Abstract: We study a flaring activity of the HSP Mrk421 that was characterized from radio to very-high-energy (VHE; E $>0.1$TeV) gamma rays with MAGIC, Fermi-LAT, Swift, XMM-Newton and several optical and radio telescopes. These observations included, for the first time for a gamma-ray flare of a blazar, simultaneous X-ray polarization measurements with IXPE. We find substantial variability in both X-rays and VHE gamma rays throughout the campaign, with the highest VHE flux above 0.2 TeV occurring during the IXPE observing window, and exceeding twice the flux of the Crab Nebula. However, the VHE and X-ray spectra are on average softer, and the correlation between these two bands weaker that those reported in previous flares of Mrk421. IXPE reveals an X-ray polarization degree significantly higher than that at radio and optical frequencies. The X-ray polarization angle varies by $\sim$100$^\circ$ on timescales of days, and the polarization degree changes by more than a factor 4. The highest X-ray polarization degree reaches 26%, around which a X-ray counter-clockwise hysteresis loop is measured with XMM-Newton. It suggests that the X-ray emission comes from particles close to the high-energy cutoff, hence possibly probing an extreme case of the Turbulent Extreme Multi-Zone model. We model the broadband emission with a simplified stratified jet model throughout the flare. The polarization measurements imply an electron distribution in the X-ray emitting region with a very high minimum Lorentz factor, which is expected in electron-ion plasma, as well as a variation of the emitting region size up to a factor of three during the flaring activity. We find no correlation between the fluxes and the evolution of the model parameters, which indicates a stochastic nature of the underlying physical mechanism. Abstract: OT 081 is a well-known, luminous blazar that is remarkably variable in many energy bands. We present the first broadband study of the source which includes very-high-energy (VHE, $E>$100\,GeV) $\gamma$-ray data taken by the MAGIC and H.E.S.S. imaging Cherenkov telescopes. The discovery of VHE $\gamma$-ray emission happened during a high state of $\gamma$-ray activity in July 2016, observed by many instruments from radio to VHE $\gamma$-rays. We identify four states of activity of the source, one of which includes VHE $\gamma$-ray emission. Variability in the VHE domain is found on daily timescales. The intrinsic VHE spectrum can be described by a power-law with index $3.27\pm0.44_{\rm stat}\pm0.15_{\rm sys}$ (MAGIC) and $3.39\pm0.58_{\rm stat}\pm0.64_{\rm sys}$ (H.E.S.S.) in the energy range of 55--300\,GeV and 120--500\,GeV, respectively. The broadband emission cannot be sucessfully reproduced by a simple one-zone synchrotron self-Compton model. Instead, an additional external Compton component is required. We test a lepto-hadronic model that reproduces the dataset well and a proton-synchrotron dominated model that requires an extreme proton luminosity. Emission models that are able to successfully represent the data place the emitting region well outside of the Broad Line Region (BLR) to a location at which the radiative environment is dominated by the infrared thermal radiation field of the dusty torus. In the scenario described by this flaring activity, the source appears to be an FSRQ, in contrast with past categorizations. Abstract: Imaging atmospheric Cherenkov telescopes (IACTs) are used to observe very high-energy photons from the ground. Gamma rays are indirectly detected through the Cherenkov light emitted by the air showers they induce. The new generation of experiments, in particular the Cherenkov Telescope Array Observatory (CTAO), sets ambitious goals for discoveries of new gamma-ray sources and precise measurements of the already discovered ones. To achieve these goals, both hardware and data analysis must employ cutting-edge techniques. This also applies to the LST-1, the first IACT built for the CTAO, which is currently taking data on the Canary island of La Palma. This paper introduces a new event reconstruction technique for IACT data, aiming to improve the image reconstruction quality and the discrimination between the signal and the background from misidentified hadrons and electrons. The technique models the development of the extensive air shower signal, recorded as a waveform per pixel, seen by CTAO telescopes' cameras. Model parameters are subsequently passed to random forest regressors and classifiers to extract information on the primary particle. The new reconstruction was applied to simulated data and to data from observations of the Crab Nebula performed by the LST-1. The event reconstruction method presented here shows promising performance improvements. The angular and energy resolution, and the sensitivity, are improved by 10 to 20% over most of the energy range. At low energy, improvements reach up to 22%, 47%, and 50%, respectively. 4. Effective viscosity of a two dimensional passive suspension in a liquid crystal solvent

