Background Gastric stump ("remnant") cancer is the development of a malignancy related to previous gastric surgery. Prognosis in gastric stump cancer, compared with that in primary gastric cancer, is still controversial. Methods From January 1988 to December 2012 at a single medical centre in Taiwan, 105 patients with gastric stump cancer, including 85 with previous peptic ulcer disease and 20 with previous gastric cancer, were analyzed for clinicopathologic characteristics and overall survival (os). Results The 5-year os rates for patients with gastric stump cancer and with primary gastric cancer were 51.2% and 54.5% respectively (p = 0.035). Analysis of clinicopathologic characteristics indicated that, compared with patients having primary gastric cancer, those with gastric stump cancer had more lymph node metastasis (p < 0.001) and had been diagnosed at a more advanced stage (p = 0.047). Multivariate analysis with os as an endpoint showed that age [p = 0.015; hazard ratio (hr): 2.300; 95% confidence interval (ci): 1.173 to 4.509], tumour size (p = 0.037; hr: 1.700; 95% ci: 1.031 to 2.801), stromal reaction (p = 0.021; hr: 1.802; 95% ci: 1.094 to 2.969), and pathologic N category (p = 0.001; hr: 1.449; 95% ci: 1.161 to 1.807) were independent predictors in gastric stump cancer. The os rates for patients with gastric stump cancer who previously had gastric cancer or peptic ulcer disease were 72.9% and 50.0% respectively (p = 0.019). The Borrmann classification was more superficial (p = 0.005), lymph node metastases were fewer (p = 0.004), and staging was less advanced (p = 0.025) in patients with gastric stump cancer who previously had gastric cancer than in their counterparts who previously had peptic ulcer disease. Conclusions Survival is poorer in patients with gastric stump cancer who previously had peptic ulcer disease than in those who previously had primary gastric cancer. Patients with gastric stump cancer who previously had gastric cancer and could receive curative gastrectomy tended to have a better prognosis because of a more superficial Borrmann classification. Regular follow-up in patients who have undergone gastric surgery is recommended for the early detection of gastric stump cancer. [ABSTRACT FROM AUTHOR]

Management of anticoagulant therapy for the treatment of venous thromboembolism (VTE) in cancer patients is complex because of an increased risk of recurrent VTE and major bleeding complications in those patients relative to the general population. Subgroups of patients with cancer also show variation in their risk for recurrent VTE and adverse bleeding events. Accordingly, a committee of 10 Canadian clinical experts developed the consensus riskstratification treatment algorithm presented here to provide guidance on tailoring anticoagulant treatment choices for the acute and extended treatment of symptomatic and incidental VTE, to prevent recurrent VTE, and to minimize the bleeding risk in patients with cancer. During a 1-day live meeting, a systematic review of the literature was performed, and a draft treatment algorithm was developed. The treatment algorithm was refined through the use of a Web-based platform and a series of online teleconferences. Clinicians using this treatment algorithm should consider the bleeding risk, the type of cancer, and the potential for drug--drug interactions in addition to informed patient preference in determining the most appropriate treatment for patients with cancer-associated thrombosis. Anticoagulation therapy is used to treat VTE; however, patients with cancer have unique clinical circumstances that can often make decisions surrounding the administration of therapeutic anticoagulation complicated. No national Canadian guidelines on the management of established cancer-associated thrombosis have been published. We therefore aimed to develop a consensus-based, evidence-informed guideline on the topic. PubMed was searched for clinical trials and meta-analyses published between 2002 and 2013. Reference lists of key articles were hand-searched for additional publications. Content experts from across Canada were assembled to review the evidence and make recommendations. Low molecular weight heparin is the treatment of choice for cancer patients with established VTE. Direct oral anticoagulants are not recommended for the treatment of VTE at this time. Specific clinical scenarios, including the presence of an indwelling venous catheter, renal insufficiency, and thrombocytopenia, warrant modifications in the therapeutic administration of anticoagulation therapy. Patients with recurrent VTE should receive extended (>3 months) anticoagulant therapy. Incidental VTE should generally be treated in the same manner as symptomatic VTE. There is no evidence to support the monitoring of anti-factor Xa levels in clinically stable cancer patients receiving prophylactic anticoagulation; however, levels of anti-factor Xa could be checked at baseline and periodically thereafter in patients with renal insufficiency. In this work, we report the use of bio-functionalized magnetic nanoparticles (BMNs) and dynamic magnetic resonance (DMR) to characterize the time-dependent spin-spin relaxation time for sensitive bio-detection. The biomarkers are the human C-reactive protein (CRP) while the BMNs are the anti-CRP bound onto dextran-coated Fe3O4 particles labeled as Fe3O4-antiCRP. It was found the time-dependent spin-spin relaxation time, T2, of protons decreases as time evolves. Additionally, the ΔT2 of of protons in BMNs increases as the concentration of CRP increases. We attribute these to the formation of the magnetic clusters that deteriorate the field homogeneity of nearby protons. A sensitivity better than 0.1 µg/mL for assaying CRP is achieved, which is much higher than that required by the clinical criteria (0.5 mg/dL). Background: Postoperative ileus (POI) is a common postoperative clinical complication that significantly affects postoperative rehabilitation and quality of life in patients and can even produce secondary complications, leading to serious consequences. External treatment using Shenhuang Plaster (SHP) (Shenque acupoint administration) has definite effects and unique advantages in the prevention and treatment of POI, but its mechanism is not completely clear. In this study, we investigated the therapeutic mechanism behind the effect of Shenhuang Plaster applied to the Shenque acupoint on gastrointestinal motility in POI mice based on metabolomics., Materials and Methods: C57BL/6 mice were divided into three groups: blank control (Ctrl), model (POI), and intervention (POI + SHP) groups. SHP treatment was started 3 days before modeling. We employed several behavioral tests and gastrointestinal transit function measurements and performed qRT-PCR analysis, 16S rRNA gene sequencing, and metabolomics analysis on serum metabolites., Results: We found that SHP could reduce the mRNA expression of inflammatory mediators in the smooth muscle tissue of the small intestine, regulate the structure and function of the intestinal microbiota, and modulate serum phenylalanine, carnitine, and glutamic acid levels., Conclusions: POI mice had obvious intestinal flora disorders and metabolic disorders of amino acids and their derivatives, and there was a significant correlation between differential flora and differential metabolites. SHP could effectively regulate the concentration of intestinal flora and serum metabolites and the metabolic pathway related to amino acids in vivo and, ultimately, achieve a therapeutic purpose in POI. In this study, it was found, for the first time, that applying SHP to the Shenque acupoint could effectively regulate the serum metabolites of phenylalanine, carnitine, and glutamate, and improve postoperative intestinal motile disturbance through association with the intestinal flora.

Current stress sensors for microsystems face integration challenges and complex signal decoding. This paper proposes a real-time uniaxially sensitive stress sensor. It is obtained by simple combinations of bar resistors using their sensitivity differences in different axes. With the aid of a Wheatstone bridge, the sensor can measure the uniaxial stress magnitude by simple calibration of the stress against the output voltage and detect the bidirectional stress magnitude and direction in a micro-zone by simple rotation. The theoretical sensitivity obtained from simulation is 0.087 mV/V·MPa when the X-bridge is stressed in the X-direction under 1 V of excitation, and the test sensitivity of the X-bridge prepared in this paper is 0.1 mV/V·MPa. The design is structurally and procedurally simple, exhibits better temperature stability, and reduces interface requirements, making it suitable for the health monitoring of multi-chip microsystem chips.

