1. Potential Chromospheric Evaporation in A M-dwarf's Flare Triggered by Einstein Probe Mission

Abstract: Although flares from late-type main-sequence stars have been frequently detected in multi-wavelength, the associated dynamical process has been rarely reported so far. Here, we report follow-up observations of an X-ray transient triggered by WXT onboard the Einstein Probe at UT08:45:08 in 2024, May 7. The photometry in multi-bands and time-resolved spectroscopy started at 3 and 7.5 hours after the trigger, respectively, which enables us to identify the transient as a flare of the M-dwarf 2MASS J12184187-0609123. The bolometric energy released in the flare is estimated to be $\sim10^{36}\ \mathrm{erg}$ from its X-ray light curve. The H$\alpha$ emission-line profile obtained at about 7 hours after the trigger shows an evident blue asymmetry with a maximum velocity of $200-250\ \mathrm{km\ s^{-1}}$. The blue wing can be likely explained by the chromospheric temperature (cool) upflow associated with chromospheric evaporation, in which the mass of the evaporating plasma is estimated to be $1.2\times10^{18}$g. Abstract: Massive stars end their life as core-collapse supernovae, amongst which some extremes are Type Ic broad-lined supernovae associated with long-duration gamma-ray bursts (LGRBs) having powerful relativistic jets. Their less-extreme brethren make unsuccessful jets that are choked inside the stars, appearing as X-ray flashes or low-luminosity GRBs. On the other hand, there exists a population of extragalactic fast X-ray transients (EFXTs) with timescales ranging from seconds to thousands of seconds, whose origins remain obscure. Known sources that contribute to the observed EFXT population include the softer analogs of LGRBs, shock breakouts of supernovae, or unsuccessful jets. Here, we report the discovery of the bright X-ray transient EP240414a detected by the Einstein Probe (EP), which is associated with the Type Ic supernova SN 2024gsa at a redshift of 0.401. The X-ray emission evolution is characterised by a very soft energy spectrum peaking at < 1.3 keV, which makes it distinct from known LGRBs, X-ray flashes, or low-luminosity GRBs. Follow-up observations at optical and radio bands revealed the existence of a weak relativistic jet that interacts with an extended shell surrounding the progenitor star. Located on the outskirts of a massive galaxy, this event reveals a new population of explosions of Wolf-Rayet stars characterised by a less powerful engine that drives a successful but weak jet, possibly owing to a progenitor star with a smaller core angular momentum than in traditional LGRB progenitors., Comment: 43 pages, 9 figures, 4 tables, submitted. 3. Homological properties of homologically smooth connected cochain DGAs

Abstract: Assume that $\mathscr{A}$ is a connected cochain DG algebra. We show that $\mathscr{A}$ is homologically smooth and Gorenstein if and only if its $\mathrm{Ext}$-algebra $H(R\Hom_{\mathscr{A}}(\mathbbm{k},\mathbbm{k}))$ is a Frobenius graded algebra. Moreover, $\mathscr{A}$ is Calabi-Yau if and only if the $\mathrm{Ext}$-algebra $H(R\Hom_{\mathscr{A}}(\mathbbm{k},\mathbbm{k}))$ is a symmetric Frobenius graded algebra. These generalize the corresponding results in \cite{HW1} and \cite{HM}, where the additional Koszul hypothesis is needed.

4. Homologically smooth connected cochain DGAs

Abstract: Let $\mathscr{A}$ be a connected cochain DG algebra such that $H(\mathscr{A})$ is a Noetherian graded algebra. We give some criteria for $\mathscr{A}$ to be homologically smooth in terms of the singularity category, the cone length of the canonical module $k$ and the global dimension of $\mathscr{A}$. For any cohomologically finite DG $\mathscr{A}$-module $M$, we show that it is compact when $\mathscr{A}$ is homologically smooth. If $\mathscr{A}$ is in addition Gorenstein, we get $$\mathrm{CMreg}M = \mathrm{depth}_{\mathscr{A}}\mathscr{A} + \mathrm{Ext.reg}\, M<\infty,$$ where $\mathrm{CMreg}M$ is the Castelnuovo-Mumford regularity of $M$, $\mathrm{depth}_{\mathscr{A}}\mathscr{A}$ is the depth of $\mathscr{A}$ and $ \mathrm{Ext.reg}\, M$ is the Ext-regularity of $M$., Comment: arXiv admin note: substantial text overlap with arXiv:1301.4382

Abstract: Long gamma-ray bursts (GRBs) are believed to originate from core collapse of massive stars. High-redshift GRBs can probe the star formation and reionization history of the early universe, but their detection remains rare. Here we report the detection of a GRB triggered in the 0.5--4 keV band by the Wide-field X-ray Telescope (WXT) on board the Einstein Probe (EP) mission, designated as EP240315a, whose bright peak was also detected by the Swift Burst Alert Telescope and Konus-Wind through off-line analyses. At a redshift of $z=4.859$, EP240315a showed a much longer and more complicated light curve in the soft X-ray band than in gamma-rays. Benefiting from a large field-of-view ($\sim$3600 deg$^2$) and a high sensitivity, EP-WXT captured the earlier engine activation and extended late engine activity through a continuous detection. With a peak X-ray flux at the faint end of previously known high-$z$ GRBs, the detection of EP240315a demonstrates the great potential for EP to study the early universe via GRBs., Comment: 41 pages, 8 figures, 7 tables

