Preventing eutrophication requires a deep understanding of nutrient sources and migration processes. The Guanshan River in the Danjiangkou Reservoir was selected as a typical agricultural basin. River water, sediment, and soil samples were collected to determine nitrogen (N) and phosphorus (P) contents and forms. Multivariate statistical analysis, buffer analysis, and extreme gradient boosting regression (XGBoost) were used to investigate the nutrient sources, the sources' scale effects, and migration. The results showed that the exogenous sources of river nutrients were fertilizer (provided phosphate (PO4-P) and ammonium (NH4-N)), domestic wastewater (provided nitrate (NO3-N) and organic nitrogen), and natural soil and rock leaching (provided PO4-P and organic phosphorus). Fertilization within 300–1200 m and domestic wastewater discharge within 150 m of the river affected the contents of the river nutrients significantly (with R ranging between 0.40 and 0.73, p ≤ 0.01). The sediment was the N source and P sink of the overlying water. With NH4-N/PO4-P compound fertilization, the NO3-N and calcium-bound phosphorus (Ca-P) exhibited co-enrichment. Appropriately increasing NH4-N application could stimulate P biological uptake, thus inhibiting P emigration from agricultural soil under N-limited states. In conclusion, this study effectively recognized river nutrient sources and their scale impacts and also explored more effective fertilization strategies, which are beneficial for the optimized management of agricultural activities. Elymus sibiricus plays a crucial role in ecological protection and animal husbandry. However, after many years of growth, the biomass of E. sibiricus decreases, and the plants degrade. Moreover, there is no good solution to the problem of degradation of Elymus sibiricus; the addition of nitrogen (N) and phosphorus (P) fertilizers is the primary measure of cultivation management to improve yield, so it is crucial to find the appropriate level of fertilization. This study performed a two-factor split-plot experiment, including four levels of N (0, 45, 60, and 75 kg·hm−2) and four levels of P (0, 60, 75, and 90 kg·hm−2), to investigate the effect of N and P fertilizers on yield, yield components, and photosynthesis characteristics of E. sibiricus. The results showed that the forage yield in 2017 was higher than in 2018. The forage yield in 2017 was highest at N75P0 with a value of 29,926 kg·hm−2, and in 2018 it was highest at N45P0 and N75P0 with a value of 12,266 kg·hm−2 and 12,233 kg·hm−2, respectively, which demonstrates the large impact of year effects on the forage yield. All traits increased with the increase in N and P fertilizer application, but with excess fertilizer application, the photosynthesis was limited, leading to a slowdown in growth and a decrease in yield. In addition, under adequate N fertilization, the role of P fertilization was not significant (p > 0.05). N, P, and N × P can significantly (p < 0.05) affect the yield traits and forage yield of E. sibiricus. According to the PCA, it is clear that N fertilizer has the largest effect, and the growth capacity of degraded E. sibiricus grassland can be restored by adding 75 kg·hm−2 of nitrogen fertilizer. Understanding the optimal extraction methods for flavonoids from Abelmoschus manihot flowers (AMF) is crucial for unlocking their potential benefits. This study aimed to optimize the efficiency of flavonoid extraction from AMF. After comparing extraction methods, the ultrasonic cell crusher demonstrated superior performance over conventional techniques. Four key factors—solid-to-liquid ratio (1:10 to 1:50 g·mL−1), ethanol concentration (55% to 95%), ultrasonic time (10 to 50 min), and ultrasonic power (5% to 25% of 900 W)—were investigated and normalized using the entropy weight method. This led to a comprehensive evaluation (CE). Optimization of extraction conditions for the ultrasonic cell crusher was achieved through response surface methodology and a deep neural network model, resulting in optimal parameters: ethanol volume fraction of 66%, solid-to-liquid ratio of 1:21 g/mL, extraction efficiency of 9%, and extraction duration of 35 min, yielding a CE value of 23.14 (RSD < 1%). Additionally, the inhibitory effects of the optimized extracts against Streptococcus mutans (S. mutans) were assessed. The results revealed that AMF extract (AMFE) exhibits inhibitory effects on S. mutans, with concomitant inhibition of sucrase and lactate dehydrogenase (LDH). The MIC of AMFE against planktonic S. mutans is 3 mg/mL, with an MBC of 6 mg/mL. Within the concentration range of 1/8 MIC to 2 MIC of AMFE, the activities of sucrase and LDH decreased by 318.934 U/mg prot and 61.844 U/mg prot, respectively. The antioxidant activity of AMFE was assessed using the potassium ferricyanide reduction and phosphomolybdenum methods. Additionally, the effect of AMFE on DPPH, ABTS, and ·OH free radical scavenging abilities was determined. Simple Summary: Ketosis (KET), a metabolic disorder frequently observed in dairy cows, is characterized by increased levels of ketone bodies. The "gold standard" for diagnosing ketosis is the concentration of blood β-hydroxybutyrate (BHB). The increasing number of studies focusing on BHB highlights the increasing significance of metabolic disorders in both the dairy industry and the scientific community. Additionally, the surge in research on the genetic and economic facets of the KET is likely correlated with the recent accessibility of extensive datasets containing blood BHB concentrations as routinely recorded traits in certain production systems. Such data are essential to accurately estimating the genetic parameters associated with these traits. Consequently, the objective of this study was to estimate genetic parameters and identify candidate genes related to blood BHB through genome-wide association analysis to provide research directions for dairy ketosis. Ketosis is a common metabolic disorder in the early lactation of dairy cows. It is typically diagnosed by measuring the concentration of β-hydroxybutyrate (BHB) in the blood. This study aimed to estimate the genetic parameters of blood BHB and conducted a genome-wide association study (GWAS) based on the estimated breeding value. Phenotypic data were collected from December 2019 to August 2023, comprising blood BHB concentrations in 45,617 Holstein cows during the three weeks post-calving across seven dairy farms. Genotypic data were obtained using the Neogen Geneseek Genomic Profiler (GGP) Bovine 100 K SNP Chip and GGP Bovine SNP50 v3 (Illumina Inc., San Diego, CA, USA) for genotyping. The estimated heritability and repeatability values for blood BHB levels were 0.167 and 0.175, respectively. The GWAS result detected a total of ten genome-wide significant associations with blood BHB. Significant SNPs were distributed in Bos taurus autosomes (BTA) 2, 6, 9, 11, 13, and 23, with 48 annotated candidate genes. These potential genes included those associated with insulin regulation, such as INSIG2, and those linked to fatty acid metabolism, such as HADHB, HADHA, and PANK2. Enrichment analysis of the candidate genes for blood BHB revealed the molecular functions and biological processes involved in fatty acid and lipid metabolism in dairy cattle. Nitrogen (N) application significantly increases tiller numbers and is accompanied by changes in endogenous hormone content. We treated seedlings of Festuca kirilowii—a perennial forage grass—with nitrogen, determined the endogenous hormone content in the tiller buds, and performed a transcriptome analysis. The application of N reduced GA3, ABA, and 5-DS content and increased ZT and IAA content. By screening DEGs in the transcriptome results, we obtained DEGs annotated to 25 GO entries and 8 KEGG pathways associated with endogenous hormones. Most of these GO entries and KEGG pathways were associated with IAA, GAS, and ABA. We conducted a validation analysis of hormone-related DEGs using qRT-PCR to demonstrate that nitrogen controls the content of endogenous hormones by regulating the expression of these DEGs, which further affects tillering in F. kirilowii. The deep shale of the Wufeng–Longmaxi Formation in the southern Sichuan Basin has high gas potential. The development characteristics of lamination could significantly impact reservoir property. Samples were investigated using microscopic observation, element analysis, organic petrology analysis, mineralogy analysis, and pore structure analysis to determine the types of laminae and laminasets, clarify the formation conditions of argillaceous lamina and silty lamina as well as their relationships with the sedimentary environment, and explore the influence of laminae on shale reservoir property. Results indicate that the Wufeng Formation shale exhibits weak development of laminae due to bioturbation, while the Longmaxi Formation shale develops