Following the ammonia treatment, the pores created by the folds allow for electrolyte storage and facilitate ion transport. The interlayer spacing between MXene nanosheets widens due to the volume increase when water freezes. The generous ion transport paths and redox-active ammonium ions offer higher specific capacity and faster reaction kinetics. However, the narrow interlayer spacing and self-stacking phenomenon limit its applicability. In this study, we demonstrate an easy two-step method involving freezing and crumpling of MXene nanosheets to improve their Na-ion storage via the addition of ammonium ions (referred to as FCM nanosheets). Flat MXene particles aggregate and undergo folding in an alkaline solution. Ammonium ions can penetrate the gaps between MXene nanosheets, expanding interlayer spaces and inducing the formation of folds. Compared to MXene nanosheets, FCM nanosheets exhibit improved ion transfer kinetics and additional high capacity owing to the intercalated ammonium ions. [Display omitted] Hydrogen peroxide (H 2 O 2) emerges as a viable oxidant for fuel cells, necessitating the development of an efficient and cost-effective electrocatalyst for the hydrogen peroxide reduction reaction (HPRR). In this study, we synthesized a self-supporting, highly active HPRR electrocatalyst comprising two morphologically distinct components: CeO 2 -NiCo 2 O 4 nanowires and CeO 2 -NiCo 2 O 4 metal organic framework derivatives, via a two-step hydrothermal process followed by air calcination. X-ray diffraction and transmission electron microscopy analysis confirmed the presence of CeO 2 and NiCo 2 O 4 , revealing the amalgamated interface between them. CeO 2 exhibits multifunctionality in regulating the surface electronic configuration of NiCo 2 O 4 , fostering synergistic connections, and introducing oxygen deficiencies to enhance the catalytic efficacy in HPRR. Electrochemical measurements demonstrate a reduction current density of 789.9 mA·cm−2 at −0.8 V vs. Ag/AgCl. The assembly of direct borohydride-hydrogen peroxide fuel cell (DBHPFC) exhibits a peak power density of 45.2 mW·cm−2, demonstrating durable stability over a continuous operation period of 120 h. This investigation providing evidence that the fabrication of heterostructured catalysts based on CeO 2 for HPRR is a viable approach for the development of high-efficiency electrocatalysts in fuel cell technology. Abstract: 2A Peptide sequences are now being widely used to construct multicistronic expression vectors. It is suggested that when only the first 2A-linked protein bears a signal sequence, the signal-less protein(s) downstream of 2A can also be translocated into the mammalian endoplasmic reticulum system through a “slipstreaming” mechanism. By using flow cytometry and cell surface CD90 as a localization indicator, we show here that slipstreaming translocation does not occur in mammalian cells; that is, the second protein downstream of 2A still requires signal sequence for secretary or membrane-anchored expression. Tributyltin (TBT), a widely and persistently distributed organontin, has been well documented to disrupt reproduction and behaviors in animals due to its anti-aromatase activity. TBT has been also reported to enhance anxiety in several fish species, whereas the mechanism underlying remains largely unknown. To investigate the disruption of TBT on fish anxiety and the mechanisms possibly involved, adult male zebrafish (Danio rerio) were treated with TBT (100 and 500 ng/L) for 28 days and anxiety behavior was further investigated using a novel tank dive test. Result showed that TBT treatment significantly enhanced the total time of the fish spent in the lower half, delayed the onset time to the higher half of the tank and increased the total duration of freezing of the fish, indicating an enhanced anxiety in TBT-treated fish. Accordingly, TBT sharply elevated the cortisol levels in plasma in a concentration-dependent manner, suggesting that the elevated cortisol level might be involved in the enhanced anxiety. Although the expression of crha was significantly increased and crhbp was significantly decreased in the brain of TBT-treated fish which is consistent to the elevated cortisol level, the expressions of actha and acthb were sharply down-regulated. In contrast, the expressions of genes responsible for the synthesis and action of serotonin (5-HT) (pet1 , thp2 and htr1aa), dopamine (DA) (th1 , slc6a3 , drd2a and drd2b) and gamma-aminobutyric acid (GABA) (gad2 and gabrg2) were all significantly inhibited. The down-regulation of these pivotal genes acting in 5-HT, DA and GABA neurotransmitter systems in response to TBT corresponded well with the TBT-enhanced anxiety in fish. It was thus strongly suggested that these neurotransmitters might be also involved in TBT-enhanced anxiety in adult male zebrafish. As an important task in digital preventive healthcare management, especially in the secondary prevention stage, active medication stocking refers to the process of preparing necessary medications in advance according to the predicted disease progression of patients. However, predicting preventive or even life-saving medicine for each patient is a non-trivial task. Existing models usually overlook the implicit hierarchical relation between patient's predicted diseases and medications, and mainly focus on single tasks (medication recommendation or disease prediction). To tackle this limitation, we propose a relation augmented hierarchical multi-task learning framework, named RAHM. which is capable of learning multi-level relation-aware patient representation for reasonable medication stocking. Specifically, the framework first leverages the underlying structural relations of Electronic Health Record (EHR) data to learn the low-level patient visit representation. Then, it uses a regular LSTM to encode the historical temporal disease information for disease-level patient representation learning. Further, a relation-aware LSTM (R-LSTM) is proposed to handle the relations between diseases and medication in longitudinal patient records, which can better integrate the historical information into the medication-level patient representation. In the learning process, two pseudo residual structures are introduced to mitigate the error propagation and preserve the valuable relation information of EHRs. To validate our method, extensive experiments have been conducted based on the real-world clinical dataset. [Display omitted] In this paper, we have developed a simple and efficient sulfur-amine chemistry strategy to prepare a three-dimensional (3D) porous Ti 3 C 2 T x composite with large amounts of N and S terminal groups. The well-designed 3D