However, the mechanisms through which CAFs influence the progression of HCC remain incompletely understood.Methods: Single-cell RNA sequencing datasets (GSE158723 and GSE112271) were retrieved from the Gene Expression Omnibus (GEO) database at the National Center for Biotechnology Information (NCBI) and analyzed using R software. Our analysis suggested that CAFs may promote liver cancer cell development, possibly through the interaction of pleiotrophin (PTN) and syndecan-2 (SDC2). Clinical samples from HCC patients were collected and processed into frozen sections and single-cell suspensions for Masson staining, immunofluorescence staining, and flow cytometry. Additionally, Huh7 liver cancer cells and LO2 normal liver cells were cultured and subjected to immunofluorescence assays using cell slides.Results: The proportion of CAFs in cancerous tissues was higher than in adjacent non-cancerous tissues, and pleiotrophin (PTN) expression was elevated in cancer tissues compared to adjacent tissues. These findings aligned with the results of the single-cell RNA sequencing (scRNA-seq) analysis. Furthermore, SDC2 expression was significantly upregulated in Huh7 liver cancer cells compared to LO2 normal liver cells.Discussion: This study suggests that CAFs may contribute to HCC progression via the PTN/SDC2 signaling pathway. We aimed to prospectively explore the associations between serum prealbumin levels and adverse clinical outcomes after ischemic stroke in a large-scale cohort study.Methods: We measured serum prealbumin levels among 6609 ischemic stroke patients admitted at Minhang hospital. The primary outcome was composite of death and major disability (modified Rankin Scale [mRS] score ≥ 3) at 3 months after stroke onset, and secondary outcomes included death and the ordered 7-level categorical score of mRS.Results: During 3 months of follow-up, a total of 2118 patients developed the primary outcome. After multivariable adjustment, high prealbumin levels were associated with a decreased risk of primary outcome (odds ratio, 0.71; 95% CI, 0.59– 0.85; Ptrend< 0.0001) when 2 extreme quartiles were compared. Each unit increase of log-transformed prealbumin was associated with a 42% (95% CI, 28-53%) decreased risk of primary outcome. There was a better shift in the distribution of mRS score at 3 months with higher quartiles of serum prealbumin in ischemic stroke patients (Ptrend< 0.0001). Multivariable-adjusted spline regression model showed a linear relationship between prealbumin and the risk of primary outcome (P for linearity = 0.0036).Conclusion: High serum prealbumin level was independently associated with decreased risks of adverse clinical outcomes among ischemic stroke patients. Our findings suggested that prealbumin may be a valuable prognostic biomarker and indicated the importance of keeping nourished in the daily life.Keywords: prealbumin, ischemic stroke, prognosis, cohort study

