Climate change is dramatically increasing the overall area of saline soils around the world, which is increasing by approximately two million hectares each year. Soil salinity decreases crop yields and, thereby, makes farming less profitable, potentially causing increased poverty and hunger in many areas. A solution to this problem is increasing the salt tolerance of crop plants. Transcription factors (TFs) within crop plants represent a key to understanding salt tolerance, as these proteins play important roles in the regulation of functional genes linked to salt stress. The basic leucine zipper (bZIP) TF has a well-documented role in the regulation of salt tolerance. To better understand how bZIP TFs are linked to salt tolerance, we performed a genome-wide analysis in wheat using the Chinese spring wheat genome, which has been assembled by the International Wheat Genome Sequencing Consortium. We identified 89 additional bZIP gene sequences, which brings the total of bZIP gene sequences in wheat to 237. The majority of these 237 sequences included a single bZIP protein domain; however, different combinations of five other domains also exist. The bZIP proteins are divided into ten subfamily groups. Using an in silico analysis, we identified five bZIP genes (ABF2, ABF4, ABI5, EMBP1, and VIP1) that were involved in regulating salt stress. By scrutinizing the binding properties to the 2000 bp upstream region, we identified putative functional genes under the regulation of these TFs. Expression analyses of plant tissue that had been treated with or without 100 mM NaCl revealed variable patterns between the TFs and functional genes. For example, an increased expression of ABF4 was correlated with an increased expression of the corresponding functional genes in both root and shoot tissues, whereas VIP1 downregulation in root tissues strongly decreased the expression of two functional genes. Identifying strategies to sustain the expression of the functional genes described in this study could enhance wheat’s salt tolerance.

Monitoring crop height during different growth stages provides farmers with valuable information important for managing and improving expected yields. The use of synthetic aperture radar Sentinel-1 (S-1) and Optical Sentinel-2 (S-2) satellites provides useful datasets that can assist in monitoring crop development. However, studies exploring synergetic use of SAR S-1 and optical S-2 satellite data for monitoring crop biophysical parameters are limited. We utilized a time-series of monthly S-1 satellite data independently and then used S-1 and S-2 satellite data synergistically to model wheat-crop height in this study. The polarization backscatter bands, S-1 polarization indices, and S-2 spectral indices were computed from the datasets. Optimized Random Forest Regression (RFR), Support Vector Machine Regression (SVMR), Decision Tree Regression (DTR), and Neural Network Regression (NNR) machine-learning algorithms were applied. The findings show that RFR (R2 = 0.56, RMSE = 21.01 cm) and SVM (R2 = 0.58, RMSE = 20.41 cm) produce a low modeling accuracy for crop height estimation with S-1 SAR data. The S-1 and S-2 satellite data fusion experiment had an improvement in accuracy with the RFR (R2 = 0.93 and RMSE = 8.53 cm) model outperforming the SVM (R2 = 0.91 and RMSE = 9.20 cm) and other models. Normalized polarization (Pol) and the radar vegetation index (RVI_S1) were important predictor variables for crop height retrieval compared to other variables with S-1 and S-2 data fusion as input features. The SAR ratio index (SAR RI 2) had a strong positive and significant correlation (r = 0.94; p < 0.05) with crop height amongst the predictor variables. The spatial distribution maps generated in this study show the viability of data fusion to produce accurate crop height variability maps with machine-learning algorithms. These results demonstrate that both RFR and SVM can be used to quantify crop height during the growing stages. Furthermore, findings show that data fusion improves model performance significantly. The framework from this study can be used as a tool to retrieve other wheat biophysical variables and support decision making for different crops.

The study aimed to quantify carcass fat and protein retention, and the efficiency of carcass energy utilization (Kre) resulting from feeding broiler chickens diets containing wheat, maize or mixtures of both as the major cereal ingredient. The apparent metabolizable energy (AME) of the four cereal samples was determined in adult cockerels. There was a linear (p < 0.001) increase in AME with increasing amounts of maize within the four cereal mixtures, with analyses indicating that the AME of maize was 1.4 MJ/kg greater than that of wheat. A second bioassay with growing chickens was used to determine Kre in each cereal, measured as carcass fat and protein from 7 to 21d age. Increasing proportions of maize resulted in linear increases in carcass fat and energy retained from fat (p < 0.001). However, the carcass protein and energy retained from protein did not follow the same pattern as fat (p = 0.121), but rather decreased numerically (L = 0.032). The Kre tended (p = 0.060) to increase with greater proportion of maize in a linear fashion (L = 0.009). Although AME values of cereals were confirmed to be additive, this could not be confirmed for Kre. This data can be used for optimizing energy utilization models for growing broilers.

Wheat is one of the major sources of protein worldwide. Its hexaploidy significantly complicates the identification of genes that may be crucial for improving wheat production to meet the challenges of an increased world population and climate change. Virus-induced gene silencing (VIGS) using Barley stripe mosaic virus (BSMV)-based constructs has proven to be a very useful tool in the analysis of gene function in the hexaploid plant, wheat. However, most published applications of this technique focus on phenotypes that can be observed in the leaves of wheat. A few studies have reported successful VIGS in the spikes of wheat, but this has proven to be more difficult than the seedling leaf assays. This study reports a time course analysis of the movement of BSMV from the site of inoculation into the meristematic region of wheat. It also describes how the photobleaching phenotype resulting from silencing phytoene desaturase (PDS), which is often used as a reporter for VIGS, does not indicate the full extent of where VIGS occurs, and this can mislead scientists as they design silencing studies. These findings provide guidance for more effective VIGS studies to determine the function of genes expressed in the spikes of wheat and may be important for wheat improvement.

Vascular dementia (VaD) is the second most common type of dementia after Alzheimer’s disease. In our previous studies, we showed that wheat bran extract (WBE) reduced white matter damage in a rat VaD model and improved memory in a human clinical trial. However, starch gelatinization made the large-scale preparation of WBE difficult. To simplify the manufacturing process and increase efficacy, we attempted to find a decoction containing an optimum ratio of wheat bran, sliced citrus peel, and sliced jujube (WCJ). To find an optimal ratio, the cell survival of C6 (rat glioma) cultured under hypoxic conditions (1% O2) was measured, and apoptosis was assessed. To confirm the efficacies of the optimized WCJ for VaD, pupillary light reflex, white matter damage, and the activation of astrocytes and microglia were assessed in a rat model of bilateral common carotid artery occlusion (BCCAO) causing chronic hypoperfusion. Using a combination of both searching the literature and cell survival experiments, we chose 6:2:1 as the optimal ratio of wheat bran to sliced citrus peel to sliced jujube to prepare WCJ. We showed that phytic acid contained only in wheat bran can be used as an indicator component for the quality control of WCJ. We observed in vitro that the WCJ treatment improved cell survival by reducing apoptosis through an increase in the Bcl-2/Bax ratio. In the BCCAO experiments, the WCJ-supplemented diet prevented astrocytic and microglial activation, mitigated myelin damage in the corpus callosum and optic tract, and, consequently, improved pupillary light reflex at dosages over 100 mg/kg/day. The results suggest that the consumption of WCJ can prevent VaD by reducing white matter damage, and WCJ can be developed as a safe, herbal medicine to prevent VaD.

The lateral root (LR) is an essential component of the plant root system, performing important functions for nutrient and water uptake in plants and playing a pivotal role in cereal crop productivity. Nitrate (NO3−) is an essential nutrient for plants. In this study, wheat plants were grown in 1/2 strength Hoagland’s solution containing 5 mM NO3− (check; CK), 0.1 mM NO3− (low NO3−; LN), or 0.1 mM NO3− plus 60 mg/L 2,3,5-triiodobenzoic acid (TIBA) (LNT). The results showed that LN increased the LR number significantly at 48 h after treatment compared with CK, while not increasing the root biomass, and LNT significantly decreased the LR number and root biomass. The transcriptomic analysis showed that LN induced the expression of genes related to root IAA synthesis and transport and cell wall remodeling, and it was suppressed in the LNT conditions. A physiological assay revealed that the LN conditions increased the activity of IAA biosynthesis-related enzymes, the concentrations of tryptophan and IAA, and the activity of cell wall remodeling enzymes in the roots, whereas the content of polysaccharides in the LRP cell wall was significantly decreased compared with the control. Fourier-transform infrared spectroscopy and atomic microscopy revealed that the content of cell wall polysaccharides decreased and the cell wall elasticity of LR primordia (LRP) increased under the LN conditions. The effects of LN on IAA synthesis and polar transport, cell wall remodeling, and LR development were abolished when TIBA was applied. Our findings indicate that NO3− starvation may improve auxin homeostasis and the biological properties of the LRP cell wall and thus promote LR initiation, while TIBA addition dampens the effects of LN on auxin signaling, gene expression, physiological processes, and the root architecture.

