Ice machines can harbor water-related organisms, and the use of ice or tap water for clinical care activities has been associated with infections in health care settings. During 2021–2022, a total of 23 cases of infection by Burkholderia multivorans (sequence type ST659) were reported at two southern California hospitals and linked to contaminated ice and water from ice machines. In addition to these 23 cases, this report also includes 23 previously unreported cases of B. multivorans ST659 infections that occurred during 2020–2024: 13 at a northern California hospital, eight at a hospital in Colorado, and two additional cases at one of the southern California hospitals. The same brand of ice machine and brands of filters, descaling, and sanitizing products were used by all four hospitals; B. multivorans was isolated from samples collected from ice machines in two of the hospitals. Whole genome sequencing indicated that all clinical and ice machine isolates were highly genetically similar (0–14 single nucleotide variant differences across 81% of the selected reference genome). Recommendations from public health officials to halt the outbreak included avoiding ice and tap water during clinical care activities. An investigation is ongoing to determine possible sources of ice machine contamination. Shortwave radiation penetrating beneath an ice-sheet surface can cause internal melting and the formation of a near-surface porous layer known as the weathering crust, a dynamic hydrological system that provides home to impurities and microbial life. We develop a mathematical model, incorporating thermodynamics and population dynamics, for the evolution of such layers. The model accounts for conservation of mass and energy, for internal and surface-absorbed radiation, and for logistic growth of a microbial species mediated by nutrients that are sourced from the melting ice. It also accounts for potential melt-albedo and microbe-albedo feedbacks, through the dependence of the absorption coefficient on the porosity or microbial concentration. We investigate one-dimensional steadily melting solutions of the model, which give rise to predictions for the weathering crust depth, water content, melt rate, and microbial abundance, depending on a number of parameters. In particular, we examine how these quantities depend on the forcing energy fluxes, finding that the relative amounts of shortwave (surface-penetrating) radiation and other heat fluxes are particularly important in determining the structure of the weathering crust. The results explain why weathering crusts form and disappear under different forcing conditions, and suggest a range of possible changes in behaviour in response to climate change. To better understand the characteristics of simulated raindrop size distributions (DSDs) and ice microphysical processes for convective systems in the East Asia monsoon region, a typical Meiyu event is simulated with the Weather Research and Forecasting (WRF) model using three two‐moment bulk microphysics schemes in this study. The simulated microphysical characteristics are then evaluated using polarimetric radar observations and retrievals. Although the observed linear storm structures are well simulated in terms of their location, there are significant deviations regarding simulated polarimetric radar variables and DSD parameters when compared to the observations. Compared to radar retrievals, all the simulated low‐level raindrops are found to have lower number concentration and larger mean sizes. To investigate the sources of the simulated DSD biases, vertical distributions of radar reflectivity and specific ice hydrometers (snow, graupel), as well as profiles of liquid/ice water contents from radar retrievals and simulations are further compared. In addition, variations about the occurrence frequency and transfer rate for four categories of ice processes (deposition, aggregation, riming, and melting) among the three schemes are analyzed. Results indicate that the overprediction of snow and graupel from riming processes is likely to be responsible for the production of extremely large raindrops at low levels. Paradana is one of the biggest ice caves in Slovenia, with an estimated ice volume of 8,000 m3. Reflecting climatological conditions, the cave ice undergoes repeated freeze-thaw cycles and regular yearly deposition of fresh ice. Three distinct ice block samples, collected from the frozen lake in May 2016, were analysed to obtain data on ice physicochemical properties and the composition of associated microbiota. Isotopic composition of the ice samples (18O, 2H) and a local meteoric water line (LMWL) constructed for monthly precipitation at Postojna were used to estimate the isotopic composition of the water that formed the ice, which had high values of deuterium excess and low concentrations of chloride, sulphate and nitrate. The values of total organic carbon (1.93–3.95 mg/l) within the ice blocks fall within the range of those measured in karst streams. Total cell count in the ice was high and the proportion of cell viability increased along the depth gradient and ranged from 4.67 × 104 to 1.52 × 105 cells/ml and from 51.0 to 85.4%, respectively. Proteobacteria represented the core of the cave-ice microbiome (55.9–79.1%), and probably play an essential role in this ecosystem. Actinobacteria was the second most abundant phylum (12.0–31.4%), followed