Abstract: International audience; Escherichia coli is a versatile commensal species of the animal gut that can also be a pathogen able to cause intestinal and extraintestinal infections. The plasticity of its genome has led to the evolution of pathogenic strains, which represent a threat to global health. Additionally, E. coli strains are major drivers of antibiotic resistance, highlighting the urgent need for new treatment and prevention measures. The antigenic and structural heterogeneity of enterohaemorrhagic E. coli colonisation factors has limited their use for the development of effective and cross-protective vaccines. However, the emergence of new strains that express virulence factors deriving from different E. coli diarrhoeagenic pathotypes suggests that a vaccine targeting conserved proteins could be a more effective approach. In this study, we conducted proteomics analysis and functional protein characterisation to identify a group of proteins potentially involved in the adhesion of E. coli O157:H7 to the extracellular matrix and intestinal epithelial cells. Among them, OmpA has been identified as a highly conserved and immunogenic antigen, playing a significant role in the adhesion phenotype of E. coli O157:H7 and in bacterial aggregation. Furthermore, antibodies raised against recombinant OmpA effectively reduced the adhesion of E. coli O157:H7 to intestinal epithelial cells. The present work highlights the role of OmpA as a potent antigen for the development of a vaccine against intestinal pathogenic E. coli.

Abstract: International audience; ONERA – The French Aerospace Lab – develops new concepts of 3D-LiDAR imaging systems including new sensor technologies and data processing. Here, we present a more efficient strategy than existing solution to numerically enhance the lateral resolution of low photon 3D-LiDAR operating in Geiger mode. Our pipeline makes it possible to reconstruct 3D-images with an unprecedented lateral-resolution, simultaneously at low photon count and Hertz level framerates. It is applied on simulated GmAPD 3D-LiDAR signals. Signals acquired using this kind of sensors are unsuitable for direct applications of Compressive Sensing algorithms. Our contribution focuses on a more efficient strategy for waveform denoising and reconstruction. For each pixel, we reconstruct sub-pixels with a Compressive Sensing approach. Compressive Sensing has already been used for single-photon applications with single-pixel cameras. In our pipeline, we extend this method to focal plane arrays in Geiger-mode. This process can be summarized as a set of signal processing techniques to enhance the incoming signal and improve the Compressive Sensing reconstruction. Our goal is to recover a complete noise-free waveform. We distinguish two main parts: a reconstruction part which compensates the low dynamic range of the signal induced by the Geiger mode; a denoising part which uses a new denoising strategy by statistical comparison. This pipeline can be parallelized on GPU as each pixel in the focal plane array is independent from the others. In this talk, we will detail the pipeline and then demonstrate its applicability on realistic simulated data.; L'ONERA – Laboratoire français de l'aérospatiale – développe de nouveaux concepts de systèmes d'imagerie 3D-LiDAR incluant de nouvelles technologies de capteurs et de traitement de données. Ici, nous présentons une stratégie plus efficace que la solution existante pour améliorer numériquement la résolution latérale du LiDAR 3D à faible photon fonctionnant en mode Geiger. Notre pipeline permet de reconstruire des images 3D avec une résolution latérale sans précédent, simultanément à un faible nombre de photons et à des fréquences d'images de niveau Hertz. Il est appliqué sur des signaux GmAPD 3D-LiDAR simulés. Les signaux acquis à l'aide de ce type de capteurs ne conviennent pas aux applications directes des algorithmes de détection compressive. Notre contribution se concentre sur une stratégie plus efficace pour le débruitage et la reconstruction des formes d'onde. Pour chaque pixel, nous reconstruisons des sous-pixels avec une approche Compressive Sensing. La détection compressive a déjà été utilisée pour des applications à photon unique avec des caméras à un pixel. Dans notre pipeline, nous étendons cette méthode aux réseaux plan focal en mode Geiger. Ce processus peut être résumé comme un ensemble de techniques de traitement du signal pour améliorer le signal entrant et améliorer la reconstruction de détection de compression. Notre objectif est de récupérer une forme d'onde complète sans bruit. On distingue deux parties principales : une partie reconstruction qui compense la faible dynamique du signal induite par le mode Geiger ; une partie débruitage qui utilise une nouvelle stratégie de débruitage par comparaison statistique. Ce pipeline peut être parallélisé sur GPU car chaque pixel du réseau plan focal est indépendant des autres. Dans cet exposé, nous détaillerons le pipeline, puis démontrerons son applicabilité sur des données simulées réalistes.

Abstract: International audience; Abstract Microbacterium sp. C448, isolated from a soil regularly exposed to sulfamethazine (SMZ), can use various sulphonamide antibiotics as the sole carbon source for growth. The basis for the regulation of genes encoding the sulphonamide metabolism pathway, the dihydropteroate synthase sulphonamide target (folP), and the sulphonamide resistance (sul1) genes, is unknown in this organism. In the present study, the response of the transcriptome and proteome of Microbacterium sp. C448 following exposure to subtherapeutic (33 µM) or therapeutic (832 µM) SMZ concentrations was evaluated. Therapeutic concentration induced the highest sad expression and Sad production, consistent with the activity of SMZ degradation observed in cellulo. Following complete SMZ degradation, Sad production tended to return to the basal level observed prior to SMZ exposure. Transcriptomic and proteomic kinetics were concomitant for the resistance genes and proteins. The abundance of Sul1 protein, 100-fold more abundant than FolP protein, did not change in response to SMZ exposure. Moreover, non-targeted analyses highlighted the increase of a deaminase RidA and a putative sulphate exporter expression and production. These two novel factors involved in the 4-aminophenol metabolite degradation and the export of sulphate residues formed during SMZ degradation, respectively, provided new insights into Microbacterium sp. C448 SMZ detoxification process.