continuous, parallel, and plate-like laminae. Compared with light silty lamina-rich shale, dark argillaceous lamina-rich shale usually develops in an anoxic reduction environment, with higher total organic carbon content, porosity, pore volume, specific surface area, and more developed organic matter pores, which can provide greater space for shale gas adsorption and storage. Shales in the middle section of the Longmaxi Formation are characterized by the development of silty-argillaceous interbedded type laminaset, which have good reservoir performance, making them the primary target for deep shale gas exploration and development. Flexible anodes are a common form of anode ground bed for the cathodic protection of buried pipes in station areas, especially in new stations. In most cases, flexible anode ground beds could obtain uniform potential distribution and good protection. However, in the process of operation, there are also failure conditions such as anode body or cable breakage, electronic shorting between anode and pipes and other situations, resulting in poor protection. How to troubleshoot failures has become a difficult problem restricting the application of flexible anodes in actual production. In this paper, the failures of a flexible anode cathodic protection system in a station were assessed and analyzed in detail. The main reasons for the failures were the electronic shorting between the flexible anode and buried pipe in local area and the breaking of a partial flexible anode. The troubleshooting methods for two kinds of failures were explored. By measuring the potentials of both the flexible anode and pipes in different areas and the excavation test, the location of electronic shoring was determined. And through measuring the grounding resistance of the flexible anode and excavation test, the breaking location of the flexible anode was found. By repairing the failure sites, the failed system was restored to normal, which could provide guidance for the failure analyses of the flexible anode cathodic protection system. The choice of appropriate proteases and pretreatment methods significantly influences the preparation of bioactive peptides. This study aimed to investigate the effects of different pretreatment methods on the hydrolytic performance of diverse proteases during the production of dipeptidyl peptidase-IV (DPP-IV) inhibitory peptides derived from wheat and their foaming and emulsion properties. Dry heating, aqueous heating, and ultrasound treatment were employed as pretreatments for the protein prior to the enzymatic hydrolysis of wheat gluten. FTIR analysis results indicated that all pretreatment methods altered the secondary structure of the protein; however, the effects of dry heating treatment on the secondary structure content were opposite to those of aqueous heating and ultrasound treatment. Nevertheless, all three methods enhanced the protein solubility and surface hydrophobicity. By using pretreated proteins as substrates, five different types of proteases were employed for DPP-IV inhibitory peptide production. The analysis of the DPP-IV inhibitory activity, degree of hydrolysis, and TCA-soluble peptide content revealed that the specific pretreatments had a promoting or inhibiting effect on DPP-IV inhibitory peptide production depending on the protease used. A new approach is presented in this paper for the dynamic modeling of the chemical and isotopic evolution of C1–3 during the hydrocarbon generation process. Based on systematic data obtained from published papers for the pyrolysis of various hydrocarbon sources (type I kerogen/source rock, type II kerogen/source rock, type III kerogen/source rock, crude oil, and asphalt, etc.), the empirical evolution framework of the chemical and isotopic composition of C1–3 during the hydrocarbon generation process was built. Although the empirical framework was built only by fitting a large amount of pyrolysis data, the chemical and isotopic composition of C1–3 derived from the pyrolysis experiments all follow evolution laws, convincing us that it is applicable to the thermal evolution process of various hydrocarbon sources. Based on the simplified formula of the isotopic composition of mixed natural gas at different maturities (δ13Cmixed), δ13Cmixed = X × n i A × δ 13 C i A + Y × n i B × δ 13 C i B X × n i A + Y × n i B , it can be derived that the cumulative isotopic composition of alkane generated in a certain maturity interval can be expressed by the integral of the product of the instantaneous isotopic composition and instantaneous yield at a certain maturity point, and then divided by the cumulative yield of alkane generated in the corresponding maturity interval. Thus, the cumulative isotopic composition (A(X)), cumulative yield (B(X)), instantaneous isotope (C(X)), and instantaneous yield (D(x)) in the dynamic model, comply with the following formula during the maturity interval of (X0~X). A(X) = ∫ X 0 X C X × D X d x B (X) , where A(X) and B(X) can be obtained by the fitting of pyrolysis data, and D(x) can also be obtained from the derivation of B(X). The dynamic model was applied on the pyrolysis data of Pingliang Shale to illustrate the quantitative evolution of the cumulative yield, instantaneous yield, cumulative isotope, and instantaneous isotope of C1–3 with increasing maturity. The dynamic model can quantify the yield of methane, ethane, and propane, as well as δ13C1, δ13C2, and δ13C3, respectively, during the hydrocarbon generation process. This model is of great significance for evaluating the natural gas resources of hydrocarbon source rock of different maturities and for identifying the origin and evolutionary process of hydrocarbons by chemical and isotopic data. Moreover, this model provides an approach to study the dynamic evolution of the isotope series of C1–3 (including reversed isotopic series), which is promising for revealing the mechanism responsible for isotopic reversal when combined with post-generation studies. Interactive depositional systems of marine carbonates and gypsum salt rocks are closely related to natural gas reservoirs. Despite continuous progress in the exploration of new areas of marine carbonate genesis within the Ordos Basin, the source and mechanism of "sub-salt" natural gas genesis remains controversial. In this study, we investigated natural gas genesis through geochemical analysis of Lower Paleozoic natural gas samples from the mid-eastern Ordos Basin, obtaining natural gas composition data and carbon/hydrogen isotope compositions. We found evident differences between the geochemical characteristics of "sub-salt" and "post-salt" natural gas; the methane carbon isotope signature of "sub-salt" natural gas was lighter overall than that of "post-salt" natural gas, while the ethane carbon isotope composition of the former was more widely distributed and partially lighter than that of the latter. Combining these data with the regional geological background and existing geochemical data, it is evident that Ordovician "post-salt" natural gas comprises a composite of Upper Paleozoic coal-type gas and Lower Paleozoic oil-type gas, with the oil-type gas accounting for the largest proportion. In contrast, the "sub-salt" natural gas was formed and preserved within the Ordovician marine carbonates or sourced from deeper and more ancient hydrocarbon source rocks. Geochemical anomalies, including light methane carbon isotopes and ethane carbon isotopes with coal-type gas characteristics, are closely related to the prevalence of thermochemical sulfate reduction during hydrocarbon formation and reservoir formation of natural gas in "sub-salt" strata. Epichloë endophytes not only affect the growth and resistance of their host plants but also confer nutrient benefits to parasitized hosts. In this study, we used Pedicularis kansuensis to parasitize Stipa purpurea, both with and without endophytic fungi, and to establish a parasitic system. In this study, endophytic fungal infection was found to increase the dry weight of the leaf, stem, and leaf sheath, as well as the plant height, root length, tiller number, aboveground biomass, and underground biomass of S. purpurea under root hemiparasitic stress. Meanwhile, the 13C allocation of the leaf sheaths and roots of S. purpurea increased as the density of P. kansuensis increased, while the 13C allocation of the leaf sheaths and roots of E+ S. purpurea was lower than that of E− S. purpurea. The 13C allocation of the stem, leaf sheath, and root of E+ S. purpurea was higher than that of its E− counterpart. Furthermore, the content of photosynthetic 13C and the 13C partition rate of the stems, leaves, roots, and entire plant of S. purpurea and P. kansuensis transferred from S. purpurea increased as the density of P. kansuensis increased. Aimed at discovering small molecules as anticancer drugs or lead compounds from plants, a lindenane-type