This study introduces a novel paper coating approach using modified zinc oxide (ZnO), providing a comparison with conventional materials used in the paper industry. The research focused on determining the concentration for effective microbial growth inhibition and evaluates the impact of different ZnO types on coated-paper properties, including antimicrobial activity, surface morphology, tensile strength, and water absorption. Specifically, ZnO microparticles (ZnOws), ZnO nanoparticles (ZnOnp), and modified ZnOnp (ZnOnp-CaCO3, with a core–shell structure composed of calcium carbonate [CaCO3] and nano-zinc oxide) were incorporated into coating formulations at varying concentrations (0 × MIC, 1 × MIC, 2 × MIC, and 3 × MIC, based on minimum inhibitory concentrations [MICs]). The results demonstrated that among all tested microorganisms, ZnOnp-CaCO3 showed the lowest MIC values. ZnOnp-CaCO3-coated paper exhibited superior antimicrobial activity against both Gram-positive and Gram-negative bacteria, as well as fungi, outperforming ZnOws and ZnOnp. At 1 × MIC, %inhibition for E. coli, S. aureus, and A. niger were 98.3%, 99.1%, and 90.8%, respectively. Additionally, ZnOnp-CaCO3 coatings caused minimal color change in the paper compared to the other ZnO variants. The coating did not negatively impact the mechanical properties of the paper across all ZnO types and concentrations. Water absorption tests showed increased hydrophobicity with higher ZnO content, with ZnOnp and ZnOnp-CaCO3 exhibiting greater reductions in water absorption than ZnOws. Overall, ZnOnp-CaCO3 showed strong potential as an antimicrobial agent for paper surfaces, making it ideal for packaging and hygiene products. By partially replacing ZnOnp with inexpensive CaCO3 core particles, ZnOnp-CaCO3 delivers enhanced performance, reduced costs, and greater sustainability for large-scale applications. Nowadays, thanks to nanotechnological progress, which itself guides us more and more closely toward not only the efficient design of innovative nanomaterials or nanostructures, but to the improvement of their functionality, we benefit from an important asset in the battle against pathogenic illnesses. Herein, we report a versatile biocompatible plasmonic nanoplatform based on a Whatman paper incorporating positively-charged gold nanospherical particles via the immersion approach. The morphological characterization of the as-engineered-plasmonic paper was examined by SEM (scanning electron microscopy) and HRTEM (high-resolution transmission electron microscopy) investigations, while its surface chemical modification with a synthetic polypeptide, specifically RRWHRWWRR-NH2 (P2), was proved by monitoring the plasmonic response of loaded gold nanospheres and the emission signal of P2 via fluorescence spectroscopy. The as-functionalized plasmonic paper is non-cytotoxic towards BJ fibroblast human cells at bactericidal concentrations. Finally, the antimicrobial activity of the P2-functionalized plasmonic paper on both planktonic bacteria and biofilms was tested against two reference strains: Gram-positive Bacteria, i.e., Staphylococcus aureus and the Gram-negative Bacteria, i.e., Escherichia coli , determining microbial inhibition of up to 100% for planktonic bacteria. In line with the above presented nanoplatform's proper design, followed by their functionalization with active antimicrobial peptides, new roads can be open for determining antibiotic-free treatments against different relevant pathogens.

Non-recyclable fiber rejects from paper mills, particularly those from recycled linerboard mills, contain high levels of structural carbohydrates but are currently landfilled, causing financial and environmental burdens. The aim of this study was to develop efficient and sustainable bioprocess to upcycle these rejects into polyhydroxybutyrate (PHB), a biodegradable alternative to degradation-resistant petroleum-based plastics. To achieve high yields of PHB per unit biomass, the specific objective of the study was to investigate various approaches to enhance the hydrolysis yields of fiber rejects to maximize sugar recovery and evaluate the fermentation performance of these sugars using Escherichia coli LSBJ. The investigated approaches included size reduction, surfactant addition, and a chemical-free hydrothermal pretreatment process. A two-step hydrothermal pretreatment, involving a hot water pretreatment (150 °C and 15% solid loading for 10 min) followed by three cycles of disk refining, was found to be highly effective and resulted in an 83% cellulose conversion during hydrolysis. The hydrolysate obtained from pretreated biomass normally requires a detoxification step to enhance fermentation efficiency. However, the hydrolysate obtained from the pretreated biomass contained minimal to no inhibitory compounds, as indicated by the efficient sugar fermentation and high PHB yields, which were comparable to those from fermenting raw biomass hydrolysate. This research investigated the inhibition of *Escherichia coli* ATCC 25922 (E. coli) bacterial growth in situ, specifically on the stems and aerial parts of *Agastache mexicana* subsp. mexicana (Amm) or "purple toronjil" and on food-grade paper, both contained within Kraft paper bags with a plastic window. The qualitative phytochemical profile of an aqueous extract of Amm revealed the presence of various compounds including alkaloids, coumarins, tannins, flavonoids, saponins, triterpenes, and sterols. The results indicate that these secondary metabolites exhibit a synergistic bactericidal effect, especially when combined with temperature and starvation stress. This was quantified using a decay equation referred to as the bacterial growth inhibition profile of E. coli (BGIPEc). Calculations, which included first derivatives, gradients based on substrate effects and temperature as well as the area under the curve of BGIPEc, demonstrated that higher temperatures led to the greater inhibition of colony forming units (CFUs), further enhanced by the presence of secondary metabolites. Additionally, a shorter half-life corresponded to a faster change rate and a lower area under the curve, indicating a reduced survival rate over time. In the pursuit of enhancing food packaging, nanotechnology, particularly green silver nanoparticles (G-AgNPs), have gained prominence for its remarkable antimicrobial properties with high potential for food shelf-life extension. Our study aims to develop corn starch-based coating materials reinforced with G-AgNPs. The mechanical properties were examined using a uniaxial tensile tester, revealing that starch coated with the highest G-AgNPs concentration (12.75 ppm) exhibited UTS of 87.6 MPa compared to 48.48 MPa of control paper, a significant (p < 0.02) 65% increase. The assessment of the WVP showcased a statistical reduction in permeability by up to 8% with the incorporation of the hydrophobic layer. Furthermore, antibacterial properties were assessed following ISO 22196:2011, demonstrating a strong and concentration-dependent activity of G-AgNPs against E. coli. All samples successfully disintegrated in both simulated environments (soil and seawater), including samples presenting G-AgNPs. In the food trial analysis, the presence of starch and G-AgNPs significantly reduced weight loss after 6 days, with cherry tomatoes decreasing by 8.59% and green grapes by 6.77% only. Escherichia coli (E. coli) detection suffers from slow analysis