Abstract: International audience; Objective: To compare the efficacy of, and complications from, the 2 main treatments for achalasia: endoscopic dilatation and surgical cardiomyotomy (Heller's myotomy).Study design: We retrospectively collected data on children treated for achalasia over an 11-year period from 8 tertiary pediatric centers. A line of treatment was defined as performing either Heller's myotomy or 1-3 sessions of endoscopy dilatation over 3 months. Treatment success was a priori defined as clinical improvement and no need for new treatment.Results: Ninety-seven children (median age, 12 years; 57% boys) were included. The median time to diagnosis was 10.5 months, and the median follow-up period was 27 months. Thirty-seven children were treated by Heller's myotomy and 60 by endoscopy dilatation as the first-line treatment. After adjustment for potentially confounding factors, Heller's myotomy was significantly more successful than endoscopy dilatation (hazard ratio, 3.93 [1.74; 8.88]; P = .001), with a median survival without failure of 49 and 7 months, respectively, and with no significant difference in the occurrence of complications (35.2% for Heller's myotomy, 29.7% for endoscopy dilatation, P = .56). Hydrostatic dilatation was as successful as pneumatic dilatation (hazard ratio, 1.35 [0.56; 3.23]; P = .50).Conclusions: Heller's myotomy is more successful than endoscopy dilatation, with no significant difference in the occurrence of serious complications. This raises the potential role of peroral endoscopic myotomy as an alternative treatment to Heller's myotomy.

Abstract: Essential fatty acids (EFA) and conjugated linoleic acids (CLA) are unsaturated fatty acids with immune-modulatory effects, yet their synergistic effect is poorly understood in dairy cows. This study aimed at identifying differentially abundant proteins (DAP) and their associated pathways in dairy cows supplied with a combination of EFA and CLA during the transition from antepartum (AP) to early postpartum (PP). Sixteen Holstein cows were abomasally infused with coconut oil as a control (CTRL) or a mixture of EFA (linseed + safflower oil) and CLA (Lutalin, BASF) (EFA + CLA) from − 63 to + 63 days relative to parturition. Label-free quantitative proteomics was performed on plasma samples collected at days − 21, + 1, + 28, and + 63. During the transition time, DAP, consisting of a cluster of apolipoproteins (APO), including APOE, APOH, and APOB, along with a cluster of immune-related proteins, were related to complement and coagulation cascades, inflammatory response, and cholesterol metabolism. In response to EFA + CLA, specific APO comprising APOC3, APOA1, APOA4, and APOC4 were increased in a time-dependent manner; they were linked to triglyceride-enriched lipoprotein metabolisms and immune function. Altogether, these results provide new insights into metabolic and immune adaptation and crosstalk between them in transition dairy cows divergent in EFA + CLA status.

Abstract: International audience; Playing wind historical instruments is often conflicting with conservation and protection of museum collections. Some musical and acoustical properties of these instruments therefore remain unknown. Theoretical models can predict some of these properties from the only knowledge of the instruments geometry. A collaboration between Makutu Inria Bordeaux Sud Ouest team, Musée de la Musique - Philharmonie de Paris, Institut Technologique Européen des Métiers de la Musique (ITEMM) at Le Mans and the Centre de Recherche et de Restauration des Musées de France (C2RMF), lead to a procedure hereby applied to the museum collection Besson, which was a leading manufacture of wind instruments, and more specifically to several natural trumpets dating from the early 20th century. The geometry of the instruments has been measured non invasively by means of X-ray tomography in C2RMF. After extracting their bore (evolution of the inner diameter), their entry impedance has been computed. For one specific trumpet (E.0925), simulated data have been compared to measurements with excellent agreement. Despite many uncertainties on the way these instruments were played, simulated sounds can be computed. An « acoustical facsimile » of E.0925 has been crafted from the X-ray and impedance data, and played by a professional natural trumpet player. This allows to compare simulated sounds with human played sounds.

Abstract: International audience; Monoclonal immunoglobulin M (IgM) anti-myelin-associated glycoprotein (MAG) neuropathy is a rare disabling condition, most commonly treated with rituximab monotherapy (R), which leads to neurological improvement in only 30%-50% of patients. The combination of rituximab plus chemotherapy has been proven to improve the level of responses. We studied the outcomes of anti-MAG neuropathy patients treated either by R, or by immunochemotherapy (ICT) in our centre, focusing on the incidence of the first neurological response evaluated by the modified Rankin Scale (mRS). From 2011 to 2018, 64 patients were studied: 34 were treated with R and 30 with ICT. According to our treatment decisionmaking process, the median mRS was higher in the ICT group (mRS 2) compared to the R group (mRS 1). At 1 year, mRS improvement rates were 46% and 18% of the ICT and R groups of patients respectively, with a median time to response of 8 and 13 months (p=0.023). Adverse effects were higher in the ICT group: 62% vs 15% (p˂0.01) all grades included. One secondary acute leukaemia occurred 5 years after treatment by ICT. In conclusion, ICT may be used as a valid option for patients with rapidly progressive and/or severe anti-MAG neuropathy symptoms.