Abstract: Suspension of particles in a fluid solvent are ubiquitous in nature, for example, water mixed with sugar or bacteria self-propelling through mucus. Particles create local flow perturbations that can modify drastically the effective (homogenized) bulk properties of the fluid. Understanding the link between the properties of particles and the fluid solvent, and the effective properties of the medium is a classical problem in fluid mechanics. Here we study a special case of a two dimensional model of a suspension of undeformable particles in a liquid crystal solvent. In the dilute regime, we calculate asymptotic solutions of the perturbations of the velocity and director fields and derive an explicit formula for an effective shear viscosity of the liquid crystal medium. Such effective shear viscosity increases linearly with the area fraction of particles, similar to Einstein formula but with a different prefactor. We provide explicit asymptotic formulas for the dependence of this prefactor on the material parameters of the solvent. Finally, we identify a case of decreasing the effective viscosity by increasing the magnitude of the shear-flow alignment coefficient of the liquid crystal solvent., Comment: 15 pages, 3 figures

Abstract: We present a proposal for a future light dark matter search experiment at the Electron Stretcher Accelerator ELSA in Bonn: Lohengrin. It employs the fixed-target missing momentum based technique for searching for dark-sector particles. The Lohengrin experiment uses a high intensity electron beam that is shot onto a thin target to produce mainly SM bremsstrahlung and - in rare occasions - possibly new particles coupling feebly to the electron. A well motivated candidate for such a new particle is the dark photon, a new massive gauge boson arising from a new gauge interaction in a dark sector and mixing kinetically with the standard model photon. The Lohengrin experiment is estimated to reach sensitivity to couplings small enough to explain the relic abundance of dark matter in various models for dark photon masses between approximately 1 MeV and approximately 100 MeV.

Abstract: Instruments for gamma-ray astronomy at Very High Energies ($E>100\,{\rm GeV}$) have traditionally derived their scientific results through proprietary data and software. Data standardisation has become a prominent issue in this field both as a requirement for the dissemination of data from the next generation of gamma-ray observatories and as an effective solution to realise public data legacies of current-generation instruments. Specifications for a standardised gamma-ray data format have been proposed as a community effort and have already been successfully adopted by several instruments. We present the first production of standardised data from the Major Atmospheric Gamma-ray Imaging Cherenkov (MAGIC) telescopes. We converted $166\,{\rm h}$ of observations from different sources and validated their analysis with the open-source software Gammapy. We consider six data sets representing different scientific and technical analysis cases and compare the results obtained analysing the standardised data with open-source software against those produced with the MAGIC proprietary data and software. Aiming at a systematic production of MAGIC data in this standardised format, we also present the implementation of a database-driven pipeline automatically performing the MAGIC data reduction from the calibrated down to the standardised data level. In all the cases selected for the validation, we obtain results compatible with the MAGIC proprietary software, both for the manual and for the automatic data productions. Part of the validation data set is also made publicly available, thus representing the first large public release of MAGIC data. 7. Predicting Expert Evaluations in Software Code Reviews

Abstract: Manual code reviews are an essential but time-consuming part of software development, often leading reviewers to prioritize technical issues while skipping valuable assessments. This paper presents an algorithmic model that automates aspects of code review typically avoided due to their complexity or subjectivity, such as assessing coding time, implementation time, and code complexity. Instead of replacing manual reviews, our model adds insights that help reviewers focus on more impactful tasks. Calibrated using expert evaluations, the model predicts key metrics from code commits with strong correlations to human judgments (r = 0.82 for coding time, r = 0.86 for implementation time). By automating these assessments, we reduce the burden on human reviewers and ensure consistent analysis of time-consuming areas, offering a scalable solution alongside manual reviews. This research shows how automated tools can enhance code reviews by addressing overlooked tasks, supporting data-driven decisions and improving the review process.