in abundance by Bacteroidetes (2.8–4.3%). Ice phylotypes recorded amounted to 442 genera, but only 43 genera had abundances greater than 0.5%. Most abundant were Pseudomonas, a well-known ice dweller, and Lysobacter, which previously was not reported in this context. Finally, two xanthophytes, Chloridella glacialis and Ellipsoidion perminimum, known from polar environments, were cultured from the ice. Although Vietnamese residents frequently harbor extended-spectrum β-lactamase-producing Escherichia coli (ESBL-E), it is unclear which foods/beverages are risk factors for acquiring these bacteria. The aim of this study was to evaluate the frequency with which edible ice served in restaurants is contaminated with antibiotic-resistant bacteria, and thereby clarify whether this product poses a risk for ESBL-E carriage in humans. Ice from restaurants in Vietnam and Japan was screened for bacteria capable of growing on agar containing cefotaxime (BG-CTX). Of the 119 BG-CTX strains isolated in Vietnam, 40%, 39%, and 12% were identified as Pseudomonas spp., Acinetobacter spp., and Stenotrophomonas maltophilia, respectively. Meanwhile, of the six such strains isolated in Japan, five were identified as Acinetobacter spp. and one as Pseudomonas spp. More than 10% of the Acinetobacter isolates exhibited cefotaxime, ceftazidime, and sulfa/trimethoprim resistance, while 21% of Pseudomonas and 14% of S. maltophilia isolates exhibited meropenem and sulfa/ trimethoprim resistance, respectively. Subsequent multiplex polymerase chain reaction (PCR) analyses detected ESBL-encoding genes in 10% of the BG-CTX. Notably, feces harvested from mice administered water contaminated with BG-CTX contained E. coli harboring the blaCTX-M-9 gene. The ice used in the food industry has to be safe and the water used in ice production should have the quality of drinking water. The consumption of contaminated ice directly or indirectly may be a vehicle for transmission of pathogenic bacteria to humans producing outbreaks of gastrointestinal diseases. The objective of this study was to monitor the microbiological quality of ice, the water used in producing ice and the hygienic conditions of ice making machines in various food enterprises. Escherichia coli was detected in seven (6.7%) ice and 23 (21.9%) ice chest samples whereas E. coli was negative in all examined water samples. Psychrophilic bacteria were detected in 83 (79.0%) of 105 ice chest and in 68 (64.7%) of 105 ice samples, whereas Enterococci were detected only in 13 (12.4%) ice samples. Coliforms were detected in 13 (12.4%) water, 71 (67.6%) ice chest and 54 (51.4%) ice samples. In order to improve the microbiological quality of ice, the maintenance, cleaning and disinfecting of ice machines should be carried out effectively and periodically. Purpose – In oil/gas exploitations of ice-covered cold regions, conical offshore structures are designed to reduce ice force and to avoid the ice-induced intense vibrations of vertical structures. The purpose of this paper is to investigate the interaction between ice cover and conical offshore structures, the discrete element method (DEM) is introduced to determine the dynamic ice loads under different structure parameters and ice conditions. Design/methodology/approach – The ice cover is dispersed into a series of bonded spherical elements with the parallel bonding model. The interaction between ice cover and conical offshore structure is obtained based on the DEM simulation. The influence of ice velocity on ice load is compared well with the experimental data of Hamburg Ship Model Basin. Moreover, the ice load on a conical platform in the Bohai Sea is also simulated. The ice loads on its upward and downward ice-breaking cones are compared. Findings – The DEM can be used well to simulate the ice loads on conical structures. The influences of ice velocity, ice thickness, conical angle on ice loads can be analyzed with DEM simulations. Polar Oceans are natural CO2 sinks because of the enhanced solubility of CO2 in cold water. The Arctic Ocean is at additional risk of accelerated ocean acidification (OA) because of freshwater inputs from sea ice and rivers, which influence the carbonate system. Winter conditions in the Arctic are of interest because of both cold temperatures and limited CO2 venting to the atmosphere when sea ice is present. Earlier OA experiments on Arctic microbial communities conducted in the absence of ice cover, hinted at shifts in taxa dominance and diversity under lowered pH. The Catlin Arctic Survey provided an opportunity to conduct in situ, under-ice, OA experiments during late Arctic winter. Seawater was collected from under the sea ice off Ellef Ringnes Island, and communities were exposed to three CO2 levels for 6 days. Phylogenetic diversity was greater in the attached fraction compared to the free-living fraction in situ, in the controls and in the treatments. The dominant taxa in all cases were Gammaproteobacteria but acidification had little effect compared to the effects of containment. Phylogenetic net relatedness indices suggested that acidification may have decreased the diversity within some bacterial orders, but overall there was no clear trend. Within the experimental communities, alkalinity best explained the variance among samples and replicates, suggesting subtle changes in the carbonate system need to be considered in such experiments. Nitrogen