time and high costs, along with the need for specificity. While state-of-the-art electrochemical biosensors are cost-efficient and easy to implement, their sensitivity and analysis time still require improvement. In this work, we present a paper-based electrochemical biosensor utilizing magnetic core-shell Fe2O3@CdSe/ZnS quantum dots (MQDs) to achieve fast detection, low cost, and high sensitivity. Using electrochemical impedance spectroscopy (EIS) as the detection technique, the biosensor achieved a limit of detection of 2.7 × 102 CFU/mL for E. coli bacteria across a concentration range of 102–108 CFU/mL, with a relative standard deviation (RSD) of 3.5781%. From an optical perspective, as E. coli concentration increased steadily from 104 to 107 CFU/mL, quantum dot fluorescence showed over 60% lifetime quenching. This hybrid biosensor thus provides rapid, highly sensitive E. coli detection with a fast analysis time of 30 min. This study, which combines the detection advantages of electrochemical and optical biosensor systems in a graphite-based paper sensor for the first time, has the potential to meet the needs of point-of-care applications. It is thought that future studies that will aim to examine the performance of the production-optimized, portable, graphite-based sensor system on real food samples, environmental samples, and especially medical clinical samples will be promising. The freshness and safety of fruits and vegetables affect our daily life. Paper products are often used in the packaging and transportation of fruits and vegetables, and these can provide other functions besides packaging after certain modifications and additions. In this study, the AgNPs/1-MCP antibacterial fresh-keeping composite paper was prepared by in-situ loaded silver nanoparticles and spraying 1-MCP solution. Moreover, the prepared paper was used to preserve sweet cherries. It was found that the prepared AgNPs/1-MCP antibacterial fresh-keeping composite paper could effectively inhibit E. coli and S. aureus. When the addition of 1-MCP in the paper was 0.05 g, the fresh-keeping effect on cherries was the best. Under this optimal condition, the weight loss ratio of the cherries was reduced by 1.93%, the firmness was increased by 27.7%, and the soluble solid content was increased by 25%. The preservation time was extended from 4 days to 12 days, three times that of the untreated ones. The prepared fresh-keeping material is environmentally friendly, non-toxic and harmless, simple to prepare and convenient to use, and is expected to become one of the important fresh-keeping methods for fruits. Microbial xylanase has wide application in bioenergy, animal feed, environmental protection, the pulp and paper industry, and agricultural development. In this study, three xylanases from the microbiota of T. fuciformis with its companion strains were identified by metagenomics sequencing. The three enzymes were subjected to cloning and expression in E. coli or P. pastoris, purification, and characterization for their properties. The results showed that AsXyn1, from Annulohypoxylon stygium, among the three enzymes possessed high thermostability at 40 °C and broad pH tolerance in the range of 2.0–10.0, exhibiting its application potential. Furthermore, it was found that post-translational modification (such as glycosylation) of AsXyn1 enzyme modulated its activity, kinetic parameters, and thermostability. The rapid and specific point-of-care (POC) analysis of virulent pathogenic strains plays a key role in ensuring food quality and safety. In this work, a paper-based fluorescent phage biosensor was developed for the detection of the virulent E. coli O157:H7 strain (as the mode of virulent pathogens) in food samples. Firstly, phages that can specifically combine with E. coli O157:H7 (E. coli) were stained with SYTO-13 dye to prepare a novel fluorescent probe (phage@SYTO). Simultaneously, a micro-porous membrane filter with a pore size of 0.45 μm was employed as a paper chip so as to retain the E. coli-phage@SYTO complex (>1.2 μm) on its surface. The phage@SYTO (200 nm in size) was able to pass through the pores of the chip, and the complex could be retained on the paper chip using the free phage@SYTO probes. The E. coli-phage@SYTO could emit a visual fluorescent signal (excited at 365 nm; emitted at 520 nm) onto the chip, which could be detected by a smartphone to reflect the concentration of E. coli. Under optimized conditions, the detection limit was as low as 50 CFU/mL (S/N = 3) and exhibited a wide linear range from 102 to 106 CFU/mL. Biological toxicity testing plays an essential role in identifying the possible negative effects induced by substances such as organic pollutants or heavy metals. As an alternative to conventional methods of toxicity detection, paper-based analytical device (PAD) offers advantages in terms of convenience, quick results, environmental friendliness, and cost-effectiveness. However, detecting the toxicity of both organic pollutants and heavy metals is challenging for a PAD. Here, we show the evaluation of biotoxicity testing for chlorophenols (pentachlorophenol, 2,4-dichlorophenol, and 4-chlorophenol) and heavy metals (Cu2+, Zn2+, and Pb2+) by a resazurin-integrated PAD. The results were achieved by observing the colourimetric response of bacteria (Enterococcus faecalis and Escherichia coli) to resazurin reduction on the PAD. The toxicity responses of E. faecalis-PAD and E. coli-PAD to chlorophenols and heavy metals can be read within 10 min and 40 min, respectively. Compared to the traditional growth inhibition experiments for toxicity measuring which takes at least 3 h, the resazurin-integrated PAD can recognize toxicity differences between studied chlorophenols and between studied heavy metals within 40 min. Guanidinylated chitosan (GCS) was prepared by grafting guanidine oligomers onto chitosan under microwave irradiation. The structure of GCS characterized by FT-IR and 1H NMR verified the covalent bonding between the guanidine oligomers and chitosan; the effects of molar ratio, reaction temperature, and time were investigated and the degree of substitution of GCS reached a maximum of 25.5% under optimized conditions in this work. The resulting GCS showed significantly enhanced antimicrobial activities. The results obtained from the dynamic UV absorption of Escherichia coli (E. coli) and atomic force microscopy (AFM) revealed that the deactivation of E. coli by GCS was due to the destructing of the cell membrane and the prompt release of cytoplasm from the bacterial cells. The adsorption of GCS onto cellulose fibers and the antimicrobial efficiency of the hygiene papers with GCS were also investigated. Microwave irradiation as a green assisted method was applied to promote this reaction. Escherichia coli has been known to cause a variety of infectious diseases. The conventional enzyme-linked immunosorbent assay (ELISA) is a well-known method widely used to diagnose a variety of infectious diseases. This method is expensive and requires considerable time and effort to conduct and complete multiple integral steps. We previously proposed the use of paper-based ELISA to rapidly detect the presence of E. coli. This approach has demonstrated utility for point-of-care (POC) urinary tract infection diagnoses. Paper-based ELISA, while advantageous, still requires the execution of several procedural steps. Here, we discuss the design and experimental