Abstract: Gaia DR3 opens a new era of all-sky spectral analysis of stellar populations thanks to the nearly 5.6 million stars observed by the RVS and parametrised by the GSP-spec module. The all-sky Gaia chemical cartography allows a powerful and precise chemo-dynamical view of the Milky Way with unprecedented spatial coverage and statistical robustness. First, it reveals the strong vertical symmetry of the Galaxy and the flared structure of the disc. Second, the observed kinematic disturbances of the disc -- seen as phase space correlations -- and kinematic or orbital substructures are associated with chemical patterns that favour stars with enhanced metallicities and lower [alpha/Fe] abundance ratios compared to the median values in the radial distributions. This is detected both for young objects that trace the spiral arms and older populations. Several alpha, iron-peak elements and at least one heavy element trace the thin and thick disc properties in the solar cylinder. Third, young disc stars show a recent chemical impoverishment in several elements. Fourth, the largest chemo-dynamical sample of open clusters analysed so far shows a steepening of the radial metallicity gradient with age, which is also observed in the young field population. Finally, the Gaia chemical data have the required coverage and precision to unveil galaxy accretion debris and heated disc stars on halo orbits through their [alpha/Fe] ratio, and to allow the study of the chemo-dynamical properties of globular clusters. Gaia DR3 chemo-dynamical diagnostics open new horizons before the era of ground-based wide-field spectroscopic surveys. Abstract: International audience; ONERA – The French Aerospace Lab – develops new concepts of LiDAR imaging systems including new sensor technologies and data processing. The rising complexities and costs of high performance systems, and the shrinking time to design drove the ONERA approach. The home-grown MATLIS software has been evolving for the past decade. It allows both linear mode LiDAR and single photon electro-optical systems simulation (both GmAPD and SPL) embedded on dynamic platforms (eg. UAVs, Aircrafts).In this paper, we present an algorithm for 3D scene reconstruction with unprecedented lateral resolution. This method is suitable for Geiger mode Avalanche Photodiode (GmAPD) 3D focal plane arrays. The main idea behind of this work is reconstruction of higher resolution 3D images by sub-pixel spatial modulation.We manage to reconstruct sub-pixel information by using Compressive Sensing (CS) algorithms, such as Orthogonal Matching Pursuit (OMP). Then we have verified our theoretical approach using simulated 3D images generated using the MATLIS software from a facetted three-dimensional scene.; L'ONERA - Laboratoire français de l'aérospatiale - développe de nouveaux concepts de systèmes d'imagerie LiDAR incluant de nouvelles technologies de capteurs et de traitement de données. La complexité et les coûts croissants des systèmes hautes performances et la réduction des délais de conception ont conduit l'approche de l'ONERA. Le logiciel maison MATLIS a évolué au cours de la dernière décennie. Il permet à la fois la simulation de LiDAR en mode linéaire et de systèmes électro-optiques à photon unique (à la fois GmAPD et SPL) embarqués sur des plates-formes dynamiques (par exemple, drones, avions).Dans cet article, nous présentons un algorithme de reconstruction de scène 3D avec une résolution latérale sans précédent. Cette méthode convient aux matrices de plans focaux 3D à photodiode à avalanche en mode Geiger (GmAPD). L'idée principale derrière ce travail est la reconstruction d'images 3D à plus haute résolution par modulation spatiale sous-pixel.Nous parvenons à reconstruire des informations sous-pixels en utilisant des algorithmes de détection de compression (CS), tels que Orthogonal Matching Pursuit (OMP). Ensuite, nous avons vérifié notre approche théorique à l'aide d'images 3D simulées générées à l'aide du logiciel MATLIS à partir d'une scène tridimensionnelle à facettes.

Abstract: The Gaia DR3 Catalogue contains for the first time about eight hundred thousand solutions with either orbital elements or trend parameters for astrometric, spectroscopic and eclipsing binaries, and combinations of them. This paper aims to illustrate the huge potential of this large non-single star catalogue. Using the orbital solutions together with models of the binaries, a catalogue of tens of thousands of stellar masses, or lower limits, partly together with consistent flux ratios, has been built. Properties concerning the completeness of the binary catalogues are discussed, statistical features of the orbital elements are explained and a comparison with other catalogues is performed. Illustrative applications are proposed for binaries across the H-R diagram. The binarity is studied in the RGB/AGB and a search for genuine SB1 among long-period variables is performed. The discovery of new EL CVn systems illustrates the potential of combining variability and binarity catalogues. Potential compact object companions are presented, mainly white dwarf companions or double degenerates, but one candidate neutron star is also presented. Towards the bottom of the main sequence, the orbits of previously-suspected binary ultracool dwarfs are determined and new candidate binaries are discovered. The long awaited contribution of Gaia to the analysis of the substellar regime shows the brown dwarf desert around solar-type stars using true, rather than minimum, masses, and provides new important constraints on the occurrence rates of substellar companions to M dwarfs. Several dozen new exoplanets are proposed, including two with validated orbital solutions and one super-Jupiter orbiting a white dwarf, all being candidates requiring confirmation. Abstract: International audience; Dans un objectif applicatif de fournir aux facteurs d’instruments à vent des outils de conception et de simulation, de nombreuses méthodes, à la fois de caractérisation et de simulation du comportement des propriétés acoustiques ont été développées. Certains de ces outils sont mis à la disposition des artisans via la Plateforme d’Aide à la Facture Instrumentale (PAFI). En parallèle d’un benchmark des modèles numériques pour leur vérification, une campagne expérimentale est ici présentée afin de créer une base de données exhaustive et de référence pour la validation des modèles (les notions de vérification et validation se basent sur les critères de l’ASME), et l’estimation des incertitudes associées aux mesures. En particulier, l’incertitude aléatoire inter/intra mesures et inter/intra échantillons est quantifiée. La campagne expérimentale se base sur de nombreux lots de cinq éprouvettes chacun. Le plan d’expérience permet d’obtenir des résultats expérimentaux consolidés concernant la conicité, les trous latéraux, les effets de bords, les conditions aux limites et les matériaux. Les éprouvettes fabriquées vont être testées dans au moins huit lieux différents présentant des méthodes de caractérisation variables. La première campagne ayant eu lieu permet d’établir un protocole de mesure reproductible. Cette campagne donne de premières estimations de la variabilité des mesures et permet de tester différents aspects du plan d’expérience.

Abstract: International audience; Dans l’objectif de mettre à jour la Plateforme d’Aide à la Facture Instrumentale Pafi, nous proposons ici de recenser et comparer les outils existants permettant de calculer l’impédance d’entrée d’instruments à vent. Un groupe de travail a été constitué, réunissant une dizaine d’équipes de recherche françaises expertes dans la modélisation et les méthodes de résolution par matrice de transfert, éléments finis 1D, éléments finis 3D, méthodes multimodales, guides d’ondes, etc. Ce groupe a défini un ensemble de géométries étalons de résonateurs mettant en jeu différentes conditions de propagation (cylindre, cône, coude, trou latéral, etc.) ou de rayonnement (fermé, avec baffle, etc.). Les impédances d’entrée de ces tubes ont été calculées en utilisant l'expertise des membres du groupe de travail puis mises en commun sur une plateforme collaborative. La base de données ainsi constituée peut être utilisée pour mener des études comparatives, la vérification de modèles et la comparaison avec une base expérimentale. Son analyse permettra notamment de faire un choix éclairé des méthodes et modèles à implémenter dans un outil d’aide à la facture en considérant leur validité, leur versatilité, leur complexité d’implémentation et les ressources de calcul nécessaires. Après avoir présenté la mise en place de ce projet et l’accessibilité de la base de données, nous discuterons des résultats obtenus sur quelques cas particuliers.