inputs and microbial nitrogen cycling were investigated along a 79 km transect into the Greenland Ice Sheet (GrIS) during the main ablation season in summer 2010. The depletion of dissolved nitrate and production of ammonium (relative to icemelt) in cryoconite holes on Leverett Glacier, within 7.5 km of the ice sheet margin, suggested microbial uptake and ammonification respectively. Positive in situ acetylene assays indicated nitrogen fixation both in a debris-rich 100 m marginal zone and up to 5.7 km upslope on Leverett Glacier (with rates up to 16.3 µmoles C2H4 m-2 day-1). No positive acetylene assays were detected > 5.7 km into the ablation zone of the ice sheet. Potential nitrogen fixation only occurred when concentrations of dissolved and sediment-bound inorganic nitrogen were undetectable. Estimates of nitrogen fluxes onto the transect suggest that nitrogen fixation is likely of minor importance to the overall nitrogen budget of Leverett Glacier and of negligible importance to the nitrogen budget on the main ice sheet itself. Nitrogen fixation is however potentially important as a source of nitrogen to microbial communities in the debrisrich marginal zone close to the terminus of the glacier, where nitrogen fixation may aid the colonization of subglacial and moraine-derived debris. Abstract: Ice melt water from a 22.27 m ice core which was drilled from the East Rongbuk Glacier, Mt. Everest was incubation in two incubation ways: plate melt water directly and enrichment melt water prior plate, respectively. The abundance of cultivable bacteria ranged from 0–295 CFU mL−1 to 0–1720 CFU mL−1 in two incubations with a total of 1385 isolates obtained. Comparing to direct cultivation, enrichment cultivation recovered more bacteria. Pigment-producing bacteria accounted for an average of 84.9% of total isolates. Such high percentage suggested that pigment production may be an adaptive physiological feature for the bacteria in ice core to cope with strong ultraviolet radiation on the glacier. The abundances of cultivable bacteria and pigment-producing isolates varied synchronously along depth: higher abundance in the middle and lower at the top and bottom. It indicated that the middle part of the ice core was hospitable for the microbial survival. Based on the physiological properties of the colonies, eighty-nine isolates were selected for phylogenetic analysis. Obtained 16S rRNA gene sequences fell into four groups: Firmicutes, Alpha-Proteobacteria, Gamma-Proteobacteria, and Actinobacteria, with the Firmicutes being dominant. Microbial compositions derived from direct and enrichment cultivations were not overlapped. We suggest that it is a better way to explore the culturable microbial diversity in ice core by combining the approaches of both direct and enrichment cultivation. [Copyright &y& Elsevier]

The discovery of polygonal terrain on Mars underlain by ice heightens interest in the possibility that this water-bearing habitat may be, or may have been, a suitable habitat for extant life. The possibility is supported by the recurring detection of terrestrial microorganisms in subsurface ice environments, such as ice wedges found beneath tundra polygon features. A characterization of the microbial community of ice wedges from the high Arctic was performed to determine whether this ice environment can sustain actively respiring microorganisms and to assess the ecology of this extreme niche. We found that ice wedge samples contained a relatively abundant number of culturable cells compared to other ice habitats (∼105 CFU·mL−1). Respiration assays in which radio-labeled acetate and in situ measurement of CO2 flux were used suggested low levels of microbial activity, though more sensitive techniques are required to confirm these findings. Based on 16S rRNA gene pyrosequencing, bacterial and archaeal ice wedge communities appeared to reflect surrounding soil communities. Two Pseudomonas sp. were the most abundant taxa in the ice wedge bacterial library (∼50%), while taxa related to ammonia-oxidizing Thaumarchaeota occupied 90% of the archaeal library. The tolerance of a variety of isolates to salinity and temperature revealed characteristics of a psychrotolerant, halotolerant community. Our findings support the hypothesis that ice wedges are capable of sustaining a diverse, plausibly active microbial community. As such, ice wedges, compared to other forms of less habitable ground ice, could serve as a reservoir for life on permanently cold, water-scarce, ice-rich extraterrestrial bodies and are therefore of interest to astrobiologists and ecologists alike. Background. This study investigates the viable persistence of avian influenza viruses (AIVs) in various types of artificially frozen environmental water and evaluates the feasibility of similar occurrence taking place in nature, and allowing for prolonged abiotic virus survival, with subsequent biotic viral recirculation. Methods. Fresh, brackish, and salty water, taken in Japan from aquatic biotopes regularly visited by migratory waterfowl, were seeded with AIVs.Wemonthly monitored the viability of the seeded viruses in the frozen state at -20°C and -30°C, for 12 months. We also monitored virus viability following repeatedly induced freezing and thawing. Results. The viruses exhibited considerable viable persistence all along that period of time, as well as during freezing thawing