Aims: The screening and diagnosis of dengue virus infection play a crucial role in controlling the epidemic of dengue fever, highlighting the urgent need for a highly sensitive, simple, and rapid laboratory testing method. This study aims to assess the clinical performance of MAGLUMI Denv NS1 in detecting dengue virus NS1 antigen., Methods: A retrospective study was conducted to assess the sensitivity and specificity of MAGLUMI Denv NS1 using residual samples. Dengue-confirmed and excluded samples, validated by qPCR, were subjected to testing with MAGLUMI Denv NS1 in accordance with the manufacturer's instructions. The linear range, endogenous interference, and cross-reactivity of MAGLUMI Denv NS1 were verified, and a consistency analysis with commercial comparator products was carried out., Results: The diagnostic specificity of MAGLUMI Denv NS1 is 98.41% (62/63), and the sensitivity is 98.32% (117/119). It effectively detects various serotypes of dengue virus, with no observed endogenous interference or cross-reactivity. Additionally, the consistency of NS1, IgM, and IgG tests on the MAGLUMI platform compared to commercial comparator reagents reaches 85.71%, 99.25%, and 98.97%, respectively., Conclusions: The MAGLUMI Denv NS1 represents a highly sensitive laboratory testing method capable of enhancing the diagnostic accuracy and efficiency of dengue virus infection detection.

In response to the intensifying competition in the mold market and the increasingly stringent specifications of die forgings, the existing 55NiCrMoV7 (MES 1 steel) material can no longer meet the elevated demands of customers. Consequently, this study systematically optimizes the alloy composition of MES 1 steel by precisely adjusting the molybdenum (Mo) and vanadium (V) contents. The primary objective is to significantly enhance the microstructure and thermal-mechanical fatigue performance of the steel, thereby developing a high-performance, long-life hot working die steel designated as MES 2 steel. The thermal-mechanical fatigue (TMF) tests of two test steels were conducted in reverse mechanical strain control at 0.6% and 1.0% strain levels by a TMF servo-hydraulic testing system (MTS). The microstructures of the two steels were characterized using scanning electron microscopy (SEM), electron backscatter diffraction (EBSD), and transmission electron microscopy (TEM). The results indicate that throughout the entire thermomechanical fatigue cycle, both steels exhibit initial hardening during the low-temperature half-cycle (tension half-cycle) and subsequent continuous softening during the high-temperature half-cycle (compression half-cycle). Furthermore, under the same strain condition, the cumulative cyclic softening damage of MES 1 steel is more pronounced than that of the newly developed MES 2 steel. The number, width, and length of cracks in MES 2 steel are smaller than those in MES 1 steel, and the thermomechanical fatigue life of MES 2 steel is significantly longer than that of MES 1 steel. The microstructures show that the main precipitate phase in MES 1 steel is Cr-dominated rod-shaped carbide. It presents obvious coarsening and is prone to inducing stress concentration, thus facilitating crack initiation and propagation. The precipitate phase in MES 2 steel is mainly MC carbide containing Mo and V. It has a high thermal activation energy and is dispersed in the matrix in the form of particles, pinning dislocations and grain boundaries. This effectively delays the reduction in dislocation density and grain growth, thus contributing positively to the improvement in thermomechanical fatigue performance.

Macrophages play important roles in metabolic dysfunction-associated steatohepatitis (MASH), an advanced and inflammatory stage of metabolic dysfunction-associated steatotic liver disease (MASLD). In humans and mice, the cellular heterogeneity and diverse function of hepatic macrophages in MASH have been investigated by single cell RNA sequencing (scRNA-seq). However, little is known about their roles in rats. Here, we collected liver tissues at the postnatal week 16, when our previously characterized Lep ∆I14/∆I14 rats developed MASH phenotypes. By scRNA-seq, we found an increase in the number of macrophages and endothelial cells and a decrease in that of NK and B cells. Hepatic macrophages in rats underwent a unique M1 to M2 transition without expression of the classical markers such as Arg1 and Nos2, except for Cd163. Lipid-associated macrophages (LAMs) were increased, which could be detected by the antibody against Cd63. In the microenvironment, macrophages had an increased number of interactions with hepatocytes, myofibroblasts, T cells, neutrophils, and dendritic cells, while their interaction strengths remained unchanged. Finally, the macrophage migration inhibitory factor (MIF) pathway was identified as the top upregulated cell-communication pathway in MASH. In conclusion, we dissected hepatic macrophage dynamics during MASH at single cell resolution and provided fundamental tools for the investigation of MASH in rat models.

The global epidemic of bovine coronavirus (BCoV) has caused enormous economic losses. The characterisation and genetic composition of endemic strains in Southwest China remain elusive. This study aimed to fill this gap by isolating three BCoV strains from this region and sequencing their whole genomes. To elucidate the genetic evolution and characterisation of the prevalent strains, the results of BCoV sequences were compared in GenBank, with a focus on genetic evolution, mutation, and recombination patterns. The results showed close homology between strains NN190313 and NN230328, while strain NN221214 showed less similarity to these two strains but clustered with the French strain of the European branch. Intriguingly, NN190313 and NN230328 were grouped with goat-derived BCoV strains from Jiangsu Province in Eastern China in the Asian-American branch. In addition, recombination analyses revealed significant signals between NN230328 and either a Chinese goat-derived strain (XJCJ2301G) or a Shandong strain (ShX310). This study highlights the importance of monitoring cross-species transmission between cattle and goats, especially in the mountainous areas of Southwest China where mixed farming occurs, and thus, the monitoring of cross-species transmission between cattle and goats is important for preventing new public health challenges, providing important insights for research on cross-species transmission, early prevention, and control measures, with potential applications in vaccine development.

Neutrophil extracellular traps (NETs) formation is a key process in inflammatory diseases like gout, but the underlying molecular mechanisms remain incompletely understood. This study aimed to establish a model to examine the formation of NETs induced by monosodium urate (MSU) and phorbol 12-myristate 13-acetate (PMA) and to elucidate their molecular pathways. Laser confocal microscopy was used to visualize NET formation, while flow cytometry was employed to detect reactive oxygen species (ROS) production. The microstructure of neutrophils was observed by transmission electron microscopy, and the expression of key proteins was determined by Western blotting. Additionally, the effect of various inhibitors targeting the MAPK signaling pathway on NET formation was evaluated. They include the Ras inhibitor Salirasib, Raf inhibitor Vemurafenib, ERK inhibitor PD98059, and p38 MAPK inhibitor SB203580, as well as NADPH oxidase inhibitor DPI and neutrophil elastase inhibitor Alvelestat. The results showed that MSU and PMA triggered significant NET formation, which was accompanied by increased ROS levels, lactate dehydrogenase release, dsDNA, and IL-8. Notably, selective MAPK pathway inhibitors and DPI and Alvelestat, except for SB203580, effectively down-regulated these indicators. These data indicated that the activation of a signaling pathway involving Ras-Raf-ERK, which is dependent on ROS, is crucial for the induction of NET formation by MSU and PMA. Given the involvement of NETs in multiple pathologies, our findings could potentially serve as molecular targets for the intervention and treatment of crystal-related diseases, especially for gout.

