In domestic hens, the atresia of large white follicles (LWFs) directly affects the number of follicles that enter the hierarchical development and ovulation. Figuring out factors responsible for LWFs atresia is helpful to improve egg production of hens. At the LWF stage, yellow yolk begins to be deposited into the follicles via receptor mediated endocytosis, which requires large amounts of ATP. Mitochondrial oxidative phosphorylation (OXPHOS) is the primary source of ATP for follicular development. However, it is not clear whether the OXPHOS is changed along LWFs atresia. In this study, firstly, differences in morphological appearance, histology, cell proliferation, apoptosis, OXPHOS and mitochondrial dynamics between LWFs and atretic large white follicles (ALWFs) in hens at the peak laying stage (35W) were determined to elucidate whether OXPHOS changes in ALWFs. Then, these differences of LWFs between the peak laying hens (35W-LWFs) and the late laying hens (70W-LWFs) were detected to confirm whether OXPHOS changes during LWFs atresia. The results showed that ALWFs exhibited a wrinkled surface with several hemorrhage spots, and numerous intercellular vacuoles, as well as severe nuclear pyknosis. Compared to LWFs, a higher cell apoptosis rate and a lower proliferation rate were observed in ALWFs. In ALWFs, OXPHOS declined as manifested by reductions in ATP levels, ATP synthetase abundance, NAD+, NADH and NAD+/NADH ratio, and mRNA levels of genes associated with OXPHOS complexes I-V. Meanwhile, mitochondrial dynamics disequilibrium was detected in ALWFs as the expression levels of proteins and genes related to mitochondrial fusion (MFN1, MFN2, and OPA1) decreased, while the expression levels of proteins and genes related to mitochondrial fission (DRP1 and FIS1) increased. Further, compared to 35W-LWFs, 70W-LWFs showed a histology resembling to ALWFs, manifested as a slightly loosen structure of granulosa layers, and a lower cell proliferation rate. Moreover, both lower OXPHOS and impaired mitochondrial dynamics were detected in 70W-LWFs. In mammalian ovaries, most follicles do not ovulate and are eliminated by atresia, which primarily depends on granulosa cell (GC) apoptosis. Autophagy is an alternative mechanism involved in follicle depletion in mammals through independent or tandem action with apoptosis. However, follicular autophagy has not yet been investigated in sheep; therefore, the present study aimed to investigate the involvement of autophagy in atresia among a pool of growing antral follicles in ewe ovaries. The abundance of the autophagic marker LC3B-II was determined using western blotting in GCs collected from ewe antral follicles. The antral follicles were classified as healthy or atretic based on morphological criteria and steroid measurements in follicular fluid (FF). Immunofluorescence and confocal microscopy analyses were performed on GCs to evaluate the presence of autophagic proteins and their subcellular localisation. Caspase-3 and DNA fragmentation were assessed using western blotting and TUNEL assays, respectively, in the same GC population to investigate the simultaneous apoptosis. The novel results of this study demonstrated enhanced LC3B-II protein expression in GCs of atretic follicles compared to that of healthy ones (1.3-fold increase; P = 0.0001, ANOVA), indicating a correlation between autophagy enhancement in GCs and antral follicular atresia. Autophagy, either functioning independently or in tandem with apoptosis, may be involved in the atresia of growing antral follicles in ewe ovaries because atretic GCs also showed high levels of apoptotic markers. • ICG-FA is used to determine anastomotic perfusion rates, thereby reducing surgical complications and aiding in intraoperative decision-making in adults, but not yet extensively described in pediatric surgery. • This systematic review analyzes the current knowledge on usefulness of ICG for intestinal perfusion assessment in pediatric gastrointestinal surgery and its safety in neonates. • ICG might be useful for intraoperative intestinal perfusion assessment, perhaps even more than conventional clinical assessment. • The safety profile of ICG looks promising in neonates. This systematic review analyzes whether ICG is useful for intestinal perfusion assessment in pediatric gastrointestinal surgery and safe to use in neonates. Systematic searches of PubMed, EMBASE & MEDLINE and CENTRAL were performed (last conducted December 6, 2021). The main inclusion criteria were (1) use of ICG for intestinal perfusion assessment and (2) use of ICG in young infants. Exclusion criteria were lack of an English or Dutch full-text and MINORS quality score <60%. Data was presented in overview tables. The usefulness in pediatric gastrointestinal surgery was assessed by surgical outcome. Safety of ICG in neonates was assessed by complication or adverse event occurrence. Regarding intestinal perfusion assessment, four studies were included, reporting 45 patients (median age 1.5 years). ICG was considered useful for anastomotic blood flow evaluation and intraoperative determination of resection length. Regarding ICG safety in neonates, eight studies were included, reporting 46 infants (median age 24.9 days), of which 18 neonates. All but one studies reported the absence of complications or adverse events. Two studies reported subcutaneous dye retention, which fully disappeared within two weeks. Although the number of available studies is small, ICG might be useful for intraoperative intestinal perfusion assessment, perhaps even more than conventional clinical assessment. Furthermore, its safety profile looks promising in neonates. Larger prospective studies are necessary to confirm these assumptions and seem warranted given the safety profile. Intestinal atresia is a congenital condition characterized by the blockage or constriction of an intestine portion. It is caused by either a vascular issue or a failure of recanalization during fetal development. Surgery is essential for both a definitive diagnosis and therapy. A preterm neonate (gestational age 34 weeks) was delivered by cesarean section due to polyhydramnios and complained of recurrent episodes of non-bilious vomiting after breastfeeding. Prenatal ultrasound revealed a double bubble sign, indicating intestinal atresia, while on contrast imaging, the contrast was refluxed from the stomach to the esophagus, the absence of gases in the intestine or colon suggested a gastric outlet obstruction. The atretic intestine was identified during laparotomy, from the second section of the duodenum to the rectum with no lumen. There were no surgical options available. Atresia is typically treated by excision and anastomosis of the remaining normal canalized intestine. However, in our case, a total intestinal atresia was discovered, and no intestinal lumen was identified to undergo surgery. It was found that the atretic intestine in our case extended from the duodenum to the rectum. However, in the two previously documented cases of atresia, the atresia began in the jejunum and progressed to the rectum in one instance and the sigmoid colon in the other. Because there were no alternative surgical options, the intestine was relocated, and the abdomen was closed. The objective of this study was to investigate the early follicular apoptosis in canine ovarian follicles by examining the expression of anti-apoptotic BCL-2 and pro-apoptotic BAX proteins throughout the estrous cycle associated with oocyte maturation. Follicular cells from preantral and antral follicles of varying sizes were isolated and grouped based on follicle type and estrous phase. Antral follicles underwent flow cytometry analysis, whereas preantral follicles were subjected to Western blotting. The meiotic capacity of oocytes from these different follicle types was evaluated through in vitro maturation. Statistical analysis included ANOVA and Duncan's test. Results showed fluctuations in BCL-2 and BAX levels across different follicular stages and estrous phases. BCL-2 levels increased (P <0.05) with follicular development in antral follicles, particularly during estrus, while BAX exhibited variations peaking (P <0.05) during estrus. The BCL-2/BAX ratio in antral follicles was higher (P <0.05) in estrus and diestrus compared to anestrus and proestrus. Additionally, BCL-2 and BAX proteins were detected in preantral follicles, with varying expression levels (P <0.05) across estrous phases. The BCL-2/BAX ratio in preantral follicles was highest (P <0.05) during anestrus and estrus and decreased (P <0.05) in proestrus and diestrus. Oocytes from preantral follicles did not reach the MII stage, regardless of the levels of BCL-2/BAX. Conversely, oocytes obtained from large follicles during estrus showed the highest (P <0.05) maturation percentages, which were associated with the highest BCL-2/BAX ratio. Intestinal atresia is an uncommon cause of intestinal obstruction. In the vast majority of the cases, signs and symptoms develop early in the neonatal period. Presentations beyond the neonatal period are rare. A former premature female infant who was otherwise