Abstract: International audience; Le jeu des instruments à vent historiques est bien souvent incompatible avec la déontologie et les pratiques de conservation protégeant les collections muséales. Certaines des propriétés musicales et sonores de ces instruments restent donc inconnues. Les modèles acoustiques peuvent prédire certaines de ces propriétés à partir de connaissances sur leurs géométries. Grâce à une collaboration entre l'équipe Makutu d'Inria Bordeaux Sud-Ouest, la Cité de la Musique – Philharmonie de Paris, l'Institut Technologique Européen des Métiers de la Musique (ITEMM) du Mans, et le Centre de Recherche et de Restauration des Musées de France (C2RMF), nous proposons une démarche ici appliquée au fonds muséal issu de la famille Besson (grande entreprise de facture instrumentale française), sur une collection de trompettes naturelles du début du XXe siècle. La géométrie des instruments a été mesurée de manière non invasive par tomographie à rayons X réalisée au C2RMF. Après extraction de leurs perces (évolution du diamètre interne), leur impédance d'entrée a été calculée. Pour l'une des trompettes (numéro: E.0925) le résultat des simulations a pu être comparé à des mesures avec un excellent accord. Malgré de nombreux inconnus sur le contrôle de ces instruments, il est également possible de simuler le son de ces instruments. Un fac-similé de cette même trompette a été construit à partir de l'imagerie par rayon X. Cela permet de comparer ces sons de synthèses à ceux produits par un musicien. Au-delà de l'intérêt patrimonial, cette étude permet de confronter les outils numériques de modélisation à un cas concret et d'en tester les limites.

Abstract: International audience; This study aimed at investigating the synergistic effects of essential fatty acids (EFA) and conjugated linoleic acids (CLA) on the liver proteome profile of dairy cows during the transition to lactation. 16 Holstein cows were infused from 9 wk. antepartum to 9 wk. postpartum into the abomasum with either coconut oil (CTRL) or a mixture of EFA (linseed + safflower oil) and CLA (EFA + CLA). Label-free quantitative proteomics was performed in liver tissue biopsied at days -21, +1, +28, and + 63 relative to calving. Differentially abundant proteins (DAP) between treatment groups were identified at the intersection between a multivariate and a univariate analysis. In total, 1680 proteins were identified at each time point, of which between groups DAP were assigned to the metabolism of xenobiotics by cytochrome P450, drug metabolism - cytochrome P450, steroid hormone biosynthesis, glycolysis/gluconeogenesis, and glutathione metabolism. Cytochrome P450, as a central hub, enriched with specific CYP enzymes comprising: CYP51A1 (d - 21), CYP1A1 & CYP4F2 (d + 28), and CYP4V2 (d + 63). Collectively, supplementation of EFA + CLA in transition cows impacted hepatic lipid metabolism and enriched several common biological pathways at all time points that were mainly related to ω-oxidation of fatty acids through the Cytochrome p450 pathway. SIGNIFICANCE: In three aspects this manuscript is notable. First, this is among the first longitudinal proteomics studies in nutrition of dairy cows. The selected time points are critical periods around parturition with profound endocrine and metabolic adaptations. Second, our findings provided novel information on key drivers of biologically relevant pathways suggested according to previously reported performance, zootechnical, and metabolism data (already published elsewhere). Third, our results revealed the role of cytochrome P450 that is hardly investigated, and of ω-oxidation pathways in the metabolism of fatty acids with the involvement of specific enzymes.

Abstract: International audience; Repeated measurements analysis of variance - simultaneous component analysis (ASCA) has been developed to handle complex longitudinal omics datasets and combine novel information with existing data. Herein, we aimed at applying ASCA to 64 liver proteomes collected at 4-time points (day -21, +1, +28, and + 63 relative to parturition) from 16 Holstein cows treated from 9 wk. antepartum to 9 wk. postpartum (PP) with coconut oil (CTRL) or a mixture of essential fatty acids (EFA) and conjugated linoleic acid (CLA) (EFA + CLA). The ASCA modeled 116, 43, and 97 differentially abundant proteins (DAP) during the transition to lactation, between CTRL and EFA + CLA, and their interaction, respectively. Time-dependent DAP were annotated to pathways related to the metabolism of carbohydrates, FA, and amino acid in the PP period. The DAP between FA and the interaction effect were annotated to the metabolism of xenobiotics by cytochrome P450, drug metabolism - cytochrome P450, retinol metabolism, and steroid hormone biosynthesis. Collectively, ASCA provided novel information on molecular markers of metabolic adaptations and their interactions with EFA + CLA supplementation. Bioinformatics analysis suggested that supplemental EFA + CLA amplified hepatic FA oxidation; cytochrome P450 was enriched to maintain metabolic homeostasis by oxidation/detoxification of endogenous compounds and xenobiotics.Significance: This report is among the first ones applying repeated measurement analysis of variance-simultaneous component analysis (ASCA) to deal with longitudinal proteomics results. ASCA separately identified differentially abundant proteins (DAP) in 'transition time', 'between fatty acid treatments', and 'their interaction'. We first identified the molecular signature of hepatic metabolic adaptations during postpartum negative energy balance; the enriched pathways were well-known pathways related to mobilizing fatty acids (FA) and amino acids to support continuous energy production through fatty acid oxidation, TCA cycle, and gluconeogenesis. Some of the DAP were not previously reported in transition dairy cows. Secondly, we provide novel information on the mechanisms by which supplemented essential FA and conjugated linoleic acids interact with hepatic metabolism. In this regard, FA amplified hepatic detoxifying and oxidation capacity through ligand activation of nuclear receptors. Finally, we briefly compared the strengths and weaknesses of the ASCA model with PLS-DA and outlined why these methods are complementary.