Understanding the mechanisms that regulate plant root growth under soil drying is an important challenge in root biology. We observed that moderate soil drying promotes wheat root growth. To understand whether metabolic and hormonic changes are involved in this regulation, we performed transcriptome sequencing on wheat roots under well-watered and moderate soil drying conditions. The genes upregulated in wheat roots under soil drying were mainly involved in starch and sucrose metabolism and benzoxazinoid biosynthesis. Various plant hormone-related genes were differentially expressed during soil drying. Quantification of the plant hormones under these conditions showed that the concentrations of abscisic acid (ABA), cis-zeatin (CZ), and indole-3-acetic acid (IAA) significantly increased during soil drying, whereas the concentrations of salicylic (SA), jasmonic (JA), and glycosylated salicylic (SAG) acids significantly decreased. Correlation analysis of total root length and phytohormones indicated that CZ, ABA, and IAA are positively associated with wheat root length. These results suggest that changes in metabolic pathways and plant hormones caused by moderate soil drying help wheat roots grow into deeper soil layers.

Eukaryotic gene transcription is fine-tuned by precise spatiotemporal interactions between cis-regulatory elements (CREs) and trans-acting factors. However, how CREs individually or coordinated with epigenetic marks function in regulating homoeolog bias expression is still largely unknown in wheat. In this study, through comprehensively characterizing open chromatin coupled with DNA methylation in the seedling and spikelet of common wheat, we observed that differential chromatin openness occurred between the seedling and spikelet, which plays important roles in tissue development through regulating the expression of related genes or through the transcription factor (TF)-centered regulatory network. Moreover, we found that CHH methylation may act as a key determinant affecting the differential binding of TFs, thereby resulting in differential expression of target genes. In addition, we found that sequence variations in MNase hypersensitive sites (MHSs) result in the differential expression of key genes responsible for important agronomic traits. Thus, our study provides new insights into the roles of CREs in regulating tissue or homoeolog bias expression, and controlling important agronomic traits in common wheat. It also provides potential CREs for genetic and epigenetic manipulation toward improving desirable traits for wheat molecule breeding.

A timely and comprehensive understanding of winter wheat maturity is crucial for deploying large-scale harvesters within a region, ensuring timely winter wheat harvesting, and maintaining grain quality. Winter wheat maturity prediction is limited by two key issues: accurate extraction of wheat planting areas and effective maturity prediction methods. The primary aim of this study is to propose a method for predicting winter wheat maturity. The method comprises three parts: (i) winter wheat planting area extraction via phenological characteristics across multiple growth stages; (ii) extraction of winter wheat maturity features via vegetation indices (VIs, such as NDVI, NDRE, NDII1, and NDII2) and box plot analysis; and (iii) winter wheat maturity data prediction via the selected VIs. The key findings of this work are as follows: (i) Combining multispectral remote sensing data from the winter wheat jointing-filling and maturity-harvest stages can provide high-precision extraction of winter wheat planting areas (OA = 95.67%, PA = 91.67%, UA = 99.64%, and Kappa = 0.9133). (ii) The proposed method can offer the highest accuracy in predicting maturity at the winter wheat flowering stage (R2 = 0.802, RMSE = 1.56 days), aiding in a timely and comprehensive understanding of winter wheat maturity and in deploying large-scale harvesters within the region. (iii) The study’s validation was only conducted for winter wheat maturity prediction in the North China Plain wheat production area, and the accuracy of harvesting progress information extraction for other regions’ wheat still requires further testing. The method proposed in this study can provide accurate predictions of winter wheat maturity, helping agricultural management departments adopt information-based measures to improve the efficiency of monitoring winter wheat maturation and harvesting, thus promoting the efficiency of precision agricultural operations and informatization efforts.

Quantifying how climatic change affects wheat production, and accurately predicting its potential distributions in the face of future climate, are highly important for ensuring food security in Ethiopia. This study leverages advanced machine learning algorithms including Random Forest, Maxent, Boosted Regression Tree, and Generalised Linear Model alongside an ensemble approach to accurately predict shifts in wheat habitat suitability in the Central Ethiopia Region over the upcoming decades. An extensive dataset consisting of 19 bioclimatic variables (Bio1–Bio19), elevation, solar radiation, and topographic positioning index was refined by excluding collinear predictors to increase model accuracy. The analysis revealed that the precipitation of the wettest month, minimum temperature of the coldest month, temperature seasonality, and precipitation of the coldest quarter are the most influential factors, which collectively account for a significant proportion of habitat suitability changes. The future projections revealed that up to 100% of the regions currently classified as moderately or highly suitable for wheat could become unsuitable by 2050, 2070, and 2090, illustrating a dramatic potential decline in wheat production. Generally, the future of wheat cultivation will depend heavily on developing varieties that can thrive under altered conditions; thus, immediate and informed action is needed to safeguard the food security of the region.

Wheat (Triticum aestivum, L.) producers have the choice to retain or remove residue from the cropping system following grain harvest. In the U.S. Pacific Northwest and other regions, wheat residue is often sold to increase operational profitability, especially from higher-yielding systems. But there are several benefits to retaining residue, including recycling of mineral nutrients contained therein, though this is understudied. Therefore, the primary objectives of this research were to collect and analyze a large and diverse dataset on wheat residue nutrient uptake (N, P, K, Ca, Mg, S, Fe, Zn, Mn, Cu), develop tools to estimate nutrient amounts in residue, and make economic estimates of the fertilizer replacement value of those nutrients. This was accomplished by conducting replicated variety trials on five classes of wheat across many Pacific Northwest sites over two years, then collecting and analyzing data on wheat residue biomass, residue nutrient concentrations, and grain yield. The results showed that wheat residue contained a significant amount of nutrients, but was particularly concentrated in K. Production environment had the most substantial effect on residue mineral uptake amounts, due to site differences in yield and soil nutrient availability. To enable simple estimation of residue nutrient uptake across a broad range of wheat production levels, two estimation tools are presented herein. Economic analysis showed the substantial monetary value of residual nutrients. For example, in a high-yielding wheat crop (9 Mg ha−1), the average fertilizer replacement value of just residue N, P, K, and S was similar to the entire fertilizer budget to grow the crop (~$211 vs. $205 ha−1), not considering micronutrients in the residue or any nutrients removed through grain harvest. In making residue management decisions, wheat producers should consider the tradeoff between the immediate economic gains of residue sale and the multifaceted benefits of residue retention, including savings on future nutrient costs.

Increasing wheat (Triticum aestivum L.) planting density is the most effective production management method for increasing yields; however, excessive crop populations under high planting densities may experience elevated risk of stem lodging. We conducted this study to assess the relationship between reduced lodging and increased yield, investigate the effects of planting density on wheat population structure, stem strength, and material transport, and provide a basis for rationale planting densities. The experiments were carried out using a split-plot design with three replicates. The main plots contained two wheat varieties: Bainong 5819 (BN5819) and Bainong 4199 (BN4199), and the sub-plots contained four planting density treatments: 90 × 104 plants/ha (D1), 180 × 104 plants/ha (D2), 270 × 104 plants/ha (D3), and 360 × 104 plants/ha (D4). A two-year field trial was conducted in 2021–2023. The relationships between population structure characteristics, changes in stem strength, activation, and retransport of stem material after anthesis, and achievement of high and stable yields were investigated at the different planting densities. When the planting density of wheat increased from D1 to D4 treatment, the activity of fructan hydrolase was significantly increased. Compared with D1 treatment, the highest activity of fructan hydrolase was increased by 457.47 μg/h/g under D4 treatment. At the same time, the increase of density also increased the contribution rate of dry matter accumulation (CDMA) to grain after anthesis increased, with the highest increase in CDMA at 33.67%, which significantly reduced stem strength. Correlation analysis revealed a significant negative association between CDMA and stem strength. Specifically, CDMA levels were significantly lower with the D3 treatment than the D4 treatment, while stem strength remained higher after anthesis as an adaptive response to mitigate lodging risk. Stem storage compounds can promote grain filling and a weight increase in inferior grains. The number of spikes per unit area increased significantly with increasing planting density, but the number of grains per spike and 1000-grain weight decreased significantly. In two years, the number of spikes in D3 treatment increased by a maximum of 211.67 × 104 ha−1 and 99.17 × 104 ha−1, respectively, compared to D1 and D2 treatments. The number of grains per spike was significantly higher than that of D4 treatment, the highest being 3.68 grains. Therefore, in the North China Plain with suitable water, fertilizer, and temperature, the sowing density of 270 × 104 plants/ha established population structure, significantly reduced CDMA, maintained post-anthesis stem strength, enhanced resilience of stems against post-anthesis lodging, and resulted in high yields by stabilizing the number of grains per spike and increasing the number of wheat spikes.