healthy developed recurrent, non-bilious vomiting associated with feedings at the age of 8 months. Up until that time her weight gain had been appropriate, and she had regular bowel movements. Over the course of the following weeks, the vomiting became more frequent. A few days after her 9-month well-child visit during which she still appeared to be in good health, her vomiting turned bilious and was not related to feedings. Her parents brought her to the emergency room. She was admitted to the hospital and underwent imaging studies to investigate the source of what seemed to be an intestinal obstruction. The upper gastrointestinal series (UGI) showed delayed passage of contrast, and a caliber change in the proximal small bowel. The computerized tomography (CT) confirmed the findings, which were suggestive of a partial proximal intestinal obstruction. She underwent a laparotomy. We identified an area of caliber change 15 cm distal to the ligament of Treitz. We opened the proximal side and identified a jejunal membrane that had a pinhole opening in the center. We resected a 4-cm segment of jejunum that included the caliber transition and the membrane and did an end-to-end anastomosis. She tolerated the operation well and was discharged home 12 days later. At the age of 1 year, she is thriving well. Although type-I jejunal atresia typically presents in the neonatal period, it should be included in the differential diagnosis of infants with recurrent vomiting and signs of a partial bowel obstruction. Esophageal atresia (EA) with or without trachea-esophageal fistula is relatively common congenital malformation with most patients living into adulthood. As a result, care of the adult patient with EA is becoming more common. Although surgical repair has changed EA from a fatal to a livable condition, the residual effects of the anomaly may lead to a lifetime of complications. These include effects related to the underlying deformity such as atonicity of the esophageal segment, fistula recurrence, and esophageal cancer to complications of the surgery including anastomotic stricture, gastroesophageal reflux, and coping with an organ transposition. This review discusses the occurrence and management of these conditions in adulthood and the role of an effective transition from pediatric to adult care to optimize adult care treatment. Biliary atresia (BA) is an idiopathic, progressive, fibroinflammatory, and obliterative disease of the biliary tract that manifests in the neonatal period with scleral icterus, acholic stools, conjugated hyperbilirubinemia, and elevated serum-gamma glutamyl transferase. Definitive diagnosis is established by a multi-disciplinary team comprising of pediatricians, hepatologists, radiologists, surgeons, and pathologists. It is currently the most common cause of pediatric liver transplantation in the USA and worldwide. More than 100 diseases can present with neonatal cholestasis mimicking BA. Early diagnosis (<60 days of age) with a timely Kasai portoenterostomy (KP) is crucial to relieve biliary obstruction and achieve longer native liver survival. Histopathologic features of BA can differ based on age and specific surgical interventions. This review describes histopathologic features of BA at various stages and focuses on a variety of common and uncommon diseases mimicking BA. With the increasing use of molecular testing and comprehensive genetic cholestasis panels, several patients who would have otherwise been diagnosed as BA, are now classified with specific genetic conditions leading to cholestasis. Infertility in hyperprolactinemic females is attributed to the dysregulation of GnRH release, subsequently affecting gonadotropin levels, and ultimately leading to anovulation. However, in addition to the hypothalamus, prolactin receptor (PRLR) is expressed in ovaries as well, suggesting potential local effects of PRL in cases of hyperprolactinemia. We have developed an experimental model of sulpiride (SPD)-induced hyperprolactinemia using a wild rodent, the plains vizcacha, and studied the implications of pharmacological PRL levels on folliculogenesis and steroid production. Ovaries of SPD females showed a striking number of atretic follicles along with a reduction in the collective number of viable follicles leading to a higher atretic/viable follicle ratio compared to that of control females (CTL) (P < 0.05). In terms of sensitivity to the hormonal environment, SPD ovaries substantially changed their potential responsiveness to pituitary PRL, as evidenced by the three-fold increase in PRLR expression alongside heightened expression of both gonadotropic receptors in comparison to CTL ovaries (P < 0.05). Circulating estradiol (E2) values doubled post-treatment in the SPD females, which also showed higher expressions of aromatase and 17β-hydroxysteroid dehydrogenase, along with both E2 receptors, ERα and ERβ than the CTL group (P < 0.05). Our findings strongly suggest that hyperprolactinemia-dependent dysregulation of ovarian function can be explained at least partially, by PRL direct actions facilitated by the heightened expression of PRLR in follicles and corpora lutea. Possibly these PRL actions synergize with those triggered by gonadotropic hormones ultimately leading to alteration of the steroidogenic pathway, folliculogenesis disruption and increased atresia. Sacrococcygeal teratomas (SCTs) and jejunoileal atresias are both rare (1 in 35,000–40,000 births and 1 in 5000 births, respectively). We present a case of a concomitant diagnosis of a cystic sacrococcygeal teratoma and a jejunoileal atresia. A twenty-week screening ultrasound identified a cystic pelvic mass (5.5 x 2.8 × 2.2 cm), dilated bowel, and mild polyhydramnios in an otherwise healthy G1P0 woman. Fetal magnetic resonance image showed a multiseptated presacral cystic mass suspicious for an internal sacrococcygeal teratoma and dilated bowel, thought to represent dilated ascending and transverse colon. Scheduled delivery at 39 weeks of gestation was uncomplicated. Postnatal ultrasound and MRI were consistent with prenatal findings. Given the rarity of both congenital anomalies and no known association between SCTs and jejunoileal atresia, the prenatal bowel dilation was thought to be due to partial obstruction secondary to the SCT. The baby thus underwent SCT resection on day-of-life 2. Ongoing obstruction after resection prompted a contrast enema which demonstrated a microcolon with reflux into the terminal ileum but no reflux into more dilated proximal loops concerning for intestinal atresia. On day-of-life 5, the baby underwent exploratory laparotomy which identified a type-2 ileal atresia, and a primary anastomosis was performed. The patient had an uneventful recovery and was discharged home on day-of-life 22. Prenatal findings of dilated bowel should prompt providers to have a high index of suspicion for jejunoileal atresia, even if other diagnoses exist. [ABSTRACT FROM AUTHOR]

To adequately counsel future patients with auricular abnormalities, this study aimed to analyze the therapeutic choices patients make after consultation concerning their auricle and/or hearing. All patients who visited the auricle consultation between January 2010 and January 2020 were included. This comprises patients with all types of auricular anomalies of both congenital and acquired etiology. Follow-up was performed by telephone to obtain informed consent and to take a short nonvalidated questionnaire regarding their therapeutic choices and satisfaction with these choices. Patient characteristics and information regarding therapeutic choices after consultation were subsequently obtained from the patients' medical files. The main outcome parameters were the patients' therapeutic choices regarding the auricle and hearing. A total of 134 patients (61.2%) were included with a mean follow-up of 3.85 years. For congenital and acquired etiologies, 72.8% and 18.2% declined reconstruction, 14.0% and 18.2% chose autologous, 5.3% and 0% chose alloplastic, 6.1% and 45.4% chose prosthetic, and 1.8% and 18.2% chose another type of reconstruction, respectively. A total of 54.9% declined hearing rehabilitation, while 38.2% chose percutaneous bone conduction device (BCD), 2.0% transcutaneous BCD, and 4.9% BCD on a softband. The overall satisfaction score was 8.7/10 concerning the auricle and 8.3/10 regarding hearing. In conclusion, most patients with a congenital etiology chose to wait or decided against auricular reconstruction, while patients with acquired etiologies mostly opted for reconstruction by a prosthesis. Patients or their caregivers were very satisfied with their choice, regardless of whether this implies reconstruction or no reconstruction, which emphasizes the importance of shared decision-making and thorough counseling. Patterns of quantitative production of follicles, their growth, and degeneration in the adult ovary of zebrafish (Danio rerio) in response to long-term (80 days) exposure to environmental estrogens (EE) in the laboratory, were studied. Experimentally naive female D. rerio procured from fish farm were acclimated to the laboratory (natural temperature, 26 ± 1° C, photoperiod, 11.30 L:12.30 D) for two weeks and divided into 10 groups. Each group (n = 20) was housed in a separate glass aquarium containing 10 L of conditioned water (physico-chemical parameters maintained within the permissible range prescribed for zebrafish) along with either 5 ng or 10 ng/L of 17α-ethynylestradiol (EE 2) or diethylstilbestrol (DES) or bisphenol A (BPA) or estradiol 17-β (positive control) or water with no chemical (negative control). All experimental fish were fed twice daily on commercial pellets (ad libitum) supplemented with Artemia nauplius, the exposure was semi-static and chemical residues in media samples were determined by ultra-performance liquid chromatography (UPLC). Exposure of fish to estrogens increased (p < 0.05) (i) body mass and gonadosomatic indices (GSI) in E 2 , EE 2 and DES groups (ii) previtellogenic and vitellogenic follicles in E 2 and EE 2 groups (iii) atretic follicles (AF) in DES and BPA groups compared to controls and (iv) decrease in total oocyte volumes (V = 4/3. π. r3) compared to those of E 2 group. During the hierarchical development of follicles in laying hens, a large number of small white follicles (SWFs) undergo atresia caused by apoptosis rather than growing into large white follicles (LWFs), which reduces the number of follicles developing into hierarchical preovulatory follicles and results in declines in egg production. Therefore, figuring out factors responsible for SWFs atresia is necessary to improve egg production of laying hens. Endoplasmic reticulum stress (ERS) is one of the possible causes of cell apoptosis. However, the association between SWFs atresia and ERS in hens is yet to be answered. In this study, cell proliferation and apoptosis, expression levels of proteins and genes related to ERS, and cell ultrastructure were compared to elucidate whether ERS occurred in atretic small white follicles (ASWFs). Subsequently, a tunicamycin-induced ERS model and a serum withdrawal-induced atresia model of SWFs in vitro were established to investigate whether ERS could lead to follicular atresia. The results showed that the cell apoptosis increased significantly while the cell proliferation decreased evidently in ASWFs compared with SWFs. Meanwhile, swollen and vacuolization deformation of ERs was observed in the GCs but not TCs of ASWFs. Results of experiments in vitro demonstrated that treatment with serum withdrawal or tunicamycin inhibited cell proliferation and promoted cell apoptosis in SWFs, and the adverse effects of tunicamycin on SWFs manifested a dose-dependent increase. In comparison with the controls, treatment with serum withdrawal or tunicamycin upregulated expression levels of proteins and genes in ERS pathway. Moreover, the dose of 5 μg/mL tunicamycin seemed to be the optimal concentration for establishing the tunicamycin-induced ERS model. Meanwhile, the results of cell ultrastructure observation showed that ERs in GCs of SWFs of serum withdrawal group and treatment with 5 μg/mL tunicamycin group became swollen, dilatation and vacuolization. Recently, we reported the differential expression of kisspeptinergic system in the bubaline hypothalamus and corpus luteum. Here, we document the expression of kisspeptin (Kp) and its receptor (Kiss1r) in the ovarian follicles of the buffalo with respect to the functional status. Follicles of ≥10 to ≤13 mm diameter (n = 45) were retrospectively categorized into active (n = 18), intermediate (n = 16) and atretic (n = 11) follicles based on the concentrations of intrafollicular progesterone (P 4) and estradiol (E 2). The P 4 :E 2 ratio was significantly lower in the active follicle (0.43 ± 0.08) than that of the intermediate (3.46 ± 0.53) and atretic (28.4 ± 10.6) follicles (P < 0.05). Relative fold change in the transcripts of kisspeptin (Kiss1), Kiss1r, gonadotrophin receptors, steroid acute regulatory protein (StAR), cytochrome P450 family 11 subfamily A member 1 (CYP11A1), cytochrome P450 Family 19 subfamily A member 1 (CYP19A1), insulin like growth factor −1 (IGF-1), apoptotic factors (caspase 3 and B-cell lymphoma 2, BCL2) was calculated using qPCR in the follicular wall of the three categories of follicle (n = 8/group). In another experiment, histological sections of the ovary (n = 41) were used to group the follicles as described above and immunostaining of Kp, Kiss1r and aromatase was done. A significant upregulation of StAR, CYP11A1 and CYP19A1 in the active follicles supported the endocrine basis of follicular classification. The transcripts of Kiss1 and Kiss1r were upregulated by 19.45 fold and 4.25 fold, respectively in the active follicle as compared to other groups. Immunolocalization studies revealed that Kp and Kiss1r were localized to the basal and antral granulosa cells (GC) of the active and intermediate follicles; however, the staining intensity was stronger in the former group. Strong expression of CYP19A1 in the GC layer of active follicle supported the histological basis of defining the functional status of the follicle. It is concluded that the follicular compartment of the bubaline ovary expressed the constituents of kisspeptinergic system. This case report describes a rare disorder of a left pulmonary artery interruption with concurrent coarctation of the right pulmonary artery in a dog. A 5-year-old, male neutered, mixed-breed dog presented for evaluation of an asymptomatic heart murmur. Echocardiography and computed tomography revealed complete interruption of the proximal left pulmonary artery and coarctation of the right pulmonary artery. The oesophagus is subject to a number of disorders affecting motility (swallowing) including gastro-oesophageal reflux disease, malignancy and disorders of neuromuscular function, such as achalasia. Most neoplasms of the oesophagus and stomach are epithelial in nature. Benign epithelial neoplasms usually take the form of polypoid lesions, such as oesophageal squamous papillomas, gastric adenomas, hyperplastic and fundic gland polyps. Malignant epithelial neoplasms of the oesophagus are divided into squamous cell carcinoma and adenocarcinoma, whereas malignant gastric neoplasms are predominantly adenocarcinomas. Each tends to develop in the context of dysplastic epithelial changes. Gastric carcinomas and lymphomas are associated with Helicobacter pylori infection. Stromal tumours of the stomach are important in that they have a distinctive molecular pathology and linked targeted therapy. This educational article provides an overview of the incidences, aetiologies and histological features of some of the most common mechanical, inflammatory and neoplastic pathologies encountered in the oesophagus and stomach. Acquired external auditory canal atresia is characterized by fibrous tissue formation in the ear canal, hearing loss and chronic otorrhea. Although the disease can be treated surgically, the recurrence rate is high. This study explored whether autologous oral mucosal epithelial cell sheets could be used as a novel therapy for ear canal atresia. We succeeded in generating a rabbit model of acquired external auditory canal atresia by dissecting the skin of the ear canal. Endoscopic and histological findings in this model indicated that atresia developed over a 4-week period and was not inhibited by the placement of polyglycolic acid sheets immediately after skin dissection. By contrast, transplantation of autologous oral mucosal epithelial cell sheets, which had been fabricated by culture on temperature-responsive inserts without a feeder layer, prevented the development of atresia during the 4-week period after skin dissection. Transplantation of autologous epithelial cell sheets after surgical treatment of acquired external auditory canal atresia could be a promising new method to reduce the risk of disease recurrence. In salmonids, exposure to elevated temperature impairs oogenesis. As such, there is a need to understand the molecular mechanisms that underpin this process, and develop mitigation strategies that maintain or rescue reproductive development in broodstock. In this study, follicle stimulating hormone (Fsh) and/or insulin-like growth factor 1 (Igf1) treatment were assessed for their ability to promote reproductive function at 14 and 22 °C in ovarian follicles from coho salmon in vitro. Maintenance at 22 °C resulted in the downregulation of fsh receptor , 17α -hydroxylase/C17,20-lyase and p450 aromatase a (cyp19a1a), and connexin 34.3 (cx34.3). While combined treatment with Fsh and Igf1 stimulated the expression of cyp19a1a at 14 °C, this treatment was not effective at 22 °C. Upregulation of cx34.3 occurred in response to treatments that contained Igf1 regardless of temperature, and there is evidence to suggest that apoptosis was inhibited to some extent at 22 °C following combined treatment with Fsh and Igf1. Congenital unilateral absence of the pulmonary vein (UCAPV) is a rare entity with characteristic clinical and imaging findings. Despite its congenital nature, the radiographic findings and symptoms of UCAPV may not be recognized at birth and patients may present in childhood or