This study aimed to develop and validate a pain assessment scale for guinea pigs to be performed in clinics by veterinarians. The scale was established using pre-existing pain scales from other species and was based on signs expressed by guinea pigs in pain already identified in the literature. The scale consists of 14 descriptors grouped into 4 categories: appearance, functional, physiological, and behavioral. Each item on the scale was assigned a score based on the observation and handling of the animals. The total score obtained ranged from 0 to 26, with more intense pain scoring higher. Fifty-eight adult guinea pigs presenting at the authors' Teaching Hospital between March 2021 and November 2022 were assessed. Guinea pigs were filmed during their consultation and an information sheet was completed. The information sheet provided details on the animals' appetite, fecal production, and behavior at home. The videos and corresponding information sheets were then reviewed to attribute a pain score to each animal. The pain scores were measured twice by a same observer 1 month apart to assess repeatability and by a second observer to assess reproducibility. The validity was assessed by correlating the pain scores given by the novel scale with scores using a numerical rating scale (NRS) as a gold standard method. The initial Cronbach's alpha obtained was 0.77, which was satisfactory. However, it highlighted that items concerning pruritus and spasmodic movements should be deleted to strengthen internal consistency. Then, Lin's coefficient and Bland and Altman's graphical method highlighted that the scale had an almost strong repeatability and intra-operator reproducibility. Moreover, the correlation between pain intensities obtained with the novel scale and the scores given using NRS was correct (Kappa = 0.72). The specificity was 87.5%, but the sensitivity of the scale was 52.0%. Each item's repeatability and reproducibility were assessed to identify items that would benefit from added clarification. Our work is the first step in establishing a pain scale in guinea pigs. However, it should not yet be used in a clinical context. Indeed, the scale was not good at detecting pain when a guinea pig was actually painful. Further studies, including improvement of certain items and testing a larger sample of animals, are ongoing. A 2-year-3-month-old female guinea pig (Cavia porcellus) was referred for lethargy, weight gain and severe abdominal pain. Ultrasound examination revealed thickening and enlargement of the left uterine horn and an unremarkable right uterine horn. The animal died during ovariohysterectomy due to respiratory and cardiac arrest. Necropsy revealed unilateral segmental uterine horn hypoplasia and aplasia of the fallopian tube of the corresponding side. This is the first report of a congenital uterine malformation in the guinea pig and we describe the clinical and pathological presentation of segmental uterine horn hypoplasia and fallopian tube aplasia. Congenital uterine malformations are rare in guinea pigs but should be considered as differential diagnoses for the more common acquired uteropathies. [ABSTRACT FROM AUTHOR]

In vitro maturation (IVM) system is an alternative method to superovulation protocols to obtain mature oocytes. Epidermal Growth Factor (EGF) and Insulin-like Growth Factor I (IGF-I) have been widely used in IVM medium in different species. Although the guinea pig is a valuable animal model for reproductive studies, IVM is rarely used. We aimed to establish a suitable in vitro production system using EGF and/or IGF-I during IVM to improve oocyte competence. Firstly, immunolocalization of EGF and IGF-I receptors in the ovary was assessed. An IVM dose-response experiment was performed with cumulus-oocyte complexes (COCs) supplemented with: 1) EGF [0, 10, 50, 100 ng/mL or 10% fetal calf serum (FCS)]; 2) IGF-I [0, 50, 100, 200 ng/mL or 10% FCS]; or 3) the concentrations of EGF and IGF-I which showed the best IVM index in the previous experiments, with or without Fetal Calf Serum (FCS). Cortical granule and mitochondria distribution patterns were determined in in vivo and in vitro -matured oocytes for the first time in this species. Apoptotic rate after IVM and oocyte competence by in vitro embryo development were evaluated. Immunohistochemistry results showed positive immunostaining of EGF and IGF receptors in corpus luteum, oocytes, granulosa and theca cells in follicles in all stages of development. Supplementation of IVM medium with 50 ng/mL EGF or 100 ng/mL IGF-I or their combination with FCS successfully led to oocyte nuclear and cytoplasmic maturation and reduced the apoptotic rate. Both growth factors improved oocyte competence during IVM in this species since early embryos were in vitro developed, showing better results when FCS was used in the IVM medium. • EGF and IGF-I receptors were identified in all guinea pig follicle stages and corpus luteum. • Cortical granule and mitochondria migration patterns were first studied in guinea pig oocytes. • EGF, IGF-I, or their combination improve in vitro nuclear and cytoplasmic oocyte maturation. • EGF and IGF-I enhance oocyte competence during IVM, enabling in vitro embryo development. This manuscript describes the mechanism by which supplementation with virgin coconut oil prevents the reduction of the relaxant reactivity of the trachea of guinea pigs with chronic allergic lung inflammation. [Display omitted] • Virgin coconut oil (VCO) improves impaired relaxation by asthma in rats. • VCO promotes formation of relaxing prostanoids or leukotrienes decrease. • VCO prevents negative modulation of β receptors/BKCa pathway by TGF-β. • VCO is a candidate for alternative therapy against allergic asthma. Asthma is a chronic inflammatory disease with many patients failing to respond appropriately to pharmacotherapy. Thus, virgin coconut oil (VCO) can be an alternative therapy. Guinea pigs were divided into control (Ctrl), chronic allergic lung inflammation (Asth), Asth treated with dexamethasone (Asth + dexa), and Asth supplemented with VCO (Asth + VCO4). The tested animals were sensitized and challenged with ovalbumin. Relaxation induced by arachidonic acid (AA) was impaired in Asth, and this reduction was prevented by VCO (p = 0.0004), indicating formation of relaxing prostanoids, or the decrease of cysteinyl-leukotrienes by VCO. Furthermore, a possible negative modulation of β receptors/BK Ca pathway by TGF–β in Asth was prevented by VCO (p = 0.0073). Finally, in Asth, Masson's trichrome staining revealed airway remodeling and immunohistochemistry showed increased TGF-β expression in the lungs of Asth (p < 0.0001)), which was prevented by VCO and dexamethasone (p < 0.0001), concluding VCO as a possible agent against asthma.
