[Display omitted] The van der Waals interaction between colloids and nanoparticles is one of the key components to understanding particle aggregation, attachment, and assembly. While the ubiquity of anisotropic particle shapes and surface roughness is well-recognized in nanocrystalline materials, the effects of both on van der Waals forces and torques have not been adequately investigated. In this study, we develop a numerical scheme to determine the van der Waals forces and torques between cubic particles with multiple configurations and relative orientations. Our results show that the van der Waals torque due to anisotropic particle shapes is appreciable at nearly all configurations and mutual angles, outcompeting Brownian torque for various materials systems and conditions. Surface roughness enhances this particle shape effect, resulting in stronger van der Waals interactions ascribed to protrusions on the surfaces. Moreover, a scaling analysis indicates that the surface roughness alters the separation dependence of the van der Waals force and, more importantly, significantly influences the dynamics of two approaching particles. Our results clearly demonstrate that surface roughness and anisotropic shape play a crucial role in the energetics and kinetics of various particle-scale and emergent phenomena, such as crystal growth by oriented attachment, nanomaterials synthesis and assembly, mud flow rheology, as well as the deposition of natural nanocrystals within the subsurface. This paper investigates the self-assembly behavior of a new amphiphilic block copolymer, PPEGMA-PPC-PPEGMA, in dilute aqueous solution and at the air–water interface. In PPEGMA-PPC-PPEGMA, the hydrophilic PEG moieties exist as side chains attached to the PMA backbone. Because of this unique non-linear architecture, the morphological and conformational properties of self-assembled PPEGMA-PPC-PPEGMA polymers are expected to be different from those of conventional linear PEG-based polymer surfactants. For this study, three PPE G MA-PP C -PPE G MA samples having an identical PPC molecular weight (5.6 kDa) and different PPEGMA molecular weights (7.2 , 2.8 and 2.1 kDa on either side) (named " G7C6G7 ", " G3C5G3 ", and " G2C6G2 ", respectively) were synthesized. The micellar self-assembly behaviors of these materials were investigated by cryo-TEM, rheology, DLS, and visual observation. Langmuir monolayers of these materials were characterized by surface mechanical testing. PPEGMA-PPC-PPEGMA micelles were found to have a spherical geometry, irrespective of copolymer composition. Interestingly, G2C6G2 and G3C6G3 micelles formed weakly-bound clusters, whereas G7C6G7 micelles predominantly existed as isolated micelles. Detailed analysis suggests that this unexpected trend in micelle morphology originates from the fact that the PPEGMA blocks are only partially hydrated at aqueous interfaces. Detailed features of the surface pressure-area isotherms obtained from Langmuir PPEG-PPC-PPEGMA monolayers further supported this notion. [ABSTRACT FROM AUTHOR]

The development of high technology electrical devices increased the importance of higher power density or higher capacity at high current density. Especially, rapid charge/discharge issues remain problematic for electric vehicle commercialization. After extensive investigation, researchers introduced hetero-nanostructured materials to the field of lithium ion batteries (LIBs), aiming to enhance the power density or improve the capacity at high current density and life cycle capability. Hetero-nanostructured materials consist of current collectors and directly attached active nanomaterials. Carbon, carbon nanotube (CNT), graphene, Nickel (Ni), Copper (Cu) and Aluminum (Al) were used for current collector, aiming to improve the electron transfer and the cyclability, due to high electrical conductivity and superior buffering effects. Also, Hetero-nanostructure can produce a favorable lithium diffusion condition by creating a lithium diffusion pathway. This article presents an explanation of important factors for high power density or high capacity at high current density. Telomerase is a reverse transcriptase that adds repeats of a six-base DNA sequence to chromosome ends and thereby prevents their shortening during successive cell divisions. Telomerase activity and expression of the catalytic subunit of telomerase (TERT) are