early adulthood with findings that may mimic other diagnoses. The evolution of imaging findings in UCAPV is presented through two cases, one of which demonstrates the progression of findings over several years. The embryologic basis of this entity is reviewed and the clinical presentation and characteristic imaging findings including radiographs, nuclear scintigraphy, computed tomography, magnetic resonance imaging and cardiac catheterization are demonstrated. Characteristically, normal at birth, radiographs demonstrate the gradual development of a small lung and ipsilateral pulmonary artery over time. In addition to unilateral absence of the pulmonary veins on CT or MRI, a mediastinal "soft tissue mass" reflecting the development of mediastinal collaterals is a common finding and should be recognized as secondary to the absent ipsilateral pulmonary veins rather than as a primary process causing occlusion of the pulmonary veins. Scintigraphy will show absent perfusion to the affected lung. Abstract Acquired urethrovaginal fistulae and urethral atresia are rare findings in pediatric patients, but have been described in adult patients related to trauma or iatrogenic injury. Little exists in the published literature to guide management of such conditions in children, but lessons learned from congenital causes can help. Herein we discuss the preoperative evaluation and management of a child with an acquired urethrovaginal fistula and urethral atresia likely related to in utero compression from an intrapelvic sacrococcygeal teratoma and provide several images detailing the complex anatomy. Abstract Many teleost fishes exhibit sequential hermaphroditism, where male or female gonads develop first and later undergo sex change. Model sex change species are characterized by social hierarchies and coloration changes, which enable experimental manipulations to better understand these processes. However, other species such as the protogynous black sea bass (Centropristis striata) do not exhibit these characteristics and instead receive research attention due to their importance in fisheries or aquaculture. Black sea bass social structure is unknown, which makes sex change sampling difficult, and few molecular resources are available. The purpose of the present study was to induce sex change using exemestane, an aromatase inhibitor, and assess gonadal gene expression using sex markers (amh , zpc2) and genes involved in steroidogenesis (cyp19a1a , cyp11b), estrogen signaling (esr1 , esr2b), and apoptosis or atresia (aen , casp9 , fabp11 , parg , pdcd4 , rif1). Overall, dietary exemestane treatment was effective, and most exposed females exhibited early histological signs of sex change and significantly higher rates of ovarian atresia relative to control females. Genes associated with atresia did not reflect this, however, as expression patterns in sex changing gonads were overall similar to those of ovaries, likely due to a whole ovary dilution effect of the RNA. Still, small but insignificant expression decreases during early sex change were detected for ovary-related genes (aen , casp9 , fabp11 , zpc2) and anti-apoptotic factors (parg , rif1). Exemestane treatment did not impact spermatogenesis or testicular gene expression, but testes were generally characterized by elevated steroidogenic enzyme and estrogen receptor mRNAs. Further research will be needed to understand these processes in black sea bass, using isolated ovarian follicles and multiple stages of sex change. Abstract Purpose The diagnosis of "closing" or "closed gastroschisis" is made when bowel is incarcerated within a closed or nearly closed ring of fascia, usually with associated bowel atresia. It has been described as having a high morbidity and mortality. Methods A retrospective review of closing gastroschisis cases (n = 53) at six children's hospitals between 2000 and 2016 was completed after IRB approval. Results A new classification system for this disease was developed to represent the spectrum of the disease: Type A (15%): ischemic bowel that is constricted at the ring but without atresia; Type B (51%): intestinal atresia with a mass of ischemic, but viable, external bowel (owing to constriction at the ring); Type C (26%): closing ring with nonviable external bowel +/− atresia; and Type D (8%): completely closed defect with either a nubbin of exposed tissue or no external bowel. Overall, 87% of infants survived, and long-term data are provided for each type. Conclusions This new classification system better captures the spectrum of disease and describes the expected long-term results for counseling. Unless the external bowel in a closing gastroschisis is clearly necrotic, it should be reduced and evaluated later. Survival was found to be much better than previously reported. Currently, the most important determinant of gastroschisis outcomes in high resource settings is whether the condition is associated with intestinal complications, such as atresia, necrosis, perforation, or volvulus. This form of the anomaly, known as complex gastroschisis, accounts for most of the mortality and a disproportionate burden of the morbidity from gastroschisis. There is some disagreement about what constitutes complex gastroschisis, and little consensus on the type and timing of surgical interventions. This article establishes a clear definition of complex gastroschisis. Surgical approaches to treatment of the diverse presentations of complex gastroschisis will be described and the timing of such interventions will be discussed. The negative effects of ambient ultraviolet (UVA) on the water environment have been recently highlighted; UVA can create deleterious effects by stimulating stress on pelagic organisms. Little is known about UVA effects on oocyte characteristics of female fish. In the present study we explored the effects of exposure to ecologically relevant levels of simulated UVA radiation on ovaries of two major strains WT (HdrR) and P53 (-/-) of medaka ( Oryzias latipes ) mature female. Fish were assigned to control and three UVA-exposed groups as (15 min, 30 min, and 60 min/day) for three days and sample selection was 24 h and 14 days after exposure. Histological alterations and oocyte atresia percentage were analyzed in the UVA-exposed fish compared to control. Alteration comprised hyperthrophied follicular cells with increased thickness, breakdown of egg chorion (zona radiata), damage of cortical alveoli, and distorted nucleus and cytoplasm. The atresia percentages significantly increased with higher UVA exposure dose and time for both the wild type and the p53 deficient fish. The wild type displayed significantly higher oocyte atresia percentage than the p53 mutant. These results suggested that UVA exposure provoked histological alterations in both p53 and WT medaka oocytes leading to follicular atresia, which reduce female reproductive ability and larval production. UVA oocyte response showed p53 dependent and independent histological alteration, however, the p53 mutant was less sensitive to UVA than the wild type in medaka fish. [ABSTRACT FROM AUTHOR]

Jejunoilial atresia is a frequent cause of bowel obstruction in neonates, with prevalence around 0.7 per 10000 livebirths. It is believed to be related to an intrauterine vascular accident that commonly presents as isolated pathology. Although, other associated anomalies have been reported on rare occasions, the association between the atresia and jejunal diverticulosis in newborn has never been described in literature. Furthermore, jenunal diverticulosis is rare below 40 and never been reported in children. Here in, we share the first reported case of this association and the first case of jejunal diverticulosis in this age category, the challenge in its management and provide review on literature. Objective(s): We aimed to analyze if anti-apoptotic agent sphingosine-1-phosphate offers protection against in vitro follicle atresia during culture of human ovarian cortical samples.Study Design: A translational research study of ex-vivo and in-vitro models of human ovarian tissue.Material and Methods: Ovarian cortical tissue fragments (1 × 0.5 cm) were obtained from young patients (n = 15 mean age ± SD: 29.4 ± 2.5) undergoing laparoscopic excision of benign ovarian cysts. The samples were cultured for 4 days in 24-well format culture plate using conventional culture techniques. S1P was added to culture media at 200 and 400 μM concentrations. At the end of culture period the samples were processed for both histomorphological assessment and detection of apoptosis with immunohistochemistry and western blot methods using apoptosis marker cleaved caspase-3. In vitro estradiol (E2) and AMH productions of the samples were measured with ELISA. Follicle counts were expressed as the mean number of follicles per mm2.Results: The mean numbers of primordial and secondary follicles were 3.2 ± 0.4 and 0.7 ± 0.2 respectively, in the fresh fixed uncultured samples. After four days of culture their numbers were significantly decreased to 0.8 ± 0.2 (p < 0.01) and 0.1 ± 0.05 (p < 0.05) respectively, in the control samples