cycles. Appreciable, yet noncrucial variances were observed in relation to some of the parameters examined. Conclusions. As typical waterborne pathogens of numerous northerly aquatic birds, AIVs are innately adapted to both the body temperature of their hosts (40°C to 42°C) and, presumably, to subzero temperatures of frozen lakes (down t-54°C in parts of Siberia) occupied and virus-seeded by subclinically infected birds, prior to freezing. Marked cryostability of AIVs appears to be evident. Summary The habitability of icy environments may be limited by low temperature, low nutrient concentrations, high solute concentrations and the physical ice matrix. The basal ice of ice sheets and glaciers contains sediments that may be a source of nutrients for microbial activity. Here we quantify microbial respiration and active cell populations of Antarctic glacial isolates Paenisporosarcina sp. B5 and Chryseobacterium sp. V3519-10 in laboratory ices with abundant nutrients at temperatures from −4°C to −33°C. At all temperatures, initial high rates of metabolism were followed by lower rates suggestive of a non-reproductive metabolic state such as maintenance or dormancy. Metabolism was sustained by viable cells as quantified via culturability, CTC reduction and LIVE/DEAD staining. Respiration rates based on active cell populations did not correspond to rates representative of reproductive growth from the literature, but suggested lower levels of metabolism. Our data demonstrated that bacteria actively respired acetate in polycrystalline ice with abundant nutrients despite low temperatures and the physical ice matrix. Our results suggest that the debris-rich basal ice that exists at temperatures just below the freezing point and underlies portions of both the Greenland and Antarctic ice sheets represents a significant potential habitat for metabolically active microbial communities. Abstract: This paper reviews the utility of analog environments in preparations for a Europa lander mission. Such analogs are useful in the demonstration and rehearsal of engineering functions such as sample acquisition from an icy surface, as well as in the exercise of the scientific protocols needed to identify organic, inorganic and possible biological impurities in ice. Summary Metabolic activity, but not growth, has been observed in ice at temperatures from −5°C to −32°C. To improve understanding of metabolism in ice, we simultaneously examined various aspects of metabolism (14C-acetate utilization, macromolecule syntheses and viability via reduction of CTC) of the glacial isolates Sporosarcina sp. B5 and Chryseobacterium sp. V3519-10 during incubation in nutrient-rich ice and brine at −5°C for 50 days. Measured rates of acetate utilization and macromolecule syntheses were high in the first 20 days suggesting adjustment to the lower temperatures and higher salt concentrations of both the liquid vein network in the ice and the brine. Following this adjustment, reproductive growth of both organisms was evident in brine, and suggested for Sporosarcina sp. B5 in ice by increases in cell numbers and biomass. Chryseobacterium sp. V3519-10 cells incubated in ice remained active. These data indicate that neither low temperature nor high salt concentrations prohibit growth in ice, but some other aspect of living within ice slows growth to within the detection limits of current methodologies. These results imply that microbial growth is plausible in natural ice systems with comparable temperatures and sufficient nutrients, such as debris-rich basal ices of glaciers and ice masses. The objective quality indices of squid (Photololigo duvaucelii) and cuttlefish (Sepia aculeata) stored in ice were compared with the subjective counterparts. Sensory (overall quality rating, quality index method [QIM], and multisample difference test), microbiological (total viable count [TVC], psychrophilic count), chemical (trichloroacetic acid-soluble peptide [TCA-soluble peptide], trimethylamine nitrogen [TMA-N], total volatile bases nitrogen [TVB-N], ammonia content, and protein pattern), and physical analyses (expressible drip, color, and texture) were determined in both species during 16 days of iced storage. As storage time increased, TCA-soluble peptide, TVB, ammonia content, and expressible drip were increased (p < 0.05). TMA content was markedly increased after 10 and 8 days of storage in squid and cuttlefish, respectively. Both TVC and psychrophilic count increased as the storage time increased (p < 0.01). Myosin heavy chain was degraded with coincidental decrease in shear force and sensory texture during storage (p < 0.05). According to the overall rating score, shelf life of both species in ice was estimated to be 6 days. The increases in ammonia content and expressible drip were highly correlated with the decrease in overall quality rating and increase in quality index score of squid and cuttlefish (p < 0.01). He presents a brief overview of some terminology regarding the origin of the word bacteria and a brief background on eubacteria and archaea. In-depth discussions on several types of eubacteria and archaea, which includes information on the environments where they can be found are presented, including mineral-loving bacteria, such as acidophiles, thermophiles, which live in the hydrothermal vents in the deep ocean, and those that live under ice sheets in the Arctic and Antarctica regions.