Abstract: International audience; Speciation of Cd in soil solutions strongly determines the fate of this toxic metal in the environment. Generally, in soil solutions, Cd predominantly binds to the dissolved organic matter (DOM). The determination of the quantity and reactivity of DOM that actually complexes Cd in soil solutions is challenging for operational purposes. Therefore, this study tested whether Cd2+ concentration in soil solutions could effectively be predicted by considering complexation with a single mean organic ligand assumed to be a fraction of DOM of unspecified nature or assumed to be purely fulvic acids (FA) with reactivity as described in WHAM VII. The reactivity of the unspecified ligand and the concentration of FA were modelled and fitted to experimental data from 76 agricultural soils with low Cd contents. The optimal reactivity and FA concentration that minimized the relative error (RE) of predictions of the concentration of Cd2+ in soil solutions were either considered constant across soils or modelled from soil properties by multiple linear regressions (MLR) or random forests (RF), giving 6 models, the predictive value of which was assessed by 10-folds cross-validation. When the reactivity of the mean ligand and the optimal FA concentration were considered constant across soils, the models were biased and 66.9% of predictions had relative errors below a factor of 2. By contrast, if the reactivity of the mean ligand or the optimal FA concentration were allowed to vary with soil characteristics, these performances increased to 95.5%, soil pH being the main predictor and RF being slightly more efficient than MLR. With more than 95% of the relative errors of prediction below a factor of 2, the models developed in this work could be valuable for assessing Cd speciation in the solution of soils having a low Cd content.

Abstract: Bacteria adapt to versatile environments by modulating gene expression through a set of stress response regulators, alternative Sigma factors, or two-component systems. Among the central processes that must be finely tuned is membrane homeostasis, including synthesis of phospholipids (PL). However, few genetic regulations of this process have been reported. We have previously shown that the gene coding the first step of PL synthesis is regulated by σE and ppGpp, and that the BasRS (PmrAB) two component system controls the expression of the DgkA PL recycling enzyme. The gene coding for phosphatidylserine decarboxylase, the last step in phosphatidylethanolamine synthesis is another gene in the PL synthesis pathway susceptible of stress response regulation. Indeed, psd appears in transcriptome studies of the σE envelope stress Sigma factor and of the CpxAR two component system. Interestingly, this gene is presumably in operon with mscM coding for a miniconductance mechanosensitive channel. In this study, we dissected the promoter region of the psd-mscM operon and studied its regulation by σE and CpxR. By artificial activation of σE and CpxRA stress response pathways, using GFP transcriptional fusion and western-blot analysis of Psd and MscM enzyme production, we showed that the operon is under the control of two distinct promoters. One is activated by σE, the second is activated by CpxRA and also responsible for basal expression of the operon. The fact that the phosphatidylethanolamine synthesis pathway is controlled by envelope stress responses at both its first and last steps might be important for adaptation of the membrane to envelope perturbations.

Abstract: International audience; The Microbacterium sp. C448 was isolated from a soil regularly exposed to sulfamethazine(SMZ), for its ability to partly mineralise this antibiotic and other related sulfonamides.The aim of our study was to explore its metabolic adaptation towards exposure to SMZenvironmental (10 mg/L) and medicinal (250 mg/L) concentrations. Its transcriptomic andproteomic responses were analysed by focusing on the degradation regulon (sad genes) andresistance genes (folP and sul1).The transcriptomic and proteomic results were essentially congruent whatever theconcentrations tested. In culture conditions, exposure to the highest concentration of SMZ led tothe highest sad expression and Sad production, confirming these proteins’ role insulfamethazine degradation in cellulo. Moreover, when sulfamethazine was completelydegraded, Sad production tends to return to the basal level observed under control conditions.Congruence of transcriptomic and proteomic results was also observed for resistance genes.Although Dihydropteroate synthase (DHPS)-Sul1 protein was 100-fold more produced than thatof DHPS-FolP, both were expressed at a basal level and were not affected by SMZ exposure.Moreover, non-targeted analyses showed overexpression and overproduction of a putativesulphate exporter W0Z8D9 in Microbacterium sp. C448 exposed to the highest SMZconcentration. Abstract: The Gaia DR3 Catalogue contains for the first time about eight hundred thousand solutions with either orbital elements or trend parameters for astrometric, spectroscopic and eclipsing binaries, and combinations of them. This paper aims to illustrate the huge potential of this large non-single star catalogue. Using the orbital solutions together with models of the binaries, a catalogue of tens of thousands of stellar masses, or lower limits, partly together with consistent flux ratios, has been built. Properties concerning the completeness of the binary catalogues are discussed, statistical features of the orbital elements are explained and a comparison with other catalogues is performed. Illustrative applications are proposed for binaries across the H-R diagram. The binarity is studied in the RGB/AGB and a search for genuine SB1 among long-period variables is performed. The discovery of new EL CVn systems illustrates the potential of combining variability and binarity catalogues. Potential compact object companions are presented, mainly white dwarf companions or double degenerates, but one candidate neutron star is also presented. Towards the bottom of the main sequence, the orbits of previously-suspected binary ultracool dwarfs are determined and new candidate binaries are discovered. The long awaited contribution of Gaia to the analysis of the substellar regime shows the brown dwarf desert around solar-type stars using true, rather than minimum, masses, and provides new important constraints on the occurrence rates of substellar companions to M dwarfs. Several dozen new exoplanets are proposed, including two with validated orbital solutions and one super-Jupiter orbiting a white dwarf, all being candidates requiring confirmation. Abstract: International audience; CANVAS caused by RFC1 biallelic expansions is a major cause of inherited sensory neuronopathy. Detection of RFC1 expansion is challenging and CANVAS can be associated with atypical features. We clinically and genetically characterized 50 patients, selected based on the presence of sensory neuronopathy confirmed by EMG. We screened RFC1 expansion by PCR, repeat-primed PCR, and Southern blotting of long-range PCR products, a newly developed method. Neuropathological characterization was performed on the brain and spinal cord of one patient. Most patients (88%) carried a biallelic (AAGGG)n expansion in RFC1. In addition to the core CANVAS phenotype (sensory neuronopathy, cerebellar syndrome, and vestibular impairment), we observed chronic cough (97%), oculomotor signs (85%), motor neuron involvement (55%), dysautonomia (50%), and parkinsonism (10%). Motor neuron involvement was found for 24 of 38 patients (63.1%). First motor neuron signs, such as brisk reflexes, extensor plantar responses, and/or spasticity, were present in 29% of patients, second motor neuron signs, such as fasciculations, wasting, weakness, or a neurogenic pattern on EMG in 18%, and both in 16%. Mixed motor and sensory neuronopathy was observed in 19% of patients. Among six non-RFC1 patients, one carried a heterozygous AAGGG expansion and a pathogenic variant in GRM1. Neuropathological examination of one RFC1 patient with an enriched phenotype, including parkinsonism, dysautonomia, and cognitive decline, showed posterior column and lumbar posterior root atrophy. Degeneration of the vestibulospinal and spinocerebellar tracts was mild. We observed marked astrocytic gliosis and axonal swelling of the synapse between first and second motor neurons in the anterior horn at the lumbar level. The cerebellum showed mild depletion of Purkinje cells, with empty baskets, torpedoes, and astrogliosis characterized by a disorganization of the Bergmann's radial glia. We found neuronal loss in the vagal nucleus. The pars compacta of the substantia nigra was depleted, with widespread Lewy bodies in the locus coeruleus, substantia nigra, hippocampus, entorhinal cortex, and amygdala. We propose new guidelines for the screening of RFC1 expansion, considering different expansion motifs. Here, we developed a new method to more easily detect pathogenic RFC1 expansions. Abstract: International audience; Objectives: Digestive perianastomotic ulcerations (DPAU) resembling Crohn disease lesions are long-term complications of intestinal resections, occurring in children and young adults. They are known to be uncommon, severe and difficult to treat.Methods: In the absence of recommendations, we performed a large European survey among the members of the ESPGHAN working group on inflammatory bowel disease (IBD) in order to collect the experience of expert pediatric gastroenterologists on DPAU.Results: Fifty-one patients (29 boys and 22 girls) were identified from 19 centers in 8 countries. Most patients were followed after necrotizing enterocolitis (n = 20) or Hirschsprung disease (n = 11). The anastomosis was performed at a median age (interquartile range) of 6 [1–23] months, and first symptoms occurred 39 [22–106] months after surgery. Anemia was the most prevalent symptom followed by diarrhea, abdominal pain, bloating, and failure to thrive. Hypoalbuminemia, elevated CRP, and fecal calprotectin were common. Deep ulcerations were found in 59% of patients usually proximally to the anastomosis (68%). During a median follow-up of 40 [19–67] months, treatments reported to be the most effective included exclusive enteral nutrition (31/35, 88%), redo anastomosis (18/22, 82%), and alternate antibiotic treatment (37/64, 58%).Conclusions: Unfortunately, persistence of symptoms, failure to thrive, and abnormal laboratory tests at last follow-up in most of patients show the burden of DPAU lacking optimal therapy and incomplete understanding of the pathophysiology.

