of Euphorbia kansui S.L.Liou ex S.B.Ho have long been used to treat edema in China. However, the severe toxicity caused by Euphorbia kansui (EK) has seriously restricted its clinical application. Although EK was processed with vinegar to reduce its toxicity, the detailed mechanisms of attenuation in toxicity of EK stir-fried with vinegar (VEK) have not been well delineated. Diterpenoids are the main toxic ingredients of EK, and changes in these after processing may be the underlying mechanism of toxicity attenuation of VEK. 3-O-(2′E,4′Z-decadienoyl)-20-O-acetylingenol (3-O-EZ) is one of the diterpenoids derived from EK, and the content of 3-O-EZ was significantly reduced after processing. This study aims to explore the underlying mechanisms of toxicity reduction of VEK based on the change of 3-O-EZ after processing with vinegar. Based on the chemical structure of 3-O-EZ and the method of processing with vinegar, simulation experiments were carried out to confirm the presence of the product both in EK and VEK and to enrich the product. Then, the difference of peak area of 3-O-EZ and its hydrolysate in EK and VEK were detected by ultra-high-performance liquid chromatography (UPLC). Furthermore, the toxicity effect of 3-O-EZ and its hydrolysate, as well as the underlying mechanism, on zebrafish embryos were investigated. The findings showed that the diterpenoids (3-O-EZ) in EK can convert into less toxic ingenol in VEK after processing with vinegar; meanwhile, the content of ingenol in VEK was higher than that of EK. More interestingly, the ingenol exhibited less toxicity (acute toxicity, developmental toxicity and organic toxicity) than that of 3-O-EZ, and 3-O-EZ could increase malondialdehyde (MDA) content and reduce glutathione (GSH) content; cause embryo oxidative damage by inhibition of the succinate dehydrogenase (SDH) and superoxide dismutase (SOD) activity; and induce inflammation and apoptosis by elevation of IL-2 and IL-8 contents and activation of the caspase-3 and caspase-9 activity. Thus, this study contributes to our understanding of the mechanism of attenuation in toxicity of VEK, and provides the possibility of safe and rational use of EK in clinics.

Background: The cell cycle checkpoint protein RAD9 is a vital cell cycle regulator in eukaryotic cells. RAD9 is involved in diverse cellular functions by oligomer or monomer. However, the specific mechanism of its activity remains unknown in crustaceans, especially in embryonic diapause resumption of the brine shrimp Artemia sinica. Methods and Results: In the present article, a 1238 bp full-length cDNA of As−RAD9 gene, encoding 376 amino acids, was obtained from A. sinica. The expression pattern of As−RAD9 was analyzed by qPCR and Western blot. The mRNA expression level climbs to the top at the 10 h stage of embryo development, while the protein expression pattern is generally consistent with qPCR results. Moreover, the As−RADd9 related signaling proteins, As−RAD1, As−HUS1, As−RAD17, and As−CHK1, were also detected. Immunofluorescence assay showed that the location of As−RAD9 did not show tissue or organ specificity, and the intracellular expression was concentrated in the cytoplasm more than in the nucleus. We also explored the amount of As−RAD9 under the stresses of cold and high salinity, and the results indicate that As−RAD9 is a stress-related factor, though the mechanisms may be different in response to different stresses. Knocking down of the As−RAD9 gene led to embryonic development delay in A. sinica. Conclusions: All these results reveal that As−RAD9 is necessary for post-diapaused embryonic development in A. sinica.

Thermal Fatigue of flip chip component solder joints is widely existing in thermal energy systems, which imposes a great challenge to operational safety. In order to investigate the influential factors, this paper develops a model to analyze thermal fatigue, based on the Darveaux energy method. Under cyclic thermal loading, a theoretical heat transfer and thermal stress model is developed for the flip chip components and the thermal fatigue lives of flip chip component solder joints are analyzed. The model based simulation results show the effects of environmental and power parameters on thermal fatigue life. It is indicated that under cyclic thermal loading, the solder joint with the shortest life in a package of flip chip components is located at the outer corner point of the array. Increment in either power density or ambient temperature or the decrease in either power conversion time or ambient pressure will result in short thermal fatigue lives of the key solder joints in the flip chip components. In addition, thermal fatigue life is more sensitive to power density and ambient temperature than to power conversion time and ambient air pressure.

