In this paper, an improved single-phase second order generalized integrator (SOGI) fixed-frequency phase-locked loop (FFPLL) is presented. The proposed improvement comprises the modification of the PLL input signal estimated phase angle correction factor, which is in this paper calculated and implemented with the exactly accurate value, while in the existing literature the approximated correction value is employed. Also, in this paper, the FFPLL with DC offset is presented, together with the corresponding estimated angle correction technique. Furthermore, the PLL with the positive sequence separation is outlined, based on the new FFPLL structure. The proposed technique is analyzed and verified by simulation and experimental runs, which proved the accuracy and efficiency of the proposed PLL technique. Furthermore, a corresponding PLL parameter values tuning procedure is presented that illustrates the dynamic performance improvements that SOGI based FFPLL introduces when compared with SOGI based PLL. Consequently, FFPLL combined with the proposed new estimated angle correction factor represents a significant improvement when compared to the conventional SOGI based PLL. [ABSTRACT FROM AUTHOR]

This paper proposes an auto-tuned constrained state-feedback controller (SFC) to attenuate periodic disturbances present in permanent magnet synchronous (PMSM) motor drives. An online auto-tuning process of SFC has been made using a powerful nature-inspired optimization algorithm—artificial bee colony (ABC)—to achieve high-performance operation of the drive. A novel performance index is proposed to minimize the impact of pulsating torque and obtain smooth-velocity of the drive. The proposed approach is a practical application of classic control theory with novel engineering-tools for improving the operational quality of a PMSM drive system. The obtained results are compared with a classical cascade control structure (CCS) based on proportional-integral (PI) regulators and disturbance observer-based control (DOBC). A detailed time- and frequency-domain analysis has been conducted in respect to periodic disturbances present in a PMSM drive system. The paper describes the cascaded voltage and current control for a bidirectional DC/DC converter in Dual Active Bridge (DAB) topology. The typical DAB converter circuit was extended by additional current filters, which allow it to operate in application fields with high requirements on current ripples. The core concept of the presented solution is usage of the modified Single Phase Shift (SPS) modulation, which allows to compensate for the DC-bias current occurring in dynamic states and provides a settling time of half switching cycle during transients. Its features were utilized to build a simplified dynamic model of the converter. The linear Proportional-Integral (PI) controllers are used in both voltage and current control loops. Based on the developed dynamic models, the tuning rules for both controllers were derived. In both cases, a number of the tuned parameters were reduced from two to one (which can present a great practical value for application engineers). The proposed solutions are validated based on a laboratory prototype. An important part of the experiments was devoted to non-linear effects occurring near the current limitation boundary of the system. This paper deals with a well-known problem of the transient DC-bias current occurring during a phase shift transition in dual active bridge (DAB) DC/DC converters. This phenomenon, if not compensated, can cause damage to the converter or deteriorate its performance. One aim of this paper is to present a solution which allows for the elimination of the undesired transient DC-bias component in current waveforms. This solution is the dual rising edge shift (DRES) compensation algorithm. It provides a very simple implementation and fast settling time within the first half of a switching period. Moreover, the solution is independent on any measurements or system parameter values. It is based on the double-sided single phase shift (DSSPS) modulation, which is described in detail along with a converter model in steady-state. Then, the mechanisms leading to the transient DC-bias are explained, and the compensation algorithm is derived. The performance of the algorithm has been tested using a laboratory prototype. A comprehensive set of tests, involving rapid step changes in power flow and frequency sweep, are provided. Finally, the features of the proposed algorithm are briefly discussed. This paper presents a finite element method (FEM)-based model, which describes the magnetic circuit of the BMW i3 traction machine. The model has been reconstructed based on data available in the public domain. The reader is provided with numerical data regarding flux linkage surfaces in d- and q-axes, as well as with all the information needed to develop a space-vector model of the machine in steady-state, taking into consideration the non-linearity of the magnetic circuit. Hence, the data of a highly-saturated machine from a renowned product are provided, which can serve as a reference design for research. After that, torque curve and partial load operation points are calculated. Finally, the machine model is linearized and the calculations are repeated with the simplified linearized model. The results from both models are then compared with each other. This comparison is intended to assess the magnitude of the expected inaccuracies, when simplified analytical tools are applied to highly-saturated machines (which are the backbone of automotive electrical drivetrains). It is especially important with regard to preliminary design of electrical drivetrains, as at this stage detailed machine geometry and materials are not known. [ABSTRACT FROM AUTHOR]