8. Detailed Analysis of Local Climate at the CTAO-North Site on La Palma from 20 Years of MAGIC Weather Station Data

Abstract: The Observatorio del Roque de los Muchachos will host the northern site of the Cherenkov Telescope Array Observatory (CTAO), in an area about 200 m below the mountain rim, where the optical telescopes are located. The site currently hosts the MAGIC Telescopes, which have gathered a unique series of 20 years of weather data. We use advanced profile likelihood methods to determine seasonal cycles, the occurrence of weather extremes, weather downtime, and long-term trends correctly taking into account data gaps. The fractality of the weather data is investigated by means of multifractal detrended fluctuation analysis. The data are published according to the Findable, Accessible, Interoperable, and Reusable (FAIR) principles. We find that the behaviour of wind and relative humidity show significant differences compared to the mountain rim. We observe an increase in temperature of $0.55\pm0.07\mathrm{(stat.)}\pm0.07\mathrm{(syst.)}^\circ C$/decade, the diurnal temperature range of $0.13\pm0.04\mathrm{(stat.)}\pm0.02\mathrm{(syst.)}^\circ C$/decade (accompanied by an increase of seasonal oscillation amplitude of $\Delta C_m=0.29\pm0.10\mathrm{(stat.)}\pm0.04\mathrm{(syst.)}^\circ C$/decade) and relative humidity of $4.0\pm0.4\mathrm{(stat.)}\pm1.1\mathrm{(syst.)}$%/decade, and a decrease in trade wind speeds of $0.85\pm0.12\mathrm{(stat.)}\pm0.07\mathrm{(syst.)}$(km/h)/decade. The occurrence of extreme weather, such as tropical storms and long rains, remains constant over time. We find a significant correlation of temperature with the North Atlantic Oscillation Index and multifractal behaviour of the data. The site shows a weather-related downtime of 18.5%-20.5%, depending on the wind gust limits employed. 9. Src Kinase Slows Collective Rotation of Confined Epithelial Cell Monolayers

Abstract: Collective cell migration is key during development, wound healing and metastasis and relies on coordinated cell behaviors at the group level. Src kinase is a key signalling protein for physiological functions of epithelia, as it regulates many cellular processes, including adhesion, motility, and mechanotransduction. Its over-activation is associated to cancer aggressiveness. Here, we take advantage of optogenetics to precisely control Src activation in time and show that its pathological-like activation slows collective rotation of epithelial cells confined into circular adhesive patches. We interpret velocity, force and stress data during period of non-activation and period of activation of Src thanks to an hydrodynamic description of the cell assembly as a polar active fluid. Src activation leads to a 2-fold decrease in the ratio of polar angle to friction, which could result from increased adhesiveness at the cell-substrate interface. Measuring internal stress allows us to show that active stresses are subdominant compared to traction forces. Abstract: Context: There are currently three pulsars firmly detected by imaging atmospheric Cherenkov telescopes (IACTs), two of them reaching TeV energies, challenging models of very-high-energy (VHE) emission in pulsars. More precise observations are needed to better characterize pulsar emission at these energies. The LST-1 is the prototype of the Large-Sized Telescope, that will be part of the Cherenkov Telescope Array Observatory (CTAO). Its improved performance over previous IACTs makes it well suited for studying pulsars. Aims: To study the Crab pulsar emission with the LST-1, improving and complementing the results from other telescopes. These observations can also be used to characterize the potential of the LST-1 to study other pulsars and detect new ones. Methods: We analyzed a total of $\sim$103 hours of gamma-ray observations of the Crab pulsar conducted with the LST-1 in the period from September 2020 to January 2023. The observations were carried out at zenith angles less than 50 degrees. A new analysis of the Fermi-LAT data was also performed, including $\sim$14 years of observations. Results: The Crab pulsar phaseogram, long-term light-curve, and phase-resolved spectra are reconstructed with the LST-1 from 20 GeV to 450 GeV for P1 and up to 700 GeV for P2. The pulsed emission is detected with a significance of 15.2$\sigma$. The two characteristic emission peaks of the Crab pulsar are clearly detected (>10$\sigma$), as well as the so-called bridge emission (5.7$\sigma$). We find that both peaks are well described by power laws, with spectral indices of $\sim$3.44 and $\sim$3.03 respectively. The joint analysis of Fermi-LAT and LST-1 data shows a good agreement between both instruments in the overlapping energy range. 11. Generation of ‘semi-guided’ cortical organoids with complex neural oscillations

