Abstract

The conjugation of carbohydrate haptens to protein carriers leading to potential vaccines using traceless Staudinger ligation is reported. The use of a borane-protected phosphane, which is deprotected at the time of the ligation reaction, led to the best results in terms of stability toward oxidation and reactivity.

Results indicate that carbon dioxide reduces the expression of capsular polysaccharide type 5 at the transcriptional level in Staphylococcus aureus. Regulation of capsular polysaccharide type 8 expression is strain dependent, suggesting that genetic background determines the carbon dioxide response.

Abstract

An Ogawa-Inaba common antigenic determinant of Vibrio cholerae 01 is discussed. The determinant has been immunochemically characterized. Both the core and the O-specific polysaccharide are involved in a common anitigenic determinant. Thus the presence/absence of the Ogawa-specific antigenic determinant would lead to expression of two independent antigenic determinants of V. cholerae O1.

Abstract

The influence of an agr mutation on expression of type 5 capsular polysaccharide (5 CP) was studied in several intricate and synthetic media by Staphylococcus aureus Newman. CP synthesis is positively regulated by agr because CP expression by the agr mutant was powerfully reduced in some media but only marginally in others. Other regulatory mechanisms could function together with agr because Cp expression took place in the post-exponential growth stage in wild-type and mutant strains.

Abstract

Abstract Background Early detection of cholera outbreaks is crucial for the implementation of the most appropriate control strategies. Methods The performance of an immunochromatographic dipstick test (Institute Pasteur, Paris, France) specific for Vibrio cholerae O1 was evaluated in a prospective study in Beira, Mozambique, during the 2004 cholera season (January-May). Fecal specimens were collected from 391 patients with acute watery nonbloody diarrhea and tested by dipstick and conventional culture. Results The overall sensitivity and specificity of the rapid test compared to culture were 95% (95% confidence interval [CI]: 91%–99%) and 89% (95% CI: 86%–93%), respectively. After stratification by type of sample (rectal swab/bulk stool) and severity of diarrhea, the sensitivity ranged between 85% and 98% and specificity between 77% and 97%. Conclusion This one-step dipstick test performed well in the diagnosis of V. cholerae O1 in a setting with seasonal outbreaks where rapid tests are most urgently needed.

Abstract

Endocarditis, or inflammation of the lining membrane of the heart, is found among some patients whose blood is infected with the bacterium Staphylococcus aureus, and it is in these cases that the highest mortality from Staphylococcus aureus infection is seen. It has been difficult, however, to devise a blood test to distinguish those patients with endocarditis from those who simply have Staphylococcus aureus infection in their blood. It is known that capsular polysaccharides (CPs), which are a complex chain of sugars that form a protective layer over each bacterial cell, are present in S. aureus and that there are two major types: type 5 and type 8 CPs. Antibodies to these polysaccharide types have been discovered in the blood and are easily measured. Patients with endocarditis more often have measurable antibodies to CPs than those with uncomplicated Staphylococcus infection. In a study of the value of using the measured antibody levels to CP types 5 and 8 as an indication of the presence of endocarditis in patients infected with S. aureus, blood was collected from 33 infected patients and uninfected controls. Of the infected patients, 23 tested positive for antibodies to CP type 8, and five tested positive for antibodies to CP type 5. These results were significantly correlated with other diagnostic information confirming a diagnosis of endocarditis. It is therefore proposed that a blood test for antibodies to CPs types 8 and 5 is the first known to correctly diagnose patients with endocarditis.
