Modeling and simulation of complex systems frequently requires capturing probabilistic dynamics across multiple scales and/or multiple domains. Cyber–physical, cyber–social, socio–technical, and cyber–physical–social systems are common examples. Modeling and simulating such systems via a single, all-encompassing model is often infeasible, and thus composable modeling techniques are sought. Co-simulation and closure modeling are two prevalent composable modeling techniques that divide a multi-scale/multi-domain system into sub-systems, use smaller component models to capture each sub-system, and coordinate data transfer between component models. While the two techniques have similar goals, differences in their methods lead to differences in the complexity and computational efficiency of a simulation model built using one technique or the other. This paper presents a probabilistic analysis of the complexity and computational efficiency of these two composable modeling techniques for multi-scale/multi-domain complex system modeling and simulation applications. The aim is twofold: to promote awareness of these two composable modeling approaches and to facilitate complex system model design by identifying circumstances that are amenable to either approach. Paratemporal methods based on tree expansion have proven to be effective in efficiently generating the trajectories of stochastic systems. However, combinatorial explosion of branching arising from multiple choice points presents a major hurdle that must be overcome to implement such techniques. In this paper, we tackle this scalability problem by developing a systems theory-based framework covering both conventional and proposed tree expansion algorithms for speeding up discrete event system stochastic simulations while preserving the desired accuracy. An example is discussed to illustrate the tree expansion framework in which a discrete event system specification (DEVS) Markov stochastic model takes the form of a tree isomorphic to a free monoid over the branching alphabet. We derive the computation times for baseline, non-merging, and merging tree expansion algorithms to compute the distribution of output values at any given depth. The results show the remarkable reduction from exponential to polynomial dependence on depth effectuated by node merging. We relate these results to the similarly reduced computation time of binomial coefficients underlying Pascal's triangle. Finally, we discuss the application of tree expansion to estimating temporal distributions in stochastic simulations involving serial and parallel compositions with potential real-world use cases. The kefir‐based smoothies with kale and spinach were designed as a ready‐to‐drink and innovative functional snack. Microbiological, physicochemical, as well as pre‐ and postgastrointestinal total antioxidant capacity (TAC; CUPRAC, DPPH, and FRAP) analyses were conducted. It was determined that the kefir‐based smoothies with vegetables had higher ash, carbohydrate, and dietary fiber values. Fructose and glucose contents of smoothy with kale were high, while smoothy with spinach included high sucrose and maltose. The microbiology results revealed that kefir‐based vegetable smoothies had minimum Lactobacillaceae viability (>log 7 cfu g−1) for the required functional effect after 14‐day storage. Moreover, the addition of kale significantly increased (p <.01) the level of initial TAC (CUPRAC, DPPH, and FRAP) and total phenolic content (TPC) values. After in vitro gastric digestion analysis, smoothie with spinach demonstrated higher TAC and TPC values and the control sample had higher TAC and TPC values compared with a predigestion step. It was found that in vitro intestinal DPPH values were higher for the sample with spinach samples, while the sample with kale had the highest FRAP values. It was also found that the bioaccessibility indexes of plain kefir were determined to be the highest in both in vitro gastric and intestinal procedures. The present study provided novel insights into the in vitro digestion properties of kefir fortified with vegetables. Industry 4.0 enables the modernisation of machines and opens up the digitalisation of processes in the manufacturing industry. As a result, these machines are ready for predictive maintenance as part of Industry 4.0 services. The benefit of predictive maintenance is that it can significantly extend the life of machines. The integration of predictive maintenance into existing production environments faces challenges in terms of data understanding and data preparation for machines and legacy systems. Current AI frameworks lack adequate support for the ongoing task of data integration. In this context, adequate support means that the data analyst does not need to know the technical background of the pilot's data sources in terms of data formats and schemas. It should be possible to perform data analyses without knowing the characteristics of the pilot's specific data sources. The aim is to achieve a seamless integration of data as information for predictive maintenance. For this purpose, the developed data-sharing infrastructure enables automatic data acquisition and data integration for AI frameworks using interoperability methods. The evaluation, based on two pilot projects, shows that the step of data understanding and data preparation for predictive maintenance is simplified and that the solution is applicable for new pilot projects. The practice of deceased-donor solid-organ transplantation is unique among clinical therapies because it substantially benefits recipients, The allocation process is a complex System-of-Systems (SoS) with multiple stakeholders, components, rules, and policies. While we model and simulate such SoS, they are challenging to verify and validate thoroughly due to a large number of use cases and the sensitivity to initial conditions. Furthermore, subject matter experts within communities of practice do not trust such models because they are difficult to explain. This paper proposes a trust-centric approach to validation focusing on experimentally exploring the