We proposed a non-contact photoacoustic (PA) detection method using spectral domain optical coherence tomography (SDOCT). Two interference spectrums (A-lines) were acquired before and after the PA excitation with SDOCT. PA signal propagated within the sample causing the vibration. The vibration inner the sample introduced phase change between the acquired two A-lines. Thus, the PA signal can be detected by evaluating the difference in phase between the two A-lines. Based on the method, an OCT-PAM dual-mode imaging system was constructed. In the system, SDOCT served as the detection unit for PAM. Thus, the combination of the two imaging modalities was simplified. Another advantage of the system is that it realizes non-contact all-optic detection, which is attractive for biomedical imaging. Using the system, we imaged phantoms of carbon fibers, asparagus leaves and human hairs. Furthermore, the cortical vasculature of rat was imaged in vivo and the flow status was evaluated quantitatively.

In the study of cavitation in room-temperature fluids, the heat transfer between gas and liquid in bubble oscillation is usually assumed to be an adiabatic process for simplification. However, this heat transfer and thermodynamic mechanism is not yet understood in cryogenic fluids, especially under small amplitude oscillation conditions excited by ultrasonic field. This article studies bubble thermodynamic model under an external ultrasonic field based on the heat transfer equation for cryogenic fluids. The temperature changes inside bubbles are calculated, and the heat transfer mechanism is briefly analyzed. The results indicate that the heat transfer mechanism of bubbles depends on the relationship between ultrasonic frequency and bubble resonance frequency. By analyzing two special cases of dual-bubble and high-pressure environment, it is believed that heat transfer can be approximated as an adiabatic process under high-pressure conditions with ultrasonic frequency far from the resonance frequency. This conclusion can provide a theoretical basis for subsequent accurate calculation of heat-transfer polytropic coefficient, or void faction measurement in cryogenic two-phase flow.

Purpose/Objectives: We sought to create nomograms to predict individual risk of early mortality, which can identify patients who require interventions to prevent early death. Methods: We included patients in the National Cancer Database with non-metastatic squamous cell carcinoma of the head and neck who received radiation and systemic therapy between 2004 and 2017 in the definitive or adjuvant setting. Early mortality was defined as any death less than 90 days after starting radiation. Multivariable logistic regression was used to assess the relationship between covariates and early mortality. Nomograms to predict the risk of early death were created for both the definitive and adjuvant settings. Results: Among 84,563 patients in the definitive group and 18,514 patients in the adjuvant group, rates of early mortality were 3.5 % (95 % CI 3.4–3.7 %) and 2.2 %, (95 % CI 1.9–2.4 %), respectively. Patients above the age of 70 had an early mortality rate of 7.8 % (95 % CI 7.3–8.2 %) in the definitive group and 4.4 % (95 % CI 3.6–5.4 %) in the adjuvant group. We study the Zcs(3985) and Zcs(4000) exotic states in the decays of Λb baryons through a molecular scenario. In the final state interaction, the Λb→Λc+Ds(⁎)− decays are followed by the Λc+Ds(⁎)− to Zcs−p rescatterings via exchange of a D(⁎) meson. We predict a branching fraction of (3.1−2.6+1.4)×10−4 for Λb→Zcs−p, which can be measured in the Λb→J/ψK(⁎)−p decay. This study proposes a new approach to test the molecular model, and guides future experimental searches for the Zcs(3985) and Zcs(4000).

Although biodegradable plastic film is a promising alternative product for reducing polyethylene plastic pollution in agricultural soils, the effects of its residues on plant growth and soil properties remain unclear. In this study, we conducted an experiment to investigate root properties and soil enzyme activities in Poly (butylene adipate-co-terephthalate) microplastics (PBAT-MPs) contaminated soil (0 % (CK), 0.1 %, 0.2 %, 0.5 % and 1 % of dry soil weight) with soybean (Glycine max (Linn.) Merr.) and maize (Zea mays L.). The results show that PBAT-MP accumulation in soil negatively affects root growth, and alter soil enzyme activities, which may then constrain C/N cycling and potential yields. For soybean, the total root length, total root surface area and root biomass decreased by 34 %− 58 %, 34 %− 54 % and 25 %− 40 % at the harvesting stage compared to CK, respectively. The negative effects of PBAT-MPs on maize roots were greater than on soybean roots. An analysis of the J/ψ→ηη′γ decay by the BESIII collaboration claims the observation of an exotic state η1(1855) with IGJPC=0+1−+. To establish its C-parity partner η1′(1855) in the picture of the KK¯1(1400) molecular state, we propose that J/ψ→η1′(1855)η(′) receives the main contributions from the final state interaction of KK⁎(K+K⁎−, K−K⁎+, K0K¯⁎0, and K¯0K⁎0). Specifically, K and K⁎ in J/ψ→KK⁎ decays transform as η1′(1855)η(′), by exchanging a K1(1400). We predict B(J/ψ→η1′(1855)η)=(6.3−3.5+12.6)×10−6, and B(J/ψ→η1′(1855)η′)=(6.5−4.6+6.6)×10−6, which can be studied in the J/ψ→K⁎K¯⁎η(′) decays.