sesquiterpene dimer, chlorahololide D, was isolated from Chloranthus holostegius. The literature review showed that there were few reports on the antitumor effects and mechanisms of chlorahololide D. Our biological assay suggested that chlorahololide D blocked the growth and triggered apoptosis of MCF-7 cells by stimulating the reactive oxygen species (ROS) levels and arresting the cell cycle at the G2 stage. Further mechanism exploration suggested that chlorahololide D regulated apoptosis-related proteins Bcl-2 and Bax. Moreover, chlorahololide D inhibited cell migration by regulating the FAK signaling pathway. In the zebrafish xenograft model, chlorahololide D was observed to suppress tumor proliferation and migration significantly. Considering the crucial function of angiogenesis in tumor development, the anti-angiogenesis of chlorahololide D was also investigated. In order to solve the problem of uniform and precise fertilizer discharge, based on experimental analysis of the uneven nature of single-screw fertilizer discharge, a double arc groove screw fertilizer discharger was designed based on the principle of the half-cycle superposition of the fertilizer discharge curve. The fertilizer discharge amount and the instantaneous fertilizer discharge characteristics of the double arc groove screw fertilizer discharger were theoretically analyzed, and the factors affecting the fertilizer discharge uniformity of the double arc groove screw fertilizer discharger were obtained, taking the pitch S, arc groove radius Rp and center distance as the test factors. Using the uniformity variation coefficient and fertilization accuracy as test indexes, the experimental indicators were evaluated through a quadratic universal rotation combination design experiment with three factors and five levels. The optimal parameters were pitch S = 35 mm, arc groove radius Rp = 17 mm and center distance a = 40 mm. The fertilizer discharger was produced based on the optimal parameter combination, and a bench verification test and a comparative test were carried out. The test results show that the uniformity variation coefficient of the bench test and the relative error between the fertilization accuracy and the simulation test are 5.60% and 5.52%, respectively, and there is little difference between them, which verifies the correctness of the simulation. The comparative test results show that the uniformity variation coefficient of the optimized double arc groove screw fertilizer discharger is 7.16%, the fertilization accuracy is 3.44% and the fitting curve equation R2 of fertilizer discharge flow is 0.998, all of which are significantly better than in the single-screw fertilizer discharger. We developed an electronic fertilizer discharge controller and conducted bench verification tests on it. The test results show that the average deviation between the measured fertilizer discharge capacity and the preset value of the double arc groove screw fertilizer discharger based on our self-developed controller is 2.78%. This fertilizer discharge device can precisely control fertilizer discharge, effectively solving the problem of uneven fertilizer discharge in single-screw fertilizer dischargers. Arbuscular mycorrhizal fungi (AMF) have established themselves as pivotal allies in the realm of plant physiology, renowned for their remarkable contributions to augmenting both growth and resilience against environmental stresses. In this study, we embarked on a comprehensive investigation into the discernible impact of two distinct AMF species on a widely planted oat cultivar, 'Qingyan No. 1', when subjected to the austere conditions of a drought. The experimental design included three distinct AMF treatments (inoculation with Rhiaophagus intraradices, Funneliformis mosseae, or not), and the three water treatments were 75% of field capacity (well watered), 50% of field capacity (moderate drought), and 30% of field capacity (severe drought). The obtained results showed that the rate of inoculation under 75% FC for both AMF species was over 74%. Drought stress limited the growth and osmotic regulation of the oat plants. However, AMF inoculation observably increased the above-ground biomass under 75% FC and increased the root biomass under 30% FC. AMF inoculation also increased the root traits under 75% FC and 50% FC. R. intraradices inoculation increased the above-ground soluble sugar and soluble protein concentrations, and both AMF species showed decreased malondialdehyde (MDA) concentrations in the roots. Furthermore, the pervasive influence of drought stress exerted a discernible stranglehold on nutrient uptake in the oat plants, profoundly impacting the distribution of nutrients within the shoots and roots. Regardless of the drought stress treatment, the inoculation with both AMF species increased the P concentrations in the roots and the K and Mg concentrations in the roots, and the inoculation with R. intraradices increased the Ca concentration in the whole oat plant. Under 75% FC, the N concentration of the whole oat plant was significantly reduced by both AMF species. However, under 50% FC and 30% FC, the N concentrations in the shoots inoculated with both AMF species were close to that of the non-inoculated shoots. In summary, AMF improved the osmotic regulation and nutrient absorption and distribution of oat plants under drought stress and thus promoted the growth and biomass accumulation of oat plants. Mammalia cysteamine (2-aminoethanethiol) dioxygenase (ADO) controls the stability of the regulator of G protein signaling 4 (RGS4) through the Cys branch of the Arg/N-degron pathway, thereby affecting the response of the body to hypoxia. However, the oxygen-sensing function of ADO remains unknown in teleost fish. Mandarin fish (Siniperca chuatsi) is one of the most important freshwater economic fishes in China. As the scale of the rearing density continues to increase, hypoxia has become an important factor threatening the growth of mandarin fish. Herein, the molecular characterization, the oxygen-sensing enzyme function, and the role in virus infection of ADO from mandarin fish (scADO) were explored. Bioinformation analysis results showed that scADO had all the molecular foundations for achieving thiol dioxygenase function: three histidine residues coordinated with Fe(II), PCO/ADO domain, and a "jelly roll" β-barrel structure. The expression pattern analysis showed that scAdo was highly expressed in the immune-related tissues, liver, and kidneys and responded to hypoxia on the expression level. Protein degradation experiment results revealed that scADO could lead to the degradation of RGS4 protein through the Cys branch of the Arg/N-degron pathway. Furthermore, the expression levels of scADO responded to fish virus infection. scADO could significantly promote the replication of Siniperca chuatsi rhabdovirus, and this was associated with its thiol dioxygenase activity. These findings not only demonstrate scADO as an oxygen-sensing protein in teleost fish, but are also of considerable importance for clarifying the contribution of the mechanism of hypoxia to the outbreaks of fish viruses. In this study, ultra–high–pressure sterilization (UHPS) of Xinli No. 7 juice (XL7) was explored and optimized. A challenge to implement UHPS in juice as a full alternative to thermal treatment could be represented by the adoption of a pressure level of up to 500 MPa for 20 min at one cycle followed by the packaging in aseptic conditions. It was found that UHPS and HS treatments could effectively kill the microorganisms in XL7 juice but HS treatment would inevitably lose the nutritional quality in the juice, while UHPS treatment could better maintain the glyconic acid content, functional components, and antioxidant activity and reduce Browning degree and improve the stability of XL7 juice. The deterioration rate of UHPS and HS–treated XL7 juice increased with the increased storage temperature. The predicted shelf life of UHPS and HS–treated XL7 juice was 68 and 41 days at 4 °C, respectively. The GRAS protein family is involved in plant growth and development, plant disease resistance, and abiotic stress response. Although the GRAS protein family has been systematically studied and reported in many plants, it has not been reported in oat, an excellent foodstuff crop of Gramineae. We identified 90 AsGRAS genes and all of the AsGRAS genes were randomly distributed on 21 chromosomes with 6 tandem duplicated genes and 49 pairs of segmental duplications, which may be the main reason for the expansion of the GRAS gene family. According to the phylogenetic tree, 90 AsGRASs were classified into 10 distinct subfamilies. Gene structure revealed introns varying from zero to seven, and all genes have conserved motifs and GRAS structure domain. Protein–protein interaction and miRNA prediction analysis showed that AsGRAS proteins mainly interacted with GA