macroporous architecture presents enlarged interlayer spacing, large specific surface area, and unique porous structure, which successfully solves the re-stacking issue of MXene during storage and electrode fabrication. It is the amount of concentrated hydrochloric acid added to the S-EDA (ethylenediamine)/MXene colloidal suspension that is critical to the formation of 3D morphology. In addition, N and S terminals on MXene could improve the adsorption ability of K+. Owing to the synergistic effect of the structure design and terminal modification, the N, S codoped three-dimensional porous Ti 3 C 2 T x (3D-NSPM) material shows a high surface capacitive contribution and rapid diffusion kinetics for K+ and Na+. As a result, the as-prepared 3D-NSPM delivers high reversible capacity (237 and 273 mAh g−1 at 0.1 A g−1 for PIBs and SIBs, respectively), superb cycling stability (84.9% capacity retention after 10,000 cycles at 1 A g−1 in PIBs and 74.0% capacity retention after 2200 cycles at 1 A g−1 in SIBs), and excellent rate capability (111 and 196 mAh g−1 at 5 A g−1 for PIBs and SIBs, respectively), which are superior to other MXene-based anodes for PIBs and SIBs. Multi-omics strategy contributes as an indispensable and efficient approach for the investigation of the innate immunity in vertebrates. To explore the crucial genes and pathways involved in the antiviral innate immunity of black carp (Mylopharyngodon piceus), the comparative phosphoproteomics and transcriptomics of Mylopharyngodon piceus kidney (MPK) cells with/without GCRV infection were performed in this manuscript. In phosphoproteomics analysis, 2637 phosphosites corresponding to 1532 proteins were identified and quantified, in which 1372 proteins were identified as differentially expressed proteins (DEPs) with 683 upregulated and 689 downregulated in GCRV infected cells. Functional annotation, enrichment analysis and pathway analysis highlighted that a large number of DEPs were enriched in immune related pathways including TLR pathway and NLR pathway. In transcriptomics analysis, a total of 2936 genes were identified as differentially expressed genes (DEGs), in which 2290 and 646 genes were upregulated and downregulated respectively after GCRV infection. As expected, pathway analysis based on DEGs also showed that a large proportion of DEGs were enriched in immune related pathways including TLR and RLR pathway. A combined list of DEPs and DEGs that enriched in above pathways were imported in Cytoscape for network analysis, reconstruction and visualization. The integrative study suggested that several significant DEPs and DEGs, such as MAP3K7 (TAK1), JUN, MAP2K2, CASP8, IL8 and IRF7 might be functionally crucial in host antiviral innate immunity. Based on a fixed point argument, we give a dynamical representation of the viscosity solution to Cauchy problem of certain weakly coupled systems of Hamilton-Jacobi equations with continuous initial datum. Using this formula, we obtain some regularity results related to the viscosity solution, including a partial extension of Lions' regularizing effect [17] to the case of weakly coupled systems. Also, the solubility of Disperse Red 11 in SC-CO 2 was obtained at the temperature of 343.15 K and pressure ranges of 14 MPa to 22 MPa and at the pressure of 14 MPa and temperatures of 343.15 K to 373.15 K, for an extension of the solubility results previously published. The neural network ANN formulated the experimental data in terms of temperature and pressure as input layers, solubility as the output layer, and the number of neurons in the hidden layer. The calculation results show that five different kinds of dyestuff solubility data at the temperature ranges of 343.15 K to 373.15 K, pressures of 14 MPa to 22 MPa as well as the solubility data of Disperse Red 4 and Disperse Red 15 taken from the previous work could be estimated accurately by BP neural network with a fitting level of 0.999. Moreover, the solubility data were correlated with eight different kinds of empirical models proposed previously and compared with those calculated by the ANN. The direct urea fuel cell (DUFC) is a power generation equipment with urea-rich wastewater or urine as fuel source. It has the unique ability to purify sewage while simultaneously generating electricity, making it a highly efficient and environmentally friendly option. In this paper, pomegranate seed-like Ni nano-blocks and Fe nanosheets were synthesized by electrodeposition and chemical reduction and attached to the carbonized melamine sponge matrix. The N -doped carbon sponge (NCS) provided a large number of polyhedral holes, which allowed for efficient gas escape through channels. The combination of Fe reduces the initial urea oxidation potential, reaction activation energy and reaction resistance. The synthesized FeNi supported on N -doped carbon sponge composite (FeNi@NCS) has a catalytic current density of 625 mA cm−2 and a Tafel slope of 42.51 mV dec−1 for urea electrooxidation reaction (UOR). Assembling the direct urine-hydrogen peroxide fuel cell (DUrHPFC) resulted in the highest performance output. The open circuit voltage (OCV) was 0.98 V, and the peak power density reached 9.61 mW cm−2. [Display omitted] The emerging aqueous Zn-ion batteries (ZIBs) with their low price and inherent safety are showing strong competitiveness in the energy storage field. In this work, layer-by-layer stacked amorphous V 2 O 5 @Ti 3 C 2 T x heterostructures anchored on three-dimensional carbon cloth (3D CC), synthesized by annealing process and subsequent in-situ electrochemical induction, are reported as high capacity and stable cathode for ZIBs. In this composite cathode, amorphous V 2 O 5 @Ti 3 C 2 T x synergistic heterostructure provides isotropic Zn2+ migration channels and rapid electron transfer kinetics, while the 3D CC substrate can ensure that the bulk phase in the electrode material is also utilized to maximize the synergistic effect. As a result, the CC/a-V 2 O 5 @Ti 3 C 2 Ti x cathode achieves a high specific capacity of 567 mAh/g at 0.1 A/g, a satisfactory rate performance of 213 mAh/g at 10 A/g, together with a long cycle life of 96.2 % capacity retention after 2000 cycles. Ex-situ characterizations show that the CC/a-V 2 O 5 @Ti 3 C 2 T x cathode undergoes a highly reversible Zn2+/H+ co-intercalation/deintercalation mechanism during the discharge/charging process. In addition, based on the CC/a-V 2 O 5 @Ti 3 C 2 T x cathode, the assembled flexible ZIB can operate properly under different bending degrees, showing its practicality in flexible devices. A novel chymotrypsin inhibitor, named ClCI, was purified from coix seed (Coix lacryma-jobi L.) by aqueous two-phase