Background: This study aimed to examine the effect of 7-Ketositosterol (7-KSS), on sphingomyelin/ceramide metabolites and apoptosis in human breast MCF-7 and human liver HepG2 cancer cells. Methods: Anti-proliferative effects of 7-KSS treatment were assessed at different concentrations and periods. Cell viability was assessed through MTT analysis, whereas the levels of sphingosine-1-phosphate (S1P), sphingomyelins (SMs), and ceramides (CERs) were measured using LC-MS/MS. Phosphorylated 44/42 ERK1/2 and NF-κB p65 (Ser536) protein levels were measured by Western blot analysis and immunofluorescence staining. Apoptosis was evaluated by TUNEL staining and flow cytometric assessment of annexin-V and propidium iodide (PI) labeling. Results: Treatment with 7-KSS significantly decreased cell survival and S1P, p-44/42 ERK1/2, and p-NF-κB p65 protein levels in cancer cells compared to controls. A substantial rise was detected in intracellular amounts of C16-C24 CERs and apoptosis in cancer cells incubated with 7-KSS. Conclusions: 7-KSS stimulated ceramide accumulation and apoptosis while decreasing cell proliferation via downregulating S1P, p-44/42 ERK1/2, and p-NF-κB p65 protein levels.

The definition and nature of information have perplexed scientists due to its dual nature in measurements. The information is discrete and continuous when evaluated on a metric scale, and the Laplace-Beltrami operator and Gauss-Bonnet Theorem can map one to another. On the other hand, defining the information as a discrete entity on the surface area of an n-dimensional discrete digital manifold provides a unique way of calculating the entropy of a manifold. The software simulation shows that the surface area of the discrete n-dimensional digital manifold is an effectively computable function. Moreover, it also provides the information-geometrical evaluation of Shannon information metrics.

Technologies providing copyright-infringing IPTV content are commonly used as an illegal alternative to legal IPTV subscriptions and services, as they usually have lower monetary costs and can be more convenient for users who follow content from different sources. These infringing IPTV technologies may include websites, software, software add-ons, and physical set-top boxes. Due to the free or low cost of illegal IPTV technologies, illicit IPTV content providers will often resort to intrusive advertising, scams, and the distribution of malware to increase their revenue. We developed an automated solution for collecting and analysing malware from illegal IPTV technologies and used it to analyse a sample of illicit IPTV websites, application (app) stores, and software. Our results show that our IPTV Technologies Malware Analysis Framework (IITMAF) classified 32 of the 60 sample URLs tested as malicious compared to running the same test using publicly available online antivirus solutions, which only detected 23 of the 60 sample URLs as malicious. Moreover, the IITMAF also detected malicious URLs and files from 31 of the sample’s websites, one of which had reported ransomware behaviour.

This article provides experimental and numerical data for the flow and thermal distributions inside an air-cooled data center. The experimental data contains the exhaust temperature profile obtained from an experimental campaign and the numerical data contains OpenFOAM and script files for the simulation of the thermal structure based on the experimental study. Experimental measurements were conducted using temperature sensors located at the rear of the rack under a working scenario of 2 kW. Publically available experimental data, numerical model and results can be used for the validation of numerical models under the thermal scenario given in the present study. Flow and thermal structures inside the data center are exhibited using the validated numerical model.

Small-scale data centers suffering from low cooling efficiency consume an intense power for reliable operation of IT equipment. In this study, thermal distribution in an air-cooled data center is simulated using an open source Computational Fluid Dynamics (CFD) model to examine the underlying mechanism that reduces thermal and cooling efficiencies. The numerical model is validated with the temperature measurements conducted in the data center. Numerical simulations have revealed that recirculating hot flows observed at the top of the racks increased the air temperature beyond the allowable maximum temperature. To this end, the data center has been retrofitted by creating a hot aisle with the implementation of a moving baffle at the rear of the rack. Numerical simulations conducted for two working scenarios have demonstrated that such a minor modification could result in remarkable enhancement in the cooling efficiency. Efficiency of the data center has improved by 47.2% and 22.7% with respect to the RCI (Rack Cooling Index) and RHI (Return Heat Index), respectively. The present numerical model can capture distributions of the efficiency metrics over the racks. Present methodology can be used to reduce power consumption by the cooling and ventilation systems in existing data centers.