implementation of a turntable paper-based device to simplify the paper-based ELISA protocols for the detection of E. coli. In this process, antibodies or reagents are preloaded onto zones of a paper-based device and allowed to dry before use. Here, we aimed to boost antibacterial performance of cellulose fibers for paper sheet application. Therefore, TiO2 nanoparticles have been used with controlled loading onto the surface of the fibers. A simple and facile composite preparation route based on ultrasound and mechanical assisted stirring has been developed. We tested cellulose paper enriched by TiO2 from 1.0 wt% to 8.0 wt%, respectively. Antibacterial performance has been studied against Staphylococcus aureus and Escherichia coli bacteria. Studies showed that all composites exhibit significant capability to reduce living cells of S. aureus and E. coli bacteria at least 60%. A colorimetric polydiacetylene (PDA) paper strip sensor that can specifically recognize Bacillus thuringiensis (BT) HD-73 spores is described in this work. The target-specific aptamer was combined with PDA, and the aptamer-conjugated PDA vesicles were then coated on polyvinylidene fluoride (PVDF) paper strips by a simple solvent evaporation method. The PDA-aptamer paper strips can be used to detect the target without any pre-treatment. Using the paper strip, the presence of BT spores is directly observable by the naked eye based on the unique blue-to-red color transition of the PDA. Quantitative studies using the paper strip were also carried out by analyzing the color transitions of the PDA. The specificity of this PDA sensor was verified with a high concentration of Escherichia coli, and no discernable change was observed. The observable color change in the paper strip occurs in less than 1 h, and the limit of detection is 3 × 107 CFU/mL, much below the level harmful to humans. The PDA-based paper sensor, developed in this work, does not require a separate power or detection device, making the sensor strip highly transportable and suitable for spore analysis anytime and anywhere. Rapid detection of pathogenic bacteria is extremely important for public health and safety. Here, we describe for the first time an integrated origami paper-based analytical device (PAD) incorporating cell lysis, molecular recognition, amplification and visual detection of Escherichia coli (E. coli). The device features three components: paper for its ability to extract protein molecules nonspecifically from cells, DNA superstructures for their ability to immobilize RNA-cleaving DNAzymes (RCDs) but undergo target-induced RNA cleavage on paper, and isothermal rolling circle amplification (RCA) for its ability to amplify each cleavage event into repetitive sequence units that can be detected by naked eye. This device can achieve detection of E. coli K12 with a detection limit of as low as 103 CFU·mL−1 in a total turnaround time of 35 min. Furthermore, this device allowed the sensitive detection of E. coli in complex sample matrices such as juice and milk. Given that more specific RCDs can be evolved for diverse bacteria, the integrated PAD holds great potential for rapid, sensitive and highly selective detection of pathogenic bacteria in resource-limited settings. For this study, a paper impregnated with silver nanoparticles (AgNPs) was prepared. To prepare the substrates, aqueous suspensions of pulp fines, a side product from the paper production, were mixed with AgNP suspensions. The nanoparticle (NP) synthesis was then carried out via laser ablation of pure Ag in water. The continuous emergence of new illegal compounds, particularly psychoactive chemicals, poses significant challenges for current drug detection methods. Developing new protocols and kits for each new drug requires substantial time, effort, and dedicated manpower. Whole-cell bacterial bioreporters have been proven capable of detecting diverse hazardous compounds in both laboratory and field settings, identifying not only single compounds but also chemical families. We present the development of a microbial bioreporter for the detection of cocaine, the nervous system stimulant that is the second-most widely used illegal drug in the US. Escherichia coli was transformed with a plasmid containing a bacterial luxCDABEG bioluminescence gene cassette, activated by a cocaine-responsive signaling cascade. The engineered bioreporter is demonstrated to be a sensitive and specific first-generation detection system for cocaine, with detection thresholds of 17 ± 8 μg/L and 130 ± 50 μg/L in a buffer solution and in urine, respectively. Further improvement of the sensor's performance was achieved by altering the nucleotide sequence of the PBen gene promoter, the construct's sensing element, using accelerated site-directed evolution. Hybrid kinetic models, linking structured models of cell (nano-scale) metabolic processes to the dynamics of macroscopic variables of the bioreactor, are proven to lead to more precise predictions of all key-species dynamics under variable operating conditions, being of an exceptional importance in engineering evaluations to in-silico (math-model-based) determine the optimal operating mode of a fed-batch bioreactor (FBR). Even if such extended dynamic models require more experimental and computational efforts, their use has proven to be advantageous. The approached probative example refers to the simulation of the dynamics of some key species of the central carbon metabolism (CCM) of a modified E. coli cell, linked to the state variables of a FBR used for the tryptophan (TRP) production. By using several optimization algorithms, and an original application of the Pareto-optimal front technique, this paper compares various operating alternatives by using multiple control variables, aiming to maximize TRP production, with minimum substrate consumption. (1) Background: The complexity, amount of time, and the large amount of resource required to perform gold-standard bacteria culture procedures makes it difficult to perform timely pathogenic analyses, especially in areas where such resources are not readily available. Bacterial contamination is a major public health concern on a global scale. Treatment resistance in bacterial infections is becoming a significant problem that requires solutions. We were interested in obtaining new polymeric functionalized compounds with antibacterial properties. Three components (polymeric amine, aldehyde, and phosphite) were used in the paper in a modified "one-pot" Kabachnik-Fields reaction, in tetrahydrofuran at 60 °C, to create the N-C-P skeleton in aminophosphonate groups. Two copolymers were thus prepared starting from an acrylonitriledivinylbenzene (AN-15%DVB) copolymer containing pendant primary amine groups modified by grafting aminophosphonate groups, i.e., aminobenzylphosphonate (Bz-DVB-AN) and aminoethylphosphonate (Et-DVB-AN). The two copolymers were characterized by FT-IR spectroscopy, SEM-EDX, TGA, and antibacterial properties. It was shown that the novel products have antibacterial qualities against S. aureus and E. coli bacteria. The sample with the strongest antibacterial activity was Et-DVB-AN. We assessed how well the Weibull model and the first-order kinetic model represent the inactivation of microbial cells in our samples. The main advantage of the new antibacterial agents developed in this work is their easy recovery, which helps to avoid environmental contamination.