Abstract: Temporal development of neural electrophysiology follows genetic programming, similar to cellular maturation and organization during development. The emergent properties of this electrophysiological development, namely neural oscillations, can be used to characterize brain development. Recently, we utilized the innate programming encoded in the human genome to generate functionally mature cortical organoids. In brief, stem cells are suspended in culture via continuous shaking and naturally aggregate into embryoid bodies before being exposed to media formulations for neural induction, differentiation and maturation. The specific culture format, media composition and duration of exposure to these media distinguish organoid protocols and determine whether a protocol is guided or unguided toward specific neural fate. The 'semi-guided' protocol presented here has shorter induction and differentiation steps with less-specific patterning molecules than most guided protocols but maintains the use of neurotrophic factors such as brain-derived growth factor and neurotrophin-3, unlike unguided approaches. This approach yields the cell type diversity of unguided approaches while maintaining reproducibility for disease modeling. Importantly, we characterized the electrophysiology of these organoids and found that they recapitulate the maturation of neural oscillations observed in the developing human brain, a feature not shown with other approaches. This protocol represents the potential first steps toward bridging molecular and cellular biology to human cognition, and it has already been used to discover underlying features of human brain development, evolution and neurological conditions. Experienced cell culture technicians can expect the protocol to take 1 month, with extended maturation, electrophysiology recording, and adeno-associated virus transduction procedure options.

This energy dependence, although suppressed by the massive QG energy scale $E_\mathrm{QG}$, expected to be on the level of the Planck energy $1.22 \times 10^{19}$ GeV, is potentially detectable in astrophysical observations. In this scenario, the cosmological distances traversed by photons act as an amplifier for this effect. By leveraging the observation of a remarkable flare from the blazar Mrk\,421, recorded at energies above 100 GeV by the MAGIC telescopes on the night of April 25 to 26, 2014, we look for time delays scaling linearly and quadratically with the photon energies. Using for the first time in LIV studies a binned-likelihood approach we set constraints on the QG energy scale. For the linear scenario, we set $95\%$ lower limits $E_\mathrm{QG}>2.7\times10^{17}$ GeV for the subluminal case and $E_\mathrm{QG}> 3.6 \times10^{17}$ GeV for the superluminal case. For the quadratic scenario, the $95\%$ lower limits for the subluminal and superluminal cases are $E_\mathrm{QG}>2.6 \times10^{10}$ GeV and $E_\mathrm{QG}>2.5\times10^{10}$ GeV, respectively.