high in brain cells during embryonic development, but are undetectable in the adult. We now report that telomerase activity and expression of TERT are induced in the hippocampus of adult mice after administration of the seizure-inducing excitotoxin kainate. Telomerase activity is present at 24 h and 7 days after kainate administration, but is no longer detectable at 4 weeks. Because the time course of telomerase induction was similar to the time course of microglial activation, we performed studies to determine whether microglia were the source of the telomerase activity. Examination of brain sections immunostained with a TERT antibody and an antibody against a microglia-specific antigen revealed that TERT was not detectable in the uninjured brain, and was present in microglia 24 h and 7 days after kainate administration. This is the first evidence that telomerase can be induced by brain injury and the first evidence that TERT can be expressed in microglia, suggesting roles for telomerase in microglial responses to brain injury. Phenformin is a biguanide compound that can modulate glucose metabolism and promote weight loss and is therefore used to treat patients with type-2 diabetes. While phenformin may indirectly affect neurons by changing peripheral energy metabolism, the possibility that it directly affects neurons has not been examined. We now report that phenformin suppresses responses of hippocampal neurons to glutamate and decreases their vulnerability to excitotoxicity. Pretreatment of embryonic rat hippocampal cell cultures with phenformin protected neurons against glutamate-induced death, which was correlated with reduced calcium responses to glutamate. Immunoblot analyses showed that levels of the N-methyl-d-aspartate (NMDA) subunits NR1 and NR2A were significantly decreased in neurons exposed to phenformin, whereas levels of the AMPA receptor subunit GluR1 were unchanged. Whole-cell patch clamp analyses revealed that NMDA-induced currents were decreased, and AMPA-induced currents were unchanged in neurons pretreated with phenformin. Our data demonstrate that phenformin can protect neurons against excitotoxicity by differentially modulating levels of NMDA receptor subunits in a manner that decreases glutamate-induced calcium influx. These findings show that phenformin can modulate neuronal responses to glutamate, and suggest possible use of phenformin and related compounds in the prevention and/or treatment of neurodegenerative conditions. Photoelectrochemical water splitting is the most efficient green engineering approach to convert the sun light into hydrogen energy. The formation of high surface area core-shell heterojunction with enhanced light-harvesting efficiency, elevated charge separation, and transport are key parameters in achieving the ideal water splitting performance of the photoanode. Herein, we demonstrate a first green engineering interfacial growth of the BiVO 4 nanoparticles onto self-assembled WO 3 nanoplates forming WO 3 /BiVO 4 core-shell heterojunction for efficient PEC water splitting performance. The three different WO 3 nanostructures (nanoplates, nanobricks, and stacked nanosheets) were self-assembled on fluorine doped tin oxide glass substrates via hydrothermal route at various pH (0.8–1.2) of the solutions. In comparison to nanobricks and stacked nanosheets, WO 3 nanoplates displayed considerably elevated photocurrent density. Moreover, a simple and low cost green approach of modified chemical bath deposition technique was established for the optimal decoration of a BiVO 4 nanoparticles on vertically aligned WO 3 nanoplates. The boosted photoelectrochemical current density of 1.7 mA cm−2 at 1.23 V vs. reversible hydrogen electrode (RHE) under AM 1.5 G illumination was achieved for the WO 3 /BiVO 4 heterojunction which can be attributed to a suitable band alignment for the efficient charge transfer from BiVO 4 to WO 3 , increased light harvesting capability of outer BiVO 4 layer, and high charge transfer efficiency of WO 3 nanoplates. We developed a new strategy for construction of a biosensor for the neurotransmitter dopamine. The biosensor was constructed by one-step electrochemical deposition of a nanocomposite in aqueous solution at pH 7.0, consisting of molybdenum disulfide, multi-walled carbon nanotubes, and polypyrrole. A series of analytical methods was performed to investigate