signalling, cell division, etc., and that the AsGRAS genes were targeted by miRNA171. RNA-seq and qRT–PCR data showed that GRAS genes were expressed at different growth and developmental stages and under different abiotic stresses in oat, indicating the potential role of GRAS genes in promoting growth and stress tolerance in oat. Overall, our evolutionary and expression analysis of AsGRAS genes contributes to the elucidation of a theoretical basis for the GRAS gene family. Dry eye disease is a chronic disease of the ocular surface characterized by abnormal tear film composition, tear film instability, and ocular surface inflammation, affecting 5% to 50% of the population worldwide. Autoimmune rheumatic diseases (ARDs) are systemic disorders with multi-organ involvement, including the eye, and play a significant role in dry eye. To date, most studies have focused on Sjögren's syndrome (one of the ARDs) since it manifests as two of the most common symptoms–dry eyes and a dry mouth-and attracts physicians to explore the relationship between dry eye and ARDs. Many patients complained of dry eye related symptoms before they were diagnosed with ARDs, and ocular surface malaise is a sensitive indicator of the severity of ARDs. In addition, ARD related dry eye is also associated with some retinal diseases directly or indirectly, which are described in this review. This review also summarizes the incidence, epidemiological characteristics, pathogenesis, and accompanying ocular lesions of ARD's related dry eye, emphasizing the potential role of dry eye in recognition and monitoring among ARDs patients. Bioinformation analysis showed that scPHD2 had the conserved prolyl 4-hydroxylase alpha subunit homolog domains at its C-terminal and was more closely related to other Perciformes PHD2 than other PHD2. Tissue-distribution results revealed that scphd2 gene was expressed in all tissues tested and more highly expressed in blood and liver than in other tested tissues. Dual-luciferase reporter gene and RT-qPCR assays showed that scPHD2 overexpression could significantly inhibit the HIF-1 signaling pathway. Co-immunoprecipitation analysis showed that scPHD2 could interact with scHIF-1α. Protein degradation experiment results suggested that scPHD2 could promote scHIF-1α degradation through the proteasome degradation pathway. This study advances our understanding of how the HIF-1 signaling pathway is regulated by scPHD2 and will help in understanding the molecular mechanisms underlying hypoxia adaptation in teleost fish. With the targets of petroleum exploration transferred to the deep and ancient strata, abundant oil and gas resources have been found in Lower Paleozoic and older strata in central and western China. Due to complex evolutionary processes including multiple episodes of hydrocarbon accumulation and ubiquitously accompanied by secondary alterations, significant uncertainties remain concerning the generation time and accumulation processes of these revealed petroleum sources. In this paper, relative pure Re and Os elements existing in the asphaltene fractions of Lower Cambrian solid bitumen collected from the Guangyuan area, western Sichuan Basin, SW China and Middle–Lower Ordovician heavy oils in the Aiding area of the Tahe oilfield in the Tarim Basin, NW China were successfully obtained by sample pretreatments, and Re–Os isotopic analysis was subsequently carried out for the dating of these. The Re–Os isotopic composition indicates a generation time of Guangyuan bitumen to between 572 Ma and 559 Ma, corresponding to the late Sinian period of the Neoproterozoic era. By the means of Re–Os isochron aging, initial 187Os/188Os ratios, and carbon isotopic compositions, the Lower Cambrian bitumen is supposed to originate from source rocks of the Doushantuo Formation in the Sinian strata and subsequently migrated into the reservoirs of the Dengying Formation. This previously reserved petroleum was transformed into its present bitumen state by the destruction of reservoirs caused by tectonic uplift. The Re–Os dating results of Middle–Lower Ordovician heavy oil of Tarim Basin suggest that it was formed between 450 Ma to 436 Ma, corresponding to the Late Ordovician–Early Silurian system, and the generated petroleum likely migrate into the Middle–Lower Ordovician karst reservoirs to form early oil reservoirs. The SHPB experimental device was adopted to carry out the uniaxial impact tests in studying the failure characteristics of the combination of surrounding rocks and primary support for the tunnel, so as to explore the key factors affecting its dynamic strength and deforming properties. To simplify the practice model of the tunnel construction, the specimens combined by the surrounding rock and primary support were simulated by the concentric annular sandstone and cement mortar in this paper. The results indicate that the thickness of the primary support, the thickness of the surrounding rock and the proportion of the thickness of the primary support are the key factors affecting the dynamic strength of the combination of the surrounding rock and the primary support. The influence of dynamic loads on tunnel construction can be weakened by adjusting the above three factors, so that to improve the supporting capacity. Since the 1950s, major breakthroughs have been made in the field of carbonate oil and gas exploration, and large reservoirs have been found in carbonate strata in many countries; however, the validity of highly evolved carbonate source rocks has been controversial. Because of the loss of organic carbon during acid-solution processing in conventional total organic carbon (TOC) determination, we decided to use a new method of reducing acid-solution losses in order to more effectively quantify acid-soluble organic matter in carbonate rock samples. Different levels of acid-soluble organic matter were present in different types of samples, and there was no positive correlation between the content of acid-soluble organic matter and conventionally measured TOC values. However, the thermal evolutionary maturity of samples with high acid-soluble organic matter content is relatively low, and the high hydrocarbon generation conversion rate of acid-soluble organic matter and conventional TOC in carbonate rocks is an important reason for the low residual TOC in highly evolved carbonate rocks. The new method is helpful in re-evaluating the abundance of organic matter in carbonate rocks and in providing evidence for studying hydrocarbon generation and the hydrocarbon capacity of carbonate source rocks. Background: Elymus L. is the largest genus in the Triticeae tribe. Most species in this genus are highly stress resistant, with excellent forage value. Elymus breviaristatus, a rare species endemic to the Qinghai-Tibet Plateau (QTP), is declining due to habitat fragmentation. However, genetic data for E. breviaristatus are limited, with expressed sequence tag (EST) markers being particularly rare, hampering genetic studies and protection measures. Results: We obtained 9.06 Gb clean sequences from the transcriptome of E. breviaristatus, generating 171,522 unigenes, which were assembled and functionally annotated against five public databases. We identified 30,668 SSRs in the E. breviaristatus transcriptome, from which 103 EST-SSR primer pairs were randomly selected. Of these, 58 pairs of amplified products of the expected size, and 18 of the amplified products were polymorphic. Model-based Bayesian clustering, the unweighted pair group method with arithmetic average (UPGMA), and principal coordinate analysis (PCoA) of 179 wild E. breviaristatus in 12 populations using these EST-SSRs were generally consistent, grouping the 12 populations into two major clades. Analysis of molecular variance (AMOVA) found 70% of the genetic variation among the 12 populations and 30% within the populations, indicating a high level of genetic differentiation (or low gene exchange) among the 12 populations. The transferability of the 58 successful EST-SSR primers to 22 related hexaploid species was 86.2–98.3%. UPGMA analysis generally grouped species with similar genome types together. Conclusions: Here, we developed EST-SSR markers from the transcriptome of E. breviaristatus. The transferability of these markers was evaluated, and the genetic structure and diversity of E. breviaristatus were explored. Our results provide a basis for the conservation and management of this endangered species, and the obtained molecular markers represent valuable resources for the exploration of genetic relationships among species in the Elymus genus. Near-surface energy budget closure has been a trending topic in land surface processes research, especially on the underlying surfaces of heterogeneous wetlands. In this investigation, the horizontal thermal advection caused by thermal inhomogeneity over the alpine wetland is calculated based on the eddy