Constructing a restoration strategy from bauxite residue to Technosols is a cost-effective and sustainable strategy for addressing the ecological and environmental issues caused by high alkalinity, salinity, and fine-grained bauxite residues. However, the quantitative contribution of restoration strategies on the upper bauxite residue-derived Technosols to the underlying untreated bauxite residue in the short term remains poorly understood. This study investigated the mediating mechanisms of vegetation and microbial metabolic effects on the alkalinity, nutrient content, and structure of the underlying bauxite residue (20-50 cm) through a simulated ecological reconstruction of the bauxite residue stockpile. Results indicated that implementing plant restoration strategies resulted in the content of polyphenolic compounds, lipids, tannins, and carbohydrates in bauxite residue dissolved organic matter (DOM) increased significantly from 52.5, 8.2, 3.3, and 2.0 % to 54.4, 10.4, 5.6, and 2.8 %, respectively, while the content of condensed aromatics, unsaturated hydrocarbons, and proteins/amino sugars decreased significantly from 15.5, 12.0, and 6.5 % to 12.1, 9.7, and 5.1 %, respectively. The newly produced molecules were concentrated in regions with low O/C and high H/C ratios, suggesting that short-term vegetation restoration strategies facilitate the transformation of substrate DOM towards easily decomposable and highly bioavailable substances. This led to the migration of the newly produced molecules to the underlying bauxite residue, and as a result, the protein and soluble microbial products of the underlying bauxite residue increased significantly, as well as the pH, exchangeable Na, and < 0.054 mm particles decreased from 10.2, 44.2 cmol kg -1 , and 28.1 % to 9.7, 27.1 cmol kg -1 , and 19.4 %, respectively, available nitrogen, urease, and 1-2 mm particles increased from 7.3 mg kg -1 , 0.2 U mg -1 , and 14.5 % to 7.6 mg kg -1 , 0.3 U kg -1 , and 21.7 %, respectively. It is predicted that oxygen minimum zones (OMZs) in the ocean will expand as a consequence of global warming and environmental pollution. This will affect the overall microbial ecology and microbial nitrogen cycle. As one of the world's largest alluvial estuaries, the Yangtze Estuary has exhibited a seasonal OMZ since the 1980s. In this study, we have uncovered the microbial composition, the patterns of community assembly and the potential for microbial nitrogen cycling within the water column of the Yangtze Estuary, with a particular focus on OMZ. Based on the 16 S rRNA gene sequencing, a specific spatial variation in the composition of prokaryotic communities was observed for each water layer, with the Proteobacteria (46.1%), Bacteroidetes (20.3%), and Cyanobacteria (10.3%) dominant. Stochastic and deterministic processes together shaped the community assembly in the water column. Further, pH was the most important environmental factor influencing prokaryotic composition in the surface water, followed by silicate, PO 4 3- , and distance offshore (p < 0.05). Water depth, NH 4 + , and PO 4 3- were the main factors in the bottom water (p < 0.05). At last, species analysis and marker gene annotation revealed candidate nitrogen cycling performers, and a rich array of nitrogen cycling potential in the bottom water of the Yangtze Estuary. The determined physiochemical parameters and potential for nitrogen respiration suggested that organic nitrogen and NO 3 - (or NO 2 - ) are the preferred nitrogen sources for microorganisms in the Yangtze Estuary OMZ. Background and Purpose: Rheumatoid arthritis (RA) is called "immortal cancer", and it affects the quality of life, disability rate and even the survival of patients. This study aimed to observe the clinical efficacy, and adverse reactions of intradermal acupuncture (IA) in the treatment of RA patients with liver and kidney deficiency syndrome., Materials and Methods: 132 RA patients were split into an IA group and a sham IA group at a 1:1 ratio. Both groups were assessed before and after the intervention with the assessments: a traditional Chinese medicine (TCM) syndrome evaluation, the Health Assessment Questionnaire (HAQ), the Disease Activity Score 28 (DAS28) and serum C-reactive protein (CRP)., Results: There was a statistically significant difference in TCM syndrome evaluation, HAQ, DAS28, and CRP between both groups before and after treatment (P < 0.01). The improvement of TCM syndrome evaluation (95% CI [1.14(0.38-1.89)]; P = 0.001), HAQ (95% CI [2.00(1.00-3.00)]; P = 0.003), and DAS28 (95% CI [0.11(0.02-0.20)]; P = 0.021) in the IA group was more obvious than that in the sham IA group (P < 0.05), except for CRP (95% CI [0.50(- 2.09 to 7.08)], P = 0.786). The difference in CRP outcome changes between the two groups was not statistically significant (P > 0.05). Both groups had comparable results in the implementation of RA in the upper and lower extremity acupoints and did not differ due to different sites (IA group: P = 0.852; sham IA group: P = 0.861). The comparison of effective rate of the upper limb as well as that of the lower limb was statistically significant (P = 0.001). The long-term accumulation of coal waste on the surface during natural weathering leads to the inevitable migration of heavy metals contained in the coal waste, which increases the likelihood of environmental contamination and health risks. Dissolved organic matter (DOM) and Fe/Al oxides play crucial roles in the transformation and bioavailability of heavy metals. Thus, we analyzed the Fe/Al oxide content and DOM molecular composition in coal waste with different degrees of weathering and explored the influence of DOM chemical diversity and Fe/Al oxides on the potential mobility of heavy metals. Results showed that weathering-driven decrease in Fe oxides (Fe d , Fe O , and Fe p decreased from 82.4, 37.5, and 3.6 mg∙L -1 to 41.3, 24.7, and 2.3 mg∙L -1 , respectively) led to decreases in the reducible fractions of V and Cr. The potential environmental risks of more toxic metals of Cd and As, also increased as a result of the residual fractions decreased to 32.6 % and 41.3 %, respectively. Weathering caused an increase in oxygen-to‑carbon ratio, double-bond equivalent, modified aromaticity index, nominal oxidation state of carbon, and molecular diversity and a decrease in (m/z) w and (H/C) w , suggesting that the DOM of highly weathered coal waste possessed high unsaturation, aromatic structures, hydrophilicity, and strong oxidative characteristics. Additionally, although V MF and Cr MF showed significant negative