Purpose: Proton therapy is an emerging therapy for several malignancies owing to its favorable therapeutic ratio. There are very limited data on the use of proton therapy in the management of thyroid carcinoma. Our objective was to review the safety, feasibility, and outcomes of proton therapy for patients with thyroid cancer treated to the head and neck. Methods: From our institution's proton database from 2012 to 2021, we identified 22 patients with thyroid cancer treated with proton beam therapy. We evaluated outcomes and toxicities. Results: Median follow-up was 26 months. Of the 22 patients, 50% were female. The mean age was 65 years. Three patients had anaplastic cancer; 13, papillary carcinoma; 2, follicular carcinoma; and 2, poorly differentiated carcinoma. Forty-six percent had T4 disease. Primary targets were the central neck compartment, level VI, and upper mediastinum. Radiation dose was 60 GyRBE adjuvantly, and 70 GyRBE for gross disease (range, 6000-7600 GyRBE). Eight patients underwent upfront adjuvant radiation, and 3 received definitive radiation for unresectable disease upfront. Eleven patients received either salvage or palliative radiation. Fifty-nine percent of patients had extrathyroidal extension, and 64% of patients had gross disease in the neck before treatment. Fifty percent of patients had metastatic disease before treatment. Sixteen patients received concurrent chemotherapy, 63% of these patients received doxorubicin. For all patients, 1-year local regional recurrence (LRR) was 0%, and overall survival (OS) was 90%. Acute grade 3+ toxicities occurred in 27% of patients, the most frequent being dermatitis (27%). Three patients required a percutaneous endoscopic gastrostomy tube after radiation therapy (RT), 2 owing to progression. There were no grade 4+ toxicities. Conclusions: Proton therapy for thyroid cancer appears feasible and effective with minimal toxicities. Prospective studies comparing proton therapy with intensity-modulated RT, to evaluate the clinical efficacy of using proton therapy to reduce toxicities in patients undergoing radiation for thyroid cancer, are warranted.

Transcription factors are protein molecules that act as regulators of gene expression. Aberrant protein activity of transcription factors can have a significant impact on tumor progression and metastasis in tumor patients. In this study, 868 immune-related transcription factors were identified from the transcription factor activity profile of 1823 ovarian cancer patients. The prognosis-related transcription factors were identified through univariate Cox analysis and random survival tree analysis, and two distinct clustering subtypes were subsequently derived based on these transcription factors. We assessed the clinical significance and genomics landscape of the two clustering subtypes and found statistically significant differences in prognosis, response to immunotherapy, and chemotherapy among ovarian cancer patients with different subtypes. Multi-scale Embedded Gene Co-expression Network Analysis was used to identify differential gene modules between the two clustering subtypes, which allowed us to conduct further analysis of biological pathways that exhibited significant differences between them. Finally, a ceRNA network was constructed to analyze lncRNA-miRNA-mRNA regulatory pairs with differential expression levels between two clustering subtypes. We expected that our study may provide some useful references for stratifying and treating patients with ovarian cancer.

Summary: A high point of Tibetan Plateau (TP) civilization, the expansive Tubo Empire (618–842 AD) wielded great influence across ancient western China. However, whether the Tubo expansion was cultural or demic remains unclear due to sparse ancient DNA sampling. Here, we reported ten ancient genomes at 0.017- to 0.867-fold coverages from the Dulan site with typical Tubo archaeological culture dating to 1308–1130 BP. Nine individuals from three different grave types have close relationship with previously reported ancient highlanders from the southwestern Himalayas and modern core-Tibetan populations. A Dulan-related Tubo ancestry contributed overwhelmingly (95%–100%) to the formation of modern Tibetans. A genetic outlier with dominant Eurasian steppe-related ancestry suggesting a potential population movement into the Tubo-controlled regions from Central Asia. Together with archeological evidence from burial styles and customs, our study suggested the impact of the Tubo empire on the northeast edge of the TP involved both cultural and demic diffusion.

Summary: Gold nanoclusters (AuNCs) have become a promising material for bioimaging detection because of their tunable photoluminescence, large Stokes shift, low photobleaching, and good biocompatibility. Last decade, great efforts have been made to develop AuNCs for enhanced imaging contrast and multimodal imaging. Herein, an updated overview of recent advances in AuNCs was present for visible fluorescence (FL) imaging, near-infrared fluorescence (NIR-FL) imaging, two-photon near-infrared fluorescence (TP-NIR-FL) imaging, computed tomography (CT) imaging, positron emission tomography (PET) imaging, magnetic resonance imaging (MRI), and photoacoustic (PA) imaging. The justification of AuNCs applied in bioimaging mentioned above applications was discussed, the performance location of different AuNCs were summarized and highlighted in an unified parameter coordinate system of corresponding bioimaging, and the current challenges, research frontiers, and prospects of AuNCs in bioimaging were discussed. This review will bring new insights into the future development of AuNCs in bio-diagnostic imaging.

Summary: We present here a detailed protocol for PANINI (protein and nucleic acid in situ imaging), a technique that enables the concurrent staining of protein and nucleic acids in archival tissue sections. PANINI utilizes an optimized antigen retrieval strategy that forgoes protease treatment while retaining high sensitivity of nucleic acid detection down to single genomic events. While the protocol here is geared toward standard fluorescent microscopes with 3–4 available channels, PANINI is compatible with many commercial multiplexed tissue imaging modalities.For complete details on the use and execution of this protocol, please refer to Jiang et al. This study explored the application of Kluyveromyces marxianus and Saccharomyces cerevisiae (commercial and wild type) in the alcoholic fermentation of Fuji apple juice under static conditions. Metabolome analyses revealed that ethyl esters, including ethyl hexanoate, ethyl decanoate, ethyl octanoate, octanoic acid and decanoic acid, were the dominant components in ciders fermented by the Saccharomyces yeasts. In the K. marxianus ciders, ethyl acetate, hexyl acetate, propyl acetate and acetic acid were the most abundant volatiles, suggesting that the cider fermented by K. marxianus might have a fruitier smell. Transcriptome analyses were adapted to gain insight into the differential metabolite patterns between K. marxianus and S. cerevisiae during cider fermentation. GO and KEGG enrichments revealed that the metabolic pathways of glucose, organic acids and amino acids during cider fermentation were quite different between these two yeasts. The K. marxianus strain exhibited a higher rate of glycolysis and ethanol fermentation than did Saccharomyces yeasts under oxygen-limited conditions. It also reduced the metabolic flux of acetate into acetyl-CoA and then into the TCA cycle, increasing the syntheses of ethyl acetate and relevant esters, which may affect its cell growth under anaerobic conditions but enriched the taste and variety of aromas in apple cider.