In order to solve the problems of high planting density, similar color, and serious occlusion between spikes in sorghum fields, such as difficult identification and detection of sorghum spikes, low accuracy and high false detection, and missed detection rates, this study proposes an improved sorghum spike detection method based on YOLOv8s. The method involves augmenting the information fusion capability of the YOLOv8 model's neck module by integrating the Gold feature pyramid module. Additionally, the SPPF module is refined with the LSKA attention mechanism to heighten focus on critical features. To tackle class imbalance in sorghum detection and expedite model convergence, a loss function incorporating Focal-EIOU is employed. Consequently, the YOLOv8s-Gold-LSKA model, based on the Gold module and LSKA attention mechanism, is developed. Experimental results demonstrate that this improved method significantly enhances sorghum spike detection accuracy in natural field settings. The improved model achieved a precision of 90.72%, recall of 76.81%, mean average precision (mAP) of 85.86%, and an F1-score of 81.19%. Comparing the improved model of this study with the three target detection models of YOLOv5s, SSD, and YOLOv8, respectively, the improved model of this study has better detection performance. This advancement provides technical support for the rapid and accurate recognition of multiple sorghum spike targets in natural field backgrounds, thereby improving sorghum yield estimation accuracy. It also contributes to increased sorghum production and harvest, as well as the enhancement of intelligent harvesting equipment for agricultural machinery.

A series of Si-H- or Si-Vi-terminated, branched and linear oligomers containing Me 2 SiO segments were prepared by equilibrium polymerization or non-equilibrium polymerization initiated by living anions, respectively. These oligomers were used to improve the defects of concentrated crosslinking points and the high hardness of crosslinked products when using phenyltris(dimethylsiloxy)silane or 1,1,5,5-tetramethyl-3,3-diphenyl trisiloxane as crosslinking agents in the preparation of silicone gel. NMR, FT-IR, and GPC characterized the structure and molecular weight information of the prepared oligomers. The effects of equilibrium polymerization and the anionic non-equilibrium ring-opening polymerization methods on the structure of oligomers were investigated in detail, together with the structure, the molar ratio of SiH to SiVi, and the phenyl content on the thermal properties and the transmittance retention yield of the silicone gel. The introduction of phenyl groups increases the glass transition temperature of silicone gel from -121.29 °C to -117.71 °C when the phenyl content increased from 0.88 wt% to 3.17 wt%. Meanwhile, the thermal decomposition temperature of silicone gel at 10% weight loss in the N 2 atmosphere increased from 440.5 °C to 480.0 °C. When the SiH/SiVi molar ratio is close to 1.0, the transmittance retention yield of the prepared silicone gel using Si-Vi-terminated phenyl T-shaped polysiloxane as the matrix and α , ω -dimethylsiloxyl-terminated PDMS as the crosslinking agent could reach 88.9% after 25 min of UV irradiation.

Based on the background of the continuously rising global demand for clean energy, offshore wind power, as an important form of renewable energy utilization, is booming. However, the pile foundations of offshore wind turbines are subject to long-term erosion in the harsh marine environment, and the problem of corrosion damage is prominent, which seriously threatens the safe and stable operation of the wind power system. In view of this, a meshless numerical simulation method based on smoothed particle hydrodynamics (SPH) and a method for generating the concrete meso-structures are developed. Concrete pile foundation models with different aggregate contents, particle sizes, and ion concentration diffusion coefficients are established to simulate the corrosion damage processes under various conditions. The rationality of the numerical algorithm is verified by a typical example. The results show that the increase in the aggregate percentage gradually reduces the diffusion rate of chemical ions, and the early damage development also slows down. However, as time goes, the damage will still accumulate continuously; when the aggregate particle size increases, the ion diffusion becomes more difficult, the damage initiation is delayed, and the early damage is concentrated around the large aggregates. The increase in the ion diffusion coefficient significantly accelerates the ion diffusion process, promotes the earlier and faster development of damage, and significantly deepens the damage degree. The research results contribute to a deeper understanding of the corrosion damage mechanisms of pile foundations and providing important theoretical support for optimizing the durability design of pile foundations. It is of great significance for ensuring the safe operation of offshore wind power facilities, prolonging the service life, reducing maintenance costs, and promoting the sustainable development of offshore wind power.

Within the domain of traditional art, Chinese Wuhu Iron Painting distinguishes itself through its distinctive craftsmanship, aesthetic expressiveness, and choice of materials, presenting a formidable challenge in the arena of stylistic transformation. This paper introduces an innovative Hierarchical Visual Transformer (HVT) framework aimed at achieving effectiveness and precision in the style transfer of Wuhu Iron Paintings. The study begins with an in-depth analysis of the artistic style of Wuhu Iron Paintings, extracting key stylistic elements that meet technical requirements for style conversion. Furthermore, in response to the unique artistic characteristics of Wuhu Iron Paintings, this research constructs a multi-layered network structure capable of effectively capturing and parsing style and content features. Building on this, we have designed an Efficient Local Attention Decoder (ELA-Decoder) that adaptively decodes the style and content features through correlation, significantly enhancing the length dependency of local and global information. Additionally, this paper proposes a Content Correction Module (CCM) to eliminate redundant features generated during the style transfer process, further optimizing the migration results. In light of the scarcity of existing datasets for Wuhu Iron Paintings, this study also collects and constructs a dedicated dataset for the style transfer of Wuhu Iron Paintings. Our method achieves optimal performance in terms of loss metrics, with a reduction of at least 4% in style loss and 5% in content loss compared to other advanced methods. Moreover, expert evaluations were conducted to validate the effectiveness of our approach, and the results show that our method received the highest number of votes, further demonstrating its superiority.

The shear failure of rock masses is one of the primary causes of underground engineering instability. The shear mechanical behavior of rocks at different sizes is of great significance for studying the shear failure pattern of engineering rock masses. However, due to the presence of various joints and defects in natural rocks, the obtained rock specimens exhibit significant discreteness, making it difficult to customize specimen sizes for size effect studies. In recent years, 3D printing (3DP) technology has gained widespread application in rock mechanics tests due to its high printing precision and ability to form specimens in a single step with minimal discreteness. Among these, specimens prepared using sand-powder 3DP exhibit elastoplastic mechanical characteristics similar to those of natural rocks. Therefore, this study utilized sand-powder 3DP to prepare rock-like specimens of four different sizes and conducted compression shear tests under three different shear velocities. The shear strength, shear strain, and wear of the shear surfaces were analyzed as functions of specimen size and shear velocities. The results indicate that under the same shear velocity, the shear strength of the specimens is negatively correlated with specimen size. The peak shear strain is generally unaffected by shear velocities, but it increases initially and then decreases with increasing specimen size. As specimen size increases, the degree of specimen damage intensifies, and larger specimens are more prone to developing derived fractures. This study broadens the application of sand-powder 3DP technology in investigating the shear mechanical properties of soft rocks, offering novel insights into the study of size effects in rock mechanics. However, the current research does not encompass tests on 3D-printed rock specimens with varying printing directions, nor does it delve into the role of fractures in size effect analyses. Future investigations will aim to address these limitations, thereby advancing the applicability of 3D printing technology in rock mechanics research and enhancing its contributions to the field.

