11. BIOSYNTHESIS OF FATTY ACIDS BY SUBCELLULAR FRACTIONS OF NEWBORN MOUSE SKIN.

Homogenates of newborn mouse skin were centrifuged at 900 × g, 15,000 × g, and 100,000 × g. Sediment from the last two centrifugations, as well as the 100,000 × g supernatant fraction incorporated [1-14C]-acetyl CoA and [1, 3-14C]-malonyl CoA into fatty acids which were analyzed by chromatographic procedures and scintillation counting. All three fractions, especially the supernatant fraction, utilized malonyl CoA in labeling saturated acids. The particulate fractions used both malonyl CoA and acetyl CoA in labeling unsaturated acids. Several cofactor requirements were established. Coenzyme A was markedly inhibitory. When oxygen was replaced by nitrogen, incorporation of both precursors was increased by 50%. Comparison of the incorporation of [1-14C]-acetyl CoA into fatty acids of the 15,000 × g and 100,000 × g pellet fractions was studied with and without oxygen. Results showed that chain elongation predominated as the mode of labeling of unsaturated acids. This was confirmed by estimating the extent of carboxyl carbon labeling. The principal chain extensions were: 16: 1→ 18: 1 → 20: 1, 18: 2 → 20:2, and 20:4 → 22:4. Desaturase activity was not observed, even in the 100,000 × g (microsomal) pellet fraction. The distribution of radioactivity among polyunsaturated acids of whole skin in after exposure to [14C]-acetate was compared to the pattern observed with a 15,000 × g pellet fraction after incubation with [14C]-acetyl CoA. Results indicated that 18:2ω6 (linoleic acid), 20:3ω6, and other were probably labeled in whole skin by a retroconversion mechanism which was inoperative under the conditions used with the subcellular fractions. Newborn mouse skin was separated into dermis and epidermis by four procedures, each of which was found to reduce the ability of both skin components to incorporate 14C-acetate into fatty acids. For comparison, skin was labeled with acetate under the same conditions prior to separation by each method. Distribution of polyunsaturated fatty acids was studied after separation of prelabeled skin by trypsin and collagenase. In epidermis both unlabeled and labeled fatty acids were concentrated in the trypsin-released cell. Results indicated that C22 polyunsaturated fatty acids are possibly not synthesized by skin, except 22:4ω6 which is a dermal product. Collagenase-released cells contain mainly saturated acids. Linoleic acid was the only C18 diene observed. The fatty acid composition of both epidermis and dermis depended on the enzymatic method used. [ABSTRACT FROM AUTHOR]

Newborn mouse skin and human preputial skin were incubated for varying time periods in different media containing 1-C14-acetate. Fatty acids were isolated by conventional chromatographic procedures and their radioactivity monitored. Complete tissue culture medium and serum or an oxygen atmosphere enhanced acetate uptake compared to that in phosphate buffer. This increase affected saturated acids more than unsaturated. Insulin increased incorporation, especially into PUFA, but prostaglandin E2 exerted the opposite effect in some experiments. Incorporation into all acids increased with time, although there was a tendency to fall off or even plateau except in the case of polyunsaturated (PUFA) fractions. Individual PUFA, including linoleic and linolenic acids, showed a linear increase at widely different rates. The labeling rate of arachidonic acid tended to increase after about five hours. Epidermal cells were obtained from human skin epidermis by trypsinization and exposed to acetate-1-C14 while maintained in a variety of culture media. Maximum amounts of radioactivity were incorporated into all lipids when a complete medium was used. Acetate-C14 proved to be a more effective precursor than glucose-C14. Saponifiable lipids were converted to their methyl esters, which were resolved according to unsaturation, using silver TLC, and then by GLC. A portion of each ester was condensed and its radioactivity assayed by scintillation counting. Another portion was degraded to find the per cent of activity in the carboxyl carbon. Long-chain acids, with 20 carbons or more, were found to be heavily labelled, with high specific activities, with the exception of 20:4. The distribution of radioactivity did not alter greatly with time. The specific activities of long-chain acids increased several fold with time. Data on carboxyl carbon activity suggested the operation of a de novo synthesis of saturated acids to C16 and a stepwise elongation of C16 by C2 units. [ABSTRACT FROM AUTHOR]

20. Inhibition of Ribosomal Ribonucleic Acid Maturation in Novikoff Hepatoma Cells by 5-Fluorouracil and 5-Fluorouridine

- Abstract
5-Fluorouracil, at 10-4mconcentrations, inhibited ribosomal RNA maturation only slightly in Novikoff hepatoma cells growing in suspension culture. On a molar basis, 5-fluorouridine was a much more potent inhibitor of the maturation process. The difference between the effectiveness of 5-fluorouracil and 5-fluorouridine is probably a result of the greater transport and incorporation of the latter into RNA. The nucleoside analogue strongly inhibited the production of mature 18 S and 28 S ribosomal RNA, while apparently only slightly inhibiting the formation of the initial 45 S ribosomal precursor RNA or the 38 S ribosomal RNA intermediate. Inhibition of 32 S ribosomal precursor RNA production was greater than that of the larger precursor molecules, but less than that of the mature ribosomal RNA molecules. 5-Fluorouridine did not affect the methylation of 45 S ribosomal precursor RNA, indicating that the analogue exerts its inhibitory effect on some other aspect of the ribosomal RNA maturation process.

- Abstract
The chromatographic separation of cholesteryl esters and wax esters has been reexamined. A convenient and satisfactory procedure involves partial removal of wax esters with urea, followed by TLC of the residual material on alumina. Examination of the double bond isomers present in the monounsaturated fatty acids in the cholesteryl ester fraction supports an earlier conclusion that most of the cholesteryl esters are of sebaceous origin.

The patent relates to a solid fuel ramjet powered missile having the air inlets positioned adjacent the aft end of the fuel grain with a channel provided adjacent the fuel grain to provide primary air into the forward end of the fuel grain. The secondary air is supplied directly to the secondary combustor. A valve is provided in the primary air flow channel to control the air flow into the fuel grain., Supersedes AD-164 592.

The purpose of the study was to assess the potential effects of radiation from Electromagnetic Pulse (EMP) simulators located at Kirtland AFB, Albuquerque, New Mexico, on the FAA Control Tower which is co-located on the Air Base. In particular, one simulator which is located only 300 meters from the tower was of prime concern. The report covers possible coupling paths, an investigation of possible spurious outages during simulator operation. Specific measurements of the signal coupled onto signal lines were made, with the simulator being fired on countdown at FAA request. An investigation of the field inside the tower building was also made. (Author)