13. Emergence of bidirectional cell laning from collective contact guidance

Abstract: Directed collective cell migration is central in morphogenesis, wound healing and cancer progression1,2. Although it is well-accepted that the molecular anisotropy of the micro-environment guides this migration3,4, its impact on the pattern of the cell flows remains largely unexplored. Studying confluent human bronchial epithelial cells (HBECs) in vitro, we show that subcellular microgrooves elicit a polar mode of collective migration in millimeter-long bidirectional lanes that are much wider than a cell size, even though cell flows are highly disordered on featureless surfaces 5. This directed flocking-like transition6,7 can be accounted for by a hydrodynamic theory of active polar fluids and corresponding numerical simulations. This model further predicts that anisotropic friction resulting from the grooves lowers the threshold of the transition, which we confirm experimentally. Therefore, microscopic anisotropy of the environment not only directs the collective motion of the cells in the easy direction, but also shapes the cell migration pattern. Flow patterns induced by collective contact guidance are thus markedly different from those induced by supracellular confinement8, demonstrating that all length-scales of the micro-environment must be considered in a comprehensive description of collective migration. Abstract: Anyons are two dimensional particles with fractional exchange statistics that emerge as elementary excitations of fractional quantum Hall phases. Experimentally, anyonic statistics manifest directly in the edge-state Fabry-P\'erot interferometer geometry, where the presence of $N_{qp}$ localized anyons in the interferometer bulk contributes a phase $N_{qp} \theta_a$ to the observed interference pattern, where $\theta_a$ is twice the statistical exchange phase. Here, we report a measurement of $\theta_a$ in a monolayer graphene Fabry-P\'erot interferometer at $\nu$ = 1/3. We find a preponderance of phase slips with magnitudes $\Delta \theta \approx 2 \pi / 3$, confirming the result of past experiments in GaAs quantum wells and consistent with expectations for the tunneling of Abelian anyons into the interferometer bulk. In contrast to prior work, however, single anyon tunneling events manifest as instantaneous and irreversible phase slips, indicative of quasiparticle equilibration times exceeding 20 minutes in some cases. We use the discrepancy between the quasiparticle equilibration rate and our measurement speed to vary the interferometer area and $N_{qp}$ independently, allowing us to precisely determine the interferometer phase and monitor the entry and exit of individual anyons to the interferometer loop in the time domain. Besides providing a replication of previous interferometric measurements sensitive to $\theta_a$ in GaAs, our results bring anyon dynamics into the experimental regime and suggest that the average `topological charge' of a mesoscopic quantum Hall device can be held constant over hour long timescales., Comment: Main Text: 9 pages, 4 figures. Abstract: Monochromatic gamma-ray signals constitute a potential smoking gun signature for annihilating or decaying dark matter particles that could relatively easily be distinguished from astrophysical or instrumental backgrounds. We provide an updated assessment of the sensitivity of the Cherenkov Telescope Array (CTA) to such signals, based on observations of the Galactic centre region as well as of selected dwarf spheroidal galaxies. We find that current limits and detection prospects for dark matter masses above 300 GeV will be significantly improved, by up to an order of magnitude in the multi-TeV range. This demonstrates that CTA will set a new standard for gamma-ray astronomy also in this respect, as the world's largest and most sensitive high-energy gamma-ray observatory, in particular due to its exquisite energy resolution at TeV energies and the adopted observational strategy focussing on regions with large dark matter densities. Throughout our analysis, we use up-to-date instrument response functions, and we thoroughly model the effect of instrumental systematic uncertainties in our statistical treatment. Abstract: PG 1553+113 is one of the few blazars with a convincing quasi-periodic emission in the gamma-ray band. The source is also a very high-energy (VHE; >100 GeV) gamma-ray emitter. To better understand its properties and identify the underlying physical processes driving its variability, the MAGIC Collaboration initiated a multiyear, multiwavelength monitoring campaign in 2015 involving the OVRO 40-m and Medicina radio telescopes, REM, KVA, and the MAGIC telescopes, Swift and Fermi satellites, and the WEBT network. The analysis presented in this paper uses data until 2017 and focuses on the characterization of the variability. The gamma-ray data show a (hint of a) periodic signal compatible with literature, but the X-ray and VHE gamma-ray data do not show statistical evidence for a periodic signal. In other bands, the data are compatible with the gamma-ray period, but with a relatively high p-value. The complex connection between the low and high-energy emission and the non-monochromatic modulation and changes in flux suggests that a simple one-zone model is unable to explain all the variability. Abstract: In recent years, a new generation of optical intensity interferometers has emerged, leveraging the existing infrastructure of Imaging Atmospheric Cherenkov Telescopes (IACTs). The MAGIC telescopes host the MAGIC-SII system (Stellar Intensity Interferometer), implemented to investigate the feasibility and potential of this technique on IACTs. After the first successful measurements in 2019, the system was upgraded and now features a real-time, dead-time-free, 4-channel, GPU-based correlator. These hardware modifications allow seamless transitions between MAGIC's standard very-high-energy gamma-ray observations and optical interferometry measurements within seconds. We establish the feasibility and potential of employing IACTs as competitive optical Intensity Interferometers with minimal hardware adjustments. The measurement of a total of 22 stellar diameters are reported, 9 corresponding to reference stars with previous comparable measurements, and 13 with no prior measurements. A prospective implementation involving telescopes from the forthcoming Cherenkov Telescope Array Observatory's northern hemisphere array, such as the first prototype of its Large-Sized Telescopes, LST-1, is technically viable. This integration would significantly enhance the sensitivity of the current system and broaden the UV-plane coverage. Abstract: Axion-like particles (ALPs) are pseudo-Nambu-Goldstone bosons that emerge in various theories beyond the standard model. These particles can interact with high-energy photons in external magnetic fields, influencing the observed gamma-ray spectrum. This study analyzes 41.3 hrs of observational data from the Perseus Galaxy Cluster collected with the MAGIC telescopes. We focused on the spectra the radio galaxy in the center of the cluster: NGC 1275. By modeling the magnetic field surrounding this target, we searched for spectral indications of ALP presence. Despite finding no statistical evidence of ALP signatures, we were able to exclude ALP models in the sub-micro electronvolt range. Our analysis improved upon previous work by calculating the full likelihood and statistical coverage for all considered models across the parameter space. Consequently, we achieved the most stringent limits to date for ALP masses around 50 neV, with cross sections down to $g_{a\gamma} = 3 \times 10^{-12}$ GeV$^{-1}$., Comment: 25 pages, 10 figures, accepted for publication in Physics of the Dark Universe