covariance data observed at the Flower Lake observation field and WRF modelling data over the Zoige alpine wetland, China. The contribution of horizontal thermal advection to the near-surface energy closure is analysed. The results show that the mean horizontal heat advection of the Zoige wetland is 20.2 W·m−2, and the maximum value reached 55.0 W·m−2 in the summer of 2017. An anti-reflection coating on a back-illuminated 128 × 128 array Si-based blocked impurity band (BIB) detector in a very-long-wave infrared range was designed in this work. The reflectance and transmittance spectra of ZnS films with different thicknesses on intrinsic Si substrates were studied with a FDTD simulation and experiment. Compared to bare Si substrate, the reflectance of Si coated with 1.5, 2.0, 2.5, and 3.0 μm thick ZnS significantly decreased, while the transmittance increased in the range of 10.0~25.0 μm band. The transmittance enhancement ratio reached approximately 32%, 32%, 28%, and 29%, respectively. It was evidenced that the enhanced transmission at a specific wavelength was caused by the effective interference cancellation effect. Then, a 2.0 μm thick ZnS thin film was deposited on the backside of the 128 × 128 array Si-based BIB detector. The spectral responsivity of the detector increased significantly. Additionally, the blackbody responsivity increased by approximately 36%, suggesting that the ZnS film is an ideal anti-reflection material for VLWIR detectors in the range of 10.0~25.0 μm band. In the application of control charts, most of the research in profile monitoring is based on accurate measurements. Measurement errors, however, often exist in many manufacturing and service environments. In this paper, we apply linear mixed models in the presence of measurement errors in fixed effects. We discuss three modified multivariate charts, namely Hotelling's T 2 , multivariate exponential weighted moving average (MEWMA) control chart, and multivariate cumulative sum (MCUSUM) control chart. Performance comparisons are made in terms of the average run length (ARL) and average extra quadratic loss (AEQL). Graptolite-rich shale is the main layer of shale gas resources in the southern marine sedimentary basin. Recently, shale gas resources were discovered in the Ordovician marine graptolite-rich strata in the Ordos Basin. The graptolite shale in the study area is different from the marine graptolite shale in the Yangtze plate in southern China, and further exploration is needed. This paper presents core samples of the graptolite-rich shale of the Pingliang Formation in the southwest Ordos Basin as research objects. The graptolite genus and graptolite shale characteristics were studied using core observation, electron microscopy, scanning electron microscopy, and geochemical analysis. We determined the role of the sedimentary environment and thermal maturation of graptolite shale in hydrocarbon formation and explored the possibility of hydrocarbon generation. Many graptolite epidermises provide buried organic matter. The quiet sea and low-energy marine environment create favorable conditions for preserving organic matter. The tectonic process resulted in the evolution stage in the oil generation window. Different types of pores formed the spaces of hydrocarbons. Therefore, the shale of the Pingliang Formation has shale oil exploration potential, which complements the shale gas in the northwestern margin of the basin, and provides new venues for shale oil and gas exploration in northern China. Although nitrogen (N) and phosphorus (P) fertilization are important for maintaining the health and productivity of Leymus chinensis steppe, their impact on the abundance and community structure of ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) in this ecosystem is still unclear. We used the amoA gene as a molecular marker to monitor changes in AOA and AOB under low and high N and P fertilization and applied the resin-core technique to measure soil N mineralization. We found that the abundance of AOA based on amoA gene copies, ranging from 7.51 × 107 to 1.23 × 108 copies g−1 soil, was higher than that of AOB. Fertilization, especially high N and P, increased the number of amoA copies associated with AOB. AOA and AOB communities were dominated by Crenarchaeota and Proteobacteria, respectively. Fertilization decreased the relative abundances of Thaumarchaeota in the AOA community and Proteobacteria in the AOB community and increased the relative abundance of Ammonia_oxidizing_bacteria_ensemble. In N and P fertilization treatments, soil N transformation was closely related to AOB but not AOA. Soil ammonium N and nitrate N reshape the structure of ammonia oxidizers of AOB but not those of AOA on L. chinensis steppes. [ABSTRACT FROM AUTHOR]

The WRKY family is widely involved in the regulation of plant growth and stress response and is one of the largest gene families related to plant environmental adaptation. However, no systematic studies on the WRKY family in oat (Avena sativa L.) have been conducted to date. The recently published complete genome sequence of oat enables the systematic analysis of the AsWRKYs. Based on a genome-wide study of oat, we identified 162 AsWRKYs that were unevenly distributed across 21 chromosomes; a phylogenetic tree of WRKY domains divided these genes into three groups (I, II, and III). We also analyzed the gene duplication events and identified a total of 111 gene pairs that showed strong purifying selection during the evolutionary process. Surprisingly, almost all genes evolved after the completion of subgenomic differentiation of hexaploid oat. Further studies on the functional analysis indicated that AsWRKYs were widely involved in various biological processes. Notably, expression patterns of 16 AsWRKY genes revealed that the response of AsWRKYs were affected by stress level and time. The outdoor wind environment and thermal environment are important factors affecting human comfort in cold winter conditions. The spatial layout of plant communities plays an important role in improving the outdoor microclimate and improving outdoor comfort. In order to explore the positive effect of plant layout on outdoor comfort in cold winter, this study took Xuzhou Tangfang Middle School with typical layout characteristics as the research object. In this study, we simulated the wind environment of these models using computational fluid dynamics (CFD) methods and the outdoor thermal environment using Ecotect (2011), and used linear regression and one-way ANOVA for mathematical statistics. The wind environment and Universal Thermal Climate Index (UTCI) of campus outdoor activities distributed in different spaces were analyzed and evaluated. The research results showed that the superposition of wind and thermal environments identified the key areas of the campus (cross-flow area and corner flow area) and showed a negative correlation. The staggered layout of the three plant combinations increases the wind prevention efficiency by 39.4%. At the same time, this study established the linkage mechanism of campus plant layout, environmental microclimate, and activity area comfort, which effectively improved outdoor human comfort in cold winter. This research can provide a reference for the remediation and improvement of the comfort of the same type of campus, and also provide data support and reference significance for the research on the outdoor pedestrian environment in winter. For the application of X80 pipelines in Northeast China, it is important to establish the correct cathodic protection (CP) potential. To achieve this, potentiodynamic polarization; electrochemical impedance spectroscopy (EIS); a slow strain rate test (SSRT); and a scanning electron microscopy (SEM) fracture morphology analysis were carried out for an X80 steel gas pipeline at several temperatures in Heilongjiang Province, China. The results show that the hydrogen evolution potential of X80 steel in soil at different temperatures was about −900 mV (vs. CSE). The generated hydrogen atoms can be adsorbed on the surface of the pipelines to reduce the surface energy, or they can be diffused into the substrate and accumulate to the critical concentration, inducing the decohesion between different structures and generating additional plastic deformation through dislocation motion. With the peak impedance potential as the minimum potential and the hydrogen embrittlement potential as the maximum potential, the CP potential of X80 steel in the soil at 30 °C, 45 °C, and 60 °C ranged from −900 mV to −1100 mV (vs. CSE), temperatures at which the X80 steel does not corrode or cause hydrogen embrittlement. 