The Bletilla striata polysaccharides (BSP) extracted through alkali-assisted method exhibit significant antioxidant activity, but its bioaccessibility was inadequate due to its tightly filamentous reticulation structure and high molecular weight. The anti-photoaging and anti-melanogenesis effects of degraded BSP (DBSPs) against UVB-induced oxidative stress on the skin were investigated. The molecular weights of the DBSPs were reduced to 153.94 kDa, 66.96 kDa, and 15.54 kDa from an initial value of 298.82 kDa. The degradation treatment altered the branched chain structure of the DBSPs, while the backbone structure, triple-helix structure, and crystallinity remained. DBSPs with a lower molecular weight exhibit better in vitro antioxidant activity. DBSPs did not show cytotoxicity to HSF cells but inhibited B16F10 cell proliferation. The addition of DBSPs protected HSF and B16F10 cells from oxidative stress and reduced ROS levels, B16F10 melanin content, and B16F10 tyrosinase activity after UVB damage, but DBSP-10 particles were slightly less effective due to aggregation. Local drug delivery has generated considerable interest due to its controlled and sustained drug release at the target site on demand. Nanoaggregate-incorporated composite hydrogels are desirable as local drug delivery systems; however, it is difficult to achieve sustained and controlled hydrophobic drug release and superior mechanical properties in one system. Herein, a "smart" composite hydrogel was synthesized by incorporating hemicellulose-based nanoaggregates into a double network consisting of alginate/Ca 2+ and polyacrylic acid-co-dimethylaminoethyl methacrylate [P(AA-co-DMAEMA)]. Hemicellulose-based nanoaggregates were assembled from xylan-rich hemicellulose laurate methacrylate (XH-LA-MA) polymers and entrapped into the hydrogel framework via chemical fixation. Another composite hydrogel with physically embedded hemicellulose laurate (XH-LA) nanoaggregates was prepared as a comparison. Accordingly, covalently cross-linked XH-LA-MA nanoaggregates in hydrogels resulted in a denser pore structure and reinforced mechanical properties. Nanoaggregate diffusion analysis revealed that covalent bonding between the nanoaggregates and the hydrogel framework contributed to prolonged diffusion behavior. Curcumin (Cur)-loaded XH-LA-MA composite hydrogels enabled sustained Cur release in simulated body fluid and showed stimulus responsiveness toward ethylenediaminetetraacetic acid (EDTA) and/or glutathione (GSH). All the composite hydrogels were biocompatible, as verified by Cell Counting Kit-8 (CCK-8) assay against NIH/3T3 cells. Selective fractionation of hemicelluloses is of great significance for realizing high-value application of hemicelluloses and comprehensive utilization of lignocellulosic biomass. Tetramethylammonium hydroxide (TMAH) solvent has been confirmed as a promising solvent to selectively fractionate hemicelluloses from holocellulose. Herein, TMAH solvent was adopted to pretreat poplar thermomechanical pulp (PTMP) for the selective fractionation of hemicelluloses and enhancement of enzymatic hydrolysis performance of residues. The maximal hemicelluloses yield (65.0 %) and excellent cellulose retention rate (93.3 %) were achieved after pretreatment by the 25 wt% TMAH solvent, while the delignification was only 33.9 %. The hemicelluloses fractions could be selectively fractionated with high molecular weights (109,800-118,500 g/mol), the contents of Klason lignin in them were low (3.2-5.9 %), and the dominating structure of them was 4-O-methylglucurono-β-D-xylan. Compared to the H 2 SO 4 and NaOH methods, the hemicelluloses fractionated by TMAH method exhibited higher yields, more complete structures and higher molecular weights. Furthermore, the crystalline structure of cellulose practically remained stable, and the highest yield of enzymatic hydrolysis glucose was 57.5 %, which was 3.3 times of that of PTMP. The fractionation effectiveness of TMAH solvent was not significantly reduced after repeatedly recycling. It is believed that hemicellulose plays a crucial role in binding cellulose and lignin in plant cells. It may provide significant implications through figuring out the interaction between hemicellulose and microfibers and gaining insights how the structure of hemicellulose affects its association with cellulose nanofibers. Herein, the hemicellulose and nanocellulose fractions from pulps obtained by controlling the H-factors of kraft pulping process were quantitatively evaluated for their adsorption behavior using QCM-D. The results showed that harsher cooking (corresponding