A study was conducted to evaluate the microbiological and chemical characteristics of ice collected from a commercial poultry further processing facility. During each of 3 visits, the following ice samples were collected: 1) freshly prepared, unused ice; 2) product-contact ice from ice-packed poultry parts; 3) product-contact ice from ice-packed poultry that had been visibly inspected and condemned as not for reuse; and 4) product-contact ice from ice-packed poultry that had passed visible inspection and had been prepared for reuse by washing (rinse with potable water and drain). The overall pattern for lowest to highest numbers of total aerobic microorganisms, coliforms, Escherichia coli, and Enterobacteriaceae was as follows: unused ice < washed ice < product-contact ice < condemned ice. Mean levels of total aerobic microorganisms, coliforms, and Eriterobacteriaceae in the unused ice were 0.3, 0.4, and 0.4 log10 cfu/mL, respectively. No E. coli was detected in the unused or washed ice, and levels were 0.5 and 1.5 log10 cfu/mL in the product-contact and condemned ice samples, respectively. Mean levels of bactena enumerated in condemned ice were 0.8, 1.0, and 0.6 log10 cfu/mL higher than the levels of bacteria found in product-contact ice for coliforms, E. coli, and Enterobacteriaceae, respectively. Washing and draining the product-contact ice decreased counts by 0.9, 0.7, 0.5, and 1.7 log10 cfu/mL for total aerobic microorganisms, coliforms, E. coli, and Enterobacteriaceae, respectively. All of the ice samples had similar pH values (pH 6.1 to 6.4). Unused and washed ice were not significantly different for total solids, total suspended solids, total Kjeldahl nitrogen, and chemical oxygen demand. Condemned ice contained the highest concentration of total solids, total suspended solids, total Kjeldahl nitrogen, and chemical oxygen demand, with levels more than 3 times that found in product contact ice. Data from the present study demonstrate that visible contamination in ice corresponds with increased microbiological and chemical contamination. AbstractLaser-induced fluorescence emission (L.I.F.E.) images were obtained in situfollowing 532 nm excitation of cryoconite assemblages in the ice covers of annual and perennially frozen Antarctic lakes during the 2008 Tawani International Expedition to Schirmacher Oasis and Lake Untersee in Dronning Maud Land, Antarctica. Laser targeting of a single millimeter-scale cryoconite results in multiple neighboring excitation events secondary to ice/air interface reflection and refraction in the bubbles surrounding the primary target. Laser excitation at 532 nm of cyanobacteria-dominated assemblages produced red and infrared autofluorescence activity attributed to the presence of phycoerythrin photosynthetic pigments. The method avoids destruction of individual target organisms and does not require the disruption of either the structure of the microbial community or the surrounding ice matrix. L.I.F.E. survey strategies described may be of interest for orbital monitoring of photosynthetic primary productivity in polar and alpine glaciers, ice sheets, snow, and lake ice of Earth's cryosphere. The findings open up the possibility of searching from either a rover or from orbit for signs of life in the polar regions of Mars and the frozen regions of exoplanets in neighboring star systems. Arguments are given that terrestrial RNA and DNA may have originated in a frozen environment more than 4 billion years ago. Scenarios are developed for atmospheric transport of microbes onto glacial ice, their adaptation to subzero temperatures in the ice, and their incorporation into one of three habitats - liquid veins, mineral grain surfaces, or isolated inside 1 of the crystals that make up polycrystalline ice. The Arrhenius dependence of microbial metabolic rate on temperature is shown to match that required to repair damage owing to spontaneous DNA depurination and amino acid racemization. Even for the oldest glacial ice, microbial lifetime is shown not to be shortened by radiation damage from 238U, 232Th, or 40K in mineral dust in ice, by phage-induced lysis, or by penetrating cosmic radiation. Instead, death of those cells adapted to the hostile conditions in glacial ice is probably due to exhaustion of available nutrients. By contrast, in permafrost microbial death is more likely due to α-particle radiation damage from U and Th in the soil and rocks intermixed with ice. For residence times in ice longer than a million years, spore formers may be unable to compete in longevity with vegetative cells that are able to repair