Maize, one of the most important cereal crops in Bangladesh, is severely contaminated by fumonisin, a carcinogenic secondary metabolite produced by Fusarium including Fusarium proliferatum. Biocontrol with Bacillus strains is an effective approach to controlling this F. proliferatum as Bacillus has proven antagonistic properties against this fungus. Therefore, the present study aimed to determine how native Bacillus strains can reduce fumonisin in maize cultivated in Bangladesh, where BDISO76MR (Bacillus subtilis) strains showed the highest efficacy both in vitro in detached cob and in planta under field conditions. The BDISO76MR strain could reduce the fumonisin concentration in detached cob at 98.52% over untreated control, by inhibiting the conidia germination and spore formation of F. proliferatum at 61.56% and 77.01%, respectively in vitro. On the other hand, seed treatment with formulated BDISO76MR showed higher efficacy with a reduction of 97.27% fumonisin contamination compared to the in planta cob inoculation (95.45%) over untreated control. The inherent volatility of PV power introduces unpredictability to the power system, necessitating accurate forecasting of power generation. In this study, a machine learning (ML) model based on Gaussian process regression (GPR) for short-term PV power output forecasting is proposed. With its benefits in handling nonlinear relationships, estimating uncertainty, and generating probabilistic forecasts, GPR is an appropriate approach for addressing the problems caused by PV power generation's irregularity. Additionally, Bayesian optimization to identify optimal hyper-parameter combinations for the ML model is utilized. The research leverages solar radiation intensity data collected at 60-min and 30-min intervals over periods of 1 year and 6 months, respectively. Comparative analysis reveals that the data set with 60-min intervals performs slightly better than the 30-min intervals data set. The proposed GPR model, coupled with Bayesian optimization, demonstrates superior performance compared to contemporary ML models and traditional neural network models. This superiority is evident in 98% and 90% improvements in root mean square errors compared to feed-forward neural network and artificial neural network models, respectively. The application of Trichoderma is a form of biological control that has been effective in combating Xanthomonas oryzae pv. oryzae, the causative agent of the devastating disease known as bacterial blight of rice. In this present study, four strains of Trichoderma, viz., T. paraviridescens (BDISOF67), T. erinaceum (BDISOF91), T. asperellum (BDISOF08), and T. asperellum (BDISOF09), were collected from the rice rhizosphere and used to test their potentiality in reducing bacterial blight. The expression patterns of several core defense-related enzymes and genes related to SA and JA pathways were studied to explore the mechanism of induced resistance by those Trichoderma strains. The results primarily indicated that all Trichoderma were significantly efficient in reducing the lesion length of the leaf over rice check variety (IR24) through enhancing the expression of core defense-related enzymes, such as PAL, PPO, CAT, and POD activities by 4.27, 1.77, 3.53, and 1.57-fold, respectively, over control. Moreover, the results of qRT-PCR exhibited an upregulation of genes OsPR1, OsPR10, OsWRKY45, OsWRKY62, OsWRKY71, OsHI-LOX, and OsACS2 after 24 h of inoculation with all tested Trichoderma strains. However, in the case of RT-PCR, no major changes in OsPR1 and OsPR10 expression were observed in plants treated with different Trichoderma strains during different courses of time. Collectively, Trichoderma induced resistance in rice against X. oryzae pv. oryzae by triggering these core defense-related enzymes and genes associated with SA and JA pathways. In Bangladesh, Fusarium proliferatum is a prevalent pathogen of maize plants that poses a significant threat to human health and livestock by producing carcinogenic fumonisin. However, the use of the Bacillus species, which can colonize the infected plant parts, has been practiced globally to mitigate fumonisin contamination in maize. In this study, B. subtilis MMM1, a native isolate from the maize rhizosphere, was found to be the most effective antagonist against F. proliferatum, which reduced their mycelial growth and total fumonisin accumulation by 85.31% and 85.12%, respectively, over control through a fivefold reduction in fungal population (1.1 × 104 CFU/g maize grain) in vitro. Furthermore, out of the five bacilli used in this study, B. subtilis MMM1 was able to increase the root and, shoot length, and the vigor index of maize seedlings, by colonizing the fresh roots at 82 × 106 CFU/g root tissue. This suggests that B. subtilis MMM1 might be put forth both as a biocontrol agent and a plant growth promoter in Bangladesh to combat F. proliferatum. Late