Ta/Re layered composite material is a high-temperature material composed of the refractory metal tantalum (Ta) as the matrix and high-melting-point, high-strength rhenium (Re) as the reinforcement layer. It holds significant potential for application in aerospace engine nozzles. Developing the Ta/Re potential function is crucial for understanding the diffusion behavior at the Ta/Re interface and elucidating the high-temperature strengthening and toughening mechanism of Ta/Re layered composites. In this paper, the embedded atom method (EAM) potential function for tantalum/rhenium binary alloys (Ta-Re alloys) is derived using the force-matching method and validated through first-principles calculations and experimental characterization. The results show that for the lattice constant of a bcc structure containing 54 atoms, surface formation energies per unit area of Ta-Re alloys obtained based on the potential function are 12.196 Å, E 100 = 0.16 × 10 -2 eV, E 110 = 0.10 × 10 -2 eV, and E 111 = 0.08 × 10 -2 eV, with error values of 0.015 Å, 0.04 × 10 -2 eV, 0.02 × 10 -2 eV, and 0.01 × 10 -2 eV, respectively, compared with the calculations from first principles calculations. It is noteworthy that the errors in the average binding energies of Ta-rich (Ta 39Re20 , where the number of Ta atoms is 39 and Re atoms is 20) and Re-rich (Ta 20 Re 39 , where the number of Ta atoms is 20 and Re atoms is 39) cluster atoms, calculated by the potential function and first-principles methods, are only 1.64% to 1.98%. These results demonstrate the accuracy of the constructed EAM potential function. Based on this, three compositions of Ta-Re alloys (Ta 48 Re 6 , Ta 30 Re 24 , and Ta 6 Re 48 ; the numerical subscripts represent the number of atoms of each corresponding element) were randomly synthesized, and a comparative analysis of their bulk moduli was conducted. The results revealed that the experimental values of the bulk modulus showed a decreasing and then an increasing tendency with the calculated values, which indicated that the potential function has a very good generalization ability. This study can provide theoretical guidance for the modulation of Ta/Re laminate composite properties.

There are many applications of soybean lecithin (SL) and cholesterol-loaded cyclodextrin (CLC) in sperm freezing processes. To the best of our knowledge, there have been few cases of the combined use of SL and CLC in freezing rooster semen. We investigated the effects of CLC, SL, and their combination on rooster sperm cryodamage. Three experiments were conducted: experiment 1, SL (0.5%, 1%, 1.5%, 2.0%); experiment 2, CLC (1.25 mg, 2.5 mg, 5 mg, 10 mg, 15 mg); and experiment 3, CLC + SL (2.5 mg + 0.25%, 2.5 mg + 0.5%, 2.5 mg + 1%, 2.5 mg + 1.5%). Semen samples were cryopreserved using a programmed cryostat, followed by the determination of post-thaw sperm quality, antioxidant indices, and hatching. The results showed that the combination of 2.5 mg CLC + 0.5% SL had the most significant synergistic effect on cryodamage, and the viability (56.69%), motility (54.35%), mitochondrial activity (54.23%), plasma membrane integrity (53.52%), acrosome integrity (54.71%), and antioxidant activity (MDA concentration: 5.65 nmol/mL; SOD activity: 152.73 U/mL) were significantly greater than those of the other combinations ( p < 0.05). Nevertheless, the combined CLC and SL addition group did not substantially increase the fertilization and hatching rates of frozen semen compared with the addition of 2.5 mg CLC. In conclusion, the addition of 2.5 mg CLC and 2.5 mg CLC + 0.5% SL enhanced the quality and fertility of frozen rooster sperm.

In response to the increasingly strict performance requirements of large molds, a novel Cr-Mo-V hot-work die steel has been developed. In order to study the high-temperature hot deformation behavior and plasticity of the novel steel, hot compression tests were conducted on the Gleeble-1500D thermal simulation testing machine at a deformation temperature of 950~1200 °C and a strain rate of 0.001~5 s -1 . Based on the Arrhenius constitutive model, a novel Cr-Mo-V steel high-temperature constitutive model considering strain was established. The reliability and applicability of this modified model, which includes strain compensation, were assessed using the phase relationship coefficient (R) and the average absolute relative error (AARE). The values of R and AARE for comparing predicted outcomes with experimental data were 0.98902 and 3.21%, respectively, indicating that the model demonstrated high precision and reliability. Based on the Prasad criterion, a 3D hot processing map of the novel Cr-Mo-V steel was established, and the instability zone of the material was determined through the hot processing map: the deformation temperature (950~1050 °C) and strain rate (0.001~0.01 s -1 ) were prone to adiabatic shear and crystal mixing. The suitable processing range was determined based on the hot processing map: The first suitable processing area was the strain range of 0.05~0.35, the temperature range was 1100~1175 °C, and the strain rate was 0.001~0.009 s -1 . The second suitable processing area was a strain of 0.45~0.65, a temperature of 1100~1200 °C, and a strain rate of 0.0024~0.33 s -1 . Finally, the forging process of hundred-ton die steel forging was developed by combining 3D hot processing maps with finite element simulation, and the forging trial production of 183 t forging was carried out. The good forging quality indicated that the established hot processing map had a good guiding effect on the production of 100-ton test steel forging.

Rolling bearings are critical rotating components in machinery and equipment; they are essential for the normal operation of such systems. Consequently, there is a pressing need for a highly efficient, applicable, and reliable method for bearing fault diagnosis. Currently, one-dimensional data-driven fault diagnosis methods, which rely on one-dimensional data, represent a mainstream approach in this field. However, these methods exhibit weak diagnostic capabilities in noisy environments and when confronted with insufficient sample sizes. In order to solve these limitations, a new fault diagnosis method for rolling bearings is proposed, which combines the ConvNeXt network and improved DenseBlock into a parallel network with a feature fusion function. The network can fully extract the global feature and the detail feature of the signal and integrate them, which shows a good diagnostic ability in the face of a strong noise environment. Additionally, the Dy-ReLU function is introduced into the network, which enhances the generalization ability of the network and improves the convergence speed. Comparative experiments show that this method still has strong fault diagnosis capability under the condition of noise pollution and insufficient training samples.

Biological tissue defects are typically characterized by various shaped defects, and they are prone to inflammation and the excessive accumulation of reactive oxygen species. Therefore, it is still urgent to develop functional materials which can fully occupy and adhere to irregularly shaped defects by injection and promote the tissue repair process using antioxidant and anti-inflammatory mechanisms. Herein, in this work, phenylboronic acid modified oxidized hyaluronic acid (OHAPBA) was synthesized and dynamically crosslinked with catechol group modified glycol chitosan (GCHCA) and guar gum (GG) into a hydrogel loaded with curcumin liposomes (Cur-LPs) which were relatively uniformly distributed around 180 nm. The hydrogel possessed rapid gelation within 30 s, outstanding injectability and tissue-adaptive properties with self-healing properties, and the ability to adhere to biological tissues and adapt to tissue movement. Moreover, good biocompatibility and higher DPPH scavenging efficiency were illustrated in the hydrogel. And a more sustainable release of curcumin from Cur-LPs-loaded hydrogels, which could last for 10 days, was achieved to improve the bioavailability of curcumin. Finally, they might be injected to fully occupy and adhere to irregularly shaped defects and promote the tissue repair process by antioxidant mechanisms and the sustained release of curcumin for anti-inflammation. And the hydrogel would have potential application as candidates in tissue defect repair.