GDF15, also known as MIC1, is a member of the TGF-beta superfamily. Previous studies reported elevated serum levels of GDF15 in patients with kidney disorder, and its association with kidney disease progression, while other studies identified GDF15 to have protective effects. To investigate the potential protective role of GDF15 on podocytes, we first performed in vitro studies using a Gdf15 -deficient podocyte cell line. The lack of GDF15 intensified puromycin aminonucleoside (PAN)-triggered endoplasmic reticulum stress and induced cell death in cultivated podocytes. This was evidenced by elevated expressions of Xbp1 and ER-associated chaperones, alongside AnnexinV/PI staining and LDH release. Additionally, we subjected mice to nephrotoxic PAN treatment. Our observations revealed a noteworthy increase in both GDF15 expression and secretion subsequent to PAN administration. Gdf15 knockout mice displayed a moderate loss of WT1+ cells (podocytes) in the glomeruli compared to wild-type controls. However, this finding could not be substantiated through digital evaluation. The parameters of kidney function, including serum BUN, creatinine, and albumin-creatinine ratio (ACR), were increased in Gdf15 knockout mice as compared to wild-type mice upon PAN treatment. This was associated with an increase in the number of glomerular macrophages, neutrophils, inflammatory cytokines, and chemokines in Gdf15 -deficient mice. In summary, our findings unveil a novel renoprotective effect of GDF15 during kidney injury and inflammation by promoting podocyte survival and regulating endoplasmic reticulum stress in podocytes, and, subsequently, the infiltration of inflammatory cells via paracrine effects on surrounding glomerular cells.

Periodontal disease in ruminants is common and occurs in farmed and wild animals. Periodontal lesions can result from the secretion of endotoxins by pathogenic bacteria and as consequences of immune system activity. Three main types of periodontitis have been described. The first is chronic inflammation involving mainly premolars and molars-periodontitis (PD). The second type is an acute inflammatory reaction occurring with calcification of the periosteum of the jawbone and swelling of the surrounding soft tissues (Cara inchada, CI-"swollen face"). Finally, a third type, similar to the first but located in the incisor area, is called "broken mouth" (BM). Etiological variation between the different types of periodontitis is indicated. This particularly manifests in the composition of the microbiome, which is characteristic of the different forms of periodontitis. The widespread detection of lesions has drawn attention to the current nature of the problem.

In this study, we investigated the impact of the uremic toxin indoxyl sulfate on macrophages and tubular epithelial cells and its role in modulating the response to lipopolysaccharide (LPS). Indoxyl sulfate accumulates in the blood of patients with chronic kidney disease (CKD) and is a predictor of overall and cardiovascular morbidity/mortality. To simulate the uremic condition, primary macrophages and tubular epithelial cells were incubated with indoxyl sulfate at low concentrations as well as concentrations found in uremic patients, both alone and upon LPS challenge. The results showed that indoxyl sulfate alone induced the release of reactive oxygen species and low-grade inflammation in macrophages. Moreover, combined with LPS (proinflammatory conditions), indoxyl sulfate significantly increased TNF-α, CCL2, and IL-10 release but did not significantly affect the polarization of macrophages. Pre-treatment with indoxyl sulfate following LPS challenge induced the expression of aryl hydrocarbon receptor ( Ahr ) and NADPH oxidase 4 ( Nox4 ) which generate reactive oxygen species (ROS). Further, experiments with tubular epithelial cells revealed that indoxyl sulfate might induce senescence in parenchymal cells and therefore participate in the progression of inflammaging. In conclusion, this study provides evidence that indoxyl sulfate provokes low-grade inflammation, modulates macrophage function, and enhances the inflammatory response associated with LPS. Finally, indoxyl sulfate signaling contributes to the senescence of tubular epithelial cells during injury.

