Abstract

Ultrahigh intensity lasers face thermal management issues that limit their repetition rates. The key challenge is to efficiently evacuate the heat deposited in the amplifier by the optical pumping without impacting the output laser beam quality. The amplifier can have a multislab geometry where the laser beam crosses successive amplifying slabs and the cooling channels that separate them. This work investigates numerically how a cryogenic cooling of the amplifier by turbulent channel flows may affect the wavefront of the laser beam. To this end, large eddy simulations (LESs) representative of the amplifier cooling are performed using TrioCFD, a code developed by the CEA. First, validation simulations are carried out for heated channel flows, allowing comparisons to direct numerical simulation (DNS) results from the literature. Then, LESs of an open turbulent channel flow cooling two slabs are conducted using conjugated heat transfer between the solid and the fluid. The phase distortions, mean and fluctuations, induced by the inhomogeneous and turbulent temperature field are computed directly from the LES. A moderate although non-negligible effect of the turbulence on the laser wavefront was found. This optical effect increases when the slab heating increases. A comparison to the Sutton model, widely used in aero-optic studies, was performed, and its applicability was found limited for this problem. For the first time, TrioCFD is used to address the question of the beam impact of the cooling of laser amplifiers, and it has proven to be a valuable tool for such application.

Abstract

Alors que nous avons la certitude désormais que continuer le programme civilisationnel de la croissance n’est plus possible, pourquoi nous est-il en même temps impossible de ne pas le poursuivre ? La théorie de l’accélération nous aide à le comprendre. Si seule une catastrophe peut conduire à l’arrêt du « camion fou » de la croissance illimitée, alors la catastrophe de la COVID-19 pourrait être le « miracle » qui, en générant la possibilité de ce qui était impossible, ouvrirait une voie vers une « réinvention de la société » et un « changement de paradigme sociétal ». En prenant la mesure de tout ce qui, dans cet épisode de la COVID-19, pourrait, au contraire, renforcer le programme visant à nous rendre le monde entièrement « disponible », nous examinons en quoi ce moment particulier peut prendre valeur d’avertissement par rapport à une catastrophe écologique prévisible, appliquant ensuite la réflexion au domaine du tourisme. Enfin, il s’agit d’esquisser les changements institutionnels, les « gestes-barrières », mais aussi les « gestes-passerelles » qui permettraient une transformation du tourisme afin qu’il devienne vraiment compatible avec la durabilité., Whereas we do know with certainty that continuing the civilizational program of growth is no longer possible, why is it impossible at the same time not to keep on with it? The acceleration theory helps us understand this gridlock. If only a catastrophe can manage to stop the “careering juggernaut” of unlimited growth, then the COVID-19 disaster could be the “miracle” which, by rendering possible what was unthinkable, would open a way toward a reinvention of society, toward a societal paradigm shift. By taking the measure of everything in this episode of COVID-19 that, on the contrary, would help to strengthen the program of making the world fully “available,” we examine how this particular time can serve as a warning to a foreseeable ecological disaster, and then apply it to the tourism sector. Finally, we outline the institutional changes, the “barrier gestures,” but also the “bridge gestures” which would allow a transformation of tourism in order to make it truly compatible with sustainability.