blight of potato caused by an oomycete, Phytophthora infestans (Mont.) De Bary limits the production of potato worldwide. Late blight management has been based on chemical fungicide application, and the repeated use of these fungicides introduces new and more aggressive genotypes, which can rapidly overcome host resistance. Therefore, innovative and effective control measures are needed if fungicide use is to be reduced or eliminated. Some potential formulated bacterial bioagents viz. Pseudomonas putida (BDISO64RanP) and Bacillus subtilis (BDISO36ThaR), and fungal bioagents viz. Trichoderma paraviridicens (BDISOF67R) and T. erinaceum (BDISOF91R), were evaluated for their performance in controlling late blight of potato under growth chamber and field conditions. Both artificial inoculation and field experiments revealed that eight sprays of these bacterial (P. putida and B. subtilis) and fungal (T. erinaceum) bioagents were found to be most effective at reducing late blight severity by 99% up until 60 days after planting (DAP), whereas these bioagents were found to be partially effective until 70 DAP, reducing late blight severity by 46 to 60% and 58 to 60% in the field and growth chamber conditions, respectively. However, these bioagents can reduce the spray frequencies of Curzate M8 by 50% (four sprays instead of eight) when applied together with this fungicide. Economic analysis revealed that T6 (eight sprays of formulated P. putida + B. subtilis + four sprays of Curzate M8) and T16 (eight sprays of formulated P. putida, B. subtilis, and T. erinaceum + four sprays of Curzate M8) performed better in consecutive two years, applying less fungicidal spray compared to T1 (eight sprays of Curzate M8 (Positive control)), which indicated that the return ranged, by Bangladeshi Currency (Taka), from 0.85 to 0.90 over the investment of Bangladeshi Currency (Taka) 1.00 in these treatments, and these results together highlight the possibility of using bioagents in reducing late blight of potato under a proper warning system to reduce the application frequency of chemical fungicide. NADPH oxidase (Nox) genes are responsible for Reactive Oxygen Species (ROS) production in living organisms such as plants, animals, and fungi, where ROS exert different functions. ROS are critical for sexual development and cellular differentiation in fungi. In previous publications, two genes encoding thioredoxin and NADH-ubiquinone oxidoreductase involved in maintaining ROS balance were shown to be remarkably induced in a highly versus a weakly aggressive Verticillium dahliae isolate. This suggested a role of these genes in the virulence of this pathogen. NoxA (NADPH oxidase A) was identified in the V. dahliae genome. We compared in vitro expression of NoxA in highly and weakly aggressive isolates of V. dahliae after elicitation with extracts from different potato tissues. NoxA expression was induced more in the weakly than highly aggressive isolate in response to leaf and stem extracts. After inoculation of potato detached leaves with these two V. dahliae isolates, NoxA was drastically up-regulated in the highly versus the weakly aggressive isolate. We generated single gene disruption mutants for NoxA genes. noxa mutants had significantly reduced virulence, indicating important roles in V. dahliae pathogenesis on the potato. This is consistent with a significant reduction of cellophane penetration ability of the mutants compared to the wild type. However, the cell wall integrity was not impaired in the noxa mutants when compared with the wild type. The resistance of noxa mutants to oxidative stress were also similar to the wild type. Complementation of noxa mutants with a full length NoxA clones restored penetration and pathogenic ability of the fungus. Featured Application: Gelatins extracted from the sturgeon head would be applicable in the foods, cosmetics, and biomedical industries. To develop a new method for extracting gelatin from the sturgeon head, the conditions for pretreatment and extraction were optimized. Treatment at 65 °C (3–3.5 h) was enough to separate the head into mixed tissues (skin, scales, pectoral fins, muscle, bones, gills, and small cartilage pieces), skull cartilage, and liquid. From the intensities of α-, β- and γ-bands and yields, the optimized pretreatment conditions for type A and type B gelatin were 0.05 M HCl (1 h) and 0.1 M NaOH (1 h), respectively. The best extraction conditions were: (1) for type A gelatin: with distilled water (DW) (w/v 1:5) at 35 °C, pH 7 when stirring at 200 rpm for 3 h, and (2) for type B gelatin: with DW (w/v 1:5) at 50 °C, pH 8 when stirring at 200 rpm for 1 h. After the decalcification of extracted residues with 0.05 M HCl for 3 h, re-extraction of gelatin was possible. Under the best extraction conditions, yields of type A and B gelatins were 5.01 and 7.25% (dry gelatin weight/wet sample weight), respectively.