There is a growing demand for rapid and sensitive detection approaches for pathogenic bacteria that can be applied by non-specialists in non-laboratory field settings. Here, the detection of the typical E. coli enzyme β-glucuronidase using a chitosan-based sensing hydrogel-coated paper sensor and the detailed analysis of the reaction kinetics, as detected by a smartphone camera, is reported. The chromogenic reporter unit affords an intense blue color in a two-step reaction, which was analyzed using a modified Michaelis–Menten approach. This generalizable approach can be used to determine the limit of detection and comprises an invaluable tool to characterize the performance of lab-in-a-phone type approaches. For the particular system analyzed, the ratio of reaction rate and equilibrium constants of the enzyme–substrate complex are 0.3 and 0.9 pM−1h−1 for β-glucuronidase in phosphate buffered saline and lysogeny broth, respectively. The minimal degree of substrate conversion for detection of the indigo pigment formed during the reaction is 0.15, while the minimal time required for detection in this particular system is ~2 h at an enzyme concentration of 100 nM. Therefore, this approach is applicable for quantitative lab-in-a-phone based point of care detection systems that are based on enzymatic substrate conversion via bacterial enzymes. Circular RNA (circRNA) exhibits a higher stability and intracellular half-life than linear RNA and has better potential in the fields of RNA vaccines and RNAi drugs. The current strategies for circRNA preparation have low efficiency, high costs, and high complexity, which significantly limits their applications. In this paper, we propose a one-step synthesis of circRNA based on E. coli. The four RNA sequence lengths of 1700, 1400, 500, and 64 nt were connected to group II intron elements from the surface protein region of Clostridium tetani and then inserted downstream of the T7 promoter in the pET28a plasmid to assist in cyclization. Then, circRNA was produced in HT115, where the yields of pET28-1700, pET28-1400, pET28-500, and pET28-64 were improved to 820, 783, 691, and 460 ng/1 mL, respectively. Consequently, this system could achieve the mass production of circRNA using only a simple E. coli culture and inducible expression. Meanwhile, the overexpressed circRNA and small circular interference RNA (sciRNA) maintained their biological functions in the protein translation and RNAi. Therefore, this simple and efficient one-step synthesis system can be applied to the functional study and preparation of circRNA in the future.

This paper presents results of a study on changes in the physical and chemical properties of papain-disintegrated sewage sludge subjected to methane fermentation and the effect of enzymes on the colony number of Escherichia coli, Salmonella spp. and Legionella spp. Fermentation studies were conducted in five batches, in which papain was introduced in amounts representing 0, 1, 2, 3 and 4% (w/w) by weight of the sludge dry matter content. The use of enzymatic disintegration allowed us to intensify the decomposition of organic compounds in the digestion process up to 31% and increase biogas production up to 41% compared to control sludge. At the same time, an almost complete reduction (99–100%) in the colony number of E. coli and Salmonella spp. could be achieved. This paper presents the results of a study on the effect of lipase on the methane fermentation of sewage sludge. The process was conducted at 37 °C for 20 days for five sludge mixtures. Excess sludge inoculated with digested sludge constituted the control sample. The other four samples are the aforementioned mixtures with the addition of lipase in amounts representing 0, 1, 2, 3, and 4% (w/w) with respect to sludge dry weight. The organic matter decomposition rate was 27.1% in the control sludge and from 33.5 to 46.7% in the disintegrated sludge. During the digestion of the control sludge, the total amount of biogas was 5802 mL·L−1. In sewage sludge enzymatically disintegrated by lipase, there was an increase in biogas from 15 to 26%. In the disintegrated sludge, an almost complete (95–100%) reduction in E. coli and Salmonella spp. was achieved. This paper presents an effective hybrid metaheuristic algorithm combining the genetic algorithm (GA) and a simple algorithm based on evolutionary computation. The evolutionary approach (EA) is applied to form the initial population of the GA, thus improving the algorithm's performance, especially its convergence speed. To assess its effectiveness, the proposed hybrid algorithm, the EAGA, is evaluated on selected benchmark functions, as well as on a real optimisation process. The EAGA is used to identify parameters in a nonlinear system of differential equations modelling an E. coli fed-batch fermentation process. The obtained results are compared against published results from hybrid metaheuristic algorithms applied to the selected optimisation problems. The EAGA hybrid outperforms the competing algorithms due to its effective initial population generation strategy. The risk of premature convergence is reduced. Better numerical outcomes are achieved. Cyclodextrins (CDs) are natural cyclic oligosaccharides with the ability to form inclusion complexes with various organic substances. In this paper, we investigate the potential of CD complex formation to enhance the antibacterial activity and antioxidant properties of poorly soluble bioactive agents, such as chalcones, chromenes, stilbenoids and xanthylium derivatives, serving as potential adjuvants, in comparison with standard antiseptics. The interaction of these bioactive agents with the hydrophobic pocket of methyl-β-cyclodextrin (MCD) was confirmed using spectroscopic methods such as UV-vis, FTIR, 1H and 13C NMR, mass-spectrometry. CD-based delivery system allows for combining multiple active agents, improving solubility, antibacterial efficacy by enhancing penetration into target bacterial cells (E. coli selectivity demonstrated via confocal microscopy). Novel compounds of chalcones and stilbenoids derivatives additionally enhance efficacy by inhibiting bacterial efflux pumps, increasing membrane permeability, and inhibiting bacterial enzymes, and showed a synergy when used in combination with metronidazole. The intricate relationship between the structural characteristics and functional properties of chalcones and stilbenoids in terms of their antibacterial and antioxidative capabilities is revealed. The substituents within aromatic rings significantly influence this activity, where position of electron-donating methoxy groups playing a crucial role. Among chalcones, stilbenoids, ana xanthyliums, the compounds caring a benzodioxol ring, analogous to natural bioactive compounds like apiol, dillapiol, and myristicin, emerge as prominent antibacterial activity. To explore the possibility to create theranostic formulations, we used fluorescent markers to visualize target cells, antiseptics to provide antibacterial activity, and bioactive agents as chalcones acting as adjuvants. Additionally, new antioxidant compounds were found such as Xanthylium derivative (R351) and chromene derivative: 1-methyl-3-(2-amino-3-cyano-7-methoxychromene-4-yl)-pyridinium methanesulfate: the pronounced antioxidant properties of these substances are observed comparable to quercetin in the efficiency. Rhodamine 6G, gentian violet, and Congo Red exhibit good antioxidant properties, although their activity is an order of magnitude lower than that of quercetin. However, they have remarkable potential due to their multifaceted nature, including the ability to visualize target cells. The most effective theranostic formulation is the combination of the antibiotic (metronidazole) + dye/fluorophore (methylene blue/rhodamine 6G) for visualization of target cells + adjuvant (chalcones or xanthylium derivatives) for antiinflammation effect. This synergistic combination, results in a promising theranostic formulation for treating bacterial infections, with enhanced efficiency, selectivity and minimizing side effects. A green and cost-effective sonochemical synthetic method was followed for coating silver-modified copper oxide (Ag-CuO) nanoparticles (NPs) on disposable surgical mask. The NP-coated masks were systematically characterized using XRD and FT-IR for understanding the structural and surface functionalities. In addition, the field emission scanning electron microscopy (FE-SEM) analysis showed the homogeneous coating of Ag-CuO NPs over the mask fibers. The average particle size of Ag-CuO was found to be ~70 nm. The NP-coated masks are useful to combat a broad range of bacterial species by taking the unique advantage of the synergistic effect of Ag and metal oxide (CuO and ZnO) NPs for the generation of reactive oxygen species (ROS). Zone of inhibition (ZoI) studies demonstrated antibacterial activity against both Gram-positive S. aureus and Gram-negative E. coli bacteria, probably due to the elevated production of ROS by the defect structure of the Ag-modified metal oxide NPs. The material was found to be effective against both airborne and soil-borne bacteria. Often, bacterial infections delay the rate of healing of traumatic wounds, making it critical to improve antimicrobial efficiency. In this paper, titanium nanotubes (TNT) with good antimicrobial and synergistic photothermal properties were used as the core, and mesoporous polydopamine (MPDA) thin films were constructed on their surface. Gold nanoparticles (AuNPs) with excellent photothermal conversion efficiencies (PCE) were incorporated. Finally, a large number of composite nanoparticles were added to polyvinyl alcohol (PVA) and polyethylene glycol (PEG) with wound-restoring ability, and an injectable antimicrobial hydrogel was successfully prepared by a one-pot synthesis. The antimicrobial effect of TNT@MPDA@Au nanoparticles with different concentrations was assessed by in vitro antimicrobial experiments on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The higher the concentration of nanoparticles under near-infrared light irradiation (NIR), the stronger the antimicrobial effect. The in vitro cytotoxicity of TNT@MPDA and TNT@MPDA@Au nanoparticles on 293T normal cells was tested through CCK-8 assay. The results show that both nanoparticles have favourable biocompatibility. Thymol and carvacrol are some of the most important and used components of Essential oils (EOs); they are widely studied, and there are much data available in the literature. Their Minimal Inhibitory Concentration (MIC) values found in the literature from 2005 to present were used to assess the bioactivity toward yeasts, molds, Gram-positive bacteria, and Gram-negative bacteria, as well as on some bacterial species/serotypes (Salmonella sp., Escherichia coli, E. coli O157:H7, lactic acid bacteria, Listeria monocytogenes, Staphylococcus aureus, S. epidermidis, etc.) to find possible common trends or differences between the two compounds and among the tested species. The results were quite interesting and pointed out that there is a common range for the MIC of thymol and carvacrol for some bacterial species (150–400 mg/L), with some exceptions to this generalized statement. In addition, the statistics pointed out that bacteria could experience homogeneous (S. epidermidis, E. coli O157:H7) or heterogeneous trends (for example, Salmonella sp.) depending on the existence of possible sub-species or different experimental set-ups. Moreover, this paper suggests that there are some drawbacks and issues that should be solved for the effective use of EOs, which are the strong variability among the microorganisms and the lack of standard protocols and reference strains. Bacteria are known to be constantly adapting to become resistant to antibiotics. Currently, efficient antibacterial compounds are still available; however, it is only a matter of time until these compounds also become inefficient. Ribonucleases are the enzymes responsible for the maturation and degradation of RNA molecules, and many of them are essential for microbial survival. Members of the PNPase and RNase II families of exoribonucleases have been implicated in virulence in many pathogens and, as such, are valid targets for the development of new antibacterials. In this paper, we describe the use of virtual high-throughput screening (vHTS) to identify chemical compounds predicted to bind to the active sites within the known structures of RNase II and PNPase from Escherichia coli . The subsequent in vitro screening identified compounds that inhibited the activity of these exoribonucleases, with some also affecting cell viability, thereby providing proof of principle for utilizing the known structures of these enzymes in the pursuit of new antibacterials.

Community-acquired urinary tract infections account for 15% of all outpatient use of antibiotics, and women are primarily affected; the major causative microorganism is uropathogenic Escherichia coli (E. coli). Treatment is indicated for cystitis and pyelonephritis and includes B-lactams (amoxicillin-clavulanic acid or third-generation cephalosporins), fluoroquinolones (ciprofloxacin or levofloxacin), nitrofurantoin, fosfomycin, and trimethoprim–sulfamethoxazole. Resistance to antibiotic treatment is of concern; several mechanisms have been associated with the acquisition of genes that confer antimicrobial resistance to fluoroquinolones, which are often associated with other patterns of resistance, especially in extended-spectrum beta-lactamase (ESBL) producers. Several studies have addressed the prevalence of uropathogens producing ESBLs, but only a few have focused on fluoroquinolone resistance, and, to our knowledge, none have addressed the prevalence of phylotypes or genes responsible for antimicrobial resistance to fluoroquinolones. The focus of the present review was to analyze recently published papers that described the E. coli phylotype causing community-acquired UTIs in association with fluoroquinolone resistance. This paper presents the findings of a large-scale study on antibiotic resistance in bacteria found in farm animal feces across Russia. The study included 6578 samples of farm animal manure from 13 regions in Russia, with the help of citizen scientists. Molecular and microbiological methods were used to analyze 1111 samples of E. coli. The microbiological analysis focused on culturing the microorganisms present in the fecal samples on selective media for E. coli and evaluating the sensitivity of the bacteria to different antibiotics, including ampicillin, tetracycline, chloramphenicol, cefotaxime, and ciprofloxacin. The molecular analysis involved isolating the genomic DNA of the bacteria and conducting PCR assays to detect the vanA, vanB, and mcr-1 antibiotic resistance genes. The results demonstrated significant differences in antibiotic sensitivity of the samples that are morphologically identical to E. coli from different regions. For example, 98.0% and 82.5% of E. coli and other fecal bacterial isolates from the Omsk and Vologda regions lacked antibiotic resistance genes, while 97.7% of samples from the Voronezh region possessed three resistance genes simultaneously. The phenotypic antibiotic sensitivity test also revealed regional differences. For instance, 98.1% of fecal bacterial samples from cattle in the Udmurt Republic were sensitive to all five antibiotics tested, whereas 92.8% of samples from the Voronezh region showed resistance to all five antibiotics. The high level of