correlations with O/C ratio, polyphenolic, carbohydrates, and condensed aromatics, pH remained a key environmental factor determining the potential environmental risks of V and Cr by changing the residual fractions. The mobilities of Cd and As were significantly negatively correlated with those of Fe/Al oxides, particularly Fe d , Fe O , Fe p , and Al p . Formaldehyde (FA) exposure has been reported to induce or aggravate allergic asthma. Infection is also a potential risk factor for the onset and aggravation of asthma. However, no study has addressed the effects of FA exposure on asthmatic patients with respiratory infection. FA is ubiquitous in environment and respiratory infections are common in clinics. Therefore, it is necessary to explore whether FA exposure leads to the further worsening of symptoms in asthma patients with existing respiratory infection. In the present study, ovalbumin (OVA) was used to establish the murine asthma model. Lipopolysaccharide (LPS) was intratracheal administrated to mimic asthma with respiratory infection. The mice were exposed to 0.5 mg/m 3 FA. FA exposure did not induce a significant aggravation on OVA induced allergic asthma. However, the lung function of specific airway resistance (sRaw), histological changes and cytokines production were greatly aggravated by FA exposure in OVA/LPS induced murine asthma model. Monocyte-derived macrophages (MDMs) were isolated from asthmatic patients. Exposure of MDMs to FA and LPS resulted in increased TNF-α, IL-6, IL-1β, and nitric oxide (NO) production. Lactate produciton and lactate dehydrogenase A (LDHA) expression were found to be upregulated by FA in OVA/LPS induced asthmatic mice and LPS stimulated MDMs. Furthermore, glycolysis inhibitor 2-Deoxy-d-glucose attenuated FA and LPS induced TNF-α, IL-6, IL-1β, and NO production. We conclude that FA exposure can lead to the aggravation of allergic asthma with infection through induction of glycolysis. Alkalinity regulation and nutrient accumulation are critical factors in the construction of plant and microbial communities and soil formation in bauxite residue, and are extremely important for sustainable vegetation restoration in bauxite residue disposal areas. However, the establishment and succession of microbial communities driven by plant colonization-mediated improvements in the physicochemical properties of bauxite residues remain poorly understood. Thus, in this study, we determined the saline-alkali properties and dissolved organic matter (DOM) components under plant growth conditions and explored the microbial community diversity and structure using Illumina high-throughput sequencing. The planting of Elymus dahuricus (E. dahuricus) in the bauxite residue resulted in a significant decrease in total alkalinity (TA), exchangeable Na, and electrical conductivity (EC) as well as the release of more tryptophan-like protein compounds and low-molecular-weight humic substances associated with biological activities into the bauxite residue substrate. Taxonomical analysis revealed an initial-stage bacterial and fungal community dominated by alkaline-tolerant Actinobacteriota, Firmicutes, and Ascomycota, and an increase in the relative abundances of the phyla Bacteroidota, Cyanobacteria, Chloroflexi, and Gemmatimonadota. The biological activities of phylum Actinobacteriota, Bacteroidota, and Gemmatimonadota were significantly associated with protein-like and UVA-like humic substances. As eutrophic bacteria, Proteobacteria participate in the transformation of humic substances and can not only utilize small molecules of organic matter and convert them into humic substances but also promote the gradual conversion of humic acids into simple molecular compounds. Our results suggest that plant roots secrete organic matter and microbial metabolites as the main biogenic organic matter that participates in the establishment and succession of the microbial community in bauxite residues. Identifying potential sources of soil potentially toxic elements (PTEs) and developing source-oriented health risk assessments in typical mining cities are key for pollution prevention and risk management. To this end, a case study was conducted to explore the pollution characteristics, potential sources, and human health risks of PTEs in Daye City, China. Indices, including the pollution factor (PF), pollution load index (PLI), and geo-accumulation index (I geo ), were applied to assess PTE pollution. Cd had the highest value among the detected PTEs, and 82.93% of the sampling sites had moderate pollution levels, with the highest mean I geo value for Cd (2.30). Four potential sources were determined. Cr and Ni originated mainly from natural sources. Zn (91.5%) was exclusively and then Cd (33.1%) was moderately derived from industrial activities. The mixed source of various mineral exploitation smelting, and coal-fired traffic emissions leaded to the accumulation of As, Cd, and Pb. Cu was associated with Cu-related mining and smelting activities. The probabilistic health risk assessment indicated that the non-carcinogenic risks for populations were negligible. Dissolved organic matter (DOM) in soil is a key factor affecting the bioavailability of heavy metals, but very few studies have focused on the role of DOM in the use of soil amendments to mitigate heavy metal accumulation in crops. Here, eleven materials were added to cadmium (Cd)-contaminated paddy soil in greenhouse pot trials; rice was grown and harvested, the chemodiversity of post-harvest soil DOM was characterized using Fourier-Transform Ion Cyclotron Resonance Mass Spectrometry, and the specific associations between soil DOM traits and water-extractable soil Cd concentration were identified at the molecular level. The results showed that the endogenous release caused by altering soil pH had a greater effect on soil DOM concentration than did the exogenous chemical input due to the application of organic amendments, which in turn contributed to the chemodiversity of DOM. After one season of rice cultivation, soil DOM molecules were mainly dominated by relatively low molecular weight heteroatom-free lignins. C/N, C/H ratios of organic materials influenced DOM molecular fingerprint patterns, and soil pH and redox potential were the main driving forces affecting the chemodiversity of DOM. Furthermore, the low molecular weight, high saturation, low aromaticity, and heteroatom-free DOM molecules are more likely to dissolve Cd from the soil solid phase, thus increasing the potential risk of Cd to the environment. Virus-like particle (VLP) vaccines have