simulation's behavior space and demonstrating its utility to a community of experts. We describe a three-step validation process to show how modelling and simulation professionals can foster trust in simulations of a complex SoS. A major goal of healthy aging is to prevent declining resilience and increasing frailty, which are associated with many chronic diseases and deterioration of stress response. Here, we propose a loss‐or‐gain survival model, represented by the ratio of cumulative stress span to life span, to quantify stress resilience at organismal level. As a proof of concept, this is demonstrated by reduced survival resilience in Caenorhabditis elegans exposed to exogenous oxidative stress induced by paraquat or with endogenous proteotoxic stress caused by polyglutamine or amyloid‐β aggregation. Based on this, we reveal that a hidden peptide ("cryptide")—AbaPep#07 (SETYELRK)—derived from abalone hemocyanin not only enhances survival resilience against paraquat‐induced oxidative stress but also rescues proteotoxicity‐mediated behavioral deficits in C. elegans, indicating its capacity against stress and neurodegeneration. Interestingly, AbaPep#07 is also found to increase cost‐free longevity and age‐related physical fitness in nematodes. We then demonstrate that AbaPep#07 can promote nuclear localization of SKN‐1/Nrf, but not DAF‐16/FOXO, transcription factor. In contrast to its effects in wild‐type nematodes, AbaPep#07 cannot increase oxidative stress survival and physical motility in loss‐of‐function skn‐1 mutant, suggesting an SKN‐1/Nrf‐dependent fashion of these effects. Further investigation reveals that AbaPep#07 can induce transcriptional activation of immune defense, lipid metabolism, and metabolic detoxification pathways, including many SKN‐1/Nrf target genes. Together, our findings demonstrate that AbaPep#07 is able to boost stress resilience and reduce behavioral frailty via SKN‐1/Nrf‐governed transcriptional reprogramming, and provide an insight into the health‐promoting potential of antioxidant cryptides as geroprotectors in aging and associated conditions. Using collaborative autoethnography, critical incidents, and artefacts, one teacher/coach (T/C) role conflict is contextualized in the Canadian physical education & school sport (PESS) setting. The purpose of the project was to use CAE in conjunction with a systematic analysis to promote personal narratives as both an approach to practitioner reflection and as a form of research inquiry to forward the field of PESS and occupational socialization. Data analysis sought to answer the singular question: Was the experience educative? An educative experience required alignment with the Meaningful PE democratic principles of autonomy and inclusivity. Critical accounts of T/C role conflicts resulted in the undesired continuity of T/C role socialization. Aberrant regulation of unfolded protein response (UPR)/endoplasmic reticulum (ER) stress pathway is associated with cancer development, metastasis, and relapse, and the UPR signal transducer ATF6 has been proposed as a diagnostic and prognostic marker for many cancers. However, a causal molecular link between ATF6 activation and carcinogenesis is not established. Here, it is found that tumor protein D52 (TPD52) integrates ER stress and UPR signaling with the chaperone machinery by promoting S2P-mediated cleavage of ATF6. Although TPD52 has been generally considered as an oncogene, TPD52 is identified as a novel tumor suppressor in bladder cancer. Significantly, attenuation of the ER stress via depletion of TPD52 facilitated tumorigenesis in a subset of human carcinomas. Furthermore, the APC Cdc20 E3 ligase is validated as the upstream regulator marking TPD52 for polyubiquitination-mediated proteolysis. In addition, inactivation of Cdc20 sensitized cancer cells to treatment with the ER stress inducer in a TPD52-dependent manner. Care as usual has failed to stem the tide of mental health challenges in children and young people. Transformed models of care and prevention are required, including targeting the social determinants of mental health. Robust economic evidence is crucial to guide investment towards prioritised interventions that are effective and cost-effective to optimise health outcomes and ensure value for money. Mental healthcare and prevention exhibit the characteristics of complex dynamic systems, yet dynamic simulation modelling has to date only rarely been used to conduct economic evaluation in this area. This article proposes an integrated decision-making and planning framework for mental health that includes system dynamics modelling, cost-effectiveness analysis, and participatory model-building methods, in a circular process that is constantly reviewed and updated in a 'living model' ecosystem. We describe a case study of this approach for mental health system policy and planning that synergises the unique attributes of a system dynamics approach within the context of economic evaluation. This kind of approach can help decision makers make the most of precious, limited resources in healthcare. This state-of-the-art review is geared toward elucidating the molecular understanding of the carbon-based flame-retardant mechanisms for polymers via holistic characterization combining detailed analytical assessments and computational material science. The use of carbon-based flame retardants, which include graphite, graphene, carbon nanotubes (CNTs), carbon dots (CDs), and fullerenes, in their pure and functionalized forms are initially reviewed to evaluate their flame retardancy performance and to determine their elevation of the flammability resistance on various types of polymers. The early transition metal carbides such as MXenes, regarded as next-generation carbon-based flame retardants, are discussed with respect to their superior flame retardancy and multifunctional applications. At the core of this review is the utilization of cutting-edge molecular dynamics (MD) simulations which sets a precedence of an alternative bottom-up approach to fill the knowledge gap through insights into the thermal resisting process