to high H-factor) significantly affected the chemical composition of hemicellulose, leading to a decrease of its molecular weight and gradually turning it into a linear structure. Hemicellulose possesses a strong natural affinity for CNC-coated sensors. The hemicellulose from the pulp cooked by high H-factor process decreases its ability to adsorb onto nanocellulose, the adsorption rate also slows down, and the conformation of the adsorbed layer changes which makes the binding weak and reversible. In this work, we investigate a multicomponent synthetic method for combining saccharides with cellulose to produce saccharide branched cellulose (b-Cel). First, cellulose is modified conventionally using carboxymethyl to create carboxyl functional groups for multicomponent reactions. The Passerini three-component reaction (Passerini-3CR) is then used to synthesize the saccharide b-Cel, with particular attention paid to the scope of the substrate and reaction process optimization. The structure of saccharide b-Cel is regulated by modifying the carboxyl group of cellulose molecules, the kind of saccharide molecules (including glucose, galactose, lactose, cellobiose, and cellulose), and the degree of branching. The branched structure of saccharide b-Cel greatly influenced its rheological characteristics and solubility. Hemicellulose extracted from ecalyptus APMP pulping waste liquor and undergoes etherification modification to produce carboxymethyl hemicellulose (CMHC). Subsequently, CMHC undergoes esterification reaction with p-hydroxybenzoic acid to synthesize a novel polysaccharide-based preservative known as carboxymethyl hemicellulose p-hydroxybenzoate (P-CMHC). The synthesis conditions of P-CMHC were optimized using the response surface methodology, resulting in an optimal esterification condition that achieved a degree of substitution of 0.232. P-CMHC exhibits excellent antioxidant activity, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical scavenging activities. Additionally, it demonstrates favorable hygroscopic and moisturizing properties. Thiazole blue (MTT) experiments evaluating cell proliferation rate indicate that P-CMHC possesses negligible cytotoxicity, making it a promising, safe, and healthy preservative. Polysaccharides, due to their remarkable features, have gained significant prominence in the sustainable production of nanoparticles (NPs). High market demand and minimal production cost, compared to the chemically synthesised NPs, demonstrate a drive towards polysaccharide-based nanoparticles (PSNPs) benign to environment. Various approaches are used for the synthesis of PSNPs including cross-linking, polyelectrolyte complexation, and self-assembly. PSNPs have the potential to replace a wide diversity of chemical-based agents within the food, health, medical and pharmacy sectors. Nevertheless, the considerable challenges associated with optimising the characteristics of PSNPs to meet specific targeting applications are of utmost importance. This review provides a detailed compilation of recent accomplishments in the synthesis of PSNPs, the fundamental principles and critical factors that govern their rational fabrication, as well as various characterisation techniques. Noteworthy, the multiple use of PSNPs in different disciplines such as biomedical, cosmetics agrochemicals, energy storage, water detoxification, and food-related realms, is accounted in detail. Insights into the toxicological impacts of the PSNPs and their possible risks to human health are addressed, and efforts made in terms of PSNPs development and optimising strategies that allow for enhanced delivery are highlighted. To comprehend the biosynthesis processes of conifers, it is essential to investigate the disparity between the cell wall shape and the interior chemical structures of polymers throughout the development of Chinese pine. In this study, branches of mature Chinese pine were separated according to their growth time (2, 4, 6, 8 and 10 years). The variation of cell wall morphology and lignin distribution was comprehensively monitored by scanning electron microscopy (SEM) and confocal Raman microscopy (CRM), respectively. Moreover, the chemical structures of lignin and alkali-extracted hemicelluloses were extensively characterized by nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC). The thickness of latewood cell walls increased steadily from 1.29 μm to 3.38 μm, and the structure of the cell wall components became more complicated as the growth time increased. Based on the structural analysis, it was found that the content of β-O-4 (39.88-45.44/100 