become one of the dominant vaccine candidates for foot-and-mouth disease (FMD). To further enhance the immunogenicity of VLP vaccines, gold nanocages (AuNCs) were selected as an adjuvant for the vaccine. Our experiments demonstrated that AuNCs had little biotoxicity in vivo and in vitro and improved the uptake of VLP in BHK-21 and RAW264.7 cell lines. The VLP-AuNCs activated DCs mainly through toll-like receptor 4 (TLR4) and promoted the secretion of IL-6, IL-1β, and TNF-α. The conjugation of VLP and AuNCs triggered a strong immune response against FMD virus (FMDV) in mice and guinea pigs. The VLP-AuNCs significantly enhanced the proliferation of CD8 + T cells (P < 0.05) and the secretion of cellular immune-related cytokines (IFN-γ, P < 0.05; IL-12p70, P < 0.01) compared with VLP. Purpose: Contact tracing has proven successful at controlling coronavirus 2019 (COVID-19) globally, and the Center for Health Security has recommended that the United States add 100,000 contact tracers to the current workforce., Methods: To address gaps in local contact tracing, health professional students partnered with their academic institution to conduct contact tracing for all COVID-19 cases diagnosed onsite, which included identifying and reaching their contacts, educating participants, and providing social resources to support effective quarantine and isolation., Results: From March 24 to May 28, 536 laboratory-confirmed COVID-19 cases were contacted and reported an average of 2.6 contacts. Contacts were informed of their exposure, asked to quarantine, and monitored for the onset of symptoms. Callers reached 94% of cases and 84% of contacts. Seventy-four percent of cases reported at least one contact. Household members had higher rates of reporting symptoms (odds ratio, 1.65; 95% confidence interval, 1.19-2.28). The average test turnaround time decreased from 21.8 days for the first patients of this program to 2.3 days on the eleventh week., Conclusions: This provides evidence for the untapped potential of community contact tracing to respond to regional needs, confront barriers to effective quarantine, and mitigate the spread of COVID-19., (Copyright © 2020 Elsevier Inc. Using diffusive gradients in thin films (DGT) is a recently developed alternative method of rapidly evaluating the bioavailability of metals in soil. However, the method has found only limited application in systematic assessment of the bioavailability of cadmium (Cd) in red limestone paddy soils treated with different soil amendments. Of the four methods compared for estimating Cd content of rice grains from plants grown in such soils of central China treated with eleven different soil amendments in pot culture, Cd content of DGT-labile soil was significantly correlated to Cd concentrations in brown rice (R = 0.447, p < 0.01). The other three methods involved CaCl 2 , diethylenetriaminepentaacetic acid (DTPA), or NH 4 NO 3 . Some other properties of soil, such as pH, redox potential, content of dissolved organic matter, and cation exchange capacity were also determined. Comprehensive analysis of fatty acids (FAs) has long been challenging due to their poor ionization efficiency, lack of characteristic fragment ions and difficulty of identifying C=C bond locations. In this study, a high coverage ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was established for the quantification and C=C bond location characterization of FAs using two structural analogues, 2-hydrazinyl-4,6-dimethylpyrimidine (DMP) and 2-hydrazinylpyrimidine (DP), as dual derivatization reagents. DP-labeled FA standards were used as internal standards to reduced matrix effects, which guaranteed the accurate quantification of FAs. The derivatization yields of FAs were larger than 99% and the sensitivities were increased by 400-fold compared with non-derivatized FAs. Pretreatment and instrumental analysis of FAs can be completed in 20 minutes. Only 5 μL rat plasma can satisfy the quantification of 36 FAs with good linearity (r>0.99). Both intra-day and inter-day accuracies were in the range of 85-105%, and the precisions were less than 15%. The extraction recoveries were investigated to be in the range of 88-112%. No obvious matrix effects were observed for the derivatized FAs. In addition, the locations of C=C bonds in DMP-derivatized FAs could be identified by diagnostic fragment ions generated from 1,4-hydrogen elimination and allylic cleavage under low energy collision induced dissociation (CID). The new method was finally employed for FA profiling in plasma from rats with moxifloxacin-induced liver injury. Decellularized human lung fibroblast-derived matrix (hFDM) has demonstrated its excellent proangiogenic capability. In this study, we propose a self-assembled, injectable multicellular microspheres containing human umbilical vein endothelial cells (HUVECs) and mesenchymal stem cell (MSCs), collagen hydrogel (Col), and hFDM toward therapeutic angiogenesis. Those multicellular microspheres are spontaneously formed by the mixtures of cell and hydrogel after being dropped on the parafilm for several hours. The size of microspheres can be manipulated via adjusting the initial volume of droplets and the culture period. The cells in the microspheres are highly viable. Multicellular microspheres show good capability of cell migration on 2D culture plate and also exhibit active cell sprouting in 3D environment (Col) forming capillary-like structures. We also find that multiple angiogenic-related factors are significantly upregulated with the multicellular microspheres prepared via Col and hFDM (Col/hFDM) than those prepared using Col alone or single cells (harvested from cocultured HUVECs/MSCs in monolayer). For therapeutic efficacy evaluation, three different groups of single cells, Col and Col/hFDM microspheres are injected to a hindlimb ischemic model, respectively, along with PBS injection as a control group. It is notable that Col/hFDM microspheres significantly improve the blood reperfusion and greatly attenuate the fibrosis level of the ischemic regions. In addition, Col/hFDM microspheres show higher cell engraftment level than that of the other groups. The incorporation of transplanted cells with host vasculature is detectable only with the treatment of Col/hFDM. Current results suggest that hFDM plays an important role in the multicellular microspheres for angiogenic cellular functions in vitro as well as in vivo. Coastal eutrophication and its associated harmful algal blooms have emerged as one of the most severe environmental problems worldwide. Seaweed cultivation has been