of the carbon-based flame retardants, such as the formation of carbonaceous char and intermediate chemical reactions offered by the unique carbon bonding arrangements and microscopic in-situ architectures. Combining MD simulations with detailed experimental assessments and characterization, a more targeted development as well as a systematic material synthesis framework can be realized for the future development of advanced flame-retardant polymers., (© 2024 The Author(s). Xylooligosaccharides (XOS) are considered a potent source of prebiotics for humans. The global prebiotic market is expanding in size, was valued at USD 6.05 billion in 2021, and is expected to grow at a 14.9% compound annual growth rate between 2022 and 2030, indicating a huge demand. These XOS are non-digestible pentose sugar oligomers comprising mainly xylose. Xylose is naturally present in the lignocellulosic biomass (LCB), fruits and vegetables. Apart from the prebiotic effect, these XOS have been reported to reduce blood cholesterol, possess antioxidant effects, increase calcium absorption, reduce colon cancer risk, and benefit diabetic patients. The primary use of XOS is reported in the feed industry followed by health, medical use, food and drinks. LCB mainly contains glucan, xylan and lignin. After glucan, xylan is the second-highest available sugar on the globe composed of xylose. Therefore, the xylan fraction of LCB has great significance in producing food, feed and energy. Glucan has been exploited for the commercial production of ethanol, xylitol, furfural, hydroxymethyl furfural and glucose. As of now, xylan has limited applications. Therefore, xylan can be exploited to convert to XOS. The production of XOS from LCB fraction not only helps to produce these at a very low price, but also helps in the reduction of greenhouse gases. Its use in food and drinks is increasing as it can be derived from the abundantly and cheaply available LCB. With the booming development of door-to-door housekeeping service, the platform faces the problem of order assignment. Improving the matching mechanism between orders and housekeepers based on a dynamic programming (DP) algorithm can not only achieve flexible order allocation but can also improve the service efficiency and service quality. In this paper, a single objective nonlinear programming model is established, which takes the maximum total weight value as the objective function to study the order allocation problem under offline and online conditions. Under the offline condition, the number of housekeepers is taken as the decision variable. The status of order and housekeeper, order time, and action trajectory are taken as constraints. For online assignment, the order backlog status is treated as the decision variable. The reliability of the model was verified using real data from 20 groups of housekeepers and 50 groups of orders. Finally, the effect of order backlog on online allocation is discussed and the optimal threshold and maximum weight are found. The online order assignment model is compared with the nearest distance assignment model. The results show that the online assignment model with a total weighted score of 1045.14 is better than the nearest distance assignment model with a score of 810.25. [ABSTRACT FROM AUTHOR]

Scientific drilling expeditions offer a unique opportunity to characterize microbial communities in the subsurface that have long been isolated from the surface. With subsurface microbial biomass being low in general, biological contamination from the drilling fluid, sample processing, or molecular work is a major concern. To address this, characterization of the contaminant populations in the drilling fluid and negative extraction controls are essential for assessing and evaluating such sequencing data. Here, rock cores down to 2250 m depth, groundwater-bearing fractures, and the drilling fluid were sampled for DNA to characterize the microbial communities using a broad genomic approach. However, even after removing potential contaminant populations present in the drilling fluid, notorious contaminants were abundant and mainly affiliated with the bacterial order Burkholderiales. These contaminant microorganisms likely originated from the reagents used for isolating DNA despite stringent quality standards during the molecular work. The detection of strictly anaerobic sulfate reducers such as Candidatus Desulforudis audaxviator suggested the presence of autochthonous deep biosphere taxa in the sequenced libraries, yet these clades represented only a minor fraction of the sequence counts (< 0.1 %), hindering further ecological interpretations. The described methods and findings emphasize the importance of sequencing extraction controls and can support experimental design for future microbiological studies in conjunction with continental drilling operations. In this study, the viability of Lactobacillus bacteria in probiotic dairy products sold in Qazvin city within two periods was examined. Eighteen samples of dairy products (yogurt, cheese, and kefir drinks) labeled with probiotics from different brands of various companies in Iran were prepared. Dilutions were prepared appropriately from each sample and inoculated into MRS Bile Agar culture medium as a pour plate culture procedure. After incubation, the colonies were counted and checked to see if the standards of the Iranian National Standardization Organization (INSO) (min 106 CFU/gr) were met. Then, to perform probiotic confirmation tests, each colony characterized different morphological was examined regarding catalase and Gram staining tests. The average count of live Lactobacillus showed that out of eighteen samples of probiotic dairy products, only one sample (5.5%) had the minimum number of bacteria according to INSO. In the yogurt, none of the samples could grow in the bile culture medium. In the cheese, none of the samples had the minimum number of viable bacteria according to the standard. Regarding the kefir drink, only one sample had the minimum number of live bacteria based on the standards, in the first week after the production