Abstract

The French collaborative Trio4CLF project aims to understand and control the cryogenic cooling of amplifiers for high power (~1 PetaWatt) and high repetition rate (1-10 Hertz) lasers. In such amplifiers, the fluid evacuates the thermal power absorbed by the solid amplifying plates. A precise knowledge of the heat exchange and thus of the turbulent fluid flow in the amplifier is requested to evaluate its impact on the laser beam quality. As a first step, a Large Eddy Simulation is carried out in air without heating to study the development of the turbulent flow. The CFD code used is TrioCFD, a code developed by the CEA. For validation purpose, the simulation is carried out in the experimental setup configuration: a closed-loop wind tunnel called TRANSAT. Two horizontal plates, separated by 0.05 m, are put in the airflow to represent the amplifier plates. Turbulent boundary layers develop from the plates edges. Numerically, the entrance flow is a homogeneous planar flow with a constant velocity at 10 m/s. The results of this Large Eddy Simulation are presented in this paper as a study of the development of the turbulent boundary layers created by the plates., Le projet collaboratif français Trio4CLF a pour objectif de comprendre et de maitriser le refroidissement cryogénique d'amplificateurs de lasers à haute puissance (~1 PétaWatt) et à haut taux de répétition (1-10 Hertz). Dans de tels amplificateurs, le fluide évacue la puissance thermique déposée dans les plaques amplificatrices solides. Une connaissance fine de l'échange de chaleur et donc de l'écoulement turbulent est nécessaire pour évaluer l'impact du refroidissement sur la qualité du faisceau laser. Dans un premier temps, une Simulation des Grandes Échelles est conduite en air et sans chauffage afin d'étudier le développement de l'écoulement turbulent. Le code de CFD utilisé est TrioCFD, un code développé par le CEA. À des fins de validation, la simulation est menée dans la configuration de l'installation expérimentale : une soufflerie à boucle fermée appelée TRANSAT. Deux plaques séparées de 0,05 m sont placées horizontalement dans l'écoulement d'air de façon à représenter les plaques amplificatrices. Des couches limites turbulentes se développent à partir du bord d'attaque des plaques. Numériquement, l'écoulement d'entrée est un écoulement homogène plan de vitesse constante 10 m/s. Les résultats de cette Simulation des Grandes Échelles sont présentés ici sous la forme de l'étude du développement des couches limites turbulentes créées par les plaques.

A precise knowledge of the heat exchange and of the turbulent fluid flow in the amplifier is requested to evaluate its impact on the laser beam quality. Large Eddy Simulations representative of the amplifier cooling are performed using TrioCFD, a code developed by the CEA. First, validation simulations are carried out for heated periodic channel flows, allowing comparisons with Direct Numerical Simulation results from the literature. They are conducted using conjugate heat transfer calculation between the fluid and the solid. The channel flow is turbulent, with a bulk Reynolds number (based and the bulk velocity and the total height of the channel) of about 14000. Abstract

Spodoptera frugiperda est un insecte ravageur de culture polyphage qui créé d’importants dégâts sur le continent américain. Depuis quelques années, elle est devenue invasive en Afrique et en Asie et pourrait bientôt atteindre l’Europe. Pour mieux contrôler ce ravageur, de nouveaux moyens de lutte doivent être proposés aux agriculteurs. L’UMR DGIMI étudie les mécanismes d’interactions entre cette noctuelle, son cortège de pathogènes/parasites et sa plante hôte. Nous essayons notamment d’identifier des facteurs clefs impliqués dans les défenses immunitaires des insectes ou dans leur adaptation à la plante. Le RNAi n’étant pas fonctionnel chez les lépidoptères, nous ne disposions pas jusqu’alors d’outils de validation fonctionnelle de gènes in vivo chez Spodoptera frugiperda. Dans ce poster, nous vous présentons la mise en place de l’outil d’édition de génome CRISPR/Cas9 chez Spodoptera frugiperda. Nous avons généré deux lignées d’insectes portant chacune une mutation perte-de-fonction dans l’un des deux gènes pro-phénoloxidase (Sf-PPO1 et Sf-PPO2). L’activité de l’enzyme active issue du clivage de PPO, la phénoloxidase, est fortement diminuée dans l’hémolymphe des deux lignées mutantes comparée à celle observée dans l’hémolymphe de larves sauvages. L’outil « CRISPR/Cas9 » est donc fonctionnel chez Spodoptera frugiperda et les deux lignées obtenues vont maintenant nous permettre de mieux caractériser le rôle de chacune des PPO dans l’interaction de cet insecte avec les différents pathogènes étudiés au sein de l’unité DGIMI.

Abstract

A fundamental study of normal helium turbulence (He- I ) in von K\(\acute{a}\)rm\(\acute{a}\)n swirling flow generated with the help of one and only unique size facility called \(\mathbf {S}\)uperfluid \(\mathbf {H}\)igh \(\mathbf {RE}\)ynolds von \(\mathbf {K}\acute{a}\)rm\(\acute{a}\)n experiment (SHREK) available at CEA-Grenoble is presented here. This study comprises of measurements of instantaneous velocity using the hot wire anemometry technique for three different configurations of the flow such as co-rotation, clockwise counter rotation (hereafter called contra rotation) and counter-clockwise counter rotation (hereafter called anti-contra rotation). The maximum Taylor’s Reynolds Number, \(Re_{\lambda }\) for these measurements found to be up to 10000. A discussion on the aspect of intermittency is presented for the largest dataset available with the samples \(\mathcal {O}(10^{9})\). The probability density functions (PDF’s) of the velocity increments are discussed with its skewness and flatness along with the exponents of structure function up to certain order. These effects of various quantities of turbulence such as scales of turbulence, dissipation and \(Re_{\lambda }\) with respect to applied forcing is discussed. The scalings laws already established for classical turbulence are explored based on the present experimental data in cryogenic turbulence.