Drought and heat stresses represent the primary agricultural challenges in arid and semiarid regions globally. In wheat, among the most vulnerable stages to these stresses is the grain-filling process. This critical phase relies heavily on photosynthesis during the late growth stage and the mobilization of stem reserves. This study evaluated 60 spring wheat lines from the CIMMYT-Mexico Core Germplasm (CIMCOG) panel alongside four Iranian wheat cultivars under normal, drought, heat, and combined drought and heat stress conditions in two growing seasons. Several agronomic traits, including those associated with stem reserve mobilization, were assessed during the study. The combined analysis of variance revealed significant impacts of both independent and combined drought and heat stresses on the measured traits. Moreover, these stresses influenced the inter-relationships among the traits. High-yielding genotypes were identified through a combination of ranking and genotype and genotype by environment (GGE) biplot analysis. Among the top 40 genotypes, 21 were identified as environment-specific, while 19 remained common across at least two environments. Environmental dependence of grain yield responses to the sinks including stem reserve mobilization and spike reserve mobilization was found. Utilizing a machine learning algorithm, a regression tree analysis unveiled specific traits—including grain filling and canopy temperature—that contributed significantly to the high-yielding features of the identified genotypes under the various environmental conditions. These traits can serve as indirect selection criteria for enhancing yield under stressful conditions and can also be targeted for manipulation to improve wheat stress tolerance.

Wheat is the most consumed crop worldwide. Zeolite application combined with good tillage practices are good combinations that provide better soil conditions for wheat crops. Zeolite also provides a good layer for carbon to be absorbed into the soil and can retain carbon for hundreds of years. The current study aimed to investigate the effect of tillage practices and zeolite treatments on soil carbon retention and wheat crop productivity. Arranging the treatments implemented according to a factorial randomized block design which includes three replications. Tillage treatments include three levels vis: T1= 6 tillage practices with the help of cultivator (farmer practice/control), T2 (minimum tillage), and T3 (2 cultivation with cultivator + Mold-board plough). The zeolite applications consist of four levels: Z1 = 0, Z2 = 5, Z3 = 10 and Z4 = 15 t ha−1. The effect of the interaction between zeolite treatments and tillage practices on various factors related to soil and crops such as emission of carbon dioxide (CO2), dissolved organic carbon, soil organic carbon, and the productivity and components of wheat productivity. Zeolite applied at 10 t ha−1 in combination with minimum tillage gave significant differences in terms of CO2 emission, dissolved organic carbon, and on soil organic carbon. The experimental results showed that minimum CO2 emission (25.43 and 31.12 (kg CO2-C ha−1 h−1), dissolved organic carbon (4.80 and 4.90 g C kg−1), soil organic carbon (7.88 and 7.97 g C kg−1), plant height (92.14 and 92.97 cm), spike length (11.88 ad 12.11 cm), number of spikelets (20.11 and 20.98), number of tillers (278.65 and 283.93) per unit area, 1000 grain weight (50.74 and 51.54 g), biological yield (8134.87 and 8187.38 kg ha−1) and grain yield (2984.28 and 3028.96 kg ha−1) and harvest index (36.69 and 37.04%) of wheat was observed in zeolite applied at 10 t ha−1 with minimum tillage practice (T2 × Z3) compared to control and other treatments for both the years, respectively. It is therefore concluded that minimum tillage should be practiced in wheat crops with the application of zeolite at 10 t ha−1 to obtain better yield and soil carbon retention under rain-fed conditions.

Wheat gluten is responsible for the unique baking properties of wheat flour, but it also causes wheat-related disorders in predisposed individuals. Different commercially available gluten materials are commonly used for a variety of assays, but a detailed characterization of their composition is missing in many cases. This is why we aimed to provide an in-depth analysis of three commonly used gliadin and gluten materials from two different batches using gel electrophoretic and chromatographic techniques. The gliadin material did not show the typical qualitative and quantitative protein composition and does not appear to be representative of wheat gliadin. The two gluten materials had the expected protein composition, but both showed large batch-to-batch variability regarding total protein content. Since these variations result in different biochemical, immunological, and functional behaviors, it is important to analyze at least the total protein content of each material and each batch.

Wheat stem rust, caused by Puccinia graminis f. sp. tritici (Pgt), is a devastating fungal disease that affects wheat globally. The planting of resistant cultivars is the most cost-effective strategy for controlling this disease. The Huanghuai region, as a major wheat-growing area, plays a crucial role in the spread and prevalence of wheat stem rust in China. In this study, 64 wheat accessions from this region were tested at the adult stage against two major Pgt races, 34MKGQM and 21C3CTHQM. DNA markers associated with the known resistance genes Sr31, Sr24, Sr25, Sr26, and Sr38 were measured to determine their presence in the tested accessions. In the 2023 field tests, 5 (7.8%) accessions were immune to 21C3CTHQM and 34MKGQM, while 35 (54.7%) and 39 (60.9%) were moderately resistant and resistant, respectively. The remaining 20 (30.7%) accessions were moderately susceptible and susceptible. In the 2024 tests, 12 (18.8%) and 14 (21.9%) entries were immune to both races; 29 (45.3%) and 30 (46.9%) were moderately resistant and resistant, respectively. Only two cultivars, Xinong 816 and Yimai 211, were immune in both years, and three entries showed some degrees of resistance in both years. Seven cultivars, including Zhongzhimai 23, Longxing 1, Yunong 937, Huaguan 301, Wanke 800, Shaanhe 285, and Yunong 612, showed increased susceptibility. DNA markers showed that 30 entries carried Sr31, while 6 entries carried Sr38. Genes Sr24, Sr25, and Sr26, which confer good resistance to the globally prevalent cultivars TKTTF and TTTRF, were absent from the set of tested entries. While this study surveyed the resistance levels of a cross-section of wheat from the southern part of the Huanghuai region and confirmed the presence of two known resistance genes, the basis of immunity or high levels of resistance in several lines remains obscure.

Most chemical pesticides, in addition to their main functions (protection against diseases, weeds, and pests), also have a noticeable inhibitory effect on target crops. In a laboratory experiment and two-year field experiments (Russia, Trans-Urals), a study was made of the effect of the biopreparation Azolen® (Azotobacter vinelandii IB-4) on plants of the Ekada 113 wheat variety under conditions of drought and stress caused by the exposure to the herbicide Chistalan (2.4-D and dicamba). The biopreparation and the herbicide were used separately and together on wheat during the tillering phase. Treatment with the biological preparation under stressful conditions had a significant effect on the hormonal balance of plants (a decrease in the amount of abscisic acid and a normalization of the balance of indolyl-3-acetic acid and cytokinins in shoots and roots of plants was noted), while the osmoprotective, antioxidant, and photosynthetic systems of plants were activated. In drought conditions, the treatment of plants with biological preparation prevented the inhibition of root growth caused by the use of the herbicide. This, in turn, improved the absorption of water by plants and ensured an increase in wheat yield (1.6 times). The results obtained give reason to believe that microbiological preparations can be used as antidotes that weaken the phytotoxic effect of herbicidal treatments, including in drought conditions.

The wild species Thinopyrum intermedium (genome JJJSJSStSt) serves as a valuable germplasm resource providing novel diseases resistance and agronomically important genes for wheat improvement. Two wheat-Th. intermedium partial amphiploids, TAI7045 (2n = 56) and 78784 (2n = 56), exhibit high resistance to stripe rust and powdery mildew, and their chromosome constitutions have been characterized. With the aim to transfer novel resistance genes from Th. intermedium, the crosses of common wheat line MY11 with TAI7045 and 78784 were produced, and their individual F2-F5 progenies were characterized using sequential non-denaturing fluorescence in situ hybridization (ND-FISH) and molecular markers. We identified a set of wheat-Th. intermedium addition lines, involving the chromosomes 1St-JS, 2St, 2St-JS, 3St, 4J, 4St, 5St, 5J.St, 6JS.J, and 7JS. Above all, the stable wheat-Th. intermedium small segmental translocation lines with chromosomes 4DS.4DL-4StL-4DL-4JL and 4DS.4DL-4StL-4DL were selected. Combining data from specific marker amplification and resistance evaluation, we mapped the gene(s) for resistance to powdery mildew and stripe rust in the 233.56–329.88 Mb region of the long arm of the 4St chromosome from the reference Th. intermedium genome. The new wheat-Th. intermedium introgressions will be used as novel germplasm for breeding purposes.