Purpose: Radiation treatment plans undergo peer review during chart rounds, but changes to treatment volumes would require replanning. Our group implemented weekly head and neck cancer “volume rounds” to peer review all target volumes for head and neck cancer before radiation therapy (RT) planning and chart rounds. Methods and Materials: We analyzed modifications made to planning target volumes (PTVs) at volume rounds for consecutive nonproton head and neck cancer cases from May 2020 to May 2021. Nine head and neck radiation oncologists participated in weekly volume rounds during this time. Recommendations were categorized as no changes, minor changes, major changes, additional workup (eg, biopsy or imaging), and consultation or tumor board discussion needed before the start of RT. Minor changes to PTVs generally did not require a second review before treatment planning while major changes did. Results: PTVs for 511 cases involving 432 patients underwent peer review and 298 (58.3%) of these cases did not require any modifications before treatment planning. Minor and major changes were recommended in 75 (14.7%) and 86 (16.8%) cases, respectively. Forty-five (8.8%) cases were recommended to have additional workup and 23 (4.5%) required additional consultation with nonradiation surgeons or medical oncologists. Of the 45 cases that were recommended for additional workup, 40 underwent biopsy or imaging. Positive findings on imaging or biopsy occurred in 13 patients, leading to a significant change in management, including 4 patients who underwent additional surgery after positive findings before the start of RT. Conclusions: Prospective peer review during head and neck cancer volume rounds led to frequent minor and major alterations to PTVs. Significant changes in the overall treatment plan, such as additional surgery before start of RT, occurred in a minority of patients.

Summary: Pancreatic cancer is a deadly disease that accounts for approximately 5% of cancer deaths worldwide, with a dismal 5-year survival rate of 10%. Known genetic risk factors explain only a modest proportion of the heritable risk of pancreatic cancer. We conducted a whole-exome case-control sequencing study in 1,591 pancreatic cancer cases and 2,134 cancer-free controls of European ancestry. In our gene-based analysis, ATM ranked first, with a genome-wide significant p value of 1 × 10−8. The odds ratio for protein-truncating variants in ATM was 24, which is substantially higher than prior estimates, although ours includes a broad 95% confidence interval (4.0–1000). SIK3 was the second highest ranking gene (p = 3.84 × 10−6, false discovery rate or FDR = 0.032). We observed nominally significant association signals in several genes of a priori interest, including BRCA2 (p = 4.3 × 10−4), STK11 (p = 0.003), PALB2 (p = 0.019), and TP53 (p = 0.037), and reported risk estimates for known pathogenic variants and variants of uncertain significance (VUS) in these genes. The rare variants in established susceptibility genes explain approximately 24% of log familial relative risk, which is comparable to the contribution from established common susceptibility variants (17%). In conclusion, this study provides new insights into the genetic susceptibility of pancreatic cancer, refining rare variant risk estimates in known pancreatic cancer susceptibility genes and identifying SIK3 as a novel candidate susceptibility gene. This study highlights the prominent importance of ATM truncating variants and the underappreciated role of VUS in pancreatic cancer etiology.

Purpose: To demonstrate temporal lobe necrosis (TLN) rate and clinical/dose-volume factors associated with TLN in radiation-naïve patients with head and neck cancer treated with proton therapy where the field of radiation involved the skull base. Materials and Methods: Medical records and dosimetric data for radiation-naïve patients with head and neck cancer receiving proton therapy to the skull base were retrospectively reviewed. Patients with < 3 months of follow-up, receiving < 45GyRBE or nonconventional fractionation, and/or no follow-up magnetic resonance imaging (MRI) were excluded. TLN was determined using MRI and graded using Common Terminology Criteria for Adverse Events (CTCAE) v5.0. Clinical (gender, age, comorbidities, concurrentchemotherapy, smoking, radiation techniques) and dose-volume parameters were analyzed for TLN correlation. The receiver operating characteristic curve and area under the curve (AUC) were performed to determine the cutoff points of significant dose-volume parameters. Results: Between 2013 and 2019, 234 patients were included. The median follow-up time was 22.5 months (range =3.2–69.3). Overall TLN rates of any grade, ≥ grade 2, and ≥ grade 3 were 5.6% (N =13), 2.1%, and 0.9%, respectively. The estimated 2-year TLN rate was 4.6%, and the 2-year rate of any brain necrosis was 6.8%. The median time to TLN was 20.9 months from proton completion. Absolute volume receiving 40, 50, 60, and 70 GyRBE (absolute volume [aV]); mean and maximum dose received by the temporallobe; and dose to the 0.5, 1, and 2 cm3 volume receiving the maximum dose (D0.5cm3, D1cm3, and D2cm3, respectively) of the temporal lobe were associated with greater TLN risk while clinical parameters showed no correlation. Among volume parameters, aV50 gave maximum AUC (0.921), and D2cm3 gave the highest AUC (0.935) among dose parameters. The 11-cm3 cutoff value for aV50 and 62 GyRBE for D2cm3 showed maximum specificity and sensitivity. Conclusion: The estimated 2-year TLN rate was 4.6% with a low rate of toxicities ≥grade 3; aV50 ≤11 cm3, D2cm3 ≤62 GyRBE and other cutoff values are suggested as constraints in proton therapy planning to minimize the risk of any grade TLN. Patients whose temporal lobe(s) unavoidably receive higher doses than these thresholds should be carefully followed with MRI after proton therapy.

The anti-triplet charmed baryon decays with the light scalar mesons are rarely measured, whereas the recent observation of the Cabibbo-favored Λc+→Ληπ+ decay hints a possible Λc+→Λa0(980)+,a0(980)+→ηπ+ process. We hence study the Λc+→Λa0(980)+ decay. Particularly, it is found that the final state interaction can give a significant contribution, where Σ+(1385) and η in Λc+→Σ+(1385)η by exchanging a charged pion are transformed as Λ and a0(980)+, respectively. Accordingly, we predict B(Λc+→Λa0(980)+)=(1.7−1.0+2.8±0.3)×10−3, accessible to the BESIII, BELLEII and LHCb experiments.