TFE3 -rearranged Renal Cell Carcinoma (TFE3-RCC) is an aggressive subtype of RCC characterized by Xp11.2 rearrangement, leading to TFE3 fusion proteins with oncogenic potential. Despite advances in understanding its molecular biology, effective therapies for advanced cases remain elusive. This study investigates the role of ARID2 , a component of the SWI/SNF chromatin remodeling complex, in TFE3-RCC. Through a series of in vitro and in vivo experiments, we confirmed that ARID2 acts as a tumor suppressor in TFE3-RCC. ARID2 knockout (KO) enhanced TFE3-RCC cell migration, proliferation, and tumor growth. Transcriptomic analysis revealed ERBB3 as a key target gene regulated by both PRCC-TFE3 and ARID2 . Chromatin immunoprecipitation (ChIP) assays demonstrated that PRCC-TFE3 directly binds to and upregulates ERBB3 expression, with ARID2 KO further enhancing this effect. TFE3-RCC ARID2 KO cells exhibited significant gene expression enrichment in MAPK and ERBB3 signaling pathways. These cells also showed increased activation of ERBB3, EGFR, and selective activation of SRC and MAPK. TFE3-RCC ARID2 KO cells demonstrated heightened sensitivity to the ERBB3 inhibitor AZD8931 compared to their wild-type counterparts, exhibiting significantly reduced migration and proliferation rates. These findings suggest that the PRCC-TFE3- ARID2 -ERBB3 axis plays a critical role in TFE3-RCC pathogenesis and highlights the potential of targeting ERBB3 in ARID2 -deficient TFE3-RCC as a therapeutic strategy. This study provides new insights into the molecular mechanisms of TFE3-RCC and suggests avenues for precision treatment of this aggressive cancer.

Aflatoxin B 1 is a prevalent secondary hazardous metabolite generated by fungus present in feed ingredients and the surrounding environment: enzymes are currently being recognized as an efficient and promising approach to reducing the associated risks. The objective of this study was to assess the effects of varying doses of enzyme complexes on several parameters in laying hens that were exposed to aflatoxin. During an 8-week experiment, a total of 288 Yukou Jingfen No.6 laying hens were placed into four groups. These groups included a group treated with toxins (CON group) and groups supplemented with compound enzyme complexes at doses of 250 g/t (E1 group), 500 g/t (E2 group), and 1000 g/t (E3 group). The E2 and E3 groups exhibited a statistically significant 2.6% increase in egg production rate compared to the CON group ( p < 0.05). In addition, the E2 group showed significant improvements in both the feed-to-egg ratio and egg weight ( p < 0.05). In addition, the E2 and E3 groups showed improved hutch unit and egg white height compared to the control group ( p < 0.05). The E2 and E3 groups showed a substantial rise in liver health indicators, namely serum alanine transaminase (ALT) and alkaline phosphatase (ALP) activity. On the other hand, malondialdehyde (MDA) was lowered, and total superoxide dismutase (T-SOD) and total antioxidant capacity (T-AOC) were raised. These findings were statistically significant ( p < 0.05). The E2 and E3 groups showed notable enhancements in intestinal morphology, as evidenced by a rise in villus height and a decrease in crypt depth in all segments of the intestine ( p < 0.05). Furthermore, analysis of 16S rRNA sequencing revealed that these participants had a higher prevalence and variety of microorganisms in their gut microbiota. More precisely, there was a significant rise in the abundance of Bacteroidota and a decline in Firmicutes at the level of the phylum. In general, the inclusion of the enzyme complex had advantageous impacts on performance, egg quality, intestinal morphology, intestinal barrier function, and intestinal flora in laying hens. Our results indicate that toxin-degrading enzymes, when used as feed additives, play a significant role in mitigating AFB 1 contamination in diets and improving the production performance of laying hens.

Cementitious material durability is affected by the transport and adsorption of water molecules and ions in the nanopore channels of cement hydration products. Hydrated calcium silicate (C-S-H) accounts for about 70% of the hydration product. It determines the mechanical properties of cementitious materials and their internal transport properties. The molecular dynamics method provides a complementary understanding of experimental and theoretical results. It can further reveal water molecules and ions' adsorption and transport mechanisms in C-S-H gel pores. This review article provides an overview of the current state of research on the structure of C-S-H gels and the adsorption and transport properties of water molecules and ions within C-S-H gels, as studied through molecular dynamics simulations. This paper summarizes the results of the molecular dynamics-based adsorption transport properties of water molecules and ions in C-S-H gels. The deficiencies in the current study were analyzed, and the fundamental problems to be solved and further research directions were clarified to provide scientific references for revealing the structural properties of C-S-H gels using molecular dynamics.

Mesocotyl plays a key role in the seedling emergence of maize; however, the mechanism of mesocotyl elongation is still unclear. Moreover, different maize inbred lines and cultivars have varied mesocotyl lengths positively correlated with deep sowing tolerance. In this study, we selected one inbred line with long mesocotyl (LM) and two maize inbred lines with short mesocotyl (SM1 and SM2) from more than 400 maize inbred lines. The mesocotyl length of the LM line was about three-fold longer than those of the SM1 and SM2 lines. Microstructure observation showed that the reason for short mesocotyl in the SM1 and SM2 lines was few cell numbers and short cell length, respectively. Subsequently, we used RNA-seq to investigate the mechanism of mesocotyl elongation by regulating cell number and cell length at the transcriptome level. Compared with the LM line, the SM1 line displayed stronger downregulation of Cytochrome P450 and peroxidase genes than the SM2 line. Moreover, plant hormone signal transduction plays a vital role in mesocotyl elongation. Taken together, we propose a model for mesocotyl elongation of maize inbred lines with different cell lengths and cell numbers, which provide new insights into mesocotyl elongation in maize.

The measurement of downhole drilling motor rotational speed is crucial for optimizing drilling operations, improving work efficiency, and preventing equipment failures. However, traditional downhole rotational speed sensors suffer from power supply limitations, which can increase drilling costs. To address this issue, this study presents a novel multifunctional rotational speed sensor based on triboelectric nanogenerator (TENG) technology, enabling the self-powered measurement of rotational speed, direction, and angle. Our experimental results demonstrate that the sensor operates stably within a temperature range of 0 to 150 °C and a humidity range of 0 to 90%. It achieves rotational speed measurement with an accuracy of less than 2.5% error within a range of 0 to 1000 rpm, angular measurement with a resolution of 60 degrees and an error of less than 2% within a range of 0 to 360 degrees, and rotational direction measurement. Furthermore, the sensor exhibits self-powered functionality, achieving a maximum power output of 29.1 μW when the external load is 10 MΩ. Compared to conventional rotational speed sensors, this sensor possesses the unique advantage of integrating the measurement of rotational speed, angle, and direction, while simultaneously harnessing downhole working conditions for self-power generation. These characteristics make it highly suitable for practical downhole environments.