widely encouraged to control eutrophication and algal blooms. Among them, cultivated kelp (Saccharina japonica) dominates primarily by production and area. However, the responses of water quality and phytoplankton community to kelp farming remain unclear. Here, thirteen cruises were conducted in the kelp farms and control areas in the turbid, highly eutrophic Xiangshan Bay of the East China Sea from 2008 to 2015. Results indicated that kelp cultivation slightly increased dissolved oxygen and pH, but reduced dissolved inorganic nitrogen and phosphorus. We estimated that kelp harvesting would remove 297 t of nitrogen and 42 t of phosphorus from this bay annually. Because of decreased flow velocity, turbulence, and sediment resuspension, kelp farming greatly reduced suspended solids and increased transparency, resulting in increases in phytoplankton chlorophyll a and abundance. Additionally, kelp farming appreciably increased phytoplankton species number, Marglef richness, and Shannon-Wiener diversity indices by 51.6%, 40.1%, and 13.1%, respectively. Analysis of similarity and similarity percentages demonstrated that phytoplankton community composition differed significantly between the farm and control area, which was mostly attributed to long-chained diatoms and single-celled dinoflagellates. However, after the kelp harvesting, all measurements of water quality and phytoplankton biomass, diversity, and community composition exhibited no significant difference. Aims: Obstructive sleep apnea (OSA) combined with type 2 diabetes (T2DM) may lead to cognitive dysfunction. We previously reported that cognitive impairment is exacerbated in KKAy mice exposed to intermittent hypoxia (IH), during which the DNA binding protein HMGB1 mediates hippocampal neuronal apoptosis by maintaining microglia-associated neuroinflammation, but the underlying mechanism remains largely unknown., Materials and Methods: We performed immunofluorescence, Western blotting, and immunohistochemistry experiments in mouse hippocampal tissues and HT22 cells. KKAy type 2 diabetes model mice and normal C57BL/6J mice were exposed to IH or intermittent normoxia. HT22 cells were cultured in high glucose medium and exposed to IH or intermittent normoxia. We transfected HMGB1 siRNA into HT22 cells and then treated them with high glucose combined with intermittent hypoxia., Key Findings: In conclusion, IH aggravated apoptosis and autophagy defects in T2DM mice, and increased the protein expression of HMGB1 and TLR4. This was also confirmed in HG + IH-treated hippocampal HT22 cells. HMGB1 siRNA can significantly reduce the protein expression of HMGB1 and TLR4, reverse neuronal apoptosis and enhance autophagy., Significance: We believe that HMGB1 is a key factor in the regulation of hippocampal neuronal apoptosis and autophagy defects in T2DM combined with OSA. To explore the effects of coastal eutrophication and warming on phytoplankton biomass and cell size, we analyzed current and historical data for size-fractionated chlorophyll a (chla) in Xiangshan Bay, China. Results showed that micro- and nanophytoplankton overwhelmingly dominated (>84%) in all seasons. The contribution of micro-chla was significantly lower in warm than in cold seasons, whereas contribution of pico-chla showed the opposite result. Overall, the micro-chla contribution increased with decreasing pico-chla contribution from the stable, clear, eutrophic upper bay to the turbulent, turbid lower bay, indicating that phytoplankton size structure on a spatial scale was largely shaped by water column stability and light rather than by nutrients. The impacts of fish cage culture and suspended oyster culture on macrobenthic communities were investigated in Xiangshan Bay, China, on a seasonal basis from January to October of 2015. Samples were collected from a fish cage farm, a suspended oyster farm, and two corresponding reference sites. Two-way ANOVA results showed that species richness, abundance, biomass, and Shannon-Wiener diversity differed significantly between the four different investigated areas, and different seasons as well. Cluster analysis showed that macrobenthic community composition in the fish and oyster culture areas significantly differed from that in the reference sites, respectively. Trophic structure of macrobenthos in the fish and oyster culture areas mostly clustered together owing to higher abundance and biomass of surface-deposit feeders and carnivores. The macrobenthic communities and feeding guilds correlated highly with environmental factors, such as bottom water chlorophyll a and nutrients, as well as sediment total organic carbon. Although integrated multi-trophic aquaculture is regarded as a suitable approach to coordinate desirable economic, social, and environmentally sustainable outcomes, coastal ecosystems may suffer from large-scale nutrient enrichment due to aquaculture and other human activities, which should not be considered in isolation., (Copyright © 2019. Commercial marine organisms were collected from the coast of Xiangshan Bay to investigate the concentrations of eight heavy metals (As, Cd, Cr, Cu, Hg, Ni, Pb, and Zn) and evaluate the potential health risks. The results indicated considerable variations in the heavy metal concentrations among six species groups, of them mollusks (seasnail, benthic bivalve, and oyster) generally contained relative high levels of most metals, followed by crustaceans (crab and shrimp), by contrast, fish had low concentrations of all metals, except Hg. Three heavy metal groups were identified to interpret the accumulative characteristics in the marine organisms. Spatial distributions illustrated the geographical variations of heavy metal concentrations in the sampling areas. Moreover, maricultured organisms demonstrated lower heavy metal concentrations than did the wild. Health risks of most heavy metals exposed from marine organism consumption were safe, except for As which is associated with the high target cancer risk values., (Copyright © 2019. The existence of bauxite residue disposal area (BRDA) is a serious problem in China owing to the huge quantity as well as toxicity and high alkalinity of bauxite residue. To assess the impact of uncontrolled release of bauxite residue on soil, 80 surface soil samples from areas nearby the BRDA in China, were tested to obtain the levels of heavy metals, as well as exchangeable sodium percentage, pH, electrical conductivity (EC), and total alkalinity (TA). High levels of total concentrations of Cd, V, Pb, and Mo were detected in the study area, along with high pH and