Based on the results of pneumatic reduction, the patients were divided into two groups: the successful reduction group (control group) and the failed reduction group (operation group). The total sample was randomly divided into a training set and a validation set in a 7:3 ratio. Logistic regression analysis was performed to establish a predictive model for surgical risk.Results: A total of 1041 samples were included in this study, with 852 in the control group and 189 in the operation group. Among the total sample, 728 cases were used for training and 313 cases were used for validation. Logistic regression analysis of the training set identified age, time of abdominal pain, presence or absence of hematostoecia, C-reactive protein value from blood test on admission, and nested position indicated by B-ultrasound as independent predictors of intussusception intervention. Based on the five independent risk factors identified through multivariate logistic regression, a nomogram was successfully constructed to predict the failure of resetting by air enema under X-ray.Conclusion: A nomogram was developed to predict the need for surgical intervention after intussusception pneumatic reduction in children. The nomogram was based on clinical risk factors including age, time of abdominal pain, presence or absence of blood in stool, value of C-reactive protein in blood test on admission, and nested position indicated by B-ultrasound. The association rules between the acupoints were further described using SPSS Modeler 18.0.Results: A total of 67 papers were included, involving 51 acupoints for Guasha against primary headaches. The most commonly used acupoints were Fengchi, Baihui, Taiyang, Shuaigu, Tianzhu, and Hegu. The goal was to enhance treatment guidance through precise subtype delineation.Methods: Study Cohort: This study encompassed a cohort of 119 patients diagnosed with MMRd-EC between 2015 and 2022. Analyses using t-tests and Mann–Whitney U-tests were performed to assess prognostic markers and peripheral blood immune cell profiles in patients with MutS deficiency (MutS-d) versus those with MutL deficiency (MutL-d). Logistic regression analysis was used to identify independent risk factors. Bioinformatics Analysis: An online database was used to assess the prognostic implications, immune cell infiltration, and immune checkpoint involvement associated with the deficiency of MutS versus MutL in EC.Results: Patients with MutL-d exhibited heightened risk factors, including elevated cancer grade and increased myometrial invasion, leading to poorer prognosis and shorter overall survival and progression-free survival. Regarding systemic immune status, patients with MutL-d demonstrated decreased peripheral blood lymphocyte percentage, lymphocyte count, and CD8+ T cell percentage. For local immunity, the infiltration of natural killer cells, CD8+ T cells, and cytotoxic T lymphocytes in the tumor tissue was reduced in patients with MutL-d. Additionally, patients with MutL-d exhibited lower expression of immune checkpoint markers. The composition of immune subtypes and survival outcomes also indicate that patients with MutL-d have a poorer immune status and prognosis than the patients with MutS-d.Conclusion: Patients with MMRd-EC can be subclassified according to MutS or MutL deficiency. Patients with MutS-d exhibited better immune status, prognosis, and immunotherapy benefits than those with MutL-d. The association between epithelial cell heterogeneity, prognosis, and immune response in this cancer remains uncertain. This study aimed to investigate epithelial cell heterogeneity in ESCC and develop a predictive risk model using the identified cell types.Methods: Single-cell RNA sequencing (scRNA-seq) and differential ESCC gene data were accessed from the Gene Expression Omnibus. Functional enrichment analysis, inferCNV, cell development trajectories, and intercellular communication were analyzed following epithelial cell characterization. Differentially expressed ESCC (n = 773) and epithelial cell marker genes (n = 3407) were intersected to obtain core genes, and epithelial cell-related prognostic genes were identified. LASSO regression analysis was used to construct a prognostic model. The external dataset GSE53624 was used to further validate the stability of the model. Drug sensitivity predictions, and immune cell infiltration were analyzed. Molecular docking clarified the possible therapeutic role of β-sitosterol in ESCC. Finally, wound healing assay, cell colony, and transwell assay were constructed to detect the effects of the core gene PDLIM2 on ESCC cell proliferation, invasion, and migration.Results: Eight cell clusters were identified, and epithelial cells were categorized into tumor and paratumor groups. The tumor group possessed more chromosomal variants than the paratumor group. Epithelial cells were associated with multiple cell types and significantly correlated with the Wnt, transforming growth factor, and epidermal growth factor signaling pathways. From 231 intersected genes, five core genes were screened for use in the risk model: CTSL, LAPTM4B, MYO10, NCF2, and PDLIM2. These genes may contribute to the cancerous transformation of normal esophageal epithelial cells and thereby act as biomarkers and potential therapeutic targets in patients with ESCC. β-Sitosterol furthermore displayed excellent docking potential with these genes. Pregnant women who delivered at two tertiary referral hospitals from January 2013 to December 2022 and were ultimately pathologically diagnosed with placenta accreta spectrum were included. They were divided into three groups based on different placental locations (anterior, posterior, and lateral wall/fundus). The differences in prenatal diagnosis, clinical characteristics, and perinatal outcomes among the three groups were compared.Results: There were 115,470 deliveries in a ten-year period at the two hospitals, and 118 case patients were confirmed to have a pathologically diagnosed placenta accreta spectrum. The posterior placenta group had a lower rate of placenta previa (76.9% vs 94.9% vs 100%, p< 0.05) and a higher gestational age at delivery (36.4± 2.45 vs 34.91± 1.76 vs 34.31± 3.41, p< 0.05) compared to the other two groups. The anterior placenta group had a significantly higher rate of invasive (increta/percreta) form placenta accreta spectrum (81.4% vs 36.5% vs 28.6%, p< 0.05) and planned cesarean section (96.6% vs 80.8% vs 71.4%, p< 0.05) compared to the other two groups. In terms of prenatal diagnosis, the anterior placenta group had a significantly higher rate of placenta accreta spectrum prenatal suspicion rate compared to the other two groups (86.4% vs 36.5% vs 57.1%, p< 0.05). The posterior placenta group had a lower rate of preoperative abdominal aortic balloon placement compared to the other two groups (5.8% vs 28.8% vs 28.6%, p< 0.05). There were no statistically significant differences among the three groups in primary perinatal outcomes, though the anterior placenta group had a longer postoperative hospital stay.Conclusion: The prenatal diagnosis rate and proportion of invasive form of placenta accreta spectrum occurring in non-anterior placenta are relatively lower than anterior placenta. DRA typically occurs in pregnant and postpartum women. We aimed to determine the risk factors and patient-reported outcomes (PROs) of DRA in Chinese postpartum women.Methods: This observational study was conducted in Hangzhou Hospital of Traditional Chinese Medicine, and involved 534 women who filled out the following risk-factor and PRO questionnaires: SF-MPQ-2, SF-ICIQ, LDQ, EPDS, MBIS, HerQles, and SF-36 (all Chinese versions). The inter-recti distance was measured by palpation. Statistical analyses were performed using SPSS v25.0 software and the Mann–Whitney U-test, chi-square test, binary logistic regression analysis (for risk factors of DRA), and the Kendall and Spearman tests (for correlation analysis).Results: After childbirth, 78.1% (417/534) of the enrolled women had DRA. Abdominal surgery (P = 0.002), number of pregnancies (P = 0.035), parity (P = 0.012), number of births (P = 0.02), fetal birth weight (P = 0.014), and waist-to-hip ratio in the supine position (P = 0.045) significantly differed between the DRA and non-DRA groups. Caesarean delivery was an independent risk factor for DRA. The PROs were significantly worse in the DRA group than in the non-DRA group.Conclusion: Caesarean delivery was an independent risk factor for DRA. Multivariable logistic regression analysis was performed to explore the associated factors of high PR knowledge scores (≥ 18 points).Results: From the 858 valid questionnaires, the routine implementation of PR was only reported for 16.95% of physicians. The main reason hindering the implementation of PR for patients was the limited knowledge and awareness of PR among the physicians involved (69.1%). A total of 618 and 240 physicians had high and low knowledge scores, respectively. Multivariable analysis suggests that the self-perception of PR knowledge (OR = 1.89, 95% CI: 1.32– 2.771, P = 0.001) was independently associated with high knowledge scores, while having no theoretical knowledge of PR was associated with poor knowledge scores (OR = 0.43, 95% CI: 0.26– 0.72, P = 0.001).Conclusion: Inadequate knowledge of pulmonary rehabilitation is evident among physicians who are involved in pulmonary disease management in China. ApoVs have shown promising therapeutic potential for inflammation, tumors, immune disorders, and tissue regeneration. Abstract: Objective: To investigate whether immediate frozen–thawed embryo transfer (FET) in the next month following coronavirus disease 2019 (COVID‐19) recovery affects ongoing pregnancy outcome. Methods: This was a retrospective cohort study carried out at a university‐affiliated reproductive medicine center. The study group (post‐COVID‐19 group) comprised women who were affected by COVID‐19 in December 2022 and immediately underwent FET in January 2023 after recovery, with transferred embryos not exposed to the infection. The control group comprised women treated during the pre‐COVID‐19 period (January 2019). Multivariable logistic regression analysis and a propensity score matching (PSM) approach were used to control for potential confounders and selection bias. Results: A total of 200 women were included in the post‐COVID‐19 group and 641 women were enrolled in the control group. The rate of ongoing pregnancy was comparable between the study cohorts in both the unadjusted and confounder‐adjusted logistic regression models. Other reproductive outcomes, including the odds of a positive pregnancy test, implantation, clinical pregnancy and early pregnancy loss, were similar between the comparison groups. PSM models further confirmed the lack of significant differences in pregnancy outcome between the post‐COVID‐19 group and the control group. Conclusions: Among patients affected by COVID‐19 for whom the transferred embryos were generated prior to infection, an immediate FET cycle in the next month after recovery does not seem to compromise ongoing pregnancy outcome. Thus, women who have frozen embryos from preinfection cycles should be counseled and encouraged to undergo FET as soon as possible after COVID‐19 recovery. © 2024 International Society of Ultrasound in Obstetrics and Gynecology. Abstract: The core-collapse supernova (CCSN) is considered one of the most energetic astrophysical events in the universe. The early and prompt detection of neutrinos before (pre-SN) and during the supernova (SN) burst presents a unique opportunity for multi-messenger observations of CCSN events. In this study, we describe the monitoring concept and present the sensitivity of the system to pre-SN and SN neutrinos at the Jiangmen Underground Neutrino Observatory (JUNO), a 20 kton liquid scintillator detector currently under construction in South China. The real-time monitoring system is designed to ensure both prompt alert speed and comprehensive coverage of progenitor stars. It incorporates prompt monitors on the electronic board as well as online monitors at the data acquisition stage. Assuming a false alert rate of 1 per year, this monitoring system exhibits sensitivity to pre-SN neutrinos up to a distance of approximately 1.6 (0.9) kiloparsecs and SN neutrinos up to about 370 (360) kiloparsecs for a progenitor mass of 30 solar masses, considering both normal and inverted mass ordering scenarios. The pointing ability of the CCSN is evaluated by analyzing the accumulated event anisotropy of inverse beta decay interactions from pre-SN or SN neutrinos. This, along with the early alert, can play a crucial role in facilitating follow-up multi-messenger observations of the next galactic or nearby extragalactic CCSN.