Composites are well-known and widely used materials due to their anisotropic nature and high strength-to-weight ratio; therefore, the mechanical performance of these materials is crucial. Precise tensile testing is essential to obtain material properties that are crucial for the design stage of composite structures. This study is an effort to investigate the effect of adhesive materials used for tabbing process, which is necessary for the tensile testing procedure. Araldite and AF 163-2k film are used as the adhesive film, whereas in the case of AF 163-2k, tabbing is done through two different procedures (Jig and corner holes method). Apart from the tensile performance, strain distribution and damage progression are monitored simultaneously using digital image correlation (DIC) and acoustic emission (AE) analysis. It is observed that there is no significant difference in the ultimate tensile strength of these composites tabbed with different adhesives and procedures. Nevertheless, the first major failure strength is much higher in Araldite tabbed specimens compared to AF 163-2k film (the first major failure activity is defined as a point at which material loses its integrity, especially when considering structural or aerospace applications). Also, strain distribution throughout the gauge length recorded via DIC is appreciably different, which is attributed to damage accumulation and progression monitored by AE analysis. The frequency-based analysis of AE data is performed to classify the damage, and cumulative energy is correlated with the DIC to navigate the failure activity at different times and stress levels.

The introduction of Data Protection by Default and Design (DPbDD) brought in as part of the General Data Protection Regulation (GDPR) in 2018, has necessitated that businesses review how best to incorporate privacy into their processes in a transparent manner, so as to build trust and improve decisions around privacy best practice. To address this issue, this paper presents a 7-stage data lifecycle, supported by nine privacy goals that together, will help practitioners manage data holdings throughout data lifecycle. The resulting data lifecycle (7-DL) was created as part of the Ideal-Cities project, a Horizon-2020 Smart-city initiative, that seeks to facilitate data re-use and/or repurposed. We evaluate 7-DL through peer review and an exemplar worked example that applies the data lifecycle to a real-time life logging fire incident scenario, one of the Ideal-Cities use cases to demonstrate the applicability of the framework.

Modeling IT equipment is of critical importance for the simulations of flow and thermal structures in air cooled data centers. Turbulent flow undergoes a significant pressure drop through the server due to the energy losses originating from the internal components. Therefore, there is an urgent need to develop a fast and an accurate method for the calculation of pressure losses inside server components for data center applications. In this study, high resolution numerical simulations were performed on an OCP (Open Compute Project) server under various inlet flow rates for inactive and active conditions. Meanwhile, one key challenge of modeling complete geometry of the server results from using an intense mesh even for a single server. To address this challenge, the server was modeled as a porous zone to mimic inertia and viscous resistance in a realistic way. Comparison of the results of porous and complete models showed that the proposed model could calculate pressure drop accurately even when the number of cells in the server was reduced to 0.3% of the complete model. Porosity coefficients were determined from the numerical simulations conducted in a broad range of air discharge for both active and inactive conditions. Errors in the calculation of pressure drop may result in a significant deviation in the prediction of the temperature rise over the server. Thus, the present model can effectively be used for the fast and accurate prediction of pressure drop inside a server component rather than solving internal flow on an intense mesh, while simulating airflow inside an air-cooled data center, which is crucial for the design safety of data centers. Finally, calculated porosity coefficients can be used for the prediction of the pressure drop in a server, while designing data centers based on numerical simulations.

Nanoporous ceramic coatings such as titania are promoted to produce drug-free cardiovascular stents with a low risk of in-stent restenosis (ISR) because of their selectivity towards vascular cell proliferation. The brittle coatings applied on stents are prone to cracking because they are subjected to plastic deformation during implantation. This study aims to overcome this problem by using a unique process without refraining from biocompatibility. Accordingly, a titanium film with 1 µm thickness was deposited on 316 LVM stainless-steel sheets using magnetron sputtering. Then, the samples were anodized to produce nanoporous oxide. The nanoporous oxide was removed by ultrasonication, leaving an approximately 500 nm metallic titanium layer with a nanopatterned surface. XPS studies revealed the presence of a 5 nm-thick TiO2 surface layer with a trace amount of fluorinated titanium on nanopatterned surfaces. Oxygen plasma treatment of the nanopatterned surface produced an additional 5 nm-thick fluoride-free oxide layer. The samples did not exhibit any cracking or spallation during plastic deformation. Cell viability studies showed that nanopatterned surfaces stimulate endothelial cell proliferation while reducing the proliferation of smooth muscle cells. Plasma treatment further accelerated the proliferation of endothelial cells. Activation of blood platelets did not occur on oxygen plasma-treated, fluoride-free nanopatterned surfaces. The presented surface treatment method can also be applied to other stent materials such as CoCr, nitinol, and orthopedic implants.