In no-till cropping systems, banding of phosphorus (P) fertiliser at seeding results in low use efficiency due to chemical reactions in soil. Foliar P has the potential to allow grain producers to respond tactically with P application after sowing when P supply from soil and fertiliser is not meeting crop demand. The objective of this study was to evaluate the effectiveness of foliar P on wheat grain yield, grain quality, biomass yield, P uptake and P use efficiency indices. Nine field experiments were conducted to investigate the response of wheat to foliar P. Three rates of P, 0, 2.5 and 5.0 kg/ha, as phosphoric acid (H3PO4 85%) were applied to wheat at three different growth stages: first tiller emergence (Z21), first node detection (Z31) and flag leaf emergence (Z39). Grain yield responses ranging from 176 kg/ha to 505 kg/ha to foliar-applied P were observed in six out of nine experiments. The percent grain yield response to foliar P was negatively related to the P buffering index (PBI, 0–10 cm soil depth), which is attributed to greater sorption by soil of the foliar P at the higher PBI levels. Mean agronomic efficiency (AE) across the experiments was 111 kg/kg P but reached up to 232 kg/kg P. It was also evident that foliar P has the potential to improve P concentration in shoots and grains and increase P uptake but with no or minimal effect on grain quality. Our results suggest that a combination of tissue testing at the seedling stage and soil P buffering can be used to guide when foliar P application is likely to increase grain yield in wheat.

In light of projected climate changes that are expected to disrupt crop yields in previously stable regions, it is essential to understand how these changes will affect food supplies. This study investigated the combined effect of light and temperature on the yield of wheat (Triticum aestivum L.) and rice (Oryza sativa L.) in a controlled environment. Three treatment combinations were tested: 30.2 mol.m−2.d−1 with a 17.5 °C average (“low”), 38.9 mol.m−2.d−1 and 22.5 °C (“medium”), and 47.5 mol.m−2.d−1 and 27.5 °C (“high”). The low treatment resulted in the highest wheat yield, with total dry biomasses of 233 g, 140 g, and 115 g for the low, medium, and high treatments. Rice showed the highest growth in the medium and high treatments, with total dry biomasses of 145 and 144 g. The medium and high treatment biomasses were not statistically significant for wheat or rice at 95% confidence. However, the wheat’s edible biomass did significantly decrease between every treatment. These results demonstrate that wheat thrives in colder climates, even with less light available, while an increase in temperature will have either a positive or insignificant effect on rice within this study’s temperature range.

Cadmium (Cd) contamination in agricultural soils has emerged as a significant concern, particularly due to its potential impact on plant-based food. Soil pH reductions can exacerbate Cd mobility, leading to excessive accumulation in crops. While liming has been demonstrated as an effective method to mitigate Cd accumulation in rice grains in acid soils of southern China, its efficacy in remediating acid soils in northern China remains unclear. In this study, a multi-year field experiment was conducted on farmland impacted by zinc ore smelting at coordinates of 33.92° N 112.46° E to investigate the use of limestone for controlling Cd accumulation in wheat and maize grains. The results indicated that applying 7.5 t ha−1 of limestone significantly raised the soil pH from 4.5 to 6.8 as anticipated. Different rates of limestone application (2.25, 4.45, and 7.50 t ha−1) reduced Cd bioavailability in the soil by 20–54%, and Cd accumulation in wheat grains by 5–38% and maize grains by 21–63%, without yield penalty. The remediation effects were sustained for at least 27 months, highlighting limestone as a promising ameliorant for smelting-affected farmland in northern China.

Salt stress is a serious problem, because it reduces the plant growth and seed yield of wheat. To investigate the salt-tolerant mechanism of wheat caused by plant-derived smoke (PDS) solution, metabolomic and proteomic techniques were used. PDS solution, which repairs the growth inhibition of wheat under salt stress, contains metabolites related to flavonoid biosynthesis. Wheat was treated with PDS solution under salt stress and proteins were analyzed using a gel-free/label-free proteomic technique. Oppositely changed proteins were associated with protein metabolism and signal transduction in biological processes, as well as mitochondrion, endoplasmic reticulum/Golgi, and plasma membrane in cellular components with PDS solution under salt stress compared to control. Using immuno-blot analysis, proteomic results confirmed that ascorbate peroxidase increased with salt stress and decreased with additional PDS solution; however, H+-ATPase displayed opposite effects. Ubiquitin increased with salt stress and decreased with additional PDS solution; nevertheless, genomic DNA did not change. As part of mitochondrion-related events, the contents of ATP increased with salt stress and recovered with additional PDS solution. These results suggest that PDS solution enhances wheat growth suppressed by salt stress through the regulation of energy metabolism and the ubiquitin-proteasome system related to flavonoid metabolism.

To non-destructively and rapidly monitor the chlorophyll content of winter wheat leaves under CO2 microleakage stress, and to establish the quantitative relationship between chlorophyll content and sensitive bands in the winter wheat growing season from 2023 to 2024, the leakage rate was set to 1 L/min, 3 L/min, 5 L/min, and 0 L/min through field experiments. The dimensional reduction was realized, fractional differential processing of a wheat canopy spectrum was carried out, a multiple linear regression (MLR) and partial least squares regression (PLSR) estimation model was constructed using a SPA selection band, and the model’s accuracy was evaluated. The optimal model for hyperspectral estimation of wheat SPAD under CO2 microleakage stress was screened. The results show that the spectral curves of winter wheat leaves under CO2 microleakage stress showed a “red shift” of the green peak and a “blue shift” of the red edge. Compared with 1 L/min and 3 L/min, wheat leaves were more affected by CO2 at 5 L/min. Evaluation of the accuracy of the MLR and PLSR models shows that the MLR model is better, where the MLR estimation model based on 1.1, 1.8, 0.4, and 1.7 differential SPAD is the best for leakage rates of 1 L/min, 3 L/min, 5 L/min, and 0 L/min, with validation set R2 of 0.832, 0.760, 0.928, and 0.773, which are 11.528, 14.2, 17.048, and 37.3% higher than the raw spectra, respectively. This method can be used to estimate the chlorophyll content of winter wheat leaves under CO2 trace-leakage stress and to dynamically monitor CO2 trace-leakage stress in crops.

The application of phosphorus-solubilizing bacteria is an effective method for increasing the available phosphorus content and inhibiting wheat uptake of heavy metals. However, further research is needed on the mechanism by which phosphorus-solubilizing bacteria inhibit cadmium (Cd) uptake in wheat roots and its impact on the expression of root-related genes. Here, the effects of strain Klebsiella aerogenes M2 on Cd absorption in wheat and the expression of root-related Cd detoxification and immobilization genes were determined. Compared with the control, strain M2 reduced (64.1–64.6%) Cd uptake by wheat roots. Cd fluorescence staining revealed that strain M2 blocked the entry of exogenous Cd into the root interior and enhanced the immobilization of Cd by cell walls. Forty-seven genes related to Cd detoxification, including genes encoding peroxidase, chalcone synthase, and naringenin 3-dioxygenase, were upregulated in the Cd+M2 treatment. Strain M2 enhanced the Cd resistance and detoxification activity of wheat roots through the regulation of flavonoid biosynthesis and antioxidant enzyme activity. Moreover, strain M2 regulated the expression of genes related to phenylalanine metabolism and the MAPK signaling pathway to enhance Cd immobilization in roots. These results provide a theoretical basis for the use of phosphorus-solubilizing bacteria to remediate Cd-contaminated fields and reduce Cd uptake in wheat.

Waterlogging stress causes substantial destruction to plant growth and production under climatic fluctuations globally. Plants hormones have been widely explored in numerous crops, displaying an imperative role in crop defense and growth mechanism. However, there is a paucity of research on the subject of plant hormones regulating waterlogging stress responses in wheat crop. In this study, we clarified the role of 6-BA in waterlogging stress through inducing phenylpropanoid biosynthesis in wheat. The application of 6-BA (6-benzyladenine) enhanced the growth and development of wheat plants under waterlogging stress, which was accompanied by reduced electrolyte leakage, high chlorophyll, and soluble sugar content. ROS scavenging was also enhanced by 6-BA, resulting in reduced MDA and H2O2 accumulation and amplified antioxidant enzyme activities. Additionally, under the effect of 6-BA, the acceleration of lignin content and accumulation in the cell walls of wheat tissues, along with the activation of PAL (phenylalanine ammonia lyase), TAL (tyrosine ammonia lyase), and 4CL (4-hydroxycinnamate CoA ligase) activities and the increase in the level of transcription of the TaPAL and Ta4CL genes, were observed under waterlogging stress. Also, 6-BA improved the root growth system under waterlogging stress conditions. Further qPCR analysis revealed increased auxin signaling (TaPR1) in 6-BA-treated plants under waterlogging stress that was consistent with the induction of endogenous IAA hormone content under waterlogging stress conditions. Here, 6-BA also reduced yield loss, as compared to control plants. Thus, the obtained data suggested that, under the application of 6-BA, phenylpropanoid metabolism (i.e., lignin) was stimulated, playing a significant role in reducing the negative effects of waterlogging stress on yield, as evinced by the improved plant growth parameters.