Abstract

Due to its very low viscosity, cryogenic helium has been used for years to generate high Reynolds turbulent flows. The measurement of velocity fluctuations in such high Reynolds flows is however a challenging issue, as it is necessary to develop small size (typically micron-length), robust sensors, to measure the whole spectrum of fluctuations of the velocity, which may extend to hundreds of kHz and possibly higher. SBT has developed for years different facilities, in particular taking benefit of the refrigeration capacities available at CEA Grenoble. In this article we present the current status of developments of hot wire sensors at CEA. Abstract

Background Spodoptera frugiperda (Noctuidae) is a major agricultural pest throughout the American continent. The highly polyphagous larvae are frequently devastating crops of importance such as corn, sorghum, cotton and grass. In addition, the Sf9 cell line, widely used in biochemistry for in vitro protein production, is derived from S. frugiperda tissues. Many research groups are using S. frugiperda as a model organism to investigate questions such as plant adaptation, pest behavior or resistance to pesticides. Results In this study, we constructed a reference transcriptome assembly (Sf_TR2012b) of RNA sequences obtained from more than 35 S. frugiperda developmental time-points and tissue samples. We assessed the quality of this reference transcriptome by annotating a ubiquitous gene family - ribosomal proteins - as well as gene families that have a more constrained spatio-temporal expression and are involved in development, immunity and olfaction. We also provide a time-course of expression that we used to characterize the transcriptional regulation of the gene families studied. Conclusion We conclude that the Sf_TR2012b transcriptome is a valid reference transcriptome. While its reliability decreases for the detection and annotation of genes under strong transcriptional constraint we still recover a fair percentage of tissue-specific transcripts. That allowed us to explore the spatial and temporal expression of genes and to observe that some olfactory receptors are expressed in antennae and palps but also in other non related tissues such as fat bodies. Similarly, we observed an interesting interplay of gene families involved in immunity between fat bodies and antennae. Abstract

International audience; The participatory illusion and the culturalist illusion are reducing the potential of tourism to contribute to achieving the Millennium Development Goals and to develop forms of wealth other than those that underpin utilitarianism, including decision making, sociability, and the experience of otherness. We highlight the possible effects of these two illusions on the basis of an analysis of the community-based tourism experience conducted in Benin by a French association for responsible tourism.; L’illusion participative et l’illusion culturaliste réduisent la capacité du tourisme à contribuer aux Objectifs du Millénaire pour le développement et à développer d’autres richesses que celles retenues par la tradition utilitariste, comme le pouvoir de décision, la sociabilité, l’expérience de l’altérité... Abstract

We present velocity spectra measured in three cryogenic liquid 4He steady flows: grid and wake flows in a pressurized wind tunnel capable of achieving mean velocities up to 5 m/s at temperatures above and below the superfluid transition, down to 1.7 K, and a "chunk" turbulence flow at 1.55 K, capable of sustaining mean superfluid velocities up to 1.3 m/s. Depending on the flows, the stagnation pressure probes used for anemometry are resolving from one to two decades of the inertial regime of the turbulent cascade. We do not find any evidence that the second order statistics of turbulence below the superfluid transition differ from the ones of classical turbulence, above the transition., Comment: 13 pages, 9 figures