the surface characteristics and the improved electrocatalytic effect of the nanocomposite, including cyclic voltammetry, electrochemical impedance spectroscopy, field-emission scanning electron microscopy, atomic force microscopy, and Raman spectroscopy. The constructed biosensor showed high sensitivity (1.130 μAμM −1 cm −2 ) with a dynamic linearity range of 25–1000 nM and a detection limit of 10 nM. Additionally, the designed sensor exhibited strong anti-interference ability and satisfactory reproducibility. The practical application of the sensor was manifested for the ex vivo determination of dopamine neurotransmitters using brain tissue samples of a mouse Parkinson's disease model. Background Peroxisome proliferator-activated receptor (PPAR)-δ is a nuclear receptor regulating cell metabolism. The role of PPAR-δ in late endothelial progenitor cells (EPCs) has not been fully elucidated. We aim to understand the effects of PPAR-δ activation on late EPC and to reveal the underlying mechanism. Methods and results Treatment with a highly selective PPAR-δ agonist (GW501516) induced proliferation of late EPCs and enhanced their vasculogenic potential. Search for the target molecule of PPAR-δ activation revealed endothelial differentiation gene (Edg)-2. Chromatin immunoprecipitation and promoter assays demonstrated that Edg-2 gene was specifically induced by PPAR-δ through direct transcriptional activation. Lysophosphatidic acid (LPA), an Edg ligand, mimicked the pro-vasculogenic effects of GW501516 in late EPCs whereas Edg antagonist (Ki16425) blocked these effects. Edg-2 is a membrane receptor for LPA which is a major growth factor from activated platelets. Thus, the interaction between platelets and late EPCs via the LPA-Edg-2 axis was assessed. Platelet supernatant boosted the pro-vasculogenic effects of GW501516, which was reversed by antagonist to PPAR-δ (GSK0660) or Edg (Ki16425). Both of in vivo Matrigel plug model and mouse skin punch-wound model demonstrated that the combination of platelets and PPAR-δ-activated late EPCs synergistically enhanced vascular regeneration. Conclusions There exists a synergistic interaction between human platelets and late EPCs leading to vascular regeneration. This interaction consists of LPA from platelets and its receptor Edg-2 on the surface of EPCs and can be potentiated by PPAR-δ activation in EPCs. Platelets not only play a role in hemostasis, but they also promote angiogenesis and tissue recovery by releasing various cytokines and making an angiogenic milieu. Here, we examined autologous ‘activated platelet supernatant (APS)’ as a priming agent for stem cells; thereby enhance their pro-angiogenic potential and efficacy of stem cell-based therapy for ischemic diseases. The mobilized peripheral blood stem cells ( mob PBSCs) were isolated from healthy volunteers after subcutaneous injection of granulocyte-colony stimulating factor. APS was collected separately from the platelet rich plasma after activation by thrombin. mob PBSCs were primed for 6 h before analysis. Compared to naive platelet supernatants, APS had a higher level of various cytokines, such as IL8, IL17, PDGF and VEGF. APS-priming for 6 h induced mob PBSCs to express key angiogenic factors, surface markers (i.e. CD34, CD31, and CXCR4) and integrins (integrins α5, β1 and β2). Also mob PBSCs were polarized toward CD14(++)/CD16(+) pro-angiogenic monocytes. The priming effect was reproduced by an in vitro reconstruction of APS. Through this phenotype, APS-priming increased cell–cell adhesion and cell–extracellular matrix adhesion. The culture supernatant of APS-primed mob PBSCs contained high levels of IL8, IL10, IL17 and TNFα, and augmented proliferation and capillary network formation of human umbilical vein endothelial cells. In vivo transplantation of APS-primed mob PBSCs into athymic mice ischemic hindlimbs and Matrigel plugs elicited vessel differentiation and tissue repair. In safety analysis, platelet activity increased after mixing with mob PBSCs regardless of priming, which was normalized by aspirin treatment. Collectively, our data identify that APS-priming can enhance the angiogenic potential of mob PBSCs, which can be used as an adjunctive strategy to improve the efficacy of cell therapy for ischemic diseases. Abstract: Alzheimer''s disease (AD) is the major form of age-related dementia and is characterized by