For the characterization of soil stiffness anisotropy at small strains and the calculation of soil elastic constants derived from the cross-anisotropic model, it is important to obtain stress wave phase velocities of soils in both principal and oblique directions. This study developed an original eight-prismatic shape apparatus equipped with disk-shaped shear plates to measure shear (S-) wave phase velocities (Vphase) in multiple directions, and four granular materials of various shapes were tested by this apparatus under isotropic confinement. Experimental results confirm the capability of the new apparatus and reveal that both S-wave propagation and oscillation directions are sensitive to soil inner fabric, i.e., Vs changes with the variation of either S-wave propagation or oscillation direction. Based on the experimental observations, it is suggested to keep the same S-wave oscillation direction when measuring Vs in multiple propagation directions so that the corresponding shape of the S-wave surface (polar plots of Vs in arbitrary propagation directions) is more precise to reflect the small-strain stiffness anisotropy of soils.

It is empirically known that the packing property and mechanical responses of cohesionless granular materials are influenced by grain shape. For example, the attainable range of void ratio depends on grain shape; angular materials tend to exhibit greater friction angles. Besides, shear wave velocity (Vs ) and small-strain shear modulus (G0 ) of sphere assemblies are affected by surface roughness. However, consensus has yet to be reached on the combined effect of grain shape and surface roughness on G0 and stress-strain relation. This contribution aims to evaluate the shape-roughness combined effect on the strain-dependent mechanical responses of granular materials. Three groups of glass beads having different grain shapes and a silica sand were used, and their grain surfaces were roughened through a systematic procedure using a milling machine. In total, eight materials were subjected to triaxial compression after measurement of Vs. The experimental results reveal that the stress-strain relation of angular particles is remarkably affected by the surface roughness, whereas the roughness effect on the stress-dependent variation in G0 is limited for angular particles.

Pumices with high pore voids of volcanic origin are distributed throughout Japan and are the causal layer of slope failures. In many cases of surface failures, it is difficult to assume that the resulting layers are fully saturated. The high waterholding capacity of the pumice suggests that they were deposited in an unsaturated state with a high degree of saturation. In this study, saturated triaxial compression tests and fully undrained unsaturated triaxial compression tests were conducted on artificially produced pumice and natural pumice while measuring the amount of crushing. This is to clarify the relationship between the crushing and mechanical properties of pumice with porous particles, which are often the cause of such disasters, and their behaviour under unsaturated conditions. The results showed that the pumice stones have an ultra-high pore structure. Moreover, pumice with porous particles reached a steady state under both saturated and highly saturated unsaturated conditions, and the amount of crushing increased under highly saturated unsaturated conditions.

Acute lung injury (ALI) is characterized by respiratory failure resulting from the disruption of the epithelial and endothelial barriers as well as immune system. In this study, we evaluated the therapeutic potential of airway epithelial cell-derived extracellular vesicles (EVs) in maintaining lung homeostasis. We isolated human bronchial epithelial cell-derived EVs (HBEC-EVs), which endogenously express various immune-related surface markers and investigated their immunomodulatory potential in ALI. In ALI cellular models, HBEC-EVs demonstrated immunosuppressive effects by reducing the secretion of proinflammatory cytokines in both THP-1 macrophages and HBECs. Mechanistically, these effects were partially ascribed to nine of the top 10 miRNAs enriched in HBEC-EVs, governing toll-like receptor-NF-κB signaling pathways. Proteomic analysis revealed the presence of proteins in HBEC-EVs involved in WNT and NF-κB signaling pathways, pivotal in inflammation regulation. ANXA1, a constituent of HBEC-EVs, interacts with formyl peptide receptor (FPR)2, eliciting anti-inflammatory responses by suppressing NF-κB signaling in inflamed epithelium, including type II alveolar epithelial cells. In a mouse model of ALI, intratracheal administration of HBEC-EVs reduced lung injury, inflammatory cell infiltration, and cytokine levels. Collectively, these findings suggest the therapeutic potential of HBEC-EVs, through their miRNAs and ANXA1 cargo, in mitigating lung injury and inflammation in ALI patients.

