Excessive growth hormone (GH) has been shown to promote joint degeneration in both preclinical and clinical studies. Little is known about the effect of disrupted GH or GH receptor (GHR) on joint health. The goal of this study is to investigate joint pathology in mice with either germline (GHR-/-) or adult inducible (iGHR-/-) GHR deficiency. Knee joints from male and female GHR-/- and WT mice at 24 months of age were processed for histological analysis. Also, knee joints from male and female iGHR-/- and WT mice at 22 months of age were scanned by micro-CT (μCT) for subchondral bone changes and characterized via histology for cartilage degeneration. Joint sections were also stained for the chondrocyte hypertrophy marker, COLX, and the cartilage degeneration marker, ADAMTS-5, using immunohistochemistry. Compared to WT mice, GHR-/- mice had remarkably smooth articular joint surfaces and an even distribution of proteoglycan with no signs of degeneration. Quantitatively, GHR-/- mice had lower OARSI and Mankin scores compared to WT controls. By contrast, iGHR-/- mice were only moderately protected from developing aging-associated OA. iGHR-/- mice had a significantly lower Mankin score compared to WT. However, Mankin scores were not significantly different between iGHR-/- and WT when males and females were analyzed separately. OARSI scores did not differ significantly between WT and iGHR-/- in either individual or combined sex analyses. Both GHR-/- and iGHR-/- mice had fewer COLX+ hypertrophic chondrocytes compared to WT, while no significant difference was observed in ADAMTS-5 staining. Compared to WT, a significantly lower trabecular thickness in the subchondral bone was observed in the iGHR-/- male mice but not in the female mice. However, there were no significant differences between WT and iGHR-/- mice in the bone volume to total tissue volume (BV/TV), bone mineral density (BMD), and trabecular number in either sex. This study identified that both germline and adult-induced GHR deficiency protected mice from developing aging-associated OA with more effective protection in GHR-/- mice.

BackgroundThe calcaneal enthesis, an osseous footprint where the Achilles tendon seamlessly integrates with the bone, represents a complex interface crucial for effective force transmission. Bone adapts to mechanical stress and remodels based on the applied internal and external forces. This study explores the relationship between the elasticity of the Achilles tendon enthesis and the bone microstructure in the calcaneal crescent.MethodsIn total, 19 calcaneal-enthesis sections, harvested from 10 fresh-frozen human cadaveric foot-ankle specimens (73.8 ± 6.0 years old, seven female), were used in this study. Indentation tests were performed at the enthesis region, and Hayes' elastic modulus was calculated for each specimen. Micro-CT scanning was performed at 50-micron voxel size to assess trabecular bone microstructure within six regions of interest (ROIs) and the cortical bone thickness along the calcaneal crescent.ResultsSignificant Spearman correlations were observed between the enthesis elastic modulus and trabecular bone thickness in the distal entheseal (ROI 3) and proximal plantar (ROI 4) regions (R = 0.786 and 0.518, respectively).ConclusionThis study highlights the potential impacts of Achilles tendon enthesis on calcaneal bone microstructure, which was pronounced in the distal calcaneal enthesis, suggesting regional differences in load transfer mechanism that require further investigation.

Considering the lack of consensus related to the impact of selective IL-6 receptor inhibition on bone remodeling and the scarcity of reports, especially on large bone defects, this study proposed to evaluate the biological impact of the selective inhibitor of interleukin-6 receptor (tocilizumab) in an experimental model of critical calvarial defect in rats. In this preclinical and in vivo study, 24 male Wistar rats were randomly divided into two groups (n=12/group): defect treated with collagen sponge (CG) and defect treated with collagen sponge associated with 2 mg/kg tocilizumab (TCZ). The defect in the parietal bone was created using an 8-mm diameter trephine drill. After 90 days, the animals were euthanized, and tissue samples (skull caps) were evaluated through micro-CT, histological, immunohistochemistry, cytokines, and RT-qPCR analyses. Tocilizumab reduced mononuclear inflammatory infiltration (P0.05). The bone cells (osteoblasts, osteoclasts, and osteocytes) in the defect area were similar in both groups (P>0.05). Tocilizumab reduced inflammatory cytokines, decreased osteogenic protein, and increased proteases in a critical bone defect in rats. Ninety days after the local application of tocilizumab in the cranial defect, we did not find a significant formation of bone tissue compared with a collagen sponge.

Osteocytes locally remodel their surrounding tissue through perilacunar canalicular remodeling (PLR). During lactation, osteocytes remove minerals to satisfy the metabolic demand, resulting in increased lacunar volume, quantifiable with synchrotron X-ray radiation micro-tomography (SRµCT). Although the effects of lactation on PLR are well-studied, it remains unclear whether PLR occurs uniformly throughout the bone and what mechanisms prevent PLR from undermining bone quality. We used SRµCT imaging to conduct an in-depth spatial analysis of the impact of lactation and osteocyte-intrinsic MMP13 deletion on PLR in murine bone. We found larger lacunae undergoing PLR are located near canals in the mid-cortex or endosteum. We show lactation-induced hypomineralization occurs 14 µm away from lacunar edges, past a hypermineralized barrier. Our findings reveal that osteocyte-intrinsic MMP13 is crucial for lactation-induced PLR near lacunae in the mid-cortex but not for whole-bone resorption. This research highlights the spatial control of PLR on mineral distribution during lactation.