Cadmium (Cd) exposure can induce follicular atresia and laying performance reduction in hens, which is linked to autophagy within the granulosa cells. Selenium (Se) can influence autophagy and counteract Cd toxicity. This study aimed to investigate the protective effect of Se on Cd-induced follicular atresia in laying hens. Sixty-four laying hens were randomly allocated into 4 treatments: control group: basal diet; Se group: basal diet + 0.4 mg/kg Se from selenized yeast; Cd group: basal diet + 25 mg/kg Cd from CdCl 2 ; and Cd+Se group: basal diet + 25 mg/kg Cd + 0.4 mg/kg Se. Compared to the Cd group, Se supplementation alleviated the ovarian pathological changes and oxidative stress in the follicles, serum, liver, and ovary, increased daily laying production, ovarian weight and F5-F1 follicle amounts, serum levels of progesterone and oestradiol, and up-regulated mTOR expression ( p < 0.05), while decreasing the count of autophagic vacuoles, ovarian atresia follicle numbers, and Cd deposition, and down-regulated expression levels of autophagy-related mRNAs, including ATG5, LC3-I, and LC3-II, Beclin1, and Dynein in the follicles ( p < 0.05). In conclusion, 0.4 mg/kg Se supplementation protected against Cd-induced laying performance reduction and follicular atresia, which were achieved via decreasing oxidative stress and inhibiting mTOR pathways of autophagy.

Background/Objectives : Ricin's high toxicity and potential as a bioweapon underscore the need for effective antidotes. Monoclonal antibodies, though effective, are limited by complex production. This study aimed to develop a graphene oxide-based aptamer nanoarray (ARMAN) for improved neutralization and protection against ricin. Methods: High-affinity aptamers targeting ricin's RTA and RTB subunits were selected using SELEX technology and conjugated to graphene oxide (GO) via click chemistry. ARMAN's characteristics, including morphology, stability, and biosecurity, were assessed. Its performance was evaluated in terms of affinity for ricin, neutralization capacity, and therapeutic effects in cellular assays and a mouse model of ricin poisoning. Results : ARMAN exhibited a uniform morphology with an average particle size of 217 nm and demonstrated significantly enhanced affinity for ricin compared to free aptamers. ARMAN showed rapid and effective neutralization ability, significantly increasing cell viability in BEAS-2B, GES-1, and HL7702 cell lines exposed to ricin. In vivo, ARMAN treatment led to a notable prolongation of survival in ricin-poisoned mice, highlighting its potential for both pre- and post-exposure treatment. These findings indicate that ARMAN not only neutralizes ricin effectively but also provides a therapeutic window for treatment. Conclusions : ARMAN's superior binding affinity, serum stability, biocompatibility, and broad therapeutic efficacy make it a promising new antidote against ricin poisoning. This study's findings represent significant progress in the development of rapid-response antidotes, with ARMAN offering a potential solution for both military and civilian emergency response scenarios.

Protein arginine methyltransferase 5 (PRMT5) is a critical oncogenic factor in various cancers, and its inhibition has shown promise in suppressing tumor growth. However, the role of PRMT5 in squamous cell carcinoma (SCC) remains largely unexplored. In this study, we analyzed SCC patient data from The Cancer Genome Atlas (TCGA) and the Cancer Dependency Map (DepMap) to investigate the relationship between PRMT5 and SCC proliferation. We employed competition-based cell proliferation assays, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assays, flow cytometry, and in vivo mouse modeling to examine the regulatory roles of PRMT5 and its binding partner WDR77 (WD repeat domain 77). We identified downstream targets, including the p63 isoform ΔNp63α and the cyclin-dependent kinase inhibitor p21, through single-cell RNA-seq, RT-qPCR, and Western blot analyses. Our findings demonstrate that upregulation of PRMT5 and WDR77 correlates with the poor survival of head and neck squamous cell carcinoma (HNSCC) patients. PRMT5/WDR77 regulates the HNSCC-specific transcriptome and facilitates SCC proliferation by promoting cell cycle progression. The PRMT5 and WDR77 stabilize the ΔNp63α Protein, which in turn, inhibits p21. Moreover, depletion of PRMT5 and WDR77 repress SCC in vivo. This study reveals for the first time that PRMT5 and WDR77 synergize to promote SCC proliferation via the ΔNp63α-p21 axis, highlighting a novel therapeutic target for SCC.

The structure-tissue exposure/selectivity relationship (STR) aids in lead optimization to improve drug candidate selection and balance clinical dose, efficacy, and toxicity. In this work, butyrocholinesterase (BuChE)-targeted cannabidiol (CBD) carbamates were used to study the STR in correlation with observed efficacy/toxicity. CBD carbamates with similar structures and same molecular target showed similar/different pharmacokinetics. L2 and L4 had almost same plasma exposure, which was not correlated with their exposure in the brain, while tissue exposure/selectivity was correlated with efficacy/safety. Structural modifications of CBD carbamates not only changed drug plasma exposure, but also altered drug tissue exposure/selectivity. The secondary amine of carbamate can be metabolized into CBD, while the tertiary amine is more stable. Absorption, distribution, metabolism, excretion, and toxicity (ADMET) parameters can be used to predict STR. Therefore, STR can alter drug tissue exposure/selectivity in normal tissues, impacting efficacy/toxicity. The drug optimization process should balance the structure-activity relationship (SAR) and STR of drug candidates for improving clinical trials.

Idiopathic pulmonary fibrosis (IPF) is a chronic, progressive lung disease with limited effective therapies. Interstitial macrophages (IMs), especially those derived from monocytes, play an unknown role in IPF pathogenesis. By using single-cell RNA sequencing (scRNA-seq), bleomycin (BLM)-induced pulmonary fibrosis mouse lungs were analyzed to characterize the cellular landscape and heterogeneity of macrophages in this model. scRNA-seq was used to identify distinct interstitial macrophage subpopulations in fibrotic lungs, with monocyte-derived macrophages exhibiting a pro-fibrotic gene expression profile enriched in wound healing, extracellular matrix (ECM) remodeling, and pro-fibrotic cytokine production functions. A pseudotime analysis revealed that IMs originated from monocytes and differentiated along a specific trajectory. A cell-cell communication analysis demonstrated strong interactions between monocyte-derived interstitial macrophages (Mo-IMs) and fibroblasts through the transforming growth factor beta (TGFβ), secreted phosphoprotein 1 (SPP1), and platelet-derived growth factor (PDGF) signaling pathways. Flow cytometry validated the presence and expansion of Mo-IMs subpopulations in BLM-treated mice. This study reveals the cellular heterogeneity and developmental trajectory of lung macrophages in early BLM-induced pulmonary fibrosis, highlighting the crucial role of Mo-IMs with a pro-fibrotic phenotype in IPF pathogenesis via interactions with fibroblasts. Targeting these specific macrophage subpopulations and associated signaling pathways may provide novel therapeutic strategies for IPF.