Background: We aimed to determine sphingolipid levels and examine apoptotic pathways in human retinal pigment epithelial cells (ARPE-19) undergoing endoplasmic reticulum (ER) stress. Methods: Cells were treated with tunicamycin (TM) to induce ER stress and tauroursodeoxycholic acid (TUDCA), an ER stress inhibitor, was administered to decrease cytotoxicity. Cell viability was measured by MTT assay. Levels of C16–C24 sphingomyelins (SM) and C16–C24 ceramides (CERs) were determined by LC-MS/MS. Glucose-regulated protein 78-kd (GRP78) and nuclear factor kappa-b subunit 1 (NFκB1) gene expressions were evaluated by quantitative PCR analysis, while GRP 78, NF-κB p65, cleaved caspase-3 and caspase-12 protein levels were assesed by immunofluorescence. Ceramide-1-phosphate (C1P) levels were determined by immunoassay, while caspase −3 and −12 activity in cell lysates were measured via a fluorometric method. Results: Induction of ER stress in TM treated groups were confirmed by significantly increased mRNA and protein levels of GRP78. TM significantly decreased cell viability compared to controls. Treatment with TUDCA along with TM significantly increased cell viability compared to the TM group. A significant increase was observed in C22–C24 CERs, C1P, caspase-3, caspase-12, NFκB1 mRNA and NF-κB p65 protein levels in cells treated with TM compared to controls. Administration of TUDCA lead to a partial decrease in GRP78 expression, NFκB1 mRNA, NF-κB p65 protein, C22–C24 CERs and C1P levels along with a decrease in caspase-3 and -12 activity. Conclusions: The results of this study reveal the presence of increased long chain CERs, C1P and apoptotic markers in retinal cells undergoing ER stress. In this work we present a real time SIP network simulation and monitoring system. The SIP network simulator is based on a probabilistic generative model that mimics a social network of VoIP subscribers calling each other at random times. The monitoring system, installed at a SIP server, provides services for collecting network data and server statistics in real time. A structural health monitoring (SHM) study of biaxial glass fibre-reinforced epoxy matrix composites under a constant, high strain uniaxial fatigue loading is performed using fibre Bragg grating (FBG) optical sensors embedded in composites at various locations to monitor the evolution of local strains, thereby understanding the damage mechanisms. Concurrently, the temperature changes of the samples during the fatigue test have also been monitored at the same locations. Close to fracture, significant variations in local temperatures and strains are observed, and it is shown that the variations in temperature and strain can be used to predict imminent fracture. It is noted that the latter information cannot be obtained using external strain gages, which underlines the importance of the tracking of local strains internally.

The significance of strain measurement is obvious for the analysis of Fiber-Reinforced Polymer (FRP) composites. Conventional strain measurement methods are sufficient for static testing in general. Nevertheless, if the requirements exceed the capabilities of these conventional methods, more sophisticated techniques are necessary to obtain strain data. Fiber Bragg Grating (FBG) sensors have many advantages for strain measurement over conventional ones. Thus, the present paper suggests a novel method for biaxial strain measurement using embedded FBG sensors during the fatigue testing of FRP composites. Poisson’s ratio and its reduction were monitored for each cyclic loading by using embedded FBG sensors for a given specimen and correlated with the fatigue stages determined based on the variations of the applied fatigue loading and temperature due to the autogenous heating to predict an oncoming failure of the continuous fiber-reinforced epoxy matrix composite specimens under fatigue loading. The results show that FBG sensor technology has a remarkable potential for monitoring the evolution of Poisson’s ratio on a cycle-by-cycle basis, which can reliably be used towards tracking the fatigue stages of composite for structural health monitoring purposes.