date. In most of the dairy products labeled as probiotic, the average count of Lactobacillus with the ability to grow in bile, is lower than INSO measurements, which is at least one million bacteria per gram of a product. Sugarcane bagasse is an agro-industrial waste produced from the sugar industry, which is composed of cellulose, hemicellulose, and lignin. It can be used as a raw material to get xylose, which is used to obtain various products of industrial interest, such as bioethanol and xylitol, among others. The objective of this work was to optimize the acid hydrolysis stage of sugarcane bagasse to obtain the maximum of xylose and the minimum of acetic acid concentration. The response surface methodology and the desirability criterion were used, evaluating the concentration of sulfuric acid (H2SO4: 1, 2, 3% v/v), the solid-liquid ratio (LSR: 6, 9, 12 ml/g), and reaction time (t: 10, 20, 30 min), as dependent variables and as response variables the maximum of xylose and the minimum of acetic acid concentration. The optimal conditions found by acid hydrolysis were 0.86% v/v of H2SO4, 22.7 min of reaction time, and 6.2 of LSR showing values of 20.0 g/L of xylose and 3.05 g/L of acetic acid. These conditions were experimentally validated, obtaining 20.37 ± 0.12 g/L, while the acetic acid concentration was 2.82 ± 0.05 g/L. These results showed that the optimization method used is good, since it was possible to accurately validate the conditions obtained, achieving similar results to those found by the optimization model, and also 75% of removal of hemicellulose was revealed. Annuities provide a lifelong income stream and can therefore help individuals to mitigate the risk of outliving their savings, a highly topical issue in the context of increasing life expectancies. Given that real-world annuitization rates remain relatively low, we investigate the influence of behavioral biases on people's choice between a lifelong annuity and a lump sum payout. In so doing, we focus on the impact of default effects due to a preselected annuity option (default option) and the impact of the decision's timing (decisions on annuitization taken by younger individuals refer to a distant future, those taken by older individuals to a near future) on annuity uptake and health consciousness. We used a scientific survey panel to conduct an online experiment with a diversified sample of 339 participants (55.2% female, average age = 42.5 years). Our results show that the timing of the decision moderates the default effect on annuity uptake, in that the effect of a preselected (default) annuity option is stronger for distant-future decisions (i.e., choice of annuity instead of lump sum at retirement made by younger participants purchasing a deferred annuity) than for near-future decisions made by older individuals who are closer to retirement. We further find that the default effect moderates health consciousness after choosing an annuity. Health consciousness is stronger in the no-default condition than in the annuity default condition. Membrane-aerated biofilm reactor (MABR) has been considered as an innovative technology to solve aeration issues in conventional bioreactors. MABR uses a membrane to supply oxygen to biofilm grown on the membrane surface. MABR can perform bubbleless aeration with high oxygen transfer rates, which can reduce energy requirements and expenses. In addition, a unique feature of counter-diffusion creates a stratified biofilm structure, allowing the simultaneous nitrification–denitrification process to take place in a single MABR. Controlling the biofilm is crucial in MABR operation, since its thickness significantly affects MABR performance. Several approaches have been proposed to control biofilm growth, such as increasing shear stress, adding chemical agents (e.g., surfactant), using biological predators to suppress microorganism growth, and introducing ultrasound cavitation to detach biofilm. Several studies also showed the important role of membrane properties and configuration in biofilm development. In addition, MABR demonstrates high removal rates of pollutants in various wastewater treatments, including in full-scale plants. This review presents the basic principles of MABR and the effect of operational conditions on its performance. Biofilm formation, methods to control its thickness, and membrane materials are also discussed. In addition, MABR performance in various applications, full-scale MBRs, and challenges is summarized. Coating surfaces with bioactive glass can be defined as depositing fine bioactive glasses on biomaterial substrates. Cobalt-chromium is a viable alternative to stainless steel for long-term applications with superior ductility. The mechanical properties of cobalt-chromium alloys are high strength with elastic modulus of 220-2300 GPa, more significant than the 30 GPa of bones. Combining metals and bioactive glass results in high biocompatibility and improved bioactivity of implant surfaces. In addition, it triggers new bone tissue to regenerate through osteogenesis and mineralisation. However, implantation failure still occurs and requires surgery revision due to a lack of adequate bone bonding and delamination at the coating surface of the implant. The improved bulk and surface function of manufactured implants has advanced implantation procedures, leading to a decline in surgical risks. Many current techniques discussed in the literature are related to additive manufacturing (AM) of lightweight implants based on reliable, precise, flexible scaffolds and capable of mimicking bone properties while incorporating other useful features. These techniques have evolved for the production of a variety of biocompatible materials. AM has progressed beyond prototype to full-scale manufacturing of metals, polymers and ceramic products. However, metallic implants often fail in vivo due to biocorrosion and deterioration, limiting implant longevity. This study reviews current trends and approaches to enhancing the surface corrosion resistance of porous metallic implants and the effect of interfacial films on biological