exchangeable Na, K, Ca, and Mg. Spatial distribution generated by Kriging interpolation of data on surface soils indicated variabilities in the concentrations of heavy metals, alkalinity, and salinity. Factor analyses confirmed the spatial distribution variance and the influence of prevailing winds. The enrichment factors of soil showed extreme enrichment of Mo, moderate enrichment of Cd and V; and high synthesis scores for soil salinization degree were noted from the eastern to southeastern region of the BRDA. Background: American adolescents and young adults between the ages of 15 and 24 account for 50% of all sexually transmitted diseases (STDs) annually. Rural populations in this age group are often understudied, despite having factors that place them at higher risk for STDs., Purpose: The purpose of this study was to evaluate the utility of time series analysis in the assessment of rural Pennsylvania county-level chlamydia and gonorrhea rates overtime (2004-2014) for 15- to 19- and 20- to 24-year-old age groups by gender., Methods: An exploratory analysis was completed using Pennsylvania STD surveillance case report and census data, to develop a linear mixed-effects model of the STD rate for each Pennsylvania county for the years 2004 through 2014 using 3-month increments. A cubic polynomial spline regression model was assumed over the 44 time points for each county to account for possible oscillations in the STD rate during the 11-year period., Results: Eight out of 12 rural counties had a significant increase in chlamydia or gonorrhea rates, and five rural counties had significant decreases in chlamydia or gonorrhea rates from 2004 to 2014., Conclusions: Results from this study provide the first analysis of change in rates of STDs in rural settings and demonstrate the utility of time series analysis for populations with small sample sizes., (Copyright © 2017 Elsevier Inc. The purpose of this study was to demonstrate that the flow function (FFc) of pharmaceutical powders, as measured by rotational shear cell, is predominantly governed by cohesion but not friction coefficients. Driven by an earlier report showing an inverse correlation between FFc and the cohesion divided by the corresponding pre-consolidation stress (Wang et al. 2016. Powder Tech. 294:105-112), we performed analysis on a large data set containing 1130 measurements from a ring shear tester and identified a near-perfect inverse correlation between the FFc and cohesion. Conversely, no correlation was found between FFc and friction angles. We also conducted theoretical analysis and estimated such correlations based on Mohr-Coulomb failure model. We discovered that the correlation between FFc and cohesion can sustain as long as the angle of internal friction at incipient flow is not significantly larger than the angle of internal friction at steady-state flow, a condition covering almost all pharmaceutical powders. The outcome of this study bears significance in pharmaceutical development. Because the cohesion value is strongly influenced by the interparticle cohesive forces, this study effectively shows that it is more efficient to improve the pharmaceutical powder flow by lowering the interparticle cohesive forces than by lowering the interparticle frictions., (Copyright © 2017 American Pharmacists Association®. Extracellular matrix (ECM) is crucial to many aspects of vascular morphogenesis and maintenance of vasculature function. Currently the recapitulation of angiogenic ECM microenvironment is still challenging, due mainly to its diverse components and complex organization. Here we investigate the angiogenic potential of human lung fibroblast-derived matrix (hFDM) in creating a three-dimensional (3D) vascular construct. hFDM was obtained via decellularization of in vitro cultured human lung fibroblasts and analyzed via immunofluorescence staining and ELISA, which detect multiple ECM macromolecules and angiogenic growth factors (GFs). Human umbilical vein endothelial cells (HUVECs) morphology was more elongated and better proliferative on hFDM than on gelatin-coated substrate. To prepare 3D construct, hFDM is collected, quantitatively analyzed, and incorporated in collagen hydrogel (Col) with HUVECs. Capillary-like structure (CLS) formation at 7day was significantly better with the groups containing higher doses of hFDM compared to the Col group (control). Moreover, the group (Col/hFDM/GFs) with both hFDM and angiogenic GFs (VEGF, bFGF, SDF-1) showed the synergistic activity on CLS formation and found much larger capillary lumen diameters with time. Further analysis of hFDM via angiogenesis antibody array kit reveals abundant biochemical cues, such as angiogenesis-related cytokines, GFs, and proteolytic enzymes. Significantly up-regulated expression of VE-cadherin and ECM-specific integrin subunits was also noticed in Col/hFDM/GFs. In addition, transplantation of Col/hFMD/GFs with HUVECs in skin wound model presents more effective re-epithelialization, many regenerated hair follicles, better transplanted cells viability, and advanced neovascularization. We believe that current system is a very promising platform for 3D vasculature construction in vitro and for cell delivery toward therapeutic applications in vivo., Statement of Significance: Functional 3D vasculature construction in vitro is still challenging due to the difficulty of recapitulating the complex angiogenic extracellular matrix (ECM) environment. Herein, we present a simple and practical method to create an angiogenic 3D environment via incorporation of human lung fibroblast-derived matrix (hFDM) into collagen hydrogel. We found that hFDM offers a significantly improved angiogenic microenvironment for HUVECs on 2D substrates and in 3D construct. A synergistic effect of hFDM and angiogenic growth factors has been well confirmed in 3D condition. The prevascularized 3D collagen constructs also facilitate skin wound healing. The goal of this project was to develop an easily operable and scalable process for the recovery and purification of foot-and-mouth disease virus (FMDV) from cell culture. Heparin resins HipTrap Heparin HP and AF-Heparin HC-650 were utilized to purify FMDV O/HN/CHA/93. Results showed that the purity of AF-Heparin HC-650 was ideal. Then, the O/HN/CHA/93, O/Tibet/CHA/99, Asia I/HN/06, and A/CHA/HB/2009 strains were purified by AF-Heparin HC-650. Their affinity/virus recoveries were approximately 51.2%/45.8%, 71.5%/70.9%, 96.4%/73.5, and 59.5%/42.1%, respectively. During a stepwise elution strategy, the viral particles were mainly eluted at 300mM ionic strength