Purpose: The incidence of brain metastases is increasing as a result of more routine diagnostic imaging and improved extracranial systemic treatment strategies. As noted in recent consensus guidelines, postoperative stereotactic radiosurgery (SRS) to the resection cavity has lower rates of local control than whole brain radiation therapy but improved cognitive outcomes. Further analyses are needed to improve local control and minimize toxicity. Methods and materials: Patients receiving SRS to a resection cavity between 2006 and 2016 were retrospectively analyzed. Presurgical variables, including tumor location, diameter, dural/meningeal contact, and histology, were collected, as were SRS treatment parameters. Patients had routine follow-up with magnetic resonance imaging, and those noted to have local failure were further assessed for the recurrence location, distance from the target volume, and dosimetric characteristics. Results: Overall, 82 patients and 85 resection cavities underwent postoperative SRS during the study period. Of these, 58 patients with 60 resection cavities with available follow-up magnetic resonance imaging scans were included in this analysis. With a median follow-up of 19.8 months, local recurrence occurred in 12 of the resection cavities for a 15% 1-year and 18% 2-year local recurrence rate. Pretreatment tumor volume contacted the dura/meninges in 100% of cavities with recurrence versus 67% of controlled cavities (P = .025). A total of 5 infield, 5 marginal, and 4 out-of-field recurrences were found, with a median distance to the centroid from the target volume of 3 mm. The addition of a 10-mm dural margin increased the target volume overlap with the recurrence contours for 10 of the 14 recurrences. Conclusions: Dural contact was associated with an increased rate of recurrence for patients who received SRS to a surgical cavity, and the median distance of marginal recurrences from the target volume was 3 mm. These results provide evidence in support of recent consensus guidelines suggesting that additional dural margin on SRS volumes may benefit local control.

Genome shuffling is a process to combine advantage traits by the recombination of the entire genome and it has been successfully applied in the laboratory evolution of various industrial microorganisms. However, genome shuffling has not been described in Kluyveromyces marxianus (KM), a promising yeast host for the expression of heterologous proteins. In this protocol, genome shuffling in KM is performed by sexual reproduction and is combined with high-throughput screening to obtain high-yielding strains. Notably, the screening of diploid clones risen from one mating mixture is carried out to improve the effectiveness of evolution. Mating-sporulation-mating cycles are repeated to obtain KM strain with ideal traits. • The method combines genome shuffling with high-throughput to achieve strains displaying high yielding of heterologous proteins. • This method can be applied to the genome shuffling of other species when only a few starting strains are available for sexual reproduction.

As the second part of the investigation on vibration characteristics of ± 800 kV converter transformers, this part focuses on load conditions. A full-load thermal stability test is carried out on 12 YY and YD converter transformers in a converter station. Vibration, voltage, current, and other important information are collected by online monitoring devices. Vibration amplitude, vibration spectrum, dominant frequency, vibration power ratio of frequency band, vibration power entropy, and odd–even harmonic ratio under different load conditions are calculated and studied to evaluate the vibration state. Theoretical analyses of winding electromagnetic force and vibration acceleration are carried out and results show that the harmonic currents make the dominant frequency shift to 300 Hz or 400 Hz. Most vibration characteristics are sensitive to load rate changes. The odd–even harmonic ratio is sensitive to filter disconnection and reconnection. The results of this study reveal the vibration regularity and corresponding mechanism and can contribute to further study on vibration assessment of ± 800 kV converter transformers under load conditions.

For lack of study on the vibration characteristics of converter transformers, this paper studied the vibration signal analysis method and investigated the vibration characteristics of converter transformers during the commissioning of a ±800 kV converter station. The study comprised two parts including no-load and load conditions. In Part I, converter transformers are under no-load conditions. Vibration data are collected under conditions of no-load closing, no-load voltage rise with converters, and no-load voltage rise with DC lines. The vibration characteristics and mechanisms are studied based on the collected data. Several different vibration characteristics are introduced. Vibration power entropy, vibration total harmonic distortion, and recurrence rate are calculated under no-load conditions. Results show that vibration amplitude, vibration total harmonic distortion, and recurrence rate are sensitive to no-load closing. Vibration power entropy is sensitive to no-load voltage rise with DC lines. All these vibration characteristics are sensitive to converter unlocking. These characteristics show different and significant changes under different no-load conditions. Thus, it is vital to choose appropriate vibration characteristics when assessing the vibration state of converter transformers under different operating conditions. The study examines the vibration characteristics’ physical meanings and their changing mechanisms. It also summarizes distribution intervals and typical values of vibration characteristics. This research reveals the vibration regularity and corresponding mechanism. It provides a reference for vibration state assessment and early fault detection of ±800 kV converter transformers under no-load conditions.

Rice blast is regarded as one of the major diseases of rice. Screening rice genotypes with high resistance to rice blast is a key strategy for ensuring global food security. Unmanned aerial vehicles (UAV)-based imaging, coupled with deep learning, can acquire high-throughput imagery related to rice blast infection. In this study, we developed a segmented detection model (called RiceblastSegMask) for rice blast detection and resistance evaluation. The feasibility of different backbones and target detection models was further investigated. RiceblastSegMask is a two-stage instance segmentation model, comprising an image-denoising backbone network, a feature pyramid, a trinomial tree fine-grained feature extraction combination network, and an image pixel codec module. The results showed that the model combining the image-denoising and fine-grained feature extraction based on the Swin Transformer and the feature pixel matching feature labels with the trinomial tree recursive algorithm performed the best. The overall accuracy for instance segmentation of RiceblastSegMask reached 97.56%, and it demonstrated a satisfactory accuracy of 90.29% for grading unique resistance to rice blast. These results indicated that low-altitude remote sensing using UAV, in conjunction with the proposed RiceblastSegMask model, can efficiently calculate the extent of rice blast infection, offering a new phenotypic tool for evaluating rice blast resistance on a field scale in rice breeding programs.