Abstract

International audience; Cycle inhibiting factors (Cif) constitute a broad family of cyclomodulins present in bacterial pathogens of invertebrates and mammals. Cif proteins are thought to be type III effectors capable of arresting the cell cycle at G2/M phase transition in human cell lines. We report here the first direct functional analysis of CifPl, from the entomopathogenic bacterium Photorhabdus luminescens, in its insect host. The cifPl gene was expressed in P. luminescens cultures in vitro. The resulting protein was released into the culture medium, unlike the well characterized type III effector LopT. During locust infection, cifPl was expressed in both the hemolymph and the hematopoietic organ, but was not essential for P. luminescens virulence. CifPl inhibited proliferation of the insect cell line Sf9, by blocking the cell cycle at the G2/M phase transition. It also triggered host cell death by apoptosis. The integrity of the CifPl catalytic triad is essential for the cell cycle arrest and pro-apoptotic activities of this protein. These results highlight, for the first time, the dual role of Cif in the control of host cell proliferation and apoptotic death in a non-mammalian cell line.

Abstract

International audience; Superfluid turbulence (TSF) project uses liquid helium for the fundamental study of turbulent phenomena behind a passive grid and is able to work both in HeI and in HeII. Local and semi-local instrumentation was developed specifically for the purpose of this experiment(e.g. sub-micrometer anemometer, total head pressure tube and second sound tweezer). The difficulties encountered with this local and fragile instrumentation are discussed. Global characterization of the flow is presented including velocity, pressure, temperature stability and turbulence intensity. Finally, first results obtained with semi local measurements (total head pressure tube and second sound tweezer) both in the two phases of helium are presented.

Abstract

International audience; Lysozyme is an important component of the insect non-specific immune response against bacteria that is characterized by its ability to break down bacterial cell-walls. By searching an EST database from the fall armyworm, Spodoptera frugiperda (Negre et al., 2006), we identified five sequences encoding proteins of the lysozyme family. The deduced protein sequences corresponded to three classical c-type lysozymes Sf-Lys1, Sf-Lys2 and Sf-Lys3, and two lysozyme-like proteins, Sf-LLP1 and Sf-LLP2. Sf-Lys1 was purified from the hemolymph of Escherichia coli-challenged S. frugiperda larvae. The mature protein had a molecular mass of 13.975 Da with an isoelectric point of 8.77 and showed 98.3% and 96.7% identity with lysozymes from Spodoptera litura and Spodoptera exigua, respectively. As the other insect lysozymes, Sf-Lys1 was active against Gram positive bacteria such as Micrococcus luteus but also induced a slight permeabilization of the inner membrane of E. coli. Genes encoding these five Sf-Lys or Sf-LLPs were differentially up-regulated in three immune-competent tissues (hemocytes, fat body and gut) after challenges with non-pathogenic bacteria, E. coli and M. luteus, or entomopathogenic bacterium, Photorhabdus luminescens. Sf-Lys1 and Sf-Lys2 were mainly induced in fat body in the presence of E. coli or P. luminescens. Sf-Lys3, which had an acidic isoelectric point, was found to be the most up-regulated of all five Sf-Lys or Sf-LLPs in hemocytes and gut after challenge with P. luminescens. More molecular data are now available to investigate differences in physiological functions of these different members of the lysozyme superfamily.

Abstract

Photorhabdus species are gram-negative entomopathogenic bacteria of the family Enterobacteriaceae . Among the different members of the genus, one species, Photorhabdus asymbiotica , is a pathogen of both insects and humans. The pathogenicity mechanisms of this bacterium are unknown. Here we show that P. asymbiotica is a facultative intracellular pathogen that is able to replicate inside human macrophage-like cells. Furthermore, P. asymbiotica was shown for the first time in an intracellular location after insect infection. We also demonstrated that among Australian and American clinical isolates, only the Australian strains were able to invade nonphagocytic human cells. In cell culture infection experiments, Australian clinical isolates as well as cell-free bacterial culture supernatant induced strong apoptosis of a macrophage cell line at 6 h postinfection. American isolates also induced cellular death, but much later than that induced by Australian ones. Mammalian cultured cells analyzed for key features of apoptosis displayed apoptotic nuclear morphology, activation of the initiator caspases 8 and 9 and the executioner caspases 3 and 7, and poly(ADP-ribose) polymerase proteolysis, suggesting activation of both the intrinsic and extrinsic apoptotic pathways.