Abstract: Glioblastoma multiforme (GBM) is characterized by its infiltrative growth pattern and high recurrence rate despite treatment. While local progression within the central nervous system (CNS) is the rule, manifestations outside the CNS, particularly skin and subcutaneous metastases, are very infrequent and seldom reported in the literature. The authors reviewed the current understanding of this rare condition, with the main purpose of giving visibility to its clinical presentation and prognostic implications, thus improving clinical management and encouraging research in this area. A PubMed, Cochrane Library, and EMBASE search from database inception through March 2024 was conducted. In this way, we compiled a total of thirty-five cases in our review. As far as we know, our work gathers the largest number of patients with this condition. Remarkably, we observed that the typical presentation of soft-tissue high-grade glioma metastases is the finding of subcutaneous erythematous nodules in patients previously operated on for a primary CNS tumor, within the craniotomy site and nearby, mostly in the first year after the initial surgery. It was also noted that there is a trend of developing a concomitant CNS recurrence and/or other metastases in different locations, either simultaneously or subsequently. From here, we propose some possible mechanisms that explain the extracranial spread of GBM. We concluded that a poor outcome is expected from the diagnosis of skin and subcutaneous metastases: the mean overall survival was 4.38 months. Yet, assessing individual characteristics is always mandatory; a palliative approach seems to be the best option for the majority of cases.

Abstract: Glioblastoma multiforme (GBM) is the most common malignant primary central nervous system (CNS) tumor. It presents an aggressive pattern, with a tendency for intracranial progression despite optimal treatment. On the other hand, metastases and manifestations outside the CNS are exceptional, and very few cases have been described. The authors submitted a case of a 48-year-old male diagnosed with a left parietooccipital GBM isocitrate dehydrogenase (IDH) wild-type, non-methylated O6-methylguanine-DNA methyltransferase (MGMT). Six months after surgical treatment, followed by a chemotherapy (CT) and radiotherapy scheme, he developed a few subcutaneous erythematous nodules within the surgical scar. A punch biopsy confirmed the histopathology of GBM. The CT scan showed concomitant intracranial progression. We ruled out systemic metastases. As the performance status was good [Karnofsky Performance Status 90 (KPS 90)] and the subcutaneous implants were growing rapidly, limiting the quality of life, we decided to perform palliative surgery to remove the implants. The result was good. Unfortunately, the patient worsened during the following week, after ruling out medical complications, we attributed the worsening to cerebral tumor swelling, revealed in the CT scan, as unresponsive to steroids. He passed away a few days later. Based on the analysis of our case, we intend to provide useful information for the approach to this peculiar manifestation of GBM.

39. Beak and feather disease virus (BFDV) persists in tissues of asymptomatic wild Crimson Rosellas

Abstract: Abstract Infectious diseases can drive populations and species to extinction. Beak and feather disease virus (BFDV) is a circovirus of global conservation concern that can infect all Psittaciformes and some other species. Yet some parrot species, such as Crimson rosellas (Platycercus elegans), can live successfully with high BFDV prevalence (>40%) with no clinical signs reported in infected individuals. We assessed BFDV load in 10-12 tissues per bird, from n = 66 P. elegans, to reveal tissue tropism and BFDV persistence in tissues. Here we show that in 94% of individuals, BFDV was detected in one or more tissues. While BFDV replicated to high levels in subadults, in adults (some confirmed seropositive) the virus persisted in various tissues at much lower levels. Our findings reveal that BFDV is much more common in wild P. elegans than previously thought and suggest that current screening practices (mostly on blood) may substantially underestimate BFDV infection estimates, with implications for biosecurity and conservation programs globally.