extraction, chymotrypsin–Sepharose 4B affinity chromatography and centrifugal ultrafiltration. ClCI was a 7.9 kDa competitive inhibitor with pI 6.54. The inhibition constants (K i) for bovine pancreatic chymotrypsin and bacterial subtilisin were 1.27 × 10−10 M and 1.57 × 10−9 M respectively. ClCI had no inhibitory activity against bovine trypsin and porcine elastase. ClCI had wide pH stability and good heat resistance. It can maintain >90% inhibition activity against chymotrypsin at 20–80 °C for 1 h. The primary structure of ClCI was highly similar (57%–92%) to those of several inhibitors belonging to the Gramineae crop potato protease inhibitor- I superfamily and showed the typical sequence motif of the protease inhibitor of the seed storage protein group. ClCI (12.5 mg) inhibited mycelial growth of the phytopathogenic fungi Mycosphaerella melonis, Helminthosporium turcicum , Alternaria solani , Phytophthora capsici, Isariopsis griseola, and Colletotrichum gloeosporioides , and caused 89% inhibition of the proteases from spore germination of plant-pathogenic fungi. The results of the present study indicate that ClCI had biotechnological potential as an alternative agent to combat the important phytopathogenic fungi. Spinal cord injury (SCI) is terrible damage leading to the deficiencies and results in infinite inconvenience to sufferers. The effective treatment for SCI still meets a larger number of problems. Herein, the underlying molecular mechanism and novel therapy of SCI are urgently to investigate. Arachidonate 12-lipoxygenase (ALOX12) is widely expressed in various cell types and plays important role in modulating different cellular processes, such as platelet aggregation, cell migration and cancer cell proliferation. Nevertheless, the effects of ALOX12 on SCI are unclear. In the study, SCI model was established in wild type (WT) mice and ALOX12 knockout mice. First, ALOX12 expression was up-regulated in spinal cord tissues of WT mice after SCI. ALOX12-knockout mice exhibited improved behavior after SCI operation. Glial activation triggered by SCI was also alleviated in mice with the loss of ALOX12, as evidenced by the down-regulated expression of glial fibrillary acidic protein (GFAP) and Iba-1 in spinal cord samples. Further, SCI-induced inflammation was markedly prevented in ALOX12-knockout mice through blocking inhibitor of NF-κB α (IκBα)/nuclear factor-κB (NF-κB) pathway signaling. Additionally, reducing ALOX12 expression attenuated apoptosis in spinal cord tissues of SCI mice by decreasing Cyto-c, cleaved Caspase-3 and poly (ADP-ribose) polymerases (PARP) expression. The protective role of ALOX12-decrease against SCI was verified in LPS-incubated glial cells through repressing inflammatory response and apoptotic formation. Moreover, transgenic mice with ALOX12 over-expression showed accelerated SCI, associated with intensified inflammation and apoptosis. The N, S dual-doped porous graphitic carbon materials with the high graphitization degree were synthesized from the light fraction of coal tar pitch and waste sponge through a facile "killing two birds with one stone" process. Emerging aqueous zinc–ion hybrid capacitors (AZICs) are considered a promising energy storage because of their superior electrochemical performance. The pore structure, suitable heteroatom content, and graphitization degree (GD) of carbon-based cathodes significantly influence the electrochemical performance of AZICs. The N, S dual-doped porous graphitic carbon materials (LC-750) with the combined characteristics of high GD (1.11) and large specific surface area (1678.38 m2 g−1) are successfully developed by a facile "killing two birds with one stone" strategy using K 3 Fe(C 2 O 4) 3 ·3H 2 O as the activating and graphitizing agent, and waste sponge (WS) and coal tar pitch (CTP) as the heteroatom and carbon resource, respectively. Results show that the LC-750 cathode displays high capacities of 185.3 and 95.2 mAh g−1 at 0.2 and 10 A g−1. Specifically, the assembled LC-750//Zn capacitor can offer a maximal energy density of 119.5 Wh kg−1, a power density of 20.3 kW kg−1, and a capacity retention of 87.8% after 15,000 cycles at 10 A g−1. Density functional theory simulations demonstrate that N and S dual-doping can promote the adsorption kinetics of Zn ions. This design strategy is a feasible and cost-effective method for the preparation of dual heteroatom-doped graphitic carbon electrodes, which enables recycling of WS and CTP into high-valued products. The study was designed to elucidate roles of PKCepsilon, ERK1/2, STAT3 in regulations of endogenous H2S in rat hepatocyte apoptosis induced by LPS. The cultured rat hepatocytes were treated with vehicle or different dose of LPS respectively. In the separate experiments, hepatocytes were treated with CBS inhibitor AOAA,CSE inhibitor PAG or PKC epsilon, ERK1/2, STAT3 specific inhibitors SCP0213, A6355, S31-201,respectively or in a different combination manner. PKC epsilon, ERK1/2 and STAT3 phospharylations, PKCepsilon membranous translocation, STAT3 nuclear translocation were detected with Western blotting. Apoptosis rates were detected with flow cytometry. PKC epsilon, ERK1/2 and STAT3 phosphorylation expression in LPS-treated hepatocytes were significantly up-regulated, obviously in 30min compared with vehicle,furthermore membranous tranlocation of PKCepsilon and nuclear tranlocation of STAT3 were found. Compared with LPS, AOAA or PAG combined with LPS respectively or jointly could cause further more obvious up-regulation in phosphorylative levels of PKCepsilon, ERK1/2 or STAT3 protein expression. SCP0213, A6355 or S31-201 combined with LPS respectively could cause increased apoptosis rates in 4h and 12h compared with LPS alone. When hepatocytes were treated with SCP0213, A6355 or S31-201 combined with AOAA or PAG respectively, apoptosis rates increased in 12h but no significant changes in 4h compared with AOAA or PAG+LPS treatments.Compared with LPS,SCP0213+LPS could cause down-regulation in phosphorylative levels of ERK1/2 or STAT3 protein expressions, A6355+LPS could cause down-regulation in the phosphorylative level of STAT3 protein expression without change in PKCepsilon, S31–201+LPS could not cause any changes of phosphorylative level of protein expression in PKCepsilon or ERK1/2. A broad range of cytokines are secreted during the inflammatory response by the immune system. Some cytokines promote inflammation, while others inhibit inflammation. Inflammatory cytokines work in harmony when they encounter external pathogens or internal dangers. Inflammation is resolved after the cause is eliminated. However, if the cause persists, it can lead to significant diseases. The