BackgroundWe investigated the relationship of two commonly used quantitative ultrasound (QUS) parameters, speed of sound (SoS) and attenuation coefficient (α), with water and macromolecular contents of bovine cortical bone strips as measured with ultrashort echo time (UTE) magnetic resonance imaging (MRI).MethodsSoS and α were measured in 36 bovine cortical bone strips utilizing a single-element transducer with nominal 5 MHz center frequency based on the time of flight principles after accommodating for reflection losses. Specimens were then scanned using UTE MRI to measure total, bound, and pore water proton density (TWPD, BWPD, and PWPD) as well as macromolecular proton fraction and macromolecular transverse relaxation time (T2-MM). Specimens were also scanned using microcomputed tomography (μCT) at 9-μm isometric voxel size to measure bone mineral density (BMD), porosity, and pore size. The elastic modulus (E) of each specimen was measured using a 4-point bending test.Resultsα demonstrated significant positive Spearman correlations with E (R = 0.69) and BMD (R = 0.44) while showing significant negative correlations with porosity (R = -0.41), T2-MM (R = -0.47), TWPD (R = -0.68), BWPD (R = -0.67), and PWPD (R = -0.45).ConclusionsThe negative correlation between α and T2-MM is likely indicating the relationship between QUS and collagen matrix organization. The higher correlations of α with BWPD than with PWPD may indicate that water organized in finer structure (bound to matrix) provides lower acoustic impedance than water in larger pores, which is yet to be investigated thoroughly.Relevance statementThis study highlights the importance of future investigations exploring the relationship between QUS measures and all major components of the bone, including the collagenous matrix and water. Background and Purpose: Spinal cord stimulation (SCS) is approved by the Food and Drug Administration for treating chronic intractable pain in the back, trunk, or limbs through stimulation of the dorsal column. Numerous studies have used swine as an analog of the human spinal cord to better understand SCS and further improve its efficacy. We performed high‐resolution imaging of the porcine spinal cord with intact dura mater using micro‐computed tomography (μCT) to construct detailed 3‐dimensional (3D) visualizations of the spinal cord and characterize the morphology of the dorsal and ventral rootlets. Methods: We obtained spinal cords from Yorkshire/Landrace crossbred swine (N = 7), stained samples with osmium tetroxide, and performed μCT imaging of the T12‐T15 levels at isotropic voxel resolutions ranging from 3.3 to 50 μm. We measured the anatomical morphology using the 3D volumes and compared our results to measurements previously collected from swine and human spinal cords via microdissection techniques in prior literature. Results: While the porcine thoracic‐lumbar spinal cord is a popular model for SCS, we highlight multiple notable differences compared to previously published T8‐T12 human measurements including rootlet counts (porcine dorsal/ventral: 12.2 ± 2.6, 26.6 ± 3.4; human dorsal/ventral: 5.3 ± 1.3, 4.4 ± 2.4), rootlet angles (porcine ventral‐rostral: 161 ± 1°, ventral‐caudal: 155 ± 6°, dorsal‐rostral: 148 ± 9°, dorsal‐caudal: 142 ± 6°; human ventral‐rostral: 170 ± 3°, ventral‐caudal: 22 ± 10°, dorsal‐rostral: 171 ± 3°, dorsal‐caudal: 15 ± 7°), and the presence and count of dorsal rootlet bundles. Conclusions: Detailed measurements and highlighted differences between human and porcine spinal cords can inform variations in modeling and electrophysiological experiments between the two species. In contrast to other approaches for measuring the spinal cord and rootlet morphology, our method keeps the dura intact, reducing potential artifacts from dissection. [ABSTRACT FROM AUTHOR]

Background: Proximal caries datasets for training artificial intelligence (AI) algorithms commonly include clinician-annotated radiographs. These conventional annotations are susceptible to observer variability, and early caries may be missed. Micro-computed tomography (CT), while not feasible in clinical applications, offers a more accurate imaging modality to support the creation of a reference-standard dataset for caries annotations. Herein, we present the Academic Center for Dentistry Amsterdam—Diagnostic Insights for Radiographic Early-caries with micro-CT (ACTA-DIRECT) dataset, which is the first dataset pairing dental radiographs and micro-CT scans to enable higher-quality annotations. Methods: The ACTA-DIRECT dataset encompasses 179 paired micro-CT scans and radiographs of early proximal carious teeth, along with three types of annotations: conventional annotations on radiographs, micro-CT-assisted annotations on radiographs, and micro-CT annotations (reference standard). Three dentists independently annotated proximal caries on radiographs, both with and without micro-CT assistance, enabling determinations of interobserver agreement and diagnostic accuracy. To establish a reference standard, one dental radiologist annotated all caries on the related micro-CT scans. Results: Micro-CT support improved interobserver agreement (Cohen's Kappa), averaging 0.64 (95% confidence interval [CI]: 0.59–0.68) versus 0.46 (95% CI: 0.44–0.48) in its absence. Likewise, average sensitivity and specificity increased from 42% (95% CI: 34–51%) to 63% (95% CI: 54–71%) and from 92% (95% CI: 88–95%) to 95% (95% CI: 92–97%), respectively. Conclusion: The ACTA-DIRECT dataset offers high-quality images and annotations to support AI-based early caries diagnostics for training and validation. Background and Aims Methods Results Interpretations Chemotherapy‐induced peripheral neurotoxicity (CIPN), with paraesthesia, numbness, dysesthesia and neuropathic pain ranks among the most common dose‐limiting toxicity of several widely used anticancer drugs. Recent studies revealed the microvascular angiogenesis as a new important actor, beside peripheral neurons, in the neurotoxicity and neuropathic pain development and chronicisation. The aim of this work is to elucidate the role of vascular alterations in CIPN.We evaluated the severity of CIPN with neurophysiological, behavioural and neuropathological analysis together with the microvascular network in central and peripheral nervous systems of rats in order to correlate the features of the CIPN and the vascular abnormalities. The vascular network was quantitatively evaluated through synchrotron radiation‐based X‐ray phase‐contrast micro‐tomography imaging, measuring four specific parameters: vascular density, vessel diameter, vessel tortuosity and branching.Rats exposed to paclitaxel and affected by a severe painful sensory axonopathy showed an increased vascular density (putative sprouting angiogenesis) in the crucial districts of the central (somatosensory cortex and lumbar spinal cord) and peripheral nervous system (lumbar dorsal root ganglia). In addition, the complexity of the vascular network and the size of neo‐formed vessels were significantly decreased in specific regions. On the other hand, less significant changes were observed in rats exposed to cisplatin, affected by a painless peripheral neuropathy, suggesting a specific involvement of neo‐angiogenesis in the development of severe neurotoxicity and neuropathic pain.These new ground‐breaking results can shed light on new pathogenetic mechanisms and potential novel therapeutic approaches for painful‐CIPN. Using high-resolution micro-CT imaging at 2.98 μm/voxel, we compared the percolation of hydrogen in gas injection with gas expansion for a hydrogen-brine system in Bentheimer sandstone at 1 MPa and 20 °C, representing hydrogen storage in an aquifer. We introduced dimensionless numbers to quantify the contribution of advection and expansion to displacement. We analysed the 3D spatial distribution of gas and its displacement in both cases and demonstrated that in gas injection, hydrogen can only advance from a connected cluster in an invasion-percolation type process, while in gas expansion, hydrogen can access more of the pore space even from disconnected clusters. The average gas saturation in the sample increased from 30% to 50% by gas expansion, and we estimated that 10% of the expanded volume is attributed to hydrogen exsolution from the brine. This work emphasises the importance of studying the combined effects of pressure decline and gas withdrawal in hydrogen storage to assess the influence of gas expansion on remobilising trapped gases. The nature of the magma plumbing system of Large Igneous Provinces is still poorly understood. Among these exceptional magmatic events from Earth's past, the end-Triassic Central Atlantic Magmatic Province (CAMP) and the end-Cretaceous Deccan Traps (Deccan) coincided in time with two of the most catastrophic biotic crises during the Phanerozoic. In order to constrain the architecture of their magma plumbing system, glomerocrysts containing abundant bubble-bearing melt inclusions from basaltic lava flows of both CAMP and Deccan were investigated via in situ geochemical and microstructural analyses. The analysed glomerocrysts, dominated by augitic clinopyroxene crystals, represent fragments of a crystal mush entrained by basaltic magmas before eruption. The analysed melt inclusions, consisting of an intermediate to felsic composition glass and CO2-bearing bubbles, represent relics of interstitial melts and fluids within a porous crystal framework forming the crystal mush. The different volume proportions between bubbles and whole inclusions reveal that melt entrapment occurred after volatile exsolution. The minimum observed bubble/inclusion fraction indicates that the CO2 concentration in CAMP and Deccan melts was at least 0.3 wt.