antibiotic resistance observed may be attributed to their use in farming practices. The distinctive feature of our research is that comprehensive geographical coverage was achieved by using a citizen science platform. Citizen scientists, specifically students from colleges and universities, were responsible for the collection and initial analysis of samples. Background and Objectives: Diabetic patients are more likely to develop infections compared to the general population, especially urinary tract infections (UTIs). The aim of this study was to assess the prevalence of UTIs in a population of patients with diabetes (DM) from Romania, to identify the most common uropathogens and their antimicrobial resistance (AMR) patterns, as well as to determine the correlations between resistance behavior and particularities of patients with UTIs according to DM type. Materials and Methods: The hospital records of 1282 type 1 (T1D) and type 2 DM (T2D) adult inpatients who were ordered urine cultures during hospitalization were reviewed, and all 241 patients who presented a positive urine culture were included in the present study analysis. Results: The prevalence of UTIs in diabetic patients was 18.8% and higher in patients with T2D vs. T1D. Patients with UTIs and T2D had a significantly older age, longer duration of DM, higher waist circumference and body mass index, lower levels of estimated glomerular filtration rate, and more frequent chronic complications of DM than patients with T1D. E. coli was the most frequently isolated uropathogen (56.4%), with a significantly higher incidence for T2D, followed by K. pneumoniae (12.9%) and Enterococcus spp. (9.5%). Although the acquired resistance phenotypes were more frequently isolated in T2D patients (over 90% of the multidrug-resistant and extended-spectrum beta-lactamase-producing isolates, respectively, and 75% of the total carbapenem-resistant organisms), no statistically significant correlation was found regarding the distribution of AMR patterns in the two types of DM. Conclusions: The present study brings new data regarding the prevalence of UTIs in diabetic patients from Western Romania. By identifying the spectrum of uropathogens and their AMR pattern, this paper may contribute to improving UTI management in diabetic patients, thus reducing antibiotic overuse and preventing recurrent UTIs. Simple Summary: Endometritis is a prevalent disease in perinatal dairy cows, which leads to compromised reproductive function. Studies have confirmed that A20, also known as TNFAIP3, can regulate inflammatory responses in various cells. However, it remains unclear whether A20 regulates the inflammatory response of bovine endometrial cells and if autophagy plays a role in A20's regulation of inflammatory responses. In this paper, we show that elevated expression of A20 is linked to the progression of endometritis in dairy cows. A20 inhibited the activation of the NF-κB pathway and the mRNA expression levels of proinflammatory cytokines in LPS-induced bovine endometrial epithelial cells. Moreover, it enhanced autophagic activity in the bovine endometrial epithelial cells inhibited by LPS. Our study further showed that A20 mitigated the inflammatory response of bovine endometrial epithelial cells by promoting autophagy. These findings will enhance our understanding of the pathogenesis and provide novel insights for the prevention and treatment of endometritis in dairy cows. Endometritis represents a prevalent condition in perinatal dairy cows. Bovine endometrial epithelial cells (BEECs), as the primary interface between cavity and the external environment, are particularly vulnerable to infection by pathogenic bacteria following parturition. A20 is essential for regulating inflammation and modulating immune responses. Nevertheless, the exact role of A20 in the BEECs in response to inflammatory response is not fully understood. An endometritis model infected by Escherichia coli (E. coli) in vivo and a BEECs inflammation model induced with lipopolysaccharide (LPS) in vitro were built to investigate the function and governing mechanisms of A20 in endometritis. The results showed that infection with E. coli resulted in endometrial damage, inflammatory cell infiltration, and upregulation of inflammatory factors in dairy cows. Furthermore, A20 expression was upregulated in the endometrium of cows with endometritis and in BEECs following LPS stimulation. A20 overexpression attenuated the level of proinflammatory cytokines in LPS-stimulated BEECs; conversely, A20 knockdown lead to an exacerbated response to LPS stimulation. The overexpression of A20 was shown to activate autophagy and suppress the NF-κB signaling pathway in LPS-stimulated BEECs. However, blocking autophagy with chloroquine notably attenuated the anti-inflammatory effect of A20, leading to the activation of the NF-κB signaling pathway. In summary, the study demonstrated that A20's suppression of inflammation in LPS-stimulated BEECs is associated with the activation of autophagy. In this paper, a series of tetrameric surfactants (4CnSAZs, n = 12, 14, 16) endowed with zwitterionic characteristic were synthesized by a simple and convenient method and their structures were characterized by FT-IR, 1H NMR and elemental analysis. Their physicochemical properties were studied using the Wilhelmy plate method, fluorescence spectra and dynamic light scattering technique. 4CnSAZs have higher surface activities and tend to adsorb at the air/water surface rather than self-assembling in aqueous solution. The thermodynamic parameters obtained from surface tension measurements show that both processes of adsorption and micellization of 4CnSAZs are spontaneous and that the micellization processes of 4CnSAZs are entropy-driven processes. Both adsorption and micellization of 4CnSAZs are inclined to occur with the increase of alkyl chain length or temperature. For 4C12SAZs, there are only small-size aggregates (micelles), while the large aggregates (vesicles) are observed at the alkyl length of 4CnSAZs of 14 or 16. This shows that the alkyl chain length for oligomeric surfactants has a greater sensitivity for aggregate growth. The aggregate morphologies obtained from the calculated values of critical packing parameter (p) for 4C14SAZs and 4C16SAZs can be supported by the DLS measurement results. The test results obtained by the separation-water-time method show that 4CnSAZs have good emulsification performance and that the prepared emulsions appear to exit in the form of multiple emulsions. In addition, 4CnSAZs have good antibacterial activities against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The present study reveals the unique behavior of a zwitterionic tetrameric surfactant and may give new insights into molecular design and synthesis of a high degree of surfactants with different structure characteristics for potential application in various industrial fields. The tricyclic-aromadendrene-type sesquiterpenes are widely distributed and exhibit a range of biological activities, including anti-inflammatory, analgesic, antioxidant, antibacterial, insecticidal and cytotoxic properties. Several key sesquiterpene synthases (STSs) of this type have been identified, of which, viridiflorol synthase has been engineered for efficiently biosynthesizing viridiflorol in an Escherichia coli strain. This paper comprehensively summarizes the distribution and biological activity of aromadendrene-type sesquiterpenes in plant essential oils and microorganisms. The progress in aromadendrene-type sesquiterpene biosynthesis research, including the modifications of key STSs and the optimization of synthetic pathways, is reviewed. This paper presents an optimal tuning of a proportional integral differential (PID) controller used to maintain glucose concentration at a desired set point. The PID controller synthesizes an appropriate feed rate profile for an E. coli fed-batch cultivation process. Mathematical models are developed based on dynamic mass balance equations for biomass, substrate, and product concentration of the E. coli BL21(DE3)pPhyt109 fed-batch cultivation for bacterial phytase extracellular production. For model parameter identification and PID tuning, a hybrid metaheuristic technique—chaotic electromagnetic field optimization (CEFO)—is proposed. In the hybridization, a chaotic map is used for the generation of a new electromagnetic particle instead of the electromagnetic field optimization (EFO) search strategy. The CEFO combines the exploitation capability of the EFO algorithm and the exploration power of ten different chaotic maps. The comparison of the results with classical EFO shows the superior behaviour of the designed CEFO. An improvement of 30% of the objective function is achieved by applying CEFO. Based on the obtained mathematical models, 10 PID controllers are tuned. The simulation experiments show that the designed controllers are robust, resulting in a good control system performance. The closed-loop transient responses for the corresponding controllers are similar to the estimated models. The settling time of the control system based on the third PID controller for all estimated models is approximately 9 min and the overshoot is approximately 15%. The proposed CEFO algorithm can be considered an effective methodology for mathematical modelling and achievement of high quality and better performance of the designed closed-loop system for cultivation processes. Given the widespread prevalence of viruses, there is an escalating demand for antimicrobial composites. Although the composite of dialdehyde cellulose and silver nanoparticles (DAC@Ag1) exhibits excellent antibacterial properties, its weak mechanical characteristics hinder its practical applicability. To address this limitation, cellulose nanofibers (CNFs) were initially ammoniated to yield N-CNF, which was subsequently incorporated into DAC@Ag1 as an enhancer, forming DAC@Ag1/N-CNF. We systematically investigated the optimal amount of N-CNF and characterized the DAC@Ag1/N-CNF using FT-IR, XPS, and XRD analyses to evaluate its additional properties. Notably, the optimal mass ratio of N-CNF to DAC@Ag1 was found to be 5:5, resulting in a substantial enhancement in mechanical properties, with a 139.8% increase in tensile elongation and a 33.1% increase in strength, reaching 10% and 125.24 MPa, respectively, compared to DAC@Ag1 alone. Furthermore, the inhibition zones against Escherichia coli and Staphylococcus aureus were significantly expanded to 7.9 mm and 15.9 mm, respectively, surpassing those of DAC@Ag1 alone by 154.8% and 467.9%, indicating remarkable improvements in antimicrobial efficacy. Mechanism analysis highlighted synergistic effects from chemical covalent bonding and hydrogen bonding in the DAC@Ag1/N-CNF, enhancing the mechanical and antimicrobial properties significantly. The addition of N-CNF markedly augmented the properties of the composite film, thereby facilitating its broader application in the antimicrobial field.

The evaluation of the potentiality of lactic acid bacteria (LAB) strains isolated from fermented products to inhibit Botrytis cinerea and Escherichia coli O157:H7 growth on spinach and lettuce was conducted. From a total of forty LAB strains tested, three were selected due to their high inhibitory effect on plant pathogenic fungi. The identification of these isolates based on a 16S rRNA gene fragment sequence analysis confirmed the genus of Levilactobacillus sp. and Lactiplantibacillus sp. An effective method of coating LAB isolates on the lettuce and spinach surface was developed. The leaves were immersed in bacterial suspension (5.0 × 106 cfu mL−1) for 4 s and drained on tissue paper. LAB survived on lettuce and spinach leaves for 8 days at 6 log10 cfu g−1. Additionally, these bacteria decreased the number of filamentous fungi on the leaves. These isolates were found to inhibit the growth of B. cinerea and E. coli O157:H7 in vitro conditions in growing microbiological media. Their efficacy was confirmed in vivo conditions. These isolates inhibited the development of grey mould caused by B. cinerea on lettuce leaves. Two LAB isolates reduced the abundance of the pathogenic bacterium E. coli on spinach leaves by about 0.7 log10 cfu g−1. In glasshouse conditions, LAB stimulated the growth of examined plants. The lactic acid bacteria used in this study showed the capacity to be used as possible alternatives to chemical compounds in the protection of leafy vegetables against grey mould and for a decrease in E. coli O157:H7 contamination. The detection of pathogens in medical wastewater is crucial due to the high content of pathogenic microorganisms that pose significant risks to public health and the environment. Medical wastewater, which includes waste from infectious disease and tuberculosis facilities, as well as comprehensive medical institutions, contains a variety of pathogens such as bacteria, viruses, fungi, and parasites. Traditional detection methods like nucleic acid detection and immunological assays, while effective, are often time-consuming, expensive, and not suitable for rapid detection in underdeveloped areas. Electrochemical biosensors offer a promising alternative with advantages including simplicity, rapid response, portability, and low cost. This paper reviews the sources of pathogens in medical wastewater, highlighting specific bacteria (e.g., E. coli, Salmonella, Staphylococcus aureus), viruses (e.g., enterovirus, respiratory viruses, hepatitis virus), parasites, and fungi. It also discusses various electrochemical biosensing techniques such as voltammetry, conductometry, impedance, photoelectrochemical, and electrochemiluminescent biosensors. These technologies facilitate the rapid, sensitive, and specific detection of pathogens, thereby supporting public health and environmental safety. Future research may should pay more attention on enhancing sensor sensitivity and specificity, developing portable and cost-effective devices, and innovating detection methods for diverse pathogens to improve public health protection and environmental monitoring. In this paper, we describe the supervised dynamic correlated topic model (sDCTM) for classifying categorical time series. This model extends the correlated topic model used for analyzing textual documents to a supervised framework that features dynamic modeling of latent topics. sDCTM treats each time series as a document and each categorical value in the time series as a word in the document. We assume that the observed time series is generated by an underlying latent stochastic process. We develop a state-space framework to model the dynamic evolution of the latent process, i.e., the hidden thematic structure of the time series. Our model provides a Bayesian supervised learning (classification) framework using a variational Kalman filter EM algorithm. The E-step and M-step, respectively, approximate the posterior distribution of the latent variables and estimate the model parameters. The fitted model is then used for the classification of new time series and for information retrieval that is useful for practitioners. We assess our method using simulated data. As an illustration to real data, we apply our method to promoter sequence identification data to classify E. coli DNA sub-sequences by uncovering hidden patterns or motifs that can serve as markers for promoter presence. Klebsiella spp. and Enterobacter, classified as non-pathogenic bacteria, are often found. Good sanitary