The use of high-value fatty acids (FA), omega-3 (n-3), and omega-6 (n-6) from seaweed could relieve the pressure from natural wild fish sources and reduce overfishing worldwide. This research is designed to explore how the harvest season (winter, spring, and summer) and culture period influence the biomass production yield, lipid content, and FA composition in the green seaweed Ulva sp. The studied seaweed was grown in plastic sleeves with flowthrough seawater in Mikhmoret (East Mediterranean, Israel) from July 2019 to December 2020. The Ulva species was identified as Ulva lactuca Linnaeus by DNA barcoding using rbcL, ITS, and tufA markers. No detectable genetic variability in U. lactuca samples was found throughout the research period. A quantitative examination of the lipid and FA content in U. lactuca in different harvest seasons revealed that the maximum content of lipids was in the summer, and that of polyunsaturated fatty acids (PUFAs) was in the winter. The PUFA profile included eicosapentaenoic acid, docosapentaenoic acid, docosahexaenoic acid, n-3, and n-6, where the n-6/n-3 ratio was comparable with the desired range for a balanced nutritional diet. Cold plasma (CP) is a non-thermal preservation technology that has been successfully used to decontaminate and extend the shelf life of aquatic products. However, the preservation effect of CP treatment is determined by several factors, including voltage, time, and gas compositions. Therefore, this study aimed to investigate the effects of gas composition (GasA: 10% O2, 50% N2, 40% CO2; GasB: air; GasC: 30% O2, 30% N2, 40% CO2) on the lipid oxidation of tilapia fillets treated after CP treatment. Changes in the lipid oxidation values, the percentages of fatty acids, and sensory scores were studied during 8 d of refrigerator storage. The results showed that the CP treatment significantly increased all the primary and secondary lipid oxidation values measured in this study, as well as the percentages of saturated fatty acids, but decreased the percentages of unsaturated fatty acids, especially polyunsaturated fatty acids. The lipid oxidation values were significantly increased in the GasC-CP group. After 8 d, clearly increased percentages of saturated fatty acids, a low level of major polyunsaturated fatty acids (especially linoleic (C18:2n-6)), and a decrease in the percentages of eicosapentaenoic acid (C20:5n-3) and docosahexaenoic acid (C22:6n-3) were found in GasC-CP; that is, the serious oxidation of lipids was found in the high O2 concentration group. In addition, the sensory score was also lower than that of the hypoxia CP group. Therefore, high O2 concentrations can enhance lipid oxidation and the changes in the fatty acid concentration. This review focuses on an understanding of lipid distribution and deposition dynamics in Atlantic salmon and explores the extent to which these can be predicted through multi-dimensional modeling. The methodology used to measure lipids can have an impact on their assessment and considerable work has been done to standardize and develop robust rapid assessment measures. The distribution of lipids in salmon fillets is spatially variable, with levels consistently highest in the belly region and lowest in the tail section. The level of lipids in the whole-body, fillet and Norwegian Quality Cut (NQC) of Atlantic salmon increases with fish size but plateaus as size increases. Lipid levels are predictable based on fish size data although there are various other factors that influence lipid deposition in the fillet. The relationship between these factors and the deposition of n-3 LC-PUFA is mathematically complex but does lend itself to multidimensional modeling. Effects are strongly influenced by dietary fatty acid composition and for the most part a "parity" exists between levels of certain fatty acids in the diet and what ends up in the body tissues. This parity results in a "dilution effect" that generally explains the relationship between diet and whole-body fatty acid levels. Some specific fatty acids demonstrate traits of selective retention in Atlantic salmon muscle tissue. Notably, docosahexaenoic acid is preferentially retained compared to other fatty acids. Which is explained by the understanding that not all fatty acids dilute at the same rate and biologically active fatty acids, like EPA, are essential for mediating inflammatory responses and have a critical role to play in biosynthesis. Aim. The purpose of the work was to study the nutritional value of siberian sturgeon caviar Acipenser baerii, grown in aquaculture conditions in the Dnieper reservoirs of Ukraine, and to compare its quality indicators with this product produced abroad. Methods: There are identified organoleptic properties of siberian sturgeon caviar, and its energetic value, composition of amino acids in albumens and content of fatty acids in lipids of the product. The data obtained in the research were compared with the same indices of quality of caviar of sturgeons bred abroad. Results. The organoleptic properties of siberian sturgeon caviar bred in Ukraine (appearance, color, consistency, taste and aroma) conformed to its standardized indices of quality. The caviar contained all eight essential amino acids and belonged to category of products rich by albumen (21.54±2.13%), and fats eicosapentaenoic and docosahexaenoic ω-3 acids (3.46 % and 11.2 %, respectively) (13.20±0.93%). The big content of fat, especially of polyunsaturated fatty acids and ω-3 acids (the eicosapentaenoic and docosahexaenoic ones — 3.46 % and 11.2 %, respectively) is one more factor, which enables to identify the siberian sturgeon caviar as the product of high biological value. It was shown that the caviar of siberian sturgeon produced in Ukraine is in close coincidence with those that were produced in other countries. Conclusions. The totality of studied characteristics of caviar of siberian sturgeon produced in Ukraine witnesses its high nutritional value. Purpose: Human milk (HM) composition is influenced by factors, like maternal diet and body stores, among other factors. For evaluating the influence of maternal fatty acid (FA) status on milk FA composition, the correlation between FA content in HM and in maternal plasma, erythrocytes, and adipose tissue was investigated. Methods: 223 European women who delivered at term, provided HM samples over first four months of lactation. Venous blood and adipose tissue (only from mothers who consented and underwent a C-section delivery) were sampled at delivery. FAs were assessed in plasma, erythrocytes, adipose tissue, and HM. Evolution of HM FAs over lactation and correlations between FA content in milk and tissues and between mother's blood and cord blood were established. Results: During lactation, arachidonic acid (ARA) and docosahexaenoic acid (DHA) significantly decreased, while linoleic acid (LA), alpha-linolenic acid (ALA), and eicosapentaenoic acid (EPA) remained stable. Positive correlations were observed between HM and adipose tissue for palmitic, stearic, oleic, and polyunsaturated fatty acids (PUFAs). Correlations were found between milk and plasma for oleic, LA, ARA, ALA, DHA, monounsaturated fatty acids (MUFAs), and PUFAs. No correlation was observed between erythrocytes and HM FAs. LA and ALA were more concentrated in maternal blood than in infant blood, contrary to ARA and DHA, supporting that biomagnification of LCPUFAs may have occurred during pregnancy. Conclusions: These data show that maternal adipose tissue rather than erythrocytes may serve as reservoir of PUFAs and LCPUFAs for human milk. Plasma also supplies PUFAs and LCPUFAs to maternal milk. If both, adipose tissue and plasma PUFAs, are reflection of dietary intake, it is necessary to provide PUFAs and LCPUFAs during pregnancy or even before conception and lactation to ensure availability for mothers and enough supply for the infant via HM. Lipid and fatty acid datasets are commonly used to assess the nutritional composition of organisms, trophic ecology, and ecosystem dynamics. Lipids and their fatty acid constituents are essential nutrients to all forms of life because they contribute to biological processes such as energy flow and metabolism. Assessment of total lipids in tissues of organisms provides information on energy allocation and life‐history strategies and can be an indicator of nutritional condition. The analysis of an organism's fatty acids is a widely used technique for assessing nutrient and energy transfer, and dietary interactions in food webs. Although there have been many published regional studies that assessed lipid and fatty acid compositions, many only report the mean values of the most abundant fatty acids. There are limited individual records available for wider use in intercomparison or macro‐scale studies. This dataset consists of 4856 records of individual and pooled samples of at least 470 different marine consumer species sampled from tropical, temperate, and