For long-term missions in complex seas, the onboard energy resources of autonomous underwater vehicles (AUVs) are limited. Thus, energy consumption reduction is an important aspect of the study of AUVs. This paper addresses energy consumption reduction using model predictive control (MPC) based on the state space model of AUVs for trajectory tracking control. Unlike the previous approaches, which use a cost function that consists of quadratic deviations of the predicted controlled output from the reference trajectory and quadratic input changes, a term of quadratic energy (i.e., quadratic input) is introduced into the cost function in this paper. Then, the MPC control law with the new cost function is constructed, and an analysis on the effect of the quadratic energy term on the stability is given. Finally, simulation results for depth tracking control are given to demonstrate the feasibility and effectiveness of the improved MPC on energy consumption optimization for AUVs.

The metabolic and pharmacokinetic studies on complanatuside, a quality marker of a Chinese materia medicatonic, Semen Astragali Complanati, were carried out. The UHPLC-Q-TOF/MS (ultra-high performance liquid chromatography coupled with electrospray ionization tandem quadrupole-time-of-flight mass spectrometry) method was applied to identify the metabolites of complanatuside in rat plasma, bile, stool, and urine after oral administration at the dosage of 72 mg/kg. Up to 34 metabolites (parent, 2 metabolites of the parent drug, and 31 metabolites of the degradation products) were observed, including processes of demethylation, hydroxylation, glucuronidation, sulfonation, and dehydration. The results indicated glucuronidation and sulfonation as major metabolic pathways of complanatuside in vivo. Meanwhile, a HPLC-MS method to quantify complanatuside and its two major metabolites—rhamnocitrin 3-O-β-glc and rhamnocitrin—in rat plasma for the pharmacokinetic analysis was developed and validated. The Tmax (time to reach the maximum drug concentration) of the above three compounds were 1 h, 3 h, and 5.3 h, respectively, while the Cmax (maximum plasma concentrations)were 119.15 ng/mL, 111.64 ng/mL, and 1122.18 ng/mL, and AUC(0-t) (area under the plasma concentration-time curve) was 143.52 µg/L·h, 381.73 µg/L·h, and 6540.14 µg/L·h, accordingly. The pharmacokinetic characteristics of complanatuside and its two metabolites suggested that complanatuside rapidly metabolized in vivo, while its metabolites—rhamnocitrin—was the main existent form in rat plasma after oral administration. The results of intracorporal processes, existing forms, and pharmacokinetic characteristics of complanatuside in rats supported its low bioavailability.

A vapor chamber can meet the cooling requirements of high heat flux electronic equipment. In this paper, based on a proposed vapor chamber with a side window, a vapor chamber experimental system was designed to visually study its evaporation and condensation heat transfer performance. Using infrared thermal imaging technology, the temperature distribution and the vapor⁻liquid two-phase interface evolution inside the cavity were experimentally observed. Furthermore, the evaporation and condensation heat transfer coefficients were obtained according to the measured temperature of the liquid near the evaporator surface and the vapor near the condenser surface. The effects of heat load and filling rate on the thermal resistance and the evaporation and condensation heat transfer coefficients are analyzed and discussed. The results indicate that the liquid filling rate that maximized the evaporation heat transfer coefficient was different from the liquid filling rate that maximized the condensation heat transfer coefficient. The vapor chamber showed good heat transfer performance with a liquid filling rate of 33%. According to the infrared thermal images, it was observed that the evaporation/boiling heat transfer could be strengthened by the interference of easily broken bubbles and boiling liquid. When the heat input increased, the uniformity of temperature distribution was improved due to the intensified heat transfer on the evaporator surface.