2519A aluminum alloy thick plate is a promising structural material in the field of military industries, owing to its low density, high tensile strength and excellent ballistic performance. However, the nonuniformly distributed microstructure along the thickness direction of this alloy leads to delamination cracks, which restrict its further application in light armor fields. In order to understand the mechanism of delamination cracking along the thickness direction, the effect of the microstructure on the mechanical properties of 2519A aluminum alloy in the thickness direction was investigated. The results show that the elongation and critical stress intensity factor values (ΔKcr) of the alloy in the thickness direction are 45.8% and 44.1% lower than the values in the rolling direction, respectively. The low mechanical properties of the alloy may be due to the short distance between the second phase, the weak binding force of grain boundaries and the disharmonious deformation caused by the inhomogeneous distribution of the microstructure. Triosteum himalayanum, Triosteum pinnatifidum (Triosteum L., Caprifoliaceae, Dipsacales) are widely distributed in China while Triosteum sinuatum mainly occurrs in northeast China. Few reports have been determined on the genus Triosteum. In the present research, we sequenced 2 chloroplast genomes of Triosteum and analyzed 18 chloroplast genomes, trying to explore the sequence variations and phylogeny of genus Triosteum in the order Dipsacales. The chloroplast genomes of the genus Triosteum ranged from 154,579 bp to 157,178 bp, consisting of 132 genes (86 protein-coding genes, 38 transfer RNA genes, and 8 ribosomal RNA genes). Comparative analyses and phylogenetic analysis supported the division of Dipsacales into two clades, Adoxaceae and six other families. Among the six families, a clade of Valerianaceae+Dipsacaceae was recovered as a sister to a clade of Morinaceae+Linnaeaceae. A closer relationship of T. himalayanum and T. pinnatifidum among three species was revealed. Our research supported that Loniceraferdinandi and Triosteum was closely related. Zabelia had a closer relationship with Linnaea borealis and Dipelta than Morinaceae. The divergence between T. sinuatum and two other species in Triosteum was dated to 13.4 mya. In this work, the wavelength selection characteristics of metal gratings on Si-based blocked-impurity-band (BIB) detectors in the terahertz band were studied by performing experiments and a finite difference time domain (FDTD) simulation. The transmission spectra of metal gratings with different periods on 130 μm intrinsic Si substrates were measured. When the metal grating period increased from 16 to 20 to 32 μm, the peak position of the spectrum moved from 21.71 to 24.50 to 36.59 μm, which is in good agreement with the FDTD simulation results. The structure with the period of 32 μm shows the best wavelength selective transmission characteristics. Then, the bare Si-based BIB devices and metal grating/Si-based BIB hybrid devices with different thicknesses of blocking layers of 2 and 5 μm were fabricated. By covering different periods of metal gratings for the devices with a thicker blocking layer of 2 μm, we obtained more effective wavelength selection characteristics and stronger response spectra enhancement ratios that were about 1.3, 2.4, or 1.9 times. This was mainly due to the localized optical field enhancement effect of the plasmons resonance in metal gratings, which decays exponentially in a vertical direction. Because of their unique properties, antimicrobial peptides (AMPs) represent a potential reservoir of novel anticancer therapeutic agents. However, only a few AMPs can kill tumors with high efficiency, and obtaining inexpensive anticancer AMPs with strong activity is still a challenge. In our previous work, a series of original short amphiphilic triblock AMP (KnFmKn) analogues were developed which were demonstrated to exert excellent effects on bacterial infection, both in vitro and in vivo. Herein, the overall objectives were to assess the potent tumoricidal capacities of these analogues against human lung cancer cell line A549 and the underlying mechanism. The results of the CCK-8 assay revealed that the precise modification of the peptides' primary sequences could modulate their tumoricidal potency. In the tumoricidal progress, positive charge and hydrophobicity were the key driving forces. Among these peptides, K4F6K4 displayed the most remarkable tumoricidal activity. Furthermore, the excellent anticancer capacity of K4F6K4 was proven by the live/dead cell staining, colony formation assay, and tumor growth observations on xenografted mice, which indicated that K4F6K4 might be a promising drug candidate for lung cancer, with no significant adverse effects in vitro or in vivo. In addition, the cell apoptosis assay using flow cytometry, the morphology observations using the optical microscope, confocal microscopy using CellMask™ Deep Red staining, and scanning electron microscope suggested that membrane disruption was the primary mechanism of its antitumor action. Through analyzing the structure–activity relationship, it was found that the amount of positive charge required for KnFmKn to exert its optimal tumoricidal effect was more than that needed for the antimicrobial activity, while the optimal proportion of hydrophobicity was less. Our findings suggest that further analysis of the structure–activity relationship of AMPs' primary sequence variations will be beneficial. Branching is one of the key determinants of plant architecture that dramatically affects crop yield. As alfalfa is the most important forage crop, understanding the genetic basis of branching in this plant can facilitate breeding for a high biomass yield. In this study, we characterized the strigolactone receptor gene MsD14 in alfalfa and demonstrated that MsD14 was predominantly expressed in flowers, roots, and seedpods. Furthermore, we found that MsD14 expression could significantly respond to strigolactone in alfalfa seedlings, and its protein was located in the nucleus, cytoplasm, and cytomembrane. Most importantly, transformation assays demonstrated that silencing of MsD14 in alfalfa resulted in increased shoot branching and forage biomass. Significantly, MsD14 could physically interact with AtMAX2 and MsMAX2 in the presence of strigolactone, suggesting a similarity between MsD14 and AtD14. Poor indoor air quality reduces the comfort experienced in the environment and can also harm our physical health. Mechanical ventilation design plays an important role in improving the indoor environment and the safety of public toilets. Therefore, in this study, we aimed to evaluate public toilet ventilation design schemes through a digital twin to determine the most effective scheme for reducing indoor pollutant concentrations. In this study, we used Autodesk Revit to create a digital twin BIM of different ventilation systems. We simulated the diffusion of pollutants in these models using computational fluid dynamics (CFD)-based methods, and we used DesignBuilder to simulate building energy consumption. From the perspective of architectural design, we determined measures important for reducing the concentration of air pollutants by increasing the number and volume of air exchanges and controlling the installation height of exhaust vents. The results show that the ventilation design of an all-air air conditioning system with an exhaust height of 400 mm can remarkably improve the indoor environmental health and ventilation efficiency of public toilets, while consuming 20.4% less energy and reducing carbon emissions by 30,681 kg CO2. The influences of cold rolling and subsequent heat treatment on the microstructure evolution of 2524 alloy were investigated using an orientation distribution function (ODF) and electron back-scattered diffraction (EBSD). A preparation method of 2524-T3 aluminum alloy with a strong Brass texture was developed, and its effect on the fatigue properties of the alloy was investigated using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The results show that with the increase in cold rolling deformation from 0% to 80%, the volume fractions of Brass, copper, and S textures in the 2524-T3 alloy also increase, especially in the case of Brass and S textures. However, the volume fractions of cube and Goss textures are reduced significantly, especially for cube textures, which are decreased by 57.4%. Reducing coarse second-phase particles (CSPs) is conducive to the formation of a strong deformation texture during cold rolling. A 10% deformation at each rolling pass, followed by a step annealing, helps the preservation of a Brass texture even after solution treatment at 500 °C for 0.5 h, while a large cold deformation followed by high-temperature annealing helps the formation of a strong cube texture. Staphylococcal enterotoxin C2 (SEC2) is a classical superantigen (SAg), which can tremendously activate T lymphocytes at very low dosage, thus exerting its powerful antitumor activity. As an intravenous protein drug and a bacterial toxin, SEC2 has some limitations including