Mineral aerosols (i.e., dust) can affect climate and weather by absorbing and scattering shortwave and longwave radiation in the Earth's atmosphere, the direct radiative effect. Yet understanding of the direct effect is so poor that the sign of the net direct effect at top of the atmosphere (TOA) is unconstrained, and thus it is unknown if dust cools or warms Earth's climate. Here we develop methods to estimate the instantaneous shortwave direct effect via observations of aerosols and radiation made over a 3-year period in a desert region of the southwestern US, obtaining a direct effect of -14±1 and -9±6 W m−2 at the surface and TOA, respectively. We also generate region-specific dust optical properties via a novel dataset of soil mineralogy from the Airborne Visible/Infrared Imaging Spectrometer (AVIRIS), which are then used to model the dust direct radiative effect in the shortwave and longwave. Using this modeling method, we obtain an instantaneous shortwave direct effect of -21±7 and -1±7 W m−2. The discrepancy between the model and observational direct effect is due to stronger absorption in the model, which we interpret as an AVIRIS soil iron oxide content that is too large. Combining the shortwave observational direct effect with a modeled longwave TOA direct effect of 1±1 W m−2, we obtain an instantaneous TOA net effect of -8±6 W m−2, implying a cooling effect of dust. These findings provide a useful constraint on the dust direct effect in the southwestern United States.

Abstract Charcot-Marie-Tooth disease type 2Z is caused by MORC2 mutations and presents with axonal neuropathy. MORC2 mutations can also manifest as developmental delay, impaired growth, dysmorphic facies, and axonal neuropathy (DIGFAN). We report a patient exhibiting an intermediate phenotype between these diseases associated with a novel MORC2 variant. A literature review revealed that the genotype‒phenotype correlation in MORC2-related disorders is complex and that the same mutation can cause a variety of phenotypes.

Idiopathic pleuroparenchymal fibroelastosis (iPPFE) is a rare form of idiopathic interstitial pneumonia. We report a case of a patient with iPPFE in whom postural changes improved diaphragmatic excursion (DE) and exercise tolerance. Chest radiography showed a greater elevation of the diaphragm at maximum expiration in the supine position than the standing position. DE measured by ultrasonography was higher in the supine position than the standing position. The findings may suggest greater diaphragm movement in the supine position, leading to successful rehabilitation and improved exercise endurance. There is no effective treatment for iPPFE; therefore, an innovative treatment strategy is warranted.

Y-box binding protein 1 (YBX1) promotes oncogenic transformation and tumor growth. YBX1 plays a role in regulation of cell cycle promotion via upregulation of cell cycle-related genes. In ovarian cancer, YBX1 also promotes tumor growth, but the mechanisms of YBX1 in cell growth and cell cycle in ovarian cancer remain not to be fully understood. Here, we investigated whether YBX1-dependent cancer cell proliferation was specifically associated with expression of cell cycle related genes in ovarian cancer. Protein and mRNA expression levels of YBX1 and cell cycle-related genes in ovarian cancer cell lines and tissues were determined by western blot analysis, immunohistochemical analysis and reverse transcription-quantitative PCR. Cell cycle analysis was performed by flow cytometry. Luciferase assay and Chromatin immunoprecipitation assay were used to investigate a transcriptional function of YBX1. YBX1 silencing induced marked growth suppression in 4 cell lines (group A), moderate suppression in 5 cell lines (group B), and no suppression in 3 cell lines (group C) among 12 ovarian cancer cell lines in culture. The YBX1 silencing induced cell cycle arrest at G2/M phase and suppressed expression of cyclin A1 gene in group A and B cell lines, but not in group C cell lines. Cyclin A1 silencing specifically suppressed cell proliferation in group A cell lines and partially in group B cell lines, but not at all in group C cell lines. YBX1 mRNA levels were significantly correlated with cyclin A1 mRNA levels in patients with high-grade serous carcinoma. Augmented YBX1 expression plays a key role in tumor growth promotion in ovarian cancer in its close association with cyclin A1. [ABSTRACT FROM AUTHOR]