activity. The art of porous metallic implants manufactured by AM and their biocorrosion behaviour are discussed. Diagnosis of primary hyperparathyroidism (PHP) is based on blood assessments in terms of synchronous high calcium and PTH (parathormone), but further management, particularly parathyroid surgery that provides the disease cure in 95–99% of cases, requires an adequate localisation of the parathyroid tumour/tumours as the originating source, with ultrasound and 99m-Technetium (99m-Tc) sestamibi scintigraphy being the most widely used. We aimed to introduce an adult female case diagnosed with PHP displaying unexpected intra-operatory findings (ectopic thyroid tissue) in relation to concordant pre-operatory imaging modalities (ultrasound + dual-phase 99m-Tc pertechnetate and sestamibi scintigraphy + computed tomography) that indicated bilateral inferior parathyroid tumours. A sudden drop in PTH following the removal of the first tumour was the clue for performing an extemporaneous exam for the second mass that turned out to be non-malignant ectopic thyroid tissue. We overviewed some major aspects starting from this case in point: the potential pitfalls of pre-operatory imaging in PHP; the concordance/discordance of pre-parathyroidectomy localisation modalities; the need of using an additional intra-operatory procedure; and the clues of providing a distinction between pathological parathyroids and thyroid tissue. This was a case of adult PHP, whereas triple localisation methods were used before parathyroidectomy, showing concordant results; however, the second parathyroid adenoma was a false positive image and an ectopic thyroid tissue was confirmed. The pre-operatory index of suspicion was non-existent in this patient. Hybrid imaging modalities are most probably required if both thyroid and parathyroid anomalies are suspected, but, essentially, awareness of the potential pitfalls is mandatory from the endocrine and surgical perspectives. Current gaps in imaging knowledge to guide us in this area are expected to be solved by the significant progress in functional imaging modalities. However, the act of surgery, including the decision of a PTH assay or extemporaneous exam (as seen in our case), represents the key to a successful removal procedure. Sweet potato stem nematode disease is a devastating disease, which seriously affects the yield and quality of sweet potato (Ipomoea batatas (L.) Lam.). At present, soil treatment with pesticides is mainly used to prevent sweet potato stem nematode disease. In this study, Illumina MiSeq high‐throughput sequencing technology was used to analyze the diversity of soil bacterial and fungal communities treated with different pesticides. At the same time, high performance liquid chromatography was used to determine the pesticide residues in soil and sweet potato, and the impact on sweet potato yield was investigated. Kruskal–Wallis test was used to analyze the four α‐diversity indexes of soil under different pesticide treatments, and the differences were not significant (p > 0.05), indicating that there was no significant difference in the diversity and abundance of bacterial and fungal communities in soil under different pesticide treatments. The results of principal coordinate analysis showed that there was no significant difference in bacterial and fungal community structure among different pesticide treatments (p > 0.05). Pesticide residue analysis and yield statistics of sweet potato showed that the residual amount of three kinds of pesticides in sweet potato did not exceed the maximum limit stipulated by the current National Standards for Food safety (GB2763‐2021). When 5% chlorpyrifos phoxim particles were applied at 15.18 kg/acre, sweet potato was unaffected by pests and diseases basically, and the yield was the highest. [ABSTRACT FROM AUTHOR]

Background: Temperament Based Therapy with Support (TBT-S) is an emerging intervention based on empirically supported neurobiological models. Due to its novelty, only a handful of studies to date have examined TBT-S, and none of these previous studies have provided a qualitative evaluation of how TBT-S is perceived by the target population. Therefore, the aim of the current study was to provide an increased understanding of how TBT-S is experienced by patients with an eating disorder and their supports., Method: Forty-six patients with an eating disorder and 63 supports consented to be included in the study. The participants provided written responses to six open-ended questions during the post-treatment assessment, detailing their treatment experiences and offering additional feedback. Thematic analysis (TA) was used to analyze their written responses, aiming for a combination of latent and semantic themes., Results: The results reveal a substantial overlap between patients' and supports' experiences with TBT-S. In both groups, identified themes suggest increased knowledge and hopefulness as key benefits of the intervention. While both patients and support persons considered TBT-S to be worthwhile, patients also reported finding the intervention quite challenging. Additionally, both groups emphasized the neurobiological rationale as an essential component of TBT-S., Conclusions: The qualitative evaluations from this study offer new insights into how TBT-S is experienced by the target population. Advanced in situ analyses are indispensable for comprehending the catalyst aging mechanisms of Pt-based PEM fuel cell cathode materials, particularly during accelerated stress tests (ASTs). In this study, a combination of in situ small-angle and wide-angle X-ray scattering (SAXS & WAXS) techniques were employed to establish correlations between structural parameters (crystal phase, quantity, and size) of a highly active skeleton-PtCo (sk-PtCo) catalyst and their degradation cycles within the potential range of the start-up/shut-down (SUSD) conditions. Despite the complex case of the sk-PtCo catalyst comprising two distinct fcc alloy phases, our complementary techniques enabled in situ monitoring of structural changes in each crystal phase in