progressive cognitive impairment, the accumulation of extracellular amyloid β-peptide (Aβ), and intracellular hyperphosphorylated tau aggregates in affected brain regions. Tau hyperphosphorylation and accumulation in neurofibrillary tangles is strongly correlated with cognitive deficits, and is apparently a critical event in the dementia process because mutations in tau can cause a tangle-only form of dementia called frontotemporal lobe dementia. Among kinases that phosphorylate tau, glycogen synthase kinase 3β (GSK3β) is strongly implicated in AD pathogenesis. In the present study, we established an ELISA to screen for agents that inhibit GSK3β activity and found that the flavonoid morin effectively inhibited GSK3β activity and blocked GSK3β-induced tau phosphorylation in vitro. In addition, morin attenuated Aβ-induced tau phosphorylation and protected human neuroblastoma cells against Aβ cytotoxicity. Furthermore, treatment of 3xTg-AD mice with morin resulted in reductions in tau hyperphosphorylation and paired helical filament-like immunoreactivity in hippocampal neurons. Morin is a novel inhibitor of GSK3β that can reduce tau pathology in vivo and may have potential as a therapeutic agent in tauopathies. [Copyright &y& Elsevier]

Abstract: Wnt/β-catenin signaling controls a variety of cellular processes, including cell growth, oncogenesis, and development. Upon Wnt stimulation, the intracellular region of the coreceptor, LRP6 or 5, is phosphorylated by the membrane-recruited GSK3β and CK1. The cytoplasmic domain of LRP6/5 contains one Ser/Thr cluster and the PPPSP motifs, both of which are essential for propagation of the signal. While the phosphorylated PPPSP motifs are known to directly inhibit GSK3β, the biochemical role of the phosphorylated Ser/Thr cluster remains to be elucidated. Herein, we reveal that the Ser/Thr cluster plays an important role in the phosphorylation of the PPPSP motif. Interestingly, we observe that GSK3β activity on the PPPSP motif requires a high ATP concentration, close to that of the physiological condition. Taken together, these data suggest that the phosphorylated Ser/Thr cluster serves as a docking site for GSK3β to promote the phosphorylation of the PPPSP motif. Our results provide insight into the molecular mechanism for the initial events of the Wnt/β-catenin signaling. [Copyright &y& Elsevier]

Abstract: Vitamin C (VC) has a strong antioxidant function evident as its ability to scavenge superoxide radicals in vitro. We verified that this property actually exists in vivo by using a real-time imaging system in which Lucigenin is the chemiluminescent probe for detecting superoxide in senescence marker protein-30 (SMP30)/gluconolactonase (GNL) knockout (KO) mice, which cannot synthesize VC in vivo. SMP30/GNL KO mice were given 1.5g/L VC [VC(+)] for 2, 4, or 8 weeks or denied VC [VC(−)]. At 4 and 8 weeks, VC levels in brains from VC(−) KO mice were <6% of that in VC(+) KO mice. Accordingly, superoxide-dependent chemiluminescence levels determined by ischemia-reperfusion at the 4- and 8 weeks test intervals were 3.0-fold and 2.1-fold higher, respectively, in VC(−) KO mice than in VC(+) KO mice. However, total superoxide dismutase activity and protein levels were not altered. Thus, VC depletion specifically increased superoxide generation in a model of the living brain. Abstract: The transition from chemotherapy-responsive cancer cells to chemotherapy-resistant cancer cells is mainly accompanied by the increased expression of multi-drug resistance 1 (MDR1). We found that hepatitis-B-virus X protein (HBx) increases the transcriptional activity and protein level of MDR1 in a hepatoma cell line, H4IIE. In addition, HBx overexpression made H4IIE cells more resistant to verapamil-uptake. HBx stabilized hypoxia-inducible factor-1α (HIF-1α) and induced the nuclear translocation of C/EBPβ. Reporter gene analyses showed that HBx increased the reporter activity in the cells transfected with the reporter containing MDR1 gene promoter. Moreover, the luciferase reporter gene activity was significantly inhibited by HIF-1α siRNA but not by overexpression of C/EBP dominant negative mutant. These results imply that HBx increases the MDR1 transporter activity through the transcriptional activation of the MDR1 gene with HIF-1α activation, and suggest HIF-1α for the therapeutic target of HBV-mediated chemoresistance.