peaks. The heparin affinity chromatography process removed more than 94% of cellular and medium proteins. Anion exchange resin Capto Q captured four FMD virus particles; 40% of binding proteins and 80%-90% of viral particles were eluted at 450mM NaCl. Moreover, ionic strength varied from 30 to 450mM had no effect on the immunity to FMDV. The results revealed that heparin sulfate may be the main receptor for CHA/99 strain attachment-susceptible cells. Heparin affinity chromatography can reach perfect results, especially when used as a ligand of the virus. Anion exchange is useful only as previous step for further purification., (Copyright © 2016. Nuclear receptor coactivator 5 (NCOA5) is known to modulate ERα-mediated transcription and has been found to be involved in the progression of several malignancies. However, the potential correlation between NCOA5 and clinical outcome in patients with luminal breast cancer remains unknown. In the present study, we demonstrated that NCOA5 was significantly up-regulated in luminal breast cancer tissues compared with adjacent non-cancerous tissues both in validated cohort and TCGA cohort. Moreover, Kaplan-Meier analysis indicated that patients with high NOCA5 expression had significantly lower overall survival (P = 0.021). Cox regression analysis indicated that the high NOCA5 expression was independent high risk factor as well as old age (>60) and HER-2 expression (P = 0.039; P = 0.003; P = 0.005; respectively). This study provides new insights and evidences that NOCA5 over-expression was significantly correlated with progression and prognosis in luminal breast cancer. Previously, granulated lactose carriers were shown to improve uniformity and aerosolization of a low-dose model drug. In the present study, the blending uniformity and aerosol dispersion performance were assessed for 2 model drugs salbutamol sulfate (SS) and rifampicin (RIF), blended at high loadings (10% or 30% drug) with granulated lactose carriers. The model drug powders differed in particle size distribution, morphology, density, and surface energies. Content uniformity of RIF blends was better than that of SS. Aerosolization studies showed that all blend formulations had acceptable emitted fractions (>70%). The SS blends showed low induction-port deposition (6%-10%) compared to RIF (5%-30%). This difference was greater at high flow rates. At 90 L/min, the low induction port deposition of SS blends allowed high fine particle fraction (FPF) of 73%-81%, whereas the FPF of the RIF blends was around 43%-45% with higher induction port deposition. However, SS blends exhibited strong flow rate-dependent performance. Increasing the flow rate from 30 L/min to 90 L/min increased SS FPF from approximately 20% to 80%. Conversely, RIF blends were flow rate and drug loading independent. It was concluded that the aerosolization of high drug-loaded dry powder inhaler formulations using granulated lactose, particularly flow rate dependency, varies with active pharmaceutical ingredient properties., (Copyright © 2016 American Pharmacists Association®. Myeloid leukemia factor 1-interacting protein (MLF1IP) has been found to be involved in the progression of several malignancies. The potential correlation between MLF1IP and clinical outcome in patients with luminal breast cancer, however, remains unknown. In the present study, we demonstrated that MLF1IP was significantly upregulated in luminal breast cancer tissue compared with adjacent normal tissue both in validated cohort and TCGA cohort. Upregulated expression of MLF1IP was correlated with more often lymph node metastasis and negative progesterone receptor expression in TCGA cohorts. Kaplan-Meier analysis indicated that patients with high MLF1IP expression had significantly lower overall survival. Moreover, multivariate analysis revealed that high MLF1IP expression was independent high risk factor as well as old age (>60) and distant metastasis. This study provides new insights and evidences that MLF1IP over-expression plays important roles in progression of luminal breast cancer. The occurrence of 15 antibiotics classified as sulphonamides, fluoroquinolones, macrolides, tetracyclines and trimethoprim in sediment, overlying water, and pore water matrices in Taihu Lake, China was studied. The total concentrations were from 4.1 μg/kg to 731 μg/kg, from 127 ng/L to 1210 ng/L, and from 1.5 ng/L to 216 ng/L in sediment, overlying water and pore water, respectively. Antibiotics in different locations originated from various sources, depending on human, agricultural and aquacultural activities. Composition analysis indicated that human-derived and animal-derived drugs significantly contributed to the total contamination of antibiotics in the lake, indicating the high complexity of contamination sources in Taihu Lake Basin. The in situ sediment-pore water partitioning coefficients were generally greater than sediment-overlying water partitioning coefficients, suggesting continuous inputs into the lake water. This study shows that antibiotics are ubiquitous in all compartments in Taihu Lake, and their potential hazards to the aquatic ecosystem need further investigation., (Copyright © 2014 Elsevier B.V. Docetaxel lipid microsphere (DT-LM) is a novel formulation of docetaxel without Tween-80. A sensitive, robust and reproducible ultrafiltration (UF) followed by UPLC-MS/MS method was developed and validated for the quantification of unbound docetaxel in human plasma using paclitaxel as IS. Ultrafiltrate samples were chromatographed on Acquity UPLC BEH C18 column (50 mm × 2.1 mm, 1.7 μm). The mobile phase was a mixture of 10mM ammonium formate in water containing 0.2% formic acid (A) and acetonitrile containing 0.2% formic acid (B). The volume of plasma utilized was only 450 μL. The calibration curve was linear over the range of 0.2-200 ng/mL, with LLOQ of 0.2 ng/mL. The method was shown to be reliable and reproducible with intra- and inter-day precision and accuracy <±15%, and extraction recovery of 98.1-104.8%. Docetaxel was stable during stability studies, e.g., short term, post-preparation and freeze-thaw cycles. Our previous study revealed that intracerebroventricular oxytocin (OT) markedly inhibited the restraint stress-priming conditioned place preference (CPP) reinstatement induced by methamphetamine (MAP) via the glutamatergic system. In this study, the effect of microinjection with OT into mesocorticolimbic regions, the medial prefrontal cortex (mPFC) and the dorsal hippocampus (DHC), on the restraint stress-priming CPP reinstatement were further studied. The