Ar), β-β (3.20-10.02/100 Ar) and β-5 (8.09-15.35/100 Ar) linkages as well as the degree of polymerization of lignin increased with the growth time. The complication propensity increased significantly over 6 years before slowing to a trickle over 8 and 10 years. In recent years, the biological activity of polysaccharides and their derivatives has been widely studied. However, in addition to the natural polysaccharides directly extracted from plants and animals, there are rich polysaccharides in the pulping waste liquor that have not been fully utilized. The extracted polysaccharide from eucalyptus Alkaline Peroxide Mechanical Pulping (APMP) waste liquor was used as a raw material. For the production of carboxymethyl polysaccharide, the effects of temperature (T), the amount of alkali (NaOH) and the amount of etherifying agent (ClCH 2 COOH) on the degree of substitution (DS) were investigated, the optimal preparation conditions are: reaction time 2 h, temperature 75 °C, and the molar ratio of polysaccharide, NaOH and ClCH2COOH is 1:1:2, the highest DS is 1.47; FT-IR, NMR and GPC were used to characterize the structure and Molecular weight, the results show that the polysaccharide of APMP waste liquor is rich in xylan, and it was proved that the carboxymethyl substitution was successful and the positions of the substituent group were determined. The characterization and biological activity research of xylan polysaccharide (XP) and carboxymethyl xylan polysaccharide (CMXP), such as antioxidation, moisture absorption/retention, bacteriostatic action and cytotoxicity were discussed. The conductive hemicellulose hydrogels (CHHs) were achieved by incorporating carboxyl terminated aniline pentamer (CTAP) into the hydrophilic xylan-rich hemicellulose networks with epichlorohydrin. CHHs were obtained by facile one-pot reaction in water at ambient temperature. The successful synthesis of CTAP was confirmed by FT-IR, 1 H NMR, and UV-vis spectra. All the hydrogels reached their equilibrium-swollen state within 12 h. The morphology and the thermal stability of CHHs were adjusted by the addition of CTAP. The conductivities of CHHs could also be tuned from 8.92 × 10 -5 to 2.11 × 10 -3 S/m by adding the blending amounts of the CTAP from 0 to 50% mass ratio while simultaneously altering the equilibrium swelling ratio from 28.1 to 15.8. CHHs showed homogeneous porous structure, tunable swelling behavior, controllable conductivity, and good thermal stability. Eucalyptus wood was pretreated with an integrated process based on hydrothermal and alkaline pretreatments. The structural changes of hemicelluloses during the pretreatments and the components of the hydrolysates were comprehensively characterized. Sugar and spectral analyses indicated that the hemicelluloses remained in the residues obtained at hydrothermal pretreatment under low temperatures were mainly composed of a (1 → 4)-β-D-Xylp backbone with 4-O-methyl-α-D-glucuronic acids attached at O-2 of the xylose together with various monosaccharides of rhamnose, arabinose, galactose, glucose, and mannose. The pretreatments resulted in serious degradation of hemicelluloses at high pretreatment temperatures. The distribution changes of the hemicelluloses in the cell walls during the integrated pretreatments were detected by Confocal Raman Microscopy, which revealed that the dissolution of hemicelluloses in different morphological regions was inhomogeneous, and the a large portion of hemicelluloses were removed from the secondary cell wall regions during the pretreatments., (Copyright © 2019 Elsevier B.V. Huge amount of waste hemicellulose liquor annually produced in China was burned or discharged into surrounding environment without further sufficient utilization, causing severely environmental issues. In order to solve this problem, this study aimed to find a way to effectively use aqueous waste hemicellulose solution from viscose fiber mills for bio-based materials development. Hereby, polyacrylamide-hemicellulose hybrid films were synthesized through the copolymerization of acrylamide (AM) monomers and hemicellulose initiated by potassium persulfate/N,N,N',N'-tetramethylethylenediamine (KPS/TMEDA) redox initiator system with cross-linker N,N-Methylene-bis(acrylamide) (MBA). The as-prepared films had been further characterized by Fourier transform infrared (FT-IR), scanning electron microscopy (SEM), gel permeation chromatography (GPC), mechanical test, light transmittance determination and oxygen transmission rate measurement. The results showed that the hybrid films possessed many advantages, such as ambient-temperature water