practices, e.g., cleaning and remaining dry, as well as boiling crickets during the harvesting process, may be helpful for the safety of edible cricket production. Food safety for cricket production is a crucial factor in producing edible crickets with safety for consumers and sustainability for two-spotted (Gryllus bimaculatus) as well as house (Acheta domesticus) cricket production. This study was conducted by simultaneously rearing two cricket species, comprising two-spotted crickets (G. bimaculatus) and house crickets (A. domesticus). A total of 16 rearing crates were used for the present study, which were allocated into 8 rearing crates for each studied cricket species, including paper egg cartons. Cricket eggs were incubated in the rearing crates. Once the crickets hatched, tap water and powdered feed were provided ad libitum throughout the experiment. At the end of this study (35 and 42 days for the two-spotted and house crickets, respectively), all crickets were harvested, rinsed in tap water, and boiled in water for 5 min. During the rearing and harvesting processes, samples were collected from various potential contamination points for bacteria, including E. coli and Salmonella spp. There were samples of the initial input (feed, drinking water, and staff hands), rearing environment (water pipe, crate wall, living cartons, frass, and cricket surface), and harvesting crickets (harvested, washed, and boiled crickets), with a 2-week sampling interval, except for the last round of sampling for the two-spotted crickets. Subsequently, all samples were submitted to isolate and identify contaminated bacteria. The samples from the last round of sampling for both kinds of crickets were submitted to quantify the level of contamination for E. coli and Salmonella spp., including antimicrobial resistance by the disk diffusion method for the positive isolate. The results showed that bacterial contamination was found in the rearing of both cricket species, primarily involving Klebsiella spp. and Enterobacter spp., mainly found in prepared drinking water and the water pipes of drinking water supply equipment, which are potential sources of contamination with cricket frass. E. coli was found in 4.8% and 4.3% of the two-spotted and house crickets, respectively, while no presence of Salmonella spp. was detected in any submitted samples. The quantification of E. coli and Salmonella spp. indicated E. coli contamination near the water pipe and the frass of two-spotted crickets, but Salmonella spp. was undetectable in both two-spotted and house crickets. The antimicrobial resistance of isolated E. coli mainly involved penicillin G, amoxicillin, ampicillin, erythromycin, lincomycin, and tiamulin. Thus, good farm management with proper sanitation practices (such as cleaning and keeping the environment dry), as well as boiling crickets during the harvesting process, may help ensure the safety of edible cricket production. The phosphoenol pyruvate–oxaloacetate–pyruvate-derived amino acids (POP-AAs) comprise native intermediates in cellular metabolism, within which the phosphoenol pyruvate–oxaloacetate–pyruvate (POP) node is the switch point among the major metabolic pathways existing in most living organisms. POP-AAs have widespread applications in the nutrition, food, and pharmaceutical industries. These amino acids have been predominantly produced in Escherichia coli and Corynebacterium glutamicum through microbial fermentation. With the rapid increase in market requirements, along with the global food shortage situation, the industrial production capacity of these two bacteria has encountered two bottlenecks: low product conversion efficiency and high cost of raw materials. Aiming to push forward the update and upgrade of engineered strains with higher yield and productivity, this paper presents a comprehensive summarization of the fundamental strategy of metabolic engineering techniques around phosphoenol pyruvate–oxaloacetate–pyruvate node for POP-AA production, including L-tryptophan, L-tyrosine, L-phenylalanine, L-valine, L-lysine, L-threonine, and L-isoleucine. Novel heterologous routes and regulation methods regarding the carbon flux redistribution in the POP node and the formation of amino acids should be taken into consideration to improve POP-AA production to approach maximum theoretical values. The paper is a review of surface plasmon resonance (SPR) structures containing amorphous chalcogenide (ChG) films as plasmonic waveguides. The calculation method and specific characteristics obtained for four-layer SPR structures containing films made of amorphous As2S3 and As2Se3 are presented. The paper is mainly based on our previously obtained and published scattered results, to which a generalized point of view was applied. In our analysis, we demonstrate that, through proper choice of the SPR structure layer parameters, we can control the resonance angle, the sharpness of the SPR resonance curve, the penetration depth, and the sensitivity to changes in the refractive index of the analyte. These results are obtained by operating with the thickness of the ChG film and the parameters of the coupling prism. Aspects regarding the realization of the coupling prism are discussed. Two distinct cases are analyzed: first, when the prism is made of material with a refractive index higher than that of the waveguide material; second, when the prism is made of material with a lower refractive index. We demonstrated experimentally that the change in reflectance self-induced by the modification in As2S3 refractive index exhibits a hysteresis loop. Guanabara Bay has been known to be polluted with trash and sewage from the surrounding areas, but health risks from recreational contact with water in the basin have not been well characterized. In this paper, fecal indicator bacteria (FIB) monitoring data are used to predict risks in three different exposure scenarios: (1) bathing in freshwater rivers that discharge into Guanabara Bay, (2) wading in these freshwater rivers, and (3) sailing in Guanabara Bay. Concentrations of indicator bacteria in river samples were measured directly, and concentrations of indicator bacteria in bay samples were sourced from publicly available government data sets. Ratios between pathogens and fecal indicator concentrations were used to estimate risks for five selected pathogens based on the indicator concentrations. The median risk of disease estimated from E. coli indicator concentrations was 1.0, 9.9 × 10−1 and 8.2 × 10−4 for the swimming, wading, and sailing exposure pathways, respectively. Risks estimates based on concentrations of the enterococci indicator bacteria in the sailing exposure scenario were comparable, at 3.4 × 10−4. The sum of total risk estimated from the five selected pathogens was 5.9 × 10−1, 3.6 × 10−1, and 1.0 × 10−3 for the swimming, wading, and sailing exposure pathways, respectively. Estimated risks of swimming and wading in the rivers far exceeded risks associated with U.S. recreational contact standards, while estimated risks for sailing in the bay were well below these risk guidelines. The 95th percentile of the sailing risk was estimated to exceed the U.S. recreational contact risk level. This paper exemplifies an approach to conducting quantitative microbial risk assessments when only fecal indicator bacteria data are available. Context-specific data on the relevant exposure routes, exposure frequency, and site-specific indicator: pathogen ratios were lacking, which ultimately led to uncertainty in the model. This study is intended to provide a framework for estimating GI risk based on fecal indicator concentrations while acknowledging that the substantial variation in indicator:pathogen ratios make the results of such efforts uncertain.