Sleep scoring involves the inspection of multimodal recordings of sleep data to detect potential sleep disorders. Given that symptoms of sleep disorders may be correlated with specific sleep stages, the diagnosis is typically supported by the simultaneous identification of a sleep stage and a sleep disorder. This paper investigates the automatic recognition of sleep stages and disorders from multimodal sensory data (EEG, ECG, and EMG). We propose a new distributed multimodal and multilabel decision-making system (MML-DMS). It comprises several interconnected classifier modules, including deep convolutional neural networks (CNNs) and shallow perceptron neural networks (NNs). Each module works with a different data modality and data label. The flow of information between the MML-DMS modules provides the final identification of the sleep stage and sleep disorder. We show that the fused multilabel and multimodal method improves the diagnostic performance compared to single-label and single-modality approaches. We tested the proposed MML-DMS on the PhysioNet CAP Sleep Database, with VGG16 CNN structures, achieving an average classification accuracy of 94.34% and F1 score of 0.92 for sleep stage detection (six stages) and an average classification accuracy of 99.09% and F1 score of 0.99 for sleep disorder detection (eight disorders). A comparison with related studies indicates that the proposed approach significantly improves upon the existing state-of-the-art approaches.

Articulatory synthesis is one of the approaches used for modeling human speech production. In this study, we propose a model-based algorithm for learning the policy to control the vocal tract of the articulatory synthesizer in a vowel-to-vowel imitation task. Our method does not require external training data, since the policy is learned through interactions with the vocal tract model. To improve the sample efficiency of the learning, we trained the model of speech production dynamics simultaneously with the policy. The policy was trained in a supervised way using predictions of the model of speech production dynamics. To stabilize the training, early stopping was incorporated into the algorithm. Additionally, we extracted acoustic features using an acoustic word embedding (AWE) model. This model was trained to discriminate between different words and to enable compact encoding of acoustics while preserving contextual information of the input. Our preliminary experiments showed that introducing this AWE model was crucial to guide the policy toward a near-optimal solution. The acoustic embeddings, obtained using the proposed approach, were revealed to be useful when applied as inputs to the policy and the model of speech production dynamics.

Sterile inflammation either resolves the initial insult or leads to tissue damage. Kidney ischemia/reperfusion injury (IRI) is associated with neutrophilic infiltration, enhanced production of inflammatory mediators, accumulation of necrotic cells and tissue remodeling. Macrophage-dependent microenvironmental changes orchestrate many features of the immune response and tissue regeneration. The activation status of macrophages is influenced by extracellular signals, the duration and intensity of the stimulation, as well as various regulatory molecules. The role of macrophage-derived monocyte chemoattractant protein-induced protein 1 (MCPIP1), also known as Regnase-1, in kidney ischemia-reperfusion injury (IRI) and recovery from sterile inflammation remains unresolved. In this study, we showed that macrophage-specific Mcpip1 deletion significantly affects the kidney phenotype. Macrophage-specific Mcpip1 transgenic mice displayed enhanced inflammation and loss of the tubular compartment upon IRI. We showed that MCPIP1 modulates sterile inflammation by negative regulation of Irf4 expression and accumulation of IRF4+ cells in the tissue and, consequently, suppresses the post-ischemic kidney immune response. Thus, we identified MCPIP1 as an important molecular sentinel of immune homeostasis in experimental acute kidney injury (AKI) and renal fibrosis.