In response to the problem of poor detection ability of object detection models for small-scale targets in intelligent transportation scenarios, a fusion method is proposed to enhance the features of small-scale targets, starting from feature utilization and fusion methods. The algorithm is based on the YOLOv4 tiny framework and enhances the utilization of shallow and mid-level features on the basis of Feature Pyramid Network (FPN), improving the detection accuracy of small and medium-sized targets. In view of the problem that the background of the intelligent traffic scene image is cluttered, and there is more redundant information, the Convolutional Block Attention Module (CBAM) is used to improve the attention of the model to the traffic target. To address the problem of data imbalance and prior bounding box adaptation in custom traffic data sets that expand traffic images in COCO and VOC, we propose a Copy-Paste method with an improved generation method and a K-means algorithm with improved distance measurement to enhance the model's detection ability for corresponding categories. Comparative experiments were conducted on a customized 260-thousand traffic data set containing public traffic images, and the results showed that compared to YOLOv4 tiny, the proposed algorithm improved mAP by 4.9% while still ensuring the real-time performance of the model.

This study addresses the challenges of low accuracy and high computational demands in Tibetan speech recognition by investigating the application of end-to-end networks. We propose a decoding strategy that integrates Connectionist Temporal Classification (CTC) and Attention mechanisms, capitalizing on the benefits of automatic alignment and attention weight extraction. The Conformer architecture is utilized as the encoder, leading to the development of the Conformer-CTC/Attention model. This model first extracts global features from the speech signal using the Conformer, followed by joint decoding of these features through CTC and Attention mechanisms. To mitigate convergence issues during training, particularly with longer input feature sequences, we introduce a Probabilistic Sparse Attention mechanism within the joint CTC/Attention framework. Additionally, we implement a maximum entropy optimization algorithm for CTC, effectively addressing challenges such as increased path counts, spike distributions, and local optima during training. We designate the proposed method as the MaxEnt-Optimized Probabilistic Sparse Attention Conformer-CTC/Attention Model (MPSA-Conformer-CTC/Attention). Experimental results indicate that our improved model achieves a word error rate reduction of 10.68% and 9.57% on self-constructed and open-source Tibetan datasets, respectively, compared to the baseline model. Furthermore, the enhanced model not only reduces memory consumption and training time but also improves generalization capability and accuracy.

Rutin is a flavonoid compound with potential for treating Alzheimer's disease, preventing brain damage, mitigating cerebral ischemia-reperfusion injury, and exhibiting anti-glioblastoma activity. However, its efficacy is limited by its low solubility, poor bioavailability, and limited permeability across the blood-brain barrier (BBB). To enhance the bioavailability and brain-targeting ability of Rutin, transferrin-modified Rutin liposome (Tf-Rutin-Lip) was developed using liposomes as a delivery system. Rutin liposomes were prepared using the thin-film dispersion method, and the preparation conditions were optimized using the response surface methodology. Then, transferrin (Tf) was incorporated into the liposomes through covalent modification, yielding Tf-Rutin liposomes. The toxicity of these liposomes on bEnd.3 cells, as well as their impact on the tight junctions of these cells, was rigorously evaluated. Additionally, in vitro and in vivo experiments were conducted to validate the brain-targeting efficacy of the Tf-Rutin liposomes. A susceptible detection method was developed to characterize the pharmacokinetics of Tf-Rutin-Lip further. The optimized conditions for the preparation of Tf-Rutin-Lip were determined as follows: a lipid-to-cholesterol ratio of 4.63:1, a drug-to-lipid ratio of 1:45.84, a preparation temperature of 42.7 °C, a hydration volume of 20 mL, a sonication time of 10 min, a surfactant concentration of 80 mg/mL, a DSPE-MPEG-2000 concentration of 5%, and a DSPE-PEG2000-COOH to DSPE-MPEG-2000 molar ratio of 10%. The liposomes did not affect the cell activity of bEnd.3 cells at 24 h and did not disrupt the tight junction of the blood-brain barrier. Tf-modified liposomes were taken up by bEnd.3 cells, which, in turn, passed through the BBB, thus improving liposomal brain targeting. Furthermore, the results of pharmacokinetic experiments showed that the C max , AUC 0-∞ , AUC 0-t , MRT 0-∞ , and t 1/2 of Tf-Rutin-Lip increased 1.99-fold, 2.77-fold, 2.58-fold, 1.26-fold, and 1.19-fold compared to those of free Rutin solution, respectively. These findings suggest that Tf-Rutin-Lip is brain-targeted and may enhance the efficacy of Rutin in the treatment of brain disorders.

AerMet 100 steel is a new type of double-hardened high-strength steel, which is often used as landing gear material in amphibious aircraft. In the present paper, the corrosion fatigue behavior and mechanism of AerMet 100 high-strength steel in a 3.5% NaCl solution was studied by stress-controlled fatigue tests and a series of subsequent characterizations of the fracture surface, microstructure, and cracks. The results indicated that the fatigue life of AerMet 100 high-strength steel decreased with a decrease in the stress level in a 3.5% NaCl solution, satisfying the relationship lgN = -2.69 × 10 -3 σ + 6.49. The corrosion fatigue crack usually initiated from the corrosion pit and propagated across the martensitic flat noodles. Meanwhile, the corrosion fatigue crack tip was filled with Cr 2 O 3 , Fe 2 O 3 , and amorphous material; it propagated in the transgranular mode by a slip dissolution mechanism. This study provides some engineering significant for the fatigue performance of AerMet 100 steel in a 3.5% NaCl solution.

This paper introduces a novel capacitive sensor array designed for tactile perception applications. Utilizing an all-in-one inkjet deposition printing process, the sensor array exhibited exceptional flexibility and accuracy. With a resolution of up to 32.7 dpi, the sensor array was capable of capturing the fine details of touch inputs, making it suitable for applications requiring high spatial resolution. The design incorporates two multiplexers to achieve a scanning rate of 100 Hz, ensuring the rapid and responsive data acquisition that is essential for real-time feedback in interactive applications, such as gesture recognition and haptic interfaces. To evaluate the performance of the capacitive sensor array, an experiment that involved handwritten number recognition was conducted. The results demonstrated that the sensor accurately captured fingertip inputs with a high precision. When combined with an Auxiliary Classifier Generative Adversarial Network (ACGAN) algorithm, the sensor system achieved a recognition accuracy of 98% for various handwritten numbers from "0" to "9". These results show the potential of the capacitive sensor array for advanced human-computer interaction applications.

Silene viscidula Franch is mainly produced in southwest China. The region has a vast area and rich climate, which has an impact on the quality of the plants due to the differences in distribution between the origins. There is a lack of systematic research on its chemical compounds in the existing literature, and fewer studies have been reported for the active compounds of this plant. Therefore, high-resolution liquid mass spectrometry was used in this study. Sixty batches of Silene viscidula Franch samples from twenty origins in three provinces were analyzed for compounds. A database of chemical compounds of Silene viscidula Franch was established through node-to-node information in the GNPS molecular network, as well as literature records. The ion fragmentation information obtained was compared with the literature data and analyzed and identified by importing the mass spectrometry software PeakView 1.2. Then, the MarkerView t -test was applied to analyze and identify the compounds of Silene viscidula Franch from different origins. Afterwards, the antioxidant activity of Silene viscidula Franch from different origins was preliminarily evaluated using DPPH and ABTS free radical scavenging assays. The results showed a total of 78 compounds, including 34 steroids, 14 triterpenoid saponins, 30 flavonoid glycosides, and other classes of compounds, such as alkaloids. The cleavage patterns of steroids, triterpenoid saponins, and flavonoids in positive-ion mode were also summarized. Based on the p -value of the t -test ( p < 0.05), 29 differential compounds were screened out. The relative contents of saponins and steroidal compounds in these samples were found to be associated with antioxidant activity. This study provided a preliminary reference for the establishment of a comprehensive evaluation system for the quality of Silene viscidula Franch.