Objective: Recommendations for the treatment of cytokine release syndrome/macrophage activation syndrome (MAS) associated with coronavirus disease-2019 (COVID-19) are still of poor quality. IL-6 is an important therapeutic target as a main mediator of cytokine storm. The aim of our study was to evaluate the tocilizumab (TCZ) efficacy and factors affecting the therapy outcome., Methods: This retrospective study included 27 patients treated with TCZ for COVID-19-MAS. All patients in this study were treated with TCZ (intravenously, at a dose of 8 mg kg1 ) in addition to standard therapy. Clinical improvement (survival and decreased oxygen demand) on the 10-14th days and secondary infection rate were assessed., Results: In our 27 treated patients, 14 (51.8%) received TCZ in the intensive care unit (ICU) and seven (25.9%) were need to invasive mechanical ventilation (IMV). Fifteen (55.6%) of these patients revealed a good clinical response (four patients discharge from the ICU and 11 patients who followed-up in nonICU beds showed a decrease in oxygen demand). TCZ was significantly less effective in patients having high Murray lung injury score, low PO2/FiO2 ratio, IMV, and ICU admission (P < .05). Severity of hypoxemia was found as a single independent risk factor in the multivariable analysis (P < .05). Secondary bacterial infections rate was significantly higher in intubated patients (P < .01) or treated in the ICU (P ¼ .01)., Conclusion: TCZ was showed limited efficacy for COVID-19-related MAS. The most important predictive indicator for therapy outcome was found as the severity of hypoxemia. In addition, IMV and/or ICU was associated with the poor outcome and high side effect. So, controlled trials are still needed to confirm the indications and timing of TCZ therapy.

Objective: Silent myocardial involvement is associated with poor prognosis in patients with systemic sclerosis (SSc). Here we aimed to evaluate the subclinical left ventricular (LV) and right ventricular (RV) systolic dysfunction in patients with SSc without any cardiovascular diseases, by using both strain imaging methods, speckle tracking echocardiography (STE) and real-time 3D echocardiography (RT3DE)., Methods: A total of 47 patients with SSc and 20 age- and gender-matched healthy controls (HC) were studied. Conventional echocardiography, STE-based strain imaging, and real-time 3D echocardiography (Bothell, WA, USA) were performed to assess the biventricular deformation. Clinical and serological findings were sought., Results: Conventional echocardiographic LV measurements were similar between SSc and HC. Both the LV and RV longitudinal peak systolic strain/strain rates were significantly impaired in SSc, demonstrating subclinical LV and RV systolic dysfunction (p≤0.001). Systolic pulmonary artery pressure (SPAP) was negatively correlated with both the LV and RV longitudinal peak systolic strain/strain rates (LV, r=-0.554 and r=-0.642, respectively, p<0.001; and RV, r=-0.554 and r=-0.642, respectively, p=0.001). There was a trend for decreasing LV strain and increasing LEVSV in a 1-year analysis of patients with SSc., Conclusion: SSc is associated with myocardial systolic dysfunction. A deformation scrutiny conducted by both the STE-based strain imaging and end-systolic LV volume analysis by real-time 3D echocardiography are promising modalities that allow us for non-invasive, comprehensive investigation of subtle deterioration in the biventricular systolic function of patients with SSc.

In this multicenter, retrospective study, we evaluated the efficacy and safety of biologic therapies, including anti-TNFs, in secondary (AA) amyloidosis patients with ankylosing spondylitis (AS) and rheumatoid arthritis (RA). In addition, the frequency of secondary amyloidosis in RA and AS patients in a single center was estimated. Fifty-one AS (39M, 12F, mean age: 46.7) and 30 RA patients (11M, 19F, mean age: 51.7) with AA amyloidosis from 16 different centers in Turkey were included. Clinical and demographical features of patients were obtained from medical charts. A composite response index (CRI) to biologic therapy-based on creatinine level, proteinuria and disease activity-was used to evaluate the efficacy of treatment. The mean annual incidence of AA amyloidosis in RA and AS patients was 0.23 and 0.42/1000 patients/year, respectively. The point prevalence in RA and AS groups was 4.59 and 7.58/1000, respectively. In RA group with AA amyloidosis, effective response was obtained in 52.2 % of patients according to CRI. RA patients with RF positivity and more initial disease activity tended to have higher response rates to therapy (p values, 0.069 and 0.056). After biologic therapy (median 17 months), two RA patients died and two developed tuberculosis. In AS group, 45.7 % of patients fulfilled the criteria of good response according to CRI. AS patients with higher CRP levels at the time of AA diagnosis and at the beginning of anti-TNF therapy had higher response rates (p values, 0.011 and 0.017). During follow-up after anti-TNF therapy (median 38 months), one patient died and tuberculosis developed in two patients. Biologic therapy seems to be effective in at least half of RA and AS patients with AA amyloidosis. Tuberculosis was the most important safety concern.