The global coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has affected millions of lives, leading to significant morbidity and mortality. With >772 million cases and nearly seven million deaths reported worldwide to date, the development of vaccines has been a critical step in mitigating the impact of COVID-19. However, concerns have arisen regarding the potential for SARS-CoV-2 mRNA vaccination to trigger autoimmune diseases, including autoimmune hepatitis (AIH). The present single-center, retrospective study aimed to compare the clinical and pathological features of AIH in patients with or without a history of SARS-CoV-2 mRNA vaccination. A total of 72 patients with AIH were examined. Among them, 10 had received the SARS-CoV-2 mRNA vaccination prior to AIH onset. These patients exhibited more pronounced CD4+ T cell infiltration into the liver tissue compared with those who were unvaccinated. No significant differences in the levels of other liver enzymes, autoimmune antibodies, or CD8+ T cell infiltration were observed between the groups. Moreover, the AIH patients with a history of SARS-CoV-2 mRNA vaccination had more extensive CD4+ T cell infiltration in their liver tissues than the unvaccinated patients. These findings suggested that the immune response to SARS-CoV-2 mRNA vaccination may influence the pathogenesis of AIH, highlighting the need for further research into the relationship between SARS-CoV-2 mRNA vaccination and autoimmune liver diseases. In the chloroplast stroma, dynamic pH changes occur from acidic to alkaline in response to fluctuating light conditions. We investigated the pH dependency of the electron transfer reaction of ferredoxin-NADP+ reductase (FNR) with ferredoxin (Fd) isoproteins, Fd1 and Fd2, which are localized in mesophyll cells and bundle sheath cells, respectively, in the leaves of C4 plant maize. The pH-dependent profile of the electron transfer activity with FNR was quite different between Fd1 and Fd2, which was mainly explained by the opposite pH dependency of the K m value of these Fds for FNR. Replacement of the amino acid residue at position of 65 (D65N) and 78 (H78A) between the two Fds conferred different effect on their pH dependency of the K m value. Double mutations of the two residues between Fd1 and Fd2 (Fd1D65N/H78A and Fd2N65D/A78H) led to the mutual exchange of the pH dependency of the electron transfer activity. This exchange was mainly explained by the changes in the pH-dependent profile of the K m values. Therefore, the differences in Asp/Asn at position 65 and His/Ala at position 78 between Fd1 and Fd2 were shown to be the major determinants for their different pH dependency in the electron transfer reaction with FNR. The retention of hydrogen (H2) bubbles generated by water radiolysis was quantitatively studied in a high-viscous suspension of carbonate slurry consisting of a mixture of suspended solid (SS) of magnesium (Mg) and calcium (Ca) precipitates under strongly alkaline conditions, like the radioactive wastes discharged from the coagulation sedimentation (co-precipitation) process at the multinuclide removal equipment in the Fukushima Daiichi Nuclear Power Station. The H2 retention properties were evaluated in two types of carbonate slurry with different hydrophilicity: the hydrophilic "current type" and the hydrophobic "return type." Then, their properties were compared with those in another suspension of clay suspension of bentonite. The hydrophilicity of the surface of the SS particles was evaluated from two different adsorption isotherms of water (H2O) vapor on the surface, as well as those of Mg(OH)2 and CaCO3 particles, simulating the precipitates in the slurry. The H2O molecules were strongly chemically adsorbed on the Mg(OH)2, while they were weakly physically adsorbed on the CaCO3. Since the amount of chemical adsorption on the SS surface of the hydrophilic slurry was almost the same as that on the Mg(OH)2 surface, Mg(OH)2 was found to be dominant on the SS surface. From the comparison between the amounts of chemical adsorption and H2O in the slurry, it was confirmed that H2O molecules must be shared among the SS particles, and this sharing of H2O molecules formed the structural viscosity in the slurry, different from that in the clay suspension where electrostatic bonding between the fine clay minerals forms the viscosity. The retention of H2 bubbles in (by) the slurry was evaluated from the difference in the amount of H2 observed with and without stirring the slurry after 60Co γ irradiation. The observed amount increased linearly with increasing absorbed dose by the slurry to obtain H2 generation yield (G-value): Gtotal (with stirring) and Gstat (without stirring). The Gtotal was independent of the sample height, whereas the Gstat decreased with increasing the height. This indicated that the higher the height, the more difficult it was for H2 bubbles to be released. From the direct observation of H2 bubbles after irradiation, the growth of H2 bubbles with increasing the dose was evaluated to obtain the resisting force of H2 bubbles to the slurry. The H2 bubbles were confirmed to be retained in the slurry because the resistance was lower than the shear strength of the slurry (yield value at rest). The ability of the slurry to retain the H2 bubbles was evaluated from the sample height dependence of the relative G-value, Gstat/Gtotal. The log-log plot of the G-value and height gave a straight line, and the slope of the hydrophilic slurry became larger than that of the hydrophobic one. This difference indicated the effect of structural viscosity, which was observed in the H2 bubble retention in the clay suspension. In addition, the slopes for the hydrophobic slurry and treated water were significantly different, and this indicated the effect of steric hindrance by the SS particles. Therefore, H2 bubbles were retained by not only the structural viscosity but also the steric hindrance in the hydrophilic slurry. [ABSTRACT FROM AUTHOR]