DNA damage via survival metabolism. The martian polar caps are among the most dynamic regions on Mars, growing substantially in winter as a significant fraction of the atmosphere freezes out in the form of CO2 ice. Unusual dark spots, fans and blotches form as the south-polar seasonal CO2 ice cap retreats during spring and summer. Small radial channel networks are often associated with the location of spots once the ice disappears. The spots have been proposed to be simply bare, defrosted ground; the formation of the channels has remained uncertain. Here we report infrared and visible observations that show that the spots and fans remain at CO2 ice temperatures well into summer, and must be granular materials that have been brought up to the surface of the ice, requiring a complex suite of processes to get them there. We propose that the seasonal ice cap forms an impermeable, translucent slab of CO2 ice that sublimates from the base, building up high-pressure gas beneath the slab. This gas levitates the ice, which eventually ruptures, producing high-velocity CO2 vents that erupt sand-sized grains in jets to form the spots and erode the channels. The bacterial diversity and abundance in the snow of East Rongbuk glacier, Mt. Everest were examined through 16S rRNA gene clone library and flow cytometry approaches. In total, 35 16S rRNA gene sequences were obtained, which belong to a, β, γ-Proteobacteria, Actinobacteria, Firmicutes, CFB, Cyanobacteria, Eukaryotic chloroplast, and TM7 candidate phylum respectively. γ-Proteobacteria was the dominant bacterial group in this region, while the genera Acinetobacter and Leclercia were dominant on the genus level. The community structure varied seasonally. The bacterial abundance in summer snow was higher than that in winter. Moreover, the snow bacterial community structures in both seasons were diverse, with not only common species but season-specific species. The common species most likely originated from the Tibet Plateau. Bacteria in summer snow are affiliated with marine environment, whereas bacteria in winter snow are closely related to more diverse environments and show the feature of resistance to cold. Seasonal variations of abundance and bacterial diversity were most probably due to the seasonal characteristics of climate and atmospheric circulation in Mt. Everest. [ABSTRACT FROM AUTHOR]

Abstract: We are currently investigating factors that influence intracellular ice formation (IIF) in mouse oocytes and oocytes of the frog Xenopus. A major reason for choosing these two species is that while their eggs normally do not possess aquaporin channels in their plasma membranes, these channels can be made to express. We wish to see whether IIF is affected by the presence of these channels. The present Xenopus study deals with control eggs not expressing aquaporins. The main factor studied has been the effect of a cryoprotective agent [ethylene glycol (EG) or glycerol] and its concentration. The general procedure was to (a) cool the oocytes on a cryostage to slightly below the temperatures at which extracellular ice formation occurs, (b) warm them to just below the melting point, and (c) then re-cool them to −50°C at 10°C/min. In the majority of cases, IIF occurs well into step (c), but a sizeable minority undergo IIF in steps (a) or (b). The former group we refer to as low-temperature flashers; the latter as high-temperature flashers. IIF is manifested as abrupt blackening of the egg, which we refer to as “flashing.” Observations on the Linkam cryostage are restricted to Stage I and II oocytes, which have diameters of 200–300μm. In the absence of a cryoprotective agent, that is in frog Ringers, the mean flash temperature for the low-temperature freezers is −11.4°C, although a sizeable percentage flash at temperatures much closer to that of the EIF (−3.9°C). When EG is present, the flash temperature for the low-temperatures freezers drops significantly to ∼−20°C for EG concentrations ranging from 0.5 to 1.5M. The presence of 1.5M glycerol also substantially reduces the IIF temperature of the low-temperature freezers; namely, to −29°C, but 0.5 and 1M glycerol exert little or no effect. The IIF temperatures observed using the Linkam cryostage agree well with those estimated by calorimetry [F.W. Kleinhans, J.F. Guenther, D.M. Roberts, P. The IIF temperatures in Xenopus are substantially higher than those observed in mouse oocytes [P. Mazur, S. Seki, I.L. Pinn, F.W. Kleinhans, K. The detailed electrical conductivity measurement (ECM), trace chemical compositions and microparticles concentration analysis are performed for BH8 ice core from the depth of 126.0m to 130.0m at Vostok Station. At depth 128.7m, a volcanic signal 4726 a B.P. is detected. The volcanic sulphate flux is 95.8 kg·km-2, sulphate peak concentration 1352.8 ng·g-1, duration time about 10.1 years, comparable with some well-known volcanic events. The results indicate that it seems to be a relatively large scale, long lasting volcanic signal with farther volcanic origin. [ABSTRACT FROM AUTHOR]