pro-inflammatory cytokine TNFα is a biomarker for the inflammatory response. The AO herbal mixture extracted from 10 medicinal herbs has been investigated for its ability to control the inflammatory process and to inhibit TNFα activity. To find the treatment for inflammation related diseases, we examined whether the AO herbal extract is able to affect the activities of other cytokines. Here we present that the AO herbal extract is able to inhibit pro-inflammatory factor activities including IL-1α. However, it does not affect the activities of IL-1β and IL-6. Herein, we cleverly combined electrostatic self-assembly with gas-phase vulcanization method to successfully combine Ti 3 C 2 T x -MXene with ultra-long recyclability and high conductivity with MnS, which presents high specific capacity but poor conductivity. The as-prepared 3D hierarchical Ti 3 C 2 T x /MnS composites have an unique sandwich-like constituent units. The tiny MnS nanoparticles are restricted between the Ti 3 C 2 T x layers and play a key role in expanding the Ti 3 C 2 T x interlayer spacing. As a result, the 3D Ti 3 C 2 T x /MnS composites as the anode of LIBs exhibits a superior capacities of 826 and 634 mAh/g after 1000 and 3000 cycles at 0.5 and 1.0 A/g, respectively. More importantly, we reveal the reaction mechanism that the specific capacity first increases and then gradually stabilizes with the increase of charge and discharge cycle times when the as-prepared 3D Ti 3 C 2 T x /MnS was used as the anode of LIBs. In addition, we have also used this material in SIBs and PIBs and achieved remarkable electrochemical capability, with a specific capacity of 107 mAh/g after 2500 cycles at 0.5 A/g or 127 mAh/g after the 2000th cycle at 0.2 A/g, respectively. Multiple lines of evidences have unraveled the emerging role of ferroptosis in the pathophysiological process of acute lung injury (ALI). In this study, we aimed to decipher the role of BACH1 in the onset and progression of ALI with a focus on ferroptosis and elucidated potential molecular mechanism. We observed that BACH1 expression was drastically elevated in BEAS-2B cells upon exposure to LPS. In the functional aspect, BACH1 deletion exerted an anti-inflammatory property, featured by decreased the secretion of several cytokines including TNF-α, IL-1β and IL-6 in the face of LPS challenge. What's more important, BACH1 knockout evidently repressed LPS-triggered oxidative stress damage, as evidenced by reduced reactive oxygen species (ROS) production and malondialdehyde (MDA) generation, accompanied with the elevated the activities of superoxide dismutase (SOD), GSH-Px and CAT. Meanwhile, ablation of BACH1 restrained LPS-elicited ferroptosis, as characterized by decreased iron content and PTGS2 expression, accompanied with increased expression of SLC7A11 and GPX4. In terms of mechanism, Nrf2/HO-1 signaling inhibitor effectively abrogated the beneficial effects of BACH1 inhibition on LPS-stimulated inflammation, oxidative damage and ferroptosis. MoSSe nanosheets anchored carbon tubes (CTs) were prepared with facile hydrothermal method and further selenized carbonization treatment. The hollow tubular carbon skeletons with abundant functional groups can regulate the uniform growth of MoS 2 clusters. In the subsequent step, the selenation treatment can extend the interlayer spacing of MoS 2 and form chemical bond (Mo N C) between the interface of MoSSe and CTs, which can effectively facilitate the transport of electron/ions. [Display omitted] Polymer materials containing C 6 rings and C O become promising electrode materials for high-performance lithium ion batteries (LIBs). However, the poor electronic conductivity severely restricts its further application. Herein, we design and construct a pyromellitic dianhydride anhydride anthraquinone/reduced graphene oxides (PMAQ/rGO-40) composite as an anode material for LIBs. The PMAQ is uniformly wrapped by conductive rGO nanosheets. The PMAQ/rGO-40 electrode without additional conductive agents displays a discharge capacity of 253 mAh g−1 over 3000 cycles under 2A g−1, which is higher than that of the PMAQ electrode with conductive agents. Meanwhile, a capacity of 196 mAh g−1 is achieved under 5A g−1. The enhanced cycling performance and rate ability are attributed to the rGO conductive network, which promotes electronic transport capability. In addition, the lithium ion storage mechanism and kinetics in the PMAQ/rGO-40 are investigated. [Display omitted] Rechargeable metal batteries such as non-aqueous Li metal batteries and aqueous Zn metal batteries have received increasing attention due to the benefits of the high-energy metal anode. However, the dendric growth on the metal anode extremely leads to restricted cycling performance and potential safety issues. Herein, the MXene-modified conductive framework (MCF) is designed and fabricated as a universal current collector of anode for non-aqueous Li metal batteries and aqueous Zn metal batteries to suppress the dendric Li and Zn growth. The MCF exhibits the merits of good lithiophilicity and zincophilicity, enables an easy Li/Zn nucleation process, and possesses high plating/stripping reversibility. After plating Li/Zn, MCF@Li and MCF@Zn realize a dendrite-free surface, facilitating Li/Zn plating/stripping kinetics. The MCF@Li and MCF@Zn symmetric cell shows a low overpotential of 13 mV at 1 mA cm−2 after 900 cycles and 64 mV at 5 mA cm−2 after 750 cycles (1 mAh cm−2), respectively. Furthermore, the non-aqueous MCF@Li||LiFePO 4 full cells and the aqueous MCF@Zn||active carbon capacitors are assembled and present stable cycling ability and improved rate performance with low polarization. Background Vascular hyporeactivity in severe hemorrhagic shock could induce refractory hypotension and is an important cause of death. The global acute inflammatory response induced in shock triggers the over-expression of reactive oxygen species, NO, ET1 and TNF-α, which play essential roles in the pathology of vascular hyporeactivity. This leads to a hypothesis that inhibition of the complement system, the mediator of the inflammatory cascade, might be a promising therapeutic exploration for vascular hyporeactivity. Methods We use cobra venom factor (CVF) and the soluble form of CR1 (sCR1) which deplete or inhibit complement C3 respectively to examine its role in vascular hyporeactivity in a conscious hemorrhagic shock rat model. Results We first confirmed the over-activation of C3 during shock and the down-regulation effects of CVF and sCR1 on C3. Then, both CVF and sCR1 could significantly mitigate the over-expression of serum NO, ET-1, TNF-α and reactive oxygen species. Finally, the vascular reactivity of superior