solubility, good recyclability, high stretchable properties and low oxygen permeability. These properties implied their application potentials in several areas, such as packaging electronics products, food and pesticide. Galactoglucomannan (GGM) is the main hemicellulose class in wood of coniferous trees and could be potentially utilized as a possible health-promoting substance for food and pharmaceutical industry. Our aim was to evaluate effects of orally administered GGM-rich extract from Norway spruce in a rat model of chronic prostatitis associated with lower urinary tract symptoms (LUTS). Prostatic inflammation and LUTS was induced in male rats using testosterone and 17β-estradiol exposure for 18 weeks. Rats were treated with 2% GGM dissolved in drinking water during weeks 13-18. Pelvic pain response, LUT function and histopathological evaluation of the prostate were assessed. The results show that hormonal exposure induced LUTS seen as decreased urine flow rate, increased bladder pressure, voiding times, bladder capacity and residual urine volumes. GGM had positive effects on urodynamical parameters by decreasing the basal bladder pressure, increasing the urine flow rate and volume, reducing the residual volume and increasing micturition intervals. GGM reduced the extent of the hormone exposure-induced prostatic inflammation. Increase of pelvic pain induced by hormone exposure was only slightly affected by GGM treatment. In this study, an environmental-friendly hydrothermal pretreatment of Eucalyptus fiber followed with alkali post-treatment was developed to produce bioethanol efficiently. This biorefinery process allowed all major components of biomass being converted into high value-added products. The chemical and structural features of the residual hemicelluloses isolated with alkali from the hydrothermal pretreated Eucalyptus fiber, were comparatively investigated. Sugar and spectral analyses indicated that the hemicelluloses were mainly composed of glucuronoxylans, and especially hemicelluloses prepared at higher temperature (180°C) contained higher contents of glucomannans and α-glucan. Hydrothermal pretreatment resulted in a significant hydrolysis of the glycosidic linkages in xylan backbone, and thus the molecular weight of the hemicelluloses was significantly reduced from 56,520 to 7780g/mol with the increase of temperature. This suggested that a combination of hydrothermal pretreatment at low temperatures (100-140°C) and alkali post-treatment was an effective technique for isolating of hemicelluloses from Eucalyptus fiber., (Copyright © 2014 Elsevier B.V. The aim of this study was to evaluate the influence of mild alkaline pre-extraction on the hemicelluloses removal of rice straw. In addition, the performance of cellulose nanofibers (CNFs) addition on some physico-mechanical properties obtained from extracted and un-extracted (control) samples were investigated. An optimum compromise was found as operating conditions (10% soda, 50°C, and 90min) that provided moderate hemicelluloses removal (48.1%). The residual extracted rice straw particles were subjected to soda-anthraquinone pulping at 160°C for 30-60min with 6 to 18% active alkali charge. Compared with the control samples, the screened yield and Kappa number for extracted pulps decreased slightly. Extracted samples showed better air resistance compared with the un-extracted (control) pulps. However, pre-extraction was found to negatively impact on some mechanical properties such as decrease in burst and tensile indices while addition of CNFs and refining of fibers could improve the strength properties considerably. In general, the addition of CNFs in any concentrations considerably enhanced tensile and burst indices of the sheets in extracted and un-extracted conditions, compared with the unfilled ones. The improvement in mechanical properties was considered one of the key benefits brought by CNFs reinforcement. On the other hand, the stretch properties of the beaten pulps are higher than those of control samples. Hemicelluloses were extracted from holocellulose of bamboo by alkaline treatment. The phosphorylated poly(vinyl alcohol) (P-PVA) samples with various substitution degrees were prepared through the esterification of PVA and phosphoric acid. A series of hydrogels of semi-interpenetrating polymeric networks (semi-IPN) composed of hemicelluloses-g-poly(acrylic acid) (HM-g-PAA) and the phosphorylated poly(vinyl alcohol) (P-PVA) were prepared by radical polymerization using potassium persulphate (KPS) as initiator. The HM-g-PAA networks were crosslinked by N,N-methylenebisacrylamide (MBA) as a crosslinking agent in the presence of linear P-PVA. FT-IR results confirmed that the hydrogels comprised a porous crosslink structure of P-PVA and HM with side chains that carried carboxylate and phosphorylate groups. SEM observations indicated that the incorporation of P-PVA induced highly porous structure, and P-PVA was uniformly dispersed in the polymeric network. The interior network structures of the semi-IPN matrix became more porous with increasing P-PVA. The TGA results showed that the thermo-decomposing temperature and thermal stability were increased effectively for intruding the chain of P-PVA. The maximum equilibrium swelling ratio of hydrogels in distilled water and 0.9 wt% sodium chloride solutions was up to 1085 g g(-1) and 87 g g(-1), respectively. This work describes the structural changes of bagasse hemicelluloses during the cooking process involving active oxygen (O(2) and H(2)O(2)) and solid alkali (MgO). The hemicelluloses obtained from the bagasse raw material, pulp, and yellow liquor were analyzed by high-performance anion-exchange chromatography (HPAEC), gel permeation chromatography (GPC), Fourier transform infrared spectroscopy (FT-IR), and (1)H-(13)C 2D hetero-nuclear single quantum coherence spectroscopy (HSQC). The results revealed that the structure of the bagasse hemicelluloses was L-arabino-(4-O-methylglucurono)-D-xylan. Some sugar units in hemicelluloses were oxidized under the cooking conditions. Hemicellulose-based hydrophobic biomaterials with degrees of substitution ranging from 0.46 to 1.54 were synthesized under mild conditions in homogeneous media (N,N-dimethylformamide-lithium chloride) by reacting the native wheat straw hemicellulosic polymers with lauroyl chloride using 4-dimethylaminopyridine as a catalyst. Other catalysts such as N-bromosuccinimide, N-methyl pyrrolidine, N-methyl pyrrolidinone, and pyridine were also investigated. Under optimum reaction conditions (2 equiv of lauroyl chloride and triethylamine per hydroxyl group, 5% 4-dimethylaminopyridine, 40 degrees C, 35 min), a high DS value of 1.54 was obtained. The biomaterials were characterized by FT-IR spectroscopy and (13)C NMR spectroscopy as well as by thermal analysis. The results showed that the lauroylation occurred preferably at the C-3 hydroxyl group of beta-D-Xylp units in the hemicelluloses, and the thermal stability of the hydrophobic polymers increased by esterification.

A series of novel water-soluble hemicellulosic derivatives, containing carbamoylethyl and carboxyethyl groups, were heterogeneously synthesized from wheat-straw hemicelluloses with acrylamide (AA) under alkaline conditions. The factors such as reaction temperature, reaction time, molar ratio of catalysis to xylose unit in hemicelluloses and molar ratio of acrylamide to xylose unit in hemicelluloses, were investigated. The average degree of substitution (DS) was calculated by (1)H NMR spectroscopy. DS values up to 0.23 in a one-step synthesis of hemicelluloses derivatives were obtained. Under optimum conditions (60°C, NaOH to xylose unit in hemicelluloses molar ratio of 0.8, AA to xylose unit in hemicelluloses molar ratio of 8.0, reaction time of 1h) an expected ratio of carbamoylethyl group to carboxyethyl group of 4.8 in the hemicellulosic derivatives was obtained. The structural features of the hemicellulosic derivatives were characterized by FTIR, NMR spectroscopy, and by elemental analysis. The current work provides a facile method for the synthesis of hemicellulose derivatives with bifunctional groups, which could be used as wet-end additives in the papermaking industry.

The nature of water sorption to different materials has always been a complex matter to address, partly due to the different possibilities of hydrogen-bond formation. For cellulosic materials this is extremely important for its product performance. In order to gain a deeper understanding of the moisture adsorption mechanisms of cellulose and hemicelluloses, the molecular interaction between moisture and paper and between moisture and some wood polymers was studied using FTIR spectroscopy under stable humid conditions. It was found that all the moisture-sorbing sites adsorbed moisture to the same relative degree, and that the rate of adsorption was the same for all these sites. It was also noticed that the moisture is adsorbed in the form of clusters. A direct relationship was found between the moisture weight gain and the increase in the absorbance peaks for humidities up to 50% relative humidity after which the moisture gain increased faster, a fact that still remains to be explained.