cultured without S1P compared to fresh fixed samples. S1P treatment decreased follicle atresia and significantly higher number of primordials (2.3 ± 0.3, p < 0.01) and secondary follicles (0.5 ± 0.1, p < 0.05) survived in the samples after 4 day culture period compared to those cultured without S1P. In line with this there was dose-dependent decrease in the protein expression of cleaved caspase-3 on western blot and in the number of apoptotic follicles stained positive for cleaved caspase-3 on immunohistochemistry in the samples incubated with S1P at 200 and 400 μM concentrations. Furthermore, those samples incubated with S1P produced significantly higher amounts of E2 (2339 ± 321 vs. 1156 ± 125 pg/mL respectively, p < 0.01) compared to control samples.Conclusions: These results suggest that S1P promotes follicle survival in human ovarian cortical samples in vitro. The majority of activated ovarian follicles undergo atresia during reproductive life in mammals, and only a small number of follicles are ovulated. Though hormone treatment has been widely used to promote folliculogenesis, the molecular mechanism behind follicle selection and atresia remains under debate due to inconsistency among investigation models. Using a high-throughput molecular pathology strategy, we depicted a transcriptional atlas of mouse follicular granulosa cells (GCs) under physiological condition and obtained molecular signatures in healthy and atresia GCs during development. Functional results revealed hypoxia-inducible factor 1 (HIF1) as a major effector downstream of follicle-stimulating hormone (FSH), and HIF1 activation is essential for follicle growth. Energy shortage leads to prevalent AMP-activated protein kinase (AMPK) activation and drives follicular atresia. FSHR-mTOR-HIF1 signaling helps follicles escape from the atresia fate, while energy stress persists. Climate change associated temperature challenges pose a serious threat to the marine environment. Elevations in average sea surface temperatures are occurring and increasing frequency of marine heatwaves resulting in mortalities of organisms are being reported. In recent years, marine farmers have reported summer mass mortality events of the New Zealand Greenshell mussel, Perna canaliculus, during the summer months; however, the etiological agents have yet to be determined. To elucidate the role of thermal stress, adult P. canaliculus were exposed to three chronic temperature treatments: a benign control of 17 °C and stressful elevations of 21 °C and 24 °C. Eight mussels per treatment were collected each month throughout a 14-month challenge period to identify and investigate histopathological differences among P. canaliculus populations exposed to the three temperatures. Histopathology revealed several significant deleterious alterations to tissues associated with temperature and exposure time. Increasing temperature and progression of time resulted in 1) an increase in the number of focal lipofuscin-ceroid aggregations, 2) an increase in focal hemocytosis, 3) an increase in the thickness of the sub-epithelial layer of the intestinal tract and 4) a decreased energy reserve cell (glycogen) coverage in the mantle. Prolonged exposure, irrespective of temperature, impacted gametogenesis, which was effectively arrested. Furthermore, increased levels of the heat shock protein 70 kDa (HSP 70) were seen in gill and gonad from thermally challenged mussels. The occurrence of the parasite Perkinsus olseni at month 5 in the 24 °C treatment, and month 7 at 21 °C was unexpected and may have exacerbated the fore-mentioned tissue conditions. Prolonged exposure to stable thermal conditions therefore appears to impact P. canaliculus , tissues with implications for broodstock captivity. Mussels experiencing elevated, temperatures of 21 and 24 °C demonstrated more rapid pathological signs. Biological data were retrieved from 374 female Atlantic wolffish ( Anarhichas lupus ) collected from 2001–2006 at A. lupus main spawning grounds in Iceland and at two other locations with different sea temperature regimes to investigate spatial and temporal difference in fecundity and indications of atresia. There was no spatial difference in fecundity but temporal difference was found, and fecundity decreased in time, clearly indicating the presence of atresia. A. lupus that was 57 cm long was 11% more fecund in the year 2003 than in 2005. Egg numbers decreased about 20% from June to October for 67 cm long A. lupus . In aquaculture of species with a large body size such as tuna and yellowtail, a major problem still is that broodstock management for egg collection requires high feeding cost and labor. In order to reduce feeding costs, development of restricted feeding techniques without affecting the reproductive performance may be useful. To gain a better understanding of reproductive responses to food supply, we examined the effect of restricted feeding during the previtellogenic and vitellogenic phases on sexual maturation in female yellowtail, Seriola quinqueradiata . Restricted feeding during the vitellogenic phase tended to reduce ovary weight relative to normally fed controls at the end of the experiment. Histological observations revealed that both restricted feeding regimes do not affect the percentage of atretic follicles as compared to the control group. Nevertheless, the mean diameter of the most advanced follicles of the females subjected to restricted feeding during the vitellogenic phase was significantly smaller to normally fed controls. These results suggest that reduced food intake during the vitellogenic phase is likely to delay oocyte growth, and lead to low ovarian weight. Endocrine analyses showed that restricted feeding during the vitellogenic phase reduced plasma 17β-estradiol levels, but not plasma and pituitary gene expression levels of gonadotropins (follicle-stimulating hormone and luteinizing hormone) at the end of the experiment. Transcripts for ovarian steroidogenesis-related genes, such as P450 side-chain cleavage ( cyp11a1 ) and 3β-hydroxysteroid dehydrogenase ( hsd3b ) after food restriction during the vitellogenic phase tended to be high. Therefore, the effects of restricted feeding on reproduction would not be mediated by alterations in circulating gonadotropin levels, but rather through direct effects on the ovarian steroid production in yellowtail females. During the course of a QTL study involving an intercross between White Plymouth Rock chickens and Red Jungle Fowl, certain reproductive anomalies such as atresia and double ovary-oviduct became evident. Observed in reciprocal pedigreed F1 crosses were 2 full-sib pullets with dual functioning ovaries. One also had complete functioning left and right oviducts. The other had asymmetrical reproductive tracts with a typical left oviduct and a rudimentary right oviduct. In addition, there were 3 females with atresia. Although they had different sires and dams, their grandparents were related, and all 5 traced to a common ancestor. Objectives Tracheal atresia (TA) is a rare congenital condition that typically requires an unexpected and emergent resuscitation in the delivery room. The mortality rate associated is very high, with only a few long-term survival cases reported. We describe the findings of a systematic review on the clinical presentation and airway management of TA. Methods Using the keywords “tracheal atresia”, “tracheal agenesis” and “tracheal hypoplasia” a search through Embase and Pubmed databases was performed following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) protocol. Articles published from 1950 to 2015 in English, French, Italian, Portuguese and Spanish were included. Exclusion criteria were cases of stillborn, and unclear diagnosis or outcome. Results 149 cases of TA were identified after reviewing 1125 initial references. There was a male preponderance (65%), and associated malformations were described in 94.2% of patients. Prenatal ultrasound was abnormal in 56.3% of cases, with polyhydramnios being the most common finding. The most frequent type of TA was Faro Type C. 94 (41.3%) patients did not survive beyond the first 24 h of life. Only 13 (8.4%) patients survived more than three months of life, after undergoing a variety of surgical approaches. Conclusion This review, which to our knowledge is the largest one to date, confirms that TA is a rare malformation, occurs more frequently in males, and has a very high mortality rate. From the 1940s through 1977, at least 590,000 kg of polychlorinated biphenyls (PCBs) were released into the Hudson River from General Electric manufacturing plants located in Hudson Falls and Fort Edward, New York. In 1984, the U.S. Environmental Protection Agency designated a nearly 322 km reach as the Hudson River PCBs Superfund Site. Here we describe a Fish Health Assessment study, part of a Natural Resource Damage Assessment, that evaluated the prevalence of toxicopathic lesions in adult brown bullhead ( Ameiurus nebulosus ), smallmouth bass ( Micropterus dolomieu ), and yellow perch ( Perca flavescens ). In fall 2001, 29–51 fish of each species were collected