22. Efficacy and safety of topical NSAIDs combined with physiotherapy for frozen shoulder: a randomized controlled trial.

Abstract: OBJECTIVE: Frozen shoulder is a prevalent condition among individuals in their middle and later years. Invasive therapy has shown promising results in the treatment of frozen shoulders, but its widespread adoption has been hampered by high costs and the need for advanced medical technology. As a result, patients with frozen shoulders often turn to non-steroidal anti-inflammatory drugs (NSAIDs) for symptomatic relief. However, the oral administration of NSAIDs can lead to troublesome adverse effects on the gastrointestinal, cardiovascular, and urinary systems. In contrast, topical NSAIDs have gained attention for their excellent efficacy and lower adverse effects in various chronic pain conditions. Therefore, our study aimed to investigate the efficacy and safety of topical NSAIDs in improving pain and mobility among patients with frozen shoulders. PATIENTS AND METHODS: A total of 108 patients experiencing moderate to severe pain and mobility impairment due to frozen shoulder were enrolled in this study. The participants were randomly assigned to either the experimental group (n=72) or the control group (n=36). The experimental group received daily treatment with the loxoprofen hydrogel patch (LOX-P) in addition to basic rehabilitation physiotherapy. The control group was treated with flurbiprofen cataplasm (FLU-C) twice a day, along with rehabilitation physiotherapy. The primary endpoint for evaluating the efficacy of the two patches was the Constant-Murley score (CMS). Clinical symptom data, adverse events, and patient satisfaction were also recorded. RESULTS: After 14 days of treatment, the effective rate was 66.67% (n=48) in the experimental group and 41.67% (n=15) in the control group. The overall difference in the effective rates was 25.00% (95% CI=5.20-42.52; p=0.013). The safety profiles of the two topical agents were similar, with only a few adverse events reported. Abstract: Purpose: X-linked hypophosphatemia (XLH) is the most common inherited renal phosphate wasting disorder and is often misdiagnosed as vitamin D deficiency. This study aims to provide clinical and mutational characteristics of 65 XLH pediatric patients in southern China. Methods: In this work, a combination of DNA sequencing and qPCR analysis was used to study the PHEXgene in 80 pediatric patients diagnosed with hypophosphatemia. The clinical and laboratory data of confirmed 65 XLH patients were assessed and analyzed retrospectively. Results: In 65 XLH patients from 61 families, 51 different variants in the PHEXgene were identified, including 23 previously reported variants and 28 novel variants. In this cohort of XLH patients, the c.1601C>T(p.Pro534Leu) variant appears more frequently. Fourteen uncommon XLH cases were described, including four boys with de novo mosaic variants, eight patients with large deletions and a pair of monozygotic twins. The clinical manifestations in this cohort are very similar to those previously reported. Conclusion: This study extends the mutational spectrum of the PHEXgene, which will contribute to accurate diagnosis. This study also suggests a supplementary qPCR or MLPA assay may be performed along with classical sequencing to confirm the gross insertion/deletion.