mesenteric arteries (SMA) was examined in vitro, which confirmed the massive reduction of vascular reactivity in shock, which was significantly rescued by both CVF and sCR1. Conclusions Inhibition of C3 might improve the reactivity of SMA to norepinephrine during hemorrhagic shock possibly through the downregulation of NO, ET1, TNF-α and reactive oxygen radicals. Eighteen new oleanane-type triterpenoids were isolated from the stems of Sabia limoniacea , including sabialimon A (1), a triterpenoid with an unprecedented 6/6/6/7/7 pentacyclic skeleton and seventeen undescribed triterpenoids, sabialimons B−R (2 − 18), along with six previously described analogs (19 − 24). Their structures were fully elucidated via extensive spectroscopic analysis including 1D and 2D NMR, high-resolution electrospray ionization mass spectrometry (HRESIMS), experimental electronic circular dichroism measurements and X-ray crystallographic studies. Compound 1 is the first triterpenoid that possesses a rare ring system (6/6/6/7/7) with an oxygen-bearing bridge between C-17 and C-18 and a hemiketal form at C-17, which is generated a larger ring by the degradation of C-28 and D/E-ring expansion. Biological evaluation revealed that sabialimon I (9), sabialimon K (11), sabialimon P (16) and 11,13(18)-oleanadien-28-hydroxymethyl 3-one (20) exhibited significantly inhibitory activities against nitric oxide (NO) release with IC 50 values of 29.65, 23.41, 18.12 and 26.64 μM, respectively, as compared with the positive control (dexamethasone, IC 50 value: 40.35 μM). [Display omitted] The solar-driven interfacial evaporation (SDIE) is now a promising way to solve the shortage of fresh water. However, high performance SDIE for the oil contaminated seawater remains challenging. Here, we propose a facile "chitosan assisted MXene decoration" strategy to prepare a superhydrophilic and underwater superoleophobic Chitosan/MXene/fabric (CMF) for highly efficient SDIE. Benefiting from the superhydrophilicity and excellent photo-thermal conversion performance, the CMF is served as both the solar absorber and the water transportation path. Under the light illumination with one sun intensity (1 kW·m−2), a high evaporation rate of 1.50 kg·m−2·h−1 and efficiency of 88.05% are achieved. The strong interfacial interaction and outstanding salt rejection behavior of the CMF ensure the SDIE long-term stability and durability. In addition, the underwater superoleophobic CMF can effectively and quickly repel different oils and is hence suitable for high performance SDIE of the oil-in-water emulsion and the crude oil contaminated seawater. All three classes of receptors for the inhibitory neurotransmitter GABA (GABAR) are expressed in the retina. This study investigated roles of GABAR, especially GABA C R (GABA(A)-ρ), in retinal signaling in vivo by studying effects on the mouse electroretinogram (ERG) of genetic deletion of GABA C R versus pharmacological blockade using receptor antagonists. Brief full-field flash ERGs were recorded from anesthetized GABA C R −/− mice, and WT C57BL/6 (B6) mice, before and after intravitreal injection of GABA C R antagonists, TPMPA, 3-APMPA, or the more recently developed 2-AEMP; GABA A R antagonist, SR95531; GABA B R antagonist, CGP, and agonist, baclofen. Intravitreal injections of TPMPA and SR95531 were also made in Brown Norway rats. The effect of 2-AEMP on GABA-induced current was tested directly in isolated rat rod bipolar cells, and 2-AEMP was found to preferentially block GABA C R in those cells. Maximum amplitudes of dark (DA) and light-adapted (LA) ERG b-waves were reduced in GABA C R −/− mice, compared to B6 mice, by 30–60%; a-waves were unaltered and oscillatory potential amplitudes were increased. In B6 mice, after injection of TPMPA (also in rats), 3-APMPA or 2-AEMP, ERGs became similar to ERGs of GABA C R −/− mice. Blockade of GABA A Rs and GABA B Rs, or agonism of GABA B Rs did not alter B6 DA b-wave amplitude. The negative scotopic threshold response (nSTR) was slightly less sensitive in GABA C R −/− than in B6 mice, and unaltered by 2-AEMP. However, amplitudes of nSTR and photopic negative response (PhNR), both of which originate from inner retina, were enhanced by TPMPA and 3-APMPA, each of which has GABA B agonist properties, and further increased by baclofen. The finding that genetic deletion of GABA C R, the GABA C R antagonist 2-AEMP, and other antagonists all reduced ERG b-wave amplitude, supports a role for GABA C R in determining the maximum response amplitude of bipolar cells contributing to the b-wave. [Display omitted] The development of efficient catalysts for hydrogen evolution reaction (HER) from water splitting is one of the most promising strategies to achieve the goal of peak carbon dioxide emissions and carbon neutrality. Herein, Ru nanoclusters decorated MoSe 2 nanosheets supported on a Crepis tectorum fluff biomass-derived hollow carbon tube (Ru-MoSe 2 /CMT) are prepared as the HER catalysts in both alkaline and acidic conditions. The Ru modification induces the transformation of MoSe 2 from 2H phase to 1T phase. Benefiting from the strong water dissociation ability of Ru, Ru-MoSe 2 /CMT exhibits a low overpotential of 70 mV with a Tafel slope of 39 mV dec-1 in 1 M KOH. Furthermore, the assembled Ru-MoSe 2 /CMT || RuO 2 system with a low cell voltage of 1.54 V at 10 mA cm−2 exhibits outstanding overall water splitting performance superior to Pt/C || RuO 2 system. The Ru-MoSe 2 /CMT || RuO 2 system also achieves the excellent stability of up to 30 h in 1 M KOH. The synergy effect between Ru and MoSe 2 , as well as the improved electron transfer kinetics provided by the biomass-derived carbon substrate together contribute to the excellent HER activity of Ru-MoSe 2 /CMT. Epithelial mesenchymal transition (EMT) is a critical process implicated in the pathogenesis of retinal fibrosis and the exacerbation of diabetic retinopathy (DR) within retinal pigment epithelium (RPE) cells. Apigenin (AP), a potential dietary supplement for managing diabetes and its associated complications, has demonstrated inhibitory effects on EMT in various diseases. However, the specific impact and underlying mechanisms of AP on EMT in RPE cells remain poorly understood. In this study, we have successfully validated the inhibitory effects of AP on high glucose-induced EMT in ARPE-19 cells and diabetic db/db mice. Notably, our findings have identified CBP/p300 as a potential therapeutic target for EMT in RPE cells and have further substantiated that AP effectively downregulates the expression of EMT-related genes by attenuating the activity of CBP/p300, consequently reducing histone acetylation alterations within the