40. Oculomic stratification of COVID-19 patients’ intensive therapy unit admission status and mortality by retinal morphological findings

Abstract: Abstract To investigate if retinal thickness has predictive utility in COVID-19 outcomes by evaluating the statistical association between retinal thickness using OCT and of COVID-19-related mortality. Secondary outcomes included associations between retinal thickness and length of stay (LoS) in hospital. In this retrospective cohort study, OCT scans from 230 COVID-19 patients admitted to the Intensive Care Unit (ITU) were compared with age and gender-matched patients with pneumonia from before March 2020. Total retinal, GCL + IPL, and RNFL thicknesses were recorded, and analysed with systemic measures collected at the time of admission and mortality outcomes, using linear regression models, Pearson’s R correlation, and Principal Component Analysis. Retinal thickness was significantly associated with all-time mortality on follow up in the COVID-19 group (p = 0.015), but not 28-day mortality (p = 0.151). Retinal and GCL + IPL layer thicknesses were both significantly associated with LoS in hospital for COVID-19 patients (p = 0.006 for both), but not for patients with pneumonia (p = 0.706 and 0.989 respectively). RNFL thickness was not associated with LoS in either group (COVID-19 p = 0.097, pneumonia p = 0.692). Retinal thickness associated with LoS in hospital and long-term mortality in COVID-19 patients, suggesting that retinal structure could be a surrogate marker for frailty and predictor of disease severity in this group of patients, but not in patients with pneumonia from other causes.

43. Occurrence, co-occurrence and persistence of symptoms of depression and post-traumatic stress disorder in survivors of COVID-19 critical illness

Abstract: Background: Intensive care unit (ICU) admission and invasive mechanical ventilation (IMV) are associated with psychological distress and trauma. The COVID-19 pandemic brought with it a series of additional long-lasting stressful and traumatic experiences. However, little is known about comorbid depression and post-traumatic stress disorder (PTSD).Objective: To examine the occurrence, co-occurrence, and persistence of clinically significant symptoms of depression and PTSD, and their predictive factors, in COVID-19 critical illness survivors.Method: Single-centre prospective observational study in adult survivors of COVID-19 with ≥24 h of ICU admission. Patients were assessed one and 12 months after ICU discharge using the depression subscale of the Hospital Anxiety and Depression Scale and the Davidson Trauma Scale. Differences in isolated and comorbid symptoms of depression and PTSD between patients with and without IMV and predictors of the occurrence and persistence of symptoms of these mental disorders were analysed.Results: Eighty-nine patients (42 with IMV) completed the 1-month follow-up and 71 (34 with IMV) completed the 12-month follow-up. One month after discharge, 29.2% of patients had symptoms of depression and 36% had symptoms of PTSD; after one year, the respective figures were 32.4% and 31%. Coexistence of depressive and PTSD symptoms accounted for approximately half of all symptomatic cases. Isolated PTSD symptoms were more frequent in patients with IMV (p≤.014). The need for IMV was associated with the occurrence at one month (OR = 6.098, p = .005) and persistence at 12 months (OR = 3.271, p = .030) of symptoms of either of these two mental disorders.Conclusions: Comorbid depressive and PTSD symptoms were highly frequent in our cohort of COVID-19 critical illness survivors. The need for IMV predicted short-term occurrence and long-term persistence of symptoms of these mental disorders, especially PTSD symptoms. The specific role of dyspnea in the association between IMV and post-ICU mental disorders deserves further investigation.Trial registration: ClinicalTrials.gov identifier: NCT04422444.

Abstract: Este trabalho avalia comparativamente o desempenho de dois sistemas de tradução automática, a saber, DeepL e Google Tradutor. O estudo parte de uma breve visão geral sobre traduções automáticas com a utilização de redes neurais, seguido de uma reflexão sobre os resultados da tradução automática ao utilizar a avaliação automática e humana. Subsequentemente, aborda a diferença de desempenho entre ambos os sistemas, além de identificar possíveis problemas linguísticos advindos dos resultados da tradução automática gerada por eles. Para a realização dessa comparação, o trabalho emprega um trecho da obra de Machado de Assis, Dom Casmurro, do capítulo “Olhos de Ressaca”. O estudo utiliza as traduções geradas de forma automática para analisar criticamente qual sistema apresenta mais qualidade no resultado da tradução e examina comparativamente os erros linguísticos gerados pelos resultados do DeepL e Google Tradutor. Os critérios de avaliação incluem a precisão da tradução, a capacidade de manter o significado e a estrutura da frase original, a fluência e adequação da tradução resultante.