Abstract: The gamut and EOTF (Electro-Optical Transfer Function) of a display together determine its color performance. Gamut determines the range of displayable colors, and the EOTF determines the color accuracy and depth. Image reproduction often comprises two parts: the image source and the display terminal. To ensure the universality of different terminals, the image source is generally produced in a standardized gamut standard, while the terminal pursues a wider gamut and more spectacular color performance. As a widely welcomed form of display, WCGDs (Wide Color Gamut Displays) boast stunning color performance. However, certain colors in WCGDs displays are oversaturated, which is unacceptable to the human eye. This paper analyzes the oversaturation problem of WCGDs. Firstly, a display model based on EOTF and gamut parameters is established. Then the color point vector field varies with EOTF is derived and the drift of luminance and chromaticity are analyzed separately. Finally, two strategies are present to optimize the color oversaturation problem of WCGDs based on the analysis conclusions and verify its effectiveness. Abstract: Melioidosis is a serious infectious disease caused by the Gram-negative bacterium Burkholderia pseudomallei . Recently, Rab32-dependent immune vesicles emerge as a critical defense pathway to restrict the intracellular B. pseudomallei . However, B. pseudomallei can evade host immune vesicles and survive in the cytoplasm, although this mechanism is not well understood. In this study, we found Rab32-dependent vesicles could effectively combat B. pseudomallei infection, but not all intracellular B. pseudomallei were encapsulated in Rab32-positive vesicles. To explore how B. pseudomallei counteracted the Rab32-dependent defense pathway, transcriptomic profiling of B. pseudomallei was performed to characterize the response dynamics during infection. We found that the type III secretion system of B. pseudomallei was activated, and a variety of effector proteins were highly upregulated. Among them, BopE, BprD, and BipC were shown to interact with Rab32. Interestingly, BopE directly interacts with host Rab32, potentially suppressing Rab32 function by interfering with nucleotide exchange, which in turn restricts the recruitment of Rab32 to bacterial-containing vesicles. Knocking out of BopE can increase the proportion of Rab32-positive vesicles, suppressing the intracellular replication and virulence of B. pseudomallei . Collectively, our findings have demonstrated that BopE may be an important effector for B. pseudomallei to evade from the Rab32-dependent killing vesicles into the cytosol for survival and replication. Therefore, a deeper understanding of the interaction between BopE and the host Rab32-dependent restriction pathway may provide an effective therapeutic strategy for the elimination of intracellular B. pseudomallei .IMPORTANCE B. pseudomallei is facultative intracellular bacterium that has evolved numerous strategies to evade host immune vesicles and survive in the cytoplasm. Rab32-dependent vesicles are one of these immune vesicles, but the mechanism by which B. pseudomallei escape Rab32-dependent vesicles remains elusive. Here, we find B. pseudomallei infection leading the activation of the type III secretion system (T3SS-3) and increasing the expression of various effectors. Specifically, we identify that BopE, an effector secreted by T3SS-3, triggers vesicle escape to promote B. pseudomallei pathogenicity and survival. Mechanistically, BopE suppresses the activation of Rab32 by interfering with nucleotide exchange, ultimately triggering vesicle escape and intracellular survival. We also find knocking out the bopE gene can increase the proportion of Rab32-positive vesicles that trap B. pseudomallei , dampening the survival of B. pseudomallei both in vitro and in vivo . Abstract: Background: Silver Diamine Fluoride (SDF) is effective for arresting dental caries, presenting a valuable non-invasive treatment option in dentistry. Despite its therapeutic advantages, a significant drawback is the tooth discoloration that follows its application, which can affect patient acceptance. Addressing this aesthetic concern without diminishing the treatment's efficacy remains challenging in dental practice. This study explores strategies to improve the aesthetic outcomes of SDF treatments., Methods: This in vitro study assessed the efficacy of Zinc in reducing SDF-induced discoloration on dentin blocks and examined its impact on the physical properties of dentin, including hardness and roughness. Dentin blocks were pre-treated with various concentrations of Zinc, followed by SDF application. Color changes were analyzed using Image J software, and cytotoxicity was evaluated using the CytoTox-ONE™ Homogeneous Membrane Integrity Assay. Dentin surface characteristics, including micro-hardness and roughness, were assessed using scanning electron microscopy., Results: The study results revealed a dose-dependent efficacy of Zinc in reducing discoloration caused by SDF on dentin, with higher Zinc concentrations showing better improvement in color outcomes. The application of a 20 M Zinc solution prior to SDF treatment significantly reduced discoloration compared to SDF alone, measured on day 14. Additionally, no significant changes in the hardness or roughness of etched dentin were observed in Zinc + SDF group compared to the SDF alone group. Zinc treatments demonstrated a desirable outcome on mucosal cytotoxicity, comparable to that of the negative control., Conclusion: Zinc significantly reduced SDF-induced tooth discoloration in a dose-dependent manner without affecting the etched dentin's micro-hardness and roughness, potentially improving patient acceptance, especially in visible areas of the mouth. 33. CEACAM6 facilitates gastric cancer progression through upregulating SLC27A2.

Abstract: Gastric cancer (GC) is one of the most lethal cancers. However, the underlying mechanisms are not yet fully understood. Here, we investigated the role of carcinoembryonic antigen-related cell adhesion molecule 6 (CEACAM6) in tumor initiation and progression in GC and proposed therapeutic strategies for CEACAM6-positive patients. In this article, we found that CEACAM6 overexpression promoted GC initiation and progression by overactivating FAO. CEACAM6 promotes SLC27A2 expression, contributing to enhanced fatty acid incorporation. CEACAM6 interacts with both SLC27A2 and USP29, facilitating the deubiquitination of USP29 on SLC27A2. Pharmacological inhibition of SLC27A2 attenuates the tumor-initiating ability of GC. Taken together, CEACAM6 overexpression facilitates GC progression by upregulating fatty acid uptake through SLC27A2, thereby contributing to FAO. 34. Enhancers and genome conformation provide complex transcriptional control of a herpesviral gene.