polar regions around Australia and in the Southern, Indian, and Pacific Oceans from 1989 to 2018. This includes data for a diverse range of taxa (zooplankton, fish, cephalopods, chondrichthyans, and marine mammals), size ranges (0.02 cm to ~13 m), and that cover a broad range of trophic positions (2.0–4.6). When known, we provide a record of species name, date of sampling, sampling location, body size, relative (%) measurements of tissue‐specific total lipid content and abundant fatty acids, and absolute content (mg 100 g−1 tissue) of eicosapentaenoic acid (EPA, 20:5n3) and docosahexaenoic acid (DHA, 22:6n3) as important long‐chain (≥C20) polyunsaturated omega‐3 fatty acids. These records form a solid basis for comparative studies that will facilitate a broad understanding of the spatial and temporal distribution of marine lipids globally. The dataset also provides reference data for future dietary assessments of marine predators and model assessments of potential impacts of climate change on the availability of marine lipids and fatty acids. There are 480 data records within our data file for which the providers have requested that permission for reuse be granted, with the likely condition that they are included as a coauthor on the reporting of the dataset. Dungeness crabs (Metacarcinus magister) are ecologically and economically important in the coastal Northeast Pacific, yet relatively little is currently known about their feeding behaviour in the wild or their natural diet. Trophic biomarkers, such as fatty acids (FA), can be used to reveal trophic interactions. We used two feeding experiments to assess differences in FA composition of juvenile crabs fed different known foods to evaluate how they modify and integrate dietary FA into their own tissues and determine whether crab FA reflect diet changes over a six-week period. These experimental results were then compared with the FAsignatures of wild caught juvenile crab with undetermined diets. We found that juvenile Dungeness crabs fed different foods assimilated dietary FA into their tissues and were distinct in their FA signatures when analysed with multivariate statistics. Experimentally fed juvenile crabs contained greater proportions of the most abundant longchain polyunsaturated fatty acids (LCPUFA, >C20) than their foods. Crabs fed foods lacking in LCPUFA, particularly DHA (22:6ω3, docosahexaenoic acid), did not survive or grewslower than crabs fed other foods. This suggests that LCPUFA are physiologically important for this species and indicates biosynthesis of these FA does not occur or is not sufficient to meet their needs. [ABSTRACT FROM AUTHOR]

Omega-3 fatty acids, which consist of alpha-linolenic acid (ALA), docosahexaenoic acid (DHA), and eicosapentaenoic acid (EPA), are a type of polyunsaturated fatty acids that are crucial for enhancing human health. These three omega-3s are regarded as vital dietary nutrients because it cannot synthesize them on its own. Therefore, they must be obtained through dietary sources. On the other hands, there are concerns regarding the quality and quantity of omega-3 in dietary sources and supplements. In this study, 1H NMR spectroscopy and multivariate analysis were harnessed for non-destructive profiling of fatty acids in commercially available omega-3 supplements and plant-based oils. Results disclosed substantial disparities in omega-3 content from 8 to over 50 %, with some revealing unexpected adulteration. Notably, in one fish oil sample DHA could not be detected indicating the possibility of adulteration. Additionally, the research delineated the fatty acid composition of plant-based oils, emphasizing variations in alpha-linolenic acid (ALA) and linoleic acid (LA) content among flaxseed, chia seed, and walnut oils. Chia seeds emerged as a rich source of ALA (57–65 %mol), while walnuts contained considerable LA (44–53 % mol). Oncorhynchus mykiss , a significant aquaculture species, possesses compounds with numerous biological and pharmacological functions, including antioxidant, anticancer, anti-microbial, and anti-obesity effects. However, possible anti-inflammatory effects of lipids extracted from O. mykiss eggs on RAW264.7 cells induced by LPS have not been elucidated yet. The current study identified 13 fatty acids in lipids extracted from O. mykiss eggs that contained high amounts (51.92% of total fatty acids) of polyunsaturated fatty acids (PUFAs), especially DHA (33.66%) and EPA (7.77%). These O. mykiss lipids (100–400 μg/mL) showed significant anti-inflammatory effects by inhibiting NO and iNOS expression in LPS-stimulated RAW264.7 cells. They also inhibited expression of pro-inflammatory cytokines IL-1β , IL-6 , and TNF-α , while upregulating anti-inflammatory cytokines IL-10 , IL-11 , and TGF-β. These lipids from O. mykiss effectively inhibited LPS-induced expression CD86 as a surface biomarker on RAW264.7 cells. Additionally, O. mykiss lipids suppressed phosphorylation of p38, JNK, and ERK1/2 and the expression of phosphorylated NF-κB subunit p65. Accumulation of amyloid ß (Aß) peptides, the major component of amyloid fibrils in senile plaques, is one of the main causes of Alzheimer’s disease. Docosahexaenoic acid (DHA) is a fatty acid abundant in the brain, and is reported to have protective effects against Alzheimer’s disease, although the mechanistic effects of DHA against Alzheimer’s pathophysiology remain unclear. Because dietary supplementation of DHA in Aß precursor protein transgenic mice ameliorates Aß pathology and behavioral deficits, we hypothesize that DHA may affect the fibrillization and deposition of Aß. Here we studied the effect of different types of fatty acids on Aß fibril formation by in vitro Aß fibrillization assay. Formation of amyloid fibrils consists of two steps, i.e., the initial nucleation phase and the following elongation phase. We found that unsaturated fatty acids, especially DHA, accelerated the formation of Aß fibrils with a unique short and curved morphology in its nucleation phase, which did not elongate further into the long and straight, mature Aß fibrils. Addition of DHA afterwards did not modify the morphology of the mature Aß(1–40) fibrils. The short and curved Aß fibrils formed in the presence of DHA did not facilitate the elongation phase of Aß fibril formation, suggesting that DHA promotes the formation of “off-pathway” conformers of Aß. Background: Eicosapentaenoic acid (EPA) plus docosahexaenoic acid (DHA) recommendations are frequently stated at 500 mg/d; however, adherence to these recommendations would result in a large global commercial EPA/DHA production deficit. Previously, our laboratory demonstrated that acute DHA intake in rats can increase the capacity for synthesis-secretion of n-3 (ω-3) polyunsaturated fatty acids (PUFAs).Objective: We aimed to investigate the utility of a dietary DHA cycling strategy that employs 2 wk of repeated DHA feeding for a total of 3 cycles over 12 wk.Methods: Male Long-Evans rats were fed a 10% fat diet by weight comprised of either 1) a 2-wk, 2% α-linolenic acid (ALA, DHA-ALA group 18:3n-3) diet followed by a 2-wk, 2% DHA + 2% ALA diet over 3 consecutive 4-wk periods ("DHA cycling," DHA-ALA group); 2) a 2% DHA + 2% ALA diet (DHA group) for 12 wk; or 3) a 2% ALA-only diet (ALA group) for 12 wk. At 15 wk old, blood and tissue fatty acid concentrations and liver mRNA expression and 13C-DHA natural abundances were determined.Results: DHA concentrations in plasma, erythrocytes, and whole blood between the DHA-ALA group and the DHA groups were not different (P ≥ 0.05), but were 72-110% higher (P < 0.05) than in the ALA group. Similarly, DHA concentrations in liver, heart, adipose, and brain were not different (P ≥ 0.05) between the DHA-fed groups, but were at least 62%, 72%, 320%, and 68% higher (P < 0.05) than in the ALA group in liver, heart, adipose, and skeletal muscle, respectively. Compound-specific isotope analysis indicated that 310% more liver DHA in the DHA-ALA group compared with the DHA group is derived from dietary ALA, and this was accompanied by a 123% and 93% higher expression of elongation of very long-chain (Elovl)2 and Elovl5, respectively, in the DHA-ALA group compared with the ALA group.Conclusions: DHA cycling requires half the dietary DHA while achieving equal blood and tissue DHA concentrations in rats. Isochrysis galbana is a marine microalga which highlights by containing a great diversity of antioxidant biomolecules. It shows a high content of lipids that can be used in aquaculture and biofuel, its high polyunsaturated fatty acid profile such as DHA (docosahexaenoic acid) or for having high fucoxanthin content (carotenoid group). For a century, this microalga is very well known as feed for bivalves, larva of fish, crustaceans, and mollusks. Transesterification is the necessary reaction to be able to derivative the fatty acids in methylated fatty acids (AGM) and thus be able to identify and quantify them. It is a