promoter region of these genes. Taken together, our results provide novel evidence supporting the inhibitory effect of AP on EMT in RPE cells, and highlight the potential of specifically targeting CBP/p300 as a strategy for inhibiting retinal fibrosis in the context of DR. The first systematic acylated diversification of naturally scarce premyrsinane diterpenes, together with their biosynthetic precursors lathyrane diterpene were carried out. Two new series of premyrsinane derivates (1a - 32a) and lathyrane derivates (1 – 32) were synthesized from the naturally abundant lathyrane diterpene Euphorbia factor L 3 through a bioinspired approach. The cholinesterase inhibitory and neuroprotective activities of these diterpenes were investigated to explore potential anti-Alzheimer's disease (AD) bioactive lead compounds. In general, the lathyrane diterpenes showed the better acetylcholinesterase (AChE) inhibitory activity than that of premyrsinanes. The lathyrane derivative 17 bearing a 3-dimethylaminobenzoyl moiety showed the best AChE inhibition effect with the IC 50 value of 7.1 μM. Molecular docking demonstrated that 17 could bond with AChE well (−8 kal/mol). On the other hand, premyrsinanes showed a better neuroprotection profile against H 2 O 2 -induced injury in SH-SY5Y cells. Among them, the premyrsinane diterpene 16a had significant neuroprotective effect with the cell viability rate of 113.5 % at 12.5 μM (the model group with 51.2 %). The immunofluorescence, western blot and reactive oxygen species (ROS) analysis were conducted to demonstrate the mechanism of 16a. Invasive Aspergillosis is a high-risk illness with a high death rate in immunocompromised people due to a lack of early detection and timely treatment. Based on immunology study, we achieved an efficient production of anti-galactomannan antibody by Chinese hamster ovary (CHO) cells and applied it to time-resolved fluoroimmunoassay for Aspergillus galactomannan detection. We first introduced dual promoter expression vector into CHO host cells, and then applied a two-step screening strategy to screen the stable cell line by methionine sulfoximine pressurization. After amplification and fermentation, antibody yield reached 4500 mg/L. Then we conjugated the antibodies with fluorescent microspheres to establish a double antibody sandwich time-resolved fluoroimmunoassay, which was compared with the commercial Platelia™ Aspergillus Ag by clinical serum samples. The preformed assay could obtain the results in less than 25 min, with a limit of detection for galactomannan of approximately 1 ng/mL. Clinical results of the two methods showed that the overall percent agreement was 97.7% (95% CI: 96.6%–98.4%) and Cohen's kappa coefficient was 0.94. Overall, the assay is highly consistent with commercial detection, providing a more sensitive and effective method for the rapid diagnosis of invasive aspergillosis. Patients suffering from sepsis-induced acute lung injury (ALI) exhibit a high mortality rate, and their prognosis is closely associated with infiltration of neutrophils into the lungs. In this study, we found a significant elevation of CD64+ neutrophils, which highly expressed p75 neurotrophin receptor (p75NTR) in peripheral blood of mice and patients with sepsis-induced ALI. p75NTR+CD64+ neutrophils were also abundantly expressed in the lung of ALI mice induced by lipopolysaccharide. Conditional knock-out of the myeloid lineage's p75 NTR gene improved the survival rates, attenuated lung tissue inflammation, reduced neutrophil infiltration and enhanced the phagocytic functions of CD64+ neutrophils. In vitro, p75NTR+CD64+ neutrophils exhibited an upregulation and compromised phagocytic activity in blood samples of ALI patients. Blocking p75NTR activity by soluble p75NTR extracellular domain peptide (p75ECD-Fc) boosted CD64+ neutrophils phagocytic activity and reduced inflammatory cytokine production via regulation of the NF-κB activity. The signal transducer and activator of transcription 2 (STAT2), a downstream factor of type I interferons (IFNs), is a key component of the cellular antiviral immunity response. However, the role of STAT2 in the upstream of IFN signaling, such as the regulation of pattern recognition receptors (PRRs), remains unknown. In this study, STAT2 homologue of black carp (Mylopharyngodon piceus) has been cloned and characterized. The open reading frame (ORF) of bcSTAT2 comprises 2523 nucleotides and encodes 841 amino acids, which presents the conserved structure to that of mammalian STAT2. The dual-luciferase reporter assay and the plaque assay showed that bcSTAT2 possessed certain IFN-inducing ability and antiviral ability against both spring viremia of carp virus (SVCV) and grass carp reovirus (GCRV). Interestingly, we detected the association between bcSTAT2 and bcRIG-I through co-immunoprecipitation (co-IP) assay. Moreover, when bcSTAT2 was co-expressed with bcRIG-I, bcSTAT2 obviously suppressed bcRIG-I-induced IFN expression and antiviral activity. The subsequent co-IP assay and immunoblotting (IB) assay further demonstrated that bcSTAT2 inhibited K63-linked polyubiquitination but not K48-linked polyubiquitination of bcRIG-I, however, did not affect the oligomerization of bcRIG-I. Herein, hollow microtubes composed of RuNi 1 Co 1 alloy nanoparticles uniformly embedded in the carbon matrix (RuNi 1 Co 1 @CMT) are prepared through a simple impregnation followed by reduction. Benefiting from the unique mosaic structure and the synergistic effect between Ru and NiCo, RuNi 1 Co 1 @CMT achieves more exposed active sites and improved reaction kinetics. As a consequence, RuNi 1 Co 1 @CMT exhibits considerable catalytic activities with the overpotentials of 78 mV for HER and 299 mV for OER at 10 mA cm−2 in 1 M KOH. In addition, RuNi 1 Co 1 @CMT exhibits excellent stability for up to 30 h in both HER and OER processes at 20 mA cm−2, which is attributed to the protection of the RuNi 1 Co 1 alloy particles by the carbon layer. Furthermore, the assembled RuNi 1 Co 1 @CMT || RuNi 1 Co 1 @CMT overall water splitting system shows a cell voltage of 1.58 V at 10 mA cm−2. The density functional theory (DFT) calculations indicate that the addition of Ru can optimize the hydrogen adsorption free energy of Ni and Co sites. Finally, a solar panel-driven water splitting device is built, which can realize green and sustainable hydrogen production. Herein, we report a catalyst with CoSe 2 -MoSe 2 heterostructure grown on reduced graphene oxide with an optimum Co/Mo proportion of 1:1 (CoSe 2 -MoSe 2 (1–1)/rGO). It exhibits good HER activities in both acidic and alkaline conditions. The CoSe 2 -MoSe 2 (1–1)/rGO shows an overpotential of 107 mV at 10 mA cm−2 