%, consistent with a maximum entrapment pressure of about 0.5 GPa at CO2–H2O fluid-saturated conditions. The MgO-rich composition of host clinopyroxene crystals and whole rocks is in contrast with the SiO2-rich composition of (trachy-) andesitic to rhyolitic glass of melt inclusions, pointing to disequilibrium conditions. Thermodynamic and geochemical modelling shows that fractional crystallization alone cannot explain the evolved composition of glass in melt inclusions starting from their whole rock composition. On one side, the oxygen isotope composition of clinopyroxene crystals in glomerocrysts ranges from +3.9 (± 0.3) to +5.8 (± 0.3) ‰ and their sample-averaged oxygen isotope composition spans from +4.4 (N = 10) to +5.6 (N = 10) ‰, implying that glomerocrysts crystallized from mafic melts with normal (i.e. mantle-like) to slightly low δ18O values. On the other side, the oxygen isotope composition of glass in melt inclusions ranges from +5.5 (± 0.4) to +22.1 (± 0.4) ‰, implying that melt inclusions entrapped intermediate to felsic melts with normal (i.e. mantle-like) to extremely high δ18O values, typical of (meta-) sedimentary rocks. Some melt inclusions are compatible with fractionation from the same mafic melts that crystallized their host mineral phase, but most melt inclusions are compatible with variable degrees of crustal assimilation and partial mixing, potentially followed by minor post-entrapment isotope re-equilibration. In the CAMP, where sedimentary basins are abundant, (meta-) pelites and occasionally granitoids were the most likely assimilants. On the contrary, in the Deccan, where sedimentary basins are rare, granitoids and metapelites were the most likely assimilants. Oxygen isotope compositions of glass in melt inclusions, spanning from mantle-like to crust-dominated signatures, suggest that the CO2 within their coexisting bubbles likely derived partly from the mantle and partly from assimilated crustal materials. The investigated glomerocrysts and their bubble-bearing melt inclusions are relics of a multiphase (i.e. solid + liquid + gas phases) crystal mush revealing a dynamic evolution for the magma plumbing system of both CAMP and Deccan, where crystals, silicate melts and exsolved fluids coexisted and interacted through most of the transcrustal section. Background: The sea spiders (Pycnogonida Latreille, 1810) of the Hunsrück Slate (Lower Devonian, ~400 million years ago) are iconic in their abundance, exquisite pyritic preservation, and in their distinctive body plan compared to extant sea spiders (Pantopoda Gerstäcker, 1863). Consequently, the Hunsrück sea spiders are important in understanding the deep evolutionary history of Pycnogonida, yet they remain poorly characterised, impacting upon attempts to establish a time-calibrated phylogeny of sea spiders. Methods: Here, we investigated previously described and new material representing four of the five Hunsrück pycnogonids: Flagellopantopus blocki Poschmann & Dunlop, 2006; Palaeoisopus problematicus Broili, 1928; Palaeopantopus maucheri Broili, 1929; and Pentapantopus vogteli Kühl, Poschmann & Rust, 2013; as well as a few unidentified specimens. Using X-ray microtomography and Reflectance Transformation Imaging, we describe new fossils, provide evidence for newly revealed anatomical features, and interpret these data in comparison to extant species. We also reinterpret the previously published illustration of the (probably lost) holotype of Palaeothea devonica Bergström, Stürmer & Winter, 1980. Results: We provide the first detailed description of the cephalic appendages of Palaeoisopus problematicus and revise the interpretation of the organisation of its ocular tubercle. Furthermore, we provide new insights into the structure of the legs and the proboscis of Palaeopantopus maucheri, the first description of the body of Flagellopantopus blocki and describe a new specimen of Pentapantopus vogteli, demonstrating that it had eight legs, in contrast to previous interpretations. We argue that, contrary to previous suggestions, Palaeothea devonica probably had a different body plan from extant pantopods. We discuss the ecological traits of the Hunsrück pycnogonids based on their morphological adaptations, and conclude that there is no compelling evidence of Pantopoda in the Devonian. Through comparative interpretation of the legs as well as general morphology, we can divide the Hunsrück pycnogonids into two morphological groups, while Pantopoda constitutes a third morphological group. Background: To investigate the anatomic features of the root furcation of permanent mandibular first molars. Methods: A total of 50 extracted mandibular first molars (25 two-rooted and 25 three-rooted) were collected and scanned using micro-computed tomography. The digital models of teeth and root canal systems were reconstructed three-dimensionally. The tooth models were displayed in parallel projection mode from buccal and distal views. Screenshots were captured and subsequently analyzed using Image-Pro Plus 6.0 software after calibration. The furcation angle, root trunk length, maximum depth and level of distal root concaves of mesial roots, and length of enamel projections were measured, and the furcation types (classified into type V, type U and type W) were detected. Statistical analysis was performed using the Shapiro-Wilk's test, one-way analysis of variance, Student's t-test and Chi-square test. Results: The mean furcation angle between the distobuccal (DB) and distolingual (DL) roots (in distal view) was the greatest (59.2°), whereas the furcation angle between the mesial and DL roots (in buccal view) was the smallest (25.4°) among the four furcation angles (all p < 0.05). Regarding the furcation types, bucco-lingual root trunk length, maximum depth and site of the distal root concavities, and enamel projection length, no significant differences were detected between the three- and two-rooted molar groups (all p > 0.05). The frequency of type V was the highest (54.0%), followed by type U (26.0%), and type W had the lowest occurrence rate (20.0%). The mean length of distal root trunk in the three-rooted mandibular molars was significantly greater than that of the buccal/lingual one (3.7 mm vs. 3.0 mm, p < 0.01). The maximum depth of the distal concavities of the mesial roots was on average 0.66 ± 0.19 mm, and the site was located at an average of 2.8 ± 1.3 mm below furcation. The mean length of buccal enamel projections was significantly longer than that of lingual ones (3.1 mm vs. 0.7 mm, p < 0.01). Conclusions: The furcation anatomy of the mandibular first molar is complex, and the presence of the DL root may further complicate its topography. Soybean is a source of high-quality vegetable protein and a suitable substitute for meat protein. However, several factors, including a lengthy cooking process, palatability issues, and digestion concerns, have limited their consumption. Several treatments have been suggested to overcome such issues, including infrared heating, which has many advantages over traditional roasting methods. In this study, the three-dimensional microstructure of soybeans with initial moisture contents (12, 16, 20, and 24% w.b.) and treated at different micronization durations (60, 90, and 120 s) at 180 °C were studied and related to quality parameters including water absorption and hardness of both non-cooked and cooked samples. The water absorption tests demonstrated no significant difference (p > 0.05) between the moisture absorbed for 60, 90, and 120 s micronization with initial moisture levels of 12, 16, and 24%. Yet all soybeans treated for 60 s exhibited significantly different results (p < 0.05) compared to raw counterparts. The highest hardness values were achieved after 60 s treatments on 24% moisturized seeds. As expected, the 20 min cooking time resulted in a considerable reduction in soybean hardness. The X-ray micro-computed tomography (micro-CT) was a powerful non-destructive tool to explore the microstructure of individual soybean seeds. Results of X-ray micro-CT have shown an increase in open porosity with increasing infrared treatment time. Overall, the qualitative and quantitative analysis provided in this study may serve as a basis for developing improved food processing techniques. [ABSTRACT FROM AUTHOR]

Abstract Bone defects pose a significant risk to human health. Medical polyetheretherketone (PEEK) is an excellent implant material for bone defect repair, but it faces the challenge of bone osteoconduction and osseointegration. Osteoconduction describes the process by which bone grows on the surface of the implant, while osseointegration is the stable anchoring of the implant achieved by direct contact between the bone and the implant. Bone defects repair depends on the implant’s three-dimensional spatial structure, including pore size, porosity, and interconnections to a great extent. However, it is challenging to fabricate the porous structures to meet specific requirements and to characterize them without causing damage. In this study, we designed and fabricated sandwich-like PEEK implants mimicking the three-layer structures of the skull, whose defects imposes a significant burden on young adulthood and paediatric populations, and performed in-line phase-contrast synchrotron X-ray microtomography to non-destructively investigate the internal porous microstructures. The sandwich-like three-layer microstructure, comprising a dense layer, a loose layer and a dense layer in succession, exhibits structural similarity to that in a natural skull. This work demonstrated the fabrication of the sandwich-like PEEK implant that could potentially enhance osteoconduction and osseointegration. Furthermore, the interior structures and residual porogen sodium chloride particles were observed within the PEEK implant, which cannot be realized by other microscopic methods without destroying the sample. It highlights the advantages and potential of using synchrotron X-ray microtomography to analyze the structure of biomedical materials. This study provides theoretical guidance for the further design and fabrication of PEEK bone repair materials and will advance the clinical application of innovative bioactive bone repair materials.