Abstract: Complex transcriptional control is a conserved feature of both eukaryotes and the viruses that infect them. Despite viral genomes being smaller and more gene dense than their hosts, we generally lack a sense of scope for the features governing the transcriptional output of individual viral genes. Even having a seemingly simple expression pattern does not imply that a gene's underlying regulation is straightforward. Here, we illustrate this by combining high-density functional genomics, expression profiling, and viral-specific chromosome conformation capture to define with unprecedented detail the transcriptional regulation of a single gene from Kaposi's sarcoma-associated herpesvirus (KSHV). We used as our model KSHV ORF68 - which has simple, early expression kinetics and is essential for viral genome packaging. We first identified seven cis-regulatory regions involved in ORF68 expression by densely tiling the ~154 kb KSHV genome with dCas9 fused to a transcriptional repressor domain (CRISPRi). A parallel Cas9 nuclease screen indicated that three of these regions act as promoters of genes that regulate ORF68. RNA expression profiling demonstrated that three more of these regions act by either repressing or enhancing other distal viral genes involved in ORF68 transcriptional regulation. Finally, we tracked how the 3D structure of the viral genome changes during its lifecycle, revealing that these enhancing regulatory elements are physically closer to their targets when active, and that disrupting some elements caused large-scale changes to the 3D genome. Abstract: Acetaminophen (APAP) overdose is still a leading cause of drug-induced liver injury (DILI), accompanied with severe inflammatory response. However, the therapy for APAP-induced DILI is rather limited. The combined application of natural products to treat DILI induced by APAP may be a new direction of the research. This study was conducted to evaluate the dual anti-inflammatory activity of curcumin (CUR) combined with berberine (BBR) against APAP-mediated DILI. Network pharmacology found that PI3K-Akt and PPAR signaling pathways were primarily involved in anti-DILI of the combination of CUR and BBR. APAP injection enhanced the levels of ALT, AST, IL-1β, IL-6, and TNF-α in mice, while such phenomenon was significantly reversed by the cotreatment of CUR and BBR, which was more effective than either single treatment. The increase of p-NF-κB and p-IKKα/β protein expression and the decrease of p-PI3K, p-AKT, and PPARγ protein expression in APAP-treated mice were markedly inhibited by the coadministration of CUR and BBR. Molecular docking further demonstrated that both CUR and BBR could stably bind to PI3K, AKT, and PPARγ protein. In conclusion, the combination of CUR and BBR more effectively protected liver from APAP-triggered DILI than individual treatment. However, it still meets a great bottleneck for the targeted production of odd-numbered neoagarooligosaccharides (NAOSs), such as neoagarotriose (NA3), due to the lack of one-step hydrolases. In this work, the α-agarase AgaA33 and β-galactosidase BgaD were synergistically used to prepare NA3 with agarose as a substrate. Additionally, an l-arabinose isomerase CaLAI from Clostridium acetobutylicum was characterized to valorize low-value byproducts (d-galactose) by forming d-tagatose, which exhibited good thermal stability without the need for additional metal ions. Under the optimal reaction conditions, the production of NA3 and d-galactose catalyzed by these three enzymes was 0.40 and 0.15 g/L, respectively. The artificial three-enzyme-based cascade transformation system not only achieved the highest production of NA3 until now but also allowed for the valorization of d-galactose, providing a wiser application route for agarose utilization.

Abstract: Bursaphelenchus xylophilus is a pathogenic nematode responsible for pine wilt disease, which can cause the demise of pine trees and discoloration of trunks. As rotenone is an important botanical pesticide, its impact on B. xylophilus was investigated through RNA-seq to understand the response mechanism of nematode. The bioassay results yielded the 12-h LC 30 (1.35 mg L -1 ) and LC 50 (2.60 mg L -1 ) values for rotenone. Differential gene expression analysis identified 172 and 614 differentially expressed genes (DEGs) in B. xylophilus exposed to two different concentrations of rotenone (1.35 and 2.60 mg L -1 ). To validate these findings, the expression patterns of 10 DEGs were confirmed through RT-qPCR. Additionally, all DEGs were categorized into eight gene expression profiles using STEM. Notably, the gene ontology (GO) processes of "single-organism process," "metabolic process," and "catalytic activity" were prominently enriched in rotenone-treated samples, suggesting a role for metabolic and catalytic pathways in the nematode's response to rotenone stress. KEGG pathways related to "carbon metabolism," "drug metabolism-cytochrome P450," and "metabolism of xenobiotics by cytochrome P450" were similarly enriched, indicating potential mechanisms for detoxification resistance and oxidative stress resistance. The analysis pointed to the pivotal roles of detoxification- and oxidoreduction-related genes, as well as signal transduction-related genes, in enabling B. xylophilus to adapt to rotenone exposure. Abstract: This study investigated a sustainable approach through dielectric barrier discharge (DBD) enhanced Fenton technology coupling nanofiltration (NF) process for landfill leachate treatment. The DBD/Fe(II)/H 2 O 2 system exhibited significant synergistic effects, removing 55.07 % of TOC and 53.79 % of UV 254 within 60 min, respectively. Additionally, the DBD/Fe(II)/H 2 O 2 system demonstrated exceptional performance in removing fluorescent substances and large molecular organic compounds, thereby reducing the formation of cake layer on the nanofiltration membrane. Moreover, membrane flux increased by 2.34 times, with reversible and irreversible resistances decreasing by 75.79 % and 81.55 %, respectively. Quenching experiments revealed ·OH as the primary active species for perfluorooctanoic acid (PFOA) degradation in the DBD/Fe(II)/H 2 O 2 process. The degradation pathway of PFOA was also elucidated via capillary electrophoresis-quadrupole time-of-flight mass spectrometry analysis. Correlation analysis indicated that TOC and EEM were the primary fouling factors. Lastly, through an assessment of energy consumption, economic costs, and carbon dioxide emissions, the advantages and practical application potential of the DBD/Fe(II)/H 2 O 2 system were demonstrated. 40. Biomass burning in Peninsular Southeast Asia intensifies meteorological drought in Southwest China.