Ice used for human consumption or to refrigerate foods can be contaminated with pathogenic microorganisms and may become a vehicle for human infection. To evaluate the microbiological content of commercial ice and ice used to refrigerate fish and seafood, 60 ice samples collected at six different retail points in the city of Araraquara, SP, Brazil, were studied. The following parameters were determined: total plate counts (37°C and 4°C), most probable number (MPN) for total coliforms, fecal coliforms and Escherichia coli, presence of Salmonella spp., Shigella spp.,Yersinia spp., E. coli, Vibrio cholerae and Aeromonas spp.. Results suggested poor hygienic conditions of ice production due to the presence of indicator micro-organisms. Fifty strains of E. coli of different serotypes, as well as one Y. enterocolitica biotype 1, serogroup O:5, 27 and phage type Xz (Ye 1/O5,27/Xz) and oneSalmonella Enteritidis phage type 1 (PT1) were isolated. Aeromonas spp., Shigella spp. and V. cholerae were not detected. The presence of high numbers of coliforms, heterotrophic indicator micro-organisms and pathogenic strains suggested that commercial ice and ice used to refrigerate fish and seafood may represent a potential hazard to the consumer in our community. Certain organisms nucleate the crystallization of ice. This requires a small volume of water to be induced, probably by lattice-matching with a solid template, to form an 'ice embryo' -- a region sharing at least some of the characteristics of macroscopic ice. It is of particular interest to understand the structure and function of biological structures capable of lattice-matching (or otherwise inducing a quasi-crystalline state). Some strains of the Gram-negative eubacterial genera Erwinia, Pseudomonas, and Xanthomonas, and the mycobionts of certain lichens, display ice-nucleating activity. In bacteria, the activity is conferred by a protein that contains three nested periodicities of repetition, which probably reflects a hierarchy of three motifs of structural repetition. Thus the tertiary structure of the ice-nucleation protein is likely to be regular, consistent with the expectation of its forming a template for lattice-matching. Even within a clonal culture, the nucleating sites formed by bacteria and lichens vary considerably in the threshold temperatures at which they display activity; this indicates wide variations in either the size of the template, or its structural regularity, or both. However, ice-nucleating sites of lichen and bacterial origin are clearly differentiated by their sensitivities to experimental treatments. [ABSTRACT FROM AUTHOR]

Abstract: Ice used for direct human consumption or to preserve foods and cool down drinks can be contaminated with pathogenic microorganisms and may potentially become a vehicle for consumer’s infection. To evaluate physical, chemical and microbiological quality of commercial ice and ice used for fish and seafood, 100 ice samples collected at 10 different retail points in the region of Epirus were studied. The following microbiological parameters were determined: Total coliforms, fecal coliforms, Salmonella spp., Shigella spp., Yersinia spp., Escherichia coli, Campylobacter sp., Vibrio cholerae, Aeromonas spp., Pseudomonas aeruginosa and Clostridium perfringens. E. coli was detected in 22% and coliforms were detected in 31% of samples. Samples in which coliforms were detected fail to meet the microbiological criteria specified by the drinking water legislation. Aeromonas spp., Shigella spp., Campylobacter sp. and V. cholerae were not detected. Spore forms of C. perfringens were prevalent at 35% and the psychotropic bacterium’s P. aeruginosa and Yersinia spp. were found only at three samples each. The presence of large numbers of coliforms as well as of other pathogenic strains suggested that commercial ice and ice used to make cool drinks or in preservation of fish and seafood may represent a potential hazard to the consumer. In view of the results reported herein, it is highly recommended that national regulatory guidelines should be established for the production of ice as long as regular inspections.