results showed that a 15-min restraint stress significantly reinstated MAP-induced CPP, which was inhibited by the microinjection of OT (0.5 and 2.5μg/μl/mouse) into the mPFC. Atosiban (Ato), a selective inhibitor of OT receptor, could absolutely block the effect of OT (2.5μg/μl/mouse). The reinstatement was inhibited by microinjecting with OT (2.5 but not 0.5μg/μl/mouse) into the DHC, which could not be reversed by Ato. Western blotting results showed that the levels of GLT1, VGLUT2, NR2B, p-ERK1/2 and p-CREB expressions in the mPFC were increased and CaMKII was decreased markedly after the stress-priming MAP-induced CPP reinstatement test. OT blocked the changing levels of GLT1, VGLUT2, NR2B, p-CREB and CaMK II, which were reversed by Ato, but failed to affect the elevated expression of p-ERK1/2. In DHC, the levels of VGLUT2, p-ERK1/2 and CREB expressions were reduced during the stress-induced reinstatement, which could be reversed by OT and further abolished by Ato. The present results suggest that mPFC and DHC play differential roles in restraint stress-priming CPP reinstatement induced by MAP and OT via OT receptor affects the reinstatement in which the glutamatergic system is involved., (Crown Copyright © 2013. This study was to systematically investigate the effect of mobile phase additives, including ammonia water, formic acid, acetic acid, ammonium chloride and water (as a control), on qualitative and quantitative analysis of fifteen representative ginsenosides based on liquid chromatography hybrid quadrupole-time of flight mass spectrometry (LC-Q-TOF/MS). To evaluate the influence of mobile phase additives on qualitative performance, the quality of the negative mode MS/MS spectra of ginsenosides produced by online LC-Q-TOF/MS analyses, particularly the numbers and intensities of fragment ions, were compared under different adduct ion states, and found to be strongly affected by the mobile phase additives. When 0.02% acetic acid was added in the mobile phase, the deprotonated ginsenosides ions produced the most abundant product ions, while almost no product ion was observed for the chlorinated ginsenoside ions when 0.1mM ammonium chloride was used as the mobile phase additive. On the other hand, sensitivity, linear range and precision were adopted to investigate the quantitative performance affected by different mobile phase additives. Validation results of the LC-Q-TOF/MS-based quantitative performance for ginsenosides showed that ammonium chloride not only provided the highest sensitivity for all the target analytes, but also dramatically improved the linear ranges, the intra-day and inter-day precisions comparing to the results obtained using other mobile phase additives. Importantly, the validated method, using 0.1mM ammonium chloride as the mobile phase additive, was successfully applied to the quantitative analysis of ginsenosides in rat plasma after intragastric administration of Ginsenoside Extract at 200mg/kg. In conclusion, 0.02% acetic acid was deemed to be the most suitable mobile phase additive for qualitative analysis of ginsenosides, and 0.1mM ammonium chloride in mobile phase could lead to the best quantitative performance. Docetaxel lipid microsphere (DT-LM), an intravenous lipid emulsion for docetaxel without Tween 80, has demonstrated significant advantage over other conventional docetaxel formulations with respect to keeping sustained release, reducing irritation or toxicity of drug, sterile for intravenous injection and presenting targeting. A rapid, sensitive and reproducible ultra high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for determination of total docetaxel from a lipid microsphere formulation in human plasma using paclitaxel as internal standard (IS) has been developed and validated. The analytes and IS were extracted from plasma by simple liquid-liquid extraction and separated on ACQUITY UPLC BEH C18 column at a flow rate of 0.3 ml/min using gradient elution mode. The total analytical time was only 2.5 min. Detection and quantitation were performed by electrospray ionization (ESI) in the positive ionization mode by multiple reaction monitoring (MRM) of the transitions at m/z 808.3→527.1 for docetaxel and 854.0→285.9 for IS. The assay was linear over the concentration range of 2-5000 ng/ml (r(2)>0.99) with the lower limit of quantification (LLOQ) of 2 ng/ml. The intra- and inter-day precision in terms of relative standard deviation (RSD%) was within 9% and accuracy in terms of relative error (RE%) was within 12%. A novel method of ultrasonic probe-assisted ionic liquid dispersive liquid-liquid microextraction combined with electrothermal atomic absorption spectrometry (ETAAS) was developed for the determination of chromium(VI) species in water samples. In this procedure, the hydrophobic chelate of chromium(VI) with ammonium pyrrolidinedithiocarbamate (APDC) was extracted into the fine droplets of 1-hexyl-3-methylimidazolium hexafluorophosphate ([Hmim][PF(6)]), which was dispersed into the aqueous sample solution by ultrasonication using an ultrasonic probe. Several variables such as the volume of [Hmim][PF(6)], sample pH, concentration of APDC, and extraction time were investigated in detail. Under the optimum conditions, the limit of detection of the proposed method was 0.07 ng mL(-1) for Cr(VI) and the relative standard deviation for five-replicated determination of 2.0 ng mL(-1) Cr(VI) was 9.2%. A new voltammetric enzyme-linked immunoassay system using the electrochemical substrate 2,3-diaminopyridine (DAP) and horseradish peroxidase (HRP) system has been developed. DAP is oxidized with H2O2 catalyzed by HRP, and the resulting electroactive product produces a sensitive voltammetric peak at potential of -0.72V (vs. saturated calomel electrode (SCE)) in Britton-Robinson (BR) buffer solutions. The enzyme-catalyzed reaction conditions and voltammetric detection conditions have been investigated in detail. Under the selected optimum conditions, the linear range for detection of free HRP is from 6.0 x 10(-11) to 1.0 x 10(-8) g mL(-1) with a detection limit of 1.0 x 10(-12) g mL(-1). The new voltammetric detection system has been successfully applied for the assay of prostate specific antigen (PSA) in human serum ranging from 0.4 to 100 ng mL(-1) with a detection limit of 0.1 ng mL(-1), which is five times lower than that of traditional o-phenylenediamine (OPD) spectrophotometric enzyme-linked immunosorbent assay (ELISA) method. The proposed N-heterocyclic electrochemical detection system of DAP-H2O2-HRP has provided a new and much improved immunoassay method.