Based on the results of dynamic probing (DP), time-domain reflectometry (TDR/MUX/MPTS), resistivity cone penetration tests (RCPT), Marchetti dilatometer tests (DMT), and seismic dilatometer tests (SDMT), it is possible to develop a relationship to calculate the relative density ( D r ) and degree of saturation ( S r ) of selected sandy soils. Compiled databases from documented research points for selected sandy soils were used to construct and develop direct correlations between the measured pressures p 0 and p 1 from DMT and shear wave velocity ( V s ) from SDMT, along with pore water pressures ( u 0 ) and atmospheric pressure (Pa). The results allowed us to make a preliminary prediction when evaluating the parameters. Further, they allowed limiting the use of an additional device, especially in the case of multilayer heavy preconsolidated subsoils. Moreover, soil physical and mechanical characteristics (temperature, humidity, pressure, swelling, salinity) measured from TDR/MUX/MPTS (laboratory/field-operated meter for simultaneous measurements of soil moisture, matric potential, temperature, and salinity-bulk electrical conductivity) were assessed. The main achievement of this paper is the original proposal of using a new nomogram chart to determine the relative density and degree of saturation based on DMT and SDMT tests.

The emergence of innovative neurotechnologies in global brain projects has accelerated research and clinical applications of BCIs beyond sensory and motor functions. Both invasive and noninvasive sensors are developed to interface with cognitive functions engaged in thinking, communication, or remembering. The detection of eye movements by a camera offers a particularly attractive external sensor for computer interfaces to monitor, assess, and control these higher brain functions without acquiring signals from the brain. Features of gaze position and pupil dilation can be effectively used to track our attention in healthy mental processes, to enable interaction in disorders of consciousness, or to even predict memory performance in various brain diseases. In this perspective article, we propose the term 'CyberEye' to encompass emerging cognitive applications of eye-tracking interfaces for neuroscience research, clinical practice, and the biomedical industry. As CyberEye technologies continue to develop, we expect BCIs to become less dependent on brain activities, to be less invasive, and to thus be more applicable.

Polylactide and aliphatic polyurethane are biodegradable synthetic polymers which are broadly used as biomaterials in regenerative medicine for implants and scaffolds for tissue engineering. In this paper, the detailed studies of the fabrication of the electrospun fibers of polyurethane/polylactide mixtures were described. The influence of the used solvent (dimethylformamide (DMF)) and diluents (acetone and dichloromethane (DCM)) on the rheological parameters and electrospinning of the described mixtures was examined. Rheological studies showed that polyure-thane/polylactide mixtures have mostly non-Newtonian character, strongly influenced by the diluent. Solutions containing 50 wt.% or more of polyurethane became less viscous after the addition of DCM or acetone, whereas those with bigger amount of polylactide showed higher viscosity after the addition of DCM and lower viscosity after the addition of acetone. Optimized electrospinning process has been elaborated. Fibers with diameters from 250 nm up to 1 µm have been produced and compared. Pure acetone worsened the electrospinning process, but the more DCM was in the mixture, the thinner and more aligned fibers were produced.

Security is one of the major concerns of the Internet of Things (IoT) wireless technologies. LoRaWAN is one of the emerging Low Power Wide Area Networks being developed for IoT applications. The latest LoRaWAN release v.1.1 has provided a security framework that includes data confidentiality protection, data integrity check, device authentication and key management. However, its key management part is only ambiguously defined. In this paper, a complete key management scheme is proposed for LoRaWAN. The scheme addresses key updating, key generation, key backup, and key backward compatibility. The proposed scheme was shown not only to enhance the current LoRaWAN standard, but also to meet the primary design consideration of LoRaWAN, i.e., low power consumption.

Growth differentiation factor 15 (GDF15) is a member of the transforming growth factor-β (TGF-β) cytokine family and an inflammation-associated protein. Here, we investigated the role of GDF15 in murine anti-glomerular basement membrane (GBM) glomerulonephritis. Glomerulonephritis induction in mice induced systemic expression of GDF15. Moreover, we demonstrate the protective effects for GDF15, as GDF15-deficient mice exhibited increased proteinuria with an aggravated crescent formation and mesangial expansion in anti-GBM nephritis. Herein, GDF15 was required for the regulation of T-cell chemotactic chemokines in the kidney. In addition, we found the upregulation of the CXCR3 receptor in activated T-cells in GDF15-deficient mice. These data indicate that CXCL10/CXCR3-dependent-signaling promotes the infiltration of T cells into the organ during acute inflammation controlled by GDF15. Together, these results reveal a novel mechanism limiting the migration of lymphocytes to the site of inflammation during glomerulonephritis.