Results Micro-CT support improved interobserver agreement (Cohen’s Kappa), averaging 0.64 (95% confidence interval [CI]: 0.59–0.68) versus 0.46 (95% CI: 0.44–0.48) in its absence. Likewise, average sensitivity and specificity increased from 42% (95% CI: 34–51%) to 63% (95% CI: 54–71%) and from 92% (95% CI: 88–95%) to 95% (95% CI: 92–97%), respectively. Conclusion The ACTA-DIRECT dataset offers high-quality images and annotations to support AI-based early caries diagnostics for training and validation. This study underscores the benefits of incorporating micro-CT scans in lesion assessments, providing enhanced precision and reliability. Graphical Abstract

Introduction. Sound-absorbing systems are widely used in construction, reconstruction, repair of industrial and civil facilities, as well as in mechanical engineering. The most affordable and widely used systems are those using porous materials. The influence of the type, size, nature of pores, as well as phase composition of porous glass-ceramic samples obtained from zeolite-containing rocks on its sound-absorbing properties was established.Materials and methods. The results of experimental studies were obtained by the impedance method, as well as by methods of X-ray phase analysis, light microscopy, X-ray microtomography, etc.Results. Porous glass-ceramic materials with average density grades D250 and D300, compressive strength class B3.5, thermal conductivity coefficient from 0.066 to 0.079 W/m∙°C and recommended maximum application temperature up to +850 °C were investigated. The total porosity of the tested specimens of glass-ceramic materials was from 87.7 to 90.1 %, and the number of open pores from to 18.3 %. The sound-absorbing properties of porous glass-ceramic specimens are directly dependent on the open porosity of the material. With an increase of this indicator from 4.5 to 18.3 %, the sound absorption coefficient of the material increased 1.5–2 times, depending on the range of sound frequencies.Conclusions. Porous glass ceramics obtained from zeolite-containing rocks have good physical and mechanical properties and a high application temperature (at least +850 °C), and with a high open porosity index, it can be recommended for application as sound-absorbing materials in construction and mechanical engineering facilities.

Abstract Background To investigate the anatomic features of the root furcation of permanent mandibular first molars. Methods A total of 50 extracted mandibular first molars (25 two-rooted and 25 three-rooted) were collected and scanned using micro-computed tomography. The digital models of teeth and root canal systems were reconstructed three-dimensionally. The tooth models were displayed in parallel projection mode from buccal and distal views. Screenshots were captured and subsequently analyzed using Image-Pro Plus 6.0 software after calibration. The furcation angle, root trunk length, maximum depth and level of distal root concaves of mesial roots, and length of enamel projections were measured, and the furcation types (classified into type V, type U and type W) were detected. Statistical analysis was performed using the Shapiro-Wilk’s test, one-way analysis of variance, Student’s t-test and Chi-square test. Results The mean furcation angle between the distobuccal (DB) and distolingual (DL) roots (in distal view) was the greatest (59.2°), whereas the furcation angle between the mesial and DL roots (in buccal view) was the smallest (25.4°) among the four furcation angles (all p 0.05). The frequency of type V was the highest (54.0%), followed by type U (26.0%), and type W had the lowest occurrence rate (20.0%). This study presents a novel method to determine the kinetics of spherical agglomeration in-line via monitoring the stirrer torque. In order to confirm that the torque can be used to characterise the process phases, samples were taken at different times during the agglomeration process and the morphology of agglomerates is examined using X-ray microtomography. It can be shown that if the stirrer torque is decreasing, the fast growth regime starts. Additionally, the influence of binding-liquid concentration as well as the primary particle shape was studied. Both have an influence on the process kinetics as well as on the different growth mechanisms of agglomerates during spherical agglomeration. These two growth mechanisms, distribution and immersion, can be identified by the morphological parameters sphericity, packing density and fractal dimension. Evidence concerning the osteotoxic effects of chemotherapy (doxorubicin) has been previously described. Periodontitis also progressively increases in patients receiving chemotherapy; however, the beneficial effects of melatonin and metformin on the alleviation of doxorubicin-induced osteotoxicity have never been investigated. Therefore, we investigated the negative impact of doxorubicin on alveolar bone homeostasis and the benefits of melatonin and metformin on the attenuation of doxorubicin-induced alveolar bone toxicity. Male Wistar rats were divided into 4 groups to receive either 1 mL of normal saline solution as a control group, 3 mg/kg of doxorubicin, 3 mg/kg of doxorubicin plus 10 mg/kg of melatonin, or 3 mg/kg of doxorubicin plus 250 mg/kg of metformin. Doxorubicin treatment was given on days 0, 4, 8, 15, 22, and 29, while interventions were given daily on days 0 to 29. Following euthanasia, blood and alveolar bones were collected for evaluation of oxidative stress, bone remodeling, inflammation, microarchitecture, and periodontal condition. We found that doxorubicin increased systemic oxidative stress, decreased antioxidative capacity, increased inflammation, decreased bone formation, increased bone reabsorption, impaired microarchitecture, and impaired periodontal condition of the alveolar bone. Although cotreatment with melatonin or metformin resulted in some improvement in these parameters, cotreatment with melatonin was more effective than cotreatment with metformin in terms of decreasing oxidative stress, reducing bone resorption, and improving microarchitecture and periodontal condition. Context: An acidic hydrogen potential (pH) in an inflammatory condition in the periapical tissues may affect the properties of repair bioceramic cement. Aims: The aim of this study was to evaluate the effect of pH on the volumetric change of the ready-to-use bioceramic NeoPUTTY (NP) compared to the powder/liquid MTA Repair HP (MTAHP) after immersion in butyric acid (BA, pH 4.1) or phosphate-buffered saline (PBS, pH 7.35). Subjects and Methods: Dentin tubes filled with NP or MTAHP were scanned in micro-computed tomography (micro-CT) after 24 h. Then, the specimens were immersed in 1.5 mL of BA: NP/BA, MTAHP/BA or PBS: NP/PBS, MTAHP/PBS. After 7 days, new micro-CT scans were performed. The percentage of volumetric change (extremities and internal part) of the materials was assessed. Statistical Analysis Used: ANOVA/Tukey and Kruskal-Wallis tests were performed (α =0.05). Results: All materials showed a volumetric decrease after immersion in BA or PBS at the extremities in contact with the solutions. MTAHP/BA showed the highest volumetric loss. There was no difference in the volumetric change when the internal part of the materials was evaluated. Conclusions: An acid pH negatively affects the volumetric stability of MTAHP. Simple Summary: Poly (L-lactide-co-D, L-lactide) and PLDLLA have mechanical properties such as biodegradability, biocompatibility, thermoplastic capacity, and physical properties. In addition, their use eliminates the need for removal in subsequent surgeries and reduces the discomfort and risks associated with