Abstract: International audience; Background The identification of patients at high risk of a disabling disease course would be invaluable in guiding initial therapy in Crohn’s disease (CD). Our objective was to evaluate a combination of clinical, serological, and genetic factors to predict complicated disease course in pediatric-onset CD. Methods Data for pediatric-onset CD patients, diagnosed before 17 years of age between 1988 and 2004 and followed more than 5 years, were extracted from the population-based EPIMAD registry. The main outcome was defined by the occurrence of complicated behavior (stricturing or penetrating) and/or intestinal resection within the 5 years following diagnosis. Lasso logistic regression models were used to build a predictive model based on clinical data at diagnosis, serological data (ASCA, pANCA, anti-OmpC, anti-Cbir1, anti-Fla2, anti-Flax), and 369 candidate single nucleotide polymorphisms. Results In total, 156 children with an inflammatory (B1) disease at diagnosis were included. Among them, 35% (n = 54) progressed to a complicated behavior or an intestinal resection within the 5 years following diagnosis. The best predictive model (PREDICT-EPIMAD) included the location at diagnosis, pANCA, and 6 single nucleotide polymorphisms. This model showed good discrimination and good calibration, with an area under the curve of 0.80 after correction for optimism bias (sensitivity, 79%, specificity, 74%, positive predictive value, 61%, negative predictive value, 87%). Decision curve analysis confirmed the clinical utility of the model. Conclusions A combination of clinical, serotypic, and genotypic variables can predict disease progression in this population-based pediatric-onset CD cohort. Independent validation is needed before it can be used in clinical practice.

Thus, one of the technologies using combustion structures and containers of particular types, namely tunnel kilns and punnet containers, which had never been really characterized, will be analyzed with regard to its geographical distribution and a more detailed understanding of its chronology.; Les données acquises depuis plus de 20 ans sur la production protohistorique de sel dans l’Ouest de la France offrent aujourd’hui l’opportunité d’une nouvelle réflexion sur les différentes technologies mises en œuvre, parallèlement ou successivement. Ainsi, l’une des technologies mettant en œuvre des structures de combustion et des moules de types particuliers, à savoir des fours couloir ou fours à pont et des barquettes, qui n’avait jamais été réellement caractérisée, sera analysée au regard de sa distribution géographique et d’une appréhension plus fine de sa chronologie.

Abstract: International audience; Background: Preoperative COVID-19 has been associated with excess postoperative morbi-mortality. Consequently, guidelines were developed that recommended the postponement of surgery for at least 7 weeks after the infection. We hypothesised that vaccination against the SARS-CoV-2 and the large predominance of the Omicron variant attenuated the effect of a preoperative COVID-19 on the occurrence of postoperative respiratory morbidity.Methods: We conducted a prospective cohort study in 41 French centres between 15 March and 30 May 2022 (ClinicalTrials NCT05336110), aimed at comparing the postoperative respiratory morbidity between patients with and without preoperative COVID-19 within 8 weeks prior to surgery. The primary outcome was a composite outcome combining the occurrence of pneumonia, acute respiratory failure, unexpected mechanical ventilation, and pulmonary embolism within the first 30 postoperative days. Secondary outcomes were 30-day mortality, hospital length-of-stay, readmissions, and non-respiratory infections. The sample size was determined to have 90% power to identify a doubling of the primary outcome rate. Adjusted analyses were performed using propensity score modelling and inverse probability weighting.Findings: Of the 4928 patients assessed for the primary outcome, of whom 92.4% were vaccinated against the SARS-CoV-2, 705 had preoperative COVID-19. The primary outcome was reported in 140 (2.8%) patients. An 8-week preoperative COVID-19 was not associated with increased postoperative respiratory morbidity (odds ratio 1.08 [95% CI 0.48-2.13]; p = 0.83). None of the secondary outcomes differed between the two groups. Sensitivity analyses concerning the timing between COVID-19 and surgery, and the clinical presentations of preoperative COVID-19 did not show any association with the primary outcome, except for COVID-19 patients with ongoing symptoms the day of surgery (OR 4.29 [1.02-15.8]; p = 0.04).Interpretation: In our Omicron-predominant, highly immunised population undergoing general surgery, a preoperative COVID-19 was not associated with increased postoperative respiratory morbidity.

Abstract: International audience; We present a paleomagnetic study of the ~10 km diameter Karla impact structure in Russia. We sampled the target carbonate rocks, and a yet undocumented fragmental melt-bearing lithic breccia layer. This impact breccia, which contains carbonate melt, is enriched in stoichiometric magnetite by a factor of ~15 compared to the target lithologies, and carries a stable natural remanent magnetization. The weak remanent magnetization and the presence of both normal and reverse polarities down to the centimeter scale indicate that the breccia does not carry a thermoremanent magnetization (TRM), but rather a chemical remanent magnetization (CRM). The presence of stoichiometric magnetite and the absence of TRM suggest that the magnetite was formed during relatively low-temperature postimpact hydrothermalism that affected the porous impact breccia layer. During this process, the breccia acquired a CRM. The paleomagnetic direction is compatible with a Cenozoic age for the impact event, but cannot bring more precise constraint on the age because of the stable position of the Eurasian plate over the last 60 Myr. However, the presence of both polarities indicates that mild hydrothermalism took place over a period of time long enough to span at least one reversal of the geomagnetic field, that is, over a time scale of the order of 100 kyr. This confirms that protracted hydrothermal systems associated with impact craters are long lived, even in relatively small craters such as Karla, and are key features of the geologic and environmental effects of impacts on Earth.

Abstract: International audience; Monoclonal antibodies targeting the PD-1/PD-L1 immune checkpoint have considerably improved the treatment of some cancers, but novel drugs, new combinations, and treatment modalities are needed to reinvigorate immunosurveillance in immune-refractory tumors. An option to elicit antitumor immunity against cancer consists of using approved and marketed drugs known for their capacity to modulate the expression and functioning of the PD-1/PD-L1 checkpoint. Here, we have reviewed several types of drugs known to alter the checkpoint, either directly via the blockade of PD-L1 or indirectly via an action on upstream effectors (such as STAT3) to suppress PD-L1 transcription or to induce its proteasomal degradation. Specifically, the repositioning of the approved drugs liothyronine, azelnidipine (and related dihydropyridine calcium channel blockers), niclosamide, albendazole/flubendazole, and a few other modulators of the PD-1/PD-L1 checkpoint (repaglinide, pimozide, fenofibrate, lonazolac, propranolol) is presented. Their capacity to bind to PD-L1 or to repress its expression and function offer novel perspectives for combination with PD-1 targeted biotherapeutics. These known and affordable drugs could be useful to improve the therapy of cancer.