with a Tafel slope of 56 mV dec−1 under acidic condition. Meanwhile, CoSe 2 -MoSe 2 (1–1)/rGO also presents an overpotential of 182 mV at 10 mA cm−2 and with a Tafel slope of 89 mV dec−1 under alkaline condition. These impressive performances of the catalyst are mainly due to the excellent electronic transmission capability of rGO and the abundant active sites of CoSe 2 -MoSe 2 heterostructure as well as the optimized hydrogen adsorption energy of CoSe 2 -MoSe 2 interface. Abstract: One hundred and sixty-eight F6 recombinant inbred lines (RILs) derived from Chinese wheat cultivars, PH82-2 and Neixiang188, were used to determine the cumulative effects of HMW-GS and LMW-GS composition and quantity of gluten protein fractions on dough mixograph properties. A wide range of variation for all parameters in the RILs was detected. Major gene loci of HMW-GS were associated with variation in mixograph characters, but accounted for no more than 25.3% of the phenotypic variations. Glu-D1, together with Glu-B3, played the most important role in determining the properties. Additive effects of HMW-GS and LMW-GS showed major contributions to most of the variation of mixograph parameters, and epistatic effects were also important and could be counter to additive effects of individual loci. The quantity of gluten protein fractions, especially the quantity of glutenin, LMW-GS, and Glu-B3, showed highly significant correlations with most of the quality parameters, but the correlation coefficients were influenced by grain hardness, protein content, or both. Protein quality could be greatly improved through increasing the quantity of glutenin, while holding desirable composition of HMW-GS and LMW-GS alleles, with an appropriate ratio of quantity of glutenin to gliadin. Abstract: As pigs are susceptible to both human and avian influenza viruses, they have been proposed to be intermediate hosts or mixing vessels for the generation of pandemic influenza viruses through reassortment or adaptation to the mammalian host. In this study, we reported avian-like H1N1 and novel ressortant H1N2 influenza viruses from pigs in China. Homology and phylogenetic analyses showed that the H1N1 virus (A/swine/Zhejiang/1/07) was closely to avian-like H1N1 viruses and seemed to be derived from the European swine H1N1 viruses, which was for the first time reported in China; and the two H1N2 viruses (A/swine/Shanghai/1/07 and A/swine/Guangxi/13/06) were novel ressortant H1N2 influenza viruses containing genes from the classical swine (HA, NP, M and NS), human (NA and PB1) and avian (PB2 and PA) lineages, which indicted that the reassortment among human, avian, and swine influenza viruses had taken place in pigs in China and resulted in the generation of new viruses. The isolation of avian-like H1N1 influenza virus originated from the European swine H1N1 viruses, especially the emergence of two novel ressortant H1N2 influenza viruses provides further evidence that pigs serve as intermediate hosts or “mixing vessels”, and swine influenza virus surveillance in China should be given a high priority. Abstract: COUP-TFII (also known as Nr2f2), a member of the nuclear orphan receptor superfamily, is expressed in several regions of the central nervous system (CNS), including the ventral thalamus, hypothalamus, midbrain, pons, and spinal cord. To address the function of COUP-TFII in the CNS, we generated conditional COUP-TFII knockout mice using a tissue-specific NSE-Cre recombinase. Ablation of COUP-TFII in the brain resulted in malformation of the lobule VI in the cerebellum and a decrease in differentiation of cerebellar neurons and cerebellar growth. The decrease in cerebellar growth in NSECre/+/CIIF/F mice is due to reduced proliferation and increased apoptosis in granule cell precursors (GCPs). Additional studies demonstrated that insulin like growth factor 1 (IGF-1) expression was reduced in the cerebellum of NSECre/+/CIIF/F mice, thereby leading to decreased Akt1 and GSK-3β activities, and the reduced expression of mTOR. Using ChIP assays, we demonstrated that COUP-TFII was recruited to the promoter region of IGF-1 in a Sp1-dependent manner. In addition, dendritic branching of Purkinje cells was decreased in the mutant mice. Thus, our results indicate that COUP-TFII regulates growth and maturation of the mouse postnatal cerebellum through modulation of IGF-1 expression. [Copyright &y& Elsevier]

In this work, we synthesized iron molybdenum selenide (FeSe 2 -MoSe 2) loaded on reduced graphene oxide (FeSe 2 -MoSe 2 /rGO) by a one-step hydrothermal method. We further optimized the Fe/Mo ratio and determined the best ratio to be 1-1. In acidic (or alkaline) solution, the optimized FeSe 2 -MoSe 2 (1-1)/rGO has a small Tafel slope of 55 (or 80) mV dec−1 and needs an overpotential of 101 (or 178) mV to achieve 10 mA cm−2. These good properties are mainly due to the structure of bimetallic selenides combining rGO. Moreover, rGO enhances the electrical conductivity. Furthermore, the synergistic effect between FeSe 2 -MoSe 2 (1-1) and rGO results in better HER performance. [Display omitted] Electrocatalytic water splitting used for generating clean and sustainable hydrogen (H 2) can be very promising to address current energy shortage and associated environmental issues. However, this methodology is severely impeded by the tardy oxygen evolution reaction (OER). Hence, designing a preferable kinetics and thermodynamics oxidation reaction that supersede OER is very significant for the energy-saving production of H 2. Herein, hollow needle-like copper cobalt sulfide was constructed on carbon cloth (CuCo 2 S 4 /CC) as a bifunctional electrocatalyst to accelerate H 2 generation and simultaneously convert ethanol into value-added acetic acid. Thanks to the synergistic effect and unique structure of Cu and Co, CuCo 2 S 4 /CC displays superior catalytic activity and durability in ethanol oxidation reaction (EOR) with a low potential of 1.38 V vs. RHE (@10 mA cm−2). Meanwhile, it exhibits excellent hydrogen evolution reaction (HER) performance. The homemade CuCo 2 S 4 /CC//CuCo 2 S 4 /CC ethanol–water electrolyser only demands a voltage of 1.59 V to deliver 10 mA cm−2, 150 mV less than that used for ordinary water splitting. This shows that the ethanol–water electrolyser elaborated here holds encouraging potential in the energy-saving production of H 2 and oxidation of ethanol into value-added acetic acid. [Display omitted] MoS 2 is regarded as an attractive anode material for lithium-ion batteries due to its layered structure and high theoretical specific capacity. Its unsatisfied conductivity and the considerable volume change during the charge and discharge