Abstract: International audience; Context. Gaia ’s Early Third Data Release (EDR3) does not contain new radial velocities because these will be published in Gaia ’s full third data release (DR3), expected in the first half of 2022. To maximise the usefulness of EDR3, Gaia ’s second data release (DR2) sources (with radial velocities) are matched to EDR3 sources to allow their DR2 radial velocities to also be included in EDR3. This presents two considerations: (i) a list of 70 365 sources with potentially contaminated DR2 radial velocities has been published; and (ii) EDR3 is based on a new astrometric solution and a new source list, which means sources in DR2 may not be in EDR3. Aims. The two aims of this work are: (i) investigate the list in order to improve the DR2 radial velocities being included in EDR3 and to avoid false-positive hypervelocity candidates; and (ii) match the DR2 sources (with radial velocities) to EDR3 sources. Methods. Thetwo methods of this work are: (i) unpublished, preliminary DR3 radial velocities of sources on the list, and high-velocity stars not on the list, are compared with their DR2 radial velocities to identify and remove contaminated DR2 radial velocities from EDR3; and (ii) proper motions and epoch position propagation is used to attempt to match all sources with radial velocities in DR2 to EDR3 sources. The comparison of DR2 and DR3 radial velocities is used to resolve match ambiguities. Results. EDR3 contains 7 209 831 sources with a DR2 radial velocity, which is 99.8% of sources with a radial velocity in DR2 (7 224 631). 14 800 radial velocities from DR2 are not propagated to any EDR3 sources because (i) 3871 from the list are found to either not have a DR3 radial velocity or it differs significantly from its DR2 value, and five high-velocity stars not on the list are confirmed to have contaminated radial velocities, in one case because of contamination from the non-overlapping Radial Velocity Spectrometer windows of a nearby, bright star; and ( ii ) 10 924 DR2 sources could not be satisfactorily matched to any EDR3 sources, so their DR2 radial velocities are also missing from EDR3. Conclusions. The reliability of radial velocities in EDR3 has improved compared to DR2 because the update removes a small fraction of erroneous radial velocities (0.05% of DR2 radial velocities and 5.5% of the list). Lessons learnt from EDR3 (e.g. bright star contamination) will improve the radial velocities in future Gaia data releases. The main reason for radial velocities from DR2 not propagating to EDR3 is not related to DR2 radial velocity quality. It is because the DR2 astrometry is based on one component of close binary pairs, while EDR3 astrometry is based on the other component, which prevents these sources from being unambiguously matched.

Abstract: International audience; Objective: In rheumatoid arthritis, the association between advanced therapies (including biologic disease-modifying antirheumatic drugs [DMARDs] and targeted synthetic DMARDs) and methotrexate (MTX) is recommended by international societies. When MTX cannot be used, other conventional synthetic DMARDs (csDMARDs) may be proposed. We aimed to compare the safety and efficacy of MTX and non-MTX csDMARDs in combination with advanced therapies.Methods: We systematically searched the literature for studies comparing the effectiveness, retention rate, and safety of MTX versus non-MTX csDMARDs (leflunomide or others) in combination with tumor necrosis factor inhibitors (TNFi), abatacept, rituximab, tocilizumab, and JAK inhibitors. Meta-analysis was performed with RevMan, using an inverse variance approach with fixed or random-effects models. Risk ratios (RRs) and 95% confidence intervals (95% CIs) were estimated.Results: The literature search revealed 3,842 articles; 41 studies were included for the systematic literature review and 21 for the meta-analysis: 13 with TNFi, 3 with abatacept, and 5 with rituximab. For TNFi, the European Alliance of Associations for Rheumatology (EULAR) response at 6 months was lower for patients receiving non-MTX csDMARDs than for those using MTX (RR 0.93 [95% CI 0.87, 1.0], P = 0.04; n = 3,843; I2 = 28%), with a lower retention rate at 12 months. For abatacept, effectiveness and safety were similar between the 2 groups. For rituximab, a good EULAR response was higher with leflunomide than MTX (RR 1.38 [95% CI 1.13, 1.68], P = 0.001; n = 2,078; I2 = 0%), with similar adverse event rates. Meta-analysis for tocilizumab or JAK inhibitors could not be performed.Conclusion: The different csDMARDs seem safe and efficient to combine with advanced therapies in RA patients. Although MTX seems slightly superior to other csDMARDs in combination with TNFi, leflunomide might be superior to MTX in combination with rituximab.

Abstract: International audience; Label free shotgun proteomics was used to analyse plasma and Longissimus muscle biopsies of Limousin-sired bulls, classified as 5 high-quality and 5 low-quality meat based on sensory texture traits (tenderness, juiciness and chewiness). A total of 31 putative protein biomarkers (16 in plasma and 15 in muscle) differed significantly in abundance between the two quality groups. The proteins were associated with muscle structure, energy metabolism, heat shock proteins, oxidative stress and proteolysis related pathways. Among them, B2M, AHSG, APOA4 and HP-20 (plasma), PFKM, MYH2, PTER, GSTM1 and MYPN (muscle) were good predictors of the three texture quality traits. Further, significant correlations were identified for FETUB, SERPINA7, ASL, TREH, HP, HP25, AZGP1, APCS and SYT15, which are novel biomarkers from plasma that warrant further evaluation. This study is a significant step forward in elucidating proteomic profiles in bovine bio-fluids and muscle tissue, which may ultimately provide opportunities to processors for early assessment of beef sensory quality.

Abstract: International audience; Background: Few data exist concerning genotype-phenotype relationships in left ventricular noncompaction (LVNC).Methods and results: From a multicenter French Registry, we report the genetic and clinical spectrum of 95 patients with LVNC, and their genotype-phenotype relationship. Among the 95 LVNC, 45 had at least 1 mutation, including 14 cases of mutation in ion channel genes. In a complementary analysis including 16 additional patients with ion channel gene mutations, for a total of 30 patients with ion channel gene mutation, we found that those patients had higher median LV ejection fraction (60% vs 40%; P < .001) and more biventricular noncompaction (53.1% vs 18.5%; P < .001) than the 81 other patients with LVNC. Among them, both the 19 patients with an HCN4 mutation and the 11 patients with an RYR2 mutation presented with a higher LV ejection fraction and more frequent biventricular noncompaction than the 81 patients with LVNC but with no mutation in the ion channel gene, but only patients with HCN4 mutation presented with a lower heart rate.Conclusions: Ion channel gene mutations should be searched systematically in patients with LVNC associated with either bradycardia or biventricular noncompaction, particularly when LV systolic function is preserved. Identifying causative mutations is of utmost importance for genetic counselling of at-risk relatives of patients affected by LVNC.

Abstract: International audience; Objective: To investigate the impact of baseline and time-varying factors on the risk of serious adverse events (SAEs) in patients during long-term certolizumab pegol (CZP) treatment.Methods: Safety data were pooled across 34 CZP clinical trials in rheumatoid arthritis (RA), axial spondyloarthritis (axSpA), psoriatic arthritis (PsA), and plaque psoriasis (PSO). Cox proportional hazards modeling was used to investigate the association of baseline patient characteristics with risk of serious infectious events (SIEs), malignancies, and major adverse cardiac events (MACEs). Cox modeling for recurrent events assessed the impact of time-varying body mass index (BMI), systemic corticosteroid (CS) use, and disease activity on SIE risk in RA and SAE risk in PSO.Results: Data were pooled from 8747 CZP-treated patients across indications. Cox models reported a 44% increase in SIE risk associated with a baseline BMI of 35 kg/m2 or more versus a baseline BMI of 18.5 kg/m2 to less than 25 kg/m2 . Baseline systemic CS use, age of 65 years or more, and disease duration of 10 years or longer also increased SIE risk. Older age was the only identified risk factor for malignancies. The risk of MACEs increased 107% for BMI of 35 kg/m2 or more versus BMI of 18.5 kg/m2 to less than 25 kg/m2 and increased 51% for men versus women. Higher disease activity, older age, systemic CS use, BMI of 35 kg/m2 or more, and baseline comorbidities were SIE risk factors in RA. Age and systemic CS use were risk factors for SAEs in PSO.Conclusion: Age, BMI, systemic CS use, and disease activity were identified as SIE risk factors in CZP-treated patients. Risk of malignancies was greater in older patients, whereas obesity and male sex were MACE risk factors.

Abstract: Les dessins des environnements futurs dans lesquels l’être humain évoluera sont « l’instrument d’un contrôle exercé par l’architecte sur l’espace et les formes » mais aussi d’un contrôle sur l’habiter, les déplacements des individus, et le sens donné à l’environnement. L’espace est une ressource primordiale pour l’être humain, tant dans son organisation personnelle que dans ses liens sociaux (Tversky, 2003). Si la co-construction entre l’être humain et son environnement (Varela et al., 1993), pousse à considérer la cognition comme liée à son environnement et à l’état corporel de l’individu (Wilson, 2002), la space syntax (Hillier et al., 1987) considère que les déplacements dans l’espace construit dépendraient essentiellement d’indices extrasomatiques évalués par des mesures allocentriques. Nous présentons ici cette approche en regard de théories incarnées et situées de la cognition, avant d’ouvrir l’analyse aux Smart Buildings et Smart Cities, dont les technologies de l’information et de la communication (TIC) offrent de nouvelles voies d’interaction sociale et de perception de l’espace qui renouvellent le cadre physique actuellement pris en compte, à notre connaissance, dans la space syntax.