To effectively reveal the disaster-causing mechanism between water stress and yield loss under different drought combinations during multiple growth periods of winter wheat, based on biennial wheat drought experiments, a crop growth analysis method was used to quantitatively identify and assess wheat yield loss sensitivity. The results showed that there was a significant negative correlation between the total dry matter relative growth rate (RGR) of wheat and the daily average degree of drought stress. The average determination coefficients of logarithmic fitting for 2017 and 2018 were 0.7935 and 0.7683, respectively. Wheat dry matter accumulation differed under the different drought combination scenarios. The yield loss sensitivity response relationship between the decrease in the RGR of wheat dry matter (relative to no drought stress) and the daily average degree of drought stress could be quantitatively identified by an S-shaped curve, and the 2017 and 2018 average coefficients of determination R2 were 0.859 and 0.849, respectively. Mild drought stress at the tillering stage stimulates adaptability and has little effect on yield. The soil water content (SWC) can be controlled to 65–75% of the field water holding capacity; the SWC at the jointing and booting stage can be controlled to be higher than the field water holding capacity of 55%. The SWC was maintained at a level higher than 75% of the field water holding capacity during the heading and flowering stages and the grain-filling and milky stages to achieve a harmonization of yields and water savings. In addition, during the production process, continuous severe drought during the jointing and booting stage and the heading and flowering stage should be avoided. This study elucidates the response relationship between drought intensity and drought-induced losses from the perspective of physical genesis, provides effective irrigation guidance for regional wheat planting, lays the foundation for the construction of quantitative agricultural drought loss risk curves, and provides technical support for predicting the trend of yield losses in wheat under different drought stresses.

Climate change poses emerging threats to wheat growth in coming future. These threats need to be explored to ensure sustainable wheat production. To do this, the SALTMED model was calibrated using data from experiments conducted on different levels of irrigation and nitrogen doses. The performance of the SALTMED model was assessed based on values of the root mean square error (RMSE), normalized root mean square error (NRMSE), coefficient of determination (R2) and coefficient of residual mass (CRM) that ranged from 0.23–1.82, 0.09–0.17, 0.91–0.93 and −0.01–0.02, respectively for calibration and 0.31–1.89, 0.11–0.31, 0.87–0.90 and −0.02–0.01, respectively for validation. Projections for future climate scenarios for wheat growth indicated that by the end of the century, sowing dates advanced by nine days under the RCP4.5 scenario and eleven days under the RCP8.5 scenario, while harvesting dates shifted earlier by twenty-four days under RCP4.5 and twenty-eight days under RCP8.5. Consequently, the overall crop duration was shortened by fifteen days under RCP4.5 and eighteen days under RCP8.5. Further simulations revealed that the wheat yield was reduced by 14.2% under RCP4.5 and 21.0% under RCP8.5; the dry matter was reduced by 14.9% under RCP4.5 and 23.3% under RCP8.5; the irrigation amount was expected to increase by 14.9% under RCP4.5 and 18.0% under RCP8.5; and water productivity was expected to be reduced by 25.3% under RCP4.5 and 33.0% under RCP8.5 until the end of century. The hypothetical scenarios showed that adding an extra 20–40% more nitrogen can enhance wheat yield and dry matter by 10.2–23.0% and 11.5–24.6%, respectively, under RCP4.5, and by 12.0–23.4% and 12.9–29.6%, respectively, under RCP8.5. This study offers valuable insights into the effects of climate change on future wheat production so that effective contingency plans could be made by policymakers and adopted by stakeholders for higher wheat productivity.

The current study aimed to quantify the effects of two drought intensities achieved by deprivation of watering for 45 and 65% of the volumetric soil moisture content (VSMC) for 14 days after wheat anthesis to identify physio-biochemical and molecular changes associated with drought tolerance in six genotypes with different drought tolerance. Drought at 65% of VSMC induced a significant decrease in the chlorophyll a content in the drought-sensitive genotype, which indicated a strong loss of photosynthetic reaction centres. Further, in the drought-tolerant genotype, the content of carotenoids, which could play a vital role in resisting water shortage stress, tended to increase. The increased production of malondialdehyde showed that the antioxidant system in the drought-sensitive genotypes was not properly activated. A significant decrease in catalase (CAT; EC 1.11.1.6) was observed at a 45% reduction in VSMC, compared to the control, in the drought-sensitive genotype, and at a reduction in VSMC of 65%, in all medium sensitive genotypes. Further, the drought-tolerant and -medium tolerant genotypes responded to drought with a decline in total glutathione concentrations with the intention to reinforce their defence system. Thereby, dehydroascorbate reductase (DHAR; EC 1.8.5.1), monodehydroascorbate reductase (MDHAR; EC 1.6.5.4), and glutathione reductase (GR; EC 1.6.4.2) were critical enzymes involved in the ascorbate–glutathione cycle together with CAT, showing their main role in the detoxification of ROS produced with the reduction in VSMC by 65%. The results of gene expression analysis showed that severe drought increased the levels of the DHN5 and WZY2 genes (that were significantly positively correlated) in the drought-tolerant genotype, whose grain weight, area, and length did not change in maturity. Also, it was seen that DHN5 expression showed a significant positive correlation with grain length and proline content at a 45% reduction in VSMC. The identification of different mechanisms under drought can contribute to the selection of drought-tolerant genotypes.

To increase Europe’s self-sufficiency for protein sources, boosting plant protein production is a prerequisite. Yield variability is one of the main problems regarding the cultivation of protein crops. In this light, cereal–legume intercropping can offer a solution, as well-balanced intercropping systems are less prone to yield variations. Therefore, in this study the effects of (i) species/genotype combination, (ii) intercropping sowing densities and (iii) fertilizer regime were evaluated under Belgian (Northwestern European) conditions over three years (i.e., the 2020–2021, 2021–2022 and 2022–2023 seasons). Regarding the species combinations, winter barley x winter pea, winter wheat x winter faba bean and winter triticale x winter faba bean, it was observed that the best-performing combination varied from year to year depending on the prevailing weather conditions. A reduced sowing density (i.e., 130 seeds/m2 for the cereal partner and 20 seeds/m2 in the case of faba bean or 40 seeds/m2 in the case of pea) was sufficient to achieve competitive yields under the prevailing conditions. Inoculation with commercial Rhizobium strains did not result in an increased yield. Fertilization with one or two nitrogen fractions significantly increased the total yield thanks to a yield increase in the cereal partner; however, as a consequence, the proportion of legumes in the mixture decreased. In conclusion, it can be stated that with the investigated cereal–legume combinations, a competitive yield and qualitative protein yield can be achieved with a reduced fertilizer input.

The use of purpose-selected wheat in brewing is limited because it is either not suitable for the application of usual agrotechnical measures or, more often, is not adapted to specific climatic conditions. As unmalted wheat in brewing can cause process problems and problems with the quality composition of mash/wort and consequently beer, by using the appropriate type and variety, but also by adjusting the malting process, these problems can be completely avoided or significantly reduced at the start. Two varieties were selected for the research, which in previous research showed good brewing/malting properties and which have significantly different starting values for the share of total protein (TP) in the grain. The goal of this research was to determine the effect of grain milling granulation on the quality of mash obtained from a hard type of wheat, i.e., the changes in the fermentable and non-fermentable extract ratio during mashing. Varieties with higher and lower initial contents of total protein (TP) in the grain were tested. It was found that regardless of the initial difference in TP concentration, the concentrations of total soluble proteins (TSP) and high molecular protein fraction (HMW N) are similar for varieties with higher and lower initial TP and that the difference in initial TP concentrations is shown as a difference in the concentration of insoluble protein fractions, which are eliminated during fermentation. By increasing the milling granulation in both kinds of wheat, there is a significant increase in the medium molecular protein fraction (MMW N), which is more pronounced in wheat with a lower starting concentration of TP, even though the starting concentrations of TSP in fine (Ø 0.2 mm) and the superfine milling (Ø < 0.2 mm) were similar. A significant drop in fermentability at the superfine granulation of the grain was noted.

Improvements in self-pollinated crops rely on crosses between different genotypes. It has been suggested that the repeated use of “the best” genotypes may lead to the restriction of the genetic diversity of the crop. In wheat, the analysis of gliadin (storage protein) polymorphism has provided evidence that genetic diversity was high and stable throughout the 20th century. Moreover, a worldwide analysis of gliadin polymorphism shows that genetic diversity is structured spatially across countries and their regions. Therefore, the analysis of gliadin genotypes in a given grain sample can provide reliable information about the origin of grains in this sample. An unexpected finding is that many registered common wheat cultivars are genetically non-uniform and composed of authentic biotypes (genotypically related lines originated from the initial cross) in spite of current crop-registration rules that include a strict demand for each new cultivar to be genetically uniform (DUS rules). In summary, the results suggest that each cultivar is the fruit of joint effects of a breeder and of a region’s environmental factors. We believe this finding will not be restricted to wheat and suggest there may be a need to re-evaluate relevant rules of cultivar registration for crop species in general.