process, however, limits its rate performance and cycling stability. Herein, 3D tremella-like nitrogen-doped carbon encapsulated few-layer MoS 2 (MoS 2 @NC) hybrid is obtained via a unique strategy with simultaneously poly-dopamine carbonization, and molybdenum oxide specifies sulfurization. The three-dimensional porous nitrogen-doped carbon served both as a mechanical supporting structure for stabilization of few-layers MoS 2 and a good electron conductor. The MoS 2 @NC exhibits enhanced high rate performance with a specific capacity of 208.7 mAh g−1 at a current density of 10 A g−1 and stable cycling performance with a capacity retention rate of 85.7% after 1000 cycles at 2 A g−1. It seems certain that COX-2 is related to tumor and some data suggested that COX-2 might have relation to tumor malignance and angiogenesis. In order to elucidate the relationship between COX-2 and tumor invasive and angiogenic ability, we transfected human transitional cell carcinoma (TCC) cell line, EJ, permanently with a COX-2 expression vector or the mock vector. The EJ-COX2 cells, which overexpressed COX-2, acquired increased invasiveness and angiogenic ability by activation of VEGF, uPA, and MMP-2. Increased invasiveness and angiogenic ability were reversed by treatment with either selective COX-2 inhibitor, NS-398, or dual COX inhibitor, indomethacin. These results demonstrate that overexpression of COX-2 can lead to phenotypic changes that alter the metastatic and angiogenic potential of TCC cancer cells. [Copyright &y& Elsevier]

Actin filaments form unique structures with robust actin bundles and cytoskeletal networks affixed to the extracellular matrix and interact with neighboring cells, which are crucial structures for cancer cells to acquire a motile phenotype. This study aims to investigate a novel antitumor mechanism by which Tanshinone IIA (Tan IIA) modulates the morphology and migration of liver cancer cells via actin cytoskeleton regulation. 97H and Huh7 exhibited numerous tentacle-like protrusions that interacted with neighboring cells. Following treatment with Tan IIA, 97H and Huh7 showed a complete absence of cytoplasmic protrusion and adherens junctions, thereby effectively impeding their migration capability. The fluorescence staining of F-actin and microtubules indicated that these tentacle-like protrusions and cell-cell networks were actin-based structures that led to morphological changes after Tan IIA treatment by retracting and reorganizing beneath the membrane. Tan IIA can reverse the actin depolymerization and cell morphology alterations induced by latrunculin A. Tan IIA down-regulated actin and Rho GTPases expression significantly, as opposed to inducing Rho signaling activation. Preventing the activity of proteasomes and lysosomes had no discernible impact on the modifications in cellular structure and protein expression induced by Tan IIA. However, as demonstrated by the puromycin labeling technique, the newly synthesized proteins were significantly inhibited by Tan IIA. In conclusion, Tan IIA can induce dramatic actin cytoskeleton remodeling by inhibiting the protein synthesis of actin and Rho GTPases, resulting in the suppression of tumor growth and migration. Bifunctional catalyst was prepared by one-step pyrolysis of trimetallic ZIF. Herein, we proposed a novel bifunctional catalyst derived from the pyrolysis of Co/Fe/Zn trimetallic zeolitic imidazolite framework. The obtained Co@CoFe 0.01 N C catalyst displays desirable activity for both ORR and OER (E 1/2 = 0.844 V, E j=10 = 1.654 V). The Zn-air battery equipped with Co@CoFe 0.01 N C catalyst on the cathode exhibits a high peak power density of 174.1 mW cm−2, which is much superior than that of commercial 20% Pt/C (87.6 mW cm−2). Binary metal niobium oxides can offer a higher specific capacity compared to niobium pentoxide (Nb 2 O 5) and thus are ideal anode candidates for lithium ion capacitors (LICs). However, their lower electronic conductivity limits their ability to achieve high energy and power densities. In this paper, one-dimensional (1D) copper niobate (CuNb 2 O 6) nanowires are successfully prepared by electrospinning technology and then immobilized on two-dimensional (2D) reduced graphene oxide (rGO) nanosheets to form a unique 1D nanowire/2D nanosheet CuNb 2 O 6 /rGO structure. The 1D/2D CuNb 2 O 6 /rGO electrode exhibits a high specific capacity of 312.2 mAh g−1 at 100 mA g−1 as the anode of LICs. The proposed Li+ storage mechanism of the CuNb 2 O 6 anode involves CuNb 2 O 6 decomposition into lithium niobate (Li 3 NbO 4) and copper (Cu) during the initial lithium insertion process. The intercalation-type Li 3 NbO 4 will further serve as the host to Li+ and the inactive Cu phase will act as a conductive network for electron transportation. Furthermore, the energy density of the assembled CuNb 2 O 6 /rGO//activated carbon (CuNb 2 O 6 /rGO//AC) device could achieve a value as high as 92.1 Wh kg−1 and could thus be considered as a possible alternative electrode material for high energy and power LICs. 3,6-anhydro-α- L -galactose (L-AHG) is one of the main monosaccharide constituents of red macroalgae. In the recently discovered bacterial L-AHG catabolic pathway, L-AHG is first oxidized by a NAD(P)+-dependent dehydrogenase (AHGD), which is a key step of this pathway. However, the catalytic mechanism(s) of AHGDs is still unclear. Here, we identified and characterized an AHGD from marine bacterium Vibrio variabilis JCM 19239 (Vv AHGD). The NADP+-dependent Vv AHGD could efficiently oxidize L-AHG. Phylogenetic analysis suggested that Vv AHGD and its homologs represent a new aldehyde dehydrogenase (ALDH) family with different substrate preferences from reported ALDH families, named the L-AHGDH family. To explain the catalytic mechanism of Vv AHGD, we solved the structures of Vv AHGD in the apo form and complex with NADP+ and modeled its structure with L-AHG. Based on structural, mutational, and biochemical analyses, the cofactor channel and the substrate channel of Vv AHGD are identified, and the key residues involved in the binding of NADP+ and L-AHG and the catalysis are revealed. Vv AHGD performs catalysis by controlling the consecutive connection and interruption of the cofactor channel and the substrate channel via the conformational changes of its two catalytic residues Cys282 and Glu248. Comparative analyses of structures and enzyme kinetics revealed that differences in the substrate channels (in shape, size, electrostatic surface, and residue composition) lead to the different substrate preferences of Vv AHGD from other ALDHs. This study on Vv AHGD sheds light on the diversified catalytic mechanisms and evolution of NAD(P)+-dependent ALDHs.