Abstract: Immuno-therapy has become a leading strategy to fight cancer. Over the past few years, immuno-therapies using checkpoint inhibitor monoclonal antibodies (mAbs) against programmed death receptor 1 (PD-1) and programmed death ligand 1 (PD-L1) have demonstrated improved survival compared with chemotherapy. We describe the microwave-assisted synthesis and the characterization of an innovative series of synthetic compounds endowed with nanomolar activity against PD-L1. The properties of the compounds were characterized using several biophysical techniques including microscale thermophoresis (MST) and fluorescence resonance energy transfer (FRET) measurements. A few small molecules demonstrated a high affinity for human PD-L1, potently disrupted the PD-L1:PD-1 interaction and inhibited Src homology region 2 domain-containing phosphatase (SHP2) recruitment to PD-1. More than 30 molecules from the pyrazolone family have been synthesized and 5 highly potent "PD-L1 silencing compounds" have been identified, based on in vitro measurements. Structure-activity relationships have been defined and ADME properties were evaluated. The phenyl-pyrazolone unit offers novel perspectives to design PD-L1-targeting agents, potentially useful to combat cancer and other pathologies implicating the PD-1/PD-L1 checkpoint.

Abstract: While insemination at first estrus is not recommended, most studies agree on the benefits of early puberty, both for reproductive performance and sow longevity. In the swine population in France, sexual maturity is expected at ca. 200 days of age. Although age at puberty is crucial information, it is not recorded on farms. Since selection for low boar taint influences male puberty, it raises questions about its potential side effects on female reproductive ability. This study was carried out as part of a project that assessed impacts of selection against boar taint on the reproduction of Landrace females. The sexual maturity of 1878 gilts at ca. 200 days of age was evaluated on five farms using quantitative plasma progesterone analysis in two blood samples taken 10 days apart. Cutoff values of 2.5 or 5.0 ng/mL gave similar estimates of 19% immature gilts, with a large variation among farms (8-49%). Insemination was not performed for 4% of gilts, 9% were re-inseminated during the first cycle and 15% did not farrow. The rate of earlyreproduction difficulties varied greatly among farms. The influence of immaturity at 200 days was generally low, with a large farm effect. Further investigation is required to analyze long-term effects of late puberty.

13. Stress thermique prénatal ou postnatal : conséquences sur le comportement, la santé, les performances de croissance et des indicateurs de qualité de viande chez le porc

Abstract: L’objectif de ce travail exploratoire financé dans le cadre des CI PHASE était de valider l’hypothèse d’un impact du stress thermique anténatal sur le comportement, les performances et les capacités d’adaptation des porcs. Un total de 24 truies (primipares et multipares) réparties en 2 lots de 12 animaux a été utilisé. Du 6ème au 109ème jour de gestation, le lot expérimental a été soumis à un challenge thermique chronique cyclique (variation nycthémérale de 28 à 34°C ; lot GHS) et le lot témoin a été élevé dans des conditions thermoneutres (18 à 24°C ; lot GTN). Le déroulement de la mise bas et la maturité des porcelets nouveau-nés ne sont pas influencés par le traitement thermique des truies en gestation. En lactation, une température rectale plus élevée dans le lot GHS suggère que le stress thermique en gestation perturbe sur le long terme la thermorégulation des truies en particulier chez les primipares. Nos résultats indiquent que la fréquence des problèmes sanitaires immédiatement après le sevrage est plus faible chez les porcelets GTN mais que la réactivité de l’axe corticotrope à 65 j d’âge est plus forte chez les porcelets GHS. A partir de 10 semaines d’âge, 24 femelles du lot GTN et 24 femelles du lot GHS ont été sélectionnées et dans chaque lot, la moitié des animaux a été maintenue dans des conditions thermoneutres (18-24°C ; lots GTN-TN et GHS-TN) et l'autre moitié a été soumise à un stress thermique (28-34°C ; lots GTN-HS et GHS-HS). Indépendamment du traitement pendant la période anténatale, le stress thermique en phase de croissance a des effets négatifs sur la vitesse de croissance. Sur l'ensemble de la croissance, les interactions entre le traitement thermique en gestation et en phase de croissance sur le gain de poids ou l'indice de consommation ne sont pas significatives. Le stress thermique prénatal tend à réduire le poids vif à 20 semaines (-8 kg en moyenne), à augmenter les températures rectale et cutanée et la concentration de cortisol salivaire comparativement aux porcs issus des lots GTN. A l'abattage, les porcs des lots GHS sont plus gras et cet effet est accentué pour le lot GHS-HS. Dans notre étude, le stress thermique en période prénatale a très peu d’effets sur les propriétés musculaires et la qualité de viande. En revanche, les indicateurs de qualité de la viande sont influencés par la température d'élevage en phase de croissance. Le stress thermique post-natal influence les propriétés métaboliques musculaires et les indicateurs de qualité de viande, notamment dans les muscles de la longe et du jambon, à métabolisme majoritairement glycolytique : le pH ultime est augmenté, la perte en eau pendant la maturation réduite et en conséquence l’indice de qualité de viande (jambon) est amélioré, comparé aux porcs élevés à la thermoneutralité. Enfin, nos travaux indiquent que l’environnement thermique prénatal impacte la composition du microbiote mesurée en fin d’engraissement en particulier sur les animaux GHS-HS.

Abstract: International audience; Northwestern Algeria is located where the Saharan Atlas borders the Saharan Platform along the South Atlas Front. The origin of the South Atlas Front is controversial, thus a detailed gravity and magnetic analysis constrained by seismic reflection profiles and well data were conducted to determine the structural configuration of this region. A residual gravity anomaly map created using upward continuation and a reduced to the pole magnetic anomaly map indicated a series of northwest-trending maxima anomalies parallel to the Atlassic orogeny folds and faults and east-trending maxima within the Benoud foreland basin. These maxima mostly coincide with Paleozoic basement uplifts based on seismic reflection profiles. Depth analyses based on upward continuation, and two-dimensional forward modeling of the gravity and magnetic data indicated that the source of the maxima are mainly 5 km in depth with the magnetic sources being approximately 0.5 km deeper than the gravity sources. The gravity and magnetic models indicate that the source bodies are steep-sided and coincide with interpreted faults from the seismic reflection profiles. The maxima anomalies are mainly caused by basement uplifts instead of variations in density and/or magnetic susceptibility in the Paleozoic or older basement lithologies. The South Atlas Front and the Saharan Atlas on the central and eastern portions of Algeria is governed by a thin-skinned tectonics style involving the Mesozoic-Cenozoic sedimentary cover. While the western part was probably controlled by a mixed thin- and thick-skinned tectonic style with the reactivation of deeply rooted Triassic and Jurassic faults during Tertiary compressive events in the interpreted Paleozoic basement. Our gravity and magnetic models illustrate a crustal architecture model of the Paleozoic basement which is consistent with a north-dipping basement normal fault between less deformed Saharan Platform than the more deformed Atlassic domain. Such a crustal model aids in determining the nature of the Atlassic orogeny as geological and geophysical studies have been determining the variations of structural styles along its entire length.