In this paper, an extensive review of the literature is provided examining the significance of tolerance to fungal diseases in wheat amidst the escalating global demand for wheat and threats from environmental shifts and pathogen movements. The current comprehensive reliance on agrochemicals for disease management poses risks to food safety and the environment, exacerbated by the emergence of fungicide resistance. While resistance traits in wheat can offer some protection, these traits do not guarantee the complete absence of losses during periods of vigorous or moderate disease development. Furthermore, the introduction of individual resistance genes into wheat monoculture exerts selection pressure on pathogen populations. These disadvantages can be addressed or at least mitigated with the cultivation of tolerant varieties of wheat. Research in this area has shown that certain wheat varieties, susceptible to severe infectious diseases, are still capable of achieving high yields. Through the analysis of the existing literature, this paper explores the manifestations and quantification of tolerance in wheat, discussing its implications for integrated disease management and breeding strategies. Additionally, this paper addresses the ecological and evolutionary aspects of tolerance in the pathogen–plant host system, emphasizing its potential to enhance wheat productivity and sustainability.

Wheat is one of the three most important cereals in the world, along with rice and maize. It serves as the primary food and source of energy for about 30–40% of the world’s population. However, the low levels of micronutrients in wheat grains can lead to deficiencies of those micronutrients in people whose dietary habits are mostly based on cereals such as wheat. Apart from iron (Fe) and zinc (Zn), a lack of selenium (Se) is also one of the biggest problems in the world. The essentiality of Se has been confirmed for all animals and humans, and the lack of this micronutrient can cause serious health issues. Wheat dominates the world’s cereal production, so it is one of the best plants for biofortification. Due to the fact that agronomic biofortification is not an economical or environmentally acceptable approach, genetic improvement of cereals such as wheat for the enhanced content of micronutrients in the grain represents the most efficient biofortification approach.

This study systematically investigated the physiological and molecular responses of the wheat mutant ‘XC-MU201’ under high-temperature stress through comprehensive transcriptome analysis and physiological measurements. RNA sequencing of 21 samples across seven different treatment groups revealed, through Weighted Gene Co-expression Network Analysis (WGCNA), 13 modules among 9071 genes closely related to high-temperature treatments. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses showed significant enrichment of lignin biosynthesis-related modules under high-temperature conditions, especially at the H-10DAT, H-20DAT, and H-30DAT time points. Experimental results demonstrated a significant increase in lignin content in high-temperature-treated samples, confirmed by tissue staining methods, indicating wheat’s adaptation to heat damage through lignin accumulation. The phenylalanine ammonia-lyase gene (TaPAL33) was significantly upregulated under high-temperature stress, peaking at H-30DAT, suggesting its critical role in cellular defense mechanisms. Overexpression of TaPAL33 in the wheat variety ‘Xinchun 11’ enhanced lignin synthesis but inhibited growth. Subcellular localization of GFP-labeled TaPAL33 in tobacco cells showed its distribution mainly in the cytoplasm and cell membrane. Transgenic wheat exhibited higher PAL enzyme activity, enhanced antioxidant defense, and reduced oxidative damage under high-temperature stress, outperforming wild-type wheat. These results highlight TaPAL33’s key role in improving wheat heat tolerance and provide a genetic foundation for future research and applications.

Nitrogen (N), as the main component of biological macromolecules, maintains the basic process of plant growth and development. GOGAT, as a key enzyme in the N assimilation process, catalyzes α-ketoglutaric acid and glutamine to form glutamate. In this study, six GOGAT genes in wheat (Triticum aestivum L.) were identified and classified into two subfamilies, Fd-GOGAT (TaGOGAT2s) and NADH-GOGAT (TaGOGAT3s), according to the type of electron donor. Subcellular localization prediction showed that TaGOGAT3-D was localized in mitochondria and that the other five TaGOGATs were localized in chloroplasts. Via the analysis of promoter elements, many binding sites related to growth and development, hormone regulation and plant stress resistance regulations were found on the TaGOGAT promoters. The tissue-specificity expression analysis showed that TaGOGAT2s were mainly expressed in wheat leaves and flag leaves, while TaGOGAT3s were highly expressed in roots and leaves. The expression level of TaGOGATs and the enzyme activity of TaGOGAT3s in the leaves and roots of wheat seedlings were influenced by the treatment of N deficiency. This study conducted a systematic analysis of wheat GOGAT genes, providing a theoretical basis not only for the functional analysis of TaGOGATs, but also for the study of wheat nitrogen use efficiency (NUE).

The study aimed to evaluate for the first time the degree of contamination of soil and crops with major and trace elements (Cd, Co, Cr, Cu, Ni, Pb, Zn, F, Na, Mg, Si, P, Cl, Fe, Al) in agricultural lands situated in the Lower Danube Basin, Galati and Braila counties (SE Romania), impacted by the steel industry. Soil samples, as well as leaves and seeds of wheat, corn, and sunflower, were collected from two depths in 11 different sites. Along with elemental and mineralogical analyses, performed by HR-CS AAS, PIGE, SEM-EDX, and ATR-FTIR, the soil pH, texture, organic matter, electric conductivity, and CaCO3 content were investigated. The results showed that the levels of Cr (83.27–383.10 mg kg−1), Cu (17.11–68.15 mg kg−1), Ni (30.16–55.66 mg kg−1), and F (319–544 mg kg−1) in soil exceeded the Romanian regulations for sensitive use of the land. Igeo, EF, PI, and PERI pollution indices indicate that the soil is moderate to highly contaminated with Cr, Ni, and Cu, while the CSI and mERMQ indices suggested a relatively low risk for metal contamination. The elemental concentrations in plant tissues and bioaccumulation factors (BFs) provide valuable insights into the soil–plant relationship, health risks, and the selectivity of plant compartments for different elements. Thus, the results revealed that the wheat plants tended to exclude the bioaccumulation of particular elements in their tissues, while exhibiting a different bioaccumulation pattern for Zn and Cu. In the case of corn, most BFs were below one, indicating a limited phytoaccumulation capacity. However, exceptions were observed for Cd, Zn, and Cu with the sunflower BFs indicating higher bioconcentration of these elements in leaves and seeds compared to other elements. Chromium (Cr) contributes to non-carcinogenic dermal contact and ingestion hazards, children being more susceptible to the adverse effects of this contaminant.

Selenium (Se) is an essential trace element for humans. Low concentrations of Se can promote plant growth and development. Enhancing grain yield and crop Se content is significant, as major food crops generally have low Se content. Studies have shown that Se biofortification can significantly increase Se content in plant tissues. In this study, the genetic transformation of wheat was conducted to evaluate the agronomic traits of non-transgenic control and transgenic wheat before and after Se application. Se content, speciation, and transfer coefficients in wheat grains were detected. Molecular docking simulations and transcriptome data were utilized to explore the effects of selenium-binding protein-A TaSBP-A on wheat growth and grain Se accumulation and transport. The results showed that TaSBP-A gene overexpression significantly increased plant height (by 18.50%), number of spikelets (by 11.74%), and number of grains in a spike (by 35.66%) in wheat. Under normal growth conditions, Se content in transgenic wheat grains did not change significantly, but after applying sodium selenite, Se content in transgenic wheat grains significantly increased. Analysis of Se speciation revealed that organic forms of selenomethionine (SeMet) and selenocysteine (SeCys) predominated in both W48 and transgenic wheat grains. Moreover, TaSBP-A significantly increased the transfer coefficients of Se from solution to roots and from flag leaves to grains. Additionally, it was found that with the increase in TaSBP-A gene overexpression levels in transgenic wheat, the transfer coefficient of Se from flag leaves to grains also increased.

Different grain sources of whiskey have great potential for aroma expression. In this paper, four whiskeys fermented from different raw materials (barley, wheat, highland barley, and sorghum) were compared. Gas chromatography–mass spectrometry (GC-MS) and sensory evaluation were used to determine the composition of the aromatic compounds. A correlation analysis was further conducted between the aromatic compounds and sensory evaluations. Barley whiskey and wheat whiskey had more pronounced fruity, floral, and grain aromas, attributed to esters and terpenes. Barley whiskey had the most compounds (55), followed by highland barley whiskey (54). Highland barley whiskey had the greatest number of unique aroma compounds (seven). It exhibited a unique cocoa aroma related to concentrations of trans-2-nonenal, γ-nonanolactone, 1-nonanol, isoamyl lactate, 2-butanol, and 6-methyl-5-hepten-2-one. Sorghum whiskey had a specific leather and mushroom aroma attributed to 6-methyl-5-hepten-2-one, ethyl lactate, ethyl caprate, phenethyl octanoate, farnesol, α-terpineol, 3-methyl-1-pentanol, and methyleugenol. Alcohols were the main aroma components of grain whiskeys. Isoamyl alcohol (231.59~281.39 mg/L), phenylethyl alcohol (5.755~9.158 mg/L), citronellol (0.224~4.103 mg/L), β-damascenone (0.021~2.431 mg/L), geraniol (0.286~1.416 mg/L), isoamyl acetate (0.157~0.918 mg/L), phenylacetaldehyde (0.162~0.470 mg/L), linalool (0.024~0.148 mg/L), 1-octen-3-ol (0.016~0.145 mg/L), trans-2-nonenal (0.027~0.105 mg/L), and trans-2-octen-1-ol (0.011~0.054 mg/L) were all important aroma compounds in the whiskeys.