Abstract: Biomass burning in Peninsular Southeast Asia (BB-PSEA) affects the climate in downwind regions, especially precipitation (PRE) in southern China. However, the impact of BB-PSEA on the meteorological drought in Southwest China (SWC), where closes to PSEA and often occurs seasonal drought, have not been clear yet. We selected a severe drought event in SWC from January to April 2010 and conducted sensitivity simulations using WRF-Chem (Weather Research and Forecasting model coupled with Chemistry) to evaluate the impact of BB-PSEA on the meteorological drought in SWC. Comparisons with observations revealed that the model performed well in simulating the spatiotemporal evolution of the drought in SWC. BB-PSEA increased the drought severity by 0.01-0.75 levels, enlarged drought areas by about 10%, and prolonged the drought duration mainly by one month in SWC. The impact of BB-PSEA on the drought in SWC in March/April was almost tenfold that in January/February, due to the higher emissions of BB-PSEA in March/April. The mechanism that BB-PSEA influenced drought predominantly involved the reduction of PRE, potential evapotranspiration (PET), and moisture fluxes in SWC. BB-PSEA aerosols warmed the air at 600-800 hPa and cooled the air near the surface in SWC, which stabilized the atmosphere and suppressed PRE and reduced PET in SWC. BB-PSEA aerosols also increased the sea surface temperature in South China Sea and the geopotential heights in the north of the Bay of Bengal, where the moisture sources of SWC originated from. This perturbation reduced the moisture fluxes across the west and south boundaries of SWC, resulting in the reduction of the water vapor content and PRE in SWC. Abstract: Acute myeloid leukemia (AML) is one of the most common types of blood cancer in adults and is associated with a poor survival rate. NK cells play a crucial role in combating AML, and alterations in immune checkpoint expression can impair NK cell function against AML. Targeting certain checkpoints may restore this function. CD96, an inhibitory immune checkpoint, has unclear expression and roles on NK cells in AML patients. In this study, we initially evaluated CD96 expression and compared CD96 + NK with the inhibitory receptor and stimulatory receptors on NK cells from AML patients at initial diagnosis. We observed increased CD96 expression on NK cells with dysfunctional phenotype. Further analysis revealed that CD96 + NK cells had lower IFN-γ production than CD96 - NK cells. Blocking CD96 enhanced the cytotoxicity of primary NK and cord blood-derived NK (CB-NK) cells against leukemia cells. Notably, patients with a high frequency of CD96 + NK cells at initial diagnosis exhibited poorer clinical outcomes. Additionally, TGF-β1 was found to enhance CD96 expression on NK cells via SMAD3 signaling. These findings suggest that CD96 is invovled in NK dysfunction against AML blast, and might be a potential target for restoring NK cell function in the fight against AML., (Copyright © 2024 Elsevier B.V. 42. Layered Bio-Inorganic MXene Membranes: A Green Approach for Uranium Extraction from Seawater Using Genetically Modified E. coli .

Abstract: With the growing demand for clean energy, efficient uranium extraction technologies are needed, especially from seawater, where uranium reserves are huge. Here, we developed a composite membrane by inserting Escherichia coli engineered with super uranyl-binding protein (SUP) within a two-dimensional (2D) MXene (Ti 3 C 2 T x ) layer. SUP endowed the bioinorganic hybrid membrane with ultrahigh selectivity for uranyl ions, while the engineered E. coli improved the mechanical strength and economy of the membranes. Experimental results showed that the membranes achieved precise recognition of uranyl ions and excellent ion screening performance (SF U/V ≈ 43, SF Na/U ≈ 158). Excellent separation performance and cyclic stability tests demonstrated the industrial application potential of the membrane. This method offers a green and sustainable solution, combining biological engineering and nanomaterial innovation, providing an environmentally friendly and efficient approach for uranium extraction from seawater, marking a significant advancement in the field of clean energy resource development.

44. Simultaneous One-Step Preparation of β/κ-Carrapentaose and 3,6-Anhydro-D-galactose by Cascading κ-Carrageenase and an Exo-α-3,6-Anhydro-D-galactosidase.

Abstract: Carrageenan oligosaccharides have shown promising bioavailability and possess a variety of physiological activities, making them highly suitable for use in the food, pharmaceutical, and agricultural industries. The preferred method for producing carrageenan oligosaccharides is using various carrageenolytic enzymes, as it offers mild reaction conditions, high efficiency, and product specificity. However, there is still a lack of specific applications for using these enzymes to prepare odd-numbered carrageenan-oligosaccharides (OCOSs). Our previous research identified a more convenient route for simultaneously preparing OCOSs and 3,6-anhydro-D-galactose (D-AHG) using only two types of carrageenolytic enzymes: κ-carrageenase and exo-α-3,6-anhydro-D-galactosidase (D-ADAGase). In this study, we utilized a CipA-based self-assembly system to cascade κ-carrageenase CaKC16A and D-ADAGase ZuGH129A for one-step preparation of β/κ-carrapentaose, G-(DA-G4S) 2 , and D-AHG from degrading β/κ-carrageenan. This self-assembled enzyme, namely CipA-CaKC16A-ZuGH129A, can be easily obtained through a simple centrifugation process. The final optimized enzymatic process produced 0.74 g/L G-(DA-G4S) 2 and 0.13 g/L D-AHG. This cascade system of different types of carrageenolytic enzymes has the potential to achieve the preparation of various types of carrageenan oligosaccharides.

45. Engineering the Non-catalytic Domain to Enhance Catalytic Activity and Thermal Stability of a Nκ2-Producing κ-Carrageenase.

Abstract: κ-Carrageenases play an important role in achieving the high-value utilization of carrageenan polysaccharides. They can be used in the preparation of even-numbered κ-neocarrageenan oligosaccharides by degrading κ-carrageenan (KC). We previously identified and characterized a κ-carrageenase, CaKC16B, with high specificity for producing a single κ-neocarrabiose. It can produce a single κ-neocarrabiose from degrading KC. However, they also exhibited poor thermal stability and catalytic efficiency. To improve these properties, we introduced noncatalytic domains (nonCDs) from a heat-resistant κ-carrageenase, MtKC16A, into the C-terminus of CaKC16B to construct CaKC16BUN and CaKC16BUNB. Compared to the original CaKC16B, both of them exhibited improved enzymatic properties, including optimal reaction temperature, thermal stability, substrate affinity, and catalytic efficiency. Remarkably, the k cat / K m values of 16BUN and 16BUNB increased by 127 and 290 times, respectively. Importantly, the addition of nonCDs did not alter the final products of degrading KC, retaining the high product specificity of CaKC16B. Interestingly, the addition of nonCDs changed CaKC16B's substrate specificity for hydrolyzing KC and βκ-carrageenan, with mutants exhibiting higher relative activity for βκ-carrageenan. We further observed through biolayer interferometry that the binding and dissociation process between MtUN nonCD and βκ-carrageenan is faster compared to that of KC. This may explain the change in the substrate specificity observed in the mutants of CaKC16B.