Abstract: OBJECTIVE: Long-term treatment with tyrosine kinase inhibitors (TKI) represents an effective cure for chronic myeloid leukemia (CML) patients and discontinuation of TKI therapy is now proposed to patient with deep molecular responses. However, evidence demonstrating that TKI are unable to fully eradicate dormant leukemic stem cells (LSC) indicate that new therapeutic strategies are needed to control LSC and to prevent relapse. In this study we investigated the metabolic pathways responsible for CML surviving to imatinib exposure and its potential therapeutic utility to improve the efficacy of TKI against stem-like CML cells. METHODS: Using complementary cell-based techniques, metabolism was characterized in a large panel of BCR-ABL+ cell lines as well as primary CD34+ stem-like cells from CML patients exposed to TKI and L-Asparaginases. Colony forming cell (CFC) assay and flow cytometry were used to identify CML progenitor and stem like-cells. Preclinical models of leukemia dormancy were used to test the effect of treatments. RESULTS: Although TKI suppressed glycolysis, compensatory glutamine-dependent mitochondrial oxidation supported ATP synthesis and CML cell survival. Glutamine metabolism was inhibited by L-asparaginases such as Kidrolase or Erwinase without inducing predominant CML cell death. However, clinically relevant concentrations of TKI render CML cells susceptible to Kidrolase. The combination of TKI with Lasparaginase reactivates the intinsic apoptotic pathway leading to efficient CML cell death. CONCLUSION: Targeting glutamine metabolism with the FDA-approved drug, Kidrolase in combination with TKI that suppress glycolysis represents an effective and widely applicable therapeutic strategy for eradicating stem-like CML cells. ispartof: MOLECULAR METABOLISM vol:55 ispartof: location:Germany status: published

Abstract: International audience; Phyllosilicates may trap hydrogen (H2) in the crust, but they may also produce it through various processes, including oxidative dehydrogenation. The dehydrogenation temperature depends on the type and composition of the phyllosilicates considered, but it may be as low as 300 °C. Here, we document the release of H2 and CO during thermal treatment of chloritite (300 °C) and talc (500 °C) from the Trimouns deposits (Eastern Pyrenees, France). Thermal release of gases coupled to stable isotope analysis has been used to recover and characterize H2 and CO, the two detected gases. Hydrogen content may be as high as 7 ppm with δDH2 values ranging from −258‰ to – 224‰ for sub-pure chloritite and − 140‰ for pure talc. CO content ranges between 3 ppm and 35.3 ppm with very homogeneous δ13CCO values between −27.6‰ and − 25.7‰. This study supports the idea that H2 was produced during experiments by dehydrogenation. The origin of CO remains enigmatic, but its carbon isotope composition suggests a link to the few amounts of graphite documented in chloritite and talc from the deposit. This work also reports extensive hydrogen isotope fractionation between H2 produced by dehydrogenation and both talc and chloritite. Dehydrogenation of phyllosilicates is a potential source term of H2 in numerous magmatic-hydrothermal settings and must thus be accounted for in the budget of the H2 geochemical cycle

17. Caractérisation des éléments cellulaires des sécrétions lactées transférables de la mère à la progéniture chez la vache, la chèvre et la truie

Abstract: La construction des phénotypes des animaux de rente pendant les premières phases de la vie post-natale est une étape cruciale pour une expression optimale de leur phénotype au stade adulte (multiperformance agronomique). La récente mise en évidence de cellules souches et progénitrices, et de vésicules extracellulaires (VEs), dans le lait de plusieurs espèces (humaine et murine) pose la question du contenu et du devenir de ces éléments cellulaires chez la progéniture, de leurs rôles dans le développement des tissus (participation directe, régulation) et impacts sur la santé tout au long de la vie (robustesse, bien-être). Ce projet, cofinancé par les départements PHASE et GA, participe à la compréhension des mécanismes d’élaboration des phénotypes des animaux de rente en identifiant les éléments cellulaires (cellules totales, cellules souches et VEs) dans les sécrétions lactées (colostrum et lait) de truies, vaches et chèvres laitières. L'apport de ces éléments à la progéniture pouvant avoir un rôle physiologique au début de la vie post-natale, notre protocole expérimental intègre une analyse cinétique grâce à un échantillonnage de «lait» dans les premières heures (0, 3, 6, jusqu’à 24h) après la mise-bas (colostrum), puis au cours des premiers jours de la lactation (jusqu’à 7 jours pour chèvres et vaches, 18 jours pour les truies), sur 6 femelles multipares (IE production laitière et UE3P). Les cellules des sécrétions lactées ont été analysées en cytométrie en flux en utilisant des marqueurs moléculaires spécifiques de typage (combinaison d’anticorps dirigés contre des protéines de surface) ciblant les cellules souches mammaires, les cellules souches mésenchymateuses et les cellules souches hématopoïétiques. Les premiers résultats montrent que, chez la vache et la chèvre, des cellules viables se retrouvent en nombre dans le colostrum dès la mise-bas. Elles diminuent progressivement pendant les 12 heures qui suivent, puis régulièrement tout au long de la semaine de suivi cinétique. Chez la truie, en revanche, le nombre de cellules viables est maximal un jour après la mise-bas. Pour les trois espèces, l’analyse en cytométrie montre que les cellules souches mammaires (0,5% à 1,5% des cellules viables selon l’espèce) sont retrouvées principalement dans le colostrum des premières heures après la mise-bas, devenant quasi inexistantes les jours d'après (< 0,2%). Fait intéressant, une population cellulaire exprimant CD24, une protéine exprimée par certaines cellules souches hématopoïétiques et les lymphocytes B, est fortement présente chez les trois espèces pendant les six premières heures, puis évolue à la baisse au cours de la lactation. Cette population cellulaire mérite une caractérisation plus poussée. A ce jour, le retraitement a posteriori des données de cytométrie des cellules souches mésenchymateuses et hématopoïétiques se poursuit. Cela nous permettra de compléter notre connaissance des différents types de cellules évoluant dans les sécrétions lactées au cours des premiers jours de lactation. Concernant les VEs, nous avons développé la purification par ultracentrifugation différentielle et par chromatographie à exclusion de taille des petites et grandes VEs ainsi que leur analyse physico-chimique par « nanoparticle tracking analysis » et western blot. Chez le bovin, les VEs sont retrouvées dans le lait par centaines de milliard par millilitre de lait écrémé. Cette abondance sera évaluée dans les différentes espèces au cours des phases de lactation et la proportion des sous-classes de VEs sera déterminée suite à leur purification par immunocapture (criblage d’anticorps en cours). En conclusion, le dépouillement partiel des analyses cytométriques et des VEs met notamment en évidence l'existence de cellules souches mammaires dans les sécrétions lactées et suggère la présence de populations cellulaires et vésiculaires spécifiques, potentiellement avantageuses sur le plan physiologique, au(x) premier(s) jour(s) de lactation. Chez les ruminants, nos résultats pourraient venir appuyer des pratiques d’élevage permettant d’améliorer le bien-être et la santé des petits, et la robustesse des animaux adultes. Pour l’espèce porcine, nos résultats pourraient confirmer la nécessité pour les porcelets de consommer le colostrum de leur mère biologique. Enfin, la poursuite de la comparaison des 3 espèces sera déterminante pour établir si ce phénomène de transfert cellulaire et vésiculaire est générique ou spécifique à l’espèce.