Low-cadmium (Cd) accumulating wheat cultivars (LAWC-Cds) can effectively reduce the total Cd content in wheat grains (Grain-Cd). Thirteen LAWC-Cds were planted in three fields to study the enrichment, transport, and accumulation patterns of Cd in LAWC-Cds. Compared with the soil properties before planting, the soil pH and the total Cd content in the soil decreased, while the Cd content in the diethylenetriaminepentaacetic acid extract, soil conductivity, and soil organic matter increased at wheat maturity. The Cd enrichment capacity of the different organs of wheat decreased in the following order: root > leaf > rachis > stem > glume > grain. The dynamics of Cd accumulation in roots affected Grain-Cd, and these factors were negatively correlated. The Cd content and Cd accumulation in all organs of LAWC-Cds showed strong negative correlations with the lengths of the first and second internodes and highly significant positive correlations with both grains per spike and awn length. Structural equation modeling showed that the Cd content of wheat organs had the most direct effect (0.639) in determining Grain-Cd, and soil properties had the largest effect (0.744) in influencing Grain-Cd. This study is important for screening wheat cultivars with stable low Cd-accumulation characteristics.

TFC (10% thifluzamide–fludioxonil–clothianidin) is a novel wheat seed-coating agent. In the field, we confirmed that 10% TFC plays a positive role in preventing soil-borne diseases and promoting wheat seedling growth. However, its effects on rhizosphere microecology and the underlying molecular mechanism are not fully understood. Field trials revealed a positive effect on the biomass, plant height, and root length of wheat sharp eyespots in a Yingshang field, with 95.3% control efficiency. The effects of 10% TFC on the rhizosphere soil microbiome of young wheat plants were evaluated using high throughput sequencing technology. The results demonstrated that seed-coating agents significantly changed bacterial and fungal communities, and reduced the number of bacteria but increased the number of fungi. Sequence analysis revealed that the abundance of Proteobacteria, Actinobacteria, and Patescibacteria in bacteria and Ascomycota, Mortierellomycota, and Basidiomycota in fungi were significantly enriched, which have been reported as being beneficial for plant growth and pathogen resistance. In contrast, the abundance of Mucoromycota in fungi was reduced, and most of the related genera identified were pathogenic to plants. In this study, 15-day-old wheat plant tissues treated with 10% TFC were subjected to global transcriptome analysis by RNA sequencing to provide insights into the effects of 10% TFC on seedling growth. The comparative analysis of Triticum aestivum L. libraries identified 8286 differentially expressed genes (DEGs), of which 2290 and 5996 genes were up- and downregulated in seedling growth in the presence of 10% TFC, respectively. Gene ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) functional analyses were performed for up- and downregulated DEGs separately, showing that these DEGs were enriched for terms related to the phenylpropanoid biosynthesis pathway, the protein products of which promote cell differentiation and seedling growth. This research provides comprehensive insights into its effects on wheat seedling growth and the rhizosphere microecology of seed coatings and provides important insights into their regulation and into understanding the potential benefits of seed coatings in disease management and plant growth promotion.

Fusarium head blight (FHB) can cause serious yield loss and significant mycotoxin contamination, which seriously threaten global food security and safety. Breeding stable and durable cultivars that are resistant to FHB is one of the most effective approaches to controlling this disease. Fhb1 is a well-known genetic locus for FHB resistance, but its resistance is not always effective across diverse wheat genetic backgrounds. To achieve a high and durable level of resistance, the discovery and use of additional quantitative trait loci (QTL) for FHB resistance are essentially needed in breeding programs. In this study, two independent wheat natural populations of different origins were used for mining resistance QTL with a major and stable effect. Using genome-wide association analysis (GWAS), a total of 58 marker–trait associations (MTAs) on chromosomes 1A, 2B, 3A, 3B, 4A, 4B, 4D, 5A, 5B, 5D, 6A, 6B, 6D, 7A and 7B were found to be significant for type II resistance to FHB. These 58 MTAs represent 24 putative QTL. Among these QTL, Qfhb.yzu.3B.1 and Qfhb.yzu.6B.3 were stably detected in the two natural populations across three consecutive experimental years. The favorable haplotypes at the two QTL could significantly reduce the disease severity, either individually or in combination. These two QTL are also additive to Fhb1 in cultivars with different genetic backgrounds. Breeder-friendly markers were designed to differentiate the contrasting alleles at these two loci, thus proving very useful for improving FHB resistance in wheat by marker-assisted selection.

The changes in biomass (including yield), copper (Cu) concentration, and the accumulation of wheat (Triticum aestivum. L) in response to soil Cu pollution under nighttime warming had still not been explored. Hence, this study was carried out, and these variations were analyzed from a physiological perspective. Pot trials were performed at two levels of ambient temperatures (no-warming (NT) and average nighttime warming of 0.28 °C (WT)) and two levels of soil Cu concentrations (control check without Cu application (CK) and 100 mg/kg Cu application (Cu)). Soil was collected from the carbonate cinnamon soil region of central China. The warming effects of the passive nighttime warming system were prominent, and the average increment was 0.28 °C. Antioxidant enzyme activities were promoted by warming (p < 0.05) and Cu. The highest yield was achieved in NT-Cu, mainly attributed to relatively strong root activity and photosynthesis caused by supplemental Cu, but the Cu concentration in its grains was close to the threshold (10 mg/kg) for Cu concentration in foodstuff and could present a potential food safety risk. Though nighttime warming did not increase the total biomass and yield of wheat, it decreased the Cu accumulation of wheat grown in Cu-contaminated soil, especially in grains. Moreover, WT-CK and WT-Cu increased the Cu concentration in the roots and glumes and reduced the Cu concentration in grains by 13.09% and 55.84%, respectively, probably because of a lower transpiration rate. Among them, the Cu concentration of grains in WT-Cu was the lowest and significantly lower than other applications. Our findings reveal that nighttime warming has the potential to reduce the Cu risk of grains in wheat grown in the Cu-contaminated carbonate cinnamon soil region of central China and could then provide a theoretical reference for risk assessment of food quality for wheat subjected to dual stress from nighttime warming and Cu pollution.

The erect leaf plays a crucial role in determining plant architecture, with its growth and development regulated by genetic factors. However, there has been a lack of comprehensive studies on the regulatory mechanisms governing wheat lamina joint development, thus failing to meet current breeding demands. In this study, a wheat erect leaf mutant, mths29, induced via fast neutron mutagenesis, was utilized for QTL fine mapping and investigation of lamina joint development. Genetic analysis of segregating populations derived from mths29 and Jimai22 revealed that the erect leaf trait was controlled by a dominant single gene. Using BSR sequencing and map-based cloning techniques, the QTL responsible for the erect leaf trait was mapped to a 1.03 Mb physical region on chromosome 5A. Transcriptome analysis highlighted differential expression of genes associated with cell division and proliferation, as well as several crucial transcription factors and kinases implicated in lamina joint development, particularly in the boundary cells of the preligule zone in mths29. These findings establish a solid foundation for understanding lamina joint development and hold promise for potential improvements in wheat plant architecture.