additional procedures. Given these advantages, a national company is currently developing an internal fixation device manufactured for PLDLLA, and it is essential to evaluate its bone incorporation process. The PLDLLA fixation device tested in this animal study was biocompatible and facilitated the formation of new bone around the device. Therefore, to avoid a second surgical procedure, this device is a highly effective option for those seeking a reliable and safe solution for bone fixation procedures. This study evaluated the bone incorporation process of a screw-shaped internal fixation device made of poly (L-lactide-co-D, L-lactide) (PLDLLA). Thirty-two male Wistar rats received 32 fixation devices (2 mm × 6 mm) randomly assigned to either the right or left tibia and one implant in each animal. After 7, 14, 28, and 42 days, the rats were euthanized and the specimens were subjected to microtomographic computed tomography (microCT) and histomorphometric analyses to evaluate bone interface contact (BIC%) and new bone formation (NBF%) in cortical and cancellous bone areas. The animals euthanized on days 28 and 42 were treated with calcein and alizarin red, and confocal LASER microscopy was performed to determine the mineral apposition rate (MAR). Micro-CT revealed a higher percentage of bone volume (p < 0.006), trabecular separation (p < 0.001), and BIC in the cortical (p < 0.001) and cancellous (p = 0.003) areas at 28 and 42 days than at 7 and 14 days. The cortical NBF at 42 days was greater than that at 7 and 14 days (p = 0.022). No statistically significant differences were observed in cancellous NBF or MAR at 28 and 42 days. Objective: This study investigated the effects of Lactobacillus fermentum BELF11 on periodontitis in mice (LIP). Methods: Sixty mice were randomly assigned to a control group (CTL), LIP/PBS group (LIP and PBS applied), or LIP/BELF11 group (LIP and L. fermentum BELF11 applied). For 14 days, PBS or L. fermentum BELF11 was applied twice daily to the mice in the LIP/PBS or LIP/BELF11 group, respectively. After 14 days, radiographic, histological, and pro‐inflammatory cytokine assessments were conducted. Results: The LIP/PBS and LIP/BELF11 groups demonstrated greater alveolar bone loss than the CTL group (p < 0.05). The LIP/BELF11 group showed significantly reduced alveolar bone loss on the mesial side compared to the LIP/PBS group. Histologically, the LIP/BELF11 group showed consistent patterns of connective tissue fiber arrangement, lower levels of inflammatory infiltration, less alveolar bone loss, and higher alveolar bone density than the LIP/PBS group, despite showing more signs of destruction than the CTL group. The LIP/BELF11 group also exhibited significantly lower levels of pro‐inflammatory cytokines than the LIP/PBS group. Conclusions: L. fermentum BELF11 inhibits alveolar bone loss and periodontitis progression by regulating pro‐inflammatory cytokine production. Background: Eccentric instruments have been proposed as more effective and less time‐consuming for endodontic instrumentation. Aim: To compare biomechanical outcomes of different instrumentation systems and time undertaken for instrumentation in resin prototypes. Design: Sixty standardized prototypes of mandibular second primary molars were instrumented according to the following systems: K‐files, ProTaper Next (PTN), XP‐endo Shaper (XPS), XP‐endo Finisher (XPF), XP–Clean (XPC), and Sequence Baby File (SBF; n = 10/each). Irrigation was performed with saline with simultaneous aspiration, and time spent was recorded. The prototypes were micro‐CT‐scanned before and after the instrumentation, and image sets were reconstructed and registered. Non‐instrumented areas, accumulated debris, removed root material volume, and canal transportation were quantified. Data were analyzed through ANOVA, the Kruskal–Wallis test, and the Wilcoxon signed‐rank test (α = 5%). Results: K‐files and SBF resulted in more instrumentation time (p <.05). SBF, XPC and PTN removed less root dentine (p <.05), but PTN left more untouched areas (p <.05). Accumulated debris were lower for XPC and SBF (p <.05). Canal transportation was similar among the groups. Conclusion: Rotary systems reduced instrumentation time, whereas SBF and XPC resulted in more conservative instrumentation, with less debris accumulation and non‐instrumented areas. Due to their excellent specific mechanical properties, closed cell elastomeric foams are the main element in the soles of running shoes to absorb repetitive shocks from strides and to release a maximum of the absorbed energy. However, these cellular materials are gradually damaged. To enhance their mechanical durability by slowing their damage kinetics, it is critical to understand their mechanical behaviour in fatigue. The objective of this work is thus to clarify the link between the microstructure and the fatigue properties of five commercial elastomeric foams used in the midsoles of running shoes. The 3D cellular structures of each foam were finely analysed using X-ray microtomography. Foam samples were then subjected to cyclic compression which were close to running conditions. During cycling, samples exhibited a rapid densification associated with noticeable decreases of (i) the stress levels required to deform them as well as (ii) the cushioning and (iii) the rebound properties. We show that the two midsoles filled with micro-sized mineral fillers present the highest specific mechanical properties during the first compression cycle and during fatigue. However, their damage kinetics and rebound properties could probably be improved by tuning the fillers-matrix compatibility. The lightest foam, being very porous and presenting process-induced tortuous cell walls, is the best cushioning system but exhibits high damage kinetics. The densest foam presents poor specific mechanical properties, but very slow damage kinetics. Its double hierarchical architecture probably prevents the occurrence of micro-cracks in the cell walls. Abstract: The minimally invasive endodontic access is not directly associated with tooth discoloration in the presence of bioceramic or epoxy resin-based root canal sealers. This study aimed to evaluate the influence of minimally invasive access and endodontic sealer composition on the color stability of endodontically-treated teeth, the restorative material adaptation, and the presence of remaining filling material in the pulp chamber. Endodontic access surgery was performed in maxillary central incisors, either through conservative or minimally invasive approaches, and the root was filled with AH Plus or Bio-C Sealer. The crown color was measured with a spectrophotometer at baseline and after root obturation, restoration, and specimen storage for one year in an oven. The occurrence of voids in the restoration and the remaining filling material was analyzed using micro-CT scans. The Yellowness Index (YI) and color changes (∆E00) were calculated after each color measurement. Data of micro-CT were submitted to 2-way ANOVA, and YI and ∆E00 were analyzed with repeated-measures ANOVA. Pair-wise comparisons were performed with Tukey's test (α = 0.05). The experimental conditions had no effect on the presence of the remaining material. The minimally invasive access associated with Bio-C Sealer resulted in more voids between the restoration and the remaining filling material. Only the evaluation time affected YI and ∆E00 values. Specimens became more yellow after filling and storage in the oven (the highest ∆E00 values). The present study showed that sealer and minimally invasive cavities are not associated with crown color stability following endodontic treatment.

Silicon-based anodes have emerged as a promising advancement in lithium-ion battery technology, offering significantly higher lithium storage capacities than traditional graphite. However, the volumetric expansion of silicon-anodes can swell by up to 300 % during lithiation-presents serious challenges to their structural integrity and electrochemical stability. This study investigates the internal structural dynamics of silicon anodes during lithiation and delithiation cycles. A novel cell design for a 18,650 cylindrical cell featuring micro-sized internal speckles within the silicon anode is presented. This design improves the simulation of electrochemical conditions and allows for precise displacement tracking, mitigating impacts on capacity and cycle performance while enhancing Digital Volume Correlation (DVC) analysis. The research prioritizes reducing scan time and radiation exposure in micro-CT assessments of Li-ion cells, and improves the accuracy of internal strain mapping via DVC. Displacement fields over three charging and discharging cycles are documented. Notably, uneven volumetric changes are observed, with local displacements reaching up to 35 µm in areas smaller than 2.5 mm in radius, which contracted during charging and expanded during discharging. This protocol offers insights into the relationship between electrode mechanics and cell performance, promoting non-destructive evaluations of internal structures in commercial cells.

Background The sea spiders (Pycnogonida Latreille, 1810) of the Hunsrück Slate (Lower Devonian, ~400 million years ago) are iconic in their abundance, exquisite pyritic preservation, and in their distinctive body plan compared to extant sea spiders (Pantopoda Gerstäcker, 1863). Consequently, the Hunsrück sea spiders are important in understanding the deep evolutionary history of Pycnogonida, yet they remain poorly characterised, impacting upon attempts to establish a time-calibrated phylogeny of sea spiders. Methods Here, we investigated previously described and new material representing four of the five Hunsrück pycnogonids: Flagellopantopus blocki Poschmann & Dunlop, 2006; Palaeoisopus problematicus Broili, 1928; Palaeopantopus maucheri Broili, 1929; and Pentapantopus vogteli Kühl, Poschmann & Rust, 2013; as well as a few unidentified specimens. Using X-ray microtomography and Reflectance Transformation Imaging, we describe new fossils, provide evidence for newly revealed anatomical features, and interpret these data in comparison to extant species. We also reinterpret the previously published illustration of the (probably lost) holotype of Palaeothea devonica Bergström, Stürmer & Winter, 1980. Results We provide the first detailed description of the cephalic appendages of Palaeoisopus problematicus and revise the interpretation of the organisation of its ocular tubercle. Furthermore, we provide new insights into the structure of the legs and the proboscis of Palaeopantopus maucheri, the first description of the body of Flagellopantopus blocki and describe a new specimen of Pentapantopus vogteli, demonstrating that it had eight legs, in contrast to previous interpretations. We argue that, contrary to previous suggestions, Palaeothea devonica probably had a different body plan from extant pantopods. We discuss the ecological traits of the Hunsrück pycnogonids based on their morphological adaptations, and conclude that there is no compelling evidence of Pantopoda in the Devonian. Through comparative interpretation of the legs as well as general morphology, we can divide the Hunsrück pycnogonids into two morphological groups, while Pantopoda constitutes a third morphological group.

Abstract Background Chondrosarcomas are rare malignant bone tumors diagnosed by analyzing radiological images and histology of tissue biopsies and evaluating features such as matrix calcification, cortical destruction, trabecular penetration, and tumor cell entrapment. Methods We retrospectively analyzed 16 cartilaginous tumor tissue samples from three patients (51-, 54-, and 70-year-old) diagnosed with a dedifferentiated chondrosarcoma at the femur, a moderately differentiated chondrosarcoma in the pelvis, and a predominantly moderately differentiated chondrosarcoma at the scapula, respectively. We combined a hematein-based x-ray staining with high-resolution three-dimensional (3D) microscopic x-ray computed tomography (micro-CT) for nondestructive 3D tumor assessment and tumor margin evaluation. Results We detected trabecular entrapment on 3D micro-CT images and followed bone destruction throughout the volume. In addition to staining cell nuclei, hematein-based staining also improved the visualization of the tumor matrix, allowing for the distinction between the tumor and the bone marrow cavity. The hematein-based staining did not interfere with further conventional histology. There was a 5.97 ± 7.17% difference between the relative tumor area measured using micro-CT and histopathology (p = 0.806) (Pearson correlation coefficient r = 0.92, p = 0.009). Signal intensity in the tumor matrix (4.85 ± 2.94) was significantly higher in the stained samples compared to the unstained counterparts (1.92 ± 0.11, p = 0.002). Conclusions Using nondestructive 3D micro-CT, the simultaneous visualization of radiological and histopathological features is feasible. Relevance statement 3D micro-CT data supports modern radiological and histopathological investigations of human bone tumor specimens. It has the potential for being an integrative part of clinical preoperative diagnostics. In this study, digital light processing (DLP) was utilized to generate 3D-printed blends composed of photosensitive acrylate-modified polylactic acid (PLA) resin mixed with varying weight ratios of lignin extracted from softwood, typically ranging from 5 wt% to 30 wt%. The microstructure of these 3D-printed blends was examined through X-ray microtomography. Additionally, the tensile mechanical properties of all blends were assessed in relation to the weight ratio and post-curing treatment. The results suggest that post-curing significantly influences the tensile properties of the 3D-printed composites, especially in modulating the brittleness of the prints. Furthermore, an optimal weight ratio was identified to be around 5 wt%, beyond which UV light photopolymerization experiences compromises. These findings regarding acrylate-modified PLA/lignin blends offer a cost-effective alternative for producing 3D-printed bio-sourced components, maintaining technical performance in reasonable-cost, low-temperature 3D printing, and with a low environmental footprint. Void defect is prevalent during the composite manufacturing process and may reduce the mechanical properties of composite structures, while limited information about three‐dimensional void distribution and evolution can be found due to the difficulties in void observation. In this work, void defects at different positions were detected and analyzed using x‐ray microtomography (XRM), and the random forest algorithm was implemented for segmenting components of CFRP samples. Results showed a trend of gradual rising porosity in the axial direction, with an increase of ~250% of porosity at the outlet of resin injection compared to the one at the inlet during the composite manufacturing process. The voids mainly featured with microscale between carbon fibers, with less distribution between fiber bundles and resin‐rich zones. Void defects achieved aggregation at the middle position of manufactured laminate, which is probably related to pressure interception. In the axial direction, the proportion of large‐diameter voids gradually increased, and a large volume of connected voids appeared near the outlet. Due to the shear deformation of resin and fiber and the pressure gradient effect, most of the voids showed stripe shape. The spatial orientation of voids was predominantly along the direction of resin flow and fluctuated due to fiber curl. Background: Optimal dimensional stability is required for successful root canal treatment. A sealant called EndoSeal mineral trioxide aggregate (MTA) was recently introduced to the market due to its favorable physical and chemical properties. On the other hand, AH Plus (AHP) is considered the gold-standard seal. Materials and Methods: In this ex vivo quasi-experimental study, 24 single-canal premolars extracted from humans were cleaned and shaped with a motorized and rotary file, then that is divided into two groups. The teeth of each group were filled with gutta F3 and each type of sealant. The teeth were scanned by a micro-computed tomography device after 24 h. After 7 days of storage in phosphate-buffered saline solution, the samples were re-scanned. Data were analyzed using SPSS software (version 2I). Descriptive data were presented as frequency, percentage, mean, and standard deviation. The Shapiro-Wilk and Kolmogorov-Smirnov tests were used to investigate the normality of the data.The Mann-Whitney test was used to compare the two groups, and the differences were ultimately not significant. The level of significance was set at 0.05 (P < 0.05). Results: The mean differences between sealer volumes before and after the intervention were not significantly different between the two groups indicating that the EndoSeal MTA sealer is not inferior to the gold-standard root canal sealer, AHP. Synchrotron X‐ray microtomography (XMT) was used to measure total air‐water interfacial areas (Aaw) as a function of water saturation (Sw) for several soils that comprise a range of physical and geochemical properties. Measurements were also conducted for glass beads and quartz sands for comparison. Apparent near‐linear Aaw‐Sw relationships are observed for the three sands and the three sandy soils. In contrast, the measured interfacial areas for two soils that contain greater proportions of silt and clay are strongly nonlinear functions of water saturation. The greater degree of nonlinearity observed for these two soils is due to their much greater particle‐size distributions (i.e., uniformity coefficients) and their concomitant greater range in pore sizes. Interfacial areas determined with the thermodynamic method were used to benchmark the XMT measurements. XMT‐measured interfacial areas compare well to the thermodynamic‐determined values for the sands and sandy soils. In contrast, the XMT‐measured interfacial areas for the two soils with larger particle‐size distributions are not fully congruent with the thermodynamic‐determined values. Both of these soils have large fractions of pore space comprising nominal pore diameters smaller than the resolution of the XMT imaging. These results suggest that air‐water interfacial area may not always be fully characterized by standard XMT for soils with large particle‐size distributions. This work emphasizes an innovative approach utilizing 3D imaging technology based on synchrotron radiation to assess the microstructure of second-phase iron particles and the porous structure within 3D-printed PLA/magnetic iron composites at different printing angles. The study examines how these observations relate to the material's ductility when processed using fused filament fabrication. In particular, this study examines the impact of one processing parameter, specifically the printing angle, on the microstructure and mechanical behaviour of a polylactic acid (PLA)–iron (PLI) composite designed for magnetic actuation. Fused filament fabrication is employed to produce PLI tensile specimens, with varied printing angles to create different layups. X-ray microtomography is utilized to analyse the microstructure, while tensile mechanical properties are evaluated for all composites, with findings discussed in relation to printing angle conditions. Scanning Electron Microscopy is used to examine the fractography of broken specimens. Results indicate that the printing angle significantly influences the tensile properties and mechanical anisotropy of 3D-printed PLI composites, with an optimal 45°/45° layup enhancing tensile performance. These findings suggest that 3D-printed PLI composites offer a cost-efficient means of producing bio-sourced, light-adaptive materials with intricate magnetic actuation capabilities. Abstract Both root canal sealer-based and supplementary protocols may influence removal of filling material during endodontic retreatment. Mesial root canals of extracted mandibular molars were prepared using HyFlex EDM 25/.08, and filled with a calcium silicate sealer (Bio-C Sealer), or an epoxy resin (AH Plus), using the single cone technique (n = 12). Retreatment was performed using ProDesign Logic (PDL) RT and PDL 35/.05. The specimens were randomly divided into two experimental groups (n = 12), and the sealers were distributed similarly. A supplementary protocol was performed with PDL 50/.01 or XP-endo Finisher. Root canal transportation and volume, in addition to the remaining filling material percentage were evaluated using high-resolution (5 µm voxel size) micro-CT. Statistical analysis was performed using t-tests (α = 0.05). Root canals filled with AH Plus presented high residual filling material (p < 0.05). Both protocols decreased residual volume of filling material in the apical third (p < 0.05). PDL 50/.01 increased the apical root canal volume (p < 0.05). No difference was observed between the systems regarding canal transportation (p > 0.05). In conclusion, AH Plus is more difficult to remove from the apical third than Bio-C Sealer. PDL 50/.01 and XP-endo Finisher enabled greater removal of filling materials in the apical third, in the retreatment of curved root canals, without promoting apical transport.

The distribution of fibers in injection-molded thermoplastic-reinforced parts is known to significantly affect both the final mechanical properties and the appearance of defects related to undesired shrinkages and warpages. Even if the numerical modelling of the process is in the state of the art, the selection criteria of the model for the fiber orientation predictability and the influence of the models' parameters are not yet clearly understood. The complexity of the matter increases further considering that, inside the same injection-molded part, the fiber orientation distribution could differ from one region to another depending on the local melt shear flow type. In this context, the aim of this study is to investigate the orientation of fibers in regions of non-simple shear flow in an injection-molded short glass fiber-reinforced part. X-ray microcomputed tomography is used to experimentally investigate fiber distribution and orientation. Furthermore, a number of numerical simulations of the injection molding process are performed in Moldflow® by varying the mesh type (2D/3D), the selected predictive model, and the models' coefficients. The main findings indicate that, in regions with non-simple shear flow, a 2D mesh fails to capture accurate fiber orientation. A 3D mesh is essential for reliable predictions. MRD-specific parameters from the literature reduce prediction error by 37.5% compared to Moldflow's default MRD parameters. Regarding the RSC model, both Wang's optimal retarding rate parameter and AMI's default retarding rate parameter result in similar 5% prediction errors. Background: This study aimed to compare the apical transportation and centering ability of One Curve, HyFlex EDM, and EdgeFile X1 in curved mesiobuccal and mesiolingual canals of mandibular first molars. Materials and Methods: In this in vitro experimental study, 60 mesiobuccal and mesiolingual canals of the mandibular first molars with a minimum length of 19 mm and 25°-40° curvature were randomly divided into three groups (n = 20) for root canal preparation with One Curve, HyFlex EDM, and EdgeFile X1. After access cavity preparation and confirming the glide path, the baseline micro-computed tomography (micro-CT) scans were obtained, and the root canals were instrumented with the respective systems according to the manufacturers' instructions. Apical transportation and centering ability were assessed at 1, 3, 5, and 7 mm from the apex by comparing pre- and postinstrumentation micro-CT scans. One-way ANOVA, independent t-test, and Duncan's post hoc test were used to statistically compare the groups, and data were analyzed by SPSS version 24 (alpha = 0.05). Results: The three groups were not significantly different regarding apical transportation at 5 and 7 mm from the apex (P > 0.05). At 1 mm level, One Curve caused significantly lower apical transportation; while, at 3 mm level, HyFlex EDM resulted in significantly higher apical transportation (P < 0.05). No significant difference was noted in the centering ability of the three groups at 1, 3, and 5 mm from the apex (P > 0.05). At 7 mm level, EdgeFile X1 showed significantly lower centering ability (P < 0.05). Conclusion: One Curve caused lower canal transportation in the apical third compared with EdgeFile and HyFlex EDM, but no significant difference was noted among the three in the coronal third of the roots. In the Malvaceae family, dynamic solar tracking by leaves is actuated by the deformation of the pulvinus, a thickened region at the leaf blade–petiole junction. While the internal structure is believed to play a crucial role in this process, experimental verification has been challenging due to technical limitations. To address this gap, we developed a semi-automated workflow, which integrates data analysis and image processing to simultaneously analyze the shape and internal structure of a Malvaceae pulvinus using X-ray microtomography. Firstly, we found that kenaf (Hibiscus cannabinus L.), a Malvaceae species with curved pulvini, exhibited solar-tracking leaf movement and selected it as a model system. We employed diffusible iodine-based contrast-enhanced computed tomography to visualize the internal structure of the kenaf pulvinus. Analysis of the pulvini's shape revealed variations in pulvinus morphology, yet plausible prediction of the centerline was accomplished using polar polynomial regression. Upon slicing the pulvini perpendicular to the centerline, we observed distinct gray value gradients along the proximo-distal and adaxial-abaxial axes, challenging threshold-based tissue segmentation. This workflow successfully generated three modified 3D images and derived quantitative parameters. Using these quantitative parameters, we conducted network analysis and found the linkage between the size-normalized cortex cross-sectional area and curvature. Polynomial least absolute shrinkage and selection operator (LASSO) regression revealed the relationship between the size-normalized cortex cross-sectional area and curvature commonly in all three tested samples. Predicting the permeability of different regions of foundry cores and molds with complex geometries will help control the regional outgassing, enabling better defect prediction in castings. In this work, foundry cores prepared with different bulk properties were characterized using X-ray microtomography, and the obtained images were analyzed to study all relevant grain and pore parameters, including but not limited to the specific surface area, specific internal volume, and tortuosity. The obtained microstructural parameters were incorporated into prevalent models used to predict the fluid flow through porous media, and their accuracy is compared with respect to experimentally measured permeability. The original Kozeny model was identified as the most suitable model to predict the permeability of sand molds. Although the model predicts permeability well, the input parameters are laborious to measure. Hence, a methodology for replacing the pore diameter and tortuosity with simple process parameters is proposed. This modified version of the original Kozeny model helps predict permeability of foundry molds and cores at different regions resulting in better defect prediction and eventual scrap reduction. Amelogenesis imperfecta (AI) comprises a group of rare, inherited disorders with abnormal enamel formation. Ameloblastin (AMBN), the second most abundant enamel matrix protein (EMP), plays a critical role in amelogenesis. Pathogenic biallelic loss-of-function AMBN variants are known to cause recessive hypoplastic AI. A report of a family with dominant hypoplastic AI attributed to AMBN missense change p.Pro357Ser, together with data from animal models, suggests that the consequences of AMBN variants in human AI remain incompletely characterized. Here we describe 5 new pathogenic AMBN variants in 11 individuals with AI. These fall within 3 groups by phenotype. Group 1, consisting of 6 families biallelic for combinations of 4 different variants, have yellow hypoplastic AI with poor-quality enamel, consistent with previous reports. Group 2, with 2 families, appears monoallelic for a variant shared with group 1 and has hypomaturation AI of near-normal enamel volume with pitting. Group 3 includes 3 families, all monoallelic for a fifth variant, which are affected by white hypoplastic AI with a thin intact enamel layer. Three variants, c.209C>G; p.(Ser70*) (groups 1 and 2), c.295T>C; p.(Tyr99His) (group 1), and c.76G>A; p.(Ala26Thr) (group 3) were identified in multiple families. Long-read AMBN locus sequencing revealed these variants are on the same conserved haplotype, implying they originate from a common ancestor. Data presented therefore provide further support for possible dominant as well as recessive inheritance for AMBN -related AI and for multiple contrasting phenotypes. Abstract This study aimed to evaluate volumetric polymerization shrinkage, degree of conversion and Vickers hardness of four bulk-fill resin composites light-activated with their dedicated light curing units (LCUs). Four groups were evaluated, according to the type of composite and curing mode: Tetric EvoCeram Bulk-fill (TEBO) and Tetric EvoFlow Bulk-fill (TEBF) were light-activated with Bluephase Style 20i (20s, in high-mode), while Tetric Powerfill (TEPO) and Tetric Powerflow (TEPF) were light-activated with Bluephase PowerCure (3s). Volumetric polymerization shrinkage test (n = 6) was performed in standardized box-shaped class-I cavities of extracted third molars (4 x 4 x 4 mm). Teeth were scanned before and after resin composite application by micro-computed tomography, and acquired data were evaluated with Amira software. Degree of conversion (n = 5) was evaluated at the top and bottom surfaces of composite cylindric samples (4 mm diameter, 4 mm thickness) using an FT-IR spectrometer (spectra between 1,500 and 1,800 cm-1, 40 scans at a resolution of 4 cm-1). Three Vickers indentations (50 g / 15 s), spaced 500 μm apart, were performed on the top and bottom composite surfaces and averaged. One-way ANOVA was used for data evaluation. TEPF showed the lowest volumetric polymerization shrinkage (p < 0.05), while the other composites were not significantly different within each other (p > 0.05). All materials presented a significant decrease in degree of conversion and Vickers hardness when compared top to bottom surfaces (p < 0.05). Bottom to top surface ratios for degree of conversion ranged from 0.8 (TEBO and TEPO) to 0.9 (TEBF and TEPF), and from 0.4 (TEPO) to 0.7 (TEBF and TEPF) for hardness. In conclusion, resinous materials present a decrease in hardness and degree of conversion from top to bottom even when a higher power is used, while the flowable material TEPF showed the lowest volumetric shrinkage values compared to the other materials.

Abstract Acidic pH can modify the properties of repair cements. In this study, volumetric change and solubility of the ready-to-use bioceramic repair cement Bio-C Repair (BCR, Angelus, Londrina, PR, Brazil) were evaluated after immersion in phosphate-buffered saline (PBS) (pH 7.0) or butyric acid (pH 4.5). Solubility was determined by the difference in initial and final mass using polyethylene tubes measuring 4 mm high and 6.70 mm in internal diameter that were filled with BCR and immersed in 7.5 mL of PBS or butyric acid for 7 days. The volumetric change was established by using bovine dentin tubes measuring 4 mm long with an internal diameter of 1.5 mm. The dentin tubes were filled with BCR at 37°C for 24 hours. Scanning was performed with micro-computed tomography (micro-CT; SkyScan 1176, Bruker, Kontich, Belgium) with a voxel size of 8.74 µm. Then, the specimens were immersed in 1.5 mL of PBS or butyric acid at and 37 °C for 7 days. After this period, a new micro-CT scan was performed. The aim of this work was to develop a novel method for studying the 3D morphology of agglomerates obtained by spherical agglomeration. It has been found, that the combination of shock-freezing the samples in a mixture of ethanol and dry ice followed by an X-ray microtomography measurement leads to useful results. Hereby, the image quality for low absorbing material like the used graphite was enhanced by propagation-based X-ray microtomography, which results in phase contrast images. We also discuss our 3D image post-processing routine, which is used to determine the morphology parameters sphericity, fractal dimension and packing density. Furthermore, a two-dimensional kernel density estimation is used to calculate the joint probability density of agglomerate size and the morphology parameter. In future, this method will be used to determine the morphological behaviour of agglomerates during the different phases of spherical agglomeration.