poor patient compliance and toxic side effects. In this research, we devoted our attention to studying the antitumor activity and toxicity of SEC2 as a potential oral administration protein drug. We proved that His-tagged SEC2 (SEC2-His) could undergo facilitated transcytosis on human colon adenocarcinoma (Caco-2) cells and SEC2-His was detected in the blood of rats after oral administration. Furthermore, oral SEC2-His caused massive cytokine release and immune cell enrichment around tumor tissue, leading to inhibition of tumor growth in vivo. Meanwhile, although SEC2-His was dosed up to 32 mg/kg in mice, no significant toxicity was observed. These data showed that SEC2 can cross the intestinal epithelium in an immunologically integral form, maintaining antitumor activity but with reduced systemic toxicity. Poa alpigena is a dominant grass species in alpine meadows, which is sensitive to environmental conditions. In this study, we analyzed the characteristics of the anatomical structure of the stems and leaves of Poa alpigena in overgrazed and enclosed conditions in order to determine the dwarfing morphological mechanism associated with overgrazing. The results show that leaf thickness, leaf epidermal thickness, epidermal cell area, and phloem thickness increased with increased grazing intensity (p < 0.05). In contrast, xylem thickness, mesophyll cell area, and guide wall thickness decreased with an increase in grazing intensity (p < 0.05). Mesophyll cell density was relatively unaffected by grazing intensity. Additionally, the plasticity indices of leaf area, upper epidermal cutin layer thickness, and leaf xylem thickness were higher than 0.5. The plasticity indices of stem tube diameter, epidermal cell size, and epidermal cuticle thickness were greater than 0.4. The results of our study indicate that the structural stem and leaf changes in Poa alpigena are induced by the water and mechanical stresses that occur under grazing conditions. Thus, plateau plants adapt to grazing stress by increasing the thickness of their leaves, cuticles, and phloem. The mesophyll cell area, as well as the stem epidermal cell area of Poa alpigena decreased in response to minor variations in grazing intensity, yet overgrazing did not change its density. With its proposal of the "double carbon" (peak carbon dioxide emissions and carbon neutralization) goal, China has entered a new stage in creating an ecological civilization and achieving sustainable development. Based on the formation and evolution mechanism of the carbon emission efficiency network, in this study, a trilateral evolutionary game model—including efficiency groups (high- and low-efficiency groups) and local governments—was constructed, in an attempt to discuss the conditions needed for different players and trilateral interconnected systems to implement balanced and stable strategies. Furthermore, the sensitivity of the participants' evolutionary trajectories toward factors such as the initial strategy ratio, transition cost, and network capital were tested via a system simulation. The main conclusions were as follows: (1) Efficiency groups form a virtuous circle when the initial proportion of the participants' strategies reaches a certain threshold, and converge into a stable "win–win" state. Under these circumstances, high-efficiency groups tend to give full play to their efficiency advantages in terms of carbon emission reduction and green development, while low-efficiency groups tend to choose green transformation and accept the spillover effect from high-efficiency groups. (2) When efficiency groups achieve a "win–win" state or form good self-management, local governments move from active supervision to a passive supervision strategy in order to reduce supervision costs. (3) While different initial strategy proportions do not affect the stable convergence point of the evolutionary system, they have a differentiated impact on the convergence speed of the players. Under the condition of a low initial strategy ratio, transformation costs can reduce the green transformation enthusiasm of inefficient groups, while network capital can enhance the green transformation willingness of inefficient groups. The seed yield of grass species is greatly dependent on inflorescence morphological traits, starting with spikelets per inflorescence and seeds per spikelet, to kernel size, and then to awns. Previous studies have attempted to estimate the contribution of these traits on the harvested yield of major cereal crops, but little information can be accessed on the influence of awns on seed yield of forage grass species. Siberian wildrye (Elymus sibiricus L.) is a widely important perennial forage grass used to increase forage production in arid and semi-arid grasslands. The grass has long inflorescences with long awns developed at the tip end of the lemmas in the florets. In order to evaluate the effect of awns on Siberian wildrye seed production, awn excision analyses from 10 accessions were performed at flowering stage under irrigated and rainfed regimes. Overall, awn excision reduced thousand-seed weight and seed size under both irrigated and rainfed regimes, which decreased final seed yield per plant. De-awned plants produced significantly more seeds per inflorescence, but spikelets per inflorescence was not influenced by awn excision in either condition. Moreover, histological analyses showed a high degradation of the abscission layer in the awned plants than de-awned ones, and awn excision evidently improved average seed breaking tensile strength (BTS), and thus decreased the degree of seed shattering. In conclusion, the observed significant impact of awn excision on different yield-related traits mirrored the impact of awns on the performance of Siberian wildrye under diverse growing conditions. Simple Summary: The formats of bispecific antibody have been investigated for many years to enhance the stability of the structure and anti-tumor efficacy. One of the formats combining two Fabs at their C termini provides unmodified variable region and comparable activity to other fragment-based bispecific antibodies that are usually combined in a head-to-tail manner. However, the current strategy to produce the BiFab molecule is limited to a semisynthetic method that introduces unnatural amino acid to antibodies' sequences during production. To improve the application of BiFab format in investigational biodrugs, we have applied sortase A-mediated "bio-click" chemistry to generate BiFab, for facile assembly of Fab molecules that have been expressed and stored as BiFab module candidates. The BiFabs made by our method stimulate T cell proliferation and activation with favorable in vitro and in vivo anti-tumor activit. Our results indicate that BiFab made by sortase A-mediated click chemistry could be used to efficiently generate various BiFabs with high potency, which further supports personalized tumor immunotherapy in the future. Bispecific antibodies (BsAbs) for T cell engagement have shown great promise in cancer immunotherapy, and their clinical applications have been proven in treating hematological malignance. Bispecific antibody binding fragment (BiFab) represents a promising platform for generating non-Fc bispecific antibodies. However, the generation of BiFab is still challenging, especially by means of chemical conjugation. More conjugation strategies, e.g., enzymatic conjugation and modular BiFab preparation, are needed to improve the robustness and flexibility of BiFab preparation. We successfully used chemo-enzymatic conjugation approach to generate bispecific antibody (i.e., BiFab) with Fabs from full-length antibodies. Paired click handles (e.g., N3 and DBCO) was introduced to the C-terminal LPETG tag of Fabs via sortase A mediated transpeptidation, followed by site-specific conjugation between two click handle-modified Fabs for BiFab generation. Both BiFabCD20/CD3 (EC50 = 0.26 ng/mL) and BiFabHer2/CD3 exhibited superior efficacy in mediating T cells, from either PBMC or ATC, to kill target tumor cell lines while spared antigen-negative tumor cells in vitro. The BiFabCD20/CD3 also efficiently inhibited CD20-positive tumor growth in mouse xenograft model. We have established a facile sortase A-mediated click handle installation to generate homogeneous and functional BiFabs. The exemplary BiFabs against different targets showed superior efficacy in redirecting and activating T cells to specifically kill target tumor cells, demonstrating the robustness of sortase A-mediated "bio-click" chemistry in generating various potent BiFabs. Alkaloids produced by endophytic fungi can have an important influence on agricultural ecology, and can often be affected by climatic factors. At present, there are no studies that have assessed the relationship between alkaloid production and elevation or climatic factors in the Qinghai-Tibetan Plateau. To address