Nutraceuticals obtained from sprouted wheat and oat grains and processing by-products (bran and hull, respectively) naturally containing antioxidant and anti-inflammatory compounds were evaluated. The objective of this study was the development of a cereal-based nutraceutical formula combining extracts from sprouts and by-products and the exploration for potential synergetic effects in their bioactive properties. The antioxidant and anti-inflammatory capacities, glycemic index, phytic acid, and β-glucan of individual wheat bran hydrolysate (EH-WB), sprouted wheat (SW), oat hull hydrolysate (EH-OH), sprouted oat (SO), and combined ingredients (CI 1, CI 2, and CI3) were used to tailor an optimal nutraceutical formula. The three blend ingredients (CI 1, CI2, and CI3) were formulated at different ratios (EH-WB:SW:EH-OH:SO; 1:1:1:1, 2:1:2:1, and 1:2:1:2, w:w:w:w, respectively). The resulting mixtures showed total phenol (TPs) content ranging from 412.93 to 2556.66 µmol GAE 100 g−1 and antioxidant capacity values from 808.14 to 22,152.54 µmol TE 100 g−1 (ORAC) and 1914.05 to 7261.32 µmol TE 100 g−1 (ABTS•+), with Fe3+ reducing ability from 734. 02 to 8674.51 mmol reduced Fe 100 g−1 (FRAP) for the individual ingredients produced from EH-WB and EH-OH, where high antioxidant activity was observed. However, the anti-inflammatory results exhibited an interesting behavior, with a potentially synergistic effect of the individual ingredients. This effect was observed in CI2 and CI3, resulting in a higher ability to inhibit IL-6 and TNF-α than expected based on the anti-inflammatory values of their individual ingredients. Similar to the antioxidant properties, oat-based ingredients significantly contributed more to the anti-inflammatory properties of the overall mixture. This contribution is likely associated with the β-glucans and avenanthramides present in oats. To ensure the bioaccessibility of these ingredients, further studies including simulated digestion protocols would be necessary. The ingredient formulated with a 2:1 hydrolysate-to-sprout ratio was the most effective combination, reaching higher biological characteristics.

Many staple foods originate from durum wheat and its milling products; because of this, it is very important to know their characteristics. This study investigates elemental contents in these products and if differences exist because of organic farming. The concentrations of 28 elements in the whole seed and in milling products, that is, bran, semolina and flour, of durum wheat, were determined through ICP-OES. The wheats were grown under conventional or organic agronomic practices to verify the possibility of discriminating, using the elemental content, between products coming from one or the other practice. The elements were more abundant in the outer layer of the seed, the bran, but most of them were also present in the others. Traces of Sb were present only in 3% of the samples, while traces of Tl were detected in approximately half of the seed and bran samples but not in other samples. The absence of an element was more characteristic of specific products, e.g., most semolina and flour lacked Co, while other elements showed small differences between products from organic and conventional cultivation or between different milling products, which was the case, for example, for traces of Ag, B, and V. The concentrations of these elements were coupled with multivariate discriminant analysis, specifically PLS-DA, to identify the cultivation provenance of the milled products. A few elements, although different for each product, are sufficient to attain precision and accuracy of classification close to 1; small differences exist for different products. The worst is flour, where the predicted precision and accuracy are 0.92, although using only three elements: B, K, and Se. Semolina attains perfect prediction when also adding to the three previous elements, Ag, Cd, and Cu. Further elements are necessary for bran, while Fe and Mg replace K and Ag to classify seeds. In conclusion, five elements, B, Cd, Cu, K, and Se, are the most important in distinguishing between organic and conventional agriculture; these elements also permit some differentiation among products. The method could help in fraud prevention.

MIXTA-like transcription factors AtMYB16 and AtMYB106 play important roles in the regulation of cuticular wax accumulation in dicot model plant Arabidopsis thaliana, but there are very few studies on the MIXTA-like transcription factors in monocot plants. Herein, wheat MIXTA-like transcription factors TaMIXTA1 and TaMIXTA2 were characterized as positive regulators of cuticular wax accumulation. The virus-induced gene silencing experiments showed that knock-down of wheat TaMIXTA1 and TaMIXTA2 expressions resulted in the decreased accumulation of leaf cuticular wax, increased leaf water loss rate, and potentiated chlorophyll leaching. Furthermore, three wheat orthologous genes of ECERIFERUM 5 (TaCER5-1A, 1B, and 1D) and their function in cuticular wax deposition were reported. The silencing of TaCER5 by BSMV-VIGS led to reduced loads of leaf cuticular wax and enhanced rates of leaf water loss and chlorophyll leaching, indicating the essential role of the TaCER5 gene in the deposition of wheat cuticular wax. In addition, we demonstrated that TaMIXTA1 and TaMIXTA2 function as transcriptional activators and could directly stimulate the transcription of wax biosynthesis gene TaKCS1 and wax deposition gene TaCER5. The above results strongly support that wheat MIXTA-Like transcriptional activators TaMIXTA1 and TaMIXTA2 positively regulate cuticular wax accumulation via activating TaKCS1 and TaCER5 gene transcription.

Water deficit affects the growth as well as physiological and biochemical processes in plants. The aim of this study was to determine differences in physiological and biochemical responses to drought stress in two wheat cultivars—Chinese Spring (CS) and SQ1 (which are parents of a mapping population of doubled haploid lines)—and to relate these responses to final yield and agronomic traits. Drought stress was induced by withholding water for 14 days, after which plants were re-watered and maintained until harvest. Instantaneous gas exchange parameters were evaluated on the 3rd, 5th, 10th, and 14th days of seedling growth under drought. After 14 days, water content and levels of chlorophyll a+b, carotenoids, malondialdehyde, soluble carbohydrates, phenolics, salicylic acid, abscisic acid (ABA), and polyamines were measured. At final maturity, yield components (grain number and weight), biomass, straw weight, and harvest index were evaluated. Physiological and biochemical parameters of CS responded more than those of SQ1 to the 14-day drought, reflected in a greater reduction in final biomass and yield in CS. Marked biochemical differences between responses of CS and SQ1 to the drought were found for soluble carbohydrates and polyamines. These would be good candidates for testing in the mapping population for the coincidence of the genetic control of these traits and final biomass and yield.

Climate change is leading to an increase in the intensity, duration, and frequency of severe droughts, especially in southern and southeastern Europe, thus aggravating water scarcity problems. Water deficit stress harms the growth, physiology, and yield of crops like durum wheat. Hence, studying ancient wheat varieties’ stress responses could help identify genetic traits to enhance crop tolerance to environmental stresses. In this background, this study aimed to investigate the effects of PEG 6000-stimulated drought stress in the ancient wheat variety Saragolla and the modern one Svevo by analyzing various biochemical and molecular parameters that can especially condition the stomatal movement. Our data revealed that drought stress caused a significant increase in the levels of total soluble sugars, ABA, and IAA in both selected cultivars to a greater extent in the Saragolla than in the Svevo. We demonstrated that, under water deficit stress, calcium dynamics as well as the expression of ERF109, MAPK3/6, MYB60, and TaTPC1, involved in the activation of drought-related calcium-sensitive pathways, display significant differences between the two varieties. Therefore, our study provided further evidence regarding the ability of the ancient wheat variety Saragolla to better cope with drought stress compared to the modern variety Svevo.

Changes in the hydrologic regime impose great challenges for grain production. We investigated the impact of dry and wet extremes on the recent losses of crops in Severo-Kazakhstanskaya Oblast (SKO), where 25% of Kazakhstan’s wheat is produced. We reconstructed the Palmer Drought Severity Index (June–August PDSI) and average grain yields (with an explained variance of 48% and 44%, respectively) using five tree ring width chronologies. The extended history of the moisture variability and yields of spring wheat, oats, and barley shows the strong impact of hydrology, rather than the heat, on the grain production. We defined three distinctive hydrologic regimes in SKO: (1) 1886–1942, (2) 1943–1977, (3) 1978–2023. The early regime had fewer drought events, including some that covered a single year. Their duration increased up to 3 years in the second period. The latest regime is an extreme mode of hydrologic variability with events abruptly switching from extremely dry to extremely wet conditions (called “whiplash”). The 21st century regime signifies that the intensified and prolonged decade-long drought transitioned into pluvial condition. The new regime created sizable instability for grain producers. This crop yield reconstruction denotes the potential of the tree-ring proxy for understanding the impact of climate change on the agriculture and food security of Central Asia.

Over the past two decades, the silo bag system has gained popularity for storing grains and by-products under hermetic conditions. However, the impact of higher temperatures in the outer grain layer on key industrial parameters, such as wheat baking quality, remains insufficiently understood. Traditional silo bags are black on the inside and white on the outside to reflect sunlight, but colored bags, recently introduced to the market, absorb more heat, potentially warming the grain and causing damage. This study aimed to assess the effect of grain strata and bag color on grain temperature and quality under field conditions. Results showed a significant surface temperature increase in colored bags compared to white ones, approximately 3 °C, which affected the temperature of the peripheral grain layer. Moisture content slightly increased (0.2 percentage points) in the outer grain layer. However, many industrial quality parameters (protein content, P/L, W, and loaf volume) and the germination test for wheat, showed no significant differences between colored and white bags or between different strata after 120 days of storage, although the falling number increased and wet gluten decreased. These findings suggest that, despite surface temperature differences, the overall industrial quality of wheat remains unaffected by external bag coloration. The influence of ambient temperature on the peripheral layer was estimated to affect approximately 5–10% of the grain mass, indicating that adverse impacts on grain quality may go unnoticed without implementing stratified sampling techniques.