Abstract

Correspondance auteur: M. Brehélin brehelin@univ-montp2.fr; Haemocytes are the main immunocompetent cells in insect cellular immune reactions. Here, we show that in Spodoptera littoralis, granular haemocytes are the primary phagocyte haemocytes, both in vivo and in vitro. The "trigger" and "zipper" modes of engulfment known in mammal macrophages are active, in vivo, in S. littoralis granular haemocytes, together with macropinocytosis. Lipopolysaccharide as well as lipoteichoic acid inhibit the binding of both Gram-positive (Corynebacterium xerosis) and Gramnegative (Escherichia coli) bacteria on granular haemocytes. In addition, different ligands can inhibit the binding of E coli. Most of these inhibitors are known as ligands of scavenger receptors in mammal macrophages and we hypothesise that one of the receptors present on S. littoralis granular haemocytes could be a scavenger-like receptor

Abstract

International audience; Photorhabdus is an entomopathogenic bacterium belonging to the Enterobacteriaceae. The genome of the TT01 strain of Photorhabdus luminescens was recently sequenced and a large number of toxin-encoding genes were found. Genomic analysis predicted the presence on the chromosome of genes encoding a type three secretion system (TTSS), the main role of which is the delivery of effector proteins directly into eukaryotic host cells. We report here the functional characterization of the TTSS. The locus identified encodes the secretion/translocation apparatus, gene expression regulators and an effector protein - LopT - homologous to the Yersinia cysteine protease cytotoxin YopT. Heterologous expression in Yersinia demonstrated that LopT was translocated into mammal cells in an active form, as shown by the appearance of a form of the RhoA GTPase with modified electrophoretic mobility. In vitro study showed that recombinant LopT was able to release RhoA and Rac from human and insect cell membrane. In vivo assays of infection of the cutworm Spodoptera littoralis and the locust Locusta migratoria with a TT01 strain carrying a translational fusion of the lopT gene with the gfp reporter gene revealed that the lopT gene was switched on only at sites of cellular defence reactions, such as nodulation, in insects. TTSS-mutant did not induce nodule formation and underwent phagocytosis by insect macrophage cells, suggesting that the LopT effector plays an essential role in preventing phagocytosis and indicating an unexpected link between TTSS expression and the nodule reaction in insects

Die sakrale Kunst ist eine Komponente hauptsächlich von unserer westlichen Zivilisation. Ohne welche es nicht möglich ist die Schöpfung künstlerischer Kunst der jahrhunderte zu studieren. Der Zweck des Museum's ist nicht zu überzeugen, jedoch das Kulturelle wissen weiter zu geben, an die grosse Öffentlichkeit. Also hat das kunst-Museum nur eine kulturelle Aufgabe !, Le Musée d'art sacré du Gard procède d'une volonté : faire comprendre qu'au-delà des convictions des uns qu'ils transcrit en images, l'art sacré est une composante essentielle de notre civilisation occidentale sans laquelle il est impossible d'étudier la création artistique au cours des siècles. Abstract

At the beginning of the 12th century, Pope Gelasius II granted a diploma to Saint-André, in which, among other privileges, he confirmed the pre-eminence of the abbey over more than sixty obediences. This document shows that, since its foundation at the end of the 10th century, the monastery had built up a network of dependencies in Provence and had gathered a substantial temporal In order to do this, like other monasteries in the region, Saint-André was undoubtedly the depositary of part of the possessions of the aristocratic families. The relations maintained by the monks with the surrounding world are mainly based on the circulation of goods, and therefore on various transactions - donations, sales, disputes, restitutions - whose periodic renewal and the negotiations they give rise to contribute to the constitution and reproduction of the social fabric., Au début du XIIe siècle, le pape Gélase II octroie un diplôme à Saint-André, dans lequel, parmi d'autres privilèges, il confirme la prééminence de l'abbaye sur plus d'une soixantaine d'obédiences. Ce document montre que, depuis sa fondation à la fin du Xe siècle, le monastère avait constitué en Provence un réseau de dépendances et réuni un temporel conséquent. Pour ce faire, à l'instar d'autres monastères de la région, Saint-André a sans doute été le dépositaire d'une partie des possessions des familles aristocratiques. Les relations entretenues par les moines avec le monde environnant passent surtout par la circulation des biens, donc par des transactions diverses -donations, ventes, contestations, restitutions- dont le renouvellement périodique et les négociations qu'elles suscitent participent à la constitution et à la reproduction du tissu social.