In this article, colloidal gold nanoparticles (Au NPs) and carboxyl group-functionalized CdS Nanoparticles (CdS NPs) were immobilized on the Au electrode surface to fabricate a novel electrochemical DNA biosensor. Both Au NPs and CdS NPs, well known to be good biocompatible and conductive materials, could provide larger surface area and sufficient amount of binding points for DNA immobilization. Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) experiments were performed to follow the whole electrode fabrication process. DNA immobilization and hybridization were characterized with differential pulse voltammetry (DPV) by using [Co(phen)(2)(Cl)(H(2)O)]Cl.2H(2)O as an electrochemical hybridization indicator. With this approach, the target DNA could be quantified at a linear range from 2.0x10(-10) to 1.0x10(-8) M, with a detection limit of 2.0x10(-11) M by 3sigma. In addition, the biosensor exhibited a good repeatability and stability for the determination of DNA sequences.

A novel nanocomposite material of multiwalled carbon nanotubes (MWCNTs) and room temperature ionic liquid (RTIL) N-butylpyridinium hexafluorophosphate (BPPF6) was explored and used to construct a novel microperoxidase-11 (MP-11) biosensor for the determination of hydrogen peroxide (H2O2). Cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were used to characterize the performance of the biosensor. Under the optimized experimental conditions, H2O2 could be detected in a linear calibration range of 0.5 to 7.0 x 10(-7)mol L(-1) with a correlation coefficient of 0.9949 (n=9) and a detection limit of 3.8 x 10(-9) mol L(-1) at 3 sigma. The modified electrodes displayed excellent electrochemical response, high sensitivity, long-term stability, and good bioactivity and selectivity.

A new voltammetric enzyme-linked immunoassay system of 3,4-diaminobenzoic acid (DBA)-H(2)O(2)-horseradish peroxidase (HRP) for sensitive detection of prostate specific antigen (PSA) in human serum was present. In the proposed procedure, labelled HRP efficiently catalyzed the oxidation reaction of DBA by H(2)O(2) and generated the electroactive product, 2,2'-biamino-4,4'-bicarboxyl azobiphenyl, which produced a sensitive second-order derivative linear sweep voltammetric peak at potential of -0.62V (versus SCE) in Britton-Robinson (BR) buffer solution. The linear range for detection free HRP was from 4.0x10(-12) to 4.0x10(-9)gml(-1) and the detection limit was about 6.0x10(-13)gml(-1). The proposed new system could be used for detection of PSA ranging from 0.20 to 16.0ngml(-1) with a detection limit of 0.10ngml(-1), which was five times lower than that of the traditional o-phenylendiamine spectrophotometric enzyme-linked immunosorbent assay (ELISA) method. The proposed electrochemical ELISA method is simple, inexpensive, reproducible and sensitive, which shows potential for detecting PSA in clinical diagnosis.

A new porous sorbent for wastewater treatment of metal ions was synthesized by covalent grafting of molecularly imprinted organic-inorganic hybrid on silica gel. With sucrose and polyethylene glycol 4000 (PEG 4000) being synergic imprinting molecules, covalent surface coating on silica gel was achieved by using polysaccharide-incorporated sol-gel process starting from the functional biopolymer, chitosan and an inorganic epoxy-precursor, gamma-glycidoxypropyltrimethoxysiloxane (GPTMS) at room temperature. The prepared porous sorbent was characterized by using simultaneous thermogravimetry and differential scanning calorimeter (TG/DSC), scanning electron microscopy (SEM), nitrogen adsorption porosimetry measurement and X-ray diffraction (XRD). Copper ion, Cu(2+), was chosen as the model metal ion to evaluate the effectiveness of the new biosorbent in wastewater treatment. The influence of epoxy-siloxane dose, buffer pH and co-existed ions on Cu(2+) adsorption was assessed through batch experiments. The imprinted composite sorbent offered a fast kinetics for the adsorption of Cu(2+). The uptake capacity of the sorbent imprinted by two pore-building components was higher than those imprinted with only a single component. The dynamic adsorption in column underwent a good elimination of Cu(2+) in treating electric plating wastewater. The prepared composite sorbent exhibited high reusability. Easy preparation of the described porous composite sorbent, absence of organic solvents, cost-effectiveness and high stability make this approach attractive in biosorption.