Partially depolymerized, water-soluble hemicelluloses were solubilized from flax shive employing hydrothermal microwave treatment and thereafter subjected to ion-exchange chromatography, enzymatic purification and/or size-exclusion chromatography (SEC). The oligo- and polysaccharide fractions thus obtained were characterized with respect to molar mass, molar mass distribution, degree of polymerization (DP) and degree of substitution with acetyl moieties (DSAc) by employing SEC in combination with MALDI-TOF mass spectrometry. The major portion of the water-soluble flax hemicellulose consisted of an O-acetyl-4-O-methylglucuronoxylan exhibiting a DPp value (i.e., peak-average DP) of 28. When the DSAc for this O-acetyl-4-O-methylglucuronoxylan was calculated on the basis of the MALDI-MS spectra obtained without and following deacetylation, a value of 0.7 was obtained. In addition, an O-acetyl-glucomannan (DPp=9, DS=0.4) and minor quantities of small neutral O-acetyl-xylooligosaccharides were also isolated from the mixture of water-soluble hemicelluloses released from the flax shive by microwave treatment.

Partly depolymerized hemicelluloses isolated from wood chips of spruce and aspen employing microwave treatment were resolved using size-exclusion chromatography (SEC) into oligo- and polysaccharide fractions containing components with a narrow range of sizes, as determined by MALDI mass spectroscopy. The degree of substitution with acetyl moieties (DS) was also calculated on the basis of the MALDI-MS spectra obtained prior to and following deacetylation. For spruce hemicelluloses, the low molecular mass fraction contained small arabino-4-O-methylglucuronoxylan oligosaccharides, with DP values ranging from 4 to approximately 20, separated primarily on the basis of their charge density. The fraction eluted last consisted of an O-acetyl-(galacto)glucomannan polysaccharide of peak-average DP value (DP(p)) 14. The degree of substitution with acetyl groups (DS) decreased with decreasing DP, a value DS of 0.39 being obtained for the fraction with DP(p) 12. For the aspen hemicelluloses, the SEC fractions eluted first contained an acidic O-acetyl-4-O-methylglucuronoxylan polysaccharide with DP ranging from 10 to approximately 28 and an average DS of approximately 0.75. The fractions eluted last consisted of oligosaccharide mixtures composed primarily of small neutral O-acetyl-xylooligosaccharides (DP(p) 6, DS 0.41), together with minor quantities of an O-acetyl-glucomannan.

Sulfated beta-(1-->4)-galacto-oligosaccharides were prepared from an arabino-galacto-rhamno-galacturonan from Lupinus polyphyllus Lindl. by successive partial hydrolysis and SO3-pyridine sulfation in DMF. The resulting oligosaccharide polysulfates were analyzed by analytical GPC and the sulfate content was determined by ion chromatography. DP 5 and higher showed a pronounced antiangiogenic effect with scores of 0.9-1.2 for DP 7-9 using the CAM-assay. An interaction with the fibroblast growth factor FGF-2 was noticed for DP 4-12 depending on the degree of sulfation using the FGF-2-trypsin assay.

Water-soluble hemicelluloses were extracted from milled aspen wood (Populus tremula) employing microwave oven treatment at 180 degrees C for 10 min. The final pH of this extract was 3.5. From this extract oligo- and polysaccharides were isolated and subsequently fractionated by size-exclusion chromatography. The structures of the saccharides in three of the fractions obtained were determined by 1H and 13C NMR spectroscopy, using homonuclear and heteronuclear two-dimensional techniques. The polysaccharides present in the two fractions eluted first were O-acetyl-(4-O-methylglucurono)xylans. The average degree of acetylation of the xylose residues in these compounds was 0.6. The structural element -->4)[4-O-Me-alpha-D-GlcpA-(1-->2)][3-O-Ac]-beta-D-Xylp-(1 --> could also be identified. On the average, these two xylans were composed of the following (1-->4)-linked beta-D-xylopyranosyl structural elements: unsubstituted (50 mol%), 2-O-acetylated (13 mol%), 3-O-acetylated (21 mol%), 2,3-di-O-acetylated (6 mol%) and [MeGlcA alpha-(1-->2)][3-O-acetylated] (10 mol%). Most of the 4-O-methylglucuronyl and acetyl substituents in the isolated polysaccharides survived the microwave oven treatment. The third fraction, eluted last, contained acetylated xylo-oligosaccharides, with minor contamination by an acetylated mannan. In the case of these xylo-oligosaccharides, the average degree of acetylation was 0.3.