The integrated stress response, especially stress granules (SGs), contributes to host immunity. Typical G3BP1 + stress granules (tSGs) are usually formed after virus infection to restrain viral replication and stimulate innate immunity. Recently, several SG-like foci or atypical SGs (aSGs) with proviral function have been found during viral infection. We have shown that the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) nucleocapsid (N) protein induces atypical N + /G3BP1 + foci (N + foci), leading to the inhibition of host immunity and facilitation of viral infection. However, the precise mechanism has not been well clarified yet. In this study, we showed that the SARS-CoV-2 N (SARS2-N) protein inhibits dsRNA-induced growth arrest and DNA damage-inducible 34 (GADD34) expression. Mechanistically, the SARS2-N protein promotes the interaction between GADD34 mRNA and G3BP1, sequestering GADD34 mRNA into the N + foci. Importantly, we found that GADD34 participates in IRF3 nuclear translocation through its KVRF motif and promotes the transcription of downstream interferon genes. The suppression of GADD34 expression by the SARS2-N protein impairs the nuclear localization of IRF3 and compromises the host's innate immune response, which facilitates viral replication. Taking these findings together, our study revealed a novel mechanism by which the SARS2-N protein antagonized the GADD34-mediated innate immune pathway via induction of N + foci. We think this is a critical strategy for viral pathogenesis and has potential therapeutic implications.

Linear chlorinated phosphazene acid is prepared using PCl 5 and NH 4 Cl as raw materials. Using hexaethylcyclotrisiloxane as the monomer, 1,1,3,3-tetramethyldisiloxane or 1,3-divinyl-1,1,3,3-tetramethyldisiloxane as the end-capping agent, and linear chlorinated phosphazene acid as the catalyst, polydiethylsiloxane oligomers terminated with active Si-H or Si-CH=CH 2 groups have been prepared. Using hexaethylcyclotrisiloxane and 1,3,5,7-octamethylcyclotetrasiloxane as comonomers, 1,1,3,3-tetramethyldisiloxane or hexamethyldisiloxane as the end-capping agent, or using hexaethylcyclotrisiloxane and octamethylcyclotetrasiloxane as comonomers, 1,1,3,3-tetramethyldisiloxane as the end-capping agent, copolymers containing active Si-H bonds and dimethylsiloxane segments have been prepared under the catalysis of linear chlorinated phosphazene acid. The effects of catalyst dosage, reaction temperature, reaction time, end-capping agent, and polymerization monomer dosage on polymer yield and structure were investigated. Using 300 ppm of linear chlorinated phosphazene acid, oligomers and copolymers containing an active Si-H bond and diethylsiloxane segment were prepared under mild conditions. The molecular weight of the obtained polymers was close to their designed values, but their PDI values were small. The highest yield of α , ω -bisdimethylsiloxyl-terminated PDES oligomers reached 93%. Using oligomers and copolymers containing Si-H bonds and diethylsiloxane segments as crosslinkers, a silicone gel containing diethylsiloxane segments was prepared by hydrosilylation reaction. With the introduction of a diethylsiloxane segment, the glass transition temperature of the silicone gel decreased relative to that of the PDMS oligomer, but the temperature at 5% weight loss in nitrogen atmosphere decreased from 347 °C to 312 °C. The mechanism of the ring-opening polymerization of hexaethylcyclotrisiloxane catalyzed by linear chlorinated phosphazene acid is also discussed.

Electron backscattered diffraction (EBSD) characterization was conducted on the typical regions in friction-stir-welded dissimilar Al/Mg joints of 2 mm thick sheets with/without ultrasonic assistance. The effects of ultrasonic vibration (UV) on the grain size, recrystallization mechanisms, and degree of recrystallization on both sides of the Al-Mg bonding interface and the intermetallic compounds (IMCs) were investigated. It was found that on the Mg side of the weld nugget zone (WNZ), the primary dynamic recrystallization (DRX) mechanisms were discontinuous dynamic recrystallization (DDRX) and continuous dynamic recrystallization (CDRX), with geometric dynamic recrystallization (GDRX) playing a secondary role. On the Al side of the WNZ, CDRX was identified as the primary mechanism, with GDRX as a secondary contributor. While UV did not significantly alter the DRX mechanisms in either alloy within the WNZ, it promoted the aggregation and rearrangement of dislocations. This led to an increase in high-angle grain boundaries (HAGBs) and an enhanced degree of recrystallization in the welds. The average grain size in both the Al and Mg alloys of the WNZ followed a pattern of initially increasing and then decreasing along the thickness direction, reaching a maximum in the upper-middle part and a minimum at the bottom. The influence of UV on the average grain size in the WNZ was minimal, with only slight grain refinement observed, and the minimum refinement degree was only 0.9%. The Schmid factor (SF) on the WNZ and thermo-mechanically affected zone (TMAZ) boundary regions of the advancing side (AS) indicates that the application of UV increased the likelihood of basal slip and extension twinning in the crystal structure. In addition, UV reduced the thickness of IMCs and improved the strength of the Al-Mg bonding interface. These results suggest a higher probability of fracture along the TMAZ and WNZ boundary on the AS when UV was applied.

The lateral organ boundaries domain ( LBD ) genes, as the plant-specific transcription factor family, play a crucial role in controlling plant architecture and stress tolerance. However, the functions of AhLBD genes in the peanut plant ( Arachis hypogea L.) remain unclear. In this study, 73 AhLBDs were identified in the peanut plant and divided into three groups by phylogenetic tree analysis. Gene structure and conserved protein motif analysis supported the evolutionary conservation of AhLBDs . Tandem and segment duplications contributed to the expansion of AhLBDs . The evolutionary relationship analysis of LBD gene family between A. hypogaea and four other species indicated that the peanut plant had a close relationship with the soybean plant. AhLBDs played a very important role in response to growth and development as well as abiotic stress. Furthermore, gene expression profiling and real-time quantitative qRT-PCR analysis showed that AhLBD16 , AhLBD33 , AhLBD67 , and AhLBD72 were candidate genes for salt stress, while AhLBD24 , AhLBD33 , AhLBD35 , AhLBD52 , AhLBD67 , and AhLBD71 were candidate genes for drought stress. Our subcellular localization experiment revealed that AhLBD24 , AhLBD33 , AhLBD67 , and AhLBD71 were located in the nucleus. Heterologous overexpression of AhLBD33 and AhLBD67 in Arabidopsis significantly enhanced tolerance to salt stress. Our results provide a theoretical basis and candidate genes for studying the molecular mechanism for abiotic stress in the peanut plant.