detail. Remarkably, the in situ WAXS measurements uncover two primary catalyst aging processes, namely the cobalt depletion (regime I) followed by the crystallite growth via Ostwald ripening and/or particle coalescence (regime II). Additionally, in situ SAXS data reveal a continuous size growth over the AST. The Pt-enriched shell thickening based on the Co depletion within the first 100 SUSD cycles and particle growth induced by additional potential cycles were also collaborated by ex situ STEM-EELS. Aims: Papillary thyroid cancer (PTC) is a serious threat to human health worldwide, while metastasis in the early phase limits therapeutic success and leads to poor survival outcomes. The CXC chemokine receptor type 4 (CXCR4) plays an important role in many cellular movements such as transcriptional modulation, cell skeleton rearrangement, and cell migration, and the change in CXCR4 levels are crucial in various diseases including cancer. In this study, we explored the role of CXCR4 in the migration and invasion of PTC and investigated the potential mechanisms underlying its effects., Subjects and Methods: We analyzed the expression levels of CXCR4 in PTC tissues and cell lines. Would healing migration, Transwell invasion assay in vitro, and tail-vein lung metastasis assay In vivo were performed to evaluated the migration and invasion abilities of PTC cells with stable CXCR4 knockdown or overexpression. Signal transducers and activators of transcription (STAT3) signaling pathway-related protein expressions were examined by Western blotting assays., Results: The results showed that CXCR4 was highly expressed in PTC cell lines and PTC tissues. CXCR4 knockdown in PTC cells dampened the migration, invasion, and epithelial-mesenchymal transition (EMT), whereas CXCR4 overexpression enhanced these properties. In vivo, we also found that CXCR4 promoted the metastasis of PTC. Mechanistic studies showed that CXCR4 played these vital roles through the STAT3 signaling pathway. Furthermore, PTC patients with high CXCR4 or p-STAT3 expression correlated with aggressive clinical characteristics such as extrathyroidal extension (ETE), and lymph node metastasis (LNM)., Conclusions: We provided evidence that CXCR4 might activate the STAT3 signaling pathway and further promote PTC development. This work is primarily focused on overcoming the limitations of polymeric membranes in achieving the balance between permeability and selectivity of the separation performance. The filler, Zeolitic imidazole framework -67 (ZIF-67) nanoparticles were synthesised in cubical morphology using hexadecyltrimethylammonium bromide (CTAB) as a surfactant via the wet-chemical method. The uniform particles with particle sizes ranging between 120-180 nm were incorporated into the polyvinylidene fluoride (PVDF) matrix to fabricate mixed matrix membranes via the phase inversion method. These mixed matrix membranes were systematically characterised to confirm the chemical, structural and morphological properties of the materials and membranes. Furthermore, the membranes showed a 56.5% improvement in their mechanical properties. The results confirm that 5 wt.% ZIF-67/PVDF membrane showed the best separation results compared to its pure counterpart. The permeability of H₂ gas was reported to be 1,094,511 Barrer, with selectivities of 3.03 for H₂/CO₂ and 3.06 for H₂/N₂. This represents a 210.6% increase in the permeability of H₂ gas. These results demonstrate the influence of ZIF-67 loading in the PVDF polymer matrix along with the potential of ZIF-67/PVDF mixed matrix membranes in the field of hydrogen separation and purification., (© 2024. Purpose: Detection of osseous metastases by imaging can be challenging in patients with invasive lobular breast cancer (ILC). ILC may demonstrate low metabolic rate due to lower tumor cell density, decreased proliferation rate, diffuse infiltration of surrounding tissue, and low level of GLUT-1 expression. The aim of this study is to assess the diagnostic accuracy of 18F-FDG PET/CT in identifying bone metastases in ILC patients., Material and Methods: Out of 52 individuals diagnosed with lobular breast cancer and underwent 18F-FDG PET/CT for evaluation of metastases, 21 patients were included in our study population after applying inclusion and exclusion criteria. The radiological and clinical follow-up of at least 6 months served as the reference standard comparator., Results: Bone metastases were confirmed in six patients. 18F-FDG PET/CT was true positive in two and false negative in four patients. The sensitivity, specificity, positive predictive value, negative predictive value, and accuracy of 18F-FDG PET/CT were 33.33, 93.33, 66.67, 77.78, and 76.19%, respectively (95% confidence interval). The tendency of ILC osseous metastases to be more sclerotic explains the low diagnostic accuracy of 18F-FDG PET/CT in detection of bone metastases, making it a less suited method of evaluation., Conclusion: The results of the present study indicate that 18F-FDG PET/CT has a low diagnostic accuracy in detecting bone metastases in lobular breast cancer and, by inference, new functional modalities can be explored in these patients. Seafood is a prime target for fraudulent activities due to the complexity of its supply chain, high demand, and difficult discrimination among species once morphological characteristics are removed. Instances of seafood fraud are expected to increase due to growing demand. This manuscript reviews the application of DNA-based methods for commercial fish authentication and identification from 2000 to 2023. It explores (1) the most common types of commercial fish used in assay development, (2) the type of method used, (3) the gene region most often targeted, (4) provides a case study of currently published assays or primer-probe pairs used for DNA amplification, for specificity, and (5) makes recommendations for ensuring standardized assay-based reporting for future studies. A total of 313 original assays for the detection and authentication of commercial fish species from 191 primary articles published over the last 23 years were examined. The most