key step to optimize and therefore know the improvement of the obtained fatty acid profile of study biomass. Hence, this work presents the significant difference of eight methods for obtaining fatty acids from the direct and indirect transesterification reaction (MTD and MTI, respectively). Moreover, acidic and alkaline catalysts were also used (AC1, AC2 and AL1 and AL2). The results presented better contents of AGMs (methylated fatty acids) respecting dry biomass in the MTD-AL1 method with ~ 6% and a relative abundance of DHA of ~ 12 % in MTI-AL2. In addition, the most abundant fatty acid profile was MTD-AL2 with 57.66 % in polyunsaturated. On the other hand, thanks to the addition of an internal standard in the experiences, it was possible to identify that the MTD-AL1 and MTI-AC1 methods were the most efficient in the transesterification obtained with ~ 93 % and ~ 87 %, respectively. Therefore, the method that is selected for the correct reading of fatty acids present in any biomass is relevant to detect a profile with more unsaturation as has been purchased with the microalga I. galbana. Aurantiochytrium limacinum has received attention because of its abundance of polyunsaturated fatty acids (PUFAs), particularly docosahexaenoic acid (DHA). DHA is synthesized through the polyketide synthase (PKS) pathway in A. limacinum. The related enzymes of the PKS pathway are mainly expressed by three gene clusters, called pks1, pks2 and pks3. In this study, the full-length pks3 gene was obtained by polymerase chain reaction amplification and Genome Walking technology. Based on a domain analysis of the deduced amino acid sequence of the pks3 gene, 3-ketoacyl-ACP reductase (KR) and dehydratase (DH) enzyme domains were identified. Herein, A. limacinum OUC168 was engineered by gene knock-in of KR and DH using the 18S rDNA sequence as the homologous recombination site. Total fatty acid contents and the degree of unsaturation of total fatty acids increased after the kr or dh gene was knocked in. The cloning and functional study of the pks3 gene of A. limacinum establishes a foundation for revealing the DHA synthetic pathway. Gene knock-in of the enzyme domain associated with PKS synthesis has the potential to provide effective recombinant strains with higher DHA content for industrial applications. The increasing use of terrestrial plant lipids to replace of fish oil in commercial aquafeeds requires understanding synthesis and storage of long chain-polyunsaturated fatty acids (LC-PUFA) in farmed fish. Manipulation of dietary fatty acids may maximize tissue storage of LC-PUFA, through increased production and selective utilization. A data synthesis study was conducted to estimate optimal levels of fatty acids that may maximize the production and storage of LC-PUFA in the edible portion of salmonids. Data were compiled from four studies with Atlantic salmon, rainbow trout, and steelhead trout (total n = 180) which were fed diets containing different terrestrial-based oils to replace fish oil. LC-PUFA (%) were linearly correlated between diet and muscle tissue (p < 0.001; r2 > 44%), indicating proportional storage after consumption. The slope, or retention rate, was highest for docosahexaenoic acid (DHA) at 1.23, indicating that an additional 23% of DHA was stored in the muscle. Dietary saturated fatty acids were positively related to DHA stored in the muscle (p < 0.001; r2 = 22%), which may involve membrane structural requirements, as well as selective catabolism. DHA was found to be optimally stored with a dietary n-3: n-6 ratio of 1.03: 1. These new results provide a baseline of optimal dietary ratios that can be tested experimentally to determine the efficacy of balancing dietary fatty acids for maximum LC-PUFA storage. Dietary lipids strongly influence patterns of hibernation in heterotherms. Increased dietary uptake of n-6 polyunsaturated fatty acids (PUFAs), particularly of linoleic acid (LA, C18:2 n-6), enables animals to reach lower body temperatures (Tb), lengthens torpor bout duration, and results in lower energy expenditure during hibernation. Conversely, dietary n-3 PUFA impacts negatively on hibernation performance. PUFA in surrounding phospholipids (PLs) presumably modulate the temperature-dependent activity of the sarcoplasmic reticulum (SR) Ca2+ ATPase 2 (SERCA2) and thus determine the threshold Tb still allowing proper heart function during torpor. We tested the effect of diets enriched with 10% of either corn oil ("CO," high n-6 PUFA, e.g., LA) or menhaden oil ["MO," long-chain n-3 PUFA, e.g., docosahexaenoic acid (DHA)] on hibernation performance and SERCA2 activity levels during torpor in garden dormice, an insectivorous, fat-storing hibernator. Prior to hibernation, individuals fed the MO diet showed an almost nine-times higher DHA levels and 30% lower LA proportions in white adipose tissue (WAT), reflecting the fatty acid composition of SR membranes, compared to CO-diet fed animals. When fed the MO diet, dormice significantly delayed their mean onset of hibernation by almost 4 days (range: 0–12 days), compared with CO-diet fed animals. Hibernation onset correlated positively with WAT-DHA levels and negatively with WAT-LA proportions prior to hibernation. Subsequently, hibernating patterns were similar between the two dietary groups, despite a significant difference in WAT-LA but not in WAT-DHA levels in mid-hibernation. SR-PL fatty acid composition and SERCA2 activity were identical in torpid individuals from the two dietary groups in mid-hibernation. In line with our previous findings on Syrian hamsters, a granivorous, food-storing hibernator, SERCA2 activity correlated positively with LA and negatively with DHA levels of SR-PL in torpid dormice, although SERCA2 activity was about three-times higher in garden dormice than in Syrian hamsters at similar PL-DHA proportions. Similarly, minimal Tb during torpor decreased as SERCA2 activity increased. We conclude that: (1) fatty acid composition of SR membranes modulates cardiac SERCA2 activity, hence determining the minimum Tb tolerated by hibernators, and (2) high DHA levels prevent hibernators from entering into torpor, but the critical levels differ substantially between species. Unsaturated omega-3 fatty acids, especially docosahexaenoic acid (DHA), when fed to dogs improves cognitive and neurological development. Supplementation with omega-3 fatty acids such as DHA and eicosapentaenoic acid (EPA) has also been associated with lipid peroxidation, which in turn has been implicated in reduced body weight and altered bone formation. To assess the impact of omega-3 fatty acid supplementation on skeletal growth, diets containing three levels of DHA and EPA (0.01 and 0.01%, 0.14 and 0.12%, and 0.21 and 0.18%, respectively) were fed to bitches during gestation and lactation with puppies also supplemented through weaning. Thus, the subjects studied were the puppies supplemented with DHA and EPA through gestation and early postnatal life. The hip joint conformation of the puppies (n = 676) was recorded at adulthood using two radiographic, non-invasive evaluations. In this population, females had higher hip distraction indices (DI) than males. Males from the lower two levels of DHA and EPA supplementation had significantly smaller hip DI than all females and males from the highest DHA and EPA supplementation. In contrast, there were no diet effects on anatomical indicators of hip joint conformation and no visible arthritic changes. The capelin (Mallotus villosus) is a significant fish species with high nutritional value. This study comprehensively investigated the fingerprinting of the lipidome of its head, meat, roe, and viscera, based on an untargeted lipidomic strategy. Results showed that unsaturated fatty acids accounted for more than 55 % of the total fatty acids in the capelin, especially docosahexaenoic acid (DHA, 7.65 −19.83 % of total fatty acids) and eicosapentaenoic acid (EPA, 7.79 −16.91 %). Additionally, 1206 lipid molecules involving 30 major classes were detected. Among these molecules, phosphatidylcholine (PC) (12:1/26:1), PC (5:0/22:5), PC (13:0/24:4), PC (16:0/16:0), PC (19:0/20:3), and phosphatidylethanolamine (PE) (39:0/15:1) were the characteristic lipids in roe, then ceramides (Cer) (t19:1/24:4), Cer (t20:0/18:2), Cer (t22:1/18:2), Cer (t19:1/18:2), Cer (t16:1/26:1), and Cer (d16:0/24:4) were the characteristic lipid in viscera. Overall, this study provided the first interpretation of the lipidome of capelin, which could serve as a reference for future studies on nutritional value, ecology, and traceability of geographic origin. Recent studies have found that feeding high levels of long chain polyunsaturated fatty acids (LC-PUFA) influenced the steroid biosynthesis pathway of Atlantic salmon on a transcriptomic level, although research observed on a metabolic level is lacking. Dietary lipid and chronic stress have also been suggested to play a role in steroidogenesis. A study