The cell type-, organ-, and species-specific expression of the pattern-recognition receptors (PRRs) are well described but little is known about the respective expression profiles of their negative regulators. We therefore determined the mRNA expression levels of A20, CYLD, DUBA, ST2, CD180, SIGIRR, TANK, SOCS1, SOCS3, SHIP, IRAK-M, DOK1, DOK2, SHP1, SHP2, TOLLIP, IRF4, SIKE, NLRX1, ERBIN, CENTB1, and Clec4a2 in human and mouse solid organs. Humans and mice displayed significant differences between their respective mRNA expression patterns of these factors. Additionally, we characterized their expression profiles in mononuclear blood cells upon bacterial endotoxin, which showed a consistent induction of A20, SOCS3, IRAK-M, and Clec4a2 in human and murine cells. Furthermore, we studied the expression pattern in transient kidney ischemia-reperfusion injury versus post-ischemic atrophy and fibrosis in mice. A20, CD180, ST2, SOCS1, SOCS3, SHIP, IRAK-M, DOK1, DOK2, IRF4, CENTB1, and Clec4a2 were all induced, albeit at different times of injury and repair. Progressive fibrosis was associated with a persistent induction of these factors. Thus, the organ- and species-specific expression patterns need to be considered in the design and interpretation of studies related to PRR-mediated innate immunity, which seems to be involved in tissue injury, tissue regeneration and in progressive tissue scarring.

The cell type-, organ-, and species-specific expression of the Toll-like receptors (TLRs) are well described, but little is known about the respective expression profiles of their accessory molecules. We therefore determined the mRNA expression levels of LBP, MD2, CD36, CD14, granulin, HMGB1, LL37, GRP94, UNC93b1, TRIL, PRAT4A, AP3B1, AEP and the respective TLRs in human and mouse solid organs. Humans and mice displayed significant differences between their respective mRNA expression patterns of these factors. In addition, the expression profiles in transient tissue inflammation upon renal ischemia-reperfusion injury, in spleens and kidneys from mice with lupus-like systemic autoimmunity, and in progressive tissue fibrosis upon unilateral ureteral obstruction were studied. Several TLR co-factors were specifically regulated during the different phases of these disease entities, suggesting a functional involvement in the disease process. Thus, the organ- and species-specific expression patterns need to be considered in the design and interpretation of studies related to TLR-mediated innate immunity, which seems to be involved in the tissue injury phase, in the phase of tissue regeneration, and in progressive tissue remodelling.

C-type lectin receptors and their adaptor molecules are involved in the recognition of glycosylated self-antigens and pathogens. However, little is known about the species- and organ-specific expression profiles of these molecules. We therefore determined the mRNA expression levels of Dectin-1, MR1, MR2, DC-SIGN, Syk, Card-9, Bcl-10, Malt-1, Src, Dec-205, Galectin-1, Tim-3, Trem-1, and DAP-12 in 11 solid organs of human and mice. Mouse organs revealed lower mRNA levels of most molecules compared to spleen. However, Dec-205 and Galectin-1 in thymus, Src in brain, MR2, Card-9, Bcl-10, Src, and Dec-205 in small intestine, MR2, Bcl-10, Src, Galectin-1 in kidney, and Src and Galectin-1 in muscle were at least 2-fold higher expressed compared to spleen. Human lung, liver and heart expressed higher mRNA levels of most genes compared to spleen. Dectin-1, MR1, Syk and Trem-1 mRNA were strongly up-regulated upon ischemia-reperfusion injury in murine kidney. Tim3, DAP-12, Card-9, DC-SIGN and MR2 were further up-regulated during renal fibrosis. Murine kidney showed higher DAP-12, Syk, Card-9 and Dectin-1 mRNA expression during the progression of lupus nephritis. Thus, the organ-, and species-specific expression of C-type lectin receptors is different between mice and humans which must be considered in the interpretation of related studies.