this knowledge gap, we explored ergot alkaloid and peramine production in Epichloë-infected (E+) Elymus tangutorum collected from the Qinghai-Tibetan plateau and assessed the relationship between the concentration of these alkaloids and elevation. The effects of temperature and precipitation on these relationships were also investigated. The concentrations of ergonovine, ergine, and peramine ranged from 0.47–0.84, 0.35–1.72 and 9.18–13.00 µg·g−1, respectively. Fitted cubic regression curves describing an arc-pattern across the elevational series were statistically significant for ergine and peramine concentrations. The elevational trend in peramine concentration was linked to mean daily temperature, while the ergine elevational trend was linked to mean precipitation. Our results provide a new understanding of the climatic factors that influence the alkaloid production of endophytic fungi at different elevations in the Qinghai-Tibetan plateau. The effects of meteorological factors on reference evapotranspiration (ET0) are variable on different time scales, although research tends to focus only on certain time scales. Therefore, using the meteorological data from 1958 to 2017 of Beijing, China, ET0 values over the last 60 years were calculated using Penman–Monteith method. The variation in ET0 values was thus analyzed against four meteorological factors over different time scales. The sensitivity of ET0 to these factors was assessed using a sensitivity coefficient, while the contribution of each factor to ET0 change was quantified by combining this sensitivity coefficient with the factor's relative change rate over multiple time scales. The results showed that the sensitivity coefficient of relative humidity over different time scales were all negative, while the sensitivity coefficients of net radiation, temperature and wind speed were mostly positive. The main sensitivity factors of ET0 on different time scales varied. On annual time scales, the main factors were relative humidity and temperature. Over annual time scales, relative humidity and net radiation alternated as the main sensitivity factor; while over interannual time scales, the most sensitive factor was relative humidity during 1958–1979 and net radiation thereafter. The contribution of these four meteorological factors to ET0 also fluctuated greatly on intra-annual time scales. On daily time scales, the contributions of temperature and wind speed at the start and end of the year were large, while net radiation and relative humidity were dominant mid-year. On monthly to seasonal time scales, the contributions of these four meteorological factors to ET0 were notable. The contribution of relative humidity was largest in spring and autumn; net radiation was dominant in summer, while temperature and wind speed were dominant in winter. Lakes on the Qinghai–Tibetan Plateau (QTP) have experienced significant changes, especially the prevailing lake expansion since 2000 in the endorheic basin. The influence of permafrost thawing on lake expansion is significant but rarely considered in previous studies. In this study, based on Landsat images and permafrost field data, the spatial-temporal area changes of lakes of more than 5 km2 in the endorheic basin on the QTP during 2000–2017 is examined and the impact of permafrost degradation on lake expansion is discussed. The main results are that permafrost characteristics and its degradation trend have close relationships with lake changes. Lake expansion in the endorheic basin showed a southwest–northeast transition from shrinking to stable to rapidly expanding, which corresponded well with the permafrost distribution from island-discontinuous to seasonally frozen ground to continuous permafrost. A dramatic lake expansion in continuous permafrost showed significant spatial differences; lakes expanded significantly in northern and eastern continuous permafrost with a higher ground ice content but slightly in southern continuous permafrost with a lower ground ice content. This spatial pattern was mainly attributed to the melting of ground ice in shallow permafrost associated with accelerating permafrost degradation. Whereas, some lakes in the southern zones of island-discontinuous permafrost were shrinking, which was mainly because the extended taliks arising from the intensified permafrost degradation have facilitated surface water and suprapermafrost groundwater discharge to subpermafrost groundwater and thereby drained the lakes. Based on observation and simulated data, the melting of ground ice at shallow depths below the permafrost table accounted for 21.2% of the increase in lake volume from 2000 to 2016. Exploring precipitation threshold from an economic loss perspective is critical for rainstorm and flood disaster risk assessment under climate change. Based on the daily gridded precipitation dataset and direct economic losses (DELs) of rainstorm and flood disasters in the mainland of China, this paper first filtered a relatively reasonable disaster-triggering daily precipitation threshold (DDPT) combination according to the relationship between extreme precipitation days and direct economic loss (DEL) rates at province level and then comprehensively analyzed the spatial landscape of DDPT across China. The results show that (1) the daily precipitation determined by the combination of a 10 mm fixed threshold and 99.3th percentile is recognized as the optimal DDPT of rainstorm and flood disasters, and the correlation coefficient between annual extreme precipitation days and DEL rates reached 0.45 (p < 0.01). (2) The optimal DDPT decreases from southeast (up to 87 mm) to northwest (10 mm) across China, and the DDPTs of 7 out of 31 provinces are lower than 25 mm, while 5 provinces are higher than 50 mm on average. These results suggest that DDPTs exist with large spatial heterogeneity across China, and adopting regional differentiated DDPT is helpful for conducting effective disaster risk analysis. COREX is one of the commercialized smelting reduction ironmaking processes. It mainly includes two reactors, i.e., a (reduction) shaft furnace (SF) and a melter gasifier (MG). In comparison with the conventional blast furnace (BF), the COREX MG is not only equipped with a more complicated top charging system consisting of one gimbal distributor for coal and eight flap distributors for direct reduction iron (DRI), but also the growth mechanism of its burden pile is in a developing phase, rather than that in a fully-developed phase in a BF. Since the distribution of charged burden plays a crucial role in determining the gas flow and thus in achieving a stable operation, it is of considerable importance to investigate the burden distribution influenced by the charging system of COREX MG. In the present work, a mathematical model is developed for predicting the burden distribution in terms of burden layer structure and radial ore/coal ratio within the COREX MG. Based on the burden pile width measured in the previous physical experiments at different ring radii on a horizontal flat surface, a new growth mechanism of burden pile is proposed. The validity of the model is demonstrated by comparing the simulated burden layer structure with the corresponding results obtained by physical experiments. Furthermore, the usefulness of the mathematical model is illustrated by performing a set of simulation cases under various charging matrixes. It is hoped that the model can be used as a what-if tool in practice for the COREX operator to gain a better understanding of burden distribution in the COREX MG. [ABSTRACT FROM AUTHOR]

The effects of the Cu content on the microstructure and strengthening mechanisms of the Al-Mg-Si-xCu alloys were systematically investigated using scanning electron microscopy (SEM), electron probe microanalysis (EPMA), transmission electron microscopy (TEM), and mechanical tensile tests. The results show that, the strengthening mechanisms change with the Cu content. For as-quenched alloys, solution strengthening (σSS) is predominant when the Cu content ≥2.5 wt.%, and of equivalent importance as grain size strengthening (σH-P) when the Cu content ≤1.0 wt.%. With respect to peak-aged alloys, precipitation strengthening (σppt) is predominant when the Cu content ≥2.5 wt.%, but σSS becomes predominant when the Cu content is 4.5 wt.%. As the Cu content increases from 0.5 to 4.5 wt.%, the main type of precipitates in alloy tends to change from a β″ phase to Q′ phase, and then to a θ′ phase. Among the three types of precipitates, θ′-precipitate causes the largest increase in yield strength (σ0.2) and the largest decrease rate in elongation. β″-precipitate leads to the smallest increase in σ0.2 and the smallest decrease rate in elongation. The increase of Cu content reduces Si solubility in the Al matrix and thus decreases the nucleation rate of β″ phase during subsequent aging. [ABSTRACT FROM AUTHOR]