was performed on Atlantic salmon post-smolts (184 g) fed with diets of high and low levels of lipid with and without high levels of LC-PUFA under hypoxic (stressed) or normoxic (unstressed) conditions. Liver, head kidney and blood samples were collected after 35 and 116 days to determine short- and long-term effects. On day 35, dissolved oxygen affected the levels of the steroid hormones 11-deoxycortisol, cortisone and testosterone as well as the relative expression of cyp11a involved in steroidogenesis and ud2a2 involved in sex steroid metabolism. On day 116, an interaction between lipid x LC-PUFA x oxygen was found for cyp11a , hsd3b (steroidogenesis) and ud2a2 genes, whereas LC-PUFA and/or oxygen affected the steroid levels of 11-dehydrocorticosterone, corticosterone, cortisol, dehydroepiandrosterone, pregnenolone and testosterone. The observation that both LC-PUFA and oxygen affected cyp11a expression and the levels of pregnenolone, provides evidence for the influence of both parameters on the rate-limiting point of steroid production, which can take >35 days to establish. The 3-way interaction between lipid, LC-PUFA and dissolved oxygen on the expression of steroid genes suggests that the chronic stressor of hypoxia may be over-riding dietary influences of the lipid and LC-PUFA, though the relative proportion of LC-PUFA within the lipid may play a role as well. In terms of fatty acid composition, low dietary lipid and high LC-PUFA increased the levels of total n-6 and n-3 PUFA in the liver, particularly EPA and DHA, and the head kidney to a lesser extent. In terms of immune pathways, hypoxia influenced cytokine gene expression (tgfb, il10), cellular repair gene expression (gr, hsp70 and hsp90) and eicosanoid levels (PGE2 and LTB5) in the head kidney, while dietary lipid and LC-PUFA influenced cytokines (ifnγ and il1β) and eicosanoids (PGE2 and LTB5). These findings demonstrate that feeding different levels of LC-PUFA significantly alters steroidogenesis and innate immune response in Atlantic salmon post-smolts with additional interactions from dietary lipid and hypoxia stress. Endocrine therapy is the first-line targeted adjuvant therapy for hormone-sensitive breast cancer. In view of the potential anticancer property of the omega-3 polyunsaturated fatty acid docosahexaenoic acid (DHA) together with chemotherapy in estrogen receptor alpha (ERα) positive mammary tumors, we have explored the regulation by estradiol of the fatty acid desaturation and elongation enzymes involved in DHA synthesis in the human breast cancer cell line MCF7, which expresses ERα but not ERβ. We demonstrate a robust up-regulation in the expression of the fatty acid elongases Elovl2 and Elovl5 upon estradiol stimulation in MCF7 cells, which was sustained for more than 24 hours. Exposure with the ER inhibitor tamoxifen abolished specifically the Elovl2 but not the Elovl5 expression. Similarly, knock-down of ERα eliminated almost fully the Elovl2 but not the Elovl5 expression. Furthermore, ERα binds to one specific ERE within the Elovl2 enhancer in a ligand dependent manner. Fish from the Amazon Basin are affected by oscillations in the river water volume, which influences the diet of animal species. This study was aimed at evaluating seasonal variations in lipid content, fatty acid composition and nutritional profiles of five fish species from the Amazon Basin. The lipid contents of all fish species were observed to be lower in flood periods than in drought periods; Brachyplatystoma flavicans showed the largest variation (6.75-15.43 %) between these periods, while Colossoma macropomum showed no significant difference ( p > 0.05). The fatty acid composition in the five fish species varied throughout seasonal periods; saturated fatty acid (SFA) contents decreased in flood periods, whereas polyunsaturated fatty acid (PUFA) contents significantly ( p < 0.05) increased for all the species in the same period. Leporinus friderici showed the highest content of α-linolenic acid, (LNA 14.86 mg g) and Colossoma macropomum presented the highest content of docosahexaenoic acid (DHA 26.13 mg g) in flood periods. Prochilodus nigricans showed the lowest content of arachidonic acid (AA) in both periods, while Brachyplatystoma flavicans showed the greatest amount of AA, 18.77 mg g in drought period and 22.10 mg g in flood period. All the fish species presented favorable indices of nutritional quality of lipid fraction, suggesting that consumption of these species could be considered beneficial to human health. Introduction: Arachidonic acid (AA) and docosahexaenoic acid (DHA) are important long-chain polyunsaturated fatty acids for neuronal and cognitive development and are ingredients of infant formulae that are recommended but there is no evidence based minimal supplementation level available. The aim of this analysis was to investigate the effect of supplemented AA and DHA on phospholipid metabolism. Methods: Plasma samples of a randomized, double-blind infant feeding trial were used for the analyses of phospholipid species by flow-injection mass spectrometry. Healthy term infants consumed isoenergetic formulae (intervention formula with equal amounts of AA and DHA—IF, control formula without additional AA and DHA—CF) from the first month of life until the age of 120 days. A group of breast milk (BM) -fed infants was followed as a reference. Results: The plasma profile detected in newborns was different from 4 month old infants, irrespective of study group. Most relevant changes were seen in higher level of LPC16:1, LPC20:4, PC32:1, PC34:1 and PC36:4 and lower level of LPC18:0, LPC18:2, PC32:2, PC36:2 and several ether-linked phosphatidylcholines in newborns. The sum of all AA and DHA species at 4 month old infants in the CF group showed level of 40% (AA) and 51% (DHA) of newborns. The supplemented amount of DHA resulted in phospholipid level comparable to BM infants, but AA phospholipids were lower than in BM infants. Interestingly, relative contribution of DHA was higher in ether-linked phosphatidylcholines in CF fed infants, but IF and BM fed infants showed higher overall ether-linked phosphatidylcholines levels. Conclusion: In conclusion, we have shown that infant plasma phospholipid profile changes remarkably from newborn over time and is dependent on the dietary fatty acid composition. Background: Whether and how n-3 and n-6 polyunsaturated fatty acids (PUFAs) are related to type 2 diabetes (T2D) is debated. Objectively measured plasma PUFAs can help to clarify these associations.Methods and Findings: Plasma phospholipid PUFAs were measured by gas chromatography among 12,132 incident T2D cases and 15,919 subcohort participants in the European Prospective Investigation into Cancer and Nutrition (EPIC)-InterAct study across eight European countries. Country-specific hazard ratios (HRs) were estimated using Prentice-weighted Cox regression and pooled by random-effects meta-analysis. We also systematically reviewed published prospective studies on circulating PUFAs and T2D risk and pooled the quantitative evidence for comparison with results from EPIC-InterAct. In EPIC-InterAct, among long-chain n-3 PUFAs, α-linolenic acid (ALA) was inversely associated with T2D (HR per standard deviation [SD] 0.93; 95% CI 0.88-0.98), but eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) were not significantly associated. Among n-6 PUFAs, linoleic acid (LA) (0.80; 95% CI 0.77-0.83) and eicosadienoic acid (EDA) (0.89; 95% CI 0.85-0.94) were inversely related, and arachidonic acid (AA) was not significantly associated, while significant positive associations were observed with γ-linolenic acid (GLA), dihomo-GLA, docosatetraenoic acid (DTA), and docosapentaenoic acid (n6-DPA), with HRs between 1.13 to 1.46 per SD. These findings from EPIC-InterAct were broadly similar to comparative findings from summary estimates from up to nine studies including between 71 to 2,499 T2D cases. Limitations included potential residual confounding and the inability to distinguish between dietary and metabolic influences on plasma phospholipid PUFAs.Conclusions: These large-scale findings suggest an important inverse association of circulating plant-origin n-3 PUFA (ALA) but no convincing association of marine-derived n3 PUFAs (EPA and DHA) with T2D. Moreover, they highlight that the most abundant n6-PUFA (LA) is inversely associated with T2D. Background: Long chain polyunsaturated fatty acids (LCPUFAs) including docosahexaenoic acid and arachidonic acid are suspected to play a key role in the pathogenesis of diabetes. LCPUFAs are known to be preferentially concentrated in specific phospholipids termed as plasmalogens. This study was aimed to highlight potential changes in the metabolism of phospholipids, and particularly plasmalogens, and LCPUFAs at various stages of diabetic retinopathy in humans. Methodology and Principal Findings: We performed lipidomic analyses on red blood cell membranes from controls and mainly type 2 diabetes mellitus patients with or without retinopathy. The fatty acid composition of erythrocytes was determined by gas chromatography and the phospholipid structure was determined by liquid chromatography equipped with an electrospray ionisation source and coupled with a tandem mass spectrometer (LC-ESI-MS/MS). A significant decrease in levels of docosahexaenoic acid and arachidonic acid in erythrocytes of diabetic patients with or without retinopathy was observed. The origin of this decrease was a loss of phosphatidyl-ethanolamine phospholipids esterified with these LCPUFAs. In diabetic patients without retinopathy, this change was balanced by an increase in the levels of several phosphatidyl-choline species. No influence of diabetes nor of diabetic retinopathy was observed on the concentrations of plasmalogen-type phospholipids. Conclusions and Significance: Diabetes and diabetic retinopathy were associated with a reduction of erythrocyte LCPUFAs in phosphatidyl-ethanolamines. The increase of the amounts of phosphatidyl-choline species in erythrocytes of diabetic patients without diabetic retinopathy might be a compensatory mechanism for the loss of LC-PUFA-rich phosphatidyl-ethanolamines. Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are the major n-3 polyunsaturated fatty acids (PUFAs) in fish oil that decrease the risk of prostate cancer. Tumor-associated macrophages (TAMs) are the main leukocytes of intratumoral infiltration, and increased TAMs correlates with poor prostate cancer prognosis. However, the mechanism of n-3 PUFAs on prostate cancer cell progression induced by TAMs is not well understood. In this study, we investigated the effects of EPA and DHA on modulating of migration and invasion of prostate cancer cells induced by TAMs-like M2-type macrophages. PC-3 prostate cancer cells were pretreated with EPA, DHA, or the peroxisome proliferator-activated receptor (PPAR)-γ antagonist, GW9662, before exposure to conditioned medium (CM). CM was derived from M2-polarized THP-1 macrophages. The migratory and invasive abilities of PC-3 cells were evaluated using a coculture system of M2-type macrophages and PC-3 cells. EPA/DHA administration decreased migration and invasion of PC-3 cells. The PPAR-γ DNA-binding activity and cytosolic inhibitory factor κBα (IκBα) protein expression increased while the nuclear factor (NF)-κB p65 transcriptional activity and nuclear NF-κB p65 protein level decreased in PC-3 cells incubated with CM in the presence of EPA/DHA. Further, EPA/DHA downregulated mRNA expressions of matrix metalloproteinase-9, cyclooxygenase-2, vascular endothelial growth factor, and macrophage colony-stimulating factor. Pretreatment with GW9662 abolished the favorable effects of EPA/DHA on PC-3 cells. These results indicate that EPA/DHA administration reduced migration, invasion and macrophage chemotaxis of PC-3 cells induced by TAM-like M2-type macrophages, which may partly be explained by activation of PPAR-γ and decreased NF-κB p65 transcriptional activity. The objectives of these experiment were to evaluate the effects of small amounts of fish oil blend supply in partial replacement of soybean oil on dry matter intake (DMI), lactation performance and milk fatty acid composition of ewes and also on performance of their lambs. Fifty Santa Inês lactating ewes (63±6kg of initial BW; mean±SD) were penned individually and used in a randomized complete block design with 10 blocks and 5 treatments. The oils were added to a basal diet that contained 700g/kg DM of concentrate and 300g/kg DM of forage (fresh sugarcane bagasse). The treatments were as follows: (1) basal diet without added oil (CONT); (2) 40g/kg DM of soybean oil (0FO); (3) 2.5g/kg DM of fish oil blend+37.5g/kg DM of soybean oil (25FO); (4) 5g/kg DM of fish oil blend+35g/kg DM of soybean oil (50FO); and (5) 7.5g/kg DM of fish oil blend+32.5g/kg DM of soybean oil (75FO). All diets were isonitrogen (140±4g/kg DM of CP). Once a week, from the second to the eighth week of lactation (weaning time), ewes were separated from their lambs, stimulated by a 6-IU intravenous oxytocin injection, and milked to empty the udder. After 3h, milk production was obtained after the same procedure. DMI (kg/d; % of BW and g/kg BW0.75) was higher for ewes fed the control diets vs. fat-supplemented. However, no effect was observed on DMI when fish oil blend inclusion in the diets increased. Nevertheless, milk production increased linearly when fish oil blend replaced soybean oil. As a consequence, the preweaning average daily gain (ADG) of lambs increased linearly. Milk fat concentration was similar for all diets. Milk protein and total solids concentrations decreased linearly when fish oil blend addition increased. Lactose milk concentration was higher for ewes fed the fat-supplemented diets vs. the control. Stearic acid, oleic acid, vaccenic acid, linoleic acid, linolenic acid and CLA cis-9, trans-11 showed higher concentrations in milk of animals fed diets containing soybean oil and fish oil blend compared to the control diet. Vaccenic acid, CLA trans-10, cis-12, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) increased linearly with fish oil blend inclusion. Small amounts of fish oil supplementation does not exert an additional effect on the concentration of CLA C18:2 cis-9, trans-11 as compered to exclusive use of soybean oil. However, the addition of 7.5g/kg DM of fish oil blend mixed with 32.5g/kg DM of soybean oil is recommended, because it increased the concentration of EPA, DHA and C18:1 trans-11 in the milk, as well increasing the performance of the ewes and their lambs. Dietary supplementation with natural chemoprotective agents is receiving considerable attention because of health benefits and lack of toxicity. In recent in vivo and in vitro experimental studies, diets rich in n-3 polyunsaturated fatty acids have been shown to provide significant anti-tumor action. In this investigation, the effects of control fatty acids (oleic acid (OA), linoleic acid (LA)) and n-3 PUFA, e.g., docosahexaenoic acid (DHA) on the uptake and metabolism of the carcinogenic polycyclic aromatic hydrocarbon, benzo[a]pyrene (BaP) was investigated in A549 cells, a human adenocarcinoma alveolar basal epithelial cell line. A549 cells activate BaP through the cytochrome P450 enzyme system to form reactive metabolites, a few of which covalently bind to DNA and proteins. Therefore, multiphoton microscopy spectral analysis combined with linear unmixing was used to identify the parent compound and BaP metabolites formed in cells, in the presence and absence of fatty acids. The relative abundance of select metabolites was associated with altered P450 activity as determined using ethoxyresorufin-O-deethylase activity in cells cultured in the presence of BSA-conjugated fatty acids. In addition, the parent compound within cellular membranes increases significantly in the presence of each of the fatty acids, with the greatest accumulation observed following DHA treatment. DHA treated cells exhibit significantly lower pyrene-like metabolites indicative of lower adducts including DNA adducts compared to control BSA, OA or LA treated cells. Further, DHA reduced the abundance of the proximate carcinogen BaP 7,8-dihydrodiol and the 3-hydroxybenzo[a]pyene metabolites compared to other treatments. The significant changes in BaP metabolites in DHA treated cells may be mediated by the effects on the physicochemical properties of the membrane known to affect enzyme activity related to phase I and phase II metabolism. Background: Arachidonic acid (AA; C20∶4 n-6) and docosahexaenoic acid (DHA; C22∶6 n-3) are important long-chain polyunsaturated fatty acids (LC-PUFA) in maintaining pancreatic beta-cell structure and function. Newborns of gestational diabetic mothers are more susceptible to the development of type 2 diabetes in adulthood. It is not known whether low circulating AA or DHA is involved in perinatally “programming” this susceptibility. This study aimed to assess whether circulating concentrations of AA, DHA and other fatty acids are associated with fetal insulin sensitivity or beta-cell function, and whether low circulating concentrations of AA or DHA are involved in compromised fetal insulin sensitivity in gestational diabetic pregnancies. Methods and Principal Findings: In a prospective singleton pregnancy cohort, maternal (32-35 weeks gestation) and cord plasma fatty acids were assessed in relation to surrogate indicators of fetal insulin sensitivity (cord plasma glucose-to-insulin ratio, proinsulin concentration) and beta-cell function (proinsulin-to-insulin ratio) in 108 mother-newborn pairs. Cord plasma DHA levels (in percentage of total fatty acids) were lower comparing newborns of gestational diabetic (n = 24) vs. non-diabetic pregnancies (2.9% vs. 3.5%, P = 0.01). Adjusting for gestational age at blood sampling, lower cord plasma DHA levels were associated with lower fetal insulin sensitivity (lower glucose-to-insulin ratio, r = 0.20, P = 0.036; higher proinsulin concentration, r = −0.37, P <0.0001). The associations remained after adjustment for maternal and newborn characteristics. Cord plasma saturated fatty acids C18∶0 and C20∶0 were negatively correlated with fetal insulin sensitivity, but their levels were not different between gestational diabetic and non-diabetic pregnancies. Cord plasma AA levels were not correlated with fetal insulin sensitivity. Conclusion: Low circulating DHA levels are associated with compromised fetal insulin sensitivity, and may be involved in perinatally “programming” the susceptibility to type 2 diabetes in the offspring of gestational diabetic mothers. Arachidonic (ARA), eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids are the most biologically active polyunsaturated fatty acids, but their biosyntheses in mammals are very limited. The biosynthesis of DHA is the most difficult, because this undergoes the Sprecher pathway–a further elongation step from docosapentaenoic acid (DPA), a Δ6-desaturase acting on a C24 fatty acid substrate followed by a peroxisomal chain shortening step. This paper reports the successful heterologous expression of two non-mammalian genes (with modification of codon usage), coding for Euglena gracilis Δ4-desaturase and Siganus canaliculatus Δ4-desaturase respectively, in mammalian cells (HEK293 cell line). Both of the Δ4-desaturases can efficiently function, directly converting DPA into DHA. Moreover, the cooperation of the E. gracilis Δ4-desaturase with C. elegans Δ15-desaturase (able to convert a number of n-6 PUFAs to their corresponding n-3 PUFAs) in transgenic HEK293 cells made a more desirable fatty acid composition – a drastically reduced n-6/n-3 PUFAs ratio and a high level of DHA as well as EPA and ARA. Our findings provide a basis for potential applications of the gene constructs for expression of Δ15/Δ4-desaturases in transgenic livestock to produce such a fatty acid profile in the related products, which certainly will bring benefit to human health. Recent studies have proposed that n-3 polyunsaturated fatty acids (n-3 PUFAs) have direct antioxidant and anti-inflammatory effects in vascular tissue, explaining their cardioprotective effects. However, the molecular mechanisms are not yet fully understood. We tested whether n-3 PUFAs showed antioxidant activity through the activation of nuclear factor erythroid 2-related factor 2 (Nrf2), a master transcriptional factor for antioxidant genes. C57BL/6 or Nrf2−/− mice were fed a fish-oil diet for 3 weeks. Fish-oil diet significantly increased the expression of heme oxygenase-1 (HO-1), and endothelium-dependent vasodilation in the aorta of C57BL/6 mice, but not in the Nrf2−/− mice. Furthermore, we observed that 4-hydroxy hexenal (4-HHE), an end-product of n-3 PUFA peroxidation, was significantly increased in the aorta of C57BL/6 mice, accompanied by intra-aortic predominant increase in docosahexaenoic acid (DHA) rather than that in eicosapentaenoic acid (EPA). Human umbilical vein endothelial cells were incubated with DHA or EPA. We found that DHA, but not EPA, markedly increased intracellular 4-HHE, and nuclear expression and DNA binding of Nrf2. Both DHA and 4-HHE also increased the expressions of Nrf2 target genes including HO-1, and the siRNA of Nrf2 abolished these effects. Furthermore, DHA prevented oxidant-induced cellular damage or reactive oxygen species production, and these effects were disappeared by an HO-1 inhibitor or the siRNA of Nrf2. Thus, we found protective effects of DHA through Nrf2 activation in vascular tissue, accompanied by intra-vascular increases in 4-HHE, which may explain the mechanism of the cardioprotective effects of DHA. Abstract: Dietary supplementation with ω-3 long chain fatty acids including docosahexaenoic acid (DHA) has increased in popularity in recent years and adequate DHA supplementation during pregnancy and early childhood is of clinical importance. Some evidence has been built for the neuro-cognitive benefits of supplementation with long chain polyunsaturated fatty acids (LCPUFA) such as DHA during pregnancy; however, recent data indicate that the anti-inflammatory properties may be of at least equal significance. Adequate DHA availability in the fetus/infant optimizes brain and retinal maturation in part by influencing neurotransmitter pathways. The anti-inflammatory properties of LCPUFA are largely mediated through modulation of signaling either directly through binding to receptors or through changes in lipid raft formation and receptor presentation. Our goal is to review the current findings on DHA supplementation, specifically in pregnancy and infant neurodevelopment, as a pharmacologic agent with both preventative and therapeutic value. Given the overall benefits of DHA, maternal and infant supplementation may improve neurological outcomes especially in vulernable populations. The best choice for feeding the newborn child is breast milk, and when it is not possible to breastfeed a baby, the breast milk is replaced by lacteal formula, which is why numerous studies focus their attention on the analysis of the diverse components of baby formula as well as functions in the breastfed baby. Fatty acids are components of great nutritional importance in the fetus and in the newborn child. Nowadays it is estimated that the fetus, during the last trimester of the gestational period and the newborn child, during the first six months of life, need a great contribution of araquidonic and docosahexaenoic acids, due to the fact that the speed of transformation of the predecessors to the hepatic level are not sufficient to cover the metabolic requirements of these fatty acids and it is the mother who contributes them through placental transport during gestation and her milk during lactation. The Organization of Food and Agriculture (FAO), the World Health Organization (WHO), the European Society of Gastroenterology, Hepatology and Pediatric Nutrition (ESPGHAN) and the American Academy of Pediatrics recommend that lacteal formula imitate the composition of the breast milk, especially in the fatty polyunsaturated acids (PUFA). Abstract: A total of 13 species of microalgae and 14 strains of cyanobacteria, collected directly in the Portuguese coast and lagoons, were characterized for their fatty acid contents, focusing on two with a market potential — i.e. eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA); and another already with alternative (yet somehow more expensive) natural sources — i.e. α-linolenic (ALA) acid. The purpose of this work was their eventual inclusion as additives in food or feed. ALA was the most abundant PUFA in Nannochloropsis sp. (0.616±0.081mgFA.Lculture −1.d−1), and EPA in Phaeodactylum tricornutum (0.148±0.013mgFA.Lculture −1.d−1); Pavlova lutheri was particularly rich in EPA (0.290±0.005mgFA.Lculture −1.d−1) and DHA (0.140±0.037mgFA.Lculture −1.d−1). Despite several previous reports on similar topics and encompassing some of our microalgal species, the wild nature of our strains accounts for the novelty of this work — in addition to the characterization of a few wild cyanobacteria. Eustigmatophyceae class was the best producer of ALA, while Prymnesiophyceae was the best for EPA and ALA. Lipids are a complex group of biomolecules whose precise functions remain poorly understood. As a result of this poor understanding, it is difficult to make mechanistically based recommendations for appropriate dietary intakes. It is equally difficult to develop methods that are capable of diagnosing functional impairments because of insufficiencies or excesses in particular fatty acids. Lipids are abundant building blocks of cellular membranes, supply components for lipid particle assembly and substrates for metabolic fuel, and provide a precursor pool for an astonishingly diverse range of signalling molecules. In each of these broad functions, the functional consequences of different structures of fatty acids are not fully understood. According to research on membrane functions through early evolution, docosahexaenoic acid provides two biophysical properties to membranes - accelerating the lateral motion of lipids and proteins within the plane of the membrane and simultaneously slowing the rate of diffusion/leakage of charged species across the plane of the membrane. The range of fatty acid structures used as substrates for assembly of either lipoproteins or milk fat globules is broad, yet the functional consequences of differences are not known. Different lipids signal into a remarkable range of biological processes. Saturated and monounsaturated fatty acids are becoming recognized as signal molecules in their own right. The complex composition of human milk lipids implies that diets with a diversity of fatty acids in complex lipid forms and structures is more beneficial than a narrow range of any particular group of fatty acids. Abstract: Objectives: Fatty acids, being multi-functional and partially diet-dependent, are crucial for health yet optimal dietary composition remains controversial. Previous work suggests that nutritionally-dependent populations live with significant fatty acid abnormalities despite abundant polyunsaturated fatty acid intake. Design and methods: An analysis of fasting plasma phospholipids, including plasmalogens, and total fatty acids was conducted on twelve tube-fed people receiving a uniform diet which meets current polyunsaturated fatty acid intake recommendations, specifically, linoleic acid as 8.1% of energy and alpha-linolenic acid as 1.3% of energy for at least two years. Results: Eicosapentaenoic- and docosahexaenoic acid-related phospholipids were low. In addition, C16:0- and C18:0-related plasmalogens, components of phospholipids, were low. Essential fatty acid deficiency as classically defined was not present. Conclusions: Based upon extensive clinical investigations in neuro-typical people, abnormalities of these key cell-membrane components may have undesirable clinical consequences. Epidemiological studies on the association between diet and cognitive function suggested a possible role of dietary fatty acids in cognitive decline. The aim of the present study was to examine whether intake of different types of fatty acids is associated with cognitive status. A cohort of 304 (127 men and 177 women) institutionalized elderly people, with a mean age of 75.3 ± 6.7 years, were studied. Subjects were evaluated for global cognitive functions (Mini-Mental State Examination [MMSE], Spanish version). Fatty acid intake was assessed with a semi-quantitative food frequency questionnaire. Intake of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) were found to be predictors of cognitive impairment as they were negatively associated with the MMSE score. In accordance with this, fish intake was inversely associated with cognitive impairment. On the contrary, the n-6/n-3 polyunsaturated fatty acid ratio was positively related to the MMSE score. These results could not be explained by differences in age, sex, education, smoking behaviour, inactivity, alcohol, institution or energy intake. We suggest that consumption of EPA and DHA should be encouraged for reducing the risk of cognitive impairment and subsequently disability in elderly people. [ABSTRACT FROM AUTHOR]

Diets supplemented with n-3 polyunsaturated fatty acids can promote lipid peroxidation and the propagation of oxygen radicals. These effects can be prevented by taurine, a functional ingredient with antioxidant properties. Here, we examined whether there is a correlation between transepithelial taurine transport, on the one hand, and membrane fatty acid composition and peroxidation in intestinal Caco-2 cells, on the other. Differentiated Caco-2 cells were maintained for 10 days, from the day of confluence, in control conditions or in a medium enriched with docosahexaenoic acid (DHA, 100 micromol/l), taurine (10 mmol/l) or DHA plus taurine. Incubation of the monolayers in a medium enriched with DHA increased the incorporation of this fatty acid into the brush-border membrane, at the expense of total n-6 fatty acids (C20:2n-6, C20:3n-6 and C22:4n-6). This was paralleled by increased membrane lipid peroxidation, which was partially limited by the addition of taurine. Transepithelial taurine transport was estimated from taurine uptake and efflux kinetic parameters at apical and basolateral domains. Cell incubation with DHA increased basolateral taurine uptake through an increase in V (max), whereas incubation with taurine downregulated basolateral uptake as occurred for apical taurine transporter. Moreover, addition of DHA reduced the apical downregulation effect exerted on taurine transport by taurine incubation. An eight-week feeding trial was conducted to evaluate the influence of an alternating linseed oil (LO) and fish oil (FO) feeding strategy in black seabream A. schlegelii (initial weight, 8.49 ± 0.01 g). Two isonitrogenous and isolipidic feeds were formulated to contain FO or LO, and used to enable three feeding strategy treatments termed FO (feeding the FO diet for 8 weeks), LO (feeding the LO diet for 8 weeks) and FO re-feeding (feeding LO for 4 weeks then FO for 4 weeks). Fish in the FO re-feeding and FO groups had significantly higher final body weights (FBW), weight gains (WG), specific growth rates (SGR) but lower muscle lipid content than fish in the LO group. In muscle, the FO re-feeding strategy restored eicosapentaenoic and docosahexaenoic acid contents to 82.67% and 83.84%, respectively, of those in fish fed the FO treatment. In addition, the FO re-feeding strategy significantly down-regulated the expression levels of genes related to long-chain polyunsaturated fatty acid (LC-PUFA) biosynthesis and lipogenesis, including fatty acyl desaturase (fads2), elongase of very long-chain fatty acids 4a (elovl4a) and sterol regulatory element-binding protein-1 (srebp-1) compared to fish fed the LO treatment. Similarly, the hepatic Srebp-1c concentration in fish in the FO re-feeding group was markedly lower than in fish in the LO treatment group. Expression of the lipid catabolism gene adipose triglyceride lipase (atgl) was up-regulated in fish in the FO re-feeding group compared with fish in the LO group. Overall, the present study indicated that a strategy of re-feeding FO following feeding LO for four-week promoted growth, mitigated lipid accumulation in muscle, and effectively restored muscle n-3 LC-PUFA levels in A. schlegelii. • Dietary LO had an adverse effect on growth in A. schlegelii. • Growth and feed utilization were recovered by FO re-feeding in A. schlegelii. • FO re-feeding could effectively restore DHA and EPA contents in muscle. • After FO re-feeding, DHA and EPA were better retained in muscle than in liver. • FO-refeeding reduced lipid accumulation by regulating gene expression. Lipids comprize from 50-60% of the structural matter of the brain and docosahexaenoic acid (C22:6, DHA) is the most important omega-3 long-chain polyunsaturated fatty acid in the brain phospholipids comprizing 25% of the total fatty acids of the grey matter. The majority of the DHA present in the human brain is incorporated during the brain growth spurt which starts at week 26 of gestation and imposes a high demand for the fatty acid until about 2 years of age. DHA is required during brain development when neuronal and glial differentiation and migration, and active myelination and synaptogenesis take place. The fatty acid must be incorporated into the brain lipids as preformed DHA because less than 5% of its precursor (alpha linolenic acid, LNA) is converted to DHA. The human foetus has a limited ability to synthesize DHA from LNA, and therefore it must be largely supplied from maternal sources. Maternal DHA available for foetal nutrition can be provided from three main sources: adipose tissue, which is the main reservoir for the fatty acid; through biosynthesis from the precursor LNA, which occurs mainly in the liver; and as preformed DHA from dietary sources. In the postnatal period DHA is provided by the mother to the newborn through milk secretion. Western nutrition provides low LNA and DHA and Expert Nutrition Committees suggest that mothers should receive DHA supplementation during pregnancy and lactation. At present DHA supplementation can be provided from different sources: as purified free DHA, as an ethyl ester derivative, extracted from single-cell algae oils, from egg yolk phospholipids, or in the form of sn-2 DHA monoacylglycerol. In this review we revise and discuss the evidence of DHA requirements for the newborn, the need for maternal supplementation during pregnancy and nursing, and the alternatives at present for providing DHA supplementation. Abstract: A preparative reversed-phase (RP; C18) high-performance liquid chromatography (HPLC) method with gradient elution using acetonitrile (MeCN)–chloroform (CHCl3) (or dichloromethane (DCM)) and evaporative light-scattering detection (ELSD) with automatic multiple injection and fraction collection was used to purify milligram quantities of microalgal polyunsaturated fatty acids (PUFA), separated as methyl esters (ME). PUFA-ME purified included methyl esters of docosahexaenoic acid (DHA; 22:6(n-3)), eicosapentaenoic acid (EPA; 20:5(n-3)) and the unusual very long-chain (C28) highly unsaturated fatty acid (VLC-HUFA), octacosaoctaenoic acid [28:8(n-3)(4, 7, 10, 13, 16, 19, 22, 25)] from the marine dinoflagellate Scrippsiella sp. CS-295/c. Other PUFA purified from various microalgae using this RP-HPLC method to greater than 95% purity included 16:3(n-4), 16:4(n-3), 16:4(n-1) and 18:5(n-3). The number of injections required was variable and depended on the abundance of the desired PUFA-ME, and resolution from closely eluting PUFA-ME, which determined the maximum loading. The purity of these fatty acids was determined by electron impact (EI) GC–MS and