Abstract: International audience; In order to improve the outcome observed with azacitidine (AZA) in higher-risk Myelodysplastic syndrome (MDS), its combination with other drugs in MDS must be evaluated. So far, no combination has not been shown to be more effective than AZA alone. AZA-PLUS was a phase II trial that, in a "pick a winner" approach, randomly assigned patients with higher-risk MDS, CMML and low blast count AML to: AZA; AZA plus lenalidomide; AZA plus Valproic Acid or AZA plus Idarubicin. 322 patients were included. After six\,cycles, 69 (21.4%) CR\,+\,PR were observed with no benefit from any combination. Median EFS and OS were 17.2 and 19.7\,months in the whole cohort, respectively, with no difference across randomised arms. Infection and rates of hospitalisation during the first six\,cycles were higher in the AZA-LEN And AZA-IDA arm, related to increased myelosuppression. Factors associated with better response were IPSS, favourable or intermediate karyotype, haemoglobin, lower circulating blast count, fibrinogen level and lower LDH, while poorer survival was seen in therapy-related MDS and, in the case of TP53, PTPN11 or CSF3R mutation. The combinations used did not improve the outcome obtained with AZA alone. However, our "pick a winner" randomised strategy may remain useful with potentially more active drugs to be tested in combination with AZA.

Abstract: International audience; Three key factors appear relevant in view of annual optimizations of the Public Hospital management: territorial reorganization, quick change in medical technologies and maintaining balanced budgets. Finding the right tools for territorial analysis, making it possible to achieve the efficiency of the care sectors of "territorial hospital groupings" is the priority of the Health Care Directorate, and this is the context in which PoleSat has been evaluated.Method: three scenarios have been considered for ‘vascular catheterizations’ among seven hospital hubs of Nouvelle Aquitaine: the existing situation, the closure of the Rochefort hub and the transfer of activity from Rochefort to the Saintes hub. Different analyzes of "modeled catchment areas and allocations of populations and activities" have been compared. Results: After comparisons of the pre-post scenarios of the covered areas and the concerned statistic of population, consequences and impacts for the hubs are expressed in ‘risks of loss’ and ‘chances of gain’ of activity and patient numbers. Conclusion: Taking into account its first tests and good repeatability, PoleSat is proving to be robust and reliable. It will be enhanced and reused for other care services, geographic spaces and territories.; Trois facteurs s'imposent aux objectifs d'optimisation annuelle de l'Hôpital Public : la réorganisation territoriale ; l'évolution rapide des technologies médicales et l'équilibre budgétaire. Trouver les bons outils d'analyse territoriale, permettant d'atteindre l'efficience des filières de soins des groupements hospitaliers de territoires est la priorité de la direction générale de l’offre de soins, qui dans ce cadre a testé PoleSat. Méthode : trois scénarios de planification des "cathétérismes vasculaires" pour sept pôles hospitaliers en Nouvelle Aquitaine ont été élaborés ; l'état de l'offre initiale, la suppression du pôle de Rochefort et le transfert d'activité de Rochefort vers le pôle de Saintes. Les analyses des zones d'influence et des répartitions modélisées des "populations et activités" ont été comparées. Résultats : Les conséquences et impacts pour les pôles, à l'issue des comparaisons pré-post scénarios des aires couvertes et des statistiques, s'expriment en "risques de pertes" et en "chances de gains" d'activité et de patientèle. Conclusion : PoleSat est robuste au vu de ses premiers tests et de la répétabilité des résultats. Il sera enrichi et réutilisé pour d'autres filières de soins, espaces géographiques et territoires.

Abstract: International audience; Improving the housing of pregnant sows by giving them more space and access to deep straw had positive effects on their welfare, influenced their maternal behavior and improved the survival of their offspring. The present study aimed at determining whether these effects were actually due to environmental enrichment and whether the provision of straw pellets and wood can partly mimic the effects of straw bedding during gestation. Three graded levels of enrichment were used, that were, collective conventional pens on slatted floor (C, n = 26), the same pens with manipulable wood materials and distribution of straw pellets after the meals (CE, n = 30), and larger pens on deep straw litter (E, n = 27). Sows were then housed in identical farrowing crates from 105 days of gestation until weaning. Decreased stereotypies, blood neutrophils, and salivary cortisol, and increased behavioral investigation indicated that health and welfare of sows during gestation were improved in the E environment compared with the C environment. The CE sows responded as C or E sows depending on the trait. Piglet mortality rate in the first 12 h after birth was lower in E and CE litters than in C litters, but enrichment level during gestation had only small effects on lactating sow behavior and milk composition postpartum. On days 2 and 3 of lactation, E sows interrupted less often their nursing sequences than C and CE sows. On day 2, milk from both E and CE sows contained more minerals than that from C sows. In one-day-old piglets, the expression levels of genes encoding toll-like receptors (TLR2, TLR4) and cytokines (interleukin-1,-6 and-10) in whole blood after 20-h culture, were greater in E piglets than in CE or C piglets. In conclusion, housing sows in an enriched environment during gestation improved early neonatal survival, probably via moderate and cumulative positive effects on sow behavior, milk composition, and offspring innate immune response. The gradation in the effects observed in C, CE and E housing environment reinforced the hypothesis of a causal relationship between maternal environmental enrichment, sow welfare and postnatal piglet traits.

24. Effet de la supplémentation en produits à base de plantes sur la physiologie des truies et des porcelets pendant la lactation et au sevrage

Abstract: Some plant-derived products are known for their antioxidant, anti-inflammatory or anti-bacterial properties. When provided to piglets or their dam, they may improve piglets’ ability to adapt to weaning stress. The effects of a supplementation with plant-derived products on sow and piglet physiology were investigated. Sixty-four sows were assigned to a Control or Extract group. Extract sows received 25 g/d of plant extract (Eleutherococcus senticocus, Uncaria tomentosa and Rosmarinus officinalis) in feed from day of gestation (DG) 106 to day of lactation (DL) 28 and 20 mL of essential oils (from Origanum vulgare and Eucalyptus globulus) on DG109. Within each sow group, 2 mL of essential oils (EO, from Origanum vulgare and Eucalyptus globulus) were administered to all piglets of 1 litter out of 2 at DL3. Piglets were weaned on DL28. Blood was collected from sows on DG94, DG112 and DL26 and from 2piglets/litter on DL25 and 5 d post-weaning to measure indicators of the metabolic, immune, inflammatory and oxidative status. Colostrum and milk were collected at farrowing and on DL6 and DL26. Extract supplementation had no effect on sow physiology except for fewer lymphocytes on DG112 and a lower free-fatty-acid concentration on DL26 (P < 0.05). Extract supplementation reduced fat and haptoglobin concentration in milk on DL26 (P < 0.05). On DL25, EO piglets had a lower granulocyte proportion (P < 0.05), a greater lymphocyte proportion and a lower oxidative stress index (OSI, P < 0.10), regardless of sow treatment. Piglet OSI, haptoglobin concentration and leukocytes number increased after weaning but were not influenced by sow or piglet supplementation. The present combination of plant-derived products could thus modify the composition of sow milk and the immune and inflammatory status of piglets before weaning.

Abstract: International audience; Depuis 2000, nous compilons les données sur les terrains géologiques les plus superficiels, qui résultent de processus supergènes, i.e. d’interactions chimiques et/ou physiques de la lithosphère en surface avec l'hydrosphère, l'atmosphère et la biosphère. Ce sont les régolithes autochtone (altérites sensu lato) et allochtone (dépôts sédimentaires continentaux à côtiers). Nous synthétisons et restituons ces informations sous la forme de deux cartes géologiques numériques de la France métropolitaine à l’échelle du 1/1 000 000, consultables sur le site infoterre.brgm.fr dans la couche "Géologie de la surface".Ces cartes sont utiles en amont des études dans les domaines des géosciences appliquées tels qu’aménagement des territoires (ruraux et urbains), environnement, gestion des risques et ressources de proche surface (0 à 100 m). Elles intègrent les données issues de deux sources principales : les cartes géologiques à 1/50 000 et les études publiées. Elles évoluent au fil du temps, au gré des parutions et de la collecte de nouvelles données. Elles intègrent progressivement les formats de production du Référentiel Géologique de la France.La version d’origine (Lacquement et al, 2010) a été mise à jour en 2013 (Tissoux et al, inédit). La version 2021 inclut de nouvelles informations qui résultent des travaux de modélisation de la géométrie des formations meubles de surface sur les roches indurées (sédimentaires ou issues de l’altération du socle anté-triasique). Les travaux ont été réalisés à partir de l’interpolation des données de forages en Bretagne et pro parte dans les Pays-de-la-Loire (à la maille de 250 m).La compilation des informations cartographiques disponibles permet d’aboutir à la fabrication des deux cartes géologiques identifiant la source des données (données de terrain ou modélisation géométrique). Abstract: International audience; Les cartes géologiques représentent la base de toute étude en lien avec les sciences de la Terre. Rien de mieux qu’une représentation spatiale du terrain pour exprimer la diversité des roches et leur arrangement en trois dimensions. Cependant, la bonne compréhension de ces documents demande un long apprentissage. Il fallait donc les faire évoluer de manière pédagogique pour les rendre accessibles à un large public en demande d’informations sur le sous-sol afin qu’il puisse bien comprendre l’organisation des roches et les différentes étapes de leur mise en place, avec selon les cas, leur déformation, leur exhumation et leur altération qui ont jalonné la longue histoire géologique. De nombreux utilisateurs de cartes géologiques ne sont pas des experts sur le sujet (gestionnaires de territoires, bureaux d’études, vignerons, enseignants, naturalistes amateurs, etc.). Ils recherchent avant tout (1) la nature des roches présentes, (2) la présence de nappe d’eau souterraine et (3) les secteurs pouvant présenter des risques naturels (cavités, glissements, etc.). Avec les cartes traditionnelles détaillées, ces utilisateurs ne trouvent pas facilement les réponses à leurs recherches. Il nous a donc paru important de nous adapter pour répondre aux demandes d’une partie de nos utilisateurs/clients, et notamment du monde de l’enseignement où les cartes géologiques ont récemment réintégré les programmes scolaires. Par rapport aux cartes classiques à l’échelle 1/50 000, ces nouvelles représentations du soussol rendent le modelé du relief plus lisible, synthétisent légèrement les unités cartographiées et proposent de percevoir directement leurs durées de formation, les hiatus et épisodes d’érosion, de déformation tectonique, les milieux de dépôt pour les roches sédimentaires, etc. Il est également possible de visualiser directement pour les mêmes niveaux les ressources minérales et leurs usages, les aquifères et aquitards, ainsi que la nature des risques naturels potentiels. Le BRGM a initié cette nouvelle collection de cartes en proposant trois titres à la vente: Paris, Lyon et Sancerre-Pouilly. Les deux premières montrent l’évidence que l’emplacement des deux métropoles n’est pas fortuit. Elles sont situées au milieu de carrefours géologiques bien particuliers. Pour Paris, cela permet également de se rendre compte des aménagements qu’il a fallu réaliser pour apporter de l’eau potable aux habitants. Parfois, la lecture peut prendre l’aspect d’une anecdote amusante avec par exemple Pierre-Bénite près de Lyon qui correspond à un rare affleurement de granite qui représentait une “bénédiction” aux mariniers pour amarrer leurs bateaux. Pour la carte Sancerre-Pouilly, cela permet de faire immédiatement le lien entre les terroirs et les vignobles. Les spécificités des terrains les plus propices aux meilleurs crus apparaissent clairement.

Abstract: In France and elsewhere, decision-makers, healthcare professionals and health planners need to better understand and specify the provision of medical care. To this end, a hospital-based research project on a gravitational health planning modelling process was initiated in 2002. Since then, geomatics has emerged as a major scientific field for facing new challenges in medical informatics and health planning, thanks to the use of attractive interfaces, new methods and user-friendly IT technologies. Our initial 2002 model has recently been enhanced, optimized and automated as part of a spatial decision support system (PoleSat_2018). These decisive improvements and optimizations were mainly based on Delaunay triangulation, the replacement of human expertise with a heuristic dominance rule that provides a complete automated algorithm, and an online graphical user interface. Rapid, easy planning scenarios (by grouping and/or closing hospitals) give a quasi-instantaneous, strategic visualization of hospital catchment areas for decision-makers who are not experts in geomatics via ready-to-use maps and spreadsheets. This new implementation achieves our main objective, since the proposed deterministic method provides a completely automated, stable algorithm. A custom version of this tool is now being used by the French Ministry of Health for real planning issues. Consequently, PoleSat could be easily generalized as a prospective, strategic decision support tool for various health planning issues.

Abstract: International audience; Dating of Quaternary sediments is now well constrained by numerical methods such as electron spin resonance (ESR) among others, which can cover the last 2.6 Ma. Nevertheless, beyond the Quaternary timescale, the application of ESR becomes extremely challenging. At earth surface, some deposits exist, such as alluvial outwash, fluvio-lacustrine fillings or marine beaches whose age could be earlier than the Quaternary and for which dating is currently very complicated in the absence of faunal remains, plants or volcanic effluents. In ESR dating of optically-bleached quartz, the two most commonly used paramagnetic centres are [AlO4] and [TiO4/Li+]0, usually named Al and Ti-Li centres, respectively. Whether these centres can provide accurate dating results is mostly conditioned by their thermal stability and saturation to the received radiation dose. The thermal stabilities of the Al and Ti-Li centers were studied very early in the development of the method and were back then estimated to be of ca. 7.4x109 and 8x106 years respectively [1]. Similarly, the apparent saturation of the signal with the radiation dose may be reached > 10,000 Gy for both Al and Ti-Li centres [2, 3]. Consequently, this combined evidence suggests that there is no theoretical obstacle to date pre-Quaternary silico-clastic sediments by ESR. We report here a series of dating attempts performed on quartz samples whose expected age ranges from about 50 Ma to the earliest part of the Pleistocene. These samples come from various types of depositional environments (e.g. marine, fluvio-deltaic, fluvial), and independent, numerical or (bio)- stratigraphic age control is sometimes available. In all the dated samples, the multiple aliquot additive dose method was applied, while the regeneration approach was also tested for a few of them. Our preliminary result show that that these quartz samples overall display a very good dose response curve to irradiation, and do not reach apparent saturation. However, if a good adequacy is obtained between calculated ages and the preliminary estimate given by the geology for marine sediments aged between 5 and 2 Ma, a great majority of the ESR ages are younger than expected. Morevover, dating tests performed on sediment from the Miocene and Paleocene-Eocene boundaries show significant age underestimations. These dating results will be discussed in relation to the expected ages, depositional environments or the composition and geochemical history of the sediment from which the dated quartz was extracted

Abstract: International audience; The 205 Ma Rochechouart impact structure (France) is characterized by various impactite formations overlying the Hercynian crystalline basement. New constraints from downhole logging and surface electrical resistivity measurements along >100 m long profiles reveal that the top melt-bearing breccia layer is more conductive and porous than the underlying melt-poor breccia layer. The stratigraphy within the impactite and the transition with the basement are irregular at small (∼1–10 m) and medium (>100 m) scales, with vertical amplitude up to 40–50 m. At larger scale (>1 km), audio-magnetotelluric observations are able to map the lateral and vertical extent of fracturing/brecciation in the basement, reaching 200 m below the surface nearby Chassenon, in the northern part of the structure. Our results also unveil that the impactite deposits and the brecciated basement of the Rochechouart impact structure may have been shifted laterally and vertically during the modification stage of the impact event through displacements of megablocks, which may be associated with the collapse of a central uplift.

Abstract: International audience; In the field of augmented reality, there is a need for very bright color microdisplays to meet the user specifications. Today, one of the most promising technology to manufacture such displays involves a blue micro-LED technology and quantum dots-based color conversion layers. Despite recent progress, the external power conversion efficiencies (EPCE) of these layers remain under ∼25%, below the needs (>40%) to reach a white luminance of 100,000 cd/m$^2$. In this work, we have synthesized CdSe$_x$S$_{1-x}$ nanoplatelet-based conversion layers for red and green conversion, and measured their absorption properties and EPCE performances with respect to layer thickness. On this basis, a model was developed that reliably predicts the layer EPCE while using only few input data, namely the layer absorption coefficients and the photoluminescence quantum yield (PLQY) of color photoresist. It brings a new insight into the conversion process at play at a micro-LED level and provides a simple method for extensive optimization of conversion materials. Finally, this study highlights the outstanding red conversion efficiency of photoresist layers made of core-double shell CdSe$_x$S$_{1-x}$ nanoplatelets with 31% EPCE (45% external PLQY) for 8 µm-thick conversion layer.

Abstract: Heuristics are important techniques designed to find quickly good feasible solutions for hard integer programs. Most heuristics depend on a solution of the relaxed linear program. Another approach based on Lagrangian relaxation offers a number of advantages over linear programming, namely it is extremely fast for solving large problems. Wedelin heuristic is such a Lagrangian based heuristic, initially developed to solve airline crew scheduling problems. The performance of this method depends crucially on the choice of its numerous parameters. To adjust them and learn which ones have important influence on whether a solution is found and its quality, we propose to conduct a sensitivity analysis followed by an automatic tuning of the most influential parameters. We have implemented a C++ open-source solver called baryonyx which is a parallel version of a (generalized) Wedelin heuristic. We used the Morris method to find useful continuous parameters. Once found, we fixed other parameters and let a genetic optimization algorithm using derivatives adjust the useful ones in order to get the best solutions for a given time limit and training instance set. Our experimental results done mostly on crew and bus driver scheduling benchmarks tackled as set partitioning problems show the significant improvements obtained by tuning the parameters and the good performances of our approach compared to state-of-the-art exact and inexact integer programming solvers.

Abstract: Although pulmonary events are considered to be frequently associated with malignant haemopathies, they have been sparsely studied in the specific context of myelodysplastic syndromes (MDS). We aimed to describe their different types, their relative proportions and their relative effects on overall survival (OS). We conducted a multicentre retrospective cohort study. Patients with MDS (diagnosed according to the 2016 WHO classification) and pulmonary events were included. The inclusion period was 1 January 2007 to 31 December 2017 and patients were monitored until August 2019. Fifty-five hospitalized patients were included in the analysis. They had 113 separate pulmonary events. Thirteen patients (23.6%) had a systemic autoimmune disease associated with MDS. Median age at diagnosis of MDS was 77 years. Median time to onset of pulmonary events was 13 months. Pulmonary events comprised: 70 infectious diseases (62%), 27 interstitial lung diseases (23.9%), including 13 non-specific interstitial pneumonias and seven secondary organizing pneumonias or respiratory bronchiolitis–interstitial lung diseases, 10 pleural effusions (8.8%), including four cases of chronic organizing pleuritis with exudative effusion, and six pulmonary hypertensions (5.3%). The median OS of the cohort was 29 months after MDS diagnosis but OS was only 10 months after a pulmonary event. The OS was similar to that of the general myelodysplastic population. However, the occurrence of a pulmonary event appeared to be either an accelerating factor of death or an indicator for the worsening of the underlying MDS in our study. More than a third of pulmonary events were non-infectious and could be systemic manifestations of MDS.