45. The epsilon toxin from Clostridium perfringens stimulates calcium‐activated chloride channels, generating extracellular vesicles in Xenopus oocytes

Abstract: Abstract The epsilon toxin (Etx) from Clostridium perfringens has been identified as a potential trigger of multiple sclerosis, functioning as a pore‐forming toxin that selectively targets cells expressing the plasma membrane (PM) myelin and lymphocyte protein (MAL). Previously, we observed that Etx induces the release of intracellular ATP in sensitive cell lines. Here, we aimed to re‐examine the mechanism of action of the toxin and investigate the connection between pore formation and ATP release. We examined the impact of Etx on Xenopus laevis oocytes expressing human MAL. Extracellular ATP was assessed using the luciferin‐luciferase reaction. Activation of calcium‐activated chloride channels (CaCCs) and a decrease in the PM surface were recorded using the two‐electrode voltage‐clamp technique. To evaluate intracellular Ca2+ levels and scramblase activity, fluorescent dyes were employed. Extracellular vesicles were imaged using light and electron microscopy, while toxin oligomers were identified through western blots. Etx triggered intracellular Ca2+ mobilization in the Xenopus oocytes expressing hMAL, leading to the activation of CaCCs, ATP release, and a reduction in PM capacitance. The toxin induced the activation of scramblase and, thus, translocated phospholipids from the inner to the outer leaflet of the PM, exposing phosphatidylserine outside in Xenopus oocytes and in an Etx‐sensitive cell line. Moreover, Etx caused the formation of extracellular vesicles, not derived from apoptotic bodies, through PM fission. These vesicles carried toxin heptamers and doughnut‐like structures in the nanometer size range. In conclusion, ATP release was not directly attributed to the formation of pores in the PM, but to scramblase activity and the formation of extracellular vesicles.

49. Optical coherence tomography angiography analysis methods: a systematic review and meta-analysis

Abstract: Abstract Optical coherence tomography angiography (OCTA) is widely used for non-invasive retinal vascular imaging, but the OCTA methods used to assess retinal perfusion vary. We evaluated the different methods used to assess retinal perfusion between OCTA studies. MEDLINE and Embase were searched from 2014 to August 2021. We included prospective studies including ≥ 50 participants using OCTA to assess retinal perfusion in either global retinal or systemic disorders. Risk of bias was assessed using the National Institute of Health quality assessment tool for observational cohort and cross-sectional studies. Heterogeneity of data was assessed by Q statistics, Chi-square test, and I2 index. Of the 5974 studies identified, 191 studies were included in this evaluation. The selected studies employed seven OCTA devices, six macula volume dimensions, four macula subregions, nine perfusion analyses, and five vessel layer definitions, totalling 197 distinct methods of assessing macula perfusion and over 7000 possible combinations. Meta-analysis was performed on 88 studies reporting vessel density and foveal avascular zone area, showing lower retinal perfusion in patients with diabetes mellitus than in healthy controls, but with high heterogeneity. Heterogeneity was lowest and reported vascular effects strongest in superficial capillary plexus assessments. Systematic review of OCTA studies revealed massive heterogeneity in the methods employed to assess retinal perfusion, supporting calls for standardisation of methodology.

Abstract: Abstract Despite the importance of protecting forests and woodlands to achieve global climate and biodiversity goals, logging impacts persist worldwide. Forestry advocates often downplay these impacts but rarely consider the cumulative threat deforestation and degradation has had, and continues to have, on biodiversity. Using New South Wales (Australia) as a case study, we quantify the extent of deforestation and degradation from 1788 (pre‐European colonization) to 2021. We used historical loss as a baseline to evaluate recent logging (2000–2022) and the condition of the remaining native forest and woodland. Condition was quantified by measuring the similarity of a current ecosystem to a historical reference state with high ecological integrity. Using these data, we measured the impacts on 269 threatened terrestrial species. We show that possibly over half (29 million ha) of pre‐1788 native forest and woodland vegetation in NSW has been lost. Of the remaining 25 million ha, 9 million ha is estimated to be degraded. We found recent logging potentially impacted 150 species that had already been affected by this historical deforestation and degradation, but the impacts varied across species. Forty‐three species that were identified as impacted by historical deforestation and degradation and continue to be impacted by logging, now have ≤50% of their pre‐1788 extent remaining that is intact and nine species now have ≤30%. Our research contextualizes the impact of current logging against historical deforestation and highlights deficiencies in environmental assessments that ignore historical baselines. Future land management must consider both the extent and condition of remaining habitat based on pre‐1788 extents.