the chain length and location of double bonds was determined by EI GC–MS of 4,4-dimethyl oxazoline (DMOX) derivatives formed using a low temperature method. Advantages over silver-ion HPLC for purifying PUFA-ME is that separation occurs according to chain length as well as degree of unsaturation enabling separation of PUFA-ME with the same degree of unsaturation but different chain length (i.e. between 18:5(n-3) and 20:5(n-3)). In addition, PUFA-ME are not strongly adsorbed, but elute earlier than their more saturated corresponding FAME of the same chain length. This method is robust, simple, and requires only a short re-equilibration time. It is a useful tool for preparing milligram quantities of pure PUFA-ME for bioactive screening (as free fatty acids), although many multiple injections may be required for minor PUFA-ME. It also enabled dose–response and structure–activity studies to be carried out. It can be used for the enrichment of low levels of VLC-HUFA-ME to facilitate elucidation of their chemical structure and so is a useful adjunct to EI GC–MS of DMOX derivatives and other techniques such as NMR, which requires milligram quantities of purified compounds. Abstract: Lipid composition was determined for hydrothermal vent species collected by the Deep Submergence Vehicle ALVIN from chimneys at 2,500 m depth on the East Pacific Rise. These are the first lipid biomarker studies for most of these species. Lipid content was low and dominated by polar lipid in the vestimentiferan tubeworm Riftia pachyptila, mussels Bathymodiolus sp. and limpets Lepetodrilus spp. The galatheid (Munidopsis subsquamosa) and most brachyuran adult (Bythograea thermydron) crabs were characterized by higher storage lipid (triacylglycerol). Total polyunsaturated fatty acids were similar in R. pachyptila plume and body, but higher in the posterior part of the soft body, which had more docosahexaenoic acid (2–5% of total FA) compared to the anterior and plume (≤0.3%). Two sulphur-oxidizing bacterial markers, 16:1(n −7)c and 18:1(n −7)c, were high in R. pachyptila and mussel (up to 23%), but lower in both crab species (4–17%). R. pachyptila had greater nonmethylene interrupted diunsaturated fatty acids (8–13%) than all other species (2–8%). R. pachyptila may desaturate and elongate 18:1(n −7)c to obtain essential polyunsaturated fatty acids 20:5(n −3) and 20:4(n −6). The sterol composition of R. pachyptila included similar amounts of cholesterol and desmosterol, whereas the other species had a more diverse sterol composition. These differences in lipids, fatty acids and sterols reflect diverse nutritional strategies and possibly temperature regimes in these species. [Copyright &y& Elsevier]

During avian development, lipoproteins derived from yolk lipid are assembled in the yolk sac membrane (YSM) for secretion into the embryonic circulation. To investigate how yolk polyunsaturated fatty acids, essential for the development of certain tissues, are distributed among the lipid classes of the lipoproteins, pieces of YSM were incubated in vitro with [14C]arachidonic and [14C]docosahexaenoic acids (DHA). There was a marked difference in the partitioning of these two precursors among the lipid classes of the tissue. Of the radioactivity incorporated into total lipid from [14C]-arachidonic acid during 1 h of incubation, 67.3% was esterified as phospholipid and 29.5% as triacylglycerol. In contrast, only 14.6% of the label incorporated from [14C]-DHA was esterified as phospholipid, whereas 73.2% was recovered in triacylglycerol. This pattern of differential partitioning was observed at all time points and across a 20-fold range of fatty acid concentrations. There was no evidence for conversion of the radioactive arachidonic and DHAs to other fatty acids prior to incorporation into tissue lipids. It is suggested that the selective incorporation of yolk-derived DHA into the triacylglycerol of secreted lipoproteins represents part of a mechanism for directing this polyunsaturate to particular embryonic tissues. The Northeast Atlantic mackerel (Scomber scombrus) is a zooplanktivorous fish with its main summer feeding grounds in the waters around Iceland. The zooplankton in the stomachs of the caught fish causes several problems during processing due to the high enzyme activity of the zooplankton. The aim of the study was to evaluate the chemical characteristics of zooplankton that accompanies mackerel as a side-catch and stomach fullness as affected by catching year, season, catching zone, and catching method over three mackerel seasons from 2016 to 2018. Species identification by the fatty acid tropic marker method (FATM) was also applied within the zooplankton rich side-stream. FATM analysis indicated that the majority of the zooplankton mass belonged to Calanus finmarchicus. The lipid composition of the zooplankton rich side-stream varied between years but was rich in monounsaturated, as well as polyunsaturated fatty acids (PUFA), such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). The results suggest that the zooplankton rich side-stream from mackerel caught in Icelandic waters has the potential for further sustainable processing into valuable nutrients. Round scad (Decapterus maruadsi) is rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). However, there is no comprehensive information covering its lipidomic profile. In this study, lipid profiles including fatty acid composition, distribution and detailed structure information were explored using gas chromatography-flame ionization detector (GC-FID) and ultra-high-performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry (UPLC-Q-Exactive Orbitrap-MS). The results showed that triacylglycerol (TG) was the dominant class, and the EPA and DHA were concentrated as glycerophospholipid (GP). The saturated fatty acids and monounsaturated fatty acids were mainly esterified in positions 1 and 3 of TG and sn -1-position of GP, while the polyunsaturated fatty acids (PUFA) were mainly esterified in sn -2-position of TG and GP. A total of 1282 species from six classes (21 subclasses) including glycerolipids (GL), sphingolipids (SP), GP, fatty acyls (FA), saccharolipids (SL) and prenol lipids (PR) were identified. Several molecular species were characterized with PUFA, especially EPA and DHA in GP. Considering the superior fatty acids composition and distribution of round scad, it is deserved for further exploitation of its marine lipid source on account of the healthy and nutritional functions. The consumption of seafood has considerably increased over recent decades; however, as wild seafood stocks are limited, the cultured ones represent a possible valuable alternative. The purpose of this study was to compare wild and cultured scallops, Flexopecten glaber, on the basis of their marketability indices and biochemical characteristics. Wild and cultured specimens were harvested from the Ionian Sea (the Central Mediterranean Sea). Protein and lipid were significantly different between scallops, with the values of protein of 8.50 and 11.6 g/100 g and lipid of 1.45 and 1.70 g/100 g for wild and cultured scallops, respectively. Regarding fatty acids (FAs), statistical differences were also detected. The cultured species showed significantly (p < 0.05) higher polyunsaturated fatty acids (PUFAs) than its wild counterpart. Eicosapentaenoic (EPA, 20:5 n3) and docosahexaenoic acid (DHA, 22:6:3) were the major polyunsaturated fatty acids, although only DHA showed significant differences between wild and culture scallops (p < 0.05). The ratio of n3/n6 PUFA showed high values, with 2.7 and 3.1 for wild and cultured scallops, respectively. The atherogenic and thrombogenic indices and hypocholesterolemic/hypercholesterolemic fatty acid ratio indicated an import role in human diet. Long-chain omega-3 polyunsaturated fatty acids (LC-PUFA) essential for human nutrition are mostly obtained from wild-caught fish. To sustain the LC-PUFA supply from natural populations, one needs to know how environmental and intrinsic factors affect fish fatty acid (FA) profiles and contents. We studied seven Salmoniformes species from two arctic lakes. We aimed to estimate differences in the FA composition of total lipids and two major lipid classes, polar lipids (PL) and triacylglycerols (TAG), among the species and to evaluate LC-PUFA contents corresponding to PL and TAG in muscles. Fatty acid profiles of PL and TAG in all species were characterized by the prevalence of omega-3 LC-PUFA and C16-C18 monoenoic FA, respectively. Fish with similar feeding spectra were identified similarly in multivariate analyses of total lipids, TAG and PL, due to differences in levels of mostly the same FA. Thus, the suitability of both TAG and total lipids for the identification of the feeding spectra of fish was confirmed. All species had similar content of LC-PUFA esterified as PL, 1.9–3.5 mg g−1, while the content of the TAG form strongly varied, from 0.9 to 9.8 mg g−1.