Abstract: Diffusion couples of Ti and biphasic alloy Al-7wt%Si were produced and heat-treated at 535 °C for between 1 and 200 hours. The nature and thickness of the phases present in the reaction layer sequence were characterized using conventional techniques such as microprobe analysis and transmission electron microscopy. For short interaction times the interfacial reaction layer consists mainly of silicides, whereas aluminides are formed after times longer than 40h. This variation in the spatial reaction layer sequence over time is explained by the biphasic nature of one of the diffusion couple end-members, with high activity for both elements, and by the huge difference between the composition of the first phase formed at the initial interface and the average composition of the biphasic end-member. This situation, which can be observed in other systems, illustrates the difficulty of confirming that a true diffusion path has been attained in a diffusion couple. Most of the criteria that were first proposed by Kirkaldy and Brown, can apparently be fulfilled, but the system could continue shifting over time towards a more stable configuration. In some systems, as in the case of Al-Si-Ti, the slow kinetics can be problematic, making it impossible to predict a radical change in material properties.

Abstract: International audience; It is generally alleged that the design choices of acoustic guitar bracing patterns lead to a specific sound of the instrument. However, in the presence of strong uncertainties due to variability of material properties and climatic conditions, the robustness of the soundboard dynamics has yet to be investigated. In this study, three types of bracing patterns are studied using physics-based models and stochastic analyses are performed to account for material and climatic uncertainties. It is shown that the choice of a brace design leads, at least in the low frequency domain, to a dynamic behaviour that is not stackable with another design, even in the presence of strong aleatory uncertainties. This assessment supports the conjecture that guitar brace design choices have a greater impact than material variability where guitar soundboard dynamics are concerned. More generally, these results illustrate the usefulness of detailed physics-based models in the understanding, design and making of guitars.

Abstract: High-k nanocomposite photoresists are highly sought-after dielectric materials for the manufacturing and miniaturization of integrated capacitors. Herein, in-depth high voltage dielectric characterizations of Metal Polymer Composites (MPCs) having carbon-coated nickel nanoparticles (Ni@C) dispersed into epoxy-based photoresist (SU-8) are reported. The MPCs were fabricated using three-step formulation (deagglomeration, surface functionalization and dispersion in a polymeric matrix) involving simple sonochemical methods. Finally, films of MPCs were deposited on silicon wafers by spin-coating, and Metal-Insulator-Semiconductor (MIS) capacitors were fabricated. Formulations with increasing nanoparticles/polymer volume fractions were prepared in order to determine a previously unestablished percolation threshold with (Ni@C) nanoparticles. The experimental results were modeled using the generalized power-law equation of the percolation theory (PT) and were compared with effective medium approximation (EMA). An enhancement of the complex dielectric permittivity of ∼ 116% was detected keeping a reasonable value of losses of 0.32 at 5 kHz. However, no giant permittivity in vicinity of the percolation threshold was detected. The high-voltage dielectric properties showed the occurrence of two electron field emission mechanisms, but no electrical aging or dielectric breakdown below ±0.38 MV/cm occurred. This study shows a reliable wafer-scale film fabrication process of MPCs for the manufacturing of long-awaited miniaturization of high-voltage capacitors.

Abstract: International audience; Objectives Data are scarce regarding the potential clinical differences between non-late onset schizophrenia (NLOS, i.e., disorder occurring before 40 years of age), late-onset schizophrenia (LOS, occurring between ages 40 and 60 years) and very-late-onset schizophrenia-like psychosis (VLOSLP, occurring after 60 years of age). Furthermore, previous research compared LOS patients with non-age matched NLOS patients. In this study, we sought to examine potential clinical differences between patients of similar age with LOS and NLOS. Methods/Design This is a cross-sectional multicentre study that recruited in- and outpatients older adults (aged >= 55 years) with an ICD-10 diagnosis of schizophrenia or schizoaffective disorder with NLOS and LOS. Sociodemographic and clinical characteristics, comorbidity, psychotropic medications, quality of life, functioning, and mental health care utilization were drawn for comparison. Results Two hundred seventy-two participants (79.8%) had NLOS, 61 (17.9%) LOS, and 8 (2.3%) VLOSLP. LOS was significantly and independently associated with greater severity of emotional withdrawal and lower severity of depression (all p < 0.05). However, the magnitude of these associations was modest, with significant adjusted odds ratios ranging from 0.71 to 1.24, and there were no significant between-group differences in other characteristics. Conclusion In an age-matched multicenter sample of elderly patients with schizophrenia, older adults with LOS were largely similar to older adults with NLOS in terms of clinical characteristics. The few differences observed may be at least partially related to symptom fluctuation with time. Implications of these findings for pharmacological and nonpharmacological management is yet to be determined.

Abstract: Exaggerated sexually selected traits, often carried by males, are characterized by the evolution of hyperallometry, resulting in their disproportionate growth relative to the rest of the body among individuals of the same population. While the evolution of allometry has attracted much attention for centuries, our understanding of the developmental genetic mechanisms underlying its emergence remains fragmented. Here we conduct comparative transcriptomics of the legs followed by an RNA interference (RNAi) screen to identify genes that play a role in the hyperallometric growth of the third legs in the males of the water strider Microvelia longipes. We demonstrate that a broadly expressed growth factor, Bone Morphogenetic Protein 11 (BMP11, also known as Growth Differentiation Factor 11), regulates leg allometries through increasing the allometric slope and mean body size in males. In contrast, BMP11 RNAi reduced mean body size but did not affect slope either in the females of M. longipes or in the males and females of other closely related Microvelia species. Furthermore, our data show that a tissue-specific factor, Ultrabithorax (Ubx), increases intercept without affecting mean body size. This indicates a genetic correlation between mean body size and variation in allometric slope, but not intercept. Strikingly, males treated with BMP11 RNAi exhibited a severe reduction in fighting frequency compared to both controls and Ubx RNAi-treated males. Therefore, male body size, the exaggerated weapon, and the intense fighting behavior associated with it are genetically correlated in M. longipes. Our results support a possible role of pleiotropy in the evolution of allometric slope., Males of the water strider Microvelia longipes possess extremely long legs used as weapons to dominate egg-laying sites; this study shows that the growth factor BMP11 is responsible for increasing body size, leg length and fighting behaviour in these males.