Abstract: International audience; The North Pyrenean Zone corresponds to the palaeopassive margin of the North Iberia plate, at the foot of which subcontinental mantle was exhumed during Albian times. Rare bodies of exhumed mantle rocks associated with strongly sheared lenses of continental crust are scattered among the North Pyrenean Zone metasediments. Significant fluid flow occurred along a major décollement at the basement–Trias interface in the Urdach massif (Chaînons Béarnais). Fluids with a broad range of salinity (10–38 wt.% NaCl equiv.), indicative of mixing between brines and more dilute waters, produced strong silicification of breccias. The brines circulated at c. 240–280°C under lithostatic pressures at c. 6 ± 1 km depth. The fluids became increasingly saline towards the final stages. The syndeposition of Cenomano-Turonian flysch layers then progressively isolated the lower aquifers close to the décollement where Triassic brines were predominant. The release and migration of significant volumes of brines during stretching and squeezing of the Triassic evaporites played a crucial part in the mineralogical and rheological transformations that occurred during the Pyrenean Cretaceous rifting event.

Abstract: International audience; Objective : To establish recommendations for optimisation of the management of patients undergoing pulmonary lobectomy, particularly Enhanced Recovery After Surgery (ERAS).Design : A consensus committee of 13 experts from the French Society of Anaesthesia and Intensive Care Medicine (Soci,t, franOaise d'anesth,sie et de r,animation, SFAR) and the French Society of Thoracic and Cardiovascular Surgery (Soci,t, franOaise de chirurgie thoracique et cardiovasculaire, SFCTCV) was convened. A formal conflict-of-interest policy was developed at the onset of the process and enforced throughout. The entire guidelines process was conducted independently of any industry funding. The authors were advised to follow the principles of the Grading of Recommendations Assessment, Development and Evaluation (GRADE) system to guide assessment of quality of evidence.Methods : Five domains were defined: 1) patient pathway and patient information; 2) preoperative management and rehabilitation; 3) anaesthesia and analgesia for lobectomy; 4) surgical strategy for lobectomy; and 5) enhanced recovery after surgery. For each domain, the objective of the recommendations was to address a number of questions formulated according to the PICO model (Population, Intervention, Comparison, Outcome). An extensive literature search on these questions was carried out and analysed using the GRADE® methodology. Recommendations were formulated according to the GRADE® methodology, and were then voted by all experts according to the GRADE grid method.Results : The SFAR/SFCTCV guideline panel provided 32 recommendations on the management of patients undergoing pulmonary lobectomy. After two voting rounds and several amendments, a strong consensus was reached for 31 of the 32 recommendations and a moderate consensus was reached for the last recommendation. Seven of these recommendations present a high level of evidence (GRADE 1+), 23 have a moderate level of evidence (18 GRADE 2+ and 5 GRADE 2-), and 2 correspond to expert opinions. Finally, no recommendation was provided for 2 of the questions.Conclusions : A strong consensus was expressed by the experts to provide recommendations to optimise the whole perioperative management of patients undergoing pulmonary lobectomy.

Abstract: Simple Summary The interaction of programmed cell death ligand-1 (PDL1) with its receptor PD1 inhibits T-cell responses. Blockade of this interaction with monoclonal antibodies leads to major antitumor effects. However, not all cancer patients respond well to anti-(PD-1/PD-L1) immunotherapy. The PD-L1 protein is expressed at the cell plasma membrane (mPD-L1), at the surface of exosomes (exoPD-L1), in cell nuclei (nPD-L1) and as a soluble circulating protein (sPD-L1). The aim of our analysis was to highlight the multiple variants of sPD-L1 generated either by the proteolytic cleavage of m/exoPD-L1 or by the alternative splicing of PD-L1 pre-mRNA. The objective was also to underline the presence and role of circulating sPD-L1 isoforms in multiple cancer indications and many other diseases (including chronic inflammatory and viral diseases), and under non-pathological conditions (pregnancy). sPD-L1 often represents a general marker of an inflammatory status. The pool of sPD-L1 proteins is an integral part of the highly dynamic PD-1/PD-L1 signaling pathway. Abstract Upon T-cell receptor stimulation, the Programmed cell Death-1 receptor (PD-1) expressed on T-cells can interact with its ligand PD-L1 expressed at the surface of cancer cells or antigen-presenting cells. Monoclonal antibodies targeting PD-1 or PD-L1 are routinely used for the treatment of cancers, but their clinical efficacy varies largely across the variety of tumor types. A part of the variability is linked to the existence of several forms of PD-L1, either expressed on the plasma membrane (mPD-L1), at the surface of secreted cellular exosomes (exoPD-L1), in cell nuclei (nPD-L1), or as a circulating, soluble protein (sPD-L1). Here, we have reviewed the different origins and roles of sPD-L1 in humans to highlight the biochemical and functional heterogeneity of the soluble protein. sPD-L1 isoforms can be generated essentially by two non-exclusive processes: (i) proteolysis of m/exoPD-L1 by metalloproteases, such as metalloproteinases (MMP) and A disintegrin and metalloproteases (ADAM), which are capable of shedding membrane PD-L1 to release an active soluble form, and (ii) the alternative splicing of PD-L1 pre-mRNA, leading in some cases to the release of sPD-L1 protein isoforms lacking the transmembrane domain. The expression and secretion of sPD-L1 have been observed in a large variety of pathologies, well beyond cancer, notably in different pulmonary diseases, chronic inflammatory and autoimmune disorders, and viral diseases. The expression and role of sPD-L1 during pregnancy are also evoked. The structural heterogeneity of sPD-L1 proteins, and associated functional/cellular plurality, should be kept in mind when considering sPD-L1 as a biomarker or as a drug target. The membrane, exosomal and soluble forms of PD-L1 are all integral parts of the highly dynamic PD-1/PD-L1 signaling pathway, essential for immune-tolerance or immune-escape.