in fall 2001 from highly contaminated areas below the plants (Thompson Island Pool (TIP) and Stillwater Dam Pool (STW)), an upriver reference area (Feeder Dam Pool (FDP)), and a reference lake, Oneida Lake (ODA). The focus was on histopathologic lesions and observations associated with contaminant exposure: liver—neoplasms, foci of cellular alteration, bile duct hyperplasia; testes—ovotestis (testicular oocytes), germ cell degeneration, altered developmental stage; ovaries—atresia and altered developmental stage. Lesions associated with PCB exposure were defined as those with significantly greater prevalence and/or severity in TIP and STW compared with ODA and FDP. For brown bullhead and smallmouth bass, no lesions or changes in gonadal development met those criteria. In yellow perch, ovarian atresia was the only lesion associated with PCB exposure. Prevalence was 53% in FDP, 75% in ODA, and 100% in both STW and TIP; severity increased from mostly minimal to mild-moderate. Because of the high prevalence of atresia in reference collections, it is likely that factors other than PCBs are also involved. As part of a post–dredging monitoring plan, we recommend assessing gonad structure and function in yellow perch collected at the time of spawning in locations with a range of PCB contamination. This review examines the spatiotemporal integration of these maternal and embryonic signals that are required for successful reproduction. In coordination with hypothalamic-pituitary-gonadal (HPG) hormones, an intraovarian HPG-like axis regulates folliculogenesis, follicular quiescence, ovulation, follicular atresia, and corpus luteal functions. Upon conception and passage of the zygote through the fallopian tube, the contribution of maternal hormones in the form of paracrine secretions from the endosalpinx to embryonic development declines, with autocrine and paracrine signaling becoming increasingly important as instructional signals for the differentiation of the early zygote/morula into a blastocyst. These maternal and embryonic signals include activin and gonadotropin-releasing hormone 1 (GnRH1) that are crucial for the synthesis and secretion of the ‘pregnancy’ hormone human chorionic gonadotropin (hCG). hCG in turn signals pre-implantation embryonic cell division and sex steroid production required for stem cell differentiation, and subsequent blastulation, gastrulation, cavitation and blastocyst formation. Upon reaching the uterus, blastocyst hatching occurs under the influence of decreased activin signaling, while the attachment and invasion of the trophoblast into the endometrium appears to be driven by a decrease in activin signaling, and by increased GnRH1 and hCG signaling that allows for tissue remodeling and the controlled invasion of the blastocyst into the uterine endometrium. Objective This study aims to present 18 years’ experience with endoscopic treatment of bilateral congenital choanal atresia as regard to management concept, surgical technique, results, pitfalls, and complications. Patients and methods A retrospective study including 112 cases of bilateral congenital choanal atresia, treated at Mansoura University Hospital endoscopically in the period from January 1998 to March 2015. As far as we know, this is the largest study group on transnasal endoscopic choanal atresia repair in literature. Results One hundred and twelve infants (87 females, 25 males) were included in the study. Age at operation ranged between 1 day and 28 days (8.75 days in average), and body weight average was 2.76 kg. All patients were diagnosed at birth except 3 infants. In this study, 85 cases were mixed atresia, 25 cases were bony atresia, and only two cases were membranous. In all cases, obliterated choana bone and vomer bone was removed, lateral wall drilling was used in 33 cases. Follow up ranged between 6 months and 18 years (95.6 months in average). The most common complication was restenosis, occurred in 42% (47 cases). Second-look procedure was done in 68 cases. The need for second-look evaluation with stent group was 74.5% (62 out of 83 infants), whereas in non-stent group was 20.6% (6 out of 29 infants). Conclusion Endoscopic repair of bilateral choanal atresia is a safe, effective technique with minimal complication. Usage of 30 degree sinuscope permits better visualization and higher accessibility for the surgical instruments. Surgically formed wide single neochoana with removal of all intervening tissue surroundings, and good follow up permit higher success rate without stenting. Advanced learning curve permits tailoring the perfect surgery with minimal tissue injury and better outcome. The development of large spontaneous portosystemic shunts (PSS) is a common finding in liver cirrhosis. The diversion of the portal flow through PSS directly into the caval system causes progressive liver atrophy and atretic changes of the portal vein. During both living and deceased donor liver transplantation (LT), persistence of large PSS has been associated to portal flow steal phenomena causing decreased patients and graft survival. Atretic changes of the portal vein and large PSS often coexist potentially representing a technical challenge during portal vein reconstruction. Potential annual fecundity (PAF) was estimated over three years (2010–2012) for yellowtail flounder with individuals from the three stocks off the northeast U.S. coast. Down-regulation of PAF, the resorption of oocytes during development, was evident as the vitellogenic cohort advanced, so we directly measured atresia of vitellogenic oocytes using stereological techniques. PAF models including relative fish condition, stock area, year, and oocyte diameter of the leading cohort explained more variation than models with just size alone based on Akaike information criteria. In a given year, Gulf of Maine females had lower PAF at size than southern New England females. Interannual differences were evident: PAF of both stocks was higher in 2010 and lower in 2012, with 2011 showing less synchronization between these stocks. Differences in size at age and relative condition suggested that energy available for somatic and reproductive growth was lower in some years in the Gulf of Maine and Georges Bank, especially in 2011. Georges Bank PAF and condition were intermediate to the other stocks or more similar to the Gulf of Maine, varying annually. A latitudinal gradient in PAF is evident based on our results and relative to earlier studies that included Canadian stocks. The magnitude of down-regulation was variable across stocks and typically 3–25% of PAF. This can be accounted for in fecundity estimates, by the seasonal schedule of sampling and use of an oocyte diameter term in the fecundity model. Theoretical models of atresia patterns suggested variable rates over the later portion of clutch development. The timing of down-regulation varied among years, and its intensity was influenced by female relative condition. Fecundity was related to fish size, but was also affected by fish condition and oocyte diameter (a proxy for time until spawning), and spatial and temporal effects. The jumbo squid Dosidicus gigas has been considered to be semelparous by several authors. However, to date no studies to support this assumption have been conducted. The present work tests the hypothesis that female jumbo squid do not die after a single reproductive event and provides evidence of the presence of postovulatory follicles (Pof) in females of different sizes. They indicates previous spawning in females in which they are present. Histological analysis was performed on reproductive structures of 73 female jumbo squid from the northwest of Mexico using hematoxylin-eosin staining. Using image analysis five ovarian stages were identified: (I) previtellogenesis, (II) vitellogenesis, (III) postvitellogenesis, (IV) spawning (in which 3.73% show atresia and the presence of Pof) and (V) Postspawning, in which a higher proportion (4.86%) show atresia and Pof. It is already known that the jumbo squid has asynchronic ovarian development with partial spawning during the reproductive period. However, the consistent presence of postovulatory follicles and the presence of oocytes of different sizes and development support the hypothesis that the jumbo squid is a multiple spawner with more than one reproductive event during its life cycle. Maximal profit in both the commercial egg and meat industries requires that the quantity of oviposited eggs closely matches the quantity of large yellow follicles maturing in the ovary. While laying hens are genetically selected for maximal egg production and strategies for management of broiler breeders have been constructed to achieve a similar outcome, a percentage of ovarian follicles that are selected into the ovulatory hierarchy in these hens still never make it to oviposition possibly due to atresia of large yellow follicles or internal ovulation of the oocyte into the peritoneal cavity rather than the oviduct. The causes and mechanisms responsible for these processes remain unclear, however, evidence in wild birds suggests that stressful and/or territorial challenges may stimulate oocyte losses. Since testosterone and corticosterone are central to the responses to territorial intrusions and stress, respectively, and since both large yellow follicles and the oviduct that will engulf them are sensitive to hormonal cues, one or both hormones may play a role in the loss of large yellow follicles via atresia and/or internal ovulation in laying hens. To test this, broiler breeder hens were treated with corticosterone or testosterone 5 h prior to ovulation and observed to see whether these treatments influenced the likelihood that a hen would lay an egg 24 h after the predicted ovulation time. A subset of hens that did not lay an egg were killed and dissected to look for evidence of follicle atresia and internal ovulation. Testosterone treatment resulted in significantly more oocyte losses, and 60% of these occurred due to internal ovulations, as was indicated by the presence of yolk in the peritoneal cavity. Corticosterone did not influence the rate of oocyte losses, follicle atresia, or internal ovulation. With the increased use of different synchronization programs in cattle, attention is given to the progesterone concentration during development of the ovulatory follicle. It has been shown that low peripheral progesterone concentrations during follicular development may lead to decreased fertility. To investigate the effect of low progesterone concentrations on the fate of the dominant follicle, a study was conducted where cycles of dairy cows and heifers were manipulated to induce the development of the first dominant follicle without progesterone (PLACEBO) or under sub-luteal progesterone concentrations from a progesterone releasing intravaginal device (PRID Delta ® ). After insertion of the devices, daily follow up was performed by transrectal ultrasonography to identify and measure follicular development and blood samples were taken to determine the circulating progesterone concentration. Follow up was continued until the ovulation of a follicle occurred. After ovulation, the fate of the first dominant follicle was identified as arrested, atretic or ovulatory. Arrest was defined as persistence of the dominant follicle followed by ovulation whereas atresia was defined as regression of the dominant follicle and subsequent growth and ovulation of a new follicle. During PLACEBO treatment, heifers ovulated earlier and smaller follicles in comparison to cows. During PRID Delta ® treatment, heifers had greater progesterone concentrations compared to cows and arrest of the dominant follicle occurred more in cows in comparison to heifers. The follicle destiny towards ovulation or atresia is multi-factorial in nature and involves outcries, paracrine and endocrine factors that promote cell proliferation and survival (development) or unchain apoptosis as part of the atresia process. In several types of cells, sphingosine-1-phospate (S1P) promotes cellular proliferation and survival, whereas ceramide (CER) triggers cell death, and the S1P/CER ratio may determine the fate of the cell. The aim of present study was to quantify S1P and CER concentrations and their ratio in bovine antral follicles of 8 to 17 mm classified as healthy and atretic antral follicles. Follicles were dissected from cow ovaries collected from a local abattoir. The theca cell layer, the granulosa cells and follicular fluid were separated, and 17β-estradiol (E2) and progesterone (P4) concentrations were measured in the follicular fluid by radioimmunoassay. Based on the E2/P4 ratio, the follicles were classified as healthy (2.2±0.3) or atretic (0.2±0.3). In both follicular compartments (granulosa and theca cell layer), sphingolipids were extracted and S1P and CER concentrations were quantified by HPLC (XTerra RP18; 5 µm, 3.0×150 mm column). Results showed that in both follicular compartments, S1P concentrations were higher in healthy antral follicles than in atretic antral follicles (P<0.05). The concentration of CER in the granulosa cells was higher in atretic antral follicles than in healthy antral follicles, but no differences were observed in the theca cell layer. The S1P/CER ratio in both follicular compartments was also higher in healthy antral follicles. Interestingly, in these follicles, there was a 45-fold greater concentration of S1P than CER in the granulosa cells (P<0.05), whereas in the theca cell layer, S1P had only a 14-fold greater concentration than CER when compared with atretic antral follicles. These results suggest that S1P plays a role in follicle health, increasing cellular proliferation and survival. Choanal Atresia is a congenital condition that presents as a blockage from the nasal cavity to the nasopharynx. According to the German statistical Institute (Statistisches Bundesamt), the incidence in Germany in 2018 reached 2.74 in 10,000 live births. It can present unilaterally or bilaterally. As newborns are obligate nasal breathers, management of bilateral atresia has to be performed early after birth. As for unilateral atresia, the optimal age for treatment was determined to be between 6 and 12 months in a recently published consensus. The main purpose of this study is to characterize the patients treated for choanal atresia in the Department of Otorhinolaryngology and Head and Neck Surgery at a tertiary hospital in the south of Germany and, based in the demographic characteristics as well as intra- and post-operative treatment, to identify factors for success or failure of the surgery. A secondary goal was to describe the complications in the use of stents and analyse its influence in the results. The cohort-based observational study included 29 patients, with a minimum follow up of one year, who underwent surgical endoscopic correction of both unilateral and bilateral choanal atresia from 2003 to 2020. Analysis of the demographics, intraoperative, and postoperative treatment, and their results, was performed. Multinomial logistic regression was applied for categorical values. Comparisons were performed using Fischer/chi-square test where applicable. A significance level of 0.05 was reached. The population was comprised of 34.5% male and 65.5% female patients. The age varied from 2 days to 20 years old, with a mean of 4.98 years and Standard Deviation (SD) 6.88. The weight of the patients varied between 1.4 kg and 85.0 kg, with a mean of 19.36 and SD 22.58. Unilateral choanal atresia was present in 58.6%, and bilateral in 41.4%. Out of this population, 48.3% presented with associated malformations. The number of procedures per patient performed in general anesthesia varied from 1 to 9, with a mean of 2.52 and SD of 2.23: statistically higher for patients presenting with bilateral atresia with a p value of 0.001*. 20 patients had a tube (Vygon, France) placed at first surgery. In 5 patients, a drug-eluting stent (Propel mini (R); Intersect ENT, USA) was applied intraoperatively, without complications. In unilateral choanal atresia, surgical and functional success was attained in 88.2% of the cases. In bilateral cases, it reached 75 %. We observed a strong linear relationship between the weight of the patient and the size of the choana perioperatively: R quadrat 0.596, which may be a determinant factor in the wound healing. Weight, age, and concomitant pathology achieved statistical significance in the logistic regression model (p respectively 0.001*, 0.001* and 0.010*), which shows its influence in the result of the surgery. In particular, weight inferior to 3 kg (p 0.001*, chi-square test) at the time of the first surgery is a determinant demographic factor for need of revision in order to achieve patency. There were no major complications associated with the use of a stent, besides its dislodgement. We could not infer an implication of its use on the success of the surgery in the present cohort. for this study were the small size of the cohort and non-standardized records for some variables. Younger patients presenting with low weight and concomitant pathology have worse results after surgical endoscopic treatment of choanal atresia, revealing these characteristics to be determinant for success. The size of the neochoana is related to the weight of the patient, which is a limiting factor. In particular, patients under 3 kg had to undergo more procedures in order to achieve patency. Two populations of flounder ( Platichtys flesus ) with different life history traits inhabit the brackish water Baltic Sea. Both types share feeding areas in coastal waters during summer-autumn but utilise different habitats for spawning in spring, namely offshore spawning with pelagic eggs and coastal spawning with demersal eggs respectively. Fecundity regulation by atresia was assessed as prevalence (portion of fish with atresia) and intensity (calculated as the average intensity of atresia in these fish) during the reproductive cycle following start of gonad development in the autumn up to spawning in spring, and evaluated in relation to fish condition (Fulton's condition factor reflecting energy reserves of the fish) and feeding incidence of the respective population. Peaking in winter (December–February), fecundity regulation was significantly higher for coastal spawning flounder than for flounder spawning offshore. For coastal spawners, the prevalence was 45–90% with an intensity of 6.4–9.3% vs. 0–25% and an intensity of 2.1–3.4% for offshore spawners during winter. Further, fecundity regulation ceased prior to spawning for offshore spawners but continued for coastal spawners. For coastal spawners, the prevalence was 12–29% and an intensity of 2.5–6.1% during spawning. The change in fish condition was strongly related to feeding incidence and differed between populations. As feeding ceased, condition of offshore spawners decreased during winter up to spawning, whereas condition of coastal spawners decreased during autumn but was maintained as feeding started again prior to spawning. Thus, habitat utilisation according to spawning strategy affects the timing of fecundity down-regulation reflecting availability of resources, namely limited food resources in deep areas and higher availability in coastal areas. Offshore spawning flounder display characteristics typical for a capital spawner with ceasing of feeding and oocyte down-regulation well before spawning, whereas coastal spawning flounder can be characterised as intermediate between a capital and income spawner with feeding prior to and during spawning along with continuous fecundity-regulation. Abstract: Herring (Clupea harengus) is a capital breeder that stores energy reserves in muscle tissue. Individual potential fecundity relies on the size and weight of female fish. Poor condition during the maturation process can lead to a heavy down-regulation of fecundity through atresia and, in the extreme, cause skipped spawning. Herring in the Central Baltic Sea exist in a variable environment where food availability fluctuates substantially. Compared to other herring populations their condition is generally poor. In the present study, the oocyte dynamics and fecundity in relation to the condition of Central Baltic herring was investigated. A modern stereological method, the physical fractionator, was used to quantify the number of oocytes in previtellogenic (PG), cortical alveoli (CA) as well as successive vitellogenic (VT1 and VT2) stages in central Baltic herring during ovarian maturation. The potential fecundity, i.e. the number of VT2 oocytes, was low compared to other Atlantic stocks but the relative potential fecundity was higher. The latter decreased by 71% when comparing early-maturing individuals with CA oocytes and late-maturing individuals with VT2 oocytes, suggesting a substantial down-regulation of fecundity. Although determined as spring spawners by otolith hatch type, 15% of the randomly sampled females were characterized by oocytes in CA stage in the prespawning period, indicating skipped spawning. The condition of these females was poor, which might have resulted in skipped spawning. Ovary weight was a good predictor of potential fecundity within maturing stages of females. Combined with estimates of skipped spawning, this ovary weight could be used to estimate egg production thereby improving Central Baltic herring stock-recruitment models. [Copyright &y& Elsevier]

Abstract: Hexavalent chromium, CrVI, is a heavy metal endocrine disruptor, known as a mutagen, teratogen, and a group A carcinogen. Environmental contamination with CrVI, including drinking water, has been increasing in more than 30 cities in the United States. CrVI is rapidly converted to CrIII intracellularly, and CrIII can cause DNA strand breaks and cancer or apoptosis through different mechanisms. Our previous study demonstrated that lactational exposure to chromium results in a delay or arrest in follicle development and a decrease in steroid hormone levels in F1 female rats, both of which are mitigated (partial inhibition) by vitamin C. The current study tested the hypothesis that lactational exposure to CrIII accelerates follicle atresia in F1 offspring by increasing reactive oxygen species (ROS) and decreasing cellular antioxidants. Results showed that lactational exposure to CrIII dose-dependently increased follicular atresia and decreased steroidogenesis in postnatal day 25, 45, and 65 rats. Vitamin C mitigated or inhibited the effects of CrIII at all doses. CrIII increased hydrogen peroxide and lipid hydroperoxide in plasma and ovary; decreased the antioxidant enzymes (AOXs) GPx1, GR, SOD, and catalase; and increased glutathione S-transferase in plasma and ovary. To understand the effects of CrVI on ROS and AOXs in granulosa (GC) and theca (TC) cell compartments in the ovary, ROS levels and mRNA expression of cytosolic and mitochondrial AOXs, such as SOD1, SOD2, catalase, GLRX1, GSTM1, GSTM2, GSTA4, GR, TXN1, TXN2, TXNRD2, and PRDX3, were studied in GCs and TCs and in a spontaneously immortalized granulosa cell line (SIGC). Overall, CrVI downregulated each of the AOXs; and vitamin C mitigated the effects of CrVI on these enzymes in GCs and SIGCs, but failed to mitigate CrVI effects on GSTM1, GSTM2, TXN1, and TXN2 in TCs. Thus, these data for the first time reveal that lactational exposure to CrIII accelerated follicular atresia and decreased steroidogenesis in F1 female offspring by altering the ratio of ROS and AOXs in the ovary. Vitamin C is able to protect the ovary from CrIII-induced oxidative stress and follicle atresia through protective effects on GCs rather than TCs. [Copyright &y& Elsevier]

Abstract: The objective of the present study was to determine the effects of bone morphogenetic protein (BMP)-15 and FSH on the growth, viability, and expression of mRNA for FSH (FSH-R) and BMP-15 (BMPR-IB and BMPR-II) receptors in cultured bovine secondary follicles. Secondary follicles were microdissected and cultured for 12 days in minimum essential medium-α alone or supplemented with BMP-15, sequential FSH, both BMP-15 and FSH, or BMP-15 from days 0 to 6, and FSH from days 7 to 12. Thereafter, the effect of these treatments on the follicular volume, viability, and antrum formation and the levels of mRNA for BMPR-IB, BMPR-II, and FSH-R were assessed. Compared with day 0, the follicles cultured with FSH or BMP-15, or both, had a significant and progressive increase in volume (P < 0.05). However, the follicles cultured for 12 days with both BMP-15 and FSH had the greatest volume and a greater rate of antrum formation than those in control medium, but results similar to those cultured with FSH (days 0 to 12) or BMP-15 (days 0 to 6) and FSH (days 7 to 12). Together with their accelerating effect on in vitro follicle growth, the combination of FSH and BMP-15 induced ultrastructural changes in the cultured follicles and increased atresia. However, adding either BMP-15 or FSH to the culture medium, not only promoted follicular growth and follicular antrum formation, but also maintained follicular viability during culture. Except for follicles cultured in minimal essential medium-α, the levels of mRNA for BMPR-IB were reduced, and the levels of mRNA for FSH-R were significantly greater in follicles cultured in medium supplemented with BMP-15. In conclusion, all in vitro follicle treatments supported growth of bovine preantral follicles; however, adding both BMP-15 and FSH to the culture medium (minimal essential medium-α) for 12 days provided the greatest stimulation. Furthermore, the viability and ultrastructural integrity of cultured follicles were only maintained when only BMP-15 or FSH was added to the culture medium. [Copyright &y& Elsevier]

Abstract: Black scabbardfish is a deep water species of high commercial interest in the NE Atlantic; however the life history and stock structure is poorly understood. For this study, specimens were collected from commercial trawls off west of the British Isles and longliners from the Madeira Islands, between September 2008 and May 2010. Geographical differences in the reproductive state of scabbardfish were evident, supporting the theory of a north-south migration. Only immature specimens were found in the northern area, while in Madeira all maturity stages were observed, with the peak spawning in October–November. Consistently, the gonadosomatic index (GSI) showed an increase throughout the year, reaching a maximum in October and November for males and females, respectively; while for the northern area the GSI values had low variability. Oocyte development was described and characterized for each maturity stage. Histology revealed that the oocytes have a group-synchronous development and it was concluded that black scabbardfish was a total spawner and had determinate fecundity. The geographical quasi-complete separation of the immature and mature individuals necessitated the use of a novel bias-reduction GLM in the estimation of length at maturity (LC50%). Estimated LC50% for both sexes was significantly higher when data from both areas were combined (females=1156mm, males=1098mm) than using the Madeira dataset (females=1110mm, males=1010mm). The results highlight large scale dispersal in this species which needs to be treated as a highly migratory species and be managed as a single population. [Copyright &y& Elsevier]