46. Corneal asymmetry contributes decentration in both spherical and toric orthokeratology lenses.

Abstract: Purpose: To discuss the characteristics of anterior corneal elevation asymmetry in myopic eyes and clarify which kind of asymmetry most influenced lens position., Methods: In this retrospective study of 199 consecutive myopic participants, corneal topography was used to analyse asymmetry in anterior corneal elevation. Amongst them, 65 participants (65 eyes) who underwent orthokeratology (31 and 34 with spherical and toric lenses, respectively) were re-evaluated. Stepwise multiple linear regression analysis was used to identify the contributing factors that influenced lens decentration. Receiver operating characteristic curve (ROC) analysis was employed to assess how the corneal asymmetry vector could predict decentration., Results: There were no significant differences in treatment zone decentration (TZDec) between participants wearing toric and spherical lenses (p = 0.60 and 0.64 for 1 week and 1 month of wear, respectively). Amongst the underlying factors, the magnitude of TZDec was only correlated with the amount of corneal asymmetry vector (standardised β = 0.44, 0.48, p < 0.001 for all) after 1 week and 1 month of wear, and the direction of TZDec after 1 month of lens wear was associated with the angle of the asymmetry vector (r = 0.25, p = 0.04). ROC analysis showed that the magnitude of corneal asymmetry vector produced accurate discrimination between non-severe and severe decentration for 1 week and 1 month of wear (area under the curve was 0.93 ± 0.04 and 0.89 ± 0.05, respectively, p < 0.001). Amongst participants whose corneal asymmetry vector exceeded 41.06 μm and was oriented inferiorly, 35.29% showed severe decentration after 1 month of lens wear., Conclusions: In myopic participants, corneal asymmetry existed in the 8.0 mm chord diameter. If the asymmetry vector >41.06 μm and the direction was oriented inferiorly, then practitioners must be vigilant about severe decentration which would not be alleviated by a toric design., (© 2024 College of Optometrists.)

Abstract: Background: Polycystic ovary syndrome (PCOS) is highly prevalent in women of reproductive age worldwide, exhibits highly heterogeneous clinical presentation and biochemical parameters, and has no cure. This study aimed to investigate the role of miR-34a-5p in PCOS, its effect on glycolysis in granulosa cells (GCs), and its potential contribution to follicular dysregulation., Methods: Herein, Follicular follicular fluid (FF) samples were collected from six patients with PCOS and six healthy controls undergoing in vitro fertilization-embryo transfer. The isolated extracellular vesicles were characterized by transmission electron microscopy, nanoparticle tracking analysis, and western blotting. Additionally, miRNA sequencing was performed to identify differentially expressed microRNAs, and their functions were analyzed through transcriptomics. The In vitro effects of miR-34a-5p on glycolysis, cell proliferation, and apoptosis were assessed in human ovarian granulosa tumour cell line (KGN cells). Targets of miR-34a-5p were identified by dual-luciferase reporter assays, and quantitative real-time polymerase chain reaction and western blotting were performed to determine gene and protein expression., Results: The level of miR-34a-5p in FF-derived extracellular vesicles derived from patients with PCOS was significantly higher than that of the control group. Transcriptomic analysis highlighted miR-34a-5p as a key regulator of glycolysis and apoptosis. Furthermore, in vitro analysis showed that miR-34a-5p targeted lactate dehydrogenase A (LDHA), inhibited glycolysis, reduced energy supply to GCs, and promoted apoptosis of KGN cells. Conversely, miR-34a-5p inhibition restored glycolysis function and cell viability., Conclusion: The findings of this study show that miR-34a-5p mediates GC apoptosis in PCOS by targeting LDHA and inhibiting glycolysis, suggesting its crucial role in PCOS pathophysiology, and offering potential therapeutic targets for improving follicular development and fertility outcomes in patients with PCOS. Abstract: Fluorouracil-based chemotherapy responses in colorectal cancer (CRC) patients vary widely, highlighting the role of pharmacogenomics in developing better predictive models. We analyzed 379 CRC patients receiving fluorouracil-based chemotherapy, collecting data on fluorouracil metabolism-related SNPs (TYMS, MTHFR, DPYD, RRM1), blood inflammatory markers, and clinical status. Six machine learning models-K-nearest neighbors, support vector machine, gradient boosting decision trees (GBDT), eXtreme Gradient Boosting (XGBoost), LightGBM, and random forest-were compared against multivariate logistic regression and a deep learning model (i.e., multilayer perceptron, MLP). Feature importance analysis highlighted seven predictors: histological grade, N and M staging, monocyte count, platelet-to-lymphocyte ratio, MTHFR rs1801131, and RRM1 rs11030918. In a five-fold cross-validation, XGBoost and GBDT exhibited superior performance, with Area Under Curve (AUC) of 0.88 ± 0.02. XGBoost excelled in identifying favorable prognosis (recall = 0.939). GBDT demonstrated balance in recognizing both categories, with a recall for favorable prognosis of 0.908 and a precision for unfavorable prognosis of 0.863. MLP had a similar AUC (0.87) with high precision for favorable prognosis (recall = 0.946). In external validation, XGBoost model achieved an accuracy of 0.79. Abstract: An intelligent insulin delivery system targeting intestinal absorption and glucose responsiveness can enhance the bioavailability through oral insulin therapy, offering promising diabetes treatment. In this paper, a glucose and pH dual-response polymer hydrogel using carboxymethyl agarose modified with 3-amino-phenylboronic acid and l-valine (CPL) was developed as an insulin delivery carrier, exhibiting excellent biocompatibility and effective insulin encapsulation. The insulin encapsulated in the hydrogel (Ins-CPL) was released in a controlled manner in response to the in vivo stimulation of blood glucose and pH levels with higher levels of intracellular uptake and utilization of insulin in the intestinal environment simultaneously. Notably, the Ins-CPL hydrogel effectively regulated blood sugar in diabetic rats over a long period by simulating endogenous insulin, responding to changes in plasma pH and glucose levels, and overcoming the intestinal epithelium barrier. This indicates a significant boost in oral insulin bioavailability and broadens its application prospects.