Abstract: International audience; The European PROHEALTH project focused on production diseases in pigs and poultry. For the pig species, INRAE has brought new knowledge to validate the role of husbandry practices in production diseases, to propose how farming conditions could be improved to preserve the health of pigs and to understand the biological mechanisms influencing the expression of these diseases. Enrichment of the housing environment of sows during gestation improved their welfare, and reduced neonatal mortality of piglets. The response over time to the stress of weaning and the prevalence of digestive problems were modulated by the feed sequence. In growing pigs, increasing the living space could be beneficial for the prevention of locomotion disorders according to the pig genetic predisposition to increase their physical activity. Also, in growing pigs, ensuring of good hygiene of housing conditions improved health and growth performance by preventing systemic inflammation and respiratory disorders. We used a model of lines divergently selected on Residual Feed Intake (RFI), a measure of feed efficiency, to show that the genetic background affected the susceptibility of pigs to diseases such as post-weaning digestive disorders, osteochondrosis and the inflammation caused by the poor hygiene of housing conditions.; Le projet européen PROHEALTH portait sur les maladies de production des porcs et des volailles. Pour l’espèce porcine, INRAE a apporté des connaissances nouvelles permettant de comprendre les mécanismes biologiques qui conduisent ou non à l’expression de ces maladies et de proposer des voies d’amélioration des conditions d’élevage pour préserver la santé des porcs. Ainsi, enrichir l’environnement des truies pendant la gestation a permis de réduire leur stress et donc d’améliorer leur bien-être et de réduire la mortalité néonatale des porcelets. Chez les porcelets, la dynamique de réponse aux stress du sevrage et la prévalence des problèmes digestifs ont été modulées par l’alimentation. Chez les porcs en croissance, augmenter la surface disponible en élevage contribuerait à prévenir l’apparition de troubles locomoteurs selon la prédisposition génétique des porcs à augmenter leur activité physique. Chez les porcs en croissance également, le respect de bonnes conditions d’hygiène du logement permet de préserver la santé et les performances de croissance en limitant l’inflammation systémique et les troubles respiratoires. Abstract: International audience; Intensive chayote cultivation in Réunion almost disappeared in the 2000s due to significant yield losses from fruit fly attacks on this historically important crop (Dacus ciliatus, Zeugodacus cuurbitae and Dacus demmerezi). Since the late 2000s, the adoption of agroecological crop protection practices have led to the effective management of fruit fly populations, a significant reduction in pesticide use, an increase in chayote production and plantations, and the development of organic production. To assist in fruit fly management, a qualitative model which simulates fruit fly damage to chayote crops, known as IPSIM-chayote, was developed, providing satisfactory prediction results. It has a user-friendly interface and is now available free of charge online, in three languages (French, English and Spanish): https://pvbmt-apps.cirad.fr/apps/ipsim-chayote/?lang=en. The IPSIM-chayote modeling platform can be used by farmers as a diagnosis to simulate fruit fly damage to their crops and as a decision-making tool for their agricultural practices. The model can be used as a training resource in agroecological crop protection. Public authorities and local government can use it as a tool in planning and forecasting agricultural development. Finally, researchers can use it as a prediction tool and a resource for the exchange of information, allowing them to review scientific knowledge or identify new, relevant research areas suited to the context and challenges. IPSIM-chayote can be considered as a forum for exchange and can stimulate collaborative work between individuals. It is a flexible model, as it allows variables to be added. IPSIM-chayote is the first qualitative model developed for crop pests in a tropical environment. It could serve as a basis for the development of other similar models simulating crop pest incidence, thus contributing significantly to the development of agroecological crop protection.

Abstract: The physiology of the sow mammary gland is qualitatively well described and understood. However, the quantitative effect of various biological mechanisms contributing to the synthesis of colostrum and milk is lacking and more complicated to obtain. The objective of this study was to integrate physiological and empirical knowledge of the production of colostrum and milk in a dynamic model of a single sow mammary gland to understand and quantify parameters controlling mammary gland output. In 1983, Heather Neal and John Thornley published a model of the mammary gland in cattle, which was used as a starting point for the development of this model. The original cattle model was reparameterized, modified, and extended to describe the production of milk by the sow mammary gland during lactation and the prepartum production of colostrum as the combined output of immunoglobulins (Ig) and milk. Initially, the model was reparameterized to simulate milk synthesis potential of a single gland by considering biological characteristics and empirical estimations of sows and piglets. Secondly, the model was modified to simulate more accurately the responses to changes in milk removal rates. This was done by linking the ejectable milk storage capacity to the number of secretory cells rather than being constant throughout lactation. Finally, the model was extended to include the prepartum synthesis of milk and the kinetics of Ig into and out of the mammary gland. A progressive capacity of secretory cells to synthesize milk was used to differentiate the time between the onset of milk synthesis and Ig transfer. Changes in maximum milk removal rate, duration of milk ejection, and nursing interval exerted a great impact on the modeled milk output. Changes by ±60% in one of these parameters were capable of increasing milk output by 28% to 39% during the first 4 wk in lactation compared with the reference parameterization. This suggests that the ability of the piglet to remove milk from the gland exerts a key control on milk synthesis during lactation. Modeling colostrum as the combined output of Ig and milk allowed to represent the rapid decline in Ig concentration observed during the first hours after farrowing. In conclusion, biological and empirical knowledge was integrated into a model of the sow mammary gland and constitutes a simple approach to explore in which conditions and to what extent individual parameters influence Ig kinetics and milk production.

Abstract: International audience; Quantitative estimates of mammary nutrient inputs, outputs and metabolism in sows are scarce, despite being critical elements to identify parameters controlling milk synthesis central for the feeding of lactating sows. The objective of this study was to quantify the mammary gland input and output of nutrients as well as the intramammary partitioning of carbon and nitrogen with the purpose to identify mechanisms controlling mammary nutrient inputs, metabolism and milk production in lactating sows. A data set was assembled by integration of results from four studies. The data set included data on litter performance, mammary arterial-venous concentration differences (AV-difference) of energy metabolites and amino acids, and the contents of lactose, fat and amino acids in milk. Milk yield was estimated based on average litter size and litter gain, and mammary plasma flow (MPF) was estimated using the sum of phenylalanine and tyrosine as internal flow markers. The yield and composition of milk were used to estimate mammary nutrient output in milk, and MPF and AV-difference were used to estimate net mammary input of carbon and nitrogen and output of CO 2. Carbon and nitrogen used for the synthesis of lactose, fat and protein in milk and CO 2-yielding processes were represented in a static nutrient partitioning model. The origin of mammary CO 2 output was calculated using theoretical estimates of carbon released in processes supporting mammary synthesis of de novo fat, protein and lactose in milk, mammary tissue protein turnover and transport of glucose and amino acids. Results indicated that total input of carbon from glucose and lactate was partitioned into lactose (36%), fat (31%) and CO 2yielding processes (34%). Theoretical CO 2 estimates indicated that de novo fat synthesis, milk protein synthesis and mammary tissue protein turnover were the main processes related to mammary CO 2 production. More than 90% of mammary gland amino acid input was used for milk protein. The quadratic relationship between AV-difference and mammary input of essential amino acids indicated that both changes in AV-difference and MPF contributed to the regulation of mammary input of essential amino acids. The impact of the arterial supply of amino acids on mammary input may be greater for the branched-chain amino acids, arginine and phenylalanine than for other essential amino acids. In conclusion, relationships between input and output parameters indicate that AV-difference and MPF regulate mammary nutrient input to match the supply and demand of nutrients for the mammary gland.