explored DNA-based method was real-time polymerase chain reaction (qPCR), followed by DNA sequencing. The most targeted gene regions were cytb (cytochrome b) and COI (cytochrome c oxidase 1). Tuna was the most targeted commercial fish species. A case study of published tuna assays (n = 19) targeting the cytb region found that most assays were not species-specific through in silico testing. This was conducted by examining the primer mismatch for each assay using multiple sequence alignment. Therefore, there is need for more standardized DNA-based assay reporting in the literature to ensure specificity, reproducibility, and reliability of results. Taxon-specific quantitative PCR (qPCR) assays are commonly used for environmental DNA sampling-based inference of animal presence. These assays require thorough validation to ensure that amplification truly indicates detection of the target taxon, but a thorough validation is difficult when there are potentially many non-target taxa, some of which may have incomplete taxonomies. Here, we use a previously published, quantitative model of cross-amplification risk to describe a framework for assessing qPCR assay specificity when there is missing information and it is not possible to assess assay specificity for each individual non-target confamilial. In this framework, we predict assay specificity against unsampled taxa (non-target taxa without sequence data available) using the sequence information that is available for other confamilials. We demonstrate this framework using four case study assays for: (1) An endemic, freshwater arthropod (meltwater stonefly; Lednia tumana), (2) a globally distributed, marine ascidian (Didemnum perlucidum), (3) a continentally distributed freshwater crustacean (virile crayfish; Faxonius virilis, deanae and nais species complex) and (4) a globally distributed freshwater teleost (common carp; Cyprinus carpio and its close relative C. rubrofuscus). We tested the robustness of our approach to missing information by simulating application of our framework for all possible subsamples of 20-all non-target taxa. Our results suggest that the modelling framework results in estimates which are largely concordant with observed levels of cross-amplification risk using all available sequence data, even when there are high levels of data missingness. We explore potential limitations and extensions of this approach for assessing assay specificity and provide users with an R Markdown template for generating reproducible reports to support their own assay validation efforts., (Published 2024. Scope: Consumption of inulin could affect the intestinal microbiota composition. Hereby, it is aimed to investigate the intestinal microbial community restoration process when the inulin supplementation is terminated (i.e., the secondary effect)., Methods and Results: The current study investigates the response and restoration of intestinal microbiota to/after high (Inulin-H) and low (Inulin-L) dosage of inulin supplementation or sequential antibiotics and inulin (Anti-Inulin-L) supplementation, based on analysis of 16S rRNA gene sequences in C57BL/6 mice. The number of significantly changed genera in response to inulin is highest in Anti-Inulin-L (n = 66) group, followed by Inulin-H (n = 51) and Inulin-L (n = 38) group. After inulin supplementation stops, microbiota of all studied groups tend to recover to their original states, with highest percentage of inulin-responding microbes stay significantly different at Anti-Inulin-L (93.94%) group, followed by Inulin-H (74.51%) and Inulin-L (44.12%) groups. Of note, the relative abundance of some non-inulin-responding taxa significantly increases during restoration., Conclusion: Sequential antibiotics and inulin supplementation induce greatest changes in the intestinal microbial composition, followed by high and low dosage of inulin. Istanbul is a megacity with a population of 15.5 million and is one of the fastest-growing cities in Europe. Due to the rapidly increasing population and urbanization, Istanbul's daily water needs are constantly increasing. In this study, eight drinking water basins that supply water to Istanbul were comprehensively examined using remote sensing observations and techniques. Water surface area changes were determined monthly, and their relationships with meteorological parameters and climate change were investigated. Monthly water surface areas of natural lakes and dams were determined with the Normalized Difference Water Index (NDWI) applied to Sentinel-2 satellite images. Sentinel-1 Synthetic Aperture Radar (SAR) images were used in months when optical images were unavailable. The study was carried out using 3705 optical and 1167 SAR images on the Google Earth Engine (GEE) platform. Additionally, to determine which areas of water resources are shrinking, water frequency maps of the major drinking water resources were produced. Land use/land cover (LULC) changes that occurred over time were determined, and the effects of the increase in urbanization, especially on drinking water surface areas, were investigated. ESRI LULC data was used to determine LULC changes in watersheds, and the increase in urbanization areas from 2017 to 2022 ranged from 1 to 91.43%. While the basin with the least change was in Istranca, the highest increase in the artificial surface was determined to be in the Büyükçekmece basin with 1833.03 ha (2.89%). While there was a 1-12.35% decrease in the surface areas of seven water resources from 2016 to 2022, an increase of 2.65-93% was observed in three water resources (Büyükçekmece, Sazlıdere, and Elmalı), each in different categories depending on their size. In the overall analysis, total WSA decreased by 62.33 ha from 2016 to 2022, a percentage change of 0.70%. Besides the areal change analysis, the algae contents of the drinking water resources over the years were examined for the major water basins using the Normalized Difference Chlorophyll Index (NDCI) and revealed their relationship with meteorological factors and urbanization., (© 2024. Industrial waste management is critical to maintaining