Magnetically induced self-assembly technology was used to construct a label-free electrochemical biosensor based on magnetic Mg0.5Cu0.5Fe2O4-Au nanocomposites for the sensitive detection of cancer antigen 125 (CA125). To aim for this, Mg0.5Cu0.5Fe2O4 nanoparticles were first prepared via the rapid combustion process. The average diameter of the nanoparticles calcined at 800 °C with the absolute ethanol volume of 20 mL was about 169.3 nm. Then Au was used for the surface modification by NaBH4 reduction reaction approach. The magnetic glassy carbon electrode (MGCE) was modified by Mg0.5Cu0.5Fe2O4-Au via a magnetic induction self-assembly process. The reduced thiol-modified single-stranded DNA was attached to the Mg0.5Cu0.5Fe2O4-Au nanocomposites by Au-S bonds without any coupling agent. CA125 antigen was grabbed directly by its aptamer DNA due to its specific identification with the aptamer. Finally, the modified electrodes were blocked with BSA and then characterized. Finally, DPV analysis was used for CA125 detection, the novel fabricated biosensor demonstrated good detection properties for CA125 with a linear range of 5–125 U/mL and a detection limit of 4.4 U/mL. The results showed that the aptamer sensor had good specificity, repeatability, and stability. The feasibility of our sensor for the determination of CA125 was also demonstrated by measuring CA125 levels in serum using the Roche gold-standard instrument of the People’s Hospital of Danyang as the reference value. The recoveries of real serum samples were 94.65–101.71%, and RSDs were 1.26–4.65%. Moreover, the surface of the electrode could be cleaned and reused by magnetic separation, greatly reducing the cost and providing the possibility for a point-of-care test (POCT). This work demonstrated a new strategy for integrating both nanostructures and biocompatibility to build advanced cancer biomarker sensors with wide applications.

The performance of high-temperature polybenzimidazole (PBI) proton exchange membrane fuel cells (HT-PEMFCs) doped with phosphoric acid is strongly influenced by the configuration of its flow channel. In this research, genetic algorithm (GA) is adopted to optimize original three-dimensional simulation model by obtaining the optimal design of the channel configuration. Set the widths of top and bottom edges of the anode/cathode flow channels as independent variables with constrained range to optimize HT-PEMFC performance. The ratio of the pressure drop loss to the output power of the HT- PEMFC is set as the objective function. The results show that the widths of the top and bottom edges are 0.513 mm 0.635 mm at the anode side, and 0.752 mm and 1.159 mm at the cathode side, respectively. The shape of the cross-sectional of the flow channel is trapezoidal, which can achieve the best performance. In the anode side, the narrow contact surface between the flow channel and the GDL can increase the diffusion rate of the hydrogen. For cathode, the larger contact surface formed by the flow channel and the GDL can provide more gas to carry out the electrochemical reaction. The pressure drop loss and output power of the optimal model are 1.7% and 6.5% greater than those of the original model at 0.4 V, respectively. The results can contribute to improve the design and operation of HT-PEMFCs in the future.

Efficient catalysts are required for both oxidative and reductive reactions of hydrogen and oxygen in sustainable energy conversion devices. However, current precious metal-based electrocatalysts do not perform well across the full range of reactions and reported multifunctional catalysts are all complex hybrids. Here, we show that single-phase porous Co3Mo3N prepared via a facile method is an efficient and reliable electrocatalyst for three essential energy conversion reactions; oxygen evolution reaction (OER), oxygen reduction reaction (ORR), and hydrogen evolution reaction (HER) in alkaline solutions. Co3Mo3N presents outstanding OER, ORR, and HER activity with high durability, comparable with the commercial catalysts RuO2 for OER and Pt/C for ORR and HER. In practical demonstrations, Co3Mo3N gives high specific capacity (850 mA h gZn−1 at 10 mA cm−2) as the cathode in a zinc-air battery, and a low potential (1.63 V at 10 mA cm−2) used in a water-splitting electrolyzer. Availability of Co and Mo d-states appear to result in high ORR and HER performance, while the OER properties result from a cobalt oxide-rich activation surface layer. Our findings will inspire further development of bimetallic nitrides as cost-effective and versatile multifunctional catalysts that will enable scalable usage of electrochemical energy devices.

Study region: The middle and lower reaches of the Jinsha River Basin (MLJRB). Study focus: The main objective is to evaluate the impact of climate change and anthropogenic activities to the water and sediment resources of the studied region in 1960—2015. The variation of runoff and suspended sediment load is first analyzed with regard to their trends and abrupt change. Different factors attributable to the variation are then quantitatively separated. New hydrological insights for the region: The annual runoff and sediment load across the MLJRB both decreased abruptly and coincidently in the recent two decades, with the transitional years detected to be 2006 and 2002, respectively. Comparing the post-transition period to the pre-transition period, runoff decreased 14.7% and sediment load decreased 63%. Climate change is the major contribution for the decrease in runoff, which explains 56.3% of the reduction, and impoundment of large reservoirs and water consumption contributed 18.6% and 2.7% respectively. The other 22.5% was ascribed to water-soil conservation and other factors. In contrast to water resources, human activities were dominant in sediment reduction, including 44.9% caused by large reservoirs, 1% from increased water consumption and 35% by conservation measures and other potential factors. Remarkably, almost 90% of the sediment load can be trapped as the direct consequence of the construction and operation of two huge dams, Xiluodu and Xiangjiaba, since 2013. Background: Cytomegalovirus (CMV) can cause infection and critical diseases in hematopoietic stem cell transplantation (HSCT) recipients. This study aimed to explore the cumulative incidence and risk factors for CMV infection and disease among HSCT recipients in Taiwan. Methods: This retrospective cohort study using the Taiwan Blood and Marrow Transplantation Registry (TBMTR) included HSCT recipients between 2009 and 2018 in Taiwan. The primary outcome was cumulative incidence of CMV infection or disease at day 100 after HSCT. Secondary outcomes included day 180 cumulative incidence of CMV infection or disease, infection sites, risk factors for CMV infection or disease, survival analysis, and overall survival after CMV infection and disease. Results: There were 4394 HSCT recipients included in the study (2044 auto-HSCT and 2350 allo-HSCT). 【Abstract】Objective: To investigate the surgical treatment for patients with multiple injuries in ICU. Methods: Clinical data of 163 multiple injury patients admitted to ICU of our hospital from January 2006 to January 2009 were retrospectively studied, including 118 males and 45 females, with the mean age of 36.2 years (range, 5-67 years). The injury regions included head and neck (29 cases), face (32 cases), chest (89 cases), abdomen (77 cases), pelvis and limbs (91 cases) and body surface (83 cases). There were 57 cases combined with shock. ISS values varied from 10 to 54, 18.42 on average. Patients received surgical treatments in ICU within respectively 24 hours (10 cases), 24-48 hours (8 cases), 3-7 days (7 cases) and 8-14 days (23 cases). Results: For the 163 patients, the duration of ICU stay ranged from 2 to 29 days, with the average value of 7.56 days. Among them, 143 were cured (87.73%), 11 died in the hospital (6.75%) due to severe hemorrhagic shock (6 cases), craniocerebral injury (3 cases) and multiple organ failure (2 cases), and 9 died after voluntarily discharging from hospital (5.52%). The total mortality rate was 12.27%. Conclusions: The damage control principle should be followed when multiple injury patients are resuscitated in ICU. Surgical treatment strategies include actively controlling hemorrhage, treating the previously missed injuries and related wounds or surgical complications and performing planned staging operations. Constipation is one of themost common chronic gastrointestinal problems. The estimated incidence of constipation in the United States is3% to 19% in general population.1,2 Patientswith head injuries, spinal cord injuries, pelvic fractures, lower extremity fractures ormultiple traumas require a long-term bed rest, during which the incidence of constipation reached as high as 50%.3,4 Constipation always brings inconvenience and tremendous suffering to patientsand strongly influences the recovery from primary disease. Irritants or lubricants can relieve the symptoms, but long-term application of them may lead to side effects like melanosis coli5 and cathartic colon6. The absorption of fat soluble vitamins is also affected.7 Polyethylene glycol 4000 (trade name: Forlax®), a long chain polymer with a high molecular weight, can conjugate withwater molecule through hydrogen bond to increase the water content and volume of stools, thereby, facilitate bowelmovement and defecation.8,9 It is neither absorbed nor metabolized in the digestive tract, hence it is highly safe and well tolerable. Thus, long-term medication of polyethylene glycol 4000 is conducive to the reconstruction of normal defecation pattern. Therefore, polyethylene glycol 4000 is now being widely used as the mainstay adult chronic functional constipation management.10,11 The aim of this study was to verify the efficacy and safety of polyethylene glycol 4000 on adult functional constipation of posttraumatic bedridden patients.

Discontinuous perfobond rib shear (PBL) connectors can be applied to fabricated steel-concrete composite beams. In this paper, six push-out specimens were designed to explore the shear performance of PBL under static and repeated loads. Four specimens were used for static push-out test. The remaining two were used for fatigue push-out test. The static load test mainly considers the influence of concrete at steel plate end and post-cast strip on the shear bearing capacity, initial shear stiffness and load-slip curve of discontinuous PBL shear connectors. On the basis of the test results, the calculating formulas of the shear capacity and shear stiffness of discontinuous PBL connectors considering the influence of end bearing, bond group effect and post-cast strip are proposed. The predicted results of the formula are close to the experiment. From the fatigue test results, it is found that the fatigue performance of above specimens with C50 and UHPC is better than that with studs, but the fatigue resistance of discontinuous PBL connectors with UHPC is not increased in proportion to the shear capacity. In addition, the residual slip and shear stiffness of PBL specimens after cyclic loading are related to fatigue load amplitude.

Melanoma is rare in Taiwan. Asian melanoma is distinct from Western melanoma because acral and mucosal melanoma accounts for the majority of melanoma cases, leading to distinct tumor behaviors and genetic profiling. With consideration of the clinical guidelines in Western countries, Taiwanese experts developed a local clinical practice consensus guideline. This consensus includes diagnosis, staging, and surgical and systemic treatment, based only on clinical evidence, local epidemiology, and available resources evaluated by experts in Taiwan. This consensus emphasizes the importance of surgical management, particularly for sentinel lymph node biopsies. In addition, molecular testing for BRAF is mandatory for patients before systemic treatment. Furthermore, immunotherapy and targeted therapy are prioritized for systemic treatment. This consensus aimed to assist clinicians in Taiwan in diagnosing and treating patients according to available evidence.

【Abstract】 Penetrating injury to the rectum, verte-bral body and spinal cord by a steel rod is a rare condition. Treatment of this kind of injury is very challenging. Rectal injury requires repair and fecal diversion, while debride-ment of the spine is difficult, especially when the injury site is very long. Here we report a case of penetrating injury of rectum and sacral vertebra by a steel rod after falling onto the ground from 1 m height. The abscess cav-ity was irrigated with 3% hydrogen peroxide and physio-logical saline repeatedly. The bony canal was carefully debrided, curetted and bony fragments were removed. Spinal irrigation and drainage lasted for 2 months and sen-sitive antibiotic (amikacin sulfate) was given 7 days after surgery, but abscess was still formed in the vertebral canal. At 6-month follow-up, the patient was paralyzed without any neurological improvement, and the pain in low back and lower limb still continued. The assessment of total antioxidant capacity (TAC) is crucial for evaluating overall antioxidant potential, predicting the risk of chronic diseases, guiding dietary and nutritional interventions, and studying the effectiveness of antioxidants. However, achieving rapid TAC assessment with high sensitivity and stability remains a challenge. In this study, Ce/Fe-MOF with abundant oxygen vacancies was synthesized using microplasma for TAC determination. The microplasma synthesis method was rapid (30 min) and cost-effective. The presence of oxygen vacancies and the collaboration between iron and cerium in Ce/Fe-MOF not only enhanced the catalyst's efficiency but also conferred multiple enzyme-like properties: peroxidase-like, oxidase-like, and superoxide dismutase mimetic activities. Consequently, a simple colorimetric assay was established for TAC determination in vegetables and fruits, featuring a short analysis time of 15 min, a good linear range of 5–60 μM, a low detection limit of 1.3 μM and a good recovery of 91 %-107 %. This method holds promise for rapid TAC assessment in agricultural products.

Background: Postoperative delirium (POD) is a common complication following cardiac surgery and increases postoperative morbidity and mortality. Intraoperative electroencephalogram (EEG) burst suppression suggests excessively deep anesthesia and predicts POD. Use of remimazolam provides a stable hemodynamic status and an appropriate depth of anesthesia. We aim to assess remimazolam administered for anesthesia and sedation in elderly patients having cardiac surgery. Methods: This is a randomized controlled clinical trial with noninferiority design. A total of 260 elderly patients aged equal to or greater than 60 years undergoing cardiac surgery will be randomly allocated to receive remimazolam or propofol (1:1) for general anesthesia and postoperative sedation until extubation. The primary outcome is the cumulative time with EEG burst suppression which is obtained from the SedLine system. The noninferiority margin is 2.0 min. The secondary outcomes include the POD occurrence within the first 5 days postoperatively and the duration of perioperative hypotension. Discussion: This noninferiority trial is the first to evaluate the effect of perioperative remimazolam administration on EEG burst suppression, POD occurrence, and duration of hypotension in elderly patients who undergo cardiac surgery. Background: Semantic segmentation is crucial in medical image diagnosis. Traditional deep convolutional neural networks excel in image classification and object detection but fall short in segmentation tasks. Enhancing the accuracy and efficiency of detecting high-level cervical lesions and invasive cancer poses a primary challenge in segmentation model development. Methods: Between 2018 and 2022, we retrospectively studied a total of 777 patients, comprising 339 patients with high-level cervical lesions and 313 patients with microinvasive or invasive cervical cancer. Overall, 1554 colposcopic images were put into the DeepLabv3+ model for learning. Accuracy, Precision, Specificity, and mIoU were employed to evaluate the performance of the model in the prediction of cervical high-level lesions and cancer. Results: Experiments showed that our segmentation model had better diagnosis efficiency than colposcopic experts and other artificial intelligence models, and reached Accuracy of 93.29 %, Precision of 87.2 %, Specificity of 90.1 %, and mIoU of 80.27 %, respectively. Conclution: The DeepLabv3+ model had good performance in the segmentation of cervical lesions in colposcopic post-acetic-acid images and can better assist colposcopists in improving the diagnosis.

Soluble sugar content in seeds is an important quality trait of soybean. In this study, 57 quantitative trait loci (QTLs) related to soluble sugar contents in soybean seeds were collected from databases and published papers. After meta-overview-collinearity integrated analysis to refine QTL intervals, eight consensus QTLs were identified. To further verify the consensus QTLs, a population of chromosome segment substitution lines (CSSLs) was analyzed. Two lines containing fragments covering the regions of consensus QTLs and the recurrent parent were selected: one line showed high soluble sugar contents associated with a consensus QTL fragment, and the other line showed low soluble sugar contents. Transcriptome sequencing was conducted for these two lines at the early, middle, and late stages of seed development, which identified 158, 109 and 329 differentially expressed genes, respectively. Based on the analyses of re-sequencing data of the CSSLs and the consensus QTL region, three candidate genes (Glyma.19G146800, Glyma.19G122500, and Glyma.19G128500) were identified in the genetic fragments introduced from wild soybean. Sequence comparisons between the two CSSL parents SN14 and ZYD00006 revealed a single nucleotide polymorphism (SNP) mutation in the coding sequence of Glyma.19G122500, causing a non-synonymous mutation in the amino acid sequence that affected the predicted protein structure. A Kompetitive allele-specific PCR (KASP) marker was developed based on this SNP and used to evaluate the CSSLs. These results lay the foundation for further research to identify genes related to soluble sugar contents in soybean seeds and for future soybean breeding.

Background/purpose: Culture environments play a critical role in stem cell expansion. This study aimed to evaluate the effects of 2,3,5,4′-tetrahydroxystilbene-2-O-b-D-glucoside (THSG) on the proliferation and differentiation of human dental pulp stem cells (DPSCs) in 2-dimensional (2D) and 3-dimensional (3D) culture systems. Materials and methods: Human DPSCs were seeded in T25 flasks for 2D cultivation. For the 3D culture system, DPSCs were mixed with microcarriers and cultured in spinner flasks. Cells in both culture systems were treated with THSG, and cell proliferation was determined using a cell counter and a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay. In THSG-treated DPSCs, the genes associated with proliferation, adipogenesis, neurogenesis, osteogenesis, pluripotency, oncogenesis, and apoptosis were analyzed using real-time polymerase chain reactions. Results: The spinner flask time-dependently improved cell numbers, cell viability, and expansion rates in THSG-treated DPSCs. In both the T25 and spinner flasks, the messenger RNA (mRNA) levels of proliferation, osteogenesis, and pluripotent-related genes had a significant maximum expression with 10 μM THSG treatment. However, 0.1 μM of THSG may be the most suitable condition for triggering neurogenesis and adipogenesis gene expression when DPSCs were cultured in spinner flasks. Furthermore, the number of oncogenes and apoptotic genes decreased considerably in the presence of THSG in both the T25 and spinner flasks. Conclusion: The spinner flask bioreactor combined with THSG may upregulate proliferation and lineage-specific differentiation in DPSCs. Thus, the combination can be used to mass-produce and cultivate human DPSCs for regenerative dentistry.

Background/purpose: Dental pulp stem cells (DPSCs) contribute to the regeneration of various tissues and have superior proliferation, immune privilege, and anti-inflammation properties to other mesenchymal stem cells. 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside (THSG) not only enhances the aforementioned properties of DPSCs but also promotes self-renewal and reprogramming-like ability. However, whether THSG enhances the aforementioned properties and abilities through direct or indirect interaction mechanisms remains unclear. To address this knowledge gap, we examined the effects of THSG-stimulated DPSC-derived conditioned medium (THSG-CM) on the activity and anti-inflammation properties of cells. Materials and methods: DPSCs were treated with various concentrations of THSG to produce THSG-CM, which was then collected, analyzed, and lyophilized. A cytokine profiling antibody assay was used to compare protein components between THSG-treated and nontreated CM. Human skin fibroblasts (HSFs) and human gingival fibroblasts (HGFs) were used to investigate the effect of THSG-CM on cell proliferation, anti-inflammation, and wound healing abilities; for this investigation, MTS assay, quantitative real-time PCR analysis, and 2-well silicone inserts wound model were conducted. Results: We observed that THSG enhanced the secretion of growth- and immune-associated proteins in THSG-CM and increased the proliferation of HSFs and HGFs. Furthermore, THSG-CM significantly attenuated lipopolysaccharide-stimulated mRNA levels of cytokines in both cells and improved wound healing abilities. Conclusion: We conclude that THSG-CM had more beneficial effects on cell activity and anti-inflammation in the HSFs and HGFs than DPSC-derived CM. DPSC-derived CM can be developed into a cell-free regenerative strategy in the future, and its therapeutic efficacy may be improved by THSG-CM.

Bacteria are key denitrifiers in the reduction of nitrate (NO3−-N), which is a contaminant in wastewater treatment plants (WWTPs). They can also produce carbon dioxide (CO2) and nitrous oxide (N2O). In this study, the autotrophic hydrogen-oxidizing bacterium Rhodoblastus sp. TH20 was isolated for sustainable treatment of NO3−-N in wastewater. Efficient removal of NO3−-N and recovery of biomass nitrogen were achieved. Up to 99% of NO3−-N was removed without accumulation of nitrite and N2O, consuming CO2 of 3.25 mol for each mole of NO3−-N removed. The overall removal rate of NO3−-N reached 1.1 mg L−1 h−1 with a biomass content of approximately 0.71 g L−1 within 72 h. TH20 participated in NO3−-N assimilation and aerobic denitrification. Results from 15N-labeled-nitrate test indicated that removed NO3−-N was assimilated into organic nitrogen, showing an assimilation efficiency of 58%. Seventeen amino acids were detected, accounting for 43% of the biomass. Nitrogen loss through aerobic denitrification was only approximately 42% of total nitrogen. This study suggests that TH20 can be applied in WWTP facilities for water purification and production of valuable biomass to mitigate CO2 and N2O emissions.

This work explores mechanical properties, structural evolution, and mechanism of creep-life enhancement for widely used P9 heat-resistant steel. The 17-year-on-site used P9 alloy exhibit a higher tensile strength and a smaller elongation than the new P9 alloy from room temperature to 700 °C. The P9 alloy also displays a typical ductile feature with a significantly necking profile. The P9 alloy shows phase transformation sequences of α-Fe (bcc) →Ac1∼858 °C α + γ -Fe (bcc + fcc) →Ac3∼894 °C γ-Fe (fcc) upon heating and γ-Fe (fcc) →Ms∼352 °C martensite (bct) →Mf∼300 °C martensite (bct) upon cooling. The new P9-alloy tube mainly contains ∼73.5% ferrite phase (α-Fe) and ∼26.5% carbide M3C. However, the used P9-alloy tube shows four crystalline phases including ∼45.9% ferrite, ∼14.5% martensite, ∼37.5% cementite (M3C) and ∼2.7% carbide M23C6. The creep test indicates that the used P9-alloy tube has a longer creep-life (or better anti-creep ability) than the new tube. Activation energies of atomic diffusion for the new and used tubes are respectively 252.45 and 345.87 kJ/mol, indicating a decreased diffusion capability in the used tube. This work suggests that martensite laths, lath boundaries, and precipitates (such as carbides) play important roles to inhibit creep-deformation in the P9-alloy steel.

Background/purpose: Dysregulation of cell cycle checkpoint control may lead to the independence of growth regulating signals. Checkpoint protein such as the PD-1/PD-L1 immune checkpoint involving tumor cells and host immune defense lymphocytes is a well-studied therapeutic target in oncology. Acting at a cell surface receptor on plasma membrane integrin αvβ3, thyroxine stimulates intracellular accumulation of PD-L1 in cancer cells. Although resveratrol also binds to integrin αvβ3, it reduces PD-L1 expression. Materials and methods: In current studies, we investigated the roles of resveratrol and thyroxine in regulating expression of proliferation-related genes and checkpoint genes, PD-L1, BTLA in two oral cancer cell lines. Results: Thyroxine suppressed the expression of pro-apoptotic BAD but induced proliferative CCND1 expression in SSC-25 cells and OEC-M1 cells. It activated expression of PD-L1 and BTLA in both cell lines. On the other hand, resveratrol suppressed the expression of all. Alternatively, it activated BAD expression. Thus thyroxine induces checkpoint gene expression which may promote proliferation in cancer cells. Alternatively, resveratrol reverses the stimulatory effects of thyroid hormone to induce anti-proliferation. Conclusion: These findings provide new insights into the antagonizing effect of resveratrol on the thyroxine-induced expression of checkpoint genes and proliferative genes in oral cancers.