Quality control of Chinese herbal tea remains a challenge due to our poor knowledge of their complex chemical profile. This study aims to investigate the chemical composition of one of the best-selling and famous brand of beverage in China, Wanglaoji Herbal Tea (WLJHT), via a full component quantitative analysis. In this paper, a total of thirty-four representative constituents were identified or tentatively characterized using ultra-high performance liquid chromatography coupled with quadrupole tandem time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Moreover, the quantitative analyses of fourteen constituents were performed by high performance liquid chromatography with a triple quadruple tandem mass spectrometry (HPLC-MS/MS) method and saccharide compositions of WLJHT were also quantitatively determined by high performance liquid chromatography (HPLC) with evaporative light scattering detector (ELSD) on a Hilic column, separately. Using multiple chromatographic techniques presented a good precision, sensitivity, repeatability and stability, and was successfully applied to analyze 16 batches of WLJHT samples. Therefore, it would be a reliable and useful approach for the quality control of WLJHT.

The coupled effect of boiling and condensation inside a flat two-phase thermosyphon has a non-negligible influence on the two-phase fluid flow behavior and heat transfer process. Therefore, a flat two-phase thermosyphon with transparent wall was manufactured. Based on this device, a visualization experiment system was developed to study the vapor–liquid two-phase behaviors and thermal performance of the flat two-phase thermosyphon. A cross-shaped wick using copper mesh was embedded into the cavity of two-phase thermosyphon to improve the heat transfer performance. The effects of heat flux density, working medium, and wick structure on the thermal performance are examined and analyzed. The results indicated that a strong liquid disturbance is caused by the bubble motions, leading to the enhancement of both convective boiling and condensation heat transfer. More bubbles are generated as the heat flux increases; therefore, the disturbance of bubble motion on liquid pool and condensation film becomes stronger, resulting in better thermal performance of the flat two-phase thermosyphon. The addition of the wick inside the cavity effectively reduces the temperature oscillation of the evaporator wall. In addition, the wick structure provides backflow paths for the condensate owing to the effect of capillary force and enhances the vapor–liquid phase change heat transfer, resulting in the improvement of thermal performance for the flat two-phase thermosyphon.

Due to the growing interest using model predictive control (MPC), there are more and more researches about the applications of MPC on autonomous underwater vehicle (AUV), and these researches are mainly focused on simulation and simple application of MPC on AUV. This paper focuses on the improvement of MPC based on the state space model of an AUV. Unlike the previous approaches using a fixed weighting matrix, in this paper, a coefficient, varied with the error, is introduced to adjust the control increment vector weighting matrix to reduce the settling time. Then, an analysis on the effect of the adjustment to the stability is given. In addition, there is always a lag between the AUV real trajectory and the desired trajectory when the AUV tracks a continuous trajectory. To solve this problem, a simple re-planning of the desired trajectory is developed. Specifically, the point certain steps ahead from current time on the desired trajectory is treated as the current desired point and input to the controller. Finally, experimental results for depth control are given to demonstrate the feasibility and effectiveness of the improved MPC. Experimental results show that the method of real-time adjusting control increment weighting matrix can reduce settling time by about 2 s when tracking step trajectory of 1 m, and the simple re-planning of the desired trajectory method can reduce the average of absolute error by about 15% and standard deviation of error by about 17%.

In this research, a new triterpenoid, tirucalla-8,24-diene-3β,11β-diol-7-one (1), and eupha-8,24-diene-3β,11β-diol-7-one (2), which was isolated from Euphorbia kansui for the first time, together with twelve other known compounds (3–14), were isolated from the ethyl acetate extract of Euphorbia kansui. Their structures were elucidated based on High resolution electrospray ionization mass spectrometry (HR-ESI-MS), Infrared Spectroscopy (IR), 1D and 2D Nuclear Magnetic Resonance (NMR) data. Both constituents 1 and 2 exhibited moderate cytotoxicity against colon cancer HCT-116, gastric cancer MKN-45 and breast cancer MCF-7.