environmental sustainability. The dairy industry (DI), as one of the major consumers of freshwater, generates substantial whey dairy effluent, which is notably rich in organic matter and thus a significant pollutant. The effluent represents environmental risks due to its high biological and chemical oxygen demands. Today, stringent government regulations, environmental laws, and heightened consumer health awareness are compelling industries to responsibly manage and reuse whey waste. Therefore, this study investigates sustainable solutions for efficiently utilizing DI waste. Employing a systematic review approach, the research reveals that innovative technologies enable the creation of renewable, high-quality, value-added food products from dairy byproducts. These innovations offer promising sustainable waste management strategies for the dairy sector, aligning with economic interests. The main objectives of the study deal with, (a) assessing the environmental impact of dairy sector waste, (b) exploring the multifaceted nutritional and health benefits inherent in cheese whey, and (c) investigating diverse biotechnological approaches to fashion value-added, eco-friendly dairy whey-based products for potential integration into various food products, and thus fostering economic sustainability. Purpose: The upregulation of fibroblast activation protein (FAP) expression has been observed in various cancers, including metastatic breast carcinoma, prompting research into small molecule inhibitors for both diagnostic and therapeutic purposes. While the diagnostic value of PET/CT imaging using 68  Ga- or 18 F-labelled FAPi-monomers in breast cancer diagnosis is well-established, there is a significant need for therapeutic analogs. This retrospective study aimed to assess the safety and effectiveness of [ 177 Lu]Lu-DOTAGA.FAPi dimer radionuclide therapy in patients with advanced-stage breast cancer who had previously undergone [ 68  Ga]Ga-DOTA.SA.FAPi PET/CT scans to confirm the expression of FAP., Materials and Methods: Between November 2020 and March 2023, a compassionate treatment approach was utilized to administer [ 177 Lu]Lu-DOTAGA.FAPi dimer radionuclide therapy to heavily pretreated patients with advanced breast cancer. Nineteen patients (18 females, 1 male) with metastatic breast cancer participated in the study, with an average age of 44.6 ± 10.7 years. The therapy was administered at intervals of 8 to 12 weeks, and the median follow-up duration was 14 months. The primary objective of the study was to assess molecular response using [ 68  Ga]Ga-DOTA.SA.FAPi PET/CT scans, with response evaluation based on the PERCIST criteria. Secondary endpoints included overall survival (OS), progression-free survival (PFS), clinical response assessment, and safety evaluation using CTCAE v5.0 guidelines., Results: A total of 65 cycles were administered, with a mean cumulative activity of 19 ± 5.7 GBq (510 ± 154 mCi) ranging from 11 to 33.3 GBq (300 to 900 mCi) of [ 177 Lu]Lu-DOTAGA.FAPi dimer. The number of cycles ranged from 2 to 6, with a median of 3 cycles. The treatment protocol consisted of different numbers of cycles administered to the patients: specifically, two cycles were given to five patients, three cycles to nine patients, four cycles to one patient, and six cycles to four patients. Most patients had invasive/infiltrative ductal carcinoma (94.7%), while a small percentage had invasive lobular carcinoma (5.3%). All patients had bone metastases, and five of them also had liver involvement, while seven had brain metastases. Response assessment using [ 68  Ga]Ga-DOTA.SA.FAPi PET/CT scans showed that 25% of the 16 patients evaluated had partial remission, while 37.5% exhibited disease progression. According to the VAS response criteria, 26.3% achieved complete response, 15.7% had partial response, 42% showed minimal response, 11% had stable disease, and 5% had no response. The clinical disease control rate was promising, with 95% of patients achieving disease control. The clinical objective response rate was 84%. The median follow-up period was 14 months. At the time of analysis, the median overall survival was 12 months, and the median progression-free survival was 8.5 months. Notably, no severe hematological, renal, or hepatic toxicities, electrolyte imbalances, or adverse events of grade 3 or 4 were observed during the study., Conclusion: The findings suggest that [ 177 Lu]Lu-DOTAGA.FAPi dimer therapy is well-tolerated, safe, and effective for treating end-stage metastatic breast cancer patients. [ 177 Lu]Lu-DOTAGA.FAPi dimer treatment demonstrated promising efficacy in patients with advanced breast cancer, as indicated by high disease control rates, favorable response outcomes, and acceptable safety profile. Agricultural waste fiber has huge utilization potential due to its abundant sources, renewability, and low recycling costs. In this process, up to 30 wt% tobacco stalk fiber (TSF) was compounded with high-density polyethylene (HDPE) and interfacial modifiers using a grinder, melting-blending mixer, and injection molding machine to prepare environmentally friendly TSF/HDPE composites, which have excellent mechanical properties and good water resistance. On this basis, the interfacial compatibility between TSF and HDPE was enhanced by adding a silane coupling agent KH550, titanate coupling agent 201, and maleic anhydride grafted polyethylene (MA-g-PE), which led to the improvement of mechanical properties of composites. The mechanical properties of the composites with 2% MA-g-PE were the best, while the tensile strength and flexural strength increased by 72% and 38%, respectively, compared with the unmodified composites. Fourier transform infrared (FTIR) spectroscopy showed that TSF was modified by different coupling agents and new chemical bonds were formed. The scanning electron microscopy images showed that the interfacial compatibility between TSF and HDPE was improved by the interface modifiers. This work confirms that it